This study addresses the situation of the Sorbs, an indigenous minority living in Lusatia, Germany. Under the overarching rubric of ethnic and nationalist projects, women are usually symbolized as the guardians of culture and language. Women’s experiences as subjects in everyday life and in multi-facetted social practices therefore become veiled. The main concern in this book is to discuss how the women who identify themselves as/with Sorbs studied here construct their identities in the modern world, how they approach a sense of self and how they position themselves in their everyday lives, what kind of processes they undergo in their identity construction, and which factors are implicated in the formation of these identities. Investigating the Sorbian minority as the research subject and focusing on the female gender primarily involves intersections of ethnicity and gender, which are the points of departure for this study. As the research progresses, women’s gradual active construction of gender and ethnicity while living their everyday lives reveals a construction of multifarious and complex identifications across differences of gender, ethnicity, culture, religion and class. The results of research create Sorbian culture anew, craft Sorbian identity afresh and render the notion of Sorbian women in new terms. New meanings encased in these conceptions actually contain an active and transformative impetus. This thrust forces these ideas to undergo a process of redefinition. It is the life experiences people have in everyday practices that impel us to envisage identity construction as a dynamic, never-ending and open-ended articulation of one’s positionings.
Daß sich die Literaturwissenschaft mit Schriftstellerinterviews schwertut, kann nicht behauptet werden. Sie macht es sich vielmehr zu leicht mit ihnen, indem sie diese Textgattung weitgehend ignoriert. Zwar werden Interviewäußerungen gelegentlich herangezogen, um eigene Forschungsthesen zu bekräftigen, kaum einmal richtet sich der Blick aber auf die Spezifika dieser Textsorte. In dieser Hinsicht besteht zwischen dem expandierenden Angebot an Interview-Bänden und der konstant gering bleibenden wissenschaftlichen Nachfrage eine deutliche Diskrepanz.
Im Folgenden ist also zu klären:
1. warum die Romantiker mit ihrer Entdeckung, dass die Idee der Poesie Prosa sei, eine poetische Forminnovation in zweierlei Hinsicht angestoßen haben, und zwar einmal im Bereich der schreibpraktisch relevanten Poetisierung der Prosa und Prosaisierung der Poesie und zweitens im Bereich der ,Wiederaufbereitung' vergangener Poesie.
2. warum der kritischen Darstellung des prosaischen Kerns im Werk als eines "ewig nüchternen Bestandes" ästhetische und religiös-mystische Implikationen zukommen.
3. warum die Entdeckung, dass die Idee der Poesie Prosa sei, diagnostischen und resistenten Wert für die eigene Gegenwart Benjamins hatte und daher seine Fortsetzungsarbeiten (d.h. der Wahlverwandtschaften-Essay und Dichtung und Wahrheit) nachhaltig prägte.
Background Endothelium-derived nitric oxide plays an important role for the bone marrow microenvironment. Since several important effects of nitric oxide are mediated by cGMP-dependent pathways, we investigated the role of the cGMP downstream effector cGMP-dependent protein kinase I (cGKI) on postnatal neovascularization. Methodology/Principal Findings In a disc neovascularization model, cGKI -/- mice showed an impaired neovascularization as compared to their wild-type (WT) littermates. Infusion of WT, but not cGKI -/- bone marrow progenitors rescued the impaired ingrowth of new vessels in cGKI-deficient mice. Bone marrow progenitors from cGKI -/- mice showed reduced proliferation and survival rates. In addition, we used cGKI alpha leucine zipper mutant (LZM) mice as model for cGKI deficiency. LZM mice harbor a mutation in the cGKI alpha leucine zipper that prevents interaction with downstream signaling molecules. Consistently, LZM mice exhibited reduced numbers of vasculogenic progenitors and impaired neovascularization following hindlimb ischemia compared to WT mice. Conclusions/Significance Our findings demonstrate that the cGMP-cGKI pathway is critical for postnatal neovascularization and establish a new role for cGKI in vasculogenesis, which is mediated by bone marrow-derived progenitors.
Background Transplantation of vasculogenic progenitor cells (VPC) improves neovascularization after ischemia. However, patients with type 2 diabetes mellitus show a reduced VPC number and impaired functional activity. Previously, we demonstrated that p38 kinase inhibition prevents the negative effects of glucose on VPC number by increasing proliferation and differentiation towards the endothelial lineage in vitro. Moreover, the functional capacity of progenitor cells is reduced in a mouse model of metabolic syndrome including type 2 diabetes (Leprdb) in vivo. Findings The aim of this study was to elucidate the underlying signalling mechanisms in vitro and in vivo. Therefore, we performed DNA-protein binding arrays in the bone marrow of mice with metabolic syndrome, in blood-derived progenitor cells of diabetic patients as well as in VPC ex vivo treated with high levels of glucose. The transcriptional activation of ETS transcription factors was increased in all samples analyzed. Downregulation of ETS1 expression by siRNA abrogated the reduction of VPC number induced by high-glucose treatment. In addition, we observed a concomitant suppression of the non-endothelial ETS-target genes matrix metalloproteinase 9 (MMP9) and CD115 upon short term lentiviral delivery of ETS-specific shRNAs. Long term inhibition of ETS expression by lentiviral infection increased the number of cells with the endothelial markers CD144 and CD105. Conclusion These data demonstrate that diabetes leads to dysregulated activation of ETS, which blocks the functional activity of progenitor cells and their commitment towards the endothelial cell lineage.
Background: Wnt signaling controls the balance between stem cell proliferation and differentiation and body patterning throughout development. Previous data demonstrated that non-canonical Wnts (Wnt5a, Wnt11) increased cardiac gene expression of circulating endothelial progenitor cells (EPC) and bone marrow-derived stem cells cultured in vitro. Since previous studies suggested a contribution of the protein kinase C (PKC) family to the Wnt5a-induced signalling, we investigated which PKC isoforms are activated by non-canonical Wnt5a in human EPC. Methodology/Principal Findings: Immunoblot experiments demonstrated that Wnt5a selectively activated the novel PKC isoform, PKC delta, as evidenced by phosphorylation and translocation. In contrast, the classical Ca2+-dependent PKC isoforms, PKC alpha and beta2, and one of the other novel PKC isoforms, PKC epsilon, were not activated by Wnt5a. The PKC delta inhibitor rottlerin significantly blocked co-culture-induced cardiac differentiation in vitro, whereas inhibitors directed against the classical Ca2+-dependent PKC isoforms or a PKC epsilon-inhibitory peptide did not block cardiac differentiation. In accordance, EPC derived from PKC delta heterozygous mice exhibited a significant reduction of Wnt5a-induced cardiac gene expression compared to wild type mice derived EPC. Conclusions/Significance: These data indicate that Wnt5a enhances cardiac gene expressions of EPC via an activation of PKC delta.
Mitochondria are essential for respiration and oxidative phosphorylation. Mitochondrial dysfunction due to aging processes is involved in pathologies and pathogenesis of a series of cardiovascular disorders. New results accumulate showing that the enzyme telomerase with its catalytic subunit telomerase reverse transcriptase (TERT) has a beneficial effect on heart functions. The benefit of short-term running of mice for heart function is dependent on TERT expression. TERT can translocate into the mitochondria and mitochondrial TERT (mtTERT) is protective against stress induced stimuli and binds to mitochondrial DNA (mtDNA). Because mtDNA is highly susceptible to damage produced by reactive oxygen species (ROS) which are generated in close proximity to the respiratory chain, the aim of this study was to determine the functions of mtTERT in vivo and in vitro. Therefore, mitochondria from hearts of adult, 2nd generation TERT-deficient mice (TERT -/-) and wt littermates were isolated and state 3 respiration was measured. Strikingly mitochondria from TERT -/- revealed a significantly lower state 3 respiration (TERTwt: 987 +/- 72 pmol/s*mg vs. TERT-/-: 774 +/- 38 pmol/s*mg, p < 0.05, n = 5). These results demonstrated that TERT -/- mice have a so far undiscovered heart phenotype. In contrast mitochondria isolated from liver tissues did not show any differences. To get further insights in the molecular mechanisms, we reduced endogenous TERT levels by shRNA and measured mitochondrial reactive oxygen species (mtROS). mtROS were increased after ablation of TERT (scrambled: 4.98 +/- 1.1% gated vs. shTERT: 2.03 +/- 0.7% gated, p < 0.05, n = 4). We next determined mtDNA deletions, which are caused by mtROS. Semiquantitative realtime PCR of mtDNA deletions revealed that mtTERT protects mtDNA from oxidative damage. To analyze whether mitochondrial integrity is required to protect from apoptosis, vectors with mitochondrially targeted TERT (mitoTERT) and wildtype TERT (wtTERT) were transfected and apoptosis was measured. mitoTERT showed the most prominent protective effect on H2O2 induced apoptosis. In conclusion, mtTERT has a protective role in mitochondria by importantly contributing to mtDNA integrity and thereby enhancing respiration capacity of the heart.
The peroxisome proliferator activated receptor gamma (PPARgamma) plays an eminent role during alternative activation of macrophages and resolution of inflammation. As an antiinflammatory signaling molecule, it seems likely that it is tightly regulated dependent on the state of the immune response. There is growing evidence that PPARgamma expression is reduced during inflammation, whereas molecular mechanisms are illdefined. Even though, its role in immunosuppression is getting more definite. Apoptotic cells (AC) provoke an active repression of pro-inflammatory responses inter alia by the inhibition of pro-inflammatory cytokine expression or attenuated generation of reactive oxygen species (ROS). The reduced formation of ROS was attributed to PPARgamma activation, while mechanisms behind the reduced cytokine expression remained unclear. Therefore, my Ph.D. thesis addressed the role of PPARgamma during inhibited cytokine synthesis in response to AC and the regulation of PPARgamma expression during an inflammatory response, which was initiated by lipopolysaccharide (LPS) exposure. In the first part of the thesis, I investigated the role of PPARgamma in coordinating the attenuation of pro-inflammatory cytokine expression in response to AC. Exposing murine RAW264.7 macrophages to AC prior to LPS-stimulation, reduced NFKB transactivation and lowered target gene expression of e.g. TNFalpha and IL-6 compared to controls. In macrophages over-expressing a dominant negative (d/n) mutant of PPARgamma, NFKB transactivation in response to LPS was restored, while using macrophages from myeloid lineage-specific conditional PPARgamma knock-out mice proved that PPARgamma transmitted the anti-inflammatory response delivered by AC. Domain analysis revealed that amino acids 32-250 are essential for inhibition of NFKB. Mutation of a SUMOylation (SUMO: small-ubiquitin related modifier) site in this region (K77R) and interfering SUMOylation by silencing the SUMO E3 ligase PIAS1 (protein inhibitor of activated Stat1) eliminated AC-provoked NFKB inhibition and concomitant TNFalpha expression. Chromatin-immunoprecipitation assays demonstrated that AC prevented the LPS-induced removal of nuclear receptor co-repressor (NCoR) from the KB response element within the TNFalpha promoter. I concluded that AC induce PPARgamma SUMOylation to attenuate the removal of NCoR, thereby blocking transactivation of NFKB. This contributes to an anti-inflammatory phenotype shift in macrophages in response to AC, by lowering pro-inflammatory cytokine production. The second part addressed molecular mechanisms responsible for reduced PPARgamma expression upon LPS exposure. PPARgamma gained considerable interest as a therapeutic target during chronic inflammatory diseases. Remarkably, the pathogenesis of diseases such as multiple sclerosis or Alzheimer’s disease is associated with impaired PPARgamma expression. Initiation of an inflammatory response by exposing primary human macrophages to LPS revealed a rapid decline of PPARgamma1 expression. PPARgamma1 mRNA decrease was prevented by inhibition of NFKB and also after pre-treatment with the PPARgamma agonist rosiglitazone, suggesting a NFKB-dependent pathway, because activated PPARgamma is known to inhibit NFKB transactivation. Since promoter activities were not affected by LPS, I focused on mRNA stability and noticed a decreased PPARgamma1 mRNA half-life. RNA stability is often regulated via 3’ untranslated regions (UTRs). Therefore, I analyzed the impact of the PPARgamma-3’UTR by luciferase assays. LPS significantly reduced luciferase activity of pGL3-PPARgamma-3’UTR, suggesting that PPARgamma1 mRNA is destabilized. Deletion of a potential miR-27a/b binding site within the 3’UTR completely restored luciferase activity. Moreover, inhibition of miR-27b, which was induced upon LPS-exposure, partially reversed PPARgamma1 mRNA decay, whereas the mature miR-27 mimicked the effect of LPS. MiR-27b was at least partially induced by NFKB, thus correlating with NFKB-dependent PPARgamma1 mRNA decrease. Since deletion of the miR-27 site also containing an AU-rich element (ARE) completely abrogated LPS-induced reduction but inhibition of miR-27b only partially restored PPARgamma1 mRNA expression, I suggested an additional implication of an ARE-binding protein. I provide evidence that LPS induces miR-27b, which in turn destabilizes PPARgamma1 mRNA. Understanding the molecular mechanism of PPARgamma mRNA destabilization, might help to rationalize inflammatory diseases associated with impaired PPARgamma expression. Even though, further experiments are needed to clarify the potential involvement of ARE-binding proteins.
Background Although current molecular clock methods offer greater flexibility in modelling historical evolutionary events, calibration of the clock with dates from the fossil record is still problematic for many groups. Here we implement several new approaches in molecular dating to estimate evolutionary ages of Lacertidae, an Old World family of lizards with a poor fossil record and uncertain phylogeny. Four different models of rate variation are tested in a new program for Bayesian phylogenetic analysis called TreeTime, based on a combination of mitochondrial and nuclear gene sequences. We incorporate paleontological uncertainty into divergence estimates by expressing multiple calibration dates as a range of probabilistic distributions. We also test the reliability of our proposed calibrations by exploring effects of individual priors on posterior estimates. Results According to the most reliable model, as indicated by Bayes factor comparison, modern lacertids arose shortly after the K/T transition and entered Africa about 45 million years ago, with the majority of their African radiation occurring in the Eocene and Oligocene. Our findings indicate much earlier origins for these clades than previously reported, and we discuss our results in light of paleogeographic trends during the Cenozoic. Conclusions This study represents the first attempt to estimate evolutionary ages of a specific group of reptiles exhibiting uncertain phylogenetic relationships, molecular rate variation and a poor fossil record. Our results emphasize the sensitivity of molecular divergence dates to fossil calibrations, and support the use of combined molecular data sets and multiple, well-spaced dates from the fossil record as minimum node constraints. The bioinformatics program used here, TreeTime, is publicly available, and we recommend its use for molecular dating of taxa faced with similar challenges.
In vielen Tumorzellen kommt es zu einer Überexpression des Hypoxie-induzierbaren Faktor 1alpha (HIF-1alpha), was zu einer verbesserten Anpassung des Tumors an die intratumorale Hypoxie sowie zu einer Resistenz gegen Strahlen- und Chemotherapie führt. Je nach Tumor kann HIF-1alpha auf verschiedenen Wegen induziert werden. Eine Möglichkeit ist die Hemmung des Abbaus von HIF-1alpha über das 26S-Proteasom, wie z.B. beim van Hippel-Lindau (VHL)-Syndrom aufgrund einer Mutation im VHL Gen. Patienten mit VHL-Syndrom entwickeln häufig renal clearcell carcinomas (RCCs). In diesen Karzinomen kann HIF-1alpha nicht über den klassischen Weg über das 26S-Proteasom abgebaut werden. Um das Verständnis für alternative Regulationsmechanismen von HIF-1alpha zu erweitern, wurde mit RCC4-Zellen gearbeitet. Im ersten Teil der vorliegenden Arbeit konnte gezeigt werden, dass in RCC4-Zellen das HIF-1alpha-Protein unter Hypoxie, in Kombination mit NO, durch die Ca2+-abhängige Protease Calpain abgebaut wird. Unter Hypoxie kam es zu einem Anstieg der Produktion von reaktiven Sauerstoffspezies (ROS) in den Mitochondrien, die mit NO zu Peroxynitrit und weiteren reaktiven Stickstoffintermediaten (RNI) reagierten. Die kombinierte Stimulation der Zellen mit NO und O2- unter Normoxie löste ebenfalls einen Anstieg des intrazellulären Ca2+-Gehaltes und der Calpain-Aktivität aus, was gleichzeitig zu einem reduzierten HIF-1alpha-Proteingehalt führte. Der Calpain-vermittelte HIF-1alpha-Abbau konnte auch in Zellen mit funktionellem VHL-Protein (pVHL) durch NO und O2- ausgelöst werden, wenn der proteasomale Abbau gehemmt war. Diese Ergebnisse beschreiben einen neuen Regulationsmechanismus für das HIF-1alpha-Protein, der unabhängig vom Sauerstoffgehalt und vom 26S-proteasomalen Abbau durch NO/O2- und Calpain erfolgt. Bisher war noch nicht bekannt, dass HIF-1alpha anders als über das 26S Proteasom abgebaut werden kann. In der vorliegenden Arbeit konnte gezeigt werden, dass der Calpain-vermittelte Abbau neben dem proteasomalen Abbau zur Regulierung von HIF 1 beiträgt. In Tumorgeweben stellt nicht nur HIF-1, welches in den Tumorzellen aktiviert ist, einen Selektionsvorteil für die Zellen des Tumorgewebes dar. Ebenso tragen die Zellen des Tumorstromas, darunter die Makrophagen, die in den Tumor einwandern, zur Progression des Tumors durch die Anpassung an die hypoxischen Umgebung bei. Daher wurde im zweiten Teil dieser Arbeit die Regulation von HIF-1alpha in den durch konditioniertes Medium von apoptotischen Zellen (KMAZ) aktivierten Makrophagen und der Bedeutung der daraus resultierenden HIF-1-Aktivierung untersucht. Makrophagen, die durch apoptotische Zellen (AZ) aktiviert werden, stellen einen anti-inflammatorischen, pro-angiogenetischen Phänotyp dar, der vergleichbar mit dem der Tumor-assozierten Makrophagen (TAMs) ist. TAMs infiltrieren in das Tumorgewebe und sind essentiell am Übergang von einem avaskulären zu einem invasiven, vaskularisierten und malignen Tumor beteiligt. Unsere Arbeitsgruppe konnte in Vorarbeiten zeigen, dass Makrophagen zunächst Tumorzellen abtöten, wodurch die apoptotischen Tumorzellen Mediatoren (u.a. Sphingosin-1-Phosphat (S1P)) freisetzen, die eine Polarisierung zu einem alternativen, TAM-ähnlichen-Phänotyp der Makrophagen bewirken. Die Inkubation der Makrophagen mit KMAZ führte zu einer Induktion der HIF-1alpha-mRNA und des -Proteins unter Normoxie, was unabhängig von der Proteinstabilität auf eine gesteigerte Proteinsynthese zurückgeführt werden konnte. Weiterhin führte die Induktion von HIF-1alpha zu einer gesteigerten HIF-1-Aktivität. Die Differenzierung von Stammzellen zu CD31+-Endothelzellen wurde durch die Überstände von den durch KMAZ polarisierten Makrophagen HIF-1-abhängig hervorgerufen und ist ein Indiz für die Ausbildung des HIF 1-vermittelten pro-angiogenetischen Phänotyps der Makrophagen. Als Mediatoren, die von den AZ freigesetzt wurden und an der HIF-1alpha-mRNA-Induktion beteiligt sind, konnten S1P und transforming growth factor-beta (TGF-beta) identifiziert werden. Des Weiteren kommt es zu einer Aktivierung des nuclear factor of activated T-cells (NFAT), der an den HIF-1alpha-Promotor bindet und die Transkription induziert. Aufgrund der verstärkten Synthese kommt es zur Akkumulation von HIF-1alpha und zur Aktivierung von HIF-1. Bisher ist die Aktivierung von HIF-1 in TAMs durch die Lokalisation in hypoxischen Arealen erklärt und nicht weiter untersucht worden. Die Erkenntnisse über die Regulierung von HIF-1 durch AZ beschreiben einen neuen Mechanismus, der zur HIF-1-Aktivierung auch unter Normoxie führt. Dabei vermitteln AZ statt der beschriebenen hypoxischen Stabilisierung des HIF-1α-Proteins eine Induktion der HIF-1alpha-mRNA. Weiterhin zeigen die Ergebnisse eine Möglichkeit auf, wie TAMs bereits unter Normoxie zur Angiogenese Induktion in Tumoren beitragen können und erweitern damit das Verständnis, wie die Tumor-unterstützende Wirkung der TAMs vermittelt wird.
Starting from the first observation of the halo phenomenon 20 years ago, more and more neutron-rich light nuclei were observed. The study of unstable nuclear systems beyond the dripline is a relatively new branch of nuclear physics. In the present work, the results of an experiment at GSI (Darmstadt) with relativistic beams of the halo nuclei 8He, 11Li and 14Be with energies of 240, 280 and 305 MeV/nucleon, respectively, impinging on a liquid hydrogen target are discussed. Neutron/proton knockout reactions lead to the formation of unbound systems, followed by their immediate decay. The experimental setup, consisting of the neutron detector LAND, the dipole spectrometer ALADIN and different types of tracking detectors, allows the reconstruction of the momentum vectors of all reaction products measured in coincidence. The properties of unbound nuclei are investigated by reconstructing the relative-energy spectra as well as by studying the angular correlations between the reaction products. The observed systems are 9He, 10He, 10Li, 12Li and 13Li. The isotopes 12Li and 13Li are observed for the first time. They are produced in the 1H(14Be, 2pn)12Li and 1H(14Be, 2p)13Li knockout reactions. The obtained relative-energy spectrum of 12Li is described as a single virtual s-state with a scattering length of as = -22;13.7(1.6) fm. The spectrum of 13Li is interpreted as a resonance at an energy of Er = 1.47(13) MeV and a width of Gamma ~ 2 MeV superimposed on a broad correlated background distribution. The isotope 10Li is observed after one-neutron knockout from the halo nucleus 11Li. The obtained relative-energy spectrum is described by a low-lying virtual s-state with a scattering length as = -22.4(4.8) fm and a p-wave resonance with Er = 0.566(14) MeV and Gamma = 0.548(30) MeV, in agreement with previous experiments. The observation of the nucleus 8He in coincidence with one or two neutrons, as a result of proton knockout from 11Li, allows to reconstruct the relative-energy spectra for the heavy helium isotopes, 9He and 10He. The low-energy part of the 9He spectrum is described by a virtual s-state with a scattering length as = -3.16(78) fm. In addition, two resonance states with l 6= 0 at energies of 1.33(8) and 2.4 MeV are observed. For the 10He spectrum, two interpretations are possible. It can be interpreted as a superposition of a narrow resonance at 1.42(10) MeV and a broad correlated background distribution. Alternatively, the spectrum is being well described by two resonances at energies of 1.54(11) and 3.99(26) MeV. Additionally, three-body energy and angular correlations in 10He and 13Li nuclei at the region of the ground state (0 < ECnn < 3 MeV) are studied, providing information about structure of these unbound nuclear systems.
Background: Intrauterine growth restriction is associated with an increased future risk for developing cardiovascular diseases. Hypoxia in utero is a common clinical cause of fetal growth restriction. We have previously shown that chronic hypoxia alters cardiovascular development in chick embryos. The aim of this study was to further characterize cardiac disease in hypoxic chick embryos. Methods: Chick embryos were exposed to hypoxia and cardiac structure was examined by histological methods one day prior to hatching (E20) and at adulthood. Cardiac function was assessed in vivo by echocardiography and ex vivo by contractility measurements in isolated heart muscle bundles and isolated cardiomyocytes. Chick embryos were exposed to vascular endothelial growth factor (VEGF) and its scavenger soluble VEGF receptor-1 (sFlt-1) to investigate the potential role of this hypoxia-regulated cytokine. Principal Findings: Growth restricted hypoxic chick embryos showed cardiomyopathy as evidenced by left ventricular (LV) dilatation, reduced ventricular wall mass and increased apoptosis. Hypoxic hearts displayed pump dysfunction with decreased LV ejection fractions, accompanied by signs of diastolic dysfunction. Cardiomyopathy caused by hypoxia persisted into adulthood. Hypoxic embryonic hearts showed increases in VEGF expression. Systemic administration of rhVEGF165 to normoxic chick embryos resulted in LV dilatation and a dose-dependent loss of LV wall mass. Lowering VEGF levels in hypoxic embryonic chick hearts by systemic administration of sFlt-1 yielded an almost complete normalization of the phenotype. Conclusions/Significance: Our data show that hypoxia causes a decreased cardiac performance and cardiomyopathy in chick embryos, involving a significant VEGF-mediated component. This cardiomyopathy persists into adulthood.
Durch das MoMiG ist der durch das sog. Novemberurteil begründeten restriktiven Rechtsprechung des BGH zur grundsätzlichen Unzulässigkeit von Darlehen aus gebundenem Vermögen an Gesellschafter die Grundlage entzogen worden. In seiner MPS-Entscheidung vom 1. Dezember 2008 hat der BGH daraus die Konsequenzen für §§ 311 ff. AktG gezogen. Entscheidend ist nunmehr, ob der Rückerstattungsanspruch vollwertig ist oder ob ein konkretes Ausfallrisiko besteht. Der Beitrag analysiert die Implikationen der Entscheidung für die Vermögensbindung und das Recht des faktischen Konzerns. Der Schwerpunkt liegt dabei auf der Frage, wie die Kriterien der Vollwertigkeit bzw. des konkreten Ausfallrisikos zu konkretisieren sind.

Nach den erfolgreichen Workshops in den Jahren 2006 an der TU Darmstadt und 2007 an der TU München konnten wir dieses Mal einer Einladung der Universität Osnabrück folgen. Auf Vorschlag u. a. von unserem Ehrenmitglied Prof. Dr. E.-G. Mahn (Halle/Saale) wurde bereits auf der Mitgliederversammlung in Erlangen 2005 auf den Bedarf zusätzlicher Aktivitäten der Arbeitsgemeinschaft, z. B. zu praxisrelevanten Fragen hingewiesen. An dem 3. Workshop in Osnabrück nahmen über 60 Mitglieder und weitere Interessierte teil. 13 Vorträge und 9 Poster-Demonstrationen deckten verschiedenste Aspekte angewandter Fragestellungen ab. Auch dieses Mal zeigte sich die Kooperation mit der Arbeitsgemeinschaft Forstliche Standorts- und Vegetationskunde, hier in einem Vortrag über Erfassung und Bewertung von Wald-Lebensraumtypen der europäischen FFH-Richtlinie in der NWTürkei (s. Beitrag von H. WALENTOWSKI & E. BERGMEIER in diesem Band).
In this paper, we analyze economies of scale for German mutual fund complexes. Using 2002-2005 data of 41 investment management companies, we specify a hedonic translog cost function. Applying a fixed effects regression on a one-way error component model there is clear evidence of significant overall economies of scale. On the level of individual mutual fund complexes we find significant economies of scale for all of the companies in our sample. With regard to cost efficiency, we find that the average mutual fund complexes in all size quartiles deviate considerably from the best practice cost frontier. Bei der vorliegenden prospektiven und verblindeten klinischen Vergleichsstudie wurde die Wirkung prophylaktisch inhalierten Iloprosts bei 40 Patienten ohne bekannte pulmonale Hypertonie untersucht, die sich einer elektiven chirurgischen Revaskularisierung der Koronargefäße unter Verwendung einer extrakorporalen Zirkulation (EKZ) unterzogen. Bei derartigen Operationen kommt es regelhaft zu einer generalisierten Entzündungsreaktion gefolgt von einer Rechtsherzbelastung unterschiedlichen Ausmaßes, was den postoperativen Verlauf teilweise erheblich kompliziert. Zur Therapie einer in diesem Rahmen aufgetretenen Rechtsherzbelastung hat sich inhaliertes Iloprost bereits vielfach als sehr wirksam erwiesen. Zusätzlich ist seit einiger Zeit auch eine antiinflammatorische Wirkung von Iloprost bekannt. Zielsetzung dieser Studie war es daher herauszufinden, ob auch die prophylaktische Applikation eine vorteilhafte Wirkung auf perioperative Hämodynamik und Oxygenierung sowie auf perioperative Inflammationsreaktion zeigen kann. Dazu wurde vor EKZ 20 Patienten 10µg aerosolisiertes Iloprost per inhalationem appliziert, 20 Patienten der Kontrollgruppe inhalierten 0,9%ige NaCl–Lösung. Nach Ende der EKZ wurden hämodynamische Parameter mittels Pulskonturanalyse (PiCCO®) und dessen Rechtsherz–Erweiterungsmonitors (VoLEF®) protokolliert sowie arterielle und gemischtvenöse Blutgasanalysen durchgeführt. Weiterhin wurden pro– und antiinflammatorische Zytokine (Interleukin–1beta, 6, 8 und 10 sowie Tumor–Nekrose–Faktor alpha) in Blutplasma und bronchoalveolärer Lavageflüssigkeit bestimmt Die Ergebnisse bei den meisten beobachteten Parametern in Studien– und Kontrollguppe unterschieden sich nicht signifikant voneinander. Jedoch zeigte sich in der Studiengruppe ein erhöhtes HZV sowie ein höherer kardialer Funktionsindex als Marker der Inotropie bei gleichzeitig niedrigerem systemischem Gefäßwiderstand. Da diese Wirkung nach Ende der üblichen Wirkdauer von Iloprost auftrat, interpretieren wir dies als einen Hinweis auf eine verbesserte hämodynamische Stabilität durch die prophylaktische Inhalation von Iloprost. Ein weiteres Ergebnis war ein etwas besseres Sauerstoffangebot in der Studiengruppe gegen Ende der Untersuchung. Die Analyse der Zytokinbestimmungen aus Blutplasma und Bronchialsekret konnte keine signifikanten Unterschiede zwischen den beiden Gruppen zeigen, eine antiinflammatorische Wirkung inhalierten Iloprosts konnte hier nicht dargestellt werden. Auch eine relevante Rechtsherzbelastung wurde nicht beobachtet. Da inhaliertes Iloprost seine vorteilhafte Wirkung auf postoperative Hämodynamik und kardiale Pumpleistung vor allem durch Senkung pathologisch erhöhter intrapulmonaler Widerstände entfaltet und eine antiinflammatorische Wirkung erst bei einer Inflammation zum Tragen kommen kann, ist zu erwägen ob nicht der inflammatorische Reiz bei dem untersuchten verhältnismäßig gesunden Patientenkollektiv zu gering war, um die vermuteten Effekte zu zeigen. Im Verlauf der Studie ist kein schwerer Zwischenfall aufgetreten, auch konnte kein nachteiliger Effekt prophylaktisch inhalierten Iloprosts bei dem vorliegenden Patientenkollektiv nachgewiesen werden. Eine Studie, die die Auswirkung von inhaliertem Iloprost auf Hämodynamik, Oxygenierung und Inflammation an kardial vorbelasteten Patienten mit langer Operations– und EKZ–Dauer untersucht, erscheint daher sinnvoll.
Riboswitches are a novel class of genetic control elements that function through the direct interaction of small metabolite molecules with structured RNA elements. The ligand is bound with high specificity and affinity to its RNA target and induces conformational changes of the RNA's secondary and tertiary structure upon binding. To elucidate the molecular basis of the remarkable ligand selectivity and affinity of one of these riboswitches, extensive all-atom molecular dynamics simulations in explicit solvent ({approx}1 µs total simulation length) of the aptamer domain of the guanine sensing riboswitch are performed. The conformational dynamics is studied when the system is bound to its cognate ligand guanine as well as bound to the non-cognate ligand adenine and in its free form. The simulations indicate that residue U51 in the aptamer domain functions as a general docking platform for purine bases, whereas the interactions between C74 and the ligand are crucial for ligand selectivity. These findings either suggest a two-step ligand recognition process, including a general purine binding step and a subsequent selection of the cognate ligand, or hint at different initial interactions of cognate and noncognate ligands with residues of the ligand binding pocket. To explore possible pathways of complex dissociation, various nonequilibrium simulations are performed which account for the first steps of ligand unbinding. The results delineate the minimal set of conformational changes needed for ligand release, suggest two possible pathways for the dissociation reaction, and underline the importance of long-range tertiary contacts for locking the ligand in the complex.
A survey on worries of pregnant women - testing the German version of the Cambridge Worry Scale

Background: Pregnancy is a transition period in a woman's life characterized by increased worries and anxiety. The Cambridge Worry Scale (CWS) was developed to assess the content and extent of maternal worries in pregnancy. It has been increasingly used in studies over recent years. However, a German version has not yet been developed and validated. The aim of this study was (1) to assess the extent and content of worries in pregnancy on a sample of women in Germany using a translated and adapted version of the Cambridge Worry Scale, and (2) to evaluate the psychometric properties of the German version. Methods: We conducted a cross-sectional study and enrolled 344 pregnant women in the federal state of Baden-Wurttemberg, Germany. Women filled out structured questionnaires that contained the CWS, the Spielberger-State-Trait-Anxiety Inventory (STAI), as well as questions on their obstetric history. Antenatal records were also analyzed. Results: The CWS was well understood and easy to fill in. The major worries referred to the process of giving birth (CWS mean value 2.26) and the possibility that something might be wrong with the baby (1.99), followed by coping with the new baby (1.57), going to hospital (1.29) and the possibility of going into labour too early (1.28). The internal consistency of the scale (0.80) was satisfactory, and we found a four-factor structure, similar to previous studies. Tests of convergent validity showed that the German CWS represents a different construct compared with state and trait anxiety but has the desired overlap. Conclusions: The German CWS has satisfactory psychometric properties. It represents a valuable tool for use in scientific studies and is likely to be useful also to clinicians.
Gauging risk with higher moments : handrails in measuring and optimising conditional value at risk

The aim of the paper is to study empirically the influence of higher moments of the return distribution on conditional value at risk (CVaR). To be more exact, we attempt to reveal the extent to which the risk given by CVaR can be estimated when relying on the mean, standard deviation, skewness and kurtosis. Furthermore, it is intended to study how this relationship can be utilised in portfolio optimisation. First, based on a database of 600 individual equity returns from 22 emerging world markets, factor models incorporating the first four moments of the return distribution have been constructed at different confidence levels for CVaR, and the contribution of the identified factors in explaining CVaR was determined. Following this the influence of higher moments was examined in portfolio context, i.e. asset allocation decisions were simulated by creating emerging market portfolios from the viewpoint of US investors. This can be regarded as a normal decisionmaking process of a hedge fund focusing on investments into emerging markets. In our analysis we compared and contrasted two approaches with which one can overcome the shortcomings of the variance as a risk measure. First of all, we solved in the presence of conflicting higher moment preferences a multi-objective portfolio optimisation problem for different sets of preferences. In addition, portfolio optimisation was performed in the mean-CVaR framework characterised by using CVaR as a measure of risk. As a part of the analysis, the pair-wise comparison of the different higher moment metrics of the meanvariance and the mean-CVaR efficient portfolios were also made. Throughout the work special attention was given to implied preferences to the different higher moments in optimising CVaR. We also examined the extent to which model risk, namely the risk of wrongly assuming normally-distributed returns can deteriorate our optimal portfolio choice. JEL Classification: G11, G15, C61
Ziel der vorliegenden Promotionsarbeit war die Herstellung und Charakterisierung einer neuen Stat5 Reportermaus zur Analyse der transkriptionellen Aktivität von STAT5 in verschiedenen Entwicklungsstadien, Zelltypen und Organen auf Einzelzellebene in vivo. Die Zusammenfassung dieser Promotionsarbeit gibt im Folgenden einen Überblick über den JAK/STAT Signalweg und seine einzelnen Komponenten. Das Hauptaugenmerk liegt hierbei auf STAT5, da es eine wichtige Rolle in der zellulären Entwicklung, Differenzierung und Proliferation spielt. Anschließend werden die Klonierung des Stat5 Reportergenkonstruktes und die Herstellung der Reportermaus durch DNA-Mikroinjektion besprochen und die Ergebnisse sowie Schlussfolgerungen der funktionellen in vivo Analyse dieses neuen Reportermausmodells dargestellt. Signal transducer and activator of transcription (STAT) Proteine gehören zu einer Familie von Transkriptionsfaktoren, die latent im Zytoplasma vorkommen. Diese Proteinfamilie besteht aus sieben Mitgliedern: STAT1, STAT2, STAT3, STAT4, STAT5a, STAT5b und STAT6. Alle STAT Proteine weisen eine konservierte Struktur auf, bestehend aus einer N-terminalen Domäne (NTD), einer Coiled-Coil-Domäne (CCD), einer DNA-Bindedomäne (DBD), einer Linkerdomäne (LD), einer src-homology 2- Domäne (SH2) und einer Transaktivierungsdomäne (TAD). Eine Vielzahl löslicher, extrazellulärer Signalmoleküle wie zum Beispiel Hormone, Zytokine und Wachstumsfaktoren binden an ihre spezifischen Oberflächenrezeptoren und Aktivieren so die JAK/STAT Signalkaskade. Dabei führt die Ligandenbindung an den entsprechenden Rezeptor zunächst zur Dimerisierung des Rezeptors und anschließend zur Transphosphorylierung von Janus Kinasen (JAKs). Aktivierte JAKs phosphorylieren dann den Rezeptor an spezifischen Tyrosinresten. An diese können STAT Proteine über ihre SH2 Domäne binden. Die gebundenen STAT Proteine werden anschließend durch JAKs an einem Tyrosinrest (und Serinrest) in der TAD phosphoryliert und dimerisieren im Zytoplasma. Dimerisierte STAT Proteine translozieren anschließend in den Nukleus und binden an spezifische DNA-Sequenzen, die sogenannten GAS (gamma-IFN-aktivierende Seite) Elemente in der Promotorregion ihrer Zielgene. GAS Elemente sind kurze palindromische DNA Regionen mit einer TTTCCNGGAAA Konsensussequenz. Nach Bindung der aktivierten, phosphorylierten STAT Proteine an die GAS Elemente werden weitere Kofaktoren, wie zum Beispiel das CREB Bindeprotein p300/CBP rekrutiert, die gemeinsam als Transkriptionsfaktoren wirken und die Transkription ihrer Zielgene anschalten. Die Identifizierung von STAT5 erfolgte im Rahmen von Promotorstudien am β-Casein Milchgen in der murinen Brustepithelzelllinie HC11 (Schmitt-Ney et al., 1991). Kurz darauf wurde STAT5 auch im Brustgewebe von laktierenden Mäusen, Ratten und Kühen gefunden. Bevor eine Sequenzhomologie zu Proteinen der STAT Genfamilie festgestellt wurde, wurde STAT5 zunächst MGF – „mammary gland factor“ genannt (Schmitt-Ney et al., 1992b; Wakao et al., 1992). Es sind zwei Stat5 Gene bekannt, Stat5a und Stat5b, die eine Sequenzhomologie von 96 % aufweisen und ihren größten Unterschied in der TAD Domäne zeigen. Da keine STAT-ähnlichen Proteine in Hefezellen identifiziert wurden, ist der JAK/STAT Signalweg nur für multizelluläre Organismen von Bedeutung, vermutlich weil diese auf komplexe Zell-Zell Kommunikationen angewiesen sind, um im Zellverband auf Signale in der Umgebung reagieren zu können. STAT5 im Speziellen reguliert neben der Entwicklung des Brustgewebes während der Schwangerschaft, die Produktion von Blutzellen in der fötalen Leber sowie die Zellproliferation während der adulten Hämatopoese. Im Embryo ist die fötale Leber der Ort der Hämatopoese, bevor hämatopoetische Stammzellen im Knochenmark kolonialisieren und sich die Leber zu einem metabolischen Organ entwickelt. In der Maus gelangen ab dem Embryonaltag E12 hämatopoetische Stammzellen aus der Aorta, den Gonaden und dem Mesonephros (Urniere), der sogenannten AGM Region, sowie aus der Plazenta durch den Blutstrom in die fötale Leber. Die Zellen proliferieren hier und migrieren etwa zwei Tage vor der Geburt (E18) ins Knochenmark, wo die Hämatopoese nach der Geburt erfolgt. Durch die Übermittlung einer Vielzahl von Zytokinsignalen reguliert STAT5 die Differenzierung der pluripotenten Zellen in reife Blutzellen und sorgt zusätzlich für die Generierung von Zellen, die anschließend in der Lage sind, das Knochenmark zu repopulieren. Ein STAT5 Verlust führt aufgrund einer auftretenden Anämie zu einer pränatalen Letalität. Während der adulten Hämatopoese fördert STAT5 hingegen die Zellproliferation und den Zellzyklus sowie die Apoptose in hämatopoetischen Stammzellen. Im Brustgewebe ist STAT5 sowohl in der Mammogenese als auch in der Laktogenese involviert. Die Aktivierung von STAT5 erfolgt hierbei durch eine Vielzahl von Faktoren, wie zum Beispiel Prolaktin und Erythropoietin. Der Phosphorylierungsstatus von STAT5 im virgin Stadium ist hierbei gering, steigt aber während der Schwangerschaft und Laktation stetig an und führt zur Aktivierung von einer Reihe von Zielgenen wie Milchproteinen, aber auch Zellzyklusregulatoren wie CyclinD1 und negativen Regulatoren des JAK/STAT Signalweges, wie zum Beispiel SOCS3. Nach der Laktation nimmt die Phosphorylierung von STAT5 hingegen ab und aufgrund von Apoptose kommt es zu einer Rückbildung des alveolaren Gewebes. Die Regulation der Apoptose erfolgt durch eine erhöhte STAT3 Phosphorylierung. Eine Deregulierung des JAK/STAT Signalweges wird in einer Vielzahl von Tumoren beobachtet. Hier liegt STAT5 typischerweise konstitutiv aktiv vor, führt dadurch zu einer verstärkten Zellproliferation und Angiogenese und verhindert gleichzeitig die Apoptose der mutierten Zellen und eine Immunantwort, was zusammen die Tumorentstehung begünstigt. Konstitutiv aktives STAT5 spielt vor allem bei der Entstehung von soliden Tumoren wie Brustkrebs sowie verschiedenen Leukämieformen wie zum Beispiel akute und chronische myelogene Leukämien eine wichtige Rolle. Neben diesen bereits bekannten STAT5 Funktionen ist die Funktion von aktivem, phosphoryliertem STAT5 im Kontext der Mausentwicklung und in adultem Gewebe noch unklar. Um die Rolle von STAT5 während der Entwicklung näher zu charakterisieren, wurden bereits verschiedene Mausmodelle generiert. Seit dem ersten Gentransfer in Mäuse im Jahre 1980 bieten transgene Tiere eine Möglichkeit, detaillierte Einblicke in zelluläre Prozesse im Rahmen der Entwicklung, des Stoffwechsels und der Entstehung von (Krebs-) Erkrankungen zu erlangen. Transgene Mäuse wurden somit zu einem wichtigen Modellsystem, das in der Lage ist, die Mechanismen, die hinter diesen Prozessen stehen, näher zu beleuchten. STAT5a und STAT5b knock out Mäuse sind überlebensfähig, zeigen jedoch phänotypische Unterschiede. Da eine Signalweiterleitung nach Prolaktininduktion in Brustgewebszellen von STAT5a knock out Mäusen nicht erfolgt, sind diese nicht in der Lage während der Schwangerschaft zu Proliferieren und zu Differenzieren. Die Deletion von STAT5a und STAT5b hingegen ist pränatal letal und die Embryos zeigen schwere Anämien aufgrund einer erhöhten Apoptoserate der erythroiden Zellen in der fötalen Leber. Zusätzlich zu den knock-out und gain-of-function Mäusen wurde die Generierung von Reportermäusen immer wichtiger, um spezifische Signalwege im Kontext des gesamten Organismus zu untersuchen. Das Ziel dieser Promotionsarbeit war somit die Herstellung und funktionelle Analyse einer neuen Stat5 Reportermaus. Hierfür wurde zunächst ein neues Stat5 Reporterkonstrukt kloniert. Dieses Reporterkonstrukt sollte eine Vielzahl spezifischer Eigenschaften aufweisen, um speziell durch phosphoryliertes STAT5 aktiviert zu werden: (i) ein LacZ Reportergen, (ii) Stat5 Responsive-Elemente und (iii) einen minimalen Promoter. Das LacZ Reportergen wurde hierbei gewählt, um die transktiptionelle Aktivität von STAT5 in Gewebeschnitten direkt durch Blaufärbung der Zellen zeigen zu können. Bei dem gewählten Promoter handelt es sich um einen Minimalpromoter, für die Bindung genereller Transkriptionsfaktoren. Eine Aktivierung des LacZ Reportergens erfolgt jedoch nur nach vorheriger Bindung eines Transaktivators. Damit STAT5 diese Funktion übernimmt wurden zusätzliche Responsive-Elemente aus dem β-Casein Gen in das Konstrukt eingefügt. Nach erfolgreicher Klonierung von insgesamt sieben verschiedenen Stat5 Reporterkonstrukten, wurde ihre spezifische Induzierbarkeit nach STAT5 Phosphorylierung mittels transienter Transfektionsstudien in vitro analysiert und bestätigt. Das p(Stat5RE)4-CMVmin-LacZ Konstrukt wurde anschließend zwischen humane matrix attachment regions (MAR) kloniert, die als sogenannte Insulatoren fungieren. Diese sollen in der transgenen Maus verhindern, dass entfernt bindende Faktoren die Expression der Reportergenkassette positiv (enhancer) oder negativ (silencer) beeinflussen. Zusätzlich zu den sieben hier generierten Stat5 Reporterkonstrukten, wurde das p(Stat5RE)4-CMVmin-LacZ Reportergenkonstrukt im Rahmen einer Diplomarbeit in einen lentiviralen Gentransfervektor kloniert. Dieser erlaubt die stabile Transduktion von Krebszellen und Primärzellen, so dass eine ineffiziente Transfektion dieser Zellen umgangen werden kann (Gäbel, 2009). Zur Herstellung der transgenen Stat5 Reportermaus wurde das linearisierte und aufgereinigte Stat5 Reporterkonstrukt mittels DNA-Mikroinjektion in den Pronukleus von 470 Eizellen von FVB und C57BL/6 Mäusen injiziert. Die Eizellen wurden anschließend in Ammenmäuse transplantiert. Von den 470 Eizellen kamen 57 Mäuse auf die Welt. Die Integration des Transgens wurde anschließend mittels PCR und Southern Blot analysiert und die Integration des kompletten Transgens konnte in zwei der 57 Mäuse festgestellt werden. Bei beiden transgen-positiven Mäusen handelte es sich um C57BL/6 Mäuse, die anschließend mit Wildtyp C57BL/6 Mäusen verpaart wurden. Nachkommen der F2 Generation wurden dann auf die spezifische Induzierbarkeit des Stat5 Reportergenkonstruktes durch phosphoryliertes STAT5 in vivo untersucht. Da der Phosphorylierungsstatus von STAT5 im Brustgewebe bereits eingehend untersucht wurde und bekannt ist, erfolgte zunächst die Analyse der Reportergenaktivität im murinen Brustgewebe. Hierfür wurde das Brustgewebe isoliert, fixiert und über Nacht gefärbt. Anschließend wurden Paraffinschnitte hergestellt und im Detail analysiert. Im Vergleich zu Wildtyp-Kontrollmäusen konnte die Aktivierung des Reportergens im Brustgewebe in verschiedenen Entwicklungsstadien, vor allem während der späten Schwangerschaft und der Laktation, durch Blaufärbung einzelner Zellen, gezeigt werden. Eine Korrelation der Blaufärbung mit der Phosphorylierung von STAT5 in diesen Zellen wurde anhand von immunhistologischen Färbungen von Paraffinschnitten mit Antikörpern gegen Stat5 und P-Stat5 gezeigt. Zusätzlich zu der hormonell induzierten STAT5 Phosphorylierung bedingt durch eine Schwangerschaft, wurde die Aktivierung des Reportergens durch das Verabreichen von LPS gezeigt. Eine Behandlung der Stat5 Reportermäuse mit LPS führt zu einer Phosphorylierung von STAT5 in Zellen des hämatopoetischen Systems, speziell Granulozyten und Makrophagen, und sollte anschließend das LacZ Reportergen in diesen Zellen aktivieren. Dies konnte durch die Färbung von Blut- und Knochenmarkzellen mit spezifischen Oberflächenmarkern, sowie einer Färbung mit FDG (Fluoresceindi-β-D-galactopyranoside) mittels FACS Analysen bestätigt werden. Das nicht-fluoreszierende FDG wird hierbei von der exprimierten β-Galaktosidase zunächst zu Fluoreszein-monogalactosid (FMG) und anschließend zum hoch fluoreszierenden Fluoreszein hydrolysiert, was eine messbare Erhöhung der Fluoreszenz nach sich zieht. Zusammenfassend konnte das Stat5-Reportergen sowohl durch endogene Signale als auch durch extern zugeführte Signale induziert werden. Anschließend erfolgte die Analyse der Reportergenaktivierung in anderen Organen der Stat5 Reportermaus. Hierbei konnte die Aktivierung des LacZ Reportergens sowohl in der Leber (Hepatozyten), Milz (Pulpa) und Niere (Mark und Rinde) als auch im Thymus (Lymphozyten und antigen präsentierende Zellen) und im Uterus (endometrisches Epithel) bestätigt werden. Diese Ergebnisse korrelieren mit zuvor durchgeführten Western Blot Analysen, die eine Phosphorylierung von STAT5 in eben diesen Organen gezeigt haben. Zusätzlich wurde phosphoryliertes STAT5 auf Proteinebene im Herz und im Gehirn gefunden, jedoch nicht in Gewebsschnitten der β-Galactosidase gefärbten Organe. Dies deutet darauf hin, dass das Reportergen trotz der Anwesenheit von phosphoryliertem STAT5, nicht immer eingeschaltet wird und somit weitere Faktoren für die transkriptionelle Aktivität von STAT5 notwendig sind. Western Blot Analysen sind somit alleine nicht ausreichend, um eine Aussage über die transkriptionelle Aktivität von phosphoryliertem STAT5 zu treffen, so dass die im Rahmen dieser Arbeit generierte Stat5 Reportermaus einen wichtigen Beitrag zum Verständnis von aktivem STAT5 bietet. Das generierte Stat5 Reportermausmodel wurde dann im Rahmen dieser Arbeit genutzt, um die Beteiligung von aktivem STAT5 in der Entwicklung von ΔTrkA induzierter akuter myeloischer Leukämie näher zu untersuchen. Hierfür wurden lineage negative Knochenmarkszellen aus den Stat5 Reportermäusen isoliert. Dabei werden sogenannte „Lin“ Antigene (z.B. CD3, CD4, CD8, Gr-1, Ter-119) genutzt, um reife murine Blutzellen zu identifizieren. Zellen, die diese Oberflächenmarker nicht oder nur in sehr geringen Mengen exprimieren, werden als lineage negativ bezeichnet. Ein Mix monoklonaler Antikörper gegen lineage Antigene kann somit zur Isolation lineage negativer Knochenmarkszellen genutzt werden. Diese negative Selektion führt letztendlich zur Anreicherung hämatopoetischer Stammzellen oder früher Progenitorzellen, die diese Marker (noch) nicht exprimieren. Diese Progenitorzellen wurden dann retroviral mit einem ΔTrkA Konstrukt transduziert und anschließend in bestrahlte Rag-1-/- Mäuse transplantiert und repopulierten in diesen das Knochenmark. Durch die ΔTrkA Transduktion wurde in den Rag-1-/- Mäusen myeloische Leukämie induziert. Jedoch konnte im Rahmen dieser Arbeit keine Aktivierung des Stat5 Reportergenkonstruktes beobachtet werden. Dies deutet darauf hin, dass STAT5 in ΔTrkA induzierten Leukämien keine Rolle spielt und bestätigt die Annahmen von Meyer et al. Durch die hier vorgestellten Ergebnisse bestätigt sich sowohl die Generierung eines neuen Stat5 Reportermausmodels als auch ihre spezifische Induzierbarkeit sowohl durch endogene hormonelle Prozesse (Schwangerschaft) als auch durch externe Manipulation (LPS Behandlung). Diese neue Stat5 Reportermaus wird in Zukunft als wichtiges und effizientes Modell fungieren, um die Rolle von transkriptionel aktivem STAT5 näher zu beleuchten. Hierbei wird sich der Fokus nicht nur auf die Rolle einzelner Zellen bei der normalen Entwicklung von Organen während verschiedener Entwicklungsstadien beschränken, sondern sich mehr und mehr in Richtung Tumorinitiierung und Tumorentwicklung bewegen. Anhand des hier generierten Stat5 Reportermausmodels können in Zukunft weitere Brustkrebs- und Leukämie-Tumormodelle herangezogen werden, um die Rolle und Funktion von STAT5 in der Tumorentwicklung in vivo detailliert analysieren zu können. Auf diesen Ergebnissen aufbauend wird dann die Möglichkeit bestehen, dieses neue Stat5 Reportermausmodell als Plattform zu nutzen, um zahlreiche neue Krebsmedikamente zu entwickeln und zu evaluieren.
In its admissibility decision in the Al-Saadoon case the ECtHR held that the United Kingdom had jurisdiction over the applicants, who had been arrested by British forces and kept in a British-run military prison in Iraq. Just before the respective mandate of the Security Council expired on 31 December 2008, the applicants were transferred to Iraqi custody at Iraqi request and thereby exposed to the risk of an unfair trial followed by capital punishment. In this respect, the case resembles the Soering case, although the applicants were, unlike Soering, not on British territory but on occupied Iraqi soil before they were handed over. This aspect raises the question of Iraqi sovereignty as a norm competing with the UK's human rights obligations. The authors trace back the ECtHR's case law concerning the extraterritorial application of the Convention and analyse the UK judgments and the ECtHR's admissibility decision in the Al-Saadoon affair from this angle. Furthermore they consider the doctrinal consequences of the ECHR's extraterritorial effect in cases like Soering and Al-Saadoon, where contracting parties violate guarantees of the Convention by exposing a person within their jurisdiction to a risk of a treatment contrary to these guarantees by a third state. Finally, they test the argument brought forward by the UK that not transferring the applicants would have violated Iraqi sovereignty and establish patterns how the ECtHR and the UK Courts did cope in the past with international law norms potentially competing with the Convention.
The goal of this research is to develop an understanding of what causes organizations and information systems to be “good” with regard to communication and coordination. This study (1) gives a theoretical explanation of how the processes of organizational adaptation work and (2) what is required for establishing and measuring the goodness of an organization with regard to communication and coordination. By leveraging concepts from cybernetics and philosophy of language, particularly the theoretical conceptualization of information systems as social systems and language communities, this research arrives at new insights. After discussing related work from systems theory, organization theory, cybernetics, and philosophy of language, a theoretical conceptualization of information systems as language communities is adopted. This provides the foundation for two exploratory field studies. Then a formal theory for explaining the adaptation of organizations via language and communication is presented. This includes measures for the goodness of organizations with regard to communication and coordination. Finally, propositions stemming from the theoretical model are tested using multiple case studies in six information system development projects in the financial services industry.
8. Workshop der Arbeitsgruppe Vegetationsdatenbanken zum Thema "Bioindikatoren" in Greifswald

Die Arbeitsgruppe Vegetationsdatenbanken widmet sich dem Aufbau und der Auswertung von pflanzensoziologischen Datenbanken in Deutschland. Sie trifft sich seit 2002 jährlich zu bundesweiten, vom Bundesamt für Naturschutz geförderten Workshops. Seit 2008 leistet die Floristisch-soziologische Arbeitsgemeinschaft zusätzliche finanzielle Unterstützung. Das 8. Arbeitstreffen fand vom 25. bis 27. Februar 2009 auf Einladung von Michael Manthey und Florian Jansen am Institut für Botanik und Landschaftsökologie der Universität Greifswald statt und wurde von 38 TeilnehmerInnen aus 8 europäischen Ländern besucht.
This thesis investigates the jet-medium interactions in a Quark-Gluon Plasma using a hydrodynamical model. Such a Quark-Gluon Plasma represents a very early stage of our universe and is assumed to be created in heavy-ion collisions. Its properties are subject of current research. Since the comparison of measured data to model calculations suggests that the Quark-Gluon Plasma behaves like a nearly perfect liquid, the medium created in a heavy-ion collision can be described applying hydrodynamical simulations. One of the crucial questions in this context is if highly energetic particles (so-called jets), which are produced at the beginning of the collision and traverse the formed medium, may lead to the creation of a Mach cone. Such a Mach cone is always expected to develop if a jet moves with a velocity larger than the speed of sound relative to the medium. In that case, the measured angular particle distributions are supposed to exhibit a characteristic structure allowing for direct conclusions about the Equation of State and in particular about the speed of sound of the medium. Several different scenarios of jet energy loss are examined (the exact form of which is not known from first principles) and different mechanisms of energy and momentum loss are analyzed, ranging from weak interactions (based on calculations from perturbative Quantum Chromodynamics, pQCD) to strong interactions (formulated using the Anti-de-Sitter/Conformal Field Theory Correspondence, AdS/CFT). Though they result in different angular particle correlations which could in principle allow to distinguish the underlying processes (if it becomes possible to analyze single-jet events), it is shown that the characteristic structure observed in experimental data can be obtained due to the different contributions of several possible jet trajectories through an expanding medium. Such a structure cannot directly be connected to the Equation of State. In this context, the impact of a strong flow created behind the jet is examined which is common to almost all jet deposition scenarios. Besides that, the transport equations for dissipative hydrodynamics are discussed which are fundamental for any numerical computation of viscous effects in a Quark-Gluon Plasma.
In this work we study the properties of quarkonium states in a quark-gluon plasma which, due to expansion and non-zero viscosity, exhibits a local anisotropy in momentum space. We determine the hard-loop resummed gluon propagator in an anisotropic QCD plasma in general linear gauges and define a potential between heavy quarks from the Fourier transform of its static limit. This potential which arises due to one-gluon exchange describes the force between a quark and anti-quark at short distances. It is closer to the vacuum potential as compared to the isotropic Debye screened potential which indicates the reduced screening in an anisotropic QCD plasma. In addition, angular dependence appears in the potential; we find that there is stronger attraction on distance scales on the order of the inverse Debye mass for quark pairs aligned along the direction of anisotropy than for transverse alignment. The potential at long distances, however, is non-perturbative and modeled as a QCD string which is screened at the same scale as the Coulomb field. At asymptotic separation the potential energy is non-zero and inversely proportional to the temperature. With a phenomenological potential model which incorporates the different behaviors at short and long distances, we solve the three-dimensional Schrödinger equation. Our numerical results show that quarkonium binding is stronger at non-vanishing viscosity and expansion rate, and that the anisotropy leads to polarization of the P-wave states. Furthermore, we determine viscosity corrections to the imaginary part of the heavyquark potential in the weak-coupling hard-loop approximation. The imaginary part is found to be smaller (in magnitude) than at vanishing viscosity. This implies a smaller decay width of quarkonium bound states in an anisotropic plasma.
Bayessche Methoden zur Schätzung von Stammbäumen mit Verzweigungszeitpunkten aus molekularen Daten

Ein großes Ziel der Evolutionsbiologie ist es, die Stammesgeschichte der Arten zu rekonstruieren. Historisch verwendeten Systematiker hierfür morphologische und anatomische Merkmale. Mit dem stetigen Zuwachs an verfügbaren Sequenzdaten werden heute verstärkt Methoden entwickelt und eingesetzt, welche die Rekonstruktion auf Basis von molekularen Daten ermöglichen. Im Fokus der aktuellen Forschung steht die Anwendung und Weiterentwicklung Bayesscher Methoden. Diese Methoden besitzen große Popularität, da sie in Verbindung mit Markov-Ketten-Monte-Carlo-Verfahren eingesetzt werden können, um einen Stammbaum zu vorgegebenen Spezies zu schätzen und dessen Variabilität zu bestimmen. Im Rahmen dieser Dissertation wurde die erweiterbare Software TreeTime entwickelt. TreeTime bietet Schnittstellen für die Einbindung von molekularen Evolutions- und Ratenänderungsmodellen und stellt neu entwickelte Methoden bereit, um Stammbäume mit Verzweigungszeitpunkten zu rekonstruieren. In TreeTime werden die molekularen Daten und die zeitlichen Informationen, wie z.B. Fossilfunde, in einem Bayes-Verfahren simultan berücksichtigt, um die Zeitpunkte der Artaufspaltungen genauer zu datieren. Für die Anwendung Bayesscher Methoden in der Rekonstruktion von Stammbäumen wird ein stochastisches Modell benötigt, das die Evolution der molekularen Sequenzen entlang den Kanten eines Stammbaums beschreibt. Der Mutationsprozess der Sequenzen wird durch ein molekulares Evolutionsmodell definiert. Die Verwendung der klassischen molekularen Evolutionsmodelle impliziert die Annahme einer konstanten Evolutionsgeschwindigkeit der Sequenzen im Stammbaum. Diese Annahme wird als Hypothese der molekularen Uhr bezeichnet und bildet die Grundlage zum Schätzen der Verzweigungszeiten des Stammbaums. Der Verzweigungszeitpunkt, an dem sich zwei Spezies im Stammbaum aufspalten, spiegelt sich in der Ähnlichkeit der zugehörigen molekularen Sequenzen. Je älter dieser Verzweigungszeitpunkt ist, desto größer ist die Anzahl der unterschiedlichen Positionen in den Sequenzen. Häufig ist jedoch die Annahme der molekularen Uhr verletzt, so dass in gewissen Teilbereichen eines Stammbaums eine erhöhte Evolutionsgeschwindigkeit nachweisbar ist. Falls die Verletzung konstanter Evolutionsgeschwindigkeiten nicht ausgeschlossen werden kann, sollten schwankende Mutationsraten in der Modellierung explizit berücksichtigt werden. Hierfür wurden verschiedene Ratenänderungsmodelle vorgeschlagen. Bisher sind nur wenige dieser Ratenänderungsmodelle in Softwarepaketen verfügbar und ihre Eigenschaften sind nicht ausreichend erforscht. Das Ziel dieser Arbeit ist die Entwicklung und Bereitstellung von Bayesschen Modellen und Methoden zum Schätzen von Stammbäumen mit Verzweigungszeitpunkten. Die Methoden sollten auch bei unterschiedlichen Evolutionsgeschwindigkeiten im Stammbaum anwendbar sein. Vorgestellt wird ein neues Ratenänderungsmodell, eine neue Möglichkeit der Angabe von flexiblen Beschränkungen für die Topologie des Stammbaums sowie die Nutzung dieser Beschränkungen für die zeitliche Kalibrierung. Das neue Raten Änderungsmodell sowie die topologischen und zeitlichen Beschränkungen werden in einen modularen Softwareentwurf eingebettet. Durch den erweiterbaren Entwurf können bestehende und zukünftige molekulare Evolutionsmodelle und Ratenänderungsmodelle in die Software eingebunden und verwendet werden. Die vorgestellten Modelle und Methoden werden gemäß dem Softwareentwurf in das neu entwickelte Programm TreeTime aufgenommen und effzient implementiert. Zusätzlich werden bereits vorhandene Modelle programmiert und eingebunden, die nicht in anderen Softwarepaketen verfügbar sind. Des Weiteren wird eine neue Methode entwickelt und angewendet, um die Passgenauigkeit eines Modells für die Apriori-Verteilung auf der Menge der Baumtopologien zu beurteilen. Diese Methode wird zur Auswahl geeigneter Modelle benutzt, indem eine Auswertung der beobachteten Baumtopologien der Datenbank TreeBASE durchgeführt wird. Anschließend wird die Software TreeTime in einer Simulationsstudie eingesetzt, um die Eigenschaften der implementierten Ratenänderungsmodelle zu vergleichen. Die Software wird für die Rekonstruktion des Stammbaums zu 38 Spezies aus der Familie der Eidechsen (Lacertidae) verwendet. Da die zugehörigen molekularen Daten von der Hypothese der molekularen Uhr abweichen, werden unterschiedliche Ratenänderungsmodelle bei der Rekonstruktion verwendet und abschließend bewertet. ........
IL-18, a recently identified member of IL-1 family, is now recognized as an important regulator of innate and acquired immune responses. Therefore, the antitumor activities of IL-18 have been investigated. IL-18 has been shown to induce IFN-γ production by T, B, and NK cells, enhances NK cell activity, activates Fas ligandmediated apoptosis of the tumor cells, and improves the overall antitumor immunity. KG-1 cells were derived from a patient with acute myeloid leukemia (AML). IL-18 has been shown to induce IFN-γ production in those leukemic cells. TLR-3, in addition to its ability to recognize viral double stranded RNA, also can recognize the synthetic analogue poly(I:C) and induces type I IFN, inflammatory cytokine production, e.g TNF-α, and maturation of denderitic cells. In the present work the potential modulatory effect of PIC on IFN-γ and TNF-α production by KG-1 cells treated with IL-18 was investigated. Indeed, PIC strongly amplified the production of IFN-γ induced by IL-18 on mRNA and protein levels via NF-κB as well as p38 and JNK MAPK activation. Compared to IFN-γ, TNF-α showed different behaviour in KG-1 cells. On mRNA level I found only weak induction of TNF-α by IL-18 which was potentiated in the presence of PIC. Similarly, the release of TNF-α by IL-18 plus PIC required NF-κB as well as p38 and JNK MAPK activation. Furthermore, in the present work I found that TLR-3 is required for IFN-γ and TNF-α production. In addition, it is demonstrated by immunofluoresence that TLR-3 is localized in cytoplasm but not on the cell surface in KG-1 cells. Recently, it has been demonstrated that IFN-γ shows therapeutic potential as detected in AML blasts, specifically via inhibition of proliferation and induction of apoptosis. Thus our data could serve as a rationale for the clinical use of PIC and IL-18 in combination therapy. In search for new cytokines potentially modulated by the combination IL-18 plus PIC in KG-1 cells, cytokine antibody array analysis was performed. I found an upregulation of expected genes like IP-10 but most interestingly unexpected upregulation of PDGF-AA. Searching for detailed mechanisms of PDGF-AA induction, I found that neither p38 nor JNK is involved in PDGF-AA production but NF-κB is essential for the expression of PDGF-AA. Furthermore, I found that PDGF-AA is not able to increase the proliferation of KG-1 cells. PDGF and TGF-β are examples of signaling molecules which control the growth, survival, motility, and differentiation of cells. Therefore, the release of TGF-β by IL-18 plus PIC was monitored by ELISA. The level of TGF-β in cellular supernatants revealed that neither PIC nor IL-18 was able to significantly mediate release of TGF-β indicating that only PDGF-AA but not TGF-β is induced by PIC and IL-18 in KG-1 cells. To the best of our knowledge this is the first time that IL-18 or PIC is shown to induce the expression of PDGF-AA in KG-1 cells.
Analyzing interest rate risk: stochastic volatility in the term structure of government bond yields

We propose a Nelson-Siegel type interest rate term structure model where the underlying yield factors follow autoregressive processes with stochastic volatility. The factor volatilities parsimoniously capture risk inherent to the term structure and are associated with the time-varying uncertainty of the yield curve’s level, slope and curvature. Estimating the model based on U.S. government bond yields applying Markov chain Monte Carlo techniques we find that the factor volatilities follow highly persistent processes. We show that slope and curvature risk have explanatory power for bond excess returns and illustrate that the yield and volatility factors are closely related to industrial capacity utilization, inflation, monetary policy and employment growth. The mTOR kinase inhibitor rapamycin (sirolimus) is a drug with potent immunosuppressive and antiproliferative properties. We found that rapamycin induces the TGF/Smad signaling cascade in rat mesangial cells (MC) as depicted by the nuclear translocation of phospho-Smads 2, -3 and Smad-4, respectively. Concomitantly rapamycin increases the nuclear DNA binding of receptor (R)- and co-Smad proteins to a cognate Smad-binding element (SBE) which in turn causes an increase in profibrotic gene expression as exemplified by the connective tissue growth factor (CTGF) and plasminogen activator inhibitor 1 (PAI-1). Using small interfering (si)RNA we demonstrate that Smad 2/3 activation by rapamycin depends on its endogenous receptor FK-binding protein 12 (FKBP12). Mechanistically, Smad induction by rapamycin is initiated by an increase in active TGF1 as shown by ELISA and by the inhibitory effects of a neutralizing TGF antibody. Using an activin receptor-like kinase (ALK)-5 inhibitor and by siRNA against the TGF type II receptor TGF-RII) we furthermore demonstrate a functional involvement of both types of TGF receptors. However, rapamycin did not compete with TGFfor TGF-receptor binding as found in radioligand-binding assay. Besides SB203580, a specific inhibitor of the p38 MAPK, the reactive oxygen species (ROS) scavenger N-acetyl-cysteine (NAC) and a cell-permeable superoxide dismutase (SOD) mimetic strongly abrogated the stimulatory effects of rapamycin on Smad 2 and 3 phosphorylation. Furthermore, the rapid increase in Dichlorofluorescein (DCF) formation implies that rapamycin mainly acts through ROS. In conclusion, activation of the profibrotic TGFSmad signaling cascade accompanies the immunosuppressive and antiproliferative actions of rapamycin. Bioaktive Sphingolipide spielen eine wichtige Rolle bei der Regulation vieler entscheidender Prozesse in Endothelzellen. Speziell dem S1P wird eine Schlüsselfunktion in der Regulation der Proliferation und Migration von Endothelzellen beigemessen. Diese Prozesse sind notwendige Teilschritte in der Angiogenese, welche eine wichtige biologische und medizinische Bedeutung hat. So ist die umorangiogenese eine Voraussetzung für die adäquate Versorgung des Tumors mit Sauerstoff und Nährstoffen und trägt des Weiteren zu dessen Aggressivität bei. Ein weiteres Merkmal vieler solider Tumoren ist das Vorkommen hypoxischer Bereiche und erhöhter NO-Spiegel, die zudem, je nach Konzentration, als proangiogene Faktoren wirken können. Das Hauptziel dieser Arbeit lag in der Untersuchung des Einflusses hypoxischer Bedingungen und des Signalmoleküls NO auf das Sphingolipidgleichgewicht in der humanen Endothelzelllinie EA.hy 926. Ein spezieller Fokus wurde dabei auf die Regulation der S1P synthetisierenden Sphingosinkinasen (SK) und der daraus resultierenden Beeinflussung angiogener Zellantworten gelegt. In dieser Arbeit konnte gezeigt werden, dass sowohl Hypoxie als auch NO die Aktivität der SK-1, jedoch nicht der SK-2, langanhaltend erhöhten. Dieser Aktivitätsanstieg wurde durch eine Aktivierung des SK-1-Promotors mit einer nachfolgenden verstärkten mRNA- und Proteinexpression vermittelt. Dabei war unter Hypoxie der Transkriptionsfaktor HIF-1α der entscheidende SK-1 regulierende Faktor. Zusätzlich führte die durch Hypoxie ausgelöste Hochregulation der SK-1 zu einem Anstieg der zellulären S1P-Spiegel und zu einer Reduktion der Sphingosinkonzentration. Mechanistische Untersuchungen des Effekts von NO auf die SK-1 zeigten, dass dieser unabhängig von erhöhten cGMP-Konzentrationen aufgrund einer Aktivierung der löslichen Guanylatzyklase (sGC) durch NO war. Zum einen hatte die Verwendung des cGMP-Analogons 8-Bromo-cGMP und des Aktivators der sGC YC-1 keinen Einfluss auf die SK-1-Proteinexpression, und zum anderen hatte die Kostimulation mit dem sGC-Inhibitor ODQ keinen hemmenden Effekt auf die durch NO ausgelöste Hochregulation der SK-1. Demgegenüber ist die klassische MAPK-Kaskade essenziell für die Wirkung von NO auf die SK-1, da der MEK-Inhibitor U0126 die erhöhte Proteinexpression der SK-1 verhindern konnte. Die Aktivierung des vorgeschalteten kleinen GTP-bindenden Proteins p21ras ist ein möglicher Mechanismus, über den NO die Aktivierung der MAPKKaskade verursachen könnte (Lander et al, 1995). In einem weiteren Teil der Arbeit wurde die funktionelle Konsequenz einer Regulation der SK-1 durch Hypoxie und NO in EA.hy 926 Zellen untersucht. Sowohl Hypoxie als auch NO wurden als pro-angiogene Faktoren beschrieben. Daher wurde anhand von Migrationsassays und in-vitro-Angiogeneseassays untersucht, ob die SK-1 bei diesen Zellantworten eine Rolle spielt. Die Depletion der SK-1 durch die Verwendung einer spezifischen siRNA oder mit Hilfe von lentiviralen shRNA-Konstrukten zeigte, dass die Hochregulation der SK-1 unter Hypoxie und NO essenziell für die erhöhte Migration der Endothelzellen ist. Zusammenfassend zeigen die Daten, dass hypoxische Bedingungen und NO zu einer Hochregulation der SK-1 in EA.hy 926 Zellen führten und eine erhöhte Migrationsrate dieser Zellen zur Folge hatten. Somit ist die SK-1 ein interessanter therapeutischer Ansatzpunkt zur Behandlung von Krankheiten, die mit einer pathologischen Angiogenese einhergehen, wie es im Tumorgeschehen der Fall ist.
Hintergrund: Adipositas und die mit ihr assoziierten Komorbiditäten verursachen eine Verkürzung der Lebenserwartung und mindern die Lebensqualität in einer wachsenden Patienten Bevölkerung. Die bariatrische laparoskopische Chirurgie kann effizient zu einer Gewichtsreduktion beitragen und die damit verbundenen Komorbiditäten senken. In dieser Longitudinal-Studie untersuchten wir den Einfluss von präoperativem BMI, Geschlecht und operative Techniken auf den Verlauf von drei Komorbiditäten. Methodik: Alle Patienten (N=448) die sich einem bariatrischen Eingriff im Zeitraum von 2001 bis 2005 in unserem Krankenhaus unterzogen, erhielten einen Fragebogen, welcher unter anderem zur Evaluation der drei Komorbiditäten Hypertonie, Diabetes mellitus Typ II und Gelenkbeschwerden präoperativ, sowie 6 und 12 Monate nach der Operation diente. Von insgesamt 191 Patienten erhielten wir alle Fragebögen zurück. Der Rückgang der Komorbiditäten im Verlauf wurde unter Zuhilfenahme des Chochrans Q-Test evaluiert. Für die Vorhersage für eine Verbesserung der Komorbiditäten nach 12 Monaten verwendeten wir eine Diskriminanzanalyse. Ergebnisse: Es fand sich ein signifikanter Rückgang bei der Hypertonie (Q=47,26, p<0,001) sowohl nach 6 Monaten (29,3%), als auch nach 12 Monaten (20,4%), verglichen mit den Baseline Erhebungen (40,3%). In einer detaillierten Untersuchung fand sich dieser Rückgang unabhängig vom präoperativen BMI, Geschlecht und der verwendeten operativen Technik. Alter, Anzahl der eingenommen Medikamente und der Excess-weight-loss konnten als Prädiktoren für einen signifikanten Rückgang der Hypertonie nach 12 Monaten identifiziert werden. Mittels der Diskriminanzanalyse konnte unter Verwendung dieser drei Variablen zu 69,5% eine Vorhersage für das Vorhandensein einer Hypertonie nach 12 Monaten vorgenommen werden. Auch für den Diabetes mellitus Typ II (Q=26.69, p<0,001) und die Gelenkbeschwerden (Q=22,68, p<0,001) wurde ein signifikanter Rückgang herausgearbeitet. Schlussfolgerung: Zusätzlich zu dem primären Ziel der Gewichtsreduktion, war nach durchgeführtem laparoskopisch bariatrischem Eingriff ein signifikanter Rückgang der untersuchten Komorbiditäten Hypertonie, Diabetes mellitus Typ II und Gelenkbeschwerden zu belegen. Diese könnte sowohl eine Steigerung der Lebenserwartung, als auch eine Kostenreduzierung im Gesundheitssystem bedeuten. Background Treatment options for metastatic renal cell carcinoma (RCC) are limited due to resistance to chemo- and radiotherapy. The development of small-molecule multikinase inhibitors have now opened novel treatment options. The influence of the receptor tyrosine kinase inhibitor AEE788, applied alone or combined with the mammalian target of rapamycin (mTOR) inhibitor RAD001, on RCC cell adhesion and proliferation in vitro has been evaluated. Methods RCC cell lines Caki-1, KTC-26 or A498 were treated with various concentrations of RAD001 or AEE788 and tumor cell proliferation, tumor cell adhesion to vascular endothelial cells or to immobilized extracellular matrix proteins (laminin, collagen, fibronectin) evaluated. The anti-tumoral potential of RAD001 combined with AEE788 was also investigated. Both, asynchronous and synchronized cell cultures were used to subsequently analyze drug induced cell cycle manipulation. Analysis of cell cycle regulating proteins was done by western blotting. Results RAD001 or AEE788 reduced adhesion of RCC cell lines to vascular endothelium and diminished RCC cell binding to immobilized laminin or collagen. Both drugs blocked RCC cell growth, impaired cell cycle progression and altered the expression level of the cell cycle regulating proteins cdk2, cdk4, cyclin D1, cyclin E and p27. The combination of AEE788 and RAD001 resulted in more pronounced RCC growth inhibition, greater rates of G0/G1 cells and lower rates of S-phase cells than either agent alone. Cell cycle proteins were much more strongly altered when both drugs were used in combination than with single drug application. The synergistic effects were observed in an asynchronous cell culture model, but were more pronounced in synchronous RCC cell cultures. Conclusions Potent anti-tumoral activitites of the multikinase inhibitors AEE788 or RAD001 have been demonstrated. Most importantly, the simultaneous use of both AEE788 and RAD001 offered a distinct combinatorial benefit and thus may provide a therapeutic advantage over either agent employed as a monotherapy for RCC treatment.
It has been shown that certain chemokine receptor polymorphisms may correspond to certain complications after organ transplantation. Ischemic-type biliary lesion (ITBL) encounters for major morbidity and mortality in liver transplant recipients. So far, the exact cause for ITBL remains unclear. Certain risk factors for the development of ITBL like donor age and cold ischemic time are well described. In a previous study, a 32-nucleotide deletion of the chemokine receptor-5Delta32 (CCR-5Delta32) was strongly associated with the incidence of ITBL in adult liver transplantation. This study re-evaluates the association of CCR-5Delta32 gene polymorphism and the incidence of ITBL. 169 patients were included into this retrospective analysis. 134 patients were homozygous for wild-type CCR-5, 33 patients heterozygous, and 2 patients were homozygous for CCR-5Delta32 mutation. There were no major differences in donor or recipients demographics. No association was found between CCR-5Delta32 mutation and the development of ITBL.We conclude that CCR-5Delta32 is no risk factor for the development of ITBL in our patient cohort.
Despite their importance in modern electronic trading, virtually no systematic empirical evidence on the market impact of incoming orders is existing. We quantify the short-run and long-run price effect of posting a limit order by proposing a high-frequency cointegrated VAR model for ask and bid quotes and several levels of order book depth. Price impacts are estimated by means of appropriate impulse response functions. Analyzing order book data of 30 stocks traded at Euronext Amsterdam, we show that limit orders have significant market impacts and cause a dynamic (and typically asymmetric) rebalancing of the book. The strength and direction of quote and spread responses depend on the incoming orders’ aggressiveness, their size and the state of the book. We show that the effects are qualitatively quite stable across the market. Cross-sectional variations in the magnitudes of price impacts are well explained by the underlying trading frequency and relative tick size.
We introduce a regularization and blocking estimator for well-conditioned high-dimensional daily covariances using high-frequency data. Using the Barndorff-Nielsen, Hansen, Lunde, and Shephard (2008a) kernel estimator, we estimate the covariance matrix block-wise and regularize it. A data-driven grouping of assets of similar trading frequency ensures the reduction of data loss due to refresh time sampling. In an extensive simulation study mimicking the empirical features of the S&P 1500 universe we show that the ’RnB’ estimator yields efficiency gains and outperforms competing kernel estimators for varying liquidity settings, noise-to-signal ratios, and dimensions. An empirical application of forecasting daily covariances of the S&P 500 index confirms the simulation results.
We examine intra-day market reactions to news in stock-specific sentiment disclosures. Using pre-processed data from an automated news analytics tool based on linguistic pattern recognition we extract information on the relevance as well as the direction of company-specific news. Information-implied reactions in returns, volatility as well as liquidity demand and supply are quantified by a high-frequency VAR model using 20 second intervals. Analyzing a cross-section of stocks traded at the London Stock Exchange (LSE), we find market-wide robust news-dependent responses in volatility and trading volume. However, this is only true if news items are classified as highly relevant. Liquidity supply reacts less distinctly due to a stronger influence of idiosyncratic noise. Furthermore, evidence for abnormal highfrequency returns after news in sentiments is shown. JEL-Classification: G14, C32
