We study how the Eurosystem Collateral Framework for corporate bonds helps the European Central Bank (ECB) fulfill its policy mandate. Using the ECBs eligibility list, we identify the first inclusion date of both bonds and issuers. We find that due to the increased supply and demand for pledgeable collateral following eligibility, (i) securities lending market trading activity increases, (ii) eligible bonds have lower yields, and (iii) the liquidity of newly-issued bonds declines, whereas the liquidity of older bonds is una↵ected/improves. Corporate bond lending relaxes the constraint of limited collateral supply, thereby making the market more cohesive and complete. Following eligibility, bond-issuing firms reduce bank debt and expand corporate bond issuance, thus increasing overall debt size and extending maturity.
Acute and chronic inflammation play a pivotal role in various diseases, such as rheumatoid arthritis, atherosclerosis, bacterial as well as viral infections and therefore are an everyday-challenge in clinical practice. In this context, biologically active products of the cyclooxygenases and the prostanoid synthases, e.g. prostaglandins, critically contribute to various aspects of the inflammatory response in almost every tissue of the body. Emerging evidence over the past decades has demonstrated that these mediators are not only responsible for a pro-inflammatory response, but also show anti-inflammatory and pro-resolving properties. The relevance of biologically active lipids in this context is strengthened by the clinical efficacy of nonsteroidal anti-inflammatory drugs (NSAIDs), e.g. Aspirin®, which block the biosynthesis of the mediators via the cyclooxygenase (COX) enzymes. Notably, microsomal prostaglandin E synthase-1 (mPGES-1)-derived prostaglandin E2 (PGE2) is a well-studied, functionally versatile PG, which promotes its effects via specific G protein-coupled receptors (GPCRs). Activation of these receptors elicits an internal signal transduction cascade, including activation of the adenylyl cyclase (AC). Active AC contributes to an elevated intracellular cyclic adenosine monophosphate (cAMP) level, which in turn activates the transcription factor cAMP response element-binding protein (CREB) via phosphorylation.
While the role of PGE2 in the inflammatory context has been well-documented in previous literature, relatively little is known about CREB-dependent transcriptional changes in inflammation. Therefore, the aim of this study was to investigate the effect of mPGES-1-derived PGE2 on CREB-mediated transcriptional changes specifically in murine wild-type (WT) and mPGES-1 knock-out (KO) macrophages in an inflammatory context. To address this issue, bone marrow-derived macrophages (BMDMs) were treated with either the bacterial cell wall component lipopolysaccharide (LPS) in combination with interferon-γ (IFN-γ) or the yeast extract zymosan. To analyze effects on CREB activation we determined protein expression profiles of relevant PGE2-synthesizing enzymes, i.e. COX-2 and mPGES-1, as well as activity of the downstream transcription factor CREB. The activity of mPGES-1 was simultaneously determined by the analysis of the prostanoid kinetics. Under these experimental conditions we showed that COX-2 is strongly induced, and we also observed elevated activated CREB levels in WT as well as in mPGES-1 KO macrophages. Further, both LPS+IFN-γ and zymosan increased expression of mPGES-1 in WT but not in mPGES-1-deficient macrophages. These findings go in hand with largely similar alterations in the PGD2, TXB2, PGF2α profiles in WT and mPGES-1 KO macrophages upon stimulation. Of note, an elevated PGE2 production was also observed in mPGES-1-deficient macrophages at later stages upon inflammatory conditions. Subsequently, potential CREB-regulated targets were identified in macrophages upon inflammatory stimuli after 16 h by chromatin immunoprecipitation (ChIP) followed by Next-Generation-Sequencing (NGS). Surprisingly, despite equal levels of pCREB the characterization of CREB binding sites revealed different targetome profiles between WT and mPGES-1 KO macrophages. Specifically, the fatty acid metabolic processes-associated targets appeared to be selectively lost in mPGES-1-deficient vs. WT macrophages. We further validated one of those targets, i.e. the endoplasmic reticulum lipid raft-associated protein 1 (Erlin1), at the mRNA expression level, which indeed was differentially transcribed in response to different PGE2 synthesizing conditions.
Mechanistically, CREB is a well-characterized phosphorylation-dependent transcription factor in cell survival, proliferation, differentiation, and immune responses. Yet, our understanding of the functions of CREB in inflammation, specifically with respect to its activation by PGE2, is insufficient. Due to its biological relevance in inflammation it clearly requires additional studies to shed light on the details of CREB activation in macrophages to provide possibilities of therapeutic interventions.
The electron transport chain (ETC) is used by cells to create an electrochemical proton gradient which can be used by the ATP synthase to produce ATP. ETC, also called respiratory chain, is formed in mitochondria by four complexes (complex I-IV) and mediated by two electron carriers: cytochrome c and ubiquinone. Electrons are passed from one complex to another in a series of redox reactions coupling proton pumping from the negative (N) side of the membrane to the positive (P) side. Complex I can introduce electrons into the ETC by oxidizing NADH to NAD+ and reducing quinone (Q) to quinol (QH2). The process accomplishes pumping of four protons across the membrane. Complex II is another electrons entry point. It catalyzes the oxidation of succinate to fumarate while reducing Q to QH2. Complex III, also called cytochrome bc1 complex, can transfer the electrons from QH2 to cytochrome c and couple to proton pumping. In complex III the Q-cycle contributes four proton translocations: two protons are required for the reduction of one quinone to a quinol and two protons are released to the P side. Complex IV (cytochrome c oxidase), the terminal complex of the ETC, catalyzes the electron transfer to oxygen and pumps four protons to the P side. Structures of ETC complexes are available. However, the structure of a hyperthermophilic cytochrome bc1 complex has not been elucidated till now. Additionally, the dimeric crystal structure of cytochrome c oxidase from bovine has been discussed controversially.
To build up a functional complex, cofactors are required. The active site of A- and B-type cytochrome c oxidases contain the high spin heme a which is synthesized by the integral membrane protein heme A synthase (HAS). HAS can form homooligomeric complexes and its oligomerization is essential for the biological function of HAS. HAS is evolutionarily conserved among prokaryotes and eukaryotes. Despite its importance, little is known about the detailed structural properties of HAS oligomers.
During my PhD studies, I focused on the cytochrome c oxidase (AaCcO), the cytochrome bc1 complex (Aabc1) and the heme A synthase (AaHAS) from Aquifex aeolicus. This organism is one of the most hyperthermophilic ones and can live at extremely high temperatures, even up to 95 °C. Respiratory chain complexes provide energy for the metabolism of organisms, and their structures have been studied extensively in the past few years. However, there has been a lack of atomic structures of complexes from hyperthermophilic and ancient bacteria, so little is known about the mechanism of these macromolecular machines under hyperthermophilic conditions. Therefore, my PhD studies had four main objectives: 1) to structurally and functionally characterize AaCcO, 2) to reveal the mechanism of Aabc1 thermal stability based on its structure, 3) to determine the oligomerization of AaHAS, 4) to provide valuable insights into the relationship between function and oligomerization of AaHAS.
1) Structure of AaCcO
Heme-copper oxidases (HCOs) catalyze the oxygen reduction reaction being the terminal enzymes in the plasma membranes in many prokaryotes or of the aerobic respiratory chain in the inner mitochondrial membrane. By coupling this exothermic reaction to proton pumping across the membrane to the P side, they contribute to the establishment of an electrochemical proton gradient. The energy in the proton electrochemical proton gradient is used by the ATP synthase to generate ATP. HCOs are classified into three major families: A, B and C, based on phylogenetic comparisons. The well-studied aa3-type cytochrome c oxidase from Paracoccus denitrificans (P. denitrificans) represents A-family HCOs. So far, the only available structure of the ba3-type cytochrome c oxidase from Thermus thermophilus represents the B-family of HCOs. This family contains a number of bacterial and archaeal oxidases. The C-family contains only cbb3-type cytochrome c oxidases.
The AaCcO is one of the ba3-type cytochrome c oxidases. Based on the genomic DNA sequence analysis, it has been revealed that A. aeolicus possesses two operons coding for cytochrome c oxidases (two different subunit I genes, two different subunit II genes and one subunit III gene). So far, only subunits CoxB2 and CoxA2 were identified. The presence of the additional subunit IIa was reported in 2012. Moreover, a previous paper reported that AaCcO can use horse heart cytochrome c and decylubiquinol as electron donors and the typical cytochrome c oxidase inhibitor cyanide does not block the reaction completely.
In the course of my PhD studies, I performed heterologous expression of AaCcO in Pseudomonas stutzeri (P. stutzeri) and co-expression with AsHAS in Escherichia coli, respectively. The subcomplex CoxA2 and CoxB2 can be purified from P. stutzeri, however, it lacks heme A. Additionally, a protocol for the heterologous production of cytochrome c555 from A. aeolicus was established. In parallel, I also purified the AaCcO from native membranes according to previously reported methods with some modifications. The activity of AaCcO with its native substrate, cytochrome c555, was 14 times higher than with horse heart cytochrome c.
To enable a detailed investigation and comparison of AaCcO and other cytochrome c oxidases, the cryo-EM structure of AaCcO was determined to 3.4 Å resolution. It shows that the three subunits CoxA2, CoxB2, and IIa are tightly bound together to form a dimer in the membrane. Surprisingly, CoxA2 contains two additional TMHs (TMH13 and TMH14) to enhance the protein stability. The cofactors heme a3, heme b, CuA and CuB are also identified. Interestingly, two molecules of 1,4-naphthoquinone and cardiolipin were observed in the dimer interface. Based on the structure analysis, the AaCcO possesses only the K-pathway for proton delivery to the active site and proton pumping.

Aufgrund des großen Potenzials der Nanotechnologie ist in Zukunft eine Zunahme der Produktion und Verwendung von Nanomaterialien zu erwarten, wodurch mit einer steigenden Freisetzung in der Umwelt zu rechnen ist. In der vorliegenden Dissertation werden daher Methoden zur Untersuchung von Nanomaterialien betrachtet und Effekte von NP auf Algen untersucht.
In Teil I wurden Silber-, Titandioxid- und Polystyrol-Nanopartikel sowie Kohlenstoffnano-röhrchen untersucht. Von jedem Nanomaterial standen eine unmodifizierte Form sowie zwei modifizierte Partikeltypen mit geladener Oberfläche und zusätzlich Polystyrol-Mikropartikel zur Verfügung. Zunächst erfolgte eine Charakterisierung der Materialien mittels Transmissions-elektronenmikroskopie, wobei die Größe der Objekte gemessen und das Verhalten beschrieben wurde. Zudem wurde im Fall der Polystyrol-Nanopartikel der Einfluss mehrerer Chemikalien getestet, welche im Zusammenhang mit der Probenvorbereitung für das Elektronen¬mikroskop zum Einsatz kamen. In einem nächsten Schritt erfolgte die Untersuchung von Nanomaterialien in umweltrelevanten Matrices. Hierbei wurden Boden- und Wasserproben sowie humane Körperflüssigkeiten und Fischgewebe elektronenmikroskopisch auf die Anwesenheit von synthetischen Nanomaterialien untersucht und Proben mit Nanomaterialien versetzte, um die Nachweisbarkeit mit dem Elektronenmikroskop bewerten zu können. Zusätzlich wurden verschiedene Zellkulturen und Gewebe auf morphologische Auffälligkeiten im Zusammenhang mit einer Exposition gegenüber Nanomaterialien untersucht.
Die durchgeführten Versuche zeigen, dass die Transmissionselektronenmikroskopie für viele Nanomaterialien ein sinnvolles Charakterisierungswerkzeug darstellt. Die Untersuchung besonders kleiner Partikel mit einem Durchmesser im einstelligen Nanobereich gestaltet sich jedoch schwierig bis unmöglich. Für den Nachweis von Nanomaterialien in Umweltmatrices und Zellen ist die Methode nur bedingt geeignet, wobei insbesondere niedrige Partikelkonzentrationen problematisch sind. Die Methode ist somit lediglich als Ergänzung zu anderen Nachweismethoden zu betrachten, kann jedoch hilfreiche Informationen zur Lokalisation von Nanoobjekten in Zellen und zu ihrem Verhalten in Umweltproben liefern.
In Teil II wurden die beiden Grünalgen Raphidocelis subcapitata und Chlamydomonas reinhardtii sowie die Diatomee Cyclotella meneghiniana gegenüber unterschiedlich modifizierten Silber-, Titandioxid- und Polystyrol-Nanopartikeln exponiert. Die Beurteilung der Toxizität wurde anhand der über die Absorption gemessenen Zellzahl, des Chlorophyll a Gehalts, der über die Chlorophyllfluoreszenz gemessenen Parameter Fv/Fm und NPQ sowie der transmissions¬elektronenmikroskopischen Untersuchung der Algenzellen vorgenommen. Zudem wurde der Einfluss der Beschattung von Algenzellen durch die Nanopartikel experimentell untersucht.
Die Untersuchungen zeigen, dass Nanomaterialien bei Absorptionsmessungen in Abhängigkeit von ihrem Grundmaterial, ihrer Oberflächenmodifikation und dem umgebenden Medium ein mehr oder weniger starkes Streuungsverhalten zeigen. Auch die Anwesenheit von Algen kann einen deutlichen Einfluss haben. Trotz der Beeinflussung der Lichtstreuung hat die Beschattung von Algen durch die Trübung des Mediums durch Nanomaterialien keinen Einfluss auf das Wachstum der Testorganismen. Die direkte Exposition der Algen gegenüber den Nanomaterialien zeigt, dass Silber-Nanopartikel die toxischste Wirkung haben. Die Abgabe von Silberionen durch die Partikel kann hierbei die auftretenden Effekte erklären. Auch Titandioxid-Nanopartikel führen zu negativen Effekten, wobei mögliche Gründe die Toxizität des Materials und die physikalische Isolierung der Zellen sind. Die Polystyrol-Nanopartikel haben eine stimulierende Wirkung auf die Algenzellen, welche auf einer Präferenz von adhäsivem Wachsen und dem Hormesis-Effekt beruhen kann. Die Oberflächenmodifikation der Nanomaterialien hat zwar einen Effekt auf die Toxizität, ihr Einfluss wird jedoch durch andere Faktoren überlagert. In Bezug auf die unterschiedlichen Methoden zum Nachweis der Toxizität, ist die Bestimmung des Chlorophyll a-Gehalts als besonders sensitiv zu bewerten und kann zudem auf alle Partikel angewandt werden. Hinsichtlich der Absorptionsmessung besteht teilweise ein Einfluss durch die Partikelstreuung. Die Messung der Chlorophyllfluoreszenz scheint einer starken Beeinflussung durch externe Faktoren und ggf. die Nanomaterialien selbst zu unterliegen. Die elektronenmikroskopische Untersuchung ist vergleichsweise wenig sensitiv, kann jedoch ergänzende Informationen bezüglich der Wirkweise von Nanomaterialien liefern. Der Vergleich der Testorganismen zeigt, dass Raphidocelis subcapitata empfindlicher reagiert als Chlamydomonas reinhardtii. Eine allgemeingültige Sensitivitätsabstufung zwischen den Grünalgen und der Diatomee ist nicht möglich, da die Reaktionen in Abhängigkeit von Medium bzw. Partikelgrundmaterial unterschiedlich ausfallen.
The RNA cleaving catalyst tris(2-aminobenzimidazole) when attached to the 5’ terminus of oligonucleotides cuts complementary RNA strands in a highly site-specific manner. Conjugation was previously achieved by the acylation of an amino linker by an active ester of the catalyst. However, this procedure was low yielding and not reliable. Here, a phosphoramidite building block is described that can be coupled to oligonucleotides by manual solid phase synthesis in total yields around 85%. Based on this chemistry, we have now studied the impact of LNA (locked nucleic acids) nucleotides on the rates and the site-specificities of RNA cleaving conjugates. The highest reaction rates and the most precise cuts can be expected when the catalyst is attached to a strong 5’ closing base pair and when the oligonucleotide contains several LNA units that are equally distributed in the strand. However, when placed in the 5’ position, LNA building blocks tend to diminish the specificity of RNA cleavage.
RNA-protein complexes (RNPs) are essential components in a variety of cellular processes, and oftentimes exhibit complex structures and show mechanisms that are highly dynamic in conformation and structure. However, biochemical and structural biology approaches are mostly not able to fully elucidate the structurally and especially conformationally dynamic and heterogeneous nature of these RNPs, to which end single molecule Förster resonance energy transfer (smFRET) spectroscopy can be harnessed to fill this gap. Here we summarize the advantages of strategic smFRET studies to investigate RNP dynamics, complemented by structural and biochemical data. Focusing on recent smFRET studies of three essential biological systems, we demonstrate that investigation of RNPs on a single molecule level can answer important functional questions that remained elusive with structural or biochemical approaches alone: The complex structural rearrangements throughout the splicing cycle, unwinding dynamics of the G-quadruplex (G4) helicase RHAU, and aspects in telomere maintenance regulation and synthesis.
Patient therapy is based mainly on a combination of diagnosis, suitable monitoring or support devices and drug treatment and is usually employed for a pre-existing disease condition. Therapy remains predominantly symptom-based, although it is increasingly clear that individual treatment is possible and beneficial. However, reasonable precision medicine can only be realized with the coordinated use of diagnostics, devices and drugs in combination with extensive databases (4Ds), an approach that has not yet found sufficient implementation. The practical combination of 4Ds in health care is progressing, but several obstacles still hamper their extended use in precision medicine.

Dass es "Generationen" gibt, glauben viele zu wissen. Aber was genau sie darunter verstehen, fällt ihnen schwer zu beschreiben. Trotzdem erfreut sich der Generationenbegriff heute einer geradezu ubiquitären Verwendung. Er wird genutzt als Identitäts- und Kollektivbezeichnung, aber auch als Erfahrungs- und Handlungsbegriff. Selbst die Werbesprache hat sich mittlerweile seiner bemächtigt. Und in den Feuilletons deutscher Zeitungen avancierte er zuletzt zu einer Passepartout-Formel, um sozialpolitische "Generationenkonflikte" auszudeuten. ...
Die kompakte Monographie, die mittlerweile auch in englischer Sprache erschienen ist, wurde von Arvid Göttlicher verfasst, der in den vergangenen gut 30 Jahren bereits mehrere Bücher zur antiken Seefahrt vorlegte und als einer der profundesten Kenner der Materie im deutschsprachigen Raum gelten darf. Bei dem hier zu besprechenden Werk aus dem Jahre 2006 wurde allerdings ein Titel gewählt, der nicht besonders treffend erscheint. Es handelt sich hier keineswegs um eine Einführung zur antiken Schifffahrt, wie der erste Titelteil suggeriert; vielmehr steht die Auseinandersetzung mit den Teilen von Herodots Werk im Vordergrund, die diverse Aspekte der Nautik behandeln. ...
Es war eine in jeder Hinsicht außergewöhnliche Tagung, die Ende Oktober 2010 in Marbach stattfand. 20 renommierte Historiker/innen kamen zusammen, um an ihren akademischen Lehrer und Mentor Wolfgang J. Mommsen zu erinnern. Jeder sollte, so die Vorgabe der beiden Organisatoren Christoph Cornelißen und Gerhard Hirschfeld, aus seinem Forschungsfeld berichten und den Einfluss Mommsens auf diesen Themengebieten einschätzen und beschreiben. Der entsprechende Tagungsband mit den Beiträge konnte bereits auf der Konferenz vorgestellt werden. Zugegen war auch die Familie Mommsen und ebenso der Zwillingsbruder Hans Mommsen. ...
Die hier zu besprechende Monographie "Mensch und Landschaft im südwestlichen Latium in der römischen Antike" stellt die leicht überarbeitete Fassung der Dissertation von Michael Teichmann (im Folgenden T.) dar, die er im Winter 2016/17 an der Christian-Albrechts-Universität zu Kiel am Institut für Geoarchäologie eingereicht hat. Der Phoibos Verlag strebt mit der Serie "Phoibos Humanities Series" eine Doppelstrategie an und vertreibt das Werk sowohl als gedrucktes Buch (89 €) wie auch als E-Book (59 €). Möglicherweise erklärt sich so der Verzicht auf einen Index, da das maschinenlesbare E-Book einen solchen theoretisch überflüssig macht. Da der Rezensent mit dem gedruckten Buch gearbeitet hat, fiel das Fehlen eines Indexes doch negativ ins Gewicht. ...
Der hier zu besprechende Sammelband ist das Begleitbuch zu einer Ausstellung der Abguss-Sammlung des Instituts für Klassische Archäologie der Freien Universität Berlin zum Thema "Realismen in der griechischen Plastik", die von Februar bis Juli 2018 dort zu sehen war. Die Ausstellung war aus Lehrveranstaltungen der beiden Herausgeber hervorgegangen und folgte damit bekannten Vorgängern vergangener Jahrzehnte, deren begleitende Kataloge geradezu Handbuchstatus erlangt haben. ...
Am 27. Juni 2011 fand im Institut für Stadtgeschichte Frankfurt/Main eine Podiumsdiskussion zum Thema "Die Archive und die historische Forschung" statt. Etwa 35 Interessierte folgten dieser Diskussion, die vom Landesarchiv Nordrhein-Westfalen und dem VdA – Verband deutscher Archivarinnen und Archivare e. V. veranstaltet wurde.
Zu Beginn stellte die Leiterin des Instituts für Stadtgeschichte, Evelyn Brockhoff, ihre Einrichtung vor und nannte dabei auch die Angebote für Wissenschaftler. Es folgte die eigentliche Diskussion, an der zwei Vertreter der Geschichtswissenschaft – CHRISTOPH CORNELIßEN (Christian-Albrechts-Universität Kiel) und DIRK VAN LAAK (Justus-Liebig-Universität Gießen) – und zwei Vertreter des Archivwesens – CLEMENS REHM (Landesarchiv Baden-Württemberg/VdA) und WILFRIED REININGHAUS (Landesarchiv Nordrhein-Westfalen) – unter der Moderation von ANDREAS PILGER (Landesarchiv Nordrhein-Westfalen) teilnahmen. ...
Verbreitungsbilder von Artefakten regten die archäologische Forschung seit dem späten 19. Jahrhundert dazu an, "Kulturkreise" zu definieren und auf dieser Basis Siedlungsräume ethnischer Gruppen, deren (Fremd)bezeichnung in lateinischen und griechischen Schriftquellen überliefert ist, zu lokalisieren. Diese Herangehensweise provoziert seit nunmehr fast zwei Jahrzehnten eine heftige Diskussion über die Interpretierbarkeit materieller Quellen im Hinblick auf die Benennung, Definition und Lokalisierung antiker Ethnien und Identitäten. ...

There has been a burgeoning interest in the sociology of the Frankfurt School as well as the oeuvre of Theodor W. Adorno since the 2016 presidential campaign of Donald J. Trump. The objectives of this study are to both illustrate the enduring importance of Adorno and to provide an important theoretical outline in making sense of Trump’s 2016 United States presidential campaign. Using Adorno’s understudied textual analysis of the radio addresses of Martin Luther Thomas and data from Trump’s 2016 US presidential campaign, we find that Trump’s own discourse can be condensed into three of Adorno’s rhetorical devices: (1) the lone wolf device or anti-statism/pseudo-conservatism, reflecting his criticism of "special interests" and his appraisal of business and (self-)finance; (2) the movement device, which amounted to glorification of action; and (3) the exactitude of error device which amounted to xenophobic, ethnonationalist hyperbole.
In der Reihe "Bildbände zur Archäologie" des Philipp von Zabern Verlags erschien 2015 der Band "Antike Felsgräber" von Stephan Steingräber. Ziel des Autors ist es, die monumentalen Felsfassadengräber Etruriens in einem größeren Kontext darzustellen, was im Untertitel "unter besonderer Berücksichtigung der etruskischen Felsgräbernekropolen" zum Ausdruck kommt. Mit 162 überwiegend sehr qualitätvollen Abbildungen auf 144 Seiten fügt sich der Band in die bekannte Reihe, die archäologische Denkmäler für ein breites, Kultur interessiertes Publikum erschließt. Der Autor gehört zu den Kennern dieser etruskischen Gräber und zieht im Vorwort die Verbindungslinien zu seiner wissenschaftlichen Tätigkeit. In 14 Kapiteln spannt der Verfasser einen weiten Bogen (S. 7-86) mit Schlaglichtern auf Felsgräber und Felsarchitektur außerhalb Südetruriens; letzteren ist ein ausführliches Kapitel gewidmet (S. 87-128). Ein Anhang mit einer nützlichen Zusammenstellung der wichtigsten Informationen zu den südetruskischen Felsgräbernekropolen (S. 131-137) und einem Glossar beschließen den Bildband. Die Bibliographie spiegelt in ihrer Auswahl die Schwerpunktsetzung des Bandes. Nur der Titel selbst – auf dem Umschlag steht der Untertitel nicht – sowie das zentrale Titelbild der Felsgrabnekropole von Myra in Kleinasien stehen in eigenartigem Widerspruch zur eindeutig regionalen Konzeption. Um es gleich vorwegzunehmen, diese ambivalente Zielsetzung wirkt sich im Verlauf der Darlegungen ungünstig aus. ...
Aims: Of patients with atrial fibrillation (AF), approximately 10% undergo percutaneous coronary intervention (PCI). We studied the safety and efficacy of dual vs. triple antithrombotic therapy (DAT vs. TAT) in this population.
Methods and results: A systematic review and meta-analysis was conducted using PubMed, Embase, EBSCO, Cochrane database of systematic reviews, Web of Science, and relevant meeting abstracts for Phase 3, randomized trials that compared DAT vs. TAT in patients with AF following PCI. Four trials including 5317 patients were included, of whom 3039 (57%) received DAT. Compared with the TAT arm, Thrombolysis in Myocardial Infarction (TIMI) major or minor bleeding showed a reduction by 47% in the DAT arm [4.3% vs. 9.0%; hazard ratio (HR) 0.53, 95% credible interval (CrI) 0.36–0.85, I2 = 42.9%]. In addition, there was no difference in the trial-defined major adverse cardiac events (MACE) (10.4% vs. 10.0%, HR 0.85, 95% CrI 0.48–1.29, I2 = 58.4%), or in individual outcomes of all-cause mortality, cardiac death, myocardial infarction, stent thrombosis, or stroke between the two arms.
Conclusion: Compared with TAT, DAT shows a reduction in TIMI major or minor bleeding by 47% with comparable outcomes of MACE. Our findings support the concept that DAT may be a better option than TAT in many patients with AF following PCI.
Römische Grabbauten sind im Grunde kein Thema, dem es an wissenschaftlicher Aufmerksamkeit mangelt. Allerdings lag der Fokus in den vergangenen Jahrzehnten beinahe durchweg auf den Denkmälern in Rom und Italien, die sowohl aus bautypologischer als auch aus sozial- und kulturhistorischer Perspektive intensiv erforscht wurden und werden. Für die Grabbauten in den Provinzen, insbesondere den Nordwestprovinzen fehlte es dagegen lange an übergreifenden Studien. Diese Lücke wird nun durch die in Teilen auf den Ergebnissen eines EUProjektes zum Thema "Transformation" aufbauende Studie von Markus Scholz (S.) geschlossen, der sich den erhaltenen Denkmälern in detaillierter Weise anhand einer typologischen Methodik annimmt. Der Größe des Untersuchungsgebiets, das von Britannien über die Rhein- und Donauprovinzen bis ans Schwarze Meer reicht, und der damit einhergehend immensen Zahl einbezogener Objekte (ca. 3500 Grabbauten) entspricht auch der Umfang des Werkes, das mit zwei Bänden mit jeweils rund 570 Seiten bereits auf den ersten Blick gewichtig anmutet. ...
The FIRE AND ICE Trial (ClinicalTrials.gov, identifier NCT01490814) was initiated in 2012 as a multicenter, randomized, head‐to‐head comparison of radiofrequency current (RFC) and cryoballoon catheter ablation for the treatment of patients with drug‐refractory symptomatic paroxysmal atrial fibrillation (AF). Six years on, it remains the largest, randomized comparison of safety and efficacy between 2 catheter ablation modalities used in the treatment of patients with AF. This landmark trial not only established noninferiority between cryoballoon and RFC ablation for pulmonary vein isolation (PVI) with regard to the study's efficacy and safety primary end points,1 but also, it evaluated secondary end points that were critical for a representative study interpretation. ...
Das vorliegende Buch ist als Begleitband zur gleichnamigen Sonderausstellung erschienen, die vom 04.04.-14.09.2008 in der Archäologischen Staatssammlung München gezeigt wurde. Sein Titel weckt hohe Erwartungen, verspricht er doch die Betrachtung einer kurzen, in der oberflächlichen Gesamtwahrnehmung der römischen Militärgrenzen an Rhein und Donau kaum auffälligen Teilstrecke des obergermanischen Limes als Beispiel eines kulturellen Welterbes. Es handelt sich um die rund 50 km lange Strecke der militärisch gesicherten Außengrenze der römischen Provinz Germania Superior, die zwischen dem bei Hanau gelegenen Großkrotzenburg und Miltenberg entlang des westlichen Ufers des Mains verlief. Dieser Limesabschnitt unterscheidet sich einzig dadurch von allen anderen Strecken des obergermanischen Limes, dass die durchgehenden Grenzsperren nicht aus Palisaden bzw. (später) Wall und Graben bestanden, sondern der Fluss selbst die natürliche Grenzsperre zwischen dem römischen Provinzterritorium und den germanischen Stammesgebieten bildete. ...
Background: Until now, a few studies have addressed the accuracy of intraoral scanners (IOSs) in implantology. Hence, the aim of this in vitro study was to assess the accuracy of 5 different IOSs in the impressions of single and multiple implants, and to compare them.
Methods: Plaster models were prepared, representative of a partially edentulous maxilla (PEM) to be restored with a single crown (SC) and a partial prosthesis (PP), and a totally edentulous maxilla (TEM) to be restored with a full-arch (FA). These models were scanned with a desktop scanner, to capture reference models (RMs), and with 5 IOSs (CS 3600®, Trios3®, Omnicam®, DWIO®, Emerald®); 10 scans were taken for each model, using each IOS. All IOS datasets were loaded into a reverse-engineering software where they were superimposed on the corresponding RMs, to evaluate trueness, and superimposed on each other within groups, to determine precision. A statistical analysis was performed.
Results: In the SC, CS 3600® had the best trueness (15.2 ± 0.8 μm), followed by Trios3® (22.3 ± 0.5 μm), DWIO® (27.8 ± 3.2 μm), Omnicam® (28.4 ± 4.5 μm), Emerald® (43.1 ± 11.5 μm). In the PP, CS 3600® had the best trueness (23 ± 1.1 μm), followed by Trios3® (28.5 ± 0.5 μm), Omnicam® (38.1 ± 8.8 μm), Emerald® (49.3 ± 5.5 μm), DWIO® (49.8 ± 5 μm). In the FA, CS 3600® had the best trueness (44.9 ± 8.9 μm), followed by Trios3® (46.3 ± 4.9 μm), Emerald® (66.3 ± 5.6 μm), Omnicam® (70.4 ± 11.9 μm), DWIO® (92.1 ± 24.1 μm). Significant differences were found between the IOSs; a significant difference in trueness was found between the contexts (SC vs. PP vs. FA). In the SC, CS 3600® had the best precision (11.3 ± 1.1 μm), followed by Trios3® (15.2 ± 0.8 μm), DWIO® (27.1 ± 10.7 μm), Omnicam® (30.6 ± 3.3 μm), Emerald® (32.8 ± 10.7 μm). In the PP, CS 3600® had the best precision (17 ± 2.3 μm), followed by Trios3® (21 ± 1.9 μm), Emerald® (29.9 ± 8.9 μm), DWIO® (34.8 ± 10.8 μm), Omnicam® (43.2 ± 9.4 μm). In the FA, Trios3® had the best precision (35.6 ± 3.4 μm), followed by CS 3600® (35.7 ± 4.3 μm), Emerald® (61.5 ± 18.1 μm), Omnicam® (89.3 ± 14 μm), DWIO® (111 ± 24.8 μm). Significant differences were found between the IOSs; a significant difference in precision was found between the contexts (SC vs. PP vs. FA).
Conclusions: The IOSs showed significant differences between them, both in trueness and in precision. The mathematical error increased in the transition from SC to PP up to FA, both in trueness than in precision.
Der vorliegende Band umfasst vier Themenkomplexe, die in insgesamt elf Aufsätzen behandelt werden. Background and objective: Acutely ill older patients with cognitive impairment represent a major subgroup in acute care hospitals. In this context, communication plays a crucial role for patients’ well-being, healthcare decisions, and medical outcomes. As validated measures are lacking, we tested the psychometric properties of an observational instrument to assess Communication Behavior in Dementia (CODEM) in the acute care hospital setting. As a novel feature, we were also able to incorporate linguistic and social-contextual measures.
Material and methods: Data were drawn from a cross-sectional mixed methods study that focused on the occurrence of elderspeak during care interactions in two German acute care hospitals. A total of 43 acutely ill older patients with severe cognitive impairment (CI group, Mage ± SD = 83.6 ± 5.7 years) and 50 without cognitive impairment (CU group, Mage ± SD = 82.1 ± 6.3 years) were observed by trained research assistants during a standardized interview situation and rated afterwards by use of CODEM.
Results: Factor analysis supported the expected two-factor solution for the CI group, i.e., a verbal content and a nonverbal relationship aspect. Findings of the current study indicated sound psychometric properties of the CODEM instrument including internal consistency, convergent, divergent, and criterion validity.
Conclusion: CODEM represents a reliable and valid tool to examine the communication behavior of older patients with CI in the acute care hospital setting. Thus, CODEM might serve as an important instrument for researcher and healthcare professionals to describe and improve communication patterns in this environment.

Umso trauriger ist es, dass der Bearbeiter dieses bedeutenden Fundes die Publikation seines Manuskriptes nicht mehr erleben konnte, starb Dr. Hans Roland Baldus doch in 2011. ...
Previous research indicates that anxiety disorders are characterized by an overgeneralization of conditioned fear as compared with healthy participants. Therefore, fear generalization is considered a key mechanism for the development of anxiety disorders. However, systematic investigations on the variance in fear generalization are lacking. Therefore, the current study aims at identifying distinctive phenotypes of fear generalization among healthy participants. To this end, 1175 participants completed a differential fear conditioning phase followed by a generalization test. To identify patterns of fear generalization, we used a k-means clustering algorithm based on individual arousal generalization gradients. Subsequently, we examined the reliability and validity of the clusters and phenotypical differences between subgroups on the basis of psychometric data and markers of fear expression. Cluster analysis reliably revealed five clusters that systematically differed in mean responses, differentiation between conditioned threat and safety, and linearity of the generalization gradients, though mean response levels accounted for most variance. Remarkably, the patterns of mean responses were already evident during fear acquisition and corresponded most closely to psychometric measures of anxiety traits. The identified clusters reliably described subgroups of healthy individuals with distinct response characteristics in a fear generalization test. Following a dimensional view of psychopathology, these clusters likely delineate risk factors for anxiety disorders. As crucial group characteristics were already evident during fear acquisition, our results emphasize the importance of average fear responses and differentiation between conditioned threat and safety as risk factors for anxiety disorders.
Chimeric antigen receptor (CAR) T cells are in prime focus of current research in cancer immunotherapy. Facilitating CAR T cell generation is among the top goals. We have recently demonstrated direct in vivo generation of human CD19-CAR T cells by targeting CD8+ cells using lentiviral vectors (LVs). The anti-tumor potency of in vivo generated CAR T cells was assessed in human PBMC-transplanted NSG mice carrying i.v. injected CD19+ Nalm-6 tumor cells. A single injection of CD8-targeted LV delivering CD19-CAR was sufficient to completely eliminate the tumor cells from bone marrow and spleen, whereas control animals contained high levels of CD19+ cells. Tumor elimination was due to in vivo generated CAR+ cells. Notably, these were not only composed of T lymphocytes but also included CAR+ natural killer cells (NK and NKT). This is the first demonstration of tumor elimination by in vivo generated human CAR T cells.

Leigh syndrome is one of the most common neurological phenotypes observed in pediatric mitochondrial disease presentations. It is characterized by symmetrical lesions found on neuroimaging in the basal ganglia, thalamus, and brainstem and by a loss of motor skills and delayed developmental milestones. Genetic diagnosis of Leigh syndrome is complicated on account of the vast genetic heterogeneity with >75 candidate disease-associated genes having been reported to date. Candidate genes are still emerging, being identified when “omics” tools (genomics, proteomics, and transcriptomics) are applied to manipulated cell lines and cohorts of clinically characterized individuals who lack a genetic diagnosis. NDUFAF8 is one such protein; it has been found to interact with the well-characterized complex I (CI) assembly factor NDUFAF5 in a large-scale protein-protein interaction screen. Diagnostic next-generation sequencing has identified three unrelated pediatric subjects, each with a clinical diagnosis of Leigh syndrome, who harbor bi-allelic pathogenic variants in NDUFAF8. These variants include a recurrent splicing variant that was initially overlooked due to its deep-intronic location. Subject fibroblasts were found to express a complex I deficiency, and lentiviral transduction with wild-type NDUFAF8-cDNA ameliorated both the assembly defect and the biochemical deficiency. Complexome profiling of subject fibroblasts demonstrated a complex I assembly defect, and the stalled assembly intermediates corroborate the role of NDUFAF8 in early complex I assembly. This report serves to expand the genetic heterogeneity associated with Leigh syndrome and to validate the clinical utility of orphan protein characterization. We also highlight the importance of evaluating intronic sequence when a single, definitively pathogenic variant is identified during diagnostic testing.
Communication sounds are ubiquitous in the animal kingdom, where they play a role in advertising physiological states and/or socio-contextual scenarios. Human screams, for example, are typically uttered in fearful contexts and they have a distinctive feature termed as “roughness”, which depicts amplitude fluctuations at rates from 30–150 Hz. In this article, we report that the occurrence of fast acoustic periodicities in harsh sounding vocalizations is not unique to humans. A roughness-like structure is also present in vocalizations emitted by bats (species Carollia perspicillata) in distressful contexts. We report that 47.7% of distress calls produced by bats carry amplitude fluctuations at rates ~1.7 kHz (>10 times faster than temporal modulations found in human screams). In bats, rough-like vocalizations entrain brain potentials and are more effective in accelerating the bats’ heart rate than slow amplitude modulated sounds. Our results are consistent with a putative role of fast amplitude modulations (roughness in humans) for grabbing the listeners attention in situations in which the emitter is in distressful, potentially dangerous, contexts.
Ataxin-2 (human gene symbol ATXN2) acts during stress responses, modulating mRNA translation and nutrient metabolism. Ataxin-2 knockout mice exhibit progressive obesity, dyslipidemia, and insulin resistance. Conversely, the progressive ATXN2 gain of function due to the fact of polyglutamine (polyQ) expansions leads to a dominantly inherited neurodegenerative process named spinocerebellar ataxia type 2 (SCA2) with early adipose tissue loss and late muscle atrophy. We tried to understand lipid dysregulation in a SCA2 patient brain and in an authentic mouse model. Thin layer chromatography of a patient cerebellum was compared to the lipid metabolome of Atxn2-CAG100-Knockin (KIN) mouse spinocerebellar tissue. The human pathology caused deficits of sulfatide, galactosylceramide, cholesterol, C22/24-sphingomyelin, and gangliosides GM1a/GD1b despite quite normal levels of C18-sphingomyelin. Cerebellum and spinal cord from the KIN mouse showed a consistent decrease of various ceramides with a significant elevation of sphingosine in the more severely affected spinal cord. Deficiency of C24/26-sphingomyelins contrasted with excess C18/20-sphingomyelin. Spinocerebellar expression profiling revealed consistent reductions of CERS protein isoforms, Sptlc2 and Smpd3, but upregulation of Cers2 mRNA, as prominent anomalies in the ceramide–sphingosine metabolism. Reduction of Asah2 mRNA correlated to deficient S1P levels. In addition, downregulations for the elongase Elovl1, Elovl4, Elovl5 mRNAs and ELOVL4 protein explain the deficit of very long-chain sphingomyelin. Reduced ASMase protein levels correlated to the accumulation of long-chain sphingomyelin. Overall, a deficit of myelin lipids was prominent in SCA2 nervous tissue at prefinal stage and not compensated by transcriptional adaptation of several metabolic enzymes. Myelination is controlled by mTORC1 signals; thus, our human and murine observations are in agreement with the known role of ATXN2 yeast, nematode, and mouse orthologs as mTORC1 inhibitors and autophagy promoters.
Background: Tacrolimus once-daily formulation (TacOD) was introduced as an alternative to twice-daily formulations de novo. Dosing recommendations range between 0.1 to 0.2 mg/kg BW/d.
Material and Methods: Amended dosing with a simple bottom-up de novo algorithm is presented. Primary outcome measure was feasibility of establishing adequate target trough levels and avoidance of over-immunosuppression, with adequate safety and efficacy after liver transplantation (LT).
Results: TacOD was given to 101 patients. Standard steroid-free immunosuppression consisted of MMF 2 g/d, basiliximab 20 mg on day 0 and 4, and delayed bottom-up IS with TacOD starting with 1 mg/d and doubling the dosage every day until target trough levels of 5 to 8 ng/ml were reached. By day 7 after LT, all except 3 patients had received TacOD. The earliest time point of introduction was day 2. A median of 9 mg/d (range: 0 to 25 mg/d) of TacOD were necessary to establish the trough levels by day 10, which was then 5.4 ng/ml (range: 1.5 to 20 ng/ml). Incidence of adverse events (AE), in particular neurological AEs (n=3), were low. Efficacy failure (acute rejection) was low (4.9%). Renal function was stable and did not deteriorate under CNI treatment.
Conclusions: This is the first report of bottom-up, amended, and simple dosing of TacOD in LT. The algorithm is feasible, safe, and efficient, avoiding trough level peaks and top-down strategies.
Smac (second mitochondria-derived activator of caspase) mimetics are considered as promising anticancer therapeutics and used to induce apoptosis by antagonizing inhibitor of apoptosis proteins, which are often abundantly expressed in cancer cells. Here, we identify interferon regulatory factor 1 (IRF1) as a novel critical regulator of Smac mimetic BV6-induced apoptosis and proinflammatory cytokine secretion with impact on the immune response. IRF1 knockdown rescues cells from BV6-induced apoptosis and attenuates BV6-stimulated upregulation of tumor necrosis factor-α (TNFα), indicating that IRF1 mediates BV6-triggered cell death, at least in part, by inducing TNFα. This notion is supported by data showing that exogenous supply of TNFα restores BV6-induced cell death in IRF-knockdown cells. Interestingly, IRF1 selectively controls the induction of nuclear factor-κB (NF-κB) target genes, as IRF1 depletion attenuates BV6-stimulated upregulation of TNFα and interleukin-8 (IL-8) but not p100 and RelB. Concomitant knockdown of IRF1 and p65 cooperate to inhibit BV6-induced cell death, implying a cooperative interaction of IRF1 and NF-κB. In addition, IRF1 silencing hampers TNFα induction by TNFα itself as an another prototypical NF-κB stimulus. Importantly, IRF1 depletion impedes BV6-stimulated secretion of additional proinflammatory cytokines such as granulocyte-macrophage colony-stimulating factor (GM-CSF), IL-8, IL-6 and monocyte chemoattractant protein-1, and migration of primary monocytes to BV6-treated tumor cells. In conclusion, this identification of IRF1 as a dual regulator of BV6-induced apoptosis and inflammatory cytokine secretion provides novel insights into determinants of sensitivity towards Smac mimetic and possible implications of Smac mimetic treatment on tumor microenvironment and immune response.
Background: Alzheimer’s disease (AD) is the most common form of dementia, and it affects more women than men. Mitochondrial dysfunction (MD) plays a key role in AD, and it is detectable at an early stage of the degenerative process in peripheral tissues, such as peripheral mononuclear blood cells (PBMCs). However, whether these changes are also reflected in cerebral energy metabolism and whether sex-specific differences in mitochondrial function occur are not clear. Therefore, we estimated the correlation between mitochondrial function in PBMCs and brain energy metabolites and examined sex-specific differences in healthy participants to elucidate these issues.
Methods: The current pilot study included 9 male and 15 female healthy adults (mean age 30.8 ± 7.1 years). Respiration and activity of mitochondrial respiratory complexes were measured using a Clarke-electrode (Oxygraph-2k system), and adenosine triphosphate (ATP) levels were determined using a bioluminescence-based assay in isolated PBMCs. Citrate synthase activity as a mitochondrial marker was measured using a photometric assay. Concentrations of brain energy metabolites were quantified in the same individuals using 1H-magnetic resonance spectroscopy (MRS).
Results: We detected sex-associated differences in mitochondrial function. Mitochondrial complexes I, I+II, and IV and uncoupled respiration and electron transport system (ETS) capacity in PBMCs isolated from blood samples of females were significantly (p < 0.05; p < 0.01) higher compared to males. ATP levels in the PBMCs of female participants were approximately 10% higher compared to males. Citrate synthase (CS) activity, a marker of mitochondrial content, was significantly (p < 0.05) higher in females compared to males. Sex-associated differences were also found for brain metabolites. The N-acetylaspartate (NAA) concentration was significantly higher in female participants compared to males in targeted regions. This difference was observed in white matter (WM) and an area with a high percentage (> 50%) of gray matter (GM) (p < 0.05; p < 0.01). The effect sizes indicated a strong influence of sex on these parameters. Sex-associated differences were found in PBMCs and brain, but the determined parameters were not significantly correlated.
Conclusions: Our study revealed sex-associated differences in mitochondrial function in healthy participants. The underlying mechanisms must be elucidated in more detail, but our study suggests that mitochondrial function in PBMCs is a feasible surrogate marker to detect differences in mitochondrial function and energy metabolism in humans and it underscores the necessity of sex-specific approaches in therapies that target mitochondrial dysfunction.

Risk culture during the last 2000 years - from an aleatory society to the illusion of risk control

The culture of risk is 2000 years old, although the term “risk” developed much later. The culture of merchants making decisions under uncertainty and taking the individual responsibility for the uncertain future started with the Roman “Aleatory Society”, continued with medieval sea merchants, who made business “ad risicum et fortunam”, and sustained to the culture of entrepreneurs in times of industrialisation and dynamic economic changes in the 18th and 19th century. For all long-term commercial relationships, the culture of honourable merchants with personal decision-making and individual responsibility worked well. The successful development of sciences, statistics and engineering within the last 100 years led to the conjecture that men can “construct” an economical system with a pre-defined “clockwork” behaviour. Since probability distributions could be calculated ex-post, an illusion to control risk ex-ante became a pattern in business and banking. Based on the recent experiences with the financial crisis, a “risk culture” should understand that human “Strength of Knowledge” is limited and the “unknown unknown” can materialise. As all decisions and all commercial agreements are made under uncertainty, the culture of honourable merchants is key to achieve trust in long-term economic relations with individual responsibility, flexibility to adapt and resilience against the unknown.
The work of the artist and writer Gerald Nestler explores finance and its social implications since the mid-1990s. Based on his professional experience as a trader as well as on post-disciplinary research, he has developed a unique approach that brings together theory and conversation with installation, video, performance, text, and other art forms. Probing into the narrative structures of contemporary capitalism, Nestler offers a techno-political critique directly from the core of the financial markets. This interview addresses his reading of the derivative as a world-producing apparatus that shapes the experience of the present by preconfiguring the future, and that provokes a shift from representational to performative speech in the actualization of biopower based on the exploitation of volatility and leverage. In conversation with Christian Kloeckner and Stefanie Mueller, he argues for the formation of specific human/non-human alliances that directly attack algorithmic as well as socio- economic black-boxing (schemes that monopolize inherently non-scarce resources), so as to open our imagination to skills and tactics that would allow us to navigate the rich but volatile flows of social, political, and economic abundance.
This special issue explores how finance deploys time, structures the future, and interacts with actors and institutions that sometimes function according to very different temporal regimes. Finance capitalism’s logic of recurrence, repetitive cycles, and successive ruptures has long been with us, but the essays in this special issue are particularly interested in how recent decades of intensified financialization have restructured temporal experience. They interrogate the production and dissemination of agency in an age of acceleration, risk, and uncertainty, asking how the temporality inscribed in financial transactions emerges from and simultaneously shapes individual and social practice. Topics covered range from the logic of finance and foundational concepts of financial theory to the intersection between objective structures and social practice, the role of literature, and finally questions of social insecurity, political action, and the possibility of resistance within a context of competing temporalities. In this introduction, the editors delineate some fundamental concepts and questions for our financial times.
This article develops a reading of Don DeLillo’s novel Cosmopolis that differentiates between two thematic and poetological axes running through the text. On the one hand, Cosmopolis explores the future-fixation of the risk regime of finance capitalism; on the other, it stages scenes of insecurity that physically threaten the protagonist and his world. Insecurity, the article argues, is a condition that throughout the text increasingly gains in appeal because it promises to offer an alternative to a world of managed risk. The concern with security emphasizes finitude and mortality, thus enabling a turn to existential matters that the virtual abstractions of finance have seemingly made inaccessible. While proposing an opposition between a logic of risk based on virtuality and a logic of (in)security based on authenticity, DeLillo’s novel also suggests that it is impossible to break out of the logic of risk management pervading late modernity. The appeal of (in)security articulated in Cosmopolis rather lies in the promise to existentially revitalize life within the confines of financialized capitalism.
A growing body of experimental syntactic research has revealed substantial variation in the magnitude of island effects, not only across languages but also across different grammatical constructions. Adopting a well-established experimental design, the present study examines island effects in Spanish using a speeded acceptability judgment task. To quantify variation across grammatical constructions, we tested extraction from four different types of structure (subjects, complex noun phrases, adjuncts and interrogative clauses). The results of Bayesian mixed effects modelling showed that the size of island effects varied between constructions, such that there was clear evidence of subject, adjunct and interrogative island effects, but not of complex noun phrase island effects. We also failed to find evidence that island effects were modulated by participants’ working memory capacity as measured by an operation span task. To account for our results, we suggest that variability in island effects across constructions may be due to the interaction of syntactic, semantic-pragmatic and processing factors, which may affect island types differentially due to their idiosyncratic properties.
As one of the first Direct Acting Antivirals (DAA), the protease inhibitor Telaprevir (TVR) was available in the European Union from 9/2011 until 9/2016 as a new treatment option for chronic Hepatitis C.Aim. To assess the implementation of therapy stopping rules or shortening of the treatment and their impact on sustained virological response (SVR), as well as the safety and efficacy of the TVR-based therapy during routine daily treatment of patients in Germany.Materials and Methods. 802 patients were assessed (272 treatment naïve, 520 pre-treated) in the noninterventional, multi-center study.Results. 56.6 % of the patients achieved SVR. SVR rate was higher in patients with relapse after previous treatment (68.0 %) than in patients with a previous null-response (31.1 %) and in previously untreated patients (58.1 %). Stopping rule conditions were fulfilled by 3.2 % of patients and it was implemented in 65.4 % of these. 34.3 % of the patients fulfilled the conditions for a therapy shortening. This rule was adhered to in 48.4 % of these, in 34.5 % it was not adhered to. Thus recommendations were not always being followed. Therapy shortening was considered more frequently in previously untreated (54.8 %) than for previously treated patients (24.2 %). Stopping rule application but not shortened treatment reduced therapy costs.Conclusion. The TVR-based therapy represented a breakthrough at that time. Further DAAs have been added as therapeutic options since, increasing the complexity of treatment choice and correct implementation. The third dataset dedicated to the Open Guided Waves platform aims at carbon fiber composite plates with an additional omega stringer at constant temperature conditions. The two structures used in this work are representative for real aircraft components. Comprehensive measurements were recorded in order to study (I) the impact of the omega stringer on guided wave propagation, and (II) elliptical reference damages of different sizes located at three separate positions on the structure. Measurements were recorded for narrowband excitation (5-cycle toneburst with varying carrier frequencies) and broadband excitation (using chirp waveforms). The paper presents the results of a technical validation including numerical modelling, and enables further research, for example related to probability of detection (POD) analysis.
Attachment theory is commonly used to investigate children’s psychosocial development. To demonstrate cultural variability and to advance the idea of attachment as a collective resource, we assessed children’s attachment networks during middle childhood among the Nseh, a Cameroonian clan with distinct concepts of family and childhood. Using photo elicitation interviews, we used an exploratory approach to investigate the structural and functional composition of these networks and to generate a comprehensive overview. Participants were 11 children (six girls and five boys), aged 6 to 10 years. Children took photos of individuals who were important to them and with whom they felt safe, comfortable, and at ease. Then, in follow-up interviews they were asked to characterize their attachment figures on sociostructural dimensions and to elaborate how those individuals made them feel comfortable and safe. Transcripts of the interviews were coded using ethnographic strategies. Initial descriptive codes were analyzed concerning key terms, semantic relationships, and their context of meaning, before assigning higher level codes to generate distinct main categories of functionality. Children described attachment networks that were structurally adapted to concepts of social ties and interactional norms of the clan. Concerning their functionality, children differentiated between peers, responsible for overt emotional needs, and adults, providing nutritional care. We conclude that this pattern reflects sources of security and concepts of care of the distinct developmental environment. We discuss the importance of context-specific and comprehensive approaches to attachment, moving beyond Eurocentric monotropic concepts, with the goal of developing a complex understanding of childhood across ecocultural settings.
Background: Computed tomography of the head (HCT) is a widely used diagnostic tool, especially for emergency and trauma patients. However, the diagnostic yield and outcomes of HCT for patients on medical intensive care units (MICUs) are largely unknown.
Methods: We retrospectively evaluated all head CTs from patients admitted to a single-center MICU during a 5-year period for CT indications, diagnostic yield, and therapeutic consequences. Uni- and multivariate analyses for the evaluation of risk factors for positive head CT were conducted.
Results: Six hundred ninety (18.8%) of all patients during a 5-year period underwent HCT; 78.7% had negative CT results, while 21.3% of all patients had at least 1 new pathological finding. The main indication for acquiring CT scan of the head was an altered mental state (AMS) in 23.5%, followed by a new focal neurology in 20.7% and an inadequate wake up after stopping sedation in 14.9% of all patients. The most common new finding was intracerebral bleeding in 6.4%. In 6.7%, the CT scan itself led to a change of therapy of any kind. Admission after resuscitation or a new focal neurology were independent predictors of a positive CT. Psychic alteration and AMS were both independent predictors of a higher chance of a negative head CT. Positive HCT during MICU is an independent predictor of lower survival.
Conclusions: New onset of focal neurologic deficit seems to be a good predictor for a positive CT, while AMS and psychic alterations seem to be very poor predictors. A positive head CT is an independent predictor of death for MICU patients.
Vegetation responds to drought through a complex interplay of plant hydraulic mechanisms, posing challenges for model development and parameterization. We present a mathematical model that describes the dynamics of leaf water-potential over time while considering different strategies by which plant species regulate their water-potentials. The model has two parameters: the parameter λ describing the adjustment of the leaf water potential to changes in soil water potential, and the parameter Δψww describing the typical ‘well-watered’ leaf water potentials at non-stressed (near-zero) levels of soil water potential. Our model was tested and calibrated on 110 time-series datasets containing the leaf- and soil water potentials of 66 species under drought and non-drought conditions. Our model successfully reproduces the measured leaf water potentials over time based on three different regulation strategies under drought. We found that three parameter sets derived from the measurement data reproduced the dynamics of 53% of an drought dataset, and 52% of a control dataset [root mean square error (RMSE) < 0.5 MPa)]. We conclude that, instead of quantifying water-potential-regulation of different plant species by complex modeling approaches, a small set of parameters may be sufficient to describe the water potential regulation behavior for large-scale modeling. Thus, our approach paves the way for a parsimonious representation of the full spectrum of plant hydraulic responses to drought in dynamic vegetation models.
Obtaining sufficient numbers of functional natural killer (NK) cells is crucial for the success of NK-cell-based adoptive immunotherapies. While expansion from peripheral blood (PB) is the current method of choice, ex vivo generation of NK cells from hematopoietic stem and progenitor cells (HSCs) may constitute an attractive alternative. Thereby, HSCs mobilized into peripheral blood (PB-CD34+) represent a valuable starting material, but the rather poor and donor-dependent differentiation of isolated PB-CD34+ cells into NK cells observed in earlier studies still represents a major hurdle. Here, we report a refined approach based on ex vivo culture of PB-CD34+ cells with optimized cytokine cocktails that reliably generates functionally mature NK cells, as assessed by analyzing NK-cell-associated surface markers and cytotoxicity. To further enhance NK cell expansion, we generated K562 feeder cells co-expressing 4-1BB ligand and membrane-anchored IL-15 and IL-21. Co-culture of PB-derived NK cells and NK cells that were ex-vivo-differentiated from HSCs with these feeder cells dramatically improved NK cell expansion, and fully compensated for donor-to-donor variability observed during only cytokine-based propagation. Our findings suggest mobilized PB-CD34+ cells expanded and differentiated according to this two-step protocol as a promising source for the generation of allogeneic NK cells for adoptive cancer immunotherapy.
Background: This study investigated whether work ability is associated with the duration of unemployment, heart rate variability (HRV), and the level of physical activity. Methods: Thirty-four unemployed persons (mean 55.7 ± standard deviation 33.3 years, 22 female, 12 male, unemployed: range 1–22.5 years) participated in the cross-sectional study. The Work Ability Index (WAI) and International Physical Activity Questionnaire (IPAQ) were applied. Short-term (five minutes) resting HRV (Low Frequency (LF), High Frequency (HF), Total Power (TP)) was collected. Results: Work ability was positively associated with the HRV: LF (r = 0.383; p = 0.025), HF (r = 0.412; p = 0.015) and TP (r = 0.361; p = 0.036). The WAI showed a positive linear correlation with the amount of total physical activity (r = 0.461; p = 0.006) as well as with the amount of moderate to vigorous physical activity (r = 0.413; p = 0.015). No association between the WAI and the duration of unemployment occurred. Conclusions: the relation between self-perceived work ability, health-associated parameters, the HRV and the level of physical activity points out the relevance of health-care exercise and the need of stress-reducing interventions to improve perceived work ability. Our results point out the need for the further and more holistic development of healthcare for the unemployed.
