NADH:ubiquinone-oxidoreductase (complex I) is the largest membrane protein complex of the respiratory chain. Complex I couples electron transfer to vectorial proton translocation across the inner mitochondrial membrane. The L shaped structure of complex I is divided into a membrane arm and a matrix arm. Fourteen central subunits are conserved throughout species, while some 30 accessory subunits are typically found in eukaryotes. Complex I dysfunction is associated with mutations in the nuclear and mitochondrial genome, resulting in a broad spectrum of neuromuscular and neurodegenerative diseases. Accessory subunit NDUFS4 in the matrix arm is a hot spot for mutations causing Leigh or Leigh-like syndrome. In this review, we focus on accessory subunits of the matrix arm and discuss recent reports on the function of accessory subunit NDUFS4 and its interplay with NDUFS6, NDUFA12, and assembly factor NDUFAF2 in complex I assembly.
Typ 1 Diabetes mellitus (T1D), Hashimoto-Thyreoiditis (HT) und Morbus Addison (AD) sind autoimmunvermittelte Erkrankungen mit multifaktorieller und polygener Ätiologie. Die gemeinsamen prädisponierenden genetischen Merkmale (HLA Klasse II-Moleküle und CTLA-4) bedingen eine Störungen in der T-Zell-Aktivierung und Homöostase. Es mehren sich Hinweise, dass PTPN22 ein weiterer genereller Risikofaktor für Autoimmunität ist. PTPN22 kodiert für eine intrazelluläre Tyrosin-Phosphatase, die als Suppressor der T-Zell- Aktivierung wirk. In der vorliegenden Arbeit wurde die Verteilung des PTPN22 1858 C/T Polymorphismus in deutschen Patienten mit T1D (n = 220), AD (n = 121) und HT (n = 116) sowie gesunden Kontrollen (n = 239) untersucht. Hierbei konnte eine signifikante Assoziation der Genvariation und den Erkrankungen T1D und HT bestätigt werden. PTPN22 1858 T wurde häufiger bei Patienten mit T1D (19,3% vs. 11,3%; p = 0,001; OR für Allel T = 1,88, 95% CI [1,30-2,72]) und HT (18,1% vs. 11,3%; p = 0,0129; OR für Allel T = 1,74, 95% CI [1,12-2,69]) beobachtet. Erstmals konnte eine exklusive Assoziation zum weiblichen Geschlecht bei Patientinnen mit T1D beobachtet werden (p = 0,0001). Ein signifikanter Unterschied zwischen männlichen Typ 1 Diabetikern und Kontrollen war nicht nachweisbar. Für eine Assoziation von PTPN22 1858 C/T und AD fanden sich keine Hinweise. Zusammenfassend ist der Polymorphismus PTPN22 1858 C/T in der Pathogenese von HT und T1D in der deutschen Bevölkerung involviert, beim T1D durch einen geschlechtsspezifischen Mechanismus, der zu einer Risikoerhöhung im weiblichen Geschlecht führt. Diese Ergebnisse werden durch aktuelle Studien bestätigt. Der Vitamin D-Stoffwechsel ist ein weiteres System mit einem klar belegten Einfluss auf das Immunsystem. Die Gabe von aktivem Vitamin D zeigte im Tiermodell eine Protektion vor T1D. Der modulierende Einfluss auf T-Zell-Differenzierung und Aktivität von dendritischen Zellen mittels Zytokinausschüttung bewirkt ein Verschiebung des Gleichgewichts von T-Helferzellen in Richtung Th2-Zellen und regulatorischen T-Zellen. Zusätzlich belegen epidemiologische Daten eine Verbindung von Vitamin D Ernährungsstatus in der frühen Kindheit und der Prävalenz von T1D. Ein genetisches Risiko für T1D konnte für die folgenden Komponenten des Vitamin D-Systems identifiziert werden: Vitamin D bindendes Protein (DBP), CYP2R1, CYP27B1 und CYP24 sowie Vitamin D-Rezeptor (VDR). Ein weiteres Makromolekül mit essentieller Bedeutung für die Aufrechterhaltung des Vitamin D-Metabolismus ist Megalin. Der endozytotische Multiliganden-Rezeptor gehört zur Familie der low-density Lipoprotein (LDL)-Rezeptoren und wird hauptsächlich in den Zellen des proximalen Tubulus der Niere exprimiert, zusätzlich aber auch in anderen resorptiven Epithelien. Megalin liefert den größten Beitrag zur Protein-Aufnahme aus dem glomerulären Filtrat in die Zellen des proximalen Tubulus. Neben einer relativ unspezifischen Bindung vieler Proteine (z.B.: Albumin, ベ1- und ベ2-Mikroglobulin) bindet Megalin einige Liganden mit hoher Affinität. Hierzu zählen Hormone, Vitamin-bindende Proteine und Lipoproteine. Megalin gewinnt durch die zelluläre Aufnahme von Vitamin D im Komplex mit seinem Transportprotein DBP eine große Bedeutung im Vitamin D-Stoffwechsel. Die Endozytose ist notwendig um 25 OH D3 zu konservieren und um Zellen mit der Vorstufe zur Generierung von 1,25 (OH)2 D3, der biologisch aktiven Form von Vitamin D, zu versorgen. Weiterhin ist die Aufnahme möglicherweise direkt mit der Aktivierung des nukleären VDR verbunden, als Vorbereitung auf kommende Liganden In der vorliegenden Arbeit wurde erstmals die Bedeutung von genetischen Polymorphismen des Megalingens bei Patienten mit T1D und gesunden Kontrollen der deutschen Bevölkerung untersucht. Die Analyse von neun SNPs erbrachten interessante Erkenntnisse: Genvariationen von Megalin sind mit T1D assoziiert, wobei MegE36 A, ein SNP auf Exon 36, das Risiko einer Erkrankungsmanifestation erhöht. Allel »A« wurde häufiger bei Patienten mit T1D beobachtet (95,6% vs. 92,2%; OR für Allel A = 1,85, 95% CI [1,18-2,91]). In der geschlechtsgetrennten Analyse beschränkten sich die gefundenen Differenzen der Allelfrequenzen lediglich auf das männliche Geschlecht (p = 0,0014; OR für Allel A = 2,67, 95% CI [1,43 – 4,98]). Bei Frauen war kein signifikanter Unterschied nachweisbar. Da dieser exonische SNP still ist, d.h. durch einen synonymen Aminosäurenaustausch keinen Einfluss auf die Rezeptorfunktionalität hat, treten mögliche Effekte auf RNA-Ebene auf. Denkbar ist auch eine Kopplung mit einem benachbarten Marker. Drei weitere nicht synonyme exonische SNPs (MegE03 A/G, MegE66 A/G and MegE69 A/C) weisen eine grenzwertige Assoziation zu T1D auf und haben zugleich einen funktionellen Einfluss auf die Proteinstruktur. Bemerkenswerterweise zeigt MegE66, wie auch MegE36 A, lediglich bei Männern ein genetisches Risiko für T1D. Diese Geschlechtsspezifität wird möglicherweise durch eine Interaktion von Megalin mit Androgenen und Östrogenen in Verbindung mit dem Transportprotein (SHBG) begründet. Die Aufklärung des molekularen Mechanismus wird Gegenstand künftiger Studien sein.
Mehr Geld für BND und Verfassungsschutz, bessere Vernetzung der europäischen Datenbanken, die Schaffung eines Ein- und Ausreiseregisters, die Übermittlung von europäischen Fluggastdaten an die Sicherheitsbehörden der EU-Mitgliedsstaaten, mehr Videoüberwachung, Kennzeichenlesesysteme, Biometrie – und vieles mehr. Detaillierte Angaben dazu, ob die angepeilten Maßnahmen überhaupt wirksam sind, werden selten gegeben. Chromatin, RNA Polymerase, Potato Tuber Tissue, Aging Phenomenon The synthesis of RNA by chromatin-bound RNA polymerase (E.C. 2.7.7.6.) from white potato tubers proceeds at a low rate, which is enhanced after slicing the tissue, however. Concomitantly DNA template availability as measured with saturating amounts of Escherichia coli polymerase is diminished drastically. Nearest neighbor frequency analysis proved that the RNA synthesized on chromatin of intact tubers is different from that synthesized on chromatin of sliced tissue.
The RNA polymerase of white potato tubers is dependent on all four ribonucleoside triphos phates and a divalent metal ion such as Mg2+ or Mn2+ and totally inhibited by the presence of pyrophosphate. Actinomycin D blocks the formation of the RNA product, which could be shown to be a heteropolymer by nearest neighbour frequency technique. The Km of the chromatin-bound enzyme with regard to ATP, GTP, CTP and UTP was 5.1 X10-5 M, 1.6X10-5 M, 0.9X10-5 M and 0.45 X 10-5M/1 respectively, α-amanitin inhibits the overall activity to about 50%, which indicates the presence of equal amounts of polymerase I and polymerase If.
Differential derepression of the genome of potato tuber cells can be initiated by slicing the tissue into disks. The consequence of this procedure on the cells of the wound surface is dedifferentiation and cell division followed by redifferentiation to a suberized phellem cell. The drift of glucose-, glucose-1-phosphate-, glucose-6-phosphate-, fructose-6-phosphate- and 6-phospho-gluconatelevels has been determined in the derepressed tissue. With the exception of 6-phospho-gluconate all intermediates so far investigated showed a rise in concentration after derepression.
This is interpreted as a consequence of altered enzymic activities which were estimated for phosphoglucomutase, hexokinase, phosphoglucoisomerase, gluco-6-phosphate- and 6-phosphogluconatedehydrogenase. The two dehydrogenases were activated after derepression, the activation represented a de-novo-synthesis, as was demonstrated with the inhibitors Actidione (translation) and p-Fluorophenyl-alanine (protein synthesis in general). Hexokinase and phosphoglucoisomerase were not severely affected by cutting the tissue. Phosphoglucomutase was degrated rapidly, the degradation being dependent on protein synthesis. The importance of an enhanced activity of the pentose phosphate shunt for the stressed cell is emphasized and the possibility of an alteration in the osmotic pressure within the cell and especially in the nucleus — a primary consequence of wounding — as a cause of derepression in potato tuber cells is discussed.
Differential derepression of the genome of potato tuber cells causes the onset of a vigorous metabolic activity, which is initiated by rapid synthesis of different RNA species, various proteins and phospholipids. Consequently enhanced respiration and the build up of cell compartments such as ribosomes and mitochondria as well as the performance of cell divisions and suberization of new-formed cell walls occur. Although there is an activation of metabolism in general with a concomitant rise in concentration of most glycolytic metabolites — as was proved for fructose-1.6-diphosphate, dihydroxyacetone, glyceraldehade-3-phosphate, phosphoenolepyruvate and pyruvate — the activities of the corresponding enzymes do not reflect these uniform metabolic changes. Aldolase and in a pronounced manner enolase and glutamate — pyruvate — transaminase lower their activities suddenly after derepression. The activity of triosephosphateisomerase remains constant. In contrast phosphoglyceromutase, pyruvate kinase and to a lower extent malic enzyme enhance their action during the same time.
Without doubt, differential lowering and enhancing the activity of glycolytic chain constituents at the same time is an important regulatory mechanism of the cell. The activation represents de novo synthesis of the protein concerned whereas the inactivation depends largely on protein synthesis. This is clearly shown by experiments with inhibitors of protein synthesis.
It is proposed that this differential synthesis and degradation represent a “long-time-regulation” of enzymatic activity of the cell in contrast to the known “short-time-regulation” by feedback or competition.
One of the earliest consequences of slicing plant storage organs such as potato tubers into thin disks is the formation of polysomes, which in potato slices is complete after 9 hours and is dependent on transcription. Fresh disks do not incorporate 32P, 3H-uridine or 14C-leucine into their ribosomes, whereas ribosomes and polysomes of aged disks use these precursors effectively. This development can be completely blocked by actinomycin D. Among the different RNAs synthesized during aging is 28S- and 16S—rRNA, 5S—RNA, tRNA, and a component sedimenting around 15—18S with a base-composition different from 16S—rRNA, 5S- and 4S—RNA and which supports peptide formation in an in vitro incorporation system.
It is suggested that this compound represents mRNA, which is not available immediately after slicing the tissue. These findings are consistent with the view of a derepression phenomenon in sliced storage tissue.
Resting potato tuber tissue possesses only faint activity of the two dehydrogenases of the oxidative pentose phosphate cycle, glucose-6-phosphate- and 6-phosphogluconate dehydrogenase. Slicing of the tissue, however, greatly enhances the action of both enzymes. The slicing-induced increase in activity is a consequence of intensified action of at least 5 glucose-6-phosphate dehydrogenase isozymes and a more differentiated activation/inactivation of seven 6-phosphogluconate dehydrogenase isozymes.
Using density labelling and isopycnic equilibrium centrifugation it could be demonstrated, that the bulk of both enzymes appearing after slicing the tissue is the result of de novo synthesis rather than activation of pre-existing proenzymes.
Whereas ribosome preparations of freshly sliced potato disks do not show appreciable activity in an in-vitro amino acid incorporation system, aging of the tissue leads to a greatly enhanced incorporation activity which reaches its maximum 24 hours after slicing. If ribosomes from freshly excised disks are provided with polyuridylic acid, their activity in the incorporation of phenylalanine is increased about 8 fold.
Moreover, an RNA-fraction can be dissociated by EDTA from ribosomes of aged potato tuber slices, which sediments at 15 —18S, has a base composition different from that of 16S — rRNA, 5S-and 4S —RNA, and is not present on ribosomes of fresh slices. Its appearance is inhibited by actinomycin D and therefore most probably dependent on transcription. This compound, purified from sucrose gradients, enhances in vitro leucine incorporation into peptide material by ribosomes of fresh potato slices.
The possibility is discussed that this fraction-among other factors-is responsible for the enhanced protein synthesis after slicing plant storage organs, and is indicative of a general derepression phenomenon in these tissues.
Nucleic acid and histone modifications critically depend on the tricarboxylic acid (TCA) cycle for substrates and cofactors. Although a few TCA cycle enzymes have been reported in the nucleus, the corresponding pathways are considered to operate in mitochondria. Here, we show that a part of the TCA cycle is operational also in the nucleus. Using 13C-tracer analysis, we identified activity of glutamine-to-fumarate, citrate-to-succinate, and glutamine-to-aspartate routes in the nuclei of HeLa cells. Proximity labeling mass spectrometry revealed a spatial vicinity of the involved enzymes with core nuclear proteins. We further show nuclear localization of aconitase 2 and 2-oxoglutarate dehydrogenase in mouse embryonic stem cells. Nuclear localization of the latter enzyme, which produces succinyl-CoA, changed from pluripotency to a differentiated state with accompanying changes in the nuclear protein succinylation. Together, our results demonstrate operation of an extended metabolic pathway in the nucleus, warranting a revision of the canonical view on metabolic compartmentalization.
