Functional genomics of Arabidopsis thaliana heat stress transcription factors with detailed study on the transcriptional regulation and downstream target genes of HsfA2

  • 21 Hsfs belonging to classes A, B and C were identified in Arabidopsis following the sequencing of its genome. 1.) Cloning of full length and CTD chimeric constructs followed by transient reporter assays in tobacco protoplast using GUS fusion constructs of the promoters of Hsp17.4-CI, synthetic (HSE9) and APX2 showed Hsfs A1a, A1b, A1d, A1e, A2, A3 and A9 to be active. CTDs of Hsfs A7a, A7b and HsfC1 had activity but they showed poor DNA binding in reporter assays. Hsfs A1a, A1b, A1d, A1e, A2 and A3 were able to induce the expression of endogenous Hsps in tomato protoplasts. Interesting differences in promoter selectivity were observed for several Hsfs. 2.) RT-PCR and microarray analysis showed the Hsfs to be differentially expressed depending on tissue, abiotic and biotic stress, hormone and developmental s ge. Interesting patterns of coexpressed Hsfs were observed under different stresses and developmental stages. 3.) HsfA1b was found to be active on the plasmid borne PHsf:GUS reporters of Hsfs A1d, A2, A4a, A7b and B4 when tested in tobacco mesophyll protoplasts. Hsfs A1d, A2, A4a, A7b and B4 when tested in tobacco mesophyll protplasts. HsfA2 was inactive on PHsfA:GUS. HsfB1 showed repression of endogenous activity on several PHsf:GUS reporter constructs. 4.) The transcriptional regulation under heat stress and promoter organization of HsfA2 and FtSH4 (a metalloprotease gene oriented in a head to head fashion with HsfA2 in the Arabidopsis genome, sharing a common promoter region) was studied. The transcripts of FtSH4 and HsfA2 coaccumulated under heat stress. HsfA1b was active on PHsfA2:GUS and PFtSH4:GUS. Hsf binding sites on the intergenic region were determined using promoter deletion constructs in tobacco and Arabidopsis protoplasts. A bidirectional regulation of HsfA2 and FtSH4 by HsfA1b was observed in tobacco protoplast. 5.) Microarray analysis of a HsfA2 T-DNA insertion line vs. wild type Col-0 under heat stress conditions led to identification of a subset of target genes to be severely affected in the absence of HsfA2. Apart from several Hsps (heat stressproteins) and APX2 (Ascorbate peroxidase 2, oxidative stress scavenger), several other unknown genes are affected. APX2 was the most severely affected among them. HsfA2 was able to induce the transcription from its target gene promoters in fusion to GUS in transient reporter assays in tobacco protoplast. The HSE cluster to which HsfA2 binds on the APX2 promoter was also mapped by the same technique. The direct binding of HsfA2 to the promoter of selected target genes in the Arabidopsis genome was also demonstrated by chromatin immunoprecipitation studies.
  • Durch die Sequenzierung des Genoms von Arabidopsis konnten 21 Hsfs identifiziert werden, die in die Klassen A, B und C eingeteilt wurden. 1.) Die Hsfs wurden als Volllängenkonstrukte und CTD-Chimäre kloniert und mit Hilfe von transienten Reporterassays in Tabakprotoplasten mit Promotoren von Hsp17.4-CI, HSE9 und APX2 in Fusion an GUS auf ihre Aktivität als Transkriptionsfaktor hin überprüft. Es konnte gezeigt werden, dass Hsf A1a, A1b, A1d, A1e, A2, A3 und A9 aktiv waren. CTDs von Hsf A7a, A7b und HsfC1 hatten ein Transaktivierungspotential, aber die Hsfs selbst zeigten nur schwache DNA-Bindung. Außerdem waren Hsf A1a, A1b, A1d, A1e, A2 und A3 in der Lage, die Expression endogener Hsps in Tomatenprotoplasten zu induzieren. Interessante Unterschiede konnten bei der Promotorselektivität beobachtet werden. 2.) RT-PCR- und Microarray-Analysen zeigten, dass Hsfs abhängig von Gewebe, abiotischem und biotischem Stress, Hormonen und pflanzlichen Entwicklungsstadien unterschiedlich exprimiert werden. Es wurden auch interessante Muster von Koexpression verschiedener Hsfs bei unterschiedlichen Stressoren und Entwicklungsstadien gefunden. 3.) Es zeigte sich, dass HsfA1b bei Tests in Tabakprotoplasten an den Plasmid Promotor-GUS Konstrukten (PHsf:GUS) der Hsfs A1d, A2, A4a, A7b und B4 aktiv war. HsfA2 war an seinem eigenen Promotor inaktiv, während HsfB1 eine Repression endogener Aktivität an diversen Hsf Promotoren bewirkte. 4.) Die Transkriptionsregulation unter Hitzestress und die Promotororganisation von HsfA2 und FtSH4 (einer Metalloprotease, die im Genom in „head to head”-Konfiguration zu HsfA2 angeordnet ist) wurden näher untersucht. Dabei konnte gezeigt werden, dass die Transkripte von FtSH4 und HsfA2 unter Hitzestress koakkumulieren. HsfA1b war an den Promotoren beider Gene aktiv. Hsf-Bindestellen in der Region zwischen beiden Genen wurden mit Hilfe von Promotordeletionskonstrukten in Tabak- und Arabidopsis-Protoplastenassays bestimmt. Es konnte eine bidirektionale Regulation von HsfA2 und FtSH4 durch HsfA1b in Tabakprotoplasten nachgewiesen werden. 5.) Durch Microarray-Analysen mit hitzegestressten Blättern einer HsfA2-T-DNA-Insertionslinien von Arabidopsis im Vergleich zum Wildtyp konnte eine Untergruppe von Zielgenen identifiziert werden, die durch Abwesenheit von HsfA2 in ihrer Expression stark beeinflusst waren. Neben einigen Hsps und APX2 (Ascorbat Peroxidase 2, welche als Radikalfänger bei oxidativem Stress dient) waren auch mehrere Gene unbekannter Funktion betroffen. Unter ihnen war APX2 das am stärksten beeinflusste Gen. Dies wurde durch transiente Reporter-Assays validiert, in welchen HsfA2 entsprechende Promotor-GUS Konstrukte der potentiellen Zielgene induzieren konnte. Das relevante HSE-Cluster als Bindestelle für HsfA2 im APX2 Promotor wurde durch Deletionsanalysen bestimmt. Schliesslich konnte die Bindung von HsfA2 an die Promotoren seiner Zielgene im Genom von Arabidopsis mittels Chromatin-Immunpräzipitation gezeigt werden.
