Die X-chromosomal gebundene chronische Granulomatose (X-CGD) ist eine seltene Erbkrankheit, bei der die NADPH-Oxidase der Phagozyten nicht funktionell ist. Der Grund hierfür liegt meist in Mutationen in der GP91phox Untereinheit der Phagozyten-Oxidase. Hierdurch treten lebensbedrohliche Bakterien- und Pilzinfektionen bei Patienten auf, was neben einer geringen Lebensqualität zu einer erheblich verkürzten Lebenserwartung führt. Eine Stammzelltransplantation eines gesunden Spenders ist bislang der einzige heilende Therapieansatz. Für X-CGD-Patienten, die keinen passen-den Spender zur Verfügung haben, stellt die genetische Modifikation autologer hämato-poetischer Stammzellen eine alternative Form der Therapie dar. Im Jahr 2004 wurden daher in einer präklinischen Phase I/II Studie in Frankfurt zwei X-CGD-Patienten gentherapeutisch behandelt. Hierbei wurden CD34+ Stammzellen ex vivo mit einem γ-retroviralen Vektor transduziert, der eine LTR-getriebene Expressionskassette für GP91phox trägt. Nach einer nicht-myeloablativen Konditionierung wurden die genetisch modifizierten Zellen der Patienten retransplantiert. Beide behandelten Patienten zeigten schon kurz nach Therapiebeginn eine deutliche Verminderung der Infektionsanfälligkeit und somit eine stark verbesserte Lebensqualität. Auf zellulärer Ebene konnte ein gutes Engraftment der modifizierten hämatopoetischen Stammzellen im Knochenmark beobachtet werden. In funktionellen Tests konnte die Bildung superoxidproduzierender Phagozyten für die Immunabwehr gezeigt werden. Das molekulare Monitoring beider Patienten hat jedoch über die Zeit eine Verringerung der Enzymaktivität in den Phagozyten (Superoxidproduktion) gezeigt, obwohl der Anteil genetisch modifizierter Zellen nicht geringer wurde. Im Rahmen der vorliegenden Arbeit konnte durch quantitative RT-PCR-Analysen proviraler mRNA-Transkripte, eine Korrelation zwischen dem Verlust der Enzymaktivität und reduzierter Transgen-expression gezeigt werden. Durch DNA-Analysen peripherer Blutproben beider Patienten konnte eine verstärkte Methylierung an der Promotor-CpG-Insel, welche die Transgen-expression reguliert, als Ursache identifiziert werden. Weiterführende klonale Untersuchungen genmodifizierter Kolonien aus dem Knochenmark der Patienten offenbarten einen direkten Zusammenhang zwischen der Abwesenheit von Transkription bzw. Superoxidbildung und der Methylierung dieser CpG-Insel im proviralen Promotor-bereich. Somit konnte zum ersten Mal ein epigenetisches Silencing bei Patienten nach einer Behandlung mit Gentherapie nachgewiesen werden. In weiteren Versuchen konnte die vollständig ausgebildete, spezifische Methylierung des SFFV-Promotors in transduzierten Knochenmarkzellen eines Patienten durch in vitro Behandlung mit einem Methyltransferase-Inhibitor (Aza-D) in Kombination mit einem Histondeacetylase-Inhibitor (TSA) bis zu 30% reduziert werden. Dieser Teilerfolg zeigt, dass eine klinisch relevante Reaktivierung der Transgenexpression, durch Umkehrung des Silencings am SFFV-Promotor, prinzipiell möglich ist. Das Phänomen der Abschaltung der Genexpression des γ-retroviralen Vektors in der Frankfurter Gentherapiestudie, hat ein Testsytem zur Evaluierung zukünftiger Gentherapie-Vektoren erfordert. Durch Monitoring proviraler Parameter (Kopien, Transgenexpression, Proteinexpression und Promotor-CpG-Methylierung), in der murinen embryonalen Stammzelllinie P19 konnte in dieser Arbeit ein prädiktiver Silencing-Assay erfolgreich etabliert werden. Mit Hilfe dieses Systems wurden vielversprechende Silencing-resistente Vektoren mit dem UCOE (Ubiquitous Chromatin Opening Element) identifiziert. Hierdurch wurden wichtige Grundlagen geschaffen, um zukünftige virale Vektorsysteme in Bezug auf ihre Langzeitexpression testen zu können. Zusätzlich zu der Inaktivierung der transduzierten Expressionskassette konnte in beiden Patienten ein klonales Auswachsen von Subklonen beobachtet werden, das letztendlich zu einem myelodisplastischen Syndrom bei beiden Patienten führte. Der virale Enhancer war im Gegensatz zum viralen Promotor niemals methyliert, wodurch seine transaktivierenden Eigenschaften unbeeinflusst blieben. Diese enhancervermittelte Aktivierung proliferationsfördernder Gene (Mds1-Evi1-Genlokus) konnte durch RT-PCR-Analysen zunächst in Mischpopulationen aus peripherem Blut der Patienten nach-gewiesen werden. Weiterführende klonale Analysen in Knochenmarkzellen zeigten den direkten Zusammenhang zwischen der transkriptionellen Aktivierung des Mds1-Evi1-Genlokus und den proviralen Insertionen. Somit konnte die Ursache für die therapie-assoziierte, klonale Dominanz in beiden X-CGD-Patienten aufgeklärt werden. In der Frankfurter Gentherapiestudie wurde erstmals ein klinischer Erfolg für X-CGD-Patienten erzielt. Durch intensives molekulares Monitoring konnte im Rahmen dieser Arbeit aufgedeckt werden, dass der eingesetzte γ-retrovirale Vektor über das Phänomen der Insertionsmutagenese hinaus, auch in Bezug auf die epigenetische Abschaltung der Transkription (Silencing), für zukünftige Studien modifiziert werden muss. Sicherheits-verbesserte Vektoren mit einer Resistenz gegenüber Silencing in murinen embryonalen Stammzellen konnten in dieser Arbeit charakterisiert werden. Mit diesen Genfähren könnte der angestrebte Langzeittherapieerfolg in Zukunft möglich werden.
The estimation model PhytoCalc allows a non-destructive quantification of dry weight and nutrient pools of understorey plants in forests by using the relationship between species biomass, cover and mean shoot length. The model has been validated with independent samples in several German forest types and can be a useful tool in forest monitoring. However, in open areas within forests (e.g. clearcuts), the current model version underestimates biomass and produces unreliable nutrient pool estimations. Thus, tissue density, as approximated by leaf dry matter content (LDMC), is systematically higher under high light compared to low light conditions. We demonstrate that the ratio of LDMC under clearcut conditions to LDMC under forest conditions can be used to adjust the PhytoCalc model to clearcut conditions. We investigated the LDMC ratio of five exemplary species commonly occurring on clearcuts. Integrating the square of the ratio as a correction factor improved estimates of biomass to more than 70% fit between observations and predictions. Results also suggest this ratio can be used to correct nutrient concentrations modelled in PhytoCalc, which tend to be overestimated in clearcuts. As morphological groups of plant species exhibit significantly different ratios, we advise using group-specific correction factors for clearcut adjustments in the future.
European pea crabs - taxonomy, morphology, and host-ecology (Crustacea: Brachyura: Pinnotheridae)

Pinnotherids are small crabs symbiotic to a variety of invertebrates. The European species infest bivalves and sea squirts. Their way of life is parasitic and poses a threat to commercially exploited bivalves. While juveniles of both sexes still look very similar - being agile swimmers and partially free living - a metamorphosis takes place in the female after mating and results in a conspicuous sexual dimorphism. Thereafter, the female settles in its host definitely and is morphologically strongly adapted to the parasitic life phase. A very high reproductive output was demonstrated among several pea crab species infesting bivalves. Despite from that, hardly any information is present in the literature on the pinnotherids’ reproductive biology and the underlying morphology.
Due to their cryptic way of life, the sexual dimorphism, and the different morphotypes of the female, the taxonomy of the Pinnotheridae is a serious challenge. Two widely accepted species are recognized on European coasts: Pinnotheres pisum and Nepinnotheres pinnotheres. Pinnotheres pectunculi was so far only known from the bivalve Glycymeris glycymeris in its type locality Roscoff (France), while Pinnotheres ascidicola and Pinnotheres marioni were described as living exclusively in ascidians without careful comparison with the previously described species. In order to produce standardized comparative descriptions, pea crabs were collected and studied from different hosts and localities in the Northeast Atlantic and in the Mediterranean. Nepinnotheres pinnotheres and Pinnotheres pisum were redescribed with consideration to characters of female and male. According to our morphological analysis, Pinnotheres ascidicola and Pinnotheres marioni are junior synonyms of Nepinnotheres pinnotheres, whereas the status of Pinnotheres pectunculi as a valid species was ascertained. Important characters are the mouthparts, the male gonopods, and especially chelipeds that showed consistent characteristics among different crab stages of both sexes.
Based on our sampling, we estimated the host-range of the European species. Nepinnotheres pinnotheres lives in ascidians and in the pen shell Pinna nobilis. Pinnotheres pisum infests numerous bivalve species - Pinna nobilis included. For Pinnotheres pectunculi novel host records are presented, all from the bivalve family Veneridae. Furthermore, feeding of the Pinnotheres-species was observed. They use a setae comb ventrally on the claw to brush mucus (and the accumulated food particles) from the bivalve gills. Feeding strategies and host-ecology will be thoroughly discussed in consideration to other Pinnotheridae.
We investigated the reproductive systems of European pinnotherids by histological methods, scanning and transmission electron microscopy, and confocal laser scanning microscopy.
The Eubrachyura have internal fertilization: paired vaginas enlarge into storage structures, the spermathecae, which are connected to the ovaries by oviducts. Sperm is stored until the oocytes are mature and transported into the spermathecae, where fertilization takes place. In the investigated pinnotherids, the vagina is of the ‘concave pattern’. Musculature is attached alongside flexible parts of the vagina-wall to control the dimension of its lumen. The genital opening is closed by a muscular mobile operculum.
The spermatheca can be divided into two distinct regions by function and morphology. The ventral part includes the connection with vagina and oviduct and is regarded as the zone where fertilization takes place. It is lined with cuticle except where the oviduct enters the spermatheca by the ‘holocrine transfer tissue’. At ovulation, the oocytes have to pass through this multi-layered glandular epithelium, which has a holocrine mode secretion. The dorsal part of the spermatheca is lined by a highly secretory apocrine glandular epithelium, which was to date only found in fiddler crabs of the genus Uca.
The male internal reproductive system consists of paired testes and corresponding vasa deferentia. The sperm morphology of pinnotherids conforms to other thoracotremes, with slight differences between Nepinnotheres pinnotheres and Pinnotheres pisum. Spermatozoa become enveloped into spermatophores in the secretory proximal vas deferens. The medial vas deferens is strongly enlarged and stores spermatophores embedded in seminal plasma. The distal vas deferens holds tubular appendices, which extend into the ventral cephalothorax and slightly into the pleon. These appendices produce and store vast quantities of seminal plasma. The copulatory system of the Brachyura is formed by paired penes and two pairs of gonopods, which function in sperm transfer. In pinnotherids, the long first gonopods transfers the sperm mass to the female. It holds the ejaculatory canal inside, which opens proximally and distally. The second gonopod is solid, short and conical. During copulation, the penis and the second gonopod are inserted into the base of the tubular first gonopod. The second gonopod functions in the transport of the sperm mass inside the ejaculatory canal towards its distal opening. The specific shape of the second gonopod is strongly adapted for a sealing of the tubular first gonopod with longitudinal cuticle foldings that interlock inside the first gonopod. The presented results are discussed concerning their function in reproduction and in respect of the systematic account.
The role of secretion in sperm transfer, storage and fertilization among the Brachyura is still under debate. It is notable that structure and function of secretion are more complex in pinnotherids and probably more efficient than in other brachyuran crabs, which will be discussed, in view of the parasitic way of life and the high fecundity of pinnotherids.
Die Bedeutung verschiedener CRASP-Proteine für die Komplementresistenz von Borrelia burgdorferi s.s.

Die vorliegende Arbeit liefert einen wichtigen Beitrag zum Verständnis des molekularen Mechanismus der Immunevasion von B. burgdorferi s.s., insbesondere der Bedeutung einzelner CRASP-Proteine für die Komplementresistenz. Sie trägt dazu bei, die Relevanz dieser Proteine für die Pathogenese dieses Erregers zu untermauern. Im Rahmen dieser Arbeit gelang es, verschiedene Vektoren mit den ursprünglichen oder mutierten CRASP-kodierenden Genen cspA, cspZ, erpP und erpA aus B. burgdorferi s.s. zu generieren und diese in das CRASP-negative Isolat B. garinii G1 zu transformieren. Die Expression der speziesfremden Gene als auch der Transport der CRASP-Moleküle auf die Zelloberfläche von B. garinii G1 konnten nachgewiesen werden. Für die konstitutiv CRASP-1- oder CRASP-2-produzierenden Borrelienzellen konnte gezeigt werden, dass diese, auf der Zelloberfläche lokalisierten CRASP-Moleküle mit Faktor H und FHL-1 interagieren, die gebundenen Komplementregulatoren ihre funktionelle Aktivität zur C3b-Inaktivierung aufrechterhalten und die Zellen in Gegenwart von Komplement überleben. Damit wurde erstmals der Nachweis erbracht, dass beide CRASP-Moleküle unabhängig voneinander Schutz vor komplementvermittelter Lyse verleihen. Untersuchungen mit den veränderten CRASP-1-Molekülen ergaben, dass die Transformante G1/pCRASP-1 E147K eine verringerte Bindung von Faktor H und FHL-1 aufwies, welche sich jedoch nicht auf die Komplementresistenz der Zellen auswirkte. Im Gegensatz dazu führte eine Aminosäuresubstitution im C-Terminus des CRASP-1-Moleküls an Position 240 zum Verlust der Bindung von Faktor H und einer stark verminderten Bindung von FHL-1, so dass auch keine Kofaktoraktivität nachgewiesen werden konnte. Trotz des Bindungsverlustes beider Komplementregulatoren zeigte die Transformante G1/pCRASP-1 Y240A nur geringe Ablagerungen des lytischen, terminalen Komplementkomplexes (TCC) auf der Zelloberfläche und Wachstum in Gegenwart von aktiven Komplement. Mittels eines Hämolyse-Assays wurde schließlich festgestellt, dass CRASP-1 direkt mit Komponenten des Komplementsystems interagiert und dadurch die Assemblierung des TCC verhindert. Die Bedeutung der Aminosäuren an den Positionen 81, 139, 207 und 211 im CRASP-2-Molekül für die Faktor H / FHL-1-Bindung und die daraus resultierenden Auswirkungen auf die Komplementresistenz der Borrelien wurde gleichfalls nachgewiesen. Dabei wies insbesondere die Transformante G1/pCRASP-2 Y211A ein inhibiertes Wachstum in Humanserum und verstärkt Komplementablagerungen auf der Zelloberfläche auf, was auf den Verlust bzw. der sehr schwachen Bindung von FHL-1 und Faktor H zurückzuführen ist. Im Gegensatz zu den Transformanten, welche ein CRASP-2-Molekül mit nur einem Aminosäureaustausch produzierten, zeigten die Transformanten, deren CRASP-2-Molekül zwei Aminosäuresubstitutionen aufwies (G1/pCRASP-2 R139A-Y207A, G1/pCRASP-2 R139A-Y211A, G1/pCRASP-2 Y207A-Y211A) keine Bindung der beiden Regulatorproteine und keinen Schutz der Zellen vor der lytischen Wirkung von Komplement. Neue, unerwartete Erkenntnisse ergaben sich aus den Untersuchungen mit Borrelienzellen, welche das CRASP-3- oder CRASP-5-kodierende erpP- bzw. erpA-Gen enthielten. Obwohl gereinigtes als auch denaturiertes CRASP-3 und RASP-5 in der Lage war, Faktor H zu binden, wiesen die vitalen Zellen der Transformanten G1/pCRASP-3 und G1/pCRASP-5 keine Bindung von Faktor H und keinen Schutz der Zellen vor komplementvermittelter Lyse auf. Aus den durchgeführten Untersuchungen konnten für gereinigtes CRASP-3 und CRASP-5 als auch für die CRASP-3- und CRASP-5-produzierenden Transformanten neue Liganden, nämlich CFHR-2 und CFHR-5, aus Humanserum identifiziert werden. Zusammenfassend lassen sich folgende Aussagen hinsichtlich des molekularen Mechanismus der Komplementresistenz bei B. burgdorferi s.s. aus den erhobenen Daten dieser Arbeit mit transformierten Borrelienzellen formulieren: *Die Komplementresistenz der Borrelien wird durch die Faktor H- und FHL-1-bindenden Proteine CRASP-1 und CRASP-2, jedoch nicht durch CRASP-3 und CRASP-5 determiniert, *CRASP-1 als multifunktionelles Protein ist zusätzlich in der Lage, direkt mit Komplement zu interagieren, *Die C-terminalen Domänen von CRASP-1 und CRASP-2 sind für die Bindung der beiden Komplementregulatoren Faktor H und FHL-1 relevant, *CRASP-3 und CRASP-5 auf der Borrelienoberfläche lokalisiert, interagieren mit CFHR-1, CFHR-2 und CFHR-5, aber nicht mit Faktor H.
With the help of miniaturized GPS recorders I recorded 167 tracks of 48 individual pigeons during their flight from 6 different sites around Frankfurt. The experiments consisted of two main series of repeated releases from two sites 30 km north and south from the pigeons' home loft. From the site in the south the pigeons homed 12 times and from the site in the north 16 times. After the final release from these sites, the pigeons were released at 60 km distance from home. These additional sites were selected so that the pigeons would presumably fly over the previous release site with which they were highly familiar. After conclusion of the main series two additional releases were performed, one within the magnetic anomaly of the Vogelsberg and one in a magnetically quiet region. To make these releases comparable, both release sites were selected so that the distance from the home loft was 40 km. All data obtained during these experiments were subjected to a threefold analysis, mostly based on methods that I had developed by myself or adapted for this specific study. In the first step, data were analyzed traditionally, evaluating variables similar to those that can be found in current literature. I therefore calculated values that correspond to those obtained by visual observation, like virtual vanishing bearings and intervals after one minute and after 2.5 km. Additionally I calculated the efficiency of the flights and efficiencies for specific portions of each flight, to derive variables that describe the behavior after vanishing. In the second step, which served also as a preparation for the mathematical analysis, the flight of the pigeons was separated into distinctive phases of the flight by the so-called points of decision. The flight of the pigeon can usually be separated into an initial phase of flying about, a departure and/or final homing phase. In more complex cases, however, several points of decision and a multitude of intermediary phases can be defined. Yet, the initial phase, the departure phase and the final homing phase can be defined for all tracks and therefore have been selected as appropriate candidates for a thorough analysis. In the last step I employed the so-called method of time lag embedding to reconstruct the underlying navigational process of the pigeons' homing flight. This method is based on the principles of chaos theory and is regularly employed for the analysis of dynamic systems. Its application allows the reconstruction of the underlying processes from experimentally recorded data without any a priori knowledge of the underlying system itself. For these reconstructed systems I calculated characteristic properties which are unique for each system. These are the so-called correlation dimension, describing the complexity of the system, and the so-called largest Lyapunov exponent, describing its predictability. Based on the knowledge gathered from these reconstructions, I used a variation of the previous methods to identify navigational phases, by calculating the correlation dimension as a sliding mean over the complete track. From these data I then derived further characteristics of the underlying process, such as its precision and differences in complexity depending on the pigeon's current position. ...
Orthopockenviren sind große DNA-Viren, die im Zytoplasma der Wirtszelle replizieren und für über 200 Proteine kodieren. Sie besitzen ein breites Wirtszellspektrum (host-range) und modulieren auf komplexe Art und Weise zelluläre Prozesse, um ihre Replikation zu gewährleisten. Zu diesem Genus der Familie der Pockenviren gehört auch das modifizierte Vacciniavirus Ankara (MVA). MVA ist ein hoch attenuiertes, replikationsdefizientes Impfvirus, dem im Vergleich zu ursprünglichen Vacciniavirus-Stämmen viele virale Genfunktionen fehlen. Zu diesen verlorengegangenen Genen zählen so genannte host-range-Gene, die für das breite Wirtszellspektrum des Vacciniavirus (VACV) verantwortlich sind, deren molekulare Funktion aber größtenteils unbekannt ist. Diese Arbeit befasste sich zum einen mit der Untersuchung der Rolle der host-range-Gene K1L und C7L in der MVA-Infektion. Zum anderen sollte geprüft werden, ob der im MVA-Genom unvollständige Leserahmen F11L durch Wiederherstellung seiner Funktionalität den Wirtstropismus von MVA erweitern kann. Das Fehlen von K1L und C7L in MVA ist mit dem Verlust der späten viralen Genexpression verbunden. Als mögliche Ursache hierfür wurde in dieser Arbeit die Phosphorylierung des eukaryotischen Translationsinitiationsfaktors 2alpha (eIF2alpha) entdeckt, welche zum Abbruch der Proteinsynthese in der infizierten Zelle führt. Unter den möglichen Kinasen wurde die Proteinkinase R (PKR) als das verantwortliche Schlüsselenzym identifiziert und somit gezeigt, dass das K1- und C7-Protein den anti-viralen PKR-eIF2alpha-Signalweg inhibieren. Es stellte sich heraus, dass die eIF2alpha-Phosphorylierung alleine jedoch nicht für das Fehlen der späten Genexpression verantwortlich ist. Neben dem inhibitorischen Einfluss auf den PKR-eIF2alpha-Signalweg zeigte sich, dass C7 die Aktivierung des NFKB-Signalwegs reduziert, welcher für eine anti-virale Antwort der Wirtszelle wichtig ist. Ein weiterer Ansatz zur Aufklärung der K1- und C7-Funktion bestand darin, zelluläre Interaktionspartner zu identifizieren. Hierbei konnte das heterogene nukleäre Ribonukleoprotein K (HNRPK) als möglicher Interaktionspartner von C7 entdeckt werden. Neben den bisher bekannten host-range-Genen gibt es vermutlich weitere Gene, die den Wirtsbereich des VACV definieren. Das im MVA-Genom defekte F11L-Gen war ein guter Kandidat für eine solche Genfunktion, da es bei der Virionenmorphogenese, Virusausbreitung und der Migration VACV-infizierter Zellen eine Rolle zu spielen scheint. Deshalb wurde ein rekombinantes MVA mit vollständiger F11L-Gensequenz konstruiert und das Wirtszellspektrum dieses Virus untersucht. Die Reparatur des F11L-Gens ermöglichte MVA die Induktion von Zellbewegung nach Infektion, jedoch blieben seine unvollständige Morphogenese und eingeschränkte Vermehrungsfähigkeit in Säugetierzellen unbeeinflusst. F11L hat daher zumindest keine selbstständige Funktion als VACV host-range-Gen. Die Ergebnisse dieser Arbeit sind ein Beitrag zum besseren Verständnis der komplexen Virus-Wirts-Interaktionen des VACV sowie des eingeschränkten Wirtstropismus des Impfvirus MVA.
This work investigated the applicability of global pairwise sequence alignment to the detection of functional analogues in virtual screening. This variant of sequence comparison was developed for the identification of homologue proteins based on amino acid or nucleotide sequences. Because of the significant differences between biopolymers and small molecules several aspects of this approach for sequence comparison had to be adapted. All proposed concepts were implemented as the ‘Pharmacophore Alignment Search Tool’ (PhAST) and evaluated in retrospective experiments on the COBRA dataset in version 6.1. The aim to identify functional analogues raised the necessity for identification and classification of functional properties in molecular structures. This was realized by fragment-based atom-typing, where one out of nine functional properties was assigned to each non-hydrogen atom in a structure. These properties were pre-assigned to atoms in the fragments. Whenever a fragment matched a substructure in a molecule, the assigned properties were transferred from fragment atoms to structure atoms. Each functional property was represented by exactly one symbol. Unlike amino acid or nucleotide sequences, small drug-like molecules contain branches and cycles. This was a major obstacle in the application of sequence alignment to virtual screening, since this technique can only be applied to linear sequences of symbols. The best linearization technique was shown to be Minimum Volume Embedding. To the best of knowledge, this work represents the first application of dimensionality reduction to graph linearization. Sequence alignment relies on a scoring system that rates symbol equivalences (matches) and differences (mismatches) based on functional properties that correspond to rated symbols. Existing scoring schemes are applicable only to amino acids and nucleotides. In this work, scoring schemes for functional properties in drug-like molecules were developed based on property frequencies and isofunctionality judged from chemical experience, pairwise sequence alignments, pairwise kernel-based assignments and stochastic optimization. The scoring system based on property frequencies and isofunctionality proved to be the most powerful (measured in enrichment capability). All developed scoring systems performed superior compared to simple scoring approaches that rate matches and mismatches uniformly. The frameworks proposed for score calculations can be used to guide modifications to the atom-typing in promising directions. The scoring system was further modified to allow for emphasis on particular symbols in a sequence. It was proven that the application of weights to symbols that correspond to key interaction points important to receptor-ligand-interaction significantly improves screening capabilities of PhAST. It was demonstrated that the systematic application of weights to all sequence positions in retrospective experiments can be used for pharmacophore elucidation. A scoring system based on structural instead of functional similarity was investigated and found to be suitable for similarity searches in shape-constrained datasets. Three methods for similarity assessment based on alignments were evaluated: Sequence identity, alignment score and significance. PhAST achieved significantly higher enrichment with alignment scores compared to sequence identity. p-values as significance estimates were calculated in a combination of Marcov Chain Monte Carlo Simulation and Importance Sampling. p-values were adapted to library size in a Bonferroni correction, yielding E-values. A significance threshold of an E-value of 1*10-5 was proposed for the application in prospective screenings. PhAST was compared to state-of-the-art methods for virtual screening. The unweighted version was shown to exhibit comparable enrichment capabilities. Compound rankings obtained with PhAST were proven to be complementary to those of other methods. The application to three-dimensional instead of two-dimensional molecular representations resulted in altered compound rankings without increased enrichment. PhAST was employed in two prospective applications. A screening for non-nucleoside analogue inhibitors of bacterial thymidin kinase yielded a hit with a distinct structural framework but only weak activity. The search for drugs not member of the NSAID (non-steroidal anti-inflammatory drug) class as modulators of gamma-secretase resulted in a potent modulator with clear structural distiction from the reference compound. The calculation of significance estimates, emphasizing on key interactions, the pharmacophore elucidation capabilities and the unique compound rannkings set PhAST apart from other screening techniques.
In the adult mammalian central nervous system, two defined neurogenic regions retain the capacity to generate new neurons throughout adulthood, namely the subependymal zone (SEZ) at the lateral ventricles and the subgranular layer of the hippocampus (SGL). Adult neurogenesis consists of a whole set of events including proliferation, fate specification, migration, survival and finally synaptic integration of newly born neurons. Each of these events is controlled by the interplay of numerous factors. In this study two signalling systems were analysed with regard to their functional role in adult neurogenesis in vivo, namely the purinergic system and the growth factor EGF. Neither short- nor long-term application of the P2Y receptor agonists UTP and ADPβS and the P2Y receptor antagonist suramin into the lateral ventricle of adult mice altered cell responses as compared to vehicle controls in vivo. In contrast, analysis of the expansion rates of cultured neural stem cells (NSCs) from knockout mice revealed a strong increase in the number of NSCs from NTPDase2-/- mice, whereas cell numbers of NSCs from P2Y1-/- and P2Y2-/- mice were significantly reduced in comparison to wildtype levels. Notably, in vivo proliferation rates were potently elevated in the SGL and the SEZ of NTPDase2-deficient mice. However, in vivo proliferation in both neurogenic niches of the single receptor knockout mice P2Y1-/- and P2Y2-/- and P2Y1-/- P2Y2-/-double-knockout mice did not differ significantly from the wildtype. In mice lacking the P2Y2 receptor the survival of newly born neurons in the hippocampal granule cell layer was significantly increased. These data provide the first line of evidence that purinergic signalling is involved in the control of neural stem cells behaviour not only in vitro but also in vivo. In order to further characterise the role of epidermal growth factor (EGF) in adult neurogenesis, transit amplifying precursors (TAPs) and type B astrocytes were identified as EGF-responsive cell populations following ventricular EGF injection, whereas ependymal cells, neuroblasts and NG2-positive cells did not or only to a minor extent respond to EGF injection. These EGF-responsive cell populations were found on both, the septal as well as striatal lateral ventricle walls. Long-term ventricular EGF infusion for 6d, 1. increased cell proliferation of both ventricle walls revealing a gradient along the rostro-caudal axis, 2. altered the balance between neuronal and macroglial cell fates to generate oligodendrocyte precursors and 3. lead to an entire remodelling of the classical architecture of the SEZ.

Der DNA Kontrollpunkt wird bei DNA Schädigungen aktiviert, um mit Hilfe der DNA Reparatur den Schaden zu beheben und somit dafür zu sorgen, dass die DNA fehlerfrei repliziert werden kann. Der zweite Zellzyklus Kontrollpunkt, der Spindel Kontrollpunkt, stellt sicher, dass die Chromosomen während der M Phase unter gleicher Spannung innerhalb der Äquatorialplatte der Metaphase Spindel angeordnet sind. Kann in der Zeit, in der der Zellzyklus Kontrollpunkt aktiv ist, der Schaden nicht behoben werden, so wird Apoptose ausgelöst und die Zelle wird aus dem Zellverband entfernt. Krebszellen haben Strategien entwickelt, Zellzyklus Kontrollpunkte zu umgehen und darüber hinaus normalen Mechanismen der Apoptose zu entkommen. Die genauen molekularen Vorgänge der Deregulierung der Apoptose sind weitestgehend unaufgeklärt. Procaspase 8 ist ein wichtiges Schlüsselenzym des extrinsischen Apoptose Signalweges. Der extrinsische Signalweg wird extrazellulär über die Bindung von Liganden an ihre korrespondierenden Rezeptoren ausgelöst. In dieser Studie wird gezeigt, dass Procaspase 8 an Ser-387 in vitro als auch in vivo von Cdk1/Cyclin B1 phosphoryliert wird. Darüber hinaus zeigt diese Phosphorylierungsstelle die typische Struktur einer Bindungsstelle für Plk1, einer weiteren mitotischen Kinase. Die Interaktion von Procaspase 8 mit Cdk1/Cyclin B1 wird über die DE Domäne („death-effector-domain“ DED) von Procaspase 8 vermittelt. Wird Procaspase 8 an Ser 387 zu Alanin (S387A) mutiert, so wird die Phosphorylierung durch Cdk1/Cyclin B1 fast vollständig verhindert. Wird zudem diese Mutante (S387A) in humanen Krebszellen überexprimiert, so hemmt dies die Apoptose nach Stimulation des Fas Rezeptors. Wird umgekehrt Cyclin B1 mittels RNA Interferenz depletiert und dadurch Cdk1 nicht aktiviert, wird extrinsische Apoptose verstärkt. Diese Studie zeigt erstmals eine gezielte Inhibierung des extrinsischen Apoptose Signalweges durch mitotische Kinasen und schlägt ein Modell vor, in dem Serin/Threonin Kinasen extrinsische Apoptose inhibieren und somit der Tumorzelle ermöglichen, der Apoptose zu entkommen. Darüber hinaus wird ein neuartiger Mechanismus der Inhibition der autokatalytischen Spaltung von Procaspase 8 durch eine mitotische Kinase gezeigt.
Die wichtigste Aufgabe der mitotischen Spindel ist die genaue Segregation der duplizierten Chromosomen in der Mitose. Diese dynamische Struktur wird von sich teilenden Zellen gebildet, um die Bewegung der Chromosomen, das Markenzeichen der Mitose, zu dirigieren. Trotz aller Unterschiede in Form und Größe der Spindeln unterschiedlicher Zelltypen, haben alle eukaryontischen Spindeln fundamentale strukturelle Eigenschaften gemeinsam. Eine der wichtigsten Eigenschaften ist die bipolare Symmetrie. Innerhalb der Spindel sind unterschiedliche Klassen der Mikrotubuli vorhanden, die durch ihre Organisation und durch ihre dynamischen Eigenschaften unterteilt sind. Alle Klassen der Mikrotubuli zeigen dynamische Instabilität. Dennoch weisen die Kinetochor-Mikrotubuli und die Spindel-Mikrotubuli zusätzlich ein anderes Verhalten auf, das als polwärts gerichteter Mikrotubuli-Flux ("Poleward Microtubule Flux") bezeichnet wird. Dabei werden die Tubulin-Untereinheiten stetig an den Plus-Enden der Mikrotubuli eingefügt und dann zu den Minus-Enden getragen, wo sie abgebaut werden. Dieser polwärts gerichtete Mikrotubuli-Flux ist für die Segregation der Chromosomen von großer Bedeutung. Mehrere regulatorische Proteine der Mikrotubuli, einschließlich der destabilisierenden und der depolymerisierenden Proteine, steuern die Dynamik dieser Mikrotubuli, um eine fehlerfreie Bildung der Spindel und eine korrekte Segregation der Chromosomen gewährleisten zu können. Die Kinesin-13 Familie der Depolymerasen gehört zu den prominentesten Modulatoren der Dynamik der Mikrotubuli. Das am Besten charakterisierte und intensiv studierte Mitglied dieser Familie ist das Protein KIF2C/MCAK. Aufgrund der unterschiedlichen Lokalisationen von MCAK während der Mitose, an den Spindelpolen, an den Chromosomen-Armen, an den Centromeren/Kinetochoren, kann MCAK eine Reihe an wichtigen Funktionen erzielen. Allerdings bleibt die Korrektur von Kinetochor-Mikrotubuli Fehlverknüpfungen die wichtigste Aufgabe von MCAK während der Mitose. Diese wesentliche Funktion steht unter der Kontrolle von Aurora B. In dieser Arbeit konnte gezeigt werden, dass MCAK während der Mitose auch von einem wichtigen Komplex, dem Cyclin B1/CDK1 Komplex, reguliert wird. In der Tat steuert die Kinase CDK1 sowohl die Lokalisation als auch die katalytische Aktivität von MCAK. Mit Hilfe einer systematischen Reihe an Experimenten konnte nachgewiesen werden, dass MCAK sowohl in vitro als auch in vivo von CDK1 phosphoryliert wird. CDK1 phosphoryliert den katalytischen Bereich von MCAK genau am Threonin 537 und führt zu einer deutlichen Abnahme der Depolymerisierungsaktivität von MCAK in vitro und in humanen Zellen. Diese Inhibition erfolgt wahrscheinlich durch eine Reduzierung der Affinität von MCAK zu den Mikrotubuli-Enden. Die Expression der Phosphostatus-vortäuschenden Mutante T537E in HeLa-Zellen verursachte eine fehlerhafte Verteilung der Chromosomen in der Mitose. Die Chromosomen waren in Anwesenheit der T537E Mutante nicht mehr in der Lage, sich auf der Metaphaseplatte anzuordnen. Darüber hinaus führte die Expression von T537E zu einer signifikanten Reduzierung des centromerischen Abstandes, was auf eine Anhäufung von Kinetochor-Mikrotubuli Fehlverknüpfungen hindeutet. Ferner zeigt die Expression der nichtphosphorylierbaren Mutante T537A in den HeLa-Zellen eine verstärkte Lokalisation an den Spindelpolen, was zum Auftreten von erheblichen Spindel-Aberrationen führte. Basierend auf den Daten der vorliegenden Arbeit wurde ein Modell entwickelt, in dem die Phosphorylierung von MCAK durch CDK1 früh in der Mitose stattfinden muss, um einerseits MCAK zu inaktivieren und andererseits diese Depolymerase aus den Spindelpolen zu verdrängen. Beide Ereignisse sind für den Aufbau einer bipolaren Spindel unentbehrlich. Zu späteren Zeitpunkten der Mitose muss das Threonin 537 dephosphoryliert werden, um eine Reaktivierung von MCAK an den Centrosomen/Kinetochoren zu ermöglichen. Dies wird die Korrektur-Funktion für Kinetochor-Mikrotubuli Fehlverknüpfungen von MCAK wiederherstellen, was eine korrekte Anordnung der Chromosomen auf der Metaphaseplatte fördert.
