The inclusive charged particle transverse momentum distribution is measured in proton–proton collisions at s=900 GeV at the LHC using the ALICE detector. The measurement is performed in the central pseudorapidity region (|η|<0.8) over the transverse momentum range 0.15<pT<10 GeV/c. The correlation between transverse momentum and particle multiplicity is also studied. Results are presented for inelastic (INEL) and non-single-diffractive (NSD) events. The average transverse momentum for |η|<0.8 is 〈pT〉INEL=0.483±0.001 (stat.)±0.007 (syst.) GeV/c and 〈pT〉NSD=0.489±0.001 (stat.)±0.007 (syst.) GeV/c, respectively. The data exhibit a slightly larger 〈pT〉 than measurements in wider pseudorapidity intervals. The results are compared to simulations with the Monte Carlo event generators PYTHIA and PHOJET.
The first measurement of two-pion Bose–Einstein correlations in central Pb–Pb collisions at √sNN=2.76 TeV at the Large Hadron Collider is presented. We observe a growing trend with energy now not only for the longitudinal and the outward but also for the sideward pion source radius. The pion homogeneity volume and the decoupling time are significantly larger than those measured at RHIC.
Inclusive transverse momentum spectra of primary charged particles in Pb–Pb collisions at √sNN=2.76 TeV have been measured by the ALICE Collaboration at the LHC. The data are presented for central and peripheral collisions, corresponding to 0–5% and 70–80% of the hadronic Pb–Pb cross section. The measured charged particle spectra in |η|<0.8 and 0.3<pT<20 GeV/c are compared to the expectation in pp collisions at the same sNN, scaled by the number of underlying nucleon–nucleon collisions. The comparison is expressed in terms of the nuclear modification factor RAA. The result indicates only weak medium effects (RAA≈0.7) in peripheral collisions. In central collisions, RAA reaches a minimum of about 0.14 at pT=6–7 GeV/c and increases significantly at larger pT. The measured suppression of high-pT particles is stronger than that observed at lower collision energies, indicating that a very dense medium is formed in central Pb–Pb collisions at the LHC.
Die vorliegende Dissertationsschrift befasst sich mit der molekulargenetischen Analyse zweier Basalganglienerkrankungen. Zum einen wurden Patienten mit M. Parkinson genetisch untersucht, zum anderen Patienten mit autosomal dominanter zervikaler Torsionsdystonie. Die Aufgabe bestand in der passenden Wahl der Methode zur jeweiligen humangenetischen Fragestellung. Der erste Teil handelte von der Suche der krankheitsverursachenden Mutation für die autosomal dominante zervikale Torsionsdystonie mit Spätmanifestation auf Chromosom 18p (Kandidatenlokus DYT7). Die erkrankte Familie deutscher Herkunft zeigt dystone Symptome mit Betonung auf kraniozervikale und brachiale Körperabschnitte und ist somit die weltweit einzige bekannte Familie mit Vererbung dieser ansonsten sporadisch auftretenden Erkrankung. Die PCR-Sequenzierung der Kandidatengene ZFP161, LOC390828, NDUFV2 und PTPRM auf dem DYT7 Lokus erbrachte bei den sieben erkrankten Familienmitgliedern im Vergleich zu nicht verwandten Kontrollen (Ehepartner und 96 Kontrollen der Blutbank) keinen Aminosäureaustausch, der ausschließlich bei den erkrankten Probanden zu finden war. Technisch konzentrierte sich diese Untersuchung auf die Amplifizierung und anschließende Sequenzierung jedes einzelnen Exons in den zu untersuchenden Proben, und die Bestätigung einer putativen Mutation mittels Verdau der PCR-Produkte durch Restriktionsendonukleasen. Die Auswahl der Kandidatengene erfolgte aufgrund der Annahme pathobiochemischer Mechanismen, die durch andere Formen der vererbten Torsionsdystonie oder zellbiologische Experimente als krankheitsverursachend gelten. Auch wenn keine Mutation gefunden wurde, so konnten bereits bekannte und neue single nucleotide polymorphisms (SNP) etabliert werden. Die zweite Thematik befasste sich mit der Frage, ob das bereits bekannte Parkinson-Gen UCH-L1 auf dem PARK5 Lokus krankheitsverursachend für den autosomal dominanten M. Parkinson in einer spanischen Familie ist. Diese parametrische Kopplungsanalyse wurde mithilfe der heißen Polyacrylamid-Gelelektrophorese (PAGE) durchgeführt. Dabei konnte in allen Patienten und den Verwandten ersten Grades über Analyse der Mikrosatelliten nördlich und südlich der Kandidatenregion (UCH-L1) in einem Bereich sehr niedriger Rekombinationswahrscheinlichkeit eine Haplotypisierung erfolgen. Die Haplotypisierung zeigte nicht die erforderliche Identifizierung eines Krankheitsallels bei allen betroffenen Probanden. Somit ist hier neben der einzig bekannten deutschen PARK5 Familie keine weitere Familie mit UCH-L1 Mutation bestätigt worden. Dementsprechend ist die Ätiologie dieser Erkrankung in dieser Familie noch unklar, was aber der Bedeutung des Ubiquitin-Proteasom Systems in der Parkinson-Entität keinen Abbruch getan hat. Da alle anderen autosomal dominanten Parkinson-Loci ausgeschlossen sind, muss sich die Ursache für den M. Parkinson in dieser Familie in einem heute noch unbekannten Gen befinden. Weitere Untersuchungen im Rahmen eines Genomscans sind aufgrund der geringen Fallzahl nicht möglich. Die letzte Aufgabe dieser Arbeit bestand in der Durchführung einer Assoziationsstudie mit den putativen PINK1 (PARK6) Interaktoren NME4 und MTIF3 für den mehrheitlich sporadisch auftretenden M. Parkinson. Dabei wurden in zwei unabhängigen Studiengruppen mit insgesamt 453 sporadischen Parkinsonpatienten und 370 Kontrollen jeweils zwei SNPs auf gekoppelte Vererbung mit der Erkrankung untersucht. Der Unterschied zwischen den Testgruppen bestand im Studiendesign, da zum einen mit den Patienten nicht verwandte Kontrollen und zum anderen verwandte Kontrollen verwendet wurden. Die mit beiden Studientypen normalerweise auftretenden Probleme durch Stratifikation bzw. erniedrigte statistische Power konnten durch Kombination der Studien ausgeglichen werden. Das Methodenspektrum umfasste PCR und Restriktionsverdau, was zum Auffinden eines Kopplungsungleichgewichts für das Gen MTIF3 führte. Ein heterozygoter Basenaustausch für den Polymorphismus rs7669 erhöht signifikant das Relative Risiko an M. Parkinson zu erkranken, wohingegen der homozygote Basenaustausch das Krankheitsrisiko des Trägers signifikant erniedrigt. Bezüglich des Relativen Risikos wurde der Effekt der molekularen Heterosis nachgewiesen. Bei diesem mitochondrial lokalisierten Gen handelt es sich um einen Initiator der mitochondrialen Translation. Demzufolge besteht hier Einfluss auf die Homöostase und somit Funktionalität der Atmungskettenkomplexe, die als bedeutend für die Pathogenese des M. Parkinson angesehen werden. Die Verbindung zum mitochondrial lokalisierten PINK1 besteht aufgrund seiner Kinase-Aktivität in der An- und Abschaltung des mitochondrialen Translations - Initiationsfaktors. Aber auch die Wichtigkeit von NME4 konnte in dieser Studie trotz fehlender Assoziation nicht ausgeschlossen werden, da vorangehende experimentelle Ergebnisse dieses Protein bereits in den PINK1 Signalweg zuordnen konnten. MTIF3 könnte wohlmöglich ein wichtiger genetische Risikofaktor für den idiopathischen M. Parkinson sein. Es bleibt abzuwarten, ob zukünftige genetische und zellbiologische Experimente die Wichtigkeit, die in diesem Protein zu liegen scheint, bestätigen können.
Um Dynamiken und Netzwerke in den untersuchten Clubszenen und urbanen Räumen Berlins wissenschaftlich erfassen zu können, bedarf es einer multidimensionalen Herangehensweise. Ethnografi sche Methoden sind dabei von größter Bedeutung: teilnehmende Beobachtung im Alltag sowie auf Veranstaltungen vor Ort, fokussierte Gespräche mit Szenegängern und reflektierende Berichte über die eigene Positionierung als Forscherin. Das gesammelte Datenmaterial ist vielfältig: detaillierte Feldnotizen nach jedem Club- oder Barbesuch, aber auch nach relevanten Gesprächen, sei es »face-to-face«, über E-Mail- oder Chatverkehr nehmen den größten Part ein. Bildmaterial wie Plakate, Flyer und selbst gemachte Fotos in Clubs ergänzen das Schriftmaterial auch visuell. Das Verfolgen stadt- und kulturpolitischer Entwicklungen, Beobachtungen sozioökonomischer Gegebenheiten in den jeweiligen Szenen und der Einfl uss urbaner Infrastruktur auf das Ausgehverhalten sind weitere Mosaiksteine, die dazu beitragen, ein möglichst ganzheitliches Bild entstehen zu lassen. Nach der Feldforschungs- und Datenerhebungsphase wird das gesamte Datenmaterial ab Januar 2011 einer qualitativen Analyse unterzogen.
We analytically show that a common across rich/poor individuals Stone-Geary utility function with subsistence consumption in the context of a simple two-asset portfolio-choice model is capable of qualitatively explaining: (i) the higher saving rates of the rich, (ii) the higher fraction of personal wealth held in stocks by the rich, and (iii) the higher volatility of consumption of the wealthier. On the contrary, time-variant "keeping-up with the Joneses" weighted average consumption playing the role of moving benchmark subsistence consumption gives the same portfolio composition and saving rates across the rich and the poor, failing to reconcile the model with what micro data say.
Floodplains play an important role in the terrestrial water cycle and are very important for biodiversity. Therefore, an improved representation of the dynamics of floodplain water flows and storage in global hydrological and land surface models is required. To support model validation, we combined monthly time series of satellite-derived inundation areas (Papa et al., 2010) with data on irrigated rice areas (Portmann et al., 2010). In this way, we obtained global-scale time series of naturally inundated areas (NIA), with monthly values of inundation extent during 1993–2004 and a spatial resolution of 0.5°. For most grid cells (0.5°×0.5°), the mean annual maximum of NIA agrees well with the static open water extent of the Global Lakes and Wetlands database (GLWD) (Lehner and Döll, 2004), but in 16% of the cells NIA is larger than GLWD. In some regions, like Northwestern Europe, NIA clearly overestimates inundated areas, probably because of confounding very wet soils with inundated areas. In other areas, such as South Asia, it is likely that NIA can help to enhance GLWD. NIA data will be very useful for developing and validating a floodplain modeling algorithm for the global hydrological model WGHM. For example, we found that monthly NIAs correlate with observed river discharges.
From the 1980 Maitatsine uprising to the 2009 Boko Haram uprising, Nigeria was bedevilled by ethno-religious conflicts with devastating human and material losses. But the Boko Haram uprising of July 2009 was significant in that it not only set a precedent, but also reinforced the attempts by Islamic conservative elements at imposing a variant of Islamic religious ideology on a secular state. Whereas the religious sensitivity of Nigerians provided fertile ground for the breeding of the Boko Haram sect, the sect’s blossoming was also aided by the prevailing economic dislocation in Nigerian society, the advent of party politics (and the associated desperation of politicians for political power), and the ambivalence of some vocal Islamic leaders, who, though they did not actively embark on insurrection, either did nothing to stop it from fomenting, or only feebly condemned it. These internal factors coupled with growing Islamic fundamentalism around the world make a highly volatile Nigerian society prone to violence, as evidenced by the Boko Haram uprising. Given the approach of the Nigerian state to religious conflict, this violence may remain a recurring problem. This paper documents and analyses the Boko Haram uprising, as well as its links with the promotion of Islamic revivalism and the challenges it poses to the secularity of the Nigerian state.
Variants resistant to compounds specifically targeting HCV are observed in clinical trials. A multi-variant viral dynamic model was developed to quantify the evolution and in vivo fitness of variants in subjects dosed with monotherapy of an HCV protease inhibitor, telaprevir. Variant fitness was estimated using a model in which variants were selected by competition for shared limited replication space. Fitness was represented in the absence of telaprevir by different variant production rate constants and in the presence of telaprevir by additional antiviral blockage by telaprevir. Model parameters, including rate constants for viral production, clearance, and effective telaprevir concentration, were estimated from 1) plasma HCV RNA levels of subjects before, during, and after dosing, 2) post-dosing prevalence of plasma variants from subjects, and 3) sensitivity of variants to telaprevir in the HCV replicon. The model provided a good fit to plasma HCV RNA levels observed both during and after telaprevir dosing, as well as to variant prevalence observed after telaprevir dosing. After an initial sharp decline in HCV RNA levels during dosing with telaprevir, HCV RNA levels increased in some subjects. The model predicted this increase to be caused by pre-existing variants with sufficient fitness to expand once available replication space increased due to rapid clearance of wild-type (WT) virus. The average replicative fitness estimates in the absence of telaprevir ranged from 1% to 68% of WT fitness. Compared to the relative fitness method, the in vivo estimates from the viral dynamic model corresponded more closely to in vitro replicon data, as well as to qualitative behaviors observed in both on-dosing and long-term post-dosing clinical data. The modeling fitness estimates were robust in sensitivity analyses in which the restoration dynamics of replication space and assumptions of HCV mutation rates were varied.
