Marine isopods, despite being a very diverse group, have been little studied in the Mexican South Pacific. After a revision of 171 specimens collected from Guerrero and Oaxaca, six new species, belonging to five genera, were discovered: Amakusanthura guerrerensis sp. nov., Cortezura caeca sp. nov., Mesanthura antenniformis sp. nov., M. estacahuitensis sp. nov., Skuphonura oaxaquensis sp. nov., and Tinggianthura mexicana sp. nov. In this work, the genera Amakusanthura Nunomura, 1977 and Tinggianthura Chew, Abdul-Rahim & Haji Ross, 2014, are recorded for the first time in the Tropical Eastern Pacific. Also, the distribution range of the genus Skuphonura Barnard, 1925 is increased from the west coast of South America to the southern Mexican Pacific, this being the first record of this genus in the Mexican Pacific. This study doubles the number of species recorded of Anthuridae from the Mexican Pacific, of six to 12 species.
Urban reserves, like other protected areas, aim to preserve species richness but conservation efforts in these protected areas are complicated by high proportions of alien species. We examined which environmental factors determine alien species presence in 48 city reserves of Prague, Czech Republic. We distinguished between archaeophytes, i.e. alien species introduced since the beginning of Neolithic agriculture up to 1500 A. D., and neophytes, i.e. modern invaders introduced after that date, with the former group separately analysed for endangered archaeophytes (listed as C1 and C2 categories on national red list). Archaeophytes responded positively to the presence of arable land that was in place at the time of the reserve establishment, and to a low altitudinal range. In addition to soil properties, neophytes responded to recent human activities with the current proportion of built-up area in reserves serving as a proxy. Endangered archaeophytes, with the same affinity for past arable land as other archaeophytes, were also supported by the presence of current shrubland in the reserve. This suggests that for endangered archaeophytes it may have been difficult to adapt to changing agricultural practices, and shrublands might act as a refugium for them. Forty-six of the 155 neophytes recorded in the reserves are classified as invasive. The reserves thus harbour 67% of the 69 invasive neophytes recorded in the country, and particularly worrisome is that many of the most invasive species are shrubs and trees, a life form that is known to account for widespread invasions with high impacts. Our results thus strongly suggest that in Prague nature reserves there is a high potential for future invasions.
Předmětem článku je analýza struktury německých párových spojení (binominálních dvojic, binominálů), pro něž je příznačné ustálené pořadí členů a jeden celkovy význam. Analýza 754 párových spojení získaných z různých slovníků ukázala, že v roli jednotlivých komponentů vystupuje 11 slovních druhů, přičemž 84,88 % výrazů je spojeno spojkou (und, oder, aber, weder-noch, als, wie), zatímco u 15,12 % funguje jako spojující výraz předložka. Některá párová spojení nemají žádný spojující výraz (bergauf, bergab) nebo se objevují v podobě věty (kommt Zeit, kommt Rat). U 8,36 % párových spojení je využito aliterace. V 28,65 % je použito stejné slovo dvakrát (Jahr für Jahr). Dále se článek zabývá fonologickými, metrickými, sémantickými a pragmatickými znaky párových spojení.
In the qualitative analysis of solutions of partial differential equations, many interesting questions are related to the shape of solutions. In particular, the symmetries of a given solution are of interest. One of the first more general results in this direction was given in 1979 by Gidas, Ni and Nirenberg... The main tool in proving this symmetry and monotonicity result is the moving plane method. This method, which goes back to Alexandrov’s work on constant mean curvature surfaces in 1962, was introduced in 1971 by Serrin in the context of partial differential equations to analyze an overdetermined problem...
Invasive species provide a unique opportunity to evaluate factors controlling biogeographic distributions; we can consider introduction success as an experiment testing suitability of environmental conditions. Predicting potential distributions of spreading species is not easy, and forecasting potential distributions with changing climate is even more difficult. Using the globally invasive coypu (Myocastor coypus [Molina, 1782]), we evaluate and compare the utility of a simplistic ecophysiological based model and a correlative model to predict current and future distribution. The ecophysiological model was based on winter temperature relationships with nutria survival. We developed correlative statistical models using the Software for Assisted Habitat Modeling and biologically relevant climate data with a global extent. We applied the ecophysiological based model to several global circulation model (GCM) predictions for mid-century. We used global coypu introduction data to evaluate these models and to explore a hypothesized physiological limitation, finding general agreement with known coypu distribution locally and globally and support for an upper thermal tolerance threshold. Global circulation model based model results showed variability in coypu predicted distribution among GCMs, but had general agreement of increasing suitable area in the USA. Our methods highlighted the dynamic nature of the edges of the coypu distribution due to climate non-equilibrium, and uncertainty associated with forecasting future distributions. Areas deemed suitable habitat, especially those on the edge of the current known range, could be used for early detection of the spread of coypu populations for management purposes. Combining approaches can be beneficial to predicting potential distributions of invasive species now and in the future and in exploring hypotheses of factors controlling distributions.
Background: The diagnostic and pathophysiological relevance of antibodies to aquaporin-4 (AQP4-Ab) in patients with neuromyelitis optica spectrum disorders (NMOSD) has been intensively studied. However, little is known so far about the clinical impact of AQP4-Ab seropositivity.
Objective: To analyse systematically the clinical and paraclinical features associated with NMO spectrum disorders in Caucasians in a stratified fashion according to the patients' AQP4-Ab serostatus.
Methods: Retrospective study of 175 Caucasian patients (AQP4-Ab positive in 78.3%).
Results: Seropositive patients were found to be predominantly female (p < 0.0003), to more often have signs of co-existing autoimmunity (p < 0.00001), and to experience more severe clinical attacks. A visual acuity of ≤ 0.1 during acute optic neuritis (ON) attacks was more frequent among seropositives (p < 0.002). Similarly, motor symptoms were more common in seropositive patients, the median Medical Research Council scale (MRC) grade worse, and MRC grades ≤ 2 more frequent, in particular if patients met the 2006 revised criteria (p < 0.005, p < 0.006 and p < 0.01, respectively), the total spinal cord lesion load was higher (p < 0.006), and lesions ≥ 6 vertebral segments as well as entire spinal cord involvement more frequent (p < 0.003 and p < 0.043). By contrast, bilateral ON at onset was more common in seronegatives (p < 0.007), as was simultaneous ON and myelitis (p < 0.001); accordingly, the time to diagnosis of NMO was shorter in the seronegative group (p < 0.029). The course of disease was more often monophasic in seronegatives (p < 0.008). Seropositives and seronegatives did not differ significantly with regard to age at onset, time to relapse, annualized relapse rates, outcome from relapse (complete, partial, no recovery), annualized EDSS increase, mortality rate, supratentorial brain lesions, brainstem lesions, history of carcinoma, frequency of preceding infections, oligoclonal bands, or CSF pleocytosis. Both the time to relapse and the time to diagnosis was longer if the disease started with ON (p < 0.002 and p < 0.013). Motor symptoms or tetraparesis at first myelitis and > 1 myelitis attacks in the first year were identified as possible predictors of a worse outcome.
Conclusion: This study provides an overview of the clinical and paraclinical features of NMOSD in Caucasians and demonstrates a number of distinct disease characteristics in seropositive and seronegative patients
Attention-deficit/hyperactivity disorder (ADHD) is a common, highly heritable neurodevelopmental disorder. Genetic loci have not yet been identified by genome-wide association studies. Rare copy number variations (CNVs), such as chromosomal deletions or duplications, have been implicated in ADHD and other neurodevelopmental disorders. To identify rare (frequency ≤1%) CNVs that increase the risk of ADHD, we performed a whole-genome CNV analysis based on 489 young ADHD patients and 1285 adult population-based controls and identified one significantly associated CNV region. In tests for a global burden of large (>500 kb) rare CNVs, we observed a nonsignificant (P=0.271) 1.126-fold enriched rate of subjects carrying at least one such CNV in the group of ADHD cases. Locus-specific tests of association were used to assess if there were more rare CNVs in cases compared with controls. Detected CNVs, which were significantly enriched in the ADHD group, were validated by quantitative (q)PCR. Findings were replicated in an independent sample of 386 young patients with ADHD and 781 young population-based healthy controls. We identified rare CNVs within the parkinson protein 2 gene (PARK2) with a significantly higher prevalence in ADHD patients than in controls (P=2.8 × 10(-4) after empirical correction for genome-wide testing). In total, the PARK2 locus (chr 6: 162 659 756-162 767 019) harboured three deletions and nine duplications in the ADHD patients and two deletions and two duplications in the controls. By qPCR analysis, we validated 11 of the 12 CNVs in ADHD patients (P=1.2 × 10(-3) after empirical correction for genome-wide testing). In the replication sample, CNVs at the PARK2 locus were found in four additional ADHD patients and one additional control (P=4.3 × 10(-2)). Our results suggest that copy number variants at the PARK2 locus contribute to the genetic susceptibility of ADHD. Mutations and CNVs in PARK2 are known to be associated with Parkinson disease.
Nach der Revolution 1917 schlossen sich mehrere russische Avantgardekünstler den kollektiven geistigen Anstrengungen ein, um ein revolutionäres Gesamtkunstwerk der neuen Menschheit zu gestalten. Sie nutzten alle Medien der Kunst, um ihre innovativen Ideen in diesem Experimentierfeld zu verwirklichen. Die Keramik wurde von ihnen als eines der bedeutenden Experimentierfelder und eine zukunftsweisende Kunstgattung gesehen, die durch den alltäglichen Gebrauch das Bewusstsein und die Wahrnehmung der Menschen entscheidend revolutionieren sollte.
Die vorliegende Dissertation untersucht erstmals die Keramik der russischen Avantgarde als eigenständiges künstlerisches Phänomen. Alle dokumentarisch belegten Entwürfe und Keramiken der russischen Avantgardisten werden zusammengefasst sowie alle bekannten Marken der Keramik aufgeführt. Die Autorin systematisiert die Formensprache und Dekormotive der avantgardistischen Keramikwerke und hebt Hintergrunde sowie theoretische Grundlagen der jeweiligen avantgardistischen Gruppen im Bereich Keramik hervor.
Die vorliegende vergleichende Analyse der russischen und westeuropäischen Keramikkunst des ersten Drittels des 20. Jahrhunderts deckt zum ersten Mal in der kunsthistorischen Forschung ihre Gemeinsamkeiten sowie die gegenseitigen Einflüsse auf, dabei werden die Unterschiede der russischen und europäischen avantgardistischen Keramikkunst deutlich reflektiert.

URPOSE: Today, the majority of medical graduates in countries such as the UK, the US or Germany are female. This poses a major problem for workforce planning especially in urology. We here use first the first time the previously established Brüggmann Groneberg (BG) index to assess if female academic career options advance in urology.
METHODS: Different operating parameters (student population, urology specialist population, urology chair female:male (f:m) ratio) were collected from the Federal Office of Statistics, the Federal Chamber of Physicians and the medical faculties of 36 German universities. Four time points were monitored (2000, 2005, 2010 and 2015). From these data, female to male (f:m) ratios and the recently established career advancement (BG) index have been calculated.
RESULTS: The German hospital urology specialists' f:m ratios were 0.257 (499 female vs. 1944 male) for 2015, 0.195 for 2010, 0.133 for 2005 and 0.12 for 2000. The career advancement (BG) index was 0.0007 for 2000, 0,0005 for 2005, 0.094 for 2010 and 0.073 for 2015. The decrease from 2010 to 2015 was due to an increase in the f:m ratio of hospital urologists and female medical students.
CONCLUSION: The BG index clearly illustrated that there is an urgent need for special academic career funding programs to counteract gender problems in urology. The BG index has been shown to be an excellent tool to assess female academic career options and will be very helpful to assess and document positive or negative changes in the next decades.
Dendritic cells (DCs) play a pivotal role in the development of cutaneous contact hypersensitivity (CHS) and atopic dermatitis as they capture and process antigen and present it to T lymphocytes in the lymphoid organs. Recently, it has been indicated that a topical application of the sphingolipid sphingosine 1-phosphate (S1P) prevents the inflammatory response in CHS, but the molecular mechanism is not fully elucidated. Here we indicate that treatment of mice with S1P is connected with an impaired antigen uptake by Langerhans cells (LCs), the initial step of CHS. Most of the known actions of S1P are mediated by a family of five specific G protein-coupled receptors. Our results indicate that S1P inhibits macropinocytosis of the murine LC line XS52 via S1P2 receptor stimulation followed by a reduced phosphatidylinositol 3-kinase (PI3K) activity. As down-regulation of S1P2 not only diminished S1P-mediated action but also enhanced the basal activity of LCs on antigen capture, an autocrine action of S1P has been assumed. Actually, S1P is continuously produced by LCs and secreted via the ATP binding cassette transporter ABCC1 to the extracellular environment. Consequently, inhibition of ABCC1, which decreased extracellular S1P levels, markedly increased the antigen uptake by LCs. Moreover, stimulation of sphingosine kinase activity, the crucial enzyme for S1P formation, is connected not only with enhanced S1P levels but also with diminished antigen capture. These results indicate that S1P is essential in LC homeostasis and influences skin immunity. This is of importance as previous reports suggested an alteration of S1P levels in atopic skin lesions.
The theory of intertemporal consumption choice makes sharp predictions about the evolution of the entire distribution of household consumption, not just about its conditional mean. In the paper, we study the empirical transition matrix of consumption using a panel drawn from the Bank of Italy Survey of Household Income and Wealth. We estimate the parameters that minimize the distance between the empirical and the theoretical transition matrix of the consumption distribution. The transition matrix generated by our estimates matches remarkably well the empirical matrix, both in the aggregate and in samples stratified by education. Our estimates strongly reject the consumption insurance model and suggest that households smooth income shocks to a lesser extent than implied by the permanent income hypothesis. We use responses to survey questions in the 2010 Italian Survey of Household Income and Wealth that ask consumers how much of an unexpected transitory income change they would consume. We find that the marginal propensity to consume (MPC) is 48 percent on average, and that there is substantial heterogeneity in the distribution. We find that households with low cash-on-hand exhibit a much higher MPC than affluent households, which is in agreement with models with precautionary savings where income risk plays an important role. The results have important implications for the evaluation of fiscal policy, and for predicting household responses to tax reforms and redistributive policies. In particular, we find that a debt-financed increase in transfers of 1 percent of national disposable income targeted to the bottom decile of the cash-on-hand distribution would increase aggregate consumption by 0.82 percent. Furthermore, we find that redistributing 1% of national disposable income from the top to the bottom decile of the income distribution would boost aggregate consumption by 0.33%.
The single most important policy-induced innovation in the international financial system since the collapse of the Bretton-Woods regime is the institution of the European Monetary Union. This paper provides an account of how the process of financial integration has promoted financial development in the euro area. It starts by defining financial integration and how to measure it, analyzes the barriers that can prevent it and the effects of their removal on financial markets, and assesses whether the euro area has actually become more integrated. It then explores to which extent these changes in financial markets have influenced the performance of the euro-area economy, that is, its growth and investment, as well as its ability to adjust to shocks and to allow risk-sharing. The paper concludes analyzing further steps that are required to consolidate financial integration and enhance the future stability of financial markets.
Recent models with liquidity constraints and impatience emphasize that consumers use savings to buffer income fluctuations. When wealth is below an optimal target, consumers try to increase their buffer stock of wealth by saving more. When it is above target, they increase consumption. This important implication of the buffer stock model of saving has not been subject to direct empirical testing. We derive from the model an appropriate theoretical restriction and test it using data on working-age individuals drawn from the 2002 and 2004 Italian Surveys of Household Income and Wealth. One of the most appealing features of the survey is that it has data on the amount of wealth held for precautionary purposes, which we interpret as target wealth in a buffer stock model. The test results do not support buffer stock behavior, even among population groups that are more likely, a priori, to display such behavior. The saving behavior of young households is instead consistent with models in which impatience, relative to prudence, is not as high as in buffer stock models. JEL Classification: D91
We review savings trends in Italy, summarizing available empirical evidence on Italians’ motives to save, relying on macroeconomic indicators as well as on data drawn from the Bank of Italy’s Survey of Household Income and Wealth from 1984 to 2004. The macroeconomic data indicate that households’ saving has dropped significantly, although Italy continues to rank above most other countries in terms of saving. We then examine with microeconomic data four indicators of household financial conditions: the propensity to save, the proportion of households with negative savings, the proportion of households with debt, and the proportion of households that lack access to formal credit markets. By international comparison, the level of debt of Italian households and default risk are relatively low. But in light of the deep changes undergone by the Italian pension system, the fall in saving is a concern, particularly for individuals who entered the labor market after the 1995 reform and who have experienced the largest decline in pension wealth. We present an intertemporal consumption model of consumer investment in financial literacy. Consumers benefit from such investment because their stock of financial literacy allows them to increase the returns on their wealth. Since literacy depreciates over time and has a cost in terms of current consumption, the model determines an optimal investment in literacy. The model shows that financial literacy and wealth are determined jointly, and are positively correlated over the life cycle. Empirically, the model leads to an instrumental variables approach, in which the initial stock of financial literacy (as measured by math performance in school) is used as an instrument for the current stock of literacy. Using microeconomic and aggregate data, we find a strong effect of financial literacy on wealth accumulation and national saving, and also show that ordinary least squares estimates underestate the impact of financial literacy on saving. Investment in financial literacy, social security and portfolio choice : [version may 21, 2013]

We present an intertemporal portfolio choice model where individuals invest in financial literacy, save, allocate their wealth between a safe and a risky asset, and receive a pension when they retire. Financial literacy affects the excess return and the cost of stock market participation. Since literacy depreciates over time and has a cost related to current consumption, investors simultaneously choose how much to save, the portfolio allocation, and the optimal investment in literacy. This last depends on households' resources, its preference parameters and on how much financial literacy affects the returns on risky assets and the stock market participation cost, and the returns on social security wealth. The model implies one should observe a positive correlation between stock market participation (and risky asset share, conditional on participation) and financial literacy, and a negative correlation between the generosity of the social security system and financial literacy. The model also implies that the stock of financial literacy accumulated early in life is positively correlated with the individual's wealth and portfolio allocations later in life. Using microeconomic cross-country data, we find support for these predictions.
In 2000 Italy replaced its traditional system of severance pay for public employees with a new system. Under the old regime, severance pay was proportional to the final salary before retirement; under the new regime it is proportional to lifetime earnings. This reform entails substantial losses for future generations of public employees, in the range of €20,000-30,000, depending on seniority. Using a difference-in-difference framework, we estimate the impact of this unanticipated change in lifetime resources, on the current consumption and wealth accumulation of employees affected by the reform. In line with theoretical simulations, we find that each euro reduction in severance pay reduces the average propensity to consume by 3 cents and increases the wealth-income ratio by 0.32. The response is stronger for younger workers and for households where both spouses are public sector employees.
Many studies show that most people are not financially literate and are unfamiliar with even the most basic economic concepts. However, the evidence on the determinants of economic literacy is scant. This paper uses international panel data on 55 countries from 1995 to 2008, merging indicators of economic literacy with a large set of macroeconomic and institutional variables. Results show that there is substantial heterogeneity of financial and economic competence across countries, and that human capital indicators (PISA test scores and college attendance) are positively correlated with economic literacy. Furthermore, inhabitants of countries with more generous social security systems are generally less literate, lending support to the hypothesis that the incentives to acquire economic literacy are related to the amount of resources available for private accumulation. We develop a quantity-driven general equilibrium model that integrates the term structure of interest rates with the repurchase agreements (repo) market to shed light on the com-bined effects of quantitative easing (QE) on the bond and money markets. We characterize in closed form the endogenous dynamic interaction between bond prices and repo rates, and show (i) that repo specialness dampens the impact of any given quantity of asset pur-chases due to QE on the slope of the term structure and (ii) that bond scarcity resulting from QE increases repo specialness, thus strengthening the local supply channel of QE.
We show that the COVID-19 pandemic triggered a surge in the elasticity of non-financial corporate to sovereign credit default swaps in core EU countries, characterized by strong fiscal capacity. For peripheral countries with lower budgetary slackness, the pandemic had essentially no impact on such elasticity. This evidence is consistent with the disaster-induced repricing of government support, which we model through a rare-disaster asset pricing framework with bailout guarantees and defaultable public debt. The model implies that risk-adjusted guarantees in the core were 2.6 times those in the periphery, suggesting that fiscal capacity buffers provide relief to firms’ financing costs.
This work uses financial markets connected by arbitrage relations to investigate the dynamics of price and liquidity discovery, which refer to the cross-instrument forecasting power for prices and liquidity, respectively. Specifically, we seek to understand the linkage between the cheapest to deliver bond and closest futures pairs by using high-frequency data on European governments obligations and derivatives. We split the 2019-2021 sample into three subperiods to appreciate changes in the liquidity discovery induced by the COVID-19 pandemic. Within a cointegration model, we find that price discovery occurs on the futures market, and document strong empirical support for liquidity spillovers both from the futures to the cash market as well as from the cash to the futures market.

Ein solcher Aufschwung kann nach Lage der Dinge in den Bergwerken von Falun nur dem Leser zuteil werden, während der Protagonist in einem Abgrund des Sinns (und der Sinnlichkeit) sich verliert.
Man muß betonen, daß das Paradies in der Päpstin Johanna, so sehr die Reden und Lieder dem Himmel zustreben, innerweltlich konstituiert ist. Auch in diesem Sinne ist Arnims Stück Welttheater. In der fünften Periode wird ein Vorzug der Dichtung darin gesehen, daß sie es erlaube, „das große Leben der Welt zu ahnen“. Gemeint sind die Bewandtnisse der Staaten, Länder und Völker, in die die Schicksale der einzelnen verflochten sind, also die ganze Welt. Für diese Welt entscheidet sich Johanna, nachdem sie das Angebot ihrer überirdischen Mutter, „zum Elemente“ zurückzukehren, abgelehnt hat, mit den Worten: „So nimm mich Welt mit allem deinem Weh [...].“ Tatsächlich weitet sich der Horizont des Stücks im Fortgang der Handlung erheblich: von der Gelehrtensatire zur Epideiktik der Mächtigen, wie man mit einer gewissen Verkürzung sagen könnte. Immerhin trifft zu, daß die Intrige des Teufels nicht durch die Reue einer Jungfrau allein abgewendet werden kann, da es hierzu vielmehr auch der Einmischung des neuen Papstes und eines veritablen Königs und Kaisers (in sukzessiver Personalunion) bedarf. Die Konfiguration des Welttheaters ist ausschweifend, viele Sphären und Formen (und Vertauschungen) umfassend. Mit Arnims Päpstin Johanna verhält es sich, wie zum Teil schon gesehen, so, daß sich Erzählung und Figurenrede in charakteristischer Mischung abwechseln; weiterhin so, daß die Erzählung eingelagerte Märchen und Legenden kennt, während die Figurenrede sich immer wieder in Gedichte, Gebete und Lieder "verwandelt". Die für die Konfiguration des Welttheaters schlechthin typische Überschreitung der dramatischen Form in Richtung auf die epische und lyrische Verselbständigung der Teile hat Tieck mit Rücksicht auf seinen in mehr als einer Hinsicht maßgeblichen Kaiser Octavianus ausgesprochen: „Da Handlung nur ein Theil des Gedichtes seyn sollte, so sind der lyrischen Ergüsse viele, und die Erzählung wird, vorzüglich im ersten Theil, mehr wie einmal selbständig.“ So könnte man auch über Arnims Päpstin Johanna sagen; mit dem Zusatz, daß sie Verselbständigung in Teilen noch weiter geht.
Künstlererzählungen beantworten in der Regel Fragen nach der Entstehung des Talents, nach der Art und Weise der kreativen Praxis und nach der Generalisierung des Singulären, anders gesagt nach der Genealogie, nach der Typologie und nach der Theorie. Angewandt auf Arnims Erzählung, lauten die Fragen: 1. Wie wird Raphael zum Künstler? 2. Was für eine Art Künstler ist Raphael? 3. Welche allgemeinen Maximen der Kunst ergeben sich aus dem speziellen Fall? Im Gegensatz zu manchen Erzählungen, die sich mit der Thematisierung der einen oder anderen Frage begnügen, durchläuft Arnims Text den ganzen Kursus. Entscheidend ist in diesem Fall ist jedoch die Ebene der Narration. Alle genannten Fragen betreffen die Diegese, also das raum-zeitliche Universum der Erzählung. Da die Erzählung aber dazu tendiert, den Akt ihrer eigenen Hervorbringung fortwährend präsent zu halten, wird es notwendig, die Diegese im Prisma der Narration zu betrachten.
Abstraktion, Stilisierung, Symbolisierung, mit diesen Begriffen lässt sich die formale Eigenart der Goetheschen Festspiele annähernd bezeichnen. Hinzu kommt, mit Goethes eigenen Worten, das „Generische”, die zur Gattung hin verdichtete Individualität. Goethe hat allerdings nicht nur die Festspiele im Auge gehabt, sondern zugleich – oder darüber hinaus – das gesamte Spätwerk, schließlich das Charakteristische ‘später’ Werke überhaupt. Die Dramaturgie des Festspiels überlagert sich mit einer Poetik der Spätzeitlichkeit. In einem weiteren Schritt wird der Gegensatz der frühen und späten Zeit auf die Folge der Epochen angewendet, so dass die Explikation des Generischen sich als eine Erläuterung des Klassischen (bzw. der Klassik im Gegensatz zum Sturm und Drang) ausnimmt. Auf allen drei Ebenen – der formalen, der temporalen und der epochalen – zeigt sich das Generische als Visierung des Allgemeinen.

The article presents results of an empirical study which examines productive transfer in the area of text skills competence and the associated writing skill which could be attributed to the learners' contact with several languages. The research was conducted within the research project "Multilingualism in the Czech Republik: Learning and Teaching German after English." After a short presentation of the key concepts in the development of the study and a presentation of selected results, implications and a modell for further research in multilingual writing and on transfer processes between languages are introduced.
Die linguistic landscapes als alltägliches Phänomen vermitteln neben der unmittelbaren Konfrontation mit der gesellschaftlichen Wirklichkeit auch Einblicke in die spezifischen, kulturell vorstrukturierten Kommunikationssituationen, die auch für den Fremdsprachenunterricht interessante Impulse bieten können. Der Beitrag befasst sich mit den linguistic landscapes als wissenschaftlichem Paradigma in seinen verschiedenen Erscheinungsformen. Besonderes Augenmerk wird dabei auf das aktuelle Paradigma gelegt, das auch für das Forschungsfeld des Fremdsprachenlernens und -lehrens eine erweiterte empirische Basis schafft. Im Anschluss daran wird das sprachdidaktische Potential von Sprachlandschaften fokussiert, indem auf die sprachliche Vielfalt eingegangen wird, die in der Stadt Brünn überall im öffentlichen Raum feststellbar ist. Das Projekt "Linguistic landscapes von Brünn (Brno)" spiegelt nicht nur die Sprachkontakte und die Sprachenpolitik in diesem Raum wider, sondern macht auch die hier ablaufenden Kulturkontakte transparent und zeigt, wie der öffentliche Raum als Diskursplattform genutzt wird.
In den letzten Jahren wird im Hochschulfach Deutsch als Fremdsprache eine intensive Diskussion hinsichtlich gewisser Defizite bei der Kulturvermittlung geführt, die das Ziel verfolgt, kulturbezogene Inhalte in den DaF-Unterricht zu (re)integrieren. Im Beitrag wird auf die Gründe des wenig zufriedenstellenden Ist-Zustandes eingegangen und auf die wichtige Rolle kulturwissenschaftlicher Theoriebildung für den DaF-Unterricht und das Potenzial des Konzeptes der Linguistic Landscapes hingewiesen, das auf kulturwissenschaftlicher Basis bereits ausgearbeitet ist und neue Perspektiven für einen Fremdsprachenunterricht eröffnet, der den engen Zusammenhang von Sprache und Kultur akzentuieren möchte. Im zweiten Teil des Beitrags wird das didaktische Potenzial dieses Konzepts an einigen konkreten Unterrichtsvorschlägen verdeutlicht.
Adenosintriphosphat (ATP) als universelles Energieäquivalent der Zelle wird durch die oxidative Phosphorylierung synthetisiert, bei der Elektronen entlang des elektrochemischen Gefälles der Atmungskette über verschiedene Redoxkomplexe transferiert und durch die chemiosmotische Kopplung Protonen über die Membran gepumpt werden. Der Protonengradient wird dann von der ATP-Synthase genutzt, um ADP zu ATP zu phosphorylieren. Zentraler Redoxkomplex der Atmungskette vieler Pro- und Eukaryonten ist der bc1-Komplex, der Elektronen von Ubichinol auf Cytochrom c überträgt, von wo sie nachfolgend auf die Cytochrom c-Oxidase transferiert werden. Das mesophile Bodenbakterium Paracoccus denitrificans wird als Modellsystem für den mitochondrialen Elektronentransfer (ET) verwendet, da es eine aerobe Atmungskette exprimiert, die der mitochondrialen homolog ist, allerdings einen wesentlich einfacheren Aufbau der einzelnen Redoxkomplexe aufweist. Auch Thermus thermophilus als extrem thermophiles Bakterium bildet eine vergleichbare aerobe Atmungskette aus, wobei deren Proteine allerdings eine hohe Thermostabilität aufweisen, wodurch sie in das Interesse der Forschung gerückt sind. Ziel dieser Arbeit war die funktionelle Charakterisierung des ET zwischen Komplex III und IV der Atmungsketten von P. denitrificans und T. thermophilus, die aufgrund ihrer mesophilen und thermophilen Eigenschaften unterschiedliche Mechanismen in der Wechselwirkung ihrer Redoxproteine aufweisen. Es wurden lösliche Redoxfragmente verwendet, um anhand von Mutagenesestudien, Stopped-Flow (SF)- und Laserflash-Kinetiken (LF) unter pre-steady state-Bedingungen sowie FRET-Experimenten den ET zu untersuchen. Für die LF-Experimente wurde eine photoaktive Rutheniumverbindung kovalent an Cyt c552 gekoppelt, durch die bei Laseranregung das Häm photooxidiert und dadurch der ET von einem reduzierten Redoxpartner induziert werden kann. Die Strukturdaten des bc1/Cyt c-Cokomplexes aus Hefe zeigen, dass direkte Kontakte zwischen Cyt c1 und Cyt c durch unpolare Wechselwirkungen und eine zentrale Kation-p-Interaktion vermittelt werden. Daher wurden anhand von Sequenz- und Struktur-alignments äquivalente Aminosäurepositionen im Paracoccus Cyt c1 identifiziert, die an der Wechselwirkung zu Cyt c552 beteiligt sein könnten. Diese wurden durch gerichtete Mutagenese in ihrem Raumvolumen, ihrer Polarität oder Ladung variiert und in kinetischen Studien untersucht. Für die LF-Kinetiken sowie für die FRET-Experimente wurden Oberflächen-Cysteinmutanten des Cyt c1 bzw. c552 generiert, über deren SH-Gruppen kovalent der Rutheniumkomplex bzw. Fluorophore gekoppelt werden konnten. Die ET-Reaktion zeigt zwei Phasen in Abhängigkeit von der Ionenstärke. Bei niedrigen Ionenstärken ergeben sich Geschwindigkeitskonstanten von 109 M -1s-1 und eine geringe Ionenstärkeabhängigkeit (etwa eine effektive Ladung), wohingegen bei Ionenstärken ab 35 mM 2-3 effektive Ladungen pro Redoxpartner beteiligt sind und bimolekulare Geschwindigkeitskonstanten von 107 bis 109 M-1s-1 erhalten werden. Diese Ergebnisse deuten auf die Bildung eines Encounter-Komplexes bei niedrigen Ionenstärken hin, der ein Ensemble verschiedener Distanzen und relativer Orientierungen der Redoxpartner zueinander darstellt. Diese Annahme wurde ebenfalls durch FRET-Experimente bestätigt, durch die selbst bei niedrigen Ionenstärken keine definierten Abstände erhalten werden konnten. Die Geschwindigkeitskonstanten der bimolekularen Reaktion weisen auf einen diffusionskontrollierten Prozess hin, an dem 2 bis 3 effektive Ladungen an der elektrostatischen Annäherung der beiden Redoxpartner beteiligt sind. Diese Ergebnisse werden durch vorherige kinetische Untersuchungen und EPR-Studien des ET von Cyt c552 zum CuA-Fragment der aa3-Oxidase bekräftigt, für den die gleiche Anzahl effektiver Ladungen und die Bildung eines Encounter-Komplex gefunden wurden. Lediglich c1-Mutanten, deren variierte Aminosäuren sich direkt oberhalb der Hämspalte und in der Sequenz um bzw. innerhalb des Hämbindemotivs befinden, bewirken eine Verlangsamung der Gesamtreaktion und ein leicht verändertes Ionenstärkeverhalten, das auf eine leicht verschobene Interaktionsfläche hindeutet. Es wurde durch Sequenz- und Strukturalignments kein aromatischer Rest in direkter Nähe der entsprechenden Position im Paracoccus Cyt c1 gefunden, der wie im Hefekomplex eine stabilisierende Kation-p-Interaktion zwischen den Redoxpartnern vermittelt. Zur Untersuchung des ET zwischen Komplex III und Komplex IV aus T. thermophilus wurde die hydrophile Cytochrom c-Domäne der caa3-Oxidase in Analogie zum Paracoccus-System als lösliches Redoxfragment kloniert, heterolog exprimiert und charakterisiert. Dieses Fragment wurde in SF-Studien eingesetzt, um den ET zu seinen potentiellen Redoxpartnern Cyt cbc des bc-Komplexes und Cyt c552 zu charakterisieren. Es konnte gezeigt werden, dass das ccaa3-Fragment Elektronen von beiden Redoxpartnern empfängt, wobei die ET-Reaktion mit Cyt c552 so schnell verläuft, dass sie durch SF-Techniken nur schlecht aufgelöst und lediglich eine Größenordnung der Geschwindigkeitskonstanten abgeschätzt werden kann (kon ~ 1010 M-1s-1). Die Reaktion mit Cyt cbc verläuft auch noch schnell (kon = 108-109 M-1s-1) und ist nahezu unabhängig von der Ionenstärke, was eine hydrophobe Wechselwirkung zwischen beiden Redoxpartnern bestätigt. Die Ergebnisse deuten erstmals darauf hin, dass ein ET direkt zwischen zwei Redoxproteinkomplexen (bc-Komplex und caa3-Oxidase) stattfindet, ohne dass ein Elektronenüberträger (wie z. B. Cyt c552) zwischengeschaltet ist. Um die Übertragbarkeit der Erkenntnisse, die hier über den ET an löslichen Redoxfragmenten gewonnen wurden, in einem Proteinkomplex zu verifizieren, wurde ein bc1-Komplex verwendet, in dem die für P. denitrificans einzigartige saure Cyt c1-Domäne deletiert ist. Der Komplex wurde für eine erleichterte Aufreinigung mit einem His-tag versehen, homolog exprimiert und erste Aufreinigungs- und Charakterisierungsschritte unternommen. Er soll zukünftig eingesetzt werden, um die Mutationen des löslichen c1-Fragmentes in den Komplex zu überführen und die Mutanten kinetisch mittels pre-steady state- und steady state-Techniken im Vergleich zum bc1WT-Komplex zu untersuchen, ohne dass die Effekte der Punktmutationen durch die hohe negative Ladungsdichte der sauren Domäne überlagert werden.

THIS EXPLORATORY STUDY INVESTIGATES DRIVERS OF THE CRYPTOCURRENCY EX CHANGE COMPETITION. WE EXAMINE THE IMPACT OF MARKET-RELATED AND COMMUNITY-RELATED ASPECTS OF CRYPTOCURRENCY EXCHANGES ON TWO DISTINCT TYPES OF COMPETITION. OUR EMPIRICAL ANALYSIS OF THREE DATASETS INDICATES THAT THE COMPETITION FOR TRADING FREQUENCY IS DRIVEN BY BOTH THE MARKET AS WELL AS THE COMMUNITY WHEREAS THE COMPETITION FOR TRADING QUANTITY IS DRIVEN SOLELY BY THE MARKET.
Hintergrund: Die häufigsten infektiösen Ursachen für Durchfallerkrankungen sowohl im Kindes- als auch im Erwachsenenalter sind viraler Genese. Dabei werden Rota-, Noro-, Adeno- und Astroviren in absteigender Reihenfolge benannt. Die Diagnose der oft nosokomialen Infektionen erfolgt durch Virusnachweis in Stuhlproben. Material und Methodik: In dieser retrospektiven epidemiologischen Auswertung wurden anhand der Stuhlproben von Gastroenteritispatienten im Zeitraum 2000 – 2008 die Häufigkeitsverteilung der einzelnen Viren sowie saisonale Aspekte untersucht. Bestimmt wurden des Weiteren die Geschlechts- und die Altersverteilung der Patienten. Der überwiegende Anteil der eingesandten Proben entstammte der Universitätsklinik Frankfurt/Main; hinzu kamen Proben von einigen in näherer Umgebung liegenden Gesundheitsämtern, Krankenhäuser und Laborarztpraxen. Ergebnisse: Die laborchemische Effizienz beträgt ca. 10 – 20 %. In Deutschland ist die winterliche Rotavirusinfektion bei Kleinkindern die häufigste Ursache des Brechdurchfalls. An zweiter Stelle stehen im Wechsel Adeno- und Norovirusinfektionen, während Astrovirusinfektionen in den letzten Jahren selten geworden sind. Schlussfolgerung: In Übereinstimmung mit neuen Studien aus anderen Regionen wird belegt, dass Noroviren des Typ 2 heute einen wesentlichen Anteil bei der infektiösen Gastroenteritis stellen und – im Unterschied zu Rotaviren – vor allem ältere Menschen betroffen sind.
Organische Verbindungen sind für den Einsatz in der Elektronik und Optoelektronik von großem Interesse, da ihre Eigenschaften durch Derivatisierung gezielt verändert werden können. Eine vielversprechende Strategie ist hierbei der Einbau von Hauptgruppenelementen (z.B. Boratomen) in die Kohlenstoffgerüste.
Bislang fehlte jedoch ein detaillierter Vergleich der optischen und elektronischen Eigenschaften einer umfassend charakterisierten Kohlenwasserstoffspezies mit denen einer isostrukturellen Bor-dotierten Verbindung. In unserer Gruppe wird 9,10-Dihydro-9,10-diboraanthracen (DBA) als Bor-haltiger Baustein zum Aufbau lumineszierender Polymere genutzt. Das isostrukturelle Kohlenstoffanalogon zum DBA-Fragment stellt hierbei das Anthracen-Gerüst dar. Vor diesem Hintergrund wurden in dieser Arbeit die elektronischen, strukturellen und optischen Eigenschaften des DBA-Derivats 30 und des Anthracen-Derivats 31 umfassend untersucht und miteinander verglichen, wobei signifikante Unterschiede zwischen der Kohlenstoffverbindung 31 und dem Bor-dotierten Analogon 30 beobachtet wurden.
Ziel: Anliegen des Kooperationsprojektes der Klinik für Nephrologie und der Klinik für Psychosomatische Medizin und Psychotherapie ist die internistische und eine umfassende psychologische Untersuchung von152 Lebendnierenspendern, die ihre Niere zwischen 1973 und 2001 in der Universitätsklinik Frankfurt am Main spendeten. Im Rahmen dieser Studie werden aus der oben genannten Arbeitsgruppe heraus, mehrere Arbeiten und Publikationen entstehen. Ziel der vorliegenden Arbeit ist die Untersuchung der 152 Frankfurter Lebendnierenspender in Bezug auf psychosomatische und psychosoziale Aspekte des Erlebens und der Verarbeitung der Lebendnierentransplantation und ihrer Folgen. In der bisherigen empirischen Forschung zu psychischen und psychosomatischen Folgen einer Lebendnierentransplantation wurden beim Spender eher wenige und wenn, dann im Ausmaß begrenzte psychische Komplikationen berichtet. In der Regel sind die psychische Verarbeitung sowie die psycho-sozialen Auswirkungen einer Lebendnierentransplantation insgesamt positiv zu bewerten.
Methode: N= 152 Lebendnierenspender werden internistisch und psychologisch untersucht. Die psychologische Untersuchung verwendet ein breites Spektrum von Erhebungsmethoden. Neben vier standardisierten testpsychologischen Fragebögen wird ein semistrukturiertes ca. einstündiges Interview mit den Spendern geführt. Die vorliegende Arbeit befasst sich gesondert mit dem halbstrukturierten Interview. Die Erlebnisberichte der Spender werden mittels eines eigens erstellten Kategoriensystems ausgewertet.
Ergebnisse: Abschluss der Datenerhebung der vorliegenden Arbeit ist der 15. Februar 2002. Sieben Spender verstarben vor Beginn der Studie, jedoch nicht an den Folgen der Einnierigkeit.Drei Spender waren nicht auffindbar. 19 Spender wurden wegen Wohnsitz im Ausland und/oder Mangel an deutschen Sprachkenntnissen vom psychologischen Interview ausgeschlossen. Von den 123 in Frage kommenden Untersuchungsteilnehmern haben wir mit 100 Spendern Interviews führen können, was einer vergleichsweise hohen Rücklaufquote von 81,3% entspricht. Die meisten Spender trafen ihre Entscheidung sofort (84%) und bereuten ihre Spende im Nachhinein nicht.
Nahezu alle Spender (97%) würden die Entscheidung ihre Niere spenden zu wollen heute wieder treffen. Die meisten Spender bewerten die Spende als ein positives Ereignis vergleichbar mit einer Lebensrettung oder einer Geburt. Einige Spender berichten durch die Spende eine Steigerung ihres Selbstwertgefühls und Selbstbewusstseins erfahren zu haben. 75% der Spender schildern durch die Spende keine Veränderung in der Beziehung zu dem Empfänger erlebt zu haben, bei 23% habe sich die Beziehung verbessert. 3% geben an, die Beziehung zu bestimmten Familienmitgliedern sei nach der Spende schlechter geworden. 3% der Spender bereuen gespendet zu haben. 8% empfanden Druck im Entscheidungsprozess. 5% hatten starke Angst vor der Operation oder dem Leben mit einer Niere. Insgesamt 6% der Spender berichten über langfristige psychische Komplikationen (Verdacht auf: 2% Anpassungsstörung, 2% Angststörung, 1% Depression, 1% Burnout). 11% wünschen sich eine professionelle psychologische Vor- und/oder Nachbetreuung.
Diskussion: Die Ergebnisse der Untersuchung weisen insgesamt auf eine langfristig positive psychische Verarbeitung, sowie auf positive psychosoziale Auswirkungen einer Lebendnierentransplantation hin. Es gibt eine inhomogene Subgruppe mit kleiner Personenanzahl, die negative Erfahrungen mit der Lebendnierenspende machte. Dieser wird gesondert Beachtung geschenkt und die Bereitstellung von adäquaten Beratungs- und Hilfsangeboten diskutiert.
Very-long-baseline interferometry (VLBI) observations of active galactic nuclei at millimetre wavelengths have the power to reveal the launching and initial collimation region of extragalactic radio jets, down to 10–100 gravitational radii (rg ≡ GM/c2) scales in nearby sources. Centaurus A is the closest radio-loud source to Earth. It bridges the gap in mass and accretion rate between the supermassive black holes (SMBHs) in Messier 87 and our Galactic Centre. A large southern declination of −43° has, however, prevented VLBI imaging of Centaurus A below a wavelength of 1 cm thus far. Here we show the millimetre VLBI image of the source, which we obtained with the Event Horizon Telescope at 228 GHz. Compared with previous observations, we image the jet of Centaurus A at a tenfold higher frequency and sixteen times sharper resolution and thereby probe sub-lightday structures. We reveal a highly collimated, asymmetrically edge-brightened jet as well as the fainter counterjet. We find that the source structure of Centaurus A resembles the jet in Messier 87 on ~500 rg scales remarkably well. Furthermore, we identify the location of Centaurus A’s SMBH with respect to its resolved jet core at a wavelength of 1.3 mm and conclude that the source’s event horizon shadow should be visible at terahertz frequencies. This location further supports the universal scale invariance of black holes over a wide range of masses.
Background: Does the dogma of nephron sparing surgery (NSS) still stand for large renal masses? Available studies dealing with that issue are considerably biased often mixing imperative with elective indications for NSS and also including less malignant variants or even benign renal tumors. Here, we analyzed the oncological long-term outcomes of patients undergoing elective NSS or radical tumor nephrectomy (RN) for non-endophytic, large (≥7cm) clear cell renal carcinoma (ccRCC).
Methods: Prospectively acquired, clinical databases from two academic high-volume centers were screened for patients from 1980 to 2010. The query was strictly limited to patients with elective indications. Surgical complications were retrospectively assessed and classified using the Clavien-Dindo-classification system (CDS). Overall survival (OS) and cancer specific survival (CSS) were analyzed using the Kaplan-Meier-method and the log-rank test.
Results: Out of in total 8664 patients in the databases, 123 patients were identified (elective NSS (n = 18) or elective RN (n = 105)) for ≥7cm ccRCC. The median follow-up over all was 102 months (range 3–367 months). Compared to the RN group, the NSS group had a significantly longer median OS (p = 0.014) and median CSS (p = 0.04).
Conclusions: In large renal masses, NSS can be performed safely with acceptable complication rates. In terms of long-term OS and CSS, NSS was at least not inferior to RN. Our findings suggest that NSS should also be performed in patients presenting with renal tumors ≥7cm whenever technically feasible. Limitations include its retrospective nature and the limited availability of data concerning long-term development of renal function in the two groups.
Inhibition of fatty acid synthesis (FAS) stimulates tumor cell death and reduces angiogenesis. When SH-SY5Y cells or primary neurons are exposed to hypoxia only, inhibition of FAS yields significantly enhanced cell injury. The pathophysiology of stroke, however, is not only restricted to hypoxia but also includes reoxygenation injury. Hence, an oxygen-glucose-deprivation (OGD) model with subsequent reoxygenation in both SH-SY5Y cells and primary neurons as well as a murine stroke model were used herein in order to study the role of FAS inhibition and its underlying mechanisms. SH-SY5Y cells and cortical neurons exposed to 10 h of OGD and 24 h of reoxygenation displayed prominent cell death when treated with the Acetyl-CoA carboxylase inhibitor TOFA or the fatty acid synthase inhibitor cerulenin. Such FAS inhibition reduced the reduction potential of these cells, as indicated by increased NADH2+/NAD+ ratios under both in vitro and in vivo stroke conditions. As observed in the OGD model, FAS inhibition also resulted in increased cell death in the stroke model. Stroke mice treated with cerulenin did not only display increased brain injury but also showed reduced neurological recovery during the observation period of 4 weeks. Interestingly, cerulenin treatment enhanced endothelial cell leakage, reduced transcellular electrical resistance (TER) of the endothelium and contributed to poststroke blood-brain barrier (BBB) breakdown. The latter was a consequence of the activated NF-κB pathway, stimulating MMP-9 and ABCB1 transporter activity on the luminal side of the endothelium. In conclusion, FAS inhibition aggravated poststroke brain injury as consequence of BBB breakdown and NF-κB-dependent inflammation.
Ibuprofen gehört zu den am meisten verwendeten nichtsteroidalen Antiphlogistika (NSAIDs) und ist in niedriger Dosierung von 200-400 mg als sogenanntes OTC (over the counter) Analgetikum frei erhältlich. Obwohl es in den meisten Fällen als razemische Mischung aus S- und R-Ibuprofen verabreicht wird, ist mittlerweile auch das reine S-Ibuprofen (Dexibuprofen) als Medikament erhältlich. Während S-Ibuprofen ein potenter COX-Inhibitor ist, zeigt R-Ibuprofen in den klinisch relevanten Konzentrationen keine COX-Hemmung. Seit längerem ist bekannt, dass NSAIDs bei einer kontinuierlichen Einnahme über mehrere Jahre die Initiierung und Proliferation von Tumoren hemmen. Die Hemmung der Cyclooxygenase wird dabei als ein wichtiger Mechanismus für die antikarzinogene Wirkung angesehen, doch für viele NSAIDs sind auch schon COX unabhängige Mechanismen beschrieben worden. In der vorliegenden Arbeit wurde unter Verwendung zweier Tumorzelllinien mit unterschiedlicher COX-2 Expression und dem Einsatz von beiden Ibuprofen Enantiomeren untersucht, inwieweit COX unabhängige Mechanismen für die antikarzinogenen Effekte von S- und R-Ibuprofen verantwortlich sind. Sowohl die COX-2 defizienten HCT-15 als auch die COX-2 exprimierenden HCA-7 Zellen wurden durch Behandlung mit S- oder R-Ibuprofen in ihrer Proliferaton gehemmt. Dabei führte die Behandlung mit S- und R-Ibuprofen für 24 h zu einem G1-Zellzyklusblock und nach 72 h zu einem signifikanten Anstieg von apoptotischen Zellen, was im Western-Blot Assay durch eine verminderte Cyclin A und B Expression, einer erhöhten Expression des Zellzyklusinhibitors p27KIP1 und PARP-Spaltung bestätigt wurde. Dabei zeigten beide Enantiomere in den Tumorzellen eine gleich starke antiproliferative und apoptotische Wirkung. Eine Messung der intrazellulären S- und R-Ibuprofen Konzentrationen ergab außerdem, dass in den verwendeten Tumorzellen keine unidirektionale Konfigurationsinversion von R- zu S-Ibuprofen stattgefunden hat. In den COX-2 exprimierenden HCA-7 Zellen waren die antikarzinogenen Effekte von S- und R-Ibuprofen schwächer ausgeprägt als in den HCT-15 Zellen, was jedoch auf eine niedrigere intrazelluläre S- bzw. R-Ibuprofen Konzentration zurückzuführen war. Diese Ergebnisse zeigen auf jeden Fall, dass COX unabhängige Mechanismen bei der antikarzinogenen Wirkung von S- und R- Ibuprofen eine Rolle spielen. Dass auch COX abhängige Mechanismen an der antikarzinogenen Wirkung von Ibuprofen beteiligt sind, konnte in verschiedenen Studien gezeigt werden. In der vorliegenden Arbeit wurde eine Abnahme an PGE2 nach S-Ibuprofen Behandlung in den COX-2 exprimierenden Tumorzellen beobachtet. In welchem Ausmaß aber die COX Hemmung an der antikarzinogenen Wirkung von S-Ibuprofen in den COX-2 exprimierenden (Hemmung der PGE2 Synthese) und möglicherweise auch in den COX-2 defizienten Zellen (Anstieg an Arachidonsäure und daraus resultierend an Ceramid) beteiligt war, konnte anhand der Ergebnisse nicht geklärt werden. Es kann jedoch angenommen werden, dass S-Ibuprofen zumindest in den COX exprimierenden HCA-7 Zellen sowohl über COX Hemmung als auch über COX unabhängige Mechanismen antiproliferativ und apoptotisch wirkt. Auch im Tumormodell der Maus wurde bei einer täglichen i.p. Applikation von 15 mg/kg S- oder R-Ibuprofen eine Hemmung des Tumorwachstums beobachtet. Dabei betrug die unidirektionale Inversion von R- zu S-Ibuprofen ca. 54 %. Die Inkubation mit S- bzw. R-Ibuprofen führte in beiden Zelllinien außerdem zu einer Akkumulation des Tumorsuppressors p53, der an der Regulierung von Zellzyklus und Apoptose beteiligt ist. Daher wurde im zweiten Teil der Arbeit untersucht, ob S- und R-Ibuprofen die Induktion von Apoptose und Zellzyklusarrest über die Aktivierung des Transkriptionsfaktors p53 und dessen Zielgene vermitteln. Verwendet wurde eine HCT-116 Zelllinien, in der beide p53 Allele durch homologe Rekombination inaktiviert waren (p53-/-). Im Gegensatz zu den HCT-116 p53wt Zellen führte die Behandlung mit S- bzw. R-Ibuprofen in den p53 defizienten Zellen zu keinem signifikanten G1-Zellzyklusblock und auch die Apoptoserate war in diesen Zellen deutlich geringer als in p53wt Zellen. In vivo zeigte eine tägliche i.p. Injektion von 15 mg/kg S- bzw. R-Ibuprofen bei den Mäusen mit p53-/- Tumoren keine signifikante Wachstumshemmung. Im Vergleich dazu wurde die Proliferation der HCT-116 p53wt Tumore durch die R-Ibuprofen Behandlung signifikant gehemmt, und auch die mit S-Ibuprofen behandelten Tieren zeigten deutlich kleinere Tumore als die Gruppe mit den p53 defizienten Tumoren. Nach der Inkubation mit S- bzw. R-Ibuprofen wurde in beiden HCT-116 Zelllinien eine erhöhte Expression des Membranrezeptors p75NTR beobachtet. Außerdem führte die Behandlung mit S- bzw. R-Ibuprofen in den p53wt Zellen, im Gegensatz zu den p53 defizienten Zellen, zu einer vermehrten Bax Expression und einem Anstieg an Bax Protein in der mitochondrialen Fraktion. Schließlich wurde gezeigt, dass die erhöhte p53 Konzentration in den Zellen zumindest teilweise über den p75NTR Rezeptor reguliert wird, da durch die externe Zugabe des Liganden NGF die Ibuprofen induzierte Akkumulation von p53 in den HCT-116 p53wt Zellen wieder aufgehoben wurde. Da in vitro in den HCA-7 Zellen nicht nur die Behandlung mit S-Ibuprofen, sondern auch die mit R-Ibuprofen zu einer Hemmung der PGE2 Synthese führte, wurde im dritten Teil der Arbeit die Wirkung von S- und R-Ibuprofen auf die Expression der PGE-Synthasen und auf die mPGES-1 Aktivität untersucht. Dabei wurden HeLa Zellen verwendet, deren mPGES-1 Expression durch die Zugabe von IL-1ß und TNF-alpha stimmuliert wurde. Nach gleichzeitiger Inkubation mit S- bzw. R-Ibuprofen wurde keine Inhibierung der mPGES-1, mPGES-2 oder cPGES Expression beobachtet. Auch die Enzymaktivität von mPGES-1 wurde weder durch die Inkubation mit S-Ibuprofen noch mit R-Ibuprofen beeinflußt. Die bei der Inkubation mit R-Ibuprofen auftretende Hemmung der PGE2 Synthese konnte in der vorliegenden Arbeit nicht abschließend erklärt werden und bietet somit einen interessanten Ansatzpunkt für weitere Untersuchungen.
