Außer Streit steht die Existenz eines Literaturkanons, der durch sozialen Konsens ästhetische Wertungen bestimmt und vom herrschenden Kunstverständnis geprägt ist. Besonders in seinem selten umstrukturierten Kern ist der Literaturkanon eine implizite Auswahl der als normsetzend und zeitüberdauernd erachteten künstlerischen Werke. Diese gelten als vorbildhaft, zugleich meist in ihrer ästhetischen Qualität als unerreichbar. Abgesichert sind sie in den Urteilen meinungsbildender Gruppen, besonders durch Zirkel, die sich Mechanismen von Salon und Tafelrunde zu eigen gemacht haben. [...] Der Kanon bleibt nicht unverrückbar. Er ist, über einen längeren Zeitraum betrachtet, tatsächlich immer wieder in Bewegung gewesen. Die Prozesse der Kanonisierung, der Aufnahme oder des Ausschlusses verlaufen in direkten oder indirekten Zusammenhängen mit poetologischen Debatten, mit einem Wandel des Kunstbegriffes, mit kulturellen und gesellschaftlichen Veränderungen. [...] Verhandelt werden jeweils Identität und Differenz, Eingrenzung und Ausgrenzung. Eine kulturelle Macht verteilt symbolisches Kapital, auch materielle Güter. Im Kanon steckt ein Autoritätsanspruch, äußert sich ein hierarchisches Denken, das Kunst und Kunstbetrachtung in eine selbstgefällige Beziehung setzt.
Das Etikett "österreichische Literatur" scheint nach wie vor für jene AutorInnen reserviert, die den autonomen Pol des österreichischen Feldes seit den 1970er Jahren dominieren. Junge AutorInnen, die sich von dieser dominierenden Position dezidiert abgrenzen, haben inzwischen zwar auch hierzulande auf sich aufmerksam gemacht, es ist ihnen jedoch nach wie vor nicht gelungen, die Umrisse des Markenzeichens "österreichische Literatur" mit den Grenzen ihrer literarischen Universen in Einklang zu bringen. Und dies, obwohl dieses vormalige Qualitätssiegel außerhalb des österreichischen Literaturbetriebs mittlerweile nicht mehr zwangsläufig als Garant wahrer Kunst gilt. Um den Renommeeverlust der einstigen Avantgarde außerhalb Österreichs einerseits sowie die bislang nur beschränkt erfolgreichen Durchsetzungsversuche eines Teils der jungen österreichischen Literatur andererseits nachzuzeichnen, sei hier zu einem kurzen, literatursoziologischen Spaziergang in die jüngste Geschichte des deutschen und österreichischen Feldes eingeladen.
1997 wurde die Arbeitsstelle für deutschmährische Literatur an der Palacký-Universität in Olmütz gegründet. Nach den umfassenden Feldforschungen der ersten Jahre stellte sich heraus, daß das gesammelte Material ein neues methodologisches und theoretisches Gerüst erhalten muß, damit es wissenschaftlich verwertbar und auch "verkaufbar" wird. Dieses Gerüst ist am ehesten als literatursoziologisch zu bezeichnen. [...] Von der Soziologie fundiert können zahlreiche Fragen an die regionale Literatur herangetragen werden, um diese vernünftig zu erschließen, sie in ihrer strukturellen Andersartigkeit im richtigen, dazugehörigen Kontext zu begreifen und gebührend einzuschätzen und von ihr die vielfachen ideologischen Beläge abzustreifen.
Hinsichtlich des Vordringens von bürgerlichen Mittelschichten in die zentralen Machtpositionen der europäischen Staaten kann man große Unterschiede für die Zeit bis 1914 ausmachen. [...] Aus vielerlei Gründen ist in diesem Zusammenhang das Militär von besonderem Interesse. Es war das klassische Betätigungsfeld einer Aristokratie; allerdings zeigen Statistiken, dass sich der Anteil des Hoch- und Altadels zugunsten jenes von Bürgern und Neuadeligen dramatisch verringerte. [...] Die sozialen Beziehungen innerhalb dieses so gewaltigen sozialen Körpers, den eine stehende Armee seit der frühen Moderne darstellt, sind auch daher wert, studiert zu werden. [...] Was war denn das für ein sozialer Organismus, diese habsburgische Armee, die noch 1914 so durchaus feudal und einem älteren Kriegerkanon verpflichtet wirkte? Wie kommt denn dieser Eindruck zustande, wenn sich die Zusammensetzung des Militärs doch durchaus in Richtung Bürgertum veränderte? Was motivierte denn Menschen bürgerlicher Herkunft dazu, sich in einem adeligen bzw. durch adelige Sitten geprägten sozialen Umfeld durchzusetzen? [...] Was hat das mit Ferdinand von Saars Novelle Leutnant Burda zu tun? Schon seit einiger Zeit hat in den Literaturwissenschaften eine Neubewertung des literarischen Schaffens von Saar eingesetzt. Seine Variante der naturalistischen Romanliteratur wird heute auch künstlerisch hoch eingeschätzt, und zugleich mehren sich Textinterpretationen, die ihn sowohl als meisterhaften Schilderer der österreichischen Konstellation des Übergangs vom Feudalismus zum Bürgertum als auch als einen die "Sphäre des Unbewussten und Unterbewussten" darstellenden Dichter verstehen. Es liegt also nahe, sich Saars Wahrnehmungsfähigkeit zu bedienen, zumal er das Milieu, das er beschreibt, aus seiner eigenen Militärzeit als junger Offizier gut kannte.
Die Literatur "weiß" viel, und vieles wusste und weiß sie früher, als die sich ausdifferenzierenden Wissenschaften seit dem 19. Jahrhundert, im Besonderen Soziologie, Psychologie und Gesellschaftsgeschichte, auf ihre Weise erkannten. Karlheinz Rossbacher zeigt dies anhand verschiedener Beispiele, die er als kleine Materialsammlung in etwa historisch anordnet.
The interaction of fibroblast growth factors (FGFs) with their fibroblast growth factor receptors (FGFRs) are important in the signaling network of cell growth and development. SSR128129E (SSR),[1, 2] a ligand of small molecular weight with potential anti-cancer properties, acts allosterically on the extracellular domains of FGFRs. Up to now, the structural basis of SSR binding to the D3 domain of FGFR remained elusive. This work reports the structural characterization of the interaction of SSR with one specific receptor, FGFR3, by NMR spectroscopy. This information provides a basis for rational drug design for allosteric FGFR inhibitors.
The stimulation of the AMP-activated kinase (AMPK) by 5-amino-1-β-D-ribofuranosyl-imidazole-4-carboxamide (AICAR) has been associated with antihyperalgesia and the inhibition of nociceptive signaling in the spinal cord in models of paw inflammation. The attenuated nociception comes along with a strongly reduced paw edema, indicating that peripheral antiinflammatory mechanisms contribute to antinociception. In this study, we investigated the impact of AICAR on the immune cell composition in inflamed paws, as well as the regulation of inflammatory and resolving markers in macrophages. By using fluorescence-activated cell sorting (FACS) analysis and immunofluorescence, we found a significantly increased fraction of proresolving M2 macrophages and anti-inflammatory interleukin (IL)-10 in inflamed tissue, while M1 macrophages and proinflammatory cytokines such as IL-1 were decreased by AICAR in wild type mice. In AMPKα2 knock-out mice, the M2 polarization of macrophages in the paw was missing. The results were supported by experiments in primary macrophage cultures which also showed a shift to a proresolving phenotype with decreased levels of proinflammatory mediators and increased levels of antiinflammatory mediators. However, in the cell cultures, we did not observe differences between the AMPKα2+/+ and −/− cells, thus indicating that the AICAR-induced effects are at least partially AMPK-independent. In summary, our results indicate that AICAR has potent antiinflammatory and proresolving properties in inflammation which are contributing to a reduction of inflammatory edema and antinociception.
To quantify water flows between groundwater (GW) and surface water (SW) as well as the impact of Abstract. To quantify water flows between groundwater (GW) and surface water (SW) as well as the impact of capillary rise on evapotranspiration by global hydrological models (GHMs), it is necessary to replace the bucket-like linear GW reservoir model typical for hydrological models with a fully integrated gradient-based GW flow model. Linear reservoir models can only simulate GW discharge to SW bodies, provide no information on the location of the GW table and assume that there is no GW flow among grid cells. A gradient-based GW model simulates not only GW storage but also hydraulic head, which together with information on SW table elevation enables the quantification of water flows from GW to SW and vice versa. In addition, hydraulic heads are the basis for calculating lateral GW flow among grid cells and capillary rise.
G³M is a new global gradient-based GW model with a spatial resolution of 5' that will replace the current linear GW reservoir in the 0.5° WaterGAP Global Hydrology Model (WGHM). The newly developed model framework enables inmemory coupling to WGHM while keeping overall runtime relatively low, allowing sensitivity analyses and data assimilation. This paper presents the G³M concept and specific model design decisions together with results under steady-state naturalized conditions, i.e. neglecting GW abstractions. Cell-specific conductances of river beds, which govern GW-SW interaction, were determined based on the 30'' steady-state water table computed by Fan et al. (2013). Together with an appropriate choice for the effective elevation of the SW table within each grid cell, this enables a reasonable simulation of drainage from GW to SW such that, in contrast to the GW model of de Graaf et al. (2015, 2017), no additional drainage based on externally provided values for GW storage above the floodplain is required in G³M. Comparison of simulated hydraulic heads to observations around the world shows better agreement than de Graaf et al. (2015). In addition, G³M output is compared to the output of two established macro-scale models for the Central Valley, California, and the continental United States, respectively. As expected, depth to GW table is highest in mountainous and lowest in flat regions. A first analysis of losing and gaining rivers and lakes/wetlands indicates that GW discharge to rivers is by far the dominant flow, draining diffuse GW recharge, such that lateral flows only become a large fraction of total diffuse and focused recharge in case of losing rivers and some areas with very low GW recharge. G³M does not represent losing rivers in some dry regions. This study presents the first steps towards replacing the linear GW reservoir model in a GHM while improving on recent efforts, demonstrating the feasibility of the approach and the robustness of the newly developed framework.
In global hydrological models, groundwater (GW) is typically represented by a bucket-like linear groundwater reservoir. Reservoir models, however, (1) can only simulate GW discharge to surface water (SW) bodies but not recharge from SW to GW, (2) provide no information on the location of the GW table, and (3) assume that there is no GW flow among grid cells. This may lead, for example, to an underestimation of groundwater resources in semiarid areas where GW is often replenished by SW or to an underestimation of evapotranspiration where the GW table is close to the land surface. To overcome these limitations, it is necessary to replace the reservoir model in global hydrological models with a hydraulic head gradient-based GW flow model.
We present G3M, a new global gradient-based GW model with a spatial resolution of 5′ (arcminutes), which is to be integrated into the 0.5∘ WaterGAP Global Hydrology Model (WGHM). The newly developed model framework enables in-memory coupling to WGHM while keeping overall runtime relatively low, which allows sensitivity analyses, calibration, and data assimilation. This paper presents the G3M concept and model design decisions that are specific to the large grid size required for a global-scale model. Model results under steady-state naturalized conditions, i.e., neglecting GW abstractions, are shown. Simulated hydraulic heads show better agreement to observations around the world compared to the model output of de Graaf et al. (2015). Locations of simulated SW recharge to GW are found, as is expected, in dry and mountainous regions but areal extent of SW recharge may be underestimated. Globally, GW discharge to rivers is by far the dominant flow component such that lateral GW flows only become a large fraction of total diffuse and focused recharge in the case of losing rivers, some mountainous areas, and some areas with very low GW recharge. A strong sensitivity of simulated hydraulic heads to the spatial resolution of the model and the related choice of the water table elevation of surface water bodies was found. We suggest to investigate how global-scale groundwater modeling at 5′ spatial resolution can benefit from more highly resolved land surface elevation data.
The haloarchaeon Haloferax volcanii contains nearly 2800 small non-coding RNAs (sRNAs). One intergenic sRNA, sRNA132, was chosen for a detailed characterization. A deletion mutant had a growth defect and thus underscored the importance of sRNA132. A microarray analysis identified the transcript of an operon for a phosphate-specific ABC transporter as a putative target of sRNA132. Both the sRNA132 and the operon transcript accumulated under low phosphate concentrations, indicating a positive regulatory role of sRNA132. A kinetic analysis revealed that sRNA132 is essential shortly after the onset of phosphate starvation, while other regulatory processes take over after several hours. Comparison of the transcriptomes of wild-type and the sRNA132 gene deletion mutant 30 min after the onset of phosphate starvation revealed that sRNA132 controls a regulon of about 40 genes. Remarkably, the regulon included a second operon for a phosphate-specific ABC transporter, which also depended on sRNA132 for rapid induction in the absence of phosphate. Competitive growth experiments of the wild-type and ABC transporter operon deletion mutants underscored the importance of both transporters for growth at low phosphate concentrations. Northern blot analyses of four additional members of the sRNA132 regulon verified that all four transcripts depended on sRNA132 for rapid regulation after the onset of phosphate starvation. Importantly, this is the first example for the transient importance of a sRNA for any archaeal and bacterial species. In addition, this study unraveled the first sRNA regulon for haloarchaea.
