Simple Summary: Penile cancer is a rare but aggressive malignancy characterized by rapid tumor growth as well as prompt metastasis in groin lymphatics. While localized diseases can be successfully cured by surgery in most cases, no truly effective treatment options have been established for metastatic diseases as of yet. In the current investigation, we assessed the value of selected members of the PI3K/mTOR/AKT pathway to serve as tumor markers or therapeutic targets for this disease. Higher expression of AKT was significantly more prevalent in high-grade tumors and independently predictive of the worse survival parameters, while increased expression of pmTOR was associated with an inferior prognosis as well. Treatment with the pan-AKT inhibitor capivasertib in PeCa cell lines induced significant reduction of cell viability and movement capacity. These findings might aid in the understanding of the molecular tumor background as well as development of novel treatment options for advanced penile cancer.
Abstract: The PI3K/mTOR/AKT pathway might represent an intriguing option for treatment of penile cancer (PeCa). We aimed to assess whether members of this pathway might serve as biomarkers and targets for systemic therapy. Tissue of primary cancer from treatment-naïve PeCa patients was used for tissue microarray analysis. Immunohistochemical staining was performed with antibodies against AKT, pAKT, mTOR, pmTOR, pS6, pPRAS, p4EBP1, S6K1 and pp70S6K. Protein expression was correlated with clinicopathological characteristics as well as overall survival (OS), disease-specific survival (DSS), recurrence-free survival (RFS) and metastasis-free survival (MFS). AKT inhibition was tested in two primarily established, treatment-naïve PeCa cell lines by treatment with capivasertib and analysis of cell viability and chemotaxis. A total of 76 patients surgically treated for invasive PeCa were included. Higher expression of AKT was significantly more prevalent in high-grade tumors and predictive of DSS and OS in the Kaplan–Meier analysis, and an independent predictor of worse OS and DSS in the multivariate regression analysis. Treatment with pan-AKT inhibitor capivasertib in PeCa cell lines induced a significant downregulation of both total AKT and pAKT as well as decreased cell viability and chemotaxis. Selected protein candidates of the mTOR/AKT signaling pathway demonstrate association with histological and survival parameters of PeCa patients, whereas AKT appears to be the most promising one.
The quest for new and improved therapies for glioblastoma (GB) has been mostly unsuccessful in more than a decade despite significant efforts. The few exceptions include the optimization of classical alkylating chemotherapy by including lomustine in the first line regimen for GB with a methylated MGMT promoter and tumor treating fields. The PI3K/mammalian Target of Rapamycin (mTOR) pathway is often aberrantly activated in rhabdomyosarcoma (RMS) and represents a promising therapeutic target. Recent evaluation of AZD8055, an ATP-competitive mTOR inhibitor, by the Preclinical Pediatric Testing Program showed in vivo antitumor activity against childhood solid tumors, including RMS. Therefore, in the present study, we searched for AZD8055-based combination therapies. Here, we identify a new synergistic lethality of AZD8055 together with ABT-737, a BH3 mimetic that antagonizes Bcl-2, Bcl-xL, and Bcl-w but not Mcl-1. AZD8055 and ABT-737 cooperate to induce apoptosis in alveolar and embryonal RMS cells in a highly synergistic fashion (combination index < 0.2). Synergistic induction of apoptosis by AZD8055 and ABT-737 is confirmed on the molecular level, as AZD8055 and ABT-737 cooperate to trigger loss of mitochondrial membrane potential, activation of caspases, and caspase-dependent apoptosis that is blocked by the pan-caspase inhibitor Z-VAD-fmk. Similar to AZD8055, the PI3K/mTOR inhibitor NVP-BEZ235, the PI3K inhibitor NVP-BKM120 and Akt inhibitor synergize with ABT-737 to trigger apoptosis, whereas no cooperativity is found for the mTOR complex 1 inhibitor RAD001. Interestingly, molecular studies reveal a correlation between the ability of different PI3K/mTOR inhibitors to potentiate ABT-737-induced apoptosis and to suppress Mcl-1 protein levels. Importantly, knockdown of Mcl-1 increases ABT-737-induced apoptosis similar to AZD8055/ABT-737 cotreatment. This indicates that AZD8055-mediated suppression of Mcl-1 protein plays an important role in the synergistic drug interaction. By identifying a novel synergistic interaction of AZD8055 and ABT-737, our findings have important implications for the development of molecular targeted therapies for RMS.
With an increased understanding of the tumor biology of squamous cell carcinoma of the head and neck (SCCHN), targeted therapies have found their way into the clinical treatment routines against this entity. Nevertheless, to date platinum-based cytostatic agents remain the first line choice and targeting the epidermal growth factor-receptor (EGFR) with combined cetuximab and radiation therapy remains the only targeted therapy approved in the curative setting. Investigation of immune checkpoint inhibitors (ICI), such as antibodies targeting programmed cell death protein 1 (PD-1) and its ligand PD-L1, resulted in a change of paradigms in oncology and in the first approval of new drugs for treating SCCHN. Nivolumab and pembrolizumab, two anti-PD-1 antibodies, were the first agents shown to improve overall survival for patients with metastatic/recurrent tumors in recent years. Currently, several clinical trials investigate the role of ICI in different therapeutic settings. A robust set of biomarkers will be an inevitable tool for future individualized treatment approaches including radiation dose de-escalation and escalation strategies. This review aims to summarize achieved goals, the current status and future perspectives regarding targeted therapies and ICI in the management of SCCHN.
Das “Protein Associated with Myc” spielt in den verschiedenen physiologischen Vorgängen eine Rolle. Dazu zählen Prozesse der Synaptogenese und Schmerzverarbeitung ebenso wie eine Regulation des Pteridin- und cAMP-Stoffwechsels. Auf welche Weise PAM die unterschiedlichen Effekte vermittelt, ist bislang nur in Ansätzen verstanden. Um die Wirkmechanismen von PAM aufzuklären, wurden in dieser Arbeit seine biochemischen Funktionen untersucht. Die These, dass PAM als E3 Ubiquitinligase aktiv ist, konnte in vitro mittels biochemischer Versuche zweifelsfrei bestätigt werden. Sowohl das nativ aufgereinigte, humane PAM, als auch der heterolog expremierte C-Terminale Bereich (C-PAM), der die katalytisch aktive RING Finger Domäne enthält, wiesen die Fähigkeit zur Ubiquitinkettenbildung und Autoubiquitinierung auf. Bei der Identifikation eines möglichen Zielproteins rückte das Protein TSC2 und der damit verbundene TSC2 / mTOR Signalweg in den Fokus. Für das gewählte Modell-System HeLa Zellen ließ sich eine Interaktion von PAM und TSC2 durch Ko-Immunopräzipitationen und Immunzytochemie nachweisen. Es konnte erstmalig gezeigt werden, dass das vollständige, native PAM, nicht aber die isolierte RING Finger Domäne, TSC2 polyubiquitinieren und zum proteasomalen Abbau markieren kann. TSC2 ist ein negativer Regulator der mTOR Kinaseaktivität, in dem es den stimulatorischen Einfluss von Rheb auf mTOR inhibiert. PAM wird in HeLa Zellen durch das Phospholipid Sphingosin-1-Phosphat (S1P) aktiviert. Nach S1P Behandlung der Zellen war eine Phosphorylierung der Proteinkinase mTOR nachweisbar. Diese ging mit einer Aktivierung der Kinaseaktivität einher, wie die rapamycinsensitive Phosphorylierung der mTOR Zielproteine p70S6K und 4E-BP1 zeigte. Durch Gabe von Rezeptor-Agonisten/-Antagonisten konnte eine Beteiligung des S1P1 und S1P2 Rezeptors ausgeschlossen werden. Der zunächst vermutete Mechanismus eines S1P induzierten, PAM-abhängigen Abbaus von TSC2 konnte trotz vielfältiger Herangehensweisen nicht nachgewiesen werden. Eine Phosphorylierung als Indikation einer Inaktivierung war ebenfalls nicht detektierbar. Auch die GAP Aktivität von TSC2 auf Rheb, wird in in vitro Versuchen durch die Interaktion mit PAM nicht vermindert. Durch eine Verminderung der TSC2 Expression mittels spezifischer siRNA zeigte sich, dass TSC2 nicht in die S1P-abhängige mTOR Aktivierung involviert ist. Auch regulatorische Proteinkinasen wie AKT, ERK oder PI3K, die durch S1P aktiviert werden können, sind an dem Signalweg nicht beteiligt, wie die Hemmung dieser Enzyme mit spezifischen Inhibitoren zeigte. Dagegen konnte eine Beteiligung von PAM und Rheb zum einen mittels Proteintransfektion bestätigt werden, zum anderen ließen sich die S1P Effekte durch Hemmstoffe verhindern, die für eine Aktivierung von PAM, respektive Rheb, nötig sind. Durch Nukleotidbindungsstudien war ein Einfluss von PAM auf den GTP-Beladungszustand von Rheb nachweisbar. Sowohl in einem GTPS Bindungsversuch als auch in einem GDP Dissoziationsexperiment erhöhte PAM konzentrationsabhängig die GTP Bindung bzw. den GDP/GTP Austausch an Rheb. In dieser Arbeit wird damit erstmalig eine duale Funktion eines Proteins als Ubiquitinligase und GEF beschrieben. So konnte die postulierte Aktivität von PAM als Ubiquitinligase bestätigt und TSC2 als Zielprotein identifiziert werden. Gleichzeitig wurde ein TSC2 unabhängiger Weg der mTOR Aktivierung aufgeklärt, an dem PAM und Rheb beteiligt sind. Als möglicher Mechanismus kommt eine Aktivität von PAM als Guanin-Nukleotid Austausch Faktor (GEF) auf Rheb in Frage. Durch Beschreibung von PAM als negativem Regulator von TSC2 und Aktivator von Rheb trägt diese Arbeit einen wichtigen Beitrag zur TSC2 / mTOR Forschung bei. Umgekehrt ermöglicht sie eine neue Sichtweise auf partiell PAM-abhängige Vorgänge wie Synaptogenese und Nozizeption, indem sie TSC2 / mTOR in diese Prozesse integriert.
Molecular tumour targeting has significantly improved anti-cancer protocols. Still, the addition of molecular targeting to the treatment regime has not led to a curative breakthrough. Combined mammalian target of Rapamycin (mTOR) and histone deacetylase (HDAC) inhibition has been shown not only to enhance anti-tumour potential, but also to prevent resistance development seen under mono-drug therapy. This investigation was designed to evaluate whether cross-communication exists between mTOR signalling and epigenetic events regulated by HDAC. DU-145 prostate cancer cells were treated with insulin-like growth factor (IGF) to activate the Akt-mTOR cascade or with the HDAC-inhibitor valproic acid (VPA) to induce histone H3 and H4 acetylation (aH3, aH4). Subsequently, mTOR, Rictor, Raptor, p70s6k, Akt (all: total and phosphorylated), H3 and H4 (total and acetylated) were analysed by western blotting. Both techniques revealed a link between mTOR and the epigenetic machinery. IGF activated mTOR, Rictor, Raptor, p70s6k and Akt, but also enhanced aH3 and aH4. Inversely, IGFr blockade and knock-down blocked the Akt-mTOR axis, but simultaneously diminished aH3 and aH4. VPA treatment up-regulated histone acetylation, but also activated mTOR-Akt signalling. HDAC1 and 2 knock-down revealed that the interaction with the mTOR system is initiated by histone H3 acetylation. HDAC-mTOR communication, therefore, is apparent whereby tumour-promoting (Akt/mTORhigh, aH3/aH4low) and tumour-suppressing signals (Akt/mTORlow, aH3/aH4high) are activated in parallel. Combined use of an HDAC- and mTOR inhibitor might then diminish pro-tumour effects triggered by the HDAC- (Akt/mTORhigh) or mTOR inhibitor (aH3/aH4low) alone.
This study was designed to investigate whether epigenetic modulation by histone deacetylase (HDAC) inhibition might circumvent resistance towards the mechanistic target of rapamycin (mTOR) inhibitor temsirolimus in a prostate cancer cell model. Parental (par) and temsirolimus-resistant (res) PC3 prostate cancer cells were exposed to the HDAC inhibitor valproic acid (VPA), and tumor cell adhesion, chemotaxis, migration, and invasion were evaluated. Temsirolimus resistance was characterized by reduced binding of PC3res cells to endothelium, immobilized collagen, and fibronectin, but increased adhesion to laminin, as compared to the parental cells. Chemotaxis, migration, and invasion of PC3res cells were enhanced following temsirolimus re-treatment. Integrin α and β receptors were significantly altered in PC3res compared to PC3par cells. VPA significantly counteracted temsirolimus resistance by down-regulating tumor cell–matrix interaction, chemotaxis, and migration. Evaluation of integrin expression in the presence of VPA revealed a significant down-regulation of integrin α5 in PC3res cells. Blocking studies demonstrated a close association between α5 expression on PC3res and chemotaxis. In this in vitro model, temsirolimus resistance drove prostate cancer cells to become highly motile, while HDAC inhibition reversed the metastatic activity. The VPA-induced inhibition of metastatic activity was accompanied by a lowered integrin α5 surface level on the tumor cells.
Hintergrund: Epilepsie bezeichnet eine Erkrankung, welche durch eine anhaltende Prädisposition für Symptome, die im Zusammenhang mit einer ungewöhnlich starken oder synchronisierten elektrischen Aktivität des Gehirns auftreten (=epileptischer Anfall) charakterisiert ist. Mutationen in dem Gen DEPDC5 (Dishevelled, Egl-10 and Pleckstrin (DEP) domain-containing protein 5) sind mit fokalen Epilepsien assoziiert und führen in Tiermodellen und humanen Modellen zu einer Überaktivierung des mTOR-Signalweges. Auf neuromorphologischer Ebene zeigt sich die mTOR-Überaktivierung durch eine Vergrößerung des Zelldurchmessers und einer zunehmenden Verästelung der Neuriten. Ziel dieser Studie war es, die morphologischen Auswirkungen der DEPDC5-assoziierten mTOR-Überaktivierung in der SH-SY5Y-Neuroblastomzelllinie zu untersuchen. Dadurch soll eine Einschätzung getroffen werden, ob das im Zusammenhang mit neurodegenerativen Erkrankungen bereits gut etablierte SH-SY5Y-Zellmodell auch bei der Untersuchung epilepsieassoziierter Pathomechanismen zum Einsatz kommen kann.
Methoden: Unter Einsatz der CRISPR/Cas9-Methode wurden Knockout(KO)-Mutationen in Exon 2 und Exon 3 des DEPDC5-Gens erzeugt und diese mittels Sanger-Sequenzierung bestätigt. Danach wurden die Knockouts auf RNA- und Proteinebene, durch Real-time-RT-PCR und Western Blot validiert. Die bestätigten homozygoten DEPDC5-KO-Zelllinen wurden anschließend mittels Western Blot auf eine mTOR-Überaktivierung untersucht. Zuletzt erfolgte die neuromorphologische Validierung des DEPDC5-KO. Die Zellgröße proliferierender SH-SY5Y wurden mittels Durchflusszytometrie (FACS) untersucht. Zudem bestimmten wir die neuronale Architektur differenzierter SH-SY5Y unter Einsatz der Sholl-Analyse.
Ergebnisse: Es konnten vier unabhängige DEPDC5-KO-SH-SY5Y-Zelllinien mit homozygoten Indel-Mutationen und vorzeitigem Stoppcodon in Exon 3 generiert werden. Die erwartete Reduktion an DEPDC5-mRNA konnte mittels Real-time 10 RT-PCR nicht festgestellt werden. Die Abwesenheit des Proteins konnte durch Western Blot aber gezeigt werden. Funktionell konnte für alle Zelllinien eine mTOR-Überaktivierung mittels Western Blot nachgewiesen werden. Dabei konnte phosphoryliertes AKT (AKT serine/threonine kinase 1) als stabilster Marker etabliert werden. Auf neuromorphologischer Ebene ließ sich ein Trend in Richtung vergrößertem Zelldurchmesser bei verlängertem Auswachsen der Neuriten feststellen, wobei sich für das Modell Unterschiede zwischen den einzelnen Klonen ergaben.
Diskussion: In dieser Studie gelang es erstmals, den Zusammenhang zwischen DEPDC5-KO und einer mTOR-Überaktivierung in der onkogenen SH-SY5Y-Zelllinie zu replizieren. Das verlängerte Auswachsen der Neuriten, bei jedoch gleichbleibender Anzahl peripherer Verästelungen, stellt dabei einen neuen Befund dar und könnte durch die frühe neuronale Entwicklungsstufe des SH-SY5Y-Zellmodells erklärt werden. Auf Grundlage der Ergebnisse dieser Arbeit lässt sich sagen, dass das robuste und kostengünstige SH-SY5Y-Zellmodell insbesondere für high-throughput Methoden und Screeningassays ein geeignetes Modell ist. Durch die Kombination mit reiferen Zellmodellen, wie beispielsweise iPSCs (induced pluripotent stem cells), könnte der Phänotyp eines DEPDC5-KO und anderer mTOR-assoziierter Epilepsien, möglichst umfassend in-vitro dargestellt werden.
Regulation of translation is essential during stress. However, the precise sets of proteins regulated by the key translational stress responses—the integrated stress response (ISR) and mTORC1—remain elusive. We developed multiplexed enhanced protein dynamics (mePROD) proteomics, adding signal amplification to dynamic-SILAC and multiplexing, to enable measuring acute changes in protein synthesis. Treating cells with ISR/mTORC1-modulating stressors, we showed extensive translatome modulation with ∼20% of proteins synthesized at highly reduced rates. Comparing translation-deficient sub-proteomes revealed an extensive overlap demonstrating that target specificity is achieved on protein level and not by pathway activation. Titrating cap-dependent translation inhibition confirmed that synthesis of individual proteins is controlled by intrinsic properties responding to global translation attenuation. This study reports a highly sensitive method to measure relative translation at the nascent chain level and provides insight into how the ISR and mTORC1, two key cellular pathways, regulate the translatome to guide cellular survival upon stress.
The mammalian target of rapamycin and the integrated stress response are central cellular hubs regulating translation upon stress. The precise proteins and pathway specificity of translation targets of these pathways remained largely unclear. We recently described a new method for quantitative translation proteomics and found that both pathways control translation of the same sets of proteins.
