Recent research has identified significant correlations between traumatic events and depression in refugees. However, few studies have addressed the role of acculturation strategies in this relationship. This study explored the relationship between cultural orientation, traumatic events and depression in female refugees from Syria, Afghanistan, Eritrea, Iran, Iraq, and Somalia living in Germany. We expected acculturation strategies to moderate the effect of traumatic experiences on depression. The sample included 98 female refugees in Germany. The depression scale of the Hopkins Symptom Checklist (HSCL) represented the dependent measure. The trauma checklists derived from the Post-traumatic Diagnostic Scale (PDS) and the Harvard Trauma Questionnaire (HTQ) as well as the Frankfurt Acculturation Scale (FRACC) were used as independent measures for traumatic events and orientation toward the host culture as well as orientation toward the culture of origin, respectively. A moderation analysis was conducted to examine whether the relationship between the number of traumatic events and depression was influenced by the women’s orientation toward the culture of origin and the host culture. We identified a significant model explaining 26.85% of the variance in depressive symptoms (Cohen’s f2 = 0.37). The number of traumatic events and the orientation toward the host culture exerted significant effects on depressive symptoms. The moderating effect was not significant, indicating that the effect of the number of traumatic events was not influenced by cultural orientation. Based on our results, orientation toward the host culture as well as traumatic experiences exert independent effects on depressive symptoms in refugees.
Non-alcoholic fatty liver disease (NAFLD) is gaining in importance and is linked to obesity. Especially,thedevelopmentoffibrosisandportalhypertensioninNAFLDpatientsrequirestreatment. Transgenic TGR(mREN2)27 rats overexpressing mouse renin spontaneously develop NAFLD with portal hypertension but without obesity. This study investigated the additional role of obesity in this model on the development of portal hypertension and fibrosis. Obesity was induced in twelve-week old TGR(mREN2)27 rats after receiving Western diet (WD) for two or four weeks. Liver fibrosis was assessed using standard techniques. Hepatic expression of transforming growth factor-β1 (TGF-β1), collagen type Iα1, α-smooth muscle actin, and the macrophage markers Emr1, as well as the chemoattractant Ccl2, interleukin-1β (IL1β) and tumor necrosis factor-α (TNFα) were analyzed. Assessment of portal and systemic hemodynamics was performed using the colored microsphere technique. Asexpected,WDinducedobesityandliverfibrosisasconfirmedbySiriusRedandOilRed O staining. The expression of the monocyte-macrophage markers, Emr1, Ccl2, IL1β and TNFα were increasedduringfeedingofWD,indicatinginfiltrationofmacrophagesintotheliver,eventhoughthis increase was statistically not significant for the EGF module-containing mucin-like receptor (Emr1) mRNA expression levels. Of note, portal pressure increased with the duration of WD compared to animals that received a normal chow. Besides obesity, WD feeding increased systemic vascular resistance reflecting systemic endothelial and splanchnic vascular dysfunction. We conclude that transgenic TGR(mREN2)27 rats are a suitable model to investigate NAFLD development with liver fibrosis and portal hypertension. Tendency towards elevated expression of Emr1 is associated with macrophage activity point to a significant role of macrophages in NAFLD pathogenesis, probably due to a shift of the renin–angiotensin system towards a higher activation of the classical pathway. The hepatic injury induced by WD in TGR(mREN2)27 rats is suitable to evaluate different stages of fibrosis and portal hypertension in NAFLD with obesity.
Background: The purpose of this pilot study was to create a valid and reliable set of assessment questions for examining Evidence-based Dentistry (EbD) knowledge. For this reason, we adapted and validated for dental students the Berlin Questionnaire (BQ), which assesses Evidence-based Medicine (EbM) abilities.
Methods: The Berlin Questionnaire was validated with medical residents. We adapted it for use in a dentistry setting. An expert panel reviewed the adapted BQ for content validity. A cross-sectional cohort representing four training levels (EbD-novice dental students, EbD-trained dental students, dentists, and EbM−/EbD-expert faculty) completed the questionnaire. A total of 140 participants comprised the validation set. Internal reliability, item difficulty and item discrimination were assessed. Construct validity was assessed by comparing the mean total scores of students to faculty and comparing proportions of students and faculty who passed each item.
Results: Among the 133 participants (52 EbD-novice dental students, 53 EbD-trained dental students, 12 dentists, and 16 EbM-/ EbD-expert faculty), a statistically significant (p < 0.001) difference was evident in the total score corresponding to the training level. The total score reliability and psychometric properties of items modified for discipline-specific content were acceptable. Cronbach’s alpha was 0.648.
Conclusion: The adapted Berlin Questionnaire is a reliable and valid instrument to assess competence in Evidence-based Dentistry in dental students. Future research will focus on refining the instrument further.
Receptor tyrosine kinases (RTKs) orchestrate cell motility and differentiation. Deregulated RTKs may promote cancer and are prime targets for specific inhibitors. Increasing evidence indicates that resistance to inhibitor treatment involves receptor cross-interactions circumventing inhibition of one RTK by activating alternative signaling pathways. Here, we used single-molecule super-resolution microscopy to simultaneously visualize single MET and epidermal growth factor receptor (EGFR) clusters in two cancer cell lines, HeLa and BT-20, in fixed and living cells. We found heteromeric receptor clusters of EGFR and MET in both cell types, promoted by ligand activation. Single-protein tracking experiments in living cells revealed that both MET and EGFR respond to their cognate as well as non-cognate ligands by slower diffusion. In summary, for the first time, we present static as well as dynamic evidence of the presence of heteromeric clusters of MET and EGFR on the cell membrane that correlates with the relative surface expression levels of the two receptors
Direct acting antivirals (DAAs) revolutionized the therapy of chronic hepatitis C infection. However, unexpected high recurrence rates of hepatocellular carcinoma (HCC) after DAA treatment became an issue in patients with advanced cirrhosis and fibrosis. In this study, we aimed to investigate an impact of DAA treatment on the molecular changes related to HCC development and progression in hepatoma cell lines and primary human hepatocytes. We found that treatment with sofosbuvir (SOF), a backbone of DAA therapy, caused an increase in EGFR expression and phosphorylation. As a result, enhanced translocation of EGFR into the nucleus and transactivation of factors associated with cell cycle progression, B-MYB and Cyclin D1, was detected. Serine/threonine kinase profiling identified additional pathways, especially the MAPK pathway, also activated during SOF treatment. Importantly, the blocking of EGFR kinase activity by erlotinib during SOF treatment prevented all downstream events. Altogether, our findings suggest that SOF may have an impact on pathological processes in the liver via the induction of EGFR signaling. Notably, zidovudine, another nucleoside analogue, exerted a similar cell phenotype, suggesting that the observed effects may be induced by additional members of this drug class.
Background: Point of care devices for performing targeted coagulation substitution in bleeding patients have become increasingly important in recent years. New on the market is the Quantra® from HemoSonics (LC, Charlottesville, VA, US). It uses sonorheometry, a sonic estimation of elasticity via resonance (SEER), a novel ultrasound-based technology that measures viscoelastic properties of whole blood. Several studies have already shown the comparability with devices already established on the market such as the ROTEM® (TEM International GmbH, Munich, Germany).
Objective: In contrast to existing studies, the planned study will be the first prospective interventional study using the new Quantra® system in a cardiac surgical patient cohort. The aim is to investigate the non-inferiority between an already existing coagulation algorithm, based on ROTEM®/Multiplate®, and a new algorithm based on the Quantra®, for the treatment of coagulopathic cardiac surgical patients.
Methods: The study is divided into two phases. In an initial observation phase, whole blood samples of 20 patients will be analyzed using both ROTEM®/Multiplate® and Quantra® obtained at three defined points of time (prior to surgery, after completion of cardiopulmonary bypass, on arrival in the intensive care unit). The obtained threshold values will be used to create an algorithm for hemotherapy. In a second intervention phase, the new algorithm will be tested against an algorithm used routineously for years at our department for non-inferiority.
Results: The main objective of the examination is the cumulative loss of blood within 24 hours after surgery. Statistical calculations based on literature and in-house data suggest that the new algorithm is not inferior if the difference in cumulative blood loss is < 150ml/24 h.
Conclusions: Because of the comparability of the Quantra® sonorheometry system with ROTEM® rotational thromboelastometric measurement methods, the existing hemotherapy treatment algorithm can be adapted to the Quantra device with a proof of non-inferiority. Mutations in blood stem cells do not necessarily have to result in leukaemia. It was only recently discovered that clones of mutated blood cells can be identified in many healthy people in old age. Nonetheless, clonal haematopoiesis, as scientists baptised this finding, is far from innocent. It is a formidable risk factor for cardiovascular diseases – on par with smoking, excess weight or high blood pressure. Why this is, is still a riddle to be solved.

Professor Wild, one of your main areas of activity is the prevention of cardiovascular diseases. Since 2010/2011, they have accounted for approximately half of all deaths worldwide.
Cardiovascular diseases are in fact frequently multi-causal. In addition to risk factors such as high blood pressure, diabetes, and nicotine abuse, genetic predispositions, family and social influences and environmental factors also play a role – as do mental illnesses and personality. A single factor normally only has a very small influence, but several ones can lead to illness through accumulation.
Since the risk factors are known, targeted prevention is possible. Unfortunately, health insurance companies often do not offer comprehensive prevention as part of standard service...
Heart and vessels form a highly complex organ system in which extremely diverse cells have to work together correctly to provide all organs with blood. In past decades, heart biology placed its focus on whole tissues or cell isolates. Now, however, new technologies allow the tracing of a diversity of cell types and their individual responses to signals down to the level of proteins and genes. Researchers hope this will help them better support the regeneration of diseased hearts.

The healthcare industry sees great potential in consolidating medical data on one and the same person which is currently distributed across various practices and hospitals. This would not only save costs on repeating procedures; physicians also hope that artificial intelligence will help them identify new connections and thereby treat diseases earlier, or even prevent them.
So far, personal feedback in the case of lectures with hundreds of students still seems utopic – even after the digitalization boom in times of the coronavirus. Tools from the research field of »learning analytics« could in future give students feedback and at the same time provide their supervisors with clues about where help is still needed.
A question of striking the right balance : how do digital media influence how we think and act?

What influence do digital technologies have on human perception, thinking and action? Do computer games harm the development of young brains? And is there really such a thing as »digital dementia«, an increasing forgetfulness caused by the use of modern technologies? For some of these questions, answers are available that are empirically corroborated.
Through digitalization, the social importance of copyright law has grown considerably. Moreover, the culture of exclusivity established by copyright law conflicts fundamentally with the culture of access prevalent on the internet. An example for this is the dispute over the EU’s latest copyright directive. Does it ring in the end of the internet as we know it, or does it »only« see to fair remuneration for those working in the creative economy?
The issue of data security has become increasingly complex in the age of the internet and artificial intelligence. The developments seem to be almost unmanageable in some areas. Cooperation between jurisprudence and information technology is the only thing that can protect the individual and certain social groups from discrimination.
As a network researcher, sociologist Professor Christian Stegbauer also deals with communication in social media. That people prefer to stay in a bubble with like-minded others rather than get to grips with different opinions and ways of thinking was in his view inherent to digital communication from the outset. He considers many of the utopian ideas of a digital culture of participation to be exaggerated.

Was Daten angeht, setzen Marketingforschung und -lehre bisher vor allem auf Masse statt Klasse. Doch um fundierte Entscheidungen zu treffen, brauchen Unternehmen hoch entwickelte statistische Modelle. Das ist das Forschungsgebiet von Prof. Thomas Otter, der auch das gute alte Bauchgefühl in Algorithmen umzusetzen vermag.
Große Tasten, ein übersichtliches Display – die Hersteller von Mobiltelefonen haben sich geirrt, als sie spezielle Geräte für ältere Menschen entwickelten: Die Angebote sind gründlich gefloppt. Doch aus gerontologischer und erziehungswissenschaftlicher Sicht lohnt es sich durchaus, die besonderen Bedürfnisse von Seniorinnen und Senioren näher zu untersuchen. Die Erziehungswissenschaften sind eine Schlüsseldisziplin für die Zukunft unserer Gesellschaft; der Umgang der Menschen mit der Digitalisierung ist hier ein wichtiges Thema für Forschung und Lehre. An der Goethe-Universität kümmert sich eigens eine Arbeitsgruppe Medien darum, die unterschiedlichen Aktivitäten im Fachbereich und auch außerhalb zu begleiten und zu vernetzen.
Persönliches Feedback bei Vorlesungen mit Hunderten Studierenden erscheint bisher utopisch – auch nach dem Digitalisierungsschub in Corona-Zeiten. Tools aus dem Forschungsgebiet der »Learning Analytics« könnten künftig den Studierenden Rückmeldung geben und zugleich den Betreuern Hinweise liefern, wo noch Hilfestellung nötig ist.

Welchen Einfluss haben digitale Technologien auf das menschliche Wahrnehmen, Denken und Handeln? Schaden Computerspiele der Entwicklung junger Gehirne? Und gibt es tatsächlich so etwas wie eine »digitale Demenz«, eine durch die Nutzung moderner Technologien bedingte wachsende Vergesslichkeit? Auf einige dieser Fragen gibt es bereits Antworten, die empirisch belegt sind.
Durch die Digitalisierung ist die soziale Bedeutung des Urheberrechts stark gewachsen. Die vom Urheberrecht etablierte Exklusivitätskultur steht überdies in einem fundamentalen Konflikt mit der im Internet vorherrschenden Zugangskultur. Ein Beispiel hierfür ist der Streit um die jüngste Urheberrechtsrichtlinie der EU.
Das Thema Datenschutz wird in Zeiten von Internet und künstlicher Intelligenz immer komplexer. Die Entwicklungen scheinen in manchen Bereichen kaum noch beherrschbar zu sein. Nur ein Zusammenspiel von Rechtswissenschaft und Informatik kann den Einzelnen und bestimmte gesellschaftliche Gruppen vor Diskriminierung schützen.
The early-diverging oomycetes contain a large number of holocarpic obligate parasites of diatoms, algae, aquatic phycomycetes, and invertebrate animals. These organisms are diverse and widespread. However, taxonomic placement most of the early-diverging oomycetes remains provisional and unresolved, since many have not been sequenced and studied for molecular phylogeny. Here, we report the taxonomy and phylogeny of several holocarpic oomycetes that we have rediscovered and newly classified, including several new species combinations. Phylogenetic reconstructions revealed that the type species of genus Ectrogella (E. bacillariacearum) is a member of the early-diverging Saprolegniales, while the type species of Olpidiopsis (O. saprolegniae) and Pontisma (P. lagenidioides) grouped within the early-diverging lineage of oomycetes forming distinct clades. Since the monophyletic red-algae parasitoids are unrelated to the Olpidiopsis, these were reclassified to the genus Pontisma, while genus Diatomophthora was introduced to accommodate all the diatom parasitoids that were previously assigned to Olpidiopsis. In addition, four new oomycete parasitoids, Miracula helgolandica, Miracula moenusica, Diatomophthora drebesii and Olpidiopsis parthenogenetica and a single rediscovered species, Diatomophthora gillii, are also classified here, including eight new species combinations of red-algae parasites (Pontisma bostrychiae, P. heterosiphoniae, P. muelleri, P. palmariae, P. porphyrae, P. pyropiae) and diatom parasitoids (Diatomophthora drebesii, D. gillii). The results obtained in this study have further improved the resolution and expanded the knowledge on the phylogeny of the earlydiverging oomycetes, leading to the establishment of three new orders (Miraculales, Diatomophthorales, Pontismatales) and one order (Anisolpidiales) being reintroduced.
Mit der COLTRIMS-Technik können immer kompliziertere Reaktionen untersucht werden, dabei steigt aber die Zahl der zu detektierenden Reaktionsfragmente. Der Nachweis von Ionen ist üblicherweise gut möglich, da die entsprechenden Flugzeiten groß sind im Vergleich zur Totzeit der benutzten Detektoren. Elektronen hingegen sind sehr leicht und erreichen den Detektor innerhalb von wenigen 10 ns. Aktuelle Detektoren erlauben aber nur den Nachweis weniger Elektronen und es werden somit neue Detektoren benötigt, um alle Teilchen nachzuweisen. Ziel dieser Arbeit war es also, einen Detektor zu entwickeln, der dies erreicht.
Zu Beginn dieser Monografie wird die COLTRIMS-Technik vorgestellt. Die Experimente mit dieser Messmethode finden hauptsächlich mit einer Laufzeitanode statt. Diese stößt aber bei dem Nachweis von mehreren Teilchen an ihre Grenzen und manche Experimente können nur unvollständig analysiert werden.
Damit ein neuer Detektor entwickelt werden kann, muss erst verstanden werden, wie die zu detektierenden Teilchen/Signale entstehen und wie ihre Eigenschaften sind. Aus diesem Grund wird das Sekundärteilchen-erzeugende MCP ausführlich vorgestellt.
Weiterhin gibt diese Arbeit einen umfassenden Überblick über bereits realisierte Anoden. Verschiedene Repräsentanten der fünf Anodenarten (Flächen-, Streifen-/Pixel-, Laufzeit-, Kamera-, sowie Halbleiter-Anode) werden vorgestellt und bewertet.
Mit diesem Wissen konnten drei Ansätze für neue Anoden entwickelt, designt, produziert, getestet und bewertet werden. Alle neu entwickelten Anoden benutzen Leiterplatinen als Basis und werden in derselben Vakuumkammer getestet. Auch wenn die Detektionsprinzipien der drei getesteten Detektoren unterschiedlich sind, so verläuft die Auskopplung, Verarbeitung und Digitalisierung der Signale nach dem gleichen Schema. Außerdem wurden im Rahmen dieser Arbeit diverse Algorithmen entwickelt und programmiert, mit deren Hilfe die Signalauswertung und Positionsbestimmung erfolgt.
Das dritte Kapitel beschreibt die neu entwickelte Draht-Harfen-Anode. Dieser Detektor besteht aus vielen kurzen Drähten die parallel auf Rahmen aus Leiterplatinen gespannt werden. Aus dieser Anode ließ sich im Rahmen dieser Arbeit aber kein funktionsfähiger Detektor entwickeln und es wird empfohlen, diesen Ansatz nicht weiterzuverfolgen.
Im Kapitel über die Pixel-Anode mit Streifenauslese wird ein Ansatz vorgestellt, bei dem die Elektronenwolke von einem Muster aus leitenden Rauten absorbiert wird. Es wurde ein funktionsfähiger Detektor mit MAMA-Verschaltung realisiert. Die aktive Fläche ist mit einem Durchmesser von 50 mm aber zu klein. Eine große Variante der Anode ist in der realisierten Form aber nicht als Detektor geeignet.
Als dritter neuer Detektor wird die Streifen-Laufzeit-Anode beschrieben. Diese besteht aus einem rechteckigen Muster von Pixeln, die in einer Richtung über eine Zeitverzögerung ausgelesen werden. Dieser Ansatz ist sehr vielversprechend und es ließen sich nicht nur einzelne Teilchen nachweisen, sondern auch beim Aufbruch eines D2+-Moleküls konnten beide Fragmente gemessen werden.
Das letzte Kapitel befasst sich mit weiteren Konzepten, die als Detektor realisiert werden könnten.
Objective: Cerebral vasospasm (CVS) after a ruptured arteriovenous malformation (AVM) is rarely reported. This study is aimed at evaluating the predictive variables in AVM hemorrhage for CVS. Methods: A total of 160 patients with ruptured AVMs were admitted to our neurosurgical department from 2002 to 2018. The frequency of cerebral vasospasm after AVM hemorrhage and the impact of AVM-associated aneurysms were evaluated. We compared different bleeding patterns, such as intracerebral hemorrhage (ICH), subarachnoid hemorrhage (SAH) or a combination of both (ICH + SAH) and evaluated predictive variables for outcome in last follow-up. Results: A total of 62 (39%) patients had AAA, mostly located prenidal (75.8%). AVMs with ruptured aneurysms often resulted in ICH with SAH component (p < 0.001). Eighty-two patients (51%) presented a SAH component, and CVS occurred in 6 patients (7.3%), mostly due to a ruptured infratentorial AVM (p < 0.03). Infratentorial location and the amount of SAH component (p < 0.001) predicted the incidence of CVS significantly. Cerebral infarction was significantly associated with CVS (p < 0.02). Conclusion: SAH component and infratentorial location of ruptured AVMs may harbor a higher risk for CVS. Follow-up with angiographic imaging should be considered in patients with infratentorial AVM hemorrhage and delayed neurologic deterioration to rule out CVS.
Genotoxicity assessment is of high relevance for crude and refined petroleum products, since oil compounds are known to cause DNA damage with severe consequences for aquatic biota as demonstrated in long-term monitoring studies. This study aimed at the optimization and evaluation of small-scale higher-throughput assays (Ames fluctuation, micronucleus, Nrf2-CALUX®) covering different mechanistic endpoints as first screening tools for genotoxicity assessment of oils. Cells were exposed to native and chemically dispersed water-accommodated fractions (WAFs) of three oil types varying in their processing degree. Independent of an exogenous metabolic activation system, WAF compounds induced neither base exchange nor frame shift mutations in bacterial strains. However, significantly increased chromosomal aberrations in zebrafish liver (ZF-L) cells were observed. Oxidative stress was indicated for some treatments and was not correlated with observed DNA damage. Application of a chemical dispersant increased the genotoxic potential rather by the increased bioavailability of dissolved and particulate oil compounds. Nonetheless, the dispersant induced a clear oxidative stress response, indicating a relevance for general toxic stress. Results showed that the combination of different in vitro assays is important for a reliable genotoxicity assessment. Especially, the ZF-L capable of active metabolism and DNA repair seems to be a promising model for WAF testing.
Objective: Phenotypic (Sensititre Myco, pDST) and genotypic drug susceptibility testing (GenoType NTM DR, gDST) in M. avium complex (MAC) have become available as standardized assays, but comparable data is needed. This study aimed to investigate the phenotypic and genotypic drug susceptibility patterns in MAC clinical isolates.
Methods: Overall, 98 isolates from 85 patients were included. pDST and gDST were performed on all isolates and results compared regarding specificity and sensitivity using pDST as a reference method. The impact of drug instability on pDST results was studied using a biological assay over 14 days. In addition, the evolution of antimicrobial resistance was investigated in sequential isolates of 13 patients.
Results: Macrolide resistance was rare, 1.2% (95% CI 0.7–7.3) of isolates in the base cohort. No aminoglycoside resistances were found, but 14.1% of the studied isolates (95% CI 7.8–23.8) showed intermediate susceptibility. The GenoType NTM DR identified two out of four macrolide-resistant isolates. Antibiotic stability was demonstrated to be poor in rifampicin, rifabutin, and doxycycylin.
Conclusions: pDST results in NTM for unstable antibiotics must be interpreted with care. A combination of pDST and gDST will be useful for the guidance of antimicrobial therapy in MAC-disease.
Molecular and cellular research modalities for the study of liver pathologies have been tremendously improved over the recent decades. Advanced technologies offer novel opportunities to establish cell isolation techniques with excellent purity, paving the path for 2D and 3D microscopy and high-throughput assays (e.g., bulk or single-cell RNA sequencing). The use of stem cell and organoid research will help to decipher the pathophysiology of liver diseases and the interaction between various parenchymal and non-parenchymal liver cells. Furthermore, sophisticated animal models of liver disease allow for the in vivo assessment of fibrogenesis, portal hypertension and hepatocellular carcinoma (HCC) and for the preclinical testing of therapeutic strategies. The purpose of this review is to portray in detail novel in vitro and in vivo methods for the study of liver cell biology that had been presented at the workshop of the 8th meeting of the European Club for Liver Cell Biology (ECLCB-8) in October of 2018 in Bonn, Germany.
Radar technology in the millimeter-wave frequency band offers many interesting features for wind park surveillance, such as structural monitoring of rotor blades or the detection of bats and birds in the vicinity of wind turbines (WTs). Currently, the majority of WTs are affected by shutdown algorithms to minimize animal fatalities via direct collision with the rotor blades or barotrauma effects. The presence of rain is an important parameter in the definition of those algorithms together with wind speed, temperature, time of the day, and season of the year. A Ka-band frequency-modulated continuous-wave radar (33.4-36.0 GHz) installed at the tower of a 2-MW WT was used during a field study. We have observed characteristic rain-induced patterns, based on the range-Doppler algorithm. To better understand those signatures, we have developed a laboratory experiment and implemented a numerical modeling framework. Experimental and numerical results for rain detection and classification are presented and discussed here. Based on this article, a bat- and bird-friendly adaptive WT control can be developed for improved WT efficiency in periods of rain and, at the same time, reduced animal mortality.
Objective: To study the effect of total hip replacement (THR) on serum cartilage oligomeric matrix protein concentration (sCOMP) and its correlation with joint loading during gait in patients with unilateral hip osteoarthritis.
Design: In this prospective multimodal (clinical, biomechanical, biochemical) study blood samples from 15 patients were taken before and up to three times after THR (7 days, 3 months and 1 year), each after a resting period of at least 30 min, for analysis of sCOMP. Gait analysis was performed before and 1 year after THR to determine hip and knee joint moments.
Results: Seven days after THR, sCOMP decreased significantly compared to the preoperative measurement (p < 0.001). Three months and 1 year postoperatively, sCOMP reverted to concentrations in the range of the preoperative value. One year postoperatively, a linear correlation between sCOMP and the maximum hip flexion moment was indicated in the first half of the stance phase on the unaffected side (r = −0.736, p = 0.024). No further correlations could be determined.
Conclusions: Surprisingly, the removal of a joint affected by osteoarthritis did not have a sustained effect on sCOMP. Both before and after THR there was no scientifically substantiated correlation between sCOMP and joint moments from gait analysis. Consequently, the examination of sCOMP is not useful to detect altered joint loads that may influence degenerative changes of adjacent joints after THR.

With the rapid growth of technology in recent years, we are surrounded by or even dependent on the use of technological devices such as smartphones as they are now an indispensable part of our life. Smartphone applications (apps) provide a wide range of utilities such as navigation, entertainment, fitness, etc. To provide such context-sensitive services to users, apps need to access users' data including sensitive ones, which in turn, can potentially lead to privacy invasions. To protect users against potential privacy invasions in such a vulnerable ecosystem, legislation such as the European Union General Data Protection Regulation (EU GDPR) demands best privacy practices. Therefore, app developers are required to make their apps compatible with legal privacy principles enforced by law. However, this is not an easy task for app developers to comprehend purely legal principles to understand what needs to be implemented. Similarly, bridging the gap between legal principles and technical implementations to understand how legal principles need to be implemented is another barrier to develop privacy-friendly apps. To this end, this paper proposes a privacy and security design guide catalog for app developers to assist them in understanding and adopting the most relevant privacy and security principles in the context of smartphone apps. The presented catalog is aimed at mapping the identified legal principles to practical privacy and security solutions that can be implemented by developers to ensure enhanced privacy aligned with existing legislation. Through conducting a case study, it is confirmed that there is a significant gap between what developers are doing in reality and what they promise to do. This paper provides researchers and developers of privacy-related technicalities an overview of the characteristics of existing privacy requirements needed to be implemented in smartphone ecosystems, on which they can base their work.
Inclusions of breyite (previously known as walstromite-structured CaSiO3) in diamond are usually interpreted as retrogressed CaSiO3 perovskite trapped in the transition zone or the lower mantle. However, the thermodynamic stability field of breyite does not preclude its crystallization together with diamond under upper-mantle conditions (6–10 GPa). The possibility of breyite forming in subducted sedimentary material through the reaction CaCO3 + SiO2 = CaSiO3 + C + O2 was experimentally evaluated in the CaO–SiO2–C–O2 ± H2O system at 6–10 GPa, 900–1500 ∘C and oxygen fugacity 0.5–1.0 log units below the Fe–FeO (IW) buffer. One experimental series was conducted in the anhydrous subsystem and aimed at determining the melting temperature of the aragonite–coesite (or stishovite) assemblage. It was found that melting occurs at a lower temperature (∼1500 ∘C) than the decarbonation reaction, which indicates that breyite cannot be formed from aragonite and silica under anhydrous conditions and an oxygen fugacity above IW – 1. In the second experimental series, we investigated partial melting of an aragonite–coesite mixture under hydrous conditions at the same pressures and redox conditions. The melting temperature in the presence of water decreased strongly (to 900–1200 ∘C), and the melt had a hydrous silicate composition. The reduction of melt resulted in graphite crystallization in equilibrium with titanite-structured CaSi2O5 and breyite at ∼1000 ∘C. The maximum pressure of possible breyite formation is limited by the reaction CaSiO3 + SiO2 = CaSi2O5 at ∼8 GPa. Based on the experimental results, it is concluded that breyite inclusions found in natural diamond may be formed from an aragonite–coesite assemblage or carbonate melt at 6–8 GPa via reduction at high water activity.
Background: Experienced and anticipated regret influence physicians’ decision-making. In medicine, diagnostic decisions and diagnostic errors can have a severe impact on both patients and physicians. Little empirical research exists on regret experienced by physicians when they make diagnostic decisions in primary care that later prove inappropriate or incorrect. The aim of this study was to explore the experience of regret following diagnostic decisions in primary care.
Methods: In this qualitative study, we used an online questionnaire on a sample of German primary care physicians. We asked participants to report on cases in which the final diagnosis differed from their original opinion, and in which treatment was at the very least delayed, possibly resulting in harm to the patient. We asked about original and final diagnoses, illness trajectories, and the reactions of other physicians, patients and relatives. We used thematic analysis to assess the data, supported by MAXQDA 11 and Microsoft Excel 2016.
Results: 29 GPs described one case each (14 female/15 male patients, aged 1.5–80 years, response rate < 1%). In 26 of 29 cases, the final diagnosis was more serious than the original diagnosis. In two cases, the diagnoses were equally serious, and in one case less serious. Clinical trajectories and the reactions of patients and relatives differed widely. Although only one third of cases involved preventable harm to patients, the vast majority (27 of 29) of physicians expressed deep feelings of regret.
Conclusion: Even if harm to patients is unavoidable, regret following diagnostic decisions can be devastating for clinicians, making them ‘second victims’. Procedures and tools are needed to analyse cases involving undesirable diagnostic events, so that ‘true’ diagnostic errors, in which harm could have been prevented, can be distinguished from others. Further studies should also explore how physicians can be supported in dealing with such events in order to prevent them from practicing defensive medicine.
We contribute to the foundations of tropical geometry with a view toward formulating tropical moduli problems, and with the moduli space of curves as our main example. We propose a moduli functor for the moduli space of curves and show that it is representable by a geometric stack over the category of rational polyhedral cones. In this framework, the natural forgetful morphisms between moduli spaces of curves with marked points function as universal curves.
Our approach to tropical geometry permits tropical moduli problems—moduli of curves or otherwise—to be extended to logarithmic schemes. We use this to construct a smooth tropicalization morphism from the moduli space of algebraic curves to the moduli space of tropical curves, and we show that this morphism commutes with all of the tautological morphisms.
Background: Point of care devices for performing targeted coagulation substitution in patients who are bleeding have become increasingly important in recent years. New on the market is the Quantra. It is a device that uses sonorheometry, a sonic estimation of elasticity via resonance, which is a novel ultrasound-based technology that measures viscoelastic properties of whole blood. Several studies have already shown the comparability of the Quantra with devices already established on the market, such as the rotational thromboelastometry (ROTEM) device.
Objective: In contrast to existing studies, this study is the first prospective interventional study using this new system in a cardiac surgical patient cohort. We will investigate the noninferiority between an already existing coagulation algorithm based on the ROTEM/Multiplate system and a new algorithm based on the Quantra system for the treatment of coagulopathic cardiac surgical patients.
Methods: The study is divided into two phases. In an initial observation phase, whole blood samples of 20 patients obtained at three defined time points (prior to surgery, after completion of cardiopulmonary bypass, and on arrival in the intensive care unit) will be analyzed using both the ROTEM/Multiplate and Quantra systems. The obtained threshold values will be used to develop a novel algorithm for hemotherapy. In a second intervention phase, the new algorithm will be tested for noninferiority against an algorithm used routinely for years in our department.
Results: The main objective of the examination is the cumulative loss of blood within 24 hours after surgery. Statistical calculations based on the literature and in-house data suggest that the new algorithm is not inferior if the difference in cumulative blood loss is <150 mL/24 hours.
Conclusions: Because of the comparability of the Quantra sonorheometry system with the ROTEM measurement methods, the existing hemotherapy treatment algorithm can be adapted to the Quantra device with proof of noninferiority.

The present paper aims at providing empirical evidence for dialectal variation concerning the perception of the central vowel [ɐ] in European Portuguese (EP). More concretely, this study compares the perception of the contrast between [a] and [ɐ] by native speakers of two varieties of EP: 23 speakers of a northern Portuguese dialect (from the city of Braga) and 23 speakers of the Littoral Center variety of EP (from the city of Lisbon, defined as Standard European Portuguese (SEP)). Based on a discrimination test, the results show that the two groups of speakers differ with respect to the perception of the contrast between the two central vowels under investigation. The speakers of the northern variety differentiate less between the two central vowels compared to the speakers from Lisbon.
Background: Computerized virtual patients (VP) have spread into many areas of healthcare delivery and medical education. They provide various advantages like flexibility in pace and space of learning, a high degree of teaching reproducibility and a cost effectiveness. However, the educational benefit of VP as an additive or also as an alternative to traditional teaching formats remains unclear. Moreover, there are no randomized-controlled studies that investigated the use of VP in a dental curriculum. Therefore, this study investigates VP as an alternative to lecturer-led small-group teaching in a curricular, randomized and controlled setting.
Methods: Randomized and controlled cohort study. Four VP cases were created according to previously published design principles and compared with lecturer-led small group teaching (SGT) within the Oral and Maxillofacial Surgery clerkship for dental students at the Department for Cranio-, Oral and Maxillofacial Plastic Surgery, Goethe University, Frankfurt, Germany. Clinical competence was measured prior (T0), directly (T1) and 6 weeks (T2) after the intervention using theoretical tests and a self-assessment questionnaire. Furthermore, VP design was evaluated using a validated toolkit.
Results: Fifty-seven students (VP = 32; SGT = 25) agreed to participate in the study. No competence differences were found at T0 (p = 0.56). The VP group outperformed (p < .0001) the SGT group at T1. At T2 there was no difference between both groups (p = 0.55). Both interventions led to a significant growth in self-assessed competence. The VP group felt better prepared to diagnose and treat real patients and regarded VP cases as a rewarding learning experience.
Conclusions: VP cases are an effective alternative to lecture-led SGT in terms of learning efficacy in the short and long-term as well as self-assessed competence growth and student satisfaction. Furthermore, integrating VP cases within a curricular Oral and Maxillofacial Surgery Clerkship is feasible and leads to substantial growth of clinical competence in undergraduate dental students.
Rationale: Early reperfusion in patients experiencing acute ischemic stroke is critical, especially for patients with large vessel occlusion who have poor prognosis without revascularization. S olitaire™ stent retriever devices have been shown to immediately restore vascular perfusion safely, rapidly, and effectively in acute ischemic stroke patients with large vessel occlusions.
Aim: The aim of the study was to demonstrate that, among patients with large vessel, anterior circulation occlusion who have received intravenous tissue plasminogen activator, treatment with S olitaire revascularization devices reduces degree of disability 3 months post stroke.
Design: The study is a global multicenter, two‐arm, prospective, randomized, open, blinded end‐point trial comparing functional outcomes in acute ischemic stroke patients who are treated with either intravenous tissue plasminogen activator alone or intravenous tissue plasminogen activator in combination with the Solitaire device. Up to 833 patients will be enrolled.
Procedures:Patients who have received intravenous tissue plasminogen activator are randomized to either continue with intravenous tissue plasminogen activator alone or additionally proceed to neurothrombectomy using the S olitaire device within six‐hours of symptom onset.
Study Outcomes: The primary end‐point is 90‐day global disability, assessed with the modified R ankin S cale (mRS). Secondary outcomes include mortality at 90 days, functional independence (mRS ≤ 2) at 90 days, change in N ational I nstitutes of H ealth S troke S cale at 27 h, reperfusion at 27 h, and thrombolysis in cerebral infarction 2b/3 flow at the end of the procedure.
Analysis:Statistical analysis will be conducted using simultaneous success criteria on the overall distribution of modified R ankin S cale (R ankin shift) and proportions of subjects achieving functional independence (mRS 0–2).
We investigate system-size effects on the rotational diffusion of membrane proteins and other membrane-embedded molecules in molecular dynamics simulations. We find that the rotational diffusion coefficient slows down relative to the infinite-system value by a factor of one minus the ratio of protein and box areas. This correction factor follows from the hydrodynamics of rotational flows under periodic boundary conditions and is rationalized in terms of Taylor-Couette flow. For membrane proteins like transporters, channels, or receptors in typical simulation setups, the protein-covered area tends to be relatively large, requiring a significant finite-size correction. Molecular dynamics simulations of the protein adenine nucleotide translocase (ANT1) and of a carbon nanotube porin in lipid membranes show that the hydrodynamic finite-size correction for rotational diffusion is accurate in standard-use cases. The dependence of the rotational diffusion on box size can be used to determine the membrane viscosity.
Background: The MRI Breast Imaging-Reporting and Data System (BI-RADS) lexicon recommends that a breast MRI proto-col contain T2-weighted and dynamic contrast-enhanced (DCE) MRI sequences. The addition of diffusion-weighted imag-ing (DWI) significantly improves diagnostic accuracy. This study aims to clarify which descriptors from DCE-MRI, DWI, andT2-weighted imaging are most strongly associated with a breast cancer diagnosis.Purpose/Hypothesis: To develop a multiparametric MRI (mpMRI) model for breast cancer diagnosis incorporating Ameri-can College of Radiology (ACR) BI-RADS recommended descriptors for breast MRI with DCE, T2-weighted imaging, andDWI with apparent diffusion coefficient (ADC) mapping.Study Type: Retrospective.Subjects: In all, 188 patients (mean 51.6 years) with 210 breast tumors (136 malignant and 74 benign) who underwentmpMRI from December 2010 to September 2014.Field Strength/Sequence: IR inversion recovert DCE-MRI dynamic contrast-enhanced magnetic resonance imaging VIBEVolume-Interpolated-Breathhold-Examination FLASH turbo fast-low-angle-shot TWIST Time-resolved angiography withstochastic Trajectories.Assessment: Two radiologists in consensus and another radiologist independently evaluated the mpMRI data. Charac-teristics for mass (n = 182) and nonmass (n = 28) lesions were recorded on DCE and T2-weighted imaging accordingto BI-RADS, as well as DWI descriptors. Two separate models were analyzed, using DCE-MRI BI-RADS descriptors, T2-weighted imagines, and ADCmean as either a continuous or binary form using a previously published ADC cutoffvalue of ≤1.25 × 10−3mm2/sec for differentiation between benign and malignant lesions. Histopathology was the stan-dard of reference.Statistical Tests: χ2test, Fisher’s exact test, Kruskal–Wallis test, Pearson correlation coefficient, multivariate logistic regres-sion analysis, Hosmer–Lemeshow test of goodness-of-fit, receiver operating characteristics analysis.Results: In Model 1, ADCmean (P = 0.0031), mass margins with DCE (P = 0.0016), and delayed enhancement with DCE(P = 0.0016) were significantly and independently associated with breast cancer diagnosis; Model 2 identified ADCmean(P = 0.0031), mass margins with DCE (P = 0.0012), initial enhancement (P = 0.0422), and delayed enhancement with DCE(P = 0.0065) to be significantly independently associated with breast cancer diagnosis. Zoos attract millions of visitors every year, many of whom are schoolchildren. For this reason, zoos are important institutions for the environmental education of future generations. Empirical studies on the educational impact of environmental education programs in zoos are still rare. To address this issue, we conducted two studies: In study 1, we investigated students’ interests in different biological topics, including zoos (n = 1,587). Data analysis of individual topics revealed large differences of interest, with advanced students showing less interest in zoos. In study 2, we invited school classes of this age group to visit different guided tours at the zoo and tested connection to nature before and after each educational intervention (n = 608). The results showed that the guided tours are an effective tool to raise students’ connection to nature. Add-on components have the potential to further promote connection to nature. The education programs are most effective with students with a low initial nature connection.
Der vorliegende Beitrag stellt die Bedeutsamkeit emotionaler und transformationaler Führungskompetenzen im Kontext inklusiver Schulentwicklung sowie allgemein im Rahmen eines transformationalen Schulmanagements dar. Daran anknüpfend werden als Praxisbeispiele zur Weiterqualifizierung ein Master-Fernstudiengang für Schulmanagement sowie ein in weiterer Entwicklung befindliches digitales Leadership-Training für Schulentwicklung vorgestellt. Letzteres fokussiert insbesondere die Förderung emotionaler und transformationaler Führungskompetenzen, welche wesentlich dazu beitragen können, inspirierend zu motivieren, kollegiale Beziehungen gewinnbringend zu gestalten und adäquat durch inklusive Schulentwicklungsprozesse zu begleiten.
Fragen von Weiterbildungsbeteiligung – und damit Fragen von Inklusion/Exklusion in einem soziologisch-sozialstrukturellen Sinn – spielen in der Erwachsenenbildung seit jeher eine große Rolle. Inklusion unter der Perspektive von geistig-körperlicher Beeinträchtigung ist in der Erwachsenenbildungswissenschaft dagegen ein (noch) randständiges Thema. Erst seit wenigen Jahren beginnen Erwachsenenbildungseinrichtungen wie auch die Erwachsenenbildungswissenschaft, sich verstärkt mit Fragen von Inklusion auf organisationaler, professioneller und didaktischer Ebene zu befassen. Ein großes Desiderat sowohl in der wissenschaftlichen Fokussierung als auch handlungspraktischen Ausgestaltung stellt die Professionalität bzw. Qualifizierung des Personals für eine inklusive Erwachsenenbildung dar, insbesondere unter der Perspektive einer nicht isolierenden, sondern für die Gesamteinrichtung funktionalen Weise.
Der Text vertritt die These, dass gerade in der Erwachsenenbildung Fachlichkeit aus einem komplexen Zusammenspiel von makro-, meso- und mikrodidaktischen Faktoren resultiert. Die komplexe Beziehung zwischen Bedarfsanalyse, Teilnehmendengewinnung und Angebotsrealisierung verlangt in der Erwachsenenbildung eine Fachlichkeit, die in einem kooperativen Miteinander von Leitung, Planung, Kursgestaltung und administrativer Begleitung aufgespannt ist. Inklusive Fachlichkeit muss aus diesem komplexen Miteinander hervorgehen, was für die theoretische Konzeptionalisierung inklusionsorientierter Professionalitätsentwicklung von großer Bedeutung ist.
Im Folgenden wird daher der Versuch unternommen, diese inklusionsorientierte, kooperative Fachlichkeit im Rahmen öffentlicher Weiterbildung und am Beispiel von Blindheit und Sehbehinderung genauer zu bestimmen. Dazu wird zunächst in einer allgemeinen Perspektive inklusive Fachlichkeit als kooperative Gesamtleistung öffentlicher allgemeiner Erwachsenenbildung skizziert, um dann die damit verbundenen Herausforderungen am Beispiel der Adressatengruppe blinder und sehbeeinträchtigter Personen zu verdeutlichen. In dieser Perspektive zeigt sich inklusive Erwachsenenbildung als eine komplexe Bildungsdienstleistung, die professionstheoretisch als kooperative Verknüpfungsleistung im Mehrebenensystem von Erwachsenenbildung zu konzipieren ist.
Professionelle Kompetenz gilt als zentrale Handlungsressource unterrichtlichen Handelns. Berufsbezogene Überzeugungen und Werte (beliefs) können dessen Qualität maßgeblich beeinflussen und sind Gegenstand zahlreicher empirischer Untersuchungen. Eine Vielzahl von Forschungsvorhaben präzisieren den gewählten Beliefbegriff hinsichtlich fachwissenschaftlicher, fachdidaktischer oder (sonder)pädagogischer Fragestellungen. In dem vorliegenden Beitrag wird zunächst unser Begriffsverständnis von professioneller Handlungskompetenz, Inklusion und Teachers’ Beliefs theoretisch begründet. Von diesen Betrachtungen ausgehend, wird das Forschungsdesign einer qualitativen Studie abgeleitet, welches die inklusionsbezogenen Überzeugungen von angehenden Lehrkräften hinsichtlich eines gemeinsamen Unterrichts beschreibt sowie Implikationen für die Hochschullehre aus Studierendenperspektive ermöglicht. Die Ergebnisse geben Aufschluss über das Heterogenitätsverständnis von Lehramtsstudierenden der Regelschule, deren vorherrschenden Rollenbildern und ideellen Überzeugungen zu inklusivem Unterricht. Sie skizzieren außerdem hochschuldidaktische Maßnahmen, welche aus Studierendenperspektive einen Beitrag für die Vorbereitung im Unterricht inklusiver Klassen leisten können. Konsequenzen, die sich aus unserer Sicht für die Ausbildung von Lehramtsstudierenden ergeben, werden mit Fokus auf die Entmystifizierung inklusionsbezogener Überzeugungen diskutiert.
Der Beitrag arbeitet den Expertisebegriff für die inklusionsbezogene Professionalisierungsforschung auf und diskutiert diesen im Kontext des Desiderats nach einem übergreifenden Forschungsparadigma, das den strukturtheoretischen, den berufsbiografischen und den kompetenztheoretischen Ansatz verbindet. Die Überlegungen werden anhand von empirischen Befunden einer qualitativen (Teil-)Studie zur Expertise von Fortbildner*innen im Feld der inklusionsbezogenen Fortbildung exemplifiziert.
Der vorliegende Beitrag fasst Inklusion unter Bezugnahme auf Oevermanns (1996) „Skizze einer revidierten Theorie professionalisierten Handelns“ als Auftrag zur Professionalisierung entlang der therapeutischen Dimension in der Beziehung von Lehrpersonen und Schüler*innen. Dieser trifft in der beruflichen Bildung auf ein spezifisches Spannungsfeld, insofern unter dem Primat der Beruflichkeit eine starke Fokussierung auf ‚Wissen und Können‘ prozessiert wird. Vor diesem Hintergrund wird exemplarisch die Ausgestaltung der pädagogischen Praxis anhand von im BMBF-geförderten Forschungsprojekt StiEL entstandenen Datenmaterial mittels der Dokumentarischen Methode der Textinterpretation analysiert. Ziel ist es, die Orientierungen von Lehrpersonen hinsichtlich pädagogischer Fachlichkeit aus deren handlungsleitenden Wissensbeständen heraus zu rekonstruieren, um die Bedingungen für die Aufnahme des Inklusionsanspruchs in den eigenen Unterricht sowie implizite, durch den Anspruch ‚Inklusion‘ hervorgerufene Ambivalenzen des pädagogisch-professionellen Handelns sichtbar zu machen. Abschließend werden Ansatzpunkte für Fort- und Weiterbildungen abgeleitet.
Ausgehend von spezifischen Anforderungen inklusiver Settings wird in diesem Beitrag ein Ansatz zur Weiterentwicklung des Modells der professionellen Handlungskompetenz von Lehrkräften für inklusiven (Mathematik-)Unterricht vorgestellt. Inklusion wird dabei verstanden als fachliches Lernen und gemeinsame Teilhabe aller Schülerinnen und Schüler. Für die Weiterentwicklung werden zunächst Anforderungen durch inklusive Bildung betrachtet, einerseits abgeleitet aus verschiedenen Aufgaben- und Kompetenzkatalogen, andererseits basierend auf Kernbeständen für den Umgang mit Heterogenität im (Mathematik-)Unterricht im Allgemeinen und für inklusiven (Mathematik-)Unterricht im Speziellen. Anschließend wird daraus Wissen zur Bewältigung dieser Anforderungen abgeleitet und unter Rückgriff auf die verschiedenen Bereiche des Professionswissens (Fachwissen, pädagogisches Wissen, fachdidaktisches Wissen, Organisationswissen und Beratungswissen) weiter ausgeführt, wobei der Fokus auf das fachdidaktische Wissen im inklusiven Mathematikunterricht gelegt wird. Die affektiven und motivationalen Merkmale (Überzeugungen, Selbstregulation und Motivation) der professionellen Handlungkompetenz werden ebenfalls in die Weiterentwicklung des Modells einbezogen. Insgesamt erfolgt die Weiterentwicklung des Modells somit im Rahmen einer Erweiterung der einzelnen bestehenden Kompetenzbereiche um Aspekte, die unter Inklusionsgesichtspunkten besondere Bedeutung erfahren. Mit der Identifizierung von Kompetenzbereichen bei Lehrkräften zu Beginn einer Fortbildung zu inklusivem Mathematikunterricht wird ein Beispiel präsentiert, wie das weiterentwickelte Modell angewendet werden kann. In diesem Zusammenhang konnten in den Äußerungen der Lehrkräfte – abgesehen vom Fachwissen – alle Kompetenzbereiche identifiziert werden. Abschließend werden Möglichkeiten vorgestellt, wie das Modell im Kontext der Lehrerprofessionalisierungsforschung weiterhin eingesetzt werden kann.

Orientiert am Themenschwerpunkt der „theoretischen und empirischen Klärung des Verständnisses pädagogischer Fachlichkeit“ beschäftigt sich unser Beitrag mit den Barrieren und Chancen, die sich in der Hochschulstruktur und –kultur und der universitären Lehre darstellen, wobei wir uns exemplarisch der Ausbildung inklusionsbezogener pädagogischer Fachlichkeit bei angehenden Lehrkräften zuwenden. Mit Bezug zu aktuellen Entwicklungen der bildungspolitischen Landschaft in Deutschland wird zunächst die Notwendigkeit eines Umdenkens in der Lehramtsausbildung, wie auch in der gesamten Hochschullandschaft, dargelegt und diskutiert. Dabei wird das dem Beitrag zugrunde liegende Verständnis von Inklusion verdeutlicht. Die sich daraus ergebenden Anforderungen an Hochschulen, besonders in Bezug auf ihr Selbstverständnis, das Verständnis von Wissenschaftlichkeit und die gelebte Hochschulkultur, werden daraufhin formuliert. Daran schließen Überlegungen zur Umsetzung inklusionsorientierter Praxen in der Lehrer*innenbildung an. Ein besonderer Fokus liegt dabei auf der partizipativen Lehre als möglichem Werkzeug für eine inklusionssensible Hochschulentwicklung. Am Beispiel partizipativer Lehre gemeinsam mit Menschen mit Lernschwierigkeiten1 werden Möglichkeiten aufgezeigt, wie Menschen, die bisher keinen Zugang zur akademischen Wissensvermittlung hatten, als Lehrende tätig sein und so sowohl von Biografien unter der Erfahrung von Diskriminierung berichten, als auch diese Erfahrungen in die Produktion von Wissen einfließen lassen können. In der Diskussion um partizipative Lehre werden verschiedene Widersprüche und Diskurse deutlich, die geeignet sind, um über Hochschulkulturen und -praxen neu nachzudenken.
Das Spektrum kunstpädagogischer Thematisierungen von Inklusion ist – vergleichbar mit anderen Fachdidaktiken - wenig entfaltet. Die bisher vorliegenden Beiträge sind bisher kaum auf eine inklusionsbezogene Professionalisierung hin reflektiert. Die kontinuierliche Weiterentwicklung pädagogischer Professionalität zählt aber zu den grundlegenden Aufgaben der Kunstpädagogik und Bildungswissenschaft. Insbesondere da einzelne Zugänge der Professionalisierungsforschung theoretisch wie empirisch leistungsfähig sind und eine Reihe spezifischer Untersuchungen zu Aspekten der Professionalisierung hervorgebracht haben. Damit geht jedoch eine Zersplitterung der Professionalisierungsforschung in wenig miteinander kommunizierende Forschungslinien einher – diese Problematik lässt sich analog im kunstpädagogischen Diskurs wiederfinden. Unter den Erklärungsansätzen nehmen die verschiedenen Spielarten und Fortentwicklungen der struktur- und kompetenztheoretischen Argumentationen eine Schlüsselstellung ein, wobei sie jedoch eher Dissens als Konsens widerspiegeln. Infolgedessen wird das Konzept der Kontingenz im Beitrag als verbindendes Element herausgestellt und ein integrierender Vorschlag der Professionalisierung auf Basis von Kontingenzbearbeitung entwickelt.
Auf der Grundlage empirischer Rekonstruktion der Tiefenstrukturen der inklusionsbezogenen Professionalisierungsprozesse von angehenden Kunstlehrkräften (n=23) wird dazu ein Vorschlag unterbreitet, der auf zwei Pfeilern beruht: erstens der Analyse der Wahrnehmung und Deutung von Kontingenz im Professionalisierungsprozess und zweitens der Rekonstruktion von Mechanismen der Kontingenzbearbeitung. Das Moment der Kontingenzbearbeitung erweist sich zugleich als Ausgangs- wie Endpunkt entwicklungsgerichteter (kunst-)pädagogischer Professionalisierungsprozesse und als entscheidendes, anschlussfähiges Brückenglied zwischen den tradierten Argumentationslinien.
Die Qualifizierung von Sportlehrkräften für einen inklusiven Sportunterricht stellt eine zentrale Herausforderung für die sportdidaktische Lehre und Forschung dar. Obwohl die Bedeutung situationsspezifischer Fähigkeiten für die universitäre Sportlehrkräftebildung auch im Zusammenhang mit einem inklusiven Sportunterricht betont wird, fehlt es an Kompetenzmodellen, die die Ebene der situationsspezifischen Fähigkeiten berücksichtigen und somit eine Grundlage für Lehrkonzepte bilden können, die auf die Förderung situationsspezifischer Fähigkeiten für einen inklusiven Sportunterricht abzielen. Zudem mangelt es bestehenden Lehrkonzepten auch an Arbeits- und Organisationsformen, um gezielt situationsspezifische Fähigkeiten für einen inklusiven Sportunterricht anzusprechen. Schließlich existieren bisher keine Testverfahren, mit denen situationsspezifische Fähigkeiten für einen inklusiven Sportunterricht gemessen und darauf abzielende Lehrkonzepte evaluiert werden können. Ziel des vorliegenden Beitrags ist es demzufolge, konzeptionelle Überlegungen anzustellen, wie ein Kompetenzmodell für den inklusiven Sportunterricht generiert werden kann, welches die Ebene der situationsspezifischen Fähigkeiten berücksichtigt, wie ein Lehrformat angelegt sein müsste, um gezielt die situationsspezifischen Fähigkeiten für einen inklusiven Sportunterricht anzusprechen und wie diese situationsspezifischen Fähigkeiten gemessen werden können, um eine diesbezügliche Evaluation zu ermöglichen.
Die UN-Behindertenrechtskonvention bringt unter anderem eine „inklusionsorientierte Lehrer*innenbildung“ hervor, die Lehrkräfte für inklusive Settings kompetent machen soll. Damit das gelingen kann, müssen einerseits für inklusive Settings notwendige Kompetenzen identifiziert und operationalisiert werden, andererseits sind Erkenntnisse darüber notwendig, wie Studierende diese Kompetenzen entwickeln. Erst dann können wirksame Lehr- und Lernformate konzipiert werden. Inzwischen liegen Forschungsergebnisse zu inklusionsbezogenen Aufgaben- und Anforderungsprofilen vor, jedoch über vereinzelte Best-Practice-Beispiele kaum evidenzbasierte Erkenntnisse über Kompetenzentwicklungsprozesse in diesem Bereich oder gar Ableitungen für die Vermittlung solcher Kompetenzen. In diesem Artikel wird für die inklusionsbezogene Teilkompetenz „(multi-)professioneller Kooperationsfähigkeit und -bereitschaft“ ein kompetenzorientiertes Lehr- und Lernformat im Rahmen des Lehramtsstudiums an der Universität Paderborn konzipiert. In Anlehnung an das Constructive-Alignment-Modell werden dafür anhand eines Kompetenzmodells Learning Outcomes definiert, didaktisch-methodische Lernwege zugeordnet und Möglichkeiten einer Evidenzbasierung in Form von Prüfungen und Evaluation diskutiert. Dieser Artikel leistet so einen Beitrag zu einer inklusiven Hochschuldidaktik als Werkzeug, aber auch Voraussetzung und gleichzeitig Ergebnis, einer pädagogischen Fachlichkeit im Spannungsfeld inklusiver Realität.
Das MIKS-Konzept ist ein Konzept für die Schulentwicklungsarbeit mit Grundschulkollegien, die alle Sprachen ihrer Schüler*innen wertschätzen und für das Lernen nutzen wollen. Ziel ist die Professionalisierung und Qualifizierung des Personals und die Unterstützung von Schulentwicklungsprozessen. Die Grundschulkollegien entwickeln und erproben eigene Praxisvorhaben zum Einbezug von Mehrsprachigkeit, implementieren sie in die regulären Schul- und Unterrichtsabläufe und gestalten so die Schulentwicklung im Handlungsfeld Mehrsprachigkeit selbst.
Das MIKS-Konzept enthält Vorgaben über den Ablauf und die Methoden der Qualifizierung und Schulbegleitung sowie über die Inhalte, die während der Qualifizierung vermittelt und bearbeitet werden. Das MIKS-Konzept wurde 2013-2016 entwickelt und in drei Grundschulen erprobt. Im Rahmen einer Dissemination wurde das Konzept 2016-2019 durch Multiplikator*innen in die Breite getragen und an weiteren 17 Grundschulen durchgeführt. Der Artikel fokussiert zunächst die theoretische Fundierung und Konzeptionalisierung von MIKS. Im zweiten Teil werden die Inhalte, die Anlage und der Ablauf der Qualifizierung erstmals ausführlich vorgestellt und durch Einblicke in bisherige Erfahrungen illustriert.
Im Rahmen des ERASMUS+-Aktionsforschungsprojektes SHARMED wurden didaktische Werkzeuge entwickelt, um Lehrende darin zu unterstützen, ihrem Auftrag in Zeiten der Inklusion gerecht zu werden. Dabei will SHARMED zu einem Perspektivenwechsel beitragen, bei dem die Diversität der Schüler*innen nicht als eine Herausforderung gesehen wird, die bewältigt werden muss, damit die Schüler*innen bestimmte Kompetenzen erwerben und Inhalte aufnehmen können. SHARMED legt den Fokus stattdessen auf das viel zu selten wahrgenommene, kommunizierte und ausgelebte Bereicherungspotenzial, das in dieser Diversität steckt und lädt ein, Raum dafür zu schaffen. SHARMED setzt auf die dialogische Haltung, wodurch Lernende zu Protagonist*innen des Lernprozesses werden, und ihn aktiv mitgestalten. Um die dazu notwendige Agency der Schüler*innen zu fördern, wurden sowohl die Techniken der dialogischen Prozessbegleitung erforscht, als auch die folgende Methode konzipiert und getestet: Anhand eigener, zu diesem Zweck selbst ausgewählter und mitgebrachter Bilder haben Schüler*innen eigene Erinnerungen erzählt, welche dann als Ausgangspunkt für Dialoge genutzt wurden. In diesem Artikel wird ein solcher Austauschprozess konversationsanalytisch untersucht und so gezeigt, wie bestimmte, die pädagogische Praxis prägende kommunikative Handlungen der Inklusion entgegenstehen. Gleichzeitig wird auch eine konkrete Vorstellung von der konversationsanalytischen Forschungsarbeit und den Materialien vermittelt, die zum Zweck der Aus- und Weiterbildung von Lehrkräften frei zur Verfügung stehen. Die Nutzung der Aufnahmen und Transkriptionen der Dialoge ermöglicht es Lehrenden, sich über die Auswirkung kommunikativer Handlungen auf den pädagogischen Prozess bewusst zu werden. Darüber hinaus geben die Materialien einerseits Impulse zur Selbstreflexion und vermitteln andererseits Wissen und konkrete Ideen, um die Arbeit in Klassenzimmer inklusiver zu gestalten.
In der vorliegenden Evaluationsstudie wurde die Wirkung der Lehre des universitären Lehrprojektes ‚Expertise und Kooperation für eine Basisqualifikation Inklusion‘ (EKBI) auf inklusionsbezogene Einstellungen, Haltungen und Selbstwirksamkeitserwartungen untersucht. Diese Lehre schloss jeweils Lehrtandems aus mindestens zwei Dozierenden sowie interdisziplinäre Kooperationen ein. Die Teilnehmenden der Kurse (Experimentalgruppe, n = 101) sowie Studierende, die nicht daran teilgenommen hatten (Kontrollgruppe, n = 54), wurden vor und nach dem Semester, in dem die Lehre stattfand, mittels Fragebogen untersucht. Bei der Entwicklung allgemeinerer Einstellungen, Haltungen und Bedenken konnte kein signifikanter Effekt der Zeit in Abhängigkeit von der Gruppe verzeichnet werden. Es zeigte sich aber, dass sowohl die spezifischen unterrichtsbezogenen Einstellungen als auch die subjektiv eingeschätzte Lehrer*innenwirksamkeit in Bezug auf Inklusion in der Experimentalgruppe stärker anstiegen als in der Kontrollgruppe. Es ist zu schlussfolgern, dass die Tandemlehre mindestens kurzfristig wirksam in Bezug auf eine proinklusive Entwicklung ist. Zukünftige Studien könnten die Nachhaltigkeit der Effekte sowie ihren Praxistransfer untersuchen.
Ausgangspunkt ist die Beobachtung, dass sich empirisch eine inklusive Schule oftmals als eine differenzierte und differenzierende Schule darstellt. Dies bezieht sich etwa auf die Ausdifferenzierung der ‚heterogenen Lehrgruppe‘. Im zeitgemäßen inklusiven Unterricht sind allgemein- und sonderpädagogische Lehrkräfte ebenso wie Schulbegleitungen anwesend. Auch Sozialpädagog*innen und Therapeut*innen gehören vermehrt zum Schulalltag.
Vorliegende Studien dokumentieren, dass die Gestaltung inklusiver Schulen bzw. von inklusivem Unterricht von nicht intendierten Effekten und Widerständigkeiten der Akteur*innen begleitet ist. Im Beitrag werden – basierend auf der ProFiS-Studie – zwei Herausforderungen von inklusiven Schulen in den Mittelpunkt gerückt. Zum einen wird auf das Spannungsfeld von Professionalisierung und Deprofessionalisierung eingegangen. Zum anderen stellt das Verhältnis von Kategorisierung und Dekategorisierung eine Herausforderung dar. Beide Relationen sind als Spannungsverhältnisse untereinander und zueinander zu denken, die sich nicht ‚einfach‘ in eine Richtung auflösen lassen, sondern die in ihrer Widersprüchlichkeit gerade konstitutiv für die Praxis ‚inklusiver Schulen‘ wirken. Zentrale Frage des Beitrags ist, wie dieses komplexe Spannungsverhältnis der wechselseitigen Bezogenheit de/kategorisierender und de/professionalisierender Prozesse in professionellen Aktivitäten hervorgebracht wird.
Die Gestaltung eines inklusiven Mathematikunterrichts, der allen Schüler*innen Zugänge zur Mathematik eröffnet, wirft Fragen hinsichtlich einer fachbezogenen Aus- und Weiterbildung von Lehrpersonen auf. Im vorliegenden Beitrag wird eine Qualifizierungsmaßnahme vorgestellt, die darauf zielt, angehende Lehrkräfte für die Anforderungen und Herausforderungen eines inklusiven Mathematikunterrichts zu sensibilisieren sowie diesbezügliche Kritik- und Analysefähigkeiten zu entwickeln. Es wird untersucht, welche Potenziale die Analyse von Praxismaterialien in Bezug auf ihre Eignung für inklusiven Mathematikunterricht bietet. Die Datengrundlage bilden offene Fragebögen, in denen Studierende einen auf Inklusion bezogenen Praxisbeitrag reflektieren. Die Auswertung der Fragebögen erfolgt anhand einer qualitativen Inhaltsanalyse nach Mayring (2010). Auf Basis einer theoretischen Konzeptionalisierung wird somit eine empirisch erprobte Möglichkeit der Qualifizierung in der ersten Phase der Lehramtsausbildung vor- und zur Diskussion gestellt.
Die Zielsetzung der vorliegenden Studie liegt darin, Bedarfe für eine Qualifizierung und Professionalisierung von Lehrkräften in inklusiven Schulen zu identifizieren. Dabei stehen sowohl spezifische Lehrer*innengruppen als auch konkrete Inhalte bzw. Anforderungen von Inklusion im Fokus. Den Rahmen hierfür bilden Schulen mit dem Schulprofil Inklusion (N = 471 Lehrkräfte aus 49 Schulen). Mittels hierarchischer Clusteranalyse (Überprüfung durch Diskriminanzanalyse) werden Gruppen von Lehrer*innen gebildet. Diese basieren darauf, wie die Lehrkräfte berichten, die Anforderungen von Inklusion (adaptive Unterrichtsgestaltung, Förderplanung, Schulkonzeptentwicklung, innerschulische und externe Kooperation) umzusetzen. Die Betrachtung der vier Cluster zeigt eine Gruppe von Lehrer*innen, die die Anforderungen inklusiver Bildung durchwegs am intensivsten realisiert. Diese machen deutlich, dass nicht für alle Lehrenden gleichermaßen die Notwendigkeit einer Qualifizierung besteht. Gleiches gilt für die Konzeption einheitlicher Maßnahmen, denn für alle Lehrkräfte identische Maßnahmen werden der Diversität der geäußerten Einschätzungen nicht gerecht. Zielführender erscheinen mikrokontextuell orientierte Maßnahmen, die auf bestimmte Lehrkräfte und Anforderungen zugeschnitten sind. Solche Bedarfe zielen einerseits auf die Lehrer*innen ab, die die Anforderungen von Inklusion übergreifend als am geringsten verwirklicht einschätzen sowie diejenigen, die bestimmte Anforderungen nicht umsetzen. Für diese sind Gründe und folgend Maßnahmen zu diskutieren, warum die Umsetzung durchwegs eine so negative Einschätzung erfährt. Maßnahmen im Kontext von Schulentwicklung oder Supervision erscheinen zielführend. Spezifische Qualifizierungsbedarfe bestehen vor allem hinsichtlich innerschulischer Zusammenarbeit, die sich clusterübergreifend als eher gering ausgeprägt erweist.
Der vorliegende Beitrag beschreibt den theoretischen Hintergrund und das Design des Projekts GLUE (Gemeinsame Lern-Umgebungen Entwickeln). Die Konzeption der Gemeinsamen Lern-Umgebungen zielt im Sinne der Prinzipien des ‚Universal Design of Learning‘ (Hall, Meyer & Rose, 2012) darauf ab, den inklusiven Mathematikunterricht von einem gemeinsamen Lerngegenstand aus zu denken, der einerseits Zugänglichkeit für alle Lernenden schafft und andererseits Unterstützungsmaßnahmen auf unterschiedlichen Niveaustufen zulässt.
Das Projekt geht der Frage nach, wie sich die Kompetenzentwicklung von berufserfahrenen Lehrkräften der allgemeinen Schule und für sonderpädagogische Förderung durch Fortbildungsangebote zum inklusiven Mathematikunterricht wirksam unterstützen lässt. Hierzu wurde ein Blended-Learning-Angebot zur Entwicklung gemeinsamer Lernumgebungen für alle Kinder einer Lerngruppe erarbeitet, die sich auch von mehreren Lehrkräften gemeinsam entwickeln lassen.
In diesem Beitrag werden der theoretische Hintergrund, die Konzeption und die methodische Anlage des Projekts vorgestellt. Kapitel 1 befasst sich mit Differenzieren und Fördern im Mathematikunterricht der Primarstufe, Kapitel 2 diskutiert zentrale Befunde zur Wirksamkeit von Lehrerfortbildungsmaßnahmen sowie zu Blended-Learning-Angeboten. Im dritten Kapitel werden auf dieser Grundlage die Ziele des Projekts (Kap. 3.1), die Inhalte und die Struktur des Fortbildungsangebots (Kap., 3.2), die Forschungsfragen (Kap. 3.3) und das Design der Interventionsstudie dargestellt (Kap. 3.4). Die Wirksamkeit wird in einem ausbalancierten Prä-Post-Follow-Up-Test-Design im Vergleich zu unbegleiteten Online-Angeboten evaluiert, die Ergebnisse sollen in einer Folgepublikation kommuniziert werden.
Historic amphibian settlements in the northwestern Nile delta - a geoarchaeological perspective

No concise picture of the archaeological and palaeoecological evolution can be drawn for the northwestern Nile delta, and archaeological records show significant population dynamics that still need explanation and spur the need for further palaeoenvironmental research. This study delivers a set of new methods especially in the fields of remote sensing and data analytics that can be regarded as important milestones and foundations for further palaeoenvironmental research in the area. Additionally, it shows new insights for individual time slices.
This geoarchaeological project is a cooperation with the archaeological excavations of the German Archaeological Institute (DAI) in Buto and Kom el’Gir. It expands the work of Wunderlich (1989) which laid important foundations in understanding the origin of the initial landscape that was later colonized in different cultural stages showing different dynamics, settlement intensities and even long phases of abandonment or breaks in between. This forms the starting point for relating the population dynamics of the different cultural phases reaching from Predynastic (prior to 3150 before Christ) up to the Greco-Roman era (~anno Domini 650) to the environmental history and events that occurred in the area. It is very likely that environmental changes such as the shifting of major water routes, inundation or paludification of larger areas or other environmental events affected settlements and human life in the area.
In the fields of remote sensing new methods are presented to complete information on the location of ancient settlements, and complex workflows are developed that allow the tracing of subsurface structures via indirect analysis of vegetation growth in larger time series data. It was verified that a relationship exists between vegetation performance, the appearance of archaeologic material in the topsoil, and the location of former Nile river branches.
Together with a new high resolution digital elevation model (DEM) based on TanDEM-X data, new interpretations with a high spatial significance are possible. For individual time slices, namely the Late Dynastic and Greco-Roman era, this work delivers a detailed landscape description suggesting a finely ramified subdelta, with all settlements placed on alluvial levees. This explains the massive increase in settlements in the Ptolemaic, Roman and in particular late Roman periods (4th century before Christ – anno Domini 7th century).
We sampled the Nile delta clays together with the channels and the material of the archaeologic excavations in vibracores and profile walls. This geologic inspection of the subsurface together with geochemical results from a handheld portable X-ray fluorescence device (pXRF) allowed new interpretations of the landscape and environmental history. For example, we used geochemical data to distinguish between artificial and natural channels as a measure for the anthropogenic influence, a proxy for past environmental characteristics and lastly as a basis for a new dating method. Many of the channels, for instance, were dated by our own 14C datings, comparisons with the previous work ofWunderlich (1989) and application of new dating approach based on machine learning with artificial neural networks. Additionally, we run a full methodological approach, and examine the applicability of pXRF methods in general, and test the quality of the data to detect distinct geochemical differences between the main settlement phases with advanced methods in data analytics. The dating is based, for example, on the training of artificial neural networks with pXRF data from archaeological material of well-dated context to date test data of cultural layers within the vibracores. With this method the homogeneous Nile alluvium, cultural layers and channels can be dated roughly and, as a result, fundamental changes in the landscape can be linked with the settlement history of Buto and neighboring tells.
Was verstehen Lehramtsstudierende unter Inklusion? Eine Untersuchung subjektiver Definitionen

Der Begriff der Inklusion ist ein mehrdimensionales Konstrukt, das sowohl in Theorie als auch in Praxis unterschiedlich definiert ist. Für angehende Lehrkräfte bedeutet dies, dass sie sich mit unterschiedlichen Konnotationen dieses Begriffs auseinandersetzen müssen und dadurch möglicherweise eigene Begriffsdefinitionen vornehmen. Im Mittelpunkt dieses Beitrages steht die Frage nach den subjektiven Begriffsdefinitionen zum Begriff Inklusion. Im Rahmen einer schriftlichen Befragung wurden 290 Lehramtsstudierende mit Hilfe einer offenen Fragestellung zu ihrem Inklusionsverständnis befragt. Das Material wurde mittels qualitativer Inhaltsanalyse ausgewertet. Die Ergebnisse verweisen auf ein differenziertes Begriffsverständnis angehender Lehrkräfte. Es können vier Verständnistypen unterschieden werden, ausgehend von einem sehr engen bis hin zu einem sehr weiten und offenen Begriffsverständnis.
Schulbegleitung ist ein zunehmend relevantes Tätigkeitsfeld bei der Realisierung inklusiver Bildungsangebote. Kritisch diskutiert wird die Einschlägigkeit der Qualifikation der eingesetzten Personen. Aktuell sind etwa 50% der als Schulbegleiter/innen Beschäftigten pädagogisch, therapeutisch oder medizinisch-pflegerisch qualifiziert. Bislang ist wenig darüber bekannt, welchen Tätigkeiten Schulbegleiter/innen genau nachgehen und wie die Zusammenarbeit mit Lehrkräften eingeschätzt wird. In der vorliegenden Untersuchung werden deshalb Tätigkeitsprofile und Selbsteinschätzungen zur Qualität der Zusammenarbeit mit Lehrkräften bei Fachkräften und Nicht-Fachkräften vergleichend in den Blick genommen. Es wurden in einer Fragebogenbefragung n = 61 Schulbegleiter/innen befragt. Die Ergebnisse zeigen, dass wider Erwarten die Fachkräfte mit pädagogischer Qualifikation eher im Bereich der lebenspraktischen Unterstützung als im Bereich der Emotions- und Verhaltensregulation tätig sind. Die Zusammenarbeit mit Lehrkräften wird von Fachkräften und Nicht-Fachkräften gleichermaßen positiv bewertet. Es gibt Hinweise darauf, dass die Bewertung der Zusammenarbeit mit Lehrkräften von spezifischen Tätigkeitsprofilen abhängt.

This thesis discusses important questions of the beam dynamics in the proton-lead operation in the Large Hadron Collider (LHC) at CERN in Geneva. In two time blocks of several weeks in the years 2013 and 2016, proton-lead collisions have so far been successfully generated in the LHC and used by the experiments at the LHC. One reason for doubts regarding the successful operation in proton-lead configuration was the fact that the beams have to be accelerated with different revolution frequencies. There is long-range repulsion between the beams, since both beams share the beam chamber around the interaction points. Because of the different revolution frequencies, the positions of the interaction between the beams shift each revolution. This can lead to resonant excitation and to an increase in the transverse beam emittance, as was observed in the Relativistic Heavy-Ion Collider (RHIC). In this thesis, simulations for the LHC, RHIC and the High-Luminosity Large Hadron Collider (HL-LHC) are performed with a new model. The results for RHIC show relative growth rates of the emittances of the gold beam in gold-deuteron operation in RHIC from 0.1 %/s to 1.5 %/s. Growth rates of this magnitude were observed experimentally in RHIC. Simulations for the LHC show no significant increase of the emittance of the lead beam for different intensities of the counter-rotating beam. The simulation results confirm the measured stability of the beams in the LHC and the issue of strongly increasing emittances in RHIC is reproduced. Also, no significant increase of the emittance is predicted for the Future Circular Collider (FCC) and the HL-LHC.
Using a frequency-map analysis, this work verifies whether the interaction of the lead beam with the much smaller proton beam in the proton-lead operation of the LHC leads to diffusion within the lead beam. Experiences at HERA at DESY in Hamburg and at SppS at CERN have shown that the lifetime of the larger beam can rapidly decrease under certain circumstances. The results of the simulation show no chaotic dynamics near the beam centre of the lead beam. This result is supported by experimental observation.
A program code has been developed which calculates the beam evolution in the LHC by means of coupled differential equations. This study shows that the growth rates of the lead beam due to intra-beam scattering is overestimated and that particle bunches of the lead beam lose more intensity than assumed in the model. The analysis also shows that bunches colliding in a detector suffer additional losses that increase with decreasing crossing angle at the interaction point.
In this work, 2016 data from beam-loss monitors in combination with the luminosity and the loss rate of the beam intensity are used to determine the cross section of proton-lead collisions at the center-of-mass energy of 8.16 TeV. Beam-loss monitors that mainly detect beam losses that are not caused by the collision process itself are used to determine the total cross section via regression. An analysis of the data recorded in 2016 at the center-of-mass energy of 8.16 TeV resulted in a total cross section of σ=(2.32±0.01(stat.)±0.20(sys.)) b. This corresponds approximately to a hadronic cross section of σ(had)=(2.24±0.01(stat.)±0.21(sys.)) b. This value deviates only by 5.7 % from the theoretical value σ(had)=(2.12±0.01) b.
The simulation code for determining the beam evolution is also used to estimate the integrated luminosity of a future one-month run with proton-lead collisions. The result of the study shows that in the future the luminosity in the ATLAS and CMS experiments will increase from 15/nb per day in 2016 to 30/nb per day, which is a significant increase in terms of the performance. This operation, however, requires the use of the TCL collimators to protect the dispersion suppressors at ATLAS and CMS from collision fragments.
This work also gives an outlook on the expected luminosity production in proton-nucleus operation using ion species lighter than lead ions. For example, a change from proton-lead to proton-argon collisions would increase the integrated luminosity from monthly 0.8/nb to 9.4/nb in ATLAS and CMS. This is an increase of one order of magnitude and approximately a doubling of the integrated nucleon-nucleon luminosity. There may be a test operation with proton-oxygen collisions in 2023, which will last only a few days and will be operated with a low luminosity. The LHCf experiment (LHCb experiment) would achieve the desired integrated luminosity of 1.5/nb (2/nb) within 70h (35h) beam time.
Stickstoff (NO), Kohlenmonoxid (CO) und Schwefelwasserstoff (H2S) gehören zur Gruppe der Gasotransmitter. Dabei handelt es sich um kleine gasförmige Signalmoleküle, welche innerhalb des Körpers gebildet werden und dort wichtige physiologische Funktionen bei der Regulation der Apoptose, der Proliferation, der Entzündungsreaktion und der Genexpression übernehmen. Aufgrund ihrer Membranpermeabilität ist die Wirkung der Gasotransmitter nicht an die Interaktion mit spezifischen membranständigen Rezeptoren gebundenen. Je nach Organ, Gewebe und Konzentration können diese Mediatoren unterschiedliche Prozesse beeinflussen und teils sogar gegenteilige Wirkungen hervorrufen.H2S beispielsweise kann im Verlauf der Leukozytenadhäsion im Epithelium anti-inflammatorisch, bei Brandwunden oder rheumatischen Erkrankungen jedoch pro-inflammatorisch wirken. Im Kreislaufsystem hingegen bewirkt H2S durch die Aktivierung von ATP-abhängigen K+-Kanälen und die damit zusammenhängende Vasorelaxion der glatten Muskelzellen einen eindeutig protektiven Effekt.
H2S kann je nach Substrat und Zelltyp durch eines von 3 Enzymen gebildet werden. Die Cystathionin-γ-Lyase (CSE) und die Cystathionin-β-Synthase (CBS) nutzen L-Cystein als Substrat für die Synthese von H2S. Das dritte H2S-bildende Enzym, die 3-Mercaptopyruvate Sulfurtransferase (3-MST) verwendet α-Ketoglutarat als Substrat, welches zuvor von der Cystein-Aminotransferse (CAT) aus L-Cystein synthetisiert wurde. Während die beiden Enzyme CSE und CBS im Zytosol der Zelle zu finden sind, ist die 3-MST hauptsächlich in den Mitochondrien der Zelle zu finden. Im Gegensatz zur CBS, welche eher ein konstitutiv exprimiertes Protein ist, wird die Expression der CSE auf der Transkriptionsebene durch u.a. Entzündungsmediatoren wie TNF-α oder Wachstumsfaktoren wie PDGF-BB induziert.
Ein Ziel der Arbeit war es, die Wirkung von H2S bei der Wundheilung, bei entzündlichen glomerulären Erkrankungen der Niere und beim Schlaganfall zu untersuchen. Für diesephänotypische Analysen stand ein Knockoutmodell für die CSE zur Verfügung.
Zudem wurden in dieser Arbeit Untersuchungen mit einem Knockoutmodell für das zytoskeletäre Protein durchgeführt. Bei Clp36 (PDLIM1) handelt es sich um ein PDLIM-Protein (PDZ and LIM domain protein),welches durch die Gasotransmitter NO und H2S auf transkriptioneller und translationaler Ebene reguliert wird ist und aufgrund seiner Assoziation mit dem Zytoskelett dynamische Vorgänge der Zelle moduliert. Es ist bereits bekannt, dass Clp36 ein negativer Regulator des Glykoprotein VI (GPVI), welches eine wichtige Rolle bei der Aktivierung von Thrombozyten spielt, ist.
Beide Knockoutmodelle wurden in murinen Mesangiumzellen der Niere und in Krankheitsmodellen der Haut (kutane Wundheilung)und des Gehirns (Schlaganfall mit dem MCAO-Modell) analysiert.
Neben nicht signifikanten Effekten im MCAO-Modell, konnten sowohl Effekte des CSE-, als auch des CLP36-KOs auf die Migration und Proliferation und im Falle der CSE auch auf die Adhäsion der murinen Mesangiumzellen beobachtet werden. Die Depletion von Clp36 führte zu einer Verringerung der Migrations- und einer Erhöhung der Proliferationsrate, wohingegen die Depletion der CSE zu einer Erhöhung der Migrations-, Proliferations- und Adhäsionsrate führte. Die vielversprechendsten Ergebnisse konnten im Tiermodell der kutanen Wundheilung generiert werden. Untersucht wurde die Expression der H2S-produzierenden Enzyme CSE, CBS und 3-MST. Alle drei Enzyme zeigten im Tiermodell keine transkriptionelle Regulation und blieben auch während der akuten Entzündungsphase und der proliferativen Phase der Wundheilung unverändert. Es konnte jedoch gezeigt werden, dass die Expression der CSE in der späten Phase der Wundheilung signifikant anstieg, wenn die Proliferation innerhalb des Granulationsgewebes und der Neoepidermis geringer wurde. Die Vermutung, dass H2S in dieser Phase eine wichtige Rolle spielt, konnte durch die Analyse der CSE-KO Mäuse bekräftigt werden, da dort der Verlust der CSE offenbar durch die CBS kompensiert wurde.
In immunhistochemischen Untersuchungen konnten insbesondere follikuläre Keratinozyten der Neo-Epidemis als Quelle der CSE-Expression identifiziert werden. Durch in-vitro Studien auf mRNA und Proteinebene in HaCaT Zellen wurde gezeigt, dass H2S die Keratinozyten-Differenzierung beeinflusst. Der langsam freisetzendeH2S-Donor GYY4137 konnte in humanen Keratinozyten zu einer signifikanten Erhöhung der Ca2+- induzierten Expression der frühen Keratinozyten-Differenzierungsmarker Cytokeratin 10 (CK10) und Involucrin (IVN) beitragen.
Im Laufe dieser Arbeit konnte der molekulare Mechanismus hinter diesen Beobachtungen noch nicht geklärt werden.
Durch weitere Versuche meiner Arbeitsgruppe konnte jedoch gezeigt werden, dass die GYY4137-abhängige Induktion der CK10-Expression durch eine verstärkte Bindung der RNA-Polymerase II an den CK10 Promotor zustande kommt.
The events of 1968/69 initiated a dispute between Adorno and Marcuse over the (alleged) separation of theory and praxis. While Marcuse “stood at the barricades” Adorno sought recluse in the “ivory tower”. Marcuse and German students perceived Adorno’s move as departure from fundamental postulates of critical theory as laid down in Horkheimer’s 1937 essay. Adorno died amidst the process of clarifying his differences with Marcuse and thus the “unlimited discussions” between the two remain unfinished. This paper sets to examine how both Marcuse and Adorno remained dedicated to the unity of theory and praxis, albeit in different ways. I argue that Adorno did not separate theory and praxis; instead, he perceived the gap between critical theory and concrete historical situation. Adorno rejected simple and unreflective translation of theory into praxis. Hence his attempt to recalibrate critical theory. Marcuse’s and Adorno’s differences lie in their different evaluation of the student movement and this (mis)evaluation was context related. My second argument is that Marcuse/Adorno disagreement is partly caused by the absence of the two from the concrete historical context.
In this review, we focus on the ubiquitination process within the endoplasmic reticulum associated protein degradation (ERAD) pathway. Approximately one third of all synthesized proteins in a cell are channeled into the endoplasmic reticulum (ER) lumen or are incorporated into the ER membrane. Since all newly synthesized proteins enter the ER in an unfolded manner, folding must occur within the ER lumen or co-translationally, rendering misfolding events a serious threat. To prevent the accumulation of misfolded protein in the ER, proteins that fail the quality control undergo retrotranslocation into the cytosol where they proceed with ubiquitination and degradation. The wide variety of misfolded targets requires on the one hand a promiscuity of the ubiquitination process and on the other hand a fast and highly processive mechanism. We present the various ERAD components involved in the ubiquitination process including the different E2 conjugating enzymes, E3 ligases, and E4 factors. The resulting K48-linked and K11-linked ubiquitin chains do not only represent a signal for degradation by the proteasome but are also recognized by the AAA+ ATPase Cdc48 and get in the process of retrotranslocation modified by enzymes bound to Cdc48. Lastly we discuss the conformations adopted in particular by K48-linked ubiquitin chains and their importance for degradation.
Background: Native T1 may be a sensitive, contrast-free, non-invasive cardiovascular magnetic resonance (CMR) marker of myocardial tissue changes in patients with pulmonary artery hypertension. However, the diagnostic and prognostic value of native T1 mapping in this patient group has not been fully explored. The aim of this work was to determine whether elevation of native T1 in myocardial tissue in pulmonary hypertension: (a) varies according to pulmonary hypertension subtype; (b) has prognostic value and (c) is associated with ventricular function and interaction.
Methods: Data were retrospectively collected from a total of 490 consecutive patients during their clinical 1.5 T CMR assessment at a pulmonary hypertension referral centre in 2015. Three hundred sixty-nine patients had pulmonary hypertension [58 ± 15 years; 66% female], an additional 39 had pulmonary hypertension due to left heart disease [68 ± 13 years; 60% female], 82 patients did not have pulmonary hypertension [55 ± 18; 68% female]. Twenty five healthy subjects were also recruited [58 ±4 years); 51% female]. T1 mapping was performed with a MOdified Look-Locker Inversion Recovery (MOLLI) sequence. T1 prognostic value in patients with pulmonary arterial hypertension was assessed using multivariate Cox proportional hazards regression analysis.
Results: Patients with pulmonary artery hypertension had elevated T1 in the right ventricular (RV) insertion point (pulmonary hypertension patients: T1 = 1060 ± 90 ms; No pulmonary hypertension patients: T1 = 1020 ± 80 ms p < 0.001; healthy subjects T1 = 940 ± 50 ms p < 0.001) with no significant difference between the major pulmonary hypertension subtypes. The RV insertion point was the most successful T1 region for discriminating patients with pulmonary hypertension from healthy subjects (area under the curve = 0.863) however it could not accurately discriminate between patients with and without pulmonary hypertension (area under the curve = 0.654). T1 metrics did not contribute to prediction of overall mortality (septal: p = 0.552; RV insertion point: p = 0.688; left ventricular free wall: p = 0.258). Systolic interventricular septal angle was a significant predictor of T1 in patients with pulmonary hypertension (p < 0.001).
Conclusions: Elevated myocardial native T1 was found to a similar extent in pulmonary hypertension patient subgroups and is independently associated with increased interventricular septal angle. Native T1 mapping may not be of additive value in the diagnostic or prognostic evaluation of patients with pulmonary artery hypertension.
Rechtsaspekte des Forschungsdatenmanagements werfen nach wie vor viele Fragen für Forschende und Nachnutzende auf: Wer darf welche Daten zu welchen Zwecken erheben und verarbeiten? Wem können die Daten "zugeordnet" werden? Wer darf entscheiden, ob und wann sie veröffentlicht werden? Welche Lizenzen sind geeignet?
Um den Unsicherheiten im rechtlichen Umgang mit Forschungsdaten zu begegnen, hat HeFDI auf Basis des Gutachtens des DataJus-Projekts (https://tu-dresden.de/gsw/jura/igetem/jfbimd13/ressourcen/dateien/dateien/DataJus/DataJus_Zusammenfassung_Gutachten_12-07-18.pdf?lang=de) die hier publizierte Handreichung zusammengestellt. Die Handreichung dient dazu, Forschenden und Forschungsdatenmanager*innen einen gut verständlichen Zugang zu Rechtsfragen hinsichtlich Forschungsdaten zu ermöglichen. Das anschaulich konzipierte Material kann sowohl in Schulungen zum Datenmanagement als auch in konkreten Beratungsprozessen eingesetzt werden.
In einem ersten Schritt hat HeFDI die Themenfelder (I) Urheber- und Leistungsschutzrechte an Forschungsdaten sowie (II) Entscheidungsbefugnis über den Umgang mit Forschungsdaten in der hier publizierten Handreichung zusammengefasst.
Das vorliegende Material umfasst zum einen eine animierte Präsentation in editierbarem Format. Gleichzeitig stellt HeFDI den Inhalt der Präsentation sowohl editierbar als auch im PDF-Format als Druckvorlage für ein Handout o. Ä. im DIN A4-Format zur Verfügung.
Brain aging is one of the major risk factors for the development of several neurodegenerative diseases. Therefore, mitochondrial dysfunction plays an important role in processes of both, brain aging and neurodegeneration. Aged mice including NMRI mice are established model organisms to study physiological and molecular mechanisms of brain aging. However, longitudinal data evaluated in one cohort are rare but are important to understand the aging process of the brain throughout life, especially since pathological changes early in life might pave the way to neurodegeneration in advanced age. To assess the longitudinal course of brain aging, we used a cohort of female NMRI mice and measured brain mitochondrial function, cognitive performance, and molecular markers every 6 months until mice reached the age of 24 months. Furthermore, we measured citrate synthase activity and respiration of isolated brain mitochondria. Mice at the age of three months served as young controls. At six months of age, mitochondria-related genes (complex IV, creb-1, β-AMPK, and Tfam) were significantly elevated. Brain ATP levels were significantly reduced at an age of 18 months while mitochondria respiration was already reduced in middle-aged mice which is in accordance with the monitored impairments in cognitive tests. mRNA expression of genes involved in mitochondrial biogenesis (cAMP response element-binding protein 1 (creb-1), peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC1-α), nuclear respiratory factor-1 (Nrf-1), mitochondrial transcription factor A (Tfam), growth-associated protein 43 (GAP43), and synaptophysin 1 (SYP1)) and the antioxidative defense system (catalase (Cat) and superoxide dismutase 2 (SOD2)) was measured and showed significantly decreased expression patterns in the brain starting at an age of 18 months. BDNF expression reached, a maximum after 6 months. On the basis of longitudinal data, our results demonstrate a close connection between the age-related decline of cognitive performance, energy metabolism, and mitochondrial biogenesis during the physiological brain aging process.
Kinetics of circulating endothelial progenitor cells in patients undergoing carotid artery surgery

Aim: Endothelial progenitor cells (EPCs) are primitive cells found in the bone marrow and peripheral blood (PB). In particular, the potential of EPCs to differentiate into mature endothelial cells remains of high interest for clinical applications such as bio-functionalized patches for autologous seeding after implantation. The objective of this study was to determine EPCs’ kinetics in patients undergoing carotid artery thromboendarterectomy (CTEA) and patch angioplasty.
Methods: Twenty CTEA patients were included (15 male, mean age 76 years). PB samples were taken at 1 day preoperatively, and at 1, 3, and 5 days postoperatively. Flow cytometric analysis was performed for CD34, CD133, KDR, and CD45. Expression of KDR, SDF-1α, and G-CSF was analyzed by means of enzyme-linked immunosorbent assay.
Results: Fluorescence-activated cell sorting analysis revealed 0.031%±0.016% (% of PB mononuclear cells) KDR+ cells and 0.052%±0.022% CD45-/CD34+/CD133+ cells, preoperatively. A 33% decrease of CD45–/CD34+/CD133+ cells was observed at day 1 after surgery. However, a relative number (compared to initial preoperative values) of CD45-/CD34+/CD133+ cells was found on day 3 (82%) and on day 5 (94%) postoperatively. More profound upregulated levels of CD45–CD34+/CD133+ cells were observed for diabetic (+47% compared to nondiabetic) and male (+38% compared to female) patients. No significant postoperative time-dependent differences were found in numbers of KDR+ cells and the concentrations of the cytokines KDR and G-CSF. However, the SDF-1α levels decreased significantly on day 1 postoperatively but returned to preoperative levels by day 3.
Conclusion: CTEA results in short-term downregulation of circulating EPCs and SDF-1α levels. Rapid return to baseline levels might indicate participation of EPCs in repair mechanisms following vascular injury.
