Neurosphären-bildende Zellen mit dem Potential zur Selbsterneuerung, sowie zur vielseitigen Differenzierung, wurden bereits aus diversen peripheren und zentralen Geweben isoliert. Auch von der Neuralleiste abstammende NCSCs wurden in verschiedenen embryonalen und adulten, peripheren Geweben im Nager-Modell identifiziert und als Neurosphären (NS) kultiviert. In der vorliegenden Arbeit konnten nun erstmals auch aus Zellen des peripheren Nervensystems von Huhn-Embryonen NS gebildet werden. Zudem wurde das Selbsterneuerungspotential der NS-bildenden Zellen analysiert und über 10 Passagen, beziehungsweise mehrere Monate hinweg quantifiziert. Durch eine selektive Isolierung O4-positiver Gliazellen aus Huhn DRGs, konnte der Anteil NS-generierender Zellen gegenüber unsortierten DRG-Zellen signifikant erhöht werden. Aufgrund dieser Beobachtung können im Gewebe verbliebener NCSCs als Ursprung NS-generierender Zellen ausgeschlossen werden. Die von Gliazellen abstammenden NS-Zellen wurden im Weiteren auf ihr Entwicklungspotential hin untersucht. Sowohl über Antikörperfärbungen, als auch auf Nukleinsäurebasis in RT-PCR-Analysen, konnte für die NS-Zellen ein oligopotentes Differenzierungspotential nachgewiesen werden. Auch in klonalen NS-Kulturen wurde die Differenzierung zu Neuronen, Gliazellen und Melanozyte bestätigt. Aufgrund des Selbsterneuerungspotentials, sowie des oligopotenten Entwicklungspotentials O4-sortierter NS-generierender Zellen, wurden die in dieser Arbeit identifizierten und charakterisierten NS-bildende Zellen GDSCs (glial-derived stem cells) benannt. Die im Huhn-Modell beschriebenen NS-bildenden PNS-Zellen wurden anschließend auch im Nager-Modell nachgewiesen. NS-generierende Zellen sind in prä- und postnatalen Maus-DRGs vorhanden und zeigen ein Selbsterneuerungspotential, das für Stammzellen charakteristisch ist. In Zusammenarbeit mit Dr. Verdon Taylor (Freiburg) konnten die aus DRGs von transgenen EGFP-Mäusen gewonnene NS-Zellen in utero in die Hirnventrikel von E10.5 Mausembryonen injiziert werden. Die implantierten, grünfluoreszierenden Zellen wurden noch 5 Monate nach dem Eingriff in großer Zahl im Hirngewebe nachgewiesen und konnten als Neurone, Astrozyten und OLPs/Oligodendrozyten identifiziert werden. Die Entstehung neuraler ZNS-Zelltypen beruht hierbei vermutlich auf einer Änderung der regionalen Identität der Zellen durch die NS-Kultivierung. Obwohl sich einige implantierte Zellen auch 5 Monate nach dem Eingriff noch im Zellzyklus befanden, konnte in keinem der analysierten Gehirne hyperplastisches Gewebe oder tumorassoziiertes Wachstum beobachtet werden. Die in dieser Arbeit nachgewiesene Möglichkeit NS-bildende Zellen aus dem PNS zu gewinnen und durch eine selektive Sortierung für O4-positive Gliazellen zusätzlich anzureichern, stellt einen wichtigen Schritt in Richtung gezielter Generation diverser neuronaler und glialer Subtypen des peripheren Nervensystems dar. Zusammen mit der durch die Neurosphären-Kultivierung erzielten Änderung der regionalen Identität muriner NS-Zellen, könnte diese Arbeit außerdem einen wichtigen Grundstein für weiterführende Arbeiten in Richtung Stammzell-Therapien, z.B. in Multiple-Sklerose-Modellen darstellen.
Brustkrebs repräsentiert die häufigste Krebserkrankung bei Frauen. Der Estrogen Rezeptor α (ERα) ist hier als ein wichtiger prognostischer Marker für Mammakarzinome etabliert, der die Homonsensitivität des Tumors determiniert. Dieser impliziert den Einsatz von Anti-Estrogenen, die die wachstumsfördenden Funktionen von ERα blockieren können. Übergewicht erhöht deutlich das Risiko einer Brustkrebserkrankung bei postmenopausalen Frauen. Übergewicht geht generell mit einem erhöhten Serumleptinspiegel einher. Das Zytokinhormon Leptin ist ein zentraler humoraler Faktor bei der Wahrnehmung und Steuerung der körpereigenen Energiereserven im Gehirn, das darüber hinaus auch pleiotrope Funktionen in der Angiogenese, Hämatopoese, Reproduktion und im Immunsystem ausübt. Leptin bindet an den Transmembranrezeptor Ob-RL (Obese-Rezeptor, lange Isoform) und aktiviert unter anderem den Januskinase (Jak)- „signal transducer and activator of transcription“ (Stat)-Signalweg, der mit neoplastischen Veränderungen assoziiert ist. Weil der kausale Zusammenhang zwischen Übergewicht und Krebserkrankungen gut belegt ist, während die molekularen Mechanismen jedoch noch nicht vollständig aufgeklärt sind, sollte daher in der vorliegenden Arbeit die mögliche Wechselwirkung des leptininduzierten Jak-Stat-Signalwegs und des ERα´s untersucht werden. Ein Zusammenspiel der Ob-RL- und ERα-abhängigen Signalwege wurde einerseits mit Luziferase-Reporter-Konstrukten untersucht, die unter der Kontrolle eines Stat3-responsiven Elements standen. Dabei konnten zelltyp-spezifsche Unterschiede in der leptinabhängigen Stat3-Aktivierung beobachtet werden. Die endogen ERα-exprimierenden Brustkrebszellen MCF-7 wiesen eine wesentlich stärkere Stat3-Aktiverung und Phosphorylierung nach Leptinapplikation auf, als ERα-negative HeLa Zervixkarzinomzellen oder die Brustkrebslinie MDA-MB-231. Dieser Effekt konnte durch die Hemmung der ERα-Expression mittels siRNA in MCF-7 Zellen revertiert werden. Die ERα Überexpression in HeLa Zellen resultierte in einer verstärkten Stat3-Transaktiverung nach Leptinbehandlung. Diese Zunahme in der Stat3-Aktivierung konnte nicht durch eine Kostimulation der Zellen mit einem ERα-Agonisten oder Antagonisten beeinflusstwerden. Weiterhin konnte eine gesteigerte Zellviabilität nach Leptinstimulation in ERα-positiven Zellen detektiert werden. Durch Koimmunpräzipitationsstudien konnte direkte Interaktion von ERα, Jak2 und Stat3 nachgewiesen werden, was auf einen zytoplasmatisch lokalisierten Komplex hindeutet. Eine Mikroarray-Analyse von leptinstimulierten Zellen ergab eine differentielle Regulation von 133 Genen in Abhängigkeit ihres ERα-Expressionstatus. Von den 133 Zielgene sind 42 in der Literatur bereits in Zusammenhang mit malignem Wachstum beschrieben und könnten somit die erhöhte Viabilität der ERα-exprimierenden Zellen in Reaktion auf Leptin erklären. Die in dieser Arbeit gewonnen Erkenntnisse deuten auf eine entscheidende Rolle des ERα´s in der Verstärkung der leptininduzierten Stat3-Aktiverung hin. Diese Daten zur Interaktion von ERα mit einem wachstumsfördernden Signalweg können zur Entwicklung neuartiger Behandlungsmöglichkeiten in der Brustkrebstherapie von übergewichtigen Patientinnen beitragen.

Die drei Bereiche, die hier verglichen werden sollen, entsprechen in etwa der überkommenen Trias von Literatur, Musik und bildender Kunst, einer Gliederung, die im Medienzeitalters mit Videos, CDs, Installationen oder Happenings eigentlich obsolet ist. Allerdings geht es hier nur um die Eigenart der Zeichensysteme, auf denen die verschiedenen Bereiche beruhen, nicht um die Werke, die dadurch möglich werden, obgleich natürlich auch die Kunstwerke im emphatischen Sinn, die bedeutenden und die banalen, die großen und die misslungenen Gestaltungen nur möglich und verstehbar sind aufgrund der Zeichen, auf denen sie beruhen.
The main tenet of the present paper is the thesis that nominalization – like other cases of derivational morphology – is an essentially lexical phenomenon with well defined syntactic (and semantic) conditions and consequences. More specifically, it will be argued that the relation between a verb and the noun derived from it is subject to both systematic and idiosyncratic conditions with respect to lexical as well as syntactic aspects.
Venture capital exit rights

Theorists argue that exit rights can mitigate hold-up problems in venture capital. Using a hand-collected data-set of venture capital contracts from Germany we show that exit rights are included more frequently in venture capital contracts when a hold-up problem associated with the venture capitalist's exit decision is likely. Examples include drag-along and tag-along rights. Additionally, we find that almost all exit rights are allocated to the venture capitalist rather than to the entrepreneur. In addition, we show that besides the basic hold-up mechanism there are other mechanisms such as ex-ante bargaining power and the degree of pledgeable income that drive the allocation of exit rights. Governance und Vertragsstrukturen in der deutschen VC Industrie: eine empirische Einschätzung

Diese Arbeit analysiert die wesentlichen Elemente der Vertragsstrukturen in der Venture Capital-Industrie. Aufbauend auf einem sehr umfangreichen und detaillierten Datensatz, der die Verträge zwischen Venture Capital-Investoren und deren Portfoliounternehmen abbildet, werden die Kontroll-, Entscheidungs- und Vetorechte einer detaillierten Analyse unterzogen. Dabei zeigt sich eine klare Tendenz in der Entwicklung der Vertragsstrukturen in Deutschland hin zu angelsächsisch strukturierten Verträgen. Dies beinhaltet unter anderem eine verstärkte Verwendung von Kontroll- und Entscheidungsrechten aber auch ein breiteres Spektrum dieser Rechte. Außerdem finden wir eine klare Interaktionen zwischen Kontrollrechten, Cash-Flow Rechten und Liquidationsrechten. Insbesondere ist zu betonen, dass Cash-Flow und Kontrollrechte einerseits und Stimmrechte und Aufsichtsratsanteile andererseits separat alloziiert werden und viele Kontrollrechte als Komplemente und nicht als Substitute zueinander aufgefasst werden müssen. By adopting a variety of shapes, proteins can perform a wide number of functions in the cell, from being structural elements or enabling communication with the environment to performing complex enzymatic reactions needed to sustain metabolism. The number of proteins in the cell is limited by the number of genes encoding them. However, several mechanisms exist to increase the overall number of protein functions. One of them are post-translational modifications, i.e. covalent attachment of various molecules onto proteins. Ubiquitin was the first protein to be found to modify other proteins, and, faithful to its evocative name, it is involved in nearly all the activities of a cell. Ubiquitylation of proteins was believed for a long time only to be responsible for proteasomal degradation of modified proteins. However, with the discovery of various types of ubiquitylation, such as mono-, multiple- or poly-ubiquitylation, new functions of this post-translational modification emerged. Mono-ubiquitylation has been implicated in endocytosis, chromatin remodelling and DNA repair, while poly-ubiquitylation influences the half-life of proteins or modulates signal transduction pathways. DNA damage repair and tolerance are example of pathways extensively regulated by ubiquitylation. PCNA, a protein involved in nearly all types of DNA transaction, can undergo both mono- and poly-ubiquitylation. These modifications are believed to change the spectrum of proteins that interact with PCNA. Monoubiquitylation of PCNA is induced by stalling of replication forks when replicative polymerases (pols) encounter an obstacle, such as DNA damage or tight DNA-protein complexes. It is believed that monoubiquitylation of PCNA stimulates the exchange between replicative pols to one of polymerases that can synthesize DNA across various lesions, a mechanism of damage tolerance known as translesion synthesis (TLS). Our work has helped to understand why monoubiqutylation of PCNA favours this polymerase switch. We have identified two novel domains with the ability to bind Ub non-covalently. These domains are present in all the members of Y polymerases performing TLS, and were named Ub-binding zinc finger (UBZ) (in polη and polκ) and Ub-binding motif (UBM) (in polι and Rev1). We have shown that these domains enable Y polymerases to preferentially gain access to PCNA upon stalling of replication, when the action of translesion polymerases is required. While the region of direct interaction between Y pols and PCNA had been known (BRCT domain in Rev1 and PIP box motif (PIP) in three others members), we propose that Ub-binding domains (UBDs) in translesion Y pols enhance the PIP- or BRCT-domain-mediated interaction between these polymerases and PCNA by binding to the Ub moiety attached onto PCNA. Following these initial studies, we have also discovered that Y polymerases themselves undergo monoubiquitylation and that their UBDs mediate this modification. This auto-ubiquitylation is believed to lead to an intramolecular interaction between UBD and Ub attached in cis onto the UBD-containing protein. We have mapped monoubiquitylation sites in polη in the C-terminal portion of the protein containing the nuclear localization signal (NLS) and the PIP box. Beside PIP, the NLS motif is also involved in direct interaction of polη with PCNA. Based on these findings, we propose that monoubiquitylation of either NLS or PIP masks them from potential interaction with PCNA. Lastly, using several functional assays, we have demonstrated the importance of all these three motifs in the C-terminus of polη (UBZ, NLS and PIP) for efficient TLS. We have also constructed a mimic of monoubiquitylated polη by genetically fusing polη with Ub. Interestingly, this chimera is deficient in TLS as compared to the wild-type protein. Altogether, these studies demonstrate that the C-terminus of polη constitutes a regulatory module involved in multiple-site interaction with monoubiquitylated PCNA, and that monoubiquitylation of this region inhibits the interaction between polη and PCNA. Our work has also revealed that the UBDs of Y pols as well as of other proteins implicated in DNA damage repair and tolerance, such as the Werner helicase-interacting protein 1 (Wrnip1), are required for their proper sub-nuclear localization. All these proteins localize to discrete focal structures inside the nucleus and mutation of their UBDs results in inability to accumulate in these foci. Interestingly, by exchanging UBDs between different proteins we have learned that each UBD seems to have a distinct functional role, surprisingly not limited to Ubbinding ability. In fact, swapping the UBZ of Wrnip1 with the UBM of polι abolished the localization of Wrnip1 to foci despite preserving the Ub-binding ability of the chimeric protein. In summary, this work provides an overview of how post-translation modification of proteins by Ub can regulate several DNA transactions. Firstly, key regulators (e.g. PCNA) can be differentially modified by Ub. Secondly, specialized UBDs (e.g. UBM, UBZ) embedded only in a subset of proteins act as modules able to recognize these modifications. Thirdly, by means of mediating auto-ubiquitylation, UBDs can modulate the behaviour of host proteins by allowing for either in cis or in trans Ub-UBD interactions.
Seit den 1980er Jahren entwickelte sich in den Literaturwissenschaften die Fragestellung nach den literarischen Konstitutionsbedingungen der Beschreibung fremder Kulturen. Dies untersuchte vor allem die narrativen Entwürfe der Begegnung mit dem Fremden in fiktio-nalen Texten. Das Schaffen Hermann Hesses bildet ein Beispiel dafür, dass unterschiedliche Mentalitäten unterschiedliche Wahrnehmungsformen von Kulturen präformieren.
