The full-length translation-regulating add adenine riboswitch (Asw) from Vibrio vulnificus has a more complex conformational space than its isolated aptamer domain. In addition to the predicted apo (apoA) and holo conformation that feature the conserved three-way junctional purine riboswitch aptamer, it adopts a second apo (apoB) conformation with a fundamentally different secondary structure. Here, we characterized the ligand-dependent conformational dynamics of the full-length add Asw by NMR and by single-molecule FRET (smFRET) spectroscopy. Both methods revealed an adenine-induced secondary structure switch from the apoB-form to the apoA-form that involves no tertiary structural interactions between aptamer and expression platform. This strongly suggests that the add Asw triggers translation by capturing the apoA-form secondary structure in the holo state. Intriguingly, NMR indicated a homogenous, docked aptamer kissing loop fold for apoA and holo, while smFRET showed persistent aptamer kissing loop docking dynamics between comparably stable, undocked and docked substates of the apoA and the holo conformation. Unraveling the folding of large junctional riboswitches thus requires the integration of complementary solution structural techniques such as NMR and smFRET.
Viewing of ambiguous stimuli can lead to bistable perception alternating between the possible percepts. During continuous presentation of ambiguous stimuli, percept changes occur as single events, whereas during intermittent presentation of ambiguous stimuli, percept changes occur at more or less regular intervals either as single events or bursts. Response patterns can be highly variable and have been reported to show systematic differences between patients with schizophrenia and healthy controls. Existing models of bistable perception often use detailed assumptions and large parameter sets which make parameter estimation challenging. Here we propose a parsimonious stochastic model that provides a link between empirical data analysis of the observed response patterns and detailed models of underlying neuronal processes. Firstly, we use a Hidden Markov Model (HMM) for the times between percept changes, which assumes one single state in continuous presentation and a stable and an unstable state in intermittent presentation. The HMM captures the observed differences between patients with schizophrenia and healthy controls, but remains descriptive. Therefore, we secondly propose a hierarchical Brownian model (HBM), which produces similar response patterns but also provides a relation to potential underlying mechanisms. The main idea is that neuronal activity is described as an activity difference between two competing neuronal populations reflected in Brownian motions with drift. This differential activity generates switching between the two conflicting percepts and between stable and unstable states with similar mechanisms on different neuronal levels. With only a small number of parameters, the HBM can be fitted closely to a high variety of response patterns and captures group differences between healthy controls and patients with schizophrenia. At the same time, it provides a link to mechanistic models of bistable perception, linking the group differences to potential underlying mechanisms.
Steilhänge der Mittelgebirge weisen eine kleinräumige standörtliche Vielfalt auf und wurden aufgrund ihrer exponierten Lage bereits frühzeitig durch den Menschen u. a. als Befestigungsanlage genutzt. Daraus resultierte häufig eine hohe floristische und vegetationskundliche Diversität mit hohem Naturschutzwert. Durch Umwelt- und Nutzungsveränderungen hat sich aber auch an diesen Standorten in den letzten Jahrzehnten ein starker Wandel vollzogen. Durch Auswertung alter Florenlisten und Vegetationsaufnahmen und durch aktuelle Erhebungen wollen wir die Vegetationszusammensetzung der Lengder Burg, eines Steilhangs auf Unterem Muschelkalk im südlichen Göttinger Wald (Süd-Niedersachsen, Deutschland), und ihre Veränderung aufzeigen und Rückschlüsse für die zukünftige Behandlung ziehen. Dazu wurden Angaben zur Gefäßpflanzenflora aus dem Zeitraum 1950 bis 1995 mit einer aktuellen floristischen Kartierung des Gesamtgebiets von 2016 verglichen. Die Vegetationszusammensetzung wurde anhand von 37 Vegetationsaufnahmen aus dem früheren Zeitraum harakterisiert. 29 dieser Flächen wurden 2009 bis 2016 erneut aufgenommen. Räumliche und zeitliche Unterschiede in der Diversität und Artenzusammensetzung wurden hinsichtlich verschiedener standörtlicher Parameter und ökologischer Artengruppen analysiert.
Die Vegetation lässt sich zwei Gruppen zuordnen: 1. Das Carici-Fagetum und seine Kontaktgesellschaften an südlich und westlich exponierten Steilhängen. 2. Das Hordelymo-Fagetum mit verschiedenen Ausbildungen auf dem Plateau und an flacheren Süd- und Nordhangbereichen. Im Carici-Fagetum ist ein deutlicher Diversitätsverlust und eine Zunahme in der Dominanzstruktur der Krautschicht zu erkennen, der im Vergleich der Aufnahmezeiträume auf eine zunehmende Homogenisierung der Vegetation hinweist. Zurückgegangen sind dabei besonders die typischen Kenn- und Trennarten dieser Waldgesellschaft bei gleichzeitiger Zunahme der Buche in der Verjüngung. Im Hordelymo-Fagetum bewirkt neben Gehölzen in der Strauch- und Krautschicht vor allem die Zunahme von Allium ursinum eine homogenere Artenzusammensetzung, jedoch ohne Diversitätsverlust. Neben Stickstoffeinträgen, dem Klimawandel sowie einem reduzierten Rehwild-Verbiss bedingt besonders der Nutzungswandel diese Veränderungen. Vor allem im Carici-Fagetum wirkte sich der Übergang zwischen früherer Nieder- und Mittelwald-Nutzung mit Waldweide über eine fast 100jährige Hochwald-Nutzung bis zum jetzigen Schutzwald stark aus. Gleichwohl weisen die steilen Hänge weiterhin einen hohen Anteil an Rote-Liste-Arten auf und tragen wesentlich zur hohen Biodiversität des Gebiets bei. Veränderungen in der Vegetation der Lengder Burg spiegeln die Veränderungen im Göttinger Wald insgesamt wider. Kleinflächige Offenhaltungsmaßnahmen zur Erhaltung wertvoller floristischer Elemente sind teilweise erfolgreich. Kleinwüchsige, lichtbedürftige Magerkeitszeiger verschwinden jedoch zunehmend aus den sich entwickelnden, hochwüchsigen Stauden-Säumen. In den benachbarten, unbewirtschafteten Hangbuchenwäldern sorgt die fehlende Nutzung nach Jahrhunderten der Auflichtung und Aushagerung für eine Sukzession in Richtung mesophilen Kalkbuchenwalds.
Background: Although mortality after cardiac surgery has significantly decreased in the last decade, patients still experience clinically relevant postoperative complications. Among others, atrial fibrillation (AF) is a common consequence of cardiac surgery, which is associated with prolonged hospitalization and increased mortality.
Methods: We retrospectively analyzed data from patients who underwent coronary artery bypass grafting, valve surgery or a combination of both at the University Hospital Muenster between April 2014 and July 2015. We evaluated the incidence of new onset and intermittent/permanent AF (patients with pre- and postoperative AF). Furthermore, we investigated the impact of postoperative AF on clinical outcomes and evaluated potential risk factors.
Results: In total, 999 patients were included in the analysis. New onset AF occurred in 24.9% of the patients and the incidence of intermittent/permanent AF was 59.5%. Both types of postoperative AF were associated with prolonged ICU length of stay (median increase approx. 2 days) and duration of mechanical ventilation (median increase 1 h). Additionally, new onset AF patients had a higher rate of dialysis and hospital mortality and more positive fluid balance on the day of surgery and postoperative days 1 and 2. In a multiple logistic regression model, advanced age (odds ratio (OR) = 1.448 per decade increase, p < 0.0001), a combination of CABG and valve surgery (OR = 1.711, p = 0.047), higher C-reactive protein (OR = 1.06 per unit increase, p < 0.0001) and creatinine plasma concentration (OR = 1.287 per unit increase, p = 0.032) significantly predicted new onset AF. Higher Horowitz index values were associated with a reduced risk (OR = 0.996 per unit increase, p = 0.012). In a separate model, higher plasma creatinine concentration (OR = 2.125 per unit increase, p = 0.022) was a significant risk factor for intermittent/permanent AF whereas higher plasma phosphate concentration (OR = 0.522 per unit increase, p = 0.003) indicated reduced occurrence of this arrhythmia.
Conclusions: New onset and intermittent/permanent AF are associated with adverse clinical outcomes of elective cardiac surgery patients. Different risk factors implicated in postoperative AF suggest different mechanisms might be involved in its pathogenesis. Customized clinical management protocols seem to be warranted for a higher success rate of prevention and treatment of postoperative AF.
Wer durch eine Stadt wie Venedig geht, sieht, dass die Zeit überall ihre Spuren hinterlässt. Der britische Kunsthistoriker John Ruskin nennt diese Spuren in seinem 1851 bis 1853 publizierten Reisebericht 'The Stones of Venice' "time stains" und macht darin eindrücklich darauf aufmerksam, dass Spuren der Zeit ästhetisch betrachtet werden können, obwohl sie kein Produkt des Menschen, sondern zunächst einmal eines der Zeit sind. Vor diesem Hintergrund drängt sich die Frage auf, ob und inwiefern die Zeit als Gestalterin ästhetischer Objekte begriffen werden kann und wie sie dieses Potenzial in Konkurrenz zu kulturellen Gestaltungsabsichten erfüllt.
Zur Beantwortung dieser Frage lassen sich die im Kontext der Ruinenästhetik geführten Diskussionen und das seit einigen Jahren zu verzeichnende neue Interesse am Naturschönen folgendermaßen verbinden und zuspitzen: Die Zeit muss als gegenkulturelle Gestalterin der Lebenswelt genauer ins Visier und als ästhetisches Subjekt ernst genommen werden. Der Beitrag reiht sich damit in die seit einigen Jahren Konjunktur verzeichnende ästhetische Auseinandersetzung mit der Zeit ein. Dort ist bislang versäumt worden, dezidiert nach der ästhetischen Gestaltungskraft von Zeit zu fragen, während man die kulturelle Gestaltbarkeit von Zeit betont hat.
Ich spreche im Folgenden über ein Thema, das 'Hermeneutik nach Luther' heißen soll. Als Hermeneutik verstehe ich dabei im Anschluss an Friedrich Schleiermacher - also im Anschluss an einen protestantischen Theologen, der seine eigene Hermeneutikkonzeption vorwiegend mit Bezug auf die Auslegung des Neuen Testamentes entwickelt hat, das heißt in einem dezidiert christlichen und zugleich mehrfachen, noch näher zu klärenden nach-Luther'schen Sinn - die "Kunst des Verstehens". Die Bestimmung verdeutlicht, dass das Verstehen nichts Selbstverständliches ist. Verstehen versteht sich nicht von selbst. Es muss selbst verstanden werden. Hermeneutik bezeichnet nach diesem Verständnis eine Aufgabe, und zwar, wie Schleiermacher zu betonen nicht müde wird, eine niemals abgeschlossene, immer weiter fortzusetzende Aufgabe.
Beim Gebrauch einer Fremdsprache durch Anfänger handelt es sich offensichtlich um "Spielarten interkultureller Kommunikation" (FÖLDES 2007: 614). Eine bezüglich der arabischen Auslandsgermanistik ergiebige Auseinandersetzung mit der Schreibfertigkeit gelingt erst, wenn interkulturelle und handlungsorientierte Aspekte zusammen und sprachkomparatistisch berücksichtigt werden. Davon ausgehend lässt sich dann erklären, inwieweit mögliche differente Verhaltens- und Handlungsmuster in der deutschen und der arabischen Sprach- bzw. Kulturgemeinschaft das Schreiben in der deutschen Sprache beeinflussen. Neben Faktoren im sprachsystematischen und lexikalisch-semantischen Bereich lässt sich dies im pragmatischen Bereich an zwei Parametern des kommunikativen Verhaltenstyps (Grad der Expressivität sowie der der Ritualisierung) untersuchen. Mögliche differente Verhaltens- bzw. Handlungsweisen werden hier anhand von arabischen Germanistikstudierenden geschriebener Aufsätze bzw. E-Mails diskutiert.
In diesem Beitrag geht es zunächst um den aktuellen Stand der Forschung über das Verhältnis von Sprache und Emotionen mit besonderer Berücksichtigung der Untersuchungen zum Deutschen und Tschechischen. Anschließend werden einige Teilaspekte des Themas behandelt: die Unterscheidung zwischen emotionsausdrückender und emotionsbezeichnender Lexik, die Rolle der bildlichen Sprache sowie das Verhältnis vom Sprachübergreifenden und Einzelsprachlichen bei der Verbalisierung von Emotionen. Abschließend wird der Frage nachgegangen, welche Konsequenzen die im Rahmen eines Projektes ermittelten Gemeinsamkeiten, Ähnlichkeiten und Unterschiede bei der Versprachlichung von Emotionen im Deutschen und Tschechischen für die Behandlung emotionsrelevanter Lexik im DaF-Unterricht bei tschechischen Muttersprachlern haben können.
Der wissenschaftliche Artikel ist eine Textsorte, die weltweit und zahlreich von der scientific community produziert und rezipiert wird. Doch obwohl es sich um eine international bekannte und häufig verwendete Textsorte handelt, zeichnen sich wissenschaftliche Artikel je nach Herkunft und Sprache durch zahlreiche Unterschiede aus. In diesem Beitrag folgt auf eine kurze Darstellung von Unterschieden deutscher und tschechischer wissenschaftlicher Artikel eine kontrastive Betrachtung der Fußnoten innerhalb dieser Texte. Interessant ist diesbezüglich nicht nur, welche unterschiedlichen Typen von Fußnoten in deutschen und tschechischen wissenschaftlichen Artikeln Verwendung finden, sondern auch, inwiefern die sprachlichen Merkmale der Fußnoten denen der Texte selbst entsprechen bzw. von ihnen abweichen.
Werbung stellt in einer von Globalisierung und Transkulturalität geprägten Welt ein Phänomen dar, das nicht nur omnipräsent ist und uns mit Glücksversprechen und Idealbildern in Plastik(-schein-)welten verführt, sondern zunehmend als populäre Form der Kunst gepriesen wird, deren Wirkung durch ausgefeilte Formen der Rhetorik sowie eine suggestive bzw. manipulative Bildersprache gekennzeichnet ist. Fach- und insbesondere Fremdsprachen bilden einen wesentlichen Bestandteil im sprachlichen und stilistischen Repertoire der Werbesprache. Der Beitrag gliedert sich denn auch in zwei Abschnitte: Im ersten Teil der Untersuchung wird das sprachliche und stilistische Repertoire (Fachsprachen, Jugendsprache, Dialekt, Fremdsprachen (Anglizismen) der Werbesprache dargestellt. Der zweite Teil der Untersuchung beinhaltet eine exemplarische Analyse von türkischen und deutschen bzw. deutschsprachigen Werbeanzeigen im Hinblick auf die Verwendung von Anglizismen.
