Sind Nanopartikel als Trägersystem für den Gentransfer in Bronchialepithelzellen geeignet? : Untersuchungen in der Zellkultur

  • Verschiedene Nanopartikel, darunter Gelatine- und Albuminnanopartikel und Cyanoakrylatnanopartikel mit Butyl- und Hexylseitenketten, wurden in dieser Arbeit auf ihre Tauglichkeit als Trägersystem zum Einsatz für einen späteren Gentransfer in Bronchialepithelzellen erprobt. In der Zellkultur wurden dazu an primären humanen Bronchialepithelzellen und an der Zelllinie 16HBE14o Zytotoxizitätstest nach Inkubation mit oben genannten Nanopartikeln durchgeführt. Weiterhin wurden qualitative und quantitative Aspekte der Anreicherung von fluoreszenzmarkierten Gelatine- und Albuminnanopartikeln in diese Zellen mit dem konfokalen Laserrastermikroskop und dem Durchflußzytometer untersucht. Zusätzlich evaluiert wurde die inflammatorische Potenz von Gelatine- und Albuminnanopartikeln mittels einer quantitativen Interleukin-8-Bestimmung. Es konnte gezeigt werden, dass Cyanoakrylate aufgrund eines hohen zytotoxischen Effekts nicht für die Behandlung von Bronchialepithelzellen geeignet sind. Hierbei zeigten Cyanoakrylate mit kurzen Butylseitenketten eine ausgeprägtere Zytotoxizität als jene mit langen Hexylseitenketten. Gelatine- und Albuminnanopartikel hingegen zeichneten sich durch sehr geringe bis fehlende Zytotoxizität aus. Mit dem konfokalen Laserrastermikroskop konnte die Aufnahme von fluoreszenzmarkierten Gelatine- und Albuminnanopartikel in das Zellinnere von primären Bronchialepithelzellen dokumentiert werden. Erstmals wurde somit eine Nanopartikelaufnahme in Zellen nachgewiesen, die nicht zum mononukleären Phagozytensystem gehören. Die Versuche zur zellulären Aufnahme der Nanopartikel mit verschiedenen Inkubationsbedingungen zeigten, dass primäre Bronchialepithelzellen und 16HBE14o-Zellen Gelatine- und Albuminnanopartikel in einem aktiven, stoffwechselabhängigen Prozess aufnehmen, der am ehesten der Phagozytose entspricht. Ferner haben sich Albuminnanopartikel im Rahmen dieser Versuche als bioadhäsiv erwiesen. Mit dem Durchflußzytometer ließ sich schließlich eine Konzentrations­abhängigkeit der Aufnahme fluoreszenzmarkierter Gelatine- und Albuminnanopartikel zeigen und feststellen, dass alle Zellen einer Zellpopulation von primären Bronchialepithelzellen oder 16HBE14o-Zellen Nanopartikel aufnehmen. Selbst primäre Zellen, die durch die Kultur auf Membraneinsätzen auf einem hohen Differenzierungsniveau gehalten wurden, nahmen Nanoparikel auf. Eine quantitative Interleukin-8-Bestimmung zeigte schließlich, dass Gelatine- und Albuminnanopartikel keine Inflammation in primären humanen Bronchialepithelzellen und der Zelllinie 16HBE14o verursachen. Abschließend lässt sich sagen, dass die hier erprobten Nanopartikel bezüglich ihrer Zytotoxizität und ihrem inflammatorischem Potenzial als geeignet für den Gentransfer erscheinen. Die Aufnahme der Nanopartikel in das Zellinnere ist vielversprechend für eine spätere Expression von Transgenen in Bronchialepithelzellen.
  • In this work various nanoparticles including gelatine- and albuminparticles and cyanoakrylatenanoparticles with butylic and hexylic sidechains have been evaluated for their potential as a carrier-system for gene transfer to airway epithelium cells in the future. This has been achieved by determining cytotoxicity after exposure to the listed nanoparticles on primary human airway epithelium cells and the cell line 16HBE14o in culture: Furthermore confocal laserscan microscopy and flow cytometry allowed the observation of qualitative and quantitative aspects of the uptake of fluorescence labelled gelatine- and albuminnanoparticles into these cells. In addition the inflammatory potential of gelatine- and albuminenanoparticles has been evaluated performing a quantitative Interleukine-8-ELISA. Due to a high cytotoxic effect polyalkylcyanoakrylate nanoparticles appeared not to be suitable for the interaction with airway epithelium cells. Cyanoacrlyates with short butylic sidechains showed a more severe cytotoxicity than those with long hexylic sidechains. In contrast to that gelatine- and albuminenanoparticles appeared highly suitable showing little or no cytotoxic effect. Confocal laserscan microscopy allowed the documentation of the cellular uptake of fluorescence labelled gelatine- and albuminnanoparticles, for the first time showing the uptake of nanoparticles into cells not belonging to the mononuclear phagocyte system. Tests with various incubation conditions showed that nanoparticle incorporation is an active, metabolism-dependent process, most resembling phagocytosis. Albumin nanoparticles proved to be bioadhesive during these tests. Flow cytometry finally proved that the uptake of fluorescence labelled gelatine- and albuminnanoparticles is dependent on the nanoparticle concentration and that all cells of a population of primary airway epithelium cells or 16HBE14o-cells incorporated nanoparticles. Even highly differentiated primary cells cultured on membrane inserts showed nanoparticle uptake. Performing the quantitative Interleukine-8-ELISA it could be shown that gelatine- and albuminnanoparticles provoked no inflammation in primary human airway epithelium cells or 16HBE14o-cells. Concluding the tested nanoparticles appear to be suitable for gene transfer as to cytotoxicity and inflammatory potential. The uptake of nanoparticles into the cells is promising for an expression of transgenes in airway epithelium cells in the future.
