Background: Alterations in the DNA methylation pattern are a hallmark of leukemias and lymphomas. However, most epigenetic studies in hematologic neoplasms (HNs) have focused either on the analysis of few candidate genes or many genes and few HN entities, and comprehensive studies are required. Methodology/Principal Findings: Here, we report for the first time a microarray-based DNA methylation study of 767 genes in 367 HNs diagnosed with 16 of the most representative B-cell (n = 203), T-cell (n = 30), and myeloid (n = 134) neoplasias, as well as 37 samples from different cell types of the hematopoietic system. Using appropriate controls of B-, T-, or myeloid cellular origin, we identified a total of 220 genes hypermethylated in at least one HN entity. In general, promoter hypermethylation was more frequent in lymphoid malignancies than in myeloid malignancies, being germinal center mature B-cell lymphomas as well as B and T precursor lymphoid neoplasias those entities with highest frequency of gene-associated DNA hypermethylation. We also observed a significant correlation between the number of hypermethylated and hypomethylated genes in several mature B-cell neoplasias, but not in precursor B- and T-cell leukemias. Most of the genes becoming hypermethylated contained promoters with high CpG content, and a significant fraction of them are targets of the polycomb repressor complex. Interestingly, T-cell prolymphocytic leukemias show low levels of DNA hypermethylation and a comparatively large number of hypomethylated genes, many of them showing an increased gene expression. Conclusions/Significance: We have characterized the DNA methylation profile of a wide range of different HNs entities. As well as identifying genes showing aberrant DNA methylation in certain HN subtypes, we also detected six genes—DBC1, DIO3, FZD9, HS3ST2, MOS, and MYOD1—that were significantly hypermethylated in B-cell, T-cell, and myeloid malignancies. These might therefore play an important role in the development of different HNs.

Gerigk vereinigt vierzehn Essays zu Dostoevskij, darunter drei Erstpublikationen und elf Bearbeitungen von Veröffentlichungen der Jahre 1981 bis 2006. Texte Dostoevskijs kommunizieren mit solchen von Turgenev, Heidegger, Schiller, E.T.A. Hoffmann, Faulkner, Flaubert, Hauptmann, Salinger, Joyce, Sylvia Plath und anderen.
Mit der hier vorliegenden Arbeit ist das Entladungsverhalten einer Dielektrischen Barriere Entladung anhand der elektrischen Parameter untersucht worden. Dazu wurde ein planparallele Elektrodenkonfiguration entwickelt und aufgebaut, die mit einer Sinus-Spannungs von maximal 5000 Vss und einer Frequenz von 5 kHz-20 kHz angesteuert wurde. Als Arbeitsgas wurde Argon im Druckbereich von 100 mbar - 1000 mbar verwendet. Auf diese Weise konnte ein Plasma erzeugt werden, in das bei einer maximalen Transfereffiienz von 96%, eine mittlere Leistung von bis zu 845 mW eingekoppelt werden konnte.
Da die Dielektrische Barriere Entladung auf Grund der Abschirmung der Elektroden vom Gasraum einige Besonderheiten gegenüber eines klassischen kapazitiv eingekoppelten Plasmas aufweist, können keine Rückschlüsse mittels einer einfachen Strom-Spannungsmessung auf die Vorgänge im Plasma gemacht werden.
Um trotzdem Einblick in die Entladung zu erhalten, wurde das von [Tra08] vorgeschlagene Ladungstransportmodel für die Analyse herangezogen und an den hier vorliegenden experimentellen Aufbau angepasst. So konnte unter anderem der Ein
uss der auf den Dielektrika befindlichen Restladungsträger auf die Ausbildung der Entladung untersucht werden. Des Weiteren ist aus den gewonnen Größen die Elektronendichte des Plasmas bestimmt worden. Diese liegt bei der hier untersuchten dielektrischen Glimmentladung, je nach Gasdruck und Frequenz, im Bereich zwischen 5 X 10exp9 - 2 X 10exp10 1/cm3 . Auch konnten in diesem Zusammenhang qualitative Aussagen über die Entwicklung der Elektronentemperatur gemacht werden.
Zusammen mit Kurzzeitaufnahmen, die den Verlauf der Entladung dokumentieren und den in [Sch11] gemachten Untersuchungen zur Excimerstrahlung konnte so ein umfassendes Bild der Vorgänge im Plasma erstellt und Kriterien erarbeitet werden, die den für eine Dielektrische Barriere Entladung typischen Ubergang zwischen einer Glimmentladung zu einer filamentierten Entladung erklären können.
Im Rahmen dieser Diplomarbeit haben Arbeiten am Elektronen-Streuexperiment von Prof. J. Jacoby und Dr. R. Berezov an der Goethe-Universität Frankfurt stattgefunden. Am Experiment wurden durch Møller-Streuung verschränkte Elektronen auf ihre Spin-Abhängigkeit untersucht. Die Untersuchung erfolgte als Koinzidenzmessung, dessen Genauigkeit von der zeitlichen Auflösung der Detektoren abhängt. Als Koinzidenzlogik diente ein von Julian Schunk entwickeltes Datenerfassungsprogramm, das auf einem Oszilloskop installiert wurde...
Which factors determine whether a stimulus is consciously perceived or unconsciously processed? Here, I investigate how previous experience on two different time scales – long term experience over the course of several days, and short term experience based on the previous trial – impact conscious perception. Regarding long term experience, I investigate how perceptual learning does not only change the capacity to process stimuli, but also the capacity to consciously perceive them. To this end, subjects are trained extensively to discriminate between masked stimuli, and concurrently rate their subjective experience. Both the ability to discriminate the stimuli as well as subjective awareness of the stimuli increase as a function of training. However, these two effects are not simple byproducts of each other. On the contrary, they display different time courses, with above chance discrimination performance emerging before subjective experience; importantly, the two learning effects also rely on different circuits in the brain: Moving the stimuli outside the trained receptive field size abolishes the learning effects on discrimination ability, but preserves the learning effects on subjective awareness.
This indicates that the receptive fields serving subjective experience are larger than the ones serving objective performance, and that the channels through which they receive their information are arranged in parallel. Regarding short term experience, I investigate how memory based predictions arising from information acquired on the trial before affect visibility and the neural correlates of consciousness. To this end, I vary stimulus evidence as well as predictability and acquire electroencephalographic data.
A comparison of the neural processes distinguishing consciously perceived from unperceived trials with and without predictions reveals that predictions speed up processing, thus shifting the neural correlates forward in time. Thus, the neural correlates of consciousness display a previously unappreciated flexibility in time and do not arise invariably late as had been predicted by some theorists.
Admittedly, however, previous experience does not always stabilize perception. Instead, previous experience can have the reverse effect: Seeing the opposite of what was there, as in so-called repulsive aftereffects. Here, I investigate what determines the direction of previous experience using multistable stimuli. In a functional magnetic resonance imaging experiment, I find that a widespread network of frontal, parietal, and ventral occipital brain areas is involved in perceptual stabilization, whereas the reverse effect is only evident in extrastriate cortex. This areal separation possibly endows the brain with the flexibility to switch between exploiting already available information and emphasizing the new.
Taken together, my data show that conscious perception and its neuronal correlates display a remarkable degree of flexibility and plasticity, which should be taken into account in future theories of consciousness.
Julius Campe wich zwar keiner Auseinandersetzung mit der Obrigkeit aus, wenn sie ihm notwendig erschien, aber er ging dabei doch stets kalkuliert vor und war sich der Risiken immer bewusst. Kaum jemand hat im Vormärz die Grenzen, welche staatliche Repression und Überwachung der Literatur und ihrer Vermarktung setzten, stärker ausgereizt als der "Odysseus des deutschen Buchhandels", wie Heine ihn genannt hat. Seine Arbeitsweise machte ihn zum Prototypen des modernen Verlegers, war aber auch charakteristisch für den Vormärz, dessen Beschränkungen paradoxerweise seinen Aufstieg erst ermöglicht haben. [...] Das zweite Verlagsverbot gegen Hoffmann und Campe erging im Januar 1847 in Österreich, und in diesen Kontext gehören die vier Dokumente, die im Folgenden vorgestellt werden. Die Buchhandelskorrespondenzen von und an Julius Campe, Gustav Mayer, Viktor von Andrian-Werburg, Gustav Remmelmann und Carl Gustav Heinrich Welsch betreffen die Frage nach der strategisch besten Antwort auf diese Repressionsmaßnahme. Sie befinden sich im Archiv des Heinrich-Heine-Instituts, Düsseldorf (im so genannten 'Nachlass Campe') und werden hier erstmals veröffentlicht.
Kaum je zuvor unterlag im deutschsprachigen Raum ein ganzes Gewerbe so strenger obrigkeitsstaatlicher Kontrolle und Restriktion wie die Buchbranche zwischen Karlsbader Beschlüssen und Märzrevolution. Andererseits aber erlebte in jener Phase auch kaum ein anderer Geschäftszweig einen derartig starken Aufschwung wie das Verlagswesen und der Buchhandel - dieses Paradoxon gehört zu den vielen erstaunlichen Ungleichzeitigkeiten, die für den Vormärz so charakteristisch sind.

Literaturbetrieb und Verlagswesen spiegeln auf vielfältige Weise die gesellschaftlichen und politischen Entwicklungen der Vormärzzeit wider. Wie in vielen anderen Bereichen wurden in jener Epoche auch hier die Grundlagen für manche modernen Verhältnisse geschaffen, wie sie teilweise auch noch heute Bestand haben. Mit dem Schwerpunktthema Literaturbetrieb und Verlagswesen will das Jahrbuch des "Forum Vormärz Forschung" diese Prozesse nachzeichnen. Aus verschiedenen Blickwinkeln erörtern die hier versammelten Beiträge, welche neuen Publikations- und Verkaufsstrategien für die neue Literatur gefunden wurden, welche Veränderungen sich auf dem Buch- und Zeitschriftenmarkt, bei der Zusammensetzung und den Erwartungen der Leserschaft ergaben, wie sich Vertriebswege und Lesekultur wandelten. Sie untersuchen, welche Rolle das Rezensionswesen spielte, was für Rückwirkungen die Marktgesetze und die verschärfte Konkurrenzsituation auf die Literatur und ihre Inhalte sowie auf die Autoren, ihr Selbstverständnis und ihre öffentliche Selbstdarstellung hatten. Der Umgang der Presse und Buchverlage mit der Zensur im deutschsprachigen Raum ist dabei ein durchgängiges Thema.
Background: Factors and processes shaping the population structure and spatial distribution of genetic diversity across a species' distribution range are important in determining the range limits. We comprehensively analysed the influence of recurrent and historic factors and processes on the population genetic structure, mating system and the distribution of genetic variability of the pulmonate freshwater snail Radix balthica. This analysis was based on microsatellite variation and mitochondrial haplotypes using Generalised Linear Statistical Modelling in a Model Selection framework. Results: Populations of R. balthica were found throughout North-Western Europe with range margins marked either by dispersal barriers or the presence of other Radix taxa. Overall, the population structure was characterised by distance independent passive dispersal mainly along a Southwest-Northeast axis, the absence of isolation-by-distance together with rather isolated and genetically depauperated populations compared to the variation present in the entire species due to strong local drift. A recent, climate driven range expansion explained most of the variance in genetic variation, reducing at least temporarily the genetic variability in this area. Other factors such as geographic marginality and dispersal barriers play only a minor role. Conclusions: To our knowledge, such a population structure has rarely been reported before. It might nevertheless be typical for passively dispersed, patchily distributed taxa (e.g. freshwater invertebrates). The strong local drift implied in such a structure is expected to erode genetic variation at both neutral and coding loci and thus probably diminish evolutionary potential. This study shows that the analysis of multiple factors is crucial for the inference of the processes shaping the distribution of genetic variation throughout species ranges. Background: Until recently, read lengths on the Solexa/Illumina system were too short to reliably assemble transcriptomes without a reference sequence, especially for non-model organisms. However, with read lengths up to 100 nucleotides available in the current version, an assembly without reference genome should be possible. For this study we created an EST data set for the common pond snail Radix balthica by Illumina sequencing of a normalized transcriptome. Performance of three different short read assemblers was compared with respect to: the number of contigs, their length, depth of coverage, their quality in various BLAST searches and the alignment to mitochondrial genes. Results: A single sequencing run of a normalized RNA pool resulted in 16,923,850 paired end reads with median read length of 61 bases. The assemblies generated by VELVET, OASES, and SeqMan NGEN differed in the total number of contigs, contig length, the number and quality of gene hits obtained by BLAST searches against various databases, and contig performance in the mt genome comparison. While VELVET produced the highest overall number of contigs, a large fraction of these were of small size (< 200bp), and gave redundant hits in BLAST searches and the mt genome alignment. The best overall contig performance resulted from the NGEN assembly. It produced the second largest number of contigs, which on average were comparable to the OASES contigs but gave the highest number of gene hits in two out of four BLAST searches against different reference databases. A subsequent meta-assembly of the four contig sets resulted in larger contigs, less redundancy and a higher number of BLAST hits. Conclusion: Our results document the first de novo transcriptome assembly of a non-model species using Illumina sequencing data. We show that de novo transcriptome assembly using this approach yields results useful for downstream applications, in particular if a meta-assembly of contig sets is used to increase contig quality. These results highlight the ongoing need for improvements in assembly methodology. Keywords: next generation sequencing; short read assembly; Mollusca
