Das Prostate-apoptosis-response-gene-4 (Par-4) sensibilisiert neoplastische Lymphozy-ten, deren Malignität durch eine Hemmung der Apoptosefähigkeit gekennnzeichnet ist, für apoptotische Stimuli. Vorhergehende Studien konnten zeigen, daß eine Überexpres-sion von Par-4 bei der Apoptoseinduktion durch Chemotherapeutika zu einer Downregulation von Bcl-2, einer Aktivierung von Caspase-3, einem verstärkten Abfall des Mi-tochondrialen Membranpotentials (MMP), sowie einer verstärkten PARP-Spaltung und dadurch zum programmierten Zelltod führt. Im Rahmen dieser Arbeit konnten weitere molekulare Mechanismen aufgezeigt werden, über die die apoptosesensibilisierende Wirkung von Par-4 vermittelt wird. Es konnte gezeigt werden, daß bei Behandlung mit Doxorubicin die Par 4 Überexpression außer zur stärkeren Aktivierung von Caspase-3 auch zur stärkeren Aktivierung von Caspase-8 und -9 führt. Außerdem kann eine Caspase 3 Inhibition bei Par-4 Überexpression durch eine alternative Aktivierung der Caspasen-6,-7,-8 und -9 umgangen werden. Dieses veränderte Caspasenaktivitätsmuster läßt sich mit einer Downregulation von Mitgliedern der Inhibitors of Apoptosis Proteins (IAPs) erklären. So konnte gezeigt werden, daß es bei Apoptoseinduktion unter Überexpression von Par-4 zur Downregulation von XIAP, cIAP1 und Survivin kommt. Diese Downregulation bleibt auch unter Caspase-3 Inhibition, sowie von XIAP und Survivin sogar unter allgemeiner Caspaseninhibition bestehen, das heißt sie ist nicht Caspase-3-, bzw. überhaupt nicht Caspasen-vermittelt. Weiterhin konnte gezeigt werden, daß es bei Überexpression von Par-4 unter Apoptoseinduktion mit Doxorubicin außer zum Abfall des MMPs auch zur verstärkten Ausschüttung von Cytochrom c und des Apoptosis In-ducing Factors (AIF) ins Cytosol kommt. Cytochrom c führt im Cytosol unter Bildung des Apoptosoms zur Aktivierung von Caspase-9, einem weiteren Mechanismus, der zur verstärkten Caspasenaktivität unter Par-4 Überexpression beiträgt. Die vermehrte Ausschüttung von Cytochrom c ist bedingt durch eine Translokation von tBid an die Mito-chondrienmembran. So ergaben die Untersuchungen, daß eine Par 4 Überexpression in neoplastischen T-Lymphozyten bei Behandlung mit Doxorubicin zur stärkeren Spaltung von Bid, einem Mitglied der proapoptotischen Bcl-2 Familie, sowie zur stärkeren Translokation des Bid Spaltproduktes (tBid) an die Mitochondrienmembran führt. Mit Hilfe der Immunfluoreszens konnte dargestellt werden, daß Bid in Par-4 überexprimierenden Zellen ein verändertes Verteilungsmuster im Cytosol aufweist und auf einen apoptotischen Reiz hin, verglichen mit den Par-4-negativen Zellen, vermehrt an die Mitochondrienmembran transloziert wird. Weiterhin konnte gezeigt werden, daß Bak und Bad, zwei weitere proapoptotische Mitgliederr der Bcl-2 Familie, durch Par 4 Überexpression Caspasen unabhängig verstärkt aktiviert werden. Bak wird durch das Bid-Spaltprodukt tBid aktiviert und führt ebenfalls zu einer verstärkten Cytochrom c Freisetzung ins Cytosol, wohingegen Bad seine proapoptotische Wirkung durch Bindung an antiapoptotische Mitglieder der Bcl-2 Familie entfaltet. Darüber hinaus konnte demonstriert werden, daß die Par-4 Überexpression in neoplastischen Lymphozyten bei Behandlung mit Doxorubicin zu einer Downregulation von Ras, welches zu den GTPasen zählt, führt. Diese Downregulation ist Caspasen-, jedoch nicht Caspase-3-abhängig. Hierbei besteht kein Zusammenhang zwischen der Expression von Ras und dem Tumorsupressorgen P53. Das heißt Par-4 entfaltet seine proapoptotische Wirkung über einen P53-unabhängigen Weg. Weiterhin konnte im Rahmen dieser Arbeit gezeigt werden, daß Par 4 Überexpression in neoplastischen Lymphozyten zu einer verstärkten Sensibilität des Fas-Rezeptors führt, das heißt, es konnte bewiesen werden, daß der pro-apoptotische Effekt von Par 4 sowohl durch Chemotherapeutika, als auch über direkte Rezeptorstimulation ausgelöst werden kann. Insgesamt legt diese Arbeit die wesentlichen molekularen Mechanismen der pro-apoptotischen Wirkung von Par-4 in neoplastischen Lymphozyten dar.
The detailed mechanism of the 20 S proteasome from Thermoplasma acidophilum is unknown. Substrates are degraded processively to small fragments without the release of intermediates, but the basis for this unique degradation mode remains obscure. The proteasome is a molecular machine, but how the different nanocompartments interplay and whether more than one substrate can be treated simultaneously has not been elucidated yet. To address these questions we had to disable the functionality of one aperture in order to dissect whether the other pore can compensate for the loss. As it is challenging to introduce mutations solely around one pore aperture of the highly symmetrical construct, we chose a novel approach by unique orientation of the proteasome at interfaces. For this purpose we purified recombinant 20 S proteasomes, where hexahistidine tags were fused either around the entrances or at the sides. According to electron microscopic studies we immobilized these constructs uniformly either end-on or side-on at metal-chelating interfaces (lipid vesicles, lipid monolayers and self-assembled thiol monolayers). Degradation of small fluorogenic peptides and large proteins like casein was analyzed. Small substrates were degraded with comparable activity by free and immobilized proteasomes, irrespective of their orientation. Thus it can be assumed that peptides can pass the sealed entrance of the 'dead-end' proteasome. However, larger substrates like fluorescently labeled casein were processed near the temperature optimum by side-on immobilized and soluble proteasomes with threefold activity compared to end-on immobilized proteasomes. Hence it can be concluded that one pore is sufficient for substrate entry and product release. In other words, the pore and antechamber can fulfil a triple function in the import and unwinding of substrates and the egress of products. With means of surface plasmon resonance the exact substrate/proteasome stoichiometry could be determined to ~1 for 'dead-end' proteasomes and ~2 for side-on immobilized (active and inactive) proteasomes. Most importantly, a fit with the Hill equation revealed positive cooperativity for side-on immobilized (Hill coefficient ~2) in contrast to end-on immobilized proteasomes (Hill coefficient ~1). Thus in case of soluble proteasomes two substrates bind presumably in opposite antechambers with positive cooperativity. The off-rate of casein as substrate is twofold for the active side-on immobilized proteasome in comparison to the end-on immobilized proteasome. The exact 2:1 stoichiometry of the off-rates equals the ratio of exit pathways amenable in case of side-on orientated versus 'dead-end' immobilized proteasomes. Thus crevices along the cylindrical body of the 20 S proteasome seem not to participate in the egress of small products. An inactive proteasome mutant displays a concentration-dependent off-kinetic against casein. Accordingly, the off-rate of the bisubstrate:proteasome complex can be attributed around half the value of the monosubstrate:proteasome complex. Consequently, substrates exit the inactive proteasome via the route of access due to obstruction of the trans side with an entering substrate. Hence the active proteasomes have to chop substrates down to small fragments prior to release through both pores. Thus the processive degradation mode might result from positive binding cooperativity. The on-rate constants for casein suggested that substrate association represents a two-step process comprising a rate-limiting translocation step and a fast binding step. As fluorescence cross-correlation revealed that two substrates can be co-localized in the proteasome and bind successively with increasing affinity (KD,1 = 8 µM versus KD,2 = 700 nM), an allosteric transition in the proteasome can be assumed. Combining our results with the data from other research groups led to a mechanistic model for the 20 S proteasome. Accordingly, the first substrate undergoes a slow translocation step, binds in the antechamber and diffuses subsequently to the catalytic centers, where it is degraded. By switching on the catalytic activity, the pores at both termini are dilated via conformational changes. Hence entry of the second substrate into the proteasome is facilitated due to omission of the rate-determining translocation step. The second substrate is either accommodated in the antechamber before it is processed (alternating degradation) or, most probably, is directly threaded into the central cavity (simultaneous degradation). As effusing peptides compete with entering proteins for binding in the antechamber, the pores are kept in an open state. After finishing digestion the pores are closed and a new degradation cycle can be reinitiated. In summary, substrate association with the proteasome underlies an ordered alternating binding mechanism in contrast to the random mode of degradation. Thus the two-stroke engine offers the advantage of speeding up degradation without enhancing complexity.

"Weimar" war die am häufigsten gebrauchte Chiffre im Selbstfindungsprozess der Deutschen nach 1945. Dort wollte man wieder "anknüpfen", aber auch lernen, was vermieden werden sollte. Weimar, das waren Nationalversammlung und Verfassungsgebung, die "goldenen Zwanziger", die Inflation, die Blockierung der Politik, der "Parteienstaat", die Koalition der "Systemfeinde", das war leuchtendes Vorbild, aber auch Chaos und Vorhölle zum NS-Staat. Christoph Gusy hat auf seiner Bielefelder Tagung daraus die Frage formuliert, wie die frühe Bundesrepublik mit Traum und Trauma von "Weimar" umgegangen ist, wie sie die Aneignung von Historie betrieben und in Politik umgesetzt hat. Auf diese Fragen antworten zunächst Wolfram Pyta mit einem souveränen Überblick über den jahrzehntelangen schrittweisen Prozess der Historisierung von Weimar, sodann Elke Seefried über die Grübeleien der Exilpolitiker, was "falsch gelaufen" und künftig zu vermeiden sei – überraschend antiparlamentarische und autoritäre Grübeleien übrigens. ...
"Solidarität" ist ein Wieselwort von gallertartiger Konsistenz. Es ist allgegenwärtig, gibt sich bedeutungsschwer und meist auch etwas vorwurfsvoll. Sein Kontext ist durchweg normativ. Es taucht dort auf, wo es um den Appell an Personen auf der gleichen Ebene geht. Typischerweise rufen Gruppen nach "Solidarität", deren innere Bindungen bröckeln oder die sich in Gefahrenlagen zusammenscharen. Als es noch Standesgenossen gab, erinnerten sie gerne an die Solidarität, wenn es darum ging, den Zusammenhalt derselben Schicht zu wahren. Die alten Zünfte, Gilden, Gaffeln, Einungen und Genossenschaften waren Solidaritätsverbände. Die Arbeiterbewegung übernahm hiervon nicht nur das Wort "Genossen", sondern auch den Appell an die Klassensolidarität. Und selbst wer heutzutage einen Krieg führen will, erinnert an alte Dankesschulden und appelliert an die Solidarität der Bundesgenossen. ...
Background: Common warts (verrucae vulgares) are human papilloma virus (HPV) infections with a high incidence and prevalence, most often affecting hands and feet, being able to impair quality of life. About 30 different therapeutic regimens described in literature reveal a lack of a single striking strategy. Recent publications showed positive results of photodynamic therapy (PDT) with 5-aminolevulinic acid (5-ALA) in the treatment of HPV-induced skin diseases, especially warts, using visible light (VIS) to stimulate an absorption band of endogenously formed protoporphyrin IX. Additional experiences adding waterfiltered infrared A (wIRA) during 5-ALA-PDT revealed positive effects. Aim of the study: First prospective randomised controlled blind study including PDT and wIRA in the treatment of recalcitrant common hand and foot warts. Comparison of "5-ALA cream (ALA) vs. placebo cream (PLC)" and "irradiation with visible light and wIRA (VIS+wIRA) vs. irradiation with visible light alone (VIS)". Methods: Pre-treatment with keratolysis (salicylic acid) and curettage. PDT treatment: topical application of 5-ALA (Medac) in "unguentum emulsificans aquosum" vs. placebo; irradiation: combination of VIS and a large amount of wIRA (Hydrosun® radiator type 501, 4 mm water cuvette, waterfiltered spectrum 590-1400 nm, contact-free, typically painless) vs. VIS alone. Post-treatment with retinoic acid ointment. One to three therapy cycles every 3 weeks. Main variable of interest: "Percent change of total wart area of each patient over the time" (18 weeks). Global judgement by patient and by physician and subjective rating of feeling/pain (visual analogue scales). 80 patients with therapy-resistant common hand and foot warts were assigned randomly into one of the four therapy groups with comparable numbers of warts at comparable sites in all groups. Results: The individual total wart area decreased during 18 weeks in group 1 (ALA+VIS+wIRA) and in group 2 (PLC+VIS+wIRA) significantly more than in both groups without wIRA (group 3 (ALA+VIS) and 4 (PLC+VIS)): medians and interquartile ranges: -94% (-100%/-84%) vs. -99% (-100%/-71%) vs. -47% (-75%/0%) vs. -73% (-92%/-27%). After 18 weeks the two groups with wIRA differed remarkably from the two groups without wIRA: 42% vs. 7% completely cured patients; 72% vs. 34% vanished warts. Global judgement by patient and by physician and subjective rating of feeling was much better in the two groups with wIRA than in the two groups without wIRA. Conclusions: The above described complete treatment scheme of hand and foot warts (keratolysis, curettage, PDT treatment, irradiation with VIS+wIRA, retinoic acid ointment; three therapy cycles every 3 weeks) proved to be effective. Within this treatment scheme wIRA as non-invasive and painless treatment modality revealed to be an important, effective factor, while photodynamic therapy with 5-ALA in the described form did not contribute recognisably - neither alone (without wIRA) nor in combination with wIRA - to a clinical improvement. For future treatment of warts an even improved scheme is proposed: one treatment cycle (keratolysis, curettage, wIRA, without PDT) once a week for six to nine weeks. © 2004 Fuchs et al; licensee German Medical Science. Das Ziel dieser prospektiven Studie war es , die technische Durchführbarkeit sowie klinische und radiologische Ergebnisse nach interkorporeller Fusion durch einen mit autologer Spongiosa gefüllten AcroMed Cervical I/F Cage (DePuy AcroMed
international, UK) nach ventraler zervikal er Diskektomie darzustellen. Über einen Zeitraum von 32 Monaten wur den in der Abteilung für Neurochirurgie (die zum Südharz-Krankenhaus Nordhausen gGmb H gehört) 50 aufeinander folgende Patienten operiert, bei welchen klinische Zeichen einer zervikalen Radikulopathie, Radikulomyelopathie oder einem Zervikalsyndrom mit monosegmentalen Bandscheibenvorfällen und/oder Spondylose nachzuweisen waren. Die Studie schloss 24 Frauen durchschnitt- lich 43.4 Jahre alt (25.4 – 59.7) und 26 Männer durchschnitt- lich 44.0 Jahre alt (29.8 – 64.6) ein. Die Dauer der Symptome betrug 1 bis 61 Monate (Mittel: 12.5 Monate). Alle Patienten wu rden prä- und postoperativ durch mich untersucht, ihre klinischen Beschwerden detailliert und standardisiert aufge- nommen. Alle Komplikationen wurden dokumentiert. Die funktio- nelle Behinderung wurde mittels Neck Pain Disability Index (NDI, Bereich 0-100) quantifiziert. Das klinische Ergebnis wurde get rennt vom Patienten und Untersucher als ausgezeich- net, gut, befri edigend oder schlecht unter Verwendung ODOM`S Kriterien eingeschätz t. Nach einer mittleren Nachuntersu- chungszeit von 14 Monaten (12-25 Monate) betrug der mittlere NDI 12,3 (0-84) gegenüber vor Operation 65.2 (44-88). Die Untersucher-Klassifikati on ergab 48% ausgezeichnete, 38% gute, 12% befriedigende und 2% schlechte Erge bnisse, 87.5% der ausgezeichneten und 89.5% der guten Gruppe kehrten zum Arbeits platz zurück. Im Vergleich ergab die Patienten-Klassifikation 58% ausgezeichnete, 32% gute, 8% befriedigende und 2% schlechte Ergebnisse, 93% der ausgezeichneten und 68.75% der guten Gruppe kehrten zum Arbeitsplatz zurück. Das Verfahren ist technisch einfach. Der Cervical I/F Cage steigert die zur Dekompression de r Nervenwurzel notwendige Höhe des Neuroforamens wirksam. Eine knöcherne Fusion wurde in 98% erreicht, die Keilform des Cages trägt zur Wiederherstel- lung der Lordose bei.
The development of image-guided neurosurgery represents a substantial improvement in the microsurgical treatment of tumors, vascular malformations and other intracranial lesions. Despite the wide applicability and many fascinating aspects of image-guided navigation systems, a major drawback of this technology is they use images, mainly MRI pictures, acquired preoperatively, on which the planning of the operative procedure as well as its intraoperative performance is based. As dynamic changes of the intracranial contents regularly occur during the surgical procedure, the surgeon is faced with a continuously changing intraoperative field. Only intraoperatively acquired images will provide the neurosurgeon with the information he needs to perform real intraoperative image-guided surgery. A number of tools have been developed in recent years, like intraoperative ultrasound and dedicated moveable intraoperative CT units. Because of its excellent imaging qualities, combined with the avoidance of ionizing radiation, MRI currently is and definitely will be in the future for the superior imaging method for intraoperative image guidance. In this short overview, the development as well as some of the current and possible future applications of MRI-guided neurosurgery is outlined.
Dragonflies were collected at ten localities in five counties (sous-préfectures) in southern Bénin and are presently identified. A total of 73 species were found, including 45 new records for the country. The list of the odonates of Bénin currently comprises 86 species although more than 100 can probably be expected.
