Precise tune determination and split beam emittance reconstruction at the CERN PS synchrotron

In accelerator physics, the need to improve the performance and better control the operating point of an accelerator has become, year after year, an increasingly important need in order to achieve higher energies and brightness, as well as point-like particle beams. If this involves increasingly advanced technological developments (in terms, for example, of materials for more intense superconducting magnets), it can not take place in the absence of targeted studies of linear and non-linear beam dynamics. In the context of this Ph.D. thesis in physics, linear and non-linear dynamics of charged particles in circular accelerators is the topic that will be discussed and treated in detail. In particular, the presentation and discussion of the results will be divided in two main topics: the need to know the physical properties of a proton beam; and the development of innovative methods to determine and study the accelerator’s working point. With regard to the first topic, an innovative procedure will be presented to determine the transverse size of the PS beam in the beam extraction phase. Among the different ways the extraction occurs at the PS, the analysed one is based on the transverse splitting of the beam by means of non-linear fields. Thus, the knowledge of the transverse beam size is not trivial since resonant linear and non-linear beam structures (namely, core and islands) arise and, for each of them, the beam size has to be quantified. This parameter is crucial for two main reasons: the accelerator that will receive the beam exiting the upstream accelerator may have restrictions (physical or magnetic) that involve a partial or total loss of the incoming beam; and any experiments located downstream of the considered accelerator may need a beam with a transversal size as constant as possible; consequently, its monitoring and control are essential. The second topic concerns the accurate determination of the working point of an accelerator, defined as the number of transverse oscillations the particle beam travels per unit of accelerator circumference, both horizontally and vertically. This quantity is called horizontal and vertical tune, respectively. Their knowledge is also crucial to understand whether the beam will be stable or unstable. In fact, not all tune values are acceptable, as there are particular values that bring the beam into resonance. In this configuration, the amplitude of the transverse oscillations of the particles increases in an uncontrolled manner and leads to the loss of all or part of the beam. Note that, in particular operating conditions, the resonant conditions are sought and desired to model, in a suitable way, the transversal shape of the beam, such as the above mentioned PS extraction scheme. It is even clearer how much the determination of the machine working point is essential to determine the operating conditions of an accelerator. In this context, several methods (also taken from the field of applied mathematics) to calculate the tune will be demonstrated and tested numerically on different types of synthetic signals. At the end of this description, the use of experimental data will allow to obtain the benchmark of a new method for the direct calculation of some characteristic quantities of non-linear beam dynamics (namely, the amplitude detuning, i.e. the variation of tune as a function of intensity of the perturbation provided to the beam.
Cancer cells, including leukemic cells, can react to therapeutic treatment by altering their metabolic phenotype (“metabolic reprogramming”) to keep their accelerated proliferative state, eventually becoming resistant to the treatment. There is an increasing amount of evidence indicating that metabolic reprogramming is one of the key mechanisms of acquisition of drug resistance by cancer cells. In agreement, several metabolic studies targeting leukaemia and specifically acute myeloid leukaemia (AML) and chronic myeloid leukaemia (CML), have been conducted over the last decades. However, there is still a lack of understanding the metabolic features of both AML and CML leukaemia specially in the acquisition of drug resistance, that is needed for unveiling novel and effective treatments for resistant and non-resistant patients. Therefore, the main objective of this thesis was to investigate the rewiring of cell metabolism occurring in the process of acquisition of resistance to conventional therapeutic treatments in AML and CML malignancies. Next, by revealing this metabolic rewiring, we intended to highlight potential metabolic and non-metabolic targets that could be exploited to overcome resistance to treatments. To this end, we have performed a comprehensive and comparative multi-OMIC study to analyse the links between the metabolic reprogramming and the resistance acquisition of THP-1 and HL-60 AML cell models sensitive or resistant to cytarabine (AraC) and doxorubicin (Dox), and of KU812 CML cell model sensitive or resistant to imatinib, all under normoxic (21% O2) and hypoxic (1% O2) conditions. The results of this thesis are divided into two chapters. On the one hand, in Chapter 1, the multi-OMIC study performed in AML parental and resistant cells unveiled that the acquisition of AraC resistance causes the reprogramming of the glucose metabolism of THP-1 and HL-60 cells by increasing the glycolytic flux whereas it is not associated with an alteration in the mitochondrial respiration. Moreover, our results also exhibited a possible disfunction of ETC complex I as well as alterations in glutamine and serine-glycine-1C metabolism in AML cells that display a more active mitochondrial metabolism. Moreover, we have also identified that the acquisition of Dox resistance causes alterations in the glucose and amino acid metabolism. Importantly, we have observed an important loss of mitochondrial respiration capacity of AML cells resistant to Dox chemotherapeutic drug, which constitutes a potential metabolic vulnerability that can be exploited for the treatment of AML patients resistant to Dox. On the other hand, in Chapter 2 is shown that the acquisition of imatinib resistance causes the reprogramming of glucose metabolism by enhancing the glycolytic flux, PPP, and glycogen metabolism, thus highlighting these metabolic pathways as potential metabolic weaknesses of KU812 cells resistant to imatinib. Moreover, we have observed a high metabolic plasticity of KU812 cells resistant to imatinib which includes the orchestration of many metabolic routes associated with the amino acid metabolism. Importantly, the CML multi-OMIC study has also unveiled an enhanced mitochondrial respiration capacity, which constitutes another potential vulnerability that can be exploited to overcome imatinib resistance. Finally, both AML and CML multi-OMIC studies have allowed us to propose and/or validate different metabolic and non-metabolic targets. In this regard, in this thesis we have identified and validated a battery of single-hit inhibitions that were able to reduce the cell viability of both parental and resistant AML and CML cells. Finally, we have confirmed that the repurposing of Dox chemotherapeutic drug counteracts the imatinib resistance in the KU812 cells resistant to imatinib.
Na «arte de procurar soluções», as populações locais na Guiné-Bissau respondem através de modelos variados de participação no sector da educação. As escolas populares, as escolas públicas com iniciativa de associações manjacas, as escolas de autogestão e as escolas comunitárias reflectem a diversidade de respostas das comunidades face às carências educativas não supridas pelo Estado. Os modelos de desenvolvimento participativo apresentados encontram fonte de aprendizagem no período colonial, em particular durante a luta pela independência. As decisões políticas assumidas no Congresso de Cassacá, em 1964, constituem os alicerces de experiências educativas baseadas em modelos de organização e participação da sociedade guineense, que voltarão a ser experimentadas com variáveis no período pós-independente até aos nossos dias. Na região de Bafatá, a participação das comunidades manifesta-se em 156 das 252 escolas existentes em 2006. O envolvimento da população contribui para minorar os efeitos negativos da pobreza e consequentemente para que se alcance a Educação Para Todos até 2015 cujo compromisso foi assinado pela Guiné-Bissau, na Conferência de Dacar. Na luta contra a pobreza, os factores políticos, históricos e sócio-culturais determinam a participação das populações na promoção da educação na Guiné-Bissau. A mobilização da população deve-se à ausência de resposta às suas necessidades, mas também ao aumento de prestígio que experiências educativas bem sucedidas têm granjeado quer em tabancas vizinhas quer junto de entidades externas. Este estudo pretende analisar o papel das populações no desenvolvimento da educação com destaque para as escolas comunitárias da região de Bafatá, no período de 2004 a 2006. INTRODUÇÃO Esta dissertação tem por objecto de estudo os efeitos dos programas de política económica e social de estabilização e de ajustamento estrutural2 no bemestar das famílias urbanas da capital de um país africano, a cidade de Bissau, na República da Guiné-Bissau, no período de 1986 a 2001. O contexto mais geral em que a investigação se insere, respeita à evolução política, económica e social do país após a independência, em 1974. A antiga Guiné Portuguesa procurou organizar a sua economia a partir de uma governação centralizada, com intervenção significativa de instituições estatais da administração central3, nacionalização de empresas existentes ou criação de outras com o mesmo estatuto. A dinamização do processo de desenvolvimento coube ao Partido para a Independência da Guiné e Cabo Verde (PAIGC), que dirigira a luta de libertação contra o colonialismo e se tornou o partido único e o agente principal de toda a vida colectiva, social e económica do país. Os instrumentos privilegiados foram os Planos de Desenvolvimento, apoiados em investimentos de grande dimensão e na ajuda internacional de origem em países de diferentes ideologias políticas. O enquadramento político expressou-se na organização do partido único, com uma governação que se impunha ao Governo e à Assembleia Nacional Popular, com mobilização política da população para a produção, incentivo a formas cooperativas de organização empresarial no campo e na cidade, repressão à oposição e à actividade de comerciantes e empresários privados. Os resultados negativos quanto ao objectivo traçado pelo partido e governo, de conseguir um melhor nível de bem-estar para a população, estão entre as origens de um golpe de Estado ( 14 de Novembro de 1980) liderado por uma parte dos militantes do PAIGC, sobretudo de origem guineense. O novo poder enveredou por um caminho de liberalização gradual da economia, mas também não conseguiu, até 1986, cumprir os objectivos de desenvolvimento a que se propunha.
O programa de ajustamento estrutural na república da Guiné-Bissau : Uma avaliação política e ética

Os guineenses assumiram o desenvolvimento como uma das metas a atingir e a estabilização e o ajustamento foi-lhes imposta como solução para os problemas estruturais existentes. No entanto, a forma como têm vindo a ser concebidos pelo Banco Mundial e pelo Fundo Monetário Internacional, direccionada sobretudo para a área económica, acabou por limitar o papel dos Programas de Ajustamento Estrutural (PAE) tidos como indutores do desenvolvimento, tornando-os num agregado de premissas austeras, com resultados não esperados. As propostas do FMI e do BM, tendendo para a liberalização económica e estímulo dos mercados em detrimento da intervenção estatal, traduzem-se em medidas de redução de taxas de utilização dos serviços públicos, supressão de subsídios, redimensionamento da administração pública, cortes, congelamentos salariais e privatizações. Os resultados destas reformas foram catastróficos, porquanto não só não melhoraram o défice orçamental, como os efeitos negativos das restrições orçamentais sobre o bem-estar, geraram um ambiente de promiscuidade social e o agravamento do sector informal como estratégia de sobrevivência Tendo em conta o objecto em estudo, isto é, a relação de forças que encontrámos entre o relacionamento entre os actores políticos guineenses e as Instituições Financeiras Internacionais, notámos que a ausência de comportamentos éticos também influiu nos resultados. Por um lado, o BM e o FMI, perante um Estado fragilizado, apresentaram condicionalismos à obtenção de empréstimos e ajudas, por outro lado, os actores guineenses, mesmo perante este dilema, não se coibiram do exercício da corrupção, do clientelismo e do neo-patrimonialismo, como estratégia para o enriquecimento fácil. Das Ziel dieser Arbeit war es, eine neuartige, drei-dimensionale Messmethode (BodyScan) zur Körperoberflächenbestimmung an Neugeborenen zu evaluieren. Die exakte Bestimmung der Oberfläche ist, besonders in der Pädiatrie, von großer Bedeutung für die Beurteilung physiologischer Parameter und der Dosierung von Medikamenten bzw. Infusionstherapien. Bestimmungen der Körperoberfläche mit 3D-Scannern liegen zwar für Erwachsene in zahlreichen Studien vor, jedoch wurden Neugeborene aus technischen Gründen bislang nicht untersucht, da ihre komplette Oberfläche in einem Scan nicht dargestellt werden kann. Das verwendete Messgerät „BodyScan“ der Firma 3D-Shape GmbH, Erlangen, arbeitete mit einer Messmethode, die es ermöglichte eine kontaktfreie, schonende, drei-dimensionale Vermessung der Körperoberfläche vorzunehmen (Streifenprojektionstechnologie). Die Neugeborenen konnten nur in liegender Position vermessen werden. Mit Hilfe einer Spiegelkonstruktion war es möglich drei Körperseiten des Neugeborenen im gescannten Bild zu erfassen. Diese gescannte Fläche wurde mit einem Korrekturfaktor zur gesamten Körperoberfläche extrapoliert. Dazu wurde der BodyScan vor Beginn der Studie mit einem artifiziellen Objekt (Messpuppe) kalibriert. Die genaue Oberfläche der Messpuppe war zuvor von einem ähnlichen genormten Gerät (BodyScan3DShape) auf 1632,24 cm², als Zähler des Korrekturfaktors bestimmt worden. Danach fanden 20 Messungen der Puppe mit dem Studien-Scanner statt. Es konnte gezeigt werden, dass der BodyScan die Körperoberfläche der Messpuppe mit einen Fehler von unter 0,1 % genau bestimmen konnte. Die ermittelte mittlere Oberfläche war der Nenner des so erhobenen Korrekturfaktors für unseren BodyScan, der schließlich 1,9787 betrug. Danach wurden 209 gesunde Neugeborenen, geboren in der Universitätsklinik Frankfurt am Main, untersucht. Sie wurden mit dem BodyScan in Bauch- und Rückenlage abgebildet, und die Körperoberfläche wurde zu beiden Ansichtsmodi computerunterstützt ermittelt. Die Ergebnisse des BodyScan wurden mit herkömmlichen, als empirischer Standard betrachteten Methoden, und zwar aus der Literatur bekannten Formeln nach DU BOIS & DU BOIS (1916), nach BOYD (1935), nach HAYCOCK ET AL. (1978), nach MEBAN (1983), und nach MOSTELLER (1987) verglichen. Zwei Neugeborene wurden außerdem mit der Geometrischen Methode untersucht, dazu wurden verschiedene Strecken und Umfänge einzelner Körperteile vermessen und deren Oberfläche mittels mathematischer Formeln bestimmt. Zudem wurde auch ein Subkollektiv von 25 Neugeborenen aus dem Gesamtkollektiv hinsichtlich der Ergebnisse der Körperoberflächenbestimmung mit dem BodyScan und den Formeln gesondert betrachtet. Dieses Subkollektiv setzte sich aus Neugeborenen zusammen, welche unter optimalen Bedingungen abgebildet wurden und von welchen qualitativ „sehr gute“ Aufnahmen erstellt wurden. Die durchschnittliche Körperoberfläche eines unter optimalen Bedingungen vermessenen Neugeborenen betrug mit dem BodyScan 2163,66 cm². Der Vergleich mit den fünf Formeln zur Oberflächenberechnung zeigte zwar geringfügige Abweichungen , aber dennoch gute Übereinstimmung der Ergebnisse unter den optimalen Studienbedingungen [besonders mit den Formeln nach DU BOIS & DU BOIS (1916), HAYCOCK ET AL. (1978), MEBAN (1983) , und MOSTELLER (1987)]. Die durchschnittliche Abweichung des BodyScans von den Formeln lag hier bei 2,55%. Als weiteres Ergebnis erhielten wir gute Übereinstimmungen für die Körperlängenbestimmung durch den BodyScan sowohl im Vergleich zu den Messungen der Hebammen als auch zu denen des Infantometers. Er betrug weniger als 0,1 % Fehler bei der Messpuppe und 2,8 % bei den Neugeborenen. Abschließend kann die Körperoberflächenmessung an Neugeborenen mit dem BodyScan als valide Methode bezeichnet werden. Sie setzt jedoch eine exakte Einstellung des Probanden in der Apparatur voraus. Weitere Untersuchungen sollten an kranken Neugeborenen und Frühgeborenen stattfinden, da die Methode wenig belastend und durch die Verwendung von weißem Licht unschädlich ist.

Die Akupunktur ist zentraler Bestandteil der traditionellen ostasiatischen Medizin, die auch Moxibustion und Kräuterheilkunde (Herbalmedizin) umfasst [Focks et al. 2006]. Akupunkturpunkte sind auch durch eine höhere Konzentration von sensorischen Rezeptoren und Mastzellen charakterisiert [Dung 1984; Heine 1988; Hwang 1992]. So ergaben Untersuchungen, dass die Stimulation (physikalische und chemische Reize) von Akupunkturpunkten auf Rezeptoren und auch Mastzellen einwirkt [Belmonte 1996; Schmidt 2002; Yang et al. 2007; Zhang et al. 2008a; Zhang et al. 2008b; Zhao 2008].
