Background The arterial in line application of the leukocyte inhibition module (LIM) in the cardiopulmonary bypass (CPB) limits overshooting leukocyte activity during cardiac surgery. We now studied in a porcine model whether LIM may have beneficial effects on cardiac function after CPB. Methods German landrace pigs underwent CPB (60 min myocardial ischemia; 30 min reperfusion)without (group I; n=6) or with LIM (group II; n=6). The cardiac indices (CI) and cardiac function were analyzed pre and post CPB with a Swan-Ganz catheter and the cardiac function analyzer. Neutrophil labeling with technetium, scintigraphy, and histological analyses were done to track activated neutrophils within the organs. Results LIM prevented CPB-associated increase of neutrophil counts in peripheral blood. In group I, the CI significantly declined post CPB (post: 3.26 +/- 0.31; pre: 4.05 +/- 0.45 l/min/m2; p<0.01). In group II, the CI was only slightly reduced (post: 3.86 +/- 0.49; pre 4.21 +/- 1.32 l/min/m2; p=0.23). Post CPB, the intergroup difference showed significantly higher CI values in the LIM group (p<0.05) which was in conjunction with higher pre-load independent endsystolic pressure volume relationship (ESPVR) values (group I: 1.57 +/- 0.18; group II: 1.93 +/- 0.16; p<0.001). Moreover, the systemic vascular resistance and pulmonary vascular resistance were lower in the LIM group. LIM appeared to accelerate the sequestration of hyperactivated neutrophils in the spleen and to reduce neutrophil infiltration of heart and lung. Conclusions Our data provide strong evidence that LIM improves perioperative hemodynamics and cardiac function after CPB by limiting neutrophil activity and inducing accelerated sequestration of neutrophils in the spleen.
This study analyzes storyline structure in three Hausa home videos; Mai Kudi (The Rich Man), Sanafahna (with time truth shall dawn) and Albashi (Salary). The study measures storyline structure in these films against a Hollywood film industry model of story writing “the Hero's Journey”. It uses narrative analysis as its analytical tool, and narrative theory as its framework. After analyzing these videos, the study found that the major elements of storyline structure in Vogler's model formed the framework of the storyline structure in Hausa home videos analyzed. However, in spite of the preponderance of these elements within the storyline structure, there are significant variations to Vogler's model. Specifically, Vogler's model has some twelve stages spread on the universal structure of storytelling, i.e. beginning, middle and end. Few of these stages were found to exist in Hausa narrative structure, perhaps due to cultural differences between Western, Indian and Hausa cultures. The study therefore recommends screenwriters and producers to be aware of the existence of standard models of scriptwriting. It also recommends more training for script writers in the Hausa film industry.
Focus presuppositions

This paper reviews notions related to focus and presupposition and addresses the hypothesis that focus triggers an existential presupposition. Presupposition projection behavior in certain examples appears to favor a presuppositional analysis of focus. It is argued that these examples are open to a different analysis using givenness theory. Overall, the analysis favors a weak semantics for focus not including an existential presupposition.
Rhythmic changes in environmental lighting conditions have ever been the most reliable environmental cue for life on earth. Nature has therefore selected a genetically encrypted endogenous clock very early in evolution, as it provided cells and subsequently organisms with the ability to anticipate persevering periods of light and darkness. Rhythm generation within the mammalian circadian system is achieved by clock genes and their protein products. The mammalian endogenous master clock, which synchronizes the body to environmental time, is located in the suprachiasmatic nucleus (SCN) of the hypothalamus. As an integral part of the time-coding system, the pineal gland serves the need to tune the body to the temporal environment by the rhythmic nocturnal synthesis and immediate release of the hormone melatonin. In contrast to the transcriptional regulation of melatonin synthesis in rodents, a post-translational shaping is indicated in the human pineal gland. Another important mediator of circadian time and seasonality to the body is the pituitary gland. The aim of this work was to elucidate regulation of melatonin synthesis in the human pineal gland. Furthermore, presence and regulation of clock genes in the human pineal and pituitary gland, and in the SCN were analyzed. Therefore, human tissue, taken from regular autopsies, was analyzed simultaneously for different parameters involved in melatonin biosynthesis and circadian rhythm generation. Presented data demonstrate that post-mortem brain tissue can be used to detect the remnant profile of pre-mortem adaptive changes in neuronal activity. In particular, our results give strong experimental support for the idea that transcriptional mechanisms are not dominant for the generation of rhythmic melatonin synthesis in the human pineal gland. Together with data obtained for clock genes and their protein products in the pituitary, data presented here offer 1) a new working hypothesis for post-translational regulation of melatonin biosynthesis in the human pineal gland, and 2) a novel twist in the molecular competence of clock gene proteins, achieved by nucleo-cytoplasmic shuttling in neuronal and neuroendocrine human tissue. Furthermore, in this study, oscillations in abundance of clock gene proteins were demonstrated for the first time in the human SCN.
Poster presentation: The mammalian pineal organ is a peripheral oscillator, depending on afferent information from the so-called master clock in the suprachiasmatic nuclei of the hypothalamus. One of the best studied outputs of the pineal gland is the small and hydrophobic molecule melatonin. In all vertebrates, melatonin is synthesized rhythmically with high levels at night, signalling the body the duration of the dark period. Changes or disruptions of melatonin rhythms in humans are related to a number of pathophysiological disorders, like Alzheimer's disease, seasonal affective disorder or the Smith-Magenis-Syndrome. To use melatonin in preventive or curative interferences with the human circadian system, a complete understanding of the generation of the rhythmic melatonin signal in the human pineal gland is essential. Melatonin biosynthesis is best studied in the rodent pineal gland, where the activity of the penultimate and rate-limiting enzyme, the arylalkylamine N-acetyltransferase (AA-NAT), is regulated on the transcriptional level, whereas the regulatory role of the ultimate enzymatic step, achieved by the hydroxyindole O-methyltransferase (HIOMT), is still under debate. In rodents, Aa-nat mRNA is about 100-fold elevated during the night in response to adrenergic stimulation of the cAMP-signalling pathway, with AA-NAT protein levels closely following this dynamics. In contrast, in all ungulates studied so far (cow, sheep), a post-transcriptional regulation of the AA-NAT is central to determine rhythmic melatonin synthesis. AA-NAT mRNA levels are constantly elevated, and lead to a constitutive up-regulation of AA-NAT protein, which is, however, rapidly degraded via proteasomal proteolysis during the day. AA-NAT proteolysis is only terminated upon the nocturnal increase in cAMP levels. Similar to ungulates, a post-transcriptional control of this enzyme seems evident in the pineal gland of the primate Macaca mulatta. Studies on the molecular basis of melatonin synthesis in the human being are sparse and almost exclusively based on phenomenological data, derived from non-invasive investigations. Yet the molecular mechanisms underlying the generation of the hormonal message of darkness can currently only be deciphered using autoptic material. We therefore analyzed in human post-mortem pineal tissue Aa-nat and Hiomt mRNA levels, AA-NAT and HIOMT enzyme activity, and melatonin levels for the first time simultaneously within tissue samples of the same specimen. Here presented data show the feasibility of this approach. Our results depict a clear diurnal rhythm in AA-NAT activity and melatonin content, despite constant values for Aa-nat and Hiomt mRNA, and for HIOMT activity. Notably, the here elevated AA-NAT activity during the dusk period does not correspond to a simultaneous elevation in melatonin content. It is currently unclear whether this finding may suggest a more important role of the ultimate enzyme in melatonin synthesis, the HIOMT, for rate-limiting the melatonin rhythm, as reported recently for the rodent pineal gland. Thus, our data support for the first time experimentally that post-transcriptional mechanisms are responsible for the generation of rhythmic melatonin synthesis in the human pineal gland.
Ein Vergleich der Schrift Grimmelshausens mit anderen in der Fürstenspiegelliteratur vertretenen Positionen ergab einen völligen Gegensatz zum rein innerweltlichen ratio status-Begriff Machiavellis, aber auch eine weitreichende Differenz zum rational-scholastischen Denken Thomas von Aquins, das Theologie und Philosophie methodisch und inhaltlich trennt und beiden Wissensbereichen je spezifische Erkenntnisquellen zuschreibt. Grimmelshausens Schrift erweist sich als eine wesentlich theologische, die auf einer biblischexemplarischen Argumentation beruht. Für Grimmelshausen gibt es sowohl eine gebotene als auch eine verpönte ratio status. Weil die gebotene ratio status auch eine erforderliche Selbsterhaltung und Pflege des bonum commune gewährleisten soll, ist sie nicht wesentlich vom tradierten Begriff der prudentia zu unterscheiden. Die verpönte ratio status dagegen dreht die Beziehung der gebotenen ratio status zum Gemeinwohl um: Statt das bonum commune zu suchen, folgt der ungerechte Regent ausschließlich seinem bonum privatum. Um die ethische Neutralität dieses Strebens genauer zu artikulieren, wird angedeutet, dass der von der verpönten ratio status geleitete Regent per fas vel nefas handelt. Obwohl weder nach göttlichem noch nach natürlichem Recht Menschen dazu bestimmt sind, über andere Menschen zu herrschen, wird Grimmelshausen zufolge legitime politische Herrschaft in der Regel von Gott mittels des Volkes übertragen. Daher soll das Volk (mit Hilfe Gottes) seine politische Führung selbst bestimmen. Das heißt, dass das Volk berechtigt ist, über die Tugend und Aufrichtigkeit des künftigen Regenten selbst zu entscheiden. Um seine Deutung der Versuchung und Verwerfung des ersten israelitischen Königs zu begründen, stellt Grimmelshausen Sauls ursprüngliche Demut und Tugendhaftigkeit im Einzelnen dar. Saul wird anfänglich mit diesen königlichen Eigenschaften geschildert, um seine spätere Entwicklung zum Tyrannen als Ergebnis seiner Überheblichkeit gegen Gott und seiner fehlenden Selbsterkenntnis zu begründen. Diese Interpretation wird von Grimmelshausen verwendet, um seine Auffassung von der verpönten ratio status mit tradierten Vorstellungen der Tyrannen-Lehre zu verbinden. Anhand der Geschichte Davids stellt der Autor seine theologisch geprägte Erläuterung der gebotenen ratio status exemplarisch dar. Hier wird auch deutlich gemacht, dass die grundlegende Differenz zwischen David und Saul in ihrer unterschiedlichen Einstellung zu Gott besteht und nicht in ihrem äußeren Verhalten. Die Demut und Selbsterkenntnis Davids erlauben es ihm - trotz seiner moralischen Verfehlungen - seine Beziehung zu Gott zu festigen und der gebotenen ratio status zu folgen. Die biblische Darstellung Jonathans wird vom Autor benutzt, um die religiös-politische Rolle von Freundschaft in seiner Vorstellung der gebotenen ratio status näher zu bestimmen. Grimmelshausen betont die tradierte Ansicht, dass die Freundschaft tugendhafter Menschen eine wirkungsvolle Abwehr gegen die Gefahren der verpönten ratio status des Tyrannen bildet. Das unglückliche Ende Jonathans wird dann benutzt, um die politischen Dimensionen der Theodizee-Problematik zu diskutieren. Anschließend verwendet der Autor die Gestalt Joabs, des Heerführers Davids, um Standeserhalt als ein ethisches und politisches Problem einzuführen. Die Besonderheit dieser Diskussion liegt darin, dass sie versucht, die Verpflichtungen übertragener politischer Autorität zu begründen. Grimmelshausen versucht direkte Beobachtungen aus der damaligen Gesellschaft aufzuarbeiten, um die Entstehung und Wirkung der verpönten ratio status zu untersuchen. Dazu benutzt er die Gestalt Oliviers aus Simplicissimus Teutsch. Anhand dieser Verkörperung eines Räuber-Königs wird Machiavellis Vorstellung von der virtú des ethisch-autonomen Regenten in die Diskussion des Autors eingeführt, um einen Kontrast zu der politischen Tugend der frommen Regenten darzustellen. Dabei stellt sich heraus, dass die Entstehung der machiavellischen Mentalität von der luxuria und avaritia des damaligen Handelskapitalismus ermöglicht wurde. In seinen politisch-ethischen Schriften kann Grimmelshausen sich darauf verlassen, dass er einen Grundstock von christlich-biblischen und antiken Vorstellungen und Topoi mit seiner Leserschaft teilt. Dementsprechend kann man eine kontrastive Auseinandersetzung mit der Problematik der ratio status vor dem Hintergrund dieser Gemeinsamkeiten durchführen. Um Zeugnisse dieser Zeit begreiflich zu machen, muss man versuchen, sie in ihrem konkreten historischen und kulturellen Kontext zu verstehen, weil nur in diesem Kontext ihre politischen Absichten sichtbar gemacht werden können, die sich – im Falle Grimmelshausens – sowohl gegen die Entwicklung des modernen Kapitalismus als auch gegen die Herausbildung des absolutistischen Staates richten. Diese Voraussetzungen haben durch die Entwicklung der Politik als eine Erfahrungswissenschaft vieles von ihrer Verständlichkeit verloren. Die Vorstellung, dass die antike Tradition oder die Bibel zeitlos gültige Normen liefern oder dass sie übertragbares Erfahrungswissen oder gültige Analogien und brauchbare Präzedenzfälle bereitstellen, kann heute nur mit Mühe nachvollzogen werden. Die damals vertrauten Ideen, dass Politik die Lehre vom guten und gerechten Leben sei oder dass Ethik, Politik und Ökonomie nicht wahllos auseinandergetrennt werden dürfen, haben vieles von ihrer Selbstverständlichkeit verloren. Eine kontrastive Untersuchung der Gegensätze zwischen Grimmelshausens heilsgeschichtlichem Verständnis biblischer Vorbilder und den innovativen Vorstellungen einiger anderen politischen Denker der Frühen Neuzeit soll exemplarisch zeigen, dass die gleichen Topoi, Personen und Episoden unterschiedliche politische Interpretationen hervorrufen konnten. In einer Zeit, in der das tradierte Verständnis des Königtums und politische Legitimation im Allgemeinen sich grundsätzlich wandelte, konnte diese exegetische Vielfalt als Ausdruck grundlegender politischer Konflikte verstanden werden. Indem man die eher herkömmlich geprägten Beispiele politischer Argumentation in der Arbeit des Autors mit der pragmatischen Rezeption alttestamentarischer Gestalten und Episoden seitens einiger Anhänger des erstarkten Königtums vergleicht, kann man feststellen, dass bereits ein epochaler politischer Wandel sich ankündigte. Die Vermutung liegt nahe, dass durch diesen Wandel die alte sakrale Geschichtsauffassung umgestaltet werden musste, wie Bossuets Discours sur l’histoire universelle deutlich zeigt. Trotz ihrer antiabsolutistischen Absichten war die politische Wirkung der Schrift Grimmelshausens sowohl begrenzt als auch kurzlebig: Mit dem Aufkommen eines säkularen Geschichtsverständnisses verlor die heilsgeschichtlich begründete Darstellung antiabsolutistischer Argumentation weitgehend an Überzeugungskraft. Auf der einen Seite hat diese Entwicklung wohl zu einer Versachlichung politischer Argumentation geführt. Andererseits hat sie zu einer Verschleierung der politischen Intentionen des Autors beigetragen.
The high energy loss of heavy ions in matter as well as the small angular scattering makes heavy ion beams an excellent tool to produce almost cylindrical and homogeneously excited volumes in matter. This aspect can be used to pump short wavelength lasers. In an experiment performed at the GSI (Gesellschaft für Schwerionenforschung, Darmstadt, Germany) ion accelerator facility in December 2005 the well-known KrF* excimer laser was pumped with an intense high energy uranium beam. Pulses of an uranium beam with initial particle energy of 250 MeV per nucleon, provided by heavy-ion-synchrotron SIS-18, were delivered to the HHT-target station and then stopped inside a gas laser cell. The maximum beam intensity reached in the experiment was 2,5·109 particles per pulse, which resulted in 34 J/g specific energy deposited in the laser gas. By applying electron cooling and a bunch compression technique at SIS-18, the beam pulses were compressed down to 110 ns (FWHM). A mixture of an excimer laser premix gas (95,5% Kr + 0,5% F2) and a buffer gas (Ar 4.8) was used as the laser gas in proportions of 35/65 and 60/40, respectively. The gas pressure inside the laser cell was varied in the range of 1,2÷2 bar in continues flow mode. The experimental setup consisted of a 1 m long stainless steel tube with a number of diagnostic viewports and two mirror adjustment units. The optical cavity was formed by a flat, Alcoated mirror at the beam entrance and a second dielectrically coated, highly reflective mirror with 3 m radius of curvature at a distance of 1,3 m. A beam of heavy ions has been used to pump a short wavelength gas laser for the first time. Laser effect on the KrF* laser transition (λ = 248 nm) has been successfully demonstrated. Laser threshold for this specific setup was reached with a beam intensity of 1,2·109 particles per pulse. Laser action has been clearly proofed by the following methods: appearance of the laser line, spectral narrowing of the laser line, temporal narrowing of the laser signal, non-linear response of the laser output intensity on the pumping power, and cavity disalignment effect. An energy of the laser pulse of about 2 mJ was measured for an ion beam intensity of 2·109 particles per pulse. The time delay of the onset of the laser emission with respect to the pumping pulse was measured as a function of ion beam intensity. The dependence of spontaneous emission spectra on the gas pressure in a range of 1,3÷2 bar was observed and the optimal gas pressure for laser experiments in the sense of laser efficiency was concluded. As a next step in studying short wavelength lasers pumped with heavy ion beams it is planned to reduce the laser wavelength down to the VUV region of the spectrum, and to proceed to the excimer lasers of the pure rare gases: Xe2 * (λ = 172 nm), Kr2 * (λ = 146 nm), Ar2 * (λ = 126 nm), Ne2 * (λ = 83 nm) and He2 * (λ = 80 nm). We believe that the use of heavy ion beams as a pumping source may lead to new pumping schemes on the higher lying level transitions and considerably shorter wavelengths (XUV and X-ray spectral region), which rely on the high cross sections for multiple ionization of the target species.
This monograph highlights the necessity for taking preventive measures in the form of peace-building as a sustainable and long-term solution to conflicts in West Africa, with a special focus on the Mano River Union countries. Apart from the Mano River Union countries, efforts at resolving other conflicts in say, Guinea Bissau, Senegal, C?te d'Ivoire and Nigeria, have suffered from a lack of attention on the post-conflict imperatives of building peace in order to ensure that sustainable peace is achieved. Given the often intractable and inter-related nature of conflicts in this region, it argues for the need to revisit the existing mechanisms of conflict resolution in the sub-region with a view to canvassing a stronger case for stakeholders towards adopting the peace-building strategy as a more practical and sustainable way of avoiding wars in the sub-region. Peace-building in consonance with its infrastructure is a more sustainable approach to ensuring regional peace and stability and, therefore, ensuring development for the peoples of West Africa. Spatial interpolation of precipitation data is uncertain. How important is this uncertainty and how can it be considered in evaluation of high-resolution probabilistic precipitation forecasts? These questions are discussed by experimental evaluation of the COSMO consortium's limited-area ensemble prediction system COSMO-LEPS. The applied performance measure is the often used Brier skill score (BSS). The observational references in the evaluation are (a) analyzed rain gauge data by ordinary Kriging and (b) ensembles of interpolated rain gauge data by stochastic simulation. This permits the consideration of either a deterministic reference (the event is observed or not with 100% certainty) or a probabilistic reference that makes allowance for uncertainties in spatial averaging. The evaluation experiments show that the evaluation uncertainties are substantial even for the large area (41 300 km2) of Switzerland with a mean rain gauge distance as good as 7 km: the one- to three-day precipitation forecasts have skill decreasing with forecast lead time but the one- and two-day forecast performances differ not significantly.
Zur Unterscheidung von Dipsacus pilosus L. und Dipsacus strigosus WILLDENOW ex ROEMER et SCHULTES

Dipsacus strigosus WILLDENOW ex ROEMER et SCHULTES 1818 ist nach WISSKIRCHEN & HAEUPLER (1998) in Deutschland ein zumindest regional fest eingebürgerter Neophyt. Allerdings ist die Sippe in der Vergangenheit immer wieder mit der indigenen Dipsacus pilosus L. 1753 verwechselt worden und aus diesem Grunde wohl als bestimmungskritisch anzusehen, obwohl die Verschlüsselung in ROTHMALER, Band 4, seit der 9. Auflage (2002) eine eindeutige Bestimmung ermöglicht. In die verbreiteten Bestimmungswerke wurde Dipsacus strigosus erst im letzten Jahrzehnt aufgenommen. Die Sippe ist in den neueren Auflagen von SCHMEIL (2000), OBERDORFER (2001) und ROTHMALER (2002) verschlüsselt und in HAEUPLER & MUER (2000) abgebildet, fehlt aber noch in ROTHMALER, BAND 2 (1996).
