Wir untersuchten den Einfluss der Umwelt und Landnutzung auf die Vegetation artenreicher Wiesen und Weiden im nordwestdeutschen Mittelgebirgsraum in Südniedersachsen. In drei Teilgebieten (Ith, Rühler Schweiz, und Wangelnstedter Berge) wurden 152 Vegetationsaufnahmen von artenreichen Wiesen und Weiden erstellt und für jede Aufnahmefläche der pH-Wert, die Meereshöhe, Hanglage, Hangneigung, Ellenberg-Zeigerwerte für Nährstoffe, Feuchte, Reaktion und Temperatur, Nutzungswertzahlen nach Briemle sowie die aktuelle Landnutzung bestimmt. Die meisten Bestände im Ith und in den Wangelnstedter Bergen stellen magere Glatthaferwiesen des Arrhenatheretum ranunculetosum dar. In der Rühler Schweiz kommen vor allem magere Kammgrasweiden des Cynosuro-Lolietum luzuletosum vor. Magere Grasländer auf mäßig sauren Böden wurden als Borstgrasrasen des Polygalo-Nardetum sowie Rotschwingelrasen innerhalb der Arrhenatheretalia klassifiziert. Die floristischen Hauptgradienten in der Vegetation sind mit Variablen, die die Nährstoffversorgung der Standorte anzeigen, aber auch mit dem pH-Wert des Bodens und der Bodenfeuchte eng korreliert. Die Meereshöhe der Flächen (130–385 m ü. NN) hat nur einen mäßigen Einfluss auf die Variation der Vegetation, ist aber für das Vorkommen mehrerer Montanzeiger wichtig. Der Pflanzenartenreichtum der Grasländer ist positiv mit der Hangneigung der Flächen und dem pH-Wert des Bodens und negativ mit den Zeigerwerten für Nährstoffe und Feuchte korreliert. Die Nutzungswertzahlen zeigen bei Borstgrasrasen niedrige und sonst hohe Pflegebedürftigkeit. Hohe Trittzahlen der Glatthaferwiesen können auf ihre Nutzung als Extensivumtriebsweide zurückgeführt werden. Anscheinend eignet sich diese Nutzungsform in Untersuchungsgebiet zum Erhalt magerer Wiesen.
At the edge of the Harz Mountains in Lower Saxony a population of the hart's tongue fern (Asplenium scolopendrium) threatened by destruction by a gypsum quarry were transplanted into a dolina which was not populated by the species at that time, and the new population was followed over ten years. 90% of the 59 transplanted plants survived this period and grew larger during the first six years after transplantation. Progenies appeared in the third year after transplantation. Nowadays, in the tenth year after transplantation, there are 1110 progenies, 171 of which are reproducing. Overall, the population increased by 1781% in the ten years. Plants that were planted on a rocky slope or a boulder heap in the new habitat, where soil was available, grew better than plants in rock faces without soil. In contrast, in the rock faces, where substrate was not covered with autumn foliage, more juveniles established. The distance between juveniles and mother plants rarely exceeded three meters, which indicates a limited dispersal potential of the hart’s tongue fern and may explain together with low diaspore pressure as a result of local rarity of the species that the dolina had not been colonized spontaneously. We conclude that transplantations of adult plants or introduction of spores are a suitable measure for preserving hart’s tongue fern populations that are endangered by destruction. In the long run, however, such measures cannot compensate for ongoing destruction of natural habitats by mining activities in the gypsum karst region at the southern edge of the Harz Mountains.
We present the data of the 2nd research expedition of the European Dry Grassland Group (EDGG), which was conducted in 2010 in Central Podolia, Ukraine. The aim was to collect plot data to compare Ukrainian dry grasslands with those of other parts of Europe in terms of syntaxonomy and biodiversity. We sampled 21 nested-plot series (0.0001–100 m2) and 184 normal plots (10 m2) covering the full variety of dry grassland types occurring in the study region. For all plots, we recorded species composi-tion of terrestrial vascular plants, bryophytes and lichens, while for the 226 10-m2 plots we estimated and measured percentage cover of all species, structural, topographic, soil and landuse parameters. The 10-m² plots were used for phytosociological classification based on iteratively refined TWINSPAN classification as well as for DCA ordination. Differences between the derived vegetation types with respect to environmental conditions and species richness were assessed with ANOVAs. We assigned our plots to nine association-level units but refrained from placing them into formal associations with two exceptions. In the study area, dry grasslands of the Festuco-Brometea were far more common than those of the Koelerio-Corynephoretea. Among the Festuco-Brometea, xeric Festucetalia valesiacae grasslands were more frequent and represented by the Festucion valesiacae (2 associations, including the Allio taurici-Dichanthietum ischaemi ass. nova) and the Stipion lessingianae (1) compared to the Brachypodietalia pinnati with the Agrostio vinealis-Avenulion schellianae (3). The Koelerio-Corynephoretea were represented by three associations, each from a different order and alliance: basiphilous outcrops (Alysso alyssoidis-Sedetalia: Alysso alyssoidis-Sedion?), acidophilous outcrops (Sedo-Scleranthetalia: Veronico dillenii-Sedion albi?) and mesoxeric sandy grasslands (Trifolio arvensis-Festucetalia ovinae: Agrostion vinealis). We discuss the issue of the mesoxeric order Galietalia veri placed within the Molinio-Arrhenatheretea by Ukrainian authors and conclude that the content of that order would probably be better placed in the mesoxeric orders of the Koelerio-Corynephoretea and Festuco-Brometea. Other syntaxonomic questions could not be solved with our geographically limited dataset and await a supraregional analysis, e.g. whether the Ukrainian outcrop communities should be assigned to the same alliances as known from Central Europe or rather represent new vicariant units. The analysis of the biodiversity patterns showed that at a grain size of 10 m2, Podolian Koelerio-Corynephoretea communities were overall richer than Festuco-Brometea communities (46.4 vs. 40.6 species). This difference was due to the Koelerio-Corynephoretea containing twice as many bryophytes and nine times more lichens, while vascular plant species richness did not differ significantly between classes. The orders within the classes showed no real differences in species richness. The richness patterns observed in Podolia were almost the opposite of those usually found in dry grasslands, where Brachypodietalia pinnati are richer than Festucetalia valesiacae, and these richer than stands of the Koelerio-Corynpehoretea – and we do not have a good explanation for these idiosyncrasies. In conclusion, Podolian dry grasslands behave quite unexpectedly regarding biodiversity, and their syntaxonomy is still poorly understood. These knowledge gaps can only be addressed with supranational analyses based on comprehensive datasets.
The Transylvanian Plateau in Romania is well known to host large areas of a variety of dry grassland types, still traditionally managed by low-intensity mowing or grazing. While this natural heritage is now under threat from changes in agricultural practices, the diversity of Transylvanian dry grasslands is still little understood. There is a lack of both field data sampled with standardised methods and a syntaxonomic treatment with modern statistical methods and supra-regional perspective. Therefore, the European Dry Grassland Group (EDGG) carried out its first international Research Expedition in Transylvania 2009 to study syntaxonomy, vegetation-environment relationships, and biodiversity patterns of these communities. In various locations across Transylvania, we sampled 10-m² vegetation plots (n = 82) and nested-plot series from 0.0001 m² to 100 m² (n = 20), including all vascular plant, bryophyte, and lichen species, as well as structural and soil data. The vegetation classification was carried out with modified TWINSPAN, followed by determination of diagnostic species with phi values and a small-scale re-assignment of relevés with the aim of crispness maximisation. Both TWINSPAN and ordination revealed three major groups of syntaxa, which were matched to three orders from the class of basiphilous dry grasslands, Festuco-Brometea, represented by one alliance each: rocky dry grasslands (Stipo pulcherrimae-Festucetalia pallentis: Seslerion rigidae); xeric grasslands on deep soils (Festucetalia valesiacae: Stipion lessingianae) and meso-xeric grasslands on deep soils (Brachypodietalia pinnati: Cirsio-Brachypodion pinnati). We accepted nine association-level units plus two that potentially merit association status but were only represented by one relevé each. Most of the units could be identified with one or several previously described associations. To support nomenclatural stability, we provide a nomenclatural revision and designate nomenclatural types where previously there were none. Further, we used DCA ordination and analysis of variance to determine the main environmental drivers of floristic differentiation and to determine ecological and structural differences between the vegetation types. The strongest differentiation occurred along the aridity gradient with the dense, particularly diverse stands on more or less level sites on the one hand (Brachypodietalia pinnati) and the more open, less diverse stands on steep south-facing slopes on the other end of the gradient (Stipo pulcherrimae- Festucetalia pallentis, Festucetalia valesiacae). The two xeric orders were then separated along the second DCA axis, with the Stipo pulcherrimae-Festucetalia pallentis inhabiting the stone-rich sites at higher altitudes while the Festucetalia valesiacae occur on soft, deep substrata at lower altitudes. The analysed dry grassland communities have extraordinarily high -diversity at all spatial scales for all plants and for vascular plants, but are relatively poor in bryophytes and lichens. Some formerly mown stands of the Festuco sulcatae-Brachypodietum pinnati (Brachypodietalia pinnati) are even richer in vascular plant species than any other recorded vegetation type worldwide on the spatial scales of 0.1 m² (43) and 10 m² (98); the respective relevés are documented here for the first time. Also, the b-diversity of the grasslands was unexpectedly high, with a mean z-value of 0.275. Despite its limited extent, the methodological thoroughness of this study allows us to shed new light on the syntaxonomy of dry grasslands in Romania and to raise the awareness that Transylvania still hosts High Nature Value grasslands that are bio - diversity hotspots at a global scale but at the same time are highly endangered through changes in agricultural practices.
This report provides a brief review of the 20th annual meeting of the German Language Branch of the Society of Environmental Toxicology and Chemistry (SETAC GLB) held from September 7th to 10th 2015 at ETH (Swiss Technical University) in Zurich, Switzerland. The event was chaired by Inge Werner, Director of the Swiss Centre for Applied Ecotoxicology (Ecotox Centre) Eawag-EPFL, and organized by a team from Ecotox Centre, Eawag, Federal Office of the Environment, Federal Office of Agriculture, and Mesocosm GmbH (Germany). Over 200 delegates from academia, public agencies and private industry of Germany, Switzerland and Austria attended and discussed the current state of science and its application presented in 75 talks and 83 posters. In addition, three invited keynote speakers provided new insights into scientific knowledge ‘brokering’, and—as it was the International Year of Soil—the important role of healthy soil ecosystems. Awards were presented to young scientists for best oral and poster presentations, and for best 2014 master and doctoral theses. Program and abstracts of the meeting (mostly in German) are provided as Additional file 1.
Ribosome recycling orchestrated by the ATP binding cassette (ABC) protein ABCE1 can be considered as the final—or the first—step within the cyclic process of protein synthesis, connecting translation termination and mRNA surveillance with re-initiation. An ATP-dependent tweezer-like motion of the nucleotide-binding domains in ABCE1 transfers mechanical energy to the ribosome and tears the ribosome subunits apart. The post-recycling complex (PRC) then re-initiates mRNA translation. Here, we probed the so far unknown architecture of the 1-MDa PRC (40S/30S·ABCE1) by chemical cross-linking and mass spectrometry (XL-MS). Our study reveals ABCE1 bound to the translational factor-binding (GTPase) site with multiple cross-link contacts of the helix–loop–helix motif to the S24e ribosomal protein. Cross-linking of the FeS cluster domain to the ribosomal protein S12 substantiates an extreme lever-arm movement of the FeS cluster domain during ribosome recycling. We were thus able to reconstitute and structurally analyse a key complex in the translational cycle, resembling the link between translation initiation and ribosome recycling.
The endoplasmic reticulum–mitochondria encounter structure (ERMES) connects the mitochondrial outer membrane with the ER. Multiple functions have been linked to ERMES, including maintenance of mitochondrial morphology, protein assembly and phospholipid homeostasis. Since the mitochondrial distribution and morphology protein Mdm10 is present in both ERMES and the mitochondrial sorting and assembly machinery (SAM), it is unknown how the ERMES functions are connected on a molecular level. Here we report that conserved surface areas on opposite sides of the Mdm10 β-barrel interact with SAM and ERMES, respectively. We generated point mutants to separate protein assembly (SAM) from morphology and phospholipid homeostasis (ERMES). Our study reveals that the β-barrel channel of Mdm10 serves different functions. Mdm10 promotes the biogenesis of α-helical and β-barrel proteins at SAM and functions as integral membrane anchor of ERMES, demonstrating that SAM-mediated protein assembly is distinct from ER-mitochondria contact sites.
The inclusive charged particle transverse momentum distribution is measured in proton–proton collisions at s=900 GeV at the LHC using the ALICE detector. The measurement is performed in the central pseudorapidity region (|η|<0.8) over the transverse momentum range 0.15<pT<10 GeV/c. The correlation between transverse momentum and particle multiplicity is also studied. Results are presented for inelastic (INEL) and non-single-diffractive (NSD) events. The average transverse momentum for |η|<0.8 is 〈pT〉INEL=0.483±0.001 (stat.)±0.007 (syst.) GeV/c and 〈pT〉NSD=0.489±0.001 (stat.)±0.007 (syst.) GeV/c, respectively. The data exhibit a slightly larger 〈pT〉 than measurements in wider pseudorapidity intervals. The results are compared to simulations with the Monte Carlo event generators PYTHIA and PHOJET.
MTO1-deficient mouse model mirrors the human phenotype showing complex I defect and cardiomyopathy

Recently, mutations in the mitochondrial translation optimization factor 1 gene (MTO1) were identified as causative in children with hypertrophic cardiomyopathy, lactic acidosis and respiratory chain defect. Here, we describe an MTO1-deficient mouse model generated by gene trap mutagenesis that mirrors the human phenotype remarkably well. As in patients, the most prominent signs and symptoms were cardiovascular and included bradycardia and cardiomyopathy. In addition, the mutant mice showed a marked worsening of arrhythmias during induction and reversal of anaesthesia. The detailed morphological and biochemical workup of murine hearts indicated that the myocardial damage was due to complex I deficiency and mitochondrial dysfunction. In contrast, neurological examination was largely normal in Mto1-deficient mice. A translational consequence of this mouse model may be to caution against anaesthesia-related cardiac arrhythmias which may be fatal in patients.
Filoviruses infect a wide range of cell types with the exception of lymphocytes. The intracellular proteins cathepsin B and L, two-pore channel 1 and 2, and bona fide receptor Niemann–Pick Disease C1 (NPC1) are essential for the endosomal phase of cell entry. However, earlier steps of filoviral infection remain poorly characterized. Numerous plasma membrane proteins have been implicated in attachment but it is still unclear which ones are sufficient for productive entry. To define a minimal set of host factors required for filoviral glycoprotein-driven cell entry, we screened twelve cell lines and identified the nonlymphocytic cell line SH-SY5Y to be specifically resistant to filovirus infection. Heterokaryons of SH-SY5Y cells fused to susceptible cells were susceptible to filoviruses, indicating that SH-SY5Y cells do not express a restriction factor but lack an enabling factor critical for filovirus entry. However, all tested cell lines expressed functional intracellular factors. Global gene expression profiling of known cell surface entry factors and protein expression levels of analyzed attachment factors did not reveal any correlation between susceptibility and expression of a specific host factor. Using binding assays with recombinant filovirus glycoprotein, we identified cell attachment as the step impaired in filovirus entry in SH-SY5Y cells. Individual overexpression of attachment factors T-cell immunoglobulin and mucin domain 1 (TIM-1), Axl, Mer, or dendritic cell-specific intercellular adhesion molecule-3-grabbing non-integrin (DC-SIGN) rendered SH-SY5Y cells susceptible to filovirus glycoprotein-driven transduction. Our study reveals that a lack of attachment factors limits filovirus entry and provides direct experimental support for a model of filoviral cell attachment where host factor usage at the cell surface is highly promiscuous.
Background: Alzheimer's disease is a common debilitating dementia with known heritability, for which 20 late onset susceptibility loci have been identified, but more remain to be discovered. This study sought to identify new susceptibility genes, using an alternative gene-wide analytical approach which tests for patterns of association within genes, in the powerful genome-wide association dataset of the International Genomics of Alzheimer's Project Consortium, comprising over 7 m genotypes from 25,580 Alzheimer's cases and 48,466 controls.
Principal findings: In addition to earlier reported genes, we detected genome-wide significant loci on chromosomes 8 (TP53INP1, p = 1.4×10−6) and 14 (IGHV1-67 p = 7.9×10−8) which indexed novel susceptibility loci.
Significance: The additional genes identified in this study, have an array of functions previously implicated in Alzheimer's disease, including aspects of energy metabolism, protein degradation and the immune system and add further weight to these pathways as potential therapeutic targets in Alzheimer's disease.
Electroencephalography (EEG) represents a widely established method for assessing altered and typically developing brain function. However, systematic studies on EEG data quality, its correlates, and consequences are scarce. To address this research gap, the current study focused on the percentage of artifact-free segments after standard EEG pre-processing as a data quality index. We analyzed participant-related and methodological influences, and validity by replicating landmark EEG effects. Further, effects of data quality on spectral power analyses beyond participant-related characteristics were explored. EEG data from a multicenter ADHD-cohort (age range 6 to 45 years), and a non-ADHD school-age control group were analyzed (ntotal = 305). Resting-state data during eyes open, and eyes closed conditions, and task-related data during a cued Continuous Performance Task (CPT) were collected. After pre-processing, general linear models, and stepwise regression models were fitted to the data. We found that EEG data quality was strongly related to demographic characteristics, but not to methodological factors. We were able to replicate maturational, task, and ADHD effects reported in the EEG literature, establishing a link with EEG-landmark effects. Furthermore, we showed that poor data quality significantly increases spectral power beyond effects of maturation and symptom severity. Taken together, the current results indicate that with a careful design and systematic quality control, informative large-scale multicenter trials characterizing neurophysiological mechanisms in neurodevelopmental disorders across the lifespan are feasible. Nevertheless, results are restricted to the limitations reported. Future work will clarify predictive value.
Background: Simultaneous pancreas kidney transplantation (SPK), pancreas transplantation alone (PTA) or pancreas transplantation after kidney (PAK) are the only curative treatment options for patients with type 1 (juvenile) diabetes mellitus with or without impaired renal function. Unfortunately, transplant waiting lists for this indication are increasing because the current organ acceptability criteria are restrictive; morbidity and mortality significantly increase with time on the waitlist. Currently, only pancreas organs from donors younger than 50 years of age and with a body mass index (BMI) less than 30 are allocated for transplantation in the Eurotransplant (ET) area. To address this issue we designed a study to increase the available donor pool for these patients.
Methods/Design: This study is a prospective, multicenter (20 German centers), single blinded, non-randomized, two armed trial comparing outcome after SPK, PTA or PAK between organs with the currently allowed donor criteria versus selected organs from donors with extended criteria. Extended donor criteria are defined as organs procured from donors with a BMI of 30 to 34 or a donor age between 50 and 60 years. Immunosuppression is generally standardized using induction therapy with Myfortic, tacrolimus and low dose steroids. In principle, all patients on the waitlist for primary SPK, PTA or PAK are eligible for the clinical trial when they consent to possibly receiving an extended donor criteria organ. Patients receiving an organ meeting the current standard criteria for pancreas allocation (control arm) are compared to those receiving extended criteria organ (study arm); patients are blinded for a follow-up period of one year. The combined primary endpoint is survival of the pancreas allograft and pancreas allograft function after three months, as an early relevant outcome parameter for pancreas transplantation.
Discussion: The EXPAND Study has been initiated to investigate the hypothesis that locally allocated extended criteria organs can be transplanted with similar results compared to the currently allowed standard ET organ allocation. If our study shows a favorable comparison to standard organ allocation criteria, the morbidity and mortality for patients waiting for transplantation could be reduced in the future.

Bipolar disorder (BD) is a highly heritable neuropsychiatric disease characterized by recurrent episodes of mania and depression. BD shows substantial clinical and genetic overlap with other psychiatric disorders, in particular schizophrenia (SCZ). The genes underlying this etiological overlap remain largely unknown. A recent SCZ genome wide association study (GWAS) by the Psychiatric Genomics Consortium identified 128 independent genome-wide significant single nucleotide polymorphisms (SNPs). The present study investigated whether these SCZ-associated SNPs also contribute to BD development through the performance of association testing in a large BD GWAS dataset (9747 patients, 14278 controls). After re-imputation and correction for sample overlap, 22 of 107 investigated SCZ SNPs showed nominal association with BD. The number of shared SCZ-BD SNPs was significantly higher than expected (p = 1.46x10-8). This provides further evidence that SCZ-associated loci contribute to the development of BD. Two SNPs remained significant after Bonferroni correction. The most strongly associated SNP was located near TRANK1, which is a reported genome-wide significant risk gene for BD. Pathway analyses for all shared SCZ-BD SNPs revealed 25 nominally enriched gene-sets, which showed partial overlap in terms of the underlying genes. The enriched gene-sets included calcium- and glutamate signaling, neuropathic pain signaling in dorsal horn neurons, and calmodulin binding. The present data provide further insights into shared risk loci and disease-associated pathways for BD and SCZ. This may suggest new research directions for the treatment and prevention of these two major psychiatric disorders.
The most basic behavioural states of animals can be described as active or passive. However, while high-resolution observations of activity patterns can provide insights into the ecology of animal species, few methods are able to measure the activity of individuals of small taxa in their natural environment. We present a novel approach in which the automated VHF radio-tracking of small vertebrates fitted with lightweight transmitters (< 0.2 g) is used to distinguish between active and passive behavioural states.
A dataset containing > 3 million VHF signals was used to train and test a random forest model in the assignment of either active or passive behaviour to individuals from two forest-dwelling bat species (Myotis bechsteinii (n = 50) and Nyctalus leisleri (n = 20)). The applicability of the model to other taxonomic groups was demonstrated by recording and classifying the behaviour of a tagged bird and by simulating the effect of different types of vertebrate activity with the help of humans carrying transmitters. The random forest model successfully classified the activity states of bats as well as those of birds and humans, although the latter were not included in model training (F-score 0.96–0.98).
The utility of the model in tackling ecologically relevant questions was demonstrated in a study of the differences in the daily activity patterns of the two bat species. The analysis showed a pronounced bimodal activity distribution of N. leisleri over the course of the night while the night-time activity of M. bechsteinii was relatively constant. These results show that significant differences in the timing of species activity according to ecological preferences or seasonality can be distinguished using our method.
Our approach enables the assignment of VHF signal patterns to fundamental behavioural states with high precision and is applicable to different terrestrial and flying vertebrates. To encourage the broader use of our radio-tracking method, we provide the trained random forest models together with an R-package that includes all necessary data-processing functionalities. In combination with state-of-the-art open-source automated radio-tracking, this toolset can be used by the scientific community to investigate the activity patterns of small vertebrates with high temporal resolution, even in dense vegetation.

The analysis showed a pronounced bimodal activity distribution of N. leisleri over the course of the night while the night-time activity of M. bechsteinii was relatively constant. These results show that subtle differences in the timing of species' activity can be distinguished using our method.
Our approach can classify VHF-signal patterns into fundamental behavioural states with high precision and is applicable to different terrestrial and flying vertebrates. To encourage the broader use of our radiotracking method, we provide the trained random forest models together with an R package that includes all necessary data processing functionalities. In combination with state-of-the-art open-source automated radiotracking, this toolset can be used by the scientific community to investigate the activity patterns of small vertebrates with high temporal resolution, even in dense vegetation.
Wir untersuchen Zusammenhänge zwischen der Artenzusammensetzung, dem Artenreichtum und den Umweltbedingungen der Schwermetallrasen und Trockenrasen der Bottendorfer Hügel in Nord-Thüringen. In diesem Gebiet wachsen primäre und sekundäre Schwermetallrasen und verschiedene Trockenrasengesellschaften auf nicht-schwermetallhaltigen Böden mit einer artenreichen Xerothermrasen-Flora. Anhand von 206 Aufnahmen unterscheiden wir neun Pflanzengesellschaften aus sechs Verbänden der Klassen Koelerio-Corynephoretea (Sedo-Veronicion, Alysso-Sedion) und Festuco-Brometea (Festucion pallentis, Koelerio-Phleion, Festucion valesiacae, Cirsio-Brachypodion). Vier Trockenrasengesellschaften, in denen die Metallophyten Armeria maritima subsp. halleri und Minuartia verna subsp. hercynica häufig Vorkommen, werden als spezielle Subassoziationen (armerietosum halleri) von vier Assoziationen (Teucrio-Festucetum, Thymo-Festucetum, Filipendulo-Helictotrichetum, Adonido-Brachypodietum) neu beschrieben. In einer DCA mit angezeigten Umweltvektoren bilden die Gesellschaften klare Gruppen. Das Teucrio-Festucetum armerietosum und Thymo-Festucetum armerietosum sind in der DCA mit den Gehalten des Bodens an Kupfer und Blei sowie dem C/N-Verhältnis des Bodens eng korreliert, das Adonido-Brachypodietum teucrietosum und Festuco-Stipetum teucrietosum mit dem pH und Karbonatgehalt des Bodens. Der Artenreichtum an Gefäßpflanzen, Moosen und Flechten nimmt bis pH 7,5 zu und dann ab. Der Artenreichtum steigt mit dem Karbonatgehalt und der Leitfähigkeit des Bodens und nimmt mit dem C/N-Verhältnis des Bodens ab. Der Artenreichtum der Gefäßpflanzen nimmt mit dem Kupfergehalt des Bodens ab, während die Wahrscheinlichkeit des Vorkommens der Metallophyten mit dem Kupfer- und Zinkgehalt steigt. - Unsere Studie zeigt, dass Bodenfaktoren wie der pH und der Karbonatgehalt für die Zusammensetzung und den Artenreichtum der Vegetation auf kontaminierten Böden ebenso wichtig sind wie der Gehalt an Schwermetallen. Schwermetallrasen können daher Trockenrasen auf nicht-schwermetallhaltigen Böden floristisch sehr ähnlich sein.
Die Steppenrasen des NSG „Badraer Lehde–Großer Eller“ werden hier erstmalig beschrieben und analysiert. 156 Originalaufnahmen wurden über eine Clusteranalyse in sieben Assoziationen und zwei ranglose Gesellschaften innerhalb der Verbände Alysso-Sedion, Seslerio-Festucion pallentis, Festucion valesiacae, Xerobromion und Cirsio-Brachypodion gegliedert. Die Auswertung alter Luftbilder zeigt, dass die beiden ranglosen Gesellschaften junge Steppenrasen auf ehemaligen Ackerflächen darstellen, während die Assoziationen überwiegend alte Steppenrasen repräsentieren. Der erste floristische Gradient nach einer NMDS in der Vegetation wird durch Variablen erklärt, die die Temperatur und die Wasserversorgung der Standorte anzeigen. Der zweite floristische Gradient kann durch das Alter der Flächen erklärt werden. Alte Steppenrasen enthalten signifikant mehr gefährdete Pflanzenarten als junge Steppenrasen, während sich der Gesamtartenreichtum zwischen jungen und alten Beständen nicht signifikant unterscheidet. Ein CSR-Strategietypenspektrum zeigt in einer Gesellschaft auf ehemaligen Ackerflächen eine höhere Bedeutung der C-Strategie, sonst aber kaum Unterschiede zwischen den Syntaxa. Die beiden Xerobromion-Assoziationen sind besonders artenreich und enthalten überdurchschnittlich viele gefährdete Pflanzenarten. Eine Gesellschaft auf ehemaligen Ackerflächen enthält durchschnittlich viele und die andere fast keine gefährdeten Arten. Der Artenreichtum der Gefäßpflanzen ist am stärksten positiv mit der Bodengründigkeit, der Deckung der Krautschicht und dem Ellenberg-Zeigerwert für Bodenreaktion und negativ mit dem Zeigerwert für Temperatur korreliert. Unsere Studie zeigt die hohe Bedeutung des NSG für den Trockenrasenschutz. Diese beruht vor allem auf der allgemein hohen Artenvielfalt und Diversität an Gesellschaften sowie auf der sehr hohen Zahl gefährdeter Arten.
Reintroductions of plant species are increasingly popular in conservation practice. Steppe grasslands contain many rare and endangered plant species that are potential objects for such reintroductions. Most reintroduction projects, however, can only target a restricted number of species, which raises the question of how species should be prioritised. Here, we present a method to select priority species for reintroduction based on species' characteristics that are widely used in conservation practice. We first determined the local species pool containing those vascular plant species that occurred both in our target region (Thuringia, Germany) and target habitat (steppe grasslands), yielding 369 species. With the help of an a priori filter that selected currently endangered species with limited distribution, 136 potential target species were determined. These potential target species had experienced stronger decline, had a narrower phytosociological amplitude and were more likely to be species of the Festuco-Brometea class and the Festucetalia valesiacae order than non-target species. Potential target species were then ranked by a points system based on ten conservation-relevant characteristics of the species from the categories "threat and protection status", "distribution and decline", and "habitat affiliation". In the ranking, six steppe grassland plant species (Astragalus exscapus, Bothriochloa ischaemum, Prunella laciniata, Pulsatilla pratensis subsp. nigricans, Scorzonera purpurea, and Seseli hippomarathrum) achieved the highest scores. An additional seven species not specifically characteristic for steppe grasslands also scored highly. A post hoc evaluation of these 13 highest scoring species based on additional conservation criteria left five species (Astragalus exscapus, Linum leonii, Orchis morio, Pulsatilla pratensis subsp. nigricans and Scorzonera purpurea) as species with highest priority for reintroductions and another five species as highly suitable for reintroductions. Associations between the ranking order and different ranking criteria revealed that a species’ threat and rarity in Thuringia and its protection status had the highest representation in the ranking, followed by threat in Germany, regional decline and habitat affiliation. In contrast, international threat and responsibility of Thuringia for its conservation had only low representation in the ranking, probably because these characteristics applied to only a few species. The ranking list gives a selection of species for reintroductions, which combined with additional information based on comprehensive local and floristic knowledge, allows the identification of the species with the highest priority. Our method can be transferred to other regions or habitat types.
Die Pflanzengesellschaften der Felsfluren und Magerrasen im unteren Unstruttal (Sachsen-Anhalt)

Die vorliegende Arbeit gibt einen Überblick über die Vegetation von Magerrasen (Festuco-Brometea) und Felsfluren (Sedo-Scleranthetalia) im unteren Unstruttal (Sachsen-Anhalt). Auf der Grundlage von 207 Vegetationsaufnahmen werden 8 Assoziationen sowie zwei ranglose Gesellschaften unterschieden:

Arealtypenspektren der Magerrasen zeigen, daß im Gebiet submediterrane und kontinentale Trockenrasenelemente (Xerobromion, Festucion valesiacae) vorherrschen und sich stark mischen. Ein Ökogramm ermöglicht für die Magerrasen eine vergleichende Betrachtung hinsichtlich der Parameter Kontinentalität und Bodenfeuchte.
Der Harz wurde über Jahrhunderte durch intensiven Bergbau und Erzverarbeitung geprägt. In ihrer Folge entstanden zahlreiche Sekundärstandorte mit schwermetallreichen Substraten auf Halden und kontaminierten Flussschottern. Daher stellt der Harz ein klassisches Gebiet des Studiums von Pflanzengesellschaften mit schwermetalltoleranten Arten dar. Allerdings lag bis heute keine systematische Übersicht und umfassende ökologische Analyse der Schwermetallvegetation und deren Standorte vor. - Grundlagen der Arbeit sind 120 Aufnahmen der Schwermetallvegetation und Analysen der Bodenbedingungen an 23 Orten auf Bergbau- und Schlackenhalden sowie kontaminierten Flussschottern im westlichen Harz und seinem Vorland. Die Rasen besitzen insgesamt nur wenige Phanerogamen; ihre Kenntaxa Armeria maritima ssp. halleri, Minuartia verna ssp. hercynica und Silene vulgaris var. humilis kommen aber in den meisten Beständen vor. Sie werden alle dem Armerietum halleri Libbert 1930 zugeordnet, das sich neben einer Silene-Pionierphase in drei Subassoziationen: A. cladonietosum chlorophaeae, A. typicum und A. achilletosum millefoliae gliedern lässt, die auch als Phasen einer Primärsukzession gedeutet werden können. Die beiden letzteren können weiter in je eine Typische und eine bodenfeuchtere Cardaminopsis halleri-Variante aufgeteilt werden. - In einer DCA trennen sich die Subassoziationen entlang der ersten Achse. Diese Achse ist positiv korreliert mit den Ellenberg-Zeigerwerten für Reaktion, Stickstoff und Feuchte sowie den Calcium-Gehalten der Bodenlösung, negativ mit den Kupfer-Gehalten und Anteilen offener Steine. Die Schwermetall-Gehalte der Böden sind durchgehend hoch. Im A. cladonietosum und A. typicum sowie der Silene-Pionierphase gibt es besonders große Zink- und Kupfer-Gehalte; weite Schwermetall/Calcium-Verhältnisse weisen hier auf extrem zinktoxische Bedingungen hin. Die Blei-Gehalte unterscheiden sich zwischen den Gesellschaften dagegen kaum. - Ein Vergleich der aktuellen Anzahl von Halden mit einer Liste von 1928 zeigt einen starken Rückgang der Schwermetallstandorte durch menschliche Zerstörung und natürliche Sukzession. Pflegemaßnahmen zur Erhaltung oder Regeneration werden erörtert. - In der Diskussion wird die Beibehaltung eines eigenständigen Armerietum halleri befürwortet. Die Auswertung von Literaturdaten in einer Übersichtstabelle für Deutschland ergibt eine Einengung der Violetea calaminariae auf einen Kern artenärmerer Gesellschaften. Für den übrig bleibenden einzigen Verband wird vorgeschlagen, den Namen Armerion halleri Ernst 1965 als nomen conservandum beizubehalten.
Seit 11 Jahren werden von Mitgliedern der Eurasian Dry Gassland Group (EDDG) und deren Vorgängerorganisationen Grasland-Sonderteile in Tuexenia herausgegeben. Der diesjährige Sonderteil präsentiert sechs Artikel mit unterschiedlichen Fragestellungen zu Trockenrasen und verwandten Graslandtypen aus sechs europäischen Ländern: Belgien, Bosnien-Herzegowina, Deutschland, Italien, Serbien und Ungarn. Ein Artikel behandelt syntaxonomische Aspekte der Serpentin-Steppenrasen des Balkans (s. auch Umschlagfoto). Zwei Beiträge untersuchen funktionelle Eigenschaften von Pflanzenarten im Grasland, der eine (in belgischen Trockenrasen) die Variationen funktioneller Merkmale in Abhängigkeit von den Umweltbedingungen und der andere die Mengenverhältnisse von funktionellen Merkmalen entlang eines Höhengradienten in den italienischen Alpen. Ein Artikel zeigt mithilfe eines experimentellen Ansatzes, dass artenarmes ungarisches Grasland durch Diasporenzufuhr in artenreiche Wiesen überführt werden kann, während ein anderer Beitrag ökologische Theorien am Grasland in einer ungarischen Stadt überprüft. Der sechste Artikel behandelt schließlich Vegetationsveränderungen von Steppenrasen in Nordostdeutschland nach 20 Jahren.
Seit 12 Jahren werden von Mitgliedern der Eurasian Dry Grassland Group (EDGG) und deren Vorgängerorganisationen Grasland-Sonderteile in Tuexenia herausgegeben. Der diesjährige Sonderteil enthält fünf Artikel, die das Grasland verschiedener mitteleuropäischer Länder mit unterschiedlichen Zielen untersuchen. Der erste Artikel untersucht Auswirkungen von Weide und Mahd auf die Diversität des Graslands in Deutschland in Abhängigkeit von verschiedenen Umweltfaktoren, der zweite die Auswirkung kleiner Büsche auf den Artenreichtum von Gefäßpflanzen in beweideten Wiesensteppen in Ungarn. Der dritte Artikel fragt, ob sich ungarische Sandtrockenrasen mit Festuca vaginata von solchen mit F. pseudovaginata in ihrer Vegetation und ihren Standortsbedingungen unterscheiden; die letztere war erst kürzlich beschrieben worden. Der vierte Artikel modelliert Auswirkungen des Klimawandels auf geschützte Graslandbestände in Serbien und deren Arten während schließlich der fünfte Artikel die Initiative einer neuen Datenbank des deutschen Graslands (GrassVeg.DE) vorstellt und dazu aufruft, sich daran zu beteiligen. Insgesamt haben zu diesem 12. Grasland-Sonderteil 41 Autoren aus fünf Ländern (Dänemark, Deutschland, Italien, Schweiz, Serbien und Ungarn) beigetragen.
This Special Feature is the tenth in a series devoted to dry grasslands and edited by members of the European Dry Grassland Group (EDGG; http://www.edgg.org) or its prede-cessor Arbeitsgruppe Trockenrasen that have been published in Tuexenia since 2005 (GAL-VÁNEK et al. 2012). With this issue we celebrate our tenth anniversary of the Tuexenia Dry Grassland Special Features.
Der diesjährige 9. Trockenrasen-Sonderteil von Tuexenia beginnt mit einem Bericht über die aktuellen Aktivitäten der European Dry Grassland Group (EDGG). Zunächst geben wir einen Überblick über die Entwicklung der Mitgliederzahl. Dann berichten wir vom letzten European Dry Grassland Meeting in Zamość (Polen, 2013) und vom letzten European Dry Grassland Field Workshop in Chakassien (Russland, 2013) und informieren über künftige Veranstaltungen der EDGG. Anschließend erläutern wir die Publikationsaktivitäten der EDGG. Im zweiten Teil des Editorials geben wir eine Einführung zu den fünf Artikeln des diesjährigen Trockenrasen-Sonderteils: Zwei davon beschäftigen sich mit der Syntaxonomie von Trockenrasen in Osteuropa: Der eine Artikel präsentiert erstmalig eine nach internationalen Nomenklaturregeln gültige Liste der Trockenrasengesellschaften Serbiens während der andere die Ergebnisse des zweiten EDGG-Field Workshop 2010 nach Podolien (Ukraine) vorstellt. Im dritten Artikel werden kryophytische Steppen in Südsibirien (Russland) analysiert. Der vierte Artikel analysiert in Böhmen (Tschechien) die bestimmenden Kräfte für Artenreichtum und Artenzusammensetzung der Trockenrasen. Der fünfte Artikel befasst sich mit der Auswahl von Wiederansiedlungsarten am Beispiel der Steppenrasen in Thüringen (Deutschland).
Background: Available data on the incidence and outcome of invasive fungal diseases (IFD) in children with hematological malignancies or after allogeneic hematopoietic stem cell transplantation (HSCT) are mostly based on monocenter, retrospective studies or on studies performed prior to the availability of newer triazoles or echinocandins.
Procedure: We prospectively collected clinical data on incidence, diagnostic procedures, management and outcome of IFD in children treated for hematological malignancies or undergoing HSCT in three major European pediatric cancer centers.
Results: A total of 304 children (median age 6.0 years) who underwent 360 therapies (211 chemotherapy treatments, 138 allogeneic HSCTs and/or 11 investigational chemotherapeutic treatments) were included in the analysis. Nineteen children developed proven/probable IFD, mostly due to Aspergillus (n = 10) and Candida spp. (n = 5), respectively. In patients receiving chemotherapy, 11 IFDs occurred, all during induction or re-induction therapy. None of these patients died due to IFD, whereas IFD was lethal in 3 of the 8 HSCT recipients with IFD. Significant differences among centers were observed with regard to the use of imaging diagnostics and the choice, initiation and duration of antifungal prophylaxis.
Conclusion: This prospective multicenter study provides information on the current incidence and outcome of IFD in the real life setting. Practice variation between the centers may help to ultimately improve antifungal management in children at highest risk for IFDs.
Neurological diseases associated with neuronal death are also accompanied by axonal denervation of connected brain regions. In these areas, denervation leads to a decrease in afferent drive, which may in turn trigger active central nervous system (CNS) circuitry rearrangement. This rewiring process is important therapeutically, since it can partially recover functions and can be further enhanced using modern rehabilitation strategies. Nevertheless, the cellular mechanisms of brain rewiring are not fully understood. We recently reported a mechanism by which neurons remodel their local connectivity under conditions of network-perturbance: hippocampal pyramidal cells can extend spine head protrusions (SHPs), which reach out toward neighboring terminals and form new synapses. Since this form of activity-dependent rewiring is observed only on some spines, we investigated the required conditions. We speculated, that the actin-associated protein synaptopodin, which is involved in several synaptic plasticity mechanisms, could play a role in the formation and/or stabilization of SHPs. Using hippocampal slice cultures, we found that ~70 % of spines with protrusions in CA1 pyramidal neurons contained synaptopodin. Analysis of synaptopodin-deficient neurons revealed that synaptopodin is required for the stability but not the formation of SHPs. The effects of synaptopodin could be linked to its role in Ca(2+) homeostasis, since spines with protrusions often contained ryanodine receptors and synaptopodin. Furthermore, disrupting Ca(2+) signaling shortened protrusion lifetime. By transgenically reintroducing synaptopodin on a synaptopodin-deficient background, SHP stability could be rescued. Overall, we show that synaptopodin increases the stability of SHPs, and could potentially modulate the rewiring of microcircuitries by making synaptic reorganization more efficient.
Molecular analysis of the ribosome recycling factor ABCE1 bound to the 30S post-splitting complex

Ribosome recycling by the twin-ATPase ABCE1 is a key regulatory process in mRNA translation and surveillance and in ribosome-associated protein quality control in Eukarya and Archaea. Here, we captured the archaeal 30S ribosome post-splitting complex at 2.8 Å resolution by cryo-electron microscopy. The structure reveals the dynamic behavior of structural motifs unique to ABCE1, which ultimately leads to ribosome splitting. More specifically, we provide molecular details on how conformational rearrangements of the iron–sulfur cluster domain and hinge regions of ABCE1 are linked to closure of its nucleotide-binding sites. The combination of mutational and functional analyses uncovers an intricate allosteric network between the ribosome, regulatory domains of ABCE1, and its two structurally and functionally asymmetric ATP-binding sites. Based on these data, we propose a refined model of how signals from the ribosome are integrated into the ATPase cycle of ABCE1 to orchestrate ribosome recycling.
Neurons which lose part of their input respond with a compensatory increase in excitatory synaptic strength. This observation is of particular interest in the context of neurological diseases, which are accompanied by the loss of neurons and subsequent denervation of connected brain regions. However, while the cellular and molecular mechanisms of pharmacologically induced homeostatic synaptic plasticity have been identified to a certain degree, denervation-induced homeostatic synaptic plasticity remains not well understood. Here, we employed the entorhinal denervation in vitro model to study the role of tumor necrosis factor alpha (TNFα) on changes in excitatory synaptic strength of mouse dentate granule cells following partial deafferentation. Our experiments disclose that TNFα is required for the maintenance of a compensatory increase in excitatory synaptic strength at 3–4 days post lesion (dpl), but not for the induction of synaptic scaling at 1–2 dpl. Furthermore, laser capture microdissection combined with quantitative PCR demonstrates an increase in TNFα-mRNA levels in the denervated zone, which is consistent with our previous finding on a local, i.e., layer-specific increase in excitatory synaptic strength at 3–4 dpl. Immunostainings for the glial fibrillary acidic protein and TNFα suggest that astrocytes are a source of TNFα in our experimental setting. We conclude that TNFα-signaling is a major regulatory system that aims at maintaining the homeostatic synaptic response of denervated neurons.
We present a report on the second annual symposium of the Dry Grassland Working Group, which took place 26.-28.08.05 in Münster and was entitled ‘Observation scales in dry grasslands’ The Dry Grassland Working Group is a discussion forum for all who are dealing with dry grasslands in central Europe. It organises annual symposia on various topics and aims to prepare the volumes on dry grasslands and closely related vegetation classes within the the ‘Synopsis of the plant communities of Germany’ The Synopsis will be based on a TURBOVEG database. As its structure and reference lists are now almost fixed, we will probably be able to start with data input at the beginning of 2006.
In the lower siliceous uplands of Central Europe, various types of nutrient-poor grasslands are widespread and grow intermingled. These species-rich grasslands, often dominated by taxa of the Festuca ovina aggregate, comprise various phytosociological classes. They are remnants of a historic rural lands - cape and are of conservation importance. Few studies on such grasslands are available and there has been disagreement in assigning them to appropriate habitat types or syntaxa. We investigated such nutrient-poor grasslands in the lower Aar valley (Middle Hesse, Rhenish Massif). We surveyed 104 vegetation plots distributed throughout the valley and identified syntaxa to (sub)association level. We carried out supervised classification combining cluster analysis, a priori assignment to classes based on prevailing diagnostic species, and regional refinement based on phi-value maximisation of the units. As a result, we classified five associations within four classes: Polytricho piliferi-Festucetum tenuifoliae/Galio harcynici-Deschampsietum flexuosae and Festuco rubrae-Genistelletum sagittalis (Calluno- Ulicetea), Jasiono montanae-Festucetum ovinae (Koelerio-Corynephoretea), Gentiano-Koelerietum pyramidatae (Festuco-Brometea) and Arrhenatheretum elatioris (Molinio-Arrhenatheretea). Ecologically, soil acidity (resulting from Ca concentrations of the bedrock) was the main cause of floristic dissimilarity of the grasslands and thus community differentiation. Many stands grew on soils with intermediate pH and showed a peculiar mixture of basiphilous and acidophilous species. We conclude that (i) our approach of supervised classification yields convincing reproducible results when a syntaxonomic system is adapted top-down to a geographically restricted area, (ii) nutrient-poor siliceous grasslands dominated by taxa of the Festuca ovina aggregate can be well assigned to ecologically meaningful syntaxa, and (iii) the nutrient-poor siliceous grasslands of the Lahn-Dill Highlands are of high conservation relevance and in urgent need of protection.
We report on the activities of the European Dry Grassland Group (EDGG) during the last year, namely the 8th European Dry Grassland Meeting in Uman', Ukraine in June 2011, the 3rd EDGG Research Expedition in Bulgaria in August 2011, the 4th EDGG Research Expedition in Sicily in April 2012, as well as the completed and forthcoming EDGG-coordinated special features in international journals. Then we provide a brief bibliometrical analysis of the Dry Grassland Special Features in Tuexenia since 2005. The 32 contributions of the years 2005–2011 constituted approx. 17% of the overall content of Tuexenia in this period. Including this 7th Dry Grassland Special Feature, sixty-one authors from 12 countries have contributed to these Special Features, guest-edited by yearly changing teams from a total of 16 guest editors. In the years with statistically reliable data, contributions in the Dry Grassland Special Features have been cited approximately four times as much as regular Tuexenia contributions. It is likely that this fact together with the internationality of the Special Features has contributed to the final inclusion of the journal in the Web of Science in 2011. Finally, we introduce the four research articles of this 7th Dry Grassland Special Feature. Two of them are focusing on vegetation change and restoration issues of cryptogam-rich sand dunes in the Netherlands and calcareous grasslands in Bavaria (Germany), respectively. The others, dealing with siliceous grasslands in Hesse (central Germany) and the results of EDGG Research Expedition 2009 to Transylvania (Romania), focus on syntaxonomy.
In our contribution, we report on the 6th European Dry Grassland Meeting held from 31 August to 1 September 2009 in Halle (Saale), Germany. The meeting was attended by 40 participants, who gave 15 oral and 17 poster presentations. The rapid positive development of the European Dry Grassland Group (EDGG), the organiser of this conference, is mentioned: the inclusion of the EDGG in the International Association for Vegetation Science (IAVS) as a working group, the establishment of two new subgroups focussing on Mediterranean and South-East European dry grasslands, respectively, and the organisation of the first EDGG research expedition in 2010 belong to the most important events. In the last part of our contribution, we give a short introduction to the six articles of this Special Feature. Two of them deal with phytosociological classification of semi-natural grassland communities, one with vegetation- environment relationships. Two papers are concerned with conservational topics, one focussing on the population structure of endangered Pulsatilla patens, the other dealing with conservation of xeric grasslands in Transylvania. The last paper examines temporal changes in calcareous grasslands with regard to species diversity.
GrassVeg.DE – die neue kollaborative Vegetationsdatenbank für alle Offenlandhabitate Deutschlands

Der Bericht stellt die neue kollaborative Vegetationsdatenbank GrassVeg.DE (EU-DE-020; http://bit.ly/2qgX208) vor, die Vegetationsaufnahmen von Grasländern und anderen nicht-aquatischen Offenlandhabitaten Deutschlands sammelt, um sie national und international für die vegetationsökologische Forschung zur Verfügung zu stellen. GrassVeg.DE trägt die Daten zum European Vegetation Archive (EVA) und künftig auch zur globalen Vegetationsdatenbank „sPlot“ bei. Datenlieferanten von GrassVeg.DE behalten volle Verfügungsgewalt über ihre Daten und werden Mitglied des GrassVeg.DE-Konsortiums. Dadurch profitieren sie durch Co-Autorenschaften und Zitate von ihren Beiträgen und erlangen zugleich die Möglichkeit, selbst Projekte zu beantragen, die GrassVeg.DE- oder EVA-Daten nutzen. Die schnell wachsende GrassVeg.DE-Datenbank umfasste im Juli 2017 3.181 Vegetationsaufnahmen aus acht deutschen Bundesländern. Perspektivisch kann GrassVeg.DE dazu beitragen, eine konsistente Neuklassifikation der Graslandvegetationstypen Deutschlands im Rahmen der Synopsis der Pflanzengesellschaften Deutschlands zu ermöglichen. Wir schließen den Beitrag mit einem Aufruf, eigene und aus der Literatur digitalisierte Vegetationsaufnahmen zu GrassVeg.DE beizutragen.
Der diesjährige 8. Trockenrasen-Sonderteil von Tuexenia beginnt mit einen Bericht über die aktuellen Aktivitäten der European Dry Grassland Group (EDGG). Zunächst geben wir einen Überblick über die Entwicklung der Mitgliederzahl und den aktuellen Vorstand, der im Mai 2013 gewählt wurde. Dann berichten wir vom letzten European Dry Grassland Meeting in Prespa (Griechenland, 2012) und informieren über künftige Tagungen und Forschungsexpeditionen der EDGG. Schließlich war und ist die EDGG sehr aktiv darin, Special Features in internationalen Fachzeitschriften herauszugeben. Im zweiten Teil des Vorwortes geben wir eine Einführung zu den sechs Artikeln des diesjährigen Trockenrasen-Sonderteils: Zwei davon beschäftigen sich Biodiversitätsanalyen von Grasland-Ökotonen in einer Flusslandschaft in Lettland bzw. von brachgefallenen Alvar-Trockenrasen in Estland. Der dritte Artikel behandelt die Ökologie und Vergesellschaftung einer Grassippe (Avenula adsurgens subsp. adsurgens) im brachgefallenen, montanen Grasland der Karpaten (Slowakei). Die letzten drei Artikel schließlich sind der Beschreibung und Syntaxonomie von Trockenrasen gewidmet: Zwei davon bilden den Start einer neuen Serie über die pannonischen Trockenrasen Österreichs (Allgemeine Einführung und Trockenrasen des Wienerwalds), während der letzte die Ergebnisse der dritten EDGG-Forschungsexpedition im Jahr 2011 nach Bulgarien präsentiert. Schließlich geben wir einen Ausblick über künftige Pläne für das Special Feature.
During postnatal development hippocampal dentate granule cells (GCs) often extend dendrites from the basal pole of their cell bodies into the hilar region. These so-called hilar basal dendrites (hBD) usually regress with maturation. However, hBDs may persist in a subset of mature GCs under certain conditions (both physiological and pathological). The functional role of these hBD-GCs remains not well understood. Here, we have studied hBD-GCs in mature (≥18 days in vitro) mouse entorhino-hippocampal slice cultures under control conditions and have compared their basic functional properties (basic intrinsic and synaptic properties) and structural properties (dendritic arborisation and spine densities) to those of neighboring GCs without hBDs in the same set of cultures. Except for the presence of hBDs, we did not detect major differences between the two GC populations. Furthermore, paired recordings of neighboring GCs with and without hBDs did not reveal evidence for a heavy aberrant GC-to-GC connectivity. Taken together, our data suggest that in control cultures the presence of hBDs on GCs is neither sufficient to predict alterations in the basic functional and structural properties of these GCs nor indicative of a heavy GC-to-GC connectivity between neighboring GCs.
In an ongoing clinical phase I/II study, 16 pediatric patients suffering from high risk leukemia/tumors received highly purified donor natural killer (NK) cell immunotherapy (NK-DLI) at day (+3) +40 and +100 post haploidentical stem cell transplantation. However, literature about the influence of NK-DLI on recipient's immune system is scarce. Here we present concomitant results of a noninvasive in vivo monitoring approach of recipient's peripheral blood (PB) cells after transfer of either unstimulated (NK-DLI(unstim)) or IL-2 (1000 U/ml, 9–14 days) activated NK cells (NK-DLI(IL-2 stim)) along with their ex vivo secreted cytokine/chemokines. We performed phenotypical and functional characterizations of the NK-DLIs, detailed flow cytometric analyses of various PB cells and comprehensive cytokine/chemokine arrays before and after NK-DLI. Patients of both groups were comparable with regard to remission status, immune reconstitution, donor chimerism, KIR mismatching, stem cell and NK-DLI dose. Only after NK-DLI(IL-2 stim) was a rapid, almost complete loss of CD56(bright)CD16(dim/−) immune regulatory and CD56(dim)CD16(+) cytotoxic NK cells, monocytes, dendritic cells and eosinophils from PB circulation seen 10 min after infusion, while neutrophils significantly increased. The reduction of NK cells was due to both, a decrease in patients' own CD69(−) NCR(low)CD62L(+) NK cells as well as to a diminishing of the transferred cells from the NK-DLI(IL-2 stim) with the CD56(bright)CD16(+/−)CD69(+)NCR(high)CD62L(−) phenotype. All cell counts recovered within the next 24 h. Transfer of NK-DLI(IL-2 stim) translated into significantly increased levels of various cytokines/chemokines (i.e. IFN-γ, IL-6, MIP-1β) in patients' PB. Those remained stable for at least 1 h, presumably leading to endothelial activation, leukocyte adhesion and/or extravasation. In contrast, NK-DLI(unstim) did not cause any of the observed effects. In conclusion, we assume that the adoptive transfer of NK-DLI(IL-2 stim) under the influence of ex vivo and in vivo secreted cytokines/chemokines may promote NK cell trafficking and therefore might enhance efficacy of immunotherapy.
hintergrund: Männer in Deutschland sterben früher als Frauen und nehmen weniger häufig Krebsvorsorgeuntersuchungen wahr.
Fragestellung: Ziel war die prospektive Evaluation einer „Movember-Gesundheitsinitiative“ am Universitätsklinikum Frankfurt (UKF) im November 2019.
Methoden: Im Rahmen der „Movember-Gesundheitsinitiative“ wurde allen männlichen Mitarbeitern des UKF ab dem 45. Lebensjahr und bei erstgradiger familiärer Vorbelastung eines Prostatakarzinoms ab dem 40. Lebensjahr im November 2019 gemäß S3-Leitlinien der Deutschen Gesellschaft für Urologie (DGU) eine Prostatakarzinom-Vorsorgeuntersuchung angeboten.
Ergebnisse: Insgesamt nahmen 14,4 % der Mitarbeiter teil. Eine familiäre Vorbelastung gaben insgesamt 14,0 % Teilnehmer an. Das mediane Alter betrug 54 Jahre. Der mediane PSA(prostataspezifisches Antigen)-Wert lag bei 0,9 ng/ml, der mediane PSA-Quotient bei 30 %. Bei 5 % (n = 6) zeigte sich ein suspekter Tastbefund in der DRU (digital-rektale Untersuchung). Nach Altersstratifizierung (≤ 50 vs. > 50 Lebensjahre) zeigten sich signifikante Unterschiede im medianen PSA-Wert (0,7 ng/ml vs. 1,0 ng/ml, p < 0,01) und der bereits zuvor durchgeführten urologischen Vorsorge (12,1 vs. 42,0 %, p < 0,01). Vier Teilnehmer (3,3 %) zeigten erhöhte Gesamt-PSA-Werte. Bei 32,2 % der Teilnehmer zeigte sich mindestens ein kontrollbedürftiger Befund. Insgesamt wurden 6 Prostatabiopsien durchgeführt. Hierbei zeigte sich in einem Fall ein intermediate-risk Prostatakarzinom (Gleason 3 + 4, pT3a, pPn1, pNx, R0).
Schlussfolgerungung: Im Rahmen der UKF-Movember-Gesundheitsinitiative 2019 konnten durch ein Vorsorgeangebot 121 Männer für eine Prostatakrebs-Vorsorge inklusive PSA-Testung gewonnen werden. Auffällige/kontrollbedürftige Befunde zeigten sich bei 32,2 %. Bei einem Mitarbeiter wurde ein therapiebedürftiges Prostatakarzinom entdeckt und therapiert.
Background: The potential anti-cancer effects of mammalian target of rapamycin (mTOR) inhibitors are being intensively studied. To date, however, few randomised clinical trials (RCT) have been performed to demonstrate anti-neoplastic effects in the pure oncology setting, and at present, no oncology endpoint-directed RCT has been reported in the high-malignancy risk population of immunosuppressed transplant recipients. Interestingly, since mTOR inhibitors have both immunosuppressive and anti-cancer effects, they have the potential to simultaneously protect against immunologic graft loss and tumour development. Therefore, we designed a prospective RCT to determine if the mTOR inhibitor sirolimus can improve hepatocellular carcinoma (HCC)-free patient survival in liver transplant (LT) recipients with a pre-transplant diagnosis of HCC. Methods: The study is an open-labelled, randomised, RCT comparing sirolimus-containing versus mTOR-inhibitor-free immunosuppression in patients undergoing LT for HCC. Patients with a histologically confirmed HCC diagnosis are randomised into 2 groups within 4-6 weeks after LT; one arm is maintained on a centre-specific mTOR-inhibitor-free immunosuppressive protocol and the second arm is maintained on a centre-specific mTOR-inhibitor-free immunosuppressive protocol for the first 4-6 weeks, at which time sirolimus is initiated. A 3-year recruitment phase is planned with a 5-year follow-up, testing HCC-free survival as the primary endpoint. Our hypothesis is that sirolimus use in the second arm of the study will improve HCC-free survival. The study is a non-commercial investigator-initiated trial (IIT) sponsored by the University Hospital Regensburg and is endorsed by the European Liver and Intestine Transplant Association; 13 countries within Europe, Canada and Australia are participating. Discussion: If our hypothesis is correct that mTOR inhibition can reduce HCC tumour growth while simultaneously providing immunosuppression to protect the liver allograft from rejection, patients should experience less post-transplant problems with HCC recurrence, and therefore could expect a longer and better quality of life. A positive outcome will likely change the standard of posttransplant immunosuppressive care for LT patients with HCC. Introduction: Prophylaxis with factor VIII (FVIII) concentrates in children with haemophilia A (HA) is current standard of care. The benefit of prophylactic treatment for adult HA patients is not commonly accepted.
Aim: To investigate the benefit of prophylaxis over on‐demand treatment in adult and elderly patients with severe or non‐severe HA in a real‐life setting.
Methods: Data from 163 patients comprising 1202 patient‐years were evaluated for 7.5 (±5.3) years. The effects on the annual bleeding rate (ABR, including spontaneous and traumatic bleeds) of treatment with a plasma‐derived FVIII concentrate, the patient's age and disease severity were investigated. The effect of changing the treatment from on demand to continuous prophylaxis on the patients’ ABRs was further analysed.
Results: Prophylaxis had the greatest effect on the ABRs of patients of any age with severe or non‐severe HA. The difference in ABR of all patients treated on demand (median 31.4; interquartile range (IQR) 27.6; N = 83) compared with those treated prophylactically (median 1.3; IQR 3.6; N = 122) was statistically significant (P < .05), even for patients with non‐severe HA (median 8.4; IQR 15.5; N = 11) vs median 1.5; IQR 4.2 (N = 17), P < .05). Patients, aged up to 88 years, switching from on demand to continuous prophylaxis showed the lowest median ABR (1.1; N = 51) after their regimen change.
Conclusion: Any (even low‐frequency) prophylaxis results in lower ABR than on‐demand treatment. Patients switching to prophylaxis benefitted the most, irrespective of age or HA severity. Prophylactic treatment—even tertiary—is the regimen of choice for patients of any age, including elderly patients, with severe or non‐severe HA.
Das Gebiet an der unteren Unstrut nimmt hinsichtlich seiner Vegetation und Flora eine Sonderstellung ein, da sich hier kontinental und (sub-)mediterran verbreitete Sippen mischen. In den Xerothermrasen profitieren die östlichen Sippen vom subkontinentalen Klima, während der vorherrschende Muschelkalk-Untergrund und die Sommerwärme die zahlreichen kalkliebenden südlichen Sippen begünstigen. Daneben stellt das Unstruttal ein bedeutsames Refugialgebiet reliktischer Steppen- und Mittelmeerarten dar (BECKER 2000). Alles zusammen bewirkt eine überdurchschnittlich hohe Artenvielfalt und eine hohe Zahl verschiedener Xerothermrasen-Gesellschaften. Diese Gesellschaften und die mikroklimatischen und edaphischen Gründe für ihre Verteilung im Raum zu beschreiben, ist ein Ziel der vorliegenden Arbeit (VGL. BECKER 1996; s.a. BECKER 1998a, b). Kleinere Teilgebiete des Unstruttales wurden hinsichtlich ihrer Xerothermrasen-Gesellschaften bereits untersucht, so das NSG Steinklöbe von MEUSEL (1937) sowie HÖLZEL (1997), das NSG Neue Göhle von ALTEHAGE (1951) und das NSG Tote Täler von REICHHOFF et al. (1979). Auch bei KRAUSE (1940) finden sich interessante Überlegungen zu einzelnen Arten, während HENSEN (1997), BÖTTNER et al. (1997), HENSEN & KENTRUP (1998) und HEINZ & PFEIFFER (1998) mehrere Gesellschaften im Gebiet hinsichtlich der Lebensstrategien ihrer Arten untersucht haben. Schließlich sei noch die Studie von MAHN (1965) genannt, mit der man die pflanzensoziologischen Ergebnisse in einen größeren Kontext stellen kann. Mit der Rolle von Mikroklima und Boden in herzynischen Xerothermrasen haben sich u.a. HELMECKE (1972), REICHHOFF (1979, 1980) und SCHLÜTER & BALLER (1982) befaßt. Nicht unerwähnt bleiben sollen die grundlegenden Arbeiten von VOLK (1936, 1937b), HEILIG (1931) und DÖRR (1941).
In den letzten etwa 150 Jahren haben sich zahlreiche Gletscher der Alpen infolge eines wärmeren Klimas zurückgezogen. Dies gilt auch für eine lange Gletscherzunge des Obersulzbachkees, die um mehr als 3 Kilometer zurück gegangen ist. Die einzelnen Stadien des Rückzugs sind recht gut datiert und waren Anlass für eine vergleichende Untersuchung der Vegetation in 6 datierten Bereichen im Vorfeld dieses Gletschers.
Ein Grundstock von über 20 Arten wuchs 1997 im gesamten Gletschervorfeld; die meisten der insgesamt 191 Arten waren aber absolut oder mit Schwerpunkt an einzelne Stadien gebunden. In einer indirekten Gradientenanalyse (DCA) erklärte der Zeitraum der Eisfreiheit 84% der Variation der Vegetation entlang der ersten Achse; damit war er für die Vegetation der bestimmende Faktor. Die α-Diversität der Vegetation stieg bis 55 Jahre Eisfreiheit kontinuierlich an; danach zeigte sie keinen eindeutigen Trend. Die Evenness fiel durch Dominanzbildung einzelner Arten im Laufe der Entwicklung ab. Zwei Sukzessionsstadien ließen sich unterscheiden: ein Cerastium uniflorum-Luzula alpinopilosa-Pionierstadium aus Arten der Schuttfluren i. w. S. mit zahlreichen alpinen Arten auf bis zu 55 Jahre alten Substrat und ein Festuca rubra-Nardus stricta-Rasenstadium ab etwa 110 Jahren Eisfreiheit mit einer Mischung aus Rasen-, Zwergstrauch- und Hochstauden-Pflanzen. Ersteres Stadium gliederte sich weiter in eine Oxyria digyna-Doronicum clusii-Phase auf jungem Substrat und eine Racomitrium canescens-Trifolium nivale-Phase nach mindestens 55 Jahren Eisfreiheit. Trotz oftmals fließender Übergänge ließen sich im Literaturvergleich 3 Assoziationen erkennen: das Sieversio-Oxyrietum digynae bis 25 Jahre, das Agrostio rupestris-Trifolietumpallentis bis 95 Jahre und das Sieversio-Nardetum strictae ab 110 Jahren Eisfreiheit. Auffällig war im gesamten Gletschervorfeld die vollständig ausgebliebene Ansiedlung von Bäumen; mögliche Gründe hierfür werden diskutiert.
Diese Arbeit untersucht die mitteldeutschen Vorkommen von Luzula divulgata, einer Sippe aus der Artengruppe um Luzula campestris und L. multiflora, deren ökologische Ansprüche und soziologisches Verhalten in Deutschland bisher kaum bekannt waren. Die östlich verbreitete Art wächst in Mitteldeutschland im oberen Bodetal, unteren Unstruttal, mittleren Saaletal und im Kyffhäusergebirge. Die isolierten Vorkommen bilden den nordwestlichen Arealrand der Art. L. divulgata ist im Gebiet an Eichen-Trockenwälder des Luzulo luzuloidis-Quercetum petraeae und Potentillo albae-Quercetum petraeae gebunden. Mitunter wächst sie auch in waldnahen Heiden, Säumen und Magerrasen. Das Klima der Wuchsgebiete ist sommerwarm und relativ trocken. Die Böden sind nahezu kalkfrei, mäßig sauer bis sauer und meist oligotroph. Das standortökologische Verhalten von L. divulgata wird mit Hilfe von Ellenberg-Zeigerwerten zusammengefasst; das Verbreitungsbild und die soziologische Bindung der Art werden diskutiert.
Introduction: We examined if a combination of proliferation markers and estrogen receptor (ER) activity could predict early versus late relapses in ER-positive breast cancer and inform the choice and length of adjuvant endocrine therapy.
Methods: Baseline affymetrix gene-expression profiles from ER-positive patients who received no systemic therapy (n = 559), adjuvant tamoxifen for 5 years (cohort-1: n = 683, cohort-2: n = 282) and from 58 patients treated with neoadjuvant letrozole for 3 months (gene-expression available at baseline, 14 and 90 days) were analyzed. A proliferation score based on the expression of mitotic kinases (MKS) and an ER-related score (ERS) adopted from Oncotype DX® were calculated. The same analysis was performed using the Genomic Grade Index as proliferation marker and the luminal gene score from the PAM50 classifier as measure of estrogen-related genes. Median values were used to define low and high marker groups and four combinations were created. Relapses were grouped into time cohorts of 0-2.5, 0-5, 5-10 years.
Results: In the overall 10 years period, the proportional hazards assumption was violated for several biomarker groups indicating time-dependent effects. In tamoxifen-treated patients Low-MKS/Low-ERS cancers had continuously increasing risk of relapse that was higher after 5 years than Low-MKS/High-ERS cancers [0 to 10 year, HR 3.36; p = 0.013]. High-MKS/High-ERS cancers had low risk of early relapse [0-2.5 years HR 0.13; p = 0.0006], but high risk of late relapse which was higher than in the High-MKS/Low-ERS group [after 5 years HR 3.86; p = 0.007]. The High-MKS/Low-ERS subset had most of the early relapses [0 to 2.5 years, HR 6.53; p < 0.0001] especially in node negative tumors and showed minimal response to neoadjuvant letrozole. These findings were qualitatively confirmed in a smaller independent cohort of tamoxifen-treated patients. Using different biomarkers provided similar results.
Conclusions: Early relapses are highest in highly proliferative/low-ERS cancers, in particular in node negative tumors. Relapses occurring after 5 years of adjuvant tamoxifen are highest among the highly-proliferative/high-ERS tumors although their risk of recurrence is modest in the first 5 years on tamoxifen. These tumors could be the best candidates for extended endocrine therapy.
Heterogenous subtypes of breast cancer need to be analyzed separately. Pooling of datasets can provide reasonable sample sizes but dataset bias is an important concern. We assembled a combined dataset of 579 Affymetrix microarrays from triple negative breast cancer (TNBC) in Gene Expression Omnibus (GEO) series GSE31519. We developed a method for selecting comparable datasets and to control for the amount of dataset bias of individual probesets.
Background: To evaluate the impact of time to castration resistance (TTCR) in metastatic hormone-sensitive prostate cancer (mHSPC) patients on overall survival (OS) in the era of combination therapies for mHSPC.
Material and Methods: Of 213 mHSPC patients diagnosed between 01/2013-12/2020 who subsequently developed metastatic castration resistant prostate cancer (mCRPC), 204 eligible patients were analyzed after having applied exclusion criteria. mHSPC patients were classified into TTCR <12, 12-18, 18-24, and >24 months and analyzed regarding OS. Moreover, further OS analyses were performed after having developed mCRPC status according to TTCR. Logistic regression models predicted the value of TTCR on OS.
Results: Median follow-up was 34 months. Among 204 mHSPC patients, 41.2% harbored TTCR <12 months, 18.1% for 12-18 months, 15.2% for 18-24 months, and 25.5% for >24 months. Median age was 67 years and median PSA at prostate cancer diagnosis was 61 ng/ml. No differences in patient characteristics were observed (all p>0.05). According to OS, TTCR <12 months patients had the worst OS, followed by TTCR 12-18 months, 18-24 months, and >24 months, in that order (p<0.001). After multivariable adjustment, a 4.07-, 3.31-, and 6.40-fold higher mortality was observed for TTCR 18-24 months, 12-18 months, and <12 months patients, relative to TTCR >24 months (all p<0.05). Conversely, OS after development of mCRPC was not influenced by TTCR stratification (all p>0.05).
Conclusion: Patients with TTCR <12 months are at the highest OS disadvantage in mHSPC. This OS disadvantage persisted even after multivariable adjustment. Interestingly, TTCR stratified analyses did not influence OS in mCRPC patients.
The objective of the study was to test the impact of implementing standard full functional-length urethral sphincter (FFLU) and neurovascular bundle preservation (NVBP) with intraoperative frozen section technique (IFT) on long-term urinary continence in patients undergoing robotic-assisted radical prostatectomy (RARP). We relied on an institutional tertiary-care database to identify patients who underwent RARP between 01/2014 and 09/2019. Until 10/2017, FFLU was not performed and decision for NVBP was taken without IFT. From 11/2017, FFLU and IFT-guided NVBP was routinely performed in all patients undergoing RARP. Long-term continence (≥ 12 months) was defined as the usage of no or one safety- pad. Uni- and multivariable logistic regression models tested the correlation between surgical approach (standard vs FFLU + NVBP) and long-term continence. Covariates consisted of age, body mass index, prostate volume and extraprostatic extension of tumor. The study cohort consisted of 142 patients, with equally sized groups for standard vs FFLU + NVBP RARP (68 vs 74 patients). Routine FFLU + NVBP implementation resulted in a long-term continence rate of 91%, compared to 63% in standard RARP (p < 0.001). Following FFLU + NVBP RARP, 5% needed 1–2, 4% 3–5 pads/24 h and no patient (0%) suffered severe long-term incontinence (> 5 pads/24 h). No significant differences in patient or tumor characteristics were recorded between both groups. In multivariable logistic regression models, FFLU + NVBP was a robust predictor for continence (Odds ratio [OR]: 7.62; 95% CI 2.51–27.36; p < 0.001). Implementation of FFLU and NVBP in patients undergoing RARP results in improved long-term continence rates of 91%.
Aim: Pharmacoresistance is a major burden in epilepsy treatment. We aimed to identify genetic biomarkers in response to specific antiepileptic drugs (AEDs) in genetic generalized epilepsies (GGE). Materials & methods: We conducted a genome-wide association study (GWAS) of 3.3 million autosomal SNPs in 893 European subjects with GGE – responsive or nonresponsive to lamotrigine, levetiracetam and valproic acid. Results: Our GWAS of AED response revealed suggestive evidence for association at 29 genomic loci (p <10-5) but no significant association reflecting its limited power. The suggestive associations highlight candidate genes that are implicated in epileptogenesis and neurodevelopment. Conclusion: This first GWAS of AED response in GGE provides a comprehensive reference of SNP associations for hypothesis-driven candidate gene analyses in upcoming pharmacogenetic studies.
