The development of binocular vision is an active learning process comprising the development of disparity tuned neurons in visual cortex and the establishment of precise vergence control of the eyes. We present a computational model for the learning and self-calibration of active binocular vision based on the Active Efficient Coding framework, an extension of classic efficient coding ideas to active perception. Under normal rearing conditions, the model develops disparity tuned neurons and precise vergence control, allowing it to correctly interpret random dot stereogramms. Under altered rearing conditions modeled after neurophysiological experiments, the model qualitatively reproduces key experimental findings on changes in binocularity and disparity tuning. Furthermore, the model makes testable predictions regarding how altered rearing conditions impede the learning of precise vergence control. Finally, the model predicts a surprising new effect that impaired vergence control affects the statistics of orientation tuning in visual cortical neurons.

We present a model for the autonomous learning of active binocular vision using a recently developed biome-chanical model of the human oculomotor system. The model is formulated in the Active Efficient Coding (AEC) framework, a recent generalization of classic efficient coding theories to active perception. The model simultaneously learns how to efficiently encode binocular images and how to generate accurate vergence eye movements that facilitate efficient encoding of the visual input. In order to resolve the redundancy problem arising from the actuation of the eyes through antagonistic muscle pairs, we consider the metabolic costs associated with eye movements. We show that the model successfully learns to trade off vergence accuracy against the associated metabolic costs, producing high fidelity vergence eye movements obeying Sherrington’s law of reciprocal innervation.
In the development of photolabile protecting groups, it is of high interest to selectively modify photochemical properties with structural changes as simple as possible. In this work, knowledge of fluorophore optimization was adopted and used to design new coumarin- based photocages. Photolysis efficiency was selectively modulated by inactivating competitive decay channels, such as twisted intramolecular charge transfer (TICT) or hydrogen-bonding, and the photolytic release of the neurotransmitter serotonin was demonstrated. Structural modifications inspired by the fluorophore ATTO 390 led to a significant increase in the uncaging cross section that can be further improved by the simple addition of a double bond. Ultrafast transient absorption spectroscopy gave insights into the underlying solvent-dependent photophysical dynamics. The chromophores presented here are excellently suited as new photocages in the visible wavelength range due to their simple synthesis and their superior photochemical properties.
The most versatile tool for visualizing endogenous RNA is molecular beacons (MBs). MBs are modified oligonucleotides that consist of a stem-loop structure equipped with a fluorophore and a quencher at the opposite ends. They only give a fluorescent signal when hybridized to the target RNA. Here we present our recent efforts to enhance the spatiotemporal resolution of RNA visualization by refining MBs.
We first asked if we could refine MBs to visualize defined subcellular populations of RNA in living neurons. To achieve this, we utilize visible light-activatable Q-dye MBs to allow only a subcellular fraction to be activated. Here, the fluorophore at the 5’-end was linked to a second quencher via a photolabile coumarin protecting group. Therefore, the MB only gives a fluorescent signal, when activated with visible light and hybridized to the target. This architecture allowed local activation of a hybridized subpopulation in a defined area of the cell. Knowing the exact origin of the activated RNA, we were able to increase the available monitoring time for neuronal mRNA from several minutes (literature known MBs) to more than 14 hours.
We next asked if it would be possible to gain spatiotemporal control over where the MB hybridization events occur. Therefore, we developed photo-tethered MBs where two phosphates in the loop backbone are covalently linked to each other via two photocages. This prevents the MB from hybridization to the target RNA. Only when light is applied, the photo-tethers are cleaved, and the inherent hybridization function of the MB is activated. This architecture allowed us to control the hybridization of photo-tethered MBs in primary cultured neurons.
Background: Peanuts are a member of the legume family (botanical family Leguminosae) and peanut allergies are the most common cause of food anaphylaxis in many countries. The prevalence of peanut allergy is increasing.
Methods: Experts from Germany and Austria performed a standardized literature search and published their consensus recommendations in a White Paper on Peanut Allergy, which this care pathway is based upon, thus, providing a comprehensive diagnosis and treatment algorithm.
Results: The most important diagnostic key elements include a detailed clinical medical history, evidence of peanut-specific sensitization by means of skin prick testing and/or in vitro determination of the peanut (extract)-specific IgE and/or the molecular component diagnostics (most important Ara h 2-specific IgE, sometimes also Ara h1-, 3-, 6-, 8- and 9-specific IgE) as well as the gold standard, the double-blind, placebo-controlled food challenge. The diagnostic algorithms were created for the following constellations: Suspected primary peanut allergy with a clear history of systemic immediate-type reaction, suspected primary peanut allergy with questionable symptoms, suspected secondary (possibly pollen-associated) peanut allergy with a history of solely oropharyngeal symptoms and incidental finding of sensitization and no peanut ingestion so far.
Conclusions: After established diagnosis the standard of care is counseling to avoid peanut contact and prescription of emergency medications (oral antihistamines, oral steroids, inhaled β2-agonists, injectable intramuscular epinephrine) as needed. Instruction on the use of these emergency medications should be provided. A preparation for oral immunotherapy (OIT) for 4 to 17 years old peanut allergic children/ adolescents has been recently approved by the regulatory authorities. OIT for peanut allergy shows high efficacy and an acceptable safety profile, improves quality of life, and health economic aspects. Thus it offers a therapeutic option for peanut allergic children and adolescents.
Chaperone-assisted selective autophagy (CASA) initiated by the cochaperone Bcl2-associated athanogene 3 (BAG3) represents an important mechanism for the disposal of misfolded and damaged proteins in mammalian cells. Under mechanical stress, the cochaperone cooperates with the small heat shock protein HSPB8 and the cytoskeleton-associated protein SYNPO2 to degrade force-unfolded forms of the actin-crosslinking protein filamin. This is essential for muscle maintenance in flies, fish, mice and men. Here, we identify the serine/threonine protein kinase 38 (STK38), which is part of the Hippo signaling network, as a novel interactor of BAG3. STK38 was previously shown to facilitate cytoskeleton assembly and to promote mitophagy as well as starvation and detachment induced autophagy. Significantly, our study reveals that STK38 exerts an inhibitory activity on BAG3-mediated autophagy. Inhibition relies on a disruption of the functional interplay of BAG3 with HSPB8 and SYNPO2 upon binding of STK38 to the cochaperone. Of note, STK38 attenuates CASA independently of its kinase activity, whereas previously established regulatory functions of STK38 involve target phosphorylation. The ability to exert different modes of regulation on central protein homeostasis (proteostasis) machineries apparently allows STK38 to coordinate the execution of diverse macroautophagy pathways and to balance cytoskeleton assembly and degradation.
Die vorliegende Arbeit liefert ein Versatzstück zu grundlagentheoretischen Überlegungen im Themenfeld Gesundheit. Dabei wird der Fokus insbesondere auf wissenschaftstheoretische, -soziologische und sozialpsychologische Reflektionen der gesundheitsbezogenen Diskurslandschaft im wissenschaftlichen Bereich gelegt. Diese werden nach gesellschaftsdiagnostischen Anteilen der Arbeit in reflektierter Art und Weise in einen diskursiven Vorschlag überführt, der wiederum für Gesundheit im universitären Setting wegweisend sein kann.
