Das Nierenzellkarzinom (NZK) ist der häufigste maligne Tumor der Niere. In vielen Fällen sind bereits bei der Erstdiagnose Metastasen vorhanden oder entstehen im Verlauf der Therapie. Die Behandlungsmöglichkeiten für diese NZK-Patienten sind äußerst limitiert. Nahezu 2/3 der Betroffenen versterben an ihrer Erkrankung. Die Etablierung neuer Therapieansätze zur Behandlung des NZK ist dringend gefordert. Es wird postuliert, dass ein Therapiekonzept basierend auf dem Histondeacetylase (HDAC)-Inhibitor Valproat (VPA) kombiniert mit niedrig dosiertem Interferon (IFN)-alpha eine innovative und effiziente Behandlungsoption für austherapierte NZK-Patienten eröffnen könnte. In der vorliegenden Studie wurde der Einfluss einer VPA-Mono- versus VPA/IFN-alpha-Kombinationstherapie auf die malignen Eigenschaften verschiedener NZK-Zelllinien evaluiert. Mittels funktioneller Untersuchungen wurden Proliferations- und Adhäsionsphänomene unter den entsprechenden Therapien näher betrachtet. Fluorimetrische und molekularbiologische Studien dienten der detaillierten Aufklärung der den Veränderungen zugrunde liegenden Wirkmechanismen. Zur translationalen Gestaltung wurden zusätzlich tierexperimentelle Untersuchungen durchgeführt. VPA induzierte eine signifikante Reduktion der Proliferation von NZK-Zellen, die durch die additive Gabe von IFN-alpha weiter verstärkt wurde. Die anti-proliferativen Effekte korrelierten mit einer Zunahme der Zellen in der G0/G1-Phase und einer damit einhergehenden verminderten Anzahl an Zellen in der S-Phase. Die Verschiebung der Zellzyklusphasen war mit einer deutlichen Modulation relevanter regulatorischer Zellzyklusproteine assoziiert. Des Weiteren resultierte VPA und die korrespondierende Kombination mit IFN-alpha in einer signifikanten Inhibition der Adhäsion an Endothel und die extrazellulären Matrixproteine. VPA und die VPA/IFN-alpha-Kombination übten ihren Einfluss dabei offensichtlich über eine Modulation von Adhäsionsrezeptoren, implizit Integrine und CD44-Varianten, aus. Die antiproliferative und –adhäsive Wirkung war in der Regel nach längerer Inkubationszeit von 5 Tagen deutlich stärker als nach 3 Tagen. In analog behandelten normalen Nierenzellen zeigten sich im Vergleich keine solchen Effekte. Die Behandlung mit VPA und IFN-alpha scheint somit spezifisch maligne Zellen zu beeinflussen. VPA induzierte in den NZK-Zellen ferner die Reduktion von Protoonkogenen und MAP-Kinasen sowie die Zunahme von Tumorsuppressoren. Die zusätzliche Gabe von IFN-alpha resultierte in einer weiteren Wirkungsverstärkung gegenüber VPA allein. VPA und die Kombination mit IFN-alpha inhibierten zudem signifikant die HDAC6-Aktivität und -Proteinexpression der NZK-Zellen. Diese Hemmung ging mit einer Hyperacetylierung der Histone einher. Die epigenetische Modulation führte zur veränderten Genregulation und Transkription. So nahmen VPA und die korrespondierende Kombination neben den genannten funktionellen und molekularbiologischen Veränderungen maßgeblich Einfluss auf das Genexpressionsprofil der Tumorzellen. Die Expression negativer Regulatoren der Proliferation, Migration und Adhäsion sowie von Genen involviert in Differenzierung und Immunantwort wurden erhöht, wohingegen die Anzahl der Transkripte von Genen mitverantwortlich für die Ausbildung von Resistenzen und die Nährstoffversorgung der Tumoren reduziert wurde. Translationale tierexperimentelle Studien bestätigten die klinische Relevanz der VPA- und VPA/IFN-alpha-Behandlung, die in einer signifikanten Hemmung des Tumorwachstums resultierten. Die Wachstums-Inhibition war mit einer starken Modulation regulatorischer Proteine des Zellzyklus, der Apoptose und des HDAC-Systems assoziiert. Die vorliegenden Ergebnisse demonstrieren das viel versprechende Wirkungspotential von VPA und der korrespondierenden Kombination mit niedrig dosiertem IFN-alpha. VPAs anti-proliferative und -adhäsive Effekte in vitro und in vivo eröffnen die Perspektive für eine innovative Strategie in der Behandlung des NZK. Aufgrund der präsentierten Daten lässt sich postulieren, dass VPA und IFN-alpha die Grundlage für ein neues, effizientes Therapiekonzept bei austherapierten NZK-Patienten darstellen könnte.
Effekte von Neonikotinoiden auf die Aktivität des Muskels M17 und das Lernverhalten der Honigbiene

Im Rahmen dieser Arbeit wurden Untersuchungen zur Auswirkung von Neonikotinoiden auf die Muskelspikeaktivität des Muskels M17 und auf das Lernvermögen in einer komplexen Aufgabe an der Honigbiene (Apis mellifera carnica) durchgeführt. Dabei wurden drei verschiedene Substanzen verwendet: Clothianidin, Thiacloprid und Imidacloprid. Neonikotinoide stehen häufig im Verdacht, für das Sterben von Bienenvölkern verantwortlich zu sein, da die Bienen bei ihrer Nahrungssuche an behandelten Pflanzen den Substanzen ausgesetzt sind und diese möglicherweise damit auch an ihr Volk weitergeben. Die vorliegende Arbeit soll dazu beitragen, diese mögliche Gefährdung der Honigbiene durch Neonikotinoide weiter aufzuklären und deren Risiken zu beurteilen. Der Hintergrund für die Versuche zur Muskelspikeaktivität waren vorangegangene Versuche, die Auswirkungen von Neonikotinoiden auf die Motorik von sich frei bewegenden Individuen dokumentierten. In der vorliegenden Arbeit wurde untersucht, ob sich diese Effekte auch an der Spikeaktivität des Muskels M17 widerspiegeln. Dafür wurden Elektromyogramme des Muskels M17 zu verschiedenen Zeitpunkten nach der Gabe der Substanzen erstellt und deren mediane Anzahl mit einer Kontrollgruppe verglichen.
In einem ersten Versuch wurden Clothianidin (1 µM), Thiacloprid (1 µM), Imidacloprid (1 µM) oder eine Kontrollsubstanz (PBS) in die Kopfkapsel appliziert. Beim Vergleich mit der Kontrollgruppe zeigten sich für Imidacloprid keine Auswirkungen. Clothianidin verursachte eine deutlich erhöhte mediane Spikerate des Muskels M17, während Thiacloprid diese absenkte, beides im Vergleich zur Kontrollgruppe. Auch bei einer um 30 Minuten versetzten Doppelapplikation von Clothianidin (1 µM) und Thiacloprid (10 µM) stellten sich diese Effekte ein, wobei Clothianidin eine dominante Rolle einzunehmen scheint. Diese Ergebnisse stehen im Einklang mit Untersuchungen zur Laufaktivität, welche sich ebenfalls durch Clothianidin erhöhte und durch Thiacloprid absenkte.
Ein weiteres Experiment untersuchte die Auswirkung einer akuten Fütterung mit Clothianidin (1 ng in 1 µl) bzw. Thiacloprid (250 ng in 1 µl) auf die Anzahl der Muskelaktionspotenziale. Auch hier zeigt sich eine deutliche Erhöhung der Spikeanzahl durch Clothianidin und eine Absenkung der Spikeanzahl durch Thiacloprid, was die Ergebnisse des ersten Versuchs nochmals bestätigt.
Des Weiteren wurden Untersuchungen zur Auswirkung einer chronischen Fütterung mit Clothianidin (50 ppb) bzw. Thiacloprid (5000 ppb) auf die Anzahl an Spikes durchgeführt. Die Bienen wurden dabei über mehrere Wochen im Volk mit den jeweiligen Substanzen gefüttert. Dabei zeigte sich, dass auch eine chronische Fütterung der Bienen mit Clothianidin ihre Anzahl an Muskelaktionspotenzialen deutlich erhöht, während eine chronische Fütterung mit Thiacloprid diese absenkt. In einer Kombination der chronischen Fütterung mit einer zusätzlichen akuten Fütterung des jeweils anderen Neonikotinoids zeigte sich, dass auch hier Clothianidin eine dominante Rolle gegenüber Thiacloprid einnimmt und auch keine synergistischen oder Gewöhnungseffekte der beiden Substanzen eintreten. Die chronisch eingefütterten Völker entwickelten sich dagegen wie die Kontrollvölker, sodass die hier beschriebenen Auswirkungen auf die Einzelbiene keine sichtbaren Effekte auf ganze Völker zu haben scheinen.
Zusätzlich wurden Versuche zum Duftlernen in einer komplexen Lernaufgabe, dem positiven Patterning, unter Einfluss von Clothianidin durchgeführt. Die Bienen müssen dabei zwischen unbelohnten Einzeldüften und einem belohnten Duftgemisch, bestehend aus den beiden Einzeldüften, unterscheiden. In verschiedenen Versuchsdurchläufen wurden 0,25 ng oder 1 ng Clothianidin (jeweils 1 µl) in den Thorax injiziert, entweder vor der Akquisitionsphase oder vor dem ersten Abruftest nach drei Stunden. In keinem der Versuchsdurchläufe zeigten sich Effekte des Clothianidins auf den Lernvorgang, weder in der Akquisitionsphase noch in den Abruftests. Somit wurden weder das Lernen an sich, noch die Konsolidierung und damit die Überführung des Gelernten in das Langzeitgedächtnis durch das Clothianidin beeinflusst. Allerdings könnte die Immunabwehr der Bienen nach längerer Einwirkung des Clothianidins (24 h) in der höheren Konzentration herabgesetzt sein, da viele Bienen starben.
Insgesamt ergaben sich Effekte von Clothianidin und Thiacloprid auf die Anzahl der Aktionspotenziale des Muskels M17, die sich aber im gesamten Volk nicht widerspiegeln. Der Lernvorgang in der hier durchgeführten komplexen Lernaufgabe wird durch Clothianidin nicht beeinflusst. Möglicherweise entsteht dieser Unterschied in den Ergebnissen durch eine Bindung der Substanzen an verschiedene Rezeptorsubtypen, die pharmakologisch unterschiedliche Eigenschaften besitzen und somit auch unterschiedliche Auswirkungen zeigen.
In the first part of this work, the development of a novel two-dimensional native gel electrophoretic system (2-D BN/hrCNE) is described. This new system simplifies proteomics and biochemical analysis of mega protein complexes that are dissociated into the constituent complexes during 2-D electrophoresis, thereby reducing the complexity of the system considerably. This technique is exceptionally well suited for the in-gel detection of fluorescence-labeled proteins and the identification of individual enzymes and protein complexes by specific in-gel assays on native gels.
In the second part, a new technique for the native immunoblotting of blue native gels (NIBN) was developed. This new technique allows for the identification of conformation-specific antibodies and the discrimination of antibodies recognizing linear epitopes of denatured proteins. Identification of conformation-specific antibodies is becoming increasingly important not only for the electron microscopic identification of native proteins but also for structural investigations in general. For this purpose, a commonly used protocol for Western blotting of blue native gels was modified in such a way that the native state of proteins and protein complexes was retained throughout the complete protocol. Instead of using the denaturing methanol in Western blotting protocols, mild detergents such as Tween 20, digitonin and Brij 35 were used for the obligatory removal of protein bound Coomassie-dye.
The detection of respiratory complex I by activity staining on the blot membrane demonstrated that all three non-ionic detergents preserved the native state of complex I. The native state of the enzyme on the blot membrane was also monitored and confirmed with the help of a set of conformation-specific antibodies. NIBN can be used as a simple alternative method to the demanding native ELISA to screen for conformation-specific antibodies for structural studies. Unlike the time consuming native ELISA, NIBN does not require introduction of appropriate affinity tags and purification of the target protein by chromatography. Thus, the NIBN technique is especially useful for microscale projects and for proteins not easily accessible to genetic manipulation.
The third part aimed at identification of the immediate protein interaction partners of Cox26, a hydrophobic protein that has been identified by our group as a novel component of yeast respiratory supercomplex. Multi-dimensional electrophoretic techniques were applied to identify non-covalent and covalent protein-protein interactions of Cox26. Three-dimensional electrophoresis (BNE/BNE/SDS-PAGE) gave both qualitative and quantitative information on covalent and non-covalent interactions of Cox26 and subunits of cytochrome c oxidase (complex IV), and showed that most of the Cox26 protein was non-covalently bound to the complex IV moiety of the respirasomes. Four-dimensional electrophoresis (BNE/BNE/SDS/SDS-PAGE) applying reducing and non-reducing conditions revealed that a minor fraction of Cox26 used a single cysteine residue in the center of a predicted transmembrane helix to form a disulfide bond with the Cox2 subunit of complex IV. A structural role of Cox26 protein in the assembly/stability of respiratory strings or patches has been suggested.
The last part of this work focused on the isolation and characterization of native and morphologically intact nucleoids from bovine heart mitochondria, since only a few studies on nucleoid organization and composition have been carried out on mammalian tissues. The nucleoids appeared as distinct bands (apparent mass around 30-36 MDa) in blue native-PAGE on large pore gels. The moderate variation in particle size seems to reflect variations in the binding of loosely nucleoid-associated components like respiratory chain complexes. The estimated 30-36 MDa mass of nucleoids on native gels suggested that each nucleoid contains one mtDNA molecule provided that nucleoids contains equal amounts of DNA, protein and RNA (Miyakawa et al., 1987).
Electron microscopic analysis of native nucleoids, which was performed by Dr. Karen Davies from the Max-Planck-Institute of Biophysics, Department of Structural Biology, Frankfurt, showed homogenous pool of particles with dimensions in 85x100 nm (in negative stain) and 100x150 nm (in cryo-tomography). Some of the nucleoids showed dumbbell-shape indicating dimerization of nucleoids. Recent EM and high-resolution light microscopy analysis of mammalian nucleoids have reported that nucleoids have a size of 70 nm in average. We also observed the same size of 70 nm in cryo-tomogramms when we applied harsher treatment of the native nucleoid particles with dimensions 100x150 nm. This observation is in agreement with published nucleoid sizes from both EM and high-resolution light microscopy, if we assume that native nucleoids have been dissociated under harsher treatment.
The protein composition of bovine heart mt-nucleoids was analyzed by a number of complementary approaches to identify low and highly abundant, easily dissociating and tightly bound proteins, and to rank the 90 most abundant mt-nucleoid proteins. Native and denaturing gel electrophoresis techniques were coupled to LC-MS/MS to achieve a comprehensive protein component analysis. Qualitative MS analysis of highly purified nucleoids identified more than 400 proteins, including well known nucleoid proteins such as mitochondrial transcription factor and mtDNA-binding protein (TFAM), mitochondrial single-stranded DNA-binding protein (mtSSB), mitochondrial DNA polymerase subunit gamma-2 (POLG2) and mitochondrial helicase C26H10ORF2 protein (Twinkle). These proteins were ranked according to Mascot scores, and sorted according to presumed functional properties. A large group of proteins involved in protein synthesis comprised an almost complete set of subunits of mitochondrial ribosomes suggesting that the nucleoids contained significant amounts of mitochondrial ribosomes. Identification of sixty six proteins from the oxidative phosphorylation (OXPHOS) system comprising around 100 proteins in total suggested that OXPHOS proteins are also associated with mt-nucleoids.
Interestingly, TFAM, described as a main mtDNA packaging factor in human and other mammalian cells, was not confirmed here as a major nucleoid component from bovine heart mitochondria. Fluorescence staining of protein spots on 2-D IEF/SDS gels clearly identified TFAM, but according to the stain intensity, this protein did not rank in the list of the 90 most abundant nucleoid proteins. Western blot analysis of sucrose gradient fractions revealed an enrichment of putative TFAM isoform in nucleoid fractions. Unexpectedly, the uncharacterized mitochondrial protein Es1 was identified as the most abundant nucleoid protein in bovine heart nucleoids instead. This implicates that nucleoid organization may differ between species and tissues. A functional characterization of Es1 is required to clarify its role in mammalian nucleoids.
One of the earliest consequences of slicing plant storage organs such as potato tubers into thin disks is the formation of polysomes, which in potato slices is complete after 9 hours and is dependent on transcription. Fresh disks do not incorporate 32P, 3H-uridine or 14C-leucine into their ribosomes, whereas ribosomes and polysomes of aged disks use these precursors effectively. This development can be completely blocked by actinomycin D. Among the different RNAs synthesized during aging is 28S- and 16S—rRNA, 5S—RNA, tRNA, and a component sedimenting around 15—18S with a base-composition different from 16S—rRNA, 5S- and 4S—RNA and which supports peptide formation in an in vitro incorporation system.
It is suggested that this compound represents mRNA, which is not available immediately after slicing the tissue. These findings are consistent with the view of a derepression phenomenon in sliced storage tissue.
Whereas ribosome preparations of freshly sliced potato disks do not show appreciable activity in an in-vitro amino acid incorporation system, aging of the tissue leads to a greatly enhanced incorporation activity which reaches its maximum 24 hours after slicing. If ribosomes from freshly excised disks are provided with polyuridylic acid, their activity in the incorporation of phenylalanine is increased about 8 fold.
Moreover, an RNA-fraction can be dissociated by EDTA from ribosomes of aged potato tuber slices, which sediments at 15 —18S, has a base composition different from that of 16S — rRNA, 5S-and 4S —RNA, and is not present on ribosomes of fresh slices. Its appearance is inhibited by actinomycin D and therefore most probably dependent on transcription. This compound, purified from sucrose gradients, enhances in vitro leucine incorporation into peptide material by ribosomes of fresh potato slices.
The possibility is discussed that this fraction-among other factors-is responsible for the enhanced protein synthesis after slicing plant storage organs, and is indicative of a general derepression phenomenon in these tissues.
Resting potato tuber tissue possesses only faint activity of the two dehydrogenases of the oxidative pentose phosphate cycle, glucose-6-phosphate- and 6-phosphogluconate dehydrogenase. Slicing of the tissue, however, greatly enhances the action of both enzymes. The slicing-induced increase in activity is a consequence of intensified action of at least 5 glucose-6-phosphate dehydrogenase isozymes and a more differentiated activation/inactivation of seven 6-phosphogluconate dehydrogenase isozymes.
Using density labelling and isopycnic equilibrium centrifugation it could be demonstrated, that the bulk of both enzymes appearing after slicing the tissue is the result of de novo synthesis rather than activation of pre-existing proenzymes.
Differential derepression of the genome of potato tuber cells can be initiated by slicing the tissue into disks. The consequence of this procedure on the cells of the wound surface is dedifferentiation and cell division followed by redifferentiation to a suberized phellem cell. The drift of glucose-, glucose-1-phosphate-, glucose-6-phosphate-, fructose-6-phosphate- and 6-phospho-gluconatelevels has been determined in the derepressed tissue. With the exception of 6-phospho-gluconate all intermediates so far investigated showed a rise in concentration after derepression.
This is interpreted as a consequence of altered enzymic activities which were estimated for phosphoglucomutase, hexokinase, phosphoglucoisomerase, gluco-6-phosphate- and 6-phosphogluconatedehydrogenase. The two dehydrogenases were activated after derepression, the activation represented a de-novo-synthesis, as was demonstrated with the inhibitors Actidione (translation) and p-Fluorophenyl-alanine (protein synthesis in general). Hexokinase and phosphoglucoisomerase were not severely affected by cutting the tissue. Phosphoglucomutase was degrated rapidly, the degradation being dependent on protein synthesis. The importance of an enhanced activity of the pentose phosphate shunt for the stressed cell is emphasized and the possibility of an alteration in the osmotic pressure within the cell and especially in the nucleus — a primary consequence of wounding — as a cause of derepression in potato tuber cells is discussed.
Chromatin, RNA Polymerase, Potato Tuber Tissue, Aging Phenomenon The synthesis of RNA by chromatin-bound RNA polymerase (E.C. 2.7.7.6.) from white potato tubers proceeds at a low rate, which is enhanced after slicing the tissue, however. Concomitantly DNA template availability as measured with saturating amounts of Escherichia coli polymerase is diminished drastically. Nearest neighbor frequency analysis proved that the RNA synthesized on chromatin of intact tubers is different from that synthesized on chromatin of sliced tissue.
The RNA polymerase of white potato tubers is dependent on all four ribonucleoside triphos phates and a divalent metal ion such as Mg2+ or Mn2+ and totally inhibited by the presence of pyrophosphate. Actinomycin D blocks the formation of the RNA product, which could be shown to be a heteropolymer by nearest neighbour frequency technique. The Km of the chromatin-bound enzyme with regard to ATP, GTP, CTP and UTP was 5.1 X10-5 M, 1.6X10-5 M, 0.9X10-5 M and 0.45 X 10-5M/1 respectively, α-amanitin inhibits the overall activity to about 50%, which indicates the presence of equal amounts of polymerase I and polymerase If.
Pflanzenfunde aus archäologischen Ausgrabungen im Norden Burkina Fasos dokumentieren die regionale Geschichte der Pflanzennutzung und damit zusammenhängende Wechselwirkungen zwischen Mensch und Umwelt. Das untersuchte Material besteht überwiegend aus verkohlten Früchten und Samen, die durch das Schlämmen von Kultursedimenten aus mehr als 20 Fundplätzen gewonnen wurden. Sie decken einen Zeitraum von etwa 4000 Jahren ab, der von der Endsteinzeit bis in die jüngste Neuzeit reicht. Die Analysen der archäobotanischen Inventare ermöglichen es, mehrere Phasen der Pflanzennutzung auszuweisen, die mit spezifischen Lebens- und Siedlungsweisen der Bevölkerung assoziiert sind. Eine ausschließlich wildbeuterische Wirtschaftsform wird für die älteste der untersuchten Fundstellen, den Lagerplatz Corcoba, angenommen. Dort nutzten mobile Gesellschaften um 2000 BC reiche Fischressourcen und sammelten systematisch die Früchte von Gehölzen. Im Inventar des Fundplatzes Tin Akof ist um 1800 BC erstmals eine Kulturpflanze, die Perlhirse (Pennisetum glaucum) nachweisbar. Sie markiert den Beginn der produzierenden Wirtschaftsweise. Es wird ein Anbau in kleinem Maßstab rekonstruiert, wobei Feldbau nur eine von mehreren praktizierten Subsistenzstrategien war. Vermutlich handelte es sich bei den Bewohnern von Tin Akof um seminomadische Viehhalter, die aus dem Sahararaum einwanderten und das bereits domestizierte Getreide einführten. Ab der Zeitenwende tritt in der Eisenzeit eine neue, sesshafte Kultur in Erscheinung. Ihre Siedlungen befanden sich vorzugsweise auf sandigen, leicht kultivierbaren Böden in der Nähe permanenter Gewässer. Die Nahrungsproduktion basierte auf der Kultivierung von Perlhirse, daneben wurden Hibiscus cf. sabdariffa und die Hülsenfrüchte Vigna subterranea und V. unguiculata angebaut. Als Anbausysteme lassen sich Mischkulturen mit Perlhirse als Hauptfrucht und Kulturbaumparks, die vom Menschen geschätzte Gehölze (u.a. Vitellaria paradoxa) aus der ursprünglichen Savannenvegetation einbeziehen, rekonstruieren. Die gemischte Wirtschaftsweise umfasste Viehhaltung, aber auch wildbeuterische Praktiken wie das Sammeln von Wildpflanzen, insbesondere von Baumfrüchten. Hinweise auf Handelskontakte liegen aus der zweiten Hälfte des ersten Jahrtausends AD vor. Sie lassen sich zum Teil mit dem Ausbau transsaharischer Handelsnetze im Verlauf der Islamisierung Westafrikas verknüpfen. Sorghum bicolor und die Wassermelone (Citrullus lanatus) wurden möglicherweise auf diese Weise eingeführt. Die Eisenzeit erweist sich insgesamt als stabile Epoche mit langer Siedlungskontinuität. Gleichwohl zeigen die detailliert untersuchten Fundsequenzen sich wandelnde Nutzungsmuster und eine Intensivierung der Landwirtschaft. Im 14. Jahrhundert werden die für die Epoche typischen Siedlungshügel im gesamten Gebiet verlassen. Mögliche Ursachen sind politische Veränderungen in benachbarten Regionen. Erst aus der jüngsten Neuzeit liegen wieder archäobotanische Belege vor. Die Ergebnisse aus Burkina Faso bestätigen die archäobotanischen Forschungen in anderen Gebieten Westafrikas, nach denen Feldbau relativ spät um 2000 BC begann und Perlhirse die erste domestizierte Kulturpflanze darstellt. Die stabile Bedingungen in der Eisenzeit führten vielerorts zur Entstehung von Städten und Handelszentren. Diese Entwicklung ist im ländlichen Raum, zu der auch die Arbeitsregion zählt, weniger deutlich ausgeprägt, aber dennoch fassbar. Die archäobotanischen Inventare der Endsteinzeit und Eisenzeit dokumentieren ein, im Vergleich zu heute, niederschlagsreicheres Klima und einen geringeren anthropozoogenen Einfluss auf die natürliche Vegetation.
Activation of Notch1 signaling in neural progenitor cells (NPCs) induces self-renewal and inhibits neurogenesis. Upon neuronal differentiation, NPCs overcome this inhibition, express proneural genes to induce Notch ligands, and activate Notch1 in neighboring NPCs. The molecular mechanism that coordinates Notch1 inactivation with initiation of neurogenesis remains elusive. Here, we provide evidence that Prox1, a transcription repressor and downstream target of proneural genes, counteracts Notch1 signaling via direct suppression of Notch1 gene expression. By expression studies in the developing spinal cord of chick and mouse embryo, we showed that Prox1 is limited to neuronal precursors residing between the Notch1+ NPCs and post-mitotic neurons. Physiological levels of Prox1 in this tissue are sufficient to allow binding at Notch1 promoter and they are critical for proper Notch1 transcriptional regulation in vivo. Gain-of-function studies in the chick neural tube and mouse NPCs suggest that Prox1-mediated suppression of Notch1 relieves its inhibition on neurogenesis and allows NPCs to exit the cell cycle and differentiate. Moreover, loss-of-function in the chick neural tube shows that Prox1 is necessary for suppression of Notch1 outside the ventricular zone, inhibition of active Notch signaling, down-regulation of NPC markers, and completion of neuronal differentiation program. Together these data suggest that Prox1 inhibits Notch1 gene expression to control the balance between NPC self-renewal and neuronal differentiation.
