In der vorliegenden Studie wird der Enzymun-Test®-PSA der zweiten Generation mit zwei monoklonalen Antikörpern am ES 700 Analyzer mit drei radioimmunologischen und einem fluorometrischen Verfahren verglichen. Für den Enzymun-Test®-PSA würden an 150 gesunden Probanden (100 Männer und 50 Frauen) die Referenzbereiche ermittelt. Bei den Männern lag die 95 % Perzentile bei 3,8 ng/ml, bei den Frauen bei 0,46 ng/ml. Außerdem wurden mit denselben Methode 50 Patienten mit benignen Prostataerkrankungen und 50 Patienten mit einem Prostatakarzinom untersucht. Der Korrelationskoeffizienl zwischen dem Enzymun-Test®-PSA und der radioimmunologischen Methode Tandem®-R-PSA liegt bei r = 0,99, die analytische Sensitivität von Enzymun-Rest®-R-PSA liegt bei 0,05 ng/ml. Die Stabilität der Serumproben zwischen +2 °C und +8 °C ist über einen Tag garantiert. Über diesen Zeitraum hinaus sollten Serumproben bei —20 °C gelagert werden.
New reactive coenzyme analogues for affinity labeling of NAD+ and NADP+ dependent dehydrogenases

Reactive coenzyme analogues ω-(3-diazoniumpyridinium)alkyl adenosine diphosphate were prepared by reaction of ω-(3-aminopyridinium)alkyl adenosine diphosphate with nitrous acid. In these compounds the nicotinamide ribose is substituted by hydrocarbon chains of varied lengths (n-ethyl to n-pentyl). The diazonium compounds are very unstable and decompose rapidly at room temperature. They show a better stability at 0 °C. L actate and alcohol dehydrogenase do not react with any of the analogues. Glyceraldehyde-3-phosphate dehydrogenase reacts rapidly with the diazonium pentyl compound. Decreasing the length of the alkyl chain significantly decreases the inactivation velocity. 3α,20β-Hydroxysteroid dehydrogenase reacts at 0 °C with the ethyl homologue and slowly with the propyl compound. The butyl-and pentyl analogues do not inactivate at 0 °C. Tests with 14C -labeled 2-(3-diazoniumpyridinium)ethyl adenosine diphosphate show that complete loss of enzyme activity results after incorporation of 2 moles of inactivator into 1 mole of tetrameric enzyme. 4-(3-Acetylpyridinium)butyl 2 ′-phospho-adenosine diphosphate, a structural analogue of NADP +, was prepared by condensation of adenosine-2,3-cyclophospho-5′-phosphomorpholidate with (3-acetylpyridinium)butyl phosphate, followed by hydrolysis of the cyclic phosphoric acid ester with 2 ′:3′-cyclonucleotide-3′-phosphodiesterase. Because of the redox potential (-315 mV) and the distance between the pyridinium and phosphate groups, this analogue is a hydrogen acceptor and its reduced form a hydrogen donor in tests with alcohol dehyd rogenase from Thermoanaerobium brockii. The reduced form of the coenzyme analogue also is a hydrogen donor with glutathione reductase. With other NADP +-dependent dehydrogenases the com pound has been show n to be a competitive inhibitor against the natural coenzyme. The acetyl group reacts with bromine to form the bromoacetyl group. This reactive bromoacetyl analogue is a specific active-site directed irreversible inhibitor of isocitrate dehydrogenase.
From the electron and proton transfer equilibria network of quinones in solution a novel intermediate can be prepared by deprotonation of 2,5-bis(trimethylsilyl)hydroquinone to its monoanion using sodium metal. The sodium salt crystallizes in polymer strings connected via O⊖···(H)O hydrogen bridges, which are capped additionally by twofold dimethoxy-ethanesolvated Na⊕ countercations. The single crystal structure determination reveals one of the shortest O⊕ ··· HO distances observed so far of only 246 pm. MNDO calculations further confirm the assignment of hydroquinone monoanion building blocks in the polymer chain. For structural comparison as well as for attempts of its sodium reduction, 2,5-bis(trimethylsilyl)-p- benzoquinone has been synthesized. Its single crystal structure is reported, which does not show any cyanine distortion.
Semiquinone radical anions are prototype compounds for contact ion pair formation with metal counter cations. In order to investigate the still open question whether bulky alkyl groups can sterically interfere, diphenoquinone derivatives O=C(RC=CH)2C=C(HC=CR)2C=O with R = C(CH3)3, CH(CH3)2 and CH3 have been selected and the following ESR/ENDOR results are obtained for the alkaline metal cations: The tetrakis(tert-butyl)-substituted radical anion only adds Li⊕ and Na⊕, while K⊕ forms no ion pair. The 3,3ʹ,5,5ʹ-tetra(isopropyl)diphenoquinone radical anion is accessible to all cations Me⊕, although Rb⊕ and Cs⊕ seem to be present solvent-separated in solution. The tetramethyl-substituted radical anion unfortunately polymerizes rapidly. Additional information concerns the ESR/ENDOR proof for ion triple radical cation formation [Li⊕ M•⊖Li⊕]•⊕, or the difference in the coupling constants upon Me⊕ docking at one δ⊖O=C group, suggesting that about 87% of the spin density is located in the cation-free molecular half of the diphenoquinone radical anion. Based on the wealth of ESR/ENDOR information, crystallization of the contact ion pairs and their structural characterization should be attempted.
Chelate complexes of 1,2-dimesitoylbenzene radical anion with alkali metal cations exhibit in aprotic solution extremely large ESR /ENDOR metal coupling constants. For rationalization, structures of both the neutral molecule (H3C)3H2C6 - CO - C6H4 - CO - C6H2(CH3)3, in which the two carbonyl groups are twisted out of the benzene ring plane by dihedral angles of ± 3̄7̄°, and a sodium contact ion quadruple have been determined. One of the dimers [dimesitoylbenzeneH⊖ (Na⊕H2N H2C - CH2NH2)]2, although generated by Na metal mirror reduction of 1,2-dimesitoylbenzene in aprotic DME solution with added ethylendiamine for better electron transfer, surprisingly contains two 245 pm short (!) hydrogen bridges ⊖O ··· (H)O and in addition two solvation bridges e ⊖O ··· Na⊕(H2NH2C - CH2NH2) ··· O⊖. Results of MNDO calculations based on the experimental coordinates support the proposed concept.
Die hier vorgelegte Studie hat versucht, hinter einige gängige Forschungspositionen Fragezeichen zu setzen. Sie bezweifelt, ob Humboldt durchgängig "Schillers Führungsrolle unbedingt" "akzeptiert" hat. Sie stellt in Frage, ob Humboldt in seiner "Vorerinnerung" zum publizierten Briefwechsel wirklich nur eine Hommage Schillers hat schreiben wollen. Die Rekonstruktion interner und impliziter Kontroversen ist freilich nicht biographiegeschichtlich motiviert. Sie wird erstens im ästhetikgeschichtlichen Interesse unternommen, um alternativpoetische Konstellationen freizulegen. Sie ist zweitens fachgeschichtlich interessiert, indem sie fragt, was es bedeutet, dass Humboldt seine groß angelegten literatur- und kulturkomparatistischen Ansätze zugunsten sprachwissenschaftlicher Forschung zurückgestellt hat und nur noch implizit weiterführt. Sie geht drittens von der Überlegung aus, dass es um 1800 eine begrenzte Anzahl von Klassizisten und Romantiker gleichermaßen beschäftigende Schlüsselprobleme gegeben hat. Eines davon ist das produktive Verhältnis von Poesie und Prosa, Poesie und Philosophie.
Examina-se a concepção de universalismo ético a que Lawrence Kohlberg chegou em suas pesquisas sobre o desenvolvimento do pensamento moral, sobretudo a idéia de que esse desenvolvimento, em todos os seres humanos, culmina em uma moralidade "pós-convencional" concebida em termos de princípios de justiça. Discutem-se algumas das tentativas que foram feitas de submeter essa concepção à verificação empírica e a apreciação crítica que dela fez Jürgen Habermas.
Im Lößprofil der Ziegelei Glos bei Lisieux ist eine Abfolge fossiler Boden aufgeschlossen, die sich gut mit der hessischen Lößstratigraphie verbinden läßt. Über dem letztinterglazialen Boden liegen nach einer Diskordanz der Lohner Boden, der E1-, der E2- und der E4-Naß-boden. Letzterer wird häufig als Äquivalent des „Sol de Kesselt" angesehen. Somit zeigt sich auch für dieses Gebiet, daß dieser Boden stratigraphisch nicht dem Lohner Boden entsprechen kann.
Os modelos de democracia propostos pelo liberalismo e pelo republicanismo comunitarista são criticados a partir da perspectiva da política deliberativa tal como concebida pela teoria do discurso. Associando ao processo democrático conotações normativas mais fortes do que o modelo liberal, porém mais fracas do que o modelo republicano, a teoria do discurso articula elementos de ambas numa forma nova.
