Viele mikroskopische Vorgänge in Festkörpern und molekularen Verbindungen sind verbunden mit Änderungen ihres Magnetisierungszustandes. Dies macht den Einsatz externer Magnetfeldsensoren interessant, die sich über wohlbekannte Effekte kalibrieren ließen und dann im Messeinsatz quantitative Aussagen liefern können. Nun laufen viele der interessanten magnetischen Vorgänge in besagten Materialien auf sehr schnellen Zeitskalen im Piko- und Subpikosekundenbereich ab. Kein etablierter Magnetfeldsensor kann diese Anforderung leisten. Im Rahmen dieser Arbeit wurde eine systematische Untersuchung verschiedener Ansätze zum Bau ultraschneller Magnetfelddetektoren durchgeführt. Ein Teil der Arbeit beschäftigt sich mit dem Potential photokonduktiver Ringantennen als Emitter und Detektor für ultraschnelle Magnetfelder. Ein alternativer Ansatz zur Messung transienter Magnetfelder besteht in der Verwendung magnetooptischer Sensoren, wie sie in verschiedenen Anwendungen, in denen keine Zeitauflösung gefordert wird, bereits zum Einsatz kommen (z. B. in der Faradaymikroskopie). Es wird eine für ultraschnelle Magnetooptik vielversprechende Materialklasse als Sensormaterial vorgestellt: die DMS-Systeme. Das sind magnetisch dotierte Verbindungshalbleiter, die in der Umgebung ihrer exzitonischen Resonanzen gewaltige Verdetkonstanten aufweisen. Parallel zu den DMS-Systemen wird das Verhalten eines dotierten Eisengranats untersucht, der als Ferrimagnet völlig andere Voraussetzungen als Messsensor bietet. Darüber hinaus werden verschiedene experimentelle Techniken zur Messung magnetooptischer Phänomene vorgestellt und ihre Vor- und Nachteile ausführlich diskutiert. Es wird ein Verfahren entwickelt, das trotz des Einsatzes der hochempfindlichen Differenzdetektion eine gewisse spektrale Auflösung gewährleistet und deshalb den Betrieb der DMS-Systeme als magnetooptische Sensoren erst ermöglicht. Es werden für die verschiedenen Messmethoden und magnetooptischen Materialien die Grenzen der Nachweisempfindlichkeit analysiert und ihre Eignung als schnelle Detektoren untersucht. Die verschiedenen Vor- und Nachteile der beiden Sensorsubstanzen wird anhand der gemessenen magnetischen Transienten detailliert analysiert. Anschließend wird das Optimierungspotenzial der beiden Materialklassen hinsichtlich ihrer chemischen Zusammensetzung ausgearbeitet und dargestellt.
Early and adequate restoration of endothelial and tubular renal function is a substantial step during regeneration after ischemia reperfusion (IR) injury, occurring, e.g., in kidney transplantation, renal surgery, and sepsis. While tubular epithelial cell injury has long been of central importance, recent perception includes the renal vascular endothelium. In this regard, the fibrin cleavage product fibrinopeptide Bβ15-42 mitigate IR injury by stabilizing interendothelial junctions through its affinity to VE-cadherin. Therefore, this study focused on the effect of Bβ15-42 on post-acute physiological renal regeneration. For this, adult male C57BL/6 mice were exposed to a 30 min bilateral renal ischemia and reperfusion for 24 h or 48 h. Animals were randomized in a non-operative control group, two operative groups each treated with i.v. administration of either saline or Bβ15-42 (2.4 mg/kg) immediately prior to reperfusion. Endothelial activation and inflammatory response was attenuated in renal tissue homogenates by single application of Bβ15-42. Meanwhile, Bβ15-42 did not affect acute kidney injury markers. Regarding the angiogenetic players VEGF-A, Angiopoietin-1, Angiopoietin-2, however, we observed significant higher expressions at mRNA and trend to higher protein level in Bβ15-42 treated mice, compared to saline treated mice after 48 h of IR, thus pointing toward an increased angiogenetic activity. Similar dynamics were observed for the intermediate filament vimentin, the cytoprotective protein klotho, stathmin and the proliferation cellular nuclear antigen, which were significantly up-regulated at the same points in time. These results suggest a beneficial effect of anatomical contiguously located endothelial cells on tubular regeneration through stabilization of endothelial integrity. Therefore, it seems that Bβ15-42 represents a novel pharmacological approach in the targeted therapy of acute renal failure in everyday clinical practice.
5-Lipoxygenase (5-LO) is the key enzyme in the formation of pro-inflammatory leukotrienes (LT) which play an important role in a number of inflammatory diseases. Accordingly, 5-LO inhibitors are frequently used to study the role of 5-LO and LT in models of inflammation and cancer. Interestingly, the therapeutic efficacy of these inhibitors is highly variable. Here we show that the frequently used 5-LO inhibitors AA-861, BWA4C, C06, CJ-13,610 and the FDA approved compound zileuton as well as the pan-LO inhibitor nordihydroguaiaretic acid interfere with prostaglandin E2 (PGE2) release into the supernatants of cytokine-stimulated (TNFα/IL-1β) HeLa cervix carcinoma, A549 lung cancer as well as HCA-7 colon carcinoma cells with similar potencies compared to their LT inhibitory activities (IC50 values ranging from 0.1–9.1 µM). In addition, AA-861, BWA4C, CJ-13,610 and zileuton concentration-dependently inhibited bacterial lipopolysaccharide triggered prostaglandin (PG) release into human whole blood. Western Blot analysis revealed that inhibition of expression of enzymes involved in PG synthesis was not part of the underlying mechanism. Also, liberation of arachidonic acid which is the substrate for PG synthesis as well as PGH2 and PGE2 formation were not impaired by the compounds. However, accumulation of intracellular PGE2 was found in the inhibitor treated HeLa cells suggesting inhibition of PG export as major mechanism. Further, experiments showed that the PG exporter ATP-binding cassette transporter multidrug resistance protein 4 (MRP-4) is targeted by the inhibitors and may be involved in the 5-LO inhibitor-mediated PGE2 inhibition. In conclusion, the pharmacological effects of a number of 5-LO inhibitors are compound-specific and involve the potent inhibition of PGE2 export. Results from experimental models on the role of 5-LO in inflammation and pain using 5-LO inhibitors may be misleading and their use as pharmacological tools in experimental models has to be revisited. In addition, 5-LO inhibitors may serve as new scaffolds for the development of potent prostaglandin export inhibitors.
Background: Intensive Care Resources are heavily utilized during the COVID-19 pandemic. However, risk stratification and prediction of SARS-CoV-2 patient clinical outcomes upon ICU admission remain inadequate. This study aimed to develop a machine learning model, based on retrospective & prospective clinical data, to stratify patient risk and predict ICU survival and outcomes. Methods: A Germany-wide electronic registry was established to pseudonymously collect admission, therapeutic and discharge information of SARS-CoV-2 ICU patients retrospectively and prospectively. Machine learning approaches were evaluated for the accuracy and interpretability of predictions. The Explainable Boosting Machine approach was selected as the most suitable method. Individual, non-linear shape functions for predictive parameters and parameter interactions are reported. Results: 1039 patients were included in the Explainable Boosting Machine model, 596 patients retrospectively collected, and 443 patients prospectively collected. The model for prediction of general ICU outcome was shown to be more reliable to predict “survival”. Age, inflammatory and thrombotic activity, and severity of ARDS at ICU admission were shown to be predictive of ICU survival. Patients’ age, pulmonary dysfunction and transfer from an external institution were predictors for ECMO therapy. The interaction of patient age with D-dimer levels on admission and creatinine levels with SOFA score without GCS were predictors for renal replacement therapy. Conclusions: Using Explainable Boosting Machine analysis, we confirmed and weighed previously reported and identified novel predictors for outcome in critically ill COVID-19 patients. Using this strategy, predictive modeling of COVID-19 ICU patient outcomes can be performed overcoming the limitations of linear regression models. A central motivation for the development of x-ray free-electron lasers has been the prospect of time-resolved single-molecule imaging with atomic resolution. Here, we show that x-ray photoelectron diffraction—where a photoelectron emitted after x-ray absorption illuminates the molecular structure from within—can be used to image the increase of the internuclear distance during the x-ray-induced fragmentation of an O2 molecule. By measuring the molecular-frame photoelectron emission patterns for a two-photon sequential K-shell ionization in coincidence with the fragment ions, and by sorting the data as a function of the measured kinetic energy release, we can resolve the elongation of the molecular bond by approximately 1.2 a.u. within the duration of the x-ray pulse. The experiment paves the road toward time-resolved pump-probe photoelectron diffraction imaging at high-repetition-rate x-ray free-electron lasers.
