Morgan Tsvangirais appointment as Zimbabwes Prime Minister in 2009 followed many years leadership of the Zimbabwe Congress of Trades Unions and the Movement for Democratic Change. How has that experience equipped him for high national office? Does he have the personal, intellectual and political qualities required to be President? In July 2004, as he was awaiting the verdict in his treason trial, Tsvangirai spent several days in conversation with Stephen Chan. Chan was concerned to find out if Tsvangirai was more than merely a charismatic leader of the opposition; if he had his own intellectual agenda [and] political philosophy. His questions were even-handed and astute. Discussion by discussion, Morgan Tsvangirai had become more open, more human less cautious and, paradoxically, more obviously and naturally presidential. Five years later, having reviewed the events since their discussions took place, Chan writes: I have not made a saint of him, not even an Atlas. I hope I have not criticized him too much or too unfairly. Probably no one could have done for Zimbabwe what he has. Citizen of Zimbabwe is a rare and intimate portrait of political leadership in Africa.
Background: Abnormalities of 11q23 involving the MLL gene are found in approximately 10% of human leukemias. To date, nearly 100 different chromosome bands have been described in rearrangements involving 11q23 and 64 fusion genes have been cloned and characterized at the molecular level. In this work we present the identification of a novel MLL fusion partner in a pediatric patient with de novo biphenotypic acute leukemia. Methods: Cytogenetics, fluorescence in situ hybridization (FISH), molecular studies (RT-PCR and LDI-PCR), and bioinformatic sequence analysis were used to characterize the CT45A2 gene as novel MLL fusion partner in pediatric acute leukemia. Results: Fluorescence in situ hybridization of the patient G-banded metaphases demonstrated a cryptic insertion of 11q23 in Xq26.3 involving the MLL gene. Breakpoint fusion analysis revealed that a DNA fragment of 653 kb from 11q23, containing MLL exons 1-9 in addition to 16 other 11q23 genes, was inserted into the upstream region of the CT45A2 gene located at Xq26.3. In addition, a deletion at Xq26.3 encompassing the 3' region of the DDX26B gene (exons 9-16) and the entire CT45A1 gene was identified. RNA analysis revealed the presence of a novel MLL-CT45A2 fusion transcript in which the first 9 exons of the MLL gene were fused in-frame to exon 2 of the CT45A2 gene, resulting in a spliced MLL fusion transcript with an intact open reading frame. The resulting chimeric transcript predicts a fusion protein where the N-terminus of MLL is fused to the entire open reading frame of CT45A2. Finally, we demonstrate that all breakpoint regions are rich in long repetitive motifs, namely LINE/L1 and SINE/Alu sequences, but all breakpoints were exclusively identified outside these repetitive DNA sequences. Conclusion: We have identified CT45A2 as a novel spliced MLL fusion partner in a pediatric patient with de novo biphenotypic acute leukemia, as a result of a cryptic insertion of 11q23 in Xq26.3. Since CT45A2 is the first Cancer/Testis antigen family gene found fused with MLL in acute leukemia, future studies addressing its biologic relevance for leukemogenesis are warranted.
The political, the urban, and the cosmopolitan : the 1970s generation in Romanian-German poetry
(2010)
This study is an introduction to the body of work produced by the German poets who were born during or after World War II in Romania and whose almost simultaneous debut lies in the relatively liberal period 1965 – 1971. Helped onto the Romanian-German literary scene by a propitious environment and informed by the socialist ideology they were born “into,” the poets born between 1942 and 1955 formed a remarkable generation unit which sought to significantly renew German-language literature in Romania. Rejecting identification with the insulary Romanian-German communities, the young poets strove to create a socially and politically relevant verse expressing an urban and cosmopolitan attitude. The growing nationalist rhetoric and isolationist stance of Romania's regime and the material and psychological hardships endured by its population through the 1970s and 80s forced the generation to revise its incipient enthusiasm for Romanian socialism. Increasingly, the poets' work came to depict the threatened existence of the German minority and the harsh general living conditions in Romania and to provide an alternative to the absurd official proclamations of a “golden age” under Ceauşescu, despite the poetry's growing reliance on obscuring literary techniques. The emigration of most of the generation members in the mid to late 1980s brought about the eventual unravelling of the generation unit and marks the end of my study. By following the evolution of three themes – social and political engagement, the German minority, and the urban environment – which define the poets as a generation throughout their literary careers in Romania, the analysis illuminates not only the generation's development from identification with Romanian socialism and rejection of the German minority to criticism of the country's policies and a renewed interest in the fate of the German community but also the changing possibilities and limits of literary expression under communism. In addition to providing an introduction to the body of work created by the 1970s generation in Romania, the study also expands the understanding of German literature in the 20th century by providing new material on literature written under totalitarianism and of intercultural German literature.
Frauen- und Männerstimmen in Medien (Moderatorinnen und Moderatoren in Rundfunk und Fernsehen)
(2010)
Just 30 % of the effect of female and male speakers are activated by the lexems and sentences. 70 % are activated by paraverbal and extraverbal constituents. A deep voice is associated with authority and objectivity. We can realise this phenomenon at male voices. Women never reach such a deepness in their voices Their voice is sensed more expressive and it activates stereotypes like „emotional“ and „trivial“. The contents of female speakers are not taken as seriously as the contents of male speakers.
Since the introduction of radiological examination techniques, such as an angiographic computed tomography (ACT) and a computed tomographic angiography (CTA), the diagnosis and treatment planning for cerebral aneurysms became quicker and safer. Unfortunately, the state of development of these techniques, and their possible indications and benefits are still today restricted to specialized centers. The objective of this update was to analyze the current scientific evidence about their actual interventional and diagnostic use and identify the individual advantages of each through a literature review and personal experience-based data.
Sucrose- and H+-dependent charge movements associated with the gating of sucrose transporter ZmSUT1
(2010)
Background: In contrast to man the majority of higher plants use sucrose as mobile carbohydrate. Accordingly proton-driven sucrose transporters are crucial for cell-to-cell and long-distance distribution within the plant body. Generally very negative plant membrane potentials and the ability to accumulate sucrose quantities of more than 1 M document that plants must have evolved transporters with unique structural and functional features.
Methodology/Principal Findings: To unravel the functional properties of one specific high capacity plasma membrane sucrose transporter in detail, we expressed the sucrose/H+ co-transporter from maize ZmSUT1 in Xenopus oocytes. Application of sucrose in an acidic pH environment elicited inward proton currents. Interestingly the sucrose-dependent H+ transport was associated with a decrease in membrane capacitance (Cm). In addition to sucrose Cm was modulated by the membrane potential and external protons. In order to explore the molecular mechanism underlying these Cm changes, presteady-state currents (Ipre) of ZmSUT1 transport were analyzed. Decay of Ipre could be best fitted by double exponentials. When plotted against the voltage the charge Q, associated to Ipre, was dependent on sucrose and protons. The mathematical derivative of the charge Q versus voltage was well in line with the observed Cm changes. Based on these parameters a turnover rate of 500 molecules sucrose/s was calculated. In contrast to gating currents of voltage dependent-potassium channels the analysis of ZmSUT1-derived presteady-state currents in the absence of sucrose (I = Q/τ) was sufficient to predict ZmSUT1 transport-associated currents.
Conclusions: Taken together our results indicate that in the absence of sucrose, ‘trapped’ protons move back and forth between an outer and an inner site within the transmembrane domains of ZmSUT1. This movement of protons in the electric field of the membrane gives rise to the presteady-state currents and in turn to Cm changes. Upon application of external sucrose, protons can pass the membrane turning presteady-state into transport currents.
Background: Previously, we showed that glioma pathogenesis related protein (GliPR) is induced in CEM T cells upon HIV-1 infection in vitro. To examine whether GliPR plays a role as HIV dependency factor (HDF), we tested the effect of GliPR suppression by siRNA on HIV-1 replication. Results: Induction of GliPR expression by HIV-1 was confirmed in P4-CCR5 cells. When GliPR was suppressed by siRNA, HIV-1 replication was significantly reduced as measured by HIV-1 transcript levels, HIV-1 p24 protein levels, and HIV-1 LTR-driven reporter gene expression, suggesting that GliPR is a cellular co-factor of HIV-1. Microarray analysis of uninfected HeLa cells following knockdown of GliPR revealed, among a multitude of gene expression alterations, a down-regulation of syndecan-1, syndecan-2, protein kinase C alpha (PRKCA), the catalytic subunit beta of cAMP-dependent protein kinase (PRKACB), nuclear receptor co-activator 3 (NCOA3), and cell surface protein CD59 (protectin), all genes having relevance for HIV-1 pathology. Conclusions: The up-regulation of GliPR by HIV-1 and the early significant inhibition of HIV-1 replication mediated by knockdown of GliPR reveal GliPR as an important HIV-1 dependency factor (HDF), which may be exploited for HIV-1 inhibition.
