Dass eine so schillernd-unscharfe Formulierung wie die vom 'Geist der Komödie' bezogen auf ein konkretes Lustspiel der eingehenden Diskussion auf mehreren Ebenen zugleich bedarf, um überhaupt fruchtbar zu werden, versteht sich. Zum einen ist dabei natürlich eine poetologische Dimension zu erwarten, zum anderen ergeben sich aus dem angedeuteten Bezug auf Theodor als der Titelfigur des Stücks aber noch weitergehende Fragen: Steht dieser tatsächlich mit der Transzendenz in Kontakt?
Oder könnte es sich hier sogar um eine Art verdeckte Gespenstergeschichte handeln? Es wird aufmerksam zu verfolgen sein, auf welche verschiedenen Arten und Weisen die Dienerfigur durch diesen Text 'geistert'. Die beiden hier angedeuteten Extreme des 'Poetologisch-Geistigen' und des zuweilen ein wenig 'Unheimlich-Geisterhaften', denen im Folgenden zunächst separat nachgegangen werden soll, werden sich im vorliegenden Fall von Hofmannsthals "Unbestechlichem" schließlich als auf das Vielfältigste und Engste miteinander verschränkt erweisen.
Im Frühjahr 1900 verbrachte Hugo von Hofmannsthal einige Wochen in Paris. Der Aufenthalt in der französischen Metropole, die ganz im Zeichen der Weltausstellung stand, wurde für Hofmannsthal zu einer Phase dichter Erlebnisse und Begegnungen. Die hier gewonnenen Impulse schlugen sich in einer Flut von Einfallen nieder, deren Verflüchtigung Hofmannsthal nur mit Mühe vorbeugen konnte, wie es in einem Brief vom 19.1.1900 an Ria Schmujlow-Claassen heißt: Ich habe die Skizze von 4 oder 6 Erzählungen wie im Fieber hingeschrieben, ein Ballet entworfen, ein Vorspiel zur Antigone in Versen ausgeführt (für Berlin), von andern kleinen lyrischen Stücken ein Scenarium gemacht und sitze zwischen Trümmern, halbfestgehaltenen Gestalten, Details wie Rodin zwischen Gypshänden, Füßen und abgebrochenen Flügeln [ ... ]
Seine Rückkehr aus Paris kündigte Hofmannsthal in demselben Schreiben "auf einem Wagen voll Manuscripten im Mai" an. Zu den "Trümmern", die Hofmannsthal auf seinem 'Musenwagen' aus Paris mitbrachte, gehören die Bruchstücke eines "dreitheiligen Spiegels": die dramatischen Entwürfe "Leda und der Schwan", "Das Festspiel der Liebe" und "Der Besuch der Göttin", die gleichzeitig nebeneinander entstanden sind. Unter dem Titel "Spiele der Liebe u. des Lebens" sollten diese Dramen im letzten von drei geplanten Bänden mit Werken aus den Jahren 1889 und 1900 zusammengefaßt werden.
Franz Kafka'nın Dava'sının Melih Cevdet Anday'ın İsa'nın Güncesi adlı romanına etkisi açık olmakla birlikte, incelenen romanlar arasındaki ilişki, basit bir esinlenmenin ötesinde, eserlerin kaleme alındığı dönemlerdeki toplumsal ve hukuki sorunların özüne ilişkin benzerlikle dikkat çekmektedir. Romanların karşılaştırmalı analizi, modernitenin, toplumsal farklılıklar ve iki roman arasındaki altmış yıla rağmen, değişik kültürlerde hukuk felsefesine ilişkin benzer sorunlara yol açtığını göstermektedir. Ulus devletlerinin kurulması ve demokrasinin gelişmesiyle ortaya çıkan kriz kalıcı olmuş, dünya savaşları ve askeri darbeler arasında olağan hâle gelen olağanüstü hâl modern dönemin hukuki durumu hâline gelmiştir.
More than 70 human adenoviruses with type-dependent pathogenicity have been identified but biological information about the majority of these virus types is scarce. Here we employed multiple sequence alignments and structural information to predict receptor usage for the development of an adenoviral vector with novel biological features. We report the generation of a cloned adenovirus based on human adenovirus type 17 (HAdV17) with high sequence homology to the well characterized human adenovirus type 37 (HAdV37) that causes epidemic keratoconjunctivitis (EKC). Our study revealed that human CD46 (CD46) is involved in cell entry of HAdV17. Moreover, we found that HAdV17 infects endothelial cells (EC) in vitro including primary cells at higher efficiencies compared to the commonly used human adenovirus type 5 (HAdV5). Using a human CD46 transgenic mouse model, we observed that HAdV17 displays a broad tropism in vivo after systemic injection and that it transduces ECs in this mouse model. We conclude that the HAdV17-based vector may provide a novel platform for gene therapy.
Human adipose tissue derived stem cells promote liver regeneration in a rat model of toxic injury

In the light of the persisting lack of donor organs and the risks of allotransplantations, the possibility of liver regeneration with autologous stem cells from adipose tissue (ADSC) is an intriguing alternative. Using a model of a toxic liver damage in Sprague Dawley rats, generated by repetitive intraperitoneal application of retrorsine and allyl alcohol, the ability of human ADSC to support the restoration of liver function was investigated. A two-thirds hepatectomy was performed, and human ADSC were injected into one remaining liver lobe in group 1 (n = 20). Injection of cell culture medium performed in group 2 (n = 20) served as control. Cyclosporine was applied to achieve immunotolerance. Blood samples were drawn weekly after surgery to determine liver-correlated blood values. Six and twelve weeks after surgery, animals were sacrificed and histological sections were analyzed. ADSC significantly raised postoperative albumin (P < 0.017), total protein (P < 0.031), glutamic oxaloacetic transaminase (P < 0.001), and lactate dehydrogenase (P < 0.04) levels compared to injection of cell culture medium alone. Transplanted cells could be found up to twelve weeks after surgery in histological sections. This study points towards ADSC being a promising alternative to hepatocyte or liver organ transplantation in patients with severe liver failure.
Human babesiosis in Europe

Babesiosis is attracting increasing attention as a worldwide emerging zoonosis. The first case of human babesiosis in Europe was described in the late 1950s and since then more than 60 cases have been reported in Europe. While the disease is relatively rare in Europe, it is significant because the majority of cases present as life-threatening fulminant infections, mainly in immunocompromised patients. Although appearing clinically similar to human babesiosis elsewhere, particularly in the USA, most European forms of the disease are distinct entities, especially concerning epidemiology, human susceptibility to infection and clinical management. This paper describes the history of the disease and reviews all published cases that have occurred in Europe with regard to the identity and genetic characteristics of the etiological agents, pathogenesis, aspects of epidemiology including the eco-epidemiology of the vectors, the clinical courses of infection, diagnostic tools and clinical management and treatment.
Mitochondrial maintenance crucially depends on the quality control of proteins by various chaperones, proteases and repair enzymes. While most of the involved components have been studied in some detail, little is known on the biological role of the CLPXP protease complex located in the mitochondrial matrix. Here we show that deletion of PaClpP, encoding the CLP protease proteolytic subunit CLPP, leads to an unexpected healthy phenotype and increased lifespan of the fungal ageing model organism Podospora anserina. This phenotype can be reverted by expression of human ClpP in the fungal deletion background, demonstrating functional conservation of human and fungal CLPP. Our results show that the biological role of eukaryotic CLP proteases can be studied in an experimentally accessible model organism.
Constitutive Wnt activation upon loss of Adenoma polyposis coli (APC) acts as main driver of colorectal cancer (CRC). Targeting Wnt signaling has proven difficult because the pathway is crucial for homeostasis and stem cell renewal. To distinguish oncogenic from physiological Wnt activity, we have performed transcriptome and proteome profiling in isogenic human colon organoids. Culture in the presence or absence of exogenous ligand allowed us to discriminate receptor-mediated signaling from the effects of CRISPR/Cas9-induced APC loss. We could catalog two nonoverlapping molecular signatures that were stable at distinct levels of stimulation. Newly identified markers for normal stem/progenitor cells and adenomas were validated by immunohistochemistry and flow cytometry. We found that oncogenic Wnt signals are associated with good prognosis in tumors of the consensus molecular subtype 2 (CMS2). In contrast, receptor-mediated signaling was linked to CMS4 tumors and poor prognosis. Together, our data represent a valuable resource for biomarkers that allow more precise stratification of Wnt responses in CRC.
The genome, antigens of human cytomegalovirus (HCMV) are frequently found in prostatic carcinoma. However, whether this infection is causative or is an epiphenomenon is not clear. We therefore investigated the ability of HCMV to promote metastatic processes, defined by tumor cell adhesion to the endothelium, extracellular matrix proteins. Experiments were based on the human prostate tumor cell line PC3, either infected with the HCMV strain Hi (HCMVHi) or transfected with cDNA encoding the HCMV-specific immediate early protein IEA1 (UL123) or IEA2 (UL122). HCMVHi upregulated PC3 adhesion to the endothelium, to the extracellular matrix proteins collagen, laminin, fibronectin. The process was accompanied by enhancement of β1-integrin surface expression, elevated levels of integrin-linked kinase, phosphorylation of focal adhesion kinase. IEA1 or IEA2 did not modulate PC3 adhesion or β1-integrin expression. Based on this in vitro model, we postulate a direct association between HCMV infection, prostate tumor transmigration, which is not dependent on IEA proteins. Integrin overexpression, combined with the modulation of integrin-dependent signalling, seems to be, at least in part, responsible for a more invasive PC3Hi tumor cell phenotype. Elevated levels of c-myc found in IEA1-transfected or IEA2-transfected PC3 cell populations might promote further carcinogenic processes through accelerated cell proliferation.
