Progranulin deficiency in humans is associated with neurodegeneration. Its mechanisms are not yet fully understood. We performed a Yeast-2-Hybrid screen using human full-length progranulin as bait to assess the interactions of progranulin. Progranulin was screened against human fetal brain and human bone marrow libraries using the standard Matchmaker technology (Clontech). This article contains the full Y2H data table, including blast results and sequences, a sorted table according to selection criteria for likely positive, putatively positive, likely false and false preys, and tables showing the gene ontology terms associated with the likely and putative preys of the brain and bone marrow libraries. The interactions with autophagy proteins were confirmed and functionally analyzed in "Progranulin overexpression in sensory neurons attenuates neuropathic pain in mice: Role of autophagy" (C. Background: Cell salvage is commonly used as part of a blood conservation strategy. However concerns among clinicians exist about the efficacy of transfusion of washed cell salvage.
Methods: We performed a meta-analysis of randomized controlled trials in which patients, scheduled for all types of surgery, were randomized to washed cell salvage or to a control group with no cell salvage. Data were independently extracted, risk ratio (RR), and weighted mean differences (WMD) with 95% confidence intervals (CIs) were calculated. Data were pooled using a random effects model. The primary endpoint was the number of patients exposed to allogeneic red blood cell (RBC) transfusion.
Results: Out of 1140 search results, a total of 47 trials were included. Overall, the use of washed cell salvage reduced the rate of exposure to allogeneic RBC transfusion by a relative 39% (RR = 0.61; 95% CI 0.57 to 0.65; P < 0.001), resulting in an average saving of 0.20 units of allogeneic RBC per patient (weighted mean differences [WMD] = -0.20; 95% CI -0.22 to -0.18; P < 0.001), reduced risk of infection by 28% (RR = 0.72; 95% CI 0.54 to 0.97; P = 0.03), reduced length of hospital stay by 2.31 days (WMD = -2.31; 95% CI -2.50 to -2.11; P < 0.001), but did not significantly affect risk of mortality (RR = 0.92; 95% CI 0.63 to 1.34; P = 0.66). No statistical difference could be observed in the number of patients exposed to re-operation, plasma, platelets, or rate of myocardial infarction and stroke.
Conclusions: Washed cell salvage is efficacious in reducing the need for allogeneic RBC transfusion and risk of infection in surgery.

Vergleich der Veränderung der mütterlichen Einstellung zur Geburt anhand von Hebammen geführten Geburtsvorbereitungskursen oder hypnoreflexogenem Training zur Geburtsvorbereitung.
Methode:
Zu Beginn und nach Beendigung der Kurse wurde die mütterliche Einstellung zur Geburt unter Zuhilfenahme des Osgood-Ertel-Eindrucksdifferenzials gemessen. Der Gießen-Test zur Persönlichkeitsbeurteilung wurde einmalig angewendet.
Ergebnisse:
213 Frauen waren in die Studie eingeschlossen. 155 davon nahmen an, von Hebammen geführten, Geburtsvorbereitungskursen teil. 58 Frauen absolvierten ein hypnoreflexogenes Training. Es waren zu Beginn der Kurse keine statistisch signifikanten Unterschiede feststellbar in Bezug auf die Charakteristiken der Teilnehmerinnen sowie im Gießen-Test und in den Ergebnissen des Osgood-Ertel-Eindrucksdifferenzials. Nach der von Hebammen geführten Geburtsvorbereitung wurde die Geburt negativer wahrgenommen(Freudlosigkeit und Trübung in der Valenz-Dimension [p < 0,05]), während die Geburt nach dem Hypnosetraining emotional positiver bewertet wurde (Freude
und Harmonie in der Valenz-Dimension [p < 0,01] sowie Helligkeit [p < 0,05]).
Zusammenfassung:
Hypnoreflexogenes Selbsthypnosetraining zur Geburtsvorbereitung scheint stärkere und positivere mütterliche emotionale Veränderungen in Bezug auf die
Einstellung zur Geburt auszulösen als konventionelle, von Hebammen geführte Geburtsvorbereitungskurse. Weitere retrospektive randomisierte Studien sind nötig, um diese Ergebnisse zu überprüfen.
Alle lebenden Organismen sind in der Lage, sich an den re-gelmäßigen Wechsel von Licht und Dunkelheit und den zeitli-che Veränderungen im Takt der Jahreszeiten anzupassen. Die-se Synchronisierung der Aktivitäts- und Ruhephasen, sowie von physiologischen Stoffwechselprozessen an die vorgegebe-nen tageszeitlichen und saisonalen Zyklen findet beim Säu-getier in der inneren Uhr im Nucleus Suprachiasmaticus (SCN) statt. Das Licht, als wichtigster Zeitgeber für die Synchronisation der inneren Uhr, findet Eingang zum SCN über die Retina und den retinohypothalamischen Trakt (RTH), der Glutamat als Neurotransmitter nutzt. Ist dieses System fehlerhaft, führt dies zu Störung der oben beschriebenen Anpassungsprozesse. Dies hat eine gestörte Homöostase des Organismus zu Folge, aus denen sich wiederum Veränderungen im Tag/Nacht- Rhythmus, Schlafstörungen und depressive Ver-stimmungen ergeben können. Die genannten Symptome decken sich mit den Frühsymptomen den neurodegenerativen Erkran-kung Morbus Parkinson.
Ziel der vorliegenden Arbeit war es, Störungen im photoneu-roendokrinen System, insbesondere Veränderungen in der Re-tina an den photosensitiven Ganglienzellen mit dem Photo-pigment Melanopsin und dem SCN bei transgene Mäuse mit dem humanen alpha-Synuclein zu untersuchen. Hierbei wurden transgene Mäuse mit dem gesunden humanen alpha-Synuclein (Wildtyp) und transgene Mäuse mit der für Parkinson spezi-fischen Mutation im alpha-Synuclein Ala53Thr (A53T) vergli-chen.
Die immunochemischen Untersuchungen an Retina und SCN zei-gen einen signifikanten Anstieg der alpha-Synuclein Immun-reaktion bei der A53T Maus im Vergleich zum Wildtyp.
Parallel dazu wurden Unterschiede in Bezug auf das Photo-pigment Melanopsin zwischen den beiden Gruppen untersucht. Melanopsin ist lichtsensitiv und trägt, durch Übermittlung der aktuellen Lichtverhältnisse über den retinohypothalami-schen Trakt zum SCN, zur Synchronisation der circadianen Rhythmik bei. Durch den in dieser Arbeit nachgewiesene Me-lanopsindefizit und des deutlich reduzierten Vglut2 im hy-pothalamischen Trakt der A53T Maus lässt sich die Hypothese ableiten, dass möglicherweise die Überexpression des mu-tierten alpha-Synuclein in der Retina einen Untergang von melanopsinhaltigen Ganglienzellen herbeiführt und dadurch die Synchronisation der inneren Uhr durch Licht gestört ist. Diese Hypothese wird durch die Aktivitätsprofile ge-stützt, die durch die Aufzeichnung der lokomotorischen Ak-tivität der Tiere erstellt wurden.
Da in beiden Gruppen unter Dauerdunkel (DD) ein endogener zirkadianer Rhythmus beobachtet werden konnte, lässt dies auf die Funktionstüchtigkeit der inneren Uhr im SCN schlie-ßen. Im anschließenden Versuch die endogene Rhythmik an exogenen Reize anzupassen, zeigte sich bei dem A53T Stamm eine fehlende Synchronisierung an vorgegebene Lichtverhält-nisse mit gesteigerter Tagaktivität und reduzierten Schlaf-phasen. Somit trägt der fehlerhaft verarbeitete Lichtreiz bei A53T Mutanten zur Destabilisierung des zirkadianen Rhythmus der Lokomotion bei. Trotz des gestörten glutama-tergen Signalweges im retinohypothalamischen Trakt konnten keine Unterschiede in der Expression der Homerproteine zwi-schen Wildtyp und A53T unter Standard-Photoperiode und nach Schlafdeprivation nachgewiesen werden.
Die vorliegenden Befunde liefern Erkenntnisse zur Entste-hung der Frühsymptome bei Morbus Parkinson. Dies könnte neue Ansatzpunkte für die Therapie und Linderung von Schlafstörungen sowie Veränderungen im Tag/Nachtrhythmus liefern.
The mitotic spindle assembly checkpoint (SAC) maintains genome stability and marks an important target for antineoplastic therapies. However, it has remained unclear how cells execute cell fate decisions under conditions of SAC‐induced mitotic arrest. Here, we identify USP9X as the mitotic deubiquitinase of the X‐linked inhibitor of apoptosis protein (XIAP) and demonstrate that deubiquitylation and stabilization of XIAP by USP9X lead to increased resistance toward mitotic spindle poisons. We find that primary human aggressive B‐cell lymphoma samples exhibit high USP9X expression that correlate with XIAP overexpression. We show that high USP9X/XIAP expression is associated with shorter event‐free survival in patients treated with spindle poison‐containing chemotherapy. Accordingly, aggressive B‐cell lymphoma lines with USP9X and associated XIAP overexpression exhibit increased chemoresistance, reversed by specific inhibition of either USP9X or XIAP. Moreover, knockdown of USP9X or XIAP significantly delays lymphoma development and increases sensitivity to spindle poisons in a murine Eμ‐Myc lymphoma model. Together, we specify the USP9X–XIAP axis as a regulator of the mitotic cell fate decision and propose that USP9X and XIAP are potential prognostic biomarkers and therapeutic targets in aggressive B‐cell lymphoma.
Abstract:
Snakebite is an important medical emergency in rural Nepal. Correct identification of the biting species is crucial for clinicians to choose appropriate treatment and anticipate complications. This is particularly important for neurotoxic envenoming which, depending on the snake species involved, may not respond to available antivenoms. Adequate species identification tools are lacking. This study used a combination of morphological and molecular approaches (PCR-aided DNA sequencing from swabs of bite sites) to determine the contribution of venomous and non-venomous species to the snakebite burden in southern Nepal. Out of 749 patients admitted with a history of snakebite to one of three study centres, the biting species could be identified in 194 (25.9%). Out of these, 87 had been bitten by a venomous snake, most commonly the Indian spectacled cobra (Naja naja; n = 42) and the common krait (Bungarus caeruleus; n = 22). When both morphological identification and PCR/sequencing results were available, a 100% agreement was noted. The probability of a positive PCR result was significantly lower among patients who had used inadequate “first aid” measures (e.g. tourniquets or local application of remedies). This study is the first to report the use of forensic genetics methods for snake species identification in a prospective clinical study. If high diagnostic accuracy is confirmed in larger cohorts, this method will be a very useful reference diagnostic tool for epidemiological investigations and clinical studies.

Snakebite can result in life-threatening envenoming, and correct identification of the biting species is crucial for care providers to choose appropriate treatment and anticipate complications. This paper explores a number of methods, including molecular techniques, to assist care providers in identifying the species responsible for bites in rural Nepal. Out of 749 patients with a history of snakebite, the biting species could be identified in 194 (25.9%). Out of these, 87 had been bitten by a venomous snake, most commonly cobras (n = 42) and kraits (n = 22). This study is the first to report the use of molecular techniques for snake species identification. The diagnostic accuracy of this method appears high but needs to be confirmed in larger studies.
Ubiquitination plays a critical role in the activation of host immune responses to infection and serves as a signal for pathogen delivery to phagophores along the xenophagy pathway. We recently performed systematic ubiquitination site profiling of epithelial cells infected with Salmonella Typhimurium. Our findings specifically highlight components of the NFKB, membrane trafficking pathways and RHO GTPase systems as ubiquitination hubs during infection. In addition, a broad spectrum of bacterial effectors and several outer membrane proteins are ubiquitinated in infected cells. This comprehensive resource of ubiquitinome dynamics during Salmonella infection enables further understanding of the complex host-pathogen interplay and may reveal novel targets for the inhibition of Salmonella invasion and inflammation.
Infant acute leukemia still has a poor prognosis, and allogeneic hematopoietic stem cell transplantation is indicated in selected patients. Umbilical cord blood (UCB) is an attractive cell source for this population because of the low risk of chronic graft-versus-host disease (GVHD), the strong graft-versus-leukemia effect, and prompt donor availability. This retrospective, registry-based study reported UCB transplantation (UCBT) outcomes in 252 children with acute lymphoblastic leukemia (ALL; n = 157) or acute myelogenous leukemia (AML; n = 95) diagnosed before 1 year of age who received a single-unit UCBT after myeloablative conditioning between 1996 and 2012 in European Society for Blood and Marrow Transplantation centers. Median age at UCBT was 1.1 years, and median follow-up was 42 months. Most patients (57%) received a graft with 1 HLA disparity and were transplanted in first complete remission (CR; 55%). Cumulative incidence function (CIF) of day 100 acute GVHD (grades II to IV) was 40% ± 3% and of 4-year chronic GVHD was 13% ± 2%. CIF of 1-year transplant-related mortality was 23% ± 3% and of 4-year relapse was 27% ± 3%. Leukemia-free-survival (LFS) at 4 years was 50% ± 3%; it was 40% and 66% for those transplanted for ALL and AML, respectively (P = .001). LFS was better for patients transplanted in first CR, regardless of diagnosis. In multivariate model, diagnosis of ALL (P = .001), advanced disease status at UCBT (<.001), age at diagnosis younger than 3 months (P = .012), and date of transplant before 2004 were independently associated with worse LFS. UCBT is a suitable option for patients diagnosed with infant acute leukemia who achieve CR. In this cohort, patients with AML had better survival than those with ALL.
