Das photoneuroendokrine System der Vertebraten steuert die rhythmische Melatoninsynthese. Melatonin ist ein wichtiges Signal für circadiane und saisonale Rhythmen und für die Synchronisation der Föten mit dem mütterlichen Organismus. Bei Säugetieren besteht das photoneuroendokrine System aus den folgenden Komponenten: der Retina für die circadiane Lichtperzeption, dem endogenen Rhythmusgenerator im Nucleus suprachiasmaticus (SCN) und dem Pinealorgan als neuroendokrinem Effektor. Dieses System vermittelt, durch die nächtliche Abgabe des Hormons Melatonin vom Pinealorgan, Änderungen in den Belichtungsverhältnissen der Umgebung an den Körper. Bei der Synthese von Melatonin im Pinealorgan ist die Arylalkylamin Nacetyltransferase (AANAT) das geschwindigkeitsbestimmende Enzym. Die nächtlich erhöhte Expression von Aanat in Pinealozyten wird vor allem durch die Freisetzung des Neurotransmitter NA aus sympathischen Nervenendigungen angetrieben. NA bindet an adrenerge Rezeptoren in der Pinealozytenmembran und aktiviert den cAMP-Signaltransduktionsweg, der zur CRE-vermittelten gesteigerten Aanat Expression führt. In der Promoterregion von Aanat ist auch ein E-box Promoterelement vorhanden, das durch Uhrenproteine angesteuert werden kann. Bislang jedoch war die Rolle des molekularen Uhrwerkes für die Expression von Aanat noch unklar. Um zu untersuchen, wie sich eine Schwächung des negativen Regulatorkomplexes auf die Expression von Aanat im Pinealorgan und in anderen Geweben auswirkt, wurden Mäuse mit gezielter Deletion des Per1 Gen (Per1 KO) untersucht. Die Expression von Aanat im Pinealorgan von Per1 KO Mäusen, die in der Standardphotoperiode gehalten wurden, zeigte einen circadianen Rhythmus mit ähnlicher Dynamik, aber erhöhter Amplitude im Vergleich zum WT. AANAT Enzymaktivität und Melatoninkonzentration folgen dem gleichen Profil. Eine Verkürzung der Photoperiode hat bei Per1 KO Mäusen starke Auswirkungen auf dieendogene Periodenlänge der Aanat Expression, die sich gegenüber dem WT drastisch verlängert. Bei einer Verlängerung der Photoperiode kommt es zu einer Verzögerung im Rhythmus der Aanat Expression von ca. 8 h gegenüber dem WT. Dies zeigt, dass das molekulare Uhrwerk je nach Photoperiode Amplitude, Periodenlänge und Phasenlage modulieren kann. Um zu untersuchen, ob es sich dabei um Pinealorgan-intrinsische Effekte handelt, wurden in vitro Experimente durchgeführt. Im WT-Pinealorgan gibt es zum Zeitpunkt CT18 ein Sensitivitätsfenster für die NA-induzierte Aanat Expression. Überraschenderweise steigt die Aanat Expression im unstimulierten Per1 KO Pinealorgan in der Nacht signifikant gegenüber dem subjektiven Tag an. Eine weitere Induktion durch NA ist nicht möglich. Dies deutet darauf hin, dass ein abgeschwächter negativer Regulator Komplex (NRC), welcher über das E-box Element wirkt, dieselben Auswirkungen in der Per1 KO Maus hat, wie eine NA-Stimulation im WT. Im WT wird der inhibitorische Effekt des NRC offenbar durch die NA-abhängige Aktivierung von CRE überwunden. Untersuchung zur ektopischen Expression von Aanat zeigten, dass dieses Gen in der Hypophyse einen cicadianen Rhythmus aufweist, der unabhängig von einem intakten molekularen Uhrwerk abläuft. Im Gegensatz dazu findet sich in der Milz von Per1 KO Mäusen eine verstärkte Aanat Expression am subjektiven Tag im Vergleich zum WT. Offenbar hat das molekulare Uhrwerk auch einen Einfluss auf die Gewebespezifität der Aanat Expression. Weiterhin wurde in dieser Arbeit die ontogenetische Entwicklung des molekularen Uhrwerkes im SCN von Melatoninrezeptor1 und 2 defizienten (MT1,2 -/-) Mäusen untersucht. Im Gegensatz zu Mäusen mit intakten Melatoninrezeptoren, zeigen diese Mäuse im Fötalstadium noch keinen Rhythmus in der Anzahl mPER1- und mPER2-Ir Zellen. In diesem Stadium sind die einzelnen SCN-Neurone noch kaum durch Synapsen miteinander gekoppelt. Dies deutet darauf hin, dass das mütterliche Melatonin die rhythmischen Uhrengenexpression in den einzelnen fötalen SCN-Zellen synchronisiert. Erst im juvenilen SCN ist ein Rhythmus der Uhrenproteine identisch mit dem adulten Tier. Zu diesem Stadium sind die intrasuprachiasmatischen Kontakte vermutlich schon soweit ausgebildet, dass kein rhythmisches Eingangssignal für die Synchronisation der SCN-Zellen notwendig ist.
By adopting a variety of shapes, proteins can perform a wide number of functions in the cell, from being structural elements or enabling communication with the environment to performing complex enzymatic reactions needed to sustain metabolism. The number of proteins in the cell is limited by the number of genes encoding them. However, several mechanisms exist to increase the overall number of protein functions. One of them are post-translational modifications, i.e. covalent attachment of various molecules onto proteins. Ubiquitin was the first protein to be found to modify other proteins, and, faithful to its evocative name, it is involved in nearly all the activities of a cell. Ubiquitylation of proteins was believed for a long time only to be responsible for proteasomal degradation of modified proteins. However, with the discovery of various types of ubiquitylation, such as mono-, multiple- or poly-ubiquitylation, new functions of this post-translational modification emerged. Mono-ubiquitylation has been implicated in endocytosis, chromatin remodelling and DNA repair, while poly-ubiquitylation influences the half-life of proteins or modulates signal transduction pathways. DNA damage repair and tolerance are example of pathways extensively regulated by ubiquitylation. PCNA, a protein involved in nearly all types of DNA transaction, can undergo both mono- and poly-ubiquitylation. These modifications are believed to change the spectrum of proteins that interact with PCNA. Monoubiquitylation of PCNA is induced by stalling of replication forks when replicative polymerases (pols) encounter an obstacle, such as DNA damage or tight DNA-protein complexes. It is believed that monoubiquitylation of PCNA stimulates the exchange between replicative pols to one of polymerases that can synthesize DNA across various lesions, a mechanism of damage tolerance known as translesion synthesis (TLS). Our work has helped to understand why monoubiqutylation of PCNA favours this polymerase switch. We have identified two novel domains with the ability to bind Ub non-covalently. These domains are present in all the members of Y polymerases performing TLS, and were named Ub-binding zinc finger (UBZ) (in polη and polκ) and Ub-binding motif (UBM) (in polι and Rev1). We have shown that these domains enable Y polymerases to preferentially gain access to PCNA upon stalling of replication, when the action of translesion polymerases is required. While the region of direct interaction between Y pols and PCNA had been known (BRCT domain in Rev1 and PIP box motif (PIP) in three others members), we propose that Ub-binding domains (UBDs) in translesion Y pols enhance the PIP- or BRCT-domain-mediated interaction between these polymerases and PCNA by binding to the Ub moiety attached onto PCNA. Following these initial studies, we have also discovered that Y polymerases themselves undergo monoubiquitylation and that their UBDs mediate this modification. This auto-ubiquitylation is believed to lead to an intramolecular interaction between UBD and Ub attached in cis onto the UBD-containing protein. We have mapped monoubiquitylation sites in polη in the C-terminal portion of the protein containing the nuclear localization signal (NLS) and the PIP box. Beside PIP, the NLS motif is also involved in direct interaction of polη with PCNA. Based on these findings, we propose that monoubiquitylation of either NLS or PIP masks them from potential interaction with PCNA. Lastly, using several functional assays, we have demonstrated the importance of all these three motifs in the C-terminus of polη (UBZ, NLS and PIP) for efficient TLS. We have also constructed a mimic of monoubiquitylated polη by genetically fusing polη with Ub. Interestingly, this chimera is deficient in TLS as compared to the wild-type protein. Altogether, these studies demonstrate that the C-terminus of polη constitutes a regulatory module involved in multiple-site interaction with monoubiquitylated PCNA, and that monoubiquitylation of this region inhibits the interaction between polη and PCNA. Our work has also revealed that the UBDs of Y pols as well as of other proteins implicated in DNA damage repair and tolerance, such as the Werner helicase-interacting protein 1 (Wrnip1), are required for their proper sub-nuclear localization. All these proteins localize to discrete focal structures inside the nucleus and mutation of their UBDs results in inability to accumulate in these foci. Interestingly, by exchanging UBDs between different proteins we have learned that each UBD seems to have a distinct functional role, surprisingly not limited to Ubbinding ability. In fact, swapping the UBZ of Wrnip1 with the UBM of polι abolished the localization of Wrnip1 to foci despite preserving the Ub-binding ability of the chimeric protein. In summary, this work provides an overview of how post-translation modification of proteins by Ub can regulate several DNA transactions. Firstly, key regulators (e.g. PCNA) can be differentially modified by Ub. Secondly, specialized UBDs (e.g. UBM, UBZ) embedded only in a subset of proteins act as modules able to recognize these modifications. Thirdly, by means of mediating auto-ubiquitylation, UBDs can modulate the behaviour of host proteins by allowing for either in cis or in trans Ub-UBD interactions.
Ziel der vorliegenden Promotionsarbeit war die Herstellung und Charakterisierung einer neuen Stat5 Reportermaus zur Analyse der transkriptionellen Aktivität von STAT5 in verschiedenen Entwicklungsstadien, Zelltypen und Organen auf Einzelzellebene in vivo. Die Zusammenfassung dieser Promotionsarbeit gibt im Folgenden einen Überblick über den JAK/STAT Signalweg und seine einzelnen Komponenten. Das Hauptaugenmerk liegt hierbei auf STAT5, da es eine wichtige Rolle in der zellulären Entwicklung, Differenzierung und Proliferation spielt. Anschließend werden die Klonierung des Stat5 Reportergenkonstruktes und die Herstellung der Reportermaus durch DNA-Mikroinjektion besprochen und die Ergebnisse sowie Schlussfolgerungen der funktionellen in vivo Analyse dieses neuen Reportermausmodells dargestellt. Signal transducer and activator of transcription (STAT) Proteine gehören zu einer Familie von Transkriptionsfaktoren, die latent im Zytoplasma vorkommen. Diese Proteinfamilie besteht aus sieben Mitgliedern: STAT1, STAT2, STAT3, STAT4, STAT5a, STAT5b und STAT6. Alle STAT Proteine weisen eine konservierte Struktur auf, bestehend aus einer N-terminalen Domäne (NTD), einer Coiled-Coil-Domäne (CCD), einer DNA-Bindedomäne (DBD), einer Linkerdomäne (LD), einer src-homology 2- Domäne (SH2) und einer Transaktivierungsdomäne (TAD). Eine Vielzahl löslicher, extrazellulärer Signalmoleküle wie zum Beispiel Hormone, Zytokine und Wachstumsfaktoren binden an ihre spezifischen Oberflächenrezeptoren und Aktivieren so die JAK/STAT Signalkaskade. Dabei führt die Ligandenbindung an den entsprechenden Rezeptor zunächst zur Dimerisierung des Rezeptors und anschließend zur Transphosphorylierung von Janus Kinasen (JAKs). Aktivierte JAKs phosphorylieren dann den Rezeptor an spezifischen Tyrosinresten. An diese können STAT Proteine über ihre SH2 Domäne binden. Die gebundenen STAT Proteine werden anschließend durch JAKs an einem Tyrosinrest (und Serinrest) in der TAD phosphoryliert und dimerisieren im Zytoplasma. Dimerisierte STAT Proteine translozieren anschließend in den Nukleus und binden an spezifische DNA-Sequenzen, die sogenannten GAS (gamma-IFN-aktivierende Seite) Elemente in der Promotorregion ihrer Zielgene. GAS Elemente sind kurze palindromische DNA Regionen mit einer TTTCCNGGAAA Konsensussequenz. Nach Bindung der aktivierten, phosphorylierten STAT Proteine an die GAS Elemente werden weitere Kofaktoren, wie zum Beispiel das CREB Bindeprotein p300/CBP rekrutiert, die gemeinsam als Transkriptionsfaktoren wirken und die Transkription ihrer Zielgene anschalten. Die Identifizierung von STAT5 erfolgte im Rahmen von Promotorstudien am β-Casein Milchgen in der murinen Brustepithelzelllinie HC11 (Schmitt-Ney et al., 1991). Kurz darauf wurde STAT5 auch im Brustgewebe von laktierenden Mäusen, Ratten und Kühen gefunden. Bevor eine Sequenzhomologie zu Proteinen der STAT Genfamilie festgestellt wurde, wurde STAT5 zunächst MGF – „mammary gland factor“ genannt (Schmitt-Ney et al., 1992b; Wakao et al., 1992). Es sind zwei Stat5 Gene bekannt, Stat5a und Stat5b, die eine Sequenzhomologie von 96 % aufweisen und ihren größten Unterschied in der TAD Domäne zeigen. Da keine STAT-ähnlichen Proteine in Hefezellen identifiziert wurden, ist der JAK/STAT Signalweg nur für multizelluläre Organismen von Bedeutung, vermutlich weil diese auf komplexe Zell-Zell Kommunikationen angewiesen sind, um im Zellverband auf Signale in der Umgebung reagieren zu können. STAT5 im Speziellen reguliert neben der Entwicklung des Brustgewebes während der Schwangerschaft, die Produktion von Blutzellen in der fötalen Leber sowie die Zellproliferation während der adulten Hämatopoese. Im Embryo ist die fötale Leber der Ort der Hämatopoese, bevor hämatopoetische Stammzellen im Knochenmark kolonialisieren und sich die Leber zu einem metabolischen Organ entwickelt. In der Maus gelangen ab dem Embryonaltag E12 hämatopoetische Stammzellen aus der Aorta, den Gonaden und dem Mesonephros (Urniere), der sogenannten AGM Region, sowie aus der Plazenta durch den Blutstrom in die fötale Leber. Die Zellen proliferieren hier und migrieren etwa zwei Tage vor der Geburt (E18) ins Knochenmark, wo die Hämatopoese nach der Geburt erfolgt. Durch die Übermittlung einer Vielzahl von Zytokinsignalen reguliert STAT5 die Differenzierung der pluripotenten Zellen in reife Blutzellen und sorgt zusätzlich für die Generierung von Zellen, die anschließend in der Lage sind, das Knochenmark zu repopulieren. Ein STAT5 Verlust führt aufgrund einer auftretenden Anämie zu einer pränatalen Letalität. Während der adulten Hämatopoese fördert STAT5 hingegen die Zellproliferation und den Zellzyklus sowie die Apoptose in hämatopoetischen Stammzellen. Im Brustgewebe ist STAT5 sowohl in der Mammogenese als auch in der Laktogenese involviert. Die Aktivierung von STAT5 erfolgt hierbei durch eine Vielzahl von Faktoren, wie zum Beispiel Prolaktin und Erythropoietin. Der Phosphorylierungsstatus von STAT5 im virgin Stadium ist hierbei gering, steigt aber während der Schwangerschaft und Laktation stetig an und führt zur Aktivierung von einer Reihe von Zielgenen wie Milchproteinen, aber auch Zellzyklusregulatoren wie CyclinD1 und negativen Regulatoren des JAK/STAT Signalweges, wie zum Beispiel SOCS3. Nach der Laktation nimmt die Phosphorylierung von STAT5 hingegen ab und aufgrund von Apoptose kommt es zu einer Rückbildung des alveolaren Gewebes. Die Regulation der Apoptose erfolgt durch eine erhöhte STAT3 Phosphorylierung. Eine Deregulierung des JAK/STAT Signalweges wird in einer Vielzahl von Tumoren beobachtet. Hier liegt STAT5 typischerweise konstitutiv aktiv vor, führt dadurch zu einer verstärkten Zellproliferation und Angiogenese und verhindert gleichzeitig die Apoptose der mutierten Zellen und eine Immunantwort, was zusammen die Tumorentstehung begünstigt. Konstitutiv aktives STAT5 spielt vor allem bei der Entstehung von soliden Tumoren wie Brustkrebs sowie verschiedenen Leukämieformen wie zum Beispiel akute und chronische myelogene Leukämien eine wichtige Rolle. Neben diesen bereits bekannten STAT5 Funktionen ist die Funktion von aktivem, phosphoryliertem STAT5 im Kontext der Mausentwicklung und in adultem Gewebe noch unklar. Um die Rolle von STAT5 während der Entwicklung näher zu charakterisieren, wurden bereits verschiedene Mausmodelle generiert. Seit dem ersten Gentransfer in Mäuse im Jahre 1980 bieten transgene Tiere eine Möglichkeit, detaillierte Einblicke in zelluläre Prozesse im Rahmen der Entwicklung, des Stoffwechsels und der Entstehung von (Krebs-) Erkrankungen zu erlangen. Transgene Mäuse wurden somit zu einem wichtigen Modellsystem, das in der Lage ist, die Mechanismen, die hinter diesen Prozessen stehen, näher zu beleuchten. STAT5a und STAT5b knock out Mäuse sind überlebensfähig, zeigen jedoch phänotypische Unterschiede. Da eine Signalweiterleitung nach Prolaktininduktion in Brustgewebszellen von STAT5a knock out Mäusen nicht erfolgt, sind diese nicht in der Lage während der Schwangerschaft zu Proliferieren und zu Differenzieren. Die Deletion von STAT5a und STAT5b hingegen ist pränatal letal und die Embryos zeigen schwere Anämien aufgrund einer erhöhten Apoptoserate der erythroiden Zellen in der fötalen Leber. Zusätzlich zu den knock-out und gain-of-function Mäusen wurde die Generierung von Reportermäusen immer wichtiger, um spezifische Signalwege im Kontext des gesamten Organismus zu untersuchen. Das Ziel dieser Promotionsarbeit war somit die Herstellung und funktionelle Analyse einer neuen Stat5 Reportermaus. Hierfür wurde zunächst ein neues Stat5 Reporterkonstrukt kloniert. Dieses Reporterkonstrukt sollte eine Vielzahl spezifischer Eigenschaften aufweisen, um speziell durch phosphoryliertes STAT5 aktiviert zu werden: (i) ein LacZ Reportergen, (ii) Stat5 Responsive-Elemente und (iii) einen minimalen Promoter. Das LacZ Reportergen wurde hierbei gewählt, um die transktiptionelle Aktivität von STAT5 in Gewebeschnitten direkt durch Blaufärbung der Zellen zeigen zu können. Bei dem gewählten Promoter handelt es sich um einen Minimalpromoter, für die Bindung genereller Transkriptionsfaktoren. Eine Aktivierung des LacZ Reportergens erfolgt jedoch nur nach vorheriger Bindung eines Transaktivators. Damit STAT5 diese Funktion übernimmt wurden zusätzliche Responsive-Elemente aus dem β-Casein Gen in das Konstrukt eingefügt. Nach erfolgreicher Klonierung von insgesamt sieben verschiedenen Stat5 Reporterkonstrukten, wurde ihre spezifische Induzierbarkeit nach STAT5 Phosphorylierung mittels transienter Transfektionsstudien in vitro analysiert und bestätigt. Das p(Stat5RE)4-CMVmin-LacZ Konstrukt wurde anschließend zwischen humane matrix attachment regions (MAR) kloniert, die als sogenannte Insulatoren fungieren. Diese sollen in der transgenen Maus verhindern, dass entfernt bindende Faktoren die Expression der Reportergenkassette positiv (enhancer) oder negativ (silencer) beeinflussen. Zusätzlich zu den sieben hier generierten Stat5 Reporterkonstrukten, wurde das p(Stat5RE)4-CMVmin-LacZ Reportergenkonstrukt im Rahmen einer Diplomarbeit in einen lentiviralen Gentransfervektor kloniert. Dieser erlaubt die stabile Transduktion von Krebszellen und Primärzellen, so dass eine ineffiziente Transfektion dieser Zellen umgangen werden kann (Gäbel, 2009). Zur Herstellung der transgenen Stat5 Reportermaus wurde das linearisierte und aufgereinigte Stat5 Reporterkonstrukt mittels DNA-Mikroinjektion in den Pronukleus von 470 Eizellen von FVB und C57BL/6 Mäusen injiziert. Die Eizellen wurden anschließend in Ammenmäuse transplantiert. Von den 470 Eizellen kamen 57 Mäuse auf die Welt. Die Integration des Transgens wurde anschließend mittels PCR und Southern Blot analysiert und die Integration des kompletten Transgens konnte in zwei der 57 Mäuse festgestellt werden. Bei beiden transgen-positiven Mäusen handelte es sich um C57BL/6 Mäuse, die anschließend mit Wildtyp C57BL/6 Mäusen verpaart wurden. Nachkommen der F2 Generation wurden dann auf die spezifische Induzierbarkeit des Stat5 Reportergenkonstruktes durch phosphoryliertes STAT5 in vivo untersucht. Da der Phosphorylierungsstatus von STAT5 im Brustgewebe bereits eingehend untersucht wurde und bekannt ist, erfolgte zunächst die Analyse der Reportergenaktivität im murinen Brustgewebe. Hierfür wurde das Brustgewebe isoliert, fixiert und über Nacht gefärbt. Anschließend wurden Paraffinschnitte hergestellt und im Detail analysiert. Im Vergleich zu Wildtyp-Kontrollmäusen konnte die Aktivierung des Reportergens im Brustgewebe in verschiedenen Entwicklungsstadien, vor allem während der späten Schwangerschaft und der Laktation, durch Blaufärbung einzelner Zellen, gezeigt werden. Eine Korrelation der Blaufärbung mit der Phosphorylierung von STAT5 in diesen Zellen wurde anhand von immunhistologischen Färbungen von Paraffinschnitten mit Antikörpern gegen Stat5 und P-Stat5 gezeigt. Zusätzlich zu der hormonell induzierten STAT5 Phosphorylierung bedingt durch eine Schwangerschaft, wurde die Aktivierung des Reportergens durch das Verabreichen von LPS gezeigt. Eine Behandlung der Stat5 Reportermäuse mit LPS führt zu einer Phosphorylierung von STAT5 in Zellen des hämatopoetischen Systems, speziell Granulozyten und Makrophagen, und sollte anschließend das LacZ Reportergen in diesen Zellen aktivieren. Dies konnte durch die Färbung von Blut- und Knochenmarkzellen mit spezifischen Oberflächenmarkern, sowie einer Färbung mit FDG (Fluoresceindi-β-D-galactopyranoside) mittels FACS Analysen bestätigt werden. Das nicht-fluoreszierende FDG wird hierbei von der exprimierten β-Galaktosidase zunächst zu Fluoreszein-monogalactosid (FMG) und anschließend zum hoch fluoreszierenden Fluoreszein hydrolysiert, was eine messbare Erhöhung der Fluoreszenz nach sich zieht. Zusammenfassend konnte das Stat5-Reportergen sowohl durch endogene Signale als auch durch extern zugeführte Signale induziert werden. Anschließend erfolgte die Analyse der Reportergenaktivierung in anderen Organen der Stat5 Reportermaus. Hierbei konnte die Aktivierung des LacZ Reportergens sowohl in der Leber (Hepatozyten), Milz (Pulpa) und Niere (Mark und Rinde) als auch im Thymus (Lymphozyten und antigen präsentierende Zellen) und im Uterus (endometrisches Epithel) bestätigt werden. Diese Ergebnisse korrelieren mit zuvor durchgeführten Western Blot Analysen, die eine Phosphorylierung von STAT5 in eben diesen Organen gezeigt haben. Zusätzlich wurde phosphoryliertes STAT5 auf Proteinebene im Herz und im Gehirn gefunden, jedoch nicht in Gewebsschnitten der β-Galactosidase gefärbten Organe. Dies deutet darauf hin, dass das Reportergen trotz der Anwesenheit von phosphoryliertem STAT5, nicht immer eingeschaltet wird und somit weitere Faktoren für die transkriptionelle Aktivität von STAT5 notwendig sind. Western Blot Analysen sind somit alleine nicht ausreichend, um eine Aussage über die transkriptionelle Aktivität von phosphoryliertem STAT5 zu treffen, so dass die im Rahmen dieser Arbeit generierte Stat5 Reportermaus einen wichtigen Beitrag zum Verständnis von aktivem STAT5 bietet. Das generierte Stat5 Reportermausmodel wurde dann im Rahmen dieser Arbeit genutzt, um die Beteiligung von aktivem STAT5 in der Entwicklung von ΔTrkA induzierter akuter myeloischer Leukämie näher zu untersuchen. Hierfür wurden lineage negative Knochenmarkszellen aus den Stat5 Reportermäusen isoliert. Dabei werden sogenannte „Lin“ Antigene (z.B. CD3, CD4, CD8, Gr-1, Ter-119) genutzt, um reife murine Blutzellen zu identifizieren. Zellen, die diese Oberflächenmarker nicht oder nur in sehr geringen Mengen exprimieren, werden als lineage negativ bezeichnet. Ein Mix monoklonaler Antikörper gegen lineage Antigene kann somit zur Isolation lineage negativer Knochenmarkszellen genutzt werden. Diese negative Selektion führt letztendlich zur Anreicherung hämatopoetischer Stammzellen oder früher Progenitorzellen, die diese Marker (noch) nicht exprimieren. Diese Progenitorzellen wurden dann retroviral mit einem ΔTrkA Konstrukt transduziert und anschließend in bestrahlte Rag-1-/- Mäuse transplantiert und repopulierten in diesen das Knochenmark. Durch die ΔTrkA Transduktion wurde in den Rag-1-/- Mäusen myeloische Leukämie induziert. Jedoch konnte im Rahmen dieser Arbeit keine Aktivierung des Stat5 Reportergenkonstruktes beobachtet werden. Dies deutet darauf hin, dass STAT5 in ΔTrkA induzierten Leukämien keine Rolle spielt und bestätigt die Annahmen von Meyer et al. Durch die hier vorgestellten Ergebnisse bestätigt sich sowohl die Generierung eines neuen Stat5 Reportermausmodels als auch ihre spezifische Induzierbarkeit sowohl durch endogene hormonelle Prozesse (Schwangerschaft) als auch durch externe Manipulation (LPS Behandlung). Diese neue Stat5 Reportermaus wird in Zukunft als wichtiges und effizientes Modell fungieren, um die Rolle von transkriptionel aktivem STAT5 näher zu beleuchten. Hierbei wird sich der Fokus nicht nur auf die Rolle einzelner Zellen bei der normalen Entwicklung von Organen während verschiedener Entwicklungsstadien beschränken, sondern sich mehr und mehr in Richtung Tumorinitiierung und Tumorentwicklung bewegen. Anhand des hier generierten Stat5 Reportermausmodels können in Zukunft weitere Brustkrebs- und Leukämie-Tumormodelle herangezogen werden, um die Rolle und Funktion von STAT5 in der Tumorentwicklung in vivo detailliert analysieren zu können. Auf diesen Ergebnissen aufbauend wird dann die Möglichkeit bestehen, dieses neue Stat5 Reportermausmodell als Plattform zu nutzen, um zahlreiche neue Krebsmedikamente zu entwickeln und zu evaluieren.
In mitochondrial respiration, the soluble protein cytochrome c accepts an electron from the membrane bound cytochrome bc1. The interaction between cytochrome bc1 and cytochrome c is highly transient in nature, enabling turnover numbers greater than 160 s-1. Yeast cytochrome bc1 has been successfully crystallised with bound cytochrome c with the help of an antibody fragment (Lange and Hunte 2002; Solmaz and Hunte 2008). In all crystal structures of the complex, the homodimeric cytochrome bc1 binds only one cytochrome c, with the binding site located on subunit cytochrome c1. Univalent cytochrome c binding is correlated with conformational changes of the Rieske protein head domain and subunit QCR6p. The interface of the complex is small. The haem moieties are centrally located in a mainly non-polar contact site that includes a cation–! interaction and is surrounded by complementary charged residues. The crystal structure is in agreement with the general architecture of the interfaces of transient redox complexes and also reveals several interesting features unique to the cytochrome bc1. On the basis of the crystal structures, an extensive thermodynamic and kinetic characterisation of the interaction was carried out in this work to challenge the static snapshot of the bound proteins in the crystal structure as the relevant physiological electron transfer. The thermodynamic parameters of the interaction between the redox partners were determined using isothermal titration calorimetry (ITC). The association constant for cytochrome bc1 and cytochrome c in oxidised state under physiological ionic strength of 120 mM at 25 °C, was determined to be 5 " 103 M-1 by direct ITC titration. So, the partners interact with an affinity of 200 #M. In spite of the low affinity the complex has a life time ($ = 1/koff) of 5 #second, sufficiently long to enable the theoretically calculated electron transfer rates of 1.0 " 106 to 2.6 " 107 s%1 with a lifetime ($ = 1/rate) of 1-0.04 μseconds and experimentally determined rate of 7.7 " 104 s%1 with a lifetime of 13 μseconds. The low affinity makes it difficult to ascertain the stoichiometry of binding. The enthalpy of the interaction is endothermic, which is consistent with the nature of an interface where hydrophobic interactions are dominant. The enthalpy and entropy is 3.6 kJmol-1 and 83 kJmol-1K-1, respectively. The importance of key interface residues was also investigated. The role of the interface residue G89 of cytochrome c which might have a role in the dissociation of the complex has been probed by site-directed mutagenesis. The interface contains a cation-! interaction between F230 of cytochrome bc1 and R19 of cytochrome c, which is thought to provide the specificity to the interaction between the otherwise promiscuous partners. To analyse the role of this interaction pair in electron transfer, F230L and F230W mutants were used to measure direct electron transfer rates by flash photolysis and steady state kinetics. The findings indicate that another ! system can work as functional substitution of F230, while deleting the ! system has a deleterious effect on the complex formation. The inability of F230L to achieve the transient and steady state turnover rates as wild type protein indicates a scenario where the variant achieves an altered bound state with inefficient electron transfer pathways and higher edge-to-edge distance. The role of supernumerary subunit QCR6p in complex formation was investigated by steady state kinetics measurements. Subunit QCR6p does not interact directly with cytochrome c but is positioned in such a way that it could electrostatically steer cytochrome c in a reactive ensemble. The highly acidic and disordered N-terminus of QCR6p could interact with a patch of conserved lysine residues on cytochrome c. The role of subunit QCR6p has been assessed using QCR6p deleted cytochrome bc1 and a lysine variant of cytochrome c. The results show that QCR6p not only affects the kinetics of the interaction but is also important for the stability of cytochrome bc1. The kinetic and thermodynamic data obtained during this study provide evidence for the functional importance of non-catalytic cytochrome bc1 subunit QCR6p, show that the entropy driven interaction is indeed of low affinity and highly transient in nature and indicate that the interface is well suited to ensure the high turnover of the electron transfer chain where cytochrome c interacts with multiple partners using overlapping interfaces. The suggested role of the cation-! interaction as a highly specific interaction has been validated.
The documentation of life on Earth, that is, the inventorization of nature and the naming and classification of organisms found therein, is a major task for biologists today and a fundamental precondition for nature conservation efforts. This study aimed at contributing to the inventory of amphibians and reptiles in selected, previously understudied ecoregions of Bolivia. I strove to document diversity patterns and seek possible ecological and historical reasons for these patterns. Special attention was paid to the Chiquitano Region situated in the eastern lowlands of Bolivia in a climatic transition zone between the humid evergreen Amazon Forests and the deciduous thorn-scrub vegetation of the Gran Chaco. In congruence with its location in the transition zone, the Chiquitano Region displays a mosaic of habitats: The vegetation is dominated by the endemic Chiquitano Dry Forest, which is probably the largest extant patch of Seasonal Dry Tropical Forest, with enclaves of savanna, the western outliers of the Cerrado biome of central Brazil. Taxonomic revisions: The taxonomic data in this study are used as a tool to measure biodiversity, to assess biogeographic relationships, and to evaluate conservation needs. Since all is predicated on the taxonomic decisions made, an adequate taxonomy is essential, and taxonomy can be regarded as the foundation of this study. The methodology encompassed a variety of herpetological field techniques, such as different survey methods, preparation and documentation of voucher specimens, recording of frog calls, and herpetological laboratory techniques, such as morphology, molecular procedures with mtDNA, phylogenetic analyses, and bioacoustic analysis and descriptions of frog calls. A total of 1251 specimens belonging to 200 species were obtained during this study, including 87 amphibian and 123 reptile species. This constitutes about 36% of the herpetofauna currently known for Bolivia, about 34% of the amphibians currently known for Bolivia and about 40% of the reptiles, respectively. In the course of this study, a new species of frog was described from the study site Caparu in the eastern lowlands of Bolivia; this species, Hydrolaetare caparu Jansen, Gonzales & G. Köhler 2007, differs from the other two congeners in external morphology (e.g., lateral fringes and relative length of fingers, size of palmar tubercle, webbing of toes, and colouration) and advertisement call. Two new colubrid snake species were also described from the study site San Sebastián. Thus far, both are known only from the Chiquitano Region, Provincia Ñuflo de Chávez. Phalotris sansebastiani Jansen & G. Köhler 2008 differs from all the other species in the genus in having a triangular projection of the red snout colouration reaching onto the parietals. Xenopholis werdingorum Jansen, Gonzales & G. Köhler 2009 can be identified as a member of the genus Xenopholis by its vertebral morphology. It differs from the other two species of Xenopholis in having a unique uniform dorsal colour pattern, and from X. scalaris in having two prefrontals and a narrow septum within the neural spine and perpendicular to its long axis as evident in the x-ray images. A review of a small collection of pitvipers from different lowland localities and from the Inter-Andean dry valleys of the region of Pampagrande revealed one new species of Bothrops and one of Bothrocophias (both to be formally described elsewhere). The two pitviper species differ morphologically and genetically from their congeners. The results of a brief review of a small collection of frogs of the genus Scinax (Anura: Hylidae) from different localities in the lowlands, together with analyses of their bioacoustics, suggest an unknown cryptic diversity in Bolivian species of Scinax cf. fuscomarginatus and allies. However, further studies are necessary to clarify the taxonomic status of these populations. In addition, this study provides new data on the morphology (e.g., pholidosis) of snakes, many of them previously known only from few museum specimens. Keys to the Bolivian lizard species of Cercosaura and the Bolivian snake species of Chironius, Clelia, Liophis, Lystrophis, Phalotris, and Xenodon are presented here for the first time. New information on distribution includes many range extensions of amphibian and reptile species, such as five new country records (one frog species, four snake species) and six new departmental records (two frog species, four snake species). Observations on ecology and natural history: Several observations on ecology and natural history were made during field work. Visual signaling, an aspect of territorial behavior that was already known for several species of the genus Phyllomedusa, could be described for the first time for Phyllomedusa boliviana (Jansen & J. Köhler 2007). Furthermore, during audio surveys of an anuran community at the study site San Sebastián from 2005 to 2007, a decline of certain amphibian populations was observed in the rainy season 2006/2007 (Jansen et al., in press). This is possibly related to an extreme drought in the dry season of 2006 where 158 consecutive days without rainfall were recorded. In addition, a new method for measuring intensity of anuran choruses by means of a continuous sound pressure metre was developed (Jansen 2009). The method was suitable to detect calling phenology (during one night), as well as differences in calling activity (between two nights). Biodiversity and biogeographical relationships: Species lists were compiled at the six study sites Pampagrande, Los Volcanes, San Sebastián, Caparú, El Espinal und El Corbalan. The total amphibian and reptile species numbers observed ranged from 37 to 101 with the highest species numbers in San Sebastián (101) and Caparú (89) and the lowest in Los Volcanes (37) and El Espinal (41). A preliminary species list of the herpetofauna of the Chiquitano Region was presented, including 60 amphibian and 84 reptile species. The majority of the amphibians of the Chiquitano Region are classified predominantly as inhabitants of open formations (41 species, 68.3%). Interestingly, even the majority of species recorded from the Chiquitano Dry Forest (32 species) are usually associated with open formations (22 species, 66.7%), followed by the number of species associated with open and forest formations (8 species, 24.4%). Only two of the observed species (6.0%) are predominant forest dwellers. The amphibian assemblage of the Chiquitano Region is most similar in composition to that of the Cerrado biome: 46 species (76.7%) occur in the Cerrado as well, and three species are regarded as Cerrado endemics (5.0%). The Chiquitano Region shares considerably fewer amphibian species with the other biomes (Amazon: 22 species, 36.7%; Gran Chaco: 13 species, 21.7%; Caatinga: 16 species, 26.7%). The reptile assemblage also has significant affinities to the Cerrado, which can be seen in the high proportion of reptile species distributed in that biome (68 species; 81.0%). Affinities to the other biomes are as follows: Amazon (48 species, 57.1%), Chaco (37 species, 40.1%), and Caatinga (30 species, 35.7%). When arranged in mutually exclusive biome categories, reptiles and amphibians showed similar patterns so that the majority of both amphibians and reptiles of the Chiquitano Region can be regarded as widespread. The high proportion of reptile species probably endemic to this region (5 species, 6.0%) is remarkable (i.e. Tropidurus xanthochilus, Apostolepis phillipsi, Phalotris sansebastiani, Xenopholis werdingorum, and Micrurus diana). In an analysis of the biodiversity patterns and biogeographical relationships of the herpetofauna of the study sites, these sites were compared with literature data from 37 localities and included in a presence/absence matrix with a total of 657 amphibian and reptile species in the surrounding South American biomes Amazon, Cerrado and Gran Chaco. The biogeographic relationships between these sites were evaluated using the Coefficient of Biogeographic Resemblance (CBR), cluster analysis, and multidimensional scaling (MDS) of sites. The analyses were first conducted on amphibians and reptiles combined, and than group-specific each for amphibians, reptiles, lizards, and snakes, separately. A “bias-reduced analysis” was developed for a better understanding of the affinities of the amphibians. In this analysis, e.g., the distinct habitat types of the Chiquitano Region, the Chiquitano Dry Forest and the Cerrado were taken into account. Analyses of the biodiversity patterns revealed that the sites in the Amazon comprise highest species numbers, as expected, followed successively by the sites in the Cerrado biome and sites in-between the two biomes. Within the eastern lowlands of Bolivia, the Chiquitano Region is the most rich in species. Comparing it with the other South American sites, the Chiquitano Region has a surprisingly high alpha diversity, especially in amphibians. The microgeographic variation in species composition (beta diversity) in the Chiquitano Region is also remarkably high and obviously related to the mosaic character of the vegetation and habitats. However, the bias-reduced analysis revealed that the amphibian fauna of the open areas and savannas at Hacienda San Sebastián (with 36 species in the Cerrado and pastureland) was one of the most species-rich savanna sites known for amphibians in South America. Considering that the Hacienda San Sebastián site is only ca. 3300 ha (= 1.29 amphibian species per km2), this outcome is particularly suprising. The results of the analyses of the biogeographical relationships suggest that the herpetofauna of Bolivia’s lowlands, including the Beni, the Pantanal and the Chiquitano Region, is as distinct from the herpetofauna of the Gran Chaco, Amazon, and Cerrado as these biomes are from each other. The Chiquitano herpetofauna in particular represents a unique and well-defined herpetofaunal assemblage when compared to all surrounding localities and biomes. This is supported by high CBR-values, findings from the cluster analysis, as well as a clear separation of the Chiquitano sites in the MDS. Biogeographic relations exist in all the surrounding biomes, but are strongest to Cerrado, followed by the Amazon. This study strongly suggests that the Chiquitano herpetofauna is composite and has multiple affinities. This is congruent with a well-defined Chiquitano flora, avifauna and mammalian fauna, suggesting a similar history. The bias-reduced analysis revealed a more detailed picture of the biogeographic relations of the Chiquitano Region, especially the Chiquitano Dry Forest. I argue here that the Chiquitano Dry Forest herpetofauna is a “young”, and “former savanna herpetofauna”. Whereas the Chiquitano Dry Forest is rather poor in amphibian and reptile species, and endemics are lacking from this forest type, the isolated Cerrado enclaves are especially diverse in species and probably contain locally endemic species, such as Phalotris sansebastiani and Xenopholis werdingorum. The colonization of the young Chiquitano Dry Forest may have taken place from savannas by mainly open area species, and only briefly through the Amazon. The results emphasise the importance of bias-reduction in studies of biogeography, e.g., by using group-specific analyses or by taking into account criterias as area size and heterogeneity of compared sites. The different biogeographic patterns of reptiles and amphibians of the Andean valleys indicate a different history of these two groups. In regard to reptiles, dispersals and withdrawals into the valleys in warm humid and dry cool periods in the Pleistocene seem likely, supported by a relation between the valleys and the dry lowland (e.g., Chaco). However, it is more plausible that, during these climatic fluctuations, amphibians migrated to adjacent, more humid regions, such as Yungas. The study verified the known patterns of sister-species pairs in the Inter-Andean Dry Forest and the lowlands. Additionally, pairs of populations with slight differences in morphology were found in the valleys and in the lowlands (Cercosaura parkeri and Xenodon rhapdocephalus). Further studies must test the taxonomic status of these populations. The discovery of new species of Bothrops and Bothrocophias from the Andean valleys has several implications, and possible reasons for the high endemism in the dry valleys are discussed. Conservation and outlook: The high local alpha and beta diversity of the Chiquitano herpetofauna shows that this is a region of complex faunal interaction, which reflects the present heterogeneity of the region, but which is possibly also related to a complex geological and environmental history. The Chiquitano Region can be assessed as a region of distinct regional herpetofaunal diversity charaterised by small scale diversity patterns. It therefore merits recognition as a unique ecoregion, and conservation effort should be increased. Further research is necessary to solve the taxonomic problems addressed in this study. Moreover, future work should be directed towards the development and institution of longterm monitoring programs to evaluate the effects of climate change and changes in land-use on biodiversity, especially that of the Chiquitano Region.
In dieser Arbeit wurden die physiologische Funktion innerhalb der Ribosomenbiogenese und die physikalischen Interaktionen des nukleolären, essentiellen Proteins Nep1p in der Hefe Saccharomyces cerevisiae untersucht. Durch Hefe-Zwei-Hybrid-Experimente und biochemische Analysen konnte eine Homodimerisierung des Proteins festgestellt sowie eine strukturabgeleitete Dimerisierungsmutante identifiziert werden. Ebenfalls aus der Struktur des Nep1p-Homologs aus Methanocaldococcus jannaschii konnte eine Nop14p-Bindungsregion auf der der Dimerkontaktfläche abgewandten Seite des Hefeproteins vorhergesagt und nach in vitro-Mutagenese bestätigt werden. Innerhalb des Nop14-Proteins wurden zwei Domänen charakterisiert, die im Zwei-Hybrid-System mit Nep1p interagieren. Aus Strukturdaten in Kombination mit Hefe-Drei-Hybrid-Experimenten konnte die RNA-Bindungsregion an der Dimerkontaktfläche des Nep1-Proteins lokalisiert werden. In Drei-Hybrid-Selektionen wurden RNA-Sequenzen mit hoher Affinität zu dem M. jannaschii Nep1p identifiziert, die auf eine Bindung des Proteins bei Helix 35 der 16S rRNA schließen lassen. Aufgrund der hohen Konservierung dieser rRNA-Region ist eine Bindung des Hefeproteins an die 18S rRNA-Schleife von Nukleotid 1189-1196 sehr wahrscheinlich. Da Nep1p eine große Ähnlichkeit zu Proteinen der SPOUTFamilie von Methyltransferasen aufweist, war von einer rRNA-Methylierung im Verlauf der Ribosomenbiogenese als katalytische Funktion des Proteins auszugehen. Aus verschiedenen Drei-Hybrid-Experimenten zur RNA-Bindungungsspezifität ergab sich als mögliche Reaktion die N1-Methylierung des Nukleotids 1-Methyl-3-(3-Amino-3-Carboxypropyl)-Pseudouridin (m1acp3Y) 1191 der 18S rRNA. Durch eine spezifische radioaktive Markierung der acp-Gruppe konnte gezeigt werden, dass Nep1p keinen Einfluss auf die spätere Aminocarboxypropylmodifizierung hat. Diese findet auch bei einer Deletion der snoRNA35 statt, also auch an einem Uridin, und ist unabhängig von dem cytoplasmatischen Protein Tma20p. In RP-HPLC-Experimenten konnte nachgewiesen werden, dass die 18S rRNA einer Dnep1Dnop6-Doppelmutante ein Aminocarboxypropyl-modifiziertes Nukleosid enthält, dass sich in seinem Retensionsverhalten von dem m1acp3Y eines Wildtyps unterscheidet. Bei dem in diesem Stamm detektierten acp-modifizierten Nukleosid handelt es sich vermutlich um ein nicht-methyliertes acpY, was eine Funktion von Nep1p als N1-Methyltransferase des Nukleotids Y1191 der 18S rRNA höchst wahrscheinlich macht. Diese katalytische Funktion konnte in Zusammenarbeit mit Prof. Wöhnert auch für das M. jannaschii Nep1p gezeigt werden. Dass sowohl eine snr35- Deletion als auch eine 18S rRNA-Mutation des Nukleotids 1191 nicht letal sind, machte deutlich, dass die N1-Methylierung nicht die essentielle Funktion von Nep1p darstellen kann. Weiterhin konnte nachgewiesen werden, dass die Suppression der nep1-1ts-Mutante durch S-Adenosylmethionin nicht auf der Unterstützung der Methyltransferase-Aktivität des Proteins, sondern vermutlich eher auf einer generellen Stabilisierung des temperatursensitiven Proteins beruht. Sowohl im Hefe-Nep1p als auch im humanen Homolog wurden durch biochemische und genetische Experimente mehrere Phänotypen der Bowen-Conradi-Mutation (Aspartat 90 zu Glycin in ScNep1p) nachgewiesen. Diese lassen auf eine Aggregation des mutierten Proteins sowie eine dadurch bedingte Fehllokalisation innerhalb der Zelle schließen. Zusätzlich ist aber auch ein RNA-Bindungsdefekt durch den Aminosäureaustausch wahrscheinlich. Nichtsdestotrotz liegt offensichtlich ausreichend Nep1p-Protein vor, dass seine essentielle Funktion erfüllen kann, da die Mutation selbst zu keinem Wachstumsphänotyp führt. Erst bei einer partiellen Translationsrepression des mutierten Proteins unter Verwendung des artifiziellen Tetrazyklin-Aptamer-Systems ist ein verlangsamtes Wachstum von Hefezellen zu beobachten, was dieses System geeignet zur Analyse von möglichen Therapeutika macht.
The mTOR kinase inhibitor rapamycin (sirolimus) is a drug with potent immunosuppressive and antiproliferative properties. We found that rapamycin induces the TGF/Smad signaling cascade in rat mesangial cells (MC) as depicted by the nuclear translocation of phospho-Smads 2, -3 and Smad-4, respectively. Concomitantly rapamycin increases the nuclear DNA binding of receptor (R)- and co-Smad proteins to a cognate Smad-binding element (SBE) which in turn causes an increase in profibrotic gene expression as exemplified by the connective tissue growth factor (CTGF) and plasminogen activator inhibitor 1 (PAI-1). Using small interfering (si)RNA we demonstrate that Smad 2/3 activation by rapamycin depends on its endogenous receptor FK-binding protein 12 (FKBP12). Mechanistically, Smad induction by rapamycin is initiated by an increase in active TGF1 as shown by ELISA and by the inhibitory effects of a neutralizing TGF antibody. Using an activin receptor-like kinase (ALK)-5 inhibitor and by siRNA against the TGF type II receptor TGF-RII) we furthermore demonstrate a functional involvement of both types of TGF receptors. However, rapamycin did not compete with TGFfor TGF-receptor binding as found in radioligand-binding assay. Besides SB203580, a specific inhibitor of the p38 MAPK, the reactive oxygen species (ROS) scavenger N-acetyl-cysteine (NAC) and a cell-permeable superoxide dismutase (SOD) mimetic strongly abrogated the stimulatory effects of rapamycin on Smad 2 and 3 phosphorylation. Furthermore, the rapid increase in Dichlorofluorescein (DCF) formation implies that rapamycin mainly acts through ROS. In conclusion, activation of the profibrotic TGFSmad signaling cascade accompanies the immunosuppressive and antiproliferative actions of rapamycin. The physiology of our most complex organ, the brain, is still not comprehensively understood. The brain basically serves the processing, storing and binding of external and internal information, and thereby generates amazing phenomena like the understanding of oneself as an individual entitiy. How exactly information is encoded and represented, how individual neurons or networks of neurons actually interact, is a gigantic puzzle, whose pieces were collected since many decades. Subject of scientific discussions are the basic spatiotemporal structures of neuronal representations. Suggestions and observations reach hereby from simple rate coding of individual neurons to synchronous activity of larger ensembles. To approach answers to these questions, our working group has used a combination of different recording techniques that allowed for the comparison of neuronal interactions on different spatial scales. We focused on prefrontal neuronal interactions during visual short-term memory. Herefore two rhesus monkeys had been trained to perform a visual short-term memory task. We measured and recorded their neuronal activity by means of a microelectrode matrix that could be inserted into the cortex via a closable chamber, which had been previously implanted above prefrontal cortex. The acquired signal was separated into two components: a high-frequency component, that represents the spiking output activity of few neurons in the vicinity of each electrode tip (multi-unit activity), and a low-frequency component, that results from dendritic input activity of larger neuronal assemblies (local field potential). From one of the experimental animals we also recorded mass signals of even larger neuronal populations by means of small silverball electrodes, that had been implated into the skull above prefrontal cortex (skull EEG) in the context of a pilot project. In the first subproject, we analyzed the selectivity of output signals with respect to the memorized stimulus and task performance. We compared selectivities of local recording sites (multi-unit activity) with the selectivities of patterns created by the combined activity of all recording sites, thus representing the activity of large and distributed ensembles. Local neuronal activity correlated with the course of the visual short-term memory task, but was not highly discriminative with respect to different visual stimuli. We could show that the population activity was significantly more specific. Concerning task performance, we obtained the same result, albeit less pronounced. Further analyses revealed that the patterns of distributed ensemble activity were only partly based on realtime coordination of neuronal activity, and in addition, did not remain stable across the time course of the short-term memory task. In the second subproject, we focused on the oscillatory behavior of the local field potential. After a time-frequency analysis, we studied different frequency bands concerning stimulus selectivity and task performance of the monkey. We hereby found significant modulations of oscillations in the beta- and gamma-frequency range, that correlated with different periods of the task. Especially for oscillations in beta- and low-gamma-range, we observed phase-locking of oscillations between different recording sites, which could play an important role as internal clock to coordinate spatially separate activity. Local high-gamma oscillations themselves seemed to be important for the maintenance of information. These results could be partly confirmed by mass signals of EEG. In sum, our results support the hypothesis that information is represented in the brain by means of concerted activity of spatially distributed neuronal ensembles. This activity again appears to be coordinated by oscillatory activity in beta- and low-gamma-frequency ranges. A deeper understanding of central nervous information processing could contribute to better treatment of diseases like Parkinson’s, Alzheimer’s as well as epilepsy, and neuropsychiatric disorders like schizophrenia.
Die Brustdrüse (glandula mammaris) bietet ein einzigartiges Modellsystem zum Studium der adulten Stammzellen und der molekularen Signalwege, welche die Selbsterneuerung dieserZellen sowie die Proliferation und Differenzierung der Vorläuferzellen kontrollieren. Die Brustdrüse besteht aus dem Brustepithel und dem Stroma, das zum größten Teil aus dem Fettgewebe gebaut ist. Es enthält auch andere Zelltypen z. B. Fibroblasten und Makrophagen. Die Entwicklung der Brustdrüse findet hauptsächlich nach der Geburt, während der Pubertät, Schwangerschaft und Laktation statt. Ein funktionelles Brustepithel wird während der aufeinander folgenden Zyklen von Schwangerschaft, Laktation und Abstillen auf- und wieder abgebaut. Diese Regenerations-Kapazität kann für die Organrekonstitution genutzt werden. Die Transplantation der kleinen Anzahl von Brustepithelzellen oder des Drüsenfragments in das Fettgewebe einer Empfängermaus, deren eigenes Brustepithel entfernt wurde (cleared fat pad), führt zur vollständigen Epithelregeneration. Die zyklische Entwicklung und Regenerations-Fähigkeit des Epithelgewebes lässt auf die Existenz von Stammzellen schließen, die im Verbund der Epithelzellen überdauern. Diese gewebespezifischen Stammzellen sind in der Lage sich durch asymmetrische Zellteilung zu erneuern (self-renewal) und gleichzeitig die differenzierenden Vorläuferzellen zu bilden. Die während der Pubertät und Schwangerschaft erhöhten systemischen Hormone, lokalen Wachstumsfaktoren und Zytokine kontrollieren die Stammzellen-Proliferation und die Differenzierung der Vorläuferzellen in den verschiedenen Brustepithel-Zelllinien: Myoepithel-, Luminal- und Alveolarzellen. Aufgrund der Tatsache, dass die Entstehung von Brustkrebs mit aberranten Proliferations- und Differenzierungsprogrammen in malignanten Stamm-/ Vorläuferzellen (cancer stem cells) einhergeht, ist die Identifizierung der Signalwege, die diese Prozesse regulieren, für die Stammzellen- und Krebs-Forschung sehr bedeutend. Basierend auf diesen Erkenntnissen wurden im Rahmen des vorliegenden Projektes die Methoden zur genetischen Manipulation von nicht-angereicherten Brustdrüsen-Stammzellen entwickelt. Durch effiziente lentivirale Transduktion von adhärenten Primärzellen wurden nahezu 90% der Zellen, einschließlich der Stammzellen, transduziert. Diese Optimierung erfolgte durch 1) die Anwendung von konzentrierten Lentiviren mit hoher Qualität, 2) Passagierung der Primärzellen und Entfernung von Gewebeklumpen von den VIII Primärzellkulturen, und besonders 3) durch die Reduzierung der Zelldichte während der viralen Transduktion. Für Brust-Stammzellen sind keine spezifischen Oberflächen-Marker bekannt und daher ist ihre Isolierung deutlich erschwert. Man konnte sie bis jetzt nur anhand der moderaten Expression von CD24 (hitzestabilen Antigen) und hoher Expression von CD49f oder CD29 (α6- oder β1-Integrin) ungefähr 10-fach anreichern. Allerdings haben andere Studien gezeigt, dass die Transplantation der FACS-sortierten Stammzellen zu einer Schädigung der Stammzellen und folglich zu einer Reduktion der Repopulation-Frequenz führen kann. Aus diesem Grund wurden die genetisch modifizierten Stammzellen nicht sortiert. Durch die Transplantationen der transduzierten Primärzellen wurde ihr Stammzellen-Anteil in ihrer natürlichen Nische (cleared fat pad) selektiert. Die transplantierten Stammzellen sind in der Lage duktale Auswüchse zu entwickeln. Mit dieser Strategie konnten Transplantate mit homogener Expression von Fluoreszenz-Markergenen, wie z. B. GFP, erzielt werden. FACS Analysen der Zellen, die aus Transplantaten isoliert werden, haben gezeigt, dass alle drei Brustepithelzell-Populationen, nämlich Luminal-, Basal- und Stammzellen, transduziert waren und GFP exprimierten und daher aus transduzierten Zellen hervor gingen. Die Transplantationen einer Mischung der unterschiedlich fluoreszenzmarkierten Stammzellen ergaben einzelne verzweigte Auswüchse, in denen jeweils nur ein Fluoreszenz-Markerprotein exprimiert wurde. Sie stammen sehr wahrscheinlich von einzelnen transduzierten Stammzellen ab und wachsen jeweils in einem begrenzten Bereich des Brustfettgewebes aus. Die Immun-Antwort der Empfängermäuse gegen Fluoreszenz- Markerproteine könnte das Auswachsen der Transplantate inhibieren. Brustepithelium-Rekonstitutionen waren daher in den Rag2-/-γc-/- Empfängermäusen mit geschwächtem Immunsystem besonders effiziert. Die lentivirale Manipulation von Stammzellen und deren Einsatz in Brustepithelium-Rekonstitutionen kann als alternative Methode zur gewebsspezifischen Knockout-Technik angesehen werden. Für die Etablierung dieser Methode wurde im Rahmen dieser Arbeit ein zentraler Transkriptionsfaktor in der Brustentwicklung, signal transducer and activator of transcription 5 (Stat5), untersucht. Kollagen des marinen Schwammes Chondrosia reniformis Nardo: Optimierte großtechnische Herstellung, Analytik und neue pharmazeutisch-technologische Anwendungsmöglichkeiten Großtechnische Isolierung und Untersuchung von Chondrosia-Kollagen Schwämme spielen eine wichtige Rolle im Ökosystem des Meeres und ihr Vorkommen ist begrenzt. Zur schonenden und nachhaltigen Nutzung als Rohstoffquelle müssen im Hinblick auf eine industrielle Nutzung alternative Wege zur Gewinnung von marinem Schwammkollagen entwickelt werden. Ein Beispiel hierfür sind die sogenannten „Marikulturen“ direkt im Meer, aus denen die im Rahmen dieser Arbeit verwendeten Schwämme stammten. Im Gegensatz zu den früher eingesetzten Methoden zur Isolierung des Chondrosia-Kollagens, die Laborcharakter trugen, konnte in der vorliegenden Dissertationsarbeit ein stark vereinfachtes, für die großtechnische Produktion geeignetes Verfahren mit einer hohen Ausbeute (ca. 35%) beschrieben werden. Das Schwammausgangsmaterial wurde über ca. 4 Tage mit Hilfe von Natriumsulfit reduktiv behandelt und die Kollagenfasern (nach einem Filtrationsschritt) in saurem Medium gewonnen. Atomkraftmikroskopische Aufnahmen des isolierten Materials zeigten die kollagentypische Fibrillenquerstreifung, die Unlöslichkeit der Faserbündel in saurem Milieu sowie deren Abbau in die einzelnen Fibrillen in neutraler Pufferlösung. Die von tierischen Kollagenen abgeleitete Bestimmung des Proteingehaltes ergab einen im Vergleich zum kalkulierten Kollagenanteil höheren Gesamtproteingehalt. Dies ist ein Hinweis auf mögliche nichtkollagene Proteinanteile, die an Kollagen assoziiert vorliegen können. Untersuchungen zur Aminosäurenzusammensetzung des isolierten Materials ergaben im Vergleich zu den von Swatschek et al. (2002a) publizierten Ergebnissen einen deutlich höheren Glycin- und einen mehr als doppelt so hohen Hydroxyprolingehalt. Dies kann dahingehend gedeutet werden, dass die neue Isolierungsmethode die bekannten Verunreinigungen des Kollagens mit Glykoproteinen reduziert und zu einem deutlich reineren Kollagenmaterial führt. Mit Hilfe einer neu entwickelten Methode konnten erstmalig Partikel aus Chondrosia-Kollagen hergestellt werden, deren Größe im Nanometerbereich lag. Hierbei handelte es sich um eine kontrollierte alkalische Hydrolyse des isolierten, gefriergetrockneten Kollagens in 1,5 M NaOH. Es konnte eine Partikelausbeute in Höhe von ca. 10% erzielt werden. Atomkraftmikroskopische Aufnahmen zeigten gleichförmige, sphärische Partikel. Die Partikelgrößenuntersuchungen mittels Photonenkorrelationsspektroskopie (nach Resuspension der gefriergetrockneten Schwammkollagennanopartikel (SKNP)) ergaben eine Größe von 168 ± 9,1 nm. Somit sind diese Partikel im Gegensatz zu den bisher aus Chondrosia-Kollagen gewonnenen Mikropartikeln auch für intravenöse Anwendungen geeignet und haben eine größere Trägerkapazität. Die hergestellten Nanopartikel wurden adsorptiv mit dem Wirkstoff 17?-Estradiol-Hemihydrat beladen. In Abhängigkeit von der eingesetzten Stoffkonzentration (1,25 bis 5,0 mg/ml) konnten Beladungsraten von bis zu 13,1% erzielt werden. Die Stabilitätsprüfung von wässrigen Partikelsuspensionen ergab nach 180 Tagen Einlagerung eine auf 7 bis 8% reduzierte Beladungsrate. Nach Inkorporation der beladenen Partikel in eine Hydrogelgrundlage wurde die transdermale Bioverfügbarkeit des Arzneistoffs im Rahmen der Hormonersatztherapie bei postmenopausalen Frauen untersucht. SKNP-Gele mit einer Estradiolkonzentration in Höhe von 0,06% wurden mit gleichkonzentrierten partikelfreien Gelen verglichen. In den Speichelproben der Patientinnen konnten nach täglicher einmaliger Applikation des Nanopartikelgels nach 28 Tagen signifikant höhere Estradiolkonzentrationen gemessen werden. Die ermittelten Flächen unter den Konzentrations-Zeit-Kurven (AUC) über 24 h nach der einmaligen topischen Applikation von 0,75 mg Estradiol waren um das 2,3 bis 3,4-fache höher als die entsprechenden AUC-Werte des Vergleichsgels. 24 h nach Auftragen des Gels lagen die Estradiolspiegel des Vergleichsgels in etwa wieder auf Höhe der Basalwerte, wohingegen die Estradiolwerte nach Applikation des SKNP-Gels zum gleichen Messzeitpunkt mindestens doppelt so hoch waren. Somit konnten bei Anwendung der SKNP-Zubereitung eine deutlich erhöhte transdermale Estradiolabsorption und eine verlängerte Arzneistoffwirkung beobachtet werden. Das nanopartikuläre Trägersystem aus Chondrosia-Kollagen stellt damit eine vielversprechende Möglichkeit zur Erhöhung der transdermalen Bioverfügbarkeit von lipophilen, schlecht resorbierbaren Wirkstoffen dar. Im Rahmen der vorliegenden Arbeit wurde eine Formulierung für einen magensaftresistenten Überzug mit Chondrosia-Kollagen als Filmbildner entwickelt. Die Sprühsuspension bestand aus 15% gefriergetrocknetem Kollagen, dem Weichmacher Triethylcitrat (1,5%), dem Trennmittel Talkum (7,5%) und Wasser. Der Coating-Prozess wurde in einem Dragierkessel mit Placebotabletten durchgeführt. In regelmäßigen Abständen wurden Tabletten-Stichproben entnommen, um die erforderliche Kollagenschichtdicke für eine Magensaftresistenz zu bestimmen. Tabletten mit einer Kollagenauftragsmenge von 12,9 mg/cm2 entsprachen der Prüfung auf Magensaftresistenz gemäß Europäischem Arzneibuch. Die Widerstandsdauer in 0,1 M Salzsäure betrug mehr als 2 h und der Zerfall in Phosphatpufferlösung pH 6,8 höchstens 10 min. Rasterelektronenmikrosko-pische Aufnahmen bei unterschiedlichen Vergrößerungen zeigten eine gleichmäßige und glatte beschichtete Oberfläche. Das Auftragsverfahren im Dragierkessel erwies sich als reproduzierbar. Die mechanischen Eigenschaften der überzogenen Tabletten waren zufriedenstellend und die Magensaftresistenz wurde durch eine 6-monatige Lagerung nicht beeinträchtigt. Als Naturprodukt eignet sich Schwammkollagen als Überzugsmaterial für Nichtarzneimittel. Der schnelle Zerfall des Kollagenüberzugs in neutraler Pufferlösung stellt einen Vorteil gegenüber den häufig verwendeten Schellacküberzügen dar, die sich im Darmtrakt zu langsam auflösen und daher nicht selten mit synthetischen Polymeren kombiniert werden.
