Im Zentrum dieser Arbeit steht die Diagnostik eines Wasserstoff-Theta-Pinch-Plasmas hinsichtlich der integrierten Elektronen- und Neutralgasdichte mittels Zweifarben Interferometrie. Die integrierte Elektronen- und Neutralgasdichte sind essenzielle Größen, aus welchen sich die Ratenkoeffizienten der Ionisation und Rekombination bei einer Plasma-Ionenstrahl-Wechselwirkung bestimmen lassen.
Ein Theta-Pinch-Plasma ist ein induktiv gezündetes Plasma, wobei das zur Zündung notwendige elektrische Feld durch ein magnetisches Wechselfeld generiert wird. Das induzierte, azimutale elektrische Feld beschleunigt freie Elektronen im Arbeitsgas, welches durch Stoßionisation in den Plasmazustand gebracht wird. Der azimutale Plasmastrom erzeugt einen radialen magnetischen Druckgradienten, der das Plasma komprimiert. Da in axialer Richtung keine Kompressionskraft wirkt, weicht das Plasma einer weiteren Kompression aus, wodurch es zu einer axialen Expansion des Plasmas kommt. Die Expansion erzeugt eine Ionisationswelle im kalten Restgas und es wird eine lange, hoch ionisierte Plasmasäule gebildet.
Dieser hochdynamische Prozess ist mit einem Mach-Zehnder-Interferometer bei der Verwendung von zwei verschiedenen Versionen des Theta-Pinchs zeitaufgelöst untersucht worden. Der Unterschied dieser Versionen liegt in der Geometrie und Induktivität der Spulen, wobei zum einen eine zylindrische und zum anderen eine sphärische Spule eingesetzt worden ist. Das grundlegende Messprinzip beruht darauf, dass das Plasma einen Brechungsindex besitzt, welcher von den Dichten der im Plasma enthaltenen Teilchenspezies abhängt. In einem Wasserstoffplasmas sind dies der Beitrag der freien Elektronen und der des Neutralgases, wodurch ein Zweifarben-Interferometer eingesetzt wird. Um eine von den Laserintensitäten unabhängige Messung zu ermöglichen, wird das heterodyne Verfahren benutzt, bei dem die Referenzstrahlen beider Wellenlängen jeweils mit einem akusto-optischen Modulator frequenzverschoben werden. Durch einen Vergleich mit einem stationären Referenzsignal mittels eines I/Q-Demodulators wird die interferometrische Phasenverschiebung aus dem Messsignal extrahiert.
Mit diesem diagnostischen Verfahren ist die integrierte Elektronen- und Neutralgasdichte des Theta-Pinch-Plasmas bei Variation des Arbeitsdrucks und der Ladespannung der Kondensatorbank untersucht worden. Mit der zylindrischen Experimentversion ist eine optimale Kombination aus integrierter Elektronendichte und effektivem Ionisationsgrad η von (1,45 ± 0,04) · 1018 cm−2 bei η = (0,826 ± 0,022) bei einem Arbeitsdruck von 20 Pa und einer Ladespannung von 16 kV ermittelt worden. Dagegen beträgt die optimale Kombination bei einem Arbeitsdruck von 20 Pa und einer Ladespannung von 18 kV bei Verwendung der sphärischen Experimentversion lediglich (1,23 ± 0,03) · 1018 cm−2 bei η = (0,699 ± 0,019).
Des Weiteren ist bei beiden Experimentversionen nachgewiesen worden, dass die integrierte Elektronendichte dem oszillierenden Strom folgend periodische lokale Maxima zeigt, welche zeitlich mit signifikanten Einbrüchen in der integrierten Neutralgasdichte zusammenfallen. Diese Einbrüche werden durch die axiale Expansion des Plasmas und der damit verbundenen Ionisationswelle im Restgas erzeugt. Neben diesem zentralen Teil dieser Arbeit ist eine lasergestützte polarimetrische Diagnostik durchgeführt worden, mit der die longitudinale Komponente der magnetischen Flussdichte der Theta-Pinch-Spulen zeit- und ortsaufgelöst bestimmt worden ist. Als Messprinzip ist der Faraday-Effekt eines magneto-optischen TGGKristalls verwendet worden.
Vor der polarimetrischen Diagnostik ist der TGG-Kristall bezüglich seiner Verdet- Konstante kalibriert worden, wobei ein Wert von V = (−149,7 ± 6,4) rad/Tm gemessen worden ist. Die ortsaufgelöste polarimetrische Diagnostik ist durch einen Seilzug ermöglicht worden, mit dem der TGG-Kristall auf einem Schlitten an unterschiedliche Positionen entlang der Spulenachse gefahren werden konnte. An den jeweiligen Messpunkten ist für beide Experimentversionen die magnetische Flussdichte für verschiedene Ladespannungen zeitaufgelöst bestimmt worden. Als Messverfahren ist dabei das Δ/Σ-Verfahren eingesetzt worden, mit dem sich eine intensitätsunabhängige Messung erzielen ließ.
Die ortsaufgelösten Messergebnisse fallen gegenüber Simulationen allerdings zu niedrig aus. Bei der zylindrischen Spule betragen die Abweichungen im Spulenzentrum circa 14 - 16% und bei der sphärischen Spule in etwa 16 - 18%. Bei einer Normierung der Messwerte und der simulierten Werte auf den jeweiligen Wert im Zentrum ist dagegen innerhalb der Fehler eine völlige Übereinstimmung zwischen den Messwerten und der Simulation für die zylindrische Spule erzielt worden. Als Ursache der negativen Abweichungen wird die Hysterese des TGG-Kristalls diskutiert. Es zeigt sich insbesondere zu Beginn der Entladung eine zeitliche Verzögerung der gemessenen magnetischen Flussdichte gegenüber dem Strom, die in der Umgebung des Stromnulldurchgangs besonders stark ausgeprägt ist.
We performed an experiment under long-term microgravity conditions aboard the International Space Station (ISS) to obtain information on the energetics and experimental constraints required for the formation of chondrules in the solar nebula by ’nebular lightning’. As a simplified model system, we exposed porous forsterite (Mg2 SiO4) dust particles to high-energetic arc discharges. The characterization of the samples after their return by synchrotron microtomography and scanning electron microscopy revealed that aggregates had formed, consisting of several fused Mg2SiO4 particles. The partial melting and fusing of Mg2SiO4 dust particles under microgravity conditions leads to a strong reduction of their porosity. The experimental outcomes vary strongly in their appearance from small spherical melt-droplets (∅≈90 µm) to bigger and irregularly shaped aggregates (∅≈350 µm). Our results provided new constraints with respect to energetic aspects of chondrule formation and a roadmap for future and more complex experiments on Earth and in microgravity conditions.
In order to gain further insights into early solar system aggregation processes, we carried out an experiment on board the International Space Station, which allowed us to study the behavior of dust particles exposed to electric arc discharges under long-term microgravity. The experiment led to the formation of robust, elongated, fluffy aggregates, which were studied by scanning electron microscopy, electron backscatter diffraction, and synchrotron micro-computed tomography. The morphologies of these aggregates strongly resemble the typical shapes of fractal fluffy-type calcium-aluminum-rich inclusions (CAIs). We conclude that a small amount of melting could have supplied the required stability for such fractal structures to have survived transportation and aggregation to and compaction within planetesimals. Other aggregates produced in our experiment have a massy morphology and contain relict grains, likely resulting from the collision of grains with different degrees of melting, also observed in some natural CAIs. Some particles are surrounded by igneous rims, which remind in thickness and crystal orientation of Wark–Lovering rims; another aggregate shows similarities to disk-shaped CAIs. These results imply that a (flash-)heating event with subsequent aggregation could have been involved in the formation of different morphological CAI characteristics.
OXA-48-like carbapenemases are among the most frequent carbapenemases in Gram-negative Enterobacterales worldwide with the highest prevalence in the Middle East, North Africa and Europe. Here, we investigated the so far uncharacterized carbapenemase OXA-484 from a clinical E. coli isolate belonging to the high-risk clone ST410 regarding antibiotic resistance pattern, horizontal gene transfer (HGT) and genetic support. OXA-484 differs by the amino acid substitution 214G compared to the most closely related variants OXA-181 (214R) and OXA-232 (214S). The blaOXA–484 was carried on a self-transmissible 51.5 kb IncX3 plasmid (pOXA-484) showing high sequence similarity with plasmids harboring blaOXA–181. Intraspecies and intergenus HGT of pOXA-484 to different recipients occurred at low frequencies of 1.4 × 10–7 to 2.1 × 10–6. OXA-484 increased MICs of temocillin and carbapenems similar to OXA-232 and OXA-244, but lower compared with OXA-48 and OXA-181. Hence, OXA-484 combines properties of OXA-181-like plasmid support and transferability as well as β-lactamase activity of OXA-232.
Acinetobacter baumannii is a Gram-negative pathogen that causes a multitude of nosocomial infections. The Acinetobacter trimeric autotransporter adhesin (Ata) belongs to the superfamily of trimeric autotransporter adhesins which are important virulence factors in many Gram-negative species. Phylogenetic profiling revealed that ata is present in 78% of all sequenced A. baumannii isolates but only in 2% of the closely related species A. calcoaceticus and A. pittii. Employing a markerless ata deletion mutant of A. baumannii ATCC 19606 we show that adhesion to and invasion into human endothelial and epithelial cells depend on Ata. Infection of primary human umbilical cord vein endothelial cells (HUVECs) with A. baumannii led to the secretion of interleukin (IL)-6 and IL-8 in a time- and Ata-dependent manner. Furthermore, infection of HUVECs by WT A. baumannii was associated with higher rates of apoptosis via activation of caspases-3 and caspase-7, but not necrosis, in comparison to ∆ata. Ata deletion mutants were furthermore attenuated in their ability to kill larvae of Galleria mellonella and to survive in larvae when injected at sublethal doses. This indicates that Ata is an important multifunctional virulence factor in A. baumannii that mediates adhesion and invasion, induces apoptosis and contributes to pathogenicity in vivo.
OXA-48 is the most common carbapenemase in Enterobacterales in Germany and one of the most frequent carbapenemases worldwide. Several reports have associated blaOXA–48 with a virulent host phenotype. To challenge this hypothesis, 35 OXA-48-producing clinical isolates of Escherichia coli (n = 15) and Klebsiella pneumoniae (n = 20) were studied in vitro, in vivo employing the Galleria mellonella infection model and by whole-genome sequencing. Clinical isolates belonged to 7 different sequence types (STs) in E. coli and 12 different STs in K. pneumoniae. In 26/35 isolates blaOXA–48 was located on a 63 kb IncL plasmid. Horizontal gene transfer (HGT) to E. coli J53 was high in isolates with the 63 kb IncL plasmid (transconjugation frequency: ∼103/donor) but low in isolates with non-IncL plasmids (<10–6/donor). Several clinical isolates were both highly cytotoxic against human cells and virulent in vivo. However, 63 kb IncL transconjugants generated from these highly virulent isolates were not more cytotoxic or virulent when compared to the recipient strain. Additionally, no genes associated with virulence were detected by in silico analysis of OXA-48 plasmids. The 63 kb plasmid was highly stable and did not impair growth or fitness in E. coli J53. In conclusion, OXA-48 clinical isolates in Germany are diverse but typically harbor the same 63 kb IncL plasmid which has been reported worldwide. We demonstrate that this 63 kb IncL plasmid has a low fitness burden, high plasmid stability and can be transferred by highly efficient HGT which is likely the cause of the rapid dissemination of OXA-48 rather than the expansion of a single clone or gain of virulence.
The solution structure of the lantibiotic immunity protein NisI and its interactions with nisin

Many Gram-positive bacteria produce lantibiotics, genetically encoded and posttranslationally modified peptide antibiotics, which inhibit the growth of other Gram-positive bacteria. To protect themselves against their own lantibiotics these bacteria express a variety of immunity proteins including the LanI lipoproteins. The structural and mechanistic basis for LanI-mediated lantibiotic immunity is not yet understood. Lactococcus lactis produces the lantibiotic nisin, which is widely used as a food preservative. Its LanI protein NisI provides immunity against nisin but not against structurally very similar lantibiotics from other species such as subtilin from Bacillus subtilis. To understand the structural basis for LanI-mediated immunity and their specificity we investigated the structure of NisI. We found that NisI is a two-domain protein. Surprisingly, each of the two NisI domains has the same structure as the LanI protein from B. subtilis, SpaI, despite the lack of significant sequence homology. The two NisI domains and SpaI differ strongly in their surface properties and function. Additionally, SpaI-mediated lantibiotic immunity depends on the presence of a basic unstructured N-terminal region that tethers SpaI to the membrane. Such a region is absent from NisI. Instead, the N-terminal domain of NisI interacts with membranes but not with nisin. In contrast, the C-terminal domain specifically binds nisin and modulates the membrane affinity of the N-terminal domain. Thus, our results reveal an unexpected structural relationship between NisI and SpaI and shed light on the structural basis for LanI mediated lantibiotic immunity.
The present contribution is meant as an approach to the debate mentioned in the heading. It is important to find ways, to make on one side the learning and the cultivation of the German language attractive for the young people and on the other side to regain again the peoples’ conscious, here and there, for the centuries-long German culture and history. Teachers of both sexes as well as the students who are going to become teachers are the future multipliers par excellence. Due to a variety of integral pedagogical and linguistic didactical concepts –meant simultaneously for transmitting information, meeting, exchange and networking – there have to be found innovative ideas, precise proposals have to be worked out, which both also strengthen the social relevance of the local Institutions and German Departments respectively Faculties, which qualify teachers for the German language and providing in this way a contribution to the harmonization of the civil society.
