Since 2007 the concept of open online courses came up leading to many discussions of this new format in blog posts and articles especially in the US and Canada. 2011, the first German open online course was started addressing the Future of Learning.
The article discusses the concept of open online courses, the experiences with the first German course, and gives some perspectives on further developments which partly were implemented in a new course that was just started in 2012.
The pointed commentary published on Verfassungsblog over the last week—coming from different perspectives and informed from different experiences—shows the potential of such debates. In the case of Greece, they are an important addition to a discourse focusing too much on austerity or debt sustainability.
An interior delta in the lower course of the Ntem River near the sub-prefecture Ma’an was identified after interpretation of satellite images, topographical maps of SW Cameroon and geological as well as hydrological references and a reconnaissance fieldtrip to the study area. Here neotectonic processes have initiated the establishment of a ‘sediment trap’ (step fault), which in combination with environmental changes strongly generated the fluvial morphology. It transitionally led to temporary lacustrine and palustrine conditions in parts of this river section. Inside the interior delta an anastomosing multi-branched river system has developed, which contains ‘stillwater locations', periodically inundated sections, islands and rapids. Following geomorphological, physiogeographical and sedimentological research approaches, the alluvial plain has been prospected and studied extensively. 91 hand-corings, including three NE–SW transects, were carried out on river benches, levees, cut-off and periodical branches, islands as well as terraces throughout the entire alluvial plain and have unveiled multi-layered, sandy to clayey alluvia reaching up to 440 cm depth. At many locations, fossil organic horizons and palaeosurfaces were discovered, containing valuable palaeoenvironmental proxy data. At these sites, through additional detailed stratigraphical analysis (close-meshed hand-coring and exposure digging) a comprehensive insight into the stratification (lamination) of the alluvia could be gained, clarifying processes and conditions that prevailed in the catchment area during the period of their deposition. 32 Radiocarbon data of macro-rests (leafs, wood), charcoal and organic sediment sampled from these horizons provided ages between 48.230 ± 6.411 and 217 ± 46 years BP (not calibrated). This constitutes the importance of the alluvia as an additional, innovative palaeoarchive for proxy data contributing to the reconstruction of palaeoenvironment and palaeoclimate in western Equatorial Africa. The further examination of the alluvia will not only provide additional information on the dynamics of vegetation, climate and hydrology (esp. fluvial morphology) in SW Cameroon since the ‘First Millennium BC Crisis’ (around 3.000 years BP), the main focus of the DFG-research project, but also on conditions prevailing since the Late Pleistocene, during the Last Glacial Maximum (~18.000 years BP), the Younger Dryas impact (~11.000 years BP) and the ‘Humid African Period’ (~9.000–6.000 years BP). Delta13C-values (–31,4 to –26,4‰) evidence that at the particular drilling sites rain forest has prevailed during the corresponding time period (rain forest refuge theory). The sampled macrorests all indicate rain forest dominated ecosystems, which were able to persist in fluvial habitats, even during arid periods.
The article presents an interdisciplinary project that is focussed on an important cultural monument, namely a fortified hilltop settlement in the cadastre of Hradiště near Pilsen. This fortification is attested by five phases, which correspond to the main epochs of the erection of west Bohemian ramparts during the time span from the turn of the early to the Middle Bronze Age until the early medieval period. Geophysical prospection and trial excavations conducted there in 2012 and 2013 revealed important and new information on the stratigraphy and natural environment of the site. The project is particularly directed towards collaboration between archaeology and scientific studies. Geophysical and geochemical aspects of archaeological contexts, archaeobotanical and archaeozoological finds as well as geological composition of the materials from the fortification were analysed. Special attention was given to the problematic of the so-called vitrified walls (Schlackenwälle) associated with this site.
The sponge genus Latrunculia is a prolific source of discorhabdin type pyrroloiminoquinone alkaloids. In the continuation of our research interest into this genus, we studied the Antarctic deep-sea sponge Latrunculia biformis that showed potent in vitro anticancer activity. A targeted isolation process guided by bioactivity and molecular networking-based metabolomics yielded three known discorhabdins, (−)-discorhabdin L (1), (+)-discorhabdin A (2), (+)-discorhabdin Q (3), and three new discorhabdin analogs (−)-2-bromo-discorhabdin D (4), (−)-1-acetyl-discorhabdin L (5), and (+)-1-octacosatrienoyl-discorhabdin L (6) from the MeOH-soluble portion of the organic extract. The chemical structures of 1–6 were elucidated by extensive NMR, HR-ESIMS, FT-IR, [α]D, and ECD (Electronic Circular Dichroism) spectroscopy analyses. Compounds 1, 5, and 6 showed promising anticancer activity with IC50 values of 0.94, 2.71, and 34.0 µM, respectively. Compounds 1–6 and the enantiomer of 1 ((+)-discorhabdin L, 1e) were docked to the active sites of two anticancer targets, topoisomerase I-II and indoleamine 2,3-dioxygenase (IDO1), to reveal, for the first time, the binding potential of discorhabdins to these proteins. Compounds 5 and 6 are the first discorhabdin analogs with an ester function at C-1 and 6 is the first discorhabdin bearing a long-chain fatty acid at this position. This study confirms Latrunculia sponges to be excellent sources of chemically diverse discorhabdin alkaloids.
In this manuscript a comprehensive coverage of recent developments in the drug therapy of vasospasm while providing the background information that neuroscientists need to understand its rationale. The range of new agents available for treatment of cerebral vasospasm is expanding rapidly along with rapid advances in pharmacology and physiology that are uncovering the mechanisms of this disease. Although there are many publications for treatment of cerebral vasospasm, most are focusing on different aspects of vasospasm treatment and many have limited value due to insufficient quality. Moreover, the complexity of this, in many cases deleterious condition, is enormous and the information needed to understand drug effects is accordingly often not readily available in a single source. A number of pharmacological and medical therapies are currently in use or being investigated in an attempt to reverse cerebral vasospasm, but only a few have proven to be useful. Current research efforts promise the eventual production of new medical therapies. At last, recommendations for the use of different treatment stages based on currently available clinical data are provided.
The most versatile tool for visualizing endogenous RNA is molecular beacons (MBs). MBs are modified oligonucleotides that consist of a stem-loop structure equipped with a fluorophore and a quencher at the opposite ends. They only give a fluorescent signal when hybridized to the target RNA. Here we present our recent efforts to enhance the spatiotemporal resolution of RNA visualization by refining MBs.
We first asked if we could refine MBs to visualize defined subcellular populations of RNA in living neurons. To achieve this, we utilize visible light-activatable Q-dye MBs to allow only a subcellular fraction to be activated. Here, the fluorophore at the 5’-end was linked to a second quencher via a photolabile coumarin protecting group. Therefore, the MB only gives a fluorescent signal, when activated with visible light and hybridized to the target. This architecture allowed local activation of a hybridized subpopulation in a defined area of the cell. Knowing the exact origin of the activated RNA, we were able to increase the available monitoring time for neuronal mRNA from several minutes (literature known MBs) to more than 14 hours.
We next asked if it would be possible to gain spatiotemporal control over where the MB hybridization events occur. Therefore, we developed photo-tethered MBs where two phosphates in the loop backbone are covalently linked to each other via two photocages. This prevents the MB from hybridization to the target RNA. Only when light is applied, the photo-tethers are cleaved, and the inherent hybridization function of the MB is activated. This architecture allowed us to control the hybridization of photo-tethered MBs in primary cultured neurons.
