Interferon-stimulated gene-15 (ISG15) is an interferon-induced protein with two ubiquitin-like (Ubl) domains linked by a short peptide chain, and the conjugated protein of the ISGylation system. Similar to ubiquitin and other Ubls, ISG15 is ligated to its target proteins with a series of E1, E2, and E3 enzymes known as Uba7, Ube2L6/UbcH8, and HERC5, respectively. Ube2L6/UbcH8 plays a literal central role in ISGylation, underscoring it as an important drug target for boosting innate antiviral immunity. Depending on the type of conjugated protein and the ultimate target protein, E2 enzymes have been shown to function as monomers, dimers, or both. UbcH8 has been crystalized in both monomeric and dimeric forms, but the functional state is unclear. Here, we used a combined approach of small-angle X-ray scattering (SAXS) and nuclear magnetic resonance (NMR) spectroscopy to characterize UbcH8’s oligomeric state in solution. SAXS revealed a dimeric UbcH8 structure that could be dissociated when fused with an N-terminal glutathione S-transferase molecule. NMR spectroscopy validated the presence of a concentration-dependent monomer-dimer equilibrium and suggested a backside dimerization interface. Chemical shift perturbation and peak intensity analysis further suggest dimer-induced conformational dynamics at ISG15 and E3 interfaces - providing hypotheses for the protein’s functional mechanisms. Our study highlights the power of combining NMR and SAXS techniques in providing structural information about proteins in solution.

5-Lipoxygenase (5-LO) is the key enzyme in the formation of pro-inflammatory leukotrienes (LT) which play an important role in a number of inflammatory diseases. Accordingly, 5-LO inhibitors are frequently used to study the role of 5-LO and LT in models of inflammation and cancer. Interestingly, the therapeutic efficacy of these inhibitors is highly variable. Here we show that the frequently used 5-LO inhibitors AA-861, BWA4C, C06, CJ-13,610 and the FDA approved compound zileuton as well as the pan-LO inhibitor nordihydroguaiaretic acid interfere with prostaglandin E2 (PGE2) release into the supernatants of cytokine-stimulated (TNFα/IL-1β) HeLa cervix carcinoma, A549 lung cancer as well as HCA-7 colon carcinoma cells with similar potencies compared to their LT inhibitory activities (IC50 values ranging from 0.1–9.1 µM). In addition, AA-861, BWA4C, CJ-13,610 and zileuton concentration-dependently inhibited bacterial lipopolysaccharide triggered prostaglandin (PG) release into human whole blood. Western Blot analysis revealed that inhibition of expression of enzymes involved in PG synthesis was not part of the underlying mechanism. Also, liberation of arachidonic acid which is the substrate for PG synthesis as well as PGH2 and PGE2 formation were not impaired by the compounds. However, accumulation of intracellular PGE2 was found in the inhibitor treated HeLa cells suggesting inhibition of PG export as major mechanism. Further, experiments showed that the PG exporter ATP-binding cassette transporter multidrug resistance protein 4 (MRP-4) is targeted by the inhibitors and may be involved in the 5-LO inhibitor-mediated PGE2 inhibition. In conclusion, the pharmacological effects of a number of 5-LO inhibitors are compound-specific and involve the potent inhibition of PGE2 export. Results from experimental models on the role of 5-LO in inflammation and pain using 5-LO inhibitors may be misleading and their use as pharmacological tools in experimental models has to be revisited. In addition, 5-LO inhibitors may serve as new scaffolds for the development of potent prostaglandin export inhibitors.
5-lipoxygenase (5-LO) catalyzes the first two steps in leukotriene (LT) biosynthesis. In a two step reaction the enzyme oxygenates arachidonic acid (AA) to form the highly unstable epoxide leukotriene A4 (LTA4) in dehydrating a hydroperoxide intermediate (20). LTA4 can then be further metabolized by two terminal synthases yielding either the potent chemoattractant leukotriene B4 (LTB4) or the cysteinyl leukotrienes (CysLTs). 5-LO enzyme expression is primarily found in mature leukocytes (22) where it can either reside in the cytoplasm or in the nucleus associated with euchromatin (29). Its enzymatic activity is embedded in a complicated network in intact cells regulating LT synthesis by various factors dependent on the cell type and nature of stimulus. Factors such as the amount of free AA released by phospholipase A2 enzymes, levels of enzymes involved, catalytic activity per enzyme molecule and availability of different small molecules influence 5-LO activity (36).
The 5-LO derived LTs are lipid mediators which were shown to primarily mediate inflammatory and allergic reactions and their role in the pathogenesis of asthma is well defined. CysLTs are among the most potent bronchoconstrictors yet studied in man and play an important role in airway remodeling. LTB4 has no bronchoconstrictory effects in healthy and asthmatic humans but displays potent chemoattractant properties on neutrophils and increases leukocyte adhesion to the vessel wall endothelium (22). Therefore, LTB4 enhances the capacity of macrophages and neutrophils to ingest and kill microbes. In concert with LTB4, histamine and prostaglandin E2 (PGE2) CysLTs are thought to maintain the tone of the human airways (82).
Besides their well studied role in asthma, 5-LO derived LTs have also been implicated to play a role in cardiovascular diseases and cancer. In contrast to healthy tissues, LT pathway enzymes and receptors were found to be abundantly expressed in cancer tissues, atherosclerotic lesions in the aorta, heart and carotid artery (86). Pharmacological inhibition of 5-LO potently suppressed tumour cell growth by inducing cell cycle arrest and triggering cell death via the intrinsic apoptotic pathway (92, 93). In several studies LTs were found to exhibit cardiovascular actions by promotion of plasma leakage in postcapillary venules, coronary artery vasoconstriction and impaired ventricular contraction leading to reduced coronary blood flow and cardiac output (24). Unfortunately, the precise molecular mechanisms through which LTs influence carcinogenesis and cardiovascular diseases are still incompletely understood.
In contrast, an increasing number of studies questions the correlation between 5-LO and cancer (95-97) since extreme LT concentrations were applied to induce proliferative effects in the majority of the publications. A few studies exist which show susceptibility towards 5-LO products in physiological concentrations or achieve anti-proliferation by applying low concentrations of 5-LO inhibitors (98) ...
Der vorliegende Abschlussbericht besteht aus zwei Teilen: während Teil A die ausführlichen Ergebnisse der begleitend durchgeführten Evaluation der Qualifizierungsschecks in Hessen für das Jahr 2014 darstellt, fasst Teil B die Ergebnisse des Projekts „Informationssystem berufliche Weiterbildung in Hessen“ zusammen. Das Informationssystem berufliche Weiterbildung in Hessen wurde gefördert vom Hessischen Ministerium für Wirtschaft, Verkehr und Landesentwicklung und durch Mittel aus den Europäischen Sozialfonds Hessen. Die Projektdauer umfasste den Zeitraum vom 1.8.2008 bis zum 31.12.2014.
This paper examines substantive noun phrases in Niuean, a Polynesian language of the Tongic subgroup with VSO word order, isolating morphology, and an ergative case system. We describe the allowable orderings of elements in the Niuean noun phrase, which include certain variations in the placement of numerals and the genitive possessor, then we provide a phrasal movement analysis for these variations, treating first the possessor variation, then the numeral variation. Parallels will be drawn between the derivation of nominal and sentential word order.
The purpose of this paper is to provide a unified (i.e. independent of lexical categories) account of Persian stress. I show that by differentiating word- and phrase-level stress rules, one can account for the superficial differences exemplified in (1) above and many of the stipulations suggested by previous scholars. The paper is organized as follows. In section 1, I look at nouns and adjectives and propose a rule that would account for their stress pattern. In section 2, I extend the stress rule to verbs and show the problem this category poses to our generalization. The main proposal of this paper is discussed in section 3. I introduce the phrasal stress rule in Persian and show that by differentiating word-level and phrase-level stress rules, one can come to a unified account of Persian stress. Section 4 deals with some problematic eases for the proposed generalization and discusses some tentative solutions and their theoretical consequences. Section 5 concludes the paper.
In Justice and Reconciliation in World Politics Catherine Lu endorses the idea that those who contribute to the reproduction of structural injustice have responsibilities to address that injustice (Lu, 2017). However, in the book, Lu does not explore the grounds and justification for recognising such a responsibility. In order to address this deficit, this paper proposes that those likely to contribute to the reproduction of structural injustice, in the future, have precautionary duties, in the present, that require them to take action aimed at preventing their future contribution. It is proposed that these ‘collectivization duties’ (Collins, 2013) require them to act responsively with a view to forming a collective that can end the structural injustice in question. This account recommends a collective-action solution alongside recognising that each socially connected agent is obliged to act. However, it does not entail that amorphous groups bear responsibilities and is appropriate in its attribution of blame, thus avoiding both Nussbaum’s (2011) critique of perpetually forward-looking accounts and the ‘agency objection’ (Wringe, 2010).
Biologischen Systemen liegen Mechanismen zugrunde, die bis heute nicht vollständig aufgeklärt sind. Für die Untersuchung eignen sich externe Trigger, die eine Regulation von außen auf das System erlauben und Prozesse gezielt steuerbar machen. Eine Möglichkeit ist das System unter optische Kontrolle zu bringen, d.h. durch Licht eine externe Steuerung zu implementieren, was von einem internen Chromophor aufgenommen wird. In der vorliegenden Dissertation werden drei individuelle Projekte vorgestellt, die alle dieses Prinzip auf unterschiedliche Weise anwenden.
RNAs haben eine Vielzahl an verschiedenen Funktionen in der Zelle, die von der Proteinsynthese bis zur Genregulation variieren. Im ersten Projekt wurde der Photoschalter Spiropyran im Kontext von RNA eingesetzt. Mit dem Ziel das Derivat PyBIPS kovalent an ein Oligonukleotid zu binden und damit die Hybridisierung eines Duplexes zu steuern, wurden Strukturmotive gesucht, an die der Photoschalter postsynthetisch gebunden werden kann. Dabei soll die sterisch anspruchsvolle Spiropyran-Form die Watson-Crick-Basenpaarung stören und die planare, konjugierte Merocyanin-Form in den Duplex, zur zusätzlichen Stabilisierung, interkalieren. In Vorarbeiten von Clara Brieke wurde festgestellt, dass erstens nur PyBIPS, nach kovalenter Verknüpfung, noch vollständig photochemisch aktiv ist und zweitens eine Herstellung über Festphasensynthese nur in schlechter Ausbeute realisierbar ist. Ausgehend davon wurde PyBIPS an die drei nicht-nukleosidischen Linker, Aminoglykol, D-Threoninol und Serinol, postsynthetisch über eine Amidbindung angebracht, die zuvor über Oligonukleotidfestphasensynthese in 2´-OMe-RNA eingebaut wurden.
In der photochemischen Charakterisierung konnte gezeigt werden, dass PyBIPS, gebunden an alle drei Motive, noch photochemisch aktiv ist und im Vergleich zu ungebundenem PyBIPS stabiler ist gegenüber Photolyse. In Untersuchungen im Doppelstrang im Wedge Motiv, d.h. im Gegenstrang befindet sich kein Nukleotid gegenüber dem Photoschalter, wurde ein ähnliches Verhalten festgestellt. Zusätzlich zur charakteristischen Merocyanin-Bande bei 550 nm ist ein zweites, rotverschobenes Absorptionsmaximum entstanden, das die gleichen Eigenschaften besitzt. In Schaltzyklen wurde festgestellt, dass eine Isomerisierung bis 660 nm möglich ist, was eine Anwendung im therapeutischen Fenster zwischen ca. 600 und 1000 nm von Blut ermöglichen würde. Das Auftreten der zweiten Bande hängt stark vom Linker und dem Baustein im gegenüberliegenden Strang ab. Es wird vermutet, dass sich das, sonst nicht-bevorzugte, TTT-Isomer der Merocyanin-Form, durch Stabilisierung durch die Umgebung im Oligonukleotid ausbildet.
Zur Untersuchung, inwiefern die Isomerisierung des Photoschalters die Duplexstruktur beeinflusst, wurden Schmelzpunktstudien und Fluoreszenzmessungen zur KD-Bestimmung durchgeführt, ohne dass eine Veränderung zu erkennen war. In einer größeren NMR-Studie, in Kooperation mit Tom Landgraf (Arbeitsgruppe Prof. Dr. Harald Schwalbe) wurde der Fokus darauf-gelegt mehr Informationen über die strukturelle Integrität zu erhalten. Es findet eine Störung der benachbarten Basenpaarungen durch den Photoschalter statt, jedoch kann die Kraft der Isomerisierung des Photoschalters nicht übertragen warden. Der Einfluss war zunächst geringer als erwartet, was in anderen Anwendungen überprüft warden muss.
Im zweiten Projekt steht das Membranprotein OmpG aus E. Coli K12 im Fokus. Proteine besitzen viele verschiedene funktionelle Gruppen, die für selektive Biokonjugation genutzt werden können in einer Reihe von Anwendungen. OmpG gehört zur Gruppe der Porine, die in der äußeren Membran von Gram-negativen Bakterien sitzen und die seltene ß-Fassstruktur ausbilden. Die Pore besitzt einen großen Innendurchmesser ohne Selektivität. Das Molekül wurde bereits intensiv auf seine Struktur, insbesondere Loop 6, der pH-abhängig die Pore verschließt, untersucht. In Vorarbeiten von Grosse et al. wurde der Loop entfernt, sodass ein ruhiger Kanal entstanden ist, der ein optimales Modellsystem darstellt. Außerdem wurden in der Pore zwei Cysteine, die auf gegenüberliegenden Seiten auf halber Höhe des Kanals sitzen, eingeführt. An die Thiolgruppen wurden photolabile Schutzgruppen angebracht, die erst den Kanal blockieren und nach Abspaltung durch Licht wieder freigeben. Dazu wurde jeweils ein 7-Diethylaminocumarin (DEACM) postsynthetisch angebracht.
Der Linker am Cumarin-Alkohol stellt dabei einen Kompromiss dar, da er zum einen selektiv mit der Thiolgruppe des Cysteins reagieren soll, gleichzeitig aber noch photo-induziert wieder abspalten muss. Durch Belichtung findet eine Hydrolyse des Esters unter Abspaltung des Alkohols statt, der einen Carbonsäurerest am Thiol in der Pore zurück lässt. In spannungsabhängigen Einzelkanal-messungen, durchgeführt von Dr. Philipp Reiß (Universität Marburg), konnte gezeigt werden, dass die zwei DEACM Modifikationen eine Reduktion der Leitfähigkeit bewirkten und durch Licht abgespalten und aus dem Kanal entfernt werden konnten. Dabei war außerdem zu erkennen, dass die Leitfähigkeit aufgrund der Carboxylreste über das Niveau von unmodifiziertem OmpG steigt.
