Electrospinning is a versatile and promising drug delivery technology for the development of tailor-made drug delivery systems for various clinical applications. By applying high voltages to drug-loaded polymer solutions, solid polymeric nanofibers can be generated, which encapsulate active pharmaceutical ingredients (APIs) into their polymer matrix. During the electrospinning process, the fibers are deposited on a collector and form a nonwoven network of drug-loaded polymer fibers. These fibers are spatially distributed in aligned or random orientation, providing the opportunity to design highly tunable structural and mechanical properties, which can be adapted to the biological requirements of the intended application site. The mechanically flexible fiber networks can therapeutically be administered to a multitude of pharmaceutical application sites. Their highly porous fiber structure exhibits a large surface-to-volume ratio, which is ideal for controlled drug release kinetics from the polymer matrix upon contact with biological fluids, such as tear fluid, saliva, mucus, wound exudate or gastro-intestinal fluid. For application at the target site, fiber mats are cut into patches. As the patch size determines the quantity of applied API, the electrospinning process must ensure homogeneous distribution of the API throughout the entire fiber mat area.
In this thesis, electrospinning was established as a formulation technology for the rational fabrication of tailor-made multifunctional drug carrier systems for local and site-specific drug delivery to the epithelial interfaces skin, oral mucosa as well as cornea. For adequate characterization and analysis of the drug delivery systems, a broad panel of robust and predictive analytical tools, based of novel investigation techniques for physicochemical characterization of electrospun fibers, was developed.
The initial part of the thesis thematically focuses on the development of predictive analytical techniques, to determine fiber morphology and physicochemical properties, as well as fiber composition and drug release. By designing two model formulations with contrasting properties, and subsequent analysis and characterization with a set of newly developed techniques and state-of-the-art methods, a comprehensive toolset has been made available and evaluated, aiming at advancing and standardizing respective techniques in the scientific field of electrospun drug delivery systems.
Starting with the initiation of the electrospinning formulation process, which often relies on empirical data rather than analytical methods to predict successful processability, analysis of rheological properties of electrospinning solutions was used to rationally detect the minimum polymer concentration required for electrospinning.
For analysis of fiber morphology, scanning electron microscopy is a common technique. However, little attention is given to underlying readout parameters. By analyzing the fiber orientation and diameter of the respective fibers, predictive results regarding mechanical properties could be obtained, which were subsequently confirmed by measuring elongation force with tensile testing. Confocal Raman microscopy, a label-free method for chemically- selective imaging of the fiber samples, was introduced as a complementary visualization technique, enabling the detection of fiber composition and drug distribution.
A novel technique for investigation of water contact angles on the fiber surface of highly hydrophilic polymers was introduced, which provides predictive data regarding interaction with body fluids and the resulting drug release kinetics. Subsequent release testing in a newly developed setup for analyzing drug release from electrospun fibers in low-volume body compartments, confirmed the anticipated drug release kinetics from measurement of the surface hydrophilicity.
By combining complementary analytical methods, including spectral composition analysis, morphology visualization, characterization of physico-chemical properties and drug release kinetics, as well as the application of multivariate data analysis, a robust and predictive toolset has been established, which can support comparability of future electrospinning studies and the translation from the lab bench into clinics.
Based on the analytical toolset, the main part of the thesis focuses on the development and preparation of electrospun platform drug delivery systems for application on epithelial barriers. Electrospun fiber mats are thin, flat, and mechanically flexible, which allows close adherence to epithelial surfaces and reduction of diffusion paths, which enables efficient drug delivery to the skin, oral mucosa, as well as the cornea.
Electrospun fibers bear a high potential for application as wound dressings, while simultaneously controlling the local delivery of APIs to the wound area. Their close resemblance to the extracellular matrix of human skin provides a suitable microenvironment for cellular proliferation and migration for wound closure. In this work, insulin, a fragile proteohormone with growth factor characteristics, was successfully encapsulated into the core of coaxially electrospun fibers, thus maintaining bioactivity throughout and after the electrospinning process. The shell has been designed from biocompatible polymers, which, upon contact with aqueous wound exudate, partially dissolve and form pores through which bioactive insulin is released in a controlled manner. The shell layer provides a hydrophilic surface for interaction with body fluids and skin cells, and possesses substantial mechanical strength, flexibility, and high tensile elongation required for application on wounds. The biocompatibility of the wound dressing was investigated by interaction with primary human dermal fibroblasts and keratinocytes, which displayed healthy cell morphologies without indicating any elevated levels of cytotoxicity markers.
To investigate the effect of insulin on cell migration, in vitro scratch assays on human skin cells were performed. Increased cellular migration speed and wound closure could be observed, indicating improved wound healing. Bio relevance of in vitro wound healing potential results was advanced by development of 3D ex vivo human epidermal skin wound models from reduction surgery donor material. These complex wound models were treated with electrospun insulin fibers and analyzed by proteome analysis to reveal significant increases in wound healing-associated signaling pathways, which could be attributed to a material-driven remarkably positive impact on wound healing of the electrospun fibers...
Im Mittelpunkt dieser Arbeit steht eine Untersuchung zum internationalen Talent-austausch, zur Mobilität und zur Ausbildung von medizinischen und natur-wissenschaftlichen Hochschulabsolventinnen und -absolventen des institutionellen BMEP-Programms (BMEP/Biomedical-Exchange-Program) während des Studiums und beim Übergang ins Berufsleben. Das transatlantische Biomedical Exchange Programm ist ein Programm zum Talentaustausch von Graduierten und Post-Graduierten mit den USA und verankert Innovations- und Wissenstransfer-Forschung von universitärer und industrieller Welt in der Life-Science-Industrie1). Das Programm wurde im Jahr 1979 von Prof. Dr. John Boylan, Prof. of Medical and Physiology an der University of New York, Buffalo und Medical School Farmington (Connecticut) und Prof. Dr. Hilmar Stolte von der Medizinischen Hochschule Hannover ins Leben gerufen. Es handelt sich um eine soziologische Aufgabenstellung mit der thematischen Zielsetzung im Bereich der Karriereforschung. Die Untersuchung hat geklärt, ob internationale Ausbildung und interdisziplinäres Wissen einen Wissensvorsprung bedingen, der sich im Hinblick auf internationale Karrieren und Führungspositionen in unserer heutigen Wissens-gesellschaft auswirkt. Um die Arbeit in den bestehenden Forschungskontext einzuordnen, ist es notwendig, sowohl soziologische als auch ökonomische Teilgebiete der Karriere-, Qualifikations- sowie Mobilitätsforschung zu berücksichtigen, was in den Kapitel 3 und Kapitel 4 dargestellt wird. Die Arbeit ist eine erstmalige empirische Untersuchung über das institutionelle, transatlantische und interdisziplinäre Biomedical-Exchange-Program (BMEP) zum Talentaustausch von medizinischen und naturwissenschaftlichen Studenten mit den USA auf der Ebene von universitärer und industrieller Welt. Um die Effektivität und Leistungsfähigkeit des institutionellen BMEP-Programms zu evaluieren, wurden je drei ehemalige Absolventen pro Jahrgang des Programms der Jahrgänge 1979-2004 mit Hilfe eines Fragebogens befragt. Die Untersuchung befasst sich schwerpunktmäßig mit den ursprünglichen Intentionen und Motiven der Teilnehmer, den Schwierigkeiten, die sich auf diesem Wege in die USA ergeben haben und den Absichten, die verwirklicht werden konnten. Sie fragt, ob erfolgreiche internationale Karrieren durch die Teilnahme am BMEP-Programm entstanden sind, sowie Unternehmertum, Selbständigkeit und internationale Science Frontier-Kontakte und ob im Laufe des Programms Publikationen entstanden sind. ....
Helicobacter pylori (H. pylori) ist ein spiralförmiges, gram-negatives und mikroaerophiles humanpathogen. Seit seiner ersten Isolierung 1983 gilt H. pylori als eine der häufigsten Ursachen für entzündliche Prozesse des gastrointestinalen Traktes (u.a. chronische, sowie atrophische Gastritis und Ulzera). Darüber hinaus kann es gastrale Adenokarzinome und Lymphome des MALT (mucosa-associated lymphoid tissue)-Systems induzieren. Einige H. pylori-Stämme besitzen eine Gruppe von ungefähr 30 Genen, die als cag-Pathogenitätsinsel (cagPAI) bekannt ist. Diese Stämme sind häufig mit Magenschleimhautentzündungen, Geschwürbildung und einem erhöhtem Risiko von Magenkrebs verbunden. Die Gene der cagPAI kodieren für ein Typ IV-Sekretionssystem (T4SS), das Protein- Effektor-Moleküle in die Wirtszelle transloziert. In dieser cagPAI befindet sich ebenfalls das Gen für den Pathogenitätsfaktor CagA (cytotoxin associated gene A). Dieses Protein ist, neben Peptidoglykan, eines der beiden bisher bekannten Moleküle, die vom T4SS transportiert werden. In der Zielzelle wird CagA von Kinasen der Src-Familie an den Tyrosinen in den EPIYA-Sequenz-Wiederholungen phosphoryliert und induziert über einen wenig verstandenen Signalweg eine starke Elongierung und Migration der infizierten Epithelzellen. In diesem Zusammenhang wurde der Einfluss von c-Abl auf die zellulären Prozesse während einer Infektion mit H. pylori untersucht. Die erarbeiteten Daten zeigen deutlich, dass c-Abl ein wichtiges Zielmolekül in der H. pylori-induzierten Zellmigration ist. Durch den Einsatz von spezifischen Inhibitoren der c-Abl-Kinase konnte die Zellelongierung signifikant reduziert werden. Nach Transfektion eines shRNA-Vektors in eine humane Adenokarzinom- Zelllinie wurde eine stabile c-Abl-„Knockdown“-Zelllinie hergestellt. Infektion dieser Abl- AGS-Zellen mit H. pylori induzierte ebenfalls nicht mehr die Zellelongierung. Da wie erwartet die Blockierung der Src-Kinasen ebenfalls den H. pylori-vermittelten Phänotyp inhibierte, sollte in weiteren Experimenten geklärt werden, ob c-Abl- und Src- Kinasen ähnliche Mechanismen aktivieren, die zur Zellmotilität beitragen. Es konnte gezeigt werden, dass Src-Kinasen bereits nach 10-30 min nach der Infektion aktiviert und sehr früh nach 45-60 Minuten wieder inaktiviert wurden. In in-vitro Kinase-Experimenten konnte darüber hinaus beobachtet werden, dass c-Abl dagegen erst nach 60-90 min aktiviert wurde und anschließend über einen längeren Zeitraum aktiviert blieb. Diese unterschiedlichen Kinetiken deuten darauf hin, dass beide Kinasen in der H. pylori-Infektion differentiell reguliert werden und unterschiedliche Funktionen übernehmen. Es wurde bereits publiziert, dass Src-Kinasen CagA direkt in den EPIYA-Motiven phosphorylieren. In dieser Arbeit konnte mittels Ko-Immunpräzipitationen gezeigt werden, dass zwischen c-Abl und CagA eine stabile physikalische Interaktion induziert wird. Des Weiteren konnte gezeigt werden, dass c- Abl CagA direkt phosphoryliert und diese c-Abl-vermittelte Phosphorylierung ein wichtiger Schritt in der H. pylori-induzierten Zellmotilität spielt. Zusammengefasst kann ein Model aufgestellt werden, dass c-Abl die CagA Phosphorylierung über einen langen Zeitraum aufrechterhält und somit die frühe Inaktivierung von Src ersetzt. Die Aktivität von Proteinkinasen wird häufig über Tyrosinphosphorylierungen reguliert. Im Gegensatz zu Src wurden die in der Literatur schon für c-Abl beschriebenen Phosphorylierungsstellen Tyrosin 245 und 412 während einer H. pylori-Infektion nicht phosphoryliert. Eine Phosphorylierung von Tyrosin 245 ließ sich nur durch Stimulation der Zellen mit Natrium-Vanadat und H2O2 erzielen. Interessanterweise korrelierte die c-Abl Aktivität aber mit einer H. pylori-induzierten Zunahme der c-Abl-Proteinmenge. Da die mRNA-Menge nicht variierte, handelte es sich vermutlich um eine Stabilisierung von c-Abl im Komplex mit CagA. Anstelle der Tyrosin-Phosphorylierung konnte die Phosphorylierung von Threonin 735 während einer H. pylori-Infektion nachgewiesen werden. Die bisher nur in geringer Anzahl vorliegenden Veröffentlichungen beschreiben, dass die Phosphorylierung von Threonin 735 eine Rolle in der zellulären Lokalisierung der Kinase spielen könnte. Mit Hilfe von konfokaler Mikroskopie und subzellulärer Fraktionierung, konnte nach H. pylori- Infektion eine Translokation von c-Abl von der typischen zytosolischen Verteilung in die Fokalkomplexe nachgewiesen werden. Ein weiterer Aspekt dieser Arbeit war erste Anhaltspunkte zur Identifizierung der bis heute völlig unbekannten Kinase zu finden, die für die Phosphorylierung von c-Abl an diesem Lokus verantwortlich ist. Bemerkenswert dabei ist, dass diese Phosphorylierung unabhängig von CagA ist, aber trotzdem ein funktionierendes T4SS voraussetzt. Dies konnte durch den Einsatz von Mutationen im T4SS von H. pylori nachgewiesen werden. Dieser Befund deutet darauf hin, dass weitere Faktoren von H. pylori existieren, die über das T4SS in die Zielzellen injiziert werden und an der Regulierung von c-Abl beteiligt sind.
The experiments presented in my thesis were performed to resolve the following major questions: i. Initial experiments are based on the systematic characterization of the C-terminal domains of all 21 HSFs of Arabidopsis with respect to their transactivation potential as well as intracellular localization. This led to the identification of a signature motif for class A HSFs, that consists of an AHA motif (essential for activator potential), and a C-treminal NES (nuclear export signal). With this signature motif, we could identify homologues sequences of more than 90 HSFs in various plant species. ii. Analysis of developmental expression profiles of HSFs using AtGenExpress microarray data led to the identification of the unique expression of HsfA9 during late seed developmental stages. This was the starting point for the investigation of the regulation of HsfA9 as well as its function during seed development. iii. The seed specific transcription factor ABI3 was identified to be responsible for the regulation of HsfA9 by using knock out mutant lines and ectopically expressing transgenic lines for ABI3 gene. Furthermore, the importance of a RY/Sph motif, as binding site for ABI3 on HsfA9 promoter has been analyzed with transient GUS reporter assays. In addition, contribution of component(s) of ABA (abscisic acid) signaling cascade as a functional interacting partner of ABI3 on HsfA9 promoter has been shown and discussed. iv. The essential role of HsfA9 as master regulator for the expression of seed specific members of of HSP encoding genes and GolS1 was shown by analyzing transgenic plants ectopically expressing HsfA9 as well as, by carrying out transient GUS reporter assays. Correlating with this, transgenic plants with ectopic expression of HsfA9 showed a thermotolerent phenotype. Furthermore, a model where HsfA9 plays a key function for the regulation of seed expressed genes which might involved in providing dessication tolerance during seed maturation has been proposed.
