Ziel dieser Arbeit war es, die Zytotoxizität von Treosulfan und Busulfan auf Leukämiezellen von pädiatrischen Patienten mit Akuten Leukämien zu untersuchen. Im Rahmen dieser Arbeit wurden in vitro und erste in vivo Untersuchungen durchgeführt: 1. In vitro Untersuchungen Zuerst wurde eine durchflusszytometrische Methode optimiert zum Nachweis der Zytotoxizität von Treosulfan und Busulfan auf maligne Zellen pädiatrischer Patienten mit Akuten Leukämien. Mit diesem durchflusszytometrischen Assay war es möglich, die Zytotoxizität auf maligne von der auf nicht maligne Zellen zu unterscheiden. Dies war unerlässlich, da in den frisch isolierten Leukämiezellen der Patienten bis zu 55% normale Lymphozyten enthalten waren. Darüber hinaus erlaubte diese Methode die simultane Bestimmung der Zell-Apoptose in jeder Probe. An Leukämie-Zelllinien wurde dieser multiparametrische Assay anschließend mit dem MTT-Assay verglichen. Es konnte gezeigt werden, dass die Bestimmung von Zytotoxizitäten mit beiden Methoden an den Zelllinien Molt 4/8 und H9 gut korrelierte (r≥0,95). Für das Arbeiten mit Patientenmaterial wurde ausschließlich die durchflusszytometrische Methode angewendet, da in den Proben der Patienten die Differenzierung zwischen leukemischen Zellen und normalen Lymphozyten essentiell war. In den Leukämie-Zelllinien Molt4/8, H9 und K562 zeigte sich, dass Treosulfan eine stärkere Zytotoxizität zeigte als Busulfan. In die Untersuchungen frischer Leukämiezellen pädiatrischer Patienten mit Akuten Leukämien konnten 24 Proben unterschiedlicher Leukämien (cALL, reife B-ALL, reife TALL, AML) und unterschiedlicher Erkrankungszeitpunkte (bei Diagnose oder bei Rezidiv) eingeschlossen werden. In diesen Proben zeigte Treosulfan eine deutlich bessere Wirkung als Busulfan. Es wurde trotz des kleinen Patientenkollektivs deutlich, dass die T-ALL gegenüber der cALL sensitiver auf Treosulfan reagiert. Außerdem war die Zytotoxizität von Treosulfan und Busulfan gegenüber der T-ALL signifikant höher als gegenüber c-ALL (Treosulfan: p=0,02, Busulfan: p=0,03). Die IC50-Werte stiegen vom Zeitpunkt der Diagnose (Median: Treosulfan: 11,45 μM, Busulfan: 96,45 μM) über die Progression (Median: Treosulfan: 45,95 μM, Busulfan: 253,75 μM) bis zum Rezidiv (Median: Treosulfan: 153,15 μM, Busulfan: 223,3 μM) hin an, und zwar um das 8-fache bei Treosulfan und das 2,5-fache bei Busulfan. Der Unterschied der Zytotoxizität von Treosulfan auf Leukämieproben zum Zeitpunkt der Diagnose gegenüber dem Rezidiv war statistisch signifikant (p=0,02). Für Busulfan ergab sich für diese Untersuchungszeitpunkte keinen signifikanten Unterschied (p=0,13). Vergleichend zu den Ergebnissen an frisch isolierten Leukämiezellen wurde die Wirkung von Treosulfan und Busulfan auf normale Lymphozytensubpopulationen und Stammzellen untersucht. Auch hier zeigte Treosulfan eine stärkere Zytotoxizität im Vergleich zu Busulfan. Insgesamt reagierten normale Lymphozyten sensitiver auf die Alkylanzien im Vergleich zu den Leukämiezellen (Mediane IC50-Werte für Treosulfan und Busulfan auf Lymphozyten: 12,3 μM und 89,9 μM, auf Leukämieproben: 30,6 μM und 133 μM mit p=0,03 und p=0,02). In einem weiteren Experiment sollte die Interaktion von Treosulfan und Busulfan mit Fludarabin untersucht werden. Fludarabin ist ein Purin-Analogon, das in der Pädiatrie in Kombination mit Alkylanzien zur Chemo-Konditionierung eingesetzt wird. Bei der Inkubation von Fludarabin mit Treosulfan-Konzentration größer 1 μM war ein deutlicher Synergismus zu verzeichnen. Die Kombination von Fludarabin mit Busulfan ergab Antagonismus. 2. In vivo Untersuchungen Es wurde die zelluläre Immunrekonstitution bei pädiatrischen Patienten mit AML (n=9) nach allogener Knochenmarktransplantation überwacht. Ein Patient wurde mit Treosulfan konditioniert (n=1), die anderen Patienten (n=8) erhielten Busulfan zur Konditionierung. Die Rekonstitution der Leukozyten, B-Zellen, NK-Zellen und T-Helferzellen verlief in beiden Gruppen ähnlich. Ein signifikanter Unnterschied konnte für die Rekonstitution der CD3+CD8+ zytotoxischen T-Zellen gezeigt werden, die bei dem Patienten, der mit Treosulfan konditioniert wurde, signifikant niedriger war im Vergleich zur Busulfangruppe. Da bei dem Patienten, der Treosulfan erhielt, die CRP-Werte über einen längeren Zeitraum erhöht waren und Infektionen die Rekonstitution von zytotoxischen T-Zellen maßgeblich beeinflussen, bietet dies eine mögliche Erklärung für diesen Unterschied. Aussagen können jedoch nur mit einem größeren Patientenkollektiv getroffen werden.
Der Extrakt des indischen Weihrauchs (Boswellia serrata) ist eines der wenigen pflanzlichen Heilmittel, dem von der EMEA der Orphan Drug Status zur Behandlung des peritumoralen Hirnödems verliehen wurde. Boswellia serrata Extrakt und die Boswelliasäuren, die wirksamen Inhaltsstoffe des Weihrauchs, zeigten in zahlreichen in vitro-Untersuchungen antiinflammatorische und antitumorale Wirksamkeit. Diese Wirkungen konnten auch in mehreren klinischen Studien nachgewiesen werden. Untersuchungen zum pharmakokinetischen Verhalten der Boswelliasäuren zeigten, dass Weihrauch nur eine geringe orale Bioverfügbarkeit aufweist. Ziel der Arbeit war es daher, den Einfluss von Löslichkeit, Metabolismus und Permeabilität auf die Bioverfügbarkeit der Boswelliasäuren zu untersuchen. Weihrauchextrakte sind in wässrigen Medien schlecht löslich. In einer Rattenstudie wurde deshalb untersucht, inwieweit die verbesserte Löslichkeit des Extrakts in einer nanoskaligen Boswellia serrata Formulierung zu einer verbesserten Bioverfügbarkeit führt. Eine bestehende LC-MS-Methode zur Bestimmung von KBA und AKBA aus Plasma und Hirngewebe wurde optimiert und revalidiert. Zur Vervollständigung des pharmakokinetischen Profils wurden die KBA- und AKBA-Konzentrationen auch in der Leber der Ratten bestimmt. Die analytische Methode wurde hierfür nach den anerkannten FDA-Richtlinien erfolgreich validiert. Die Plasma- und Leberkonzentrationen waren jedoch bei den Ratten, die die nanoskalige Boswellia serrata Formulierung bekamen, in den ersten Stunden nach der oralen Verabreichung nicht signifikant höher als bei den Ratten, die den unbehandelten Extrakt erhielten. Die in dieser Arbeit durchgeführten Untersuchungen zur metabolischen Stabilität von KBA und AKBA in Rattenlebermikrosomen (RLM), Humanlebermikrosomen (HLM) und Rattenhepatozyten (RH) zeigten, dass KBA einer stark ausgeprägten hepatischen Metabolisierung unterliegt. AKBA hingegen erscheint metabolisch relativ stabil. Die Identifizierung der Metabolite ergab, dass Boswelliasäuren in RLM hauptsächlich Phase-I-Metabolite wie mono-, di-, und seltener auch trihydroxylierte Metabolite bilden. Von AaBA und AbBA konnten keine Metabolite detektiert werden. Das metabolische Profil von KBA und AKBA in RH war mit dem in RLM vergleichbar. In einer Rattenstudie konnten dann im Plasma und in der Leber jedoch nicht im Hirn der Ratten KBA-Metabolite nachgewiesen werden, während für AKBA in vivo keine Metabolite detektiert wurden. Phase-II-Metabolite konnten weder von KBA noch von AKBA nachgewiesen werden. Bisher war man davon ausgegangen, dass die niedrigen Plasmakonzentrationen von AKBA in vivo durch eine Deacetylierung zu KBA zustande kommen. Diese These konnte im Rahmen dieser Arbeit widerlegt werden. Im Caco-2-Zellmodell zeigte KBA eine mittlere Permeabilität. Es konnte gezeigt werden, dass KBA und AKBA offensichtlich keinem Efflux-Transport unterliegen. AKBA erwies sich sowohl in absorptiver und sekretorischer Richtung als auch bei 4° C als schlecht permeabel. Da KBA und AKBA die Aktivität des ABC-Transportproteins Pgp modulieren, wurde in dieser Arbeit überprüft, ob diese beiden Boswelliasäuren auch Substrate des Pgp sind. Die Permeation von KBA und AKBA war in Anwesenheit des Pgp-Inhibitors Verapamil jedoch nicht signifikant verändert, was darauf hindeutet, dass KBA und AKBA keine Pgp-Substrate sind. Die Ergebnisse dieser Arbeit bilden einen wichtigen Baustein zur weiteren Aufklärung des pharmakokinetischen Verhaltens von KBA und AKBA. So ist die begrenzte systemische Verfügbarkeit von KBA auf eine mittlere Absorption aus dem Gastrointestinaltrakt in Kombination mit der umfangreichen hepatischen Metabolisierung zurückzuführen. Die niedrigen systemischen Konzentrationen von AKBA hingegen liegen eher in der schlechten Absorption begründet. Auf der Basis der extensiven Metabolisierung von KBA und der schlechten Permeabilität von AKBA stellt sich im Allgemeinen die Frage nach dem tatsächlichen Wirkmechanismus von KBA und AKBA. In keiner pharmakokinetischen Studie konnten die in vitro pharmakologisch aktiven Konzentrationen dieser beiden Boswelliasäuren erzielt werden. Es ist daher nicht auszuschließen, dass auch andere Wirkmechanismen als die bisher beschriebenen existieren. Unter dem Gesichtspunkt möglicher Arzneimittelinteraktionen wurde die Wirkung von KBA und AKBA auf MRP2 und OATP1B3 in zwei zellbasierten Assays untersucht. Es konnte gezeigt werden, dass KBA und AKBA die Aktivität von MRP2 und OATP1B3 in Konzentrationen modulieren, welche im Rahmen dieser Arbeit in der Leber von Ratten nachgewiesen wurden. Da Weihrauchextrakt häufig in Comedikation verwendet wird, sollte im Hinblick auf die Arzneimittelsicherheit in Zukunft geprüft werden, ob es zu praxisrelevanten Arzneimittelinteraktionen mit klinisch relevanten MRP2- und OATP1B3-Substraten kommt.
IL-22 ist ein TH17-Signaturzytokin und wird primär von aktivierten T- und NK-Zellen produziert. Die Literatur beschreibt IL-22 als immunregulatorisches Signalmolekül, welches vor allem bei Infektionen und Entzündungsprozessen eine wichtige Rolle spielt. Typische IL-22-induzierte Gene sind beispielsweise Akut-Phase-Proteine, β-Defensine sowie Matrixmetalloproteasen. In dieser Arbeit wurde erstmals gezeigt, dass IL-22 die IFNγ-induzierte iNOS-Expression in humanen DLD-1 Kolonkarzinomzellen massiv verstärkt. Eine gestörte iNOS-Induktion mit folglich extensiver NO-Produktion trägt zu entzündlichen Veränderungen und zur Tumorentstehung bei. Insbesondere gibt es stichhaltige Belege, welche iNOS und NO in Verbindung mit Krebs als potentes Mutagen, als Angiogenesefaktor, als Verstärker der Protoonkogenexpression und als Inhibitor der Apoptose charakterisieren. Demzufolge ist gerade die Identifikation von Signaltransduk-tionswegen von Interesse, welche das Potential zur Regulation der iNOS-Expression in epithelialen Kolonkarzinomzellen besitzen. Der Nachweis des IL-22-Rezeptorkomplexes sowie einer STAT3-Phosphorylierung belegte die IL-22-Responsivität der hier verwen-deten DLD-1 Kolonkarzinomzellen. Der Transkriptionsfaktor STAT3 gilt als prominentes Signalmolekül in der Signaltransduktionskette von IL-22. In der aktuellen Literatur wird STAT3 aufgrund seiner antiapoptotischen und proliferativen Eigenschaften als Onkogen definiert. Sowohl in Patienten mit Morbus Crohn als auch im humanen Kolorektalkarzinom ist eine Überexpression von STAT3 und iNOS beschrieben. Unter Verwendung einer STAT3-spezifischen siRNA konnte der Nachweis erbracht werden, dass STAT3 ein essen-tieller Faktor in der IL-22-vermittelten Induktion der iNOS-Expression ist. Luciferase-Reporterstudien zeigten, dass IL-22 die humane iNOS-Promotoraktivität in DLD-1 Zellen, welche die Photinus pyralis (Firefly)-Luciferase unter Kontrolle eines 16kb-iNOS-Promotor-konstrukts überexprimieren (DLD-1/pXP2-16kb), erhöht. Somit kann eine Regulation der Transkription durch IL-22 angenommen werden. Dennoch vermag diese Arbeit die Frage nach den transkriptionellen Regulationsmechanismen der IL-22-vermittelten iNOS-Expres-sion in DLD-1 Zellen nicht abschließend zu beantworten. Eine Beteiligung ausgewählter Parameter der Zellaktivierung wie beispielsweise NFκB, STAT1α oder IRF-1 an der IL-22-vermittelten iNOS-Induktion konnte durch die vorliegende Arbeit ausgeschlossen werden. Neben transkriptionellen Regulationsmechanismen übernimmt auch die Regulation der iNOS mRNA-Stabilität eine wichtige Funktion bei der Induktion der iNOS-Expression. Allerdings zeigten auch hier die Ergebnisse der vorliegenden Arbeit, dass IL-22 die iNOS-mRNA nicht stabilisieren kann. Dies bekräftigt wiederum die Annahme, dass IL-22 auf transkriptioneller Ebene den iNOS-Promotor reguliert. Weitere Versuche zeigten auch, dass IL-22 spezifisch an der iNOS-Expression in Kolonkarzinomzellen beteiligt ist und keinen generellen Verstärker der IFNγ-induzierten Genexpression darstellt. Beachtet man hierbei die bedeutende Rolle von STAT3 und iNOS bei Entzündung und Karzinogenese, so repräsentiert IL-22 möglicherweise eine neue Zielstruktur für immuntherapeutische Interventionen im Rahmen dieser Erkrankungen. Der zweite Abschnitt der vorgelegten Arbeit beschäftigte sich mit der Regulation der IL-22-Sekretion im Kontext von Sepsis und systemischer Entzündung. Es stellte sich die Frage, inwiefern IL-22 in diesem komplexen klinischen Syndrom der Sepsis nachzuweisen ist. In diesem Zusammenhang zeigte sich, dass sowohl eine generelle T-Zellaktivierung sowie verschiedene Stimuli der angeborenen Immunität, hier im Besonderen ein hitze-inaktiviertes Lysat von Staphylococcus epidermidis, eine vermehrte IL-22-Produktion anre-gen. Eine wichtige Frage ist hier die pharmakologische Beeinflussung der IL-22-Sekretion. Da bei der Therapie einer Sepsis die Behandlung mit niedrig dosierten Glukokortikoiden Erfolg versprechend ist, wurde im Folgenden der Einfluss des klassischen antientzünd-lichen Pharmakons Dexamethason auf die IL-22-Sekretion in PBMC untersucht. Es konnte eindrucksvoll gezeigt werden, dass Dexamethason sowohl auf mRNA- als auch auf Proteinebene einen sehr potenten Inhibitor der S. epidermidis-induzierten IL-22-Sekretion darstellt. Um die in vitro-Daten der IL-22-Sekretion nach Immunaktivierung humaner PBMC durch in vivo-Experimente zu ergänzen, wurde ein kürzlich etabliertes Nagermodell der schweren Sepsis mit akutem Verlauf verwendet (CLI; caecum ligation and incision). In diesem Modell ist IL-22 im Vergleich zu unbehandelten Tieren signifikant erhöht. In Über-einstimmung mit den in vitro-Experimenten in humanen PBMC hat die Applikation von Dexamethason im CLI-Modell die Hemmung der IL-22-Produktion zur Folge. Zur Abrun-dung dieser Ergebnisse wurde das Serum von Patienten mit einer Peritonitis-bedingten Sepsis untersucht und es konnte erstmals eindrucksvoll gezeigt werden, dass IL-22 in dieser Patientengruppe signifikant erhöht ist. Zum aktuellen Zeitpunkt ist die Rolle von IL-22 in der Sepsis noch nahezu unbekannt. Daher ist zu klären, wie die eingehend beschriebene IL-22-vermittelte Aktivierung der epithelialen Immunabwehr, die systemische Immunsuppression über die Induktion von IL-10 sowie das proinflammatorische und destruktive Potential von IL-22 in Einklang zu bringen sind.
Der zur Familie der ionotropen Glutamatrezeptoren gehörende N-Methyl-DAspartat (NMDA)-Rezeptor ist maßgeblich an der Weiterleitung erregender Signale zwischen Nervenzellen beteiligt. Er spielt sowohl physiologisch bei z.B. Vorgängen des Lernens oder der Gedächtnisbildung, als auch pathophysiologisch bei neurologischen Erkrankungen eine entscheidende Rolle. NMDA-Rezeptoren sind tetramere Membranproteine, welche aus den homologen NR1-, NR2A-NR2D- sowie NR3A- und NR3B-Untereinheiten aufgebaut sind. Die Untereinheiten sind modular aus jeweils vier verschiedenen Domänen aufgebaut, die spezifische Rollen beim Aufbau und der Funktion der Rezeptoren erfüllen. Konventionelle NR1/NR2-NMDA-Rezeptoren bestehen aus zwei Glyzin-bindenden NR1- und zwei Glutamat-bindenden NR2-Untereinheiten. Sie werden nur durch gleichzeitiges Binden der Agonisten Glutamat und Glyzin effizient aktiviert. Ziel der vorliegenden Arbeit war, den Einfluss der extrazellulären N-terminalen Domänen (NTDs) auf die Assemblierung, Funktion und allosterische Modulation von rekombinanten NR1/NR2 NMDA-Rezeptoren mittels biochemischer und elektrophysiologischer Methoden zu untersuchen. Deletionsexperimente zeigten, dass die NTDs von NR1- und NR2A- bzw. NR2B-Untereinheiten die hochaffine, allosterische Zn2+- und Ifenprodil-Hemmung bestimmen, nicht aber für die Bildung funktioneller Rezeptoren von Bedeutung sind. Die NR2-NTDs stellen zusätzlich eine entscheidende strukturelle Determinate für die unterschiedliche Glyzinaffinität von NR1/NR2A- und NR1/NR2B-Rezeptoren dar. Ein zweiter Aspekt war die funktionelle Charakterisierung von NMDA-Rezeptoren, welche aus NR1- und NR3-Untereinheiten aufgebaut sind. Diese exzitatorischen NR1/NR3-Rezeptoren werden ausschließlich durch den Neurotransmitter Glyzin aktiviert und generieren nur sehr kleine Agonistaktivierte Ströme im Vergleich zu NMDA-Rezeptoren vom NR1/NR2-Typ. Es wurde gefunden, dass die Glyzinbindung an die NR1- und NR3-Ligandenbindungsdomänen (LBDs) entgegengesetzte Wirkungen auf die Rezeptorfunktion zur Folge hat. Während die NR3-LBD essentiell für die Aktivierung des Rezeptors ist, bewirkt Glyzin über die NR1-LBD eine Hemmung der NR1/NR3-Rezeptoren. Das erklärt die geringe Effizienz der Rezeptoraktivierung durch Glyzin. Weiterhin zeigen die Ergebnisse zum ersten Mal, dass Zn2+ an diesen Rezeptoren als Agonist und positiver Modulator wirkt und in Kombination mit einem NR1-Antagonisten die Glyzin-aktivierten Ströme >120-fach in supralinearer Weise potenzieren kann. Mutationsanalysen ergaben, dass die NR1-LBD für die Zn2+-Aktivierung und –Potenzierung verantwortlich ist. Da die physiologische Rolle von NR1/NR3-Rezeptoren noch nicht eindeutig geklärt ist, könnte die supralineare Potenzierung eine Strategie darstellen, diesen unkonventionellen NMDA-Rezeptor in zukünftigen Untersuchungen besser zu detektieren und zu charakterisieren. Zusammenfassend liefern die in dieser Arbeit gewonnenen Erkenntnisse zu Struktur-Funktionsbeziehungen in NMDA-Rezeptoren auf Ebene der NTDs und LBDs einen wichtigen Beitrag für das Verständnis der Pharmakologie dieser Rezeptorfamilie. Diese Ergebnisse können für die Entwicklung neuer neurologischer Therapeutika genutzt werden.
Cytochrome P450 epoxygenases of the 2C family (CYP2C) are highly expressed in the endothelium and metabolize arachidonic acid to different regioisomers of epoxyeicosatrienoic acids (EET). They have a number of roles in the regulation of vascular tone and homeostasis by activating different signal transduction pathways and have recently been reported to be involved in proliferation and angiogenesis. However, the exact mechanisms by which epoxygenases regulate angiogenesis are still unclear. Therefore, the initial aim of the present study was to characterize the relevance of major signalling molecules that are involved in angiogenesis and to investigate possible signalling pathways involved. Initially the effect of CYP2C9 overexpression on expression levels of EphB4, a tyrosine kinase that plays a role in a number of developmental processes, was investigated. EphB4 protein expression was increased in CYP2C9 overexpressing cells without any effects on expression levels of its ligand ephrinB2. To clarify whether EphB4 is a critical determinant of CYP2C9-induced angiogenesis, endothelial cell sprouting was assessed using a collagen gel-based in vitro angiogenesis assay. Following transfection with EphB4 antisense or scrambled oligonucleotides, capillary-like structures were clearly present after 24 hours in cells overexpressing CYP2C9, while EphB4 downregulation abolished CYP2C9-induced sprouting. In addition stimulation of human umbilical vein endothelial cells with VEGF resulted in an increase in CYP2C expression and a subsequent increase of 11,12-EET production; an effect that was abolished by the CYP epoxygenases inhibitor MSPPOH as well as when cells were infected with a dominant negative mutant of AMPK. In vivo 11,12-EET treatment increased EphB4 expression in mesenteric arteries as well as in Matrigel plugs; an effect that was abolished when plugs were impregnated at the same time with small interfering RNA (siRNA) for EphB4. Furthermore, impregnation of Matrigel plugs with VEGF resulted in endothelial cell and smooth muscle cell recruitment into a Matrigel plug and this effect was mediated by CYP2C9-derived EETs as it was prevented by 14,15-EEZE. When infiltration of EET impregnated plugs with endothelial cells and pericytes/smooth muscle cells in vivo was compared to the effects seen in VEGF treated plugs, it was apparent that only EET treatment resulted in the formation of tube like structures that were covered by smooth muscle cells. Therefore, the final aim of the study was to further define the consequences of EET signalling in vivo as well as to characterize its physiological relevance. This hypothesis could be assessed by isolectin injection through the tail-vein where isolectin was taken up only by the EET-impregnated plug. Moreover ultrasound measurements revealed accumulation of contrast agent in EET impregnated plugs compared to control plugs. Taken together our findings emphasize that CYP2C plays a crucial role in the vessel formation process by modulating the effects mediated by two important control elements of the angiogenic response, namely VEGF and EphB4. CYP2C-derived EETs not only participate as second messengers in the angiogenic response, but have the potential to influence much more than angiogenesis by enhancing smooth muscle cell/pericyte recruitment to endothelial cell tubes to promote vascular maturation.
In this thesis I have investigated the regulation of eicosanoid synthesizing-enzymes by cannabinoid receptor agonists. Rat renal mesangial cells were used as a model system. I could show that all three (CB1, CB2, and GPR55) cannabinoid receptors are expressed on the mRNA level in rat renal mesangial cells – but with differing expression profiles. The CB1 and GPR55 receptors are expressed in comparable amounts, whereas the CB2 receptor is considerably less expressed than the CB1 and the GPR55 receptors. Furthermore I could show that stimulation of renal mesangial cells with CB1 receptor agonists, such as R(+)MA or ACEA, increased IL-1β-induced cPLA2, sPLA2-IIa, and COX2 protein and mRNA expression which subsequently led to an enhanced IL-1β-induced PGE2 formation. Additionally, the IL-1β- induced sPLA2-IIa promoter activity was also increased by CB1 receptor stimulation. Besides the modulated expression of the eicosanoid synthesizing enzymes, I could show that CB1 agonists also led to an increase of IL-1β-induced iNOS expression and subsequent NO formation. In contrast, stimulation with CB2 selective agonists led to a decrease in IL-1β- induced sPLA2-IIa protein expression and PGE2 formation. Accordingly, the IL-1β-induced sPLA2-IIa promoter activity was also reduced by CB2 receptor agonists. IL-1β-induced iNOS expression and subsequent NO formation were not influenced by CB2 recptor activation. Matching the results I obtained with CB1 receptor agonists on IL-1β-induced PGE2 formation, I could observe an increased cPLA2 protein and mRNA expression with a subsequent increase in IL-1β-induced PGE2 formation by GPR55 stimulation. Stimulation with THC, an unselective CB agonist, increased the IL-1β-induced sPLA2-IIa protein expression and subsequently led to an enhanced IL-1β-induced PGE2 formation. Subjecting the cells to higher THC concentrations surprisingly led to a reduction of the IL-1b-induced sPLA2-IIa protein expression and PGE2 formation. A possible explanation may be the differential expression of the three CB receptors. At low concentrations THC may predominantly activate CB1 and GPR55 and with increasing concentration CB2 receptors may also be activated, slightly reversing the enhancing effect. Moreover, I could show that the CB1 receptor stimulation mediated phosphorylation and hence the activation of ERK1/2 MAPK. Additionally to ERK1/2, there was also a phosphorylation and activation of NFkB observed by CB1 receptor stimulation. In my thesis I could show for the first time that PPARα was activated by IL-1β in rMC. The IL-1β-induced PPARα promoter activity was completely inhibited by addition of the CB2 receptor agonist, JWH015. These findings were confirmed by inhibition of the IL-1β-induced PGE2 formation by a PPARα antagonist (MK-886). In summary, I could show that activation of CB1 receptors in our system led to a worsening of an inflammatory condition, whereas activation of the CB2 receptors led to the complete opposite; namely a reduction of the inflammatory response by reducing the sPLA2-IIa expression and PGE2 formation. GPR55 activation did not display any alteration of inflammatory conditions, since the classical inflammatory pathway was not influenced.
Glutamat ist einer der wichtigsten erregenden Neurotransmitter im zentralen Nervensystem (ZNS), der unter anderem metabotrope Glutamatrezeptoren (mGluRs) aktiviert. Die Signalübertragung der mGluRs erfolgt indirekt durch Aktivierung intrazellulär gekoppelter heterotrimerer G-Proteine, die daraufhin zelluläre Signalprozesse beeinflussen. Es sind acht verschiedene mGluRs bekannt, die in drei Gruppen gegliedert werden, welche sich in ihrer Lokalisation, ihrer Struktur und ihren pharmakologischen Eigenschaften unterscheiden. Die hauptsächlich präsynaptisch lokalisierten Gruppe III mGluRs sind an der Regulation der Neurotransmission an exzitatorischen Synapsen beteiligt. Sie fungieren als Autorezeptoren, die rückkoppelnd die Glutamatausschüttung hemmen. Um eine möglichst exakte Übersetzung der extrazellulären Glutamatsignale durch mGluRs zu gewährleisten, muss die Signaltransduktion dieser Rezeptoren sehr genau kontrolliert und reguliert werden. Als Regulatoren der Signalübertragung verschiedener G-Protein gekoppelter Rezeptoren (GPCRs) haben sich Vertreter der RGS-(Regulators of G protein Signalling)-Proteinfamilie erwiesen. RGS-Proteine können direkten Einfluss auf die Kinetik der Signalübermittlung eines GPCRs nehmen. RGS-Proteine beschleunigen mittels ihrer GAP (GTPase Accelerating Protein)-Funktion die GTP-Hydrolyse, wodurch die Geschwindigkeit der Inaktivierung der Ga-Untereinheit und somit der Signalantwort des GPCRs deutlich erhöht wird. Bis heute wurden mehr als 30 verschiedene RGS-Proteine beschrieben, die alle eine konservierte RGS-Domäne besitzen, die die Bindung an die Ga-Untereinheit des G-Proteins und die GAP-Aktivität vermittelt. Für einige GPCRs wurde bereits eine spezifische und selektive Interaktion sowie Regulation durch bestimmte RGS-Proteine gezeigt. An Gruppe III mGluRs gab es bisher keine einschlägigen Untersuchungen, außer am in der Retina exprimierten mGluR6, der jedoch einen Sonderfall innerhalb dieser Rezeptorgruppe darstellt. Deshalb sollte in dieser Dissertation untersucht werden, ob Gruppe III mGluRs, mit besonderem Augenmerk auf eine der zwei bekannten Spleißvarianten des mGluR7 (mGluR7a), durch ein oder auch mehrere Mitglieder der RGS-Familie reguliert werden können. Zwei Vertreter der RGS-Protein Familie, RGS3 und RGS4, die beide im Gehirn exprimiert werden, wurden als potentielle Kandidaten ausgewählt. Biochemische Interaktionsanalysen dieser Dissertation zeigten, dass sowohl RGS3 als auch RGS4 direkt an mGluR7a binden. Die Bindung zwischen mGluR7a und RGS3 bzw. RGS4 war in Abwesenheit von Ca2+ deutlich verstärkt. Da Calmodulin (CaM) in Anwesenheit von Ca2+ sowohl an mGluR7a als auch an RGS3 und RGS4 bindet, wurde postuliert, dass gebundenes Ca2+/CaM ein räumliches Hindernis für die Wechselwirkung zwischen Rezeptor und RGS-Protein darstellen könnte. Die Daten dieser Arbeit weisen darauf hin, dass die Bindung von Ca2+/CaM an den C-Terminus des mGluR7a die Interaktion zwischen dem Rezeptor und RGS4 abschwächt. Ein physiologischer Effekt der beiden RGS-Proteine auf die mGluR7a-vermittelte Signalübermittlung wurde zunächst in transfizierten HEK-Zellen nachgewiesen. Hierzu wurde die Signalantwort des mGluR7a in An- und Abwesenheit von RGS3 oder RGS4 durch Bestimmung der Menge der sekundären Botenstoffe gemessen und verglichen. RGS3 sowie RGS4 beschleunigten in diesem System durch ihre GAP-Aktivität die GTP-Hydrolyse an der Ga-UE und damit die Inaktivierung der Signaltransduktion des Rezeptors. Mittels hochauflösender Elektronenmikroskopie konnte gezeigt werden, dass RGS3 und RGS4 wie auch mGluR7a in vivo an der präsynaptischen Membran kortikaler Maus-Neuronen zu finden sind. Da die Proteine in den Nervenzellen kolokalisieren, sollte die Interaktion und Regulation, die in vitro nachgewiesen wurde, auch in vivo möglich sein. Ein in vivo-Nachweis des regulatorischen Effektes von RGS4 auf die Signaltransduktion der Gruppe III mGluRs bzw. des mGluR7a erfolgte durch Vergleich der Signalantworten dieser Rezeptoren in kultivierten kortikalen Neuronen aus Wildtyp- und RGS4-defizienten Mäusen. Mit dem Gruppe III mGluR-spezifischen Agonisten L-AP4 konnte im Vergleich zu den Wildtyp-Neuronen keine Antwort der mGluRs in den RGS4-defizienten Nervenzellen gemessen werden. Diese Daten lassen den Schluss zu, dass die Signalübermittlung der Gruppe III mGluRs bzw. des mGluR7a ohne das RGS4-Protein nicht korrekt ablaufen kann. Zusammenfassend kann mit den Daten der vorliegenden Arbeit das bereits bestehende Modell eines mGluR7a-Signalkomplexes erweitert werden: Dabei bildet der Rezeptor bereits vor Stimulation durch einen Agonisten einen Komplex mit dem G-Protein und nachgeschalteten Effektoren, die durch den Rezeptor beeinflusst werden. Durch eine solche lokale Organisation funktionell zusammengehöriger Proteine, die die Initiation (wie das G-Protein) und Termination (wie die RGS-Proteine) der Signaltransduktion bewirken, wird eine schnelle und feinregulierte Signalübermittlung gewährleistet.
Disruption of the complex gastrointestinal ecosystem between the resident microflora and the colonic epithelial cells has been associated with increased inflammation and altered cell growth. Possible endpoints of this disturbance are IBD and CRC. The data presented in this thesis, entitled "PPARgamma as molecular target of epithelial functions in the gastrointestinal tract", shed further light on the underlying molecular mechanisms contributing to the well ordered homeostasis of this gastrointestinal ecosystem. Except for elucidating important roles for mesalazine and the dietary HDAC inhibitors butyrate and SFN in a) the modulation of cellular growth, b) the induction of APs, and c) the control of NFkappaB signalling in CRC cells, the involvement of the nuclear hormone receptors PPARgamma und VDR as "gatekeepers" in these intricate regulatory mechanisms were established. Future work will be engaged in analysing whether these in vitro findings are also physiologically relevant in regard to prevention and therapy of gastrointestinal diseases. Within the scope of this work, in Paper I and II it could be demonstrated that butyrate and mesalazine act via PPARgamma to induce their anti-proliferative and pro-apoptotic actions along the caspase signalling pathway. Activation of the intrinsic and extrinsic signalling trail and the down-regulation of anti-apoptotic proteins are responsible for increased caspase-3 activity caused by butyrate. In contrast, mesalazine merely activates this cascade via the extrinsic trail and the IAPs. Moreover, a signal transduction pathway leading to increased cell death via p38 MAPK - PPARgamma - caspase-3 in response to butyrate was unveiled. In addition, there is strong evidence that mesalazine-mediated pro-apoptotic and growth-inhibitory abilities are controlled by PPARgamma-dependent and -independent mechanisms which appear to be triggered at least in part by the modulation of the tumor suppressor gene PTEN and the oncoprotein c-myc, respectively. In Paper III and IV the induction of the APs HBD-2 and LL-37 in response to the dietary HDAC inhibitors butyrate and SFN was pinpointed. Regarding the molecular events of this regulation, the data presented in this thesis provide strong evidence for the involvement of VDR in HBD-2- and LL-37-induced gene expression, while the participation of PPARgamma was excluded. Moreover, the role for p38 MAPK and TGF-beta1 in the up-regulation of LL-37 caused by butyrate was established. In contrast, SFN-mediated induction of HBD-2 is modulated via ERK1/2 signalling. The findings in Paper V clearly refer to the involvement of the nuclear hormone receptors PPARgamma and VDR in butyrate-mediated suppression of inducible NFkappaB activation dependent on the stimulated signalling pathway caused by LPS or TNFalpha. Moreover, an inhibitory role for VDR in the regulation of basal NFkappaB activation was revealed. On the contrary, a modulating role for PPARgamma on basal NFkappaB could be debarred. Altogether the data presented in this thesis not only provide new insights in the understanding of the fundamental gastrointestinal physiology regulated by nuclear hormone receptors, but also may offer opportunities for the development of potential drug targets and therapeutic strategies in the treatment of IBD and CRC.
Extracts of Boswellia serrata, also known as Indian frankincense, have been used to treat inflammatory diseases in the Indian ayurvedic medicine or Chinese traditional medicine (TCM) for over 3000 years, but the molecular mechanisms of the anti-inflammatory effects are still not well understood. It is obvious that the boswellic acids, the major compounds in the extracts, are responsible for the efficacy. This work employed a protein fishing technique to identify putative targets of boswellic acids at different stages within the inflammatory cascade. For fishing experiments, boswellic acids were immobilized to sepharose and incubated with cell lysates. After washing and boiling, fished proteins were separated by SDS-PAGE and analysed by MALDI-TOF-MS. CatG, DNA-PK and the protein kinase Akt were identified by protein pulldowns with immobilised BAs and characterised as selective and important targets for BAs with an IC50 in the range of physiologically achievable plasma levels up to 5 microM. In addition, the influence on several signal transductions by BAs was tested. Calcium influx, arachidonic acid release, platelet aggregation and TNFalpha-release were assayed to reveal further pharmacological effects of BAs. Celecoxib is a well-known selective COX-2 inhibitor that is in clinical use. In this work, it is demonstrated that celecoxib is also a highly potent direct 5-LO inhibitor. Celecoxib is used in arthritis and its gastro-intestinal side effects are reduced compared to non-selective NSAIDs. In patients with a familiar disposition to polyp forming, celecoxib reduced polyps and the incidence of colon cancer. Because of lowered leukotriene levels in patients under celecoxib therapy it was plausible to test whether celecoxib interferes with 5-LO. Here it is shown that the activity of 5-LO is inhibited in PMNL and cell-free assays with IC50 of 8 microM in intact cells, 20 microM with supplemented arachidonic acid and 30 microM in cell-free systems. Thus, celecoxib is a dual inhibitor of COX-2 and 5-LO. Since 2006, celecoxib has been approved as an orphan drug for the treatment of familial adenomatous polyposis. Aside from this indication, it could be useful for treatment of asthma and other diseases where 5-LO is implicated.
Crohn´s disease (CD) and Ulcerative colitis (UC) are idiopathic inflammatory disorders. Environmental factors, infectious microbes, ethnic origin, genetic susceptibility, and a dysregulated immune system can result in mucosal inflammation. However, the etiology of both CD and UC still remains largely unclear. Inflammatory bowel diseaserelated animal models suggest that a combination of genetic susceptibility factors and altered immune response driven by microbial factors in the enteric environment may contribute to the initiation and chronification of the disease. The intestinal immune system represents a complex network of different lymphoid and non-lymphoid cell populations as well as humoral factors. In inflammatory bowel disease, the controlled balance of the intestinal immune system is disturbed at all levels. In CD, naïve T cells preferably differentiate into Th1 or Th17 producing cells, while in UC, these cells differentiate into aberrant Th2 cells. Overall, in active inflammatory bowel disease effector T cell activity (Th1, Th17, Th2) predominates over regulatory T cells. Animal models of intestinal inflammation are indispensable for our understanding of the pathogenesis of CD and UC. When chosen appropriately, these models proved to be a helpful tool to investigate pathophysiological mechanisms, as well as to test emerging therapeutic options in the preclinical phase. 2,4,6-Trinitrobenzene sulfonic acid (TNBS) and oxazolone are the two major chemicals applied to induce Th1- and Th2-skewed intestinal inflammation, respectively. Colitis can be induced in susceptible strains of mice by intrarectal instillation of the haptenating substances TNBS or oxazolone in ethanol, which is necessary for an initial desintegration of the epithelial barrier. TNBS or oxazolone are believed to haptenize colonic autologous or microbiotic proteins rendering them immunogenic to the host immune system. While TNBS administration in the presence of ethanol results in a transmural infiltrative disease in the entire colon based on an IL-12/IL-23 driven, Th1-or Th17 mediated response, oxazolone instillation finally leads to a colitis caused by a polarized Th2 IL-13-dominated lymphocyte response. Rectal oxazolone instillation in ethanol produces a more superficial inflammation that affects the distal half of the colon rather than the whole colon. Therapeutic modulation of the disturbed immune response in patients with inflammatory bowel disease still represents a complex challenge in the clinic. Currently, none of the therapeutic measure are disease specific and they generally target the pathophysiology downstream of the driving immunpathology. So, there is still the need to develop a tailored approach to prevention of the initiation and perpetuation of the inflammatory cascade before tissue injury occurs. One important aspect of this approach might involve the induction or re-establishment of immunological tolerance. FTY720 following rapid phosphorylation to FTY-P by endogenous sphingosine kinases acts as a sphingosine-1-phosphate (S1P) receptor agonist and represents the prototype of a new generation of S1P receptor modulators. While changing currently its proposed mode of action still focus on the fact, that FTY720 effectively inhibits the egress of T-cells from lymph nodes, thereby reducing the number of antigen-primed/restimulated cells that re-circulate to peripheral inflammatory tissues. However, recent studies indicate, that its immunomodulatory properties might be more complex and exerted not only via interactions with other S1P receptor subtypes but also via a direct modulation of the inflammatory capacity of dendritic cells (DC) resulting in a modified regulation of T cell effector functions as well as in an induction of regulatory T cells and function. 1,25(OH)2D3, the active form of vitamin D, is a secosteroid hormone that has in addition to its central function in calcium and bone metabolism pronounced immune regulatory properties. The biological effects of calcitriol are mediated by the vitamin D receptor (VDR), a member of the superfamily of nuclear hormone receptors. A number of studies identified calcitriol/VDR as prominent negative regulators of Th1-type immune responses, whereas Th2 responses are not affected or even augmented. These effects have been mainly explained by direct activities on lymphocytes, subsequent studies clearly supported a role of calcitriol in modulating monocyte differentiation or DC maturation. However, to translate the immunosuppressive capacities of calcitriol into an effective immunointervention, a great challenge was the design of structural analogs of calcitriol that are devoid of adverse effects related to hypercalcemic activity. The intense study of the 25-oxa series generated a large number of calcitriol analogs exhibiting substantial dissociation between possible immunomodulatory capacities and undesired hypercalcemia. Especially, the combination of the 22-ene modification with the 25-oxa element as realized in ZK156979 yielded a very promising set of new analogs for further characterization in animal models resembling human autoimmune diseases. So, the overall aim of the studies presented here was to evaluate strategies of enhancing regulatory immunity in mouse models of Th1- and Th2-mediated colitis as a new therapeutic approach. To this end we used FTY720, 22-ene-25-oxa vitamin D (ZK156979), as well as the combination of calcitriol and dexamethasone to evaluate the respective pro-tolerogenic potential in intestinal inflammation models in mice. First, to induce Th1-mediated colitis a rectal enema of TNBS was given to Balb/c mice. FTY720 was administered i.p. from day 0-3 or 3-5. FTY720 substantially reduced all clinical, histopathologic, macroscopic, and microscopic parameters of colitis analyzed. The therapeutic effects of FTY720 were associated with a down-regulation of IL-12p70 and subsequent Th1 cytokines. Importantly, FTY720 treatment resulted in a prominent up-regulation of FoxP3, IL-10, TGFβ and CTLA4. Moreover, we observed a significant increase of CD25 and FoxP3 expression in isolated lamina propria CD4+ T cells of FTY720-treated mice. The impact of FTY720 on regulatory T cell induction was further confirmed by concomitant in vivo blockade of CTLA4 or IL-10R which significantly abrogated its therapeutic activity. Thus, our data provide new and strong evidence that besides its well-established migratory properties FTY720 down-regulates proinflammatory signals while simultaneously inducing the functional activity of CD4+CD25+ regulatory T cells. In a second approach, the rectal instillation of oxazolone yielded a Th2-mediated colitis. Treatment with FTY720 prominently reduced the clinical and histopathologic severity of oxazolone-induced colitis, abrogating body weight loss, diarrhea, and macroscopic and microscopic intestinal inflammation. The therapeutic effects of FTY720 were associated with a prominent reduction of the key Th2 effector cytokines IL-13, IL-4 and IL-5. Moreover, FTY720 inhibited GATA3 and T1/ST2 expression, which represent distinct markers for Th2 differentiation and Th2 effector function. Thus, our data are supportive for the view that FTY720 exhibits beneficial prophylactic as well as therapeutic effects in Th2-mediated experimental colitis by directly affecting Th2 cytokine profiles, probably by reducing GATA3 and T1/ST2. Recently, we described 22-ene-25-oxa-vitamin D (ZK156979) as a representative of a novel class of low calcemic vitamin D analogs showing prominent immunomodulative capacities. Here, we used the Th1-mediated TNBS colitis to test its anti-inflammatory properties in vivo. We found that treatment with ZK156979 clearly inhibited the severity of TNBS-induced colitis without exhibiting calcemic effects. Both early and late treatment abrogated all the clinical macroscopic and microscopic parameters of colitis severity; in addition we observed a clear down-regulation of the relevant Th1 cytokine pattern including the T-box transcription factor, T-bet. On the other hand, application of ZK156979 increased local tissue IL-10 and IL-4. Finally, as a new approach we evaluated the pro-tolerogenic potential of calcitriol and dexamethasone in acute Th1-mediated colitis. Calcitriol and/or dexamethasone were administered i.p. from day 0-3 or from day 3-5 following the instillation of the haptenating agent. The combination of these steroids most effectively reduced the clinical and histopathologic severity of TNBS colitis. Th1-related parameters were down- while Th2 markers like IL-4 and GATA3 were up-regulated. Clearly distinguishable from known steroid effects calcitriol in particular promoted regulatory T cell profiles as indicated by a marked increase of IL-10, TGFß, FoxP3 and CTLA4. Furthermore, analysis of DC mediators responsible for a pro-inflammatory differentiation of T cells revealed a clear reduction of IL-12p70, and IL23p19 as well as IL-6 and IL-17. Thus, our data suggest the concept of a steroid-sparing application of calcitriol derivatives in inflammatory bowel disease. Furthermore, the data presented suggest that early markers of inflammatory DC and Th17 differentiation might qualify as new target molecules for both calcitriol as well as for selective immune modulating vitamin D analogs. In conclusion the data of these published investigations added to the substantial progress in understanding the biology of tolerogenic DC and regulatory T cells with respect to their roles in health and disease achieved in the past years. This has led to an increasing interest in the possibility of using DC and regulatory T cells as biological therapeutics to preserve and restore tolerance to self antigens and alloantigens. Especially DC may be helpful to exert their important roles in directing tolerance and immunity by modulation of subpopulations of effector T cells and regulatory T cells. The data demonstrated in the present studies may assist to define the divergent implications of new therapeutic concepts for the treatment of inflammatory bowel disease, especially with regard to a possible auspicious impact on pro-tolerogenic DC and regulatory T cell functions. However, further studies are needed to fulfil our understanding of the complex immunomodulatory profiles of FTY720 as well as of calcitriol and its low calcemic analog ZK156979, thus accelerating their entry into the clinic as new therapeutic options for the cure of inflammatory bowel disease.
