Stereotaktische Biospien gehören seit vielen Jahren zu den Standardoperationen zahlreicher neurochirurgischer Kliniken. Hierbei werden Proben von Hirnläsionen entnommen, um diese histopathologisch zu untersuchen.
Die histopathologische Diagnose unklarer Hirnläsionen ist zwingend erforderlich, um eine adäquate Therapie durchzuführen. Eine weitere Therapie kann aus Bestrahlung, Chemotherapie, Kombination beider oder Resektion bestehen. In wenigen Fällen wird eine zweite oder dritte Biopsie benötigt, um eine endgültige Diagnose zu erhalten. Das Ziel dieser Studie war es, jene Patienten genauer zu untersuchen, bei denen die erste Biopsie kein definitives Ergebnis erbracht hatte. Die meisten dieser Patienten mussten sich einer zweiten Biopsie unterziehen. Wir haben eine umfassende Recherche der letzten 10 Jahre durchgeführt und eine Datenbank mit den Patienten erstellt, bei denen die erste Biopsie kein Ergebnis erbracht hatte.
Hierbei wurden klinische Parameter, welche einen Einfluss auf die nicht zielführenden Biopsie haben können, erhoben, beschrieben und diskutiert. Die Parameter umfassten die entnommene Probenanzahl, Kontrastmittelaufnahme der Läsion, Lokalisation der Läsion, Erfahrung des Operateurs, neuroradiologische Verdachtsdiagnose und Vorbehandlung.
Wir haben in dieser retrospektiven Arbeit unser Augenmerk auf die klinischen Aspekte der einzelnen Patienten, bei denen die erste Biopsie kein definitives Ergebnis erbrachte, gelegt.
Hier zeigten sich keinerlei Auffälligkeiten, welche positiv mit einer nichtzielführenden Biopsie einhergehen könnten.
Wir folgern, dass in den meisten Fällen eine definitive Diagnose zu erwarten ist. Unklar bleibt, bei welchen Patienten keine zielführende Biopsie erfolgen wird, so dass sie einer erneuten Biopsie unterzogen werden müssen.
Beim Auffinden menschlicher Überreste stellt sich neben der Beurteilung des postmortalen Intervalls auch konsequent die Frage nach der Möglichkeit des Vorliegens eines Tötungsdelikts. Da Weichgewebe nur in begrenztem Ausmaß Verwesung, Fäulnis oder Umwelteinflüssen standhält, ist dieses nur bedingt geeignet, auch langfristig Spuren von Gewalteinwirkung zu konservieren. Knochengewebe hingegen kann Läsionen noch nach langen Zeiträumen nahezu unverändert abbilden und stellt somit einen forensisch bedeutenden Spurenträger dar.
Im Rahmen dieser retrospektiven Studie sollte geklärt werden, inwieweit und in welchem Ausmaß bei Tötungsdelikten knöcherne Verletzungen entstehen. Ob bei definierten Formen letaler Gewalteinwirkung unterschiedliche Häufigkeiten des Auftretens knöcherner Verletzungen zu beobachten und zudem bevorzugte Körperregionen zu identifizieren sind, stellte eine weitere wesentliche Fragestellung der Arbeit dar.
Nach Auswertung der Sektionsprotokolle von insgesamt 897 im Institut für Rechtsmedizin in Frankfurt am Main obduzierten, im In- und Ausland begangenen Tötungsdelikten im Zeitraum 01.01.1994 bis 31.12.2014 zeigte sich, dass unabhängig von der Art der tödlichen Gewalteinwirkung 70,9% der Opfer mindestens eine knöcherne Verletzung aufwiesen und darüber hinaus bei insgesamt 45,5% der Opfer mehrfache knöcherne Verletzungen nachgewiesen werden konnten.
Zudem zeigte sich, dass unterschiedliche, definierte letale Gewalteinwirkungen entsprechend charakteristische Häufigkeiten und Verteilungen knöcherner Verletzungen zur Folge haben. So sind mit 92,6 % die häufigsten knöchernen Läsionen bei Schussopfern festzustellen. Nach stumpfer und scharfer Gewalt mit je 80 % und 66,3 % ließ sich auch nach tödlicher Gewalteinwirkung gegen den Hals in 53,3 % der Fälle mindestens eine knöcherne Läsion nachweisen.
Das Fehlen knöcherner Verletzungen in insgesamt 29% der im Auswertungszeitraum untersuchten 897 Tötungsdelikte zeigt auch, dass selbst bei knöchern unversehrten, vollständigen Skelettfunden ein Homizid keineswegs ausgeschlossen werden kann. Neben der gerichtlichen Leichenöffnung sind stets ergänzende forensische Aufarbeitungen der menschlichen Überreste zu fordern. Hierbei sind einerseits physikalische und chemische Methoden in Betracht zu ziehen, vor allem jedoch auch radiologische Untersuchungen. Weitere Untersuchungen der gewonnenen Ergebnisse im Rahmen einer weiteren Studie sollen klären, welcher Stellenwert der postmortalen Computertomographie zugesprochen werden kann.
Mycophenolate-Mofetil (MMF) clinically used as CellCept is inserted as immunosuppressive xenobiotic drug for preventing transplantate rejection. It is well known that MMF works selectively through inhibiting the IMPDH which is an essential enzyme in the de novo pathway for biosynthesis of guanosine Nukleotides.
In this study, we investigated the effects of MMF on the astrocytes, because astrocytes are important glial cells of the CNS with various functions in the healthy tissue, such as being involved in the neuronal differentiation, axonal growth and regulation of the environmental composition. They also build up the scared tissue during acute CNS lesions by releasing neurotoxic substances such as NO and different inflammatory Zytokines, e.g. IL-1ß and TNF-a, which even influence the microglial cell proliferation, migration and activity. Therefore, astrocytes are impressingly involved in the extent of the neuronal damage.
Our observations revealed an influence of astrocytic proliferation dependent on the concentration of the serum (10%, 5%, 2%, 1% and serum free medium) used for the in vitro cultivation: the more the serum part in the medium the more the extent of the cell proliferation. The proliferation of astrocytes cultured in serum free medium can be increased by Corticotropin Releasing Factor (CRF) 10µM and Guanosine application. Dose-dependent MMF led to a suppression of the astrocytic proliferation which is antagonisable with Guanosine only.
The number of the isolated proliferating astrocytes labeled by BrdU was significantly reduced after simultaneous treatment with serum (10% or 1%) and dose-dependent MMF (10µg/ ml, 1µg/ ml and 0,1µg/ ml) application, whereas LPS showed no effect on the proliferating rate. But, as measured by ELISA, with LPS conditioned medium contained relevant amounts of TNF-a and NO, but surprisingly not of IL-1ß. There were a significant reduction of cytokine and NO amounts determined in the astrocytic conditioned medium treated only with MMF dependent of the concentration (10µg/ml, 1µg/ml and 0,1µg/ml). Even the simultaneous treatment with LPS and MMF revealed compared to the LPS stimulated astrocytic group a measurable significant decrease of TNF-a. The treatment of the astrocytic cultures with guanosine lifted up the effects of MMF on TNF-a secretion. Therefore, we conclude, that the immunosuppressive drug MMF might have a neuroprotective and scar formation modulating effect through the antiproliferative potency on astrocytic cells and modulating character of the microenvironment by influencing the TNF-a secretion. Moreover, these regulatory effect on microglial cells are described as suppression (Hailer NP,Wirjatijana F,Roser N,Hirschebeth GT,Korf HW,Dehghani F,2001).

Die evolutionäre Voraussetzung für die Entwicklung komplexer, differenzierter Organismen bildet die Separierung der Zelle in Reaktionsräume, die so genannte Kompartimentierung. Das Prinzip der Kompartimentierung ermöglicht zahlreiche lebensnotwendige, biochemische Prozesse, wie die Konservierung von Energie durch Protonengradienten in der Atmungskette oder parallele, gegenläufige Stoffwechselwege. Zelluläre Kompartimente werden häufig durch Biomembranen gebildet, welche aus einer zweilagigen Lipidschicht bestehen. Lipidmoleküle in einer Zelle sind meistens amphipathisch, das bedeutet, sie bestehen aus einer polaren, hydrophilen Kopfgruppe und einem unpolaren, hydrophopen Ende (Abbildung 1). Ce travail, dont le titre est : Le modèle d’immersion réciproque en question : enseigner en classe bilingue à New York et à Francfort / Das Two-Way-Immersion Modell im Fokus: Zweisprachig unterrichten in New York und Frankfurt/Main, a pour objectif de répondre à la problématique suivante : Comment la conceptualisation du modèle d’enseignement bilingue d’immersion réciproque influence les pratiques éducatives (didactiques et pédagogiques) des enseignants, et pourquoi?
Afin de répondre à cette question, j’ai choisi d’étudier un modèle bilingue spécifique, le modèle bilingue d’immersion réciproque (ou « Two-Way-Immersion ») qui, dans la façon dont il intègre deux groupes d’élèves locuteurs de différentes langues qui apprennent les uns des autres, m’a semblé reposer sur une conceptualisation de l’enseignement bilingue dépassant une vision monolingue de ce dernier. Je montrerai dans la thèse, qu’en réalité, un même modèle peut se décliner différemment selon les choix didactiques et pédagogiques et selon les contextes de sa mise en œuvre et que seule une analyse approfondie peut permettre d’en révéler toute la complexité. À cette fin, j’ai observé des classes bilingues à New York et à Francfort, classes qui ont ensuite constitué mon terrain d’étude.
La thèse est organisée en 5 chapitres. Le premier chapitre propose un passage en revue de la littérature scientifique sur l’enseignement bilingue dans divers contextes. Le deuxième chapitre est consacré à la méthodologie de recherche que j’ai choisi d’utiliser pour explorer la complexité de mon objet de recherche. Les deux chapitres suivants proposent une analyse sociolinguistique de chaque contexte, New York (3) et Francfort (4). Le dernier chapitre, (5), est consacré à l’analyse des données récoltées (des entretiens, des observations de séances, des documents d’élèves et divers documents pédagogiques). Mon objectif, dans ce chapitre, est de montrer comment les orientations influencent les choix didactiques et pédagogiques des enseignants et ont un impact sur les pratiques de classe.
The individual parameter

The present thesis tackles the unification of two-dimensional semantic systems, which are designed to deal with context-dependency of a certain kind i.e. indexicality, with dynamic theories of meaning, designed to capture facts about anaphoricity and the distribution of definite and indefinite articles. The need for a more principled look at this unification is twofold. Firstly, there is an overlap of these two families of theories in terms of empirical data, namely third person personal pronouns, as well as definite descriptions. Both kinds of expressions have anaphoric as well as non-anaphoric usages, whereas some of the latter ones can be captured in terms of indexicality. But, on the other hand, no language, especially not German and English, the main sources of data in this thesis, seems to distinguish these two usages formally, i.e. by employing different expressions. Hence the need for a unified framework in which this sort of ambiguity can be treated. Secondly, the theoretical state is dissatisfactory in the sense that the families of theories take very disparate forms that are not easy to relate conceptually.
The overlap in empirical area of application strongly suggests that this dichotomy is an artifact of the way these theories traditionally are developed and justified. This thesis seeks to overcome this state of the field. It proceeds as follows.
The first chapter discusses the way in which theories indexicality are designed. After taking a closer look some hallmarks of these theories such as the notions of index- and context-dependency themselves, double indexing, etc., it develops a notion of index dependency that makes use of a more complex individual parameter than the one that is usually assumed in the literature. Apart from agents and addressees, the two standard components of indices that represent contexts, additional objects are assumed. This leads to a variant of the semantics of deictically used third person expression that is called ‘indexical theory of demonstratives’, which is then investigated further.
The second chapter discusses the classics of dynamic semantics: DRT, DPL, and FCS. It arrives at the common core of all of these theories that consists in the assumption of a novel sort of variable namely active variables as opposed to free and bound ones that are intended to model the behavior of (in)definite descriptions and pronouns. The projection behavior of these variables or discourse referents is described either in (discourse-)syntactic or semantic terms. The chapter also arrives at a new formulation of the uniqueness condition that is thought to be part of the semantics of definite descriptions and sketches an account of transparent negation.
The third chapter then combines the insights of the previous ones by developing the notion of representation that connects the entities of evaluation of the first chapter i.e. indices with those of the second namely sets of assignments, a.k.a. files. The formal language that emerged in the second chapter is endowed with two kinds of variables for situations to allow for double indexing within a dynamic setting. A novel interpretation mechanism for the so designed language is proposed, which is shown to capture not only those aspects that are known to exist in two-dimensional frameworks, but also certain other index-index interactions that are described in yet another body of literature.
The final chapter discusses potential flaws of the theory and sketches an account of allegedly bound indexicals that is compatible with Kaplan’s infamous ban on monsters.
Das Ziel dieser Dissertation war es, die biologische Relevanz der F1Fo-ATP-Synthase für den Alterungsprozess des Ascomyceten P. anserina aufzuklären sowie die Funktion der Dimerisierungsuntereinheiten PaATPE und PaATPG genauer zu untersuchen. Folgende Ergebnisse wurden dabei erzielt:
1. Der Verlust einer Dimerisierungsuntereinheit führt in P. anserina zum Verlust der Dimerisierungsfähigkeit der F1Fo-ATP-Synthase. Dieses Ereignis resultiert in einer vorzeitigen Anhäufung von Seneszenzmerkmalen, einer starken Verkürzung der Lebensspanne und einem beschleunigten Alterungsprozess. Der Phänotyp der Stämme ∆PaAtpe und ∆PaAtpg lässt sich erfolgreich revertieren. Dadurch konnte bestätigt werden, dass der beobachtete Phänotyp der Deletionsstämme auf den Verlust der Dimerisierungsgene zurückzuführen ist.
2. Die konstitutive Überexpression des PaAtpe-Gens führt zu einer Verlängerung der Lebensspanne, die allerdings nicht abhängig von einer Erhöhung der Dimer-Menge, sondern auf eine Verlängerung der Mitochondriennetzwerke und eine erhöhte Atmung zurückzuführen ist.
3. Obwohl die Lebensspannen der untersuchten PaAtpg_OEx-Stämme voneinander abweichen, ist der auf den Organismus ausgeübte Effekt der PaAtpg-Überexpression positiv. Dabei lässt sich eine Erhöhung der Dimer-Menge auch in diesen Stämmen nicht nachweisen und eine Veränderung der Mitochondrienmorphologie tritt nur in einem der fünf untersuchten Stämme auf, was hier allerdings mit dem größten Effekt auf die Lebensspanne korreliert.
4. Die gleichzeitige Überexpression der Gene PaAtpe und PaAtpg führt interessanterweise zu einer Aufhebung der durch die PaAtpe-Überexpression hervorgerufenen positiven Effekte. Die generierte Doppelmutante weist somit wildtypische Eigenschaften auf. Eine Erhöhung der Dimer-Menge kann trotz Überexpression beider Gene nicht festgestellt werden.
5. Trotz gleicher Funktion in der Dimerbildung der F1Fo-ATP-Synthase ist das Expressionsniveau der Dimerisierungsgene in P. anserina unterschiedlich. Bereits im Wildtyp wird das PaAtpe-Gen weniger transkribiert als das PaAtpg-Gen. Letzteres wird durch die PaAtpe-Überexpression sowohl in den PaAtpe_OEx-Mutanten als auch in der Doppelmutante herunterreguliert. Auch im Wildtyp kommt es zu einer altersabhängigen Herunterregulierung des PaAtpg-Gens.
6. Im Wildtyp nimmt die Dimer-Menge im Alter ab und es kommt zu einer altersbedingten Änderung der Superkomplexe von S1 zu S0. Dies sind Hinweise auf eine altersabhängige Remodellierung der Cristae-Membran, die möglicherweise zum Seneszenzphänotyp von P. anserina beiträgt. Somit ist die Aufrechterhaltung des Dimers von großer Bedeutung für die Gewährleistung mitochondrialer Funktionen des Organismus.
Die F1Fo-ATP-Synthase spielt in ihrer dimeren Form eine bedeutende Rolle in der Aufrechterhaltung der Mitochondrienfunktion und gewährleistet den Ablauf von lebenswichtigen mitochondrialen Prozessen, die für die Erhaltung der zellulären Homöostase von essentieller Bedeutung sind.
NADH:ubiquinone oxidoreductase (Complex Ⅰ) is the first and largest enzyme in the respiratory chain. It catalyzes the transfer of two electrons from NADH to ubiquinone via a series of enzyme-bound redox centers - Flavin mononucleotide (FMN) and iron-sulfur (Fe-S) clusters – and couples the exergonic reaction with the endergonic translocation of four protons across the membranes. Bacteria contain the minimal form of complex I, which is composed of 14 conserved core subunits with a molecular mass of around 550 kDa. Complex Ⅰ has an L-shaped structure which can be subdivided into two major parts (arms). The hydrophilic arm protruding into the bacterial cytosol (or mitochondrial matrix) harbors the binding site for the substrate NADH, the two- to one-electron switch FMN and all one-electron transferring Fe-S clusters and therefore considered as the catalytic unit. Auswirkung der Chemisorption von Organothiolaten auf den elektrischen Widerstand dünner Goldfilme

Hochgeordnete Monolagen von Organothiolaten auf Goldoberflächen bilden sich bei Kontakt einer Goldoberfläche mit einer Lösung eines Thiols oder Thioacetats spontan aus. Die Adsorption auf dünnen Metallfilmen mit Schichtdicken im Bereich von 25 - 100 nm führt zu einer Änderung des elektrischen Widerstandes des Films, die an Goldfilmen mit Schichtdicken von 25 - 40 nm über eine einfache Zweipunktmessung verfolgt wurde. Die Proportionalität der Widerstandsänderung mit der Menge an adsorbiertem Material konnte für die in dieser Arbeit verwendeten Dünnschichtsensoren bestätigt werden. Zu diesem Zweck wurden gleichzeitig Widerstands- und Oberflächenplasmonenresonanzmessungen an 40 nm starken Goldfilmen durchgeführt. In diesen Experimenten zeigt sich die Widerstandsmessung zur Beobachtung der Adsorptionskinetik als die überlegene Technologie.
Die durch Mikrokontaktdrucken und Freiätzen der gedruckten Strukturen hergestellten Sensoren zeigen eine individuelle Signalintensität. Die Normierung auf die maximale, durch Belegung mit Hexadecanthiol (HDT) oder Dodecanthiol erreichte, Signalstärke ermöglichte den Vergleich der maximalen Signalstärke von Thiolatmonolagen, die durch Belegung mit n-Alkanthiolen (CH_3(CH_2)_(n-1)-SH mit n = 12, 16, 19, 22 und 33, Cn), 11-Mercaptoundecyl-hexaethylenglycol (HSC11EG6OH), Adamantan-1-thiol (AdaSH), Triptycenthiol (TrpSH), Anthracen-2-thiol (Ant-0SH), Anthracen-2-alkanthiolen (Ant-(CH_2)_n-SH mit n = 1 - 5 und 10, Ant-nSH), p-Terphenyl-4-thiol (TP0SH), p-Terphenyl-4-alkanthiolen (TP-(CH_2)_n-SH mit n = 1 - 4, TPnSH) und p-Terphenyl-4-ethanthioacetat (TP2SAc) erzeugt wurden. Die Größe der Widerstandsänderung zeigt eine deutliche Abhängigkeit vom organischen Rest des Oberflächenadsorbats. Für die Verstärkung des Signals wurde die folgende Reihenfolge gefunden: Trp > Ada > Ant-0 > Ant-1 > TP0 > TP1 > Ant-2 > TP2 > Cn (n = 12 - 33) = C11EG6OH = Ant-n (n = 3 - 11) = TPn (n = 3, 4). Bei bekannter Verstärkung des Signals durch ein Adsorbat kann unabhängig von der Oberflächenrauigkeit die Oberflächenbedeckung durch Chemisorbate in einer Güte bestimmt werden, die der durch STM- und TEM-Messungen erreichten vergleichbar ist. Die Methode wurde angewendet, um Schichten von TP2SH und TP2SAc, die bei 20 und 60 °C aus ethanolischer Lösung abgeschieden wurden, zu vergleichen. Die Unterschiede in der Oberflächendichte, die durch eine Erhöhung der Abscheidungstemperatur zu beobachten sind, können durch eine Beschleunigung der Reaktion nach Arrhenius erklärt werden. Auch die Temperaturabhängigkeit der Abscheidungsgeschwindigkeit von HDT aus ethanolischer Lösung an Goldoberflächen, die in einem Bereich von -10 °C bis +30 °C betrachtet wurde, ist mit dem Arrhenius'schen Ansatz konform. Die Aktivierungsenergie der Adsorption von HDT auf Gold wurde auf E_a = 23 +-6 kJ/mol bestimmt.
Die Konzentrationsabhängigkeit der Abscheidung aus ethanolischer Lösung an Goldoberflächen wurde für HDT, AdaSH, TP2SH und TP2SAc untersucht. Um eine Präadsorption der Thiole vor dem eigentlichen Start der Messung zu verhindern, wurde eine Apparatur mit einem Diaphragma aus Aluminium entwickelt, das beim Start der Messung mit dem Sensor durchstoßen wird. Mit Ausnahme von TP2SH zeigen alle Adsorptive ein Adsorptions-Desorptions-Gleichgewicht. Die Adsorptionsisothermen bei 20 °C lassen sich am besten durch die Freundlich-Isotherme beschreiben. Während die Reaktionsordnung im Adsorbat für die Adsorption der Thiole nahe an 1 liegt, hat sie für die Adsorption von TP2SAc einen Wert von ca. 1/4. Damit ergibt sich für die Geschwindigkeitskonstante der Adsorption k_a(HDT) = (2,3 +-0,2) 10^4 L/(mol s), k_a(AdaSH) = (6,1 +-0,2) 10^4 L/(mol s), k_a(TP2SH) = (7,3 +-0,4) 10^3 L/(mol s) und k_a(TP2SAc) = (8 +-3) 10^-2 L^(1/4)/(mol^(1/4) s). Die Adsorptionskurven der Thiole weisen bei Konzentrationen unterhalb von 5 10^-5 mol/L einen linearen Bereich auf, der einer zwischenzeitlichen Diffusionskontrolle zugeordnet wird.
An die aufgenommenen Adsorptionskurven der Thiole wurden literaturbekannte Modelle numerisch angepasst und teilweise weiterentwickelt. Die Anpassung konnte durch die Einführung einer vor der Oberfläche gelagerten Diffusionsgrenzschicht, in welcher der zeitabhängige Verlauf der Analytkonzentration in einem System von 10 Schichten berechnet wurde, deutlich verbessert werden. Von allen getesteten Modellen zeigt nur die Adsorption mit Ausschlussmuster keine Konzentrationsabhängigkeit der Geschwindigkeitskonstante der Desorption. Dieses Modell bezieht den Verlust von Adsorptionsplätzen mit ein, die einem besetzten Adsorptionsplatz benachbart sind und durch das adsorbierte Teilchen verdeckt werden. Der daraus resultierende Zusammenhang zwischen der Konzentration freier Adsorptionsplätze und dem Bedeckungsgrad der Oberfläche Theta_F(Theta) ist abhängig vom Verhältnis der Stoßfrequenz zwischen den Teilchen und der Oberfläche zur Platzwechselfrequenz der Teilchen auf der Oberfläche. Zur Bestimmung von Theta_F(Theta) für die numerische Anpassung der Adsorptionskurven von HDT und TP2SH wurde die Oberflächenbesetzung in einem Monte-Carlo-Verfahren für eine Konzentrationsreihe in Zehnerpotenzschritten simuliert.
Due to the resurrection of data-hungry models (such as deep convolutional neural nets), there is an increasing demand for large-scale labeled datasets and benchmarks in the computer vision fields (CV). However, collecting real data across diverse scene contexts along with high-quality annotations is often expensive and time-consuming, especially for detailed pixel-level label prediction tasks such as semantic segmentation, etc. To address the scarcity of real-world training sets, recent works have proposed the use of computer graphics (CG) generated data to train and/or characterize performance of modern CV systems. CG based virtual worlds provide easy access to ground truth annotations and control over scene states. Most of these works utilized training data simulated from video games and pre-designed virtual environments and demonstrated promising results. However, little effort has been devoted to the systematic generation of massive quantities of sufficiently complex synthetic scenes for training scene understanding algorithms. In this work, we develop a full pipeline for simulating large-scale datasets along with per-pixel ground truth information. Our simulation pipeline constitutes of mainly two components: (a) a stochastic scene generative model that automatically synthesizes traffic scene layouts by using marked point processes coupled with 3D CAD objects and factor potentials, (b) an annotated-image rendering tool that renders the sampled 3D scene as RGB image with a chosen rendering method along with pixel-level annotations such as semantic labels, depth, surface normals etc. This pipeline is capable of automatically generating and rendering a potentially infinite variety of outdoor traffic scenes that can be used to train convolutional neural nets (CNN).
However, several recent works, including our own initial experiments demonstrated that the CV models that are trained naively on simulated data lack generalization capabilities to real-world scenes. This opens up several fundamental questions about what is it lacking in simulated data compared to real data and how to use it effectively. Furthermore, there has been a long debate since 1980’s on the usefulness of CG generated data for tuning CV systems. Primarily, the impact of modeling errors and computational rendering approximations, due to various choices in the rendering pipeline, on trained CV systems generalization performance is still not clear. In this thesis, we take a case study in the context of traffic scenarios to empirically analyze the performance degradations when CV systems trained with virtual data are transferred to real data. We first explore system performance tradeoffs due to the choice of the rendering engine (e.g., Lambertian shader (LS), ray-tracing (RT), and Monte-Carlo path tracing (MCPT)) and their parameters. A CNN architecture, DeepLab, that performs semantic segmentation, is chosen as the CV system being evaluated. In our case study, involving traffic scenes, a CNN trained with CG data samples generated with photorealistic rendering methods (such as RT or MCPT), shows already a reasonably good performance on real-world testing data from CityScapes benchmark. Use of samples from an elementary rendering method, i.e., LS, degraded the performance of CNN by nearly 20%. This result conveys that training data must be photorealistic enough for better generalizability of the trained CNN models. Furthermore, the use of physics-based MCPT rendering improved the performance by 6% but at the cost of more than three times the rendering time. This MCPT generated dataset when augmented with just 10% of real-world training data from CityScapes dataset, the performance levels achieved are comparable to that of training CNN with the complete CityScapes dataset.
The next aspect we study in the thesis involves the impact of choice of parameter settings of scene generation model on the generalization performance of CNN models trained with the generated data. Towards this end, we first propose an algorithm to estimate our scene generation model parameters given an unlabeled real world dataset from the target domain. This unsupervised tuning approach utilizes the concept of generative adversarial training, which aims at adapting the generative model by measuring the discrepancy between generated and real data in terms of their separability in the space of a deep discriminatively-trained classifier. Our method involves an iterative estimation of the posterior density of prior distributions for the generative graphical model used in the simulation. Initially, we assume uniform distributions as priors over parameters of a scene described by our generative graphical model. As iterations proceed the uniform prior distributions are updated sequentially to distributions for the simulation model parameters that leads to simulated data with statistics that are closer to the distributions of the unlabeled target data.

Universities typically offer video-based courses and internships to prepare prospective teachers for classroom management. Empirical studies have shown the effectiveness of analyzing video clips in programs for teacher training. However, no empirical studies have explored the relationship between the teaching experience of prospective teachers and the effectivity of video-based courses on classroom management. The research question was: How can classroom management competency be effectively conveyed in video-based courses regarding the differences in practical experiences of prospective teachers. Theory on cognitive load and situated learning suggests that prospective teachers with teaching experience could benefit more from a situated instructional strategy, whereas prospective teachers without this experience benefit more from a cognitive instructional strategy.
The sample consisted of a total of 87 prospective teachers belonging to a first cohort that had participated in an internship and a second cohort that had not yet participated. Participants of both cohorts were randomly assigned to a course following one of the two instructional strategies. A video test was designed to assess classroom management competence. Additionally, measurements of prospective teachers’ attentiveness and their self-efficacy at the start of the internship were included.
The study showed a significant main effect between pretest and posttest. However, neither a significant difference in the effectiveness between the two instructional strategies nor between the two cohorts was found (first order). Also, the effectiveness of the instructional strategy was not influenced by the availability of teaching experience (second order). Scores on the video test were not significantly related to prospective teachers’ attentiveness. The prospective teachers that participated in the course before participation in the internship rated their own classroom management significantly lower than the prospective teachers that participated in the course after participating in their internship.
These results firstly indicate that there was no significant difference in cognitive load between the groups, implying that both instructional strategies are suitable for the conveying of classroom management competence within universities. Secondly, the current study has found no evidence that the trait of attentiveness influences prospective teachers’ performance within video tests. Thirdly, these results give reason to believe that the valid assessment of self-efficacy on classroom management depends on the degree of familiarity with the topic, for example, through previous dealings within video-based courses. The limited reliability of the new video test should be taken into account when interpreting the results of the current study.
Identification and characterization of hypoxia-regulated long non-coding RNAs in endothelial cells

RNA deep sequencing of the human transcriptome revealed that almost ~84 % of the genome are transcribed, however, only 2 % of all transcripts encode for proteins. All remaining transcripts are referred to as non-coding RNAs and can be divided into small non-coding RNAs (<200 nt) and long non-coding RNAs (lncRNAs; >200 nt). Studies throughout the last decade suggest a broad functional spectrum for lncRNAs. Regarding the cardiovascular field, several studies could show that lncRNAs are implicated in various aspects of endothelial cell biology. The response to hypoxia and the regulation of angiogenesis are key events in the context of several diseases. Therefore, the aim of this study was to determine the influence of hypoxia on lncRNA expression in human umbilical vein endothelial cells and furthermore, to characterize the lncRNA function on a molecular level. ...
The genetic mutation of the coagulation factor VIII (fVIII) results in a defective or missing protein, leading to a malfunctioning blood coagulation. The resulting disease is called hemophilia A. Depending on the severity of the mutation, affected patients experience an increased risk of pathologic bleeding after minor trauma or even sudden bleeding events. Substitution therapies with extrinsic fVIII exist using plasmatic or recombinant fVIII products. Due to an insufficient immune tolerance towards substituted fVIII, about 30 % of patients develop allogenic neutralizing antibodies (inhibitors) against substituted fVIII products. The gold standard of treating inhibitors is the immune tolerance induction (ITI), where patients are given frequent, high doses of fVIII to induce an immune tolerance. ITI therapy fails in about 30 % of patients. Mechanisms of action of ITI are part of research, as insufficient knowledge about mechanisms and prognostic factors complicate treatment. For example, the development of anti-idiotypic antibodies, which occur naturally as a regulatory mechanism of the immune system, are being studied. Such anti-idiotypes have been detected in immunoglobuline preparations and in patients after successful ITI.
Inhibitors interfere with fVIII function in coagulation by binding functional epitopes within fVIII domains. Inhibitors against the A2 and C2 domain are predominantly found, however also the C1 domain has been shown to be highly immunogenic in some patients. The polyclonality of inhibitors aggravates the understanding and treatment of these. The present project therefore focusses on the selection of synthetic anti-idiotypic antibodies to target inhibitors in patients. The phage display method was applied to, for one, isolate anti-idiotypic single chain variable fragments (scFvs) specific against human polyclonal anti-fVIII antibodies and second against two C1 domain-specific inhibitory monoclonal antibodies (mAbs).
In the first project, anti-fVIII antibodies were purified from human plasma to serve as target molecules. A previous project showed that using full plasma as a target did not yield anti-idiotypic antibodies from phage display. For the purification, protein A chromatography and fVIII coupled Affi Gel® chromatography were applied. The isolated antibodies were next used as targets for the selection of anti-idiotypic scFvs. Analysis revealed that none of the selected phages solely bound the anti-fVIII antibody target. Consequently, the test protocol was modified, which resulted in a reduction of unspecific binders. Yet, no target-specific binders were isolated from phage pools. Reason for this may have been the high diversity of the polyclonal antibody target and the limited diversity of the phage libraries.
The aim of the second project, was the selection and characterization of scFvs, that target the paratopes of C1 domain-specific mAbs GMA8011 and LE2E9. From a therapeutic viewpoint, the preparation of an anti-idiotypic antibody pool, tailored to each patient’s inhibitor population, could help neutralize inhibitors in patients. Ultimately, one GMA8011-specific scFv-carrying phage clone (H2C1) and two specifics to LE2E9 (H3G7, H3F10) were isolated. In further experiments, only the GMA8011-specific scFv showed competitive behavior in presence of fVIII, pointing towards an anti-idiotypic binding to the inhibitor paratope. The LE2E9-specific scFvs did not prevent binding of the inhibitor to fVIII. Hence, no anti-idiotypic behavior could be determined. For further characterization, selected scFvs were genetically fused to Fc antibody fragments and recombinantly produced. In this antibody format, all three scFvs showed concentration dependent binding to the target and the isotype control. The binding specificity to the target, observed in phage context, could not be reproduced. Competition experiments with fVIII confirmed that none of the scFvs bound the paratope of their target inhibitor.
The selection of anti-idiotypic scFvs from phage display libraries proves to be effortful. Polyclonal anti-fVIII antibodies purified from hemophilic plasma appear to be unsuitable as a target for phage display, likely due to the high diversity of the target molecules. Furthermore, the preparation of an individualized anti-idiotypic pools for patients by selecting scFvs against single inhibitory mAbs proves to be difficult. The selection of scFvs against anti-C1 inhibitors GMA8011 and LE2E9 produced three promising scFv-carrying phages. However, analysis could not detect anti-idiotypic behavior. Further research with inhibitors, monoclonal and polyclonal, and anti-idiotypic antibodies should be performed to bring better insight into the highly complex paratope-epitope interaction.
Die Sepsis ist mit einer hohen Letalität im Krankenhaus verbunden. Ein wesentlicher Auslöser der Sepsis ist das Endotoxin. Es führt zu Entzündungsreaktionen im Organismus mit Ausbildung eines septischen Schocks bis hin zum Multiorganversagen. Zudem führt es zu Veränderungen im Gerinnungssystem mit unterschiedlich schweren Ausprägungen. Das Endotoxin wird physiologisch im menschlichen Körper durch IgM neutralisiert. In dieser Studie wurde der Effekt von IgM angereichertem intravenösen Immunglobulin auf die Endotoxinaktivität untersucht. Zusätzlich wurden die Auswirkungen einer IgM-IVIg-Therapie auf Entzündungsparameter und auf viskoelastische und konventionelle Gerinnungsparameter betrachtet. Patienten mit schwerer Sepsis und septischem Schock auf einer anästhesiologisch-chirurgischen Intensivstation wurden rekrutiert. Nach dem Studieneinschluss von 15 Patienten erfolgte die Implementierung einer neuen SOP, in der die Anwendung von IgM-IVIg (5g/kg/d für 3 Tage) integriert wurde. Daraus bildeten sich eine Kontroll- und eine IgM-IVIg Gruppe. Der Beobachtungszeitraum umfasste 4 Tage. Es wurde der Endotoxin Activity Assay® (EAA®) zur Messung der Endotoxinaktivität an Vollblutproben durchgeführt. Die durchgeführte Rotationsthrombelastometrie ROTEM® diente neben der Messung der Gerinnungszeiten auch zur Erfassung der mechanischen und zeitlichen Stabilität eines Blutgerinnsels. Als Maß für die Aggregation von Thrombozyten diente die Impedanzaggregometrie Multiplate®. Die Routinelaborparameter wurden nach allgemeinem Standard erhoben. Eingeschlossen wurden die Daten von 26 Patienten. Die IgM-IVIg-Gruppe zeigte am ersten Tag, 6 und 12 Stunden nach Behandlungsbeginn, eine Reduktion der Endotoxinaktivität (0,51±0,06 vs. 0,26±0,07, p<0,05) und unterschied sich signifikant im Vergleich zur Kontrollgruppe 6 Stunden nach Behandlungsbeginn (0,26±0,07 vs. 0,43±0,07, p<0,05). Die Thrombozytenzahl war signifikant höher in der IgM-IVIg- Gruppe im Beobachtungszeitraum (200/nl±43 vs. 87/nl±20, p<0,05). Die Fibrinogenkonzentration war in der Kontrollgruppe am zweiten (311mg/dl±37 vs. 475mg/dl±47 (p=0,015)) und am vierten Tag (307mg/dl±35 vs. 420mg/dl±16 (p=0,017)) signifikant niedriger. Es konnten keine Unterschiede in den thromboelastometrischen oder aggregometrischen Untersuchungen, oder bei den Entzündungsparametern beobachtet werden. Die präsentierten Ergebnisse müssen aufgrund der geringen Stichprobengröße sorgsam interpretiert werden. Dennoch könnten die Ergebnisse die Basis für weitere Studien in der Zukunft sein, die auf IgM-IVIg als eine therapeutische Option bei Patienten mit einer hohen Endotoxinaktivität abzielen. In der vorliegenden Studie fanden wir Hinweise, dass eine IgM-IVIg-Therapie bei Patienten mit schwerer Sepsis und septischem Schock die Endotoxinaktivität vermindert. Zudem scheint IgM-IVIg bei o.g. Patienten eine pathognomonische Thrombozytopenie positiv zu modelieren.
The study focuses on the introduction of a health education curriculum in Cyprus’ public schools. The curriculum’s implementation is looked at as a project of modernization and is examined ethnographically in two primary schools in the Republic of Cyprus over a period of three years. Utilizing theories and methods from Science and Technology Studies and Global Ethnography, the study examines the entanglements of Science with Culture and of Tradition with Modernity as experts, teachers, parents and children encounter the new health education curriculum. Health education is compared to a project of biological citizenship and the curriculum is seen as an actant attempting to form a personal obligation towards health by promoting “common sense” knowledge and privileging “modern” individuals.
Der Goldstandard zur Detektion eines erhöhten intrakraniellen Drucks ist das invasive Monitoring, z. B. mittels externer Ventrikeldrainage bei Intensivpatienten bzw. die Lumbalpunktion mit Messung des Liquoreröffnungsdrucks. Die am häufigsten angewendete klinische Untersuchung zum Ausschluss eines erhöhten ICP ist die Funduskopie. In den letzten 30 Jahren wurden zwei ultraschallbasierte Untersuchungsmethoden beschrieben um einen erhöhten ICP zu detektieren und die Funduskopie zu diesem Zweck zu ersetzen. Das Ziel unserer Studie war es, die Messung des Durchmessers der Sehnervenscheide (optic nerve sheat diameter, ONSD), der Undulation des Septum pellucidum (septum pellucidum undulation, SPU) hervorgerufen durch passive Rotation des Kopfes, und die Funduskopie mit einer invasiven Hirndruckmessung zu vergleichen. Der ONSD wurde mittels transbulbärer B-Mode Sonographie gemessen, für die Ermittlung der SPU wurde eine transtemporale M-Mode Sonographie durchgeführt. Zunächst schlossen wir gesunde Probanden ein, um die intrarater- und interrater- Reproduzierbarkeit der Ultraschalluntersuchungen zu berechnen. Die intrarater Reproduzierbarkeit des ONSD betrug 0,65 bis 0,84 und die interrater Reproduzierbarkeit war zwischen 0,69 und 0,91 (n=10). Die intrarater Reproduzierbarkeit der SPU war 0,29 und die interrater Reproduzierbarkeit ergab 0,26 (n=15). Anschließend untersuchten wir 45 Patienten, die für eine invasive Hirndruckmessung vorgesehen waren. Auch hier ermittelten wir die SPU und den ONSD, zusätzlich erfolgte die Funduskopie zum Ausschluss oder Bestätigung einer Stauungspapille. Die Sensitivität und Spezifität der drei Untersuchungen zur Ermittlung einer Erhöhung des intrakraniellen Drucks auf über 20 cm H2O wurden berechnet. Die Sensitivität der Funduskopie betrug 87,5% bei einer Spezifität von 25%. Im Vergleich hierzu betrug die Sensitivität der beiden Sonographieverfahren in Kombination 80% bei einer Spezifität von 100%. Die Ergebnisse der Studie zeigen, dass es zusätzlich zur Funduskopie sensitive und spezifische klinische Untersuchungsmethoden gibt zur Detektion eines erhöhten intrakraniellen Drucks im Alltag. Die Messung des ONSD und der SPU können nützliche Alternativen zur Funduskopie darstellen, optimalerweise in Kombination.
Hintergrund und Fragestellung: Nomogramme können durch individuelle Prognoseberechnungen Patienten helfen, Therapieentscheidungen zu treffen sowie ihre Gesundheitskompetenz durch ein besseres Krankheitsverständnis zu fördern. Nomogramme sind graphische Darstellungen von Regressionsgleichungen, mit denen sich verschiedene Endpunkte berechnen lassen. In der vorliegenden Arbeit sollen neben Hintergrundwissen zu Nomogrammen auch Gütekriterien für eine kritische Nutzung von Nomogrammen im klinischen Alltag sowie eine Übersicht empfehlenswerter Nomogramme vorgestellt werden.
Methode: Es fand eine systematische Literaturrecherche in der Datenbank Medline im Zeitraum von September 2014 bis Mai 2016 statt. Gesucht wurde auf englischer Sprache nach Nomogrammen in der Urologie anhand des Suchterms „Nomogram AND urology“. Einschlusskriterien für die Betrachtung waren vorhandener Abstract sowie englische oder deutsche Sprache. Die Bewertung der Nomogramme fand unter folgenden Gesichtspunkten statt: Fallzahl, Aktualität, Validierung, Gütekriterien sowie klinische Anwendung.
Ergebnis: Insgesamt hat die Literaturrecherche 311 Nomogramme in der Urologie zu verschiedenen Erkrankungen und Organsystemen identifizieren können. Für die Bereiche Urodynamik, Nebenniere, entzündliche Erkrankungen der Harnwege, Penis, Urolithiasis, Urothelkarzinome, Nierenkarzinome und Prostatakarzinome sind 122 extern validierte Nomogramme gefunden worden. Dabei wurde die fehlerhafte Nutzung des Nomogrammbegriffs deutlich, zumal Tabellen, Perzentilenkurven und Diagramme als solche beschrieben wurden. Hinsichtlich der Qualität der publizierten Modelle gibt es noch Verbesserungsbedarf. So sind viele publizierte Modelle nicht extern validiert oder nur einem Datensplitting unterzogen worden, was die Übertragbarkeit in fremde Populationen behindert. Weiterhin bestehen Mängel bei der Konzeption der Studien zur Nomogramm-Entwicklung, bei der Rekrutierung der Patienten und bei der Datenqualität.
Fazit: Die vorliegende Arbeit identifiziert die klinisch sinnvollsten und erprobten Nomogramme für den Einsatz in der Praxis (siehe Tabellen 1-9). Durch Fortschritte in Medizin und Technik können Nomogramme mit Hilfe von hochqualitativen Daten sowie einheitlicher Qualitätsstandards zur Entwicklung und Etablierung dieser in Zukunft einen wertvollen, klinisch sinnvollen Einsatz zur Verbesserung der Therapieentscheidung bei Patienten erfahren. Neue Biomarker, insbesondere auf Grundlage von Genomanalysen, sowie optimierte bildgebende Verfahren könnten zukünftig Bestandteil von Nomogrammen werden. Weiterhin sollte die Evaluation des klinischen Nutzens der Nomogramme sowie ein Vergleich der Modelle unter gleichen Bedingungen in Zukunft erfolgen, bevor diese im klinischen Alltag Anwendung finden.
Enhanced aridification of Central Asia driven by the combined effects of orogenic surface uplift, Paratethys retreat, changes in atmospheric moisture transport and global cooling is one of the most prominent Cenozoic climate change events of the Northern Hemisphere. Deciphering regional long-term patterns of Central Asian hydrology is, therefore, a key element in understanding the role of Northern Hemisphere mid-latitude drying in the global hydrological system. Pedogenic carbonates record information of the paleoecosystem, the paleogeography, hydrology, tectonic and climatic conditions as well as the sedimentary regime during the time in which they formed. The calcrete-bearing paleosols in continental sedimentary basins yield the possibility for understanding these processes, mechanisms and controlling factors. This study characterizes long-term paleoenvironmental conditions between the late Oligocene and early Miocene in SE Kazakhstan based on stable isotopes, elemental geochemistry and laser ablation U-Pb geochronology from alluvial, fluvial and pedogenic deposits. The main topics addressed in this thesis are paleosol and calcrete formation processes in the light of geochemistry, tectonic and climatic influences on both, as well as the paleoenvironmental and hydrological conditions which led to these processes.
The sedimentological succession of the 14 km-long Kendyrlisai Valley in the Ili Basin, SE Kazakhstan in Central Asia, yields exceptional well-developed paleosols which provide an insight into the relationship between tectonics and sedimentation as well as soil formation processes. The valley accommodates more than 600 m of siliciclastic sediments deposited in a fluvial to alluvial environment in the lower part which grade into lacustrine to fluvial deposition with minor gypsum accumulation in the upper part. The yet undated sediments of Kendyrlisai Valley are compared with a biostratigraphically dated and well described succession in an adjacent location. The correlation of the two successions revealed a proximal-distal relation with the Kendyrlisai Valley succession represents the basin margin position with coarse alluvial-fluvial sediments. The calcrete nodules in Kendyrlisai Valley paleosols give the opportunity of U-Pb dating with LA-ICP-MS which uses the variation in both U and Pb to directly date the calcite. The U-Pb dating revealed a late Oligocene-early Miocene age for the investigated section. The analysis of facies and geochemistry of the paleosol profiles in Kendyrlisai Valley gives insight into calcretization stages, pedogenic processes and landscape stability. Kendyrlisai Valley paleosols show a low to moderate weathering intensity with the highest weathering intensity between 0.5 and 0.8 m depth in the paleosol profile. The comparison between acid leachable and non-acid leachable fraction indicate an incorporation of Ca and Ba in incipient calcrete calcite, whereas clay minerals adsorb Mg and Sr. During the evolution from early to more mature calcrete stages, i.e. calcrete nodules, Ba is lost by dissolution and subsequently adsorbed on clay minerals. The nodules consist almost exclusively of pure calcium carbonate with rare substitution of Ca by Mg indicating a constant supply of Ca by weathering and solution during calcrete formation. The occurrence of Mg-bearing clay minerals indicates high evaporative conditions with alkaline waters (pH >9).
Sampled calcrete nodule cross sections reveal more than one weathering and calcrete formation phase with a higher supply of Ca, Mg and Fe during early phases. The organic and inorganic carbon isotope composition of the calcrete nodules reflects C3 metabolism under occasionally moisture stress, resulting in higher δ13C values and lower respiration rates. This study also presents calculated atmospheric pCO2 values from the calcrete nodules with the equation from Cerling et al. (1999). The average calculated CO2 concentration for Kendyrlisai Valley paleosols is 313 ± 110 ppmv. The comparison with data from other studies during the late Oligocene–early Miocene reveal lower values for Kendyrlisai Valley paleosols, which may stem from an underestimation of the plant-respired CO2.
The knowledge of the variability within the paleosols and calcrete nodules enables a reliable paleoclimatic interpretation for the Kendyrlisai Valley and beyond the Ili Basin. Sedimentary facies and geochemical weathering indices suggest an increased surface and groundwater discharge fed by orographically-enhanced precipitation in the Tien Shan hinterland. In contrast, pedogenic stable isotope data and elevated rates of Mg fixation in clay minerals mirror enhanced rates of evaporation in the vadose zone due to protracted aridification. This study posits that pronounced surface uplift of the Tien Shan Mountains during the Oligocene–Miocene transition promoted regionally increased orographic precipitation and the development of fluvial discharge systems.
The comparison with other studies in adjacent basins creates a precipitation pattern for Central Asia during the late Oligocene to early Miocene. The westerlies supplied Central Asia with atmospheric moisture from the West, possibly from the Eastern Paratethys. The uplifting Tien Shan Mountain ranges captured this moisture on their luv-side, resulting in a pronounced orographic rainfall in the adjacent Ili and Issyk Kul Basins. The Tarim Basin and the Valley of Lakes experienced drier conditions due to the rain shadow effect on the lee-side of the Tien Shan and Altai Mountains. In course of this findings, the thesis highlights the crucial role the Tien Shan Mountains play and had been playing in former times as an orographic barrier for the distribution of atmospheric moisture across Central Asia.
Aim: Long noncoding RNAs (lncRNAs) belong to the interface of epigenetics and exhibit diverse functions. Their features depend on their sequence, genomic location and tertiary structure. The aim was to identify novel lncRNAs and characterise their physiological functions and mechanisms in endothelial cells. Three different approaches were performed:
The hypothesis that pseudogene-annotated lncRNA NONHSAT073641 regulates the expression of their parental gene platelet activating factor acetylhydrolase 1b regulatory subunit 1 (PAFAH1B1) was examined.
The physiological functions and in vivo relevance of most lncRNAs are still unknown, therefore a part of this work aimed to identify lncRNAs in response to a pathophysiological stimulus (high amplitude stretch) in endothelial cells.
The long intergenic noncoding RNA antisense to S1PR1 (LISPR1) gene, is located within the promotor of sphingosine-1-phosphate receptor 1 (S1PR1) and shares a part of the promotor region. This study examined additionally the hypothesis that LISPR1 controls the S1PR1 expression in endothelial cells.
Methods: The angiogenic functions of NONHSAT073641 and LISPR1 were examined with spheroid-outgrowth and scratch wound assays. Furthermore, stretch experiments were performed in order to identify differently expressed lncRNAs in human umbilical vein endothelial cells (HUVECs). In addition, the in vivo relevance of both lncRNAs was examined in samples from pulmonary arterial hypertension patients. Knockdown (e.g. LNA GapmeRs), knockout (CRISPR/ Cas9) and overexpression experiments (e.g. CRISPR activation) were performed to analyse target genes. The molecular mechanism of LISPR1 was investigated with RNA and Chromatin immunoprecipitation.
Results: NONHSAT073641 and PAFAH1B1 exhibited angiogenic function in endothelial cells. It could be observed that NONHSAT073641 is not regulating the expression of PAFAH1B1. The pro-angiogenic feature of PAFAH1B1 might be attributed to the target gene matrix Gla protein (MGP). NONHSAT073641 and PAFAH1B1 were significantly induced in CTEPH samples and might be important in the development of this disease. It could be speculated that NONHSAT073641 is regulating the expression of the cell-cycle regulator BCL2L11 as has been investigated in mice.
LISPR1 is a cis-acting lncRNA which maintains S1PR1 gene transcription by intercepting the transcriptional repressor ZNF354C and enabling Polymerase II (PolII) to bind. ZNF354C regulates S1PR1 expression in HUVECs. However, the role of ZNF354C in pulmonary arterial hypertension (PAH) is unknown. LISPR1 and S1P1 receptor were both significantly depleted in COPD samples. It can be assumed that due to higher S1P production, the signalling is attenuated through reduction of the lncRNA LIPSR1 and thus the receptor S1P1.
The stretch experiments present a possible in vitro model in order to mimic the condition of endothelial cells during high blood pressure, such as in PAH. Referring to published data, it could be confirmed that stretching of endothelial cells alters the gene expression, which is on the other hand linked to cardiovascular disease. In cardiovascular disease mechanical stretch altered genes, which are participating in the vascular remodelling process. The role of differently expressed lncRNAs (TGFβ2-AS1, CTD-2033D15.2, INHBA-AS1, RP11-393I2.4, TAPT1-AS1, TPM1-AS1, CFLAR-AS1 and HIF1α-AS2) upon mechanical stretch is yet not clarified.
Conclusion: NONHSAT073641 and LISPR1 are important for the endothelial angiogenic function. Both lncRNAs were deregulated in PAH samples. The pathophysiological stimulus had an impact on the expression of different lncRNAs (e.g. TGFβ2-AS1) and pathways (e.g. TGF-β) in endothelial cells.
Metabolic engineering can serve to convert microorganisms to
microbial cell factories with the goal of producing various chemicals. Commonly used strategies to modify metabolic pathways include deletions and overexpression of genes, as well as the introduction of heterologous genes or genes which have been optimized for the host organism or for a reaction of interest. Aside from these classic metabolic engineering strategies, researchers have also implemented pathway compartmentalization strategies, which mimic nature’s strategies of colocalizing enzymes for pathway optimization.
In this thesis, classic metabolic engineering strategies were combined with pathway compartmentalization strategies. For pathway compartmentalization, mitochondria and peroxisomes were harnessed, and additionally a new strategy to create artificial subcellular organelles was evaluated. In the latter approach, the so-called Zera peptide was fused to the enzymes of interest. Zera consists of the first 113 amino acids of the plant storage protein γ-Zein (Zea mays). Natively, plant storage proteins accumulate in endoplasmic reticulum (ER)-derived vesicles in plant seeds and serve as an amino acid source for the germinating plant. In this thesis, it was shown that Zera also induces the formation of artificial, ER-derived vesicles in Saccharomyces cerevisiae. Furthermore, it was shown that Zera fusion enzymes remained active, albeit with sometimes reduced activity.
In line with the goal of compartmentalizing pathways in these artificial, Zera-induced vesicles, a new tool was developed to determine the pH in the ER of S. cerevisiae and in the ER-derived vesicles. pHluorin, a pH-sensitive green fluorescent protein (GFP) variant, is commonly used to analyze the cytosolic pH or the pH of subcellular organelles. In this thesis, it was shown that pHluorin has very low fluorescence intensity and pH sensitivity in the ER and in Zera-induced ER-derived vesicles. Therefore, a superfolder variant of pHluorin was developed which allows reliable pH measurements in these compartments and can be used to analyze whether the organellar or vesicular pH suits a pathway of interest....
A necessary requirement for a pharmacological effect is that a drug molecule tightly interacts with its disease relevant target molecule in the patient. Kinases are regulatory, signal transmitting enzymes and are a large protein family that belongs to the most frequent targets of pharmaceutical industry, as deregulation of kinases has been associated with the development of a variety of diseases, including cancer. In drug discovery, equilibrium binding metrics such as the affinity (Ki, KD) or potency (IC50, EC50) are usually applied for the systematic profiling for potent and selective drug candidates. In recent years, dynamic binding parameters, the drugs association (kon) and dissociation (koff) rates for desired primary-targets and undesired off-targets, were discussed to be better predictors than steady-state affinity per se (KD = koff / kon) for the onset and duration of the drug-target complex in the open in vivo environment and thereby for the therapeutic effect and safety of the drug. It is yet unclear whether and when the binding kinetics parameters can influence drug action in the complex context of pharmacokinetics and pharmacodynamics and how the kinetic rate constants can be optimized rationally. One major obstacle for providing proof for the hypothesis that drug binding kinetics is of importance for drug action is the generation of large and comparable binding kinetic datasets.
The aim of this thesis was the comprehensive analysis of the binding kinetic and affinity parameters of a diverse spectrum of 270 small-molecule kinase inhibitors against a panel of pharmacologically relevant kinases to study the role played by binding kinetics for drug discovery: The generated dataset was utilized to assess the effect of chemical properties on drug binding kinetics, and to evaluate the impact of kinetic rate constants on the success of compounds in the drug discovery pipeline.
Large scale profiling was made possible by a recently developed “kinetic Probe Competition Assay” (kPCA), whose evaluation is based on Motulsky’s and Mahan’s “kinetics of competitive binding” theory. Monte Carlo analyses performed in this dissertation widened the theoretical knowledge of this theory, provided new insights into its limitations and allowed to derive recommendations about how to best design assays. It was demonstrated that kPCA is indeed high-throughput compatible and that it is comparable to other biochemical and biophysical assay formats in terms of precision and accuracy.
Multivariable linear regression for the description of the determined kinase inhibitors’ target binding characteristics (kon or koff or KD) using molecular properties and/or particular kinase-inhibitor interactions as descriptors supported the assumption that molecular properties of compounds might affect binding kinetics, generated new hypothesis about molecular determinants influencing binding kinetic parameters and provided a rational basis for following structure-kinetic relationship studies. Remarkably, the binding kinetic rate constants were better described by the established models than binding affinities.
Interestingly, the systematic, quantitative analysis of kinase inhibitors’ target binding kinetics indicated that a slow dissociation rate for the main target is a feature which is more frequently observed in inhibitors that reached approval or late stage clinical testing than in earlier phases of clinical development. In addition, it was demonstrated that binding kinetics of kinase inhibitors is a better predictor for the time course of target engagement in cells as compared to affinity per se. Furthermore, in some study cases simulations using a standard pharmacokinetics model and a modified model considering the inhibitors binding kinetics lead to different in vivo kinase occupancy time profiles. It was illustrated by simulations how the concept of kinetic selectivity can be applied to turn an unselective compound in equilibrium conditions into a more selective compound in the open in vivo situation, where the thermodynamic equilibrium of drug-target binding is not necessarily reached.
Thus the generated data and models provide evidence for the importance of binding kinetics in drug discovery and represent a valuable resource for future studies in this field.
Das Ziel dieser Dissertation war es die biologische Rolle der Ubiquitinierung und die Bedeutung für Alterungsprozesse im filamentösen Ascomyceten Podospora anserina zu untersuchen. Folgende Ergebnisse wurden dabei erzielt:
1. Ubiquitinierte Proteine wurden nachgewiesen und die Deubiquitinierung von Proteinen konnte durch den Einsatz der Inhibitoren Urea, PR-619 und PIC erfolgreich inhibiert werden, was die Anwesenheit aktiver Deubiquitinasen in P. anserina beweist. Zudem wurden erstmalig ubiquitinierte Proteine in P. anserina unter den zur Aufreinigung
gewählten Bedingungen über die Technik der LC-MS/MS identifiziert.
2. Insgesamt wurden 1745 ubiquitinierte Proteine in P. anserina identifiziert, was ca. 16,4 % des gesamten Proteoms darstellt. Somit wurde erstmalig das Ubiquitinom des Ascomyceten P. anserina charakterisiert, welches Proteine aus allen zellulären Kompartimenten enthält.
3. Die erste im Rahmen dieser Arbeit durchgeführte umfassende Studie altersabhängiger Veränderungen im Ubiquitinom eines Organismus zeigt eine Herabregulation der Ubiquitinierung im Alter in der gesamten Zelle. Dem gegenüber steigt die Ubiquitinierung an den Mitochondrien leicht an und unterstreicht die Rolle einer mitochondrialen Qualitätskontrolle durch diesen Prozess.
4. Die Untersuchung der am Ubiquitinierungsprozess beteiligten E3-Ligase PaMUS10 zeigt, dass es sich bei Mus10 um ein essentielles Gen in P. anserina handelt, da die Deletion zu starken mitochondrialen Beeinträchtigungen und dem sofortigen Absterben des Organismus direkt nach der Keimung führt.
5. Zur weiteren Untersuchung von PaMus10 wurde erstmalig und vollständig ein Hygromycin-basiertes „Knockdown“-System in P. anserina etabliert, was die detaillierte Untersuchung eines essentiellen Gens in diesem Organismus ermöglichte. Durch dessen Einsatz konnte eine reduzierte Fertilität, eine verkürzte Lebensspanne sowie Veränderungen der mitochondrialen Morphologie und Funktion als direkte Folge einer PaMus10-Herabregulation nachgewiesen werden.
6. PaMUS10 ist vorwiegend cytosolisch lokalisiert wird aber unter oxidativem Stress oder in gealterten Kulturen an die Mitochondrien rekrutiert, was einen vergleichbaren Mechanismus zur menschlichen E3-Ligase PARKIN darstellt.
7. Der Verlust von PaMUS10 verursacht ein Präseneszenzsyndrom und führt zum vorzeitigen Absterben des Organismus, wohingegen die zusätzliche Expression von PaMus10::Gfp offenbar positive Effekte nach sich zieht, da hier eine deutliche Verlängerung der Lebensspanne beobachtet wurde.
8. Der Vergleich der beiden Ubiquitinome von ∆PaMus10/PaMus10 und Wthph1/2 zeigt eine massive Reduzierung der globalen Ubiquitinierung, welche offenbar durch das Fehlen von PaMUS10 ausgelöst wird und damit dessen Funktion als E3-Ligase untermauert.
9. Im Rahmen der hier durchgeführten Substratanalyse wurden insgesamt 131 Proteine identifiziert, von denen ca. 20 % dem Mitochondrium zugeordnet werden können. Aufgrund der diversen biologischen Prozesse und Funktionen dieser Substrate ist PaMUS10 sowohl in die Ubiquitinierung cytosolischer als auch mitochondrialer Proteine involviert und greift womöglich sogar als zentraler Schalter in die gesamte zelluläre Homöostase ein.
Aufgrund der nicht unerheblichen Zahl der identifizierten ubiquitinierten Proteine (Ubiquitinom) und den fatalen Auswirkungen, die der Verlust einer E3-Ligase in diesem Organismus nach sich zieht, lässt sich folgende grundlegende Erkenntnis formulieren:
Die Ubiquitinierung spielt in P. anserina eine bedeutende Rolle zur Aufrechterhaltung zellulärer Prozesse insbesondere der mitochondrialen Homöostase und beeinflusst dadurch positiv die Entwicklung und Alterung in diesem Organismus.
My study examined MMA training, and thereby the ‘back region’ of MMA, where the ‘everyday life’ of MMA takes place. I enquired into how MMA training corresponds with MMA’s self-description, namely the somehow self-contradicting notion that MMA fights would be dangerous combative goings-on of approximately real fighting, but that MMA fighters would be able to approach these incalculable and uncontrolla-ble combative dangers as calculable and controllable risks.235 Conducting an ethnog-raphy in which I focused on the combination of participation and observation, I stud-ied how the specific interaction organisations of the three core training practices of MMA training provide the training students with specific combative experiences and how they thereby construct the social reality that is MMA training....
Protein biosynthesis is a conserved process, essential for life. Proteins are assembled from single amino acids according to their genetic blueprint in the form of a messenger ribonucleic acid (mRNA). Peptide bond formation is catalyzed by ancient ribonucleic acid (RNA) residues within the supramolecular ribosomal complex, which is organized in two dynamic subunits (Ramakrishnan, 2014). Each subunit comprises large ribosomal RNA (rRNA) molecules and several dozens of peripheral proteins. mRNA translation has been divided into three phases, namely translation initiation, elongation and termination in biochemistry textbooks. During initiation, the ribosomal subunits assemble into a functional ribosome on an activated mRNA and acquire the first transfer RNA (tRNA), an adapter between the start codon on the mRNA and the N-terminal methionine of the protein (Hinnebusch and Lorsch, 2012). During elongation, the ribosome translocates along the mRNA exposing one codon after the other, and amino acids are delivered to the ribosome by the respective tRNAs, and attached to the nascent polypeptide chain. During termination, the polypeptide is released and the ribosome remains loaded with mRNA and tRNA at the end of the open reading frame for the translated gene (Hellen, 2018). Bacterial ribosomes are subsequently recycled by a specific ribosome recycling factor and the small ribosomal subunit is simultaneously consigned to initiation factors for a next round of translation – rendering bacterial translation as a cyclic process with an additional ribosome recycling phase. However, the process of ribosome recycling remained enigmatic in Eukarya and Archaea until the simultaneous discovery of the twin-ATPase ABCE1 as the major ribosome recycling factor. Strikingly, ABCE1 has initially been shown to participate in translation initiation (Nürenberg and Tampé, 2013). Thus, closing the translation cycle by revealing the detailed molecular mechanism of ABCE1 and its role for translation initiation are the two goals of this research.
Beyond the plenitude of well-studied translational GTPases, ABCE1 is the only essential factor energized by ATP, delivering the energy for ribosome splitting via two nucleotide-binding sites. Here, I define how allosterically coupled ATP binding and hydrolysis events in ABCE1 empower ribosome recycling. ATP occlusion in the low-turnover control site II promotes formation of the pre-splitting complex and facilitates ATP engagement in the high-turnover site I, which in turn drives the structural re- organization required for ribosome splitting. ATP hydrolysis and ensuing release of ABCE1 from the small subunit terminate the post-splitting complex. Thus, ABCE1 runs through an allosterically coupled cycle of closure and opening at both sites consistent with a processive clamp model. This study delineates the inner mechanics of ABCE1 and reveals why various ABCE1 mutants lead to defects in cell homeostasis, growth, and differentiation (Nürenberg-Goloub et al., 2018).
Additionally, a high-resolution cryo-electron microscopy (EM) structure of the archaeal post-splitting complex was obtained, revealing a central macromolecular assembly at the crossover of ribosome recycling and translation initiation. Conserved interactions between ABCE1 and the small ribosomal subunit resemble the eukaryotic complex (Heuer et al., 2017). The conformational state of ABCE1 at the post-splitting complex confirms the molecular mechanism of ribosome recycling uncovered in this study. Moving further along the reaction coordinate of cellular translation, I reconstitute the complete archaeal translation initiation pathway and show that essential archaeal initiation factors are recruited to the post-splitting complex by biochemical methods and cryo-EM structures at intermediate resolution. Thus, the archaeal translation cycle is closed, following its bacterial model and paving the way for a deeper understanding of protein biosynthesis.
Die Arbeit thematisiert weibliche Beiträge der graphischen Gattung der humorvollen und satirischen Zeichnung im "Golden Age" der Karikatur in Großbritannien. Ausgehend von den Begünstigungen und relativen Freiheiten weiblicher Bildproduktion in der vorviktorianischen Zeit, setzt die Untersuchung die zeichnerische Aktivität von Frauen in den Kontext von weiblicher "curiosity" und weiblichem "wit" in der Wahrnehmung neuer Interessenlagen der Aufklärung.
Object recognition is such an everyday task it seems almost mundane. We look at the spaces around us and name things seemingly effortlessly. Yet understanding how the process of object recognition unfolds is a great challenge to vision science. Models derived from abstract stimuli have little predictive power for the way people explore "naturalistic" scenes and the objects in them. Naturalistic here refers to unaltered photographs of real scenes. This thesis therefore focusses on the process of recognition of the objects in such naturalistic scenes. People can, for instance, find objects in scenes much more efficiently than models derived from abstract stimuli would predict. To explain this kind of behavior, we describe scenes not solely in terms of physical characteristics (colors, contrasts, lines, orientations, etc.) but by the meaning of the whole scene (kitchen, street, bathroom, etc.) and of the objects within the scene (oven, fire hydrant, soap, etc.). Object recognition now refers to the process of the visual system assigning meaning to the object.
The relationship between objects in a naturalistic scene is far from random. Objects do not typically float in mid-air and cannot take up the same physical space. Moreover, certain scenes typically contain certain objects. A fire hydrant in the kitchen would seem like an anomaly to the average observer. These "rules" can be described as the "grammar" of the scene. Scene grammar is involved in multiple aspects of scene- and object perception. There is, for instance, evidence that overall scene category influences identification of individual objects. Typically, experiments that directly target object recognition do not involve eye movements and studies that involve eye movements are not directly aimed at object recognition, but at gaze allocation. But eye movements are abundant in everyday life, they happen roughly 4 times per second. Here we therefore present two studies that use eye movements to investigate when object recognition takes place while people move their eyes from object to object in a scene. The third study is aimed at the application of novel methods for analyzing data from combined eye movement and neurophysiology (EEG) measurements.
One way to study object perception is to violate the grammar of a scene by placing an object in a scene it does not typically occur in and measuring how long people look at the so-called semantic inconsistency, compared to an object that one would expect in the given scene. Typically, people look at semantic inconsistencies longer and more often, signaling that it requires extra processing. In Study 1 we make use of this behavior to ask whether object recognition still happens when it is not necessary for the task. We designed a search task that made it unnecessary to register object identities. Still, participants looked at the inconsistent objects longer than consistent objects, signaling they did indeed process object and scene identities. Interestingly, the inconsistent objects were not remembered better than the consistent ones. We conclude that object and scene identities (their semantics) are processed in an obligatory fashion; when people are involved in a task that does not require it. In Study 2, we investigate more closely when the first signs of object semantic processing are visible while people make eye movements.
Although the finding that semantic inconsistencies are looked at longer and more often has been replicated often, many of these replications look at gaze duration over a whole trial. The question when during a trial differences between consistencies occur, has yielded mixed results. Some studies only report effects of semantic consistency that accumulate over whole trials, whereas others report influences already on the duration of the very first fixations on inconsistent objects. In study 2 we argue that prior studies reporting first fixation duration may have suffered from methodological shortcomings, such as low trial- and sample sizes, in addition to the use of non-robust statistics and data descriptions. We show that a subset of fixations may be influenced more than others (as is indicated by more skewed fixation duration distributions). Further analyses show that the relationship between the effect of object semantics on fixation durations and its effect on oft replicated cumulative measures is not straightforward (fixation duration distributions do not predict dwell effects) but the effect on both measures may be related in a different way. Possibly, the processing of object meaning unfolds over multiple fixations, only when one fixation does not suffice. However, it would be very valuable to be able to study how processing continues, after a fixation ends.
Study 3 aims to make such a measure possible by combining EEG recordings with eye tracking measurements. Difficulties in analyzing eye tracking–EEG data exist because neural responses vary with different eye movements characteristics. Moreover, fixations follow one another in short succession, causing neural responses to each fixation to overlap in time. These issues make the well-established approach of averaging single trial EEG data into ERPs problematic. As an alternative, we propose the use of multiple regression, explicitly modelling both temporal overlap and eye movement parameters. In Study 3 we show that such a method successfully estimates the influence of covariates it is meant to control for. Moreover, we discuss and explore what additional covariates may be modeled and in what way, in order to obtain confound-free estimates of EEG differences between conditions. One important finding is that stimulus properties of physically variable stimuli such as complex scenes, can influence EEG signals and deserve close consideration during experimental design or modelling efforts. Overall, the method compares favorably to averaging methods.
From the studies in this thesis, we directly learn that object recognition is a process that happens in an obligatory fashion, when the task does not require it. We also learn that only a subset of first fixations to objects are affected by the processing of object meaning and its fit to its surroundings. Comparison between first fixation and first dwell effects suggest that, in active vision, object semantics processing sometimes unfolds over multiple fixations. And finally, we learn that regression-based methods for combined eye tracking-EEG analysis provide a plausible way forward for investigating how object recognition unfolds in active vision.
The present study aimed to assess the tissue response to the SYMBIOS® resorbable collagen membrane SR, which is derived from bovine Achilles tendon, and compare it to the physiological wound healing of a sham operation as a control.
An ex vivo analysis was performed using injectable platelet-rich fibrin (i-PRF), that is gained by the centrifugation of human venous blood and contains fibrin, leukocytes and platelets, to elucidate the membrane permeability and interactions with human cells and plasma proteins. In the in vivo study, a subcutaneous implantation model was established in Wistar rats to evaluate the cellular reactions for up to 30 days after membrane implantation. Histochemical, immunohistochemical and histomorphometric analyses were performed to assess the cellular inflammatory response, vascularization pattern and cell infiltration capacity.
In the ex vivo study, i-PRF components including fibrin, leukocytes and platelets penetrated the membrane after just 15 minutes. Within the observation period, the cellular reaction in the early phase, which included the first 3 days, produced only mononuclear cells. From 10 to 30 days , the formation of multinucleated giant cells (MNGCs) was induced by the collagen membrane. CD-68 positive cells (macrophages) occurred in a high number on day 3, and the number decreased over time up to day 30. Along with the reduction in the number of CD-68 positive cells, the number of MNGCs increased significantly. The presence of MNGCs was accompanied by significantly increased vascularization within the central region of the membrane, and only mononuclear cells (MNCs) did not produce vascularization. In contrast, the accumulated MNGCs were located on the membrane surface. The control group reflected the physiological process of wound healing, as MNGCs did not form over the 30 day period, and a significantly lower level of vascularization was observed compared with the test group.
This finding showed dynamic changes in the cellular reaction, which indicated a relationship between macrophage fusion and MNGC formation, and vascularization of the collagen membrane is circumstantial evidence of a reaction to a foreign body. However, the collagen membrane was able to maintain its structure and integrity over time, showing no signs of premature breakdown and disintegration due to the specific porosity of its membrane structure.
Therefore, we questioned whether the biomaterial-induced formation of MNGCs should be accepted as a biomaterial-induced cellular reaction that is able to restore vascularization or as an adverse reaction. Therefore, extensive preclinical and clinical studies are needed to investigate the type of MNGCs that form in response to the membrane material studied here.
Cancer is one of the leading causes of death across all countries and its diagnosis still yields fear for the affected patient. Although treatment of cancer has made marvelous progress compared to the agents available thirty years ago, a cure for cancer, however, is still a distant prospect. Modern therapy still is a burden for many patients due to heavy side effects. With the development of agents targeting specific molecular targets on cancer cells, a new field of cancer therapy was opened and a small success story in the history of cancer began.
Aurora kinases represent a relatively new target in cancer therapy. The kinase is a essential part of mitosis and cell cycle progression and its overexpression has been shown to be related to many kinds of malignancies. Allosteric inhibition of a kinase is an increasing pre-clinical approach not yet established in the treatment of patients. In this thesis, we combine allostery with another innovative approach that is drug repurposing. If repurposed, a drug can be permitted to fast track drug admission to clinical trials.
I set up a screening of 1280 FDA approved drugs to identify small molecule compounds that affect the binding of Aurora kinase A and its main physiologic binding partner, TPX2. Further, I characterized the positive hits in vitro for their capabilities to displace TPX2 from Aurora A, to inhibit Aurora kinase activity, to thermally stabilize the protein and performed assays to determine their dissociation constant. Last but not least, I tested the compounds in cells for their effect on the cell viability and cell cycle via flow cytometry. Comparing the hit-compounds with controls I found that ATP-competitive AurA inhibitor MLN 8237 strongly displaces the interaction of Aurora A with TPX2.
Summarized, we identified eight hit compounds allosterically affecting Aurora A, but no compound proved to be active in all assays. Just one compound, PS 731, identified in another screening performed by our group and further characterized in this thesis remains interesting, especially when put in context with recent publications released in the time between the start of experiments for this thesis and its finalization.
Die Tuberkulose ist eine der bedeutendsten Infektionskrankheiten weltweit. In Deutschland spielt sie eine untergeordnete Rolle und betrifft hauptsächlich Risikogruppen. Bisher wurde noch keine mehrere Jahre umfassende Arbeit zur Epidemiologie der Tuberkulose anhand molekularbiologischer Faktoren in einer deutschen Stadt veröffentlicht. In dieser Arbeit wurden mittels 24-loci MIRU-VNTR und Spoligotypisierung die Mykobakterienstämme von Patienten in Frankfurt am Main aus dem Zeitraum von 2008 bis 2016 genetisch typisiert und aus Fällen mit übereinstimmendem DNA-Fingerabdruck bestehende molekulare Cluster identifiziert, um in Zusammenschau mit epidemiologischen Patientendaten die Übertragungswege in Frankfurt am Main besser zu verstehen. Dabei wurden mittels logistischer Regression Risikofaktoren für Clusterzugehörigkeit identifiziert, einzelne Cluster auf epidemiologische Plausibilität hin untersucht und die Bedeutung der Tuberkulose vor dem Hintergrund zunehmender Migration beschrieben.
Insgesamt wurden im Studienzeitraum 61 molekulare Cluster identifiziert. Der Clusteranteil (28,6%) und der Anteil rezenter Übertragung (18,7%) lagen in der Größenordnung anderer Niedriginzidenzländer. Es dominierten kleine Cluster, nur vereinzelt kam es zu Infektionsketten mit mehr als 3 Fällen. Obwohl 81% der Patienten aus dem Ausland stammten und nur 19% aus Deutschland, hatten Migranten ein signifikant niedrigeres Risiko, einem Cluster anzugehören als Deutsche. Unter den Clustern bestanden 42,6% sowohl aus Patienten deutscher als auch Patienten nichtdeutscher Herkunft. Im Ausland geborene Patienten in gemischten Clustern lebten zum Diagnosezeitpunkt durchschnittlich bereits 10 Jahre in Deutschland und stammten mehrheitlich aus europäischen Ländern. Eine TB-Übertragung von kürzlich eingewanderten Patienten auf einheimische Bürger fand in begrenztem Umfang statt, begründet aber keine Ängste vor einer erhöhten Gefährdung der in Deutschland geborenen Bevölkerung im Hinblick auf die Tuberkulose in Frankfurt am Main.
Nur 9,8% der molekularen Cluster konnten epidemiologisch bestätigt werden. Infektionswege, die über das familiäre Umfeld oder bestimmte Risikomilieus (Drogen, Obdachlosigkeit) hinausgehen, ließen sich anamnestisch nur schwer erfassen. Männer, in Deutschland geborene Personen und iv-drogenabhängige Personen wiesen ein erhöhtes Risiko auf, sich vor Ort mit Tuberkulose zu infizieren oder die Erkrankung auf andere zu übertragen. Dies trifft vermutlich auch auf andere deutsche Großstädte zu und betont die Notwendigkeit einer Integration von iv-Drogenabhängigen in die medizinische Regelversorgung und die Bedeutung einer Zusammenarbeit von öffentlichem Gesundheitsdienst und entsprechenden Hilfseinrichtungen.
Es ist von einer Überschätzung des Clusteranteils mittels 24-loci MIRU-VNTR auszugehen, weshalb für zukünftige Arbeiten die feinere Typisierung und somit eine zuverlässigere Identifikation von Clustern mittels Whole Genome Sequencing wünschenswert ist. Für die bundesweite Verbesserung der TB-Kontrolle ist weiterhin eine enge Zusammenarbeit zwischen den Gesundheitsämtern erforderlich. Um zu untersuchen, wie sich die TB-Epidemiologie von Frankfurt am Main im bundesweiten Vergleich darstellt, sind ähnlich angelegte Arbeiten aus anderen deutschen Großstädten notwendig.
The subject of this dissertation is the aesthetic and political significance of photography for Bertolt Brecht's theatrical work and writing. Since the investigation of the gestus concept from the perspective of the politics of images—which draws attention to the fact that every image has a memory, mostly a political one—photography stands out and reveals the precise moment of the interruption of the dramatic action and, thereby, holds a significant gestus. The dissertation highlights the importance of photography’s examination for Brecht, besides featuring the extent to which his theatre responded to and dealt with the technical condition of its own time. If a gestus is recognisable as an interruption, through the observation of the detail, two different gestus appear: the gestus of the scene and the gestus of the photograph. In other words, the actor and photographer produce a double gestus with different levels of interpretation. The comparison between such a static photo and the notion of interruption in Bertolt Brecht’s works opens new grounds in Brechtian studies, and even in more general theatre studies.
Role of npas4l and Hif pathway in endothelial cell specification and specialization in vertebrates

Cardiovascular development requires two main steps, vasculogenesis and angiogenesis. During vasculogenesis, angioblasts, the precursors of endothelial cells (ECs), specify from the mesoderm and coalesce to form the axial vessels of the vertebrate embryo. Many questions regarding the transcriptional waves initiating and sustaining angioblast specification are still unanswered. The identity of cloche, a gene essential for EC differentiation in zebrafish, was only recently discovered by our group, and very little is known about its upstream regulators or its molecular mechanism of action. I described the molecular players involved in orchestrating npas4l expression, upstream of angioblast specification. By using genetic models and chemical treatments, I identified FGF-Erk axis and BMP signaling to be involved in npas4l regulation. I also showed that eomesa is a potent inducer of npas4l expression. In addition, in vitro experiments indicated that murine Eomes promotes EC specification, acting upstream of Etv2 and Tal1. Using a combination of gain-of-function and loss-of-function models for npas4l, I identified primary and secondary downstream effectors of npas4l. I showed that Npas4l binding sites are present in the promoter of genes involved in hematoendothelial specification, such as tal1, lmo2 and etv2. Importantly, I reported that npas4l is sufficient and necessary to promote the EC specification program. By performing a combined analysis of the developed datasets, I recovered putative genes with a potential role in EC specification. One of the most promising candidates was tspan18b. I generated a mutant allele for tspan18b and observed angiogenic defects in tspan18b-/- embryos, confirming a role for this gene in zebrafish cardiovascular development. I showed that Npas4l binds etv2 promoter in zebrafish. In mammalian embryonic stem cells, however, Etv2 promoter is bound by HIF-1α, a transcription factor homolog to Npas4l. Interestingly, Eomes knockdown in vitro lead to a significant reduction of Hif-1α expression. To test the function of Hif-1α in vivo, I took advantage of a murine loss-of-function model.
Hif-1α mouse mutant embryos exhibit a significant decrease in Etv2 expression, when compared with WT siblings. These data suggest a model where mammals lost npas4l during evolution and HIF-1α acquired a new function, replacing npas4l role in EC specification. I compared the phenotype of Hif-1α mouse mutant with zebrafish hif-1α loss-of- function models. Importantly, zebrafish hif-1α mutant did not show defects in vasculogenesis or EC specification, but in EC specialization, during HSC development. I showed that hypoxia is a potent inducer of HSC formation, and hif-1α as well as hif-2α act upstream of notch1, vegfaa and evi1 in hemogenic endothelial specification.
Conclusions
In this work, I explored the molecular mechanisms underlying EC specification in vertebrates, analyzing the role of bHLH-PAS transcription factors in this biological process. I identified the upstream regulators and the downstream effectors of npas4l, describing a novel role for tspan18b in zebrafish cardiovascular development. Npas4l is a transcription factor necessary and sufficient for angioblast differentiation in zebrafish, but the gene was lost in the mammalian lineage. hif-1α and hif-2α, paralogous genes of npas4l, are involved in the establishment of EC heterogeneity and specifically in the specification of hemogenic endothelium in zebrafish. Murine Hif-1α, however, is responsible for Etv2 regulation, indicating a role for hypoxia inducible factor in initiating the EC specification program in mouse, similarly to npas4l function in zebrafish.
Ausgangspunkt der Studie ist die These, dass sprachlich und kulturell heterogene Lerngruppen, an denen Studierende mit und ohne spanischsprachigen Familienhintergrund gemeinsam teilnehmen, als eine günstige Lernsituation betrachtet werden können. Voraussetzung dafür ist, dass die unterschiedlichen sprachlichen und kulturellen Gegebenheiten als Ressourcen für den Unterricht verstanden werden.
Mit dem Fokus auf Sprachveranstaltungen in Spanisch, die von Studierenden an der Johann Wolfgang Goethe-Universität Frankfurt am Main besucht werden, und die im Rahmen ihres Lehramtsstudiums im Fach Spanisch stattfinden, wurden in dieser Studie folgende Forschungsfragen verfolgt:
1. Wie entstehen und entwickeln sich Sprachlernprozesse in sprachlich und kulturell vielfältigen Lernkontexten?
2. Wie zeichnet sich das Lernen in sprachlich und kulturell vielfältigen Lernkontexten aus?
3. Welche sprachlichen und kulturellen Ressourcen können für die sprachliche und kulturelle Förderung genutzt und bewusst eingesetzt werden?
Wichtigste Forschungsergebnisse:
Es wurde festgestellt, dass Sprachlernprozesse bei Studierenden mit und ohne spanischsprachigen Familienhintergrund sowohl im kognitiven als auch im emotionalen Bereich qualitativ unterschiedlich verlaufen und, dass in sprachlich und kulturell vielfältigen Lernkontexten individuelle und überindividuelle Sprachlern-prozesse stattfinden, die einander bedingen.
Was das gemeinsame Lernen angeht, konnte festgestellt werden, dass Studierende ohne spanischsprachigen Familienhintergrund widersprüchliche Erwartungen an die Studierenden mit spanischsprachigem Familienhintergrund haben: Einerseits schätzen sie die Präsenz ihrer Kommilitonen und sehen sie als Mittler zwischen dem Unterricht und der Realität außerhalb des Unterrichts, weil sie die „echte“ Sprache und Kultur in den Klassenraum bringen. Anderseits fühlen sie sich durch ihre Anwesenheit im Unterricht verunsichert, was ihre sprachliche Entwicklung zum Teil hemmt. Bei den Studierenden mit spanischsprachigem Familienhintergrund konnten unterschiedliche Phasen in ihrem Sprachlernprozess beobachtet werden, die ihre Lernattitüde und ihre Haltung zu den Kommilitoninnen und Kommilitonen konditionieren.
Entscheidend für die Konstruktion gemeinsamer Sprachlernprozessen ist, dass die Studierenden ihre Positionierung als peer ihrer Kommilitoninnen und Kommilitonen bewusst wahrnehmen, was nicht immer der Fall ist. Auch die Bewusstwerdung der eigenen Funktion als Mittler zwischen Sprachen und Kulturen insbesondere (aber nicht nur) bei Studierenden mit spanischsprachigem Familienhintergrund soll im Unterricht durch die Reflexion über die eigene sprachliche und kulturelle Identität gefördert werden.
Im Umgang mit solchen Lerngruppen haben Dozentinnen und Dozenten unterschiedliche Erwartungen. In der Regel werden aber Lerninhalte vermittelt, die die sprachliche und kulturelle Vielfalt im Unterricht nicht berücksichtigen. Für die Leistungsbewertung beider Studierendengruppen werden oft unterschiedliche Kriterien angewendet, woraus Konflikte zwischen den Veranstaltungsteilnehmerinnen und -teilnehmern entstehen.
Die Rolle der Dozentinnen und Dozenten selbst ist in der Gestaltung gemeinsamer Lernkontexte, in denen die Interaktion der Studierenden gefördert werden, grundlegend. Damit dies gelingt, sollen sie einerseits für Mehrsprachigkeit als Phänomen der Gesellschaft, das den Sprachunterricht im besonderen Maße betrifft, sensibilisiert sein. Andererseits soll ihnen bewusst werden, dass das Sprachenlernen aus einer kognitiven und einer emotionalen Seite besteht, die in engem Zusammenhang mit mehrsprachigen und mehrkulturellen Identitätskonstruktionen stehen. Beide Seiten sollen im Unterricht gefördert werden.
Atmospheric nanoaerosols have extensive effects on the Earth’s climate and human health. This cumulative work focuses on the development and characterization of instrumentation for measuring various parameters of atmospheric nanoaerosols, and its use to understand new particle formation from organic precursors. The principal research question is, how the chemical composition of nanoaerosol particles can be measured and how atmospheric chemistry influences aerosol processes, especially new particle formation and growth. Therefore, nanoaerosols are investigated under various aspects. More specifically, an instrument is developed to analyze nanoparticles, and field as well as chamber studies are conducted.
The main project is the instrument development of the Thermal Desorption Differential Mobility Analyzer (TD-DMA, project 1, Wagner et al. (2018)). This instrument analyzes the chemical composition of small aerosol particles. By characterization and testing in chamber experiments, it is proven to be suitable for the analysis of freshly nucleated particles.
The second project (Wagner et al. (2017)) applies a broad spectrum of aerosol measurement instruments for the characterization of aerosol particles produced by a skyscraper blasting. A comprehensive picture of the particle population emitted by the demolition is obtained.
Project 3 (K¨urten et al. (2016)) is also an ambient aerosol measurement, focusing of new particle formation in a rural area in central Germany, and the ability of a negative nitrate CI-APi-TOF to detect various substances in atmosphere. Project 4 (Heinritzi et al. (2016)) is a characterization of the negative nitrate CI-APi-TOF used in projects 1, 3, 5, 6, 7 and 8. The following projects focus on understanding new particle formation from atmospherically abundant organic precursors. Key instruments comprise the negative nitrate CI-APiTOF for gas-phase measurements of the nucleating species, and various sizing and counting instruments for quantifying the particle formation and growth. Project 5 (Kirkby et al. (2016)) shows that biogenic organic compounds formed from alpha-pinene can nucleate on their own without the influence of e.g. sulfuric acid. Project 6 (Tr¨ostl et al. (2016)) describe the subsequent growth of these particles. Project 7 (Stolzenburg et al. (2018)) covers the temperature dependence of this growth and in project 8 (Heinritzi et al. (2018)), the suppressing influence of isoprene on the new particle formation is assessed.
Die Herzinsuffizienz im hohen Lebensalter spielt wegen der demografischen Entwicklung in Deutschland eine zunehmende Rolle im perioperativen Setting der Herzchirurgie. Auch nehmen strukturelle Schäden und Dysfunktionen am Herzen aufgrund von chronischen kardiovaskulären Erkrankungen zu. Diese bedürfen oftmals einer operativen Intervention als Therapie. Zum perioperativen Monitoring werden verschiedene Laborwerte herangezogen, um mögliche Komplikationen frühzeitig zu erkennen. In der vorliegenden Studie wurde das Augenmerk auf Hochrisikopatienten mit Herzinsuffizienz gelegt. Die Fragestellung behandelt, ob sich gewisse Entzündungsmarker bei Hochrisikopatienten, vor allem im hohen Lebensalter, bei verschiedenen Risikogruppen unterscheiden. Es handelt sich um eine prospektive Beobachtungsstudie, in der 94 Patienten perioperativ untersucht wurden. Diese wurden in zwei Gruppen mit jeweils 47 Patienten unterteilt. Die eine enthielt Patienten unter 75 Jahren mit einer linksventrikulären Ejektionsfraktion <35%, einer operationsbedürftigen Mehrklappen-Erkrankung oder einer Herzklappendysfunktion, kombiniert mit einer operationswürdigen koronaren Herzkrankheit. In der zweiten Gruppe wählte man Patienten in einem Alter ab 75 Jahren mit einer operationsbedürftigen Herzkrankheit. Bestimmt wurden die Entzündungsparameter CRP, Procalcitonin, Interleukin 6 und Interleukin 8. Zusätzlich wurde NT-pro-BNP als Marker für Herzinsuffizienz bestimmt. Die Blutentnahmen erfolgten präoperativ, unmittelbar postoperativ, am 1.,2. und 5. Tag postoperativ. Die präoperativen Werte wurden mit dem Risikoprofil der Patienten in Korrelation gesetzt. Dabei stach heraus, dass bei Patienten mit stattgehabter kardialer Dekompensation die Werte von CRP, NT-proBNP, IL6 und IL8 signifikant erhöht waren. Postoperativ erlitten diese Patienten häufiger eine Rethorakotomie. Außerdem ergab sich ein signifikanter Zusammenhang zwischen der Höhe der postoperativen Entzündungsparameter und dem Auftreten einer postoperativen Sepsis oder Infektion. Hier zeigte sich, dass CRP erst ab dem 5. Tag postoperativ als Indikator für eine solche gewertet werden kann, während Procalcitonin, IL6 und IL8 schon unmittelbar postoperativ als Parameter für das Auftreten einer Sepsis oder Infektion herangezogen werden können. In der vorliegenden Studie wurden jedoch nicht die Zeitpunkte der postoperativen Komplikationen bestimmt, was eine Prüfung auf Unterschied mit den postoperativen Etnzündungswerten erschwert. Zwischen den perioperativen Verläufen der bestimmten Parameter gab es zwischen den beiden Gruppen keinen signifikanten Unterschied. Dies impliziert, dass eine Ejektionsfraktion <35%/eine Mehrklappen-Erkrankung und ein Alter über 75 Jahren bei gröstenteils normwertiger EF als ebenbürtige Risikofaktoren angesehen werden können.
The overarching goal of the thesis was to create a holistic predictive framework, a vegetation model, by improving the representations of and interactions between the biosphere, hydrosphere, atmosphere and pedosphere. Vegetation models rep- resent a crucial component of Earth system model since the properties of the land surface, via interactions with the atmosphere, can have extremely large climatic effect. Yet, there remains great uncertainty associated with the dynamics of the vegetated land surface. Various vegetation models have been critiqued for numerous reasons including overly simplistic representations of vegetation, prescribed vegetation, poor representations of diversity, inaccurate representations of competition, non-transparent model calibration, and poor responses to drought. The purpose of the creation of this "next generation" model was to address deficiencies common to current vegetation modelling paradigms.
The representation of the biosphere within this framework was improved via two separate development axes. First, ecological realism was improved by integrating concepts from community assembly theory, co-existence theory, and evolutionary theory. Explicitly, rather than defining teleonomic rules to define plant behaviour the process of natural selection is modelled. By modelling the pro- cess of natural selection and its affect on relative fitness, myriad "rules" which continually adapt to biotic and abiotic conditions "come out" as a consequence of the modelled dynamics rather than being "put in". In aDGVM2 (adaptive Dynamic Global Vegetation model 2) communities of plants and their trait values evolve through time, this evolution is constrained by trade-offs between traits. Poorly performing individuals are more likely to die and produce fewer copies of themselves, this results in a filtering of trait values. Further, the community and species’ trait values can evolve through successive generations via reproduction, mutation and crossover which we approximate by using a genetic optimisation algorithm. Thus, a plant community consisting of individuals and species with potentially novel and diverse trait values is assembled iteratively through time.
We tested the assertion that improved integration of concepts from community assembly, evolutionary, and co-existence theory could address limitations of DGVMs in Chapter 2. We demonstrated that such an approach does indeed allow diverse communities of plants to emerge from the modelling framework. We showed that the position of the emergent communities in trait space differed along abiotic gradients and that, in simulations where reproductive isolation was simulated, communities emerged which were composed of multiple co-existing clusters in trait-space. Simulated trait values of co-existing strategies emerging from aDGVM2 were often multimodal, indicative of the emergence of multiple life- history strategies.
Second, to successfully model how natural selection forms a community requires accurate representation of how resource availability affects fitness. In the majority of dynamic global vegetation models (DGVMs) there is no real representation of plant hydraulics with plant water availability being calculated as a simple function of relative soil moisture content and root fractions across a number of soil layers. Worryingly, a number vegetation models appear to under represent the magnitude of these observed responses to drought. This was deficiency was ad- dressed in Chapter 3 by designing a simplified version of the cohesion tension theory of sapwood ascent where elements determining plant conductances are considered in series and implementing a set of trait trade-offs which influence a plant’s hydraulic strategy whereby hydraulic safety trades-off against xylem and leaf conductivity.
Interactions between the biosphere, pedosphere, and hydrosphere can also potentially mediate water resource availability and thus fitness. In the majority of DGVMs the volume of soil explored and explorable by plant roots in fixed glob- ally and usually constrained to a depth not greater than 3m. However, we know that soils can have a strong effect on vegetation distributions, that soil depth is not constant globally, and that plants root to variable depths.
In Chapter 4 I explored interactions between soil depth, plant rooting and the emergent properties of communities and highlighted the importance of considering interactions between the biosphere, hydrosphere, pedosphere, and fire. Here I demonstrated that, in addition to fire and precipitation, edaphic constraints on the volume of soil explorable by plant roots (e.g. by shallow soils, lateritic layers, anoxic conditions due to water logging, toxicity resulting from heavy metal concentrations) can affect the process of plant community assembly, alter the mean values of multiple traits in communities, and the trait diversity of communities.

Cardiac trabeculation is one of the essential processes required for the formation of a competent ventricular wall, whereby clusters of ventricular cardiomyocytes (CMs) from a single layer delaminate and expand into the cardiac jelly to form sheet-like projections in the developing heart (Samsa et al., 2013). Several congenital heart diseases are associated with defects in the formation of these trabeculae and lead to embryonic lethality (Jenni et al., 1999; Zhang et al., 2013, Jenni et al., 2001; Towbin 2010). It has been experimentally shown that lack of Nrg1/ErbB2/ErbB4, Angipoetin1/Tie2, EphrinB2/B4, BMP10, or any component of the Notch signaling pathway can cause defective trabeculation. Moreover, changes in blood flow and/or contractility can also affect trabeculation (Samsa et al., 2013). Together, these observations demonstrate that cardiac trabeculation is a highly dynamic and regulated process.
Trabeculation is a morphogenetic process that requires control over cell shape changes and rearrangements, similar to those observed during EMT. Epithelial cells within an epithelium are polarized and establish cell-cell junctions with the neighboring cells (Ikenouchi et al., 2003; Ferrer-vaquer et al., 2010), thus epithelial cell polarity is an important feature to maintain cell shape and tissue structure. During developmental processes such as cell migration and cell division or in disease states epithelial polarity might be disrupted. As a consequence of this alteration, cells lose their tight cell-cell adhesions, undergo cytoskeletal rearrangements, change their shape and gain migratory properties becoming mesenchymal cells (Micalizzi et al., 2010). In epithelial cells, apicobasal polarity is regulated by a conserved set of core complexes, including the PAR, Scribble and Crumbs complexes (Kemphues et al., 1988; Bilder and Perrimon, 2000; Teppas et al., 1984). The polarity proteins composing these complexes interact in a well organized and coordinated-manner creating molecular asymmetry along the apicobasal axis of the cell. In turn, this crosstalk regulates the maturation and stabilization of the junctions between cells and cytoskeleton in order to strengthen cell polarization (Roignot et al., 2013). Amongst the different polarity complex, Crumbs has been shown to be a key regulator of apicobasal polarity during development in both vertebrates and invertebrates (Tepass et al., 1990; Fan et al., 2004).
Here, taking advantage of zebrafish as a model organism, I study in vivo at single cell resolution changes in CM apicobasal polarity during cardiac trabeculation. Moreover, I show which factors regulate CM apicobasal polarity during this process. In addition, I dissect the role of the polarity complex Crumbs in regulating CM junctional rearrangements and the formation of the trabecular network.
In the 'Golden Age of Antibiotics', between 1940 and 1970, the global pharmaceutical companies discovered many antibiotics, such as cephalosporins, tetracyclines, aminoglycosides, glycopeptides, etc., as well as antifungal and antiparisitic agents. Due to several reasons, e.g. the steady re-discovery of already known NPs and the associated high costs, many pharmaceutical companies have significantly scaled back or totally abandoned their NP discovery programs since the late 20th century. Instead those companies started to focus on drug discovery based on combinatorial synthesis and thereby on the creation of enormous synthetic libraries containing small molecules. Unfortunately, this synthetic approach dealing with the optimization of existing NP or antibiotic has its limitations. As a result, leading pharmaceutical companies are re-conducting NPs research to discover new antimicrobials for the upcoming antimicrobial resistance threat. The Natural Product Center of Excellence, a collaboration between Sanofi-Aventis and Fraunhofer IME, is advancing in this context the discovery and development of novel antimicrobial agents for the treatment of infectious diseases through the testing of Sanofi's microbial extract library and strain collection. The aim of the present PhD thesis was the discovery and isolation of novel antimicrobial compounds with improved activities and/or novel MOAs as potential lead compound for a further drug discovery.
Deep learning and isolation based security for intrusion detection and prevention in grid computing

The use of distributed computational resources for the solution of scientific problems, which require highly intensive data processing is a fundamental mechanism for modern scientific collaborations. The Worldwide Large Hadron Collider Computing Grid (WLCG) is one of the most important examples of a distributed infrastructure for scientific projects and is one of the pioneering examples of grid computing. The WLCG is the global grid that analyzes data from the Large Hadron Collider (LHC) at the European Organization for Nuclear Research (CERN), with 170 sites in 40 countries and more than 600,000 processing cores. The grid service providers grant users access to resources that they can utilize on demand for the execution of custom software applications used for the analysis of data. The code that the users can execute is completely flexible, and commonly there are no significant restrictions. This flexibility and the availability of immense computing power increases the security challenges of these environments. Attackers are a concern for grid administrators. These attackers may request the execution of software with a malicious code that gives them the possibility of compromising the underlying institutions’ infrastructure. Grid systems need security countermeasures to keep the user code running, without allowing access to critical components but whilst still retaining flexibility. The administrators of grid systems also need to be continuously monitoring the activities that the applications are carrying out. An analysis of these activities is necessary to detect possible security issues, to identify ongoing incidents and to perform autonomous responses. The size and complexity of grid systems make manual security monitoring and response expensive and complicated for human analysts. Legacy intrusion detection and prevention systems (IDPS) such as Snort and OSSEC are traditionally used for security incident monitoring in the grid, cloud, clusters and standalone systems. However, IDPS are limited due to the use of hardcoded fixed rules that need to be updated continuously to cope with different threats.
This thesis introduces an architecture for improving security in grid computing. The architecture integrates the use of security by isolation, behavior monitoring and deep learning (DL) for the classification of real-time traces of the running user payloads also known as grid jobs. The first component of the proposal, the Linux containers (LCs), are used to provide isolation between grid jobs and to gather specific traceable information about the behavior of individual jobs. LCs offer a safe environment for the execution of arbitrary user scripts or binaries, protecting the sensitive components of the grid member organizations. The containers consist of a software sandboxing technique and form a lightweight alternative to other technologies such as virtual machines (VMs) that usually implement a full machine-level emulation and can, therefore, significantly affect the performance. This performance loss is commonly unacceptable in high-throughput computing scenarios. Containers enable the collection of monitoring information from the processes running inside them. The data collected via the LCs monitoring is employed to feed a DL-based IDPS.
DL methods can acquire knowledge from experience, which eliminates the need for operators to formally specify all the knowledge that a system requires. These methods can improve IDPS by building models that are utilized to detect security incidents automatically, having the ability to generalize to new classes of issues. DL can produce lower false positive rates for intrusion detection, but also provides a measure of false negatives, which can be improved with new training data. Convolutional neural networks (CNNs) are utilized for the distinction between regular and malicious job classes. A set of samples is collected from regular production grid jobs from the grid infrastructure of “A Large Ion Collider Experiment” (ALICE) and malicious Linux binaries from a malware research website. The features extracted from these samples are utilized for the training and validation of the machine learning (ML) models. The utilization of a generative approach to enhance the required training data is also proposed. Recurrent neural networks (RNN) are used as generative models for the simulation of training data that complements and improves the real collected dataset. This data augmentation strategy is useful to supplement the lack of training data in ML processes.

How much we trust our own decisions, knowledge or perceptions influences our behavior in many everyday situations. Normally the confidence we have in our decisions is rather accurate, but under certain circumstances the subjective evaluation of a decision and its objective quality can differ heavily. Subjectively over- or underestimating the quality of decisions can lead to disadvantageous behavior. Little is known about how this feeling of confidence about a decision is generated. Is it computed automatically with the decision or does it arise in a different process?
This thesis is based on a publication that contributed to the investigation of this question by comparing the influence of two different forms of spatial attention on decision confidence. Visual spatial attention is a cognitive mechanism that serves to select parts of the visual field, leading to more accurate decisions about the attended items. It can be either voluntarily controlled (endogenous) or reflexively driven by external events (exogenous). In an orientation-matching task participants performed better in both attentional conditions than in a control condition without directed attention. Additionally, we found that only endogenous, but not exogenous attention led the subjects to overestimate the quality of their performance. The possible implications of this “relative overconfidence” were discussed with respect to the theoretical framework of spatial attention and decision confidence. The present findings support the idea that decision confidence is generated in a distinct metacognitive process. Possible ideas for further neurophysiological research are proposed. The thesis concludes with an attempt to integrate the discussion into a broader context of medical research on certain neuropsychiatric symptoms and conditions.
Die vorliegende Promotionsarbeit befasst sich mit der retrospektiven Untersuchung von dentalen Implantaten, die nach Augmentation mit einem synthetischen Knochenersatzmaterial Nanobone® (Artoss, Rostock, Germany) inseriert wurden.
Ziel der vorangegangenen Studie war die Untersuchung, ob durch die Verkürzung der Einheilzeit von 6 auf 3 Monaten ein vergleichbares Ergebnis an Knochenneubildung erreicht werden kann. Es konnte gezeigt werden, dass eine Sinusbodenaugmentation mit dem synthetischen Knochenersatzmaterial Nanobone® schon nach einer Einheilzeit von 3 Monaten ein suffizientes Implantatbett vorweist und somit eine langzeitstabile Implantatrestauration (3-Jahres Follow up) ermöglicht.
Im Rahmen des 10-Jahres Follow-up konnten 12 der insgesamt 14 Patienten klinisch nachuntersucht werden.
Somit standen für die durchgeführte Nachuntersuchung noch 24 der insgesamt 27 Implantate zur Verfügung.
Hiervon wurden 17 Implantate nach 3 Monaten und 7 Implantate nach 6 Monaten inseriert.
Zehn Jahre nach der primären Augmentation und zweizeitigen Implantation zeigten sich bei der Nachuntersuchung 22 der 24 Implantate stabil und reizfrei in situ.
Die Parameter, die in der klinischen Untersuchung erhoben wurden waren, neben der festen und stabilen in situ Insertion der Implantate, eine Periotest-Messung (Medizintechnik Gulden, Modautal, Deutschland), das Bluten auf Sondieren (Bleeding on probing, BOP), die Bestimmung des Plaqueindex sowie die Untersuchung des Weichgewebes auf Rezessionen und Weichgewebsdehiszenzen.
Zwei der 24 Implantate mussten explantiert werden, eines davon bereits ein Jahr nach Implantation aufgrund von progredienten Entzündungszeichen und Lockerung.
Dies entspricht einer mittleren Implantatüberlebensrate von 91,66% oder einer Verlustrate von 8,34%.
Anders als im 3 Jahres Follow-up zeigte sich im Gruppenvergleich 3- zu 6 Monaten ein deutlicher Unterschied mit einer Implantatüberlebensrate in der 6- Monatsgruppe von 100% (7 von 7 Implantaten in situ) zu einer Implantatüberlebensrate der 3-Monatsgruppe von 88,24% (15 von 17 Implantaten in situ).
Somit ergibt sich ein deutlich besseres Outcome 10 Jahre post implantationem im Sinne einer erhöhten Implantatüberlebensrate und einer niedrigeren Komplikationsrate für die 6-Monatskontrollgruppe im Vergleich zu den Patienten, bei denen bereits 3 Monate post augmentationem implantiert wurde.
Die vorliegenden Untersuchungen zeigen, dass die Langzeitergebnisse nach Verwendung von alloplastischen KEM NanoBone®, auch nach 10 Jahren, mit vergleichbaren Langzeitergebnissen zu autologen Knochenersatz zur Knochenaugmentation im Oberkieferbereich mit gegebenenfalls kombinierter Sinusbodenaugmentation und zweizeitiger Implantatversorgung einhergehen.
Die Vasektomie stellt eine der einfachsten und effektivsten Methoden der permanenten Empfängnisverhütung dar. Trotzdem bevorzugen mehr Paare die Tubenligatur der Frau, die verglichen mit der Vasektomie sowohl größere Kosten als auch größere Risiken mit sich bringt. Durch mangelnde Informationen haben immer noch viele Männer Angst, dass sich eine Vasektomie negativ auf ihr Sexualleben auswirken könnte. Mit dieser Studie sollten diese Auswirkungen näher untersucht werden, um Männern und auch ihren Partnerinnen die Angst vor möglichen negativen Konsequenzen zu nehmen.
Hierfür wurden 294 vasektomierten Männern und ihren Partnerinnen drei Fragebögen zugeschickt. 95 Männer beantwortete den allgemeinen Fragebogen, 90 den IIEF und 74 Frauen den FSFI Fragebogen. Die Ergebnisse wurden mit Kontrollgruppen aus der Literatur verglichen.
Im IIEF-Fragebogen erreichten die vasektomierten Männer in allen fünf Domänen signifikant höhere Scores als das Vergleichskollektiv mit einer erektilen Dysfunktion aus der Literatur. In vier der fünf Domänen erreichten sie signifikant höhere Scores als die gesunde Kontrollgruppe aus der Literatur. Nur in der Domäne „generelle Zufriedenheit“ gab es keinen signifikanten Unterschied.
Die Partnerinnen der vasektomierten Männer erreichten in allen 19 Fragen des FSFI-Fragebogens sowie in allen sechs Domänen signifikant höhere Scores als Patientinnen mit einer sexuellen Dysfunktion aus der Literatur. In allen 19 Fragen und fünf der sechs Domänen gab es keinen signifikanten Unterschied zu der gesunden Kontrollgruppe aus der Literatur. Lediglich in der Domäne „Erregung“ hatten die Partnerinnen vasektomierter Männer signifikant bessere Ergebnisse.
Die Vasektomie hat keine negativen Auswirkungen auf das Sexualleben des Paares. Die Zufriedenheit der Partnerinnen wird nicht negativ beeinflusst. Die vasektomierten Männer sind mit ihrem Sexualleben sogar zufriedener und erzielen signifikant bessere Werte im IIEF-Fragebogen als eine nicht-vasektomierte Kontrollgruppe.
Die vorliegende Arbeit befasst sich mit der Medikamentenadhärenz an die Therapie mit oralen Antikoagulanzien (OAT) in der Schlaganfall-Sekundärprophylaxe und leitet auf Basis einer prospektiven Untersuchung Rückschlüsse über das Einnahmeverhalten unter verschiedenen Behandlungsformen ab.
Orale Antikoagulanzien werden erfolgreich zur Schlaganfallprävention bei Patienten mit Vorhofflimmern eingesetzt. Über Jahrzehnte waren Vitamin K-Antagonisten (VKA) die einzige Therapieoption, in der Praxis jedoch mit Schwierigkeiten verbunden, wie etwa Medikamenten- und Nahrungsmittelinteraktionen sowie häufige Gerinnungskontrollen. Als ab dem Jahr 2011 die ersten nicht-Vitamin K oralen Antikoagulanzien (NOAK) zugelassen wurden (zunächst Rivaroxaban und Dabigatran), lag ein besonderes Augenmerk auf der Frage, ob die fehlenden Gerinnungskontrollen und dadurch selteneren Arztkontakte unter Einnahme von NOAK einen negativen Einfluss auf die Adhärenz ausüben könnte.
In der vorliegenden Studie wurden prospektiv Daten zur Adhärenz und Persistenz in Bezug auf die Therapie mit OAT zu Zwecken der Schlaganfall-Sekundärprophylaxe gesammelt. Hierbei stand die Selbsteinschätzung der Patienten anhand der 8-Punkte-Morisky Medication Adherence Scale (MMAS-8) im Vordergrund. Die Daten der Untersuchung wurden an drei großen akademischen Schlaganfallzentren der Universitätskliniken Frankfurt, Würzburg und Marburg (Klinikum Fulda) erhoben. Während des Zeitraums Oktober 2011 bis September 2012 wurden alle 596 Patienten mit der Entlassungsdiagnose eines ischämischen Schlaganfalls oder einer transient ischämischen Attacke in Kombination mit Vorhofflimmern in die Studie aufgenommen. Dabei bildeten diejenigen 324 Patienten, die nach Entlassung eine orale Antikoagulation (VKA, Dabigatran oder Rivaroxaban) erhielten, die Untersuchungskohorte dieser Arbeit. Ein Jahr nach Entlassung wurden in einem Follow-up (1) die Adhärenz bzw. Persistenz in Bezug auf die verschriebene Medikation, (2) etwaige Therapiewechsel und deren Gründe, sowie (3) patientenseitige Einflussfaktoren erfragt.
Insgesamt konnte eine sterblichkeitskorrigierte Antwortrate von 73,3% (209 Patienten) erzielt werden. Von diesen Patienten erhielten 92,8% weiterhin eine Art der oralen Antikoagulation. Auch innerhalb der spezifischen ursprünglich verschriebenen OAT konnte eine gute Persistenz festgestellt werden (VKA 80,9%; NOAK 74,8%; P=0,243), wobei Dabigatran tendenziell, aber nicht-signifikant, am schwächsten abschnitt. Sofern Wechsel zwischen verschiedenen Formen der OAT erfolgten, wurden diese zumeist mit Nachteilen der jeweiligen Wirkstoffeigenschaften, wie z.B. gastrointestinale Nebenwirkungen, eine Verschlechterung der Nierenfunktion, sowie die vereinfachte Einnahme anderer Antikoagulanzien mit nur einer täglichen Dosis, begründet.
Zusätzlich stellte sich im Rahmen einer multivariaten Analyse insbesondere der Grad der Behinderung (Modifizierte Rankin Skala, mRS) zum Zeitpunkt des Follow-ups als signifikanter Einflussfaktor auf die Adhärenz heraus. Die Wahl der OAT hatte hingegen keinen relevanten Einfluss.
Zusammenfassend wurde in der Studie eine sehr gute Einnahmetreue von über 90% beobachtet, sofern man konventionelle und neue Antikoagulanzien in Summe zählt. Zudem wurde gezeigt, dass die Wahl der spezifischen OAT keinen signifikant positiven oder negativen Einfluss auf die Adhärenz nach sich zieht. Dieses Ergebnis widerspricht der Befürchtung einer generell niedrigeren Adhärenz an NOAK. Vielmehr könnte eine Erweiterung der Therapieoptionen durch die neuen oralen Antikoagulanzien es erlauben, besser auf spezielle Patientenbedürfnisse wie beispielsweise Medikamentenverträglichkeit oder Komorbiditäten einzugehen, und in Folge die Einnahmetreue zu Zwecken der Schlaganfall-Sekundärprophylaxe zu verbessern.
Zwangsmaßnahmen stellen in der psychiatrischen Versorgung ein umstrittenes Thema dar. Obwohl soweit wie möglich eine zwangsfreie Psychiatrie angestrebt wird, können psychiatrische Erkrankungen in einzelnen Fällen die Selbstbestimmungsfähigkeit der Patienten einschränken und Zwangsmaßnahmen notwendig machen. Diese können eine erhebliche Belastung für die betroffenen Patienten darstellen und bringen das medizinische Personal unter Umständen in ein ethisches Dilemma zwischen Fürsorgepflicht für erkrankte Patienten und deren Autonomiewahrung. Da sowohl national als auch international noch kein einheitlicher Konsens bezüglich möglicher Prädiktoren einer Zwangseinweisung und deren Auswirkung auf den Therapieverlauf erreicht wurde, besteht dazu noch weiterer Forschungsbedarf.
Um diese Lücke zu verkleinern, wurde eine aktuelle, retrospektive Studie auf der Grundlage der besonderen rechtlichen Situation in Hessen bezüglich des Hessischen Freiheitsentzugsgesetzes (HFEG) durchgeführt. Hierfür wurden die Patientenakten aller volljährigen Patienten, die freiwillig oder unfreiwillig (nach §10 HFEG, §1906 BGB und §1 HFEG) auf eine der drei geschützten Stationen der Psychiatrie des Universitätsklinikums Frankfurt am Main während der Quartale I/2015 und II/2015 aufgenommen wurden, ausgewertet. Dabei wurden die Unterschiede der freiwillig und der unfreiwillig aufgenommenen Patienten in soziodemographischen Prädiktoren und die Auswirkung des Aufnahmemodus auf den Therapieverlauf durch Signifikanzprüfungen und dem Verfahren der logistischen Regression betrachtet. Analysiert wurden 543 Patienten, von denen ca. 70% freiwillig und 30% unfreiwillig aufgenommen wurden.
Es konnten vier mögliche Prädiktoren einer unfreiwilligen Aufnahme identifiziert werden: Initiierung der Aufnahme, Aufnahmezeitpunkt, Hauptdiagnose und somatische Nebendiagnosen. Eine unfreiwillige Aufnahme wurde eher durch Passanten, den Betreuer/die Einrichtung oder Nachbarn/Freunde initiiert, während freiwillige Aufnahmen eher durch die Familie oder Eigeninitiative motiviert waren. Die Betrachtung der Initiierung einer Aufnahme erfolgte in der Literatur bisher selten. Übereinstimmend mit anderen Studien wurden zwangseingewiesene Patienten eher wochentags außerhalb der Kernarbeitszeiten oder am Wochenende/Feiertag aufgenommen, während freiwillige Aufnahmen vermehrt in der Kernarbeitszeit stattfanden. Die Diagnosen F0 und F4 erwiesen sich in der Regressionsanalyse als Risikofaktoren für eine Zwangseinweisung, wodurch der in der Literatur bestehende Kenntnisstand eines Zusammenhangs zwischen der Diagnose und dem Aufnahmemodus bestätigt wurde. Unfreiwillig aufgenommene Patienten wiesen zudem signifikant weniger somatische Nebendiagnosen auf. Der Einfluss der Nebendiagnosen ist in der Literatur noch nicht ausreichend untersucht, auch wenn die Ergebnisse der vorhandenen Studien größtenteils mit der vorliegenden Studie übereinstimmen.
Hinsichtlich des Therapieverlaufs unterschieden sich die beiden Gruppen in den Variablen Aufenthaltsdauer, Unterbringung im Verlauf, Zwangsfixierung, Zwangsmedikation, medikamentöse Einstellung, Antidepressiva und andere Medikamente. Zwangseingewiesene Patienten zeigten eine kürzere Aufenthaltsdauer, was möglicherweise der besonderen rechtlichen Situation in Hessen geschuldet ist. Sie wurden zudem häufiger im Verlauf des Aufenthalts untergebracht, waren öfter von Zwangsmaßnahmen wie Fixierungen oder Zwangsmedikationen betroffen und erhielten generell weniger Medikamente. Möglicherweise wurde dies durch eine fehlende Behandlungsmotivation oder eine kürzere Aufenthaltsdauer bedingt. In zusätzlichen Analysen zeigte sich, dass sich einfach und mehrfach aufgenommene Patienten in Baseline-Parametern nicht signifikant unterschieden.
Auch wenn Limitierungen wie das Vorliegen einer unizentrischen Studie oder einem Auswertungszeitraum von einem halben Jahr vorliegen, geben die in dieser Studie gefundenen Zusammenhänge wichtige Hinweise, um das Zustandekommen und die Auswirkungen von Zwangseinweisungen besser zu verstehen. Mit dieser Studie als Vergleichsgrundlage kann in Zukunft eine erstmalige Evaluierung der PsychKHG-Einführung in Hessen durchgeführt werden.
Obesity is considered as a type of chronic inflammation. It enhances the risk of developing cardiovascular disease, diabetes, and some cancers. The key players in the induction of inflammation in adipose tissue are macrophages. However the mechanism of macrophage activation in obese fat tissue is still not fully understood. Elevated level of saturated fatty acids in adipose tissue promotes inflammation and insulin resistance. Exposure of macrophages to saturated fatty acids stimulates pro-inflammatory c-Jun N-terminal kinase (JNK), nuclear factor kappa B (NF-kB) signaling, and production of pro-inflammatory cytokines, such as IL-6, IL-8, IL-1β, and TNFα. Palmitate is a major saturated free fatty acid released by adipocytes. It activates inflammatory pathways through Toll-like receptors (TLR) 2 and 4, provokes endoplasmic reticulum (ER) stress and increases levels of diacylglycerols (DAGs) and ceramides. Saturated fatty acids also affect cellular oxidative metabolism. Thus, mitochondrial fatty acid oxidation reduces ER-stress and expression of inflammatory cytokines in palmitate-treated macrophages. On the other hand mitochondrial reactive oxygen species (ROS) promote palmitate-mediated pro-inflammatory cytokine production. Recently, mitochondrial functions were linked to their morphology. Mitochondrial fission has been reported in β-cells and myocytes in response to high levels of glucose and free fatty acids, and was associated with disruption of mitochondrial functions, increased ROS level, and cell death. The aim of this study was to investigate the role of mitochondrial fragmentation in palmitate-induced inflammation in human macrophages. In our settings fatty acids, independently of their saturation, affected mitochondrial morphology. Mixtures of long chain saturated and unsaturated fatty acids as well as triglyceride-rich lipoprotein lipolysis products promoted mitochondrial fission. Mitochondrial fragmentation in palmitate-treated macrophages revealed a time- and concentration-dependent character, and was reversible upon palmitate removal. This observation, together with unaltered levels of mitochondrial protein and DNA content, and intact mitochondrial respiration, suggested that mitochondria were not damaged and were functionally active. Mechanistically, palmitate-induced mitochondrial fragmentation was not regulated by ER stress or loss of mitochondrial membrane potential. However, inhibition of palmitate incorporation into mitochondrial membrane phospholipids decreased mitochondrial fragmentation. Other approach to prevent mitochondrial fission was the inhibition of dynamin-related protein 1 (DRP1) activity, which drives mitochondrial fission by forming ring- like structures around mitochondria and constricting mitochondrial membranes. Palmitate altered mitochondrial membrane lipid composition and promoted DRP1-oligomerization. The inhibition of palmitate-induced mitochondrial fragmentation enhanced mitochondrial ROS production, c-Jun phosphorylation, and upregulated expression of pro-inflammatory cytokines. Taken together, these results suggest that mitochondrial fragmentation is a protective mechanism attenuating palmitate-induced inflammatory responses. Future experiments will be required to investigate the role of mitochondrial fragmentation in obesity-associated diseases in vivo.
HIV ist heutzutage eine gut behandelbare, chronische Erkrankung. Insbesondere bei chronischen Erkrankungen ist es entscheidend, auch die psychischen und physischen Auswirkungen auf die Lebenssituation zu untersuchen und dabei auch geschlechtsspezifische Aspekte in der gesundheitsbezogenen Lebensqualität von PLWH mit einzuschließen.
Ziel dieser monozentrischen Beobachtungsstudie ist es, die gesundheitsbezogene Lebensqualität von Patientinnen und Patienten des HIVCENTERs Frankfurt darzustellen und diesbezügliche Einflussfaktoren zu identifizieren. Im Zuge dessen wurden zusätzlich geschlechtsspezifische Unterschiede ausgewertet. Der Mental Component Score stellte die primäre Zielgröße der Studie dar, der Physical Component Score die sekundäre.
Zur Erhebung der gesundheitsbezogenen Lebensqualität wurde der SF-12v2 Fragebogen verwendet, der insgesamt zwölf Fragen beinhaltet. Inhaltlich gliedert sich der Bogen in acht Skalen und zwölf Items, die den Mental- und Physical Component Score bilden. Zur näheren Erfassung der aktuellen Lebenssituation der Patientinnen und Patientinnen des HIVCENTERs Frankfurt, wurde ein eigens für die Studie entwickelter Fragebogen verwendet. Dieser erfasste mit 19 Fragen unter anderem soziodemographische Daten, sowie Parameter zu Religiosität oder Sexualität. Retrospektive Daten aus der Epidem-Datenbank des HIVCENTERs und aus den Patientinnen- und Patientenakten wurden ebenfalls in die Auswertung einbezogen.
Die statistische Auswertung beinhaltete neben deskriptiven Methoden, einfache Varianzanalysen für geschlechtsunabhängige Zusammenhänge und Varianzanalysen mit Interaktion für das Geschlecht zur Ermittlung von geschlechtsspezifischen Einflussgrößen. Des Weiteren wurden Spearmankorrelationen berechnet und zur Identifikation von potenziellen Prädiktoren Regressionen mit Rückwärtsausschluss durchgeführt. Für beide Zielgrößen wurde identisch verfahren. Alle statistischen Tests waren zweiseitig und nutzen ein Signifikanzniveau von alpha=5%.
Im Zeitraum von September 2016 bis Mai 2017 wurden insgesamt 275 Patientinnen und Patienten in die Studie eingeschlossen, darunter 123 Frauen, 150 Männer und 2 transgender Personen. Letztere wurden aufgrund der geringen Fallzahl nicht über die deskriptive Statistik hinaus in den Berechnungen berücksichtigt. Das durchschnittliche Alter in der Studienpopulation betrug 46 Jahre. Frauen hatten ein Durchschnittsalter von 44, Männer von 48 Jahren. 97% der Patientinnen und Patienten waren zum Erhebungszeitpunkt unter antiretroviraler Therapie. Im Durchschnitt erzielten die Teilnehmerinnen und Teilnehmer im Vergleich mit der Referenzpopulation einen unterdurchschnittlichen Mental Component Score von 46. Die Frauen der Studie erzielten einen signifikant schlechteren MCS als die Männer (45 vs. 48; p=0,02). Im Kontext mit den übrigen Prädiktoren des Regressionsmodells erreichten Frauen einen um durchschnittlich 13 Punkte schlechteren MCS als Männer (B=-13; p=<0,001). Als geschlechtsunabhängige negative Prädiktoren auf den MCS stellten sich unter anderem regelmäßiger Alkohol- und Drogenkonsum heraus, sowie das Unterlassen von regelmäßigem Sport oder eine negative Zukunftsaussicht. Als geschlechtsabhängiger negativer Prädiktor erwies sich bei den Frauen eine afrikanische versus westeuropäische Herkunft (B=-5; p=0,028). Arbeitslosigkeit stellte sich bei Männern als geschlechtsabhängiger negativer Prädiktor heraus (B=-5; p=0,033).
Vorliegende Dissertation macht deutlich, dass PLWH auch heute noch eine unterdurchschnittliche gesundheitsbezogene Lebensqualität aufweisen und darüber hinaus deutliche geschlechtsabhängige Unterschiede existieren. HIV-positive Frauen erreichten in dieser Studie signifikant schlechtere Werte für den Mental Component Score als Männer und waren hinsichtlich negativer Einflussgrößen überrepräsentiert. Anhand der hier verwendeten Fragebögen wird es behandelnden Ärztinnen und Ärzten innerhalb der Routinesprechstunde ermöglicht, ein regelmäßiges Monitoring der HRQoL durchzuführen und auch ihren Verlauf zu beurteilen. Zudem wäre ein Screening nach den in dieser Arbeit ermittelten negativen Prädiktoren der HRQoL möglich, wodurch entsprechenden Patientinnen und Patienten ein zielgruppenspezifisches Angebot erhalten könnten.
Nachdem die Sicherheit der Blutprodukte in den letzten Jahrzehnten vor allem bei transfusionsmedizinisch relevanten Viren wie HIV-1, HCV und HBV wesentlich durch die Einführung von molekularen Nachweismethoden auf Restrisiken unter 1 zu 1 Million reduziert werden konnte, liegt der aktuelle Fokus der Transfusionsmedizin auf der Vermeidung von bakteriellen Übertragungen. Dabei stehen vor allem die Thrombozytenkonzentrate im Vordergrund, da diese bei Raumtemperatur gelagert werden und somit für viele Bakterien ideale Wachstumsbedingungen darstellen. Die vorliegende Arbeit hat dabei in vier aufeinander aufbauenden Phasen systematisch die klinische Effizienz eines photochemischen Pathogeninaktivierungsverfahrens untersucht.
Phase 1: Darstellung des Wachstumsverhaltens von 8 transfusionsmedizinisch relevanten Keimen mittels der Spiking-Versuche in Thrombozytenkonzentraten. Für die nachfolgenden Phasen sind nur die Bakterienstämme ausgewählt worden, die nachweislich zu einer Vermehrung in Thrombozytenkonzentraten geeignet sind.
Phase 2: In den Experimenten mit Vollblutkonzentraten zeigt sich bei der Spikingkonzentration von 100 CFU/Beutel für alle ausgewählten Keime eine 100%ige Inaktivierungseffizienz. Bei der Anfangskonzentration von 1000 CFU/Beutel ergibt sich für den Keim Klebsiella pneumoniae (PEI-B-08-09) eine Inaktivierungseffizienz von 75% und für den Keim Bacillus cereus eine Inaktivierungseffizienz von 50%. Alle anderen Keime haben in den Experimenten mit der höheren Spikingkonzentration eine Inaktivierungseffizienz von 100%.
Phase 3: Für den Keim Klebsiella pneumoniae (PEI-B-08-09) zeigt sich in den Experimenten mit Pool-TKs schon bei der niedrigen Anfangskonzentration eine Inaktivierungseffizienz von 75%. Bei der Spikingkonzentration von 1000 CFU/Beutel ergibt sich erneut für die Keime Klebsiella pneumoniae und Bacillus cereus eine Inaktivierungseffizienz von 50%. Alle anderen Keime weisen sowohl bei der niedrigen als auch bei der hohen Bakterienkonzentration eine 100%ige Pathogeninaktivierungseffizienz auf.
Phase 4: In den Experimenten mit Apherese-TKs zeigt sich für alle untersuchten Keime sowohl bei der niedrigen als auch bei der hohen Spikingkonzentration eine 100%ige Inaktivierungseffizienz.
Aus diesen Experimenten geht hervor, dass man mittels photochemischen Pathogeninaktivierungsverfahrens keine 100%ige Inaktivierungseffizienz erreichen kann. Vielmehr ist die Inaktivierungseffizienz zum einen von der bakteriellen Ausgangskonzentration, zum anderen aber auch vom Zeitpunkt der Anwendung des Verfahrens abhängig. Somit sollte der Begriff „ Pathogeninaktivierungsverfahren“ besser durch den Begriff des Pathogenreduktionsverfahrens ersetzt werden. Ferner wird anhand der Ergebnisse deutlich, dass die Anwendung von Pathogenreduktionsverfahren möglichst schnell nach der Spende erfolgen sollte. In diesem Zusammenhang haben Apherese-Thrombozytenkonzentrate gegenüber Pool-Thrombozytenkonzentraten einen Sicherheitsvorteil.
Our understanding of human biology and disease is based on the last millennia’s gain of knowledge, which has been exponentially accelerated since the invention of optical and "biochemical" microscopes like transcriptomics and other omics technologies.
In order to broaden our knowledge of an important human transcription factor, T-Cell Acute Lymphocytic Leukemia 1 (TAL1), some of these technologies were used.
TAL1’s gene or promoter structure is altered in about 20-30% of T-ALL. In addition, there is an increase in TAL1 expression in ca. 60% of pediatric and about 45% of adult T-ALL. Physiologically, TAL1 is an indispensable factor in hematopoiesis: in the murine knockout model, blood cells vanish in the early embryonic period. In addition, the TF is also relevant in adult erythropoiesis.
Accordingly, the identification of novel TAL1 target genes was significant both for clinical reasons and in order to understand the hematopoietic functions.
We performend a combined RNA- and ChIPseq approach. After a lentiviral mediated knockdown in K562 cells RNAseq was performed using the Illumina high-throughput method. Overall, the RNAseq yielded one billion good quality sequencing fragments. They made identification of up- and downregulated transcripts as well as associated biological processes, cellular components, molecular function and dominant KEGG signaling pathways possible. Furthermore, more than 2-fold altered coding transcripts and lncRNA were analyzed for relevant TAL1-binding in the transcription start area. There were 3205 significantly altered coding transcripts and 5136 significantly altered lncRNA. By integrating an Encode TAL1-ChIPseq in K562 cells (using a cutoff fold change of 2x) a relevant TAL1 binding could be detected with 71 coding and 416 lncRNA genes.
The combination of RNA- and ChIPseq yields a wealth of relevant results. Accordingly, TAL1 has complex pro- and anti-malignant effects in all areas of oncogenesis like described by Hanahan and Weinberg. Various interactions with target genes and signaling cascades in inter alia proliferation (e.g. HEMGN, MYC, AHI1, YPEL3, BTG2), angiogenesis (e.g. EGFL7, LTBP3), apoptosis (e.g. BCL3, BCL2A1, BMF), immune evasion (e.g. CMTM6) and inflammation (e.g. IL23 and PTGS1) have been revealed, thus complementing the knowledge about pro- and anti-oncogenic effects of TAL1. In addition, it was possible to identify target genes relevant for erythropoiesis and possible osteogenesis. Concerning lncRNA, interesting potential effectors have been identified. However, they still need to be functionally characterized. Relating the results to Virchow’s first description of leukemia as "white blood" the role of TAL1 in leukemia’s genesis but also in erythropoiesis has been confirmed and extended, thus contributing to explain Virchow’s observation: "...therefore, when I speak of white blood, I mean in fact a blood in which the proportion between the red and colorless (in white) blood corpuscles is reversed ...” (Virchow R. Weisses Blut. Frorieps Notizen 1845;36:151-156).
Structured illumination microscopy (SIM) is part of the super-resolution methods developed at the beginning of this century. To produce a super-resolution image SIM requires three things: 1) illumination of the sample with a periodic pattern, 2) acquisition of multiple images per plane under different pattern’s phases and orientations and 3) the processing of these images has to be carried with a reconstruction algorithm. The result of the reconstruction is an image with a resolution gain that is proportional to the frequency of the pattern (po). The typical SIM set-up uses an epi-fluorescence configuration, thus the interference angle of the beams that create the pattern is restricted by the angular aperture of the objective. Under this restriction the maximum value of po is given by the cut-off frequency of the objective lens and sets at 2 the maximum resolution gain of SIM under linear illumination.
In the first part of this thesis we present the implementation and characterization of the 2D-SIM set-up designed by Dr. Bo-Jui Chang (B-J. Chang et al., PNAS 2017), this design exploits the concept introduced by light-sheet microscopy, i.e. separation of illumination and detection paths to obtain resolution gains larger than the usual two-fold (Chapter 3). The set-up is named coherent structured illumination light-sheet based fluorescence microscopy (csiLSFM) and it consists of a triangular array of three objectives, such that two are used for illumination and one for detection. With the independent illumination arms is possible to interfere two coherent light-sheets at angles beyond the angular aperture of the detection lens, attaining the maximum interference angle of 180° when the light-sheets counter-propagate. This condition delivers a pattern with a po 1.4 times larger than the cut-off frequency (ωo), hence our set-up provides generic resolution gains of 2.4.
The extraction of the high spatial frequencies that produce the resolution gain in the csiLSFM is a challenge due to a low pattern modulation. The low modulation inherently arises because the frequency associated to the pattern period lies beyond the cut-off frequency of the detection lens. To overcome this challenge we developed a filtering strategy that facilitates the withdrawal of information from a SIM data set, simultaneously the proposed filtering process optimizes the reconstruction algorithm by reducing the periodic artifacts that are recurrent in SIM images. In this same chapter we also performed an spectral analysis of the artifacts and determined that they originate from irregularities in the power spectrum that occur due to the partial or total lack of certain spatial frequencies (fig.4.2 and 4.3), our reconstruction reduces this information drops and diminishes the artifact occurrence. The relevance of our reconstruction pipeline is that it delivers a standardized process to enhance the SIM image in a current context in which the commonly used reconstruction algorithms employ empirical tuning to improve it (fig.4.13). Moreover, the pipeline is applicable to the csiLSFM data and also to images acquired with any other 2D-/3D-SIM set-up (fig.4.10 and 4.11).
The processing of various image data sets acquired with the csiLSFM exposed us to the question of how low the modulation of the illumination pattern can be before no super-resolution frequencies can be extracted. Answering this question is important to guarantee that the SIM data contains enough spatial frequencies to provide significant resolution gains. Thus in chapter 5 we developed a quantitative metric to indirectly determine the pattern modulation from the SIM data and find its critical value to use it as evaluation criterion. We called this metric the quality factor (Q-factor) and it represents the normalized strength (amplitude) of the extracted frequencies respect to the Gaussian noise contained in the images. Through simulations we estimated that Q=0.11 is a critical value and a SIM data set requires this as minimum value is to deliver a significant resolution gain. Q works then as an assessment tool for classifying SIM data as optimal or sub-optimal, i.e. Q≥0.11 or Q<0.11. We demonstrated such application with data acquired in various SIM commercial set-ups to prove its feasibility in the field (fig.5.6-5.11)
As mentioned at the beginning of this abstract SIM requires a specialized set-up and a processing algorithm to produce super-resolution images. This thesis contributes to these two areas in the following aspects: first, in its linear version a structured illumination microscope is highly associated to a 2-fold resolution gain. Here we demonstrated the possibility of extending this gain to 2.4 using our custom set-up the csiLSFM. Second, a reconstructed SIM image is prone to artifacts due to the mathematical process it undergoes, here we analyzed the artifact sources and identified them with drops of spatial information in the reconstructed spectrum, based on these conclusions we designed a processing pipeline to facilitate the extraction of spatial frequencies and directly reduce artifacts. A third and final outcome of this thesis is the development and practical implementation of a quantitative index to evaluate the quality of SIM data in terms of its relevant information content (Q-factor). Accordingly, the overall contributions of this work were done in the areas of SIM set-up, SIM reconstruction procedure and SIM data evaluation.
The membrane protein Green Proteorhodopsin (GPR), found in an uncultured marine γ-proteobacterium, is a retinal binding protein and contains a conserved structure of seven transmembrane helices (A-G). The retinal is bound to a conserved lysine residue (K231) in helix G via Schiff base linkage. It belongs to the widespread family of microbial rhodopsins and functions as a light dependent outward proton pump that bacteria may utilize for establishing a proton gradient across the cellular membrane. Proton pumping takes place after photon absorption, where GPR goes through a series of conformational changes, termed photocycle, causing the proton to be transported across the cellular membrane from the intra-cellular to the extracellular space. It is further mediated by the highly conserved functional residues D97 and E108, which function as the primary proton acceptor and primary proton donor for the protonated Schiff base, respectively. Another functionally important residue is the highly conserved H75 in helix B. It forms an intra-molecular cluster with D97 and is responsible for the high pKa value of the primary proton acceptor, stabilized by a direct interaction between D97 and H75.
Different Proteorhodopsin variants are globally distributed and colour tuned to their environment, depending on the water depth in which they occur. A single residue in the retinal binding pocket at position 105 is responsible for determining the absorption wavelength of the protein. GPR (from eBAC31A08) contains a leucine at position 105, while BPR (blue proteorhodopsin, from Hot75m4) in deeper waters possesses a glutamine. Although GPR shows 79% sequence identity with BPR, a single amino acid substitution (L105Q) in GPR is able to switch the absorption maximum to the one of BPR.
Protein oligomerisation describes the association of subunits (protomers) through non-covalent interactions, forming macromolecular complexes. It is an important structural characteristic of microbial rhodopsins, contributing to structural stability and promoting tight packing of the protomers in the bacterial membrane. GPR was shown to assemble into radially arranged oligomers, mainly pentamers and hexamers. No high resolution crystal structure of the whole GPR complex is available, but the structurally related BPR (Hot75m4) was successfully crystallized, showing pentameric oligomers.
The BPR crystal structure model reveals detailed information about complex assembly of the whole proteorhodopsin family. It reveals the oligomeric structures and shows residues that are part of the protomer interfaces, forming cross-protomer contacts, which is valuable information for the elaborate analysis of cross-protomer interactions of GPR oligomers.
Based on the knowledge of GPR and BPR oligomeric complexes, the aim of this study is to analyse specific cross-protomer contacts and to characterize the functional role of GPR oligomerisation. This includes the identification of residues, which are part of charged cross-protomer contacts and play an important role for the formation of the GPR oligomeric complex. Furthermore, this study deals with a detailed characterization of a potentially functional cross-protomer triad between the residues D97-H75-W34, which was detected in the BPR structural model. Hereby, the focus lies especially on the functional role H75, which is highly conserved and is positioned in between the primary proton acceptor D97 and W34 across the protomer interface. In summary, this study addresses GPR oligomerisation via specific cross-protomer contacts and its potential role for the functional mechanism of the protein.
The fundamental technique used in this study is solid-state NMR. Furthermore, an elaborate characterization of GPR oligomerisation was executed using a variety of biochemical methods and mutational approaches. Solid-state NMR is a powerful biophysical method to analyse membrane proteins in their native lipid environment and can be used to obtain diverse information about structure, molecular dynamics and orientation of the protein in the lipid bilayer.
Solid-state NMR naturally has a low sensitivity. In order to detect the low number of spins, DNP signal enhancement is of particular importance in this study. It is exhibited under cryogenic conditions and allows to drastically enhance the solid-state NMR signal by transferring magnetization from highly polarized electrons to the nuclear spins.
By applying these methods and techniques on GPR oligomers, this study reveals new insights in specific cross-protomer interactions in the complex. First the oligomeric states of GPR were determined for the specific experimental conditions used in this study. LILBID-MS, BN-PAGE and SEC analysis identified the pentameric state to be dominant for GPR. Furthermore, specific interactions across the protomer interface, which drive GPR oligomerisation, were identified. This was conducted by creating mixed 13C-15N labelled complexes. These mixed complexes show a unique isotope labelling pattern across their protomer interfaces. Solid-state NMR 13C-15N-correlation spectroscopy (TEDOR) was used to identify through-space dipole-dipole couplings, which indicate specific cross-protomer contacts. The results indicated that the residues R51, D52, E50 and T60 are important for GPR oligomerisation, and further analysis via single mutations of these residues showed a severe impact of the GPR oligomerisation behaviour.
The functional importance of GPR oligomerisation was analysed by DNP-enhanced solid-state NMR on the cross-protomer D97-H75-W34 triad. The DNP cryogenic conditions allowed to trap GPR in distinct stages of the photocycle. It could be shown that trapping GPR in a specific intermediate leads to a drastic conformational effect for the highly conserved H75 residue. Furthermore, DNP-enhanced solid-state NMR was used to characterize the cross-protomer contact between H75 and W34. Mutations of W34 could show that the cross-protomer interaction is highly important for the functionality of the protein, as negative mutants such as W34E showed a reverse proton transport across the bacterial membrane.
In summary this study represents a detailed analysis of GPR cross-protomer interactions and sheds light into the cause and functional importance of oligomeric complex formation in the microbial rhodopsin.
