(±)-Aeroplysinin-1, an optically active 1.2-dihydroarene-1.2-diol. was isolated from the marine sponges Verongia aerophoba (+-isomer) and lanthella ardis (--isomer). For the experiments presented we used the +-isomer from Verongia aerophoba. Here we describe the hitherto unknown biological and pharmacological property of this compound to display pronounced anticancer activity against L5178y mouse lymphoma cells (ED50: 0.5 μm). Friend erythroleukemia cells (ED50: 0.7μm) , human mamma carcinoma cells (ED50: 0.3μm) and human colon carcinoma cells (ED50: 3.0 μm) in vitro. Furthermore, aeroplysinin caused a preferential inhibition of [3H]thymidine (dThd) incorporation rates in L5178y mouse lymphoma cells if compared with murine spleen lymphocytes in vitro. At concentrations between 1.1 and 28.5 μm, the [3H]dThd incorporation rates in L5178y cells were suppressed to 28% -0% but only to 78% -18% in murine spleen lymphocytes. The same differential effect in vitro was found with the following epithelial cells: 14.70 μm of the compound were required to inhibit normal human fibroblasts to 50% , but only 2.9 μm in the assays with human malign keratinocytes or malignant melanoma cells to observe the same inhibitory effect. Moreover, aeroplysinin-1 displayed antileukemic activity in vivo using the L5178y cell/NMRI mouse system; administered at a dose of 50 mg/kg for five consecutive days, the T/C (% ) value was determined to be 338. Preliminary toxicology studies revealed an acute LD50 of 202 mg/kg and a subacute LD50 of 150 mg/kg. Aeroplysinin-1 is neither a direct mutagen nor a premutagen in the umu/Salmonella typhimurium test system.
Glycogen synthase kinase-3 couples AKT-dependent signaling to the regulation of p21Cip1 degradation

Signaling via the phosphoinositide 3-kinase (PI3K)/AKT pathway is crucial for the regulation of endothelial cell (EC) proliferation and survival, which involves the AKT-dependent phosphorylation of the DNA repair protein p21(Cip1) at Thr-145. Because p21(Cip1) is a short-lived protein with a high proteasomal degradation rate, we investigated the regulation of p21(Cip1) protein levels by PI3K/AKT-dependent signaling. The PI3K inhibitors Ly294002 and wortmannin reduced p21(Cip1) protein abundance in human umbilical vein EC. However, mutation of the AKT site Thr-145 into aspartate (T145D) did not increase its protein half-life. We therefore investigated whether a kinase downstream of AKT regulates p21(Cip1) protein levels. In various cell types, AKT phosphorylates and inhibits glycogen synthase kinase-3 (GSK-3). Upon serum stimulation of EC, GSK-3beta was phosphorylated at Ser-9. Site-directed mutagenesis revealed that GSK-3 in vitro phosphorylated p21(Cip1) specifically at Thr-57 within the Cdk binding domain. Overexpression of GSK-3beta decreased p21(Cip1) protein levels in EC, whereas the specific inhibition of GSK-3 with lithium chloride interfered with p21(Cip1) degradation and increased p21(Cip1) protein about 10-fold in EC and cardiac myocytes (30 mm, p < 0.001). These data indicate that GSK-3 triggers p21(Cip1) degradation. In contrast, stimulation of AKT increases p21(Cip1) via inhibitory phosphorylation of GSK-3.
Das Protein p21 (Cip1/Waf1/Sdi1), ein Mitglied der Familie der Cyclin abhängigen Kinasen-Inhibitoren, ist ein wichtiger Modulator des Zellwachstums und der Reaktion auf DNA-Schädigung. Die Funktion von p21 hängt von der Stabilität des Proteins ab. p21 ist besonders stabil in der Phase G0/G1 des Zellzyklus. Phoshorylierungsvorgänge sowie Interaktionen mit anderen Proteinen spielen in der Stabilität von Proteinen eine wichtige Rolle. Ziel der vorliegenden Arbeit war, herauszufinden, ob die Phosphorylierung von p21 durch die Proteinkinase AKT oder die durch diese Phosphorylierung beeinflusste Interaktion mit dem Proliferating cell nuclear antigen, kurz PCNA, einen Einfluß auf die Stabilität von p21 hat. Mittels Proteinhalbwertszeitbestimmung konnte demonstriert werden, daß die Phosphorylierung am Threonin 145 durch AKT keinen signifikanten Einfluß auf die Stabilität von p21 aufwies. Durch Pulse chase und Westem-Blot Versuche konnte aber nachgewiesen werden, daß die Anwesenheit von PCNA das Protein p21 stabilisierte und die Degradation beeinflusste. Es konnte mittels p21 Mutanten, deren PCNA- Bindung durch Austausch der Aminosäure (M147) inhibiert ist, gezeigt werden, daß nur eine direkte Bindung von PCNA an p21 die Degradation beeinflussen konnte. Die Bestimmung der subzellulären Lokalisation von p21, die zur weiteren Abklärung der erhöhten p21-Stabilität durch PCNA diente, zeigte in Immunopräzipitationsversuchen nach subzellulärer Fraktionierung eine Interaktion von p21 mit PCNA vorwiegend im Zytoplasma. Dies ließ sich auch durch Immunofluoreszenzuntersuchungen bestätigen. Schließlich zeigten die Untersuchungen, daß die subzelluläre Lokalisation von der direkten Bindung an PCNA abhängig war. Zusammenfassend zeigte die Arbeit auf, daß die Stabilität von p21 durch seinen Bindungspartner PCNA beeinflusst werden konnte, und dies vermutlich durch subzelluläre Translokation erfolgt.
Pluto is a Monte-Carlo event generator designed for hadronic interactions from Pion production threshold to intermediate energies of a few GeV per nucleon, as well as for studies of heavy ion reactions. The package is entirely based on ROOT, without the need of additional packages, and uses the embedded C++ interpreter of ROOT to control the event production. The generation of events based on a single reaction chain and the storage of the resulting particle objects can be done with a few lines of a ROOT-macro. However, the complete control of the package can be taken over by the steering macro and user-defined models may be added without a recompilation of the framework. Multi-reaction cocktails can be facilitated as well using either mass-dependent or user-defined static branching ratios. The included physics uses resonance production with mass-dependent Breit-Wigner sampling. The calculation of partial and total widths for resonances producing unstable particles is performed recursively in a coupled-channel approach. Here, particular attention is paid to the electromagnetic decays, motivated by the physics program of HADES. The thermal model supports 2-component thermal distributions, longitudinal broadening, radial blast, direct and elliptic flow, and impact-parameter sampled multiplicities. The interface allows angular distribution models (e.g. for the primary meson emission) to be attached by the user as well as descriptions of multi-particle correlations using decay chain templates. The exchange of mass sampling or momentum generation models is also possible. The first feature allows for consistent coupled-channel calculations, needed for a correct description of hadronic interactions. For elementary reactions, angular distribution models for selected channels are already part of the framework, based on parameterizations of existing data. This report gives an overview of the design of the package, the included models and the user interface.
We have developed a versatile software package for the simulation of di-electron production in pp and dp collisions at moderate beam kinetic energies (1-2GeV). Particular attention has been paid to incorporate different descriptions of the Dalitz decay Δ rightarrow Ne + e - via a common interface. In addition, suitable parameterizations for the virtual bremsstrahlung process NN rightarrow NNe + e - based on one-boson exchange models have been implemented. Such simulation tools with high flexibility of the framework are important for the interpretation of the di-electron data taken with the HADES spectrometer and demonstrates the wide applicability within the field of nuclear and hadronic physics.
High impact events, political changes and new technologies are reflected in our language and lead to constant evolution of terms, expressions and names. Not knowing about names used in the past for referring to a named entity can severely decrease the performance of many computational linguistic algorithms. We propose NEER, an unsupervised method for named entity evolution recognition independent of external knowledge sources. We find time periods with high likelihood of evolution. By analyzing only these time periods using a sliding window co-occurrence method we capture evolving terms in the same context. We thus avoid comparing terms from widely different periods in time and overcome a severe limitation of existing methods for named entity evolution, as shown by the high recall of 90% on the New York Times corpus. We compare several relatedness measures for filtering to improve precision. Furthermore, using machine learning with minimal supervision improves precision to 94%.
We present measurements of exclusive ensuremathπ+,0 and η production in pp reactions at 1.25GeV and 2.2GeV beam kinetic energy in hadron and dielectron channels. In the case of π+ and π0 , high-statistics invariant-mass and angular distributions are obtained within the HADES acceptance as well as acceptance-corrected distributions, which are compared to a resonance model. The sensitivity of the data to the yield and production angular distribution of Δ (1232) and higher-lying baryon resonances is shown, and an improved parameterization is proposed. The extracted cross-sections are of special interest in the case of pp → pp η , since controversial data exist at 2.0GeV; we find \ensuremathσ=0.142±0.022 mb. Using the dielectron channels, the π0 and η Dalitz decay signals are reconstructed with yields fully consistent with the hadronic channels. The electron invariant masses and acceptance-corrected helicity angle distributions are found in good agreement with model predictions.
The knowledge of baryonic resonance properties and production cross sections plays an important role for the extraction and understanding of medium modifications of mesons in hot and/or dense nuclear matter. We present and discuss systematics on dielectron and strangeness production obtained with HADES on p+p, p+A and A+A collisions in the few GeV energy regime with respect to these resonances.
New results on the differential cross section in deuteron-proton elastic scattering are obtained at the deuteron kinetic energy of 2.5 GeV with the HADES spectrometer. The angular range of 69° – 125° in the center of mass system is covered. The obtained results are compared with the relativistic multiple scattering model calculation using the CD-Bonn deuteron wave function. The data at fixed scattering angles in the c.m. are in qualitative agreement with the constituent counting rules prediction.
The web and the social web play an increasingly important role as an information source for Members of Parliament and their assistants, journalists, political analysts and researchers. It provides important and crucial background information, like reactions to political events and comments made by the general public. The case study presented in this paper is driven by two European parliaments (the Greek and the Austrian parliament) and targets an effective exploration of political web archives. In this paper, we describe semantic technologies deployed to ease the exploration of the archived web and social web content and present evaluation results.
