We study the implications on compact star properties of a soft nuclear equation of state determined from kaon production at subthreshold energies in heavy-ion collisions. On one hand, we apply these results to study radii and moments of inertia of light neutron stars. Heavy-ion data provides constraints on nuclear matter at densities relevant for those stars and, in particular, to the density dependence of the symmetry energy of nuclear matter. On the other hand, we derive a limit for the highest allowed neutron star mass of three solar masses. For that purpouse, we use the information on the nucleon potential obtained from the analysis of the heavy-ion data combined with causality on the nuclear equation of state.
The first measurement of the fluctuation of the kaon-to-proton ratio in relativistic heavy-ion collisions is presented. This thesis details the analysis procedure for identifying kaons and protons using the NA49 experiment at CERN-SPS and discusses the results in the context of the current state of the field.
Background: Focused electron beam induced deposition (FEBID) is a direct-writing technique with nanometer resolution, which has received strongly increasing attention within the last decade. In FEBID a precursor previously adsorbed on a substrate surface is dissociated in the focus of an electron beam. After 20 years of continuous development FEBID has reached a stage at which this technique is now particularly attractive for several areas in both, basic and applied research. The present topical review addresses selected examples that highlight this development in the areas of charge-transport regimes in nanogranular metals close to an insulator-to-metal transition, the use of these materials for strain- and magnetic-field sensing, and the prospect of extending FEBID to multicomponent systems, such as binary alloys and intermetallic compounds with cooperative ground states.
Results: After a brief introduction to the technique, recent work concerning FEBID of Pt–Si alloys and (hard-magnetic) Co–Pt intermetallic compounds on the nanometer scale is reviewed. The growth process in the presence of two precursors, whose flux is independently controlled, is analyzed within a continuum model of FEBID that employs rate equations. Predictions are made for the tunability of the composition of the Co–Pt system by simply changing the dwell time of the electron beam during the writing process. The charge-transport regimes of nanogranular metals are reviewed next with a focus on recent theoretical advancements in the field. As a case study the transport properties of Pt–C nanogranular FEBID structures are discussed. It is shown that by means of a post-growth electron-irradiation treatment the electronic intergrain-coupling strength can be continuously tuned over a wide range. This provides unique access to the transport properties of this material close to the insulator-to-metal transition. In the last part of the review, recent developments in mechanical strain-sensing and the detection of small, inhomogeneous magnetic fields by employing nanogranular FEBID structures are highlighted.
Conclusion: FEBID has now reached a state of maturity that allows a shift of the focus towards the development of new application fields, be it in basic research or applied. This is shown for selected examples in the present review. At the same time, when seen from a broader perspective, FEBID still has to live up to the original idea of providing a tool for electron-controlled chemistry on the nanometer scale. This has to be understood in the sense that, by providing a suitable environment during the FEBID process, the outcome of the electron-induced reactions can be steered in a controlled way towards yielding the desired composition of the products. The development of a FEBID-specialized surface chemistry is mostly still in its infancy. Next to application development, it is this aspect that will likely be a guiding light for the future development of the field of focused electron beam induced deposition.
A careful analysis of the magneto-transport properties of epitaxial nanostructured Nb thin films in the normal and the mixed state is performed. The nanopatterns were prepared by focused ion beam (FIB) milling. They provide a washboard-like pinning potential landscape for vortices in the mixed state and simultaneously cause a resistivity anisotropy in the normal state. Two matching magnetic fields for the vortex lattice with the underlying nanostructures have been observed. By applying these fields, the most likely pinning sites along which the flux lines move through the samples have been selected. By this, either the background isotropic pinning of the pristine film or the enhanced isotropic pinning originating from the nanoprocessing have been probed. Via an Arrhenius analysis of the resistivity data the pinning activation energies for three vortex lattice parameters have been quantified. The changes in the electrical transport and the pinning properties have been correlated with the results of the microstructural and topographical characterization of the FIB-patterned samples. Accordingly, along with the surface processing, FIB milling has been found to alter the material composition and the degree of disorder in as-grown films. The obtained results provide further insight into the pinning mechanisms at work in FIB-nanopatterned superconductors, e.g. for fluxonic applications.
The biological effects of energetic heavy ions are attracting increasing interest for their applications in cancer therapy and protection against space radiation. The cascade of events leading to cell death or late effects starts from stochastic energy deposition on the nanometer scale and the corresponding lesions in biological molecules, primarily DNA. We have developed experimental techniques to visualize DNA nanolesions induced by heavy ions. Nanolesions appear in cells as “streaks” which can be visualized by using different DNA repair markers. We have studied the kinetics of repair of these “streaks” also with respect to the chromatin conformation. Initial steps in the modeling of the energy deposition patterns at the micrometer and nanometer scale were made with MCHIT and TRAX models, respectively.
Quarkonia are very promising probes to study the quark-gluon plasma. The essential baseline for measurements in heavy-ion collisions is high-precision data from proton-proton interactions. However, the basic mechanisms of quarkonium hadroproduction are still being debated. The most common models, the Color-Singlet Model, the non-relativistic QCD approach and the Color-Evaporation Model, are able to describe most of the available cross-section data, despite of their conceptual differences. New measures, such as the polarization, and data at a new energy regime are crucial to test the competing models. Another issue is an eventual interplay between the production process of a quarkonium state and the surrounding pp event. Current Monte Carlo event generators treat the hard scattering independently from the rest of the so-called underlying event. The investigation of possible correlations with the pp event might be very valuable for a detailed understanding of the production processes. ALICE ist the dedicated heavy-ion experiment at the LHC. Its design has been optimized for high-precision measurements in very high track densities and down to low transverse momenta. ALICE is composed of various different detectors at forward and at central rapidities. The most important detectors for this study are the Inner Tracking System and the Time Projection Chamber, allowing to reconstruct and identify electron candidate tracks within eta < 0.9. The Transition Radiation Detector has not been utilized at this stage of the analysis; however, it will strongly improve the particle identification and provide a dedicated trigger in the upcoming beam periods. ...
Pseudo-Critical Temperature and Thermal Equation of State from Nf = 2 Twisted Mass Lattice QCD
(2012)
We report about the current status of our ongoing study of the chiral limit of two-flavor QCD at finite temperature with twisted mass quarks. We estimate the pseudo-critical temperature Tc for three values of the pion mass in the range of mPS ~ 300 and 500 MeV and discuss different chiral scenarios. Furthermore, we present first preliminary results for the trace anomaly, pressure and energy density. We have studied several discretizations of Euclidean time up to Nt = 12 in order to assess the continuum limit of the trace anomaly. From its interpolation we evaluate the pressure and energy density employing the integral method. Here, we have focussed on two pion masses with mPS ~ 400 and 700 MeV.
This work derives the value of the neutron capture cross section of 60Fe at the energy of kT = 25 meV. Iron plays an important role in stellar nucleosynthesis, because it is a seed material for the s-process, a neutron capture process in which the elements between iron and bismuth are synthesized.
To determine the thermal neutron cross section of 60Fe, an iron sample produced in the framework of the ERAWAST1 program at the Paul Scherrer Institute in Switzerland was irradiated at the research reactor TRIGA2 at Johannes Gutenberg University in Mainz, Germany. Before the irradiation, the number of 60Fe particles in the sample was calculated using the decay scheme of 60Fe. There were (8.207 ± 0:066 stat 0:298 syst) * 10 14 60Fe particles in the sample.
The irradiation of the sample took place in May 2012. The counting of the reaction product, 61Fe, was undertaken using a HPGe detector located in a laboratory at Johannes Gutenberg University in Mainz. Knowing the number of 60Fe particles in the sample, the number of produced 61Fe particles and the neutron flux from the reactor, the thermal neutron capture cross section of ρth60Fe = (0.203 ± 0:021 stat 0:024 syst) b could be determined.
The determination of the thermal neutron capture cross section at the energy of kT = 25 meV constitutes to the first at this energy. There was already a measurement of the neutron capture cross section of 60Fe at the energy of kT = 25 keV in Karlsruhe in Germany. The result of the measurement was 25 keV = (9.9 ± 2:8 syst 1:4 stat) mb and was published in the year 2009 [14].
Adding datapoints for the cross section at different energies improves extarpolation and helps verification of theoretical models for elemental synthesis. For this reason, a measurement of the 60Fe neutron capture cross section at the energy of kT = 90 keV is planned at Goethe University Frankfurt, Germany.
Having the neutron capture cross section of 60Fe at the energy of kT = 25 keV measured in the year 2009 in Karlsruhe [14], at the energy of kT = 25 meV measured in May 2012 at Johannes Gutenberg University in Mainz and at the planned energy of kT = 90 keV at Goethe University Frankfurt, there will be three points for the extrapolation of the values for the neutron capture cross section of 60Fe. This will lead to a significant reduction in uncertainty with respect to existing theoretical discrepancies.
Feedforward inhibition and synaptic scaling are important adaptive processes that control the total input a neuron can receive from its afferents. While often studied in isolation, the two have been reported to co-occur in various brain regions. The functional implications of their interactions remain unclear, however. Based on a probabilistic modeling approach, we show here that fast feedforward inhibition and synaptic scaling interact synergistically during unsupervised learning. In technical terms, we model the input to a neural circuit using a normalized mixture model with Poisson noise. We demonstrate analytically and numerically that, in the presence of lateral inhibition introducing competition between different neurons, Hebbian plasticity and synaptic scaling approximate the optimal maximum likelihood solutions for this model. Our results suggest that, beyond its conventional use as a mechanism to remove undesired pattern variations, input normalization can make typical neural interaction and learning rules optimal on the stimulus subspace defined through feedforward inhibition. Furthermore, learning within this subspace is more efficient in practice, as it helps avoid locally optimal solutions. Our results suggest a close connection between feedforward inhibition and synaptic scaling which may have important functional implications for general cortical processing.
Quantum theory is the most successful physical theory ever. About one third of the gross national product in the developed countries results from its applications. These applications range from nuclear power to most of the high-tech tools for computing, laser, solar cells and so on. No limit for its range of validity has been found up to now...
A 5-gap timing RPC equipped with patterned electrodes coupled to both charge-sensitive and timing circuits yields a time accuracy of 77 ps along with a position accuracy of 38 μm. These results were obtained by calculating the straight-line fit residuals to the positions provided by a 3-layer telescope made out of identical detectors, detecting almost perpendicular cosmic-ray muons. The device may be useful for particle identification by time-of-flight, where simultaneous measurements of trajectory and time are necessary.
We study the light scalar mesons a_0(980) and kappa using N_f = 2+1+1 flavor lattice QCD. In order to probe the internal structure of these scalar mesons, and in particular to identify, whether a sizeable tetraquark component is present, we use a large set of operators, including diquark-antidiquark, mesonic molecule and two-meson operators. The inclusion of disconnected diagrams, which are technically rather challenging, but which would allow us to extend our work to e.g. the f_0(980) meson, is introduced and discussed.
It is a long discussed issue whether light scalar mesons have sizeable four-quark components. We present an exploratory study of this question using Nf = 2+1+1 twisted mass lattice QCD. A mixed action approach ignoring disconnected contributions is used to calculate correlatormatrices consisting of mesonic molecule, diquark-antidiquark and two-meson interpolating operators with quantum numbers of the scalar mesons a0(980) (1(0++)) and k (1/2(0+)). The correlation matrices are analyzed by solving the generalized eigenvalue problem. The theoretically expected free two-particle scattering states are identified, while no additional low lying states are observed. We do not observe indications for bound four-quark states in the channels investigated.
Directed deposition of silicon nanowires using neopentasilane as precursor and gold as catalyst
(2012)
In this work the applicability of neopentasilane (Si(SiH3)4) as a precursor for the formation of silicon nanowires by using gold nanoparticles as a catalyst has been explored. The growth proceeds via the formation of liquid gold/silicon alloy droplets, which excrete the silicon nanowires upon continued decomposition of the precursor. This mechanism determines the diameter of the Si nanowires. Different sources for the gold nanoparticles have been tested: the spontaneous dewetting of gold films, thermally annealed gold films, deposition of preformed gold nanoparticles, and the use of “liquid bright gold”, a material historically used for the gilding of porcelain and glass. The latter does not only form gold nanoparticles when deposited as a thin film and thermally annealed, but can also be patterned by using UV irradiation, providing access to laterally structured layers of silicon nanowires.
Interacting ultracold gases in optical lattices: non-equilibrium dynamics and effects of disorder
(2012)
This dissertation aims at giving a theoretical description of various applications of ultracold gases. A particular focus is cast upon the dynamical evolution of bosonic condensates in non-equilibrium by means of the time-dependent Gutzwiller method. Ground state properties of strongly interacting fermionic atoms in box and speckle disordered lattices are investigated via real-space dynamical mean-field theory. ...
In this thesis I use effective models to investigate the properties of QCD-like theories at nonzero temperature and baryon chemical potential. First I construct a PNJL model using a lattice spin model with nearestneighbor interactions for the gauge sector and four-fermion interactions for the quarks in (pseudo)real representations of the gauge group. Calculating the phase diagram in the plane of temperature and quark chemical potential in QCD with adjoint quarks, it is qualitatively confirmed that the critical temperature of the chiral phase transition is much higher than the deconfinement transition temperature. At a chemical potential equal to half of the diquark mass in the vacuum, a diquark Bose–Einstein condensation (BEC) phase transition occurs. In the two-color case, a Ginzburg–Landau expansion is used to study the tetracritical behavior around the intersection point of the deconfinement and BEC transition lines which are both of second order. A compact expression for the expectation value of the Polyakov loop in an arbitrary representation of the gauge group is obtained for any number of colors, which allows us to study Casimir scaling at both nonzero temperature and chemical potential. Subsequently I study the thermodynamics of two-color QCD (QC2D) at high temperature and/or density using ZQCD, a dimensionally reduced superrenormalizable effective theory, formulated in terms of a coarse grained Wilson line. In the absence of quarks, the theory is required to respect the Z2 center symmetry, while the effects of quarks of arbitrary masses and chemical potentials are introduced via soft Z2 breaking operators. Perturbative matching of the effective theory parameters to the full theory is carried out explicitly, and it is argued how the new theory can be used to explore the phase diagram of two-color QCD.
Which are the factors underlying human information production on a global level? In order to gain an insight into this question we study a corpus of 252–633 mil. publicly available data files on the Internet corresponding to an overall storage volume of 284–675 Terabytes. Analyzing the file size distribution for several distinct data types we find indications that the neuropsychological capacity of the human brain to process and record information may constitute the dominant limiting factor for the overall growth of globally stored information, with real-world economic constraints having only a negligible influence. This supposition draws support from the observation that the files size distributions follow a power law for data without a time component, like images, and a log-normal distribution for multimedia files, for which time is a defining qualia.
Author summary: The generation of new information is limited by two key factors, by the incurring economic costs and by the capacity of the human brain to process and store data and information; the controlling agent needs to retain an overall understanding even when data is generated by semiautomatic processes. These processes are reflected in the statistical properties of the data files publicly available on the Internet. Collecting a corpus of 252–633 mil. files we find that the statistics of the file size distribution are consistent with the supposition that data production on a global level is shaped and limited by the neuropsychological information processing capacity of the brain, with economic and hardware constraints having a negligible influence.
The subject of this thesis aimed at a better understanding of the spectacular X-ray burst. The most likely astrophysical site is a very dense neutron star, which accretes H/He-rich matter from a close companion. While falling towards the neutron star, the matter is heated up and a thermonuclear runaway is ignited. The exact description of this process is dominated by the properties of a few proton-rich radioactive isotopes, which have a low interaction probability, hence a high abundance.
The topic of this thesis was therefore an investigation of the short-lived, proton-rich isotopes 31Cl and 32Ar. The Coulomb dissociation method is the modern technique of choice. Excitations with energies up to 20 MeV can be induced by the Lorentz contracted Coulomb field of a lead target. At the GSI Helmholtzzentrum für Schwerionenforschung GmbH in Darmstadt, Germany, a Ar beam was accelerated to an energy of 825 AMeV and fragmented in a beryllium target. The fragment separator was used to select the desired isotopes with a remaining energy of 650 AMeV. They were subsequently directed onto a 208 Pb target in the ALAND/LAND setup. The measurement was performed in inverse kinematics. All reaction products were detected and inclusive and exclusive measurements of the respective Coulomb dissociation cross sections were possible.
During the analysis of the experiment, it was possible to extract the energy-differential excitation spectrum of 31Cl, and to constrain astrophysically important parameters for the time-reversed 30S(p,γ)31Cl reaction. A single resonance at 0.443(37) MeV dominates the stellar reaction rate, which was also deduced and compared to previous calculations.
The integrated Coulomb dissociation cross section of this resonance was determined to 15(6) mb. The astrophysically important one- and two-proton emission channels were analyzed for 32Ar and energy-differential excitation spectra could be derived. The integrated Coulomb dissociation cross section for two proton emission were determined with two different techniques. The inclusive measurement yields a cross section of 214(29stat)(20sys) mb, whereas the exclusive reconstruction results in a cross section of 226(14stat)(23sys) mb. Both results are in very good agreement. The Coulomb dissociation cross section for the one-proton emission channel is extracted solely from the exclusive measurement and is 54(8stat)(6sys) mb.
Furthermore, the development of the Low Energy Neutron detector Array (LENA) for the upcoming R3B setup is described. The detector will be utilized in charge-exchange reactions to detect the low-energy recoil neutrons from (p,n)-type reactions. These reaction studies are of particular importance in the astrophysical context and can be used to constrain half lifes under stellar conditions. In the frame of this work, prototypes of the detector were built and successfully commissioned in several international laboratories.
The analysis was supported by detailed simulations of the detection characteristics.
Low-level laser irradiation of visible light had been introduced as a medical treatment already more than 40 years ago, but its medical application still remains controversial. Laser stimulation of acupuncture points has also been introduced, and mast-cells degranulation has been suggested. Activation of TRPV ion channels may be involved in the degranulation. Here, we investigated whether TRPV1 could serve as candidate for laser-induced mast cell activation. Activation of TRPV1 by capsaicin resulted in degranulation. To investigate the effect of laser irradiation on TRPV1, we used the Xenopus oocyte as expression and model system. We show that TRPV1 can functionally be expressed in the oocyte by (a) activation by capsaicin (K 1/2 = 1.1 μM), (b) activation by temperatures exceeding 42°C, (c) activation by reduced pH (from 7.4 to 6.2), and (d) inhibition by ruthenium red. Red (637 nm) as well as blue (406 nm) light neither affected membrane currents in oocytes nor did it modulate capsaicin-induced current. In contrast, green laser light (532 nm) produced power-dependent activation of TRPV1. In conclusion, we could show that green light is effective at the cellular level to activate TRPV1. To which extend green light is of medical relevance needs further investigation.
We discuss deviations from the exponential decay law which occur when going beyond the BreitWigner distribution for an unstable state. In particular, we concentrate on an oscillating behavior, remisiscent of the Rabi-oscillations, in the short-time region. We propose that these oscillations can explain the socalled GSI anomaly, which measured superimposed oscillations on top of the exponential law for hydrogen-like nuclides decaying via electron-capture. Moreover, we discuss the possibility that the deviations from the Breit-Wigner in the case of the GSI anomaly are (predominantely) caused by the interaction of the unstable state with the measurement apparatus. The consequences of this scenario, such as the non-observation of oscillations in an analogous experiment perfromed at Berkley, are investigated.
We present and compare new types of algorithms for lattice QCD with staggered fermions in the limit of infinite gauge coupling. These algorithms are formulated on a discrete spatial lattice but with continuous Euclidean time. They make use of the exact Hamiltonian, with the inverse temperature beta as the only input parameter. This formulation turns out to be analogous to that of a quantum spin system. The sign problem is completely absent, at zero and non-zero baryon density. We compare the performance of a continuous-time worm algorithm and of a Stochastic Series Expansion algorithm (SSE), which operates on equivalence classes of time-ordered interactions. Finally, we apply the SSE algorithm to a first exploratory study of two-flavor strong coupling lattice QCD, which is manageable in the Hamiltonian formulation because the sign problem can be controlled.
It is widely believed that chiral symmetry is spontaneously broken at zero temperature in the strong coupling limit of staggered fermions, for any number of colors and flavors. Using Monte Carlo simulations, we show that this conventional wisdom, based on a mean-field analysis, is wrong. For sufficiently many fundamental flavors, chiral symmetry is restored via a bulk, first-order transition. This chirally symmetric phase appears to be analytically connected with the expected conformal window of manyflavor continuum QCD. We perform simulations in the chirally symmetric phase at zero quark mass for various system sizes L, and measure the torelon mass and the Dirac spectrum. We find that all observables scale with L, which is hence the only infrared length scale. Thus, the strong-coupling chirally restored phase appears as a convenient laboratory to study IR-conformality. Finally, we present a conjecture for the phase diagram of lattice QCD as a function of the bare coupling and the number of quark flavors.
Analgesia is a well-documented effect of acupuncture. A critical role in pain sensation plays the nervous system, including the GABAergic system and opioid receptor (OR) activation. Here we investigated regulation of GABA transporter GAT1 by δOR in rats and in Xenopus oocytes. Synaptosomes of brain from rats chronically exposed to opiates exhibited reduced GABA uptake, indicating that GABA transport might be regulated by opioid receptors. For further investigation we have expressed GAT1 of mouse brain together with mouse δOR and μOR in Xenopus oocytes. The function of GAT1 was analyzed in terms of Na(+)-dependent [(3)H]GABA uptake as well as GAT1-mediated currents. Coexpression of δOR led to reduced number of fully functional GAT1 transporters, reduced substrate translocation, and GAT1-mediated current. Activation of δOR further reduced the rate of GABA uptake as well as GAT1-mediated current. Coexpression of μOR, as well as μOR activation, affected neither the number of transporters, nor rate of GABA uptake, nor GAT1-mediated current. Inhibition of GAT1-mediated current by activation of δOR was confirmed in whole-cell patch-clamp experiments on rat brain slices of periaqueductal gray. We conclude that inhibition of GAT1 function will strengthen the inhibitory action of the GABAergic system and hence may contribute to acupuncture-induced analgesia.
Detailed knowledge of reaction mechanisms is key to understanding chemical, biological, and biophysical processes. For many reasons, it is desirable to comprehend how a reaction proceeds and what influences the reaction rate and its products.
In biophysics, reaction mechanisms provide insight into enzyme and protein function, the reason why they are so efficient, and what determines their reaction rates. They also reveal the relationship between the function of a protein and its structure and dynamics.
In chemistry, reaction mechanisms are able to explain side products, solvent effects, and the stereochemistry of a product. They are also the basis for potentially optimizing reactions with respect to yield, enhancing the stereoselectivity, or for modifying reactions in order to obtain other related products.
A key step to investigate reaction mechanisms is the identification and characterization of intermediates, which may be reactive, short-lived, and therefore only weakly populated. Nowadays, the structures of those can in most cases only be hypothesized based on products, side products, and isolable intermediates, because intermediates with a life time of less than a few microseconds are not accessible with the commonly used techniques for structure determination such as X-ray crystallography and nuclear magnetic resonance (NMR) spectroscopy.
In this thesis, two-dimensional infrared (2D-IR) spectroscopy is shown to be a powerful complement to the existing techniques for structure determination in solution. 2D-IR spectroscopy uses a femtosecond laser setup to investigate interactions between vibrations - analogous to 2D-NMR, which investigates the interactions between spins. Its ultrafast time resolution makes 2D-IR spectroscopy particularly well suited for the two topics investigated in this thesis: Structure Determination of Reactive Intermediates and Conformational Dynamics of Proteins.
Structure Determination of Reactive Intermediates: The focus of this thesis is using polarization-dependent 2D-IR (P2D-IR) spectroscopy for structure determination of N-crotonyloxazolidinone (referred to as 1), a small organic compound with a chiral oxazolidinone, known as Evans auxiliary, and its reactive complexes with the Lewis acids SnCl4 and Mg(ClO4)2. Chiral oxazolidinones in combination with Lewis acids have frequently been used in stereoselective synthesis for over 30 years. Nevertheless, the detailed mechanisms are in many cases xvi ABSTRACT still mere hypotheses and have not yet been experimentally proven. By accurately measuring the angles between the transition dipole moments in the molecules using an optimized P2D-IR setup and comparing the results to DFT calculations, the conformation of 1 and the conformation and coordination of the main complexes with SnCl4 and Mg(ClO4)2 are unequivocally identified and analyzed in depth. Structural details, such as a slight twist in the solution structure of 1, are detected using P2D-IR spectroscopy; these cannot be inferred from NMR spectroscopy or DFT calculations. In addition to the main Lewis acid complexes, complexes in low concentration are detected and tentatively assigned to different conformations and complexation geometries. The knowledge of those structures is essential for rationalizing the observed stereoselectivities. Additionally, a method is introduced that enables structure determination of molecules in complex mixtures and even in the presence of molecules with similar spectral properties and in high concentration. This work sets the stage for future studies of other substrate-catalyst complexes and reaction intermediates for which the structure determination has not been possible to date.
Conformational Dynamics of Proteins: Exchange 2D-IR spectroscopy allows the investigation of fast dynamics without disturbing the equilibrium of the exchanging species. It is therefore well suited to investigate fast dynamics of proteins and to reveal the speed limit of those. The temperature dependence of the conformational dynamics between the myoglobin substates A1 and A3 in equilibrium is analyzed. The various substates of myoglobin can be detected with FTIR spectroscopy, if carbon monoxide is bound to the heme. From previous studies it is known that the exchange rates at room temperature are in the picosecond time range, well suited to be investigated by 2D-IR spectroscopy. In the temperature range between 0 °C and 40 °C only a weak temperature dependence of the exchange rate in the myoglobin mutant L29I is observed in the present study. The exchange rate approximately doubles from 15 ns-1 at 0 °C to 31 ns-1 at 40 °C. It turned out that the conformational dynamics correlates linearly with the solvent viscosity, which itself is temperature dependent. Comparing our results to measurements at cryogenic temperatures, the linear relation between exchange time constant for this process and the viscosity is shown for the temperature range between -100 °C and 40 °C (corresponding to a viscosity change of 14 orders of magnitude). Thus, it is proven that the dynamics of the conformational switching are mainly determined by solvent dynamics, i.e., the protein dynamics are slaved to the solvent dynamics. This is the first time slaving is observed for such fast processes (in the picosecond time range). The observation implies a long-range structural rearrangement between the myoglobin substates A1 and A3. In addition, the exchange for other mutants and wild type myoglobin is analyzed qualitatively and found to agree with the conclusions drawn from L29I myoglobin.
Proteomic profiles of myocardial tissue in two different etiologies of heart failure were investigated using high performance liquid chromatography (HPLC)/Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS). Right atrial appendages from 10 patients with hemodynamically significant isolated aortic valve disease and from 10 patients with isolated symptomatic coronary heart disease were collected during elective cardiac surgery. As presented in an earlier study by our group (Baykut et al., 2006), both disease forms showed clearly different pattern distribution characteristics. Interesting enough, the classification patterns could be used for correctly sorting unknown test samples in their correct categories. However, in order to fully exploit and also validate these findings there is a definite need for unambiguous identification of the differences between different etiologies at molecular level. In this study, samples representative for the aortic valve disease and coronary heart disease were prepared, tryptically digested, and analyzed using an FT-ICR MS that allowed collision-induced dissociation (CID) of selected classifier masses. By using the fragment spectra, proteins were identified by database searches. For comparison and further validation, classifier masses were also fragmented and analyzed using HPLC-/Matrix-assisted laser desorption ionization (MALDI) time-of-flight/time-of-flight (TOF/TOF) mass spectrometry. Desmin and lumican precursor were examples of proteins found in aortic samples at higher abundances than in coronary samples. Similarly, adenylate kinase isoenzyme was found in coronary samples at a higher abundance. The described methodology could also be feasible in search for specific biomarkers in plasma or serum for diagnostic purposes.
The intriguing effects of electroweak induced parity violation (PV) in molecules have yet to be observed, but experiments on molecular PV promise to provide fascinating insights. They potentially offer a novel testing ground for the low energy sector of the standard model and, in addition, a successful measurement of PV differences between the two enantiomers of a chiral molecule could promote a deeper understanding of molecular chirality, by essentially establishing a new link between particle physics and biochemistry. A key challenge in the design of such experiments is the identification of suitable molecules, which in turn requires widely applicable computational schemes for the prediction of PV experimental signals. To this end, a quasirelativistic density functional theory approach to the calculation of PV effects in nuclear magnetic resonance (NMR) spectra of chiral molecules has been developed and implemented during the course of this thesis. It includes relativistic as well as electron--correlation effects and has been used extensively in the screening of molecules possibly suited for a first observation of molecular PV. Some relevant compound classes have been identified, but none of their selected representatives are predicted to exhibit PV NMR frequency shifts that can be detected under current experimental restrictions. In order to advance the design of molecules which exhibit particularly large PV signals in experiments, systematic effects on PV NMR frequency splittings such as scaling with nuclear charge, conformational dependence and the impact of atomic substitution around the NMR active nucleus have been studied. Previously predicted scaling laws were confirmed and it was determined that the environment of the NMR active nucleus, both in terms of conformation and atomic composition, can be tuned to increase PV frequency shifts by several orders of magnitude. In addition to molecules suited for NMR experiments, a fascinating chiral actinide compound was studied with regard to PV frequency shifts in vibrational spectra. This compound displays the largest such shift ever predicted for an existing molecule, which lies well within the attainable experimental resolution. The challenge now lies in making it compatible with current experimental setups.
In this thesis we discussed the expansion behaviour of an ultracold bosonic gas from an initial harmonic confinement. We studied the reaction of the non-interacting system to changes of the trap frequency ω and of the strongly interacting system to changes of the number of Mott insulating particles NMI in the initial state and the interaction U/J. The total number of particles is kept constant for the different simulations, which are performed by means of the Bosonic Gutzwiller approach...
