Wenn Søren Kierkegaard in den "Schriften über sich selbst" die eigene Gegenwart als "eine Zeit der Auflösung"l bezeichnet, die eine "Radikalkur" bedürfe, so markiert er damit zugleich seinen eigenen philosophischen Entwurf existenz-dialektischen Denkens. als ein Konzept, das im Horizont jener gesellschaftlichen Modernisierungsprozesse zu verorten ist, die im Laufe des neunzehnten Jahrhunderts eine Neukonzeptualisierung des Denkens von Mensch und Welt unabweisbar fordern. Topisch geworden in diesem Sinne ist Karl Löwiths Rede vom "revolutionären Bruch im Denken des neunzehnten Jahrhunderts". Wie für viele seiner Zeitgenossen bildet dabei für Kierkegaard die Identitätsphilosophie Hegels den negativen Bezugspunkt. Während jedoch Theoretiker wie Marx in dem Sinne der Hegelschen Spur folgen, daß sie den Systemcharakter dieser ersten Theorie der Moderne beerben, ist es im Falle Kierkegaards gerade Hegels Verwissenschaftlichung der Philosophie, die zum Widerspruch herausfordert. Der Begriff der Verwissenschaftlichung bezeichnet dabei den Sachverhalt, daß die Philosophie in Gestalt einer objektivierenden Ontologie der Gegenwart dem Individuum keinerlei Handlungsorientierung mehr zu geben vermag. Kierkegaard problematisiert mit seiner HegelKritik also jene Grundtendenz der modernen Kultur, welche Max Weber als einen Prozeß der Ausdifferenzierung in füreinander gleichgültige Wertsphären und mithin als einen Vorgang der Dezentrierung der symbolischen Ordnung beschrieben hat.
Es wurden die Herstellung von protaminbasierten Oligonukleotid-Nanopartikel („binäres System“) physikochemisch charakterisiert. Vermischt man Oligonukleotide mit einer wässrigen Protaminlösung entstehen nach wenigen Sekunden sehr kleine Partikel (<20 nm). Der Partikeldurchmesser nimmt mit der Zeit zu und stabilisiert sich nach etwa 30 Minuten. Danach sind die NP für mindestens 24 Stunden bei Raumtemperatur stabil. Die Größe, die Größenverteilung und die Gestalt der Partikel wurden bestimmt. Diese runden Partikel konnten in ihrer Größe durch die Veränderung von Protamin- und ON-Konzentration in ihrer Größe gesteuert werden (etwa 100-1000 nm Durchmesser). Die Veränderung der Kettenlänge der Oligonukleotide (10-25 mer) zeigte einen nur geringen Einfluss auf die Größe und Größenverteilung der Partikel. Auch konnte die Oberflächenladung (Zetapotential) der Partikel durch die Veränderung des Protamin/OligonukleotidVerhältnisses gesteuert werden. Analytische Ultrazentrifugations-Messungen (durchgeführt vom AK Prof. Schubert) mit diesen Partikeln zeigte das diese schon in geringen Salzkonzentrationen (15 mM NaCl) nach wenigen Minuten zur Aggregation neigen. Diese konnte durch 15% Polyethylenglykol 20.000 Lösung gestoppt werden. Dies war ein wichtiger Hinweis, dass man mittels Makromolekülen die Partikelpräparationen stabilisieren kann. Ferner wurde die Zusammensetzung der Partikel, sowohl hinsichtlich der Oligonukleotide und als auch des Protamingehaltes bestimmt. Mehr als 90% des Wirkstoffes konnte in die Nanopartikel eingebunden werden. Da das von uns verwendete Protamin keine aromatische Aminosäure beinhaltet (Absorptionsmaximum kleiner 200 nm) und die Standard-Proteinnachweise wie z.B. BCA-Assay nur ungenügende Nachweisgrenzen aufweisen, wurde eine neue Protamin-Quantifizierungmethode mittels ortho-Phthaldialdehyd (OPA) und N-Acetyl-L-Cystein (NAC) für die Mikrotiterplatten erarbeitet. Damit war es möglich, auch in Anwesenheit von DNA, Protamin und Protaminsalze in einer Konzentration von 250 ng/ml sicher nachzuweisen. Ein weiteres Problem mit diesem binären Partikelsystem, neben der Aggregationsneigung, zeigte sich in verschiedenen Zellmodellen (durchgeführt von Norbert Dinauer, AK von Briesen). Die Partikel wurden zwar im großen Maße von humanen Macrophagen, humanen T-Zellen aufgenommen, aber danach langsam freigegeben. Auch wurden intrazellulär Aggregate gefunden. Humanes Serum Albumin (HSA) zeigte einen positiven Effekt auf die Aggregationsneigung (AlPrO-Nanopartikel). Es fungiert hierbei als ein Schutzkolloid, welches nur zu einem geringen Teil in die Partikelmatrix inkorporiert wird (<10%). Der Großteil des HSA befindet sich in der äußeren wässrigen Phase. Es ist zu vermuten dass es sterische die Partikel an der Sekundäraggregation hindert. Zusätzlich konnten primäre Komplexe vor der Zugabe von HSA aus HSA und Protamin beobachtet werden (Durchmesser 14 nm). Dies führt zu einem veränderten Self-Assembly der Nanopartikel gegenüber dem binären System. AlPrO-Partikel, hergestellt mit 1 mg/ml HSA, waren über Stunden stabilisiert in Zellmedium. Sie sind von runder Gestallt und 200-350 nm im Durchmesser groß. Sie weisen eine leicht negative Oberflächenladung auf, was auf eine Abdeckung der Partikeloberfläche mit HSA hinweist. Nahezu die gesamte eingesetzte Menge des Oligonukleotides wurde in die Partikelmatrix inkorporiert (> 90%). Es war möglich Partikel dieser Qualität mit unmodifizierten-, phosphorothioat modifizierten Oligonukleotiden und doppelsträngiger RNA (siRNA) herzustellen. Die von uns entwickelten AlPrO-Nanopartikel stellen eine neue Möglichkeit der peptidbasierte Transfektion von Oligonukleotiden in Zellen dar.
RDF is widely used in order to catalogue the chaos of data across the internet. But these descriptions must be stored, evaluated, analyzed and verified. This creates the need to search for an environment to realize these aspects and strengthen RDFs influence. InterSystems postrelational database Caché exposes many features that are similar to RDF and provide persistence with semantic part. Some models for relational databases exist but these lack features like object-oriented data-structures and multidimensional variables. The aim of this thesis is to develop an RDF model for Caché that saves RDF data in an object-oriented form. Furthermore an interface for importing RDF data will be presented and implemented.
In this work we examine several sentential particles, occurring in imperatives, main exclamative and interrogative sentences, which display a uniform syntactic behaviour. We analyse them as heads of high CP projections which require their specifier to be filled either by the wh-item (in sentences where there is one) or by the whole clause, yielding the sentence final position of the particle. The hypothesis that they are C°-heads accounts for their sensitivity to sentence type and for their occurrence only in matrix contexts. We also provide a first sketch of their semantic contribution, showing that they select ‘non standard’ contexts and interact with tense and modality of the verb when the whole CP has moved to their specifier.
Cytochrome P450 (CYP) enzymes oxidize, peroxidize and/or reduce cholesterol, vitamins, steroids, xenobiotics and numerous pharmacological substances in an oxygen- and NADPHdependent manner. Since many CYP isozymes are also capable of metabolizing arachidonic acid to biologically active products, CYP enzymes are often described as the third pathway of arachidonic acid metabolism i.e., in addition to cyclooxygenases and lipoxygenases. CYP enzymes are predominantly expressed in the liver while others, such as members of the CYP 2J, CYP 2C and CYP 4A subfamilies, can be detected in extrahepatic tissues, particularly in the cardiovascular system. Recent data suggest that a CYP 2C enzyme(s) expressed in coronary artery endothelial cells generate epoxyeicosatrienoic acids (5,6-; 8,9-; 11,12- and 14,15-EET) which contribute to the acute control of vascular tone and the longterm regulation of vascular homeostasis.
The expression of CYP 2C in coronary artery endothelial cells is regulated by a number of stimuli, such as cyclic stretch and fluid shear stress as well as by the corticosteroid cortisol and a number of CYP substrates (nifedipine, cerivastatin and -naphthoflavone). However, the signalling pathways and the transcription factors involved in regulating the expression of the gene are unknown.
Since most of the CYP 2C enzymes are transcriptionally regulated, we were interested in identifying the CYP 2C isoform(s) expressed in porcine coronary artery endothelial cells (PCAEC) as well as determining its/their promoter sequence(s). The overall goal was to study the involvement of different transcription factor binding elements in the regulation of the CYP 2C gene(s). Porcine coronary arteries were used given the possibility of analysing the results obtained at the cellular level with alterations in vascular function. Comparison of the porcine CYP 2C and the human CYP 2C8 and 2C9 promoters was also a major goal of this study.
To identify the relevant porcine CYP 2C isoform nested RT-PCR was performed using total RNA from porcine coronary artery endothelial cells. Comparison of the sequence of the product of this reaction with the NCBI database suggested that the CYP 2C expressed in PCAEC was approximately 85% homologous with the human CYP 2C9 enzyme. To obtain the full length CYP 2C isoform 5´ rapid amplification of cDNA end (5´ RACE) was performed using a downstream reverse gene specific primer which is conserved in all of the porcine CYP 2C isoforms. The intention behind using such a primer was to amplify all the possible CYP cDNAs expressed in PCAEC. With the 5´ RACE technology it was possible not only to identify the exact isoform (CYP 2C34) expressed in PCAEC, but it was also possible to amplify 550 bp of the 5´ upstream region. This result was authenticated by comparing the protein/nucleotide sequence with other human CYP 2C genes such as CYP 2C8 and CYP 2C9 as well as different porcine CYP 2C genes (CYP 2C34, CYP 2C49). Multiple protein/nucleotide sequence alignment revealed approximately 85-90% sequence identity. An exon1-2 specific radio-labelled probe of the CYP 2C34 gene was then used to screen a porcine genomic library for positive genomic clones containing the promoter region of the CYP 2C34 gene.
For the isolation of 5´ flanking region of CYP 2C34 gene a PCR-based directional genome walking strategy was used in which the positive porcine genomic BAC clones were taken as a DNA template. Four arbitrarily designed universal walking primers and a gene-specific primer derived from the CYP 2C34 gene sequence were employed and led to the identification and isolation of 1.4 kb of the 5´ flanking region.
The 1.4 kb 5´ flanking region of CYP 2C34 gene contains multiple transcription factor binding sites including glucocorticoid-responsive element (GRE), hypoxia-responsive element (HRE), CAAT-enhancer binding protein (C/EBP), stress responsive element (STRE) consensus sequences. CYP 2C34 promoter constructs were generated and reporter gene activity (luciferase) activity was compared with that of a promoterless vector (pGL3-Basic) at first in HEK cells and then in PCAEC. After using cortisol as a positive control to demonstrate that the promoter constructs generated were functional we determined the effects of physiologically relevant stimuli i.e., hypoxia and cyclic stretch. Additional experiments with zinc sulphate were performed in a preliminary analysis of the role of Zn2+ inducible transcription factors and might be cooperative heterodimerization formation with these transcription factor with C/EBP in the regulation of CYP 2C34 expression. With all these stimuli, reporter gene activity of CYP 2C34 promoter was significantly (3-8 fold) increased over values obtained in unstimulated cells.
Analysis of the regions that are essential for the induction of promoter activity in response to the different stimuli of interest have to be performed in combination with gel shift assays, siRNA experiments as well as site-directed mutagenesis experiments. Comparison of the regulation of the CYP 2C34 gene and correlation with changes in vascular function (in isolated porcine coronary arteries) should deliver information relevant to the regulation of the CYP 2C enzyme expressed in human coronary artery endothelial cells. The recent demonstration of a clinically relevant role for CYP 2C9 in coronary heart disease underlines the importance of such a study.
This paper investigates the magnitude and the main determinants of share price reactions to buy-back announcements of German corporations. For our comprehensive sample of 224 announcements that took place between May 1998 and April 2003 we find average cumulative abnormal returns around -7.5% for the thirty days preceding the announcement and around +7.0 % for the ten days following the announcement. We regress post-announcement abnormal returns with multiple firm characteristics and provide evidence which supports the undervaluation signaling hypothesis but not the excess cash hypothesis or the tax-efficiency hypothesis. In extending prior empirical work, we also analyze price effects from initial statements of firms that they intend to seek shareholder approval for a buy-back plan. Observed cumulative abnormal returns on this initial date are in excess of 5% implying a total average price effect between 12% and 15% from implementing a buy-back plan. We conjecture that the German regulatory environment is the main reason why market variations to buy-back announcements are much stronger in Germany than in other countries and conclude that initial statements by managers to seek shareholders’ approval for a buy-back plan should also be subject to legal ad-hoc disclosure requirements.
During the past fifty years sign languages have been recognised as genuine languages with their own syntax and distinctive phonology. In the case of sign languages, phonetic description characterises the manual and non-manual aspects of signing. The latter relate to facial expression and upper torso position. In the case of manual components these characterise hand shape, orientation and position, and hand/arm movement in three dimensional space around the signer's body. These phonetic charcaterisations can be notated as HamNoSys descriptions of signs which has an executable interpretation to drive an avatar.
The HPSG sign language generation component of a text to sign language system prototype is described. The assimilation of SL morphological features to generate signs which respect positional agreement in signing space are emphasised.
Signatur, Hieroglyphe, Wechselrepräsentation : zur Metaphysik der Schrift in Novalis' "Lehrlingen"

Die These, die ich in diesem Aufsatz entfalten möchte, besagt, dass sich die [...] Gedankenfigur einer Naturschrift aus drei Elementen zusammensetzt: Novalis verbindet zwei historische Modelle, die aus verwandten (aber nicht identischen) Traditionen stammen, mit einer aktuellen Debatte in der Naturphilosophie und -wissenschaft. Die Rede ist von der Signaturenlehre der Frühen Neuzeit, der Hieroglyphen-Diskussion des 18. Jahrhunderts und der Organismus-Debatte um 1800. Ich werde dabei so vorgehen, dass ich zuerst diese drei Größen isoliere und in einem zweiten Schritt ihre Synergie-Effekte vorführe
