Zusammenfassung - Das Kernanliegen des KredReorgG –die Internalisierung des systemischen Risikos in den Entscheidungsprozess und die Verantwortlichkeit von Bankeignern und –gläubigern – wird im Wesentlichen erreicht. - Die Wirksamkeit des Gesetzes steht und fällt mit der Möglichkeit, jede Bank in systemisch relevante (zu rettende) und systemisch nicht-relevante (abzuwickelnde) Teile zu zerlegen. Dieser Ansatz ist Ziel führend und international „state of the art“ (Bsp. UK). - Unsere Hauptkritik: Um die o.g. Wirksamkeit des Gesetzes überhaupt zu ermöglichen (und eine Unterlaufung der Gesetzesintention zu verhindern), bedarf es einer zusätzlichen und zwingenden Vorgabe, dass jede Bank eine Mindestmenge an Anleihen außerhalb des Kern-Finanzsektors dauerhaft platzieren muss, und dass diese Anleihen zu keinem Zeitpunkt von Banken erworben werden dürfen. - Um dies zu erreichen sind die Anlagevorschriften für Kapitalsammelstellen (Lebensversicherer, Pensionsfonds) und für Banken entsprechend zu ändern bzw. zu verschärfen. - Weitere Kritikpunkte betreffen die vermutete geringe Bedeutung der freiwilligen Verfahren (Sanierung und Reorganisation) und die Gestaltung der Sonderabgabe und der Restrukturierungsfonds.
The single unit doctrine proposes that each one of our percepts and sensations is represented by the activity of specialized high-level cells in the brain. A common criticism applied to this proposal is the one referred to as the "combinatorial problem". We are constantly confronted with unlimited combinations of elements and features, and yet we face no problem in recognizing patterns and objects present in visual scenes. Are there enough neurons in the brain to singly code for each one of our percepts? Or is it the case that perceptions are represented by the distributed activity of different neuronal ensembles? We lack a general theory capable of explaining how distributed information can be efficiently integrated into single percepts. The working hypothesis here is that distributed neuronal ensembles signal relations present in the stimulus by selectively synchronizing their spiking responses. Synchronization is generally associated with oscillatory activity in the brain. Gamma oscillations in particular have been linked to various integrative processes in the visual system. Studies in anesthetized animals have shown a conspicuous increase in power for the gamma frequency band (30 to 60 Hz) in response to visual stimuli. Recently, these observations have been extended to behavioral studies which addressed the role of gamma activity in cognitive processes demanding selective attention. The initial motivation for carrying out this work was to test if the binding-by-synchronization (BBS) hypothesis serves as a neuronal mechanism for perceptual grouping in the visual system. To this aim we used single and superimposed grating stimuli. Superimposed gratings (plaids) are bi-stable stimuli capable of eliciting different percepts depending on their physical characteristics. In this way, plaids can be perceived either as a single moving surface (pattern plaids), or as two segregated surfaces drifting in different directions (component plaids). While testing the BBS hypothesis, we performed various experiments which addressed the role of both stimulus and cortical architecture on the properties of gamma oscillations in the primary visual cortex (V1) of monkeys. Additionally, we investigated whether gamma activity could also be modulated by allocating attention in time. Finally, we report on gamma-phase shifts in area V1, and how they depend on the level of neuronal activation. ...
Die vorliegende Dissertation analysiert Großinvestorhandelsstrategien in illiquiden Finanzmärkten. Ein Großinvestor beeinflusst die Preise der Wertpapiere, die er handelt, so dass der daraus resultierende Feedbackeffekt berücksichtigt werden muss. Der Preisprozess wird durch eine Familie von cadlag Semimartingalen modelliert, die in dem zusätzlichen Parameter stetig differenzierbar ist. Ziel ist es, eine möglichst allgemeine Strategiemenge zu bestimmen, für die eine Vermögensdynamik definiert werden kann. Es sind dies vorhersehbare Prozesse von wohldefinierter quadratischer Variation entlang Stoppzeiten. Sie erweisen sich als laglad. Die Vermögensdynamikzerlegung zeigt, dass bei stetigen adaptierten Strategien von endlicher Variation (zahme Strategien) die quadratischen Transaktionskostenterme verschwinden und der Gewinnprozess nur noch aus einem nichtlinearen stochastischen Integral besteht. Es wird gezeigt, unter welchen Bedingungen gewisse Approximationen vorhersehbarer laglad Strategien durch adaptierte stetige Strategien von endlicher Variation möglich sind. Im Fall, dass der Approximationsfehler für die Risikoeinstellung des Großinvestors erträglich ist, kann er seine Investmentziele durch Verwendung dieser zahmen Strategien, Liquiditätskosten vermeidend, erreichen. In diesem Fall ist der Gewinnprozess durch das nicht-lineare stochastische Integral gegeben.
Stop Making Sense : USA 1984

Die Aufzeichnungen zu "Stop Making Sense" fanden an drei Abenden im Dezember 1983 im Pantages Theatre in Hollywood statt. David Byrne und seine Band machten dort im Rahmen der Promotiontour zu ihrem damals neuem Album 'Speaking in Tounges' Station. Als Regisseur holten sich die Talking Heads den filmerfahrenen Jonathan Demme, der später auch Blockbuster wie "The Silence of the Lambs" und "Philadelphia" schuf, und den Kameramann Jordan Cronenweth (Blade Runner) in die Tourneecrew. "Stop Making Sense" ist Demmes Dokumentarfilm-Debüt.

Stranger in his Homeland

Stranger in His Homeland completes the long-awaited trilogy of Linus Asong's fictitious village of Nkokonoko Small Monje, separately treated in The Crown of Thorns and its sequel A Legend of the Dead. However, it leads us back not to events after A Legend of the Dead, but to the crisis that created the passionately exciting The Crown of Thorns. Honest, enthusiastic, arrogant and self-righteous, Antony Nkoaleck, the first graduate of his tribe means well. But his society, entrenched in corruption, sees things differently and therefore judges him according to its own norms. Just one or two errors on Antony's part are enough to cost him his job with the government, the coveted throne of Nkokonoko Small Monje, and finally his life. It is a sad story, strongly reminiscent of Myshkin's fate in Dostoevysky's novel The Idiot, a story in which the Russian novelist vividly shows the inability of any man to bear the burden of moral perfection in an imperfect world.
The article analyses A. Boissier's image "Les Amants électrisés par l'amour" in view of the larger question of how something is able to arouse interest on first sight, but also in repeat encounters. Highlighting the engraving's didactic iconography, the article shows how it revolves around the solution to a riddle and uses a typical design of the Enlightenment to show the uncovering of a deception. As such, the engraving is part of a long tradition of showing (supposedly) supernatural events, more specifically the tradition of Magia naturalis. At the same time, the image contains dissonances and can be seen to simulate suspense through dichotomies that can be identified as antagonistic historical concepts. The article furthermore discusses the amalgamation of love and electricity in contemporary discourses and addresses the temporal dimension of the engraving, which constructs itself out of an absence, out of something yet unseen.
Strengthening popular participation in the African Union : a guide to AU structures and processes

The African Union (AU) has committed to a vision of Africa that is 'integrated, prosperous and peaceful - driven by its own citizens, a dynamic force in the global arena' (Vision and Mission of the African Union, May 2004). This guide is an effort to take up the challenge of achieving this vision. It is a tool to assist activists to engage with AU policies and programmes. It describes the AU decision-making process and outlines the roles and responsibilities of the AU institutions. It also contains a sampling of the experiences of those non-governmental organisations (NGOs) that have interacted with the AU.
This European Policy Analysis discusses the need to strengthen the institutions underpinning the euro and makes several policy recommendations. The Stability and Growth Pact must be reinforced, have greater automaticity and entail graduated sanctions. Fiscal surveillance must be improved through the establishment of a European Fiscal Stability Agency. Finally, the European Financial Stability Facility must be made permanent.
Employing NMR spectroscopy, it is not only possible to calculate the three dimensional structures of single proteins, but also to study dynamics and conformational changes of protein-complexes. In fact that is an important aspect, since the protein function depends on dynamics and interactions with other molecules. Therefore the study of protein-protein interactions is of highest importance for a better understanding of biological processes. Based on NMR methods, in this thesis we were able to determine protein-protein interactions within the enterobacterial Rcs signalling complex which is regulated via a phosphorelay. Originally identified as regulator of capsule synthesis, the Rcs phosphorelay is now considered to be implicated in stress response caused by disturbances in the peptidoglycan layer. Beyond that the Rcs system is involved in multiplex transcriptional networks including cell division, motility, biofilm formation and virulence. Because of such global nature and its extraordinary structural organisation involving membrane integrated sensor proteins (RcsC, RcsD), coactivators (RcsF, RcsA) and a transcription factor (RcsB), the Rcs system is one of the most remarkable phosphorelays in the family of enterobacteriacaea. During the complex phosphotransfer the histidine phosphotransferase (HPt) domain of the intermediary RcsD protein mediates the phosphotransfer between RcsC and RcsB, and probably modulates the phosphorylation state of the response regulator RcsB. Therefore the present work has been focused on the interface between RcsD and RcsB in more detail. In the first part of the thesis a new domain within the RcsD protein has been identified and structurally analysed by liquid NMR spectroscopy. RcsD is an inner membrane bound hybrid sensor like-kinase composed of a periplasmic sensor domain and a cytoplasmic portion. The cytoplasmic part contains the histidine like-kinase (HK) domain and the histidine phosphotransferase (HPt) domain. By analysis of the secondary structure in more detail, it was shown here that the two domains are intermitted by an additional 13.3 kDa domain. Corresponding to the position of the ABL (α−β−loop) domain of RcsC, located C-terminal to the RcsC-HK domain, the new identified domain was named RcsD-ABL. The central structural element of RcsD-ABL is a β-sheet composed of six strands with a β1−β2−β3−β4−β6−β5 topology and surrounded by two α-helices α1 and α2. In the second part of the thesis, RcsD-ABL is identified as a binding domain for the response regulator RcsB by NMR titration experiments. Such a binding domain for a response regulator has so far only been described for the histidine kinase CheA. In reportergene assays with β-galactosidase and ONPG as substrate it was shown that overexpression of RcsD-ABL in high amounts inhibited binding of RcsB to its target promoter. The β-galactosidase activity was reduced by 80 % with respect to cells carrying no plasmid encoding RcsD-ABL. The mapping of the binding interface was successfully achieved by chemical shift perturbations, a fast mapping protocol and selective labelling. It was shown that the interaction between RcsD-ABL and RcsB takes place via a binding interface comprising mainly the two α-helices of RcsD-ABL and the α-helices α7, α8 and α10 in the effector domain of RcsB. In the third part of the thesis, the interaction of RcsB with RcsD-ABL was related to that with RcsD-HPt. Using NMR titration experiments and ITC measurements, a comparison of the binding constants (Kd) of RcsB interacting either with the isolated RcsD-ABL (2 PM) or the isolated RcsDHPt domain (40 PM) revealed a higher affinity of RcsD-ABL to RcsB. A conjugate of RcsD-ABL-HPt interacting with RcsB decreased the Kd in the one-site fitting mode to 10 PM. However, the two-site fitting mode applied for RcsD-ABL-HPt/RcsB interaction resulted in a Kd (RcsD-ABL) of 2 PM and a Kd (RcsD-HPt) of 8 PM, indicating that RcsD-ABL enhances the binding of RcsD-HPt to RcsB. In the last part of the thesis, it was partly possible together with the data obtained from NMR titration experiments, PRE measurements and a HADDOCK protocol to develop a geometrical model for the interaction of RcsD with RcsB. In this model the receiver domain of RcsB interacts with the RcsD-HPt domain and the RcsB effector domain interacts with the RcsD-ABL domain. These results lead to surprising insights on the regulation of phosphorelays, since normally the effector domain binds to DNA. Here the effector domain is recognized by the newly identified RcsD-ABL domain. Prospectively, further investigations of phosphorylation affects and mutational studies will be of great interest.
Human Transformer2-beta (hTra2-beta) is an important member of the serine/arginine-rich protein family, and contains one RNA recognition motif (RRM). It controls the alternative splicing of several pre-mRNAs, including those of the calcitonin/calcitonin gene-related peptide (CGRP), the survival motor neuron 1 (SMN1) protein and the tau protein. Accordingly, the RRM of hTra2-beta specifically binds to two types of RNA sequences [the CAA and (GAA)2 sequences]. We determined the solution structure of the hTra2-beta RRM (spanning residues Asn110–Thr201), which not only has a canonical RRM fold, but also an unusual alignment of the aromatic amino acids on the beta-sheet surface. We then solved the complex structure of the hTra2-beta RRM with the (GAA)2 sequence, and found that the AGAA tetra-nucleotide was specifically recognized through hydrogen-bond formation with several amino acids on the N- and C-terminal extensions, as well as stacking interactions mediated by the unusually aligned aromatic rings on the beta-sheet surface. Further NMR experiments revealed that the hTra2-beta RRM recognizes the CAA sequence when it is integrated in the stem-loop structure. This study indicates that the hTra2-beta RRM recognizes two types of RNA sequences in different RNA binding modes.
Der L-Carnitin/gamma-Butyrobetain Antiporter CaiT ist ein Mitglied der Betain/Carnitin/Cholin Transporter (BCCT) Familie. Sekundärtransporter der BCCT Familie transportieren Substrate, die eine positiv-geladene quartäre Ammoniumgruppe besitzen. CaiT besteht aus 504 Amiosäuren und besitzt ein moleculares Gewicht von etwa 56 kDa. In Enterobakterien wie Escherichia coli, Proteus mirabilis und Salmonella typhimurium wird die Expression des caiTABCDE Operons unter anaeroben Bedingungen induziert. Unter diesen Bedinungen ist CaiT der Haupttransporter des Betain-Derivates L-Carnitin. In Enterobakterien wird L-Carnitin unter anaeroben Bedingungen aufgenommen und dehydratisiert wobei Crotonobetain ensteht. Crotonobetain wird anschließend zum Endprodukt gamma-Butyrobetain reduziert. Gamma-Butyrobetain ist das Gegensubstrat, das aus der Zelle hinaustransportiert wird, wenn L-Carnitin in die Zelle aufgenommen wird. Der Austauschmechanismus von LCarnitin gegen gamma-Butyrobetain geschieht ohne das Vorhandensein eines elektrochemischen Gradients, d.h. CaiT ist sowohl H+- als auch Na+-unabhängig. Ein Ziel dieser Arbeit war es die drei-dimensionale (3D) Struktur von CaiT mittels Röntgenstrukturanalyse zu lösen. Weiterhin sollten mit Hilfe der 3D-Struktur und funktionellen Studien detailiertere Erkenntnisse über den kationenunabhängigen Antiportmechanismus von CaiT ermittelt werden. Im Rahmen dieser Arbeit wurden die 3D-Röntgenkristallstrukturen von drei CaiT-Homologen der Enterobakterien P. mirabilis (PmCaiT), E. coli (EcCaiT) und S. typhimurium (StCaiT) mittels molekularem Ersatz (engl.: molecular replacement, MR) mit einem Alanin-Model des CaiT verwandten Na+/Glycinbetain Symporters BetP gelöst. PmCaiT konnte mit einer Auflösung von 2.3 Å gelöst werden. Das Protein kristallisierte in der Kristallraumgruppe H3, mit drei Molekülen in der asymmetrischen Einheit (engl.: asymmetric unit, AU). Die drei PmCaiT-Moleküle ordneten sich innerhalb der AU um eine kristallographische dreifach Symmetrieachse an. EcCaiT wurde mittels MR mit einem Alanin-Model von PmCaiT bei einer Auflösung von 3.5 Å gelöst. EcCaiT kristallisierte in der Kristallraumgruppe P32, ebenfalls mit drei Molekülen in der AU, jedoch ohne kristallographische Symmetry. Während der Verfeinerung des EcCaiT-Models wurde eine strenge dreifache nichtkristallographische Symmetry (engl.: non-crystallographic symmetry, NCS) angewandt. StCaiT, das ebenfalls mittels MR mit einem Alanin-Model von PmCaiT, aber bei einer Auflösung von 4.0 Å gelöst wurde, kristallisierte in der Kristallraumgruppe P65, ebenfalls mit drei StCaiT-Molekülen in der AU, ohne kristallographische Symmetry. Bei der Verfeinerung des StCaiT-Modells wurde wie bei EcCaiT eine strenge NCS angewandt. Da die Auflösung von 4.0 Å bei StCaiT zu niedrig ist um detailierte moleculare Erkenntnisse zu gewinnen, wurden Protein- sowie Substratinteraktionen nur an den Strukturen von PmCaiT und EcCaiT analysiert. Alle drei CaiT-Homologe weisen jedoch einen ähnlichen strukturellen Aufbau auf. In der Röntgenkristallstruktur bildet CaiT ein symmetrisches Trimer, das über ionische und polare Wechselwirkungen zwischen den Protomeren stabilisiert wird. Der trimere Oligomerisierungszustand von CaiT in Detergenzlösung sowie in zweidimensionalen Lipidmembrankristallen wurde bereits in früheren Arbeiten gezeigt. Jedes der drei CaiT-Protomere besteht aus zwölf Transmembranhelices (TMH), die N- und C-terminalen Domänen des Proteins befinden sich auf der cytoplasmatischen Seite. Zehn der TMH bilden zwei invertierte Wiederholungseinheiten aus jeweils fünf TMH. Die erste Einheit besteht aus den TMH 3 – 7, die invertierte zweite Einheit besteht aus den TMH 8 – 12. Beide Wiederholungseinheiten sind strukturell nahezu identisch und lassen sich fast vollständig übereinanderlegen, jedoch weisen die Aminosäuren der beiden Einheiten keine signifikante Sequenzidentität auf. Die ersten beiden Helices der Wiederholungseinheiten, die TMH 3 – 4 und die TMH 8 – 9, bilden ein antiparalleles vier-Helix-Bündel, in dem in CaiT zwei Substratbindestellen lokalisiert sind. Eine derartige Transporterarchitektur wurde erstmals in der Struktur des Na+/Alanin Symporters LeuTAa des thermophilen Bakteriums Aquifex aeolicus gezeigt. Bislang wurden, inklusive CaiT, sieben Sekundärtransporterstrukturen gelöst, die diese LeuT-Transporterarchitektur aufweisen. Ungewöhnlich dabei ist, dass diese sieben Sekundärtransporter fünf verschiedenen Transporterfamilien angehören und eine Verwandschaft auf Basis der Aminosäuren nicht zu finden ist. Da jedoch die tertiäre Struktur dieser Tansporter konserviert ist, kann davon ausgegangen werden, dass sie alle von einem Urprotein entstanden sind, welches zunächst aus fünf TMH bestanden haben muss. Im Laufe der Evolution hat sich das Urgen des Urproteins zunächst dupliziert und die weitere Evolution hat zwar die Aminosäuresequenz verändert und den Umweltbedingungen angepasst, jedoch ist die tertiäre Struktur erhalten geblieben. Da sich die tertiäre Struktur der sieben Sekundärtransporter so stark ähnelt, ist zu vermuten, dass auch der Transportmechanismus ähnlich, jedoch nicht identisch ist. Nach dem strukturellen Aufbau der Transporter, der Lage der Substratbindestellen in den jeweiligen Transportern und der Tatsache, dass es sich bei diesen Proteinen um Membranproteine handelt, wurde ein Transportmechanismus aufgestellt, in dem die Bindestelle des zu transportierende Substrats alternierend zu beiden Seiten der Membran zugänglich ist, ohne jedoch jemals den Substratweg innerhalb des Proteins vollständig zu öffnen. Dieser Mechanismus wurde als “alternating access mechanism” beschrieben. Anhand der unterschiedlichen Zustände, in denen einige der Transporter kristallisierten, kann abgeleitet werden, welche Konformationsänderungen erforderlich sind um das Substrat von einer Seiter der Membran auf die andere zu transportieren. Bisher kristallisierten einzelne der sechs Transporter in der nach außen gerichteten offenen Form, der nach außen gerichteten Form, in der die Substratbindestelle jedoch nicht mehr zugänglich ist, in einer Form, die keine Öffnungspräferenz der Substratbindestelle zu einer Seite der Membran hat und in der nach innen gerichteten Form, in der die Substratbindestelle jedoch nicht geöffnet ist. CaiT kristallisierte in der noch fehlenden Konformation, der nach innen gerichteten Form, in der die Substratbindestelle zugänglich ist. Mit dieser noch fehlenend Konformation kann der Transportzyklus des “alternating access mechanism” vollständig beschrieben werden. Alle drei CaiT-Homologe kristallisierten in der nach innen gerichteten, offenen Konformation. Im Gegensatz zur EcCaiT-Struktur kristallisierte PmCaiT in der substratungebundenen Form. In der StCaiT-Struktur konnte aufgrund der niedrigen Auflösung kein Substrat nachgewiesen werden. In der EcCaiT-Struktur sind zwei gamma-Butyrobetain-Moleküle gebunden. Das erste Molekül wurde in der zentralen Substratbindestelle, der sogenannten Tryptophan-Box bestehend aus vier Tryptophanen, im Zentrum des Protein lokalisiert. Das zweite gamma-Butyrobetain-Molekül wurde in einer Vertiefung an der extrazellulären Proteinoberfläche gefunden. Beide Substrate werden hauptsächlich über Kation-Pi-Interaktionen zwischen der positiv geladenen quatären Ammoniumgruppe des Substrats und des Pi-Elektronensystems der Tryptophane in den jeweiligen Bindestellen gebunden. Eine besondere Eigenschaft von CaiT ist der H+- bzw. Na+-unabhängige Substrattransport. Die CaiT-Struktur erklärt warum kein zusätzliches Kation benötigt wird um Substrat zu binden oder zu transportieren. In der EcCaiT-Struktur ist eine wichtige polare nicht-bindende Interaktion zwischen der Carboxylgruppe des gamma-Butyrobetains und dem Schwefelatom eines Methionins in der zentrale Bindestelle zu erkennen. Dieses Methionin ist konserviert in den prokaryotischen CaiTs und in den Na+-unabhängigen eukaryotischen L-Carnitin Transportern (OCTN), jedoch ist es nicht konserviert im Na+-abhängigen verwandten Glycinbetain Transporter BetP. In BetP ist diese Position des Methionins durch ein Valin ersetzt. Die Mutation des Methionins in CaiT zu Valin ermöglicht zwar immernoch die H+- bzw. Na+-unabhängige Bindung des Substrates durch die Tryptophan-Box, jedoch ist der Substrattransport nahezu vollständig zerstört. Eine derart wichtige Substratkoordinierende Funktion des Schwefelatoms eines Methionins wurde bisher nicht beschrieben. Eine weitere Stelle, die in H+- bzw. Na+-abhängigen Transporter mit H+ bzw. Na+ besetzt ist, ist in CaiT von einem positiv geladenen Arginin eingenommen. Eine positive Ladung an dieser Stelle stabilisiert den Bereich im Protein in der Nähe der zentralen Substratbindestelle. Die Mutation des Arginins zu Glutamat in CaiT erzielt eine vollständige Inaktivierung des Substrattansports. Durch Zugabe von Na+ im Transportansatz kann die Substrattransportaktivität der Glutamat-Mutante jedoch teilweise zurückerlangt werden. Diese eben beschriebenen Aminosäurereste in den beiden Stellen des Proteins erklären die Kationenunabhängigkeit von CaiT. Die Aktivierung des Antiportmechanismus in CaiT wurde mit Hilfe von Bindungsstudien an rekonstituiertem Protein ermittelt. Diese Messungen ergaben für das Wildtypprotein ein sigmoidales Substratbindungsverhalten, was auf ein positiv-kooperatives Bindungsverhalten hindeutet. Die beiden Substratbindestellen im Protein sowie die beiden unterschiedlichen Substrate, L-Carnitin und gamma-Butyrobetain, lassen auf einen heterotropen positiv-kooperativen Bindungs- und einen allosterisch regulierten Transportmechanismus schließen. Bei diesem Mechanismus erhöht die Bindung eines Substrats in der regulatorischen Bindestelle durch induzierte Konformationsänderungen die Affinität eines anderen Substrats in einer weiteren Substratbindestelle. Die regulatorische Bindestelle in CaiT befindet sich an der extrazellulären Proteinoberfläche. Eine Schwächung der Substrataffinität in dieser Bindestelle durch Einführung einer Mutation, verstärkt das sigmoidale Substratbindungsverhalten und hat einen negativen Einfluss auf den Substrattransport. Durch die in dieser Arbeit gelösten 3D-Röntgenkristallstrukturen der zwei CaiT-Homologen, PmCaiT und EcCaiT, sowie den durchgeführten funktionellen Studien sowohl an Wildtypprotein wie auch an Mutanten konnte ein L-Carnitin/gamma-Butyrobetain Antiport-Mechanismus für CaiT vorzuschlagen werden.
Transmissible spongiform encephalopathies (TSEs) are rare but fatal neurodegenerative diseases affecting human and animals. The prion protein which is the causative agent, according to “protein-only” hypothesis misfold in to rogue amyloid conformer. Despite several years of studies, the atomic structural details of the rogue conformers have not been clearly understood. This study focused on developing an in-vitro conversion method, which allows us to monitor the transition from unfolded state of prion protein to fibril state. In order to reach maximal unfolded state, we have used 8 M urea as chemical denaturant, pH 2 and prion fragment 90-230 as the model. It has been demonstrated earlier that acidic pH and mild denaturant induce the fibril formation. The mechanism underlying the structural transition from monomeric state to polymeric form is largely unknown. We have confirmed by EM and AFM that fibrils are formed in our conditions, which resemble to naturally occurring fibrils in morphologies observed. The agitation accelerates the rate of fibril formation and, which allow us to do time-resolved NMR on these preparations. The conformational flexibility is inherent to amyloid fibrils and has been observed in our preparations. We aimed to map the important segment of prion protein, which forms the rigid core in its fibrillar structured form. Our time-resolved NMR studies allowed us to monitor the changes happening from unfolded state to fibrillar state. Analysis of data identified the segment between residues 145 to 223 forming the rigid core in these fibrils, which correspond to β strand 2, helix 2 and major part of helix 3 of native prion monomeric structure. Most of the point mutations which are associated with hereditary prion disease are part of rigid core, which undergo a refolding on fibril formation. The C-terminal residues from 224 to 230 displayed peak shifting and therefore, indicate the adaptation to a fibril specific conformation. The major part of N-terminal 90-144 segment, remains dynamic, which can be understood by their accessibility to amyloid specific antibodies. This provides novel structural insight to the amyloid formation from unfolded state of prion protein fragment 90-230, which represents the proteinase-K resistant part naturally occurring prions. Earlier studies have established the core to 160-220 where hydrogen-deuterium exchange mass spectrometry or site-directed spin labeling EPR spectroscopy was used for analysis. Those studies have been initiated from either native-like or partially unfolded state of recombinant prion protein, and therefore, it is quite striking to find out that fibrils initiated from unfolded monomeric state share the same “amyloid core”. This structural insight has important implications for understanding the molecular basis of prion propagation.

Das detaillierte Verständnis der zu Grunde liegenden Regulationsmechanismen beinhaltet die Charakterisierung von beteiligten RNA-Architekturen und deren funktionaler Stabilitäten, von dynamischen Aspekten der RNA-Faltungsprozesse sowie die Korrelation dieser Charakteristika mit zellulären Funktionen. Die Genexpression in prokaryotischen Organismen unterliegt einer Vielzahl von Regulationsmechanismen, deren Aufgabe darin besteht, die Zelle an sich ändernde Umweltbedingungen anzupassen, um so das Überleben des prokaryotischen Organismus zu gewährleisten. Eine Reihe von Hitzeschock- und Virulenzgenen unterliegen temperaturabhängiger Regulation, mit dem Ziel, die Zelle an die sich ändernde Umgebung anzupassen. Die Messung der Temperatur erfolgt dabei über temperatursensitive RNA-Elemente, sogenannte RNA-Thermometer, die sich üblicherweise in der 5’-untranslatierten Region der Gene befinden, die sie regulieren. Sie unterdrücken die Translationsinitiation, indem sie die Shine-Dalgarno (SD)-Sequenz bei niedrigen Temperaturen über Basenpaarung blockieren und dadurch die Bindung des Ribosoms verhindern. In Kapitel 2 der vorliegenden Arbeit wurde die thermodynamische Stabilität der temperatursensitiven Haarnadelschleife 2 des Salmonella FourU RNA-Thermometers über einen breiten Temperaturbereich analysiert. Freie Enthalpie-, Enthalpie- und Entropie-Werte für die Basenpaaröffnung der einzelnen Nukleobasen innerhalb der RNA wurden über die temperaturabhängige Messung von Iminoprotonen-Austauschraten mittels NMR-Spektroskopie bestimmt. Die Austauschraten wurden für die Wildtyp-RNA und die A8C-Mutante bestimmt und miteinander verglichen. Es zeigte sich, dass die Wildtyp-RNA durch das außergewöhnlich stabile Basenpaar G14-C25 stabilisiert wird. Dies konnte durch die Untersuchung der Entfaltung der destabilisierenden G14A-C25U-Doppelmutante verifiziert werden. Über CD-spektroskopsiche Untersuchungen konnte der globale Entfaltungsübergang der jeweiligen RNA analysiert werden. Das Mismatch-Basenpaar innerhalb des Wildtyp-RNA-Thermometers (A8-G31) erwies sich als Ursache für die geringere Kooperativität des Entfaltungsübergangs der Wildtyp-RNA im Vergleich zur A8C-Mutante. Enthalpie- und Entropie-Werte für die Basenpaaröffnung einzelner Nukleotide sind für beide RNAs linear korreliert. Die Steigungen dieser Korrelationen stimmen mit den Schmelzpunkten der RNAs überein, die über CD-Spektroskopie bestimmt wurden. Entfaltung der RNA tritt also genau dann auf, wenn alle Nukleotide gleiche thermodynamische Stabilitäten besitzen. Die Resultate sind mit einem Reißverschluss-Mechanismus für die RNA-Helix Entfaltung konsistent und erklärbar, in dem die Stapelinteraktionen der benachbarten Nukleobasen innerhalb der RNA-Helix verantwortlich für die beobachtete Kooperativität sind. Die Ergebnisse weisen auch auf die Wichtigkeit der RNA-Lösungsmittel-Interaktion für die Stabilität der RNA-Struktur hin. So konnten langreichweitige Wechselwirkungen der A8C-Mutation auf die Stabilität der G14-Nukleobase identifiziert werden, die möglicherweise über die Hydrathülle der RNA vermittelt werden. Schließlich konnte für das FourU-Motiv eine Mg2+-Bindestelle identifiziert werden, die die temperaturabhängige Stabilität des RNA-Thermometers beeinflusst. Es besteht also die Möglichkeit, dass Änderungen der intrazellulären Mg2+-Konzentration die Expression des agsA-Gens in vivo modulierend beeinflussen. In Kapitel 3 dieser Arbeit wurden die dynamischen Eigenschaften des Phosphodiesterrückgrats einer perdeuterierten cUUCGg-Tetraloop-14mer-RNA untersucht. Dazu wurden die Relaxationseigenschaften aller 31P-Kerne dieser RNA bei magnetischen Feldstärken von 300, 600 und 900 MHz untersucht. Dipolare Relaxationsbeiträge konnten unterdrückt werden, indem eine perdeuterierte RNA-Probe in einem D2O-Puffer verwendet wurde. Um die 31P-Relaxationsdaten (R1, R2) interpretieren zu können, wurde zusätzlich mittels Festkörper-NMR die Chemische Verschiebungsanisotropie (CSA) der 31P-Kerne des Phosphodiesterrückgrats bestimmt. Die Messungen wurden bei verschiedenen Salzkonzentrationen und unter unterschiedlichen Hydratationsbedingungen durchgeführt. Aus den Daten konnte ein 31P-CSA-Wert von 178.5 ppm im statischen Zustand (S2 = 1) bestimmt werden. Auf der Grundlage der durchgeführten R1- und R2-Messungen wurde eine Modelfree-Analyse durchgeführt, um Informationen über die schnellen Dynamiken des Phosphodiesterrückgrats zu erhalten. Die Resultate zeigen, dass die Dynamiken des Phosphodiesterrückgrats auf der Subnanosekundenzeitskala stärker ausgeprägt sind als die Dynamiken der Ribofuranosylreste und der Nukleobasen. Des Weiteren konnte gezeigt werden, dass die Dynamik einer individuellen Phosphatgruppe zu der jeweiligen 5’-benachbarten Nukleobase korreliert ist. In Kapitel 4 dieser Arbeit wird die Entwicklung neuer Methoden beschrieben, mit denen Torsionswinkelinformation aus der Analyse kreuzkorrelierter Relaxationsraten gewonnen werden können. Im ersten Teil des Kapitels wird die Entwicklung einer neuen NMR-Pulssequenz beschrieben, über die der glykosidische Torsionswinkel Chi in 13C,15N-markierten Oligonukleotiden bestimmt werden kann. Mit dem neuen quantitativen Gamma-HCNCH-Experiment ist es möglich, die dipolaren kreuzkorrelierten Relaxationsraten Gamma-C6H6-C1´H1´ (Pyrimidine) und Gamma-C6H6-C1´H1´ (Purine) zu messen. Die kreuzkorrelierten Relaxationsraten wurden an einer 13C,15N-markierten cUUCGg-Tetraloop-14mer-RNA bestimmt. Die aus den Raten extrahierten Chi-Winkel wurden mit bereits vorhandener Strukturinformation verglichen. Sie stimmen bemerkenswert gut mit den Winkeln der Kristallstruktur des Tetraloops überein. Zusätzlich wurde die neue Methode an einer größeren 30mer-RNA, dem „Stemloop D“ (SLD) aus dem Coxsackievirus-B3-Kleeblatt, getestet. Für die SLD-RNA wurde der Effekt von anisotroper Rotationsdiffusion auf die Relaxationsraten untersucht. Es konnte gezeigt werden, dass die Chi-Winkelbestimmung besonders für Nukleotide in der anti-Konformation sehr genau ist und die Methode eine eindeutige Unterscheidung von syn- und anti-Konformation zulässt. Im zweiten Teil von Kapitel 4 wird die Entwicklung des Gamma-HCCCH-Experiments beschrieben. Hierbei handelt es sich um eine neue NMR-Pulssequenz zur Messung der Gamma-C1´H1´-C3´H3´-Rate in 13C-markierten RNAs. Die Funktionsfähigkeit der neuen Methode wurde an einer cUUCGg-Tetraloop-14mer-RNA demonstriert. Zusätzlich dazu wurden die analytischen Gamma-C1´H1´-C3´H3´(P,nü_max)-, Gamma-C1´H1´-C4´H4´(P,nü_max)- und Gamma-C2´H2´-C4´H4´(P,nü_max)-Abhängigkeiten mathematisch hergeleitet. Die an der 14mer-RNA gemessenen Gamma-C1´H1´-C3´H3´-Raten wurden mit Hilfe der Gamma-C1´H1´-C3´H3´(P,nü_max)-Beziehung analysiert. Die Ergebnisse für die Pseudorotationsphase P sind konsistent mit Referenzwinkeln aus der 14mer-NMR-Struktur und den bereits bekannten (Gamma-C1´H1´-C2´H2´)/(Gamma-C3´H3´-C4´H4´)-Ratenverhältnissen. Die neue Methode liefert zusätzliche Informationen, um Konformation (P, nü_max) und Dynamik S2(C1´H1´-C3´H3´) der Ribosereste in RNA-Molekülen genauer beschreiben zu können. In Kapitel 5 dieser Arbeit wird die Entwicklung des 3D-HNHC-Experiments, einer neuen NMR-Pulssequenz, beschrieben. Dieses Experiment ermöglicht es, die H2-, C2- und N1-Resonanzen in Adenin-Nukleobasen 13C, 15N-markierter RNA-Oligonukleotide miteinander zu korrelieren. Die Funktionsfähigkeit der neuen Methode wurde an einer mittelgroßen, entsprechend markierten 36mer-RNA demonstriert. Die neue Methode vereinfacht die Zuordnung der Kerne der Adenin-Nukleobasen, da Zuordnungsmehrdeutigkeiten aufgrund überlappender Resonanzen in der 1H-Dimension aufgelöst werden können. In Kombination mit dem TROSY-relayed-HCCH-COSY-Experiment liefert das neue 3D-HNHC-Experiment das fehlende Glied für die Zuordnung der Imino-H3-Resonanzen der Uracil-Nukleobasen über das AU-Basenpaar hinweg zu den H8-Resonanzen der Adenin-Nukleobasen.
Gradation and intensification can be subject to different linguistic procedures. The present paper focuses on aspects of intensification and gradation by means of word formation in adjectives. The argument is that German and Romanian, as languages belonging to different language families, operate with different means to express high intensity in adjectives. The critical analysis on dictionary items (German-Romanian) aims at disclosing how mental representations are being formally structured at language level. It argues that correct equivalence in translation work can be achieved only if the deep structure level is properly understood by the translator.
Die Diskussion um die Patientenverfügung in der Bundesrepublik Deutschland geht bis in die 1970er Jahre zurück, seit Mitte der 90er Jahre nahm sie an Intensität zu. Meilensteine ihrer Entwicklung waren die Entscheidungen des BGH im Kempten-Fall (1994), im Frankfurter Fall (1998) und im Lübeck-Fall (2003). Nach dem Lübeck-Fall mehrten sich die Stimmen, wonach der Gesetzgeber das Betreuungsrecht ändern und die Patientenverfügung gesetzlich verankern sollte. Ein Entwurf des Bundesjustizministeriums aus dem Jahr 2004 erreichte jedoch nicht den Deutschen Bundestag, er wurde dem Gesetzesentwurf unter der Leitung von Joachim Stünker angegliedert. Erst Anfang des Jahres 2007 erhielt die Diskussion mit einer Orientierungsdebatte Einzug in das Deutsche Parlament. In den Jahren 2007 bis 2008 wurden dann drei Gesetzesentwürfe zum Thema Patientenverfügung in den Bundestag eingebracht. Der erste Entwurf wurde im Mai 2007 unter Federführung von Wolfgang Bosbach (CDU) veröffentlicht. In ihm waren Formvorschriften für die Patientenverfügung verankert. Diese müsse in schriftlicher Form vorliegen sowie eine Unterschrift oder ein notariell beglaubigtes Handzeichen enthalten. Ein Widerruf der Patientenverfügung wäre jederzeit und ohne weiteres sowohl mündlich als auch zum Beispiel durch Gesten möglich gewesen. Der Gesetzesentwurf beinhaltete eine Reichweitenbegrenzung. Eine lebensverlängernde Maßnahme hätte demnach nur abgebrochen beziehungsweise nicht eingeleitet werden dürfen, wenn der Patient ein irreversibel zum Tode führendes Grundleiden gehabt hätte oder mit an Sicherheit grenzender Wahrscheinlichkeit das Bewusstsein nicht wiedererlangen würde. Eine unmittelbare Todesnähe wäre nicht notwendig gewesen. Ende 2008 wurde dieser Entwurf basierend auf einem Änderungsantrag unter Federführung von Katrin Göring-Eckardt (Bündnis90/Die Grünen) modifiziert. Es sollte ein zweistufiges Modell geben. Für eine Patientenverfügung mit Reichweitenbegrenzung sollten die gleichen Formvoraussetzungen wie im bisherigen Entwurf gelten, es sollte jedoch auch eine Patientenverfügung ohne Reichweitenbegrenzung möglich sein, für die schärfere Voraussetzungen gelten sollten, zum Beispiel eine regelmäßige Aktualisierungs- und Beratungspflicht. Ein zweiter Gesetzesentwurf entstand unter Federführung von Wolfgang Zöller (CSU) im Jahre 2007. Dieser Entwurf sah keine Reichweitenbegrenzung vor. Die Patientenverfügung sollte unabhängig von Art und Stadium der Erkrankung gelten. Weiterhin wären nicht nur schriftliche Verfügungen möglich gewesen, auch Tonträger oder Videos wären zu berücksichtigen gewesen, wenn sie eindeutig den Willen des Patienten gespiegelt hätten. Das Vormundschaftsgericht hätte nur bei Dissens zwischen Arzt und Betreuer eingeschaltet werden müssen, allerdings hätte es in diesem Fall das Gutachten eines Sachverständigen einholen müssen. Ein dritter Antrag wurde 2007 unter Federführung von Joachim Stünker (SPD) erarbeitet und im Jahre 2008 veröffentlicht. Auch dieser Gesetzesentwurf sah keine Reichweitenbegrenzung vor, eine Patientenverfügung sollte unabhängig von Art und Stadium der Erkrankung gelten können mit der formalen Voraussetzung der Schriftform. Das Vormundschaftsgericht sollte lediglich bei einem Dissens zwischen Arzt und Betreuer eingeschaltet werden, es müsste jedoch im Gegensatz zum Zöller-Entwurf vor seiner Entscheidung kein Gutachten einholen. In allen drei Entwürfen sollten jeweils §§ 1901 und 1904 BGB geändert werden. Am 29.05.2009 wurde unter der Leitung von Hubert Hüppe (CDU) zusätzlich ein Antrag auf Verzicht auf ein Gesetz eingebracht. Am 18.06.2009 entschied sich die Mehrheit des Bundestages für den Gesetzesentwurf der Gruppe um Joachim Stünker. Die anwesenden Abgeordneten der SPD, der FDP und der Partei Die Linke stimmten zum großen Teil für den Stünker-Entwurf und sicherten so die Mehrheit, die Vertreter der CDU/CSU stimmten mit nur einer Ausnahme gegen diesen Entwurf, fünf Abgeordnete enthielten sich bei dieser Abstimmung. Die Rolle des Betreuers gewann mit der Verabschiedung des Gesetzes an Bedeutung. Das Gesetz sieht vor, dass der Betreuer anhand der Patientenverfügung (bei Nichtvorliegen anhand des mutmaßlichen Willens des Betreuten) prüfen muss, ob „ diese Festlegungen auf die aktuelle Lebens- und Behandlungssituation zutreffen“297. Das Vormundschaftsgericht muss nur eingeschaltet werden, wenn zwischen behandelndem Arzt und Betreuer ein Dissens über die Behandlung des Betreuten beziehungsweise den Abbruch einer lebensverlängernden Maßnahme herrscht.

Studying the role of human parietal cortex in visuospatial attention with concurrent TMS-fMRI

Combining transcranial magnetic stimulation (TMS) with concurrent functional magnetic resonance imaging (fMRI) allows study of how local brain stimulation may causally affect activity in remote brain regions. Here, we applied bursts of high- or low-intensity TMS over right posterior parietal cortex, during a task requiring sustained covert visuospatial attention to either the left or right hemifield, or in a neutral control condition, while recording blood oxygenation-level–dependent signal with a posterior MR surface coil. As expected, the active attention conditions activated components of the well-described “attention network,” as compared with the neutral baseline. Also as expected, when comparing left minus right attention, or vice versa, contralateral occipital visual cortex was activated. The critical new finding was that the impact of high- minus low-intensity parietal TMS upon these visual regions depended on the currently attended side. High- minus low-intensity parietal TMS increased the difference between contralateral versus ipsilateral attention in right extrastriate visual cortex. A related albeit less pronounced pattern was found for left extrastriate visual cortex. Our results confirm that right human parietal cortex can exert attention-dependent influences on occipital visual cortex and provide a proof of concept for the use of concurrent TMS–fMRI in studying how remote influences can vary in a purely top–down manner with attentional demands. At the edge of the Harz Mountains in Lower Saxony a population of the hart's tongue fern (Asplenium scolopendrium) threatened by destruction by a gypsum quarry were transplanted into a dolina which was not populated by the species at that time, and the new population was followed over ten years. 90% of the 59 transplanted plants survived this period and grew larger during the first six years after transplantation. Progenies appeared in the third year after transplantation. Nowadays, in the tenth year after transplantation, there are 1110 progenies, 171 of which are reproducing. Overall, the population increased by 1781% in the ten years. Plants that were planted on a rocky slope or a boulder heap in the new habitat, where soil was available, grew better than plants in rock faces without soil. In contrast, in the rock faces, where substrate was not covered with autumn foliage, more juveniles established. The distance between juveniles and mother plants rarely exceeded three meters, which indicates a limited dispersal potential of the hart’s tongue fern and may explain together with low diaspore pressure as a result of local rarity of the species that the dolina had not been colonized spontaneously. We conclude that transplantations of adult plants or introduction of spores are a suitable measure for preserving hart’s tongue fern populations that are endangered by destruction. In the long run, however, such measures cannot compensate for ongoing destruction of natural habitats by mining activities in the gypsum karst region at the southern edge of the Harz Mountains.
Schön ist nicht nur das Spiel mit Buchstaben und Erinnerungssplittern, sondern auch das mit Zahlen. Im Jahr Zweitausendundzehn ist es dreißig Jahre her, dass ich der damals dreißigjährigen Sigrid Weigel zum ersten Mal begegnete. Mit ihr als Dozentin lernte ich - nach meinem Wechsel von der Bonner zur Hamburger Universität im Sommersemester 1980 - eine Form des leidenschaftlichen Studierens kennen, die ich noch mehr ersehnt hatte als das Großstadtleben. Es war wohl eine der glücklichen Konstellationen, in denen Lehrende und Lernende gemeinsam aufbrachen, um neue Fragen an ihr Fach, die Literaturwissenschaft, zu stellen, vergessene Texte (von Frauen) zu entdecken und bekannte (von Männern) anders zu lesen. Getrieben von der Wissbegierde, was es denn mit der Geschlechterdifferenz auf sich hatte, übten wir uns in genauer Lektüre, intensiver Analyse und theoretischer Reflexion.
Sucrose- and H+-dependent charge movements associated with the gating of sucrose transporter ZmSUT1

Background: In contrast to man the majority of higher plants use sucrose as mobile carbohydrate. Accordingly proton-driven sucrose transporters are crucial for cell-to-cell and long-distance distribution within the plant body. Generally very negative plant membrane potentials and the ability to accumulate sucrose quantities of more than 1 M document that plants must have evolved transporters with unique structural and functional features.
Methodology/Principal Findings: To unravel the functional properties of one specific high capacity plasma membrane sucrose transporter in detail, we expressed the sucrose/H+ co-transporter from maize ZmSUT1 in Xenopus oocytes. Application of sucrose in an acidic pH environment elicited inward proton currents. Interestingly the sucrose-dependent H+ transport was associated with a decrease in membrane capacitance (Cm). In addition to sucrose Cm was modulated by the membrane potential and external protons. In order to explore the molecular mechanism underlying these Cm changes, presteady-state currents (Ipre) of ZmSUT1 transport were analyzed. Decay of Ipre could be best fitted by double exponentials. When plotted against the voltage the charge Q, associated to Ipre, was dependent on sucrose and protons. The mathematical derivative of the charge Q versus voltage was well in line with the observed Cm changes. Based on these parameters a turnover rate of 500 molecules sucrose/s was calculated. In contrast to gating currents of voltage dependent-potassium channels the analysis of ZmSUT1-derived presteady-state currents in the absence of sucrose (I = Q/τ) was sufficient to predict ZmSUT1 transport-associated currents.
Conclusions: Taken together our results indicate that in the absence of sucrose, ‘trapped’ protons move back and forth between an outer and an inner site within the transmembrane domains of ZmSUT1. This movement of protons in the electric field of the membrane gives rise to the presteady-state currents and in turn to Cm changes. Upon application of external sucrose, protons can pass the membrane turning presteady-state into transport currents.
Der typische Sukzessionsverlauf auf basenreichen offenen Sandstandorten im Binnenland führt von Pionierstadien der Koelerio-Corynephoretea zu artenreichen Allio-Stipetum-Beständen (Festuco-Brometea). Innerhalb weniger Jahre können sich aber auch artenarme Dominanzbestände mit hohem Ruderalisierungsgrad entwickeln, die durch generalistische konkurrenzstarke Graminoide bestimmt werden. 32 Dauerbeobachtungsflächen, die ohne Pflegemanagement sind und mittlerweile seit bis zu 15 Jahren von uns untersucht werden, ermöglichen einen detaillierten Blick auf die unterschiedlichen Sukzessionslinien und die Geschwindigkeiten, mit denen sich Änderungen in der Artenzusammensetzung vollziehen. Dabei stehen uns neben den Daten der Vegetationsaufnahmen und den daraus abgeleiteten Informationen (wie z. B. mittleren Ellenberg-Zeigerwerten) für verschiedene Flächen auch Boden-Daten (N, P, pH) zur Verfügung. Eine Analyse aller Flächen dokumentiert die relative Stabilität offener Sandvegetation nährstoffarmer Standorte für die untersuchten Zeitfenster. Pioniergesellschaften des Sileno conicae-Cerastietum semidecandri s. l. und Jurineo-Koelerietum glaucae zeigen z. T. strukturelle Änderungen (so z. B. Zunahmen der Kryptogamendeckung). Eine Zunahme von Stipa capillata kann z. T. in 11–12 Untersuchungsjahren im Jurineo-Koelerietum festgestellt werden; dies korrespondiert mit einer Abnahme von Koeleria glauca und z. T. mit einer Ablösung von Tortula ruraliformis durch Hypnum cupressiforme var. lacunosum. Dennoch konnten sich während der Untersuchungsjahre in den Beständen die Arten des Jurineo-Koelerietum halten. Bestände des Allio-Stipetum zeigen in der spontanen Sukzession z. T. lange Stagnationen von mehr als 14 Jahren; vereinzelt tritt der Polykormonbildner Prunus spinosa auf. Stellenweise kommt es mit zunehmender Deckung von Stipa capillata jedoch zu gravierenden Abnahmen der Artenzahl. Bestände, die in ihrer Pionierphase durch höhere Bodennährstoffgehalte (besonders Phosphat) und höhere Ellenberg-Feuchtezahlen gekennzeichnet sind, entwickeln sich innerhalb weniger Jahre zu artenarmen Ruderalbeständen. Die gewonnenen Ergebnisse werden in einem Sukzessionsmodell dargestellt. Für ein Pflegemanagement ergibt sich aus den Ergebnissen, dass intakte Koelerion glaucae-Flächen oft nur sehr extensiv beweidet werden müssen. Insbesondere zur Dämpfung der Ruderalisierung in den nährstoffreicheren Flächen sowie zur Dynamisierung in größeren homogenen Beständen des Allio-Stipetum ist ein Pflegemanagement jedoch eine essentielle Maßnahme. Die in unseren Fällen eingesetzte Schaf- und/oder Eselbeweidung führt zur partiellen Förderung der besonders gefährdeten Pionierstadien im Mosaik mit dem Allio-Stipetum.

We propose a framework of individual problem-solving and communicative demands (IproCo) that bridges the gap between models from cognitive psychology and communication pragmatics. Furthermore, we present two experiments conducted to identify factors influencing the demands and to test possibilities for support. The experiments employed a remote collaborative picture-sorting task with concrete and abstract pictures and applied non-interactive conditions compared to interactive conditions. In a first experiment, the influence of the postulated demands on collaboration process and outcome was analysed, and the impact of shared applications was tested. In a second experiment, we evaluated instructional support measures consisting of model collaboration and a collaboration script. The collaboration process showed benefits of the support but the outcome did not. However, the support measures fostered the collaboration process even in the particularly difficult conditions with non-interactive communication. We discuss the impact of the IproCo framework and apply it to other tasks.
Inclusive transverse momentum spectra of primary charged particles in Pb–Pb collisions at √sNN=2.76 TeV have been measured by the ALICE Collaboration at the LHC. The data are presented for central and peripheral collisions, corresponding to 0–5% and 70–80% of the hadronic Pb–Pb cross section. The measured charged particle spectra in |η|<0.8 and 0.3<pT<20 GeV/c are compared to the expectation in pp collisions at the same sNN, scaled by the number of underlying nucleon–nucleon collisions. The comparison is expressed in terms of the nuclear modification factor RAA. The result indicates only weak medium effects (RAA≈0.7) in peripheral collisions. In central collisions, RAA reaches a minimum of about 0.14 at pT=6–7 GeV/c and increases significantly at larger pT. The measured suppression of high-pT particles is stronger than that observed at lower collision energies, indicating that a very dense medium is formed in central Pb–Pb collisions at the LHC.
Although a variety of genetic strategies have been developed to inhibit HIV replication, few direct comparisons of the efficacy of these inhibitors have been carried out. Moreover, most studies have not examined whether genetic inhibitors are able to induce a survival advantage that results in an expansion of genetically-modified cells following HIV infection. We evaluated the efficacy of three leading genetic strategies to inhibit HIV replication: 1) an HIV-1 tat/rev-specific small hairpin (sh) RNA; 2) an RNA antisense gene specific for the HIV-1 envelope; and 3) a viral entry inhibitor, maC46. In stably transduced cell lines selected such that >95% of cells expressed the genetic inhibitor, the RNA antisense envelope and viral entry inhibitor maC46 provided the strongest inhibition of HIV-1 replication. However, when mixed populations of transduced and untransduced cells were challenged with HIV-1, the maC46 fusion inhibitor resulted in highly efficient positive selection of transduced cells, an effect that was evident even in mixed populations containing as few as 1% maC46-expressing cells. The selective advantage of the maC46 fusion inhibitor was also observed in HIV-1-infected cultures of primary T lymphocytes as well as in HIV-1-infected humanized mice. These results demonstrate robust inhibition of HIV replication with the fusion inhibitor maC46 and the antisense Env inhibitor, and importantly, a survival advantage of cells expressing the maC46 fusion inhibitor both in vitro and in vivo. Evaluation of the ability of genetic inhibitors of HIV-1 replication to confer a survival advantage on genetically-modified cells provides unique information not provided by standard techniques that may be important in the in vivo efficacy of these genes.
Since the collapse of apartheid, there have been major increases in migration flows within, to and from the Southern African region. Cross-border movements are at an all-time high across the region and internal migration is at record levels. The implications of greater mobility for areas of origin and destination have not been systematically explored. Migration is most often seen as a negative phenomenon, a result of increased poverty and the failure of development. More recently, the positive relationship between migration and development has been emphasised by agencies such as the Global Commission on International Migration, the Global Forum on Migration and Development, the United Nations Development Programme and the African Union. The chapters in this publication are all based on primary research and examine various facets of the relationship between migration, poverty and development, including issues that are often ignored in the migration-development debate like migration and food security and migration and vulnerability to HIV. The book argues that the development and poverty reduction potential of migration is being hindered by national policies that fail to recognise and build on the positive aspects and potential of migration. As a result, as these studies show, migrants are often pushed to the margins where they are forced to 'survive on the move'. Their treatment violates labour laws and basic human rights and compromises the potential of migration as a means to create sustainable livelihoods, reduce poverty and food insecurity, mitigate the brain drain and promote the productive use of remittances. This book shows that migrant lives and livelihoods should be at the centre of international and African debates about migration, poverty and development.
In situ rainwater harvesting has a long history in arid and semi-arid regions of the world buffering water shortages for human consumption and agriculture. In the context of an Integrated Water Resource Management (IWRM) in the Cuvelai Basin in northern Namibia, roof top rainwater harvesting is being introduced to a rural community for the irrigation of household scale gardens for the cultivation of horticulture products. This study elaborates how harvested rainwater can be used for garden irrigation in a sustainable manner evaluating ecologic, economic and social implications. Considering local conditions eight cropping scenarios were designed, including different criteria as well as one and two annual planting seasons. These schemes were tested under present climate conditions and under three future climate change scenarios for 2050 with the help of a tank model designed to model monthly tank inflows and outflows. Special attention was laid on risk and uncertainty aspects of varying inter-annual and interseasonal precipitation and future climate change. A framework for the assessment of sustainability was adapted to the purposes of this study and indicators have been developed in order to assess the cropping and irrigation schemes for sustainability.
The study found that with the given tank size of 30 m³, depending on crop scenario, under optimized conditions a garden area of 60 to 90 m³ can be irrigated. The choice of crops highly impacts water use efficiency and economic profitability, compared to the considerably lower impact of amount of annual planting seasons and future climate change. In the case of worsening future climate conditions, adaptation measures need to be taken as especially the economic as well as the environmental situation are expected to exacerbate due to expected decreases in yields and revenues. Already under present conditions however, the economic dimension represents the most limiting factor to sustainability, particularly due to the excessive investment costs of the rainwater harvesting and gardening facility. Nonetheless, rainwater harvesting in combination with gardening can be regarded as successful in securing household nutrition, providing sufficient horticulture products for household consumption or market sale. At the same time with the optimal choice of crops the investment costs can be recovered within the end of the lifespan of the facility.
The C-type lectin-like receptor CLEC-2 signals via phosphorylation of a single cytoplasmic YXXL sequence known as a hem-immunoreceptor tyrosine-based activation motif (hemITAM). In this study, we show that phosphorylation of CLEC-2 by the snake toxin rhodocytin is abolished in the absence of the tyrosine kinase Syk but is not altered in the absence of the major platelet Src family kinases, Fyn, Lyn, and Src, or the tyrosine phosphatase CD148, which regulates the basal activity of Src family kinases. Further, phosphorylation of CLEC-2 by rhodocytin is not altered in the presence of the Src family kinase inhibitor PP2, even though PLCγ2 phosphorylation and platelet activation are abolished. A similar dependence of phosphorylation of CLEC-2 on Syk is also seen in response to stimulation by an IgG mAb to CLEC-2, although interestingly CLEC-2 phosphorylation is also reduced in the absence of Lyn. These results provide the first definitive evidence that Syk mediates phosphorylation of the CLEC-2 hemITAM receptor with Src family kinases playing a critical role further downstream through the regulation of Syk and other effector proteins, providing a new paradigm in signaling by YXXL-containing receptors.
Gamma synchronization has generally been associated with grouping processes in the visual system. Here, we examine in monkey V1 whether gamma oscillations play a functional role in segmenting surfaces of plaid stimuli. Local field potentials (LFPs) and spiking activity were recorded simultaneously from multiple sites in the opercular and calcarine regions while the monkeys were presented with sequences of single and superimposed components of plaid stimuli. In accord with the previous studies, responses to the single components (gratings) exhibited strong and sustained gamma-band oscillations (30–65 Hz). The superposition of the second component, however, led to profound changes in the temporal structure of the responses, characterized by a drastic reduction of gamma oscillations in the spiking activity and systematic shifts to higher frequencies in the LFP (~10% increase). Comparisons between cerebral hemispheres and across monkeys revealed robust subject-specific spectral signatures. A possible interpretation of our results may be that single gratings induce strong cooperative interactions among populations of cells that share similar response properties, whereas plaids lead to competition. Overall, our results suggest that the functional architecture of the cortex is a major determinant of the neuronal synchronization dynamics in V1. Dynamical systems driven by Gaussian noises have been considered extensively in modeling, simulation, and theory. However, complex systems in engineering and science are often subject to non-Gaussian fluctuations or uncertainties. A coupled dynamical system under a class of Lévy noises is considered. After discussing cocycle property, stationary orbits, and random attractors, a synchronization phenomenon is shown to occur, when the drift terms of the coupled system satisfy certain dissipativity and integrability conditions. The synchronization result implies that coupled dynamical systems share a dynamical feature in some asymptotic sense.
Innerhalb der letzten 30 Jahre haben sich photoaktivierbare Verbindungen („caged compounds“) zu einem äußerst wertvollen Werkzeug entwickelt. Seit der erstmals publizierten Anwendung von lichtaktivierbarem ATP im Jahre 1978 durch Hoffman et al. konnten viele neue Erkenntnisse im Bereich der Physiologie, Molekularbiologie und Biochemie mit Hilfe von photoaktivierbaren Verbindungen gewonnen werden. Hierunter versteht man Substanzen, deren biologische Wirksamkeit temporär inaktiviert ist, jedoch durch Bestrahlung mit Licht wiederhergestellt werden kann. Die Inaktivierung wird durch spezielle, lichtreaktive Chromophore erreicht, sogenannten photolabilen Schutzgruppen („cages“). Der Einsatz dieser Technik zur Aktivierung von Nukleinsäuren eröffnet ein weites Feld an Anwendungsmöglichkeiten. So ist auf diese Weise eine orts- und zeitaufgelöste Kontrolle der Aktivität von z.B. Antisense-DNA, siRNA oder Aptameren durch Licht möglich. Während der vorliegenden Doktorarbeit konnten zwei neuartige photolabil geschützte Desoxythymidine als Phosphoramidite zum Einsatz in der automatisierten DNA-Synthese dargestellt werden. Hierfür wurden jeweils NDBF-OH bzw. pHP-OH in O4-Position der Nukleobase eingeführt. Es konnte gezeigt werden, dass sich die beiden Nukleoside nach ihrem Einbau in DNA mittels UV-Licht entschützen lassen. Die photolabilen Schutzgruppen besaßen jedoch nur eine geringe chemische Stabilität unter verschiedenen basischen Abspaltbedingungen und sind deshalb nicht für die automatisierte DNA-Festphasensynthese unter Standardbedingungen geeignet. Durch Homologisierung der NDBF-Gruppe mit einer zusätzlichen Methyleneinheit zu hNDBF-OH wurde eine verbesserte Variante synthetisiert, welche eine ausreichende Basenstabilität und hervorragende photochemische Eigenschaften aufweist. Wie im Arbeitskreis Heckel gezeigt werden konnte, ist mittels in O6-Position hNDBF-modifiziertem Desoxyguanosin sowohl die DNA-Festphasensynthese unter Standardbedingungen möglich, als auch die spätere Photolyse dieser Schutzgruppe bei einer Wellenlänge von 420 nm. Hierdurch eröffnet sich zum ersten Mal die Möglichkeit, bei Verwendung von beispielsweise NPP und hNDBF, photolabil geschützte Bereiche in Nukleinsäuren wellenlängenselektiv durch Licht unterschiedlicher Wellenlänge zu aktivieren. In einem weiteren Projekt wurden erfolgreich photoaktivierbare Varianten des bivalenten Fusionsaptamers HD1-22 getestet. HD1-22 besitzt zwei Aptamerdomänen, HD1 und HD22, welche jeweils an eine der beiden Exosites I bzw. II von Thrombin binden. Im konkreten Fall konnten wir die Blutgerinnungsaktivität von Thrombin unter Verwendung einer photolabil geschützten Aptamersequenz von vollständig aktiv zu komplett inaktiv modulieren: Vor der UV-Bestrahlung war lediglich die Exosite II durch die HD22-Domäne blockiert. Dies erlaubte keinerlei Bindung des natürlichen Antikoagulans Antithrombin III an Thrombin – weder durch Heparin vermittelt noch ohne Beteiligung von Heparin. Der Zugang zur Exosite I war hingegen nicht eingeschränkt, die Blutgerinnungsaktivität des Thrombins somit durch die Rekrutierung von Fibrinogen an die Exosite I vollständig aktiv und nicht durch Antithrombin inhibierbar. Durch lichtvermittelte Entschützung der HD1-Domäne konnte nun eine Inaktivierung der Exosite I erfolgen, welche durch die hohe Bindungsaffinität der HD22-Aptamerdomäne stärker ausfiel als bei der ausschließlichen Verwendung eines HD1 Aptamers. Die koagulative Wirkung des Thrombins wurde somit vollständig aufgehoben. Im dritten Teil dieser Arbeit wurde der Grundstein für die Entwicklung eines kovalenten, lichtaktivierbaren Thrombin-Thrombinaptamer-Reagenzes gelegt. Dieses könnte zur gezielten Thromboembolisierung bei bestimmten malignen Tumorerkrankungen eingesetzt werden, um das entartete Gewebe von der Blut- und damit Nährstoffversorgung abzuschneiden. Zudem könnte so möglicherweise während einer Tumorexzision die Freisetzung von metastasierenden Krebszellen unterbunden werden. Das Reagenz besteht aus der von Heckel et al. publizierten „ausschaltbaren“ Variante des HD1-Aptamers. Diese besitzt einen photolabil geschützten Gegenstrang. Solange die Schutzgruppe intakt ist, bindet das Aptamer an die Exosite I von Thrombin und verhindert auf diese Weise die Rekrutierung von Fibrinogen. Wird die Sequenz jedoch mit UV-Licht bestrahlt, so erfolgt die Entschützung des Gegenstrangs und durch die folgende Basenpaarung eine Inaktivierung des HD1 Aptamers. Hieraus resultiert die Freisetzung von aktivem Thrombin, welches direkt zur Ausbildung eines Thrombus führt. Durch die – ebenfalls photospaltbare – kovalente Anbindung des „ausschaltbaren“ HD1 an die Proteinoberfläche werden eine vorzeitige Dissoziation des Aptamers und damit eine vorzeitige Wiederherstellung der Proteinaktivität verhindert.
Synthese und Optimierung von Nitroxyl-Spin-Labeln zur Analyse von Nukleinsäuren mittels EPR und NMR

Im Rahmen dieser Doktorarbeit wurde der Einsatz der Nitroxyl-Spin-Markierung für die Strukturaufklärung von Oligonukleotiden getestet. Dafür wurden im Vorfeld verschiedene Spin-Label synthetisiert und charakterisiert. Bei den Nitroxiden handelt es sich hierbei um das Spin-Label TPA (2,2,5,5-Tetramethyl-pyrrolin-1-oxyl-3-acetylene) und TEMPA (2,2,6,6-Tetramethyl-3,4-dehydro-piperidin-N-oxyl-4-acetylene). Beide Spin-Label wurden auch als deuteriert und 15N-markiert synthetisiert. Basierend auf den erhaltenen Ergebnissen auf dem Gebiet der Modell-Systeme und Modell-RNAs wurden biologisch relevante und strukturell anspruchsvolle Oligonukleinsäuren untersucht. Dabei konnten DNA-RNA-Hybride, die HCV-IRES Domain II, ein Tetra-Loop, das Neomycin-B Aptamer, das Diels-Alder Ribozym und ein Nep1-RNA-Komplex charakterisiert werden. Die Einführung des Spin-Labels erfolgte noch auf der Festphase mittels der Sonogashira-Kreuz-Kupplungs Reaktion.
PPARs gehören wie die Steroidhormon-Rezeptoren (z. B.Glucocorticoid-, Estrogen- oder Testosteron-Rezeptoren) zur Superfamilie der nukleären Rezeptoren. Es existieren drei PPAR-Subtypen, die als PPARalpha, PPARbeta/delta und PPARgamma bezeichnet werden und von drei verschiedenen Genen kodiert werden. Fibrate sind PPARalpha-Agonisten, welche die Plasma-Triglyceridspiegel reduzieren und gleichzeitig eine moderate Steigerung des HDL-Cholesterols bewirken. Thiazolidindione (Glitazone) sind PPARgamma-Agonisten, die bei Typ-2-Diabetes-mellitus indiziert sind und als Insulinsensitizer wirken. Duale PPARalpha/gamma-Agonisten stellen eine neue Klasse von Arzneistoffen dar, die zukünftig zur Behandlung von Typ-2-Diabetikern mit gestörtem Lipidprofil eingesetzt werden könnten. Im Rahmen der vorliegenden Dissertation wurde eine Leitstrukturoptimierung des selektiven PPARalpha-Agonisten Pirinixinsäure (WY 14643) durchgeführt. Die pharmakologische In-vitro-Charakterisierung der Substanzen erfolgte mit Hilfe subtypspezifischer Reportergen-Assays. Zusätzlich wurde gezeigt130, dass Chinolinderivate der Pirinixinsäure 5-LOX-inhibierende Eigenschaften in polymorphnukleären Leukozyten zeigen. Zunächst wurde eine geeignete Synthesestrategie zur Darstellung von Pirinixinsäurederivaten etabliert, was zur Charakterisierung und Identifizierung einer Serie von potenten dualen PPARalpha/gamma-Agonisten und 5-LOX-Inhibitoren führte. Die Synthesestrategie zur Darstellung von sowohl im Arylamino-Bereich (W) als auch in alpha- Position (R) modifizierten Derivaten der Pirinixinsäure bestand in einer vierstufigen Reaktionsfolge (I–IV; siehe Abb.). ... Die PPAR-Modulatoren (Carbonsäuren) 4, 8, 14, 32–38, 41 und 42 wurden in-vitropharmakologisch unter Anwendung subtypselektiver Reportergen-Assays (hPPARalpha, beta, gamma) charakterisiert. Die 5-LOX-Modulatoren (Carbonsäureester) 3, 7, 8, 13, 14, 24–29, 31–42 und 50 wurden in-vitro-pharmakologisch unter Anwendung des 5-LOX-Standardassays in intakten PMNL (polymorphnukleäre neutrophile Leukozyten) evaluiert. Die vorliegende Untersuchung deckte auf, dass der Ersatz des 2,3-Dimethylanilin-Strukturelements der Pirinixinsäure durch 6-Aminochinolin zu einem Gesamtverlust des PPARalpha/gamma-Agonismus führt. Durch Strukturmodifikationen im Arylamino-Bereich der Leitstruktur WY 14643 mit für 5-LOX-Non-Redoxinhibitoren typischen Pharmakophorresten, Tetrahydropyran-4-yl (Substanzen 13–14) und Methoxychinolin-2-yl (Substanz 43), konnte eine signifikante Steigerung des 5-LOX-inhibitorischen Potenzials erzielt werden [IC50 7 mikro M (13) bzw. 4 mikro M (43)]. Die alpha-alkylsubstituierten Carbonsäurederivate 33–38 zeigten sowohl am hPPARalpha als auch am hPPARgamma eine mit der aliphatischen Kettenlänge steigende Potenz. Am hPPARalpha erwiesen sich das alpha-Hexyl-Derivat 35 und das alpha-Butyl-Derivat 34 mit EC50-Werten von 1,9 mikro M (35) bzw. 11,5 mikro M (34) entsprechend einer Steigerung der Aktivität gegenüber WY 14643 um den Faktor 20 (35) bzw. den Faktor 4 (34) als besonders potent. Im Falle des hPPARgamma zeigten ebenso das alpha-butylsubstituierte Derivat 34 und der alpha-hexylsubstituierte Ligand 35 die höchste Aktivität mit EC50-Werten von 8,7 mikro M (34) bzw. 5,8 mikro M (35) und einer Steigerung der Aktivität gegenüber WY 14643 von Faktor 6 (35) bzw. Faktor 9 (34). Die Einführung von alpha-Alkylsubstituenten in das Grundgerüst der Pirinixinsäure führt möglicherweise zum Besetzen der linken proximalen Bindungstasche und somit zu einer im Vergleich zur Leitstruktur WY14643 stärkeren Bindung in die Ligandenbindungstasche des Rezeptors. Die Besetzung dieser proximalen Bindungstasche entscheidet jedoch nicht über die PPAR-Selektivität, da diese sowohl von hPPARalpha- als auch von hPPARgamma-Agonisten belegt wird. PPARalpha/gamma-Selektivität kann zum einen durch das Besetzen der linken bzw. rechten distalen Bindungstasche erzielt werden, zum anderen durch die Auswahl verschiedener acider Kopfgruppen. Aufgrund unterschiedlicher Aminosäuresequenzen der Bindungstaschen der einzelnen PPAR-Subtypen ergibt sich für die Kopfgruppen der PPAR-Modulatoren im hPPARalpha und hPPARgamma eine durch den sterischen Einfluss von Carboxyl- bzw. TZD-Kopfgruppen verursachtes unterschiedliches Wasserstoffbrückennetzwerk. Im Vergleich mit Pirinixinsäure konnte nur durch die Einführung der größeren Alkylketten (n-Butyloder n-Hexyl) ein stark erhöhter dualer PPARalpha/gamma-Agonismus erreicht werden. Die Substitution des sekundären Amins der Chinolin-6-ylverbindungen durch einen Ether-Sauerstoff in den Substanzen 41 und 42 sowie die Einführung eines Linkers, genauer einer Methylengruppe, zwischen dem Chinolin-6-ylrest und dem Scaffold (Derivate 29 und 36), führt zu einer verminderten Aktivität sowohl an hPPARalpha als auch an hPPARgamma. Eine mögliche Erklärung hierfür könnte sein, dass an entsprechender Position der Liganden-Bindungstasche eine Wasserstoffbrücken-Akzeptor-Position vorhanden ist, welche zur Erhöhung der Affinität beiträgt. Offensichtlich wird, durch die Ausbildung einer Wasserstoffbrücke zwischen der NH-Funktion und einem Wasserstoffbrücken-Akzeptor innerhalb der LBP im Falle von WY 14643 ein hoher Bindungsbeitrag erreicht. Daten bezüglich der Inhibierung der 5-Lipoxygenase demonstrieren, dass 6-Aminochinolinderivate potente 5-LOX-Inhibitoren sind. Die Einführung eines 3,5-Bis-(2,2,2-trifluor-ethoxy)-phenylrestes in 6-Position des alpha-hexylsubstituierten Scaffolds führt zu Ligand 38. Dieser ist ein potenter dualer PPARalpha/gamma-Agonist mit einem EC50-Wert von 11 mikro M an hPPARalpha bzw. 7,7 mikro M an hPPARgamma und ein stark wirksamer 5-LOX-Inhibitor mit einem IC50-Wert von 1 mikro M im 5-LOX-PMNL-Standardassay. Die Substanz 38 wurde in präklinischen Studien eingesetzt. Vier Stunden nach oraler Administration bei Mäusen (n = 6) wurde einen Plasmaspiegel von 40 mikro M erreicht. Im Hinblick auf eine Steigerung der PPAR-agonistischen Aktivität und der Reduktion der hepatotoxischen Eigenschaften der Leitstruktur Pirinixinsäure wurde eine Leitstrukturoptimierung durch Einführung der größeren Alkylketten in alpha-Position zur pharmakophoren Carboxyl-Funktion durchgeführt. Für die inhibierende Wirkung auf die 5-LOX scheint das Chinolin-Strukturelement verantwortlich zu sein, während die Potenz der Inhibition durch das Substitutionsmuster moduliert wird.
Die Gattung Palaua gehört zur Tribus der Malveae (Malvaceae, Malvoideae). Sie umfasst fünfzehn einjährige oder ausdauernde krautige Arten, die für die Nebeloasen („Lomas“, „Desierto Florido“) der Küstenwüste Perus und Chiles endemisch sind. Abweichend von den meisten anderen Gattungen der Malveae besitzt Palaua (mit Ausnahme von P. sandemanii) unregelmäßig übereinander angeordnete Merikapien. Dieses Merkmal ist ansonsten nur von den beiden altweltlichen Gattungen Kitaibela und Malope bekannt, weshalb diese früher mit Palaua in der Tribus Malopeae vereint wurden. Palynologische, cytogenetische und molekulare Analysen zeigten jedoch, dass die Malopeae eine polyphyletische Gruppe bilden und dass die in Südamerika verbreiteten Gattungen Fuertesimalva und Urocarpidium die nächsten Verwandten von Palaua sind. Ebenso wie im Aufbau des Gynözeums unterscheidet sich Palaua auch durch das Fehlen eines Epicalyx vom Großteil der Malveae, einschließlich ihrer Schwestertaxa. Seit der Erstbeschreibung der Gattung durch Cavanilles im Jahr 1785 sind nur zwei detaillierte Bearbeitungen der Gattung Palaua veröffentlicht worden. Die umfassendste davon stammt von Ulbrich (1909). Auf der Grundlage der umfangreichen Aufsammlungen von August Weberbauer beschrieb er mehrere neue Arten in seinem Werk „Malvaceae austro-americanae imprimis andinae“, das er in nachfolgenden Jahren (1916, 1932) vervollständigte. Die zweite bedeutsame Bearbeitung ist die Revision der Gattung durch Macbride (1956) in der „Flora of Peru“. Seit den 50er Jahren des vorherigen Jahrhunderts kamen jedoch zahlreiche Aufsammlungen hinzu, insbesondere durch den peruanischen Botaniker Ramón A. Ferreyra (1912-2005), sowie durch Ernesto Günther (1870-?) zusammen mit Otto Buchtien (1859-1946), Gerd K. Müller (1929-) und Michael O. Dillon (1947-), so dass eine Neubearbeitung von Palaua erforderlich wurde. Darin bestand das Hauptziel der hier vorgestellten Dissertation. Für die Revision der Gattung wurden 618 Herbarbelege der wichtigsten Herbarien morphologisch untersucht. In den Jahren 2002 und 2003 wurden während mehrmonatiger Geländearbeiten in den Lomas-Standorten Perus und Chiles eigene botanische Aufsammlungen durchgeführt sowie Daten zur Verbreitung der Arten und ihrer Ökologie erfasst. Des Weiteren wurde aus dem mitgebrachten Samenmaterial eine mehrere Arten einschließende Lebendsammlung angelegt, mit deren Hilfe detaillierte Untersuchungen zur Blütenmorphologie und Karyologie realisiert werden konnten. Besonders schwierig gestaltete sich die Bearbeitung nomenklatorischer Fragestellungen, da viele der in Berlin (B) aufbewahrten Typusbelege von Weberbauer im Zweiten Weltkrieg zerstört wurden und somit eine Identifizierung vieler Arten problematisch war. Auch die Ermittlung des Typusbelegs der Gattung, den Cavanilles für seine Beschreibung vorliegen hatte, war mühsam. Neben dem Studium der Originalbelege und Protologe mussten auch die historischen Begebenheiten rekonstruiert und Reiseberichte zu den Aufsammlungen durchgesehen werden, um unter anderem den Holotypus der Gattung identifizieren zu können. Die eigenen taxonomischen Studien führten zur Festlegung von insgesamt 8 Lectotypen, 3 Epitypen and 2 Ikonotypen. Im Rahmen der morphologischen Untersuchungen wurden sämtliche taxonomisch relevanten Merkmale detailliert erfasst, einschließlich der verschiedenen Behaarungstypen. Neben den für die Malvaceen bekannten Sternhaaren, sind hier auch Drüsenhaare für Palaua beschrieben und charakterisiert worden. Die anatomischen Studien konzentrierten sich auf Blatt- und Samenmerkmale. Zusätzlich zu den morphologisch-anatomischen Studien wurden molekularsystematische Analysen durchgeführt. Zwei Methoden kamen dabei zur Anwendung: DNA-Sequenzierung und Amplified Fragment Length Polymorphism (AFLP). Letztere wurde eingesetzt, um insbesondere die Verwandtschaftsverhältnisse junger Taxa, die sich mit DNA-Sequenzdaten kaum auflösen lassen, zu rekonstruieren. In umfangreichen Versuchen stellte sich jedoch heraus, dass diese Methode keine reproduzierbaren Ergebnisse hervorbrachte, vermutlich bedingt durch den sehr hohen Polysaccharidgehalt der DNA-Template, wie es von Malvaceen her bekannt ist. Selbst die Erprobung zahlreicher Reinigungsschritte und –methoden ergab kein zufriedenstellendes Resultat. Für die phylogenetische Rekonstruktion wurden daher ausschließlich DNA-Daten verwendet, und zwar Kern-DNA (Internal Transcribed Spacer, ITS) und Plastiden-DNA (psbAtrnH Intergenic Spacer). Andere getestete Marker, wie z.B. die trnL-F-Region, wiesen zu wenig phylogenetisch informative Merkmale auf. Die morphologischen Analysen ergaben, dass Merkmale wie die Behaarung der Kelch- und Laubblätter, die Blattform und die Größe der Blüten besonders hilfreich für die Abgrenzung der Arten sind. Im Gegensatz zu anderen nah verwandten Gattungen ist die Form der Merikarpien in Palaua relativ uniform und daher als diakritisches Merkmal ungeeignet. Die Größe der Blüte nimmt in der Regel mit der Anzahl der Staubgefäße und Merikarpien zu. Die Palaua-Arten zeigen einige Anpassungen an ihren extrem trockenen Lebensraum. Die meisten Arten sind Annuelle und vollziehen eine rasche Entwicklung während der kurzen Zeit, in der ausreichend Feuchtigkeit verfügbar ist. Bei solchen Pflanzen findet man als Anpassung häufig eine Tendenz zur vermehrten Samenproduktion. In diesem Zusammenhang ließe sich auch die innerhalb des Verwandtschaftskreises ungewöhnliche Stellung der Merikarpien bei Palaua interpretieren, mit der es den Arten gelingt, mehr Samen als bei Arten mit einreihiger Merikarpienanordnung zu produzieren. Als weitere Anpassung findet man bei den ausdauernden Arten größtenteils eine sehr dichte Behaarung, wobei die Sternhaare mehrjähriger Arten wesentlich mehr Strahlen besitzen als die bei den einjährigen Arten. In der hier vorgestellten Revision der Gattung werden 15 Arten anerkannt: P. camanensis, P. dissecta, P. guentheri, P. inconspicua, P. malvifolia, P. modesta, P. mollendoensis, P. moschata, P. rhombifolia, P. sandemanii, P. tomentosa, P. trisepala, P. velutina, P. weberbaueri sowie die neu zu beschreibende P. spec. nov. Die morphologisch abweichende P. sandemanii wird aufgrund der molekularen Analysen ebenfalls zu Palaua gestellt. Die auch in der jüngeren Literatur meist als getrennte Arten aufgefassten P. concinna und P. moschata lassen sich nach Durchsicht des umfangreichen Materials nicht mehr als eigenständige Arten aufrechterhalten. Die vormals als chilenischer Endemit behandelte P. concinna wird hier in die Synonymie von P. moschata gestellt. Auch die peruanische P. micrantha var. hirsuta wurde in die Synonymie der zuvor rein chilenischen P. modesta verwiesen, was bedeutet, dass sich das Vorkommen von P. modesta nun auch auf Peru ausdehnt. Auf infraspezifischem Niveau wurden einige Varietäten und eine Form neu beschrieben, um die im Sammlungsmaterial vorhandene morphologische Variabilität besser zu gliedern. Das ist der Fall bei P. dissecta (2 Varietäten), P. tomentosa (1 Varietät), P. weberbaueri (1 Varietät) und P. mollendoensis (1 Form). Die neuen Taxa werden an anderer Stelle gültig publiziert. Die von Baker (1890) and Ulbrich (1909) gewählte infragenerische Klassifikation mit der Einteilung in die Sektionen Annuae (einjährige Arten) und Perennes (mehrjährige Arten) erweist sich als nicht haltbar. Weder die morphologischen noch die molekularen Daten bieten hierfür Unterstützung. Auch die von Hochreutiner (1956) vorgeschlagene Ausgliederung von P. trisepala als eigene Untergattung Rauhia, aufgrund des Vorkommens von lediglich drei statt fünf Kelchblättern, erscheint nicht sinnvoll. Abgesehen von ihrer reduzierten Kelchblattzahl (3 statt 5 Kelchblätter) ist diese Art morphologisch P. moschata und P. velutina sehr ähnlich. Die Aufstellung einer eigenen Untergattung würde die tatsächlichen Verwandtschaftsverhältnisse verwischen und vermutlich eine paraphyletische Einheit schaffen. Im Vergleich zur Anzahl der Kelchblätter sind Merkmale wie der Aufbau der Infloreszenzen, die Blütengröße und -farbe, sowie die Blattmorphologie (geteilte vs. ungeteilte Blätter) nützlicher für eine infragenerische Unterteilung. Die Form der Stipeln, die von Ulbrich (1909) für eine weitere Unterteilung seiner Sektionen verwendet wurde, ist weniger für eine infragenerische Gliederung als für die Abgrenzung mancher Arten geeignet. Formell wurde in der hiesigen Arbeit auf eine infragenerische Unterteilung verzichtet, da zunächst abgewartet werden soll, ob weiterführende molekularsystematische Untersuchungen nicht doch zu einer besseren Auflösung und auch Unterstützung der basalen Knoten der Palaua-Phylogenie führen. Andernfalls steht zu befürchten, dass wiederum künstliche Sippen geschaffen werden. Nichtsdestotrotz, sprechen die eigenen morphologischen und zum Teil auch die molekularen Daten für eine Gliederung der Gattung in drei taxonomische Einheiten (siehe unten). Die Ergebnisse der molekularen Analysen (kombinierte Analyse von ITS- und psbA-trnHSequenzen) ergaben drei mehr oder weniger gut gestützte Kladen innerhalb einer sehr gut gestützten monophyletischen Palaua. Interessanterweise bildeten die Arten P. inconspicua und P. modesta eine Klade (88% Jackknife-Unterstützung, JK), die die Schwestergruppe zu den restlichen Arten der Gattung darstellt. Beide Arten haben eine von der restlichen Gattung abweichende Blütenmorphologie (kleine Petalen, weniger Merikarpien) und die razemösen Infloreszenzen enthalten neben Einzelblüten in den Achseln der Trägblätter auch 2-4-blütige Teilinfloreszenzen, an denen die Blüten kein Tragblatt aufweisen. Die zweite Klade (JK 97%) beinhaltet die Arten des P. dissecta-Komplexes, dessen Arten sich durch tief geteilte Blätter und große, auffällig rosarot bis violett gefärbte Blüten mit zahlreichen Merikarpien auszeichnen. In der dritten Klade (JK 73%) bildet P. guentheri die Schwestergruppe zu den restlichen Arten. Die hier vereinten Arten sind durch den Besitz ungeteilter Blätter und meist großer, auffällig rosarot bis violett gefärbter Blüten mit zahlreichen Merikarpien gekennzeichnet. Eine Ausnahme bildet P. guentheri, die geteilte Blätter hat und von daher Übereinstimmungen mit den Arten um P. dissecta aufweist. Sie weicht jedoch von den Arten des P. dissecta-Komplexes aufgrund ihrer geringeren Blütengröße und der geringeren Merikarpienanzahl ab. Außerdem sind die Blätter meist stärker reduziert und weniger regelmäßig geteilt als jene. Allerdings bedarf die Stellung von P. guentheri innerhalb der Gattung noch einer eingehenderen Überprüfung mit zusätzlichen (molekularen) Daten, da die Unterstützung für diese Klade vergleichsweise moderat ausfällt. Interessanterweise schließt diese Klade auch die aberrante P. sandemanii ein. Eine phylogenetische Rekonstruktion der Karpellanordnung ergab, dass die einreihige Anordnung der Karpelle in P. sandemanii vermutlich sekundär in Palaua entstanden ist. Allerdings zeigten Hypothesentests (Templeton-Test, Shimodaira-Hasegawa-Test), dass die Datengrundlage nicht ausreichend robust ist, um auch die Alternativhypothese einer sekundären Entstehung der unregelmäßig übereinander angeordneten Karpelle, wie sie die restlichen Arten der Gattung kennzeichnen, zu verwerfen. Innerhalb der Integrifolia-Klade lassen sich außerdem zwei Gruppen von Arten morphologisch deutlich unterscheiden. Die erste Gruppe besteht aus den einjährigen P. malvifolia und P. rhombifolia, die sich durch ihr fast kahles Indumentum auszeichnen und in Nord- bis Zentral-Peru vorkommen. Die zweite Gruppe, gebildet von den ausdauernden P. moschata, P. trisepala und P. velutina, ist durch ein samtiges Indumentum gekennzeichnet. Während sich P. moschata über das gesamte Verbreitungsgebiet der Gattung erstreckt, kommen die anderen Arten nur in Südperu vor. Die Chromosomenzahl von Palaua ist ein wichtiges Merkmal und diente Bates (1968) für deren Zuordnung zur Sphaeralcea-Allianz. Bis dato sind Chromosomenzählungen nur für zwei Arten bekannt gewesen: P. rhombifolia und P. moschata (beide mit 2n = 10 Chromosomen). In dieser Arbeit wurden weitere Zählungen durchgeführt und es wurde bestätigt, dass es neben diploiden auch tetraploide Arten mit 2n = 20 Chromosomen gibt. Polyploidie scheint dabei auf die ausdauernden Arten beschränkt zu sein. In manchen Arten, insbesondere in denjenigen des P. dissecta-Komplexes und in P. tomentosa, findet man eine ausgeprägte phänotypische Variabilität, die die Abgrenzung derselben stark erschwert. Ohne die Ursachen abschließend klären zu können, erscheint diese Variabilität zumindest teilweise als Ergebnis von Hybridisierung, Introgression und Polyploidisierung zu sein. In Bezug auf die Biogeographie der Gattung, zeigt sich, dass 11 Palaua-Arten endemisch für Peru sind und 4 Arten auch in Chile vorkommen. Das Verbreitungszentrum von Palaua ist das Gebiet der Lomas im Süden Perus (Departments Arequipa, Moquegua, Tacna), in dem 12 der 15 Arten auftreten. Die Blütezeit der Palaua-Arten variiert von Jahr zu Jahr, abhängig davon, wie viel Nebelfeuchtigkeit in der südhemisphärischen Winter-/Frühlingszeit für die Pflanzen zur Verfügung steht. Die Entstehung der Nebel variiert außerdem von Norden nach Süden, so dass sich die Blühphasen entlang dieses Gradienten verschieben. So liegt die Blütezeit in Nordperu zwischen Juli und August, in Zentral-Peru zwischen August und September und in Südperu und Chile zwischen Oktober und November. Abweichungen von diesem Schema entstehen vor allem in El Niño-Jahren, in denen auch während des südhemisphärischen Sommers die Lomaspflanzen blühen. Die Lomasvegetation ist eine bedrohte Pflanzenformation, deren Artenvielfalt bisher aber nur in Form eines recht kleinen Naturreservats geschützt wird. Da sich viele Lomasstandorte in der Nähe von Siedlungen befinden, sind etliche der lokal nur begrenzt vorkommenden Arten in ihrem Bestand bedroht. Dies betrifft insbesondere P. rhombifolia und P. malvifolia, deren Verbreitungszentrum im Gebiet der Hauptstadt Lima liegt. Eigene Beobachtungen am Standort haben zudem bestätigt, dass einige Populationen dieser Arten durch von Käfern verursachter Herbivorie nahezu vollständig zerstört werden. Weitere Schutzmaßnahmen zum Erhalt der Palaua-Arten (wie auch der anderen Lomas-Arten) wären daher dringend geboten.
This study examines the legal environment of netting agreements covering financial contracts. It concludes that an international instrument should be developed capable of improving the effectiveness of netting agreements in mitigating systemic risk. To this end, two different aspects of the enforceability of netting agreements are considered: (i) the general enforceability of netting, and (ii) the possibility of precluding the operation of netting a mechanism by way of a regulatory moratorium for considerations of systemic stability. The first part of the study presents the use of netting and the various forms it may take before going on to explain the benefits and drawbacks of enforceable netting agreements. Benefits for individual firms consist in lower counterparty risk and more favourable capital requirements. Benefits for the financial market as a whole flow from greater financial market stability since the contagion of systemically relevant institutions by the default or insolvency of another institution is limited, thus helping to avoid systemic effects. Additionally, the use of netting arrangements can improve overall market liquidity. A potential drawback of enforceability of netting, in certain situations, is that the operation of a netting mechanism could actually work against the purpose of systemic stability where the transfer of parts of the business of an insolvent financial institution to a solvent bridge entity would enhance or maintain value to a greater extent than the operation of a netting agreement would. Regulatory authorities are considering under which conditions a moratorium to halt the netting mechanism until the situation is solved could avoid this threat to systemic stability. The second part of the study examines whether there is the potential to support the purpose of enhanced systemic stability by way of international harmonisation of private and insolvency law. As regards the issue of general enforceability, the global picture of netting legislation is heterogeneous. Given the great practical relevance of the matter, an international instrument could be very useful. As to the issue of private law consequences of regulatory moratoria, the absence of a harmonised framework appears to lead to actual cross-border inconsistency and legal uncertainty as regards financial contracts that are governed by a foreign law. Taking these to aspects into account, this paper recommends that work on developing an international instrument be undertaken. The final part of the study suggests a set of preliminary guidelines for the development of suchan instrument. In the light of the findings of the previous sections, a mixed, two-step approach is recommended. First, a non-binding instrument could be developed, serving as a benchmark and reservoir of legal solutions in respect of the relevant issues. Secondly, isolated aspects relating to both the general enforceability of netting and the accommodation of a regulatory moratorium in foreign private and insolvency law could be dealt with in an international Convention, in particular where cross-border situations involving netting require uniformity of applicable legal rules.
Das C-reaktive Protein (CRP) ist ein systemischer Marker für unspezifische Infektionen und wird bei einer Entzündungsreaktion in der Leber produziert. Seine Serum-Konzentration steigt im Rahmen einer Immunreaktion innerhalb von 24 - 48 Stunden auf ein vielfaches an und induziert zahlreiche Prozesse des Immunsystems. Erhöhte CRP-Werte konnten als Risikofaktor für kardiovaskuläre Erkrankungen identifiziert werden. Zahlreiche Querschnittsstudien konnten erhöhte Serum-CRP-Werte bei Parodontitispatienten im Vergleich zu gesunden Kontrollpersonen finden. Der Anstieg des Serum-CRP könnte ursächlich an den Zusammenhängen zwischen Parodontitis und kardiovaskulären Erkrankungen beteiligt sein. Die Neutrophile Elastase ist ein weiterer systemischer Parameter, der im Rahmen chronischer Entzündungsreaktionen, wie der chronischen obstruktiven Lungenerkrankung (COPD), erhöhte Werte annimmt. Zahlreiche Studien konnten erhöhte Leukozytenzahlen bei Parodontitispatienten finden. Bisher wurde von den meisten Studien die Erhöhung des CRP-Wertes, sowie weiterer anderer Entzündungsparameter im Zusammenhang mit Parodontitis untersucht. Nach unserem Kenntnisstand konnten lediglich erhöhte Elastase-Werte im Zusammenhang mit Gingivitis gezeigt werden. Ziel der Untersuchung: Vergleich der Entzündungsparameter (CRP, Elastase, Leukozytenzahl, LBP, IL-8, IL-6) bei parodontal gesunden Probanden (Pro) und Patienten mit aggressiver (AgP) und chronischer Parodontitis (ChP). Methode: Es wurden 30 Pro (Sondierungstiefe (ST) < 3,6 mm oder < 5 mm ohne Bluten auf Sondieren (BOP), BOP < 10 %), 31 Patienten mit ChP (ST ≥ 3,6 mm und Attachmentverlust (AL) ≥ 5 mm ≥ 30 % der Stellen, Alter ≤ 35 Jahre) und 29 Patienten mit AgP (klinisch gesund (d.h. keine Allgemeinerkrankungen, die für Parodontitis prädisponieren), ST ≥ 3,6 mm > 30 % der Stellen, röntgenologischer Knochenabbau von ≥ 50 % der Wurzellänge an ≥ 2 Zähnen, Alter ≥ 18 Jahre und ≤ 35 Jahre) wurden klinisch auf folgende parodontologische Parameter untersucht: Entzündungsbefund, Plaquebefund, ST, AL, BOP. Bei allen Patienten wurde der BMI erhoben und Blutproben zur Bestimmung von CRP (untere Nachweisgrenze: 0,01 mg/dl), Elastase, Leukozytenzahl, LBP, IL-8 und IL-6 entnommen. Ergebnisse: Es wurden 30 Pro (16 Frauen, 8 Raucher), 31 Patienten mit ChP (12 Frauen, 10 Raucher) und 29 Patienten mit AgP (16 Frauen, 9 Raucher) eingeschlossen. Die Ergebnisse der CRP-Konzentration (Pro: 0,10 ± 0,12; ChP: 0,17 ± 0,23; AgP: 0,55 ± 0,98 mg/dl [p < 0,001]), der Elastase-Werte (Pro: 10,0 ± 4,7; ChP: 17,1 ± 12,3; AgP: 32,0 ± 14,6 mg/dl [p < 0,001]) unterschieden sich in allen drei Gruppen signifikant, wohingegen keine signifikanten Unterschiede in der Leukozytenzahl und der Konzentrationen von IL-8 festzustellen war. Die Werte für LBP (p < 0,01) waren signifikant höher in der AgP-Gruppe als in der Pro-Gruppe und der ChP-Gruppe (Pro = ChP < AgP). Die IL-6 Konzentration war in der Kontrollgruppe signifikant niedriger (p < 0,001) als in den Testgruppen (Pro < ChP = AgP). Die unterschiedlichen CRP-Werte in den Testgruppen sind teilweise durch den BMI zu erklären (p = 0,003). Schlussfolgerung: Die Konzentrationen für Serum-CRP und Elastase sind signifikant erhöht bei Patienten mit aggressiver Parodontitis im Vergleich zu parodontal gesunden Probanden als auch zu Patienten mit chronischer Parodontitis. Erhöhte Serum-CRP-Werte könnten bei Patienten mit aggressiver Parodontitis zu einem erhöhten kardiovaskulären Risiko beitragen.

Mais la philosophie a toujours le droit de se porter vers l’esthétique sans perdre pour autant de son essence : le philosophe se tournant de tous les côtés pour trouver la signification et le fondement de ce qui est donné n’a pas besoin de détourner son regard des faits esthétiques. S’il veut appréhender en toute rigueur conceptuelle ce qui est et se manifeste en tant que beau, laid, sublime, etc., il doit pouvoir rapporter des constats empiriques (comme par exemple le caractère satisfaisant de certains couples de couleurs) à un concept plus général (par exemple celui d’harmonie), et rapporter ce dernier à son tour à une attitude intellectuelle plus large (disons, celle de la pure contemplation gratuite). C’est par cette réduction progressive aux principes les plus généraux que peuvent apparaître les relations [Zusammenhang] qu’entretiennent entre elles les connaissances singulières, ces dernières pouvant souvent prendre alors un nouveau sens. Gene therapy is a promising therapeutic strategy that emerged from the attractive idea of targeting therapy at the molecular level. For many patients who suffer from genetic and acquired diseases that cannot be effectively treated by conventional treatment approaches gene therapy remains a huge hope of cure in spite of the hurdles regarding efficacy and safety that need to be overcome. The development of efficient gene transfer vehicles, mainly retroviral vectors, led to the first successful gene therapy trial, to treat patients suffering from X-linked severe combined immunodeficiency syndrome (X-SCID) using gene modified stem cells (Hacein-Bey-Abina, Le Deist et al. 2002). Despite the success of this trial, it revealed the danger of retroviral insertional mutagenesis as a major adverse event of gene therapy using gene-modified stem cells (Hacein-Bey-Abina, von Kalle et al. 2003). In contrast to stem cells, T cells are relatively resistant to insertional mutagenesis and transformation even after transduction with potent oncogenes using retroviral vectors (Newrzela, Cornils et al. 2008). However, mature T cells can self-renew, proliferate and survive for long periods. These criteria are supposed to render T cells prone to transformation. Therefore, the questions of mature T cells transformability and the control mechanism limiting their transformation are still elusive.
Im fünften Jahr in Folge luden das Institut für Deutsche und Niederländische Philologie der Freien Universität Berlin, das Max-Planck-Institut für Wissenschaftsgeschichte und das Exzellenzcluster 16 der Universität Konstanz am 10. Juli 2010 ein zu einem für alle Interessierten offenen Forum für NachwuchswissenschaftlerInnen, die sich im Feld von Literature and Science betätigen und vorläufige Ergebnisse noch nicht abgeschlossener Arbeiten zur Diskussion stellen möchten.
Um 1795, unter der Regentschaft des aufgeklärten Fürsten Friedrich und wenige Jahre nach dem Ausbruch der Französischen Revolution, wird in der Hofbibliothek der Fürsten zu Waldeck und Pyrmont „jedermann zu lesen gnädigst erlaubet“ (StA MR Best. 118d, Generalia I,1). Damit wird diese Fürstenbibliothek, deren Ursprünge bis ins Mittelalter zurückreichen und die in der Frühen Neuzeit zu einer prachtvollen Sammlung von Büchern und Kunstobjekten zusammenwächst, für eine breite Öffentlichkeit zugänglich. Dies legen alte Ausleihlisten und Kataloge unmissverständlich dar. Wer waren aber die Nutzer dieser und anderer Bibliotheken im Aufklärungsjahrhundert? Welche Bücher und Archivalien wurden gesammelt, gelesen oder erforscht? In welchem Maße und auf welche Art fanden das „Schöne“ und das „Nützliche“ in Bibliotheken dieser Zeit zueinander? Ausgehend von diesen Themenkomplexen fokussierte der Workshop das Spannungsverhältnis von Repräsentation, Wissen und Öffentlichkeit in (Hof)bibliotheken der Zeit zwischen Barock und Aufklärung und ging der Frage nach, wie diese drei Aspekte in der Entwicklung von der Raritätenkammer zur „modernen“ Bibliothek sich zueinander verhalten und gegenseitig beeinflussen.
Typologisches Denken gerät im 17. Jahrhundert in Unordnung, wenn die Figuren des Barock, wie der Simplicissimus, nicht (länger) das erfüllen, was sie versprechen und zugleich andere Aspekte des Figurenbegriffs an Bedeutung gewinnen. An diesem Punkt setzte die Tagung „Barocke Figuren. Zeit – Gestalt – Medien“ (27.-29. Oktober 2010, Universität Konstanz) an. Sie untersuchte eben jene (atypischen) Figuren, die sich gegen eine strenge Typologisierung sperren, deren Umwege, Metamorphosen, Begegnungen und Konflikte es aber erlauben, die Frage, was im Barock eine Figur ausmacht, in begrifflich-analytischer Perspektive neu zu stellen.
Tagungsbericht: Veranstalter: Urs Meyer/ Gabi Pahnke, Johann-Gottfried-Seume-Gesellschaft zu Leipzig e.V.; Ludwig Stockinger, Universität Leipzig Datum, Ort: 03.06.2010-05.06.2010, Leipzig Bericht von: Urs Meyer, Universität Fribourg: Obgleich Johann Gottfried Seume (1763-1810) bis heute vor allem als der "Spaziergänger nach Syrakus", als Reisender fern seiner Heimat, in Erinnerung geblieben ist, war der sozialkritische Schriftsteller, Dichter und Publizist der Spätaufklärung auch überzeugter sächsischer Landsmann – und bisweilen sogar heimwehkrank nach seiner "Vaterstadt" Leipzig. Immer wieder trieb es ihn fort aus seiner Heimat, doch kehrte er auch immer wieder zurück. Damit drängt es sich auf, Seumes Verhältnis zu Leipzig, zu der Stadt, in der er über 25 Jahre seines Lebens verbrachte, zum Gegenstand einer Tagung zu machen, die im Rahmen von Feierlichkeiten zum 200. Todestag des Dichters in Leipzig stattfand.
Szenen und Netzwerke, die für die Berliner Nachkriegsliteratur im Zeitraum der Jahre 1955 bis 1965 konstitutiv waren, bildeten den Gegenstand der Tagung, die die Johannes-Bobrowski-Gesellschaft in Zusammenarbeit mit dem Literaturforum im Brecht-Haus veranstaltete. Die von Andreas Degen konzipierte Veranstaltung fand vom 4. bis 5. März 2010 im Berliner Literaturforum im Brecht-Haus statt und wurde von der Arbeitsgemeinschaft Literarischer Gesellschaften und Gedenkstätten aus Bundesmitteln gefördert. Die Tagung begleiteten und ergänzten Lesungen aus Johannes Bobrowskis Briefwechsel mit Christoph Meckel und Lilo Fromm sowie Gespräche über "zu Unrecht vergessene" Texte Berliner Literatur aus den sechziger Jahren.
In dieser Betrachtung wird versucht, die Besonderheiten und Schwächen von Ehrensteins 'Pe-Lo-Thien' zu erläutern und aus einer chinesischen Perspektive zu bewerten, ob seine Nachdichtungen den ursprünglichen Stil und Wert von Pe Lo Thiens Gedichten beibehalten, ferner: durch die Analyse der Hauptthemen und des Sprachgebrauchs herauszufinden, ob seine Nachdichtungen expressionistische Elemente enthalten. Außerdem wird Pe Lo Thiens Gedicht 'In der Nacht einen Singenden hören. Übernachten in E Zhou' mit Ehrensteins Nachdichtung 'Ein Mensch weint in der Nacht' und mit Klabunds Nachdichtung 'Das nächtliche Lied und die fremde Frau' verglichen, um die Mehrdeutigkeit von Pe Lo Thiens Gedicht aufzuzeigen und einige Unterschiede zwischen der westlichen und der chinesischen Dichtung zu verdeutlichen.
This book is the first comprehensive contribution to understanding the character of important societal transitions in Tanzania during Benjamin Mkapa's presidency (1995- 2005). The analyses of the trajectory of these transitions are conducted against the background of the development model of Tanzanian's first president, Julius Nyerere (1961-1985), a model with lasting influence on the country. This approach enables an understanding of continuities and discontinuities in Tanzania over time in areas such as development strategy an ideology, agrarian-land, gender and forestry issues, economic liberalization, development assistance, corruption and political change. The period of Mkapa's presidency is particularly important because it represents the first phase of Tanzania's multi- party political system. Mkapa's government initially faced a gloomy economic situation. Although Mkapa's crusade against corruption lost direction, his presidency was characterised by relatively high growth rates and a stable macro-economy. Rural and agrarian transitions were dominated by diversification rather than productivity growth and transformation. Rural attitudes in favour of land markets emerged only slowly but formal land disputes showed more respect for women's rights. Some space emerged for widening local participation in forest management, but rural dynamics was mainly found in trading settlements feeding on economic liberalization and artisanal mining. The transitions documented and analysed of Mkapa's presidency, however, indicate only limited transformational change. Rural poverty is therefore likely to remain deep and the sustainability of economic development to be at risk in the future. Mkapa was, however, able to protect the legacy of peace and political stability of Nyerere, but there were nevertheless important challenges to the first multiparty elections and governance, and particularly in Zanzibar. The post- script (covering 2005 2010), indicates that the incumbent president, Jakaya Kikwete, has yet to prove that he can change this legacy of Mkapa. Co-published with the Nordic Africa Institute and the Sokoine University of Agriculture, the contributions to the eleven chapters of this book are evenly shared between Tanzanian, Nordic and other European researchers with a long-term commitment to Tanzanian development research. he book is dedicated to the youth of Tanzania.
Tanzen ist eine Form des Umgangs mit Musik, die in allen Kulturen weltweit existiert. Eine der vielen Funktionen, die dem Tanzen im Laufe der Geschichte zugeschrieben wurden, ist sein Nutzen als heilkräftiges Medium. Bisher gibt es jedoch nur wenige Untersuchungen, die sich mit den potenziellen Effekten des Tanzens als gesundheitsfördernde Tätigkeit beschäftigt haben. Ziel der vorliegenden Arbeit war es einerseits, eine Orientierung in diesem zersplitterten und gleichwohl viel versprechendem Forschungsfeld zu bieten und andererseits die subjektiven Wirkungen des Amateur Tanzens, seine psychobiologischen Korrelaten, sowie die dabei zu Grunde liegenden Einflüsse von Musikstimulation und Partner zu untersuchen. Es wurde gezeigt, dass Tanzen eine multidimensionale Tätigkeit darstellt, die verschiedene gesundheitsfördernden Komponenten, nämlich Körperbewegung, soziale Interaktion und Musikstimulation, einschließt. Wahrgenommen werden positive Wirkungen des Tanzens auf das emotionale Befinden, das körperliche Wohlergehen, das Selbstwertgefühl, das Sozialempfinden sowie das spirituelle Wohlbefinden. Tanzen wird darüber hinaus als wichtige Strategie zur Stressbewältigung anerkannt. Die Untersuchung der Wirkung des Paartanzens auf den emotionalen Zustand sowie auf Cortisol- und Testosteronkonzentrationen ergab, dass Tanzen zu einem Anstieg des positiven und einem Absinken des negativen Affektes führt. In Bezug auf die psychobiologischen Parameter wurden Cortisolreduktionen nach dem Tanzen nachgewiesen, welche von der Musikstimulation abhängig waren, während Testosteronerhöhungen zu Beginn der Untersuchung mit der Präsenz des Partners assoziiert waren. Die hier skizzierten Studien geben Hinweise auf die gesundheitsfördernde Bedeutung des Amateurtanzens sowie auf seine subjektiven und neuroendokrinen Korrelate. Darüber hinaus trägt diese Arbeit dazu bei, die differentiellen Einflüsse von Musik und Partner beim Tanzen zu verstehen. Diese Befunde sollen zukünftige Forschungsvorhaben fördern, die den Nutzen des Tanzens als gesundheitsfördernde Tätigkeit weiter verdeutlichen und das Verständnis der dahinter liegenden psychobiologischen Mechanismen erweitern.
'Übergang' ist eine Metapher für die Beschreibung ideengeschichtlicher Prozesse. Sicher ist ihr Bedeutungsfeld weiter. Kurt Röttgers (2007) nennt neben der anthropologischen Konstante: dem Übergang als Erfahrung von Tod und Geburt, den Übergang als Metamorphose, den Übergang als Transzendieren sowie dessen Negation: nämlich das Transzendieren als ein unstatthaftes, gar mit Tabu belegtes Übergehen von einer Sphäre in die andere. Hinüberzugehen kann, so Röttgers weiter, auch bedeuten, über eine Schwelle zu gehen, fortzuschreiten zu Neuem und in diesem Akt des Fortschreitens das Werden und die Geschichte, ja die Geschichtlichkeit selbst, auch in ihrer radikalen Verneinung alles Vorangegangenen, zu bejahen. Gerade im Zusammenhang mit letzteren Bedeutungszuschreibungen hat die Metapher des Übergangs im geschichtsphilosophischen beziehungsweise bewusstseinsphilosophischen Konzept des Deutschen Idealismus ihre Nobilitierung erfahren. Bewusstseinsphilosophisch ist dieses Konzept insofern, als es von den seit Descartes zentralen Prämissen der Orientierung an sich selbst sowie der Bildung von Allgemeinbegriffen ausschließlich auf der Grundlage von Erfahrung und damit auf der Grundlage von etwas zunächst in der Vorstellung Gegebenem ausgeht (vgl. Zeuch 2001). Die Frage, der ich im Folgenden nachgehe, ist, ob diesem Konzept, das seinen Kairos um 1800 hat, ein besonderes Potential zukommt, das der Ideengeschichte gegen ihre Kritiker auch in der Gegenwart eine 'raison d'etre' verleiht, wenn es um den Rekonstruktionsversuch komplexer gesellschaftlicher, politischer, philosophischer Zusammenhänge und Kontinuitäten über mehrere Epochen oder Zeitabschnitte hinweg geht.
Taxonomische Neukombinationen zur Flora Mittel- und Osteuropas, insbesondere Nordrhein-Westfalens

Im Rahmen der Erarbeitung mehrerer umfassender floristischer Inventare von Regionen Mittel- und Osteuropas wurden umfangreiche phytotaxonomische Studien durchgeführt. Als ein Ergebnis dieser Untersuchungen werden Neu- und Umkombinationen bei Sippen verschiedener Familien in den Art- und Unterartstatus vorgenommen und kommentiert sowie einige Gattungen und infragenerische Taxa neu aufgestellt.
This article discusses the function of tension in autobiographies written by eighteenth-century doctors George Cheyne, Francis Fuller, Claude Revillon, and the Viscount de Puysegur. It studies how their rhetorical strategies stir tensions in readers through the narration of their own periods of infirmity and search for a remedy. The descriptions of their recoveries offer resolution, legitimate their medical practices, and help diffuse their works. Through the staging of these reversals, the authors suggest a shift in the way the role of medical doctors was perceived as well as a fundamental change in their relationship to illness.
The article sketches a critical paradigm for interdisciplinary work that is centred on tension as a highly ambiguous and ultimately deeply paradoxical notion. It highlights that a unifying account of what tension is or a systematic classification of its diverse meanings would risk resolving tensions between different approaches and privileging a particular mode of doing so. Successively focussing on aesthetic, socio-political, and physical tensions, the essay articulates tension rather as a broad umbrella term that is stretched by multi-perspectival articulations, unified through its intensive surface tension, and at the same time full of transformative and generative potentials. In particular, it proposes that tensions between different cultural or disciplinary fields can be made productive by inducing tensions within each field so that different fields can be related to each other on the basis of tension rather than some substantial commonality.
This article shows that 'tension' cannot be conceived as a specific object of an analysis for which one could determine a precise field of enquiry. Instead, it establishes tension as a specific mode or angle of approach with which any given contingent object or set of objects can be explored. The wideness of its applicability and the specificity of its angle suggest that research on tension can help to unfold a better understanding of a classical ontological question concerning the essential value of actions and relations in the definition of what a thing is. The text follows this line of argumentation by pairing contemporary philosophical sources and specific aesthetic and political examples. Suggesting the possibility of an open classification of different modes of tension, it clarifies the extent to which the essential definition of a thing is bound to the contingent analysis of its transformations.
Das Phänomen Terrorismus ist regelmäßig Gegenstand aktueller Berichterstattung in den Nachrichten und tritt hierbei deutlich als von Medien abhängige aggressive Taktik in Erscheinung. Seit den Anschlägen auf das New Yorker World Trade Center am 11. September 2001 hat es in verstärkter Form eine zentrale Rolle im öffentlichen Diskurs inne. Darüber hinaus stellt der Themenkomplex ein wiederkehrendes Motiv in den Unterhaltungsmedien dar. Diese nehmen Bezug auf zeitgeschichtliche Kontexte, welche oft in fiktionalisierter Form Ver- oder wenigstens Bearbeitung finden.
Wenn es in Franz Kafkas Erzählung 'Eine kaiserliche Botschaft' (1919) gegen Ende heißt: "Niemand dringt hier durch und gar mit der Botschaft eines Toten", so ließe sich dies als das Scheitern eines erwünschten Nachlebens verstehen, erwünscht in Form einer nach dem eigenen Tod bei jemandem ankommenden Botschaft; als das Scheitern einer erwünschten Souveränität, die einen Toten, und sei er auch Kaiser gewesen, offenbar nicht länger auszeichnet. Am Ende der folgenden Überlegungen aber wird eine andere Lektüre stehen: Sie resultiert aus der Frage nach den Bedingungen, Praktiken und Effekten eines Schreibens, das die Idee der Souveränität in bestimmter Weise verfolgt, nämlich hinsichtlich der Organisation des Nachlebens, des Sich-Heranschreibens an die Grenze des Todes, des Sich-selbst-Überlebens - ein testamentarisches Schreiben also. Damit ist zunächst eine juristisch relevante Form bezeichnet: das Testament, das heißt eine konkrete Praxis der Adressierung an die Nachwelt. Diese Praxis ist sowohl institutionalisiert als auch individualisiert, und ihr eignet eine grundlegende Literarizität, nicht nur in formalästhetischer Hinsicht, sondern auch bezogen auf ihren Imaginations- und Fiktionalitätsstatus, der aus der Schreibhaltung des "ich bin tot" herrührt. Dies sei im Folgenden anhand eines etwas grob verfahrenden Cursus entwickelt, dessen erster Teil zum "Testament als Medium der Souveränität" zunächst im antiken römischen Recht einsetzt und dann zu Jean Paul springt; dessen zweiter Teil sich "das testierende Ich" vornimmt; und dessen dritter Teil sich den theoretischen und literarischen 'double binds' eines Testaments und eines "desire to speak with the dead" widmet.

Ad fontes. Wir wissen, daß fast alle Werke Thomas Manns ihre langen Wurzeln haben, und kaum ein anderer Roman hat längere als die Geschichte des Doktor Faustus. Sie sind, wenn man so sagen darf, über vierzig Jahre lang, und es versteht sich von selbst, daß sich der Stoff über einen so großen Zeitraum wandelte. Wir kennen den berühmten Dreizeilenplan aus dem Jahre 1901 - so wurde er von Thomas Mann selbst am 17. März 1943 benannt. Aber es war - darauf hat Peter de Mendelssohn in seinem Nachwort zur Frankfurter Ausgabe des Romans hingewiesen - nicht ein Dreizeilenplan, sondern es gab derer zwei. Der erste lautete (in Thomas Manns Notizbuch 7 festgehalten): "Zum Roman. Der syphilitische Künstler nähert sich von Sehnsucht getrieben einem reinen, süßen jungen Mädchen, betreibt die Verlobung mit der Ahnungslosen und erschießt sich dicht vor der Hochzeit". Man kann diesen Plan durchaus als Variation der Verführungsgeschichte Fausts und Gretchens lesen, auch wenn von einem Teufelspakt nicht die Rede ist und nicht Gretchen umkommt, sondern der Künstler, der Opfer einer venerischen Erkrankung ist. Aber das "reine, süße junge Mädchen": unverkennbar eine Gretchen-Gestalt. Der zweite Plan, wohl auf Ende 1904 zu datieren, ist von Mendelssohn als Variante und Fortführung der ersten Niederschrift charakterisiert worden, aber bei genauerem Hinsehen zeigt sich, daß es eigentlich ein ganz anderer Plan ist, in den der erste zwar aufgenommen ist, aber darin ein Fremdkörper bleibt.

Výsledky dosavadních výzkumŧ tohoto vztahu, jeţ vycházejí z posledních konceptŧ narace jako základní existenciální struktury bytí, naznaĉují, ţe narativní text a receptivní i produktivní narativní pracovní postupy ve vyuĉování a uĉení se jazyku indukují specifický potenciál pozitivních emocí a postojŧ ţáka.
This work investigated the applicability of global pairwise sequence alignment to the detection of functional analogues in virtual screening. This variant of sequence comparison was developed for the identification of homologue proteins based on amino acid or nucleotide sequences. Because of the significant differences between biopolymers and small molecules several aspects of this approach for sequence comparison had to be adapted. All proposed concepts were implemented as the ‘Pharmacophore Alignment Search Tool’ (PhAST) and evaluated in retrospective experiments on the COBRA dataset in version 6.1. The aim to identify functional analogues raised the necessity for identification and classification of functional properties in molecular structures. This was realized by fragment-based atom-typing, where one out of nine functional properties was assigned to each non-hydrogen atom in a structure. These properties were pre-assigned to atoms in the fragments. Whenever a fragment matched a substructure in a molecule, the assigned properties were transferred from fragment atoms to structure atoms. Each functional property was represented by exactly one symbol. Unlike amino acid or nucleotide sequences, small drug-like molecules contain branches and cycles. This was a major obstacle in the application of sequence alignment to virtual screening, since this technique can only be applied to linear sequences of symbols. The best linearization technique was shown to be Minimum Volume Embedding. To the best of knowledge, this work represents the first application of dimensionality reduction to graph linearization. Sequence alignment relies on a scoring system that rates symbol equivalences (matches) and differences (mismatches) based on functional properties that correspond to rated symbols. Existing scoring schemes are applicable only to amino acids and nucleotides. In this work, scoring schemes for functional properties in drug-like molecules were developed based on property frequencies and isofunctionality judged from chemical experience, pairwise sequence alignments, pairwise kernel-based assignments and stochastic optimization. The scoring system based on property frequencies and isofunctionality proved to be the most powerful (measured in enrichment capability). All developed scoring systems performed superior compared to simple scoring approaches that rate matches and mismatches uniformly. The frameworks proposed for score calculations can be used to guide modifications to the atom-typing in promising directions. The scoring system was further modified to allow for emphasis on particular symbols in a sequence. It was proven that the application of weights to symbols that correspond to key interaction points important to receptor-ligand-interaction significantly improves screening capabilities of PhAST. It was demonstrated that the systematic application of weights to all sequence positions in retrospective experiments can be used for pharmacophore elucidation. A scoring system based on structural instead of functional similarity was investigated and found to be suitable for similarity searches in shape-constrained datasets. Three methods for similarity assessment based on alignments were evaluated: Sequence identity, alignment score and significance. PhAST achieved significantly higher enrichment with alignment scores compared to sequence identity. p-values as significance estimates were calculated in a combination of Marcov Chain Monte Carlo Simulation and Importance Sampling. p-values were adapted to library size in a Bonferroni correction, yielding E-values. A significance threshold of an E-value of 1*10-5 was proposed for the application in prospective screenings. PhAST was compared to state-of-the-art methods for virtual screening. The unweighted version was shown to exhibit comparable enrichment capabilities. Compound rankings obtained with PhAST were proven to be complementary to those of other methods. The application to three-dimensional instead of two-dimensional molecular representations resulted in altered compound rankings without increased enrichment. PhAST was employed in two prospective applications. A screening for non-nucleoside analogue inhibitors of bacterial thymidin kinase yielded a hit with a distinct structural framework but only weak activity. The search for drugs not member of the NSAID (non-steroidal anti-inflammatory drug) class as modulators of gamma-secretase resulted in a potent modulator with clear structural distiction from the reference compound. The calculation of significance estimates, emphasizing on key interactions, the pharmacophore elucidation capabilities and the unique compound rannkings set PhAST apart from other screening techniques.

WE DEVELOP A MODEL WHICH ACCOUNTS BOTH FOR INTER- AND INTRA-ORGANIZATIONAL INFLUENCE FACTORS ON THE ADOPTION PROCESS AND EMPIRICALLY IDENTIFIES THE MOST SIGNIFICANT INFLUENCE FACTORS.
The design, construction, and commissioning of the ALICE Time-Projection Chamber (TPC) is described. It is the main device for pattern recognition, tracking, and identification of charged particles in the ALICE experiment at the CERN LHC. The TPC is cylindrical in shape with a volume close to 90 m3 and is operated in a 0.5 T solenoidal magnetic field parallel to its axis.
In this paper we describe in detail the design considerations for this detector for operation in the extreme multiplicity environment of central Pb–Pb collisions at LHC energy. The implementation of the resulting requirements into hardware (field cage, read-out chambers, electronics), infrastructure (gas and cooling system, laser-calibration system), and software led to many technical innovations which are described along with a presentation of all the major components of the detector, as currently realized. We also report on the performance achieved after completion of the first round of stand-alone calibration runs and demonstrate results close to those specified in the TPC Technical Design Report.
The Anthonomus juniperinus group, with descriptions of two new species (Coleoptera: Curculionidae)

The Anthonomus juniperinus (Sanborn) species group is defined and two new species, Anthonomus sanborni, new species, and A. rileyi new species, from the United States are described, keyed and illustrated. The three species of the group are associated with the plant genus Juniperus and the larvae of A. juniperinus are known to develop in fungal galls of Gymnosporangium spp. as well as fruits of the Eastern redcedar, Juniperus virginiana L. The biology of the group and its taxonomic relationships to other species of Anthonomus Germar are also discussed.
Moderately elevated levels of plasma plant sterols have been suspected to be causally involved in atherosclerosis. The aim of this study was to investigate whether plant sterols and other markers of sterol metabolism predicted all-cause and cardiovascular mortality in participants of the Ludwigshafen Risk and Cardiovascular health (LURIC) study. A total of 1,257 individuals who did not use statins and at baseline had a mean (± SD) age of 62.8 (± 11.0) years were included in the present analysis. Lathosterol, cholestanol, campesterol, and sitosterol were measured to estimate cholesterol synthesis and absorption. The mean (± SD) time of the follow-up for all-cause and cardiovascular mortality was 7.32 (± 2.3) years. All-cause (P = 0.001) and cardiovascular (P = 0.006) mortality were decreased in the highest versus the lowest lathosterol to cholesterol tertile. In contrast, subjects in the third cholestanol to cholesterol tertile had increased all-cause (P < 0.001) and cardiovascular mortality (P = 0.010) compared with individuals in the first tertile. The third campesterol to cholesterol tertile was associated with increased all-cause mortality (P = 0.025). Sitosterol to cholesterol tertiles were not significantly related to all-cause or cardiovascular mortality. The data suggest that high absorption and low synthesis of cholesterol predict increased all-cause and cardiovascular mortality in LURIC participants.
The Bafaw Language (Bantu A10) is a product of the language research programme of the Department of Linguistics of the University of Buea. It is the first serious piece of work on this highly endangered language, and aims to account generally for the data of Bafaw. The work therefore lays the foundation for more advanced work in the future. It provides a description of: the phonology, i.e. the sound system; the morphology or lexis; and the syntax of the Bafaw language. The work goes far beyond to provide a sociolinguistic survey of the Bafaw language community, and offers a discussion of functional literacy in Bafaw, the development of an orthography and the thematic glossary of the language. The book provides a useful resource for the Bafaw language development and an inspiration for further research and scholarship.

This paper summarizes the information published on the beetle fauna of the island of St. Vincent (excluding the Grenadine islands). The fauna contains 62 families, with 371 genera, and 536 species. The families with the largest number of species are Staphylinidae (128), Curculionidae (54), Chrysomelidae (47), Scarabaeidae (31), Tenebrionidae (30), and Cerambycidae (29). At least 17 species (3.17%) were probably accidentally introduced to the island by human activities. One hundred four species (19.40%) are endemic (restricted) to the island and likely speciated on the island. One hundred twenty species (22.39%) are shared only with other islands of the Lesser Antilles (Lesser Antillean endemics), and 41 species (7.65%) are more widespread Antilles endemics. The remaining 254 species (47.38%) in the fauna are otherwise mostly widely distributed in the Antilles and the Neotropical Region. The St. Vincent beetle fauna has thus mostly originated elsewhere than on St. Vincent and is largely an immigrant fauna from other islands of the West Indies or the continental Neotropics. Of the St. Vincent species known to occur on other islands, the largest numbers are shared with (north to south) Guadeloupe (206), Dominica (115), Martinique (76), St. Lucia (87) and Grenada (298). Undoubtedly, the real number of species on St. Vincent is higher than now reported and may actually be around 1200 or more species.
The Birds of the Highlands of South-West Saudi Arabia and adjacent parts of the Tihama: July 2010

The objective of the survey was to compare habitats and bird life in the Asir region, particularly Jebal Souda and the Raydah escarpment protected area of the Saudi Wildlife Commission, and adjacent regions of the tihama, with those observed in July 1987 (Jennings, et al., 1988). The two surveys were approximately the same length and equal amounts of time were spent in the highlands and on the tihama. A number of walked censuses were carried out during 2010 on Jebal Souda, using the same methodology as walked censuses in 1987, and the results are compared. Broadly speaking the comparison of censuses revealed that in 2010 there were less birds and reduced diversity on the Jebal Souda plateau, compared to 1987. However in the Raydah reserve the estimates of breeding bird populations compiled in the mid 1990s was little changed as far as could be assessed in 2010. The highland region of south-west Saudi Arabia, especially Jebal Souda, has been much developed since the 1987 survey and is now an important internal recreation and resort area. This has lead to a reduction in the region’s importance for terraced agriculture. These changes may be a contributing factor to changes in bird numbers on the plateau. Subsidiary tasks that arose during the 2010 survey were to help locate satellite tagged Bald Ibises Geronticus eremita from Syria which were transiting Saudi Arabia at the time. Secondly to search for the Asir subspecies of the Eurasian Magpie Pica pica asirensis, which is endemic to the south-west Saudi Arabia highlands, and is reported to be in decline. A separate team searching for the Bald Ibis located some individuals and one was found dead. Few Magpies were located and it seems clear that this very scarce bird has declined further in numbers in recent years. A number of interesting records of birds were obtained, especially on the tihama, where two new birds for Saudi Arabia were observed, Black-headed Heron Ardea melanocephala and Painted Snipe Rostratula benghalensis and one species, Glossy Ibis Plegadis falcinellus was found breeding for the first time in the Arabian Peninsula. Some recommendations for the protection and management of the Raydah reserve are presented. A systematic list of all birds seen is provided.
From the 1980 Maitatsine uprising to the 2009 Boko Haram uprising, Nigeria was bedevilled by ethno-religious conflicts with devastating human and material losses. But the Boko Haram uprising of July 2009 was significant in that it not only set a precedent, but also reinforced the attempts by Islamic conservative elements at imposing a variant of Islamic religious ideology on a secular state. Whereas the religious sensitivity of Nigerians provided fertile ground for the breeding of the Boko Haram sect, the sect’s blossoming was also aided by the prevailing economic dislocation in Nigerian society, the advent of party politics (and the associated desperation of politicians for political power), and the ambivalence of some vocal Islamic leaders, who, though they did not actively embark on insurrection, either did nothing to stop it from fomenting, or only feebly condemned it. These internal factors coupled with growing Islamic fundamentalism around the world make a highly volatile Nigerian society prone to violence, as evidenced by the Boko Haram uprising. Given the approach of the Nigerian state to religious conflict, this violence may remain a recurring problem. This paper documents and analyses the Boko Haram uprising, as well as its links with the promotion of Islamic revivalism and the challenges it poses to the secularity of the Nigerian state.
Central elements of the Bologna declaration have been implemented in a huge variety of curricula in humanities, social sciences, natural sciences and engineering sciences at German universities. Overall the results have been nothing less than disastrous. Surprisingly, this seems to be the perfect time for German universities to talk about introducing a curriculum that is fully compatible with the Bologna declaration for medical education as well. However, German medical education does not have problems the Bologna declaration is intended to solve, such as quality, mobility, internationalization and employability. It is already in the Post-Bologna age.
The bond of shame

A long time ago I suddenly realized that the country one belongs to is not, as the usual rhetoric goes, the one you love but the one you are ashamed of. Shame can be a stronger bond than love. I repeatedly tested my discovery with friends from different countries: they all reacted the same way - with surprise immediately followed by full agreement, as if my suggestion was a self-evident truth. I am not claiming that the burden of shame is always the same; in fact, it varies immensely among countries. But the bond of shame - shame as a bond - invariably works, for a larger or smaller number of individuals. Aristotle listed "shame" ('aidos') among the passions, pointing out that "it is not a virtue" ('Nicomachaean Ethics' 1108 a 30-31). This definition still makes sense. Shame is definitely not a matter of choice: it falls upon us, invading us - our bodies, our feelings, our thoughts - as a sudden illness. It is a passion placed at the intersection between biology and history: the domain which Sigrid Weigel made so distinctively her own.
The Nep1 (Emg1) SPOUT-class methyltransferase is an essential ribosome assembly factor and the human Bowen–Conradi syndrome (BCS) is caused by a specific Nep1D86G mutation. We recently showed in vitro that Methanocaldococcus jannaschii Nep1 is a sequence-specific pseudouridine-N1-methyltransferase. Here, we show that in yeast the in vivo target site for Nep1-catalyzed methylation is located within loop 35 of the 18S rRNA that contains the unique hypermodification of U1191 to 1-methyl-3-(3-amino-3-carboxypropyl)-pseudouri-dine (m1acp3Psi). Specific 14C-methionine labelling of 18S rRNA in yeast mutants showed that Nep1 is not required for acp-modification but suggested a function in Psi1191 methylation. ESI MS analysis of acp-modified Psi-nucleosides in a DeltaNep1-mutant showed that Nep1 catalyzes the Psi1191 methylation in vivo. Remarkably, the restored growth of a nep1-1ts mutant upon addition of S-adenosylmethionine was even observed after preventing U1191 methylation in a deltasnr35 mutant. This strongly suggests a dual Nep1 function, as Psi1191-methyltransferase and ribosome assembly factor. Interestingly, the Nep1 methyltransferase activity is not affected upon introduction of the BCS mutation. Instead, the mutated protein shows enhanced dimerization propensity and increased affinity for its RNA-target in vitro. Furthermore, the BCS mutation prevents nucleolar accumulation of Nep1, which could be the reason for reduced growth in yeast and the Bowen-Conradi syndrome.
The transcription factor p63 is expressed as at least six different isoforms, of which two have been assigned critical biological roles within ectodermal development and skin stem cell biology on the one hand and supervision of the genetic stability of oocytes on the other hand. These two isoforms contain a C-terminal inhibitory domain that negatively regulates their transcriptional activity. This inhibitory domain contains two individual components: one that uses an internal binding mechanism to interact with and mask the transactivation domain and one that is based on sumoylation. We have carried out an extensive alanine scanning study to identify critical regions within the inhibitory domain. These experiments show that a stretch of ~13 amino acids is crucial for the binding function. Further, investigation of transcriptional activity and the intracellular level of mutants that cannot be sumoylated suggests that sumoylation reduces the concentration of p63. We therefore propose that the inhibitory function of the C-terminal domain is in part due to direct inhibition of the transcriptional activity of the protein and in part due to indirect inhibition by controlling the concentration of p63. Efenze, the President of the Board of Directors of government companies and a member of the Central Committee of the Ruling Party, eliminates his erstwhile business contractor, Sancheu, with the complicity of the latter's wife. His aim is to inherit Sancheu's widow and wealth and to forge his way into the Political Bureau of the Party. The Call of Blood is a dramatization of evil in its multifaceted dimensions including treachery, infidelity, greed, hypocrisy, double-crossing and vaulting ambition in a postcolonial society where those who wield political and financial power thrive or perish by their involvement in obscure schemes. The play is enriched by a great sense of dramatic economy and poetic style evident in the preponderant use of local imagery.
The canopy spiders of the floodplain forest in Leipzig have become a focus of ecological studies in recent years. In 2006 we sampled 30 tree canopies in the ‘Burgaue’ nature reserve with pyrethrum knock-down fogging, recording 502 adult spiders belonging to 48 species and 11 families. Based on these data and the results of a previous fogging study, the studied spider community was dominated by forest and forest-edge species with a preference for the shrub and canopy strata as well as by spiders of the web spider feeding guild. The community structure was typical for arboreal spider communities from northern temperate forests but very different from communities in the tropics. Species richness and evenness were similar to the old growth near-primary Białowieża Forest in Poland. The checklist of 96 canopy spider species of the floodplain forest of Leipzig includes 54 additions to the spider fauna of Leipzig and vicinity by recent canopy studies and eight first canopy records for Leipzig from our field work. The theridiid Dipoena torva (Thorell, 1875) was recorded for the first time in Saxony. The floodplain forest of Leipzig sustains a large and species-rich arboreal spider community and is thus a valuable habitat for a large proportion of endangered species (12%).
The Alborz Mountains are forming a ~100 km wide, E-W trending mountain chain where individual summits are up to 5000 m in elevation. The Alborz Mountains range are part of the Alpine orogen and are straddling a 2000 km wide area S of the Caspian Sea. The rocks of the Alborz Mountains consist of Neogen sediments, which are affected by folding and faulting. In the western part of the Alborz Mountains the folds and faults are trending NW-SE, whereas in the eastern part they are trending NE-SW. GPS data confirm N-S shortening including dextral strike-slip along ESE-WNW trending faults, and sinistral strike-slip along ENE-WSW trending faults. The present thesis is focusing on the active Garmsar salt nappe, the fragmented roof of which is pierced by rock salt which extruded near the front of the Alborz Mountains Range. During the past 5 m.y. the front of the Alborz chain migrated towards SSW on top of the salt of the Garmsar basin. The salt was squeezed towards SSW and took place at the Great Kavir. The extruded salt is forming the Eyvanekey plateau between the cities of Eyvanekey and Garmsar. Both the Garmsar salt nappe and the Eyvanekey plateau are dextrally displaced for ca. 9 km along the Zirab-Garmsar fault. Structural analyses of the Garmsar salt nappe indicate three different groups of joints which are trending perpendicular and parallel to the local mechanical anisotropy. The folds of the study area are congruent (type 2 and 3 after Ramsay) resulting from viscose inhomogeneous flow. InSAR-Investigations suggest the Alborz Mountains to be lifted up by ca. 1 cm/a, while horizontal shortening is active at a rate of 8 ±2 mm/a. These values are consistent with GPS data. Based on nine „Advanced Synthetic Aperture Radar“ (ASAR) scenarios, produced by the ENVISAT satellite of the European space agency between 2003 and 2006, we used interferograms to map the displacement via 22 increments during 2 – 18 months. The results suggest that the topographic height of the surface of the salt is changing at a rate which is controlled by the season. The displacement ranges from subsidence at -40 to -50 mm/a to uplift of 20 mm/a. In order to investigate the time-dependent deformation with high spatial resolution, we used algorithms which are based on data of small base lines (SBAS). The resulting interferometric SAR time series analyses also suggest that the study area is largely subsiding at a rate that is controlled by the seasons. The map with the averaged LOS deformation velocities, on the other hand, suggests the subsidence to increase from the upper part of the salt nappe towards deeper topographic 5 levels of the agricultural lowlands. The major part of subsidence is probably caused by the annual rainfall which results in subrosion of salt. The spatial changes in the subsidence rate are probably controlled by the distribution of fountains, mining activity at the margin of the salt glacier, and faults and fractures inside the salt. Striking seasonal imprints are obvious along the agricultural areas which are surrounding the Garmsar salt nappe. These areas are rapidly subsiding in summer and spring when groundwater is used for irrigations. The maximum rate of subsidence (40-50 mm/a) is located E and W of the Eyvanekey plateau, where large areas are irrigated. The maximum displacement is 20 mm/a in the farmland and 5 mm/a in the center of the salt nappe. Depth estimates using Euler deconvolution method for gravimetric and magnetic data suggest the salt to extrude from a depth less than ca. 2000 m. The gravity field of the study area is characterized by strong anomalies in the SW and weak anomalies in the NE. A considerable negative anomaly in the N indicates that the northern part subsided, whereas the southern part was lifted up. The seismic data show three major horizons inside the Miocene sediments: the Lower Red Formation, the Qom Formation, and the Upper Red Formation. The western part of the study area seems to be free from salt domes. The layers of the upper part of the Qom Formation show thinning along the NE and NW trending faults. In some areas the seismic reflectors indicate steep faults close the saddle of the folds. NE-SW-, NW-SE and E-Wtrending faults prevail. Analogue experiments have been carried out to extend our knowledge about the evolution of the Garmsar salt dome. We used a scaled model (34 cm * 25 cm * 2.5 cm) that was shortened perpendicular to its long side. The wedge shape of the Alborz Mountains was simulated by a wedge consisting of Styrofoam. Rock salt was simulated using Polydimethylsiloxan (PDMS), a linear viscous material with a viscosity of 2.3*104 Pa s and a density of 0.96 g/cm3 at room temperature. Other sediments were modeled using dry quartz sand. The experimental results can be used to simulate the structural evolution of the study area: The Alborz deformation front was emplaced on top of the salt rocks in the Garmsar area while migrating towards SSW. A salt basin and a salt extrusion have also been produced in the model. Cross sections through the wedge shaped analogue model indicate N- and S-dipping reverse faults, which are in line with the wedge shape of the Alborz chain. Moreover, ENE-WSW trending sinistral and ESE-WNW trending dextral strike-slip faults led to N-S shortening during the Miocene. Structural marker horizons, 6 which have been turned into Z-folds on the western fold limbs and to S-folds on the eastern fold limbs, are comparable with the folds of the study area. Solving the problem of waste is one of the central tasks of environmental protection. It is becoming increasingly difficult to find suitable sites that are acceptable to the public. Salt and salt formations have relevant properties to be utilizing as a repository for each kind of waste. The favorable properties make rock salt highly suitable as a host rock, in particular for nonradioactive and radioactive wastes. The Qom and Garmsar basins are the nearest salt diapirs to the Tehran province, and there are suitable repositories for waste disposal. Based on surface and subsurface data, the Garmsar salt diapir has been investigated as a case example for its suitability as a host and repository for various types of waste. The data used are based on field studies, interferometry, and geophysical investigations. The results of this study suggest the deep bedded salt of the Garmsar Salt Basin to be an appropriate host for the deposition of industrial waste. Rock salt of surficial layers or domes, on the other hand, is not regarded as an appropriate candidate for waste disposal.
Background: Clock genes and their protein products regulate circadian rhythms in mammals but have also been implicated in various physiological processes, including bone formation. Osteoblasts build new mineralized bone whereas osteoclasts degrade it thereby balancing bone formation. To evaluate the contribution of clock components in this process, we investigated mice mutant in clock genes for a bone volume phenotype. Methodology/Principal Findings: We found that Per2Brdm1 mutant mice as well as mice lacking Cry2-/- displayed significantly increased bone volume at 12 weeks of age, when bone turnover is high. Per2Brdm1 mutant mice showed alterations in parameters specific for osteoblasts whereas mice lacking Cry2-/- displayed changes in osteoclast specific parameters. Interestingly, inactivation of both Per2 and Cry2 genes leads to normal bone volume as observed in wild type animals. Importantly, osteoclast parameters affected due to the lack of Cry2, remained at the level seen in the Cry2-/- mutants despite the simultaneous inactivation of Per2. Conclusions/Significance: This indicates that Cry2 and Per2 affect distinct pathways in the regulation of bone volume with Cry2 influencing mostly the osteoclastic cellular component of bone and Per2 acting on osteoblast parameters.
The Coming African hour is not a slogan, nor wishful thinking. It is a conclusion that derives from an insightful analysis of the current situation pertaining on the continent. Several African scholars, coming from different regions and academic backgrounds are elaborating ideas and arguments in order to explain the constraints and to illustrate the opportunities. The result of that scientific gathering is a book that synthesizes and renews the reflections on development. What is at stake is not to be pessimistic or optimistic about Africa. The epistemological challenge is to understand what is going on. By focusing on converging and diverging African realities, on the issues of state, civil society, gender and development strategies, the authors of the book show under which conditions the African hour is coming. At that level, the commitment for political science meets the commitment for Africa. The main success of this book is to overcome the preconceived ideas and self-fulfilling prophecies about Africa. Here, the analysis avoids the trap of indulgence; then hope is based on truth. Consequently, the coming African hour is not inescapable: it is, as analyzed, a possibility that its achievement depends on institutional, human, political, social and economic factors.

Weltweit berichtete die Presse von den dramatischen Elendszuständen. Eine Fülle von Hilfsorganisationen rief die Bevölkerung der Industriestaaten zur Hilfe auf. In diesem Kontext entstand auch ein Impuls, der den Gestus innergesellschaftlicher Opposition und eines allgemeinen Protestes gegen die moralischen und politischen Handlungsideale der westlichen Gesellschaften, der so typisch für die Rockmusik der späten 1960er und frühen 1970er zu sein schien, mit der Zuwendung zu den Zuständen in den Ländern der Dritten Welt verband. Der in den 1970ern so wichtige Nord-Süd-Konflikt war in der Rockmusikkultur angekommen. Das Concert for Bangladesh ging als erstes großes Wohltätigkeitskonzert in die Popgeschichte ein, wurde zum Vorbild vieler weiterer Großveranstaltungen (darunter die beiden Mammutveranstaltungen Live Aid, 1985, und Live 8, 2005). Angeleitet von dem Wunsch, die Situation der Bangladeschflüchtlinge zu verbessern, wandte sich der indische Musiker Radi Shankar an seinen Freund, den Ex-Beatle George Harrison. Dieser komponierte daraufhin die Benefiz-Single Bangla Desh und organisierte gemeinsam mit Shankar innerhalb von fünf Wochen zwei Konzerte, deren Erlöse der Flüchtlingshilfe zugute kommen sollten. Die beiden Konzerte, die am Mittag und Abend des 1. August 1971 im New Yorker Madison Square Garden vor insgesamt 40.000 Zuschauern stattfanden, wurden sowohl für ein Livealbum mitgeschnitten als auch gefilmt. Der Konzertfilm, der im Folgejahr erschien, enthielt im Gegensatz zu dem Album nicht die gesamte Setlist der Konzerte, dokumentiert dafür aber in einigen Szenen Phasen der Vorbereitung der beiden Shows.

Every man lives for himself, using his freedoms to attain his personal aims, and feels with his whole being that he can at any moment perform or not perform this or that action. The higher a man stands in the social scale, the more connections he has with others and the more power he has over them, the more conspicuous is the predestination and inevitability of every act he commits. Upon this philosophy, a former banker, Hansel Bolingo, suddenly finds [or makes] himself the regional representative of a Chinese firm that deals in crabs in Bangui. This catapults him into a position of instant wealth. His mouth-watering affluence draws immediate attention while his hypnotic powers cause hundreds of [not-so-honest]citizens to clamour for shares from which he builds up a huge fortune. But he soon discovers that he cannot deceive everybody all the time.
Studies have shown that the negative effects of credit market inefficiencies are most felt by smaller firms. Therefore, in countries such as Uganda, where micro enterprises are at the bottom of the economic pyramid, moral hazard and adverse selection severely affect their ability to access formal credit hence limiting their growth potential. Microfinance has been heralded for its use of innovative lending methods to improve access to credit. The last decade has witnessed an unprecedented increase in the outreach of micro lending institutions and the development of financial products suited to the needs of the economically active poor, who often, are unable to obtain credit from mainstream financial institutions. This book analyzes the law and economics theories on access to credit and enterprise finance and based on case studies in Uganda, presents empirical findings of the promise and limits of contractual innovations in micro credit.
This paper investigates the accuracy and heterogeneity of output growth and inflation forecasts during the current and the four preceding NBER-dated U.S. recessions. We generate forecasts from six different models of the U.S. economy and compare them to professional forecasts from the Federal Reserve’s Greenbook and the Survey of Professional Forecasters (SPF). The model parameters and model forecasts are derived from historical data vintages so as to ensure comparability to historical forecasts by professionals. The mean model forecast comes surprisingly close to the mean SPF and Greenbook forecasts in terms of accuracy even though the models only make use of a small number of data series. Model forecasts compare particularly well to professional forecasts at a horizon of three to four quarters and during recoveries. The extent of forecast heterogeneity is similar for model and professional forecasts but varies substantially over time. Thus, forecast heterogeneity constitutes a potentially important source of economic fluctuations. While the particular reasons for diversity in professional forecasts are not observable, the diversity in model forecasts can be traced to different modeling assumptions, information sets and parameter estimates. This book critically discusses missionary Christianity and colonization in Africa as twin enterprises with a common ambition. While the colonialist set out to invest capital and reap profit, the missionary desire was to tend and turn African souls from damnation. It was this desire that drove the missionaries into the interior, propelled by the belief that no land was too remote to escape their attention and vigilance. It equally kept missionary zeal buoyant. The clarification of the concept of salvation within the Roman Catholic Church during the Vatican II Council set in motion the current lethargy that has in some places crippled the mission itself. In retrospect, one can begin to wonder why Africans became Christians. What reasons motivated the early adherents to cling to this foreign religion? Were there some internal deficiencies in African traditional religions, which the Africans hoped to remedy by joining the new religion? Or was it just part of the wholesale flirting with whatever was foreign and perceived to be modern? What baits were used by the missionaries to entice Africans? Christianity posed a danger to many of the time-honoured answers to African problems. These were the values Africans converting to Christianity were expected to abandon. Why have Christians continually returned to their abandoned roots in time of crisis? This moving, well argued, richly documented and empirically substantiated study concludes by cautioning against the stubborn drive at radical conversion to Christianity with scant regard to the imperatives of enculturation.
