The last decade has seen a sharp increase in the number of scientific publications describing physiological and pathological functions of extracellular vesicles (EVs), a collective term covering various subtypes of cell-released, membranous structures, called exosomes, microvesicles, microparticles, ectosomes, oncosomes, apoptotic bodies, and many other names. However, specific issues arise when working with these entities, whose size and amount often make them difficult to obtain as relatively pure preparations, and to characterize properly. The International Society for Extracellular Vesicles (ISEV) proposed Minimal Information for Studies of Extracellular Vesicles (“MISEV”) guidelines for the field in 2014. We now update these “MISEV2014” guidelines based on evolution of the collective knowledge in the last four years. An important point to consider is that ascribing a specific function to EVs in general, or to subtypes of EVs, requires reporting of specific information beyond mere description of function in a crude, potentially contaminated, and heterogeneous preparation. For example, claims that exosomes are endowed with exquisite and specific activities remain difficult to support experimentally, given our still limited knowledge of their specific molecular machineries of biogenesis and release, as compared with other biophysically similar EVs. The MISEV2018 guidelines include tables and outlines of suggested protocols and steps to follow to document specific EV-associated functional activities. Finally, a checklist is provided with summaries of key points.
The membrane proximal external region (MPER) of the HIV-1 glycoprotein gp41 is targeted by the broadly neutralizing antibodies 2F5 and 4E10. To date, no immunization regimen in animals or humans has produced HIV-1 neutralizing MPER-specific antibodies. We immunized llamas with gp41-MPER proteoliposomes and selected a MPER-specific single chain antibody (VHH), 2H10, whose epitope overlaps with that of mAb 2F5. Bi-2H10, a bivalent form of 2H10, which displayed an approximately 20-fold increased affinity compared to the monovalent 2H10, neutralized various sensitive and resistant HIV-1 strains, as well as SHIV strains in TZM-bl cells. X-ray and NMR analyses combined with mutagenesis and modeling revealed that 2H10 recognizes its gp41 epitope in a helical conformation. Notably, tryptophan 100 at the tip of the long CDR3 is not required for gp41 interaction but essential for neutralization. Thus bi-2H10 is an anti-MPER antibody generated by immunization that requires hydrophobic CDR3 determinants in addition to epitope recognition for neutralization similar to the mode of neutralization employed by mAbs 2F5 and 4E10.
Non-standard errors

In statistics, samples are drawn from a population in a data-generating process (DGP). Standard errors measure the uncertainty in sample estimates of population parameters. In science, evidence is generated to test hypotheses in an evidence-generating process (EGP). We claim that EGP variation across researchers adds uncertainty: non-standard errors. To study them, we let 164 teams test six hypotheses on the same sample. We find that non-standard errors are sizeable, on par with standard errors. Their size (i) co-varies only weakly with team merits, reproducibility, or peer rating, (ii) declines significantly after peer-feedback, and (iii) is underestimated by participants.
The slow neutron capture process (s-process) is responsible for producing about half of the elemental abundances heavier than iron in the universe. Neutron capture cross sections on stable isotopes are a key nuclear physics input for s-process studies. The 72Ge(n, γ) cross section has an important influence on production of isotopes between Ge and Zr during s-process in massive stars and therefore experimental data are urgently required. 72Ge(n, γ) was measured at the neutron time-of-flight facility n_TOF (CERN) for the first time at stellar energies. The measurement was performed using an enriched 72GeO2 sample at a flight path of 185m with a set of liquid scintillation detectors (C6D6). The motivation, experiment and current status of the data analysis are reported.
Ziel des Beitrags ist, einen Überblick über aktuelle Entwicklungen und Ausprägungen von MOOCs sowie die Erfahrungen und Ergebnisse aus der Unter-suchung von zwei MOOCs vorzustellen, die 2011 und 2012 durchgeführt wurden. Besondere Schwerpunkte liegen dabei auf der Bedeutung und Ausprägung der Beteiligungsformen in den verschiedenen MOOC-Formaten, der Beteiligung in den beiden untersuchten MOOCs sowie auf deren Unterschiede auch aufgrund von Veränderungen des Kursdesigns.
Die Volkshochschulen sind der größte Bildungsanbieter in Deutschland. 2013 startete eine VHS-Initiative den vhsMOOC: eine offene Online-Veranstaltung zum Thema selbstbestimmtes Weblernen. Auf mehreren Plattformen wie Google+, Facebook und Twitter wurde gleichzeitig gepostet und kommentiert, auf YouTube entstanden 48 Videos zum vhsMOOC, in denen sich VHS-Mitarbeiterinnen und Mitarbeiter über Weblernen, neue Lehr- und Lernformate, unterstützende Tools und die Entwicklung der Volkshochschulen äußern. Der Band fasst den Verlauf, die Argumente und die Ergebnisse der achtwöchigen Online-Lehrveranstaltung zusammen. Passend zum offenen Format des vhsMOOC sind die Inhalte des Bandes auch auf wbv Open Access verfügbar.
The paper takes a deeper look at participation rates in cMOOCs. To get a better insight into the behavior of learners in MOOCs, studiumdigitale has developed a tool which helps to analyze the contribution of participants in the so called cMOOCs. These are MOOCs which are fostering the active participation of learners in various tools and which are based on the concept of connectivism [1]. After an approach at each part of the definition of MOOCs and the discussion of the different categories of this quite new phenomena a deeper look will be taken into the analysis of two cMOOCs, OPCO11 and OPCO12 which took place 2011 and 2012 [2].
Unter Web-based Trainings (WBTs) versteht man multimediale, interaktive und thematisch abgeschlossene Lerneinheiten in einem Browser. Seit der Entstehung des Internets in den 1990er Jahren sind diese ein wichtiger und etablierter Baustein bei der Konzeption und Entwicklung von eLearning-Szenarien. Diese Lerneinheiten werden üblicherweise von Lehrenden mit entsprechenden Autorensystemen erstellt. In selteneren Fällen handelt es sich bei deren Umsetzungen um individuell programmierte Einzellösungen. Betrachtet man WBTs aus der Sicht der Lernenden, dann lässt sich feststellen, dass zunehmend auch nicht explizit als Lerneinheiten erstellte Inhalte genutzt werden, die jedoch genau den Bedürfnissen des jeweiligen Lernenden entsprechen (im Rahmen des informellen und selbstgesteuerten Lernens). Zum einen liegt das an der zunehmenden Verfügbarkeit und Vielfalt von „alternativen Lerninhalten“ im Internet generell (freie Lizenzen und innovative Autorentools). Zum anderen aber auch an der Möglichkeit, diese Inhalte von überall aus und zu jeder Zeit einfach finden zu können (mobiles Internet, Suchmaschinen und Sprachassistenten) bzw. eingeordnet und empfohlen zu bekommen (Empfehlungssysteme und soziale Medien).
Aus dieser Veränderung heraus ergibt sich im Rahmen dieser Dissertation die zentrale Fragestellung, ob das Konzept eines dedizierten WBT-Autorensystems den neuen Anforderungen von frei verfügbaren, interaktiven Lerninhalten (Khan Academy, YouTube und Wikipedia) und einer Vielzahl ständig wachsender und kostenfreier Autorentools für beliebige Web-Inhalte (H5P, PowToon oder Pageflow) überhaupt noch gerecht wird und wo in diesem Fall genau die Alleinstellungsmerkmale eines WBTs liegen?
Zur Beantwortung dieser Frage beschäftigt sich die Arbeit grundlegend mit dem Begriff „Web-based Training“, den über die Zeit geänderten Rahmenbedingungen und den daraus resultierenden Implikationen für die Entwicklung von WBT-Autorensystemen. Mittels des gewählten Design-based Research (DBR)-Ansatzes konnte durch kontinuierliche Zyklen von Gestaltung, Durchführung, Analyse und Re-Design am Beispiel mehrerer eLearning-Projekte der Begriff WBT neudefininiert bzw. reinterpretiert werden, so dass sich der Fokus der Definition auf das konzentriert, was WBTs im Vergleich zu anderen Inhalten und Funktionen im Internet im Kern unterscheidet: dem Lehr-/Lernaspekt (nachfolgend Web-based Training 2.0 (WBT 2.0)).
Basierend auf dieser Neudefinition konnten vier Kernfunktionalitäten ausgearbeitet werden, die die zuvor genannten Herausforderungen adressieren und in Form eines Design Frameworks detailliert beschreiben. Untersucht und entwickelt wurden die unterschiedlichen Aspekte und Funktionen der WBTs 2.0 anhand der iterativen „Meso-Zyklen“ des DBR-Ansatzes, wobei jedes der darin durchgeführten Projekte auch eigene Ergebnisse mit sich bringt, welche jeweils unter didaktischen und vor allem aber technischen Gesichtspunkten erörtert wurden. Die dadurch gewonnenen Erkenntnisse flossen jeweils in den Entwicklungsprozess der LernBar ein („Makro-Zyklus“), ein im Rahmen dieser Arbeit und von studiumdigitale, der zentralen eLearning-Einrichtung der Goethe-Universität, entwickeltes WBT-Autorensystem. Dabei wurden die Entwicklungen kontinuierlich unter Einbezug von Nutzerfeedbacks (jährliche Anwendertreffen, Schulungen, Befragungen, Support) überprüft und weiterentwickelt.
Abschließend endet der letzte Entwicklungszyklus des DBR-Ansatzes mit der Konzeption und Umsetzung von drei WBT 2.0-Systemkomponenten, wodurch sich flexibel beliebige Web-Inhalte mit entsprechenden WBT 2.0-Funktionalitäten erweitern lassen, um auch im Kontext von offenen Lehr-/Lernprozessen durchgeführte Aktivitäten transparent, nachvollziehbar und somit überprüfbar zu machen (Constructive Alignment).
Somit bietet diese Forschungsarbeit einen interdisziplinären, nutzerzentrierten und in der Praxis erprobten Ansatz für die Umsetzung und den Einsatz von WBTs im Kontext offener Lehr-/Lernprozesse. Dabei verschiebt sich der bisherige Fokus von der reinen Medienproduktion hin zu einem ganzheitlichen Ansatz, bei dem der Lehr-/Lernaspekt im Vordergrund steht (Lernbedarf erkennen, decken und überprüfen). Entscheidend ist dabei, dass zum Decken eines Lernbedarfs sämtliche zur Verfügung stehenden Ressourcen des Internets genutzt werden können, wobei WBTs 2.0 dazu lediglich den didaktischen Prozess definieren und diesen für die Lehrenden und Lernende transparent und zugänglich machen.
WBTs 2.0 profitieren dadurch zukünftig von der zunehmenden Vielfalt und Verfügbarkeit von Inhalten und Funktionen im Internet und ermöglichen es, den Entwicklern von WBT 2.0-Autorensystemen sich auf das Wesentliche zu konzentrieren: den Lehr-/Lernprozess.
Purpose: The coronavirus disease 2019 (COVID-19) poses major challenges to health-care systems worldwide. This pandemic demonstrates the importance of timely access to intensive care and, therefore, this study aims to explore the accessibility of intensive care beds in 14 European countries and its impact on the COVID-19 case fatality ratio (CFR).
Methods: We examined access to intensive care beds by deriving (1) a regional ratio of intensive care beds to 100,000 population capita (accessibility index, AI) and (2) the distance to the closest intensive care unit. The cross-sectional analysis was performed at a 5-by-5 km spatial resolution and results were summarized nationally for 14 European countries. The relationship between AI and CFR was analyzed at the regional level.
Results: We found national-level differences in the levels of access to intensive care beds. The AI was highest in Germany (AI = 35.3), followed by Estonia (AI = 33.5) and Austria (AI = 26.4), and lowest in Sweden (AI = 5) and Denmark (AI = 6.4). The average travel distance to the closest hospital was highest in Croatia (25.3 min by car) and lowest in Luxembourg (9.1 min). Subnational results illustrate that capacity was associated with population density and national-level inventories. The correlation analysis revealed a negative correlation of ICU accessibility and COVID-19 CFR (r = − 0.57; p < 0.001).
Conclusion: Geographical access to intensive care beds varies significantly across European countries and low ICU accessibility was associated with a higher proportion of COVID-19 deaths to cases (CFR). Important differences in access are due to the sizes of national resource inventories and the distribution of health-care facilities relative to the human population. Our findings provide a resource for officials planning public health responses beyond the current COVID-19 pandemic, such as identifying potential locations suitable for temporary facilities or establishing logistical plans for moving severely ill patients to facilities with available beds.
The radiative capture cross section of 238U is very important for the developing of new reactor technologies and the safety of existing ones. Here the preliminary results of the 238U(n,γ) cross section measurement performed at n_TOF with C6D6 scintillation detectors are presented, paying particular attention to data reduction and background subtraction.
73Ge(n, γ ) cross sections were measured at the neutron time-of-flight facility n_TOF at CERN up to neutron energies of 300 keV, providing for the first time experimental data above 8 keV. Results indicate that the stellar cross section at kT = 30 keV is 1.5 to 1.7 times higher than most theoretical predictions. The new cross sections result in a substantial decrease of 73Ge produced in stars, which would explain the low isotopic abundance of 73Ge in the solar system.
We have measured the radiative neutron-capture cross section and the total neutron-induced cross section of one of the most important isotopes for the s process, the 25Mg. The measurements have been carried out at the neutron time-of-flight facilities n_TOF at CERN (Switzerland) and GELINA installed at the EC-JRC-IRMM (Belgium). The cross sections as a function of neutron energy have been measured up to approximately 300 keV, covering the energy region of interest to the s process. The data analysis is ongoing and preliminary results show the potential relevance for the s process.
Above 1 MeV of incident neutron energy the fission fragment angular distribution (FFAD) has generally a strong anisotropic behavior due to the combination of the incident orbital momentum and the intrinsic spin of the fissioning nucleus. This effect has to be taken into account for the efficiency estimation of devices used for fission cross section measurements. In addition it bears information on the spin deposition mechanism and on the structure of transitional states. We designed and constructed a detection device, based on Parallel Plate Avalanche Counters (PPAC), for measuring the fission fragment angular distributions of several isotopes, in particular 232Th. The measurement has been performed at n_TOF at CERN taking advantage of the very broad energy spectrum of the neutron beam. Fission events were recognized by back to back detection in coincidence in two position-sensitive detectors surrounding the targets. The detection efficiency, depending mostly on the stopping of fission fragments in backings and electrodes, has been computed with a Geant4 simulation and validated by the comparison to the measured case of 235U below 3 keV where the emission is isotropic. In the case of 232Th, the result is in good agreement with previous data below 10 MeV, with a good reproduction of the structures associated to vibrational states and the opening of second chance fission. In the 14 MeV region our data are much more accurate than previous ones which are broadly scattered.

Objective: TGF-β2 (TGF-β, transforming growth factor beta), the less-investigated sibling of TGF-β1, is deregulated in rodent and human liver diseases. Former data from bile duct ligated and MDR2 knockout (KO) mouse models for human cholestatic liver disease suggested an involvement of TGF-β2 in biliary-derived liver diseases.
Design: As we also found upregulated TGFB2 in liver tissue of patients with primary sclerosing cholangitis (PSC) and primary biliary cholangitis (PBC), we now fathomed the positive prospects of targeting TGF-β2 in early stage biliary liver disease using the MDR2-KO mice. Specifically, the influence of TgfB2 silencing on the fibrotic and inflammatory niche was analysed on molecular, cellular and tissue levels.
Results: TgfB2-induced expression of fibrotic genes in cholangiocytes and hepatic stellate cellswas detected. TgfB2 expression in MDR2-KO mice was blunted using TgfB2-directed antisense oligonucleotides (AON). Upon AON treatment, reduced collagen deposition, hydroxyproline content and αSMA expression as well as induced PparG expression reflected a significant reduction of fibrogenesis without adverse effects on healthy livers. Expression analyses of fibrotic and inflammatory genes revealed AON-specific regulatory effects on Ccl3, Ccl4, Ccl5, Mki67 and Notch3 expression. Further, AON treatment of MDR2-KO mice increased tissue infiltration by F4/80-positive cells including eosinophils, whereas the number of CD45-positive inflammatory cells decreased. In line, TGFB2 and CD45 expression correlated positively in PSC/PBC patients and localised in similar areas of the diseased liver tissue.
Conclusions: Taken together, our data suggest a new mechanistic explanation for amelioration of fibrogenesis by TGF-β2 silencing and provide a direct rationale for TGF-β2-directed drug development.
Accurate measurement of the standard 235U(n,f) cross section from thermal to 170 keV neutron energy

An accurate measurement of the 235U(n,f) cross section from thermal to 170 keV of neutron energy has recently been performed at n_TOF facility at CERN using 6Li(n,t)4He and 10B(n,α)7Li as references. This measurement has been carried out in order to investigate a possible overestimation of the 235U fission cross section evaluation provided by most recent libraries between 10 and 30 keV. A custom experimental apparatus based on in-beam silicon detectors has been used, and a Monte Carlo simulation in GEANT4 has been employed to characterize the setup and calculate detectors efficiency. The results evidenced the presence of an overestimation in the interval between 9 and 18 keV and the new data may be used to decrease the uncertainty of 235U(n,f) cross section in the keV region.
233U is the fissile nuclei in the Th-U fuel cycle with a particularily small neutron capture cross setion which is on average about one order of magnitude lower than its fission cross section. Hence, the measurement of the 233U(n, γ) cross section relies on a method to accurately distinguish between capture and fission γ-rays. A measurement of the 233U α-ratio has been performed at the n_TOF facility at CERN using a so-called fission tagging setup, coupling n_TOF 's Total Absorption Calorimeter with a novel fission chamber to tag the fission γ-rays. The experimental setup is described and essential parts of the analysis are discussed. Finally, a preliminary 233U α-ratio is presented.
We have measured the capture cross section of the 155Gd and 157Gd isotopes between 0.025 eV and 1 keV. The capture events were recorded by an array of 4 C6D6 detectors, and the capture yield was deduced exploiting the total energy detection system in combination with the Pulse Height Weighting Techniques. Because of the large cross section around thermal neutron energy, 4 metallic samples of different thickness were used to prevent problems related to self-shielding. The samples were isotopically enriched, with a cross contamination of the other isotope of less than 1.14%. The capture yield was analyzed with an R-Matrix code to describe the cross section in terms of resonance parameters. Near thermal energies, the results are significantly different from evaluations and from previous time-of-flight experiments. The data from the present measurement at n_TOF are publicly available in the experimental nuclear reaction database EXFOR.
