New measurements of the 7Be(n,α)4He and 7Be(n,p)7Li reaction cross sections from thermal to keV neutron energies have been recently performed at CERN/n_TOF. Based on the new experimental results, astrophysical reaction rates have been derived for both reactions, including a proper evaluation of their uncertainties in the thermal energy range of interest for big bang nucleosynthesis studies. The new estimate of the 7Be destruction rate, based on these new results, yields a decrease of the predicted cosmological 7Li abundance insufficient to provide a viable solution to the cosmological lithium problem.
The design and operation of innovative nuclear systems requires a better knowledge of the capture and fission cross sections of the Pu isotopes. For the case of capture on 242Pu, a reduction of the uncertainty in the fast region down to 8-12% is required. Moreover, aiming at improving the evaluation of the fast energy range in terms of average parameters, the OECD NEA High Priority Request List (HPRL) requests high-resolution capture measurements with improved accuracy below 2 keV. The current uncertainties also affect the thermal point, where previous experiments deviate from each other by 20%. A fruitful collaboration betwen JGU Mainz and HZ Dresden-Rossendorf within the EC CHANDA project resulted in a 242Pu sample consisting of a stack of seven fission-like targets making a total of 95(4) mg of 242Pu electrodeposited on thin (11.5 μm) aluminum backings. This contribution presents the results of a set of measurements of the 242Pu(n, γ) cross section from thermal to 500 keV combining different neutron beams and techniques. The thermal point was determined at the Budapest Research Reactor by means of Neutron Activation Analysis and Prompt Gamma Analysis, and the resolved (1 eV - 4 keV) and unresolved (1 - 500 keV) resonance regions were measured using a set of four Total Energy detectors at the CERN n_TOF-EAR1.
Setup for the measurement of the 235U(n,f) cross section relative to n-p scattering up to 1 GeV

The neutron induced fission of 235U is extensively used as a reference for neutron fluence measurements in various applications, ranging from the investigation of the biological effectiveness of high energy neutrons, to the measurement of high energy neutron cross sections of relevance for accelerator driven nuclear systems. Despite its widespread use, no data exist on neutron induced fission of 235U above 200 MeV. The neutron facility n_TOF offers the possibility to improve the situation. The measurement of 235U(n,f) relative to the differential n-p scattering cross-section, was carried out in September 2018 with the aim of providing accurate and precise cross section data in the energy range from 10 MeV up to 1 GeV. In such measurements, Recoil Proton Telescopes (RPTs) are used to measure the neutron flux while the fission events are detected and counted with dedicated detectors. In this paper the measurement campaign and the experimental set-up are illustrated.
The accuracy on neutron capture cross section of fissile isotopes must be improved for the design of future nuclear systems such as Gen-IV reactors and Accelerator Driven Systems. The High Priority Request List of the Nuclear Energy Agency, which lists the most important nuclear data requirements, includes also the neutron capture cross sections of fissile isotopes such as 233,235U and 239,241Pu. A specific experimental setup has been used at the CERN n_TOF facility for the measurement of the neutron capture cross section of 235U by a set of micromegas fission detectors placed inside a segmented BaF2 Total Absorption Calorimeter.
The experimental area 2 (EAR-2) at CERNs neutron time-of-flight facility (n_TOF), which is operational since 2014, is designed and built as a short-distance complement to the experimental area 1 (EAR-1). The Parallel Plate Avalanche Counter (PPAC) monitor experiment was performed to characterize the beam pro↓le and the shape of the neutron 'ux at EAR-2. The prompt γ-flash which is used for calibrating the time-of-flight at EAR-1 is not seen by PPAC at EAR-2, shedding light on the physical origin of this γ-flash.
Monte Carlo (MC) simulations are an essential tool to determine fundamental features of a neutron beam, such as the neutron flux or the γ-ray background, that sometimes can not be measured or at least not in every position or energy range. Until recently, the most widely used MC codes in this field had been MCNPX and FLUKA. However, the Geant4 toolkit has also become a competitive code for the transport of neutrons after the development of the native Geant4 format for neutron data libraries, G4NDL. In this context, we present the Geant4 simulations of the neutron spallation target of the n_TOF facility at CERN, done with version 10.1.1 of the toolkit. The first goal was the validation of the intra-nuclear cascade models implemented in the code using, as benchmark, the characteristics of the neutron beam measured at the first experimental area (EAR1), especially the neutron flux and energy distribution, and the time distribution of neutrons of equal kinetic energy, the so-called Resolution Function. The second goal was the development of a Monte Carlo tool aimed to provide useful calculations for both the analysis and planning of the upcoming measurements at the new experimental area (EAR2) of the facility.
Destruction of the cosmic γ-ray emitter 26Al in massive stars: study of the key 26Al(n,p) reaction

The 26Al(n,p)26Mg reaction is the key reaction impacting on the abundances of the cosmic γ-ray emitter 26Al produced in massive stars and impacts on the potential pollution of the early solar system with 26Al by asymptotic giant branch stars. We performed a measurement of the 26Al(n,p)26Mg cross section at the high-flux beam line EAR-2 at the n_TOF facility (CERN). We report resonance strengths for eleven resonances, nine being measured for the first time, while there is only one previous measurement for the other two. Our resonance strengths are significantly lower than the only previous values available. Our cross-section data range to 150 keV neutron energy, which is sufficient for a reliable determination of astrophysical reactivities up to 0.5 GK stellar temperature.
Purpose: Molecular diagnostics including next generation gene sequencing are increasingly used to determine options for individualized therapies in brain tumor patients. We aimed to evaluate the decision-making process of molecular targeted therapies and analyze data on tolerability as well as signals for efficacy.
Methods: Via retrospective analysis, we identified primary brain tumor patients who were treated off-label with a targeted therapy at the University Hospital Frankfurt, Goethe University. We analyzed which types of molecular alterations were utilized to guide molecular off-label therapies and the diagnostic procedures for their assessment during the period from 2008 to 2021. Data on tolerability and outcomes were collected.
Results: 413 off-label therapies were identified with an increasing annual number for the interval after 2016. 37 interventions (9%) were targeted therapies based on molecular markers. Glioma and meningioma were the most frequent entities treated with molecular matched targeted therapies. Rare entities comprised e.g. medulloblastoma and papillary craniopharyngeoma. Molecular targeted approaches included checkpoint inhibitors, inhibitors of mTOR, FGFR, ALK, MET, ROS1, PIK3CA, CDK4/6, BRAF/MEK and PARP. Responses in the first follow-up MRI were partial response (13.5%), stable disease (29.7%) and progressive disease (46.0%). There were no new safety signals. Adverse events with fatal outcome (CTCAE grade 5) were not observed. Only, two patients discontinued treatment due to side effects. Median progression-free and overall survival were 9.1/18 months in patients with at least stable disease, and 1.8/3.6 months in those with progressive disease at the first follow-up MRI.
Conclusion: A broad range of actionable alterations was targeted with available molecular therapeutics.
However, efficacy was largely observed in entities with paradigmatic oncogenic drivers, in particular with BRAF mutations. Further research on biomarker-informed molecular matched therapies is urgently necessary.
Im Zeitraum 1999 bis 2003 wurden im Harz 83 gezüchtete Auerhühner besendert und nach der Auswilderung telemetrisch überwacht. Das Hauptziel war dabei, über Daten zu Überlebenschancen, Raum- und Habitatnutzung und zur Ernährung der Auswilderungstiere eine effektive Erfolgskontrolle zu ermöglichen und Optimierungsvorschläge für zukünftige Auswilderungsbestrebungen geben zu können. Von den mit Sendern ausgestatteten Tieren überlebten ca. 23 % (n = 18) bis zum Sender- oder Kontaktverlust. Die mediane Überlebensdauer betrug 13 Tage, wobei Hennen länger überlebten (18 Tage) als Hähne (12 Tage). Die höchsten Verluste (79 %, n = 48) traten innerhalb der ersten vier Wochen nach der Ausbringung auf. Von 61 registrierten Totfunden entfielen 62 % auf den Fuchs, 10 % auf den Habicht und 7 % auf den Luchs. Neben Totfunden mit unklarer Ursache, verendeten einzelne Tiere im Verkehr, aufgrund von Unterernährung oder wurden von Hunden getötet. Die Hauptgründe für die hohen Verluste unter den ausgewilderten Auerhühnern sind in physiologischen und ethologischen Defiziten aufgrund der Gehegezucht zu suchen. Hier seien insbesondere mangelhafte Feindkenntnis, und die wahrscheinlich unzureichende Anpassung an die Freilandnahrung zu nennen. Zudem waren 5 % der Verluste auf angelockte Prädatoren im Umfeld der Eingewöhnungsvoliere zurückzuführen. Die Auswilderungstiere nutzten Aktionsräume (Suchgebiete) zwischen unter einem bis zu 17.100 ha, wobei die Mehrzahl der Aktionsräume (77 %) bis zu 1.000 ha umfasste. Die ermittelten Aktionsraumgröße und auch die maximale Entfernung zum Auswilderungsort nahmen mit der Ortungsdauer zu. Das mitunter weiträumige Verstreichen der Auswilderungstiere könnte auf die allgemein starke Dispersion juveniler Auerhühner, aber auch auf vom Habitat her ungeeignete Auswilderungsorte zurückzuführen sein. Innerhalb der Aktionsräume etablierten die Vögel räumlich kleinere Nutzungszentren die zumeist bis zum Totfund oder Senderverlust frequentiert blieben. Die Größe der Nutzungszentren (n = 17) variierte zwischen 0,5 ha und bis zu 327 ha. Täglich legten die Vögel in ca. 70 % aller Fälle nicht mehr als 500 m zurück. Etwa 67 % aller Ortungen entfielen auf einen 3-km-Radius um den Aussetzungsort. Maximalwerte verwiesen auf Distanzen bis ca. 22 km (W). Methodisch ist aufgrund der Ergebnisse die Auswilderung direkt aus den Transportkisten ohne Akklimatisierung in Eingewöhnungsvolieren zu empfehlen. So ließen sich zusätzliche Verluste am Auswilderungspunkt vermeiden, und zudem bliebe man flexibel in der Wahl des Auswilderungsortes. Weitere Optimierungsmaßnahmen in der Zucht und Auswilderungsmethodik sind unerlässlich (z. B. frühzeitig intensivierte artgerechte Ernährung, Trainingsprogramme, angemessene Krankheitsprophylaxe), ohne deren erfolgreiche Umsetzung die weitere Auswilderung von Gehegetieren nicht weiter verantwortbar bleibt.
In den hier skizzierten Kontext kollektiver Selbstverständigung zeitgenössischer Kulturdiagnostiker fügt sich auch das Mem-Konzept ein, dessen Urheber jedoch nicht im Entferntesten daran dachte, einen Beitrag zur Lösung kulturanthropologischer Grundprobleme zu leisten. Der englische Evolutionsbiologe Richard Dawkins, der den Neologismus "Mem" eingeführt hat, ist in einem ganz anderen Kontext beheimatet und verfolgt ganz andere Intentionen. Es war vor allem der Gedanke der Replikation, der Dawkins so fasziniert hat, dass er die Frage stellte, ob nicht auch die kulturelle Evolution als ein autoreproduktiver Replikationsprozess begriffen werden könnte. Aber wer oder was sollte sich replizieren? Ließ sich eine Struktureinheit mit einem variablen Komplexitätsgrad annehmen, die analog zum Gen als ein Replikator fungieren könnte? Die Antwort auf diese Frage war die Mem-Hypothese. Gene stellen als biologische Erbeinheiten eine Art von Grundbausteinen der belebten Materie dar. Daran anknüpfend führte Dawkins als Analogon zum Gen die Größe ein, die er Mem nannte. Er definierte Mem als die Einheit kultureller Vererbung, die sich dadurch auszeichnet, dass sie Kopien von sich herstellen kann. Aufgrund eines solchen Wirkungsvermögens können Meme als Quasiakteure der Replikation betrachtet werden, sie seien die Replikatoren, die die kulturelle Evolution in Gang hielten.
Der Altphilologe Johannes Stroux hat unmittelbar nach Kriegsende bemerkenswerte Anstrengungen unternommen, die Altertumswissenschaft aus ihrer Isoliertheit herauszuführen und in die von Künstlern, Wissenschaftlern, Intellektuellen unterschiedlicher Couleur, von Politikern, Gewerkschaftlern, Kirchenleuten angestrebte "Erneuerung der deutschen Kultur" zu integrieren. Stroux reagierte damit auf die Resignation vieler Altertumswissenschaftler, die sich in die Defensive gedrängt fühlten.
Wir haben das Feld der sozialen Situationen sondiert und die spektatorische Situation als eine spezifische Zeichensituation und Verkehrsform erörtert. Darüber hinaus sind problemgeschichtliche Aspekte der Figur des Zuschauers zur Sprache gekommen; gleichsam als Bestandteil konzeptioneller Vorarbeiten zur konkreten kultursemiotischen Untersuchung historischer Modelle von Spectatorship in ihrer funktionellen Typenvielfalt.
Von 1999 bis 2003 kamen im Nationalpark Harz (Niedersachsen) 83 gezüchtete juvenile Auerhühner frei, die zuvor mit Sendern ausgestattet und anschließend regelmäßig geortet wurden. Es wurde geprüft, ob die ausgewilderten Auerhühner ein ähnliches Nahrungsspektrum haben wie Wildvögel. Der Hintergrund dabei sind Erkenntnisse darüber, dass gezüchtete Hühnervögel aufgrund der hochverdaulichen und nährstoffreichen Volierennahrung ernährungsphysiologisch schlecht auf ein Leben im Freiland vorbereitet sind. Es wurden 109 Losungsproben gesammelt und die darin enthaltenen Nahrungsreste mikroskopisch aufbereitet, mit Hilfe einer Vergleichssammlung und der arttypischen Epidermiskennzeichen bestimmt und ihre Anteile geschätzt. Als Hauptnahrungskomponenten der im Herbst ausgewilderten Auerhühner sind für die Monate Oktober bis Februar Fichte (34 %), Heidelbeere (26 %) und Kräuter (20 %) zu nennen. Bei den im Frühjahr frei gelassenen Vögeln dominierten Fichte (56 %) und Gräser (20 %). Der Koniferenanteil bleibt jedoch in den Herbst- und insbesondere in den Wintermonaten hinter den Erwartungen zurück, besteht doch die Ernährung wild lebender Auerhühner im Winter nahezu ausschließlich aus Nadeln. Betrachtet man die drei Auswilderungstermine im Herbst (1999, 2002 und 2003) separat, fällt auf, dass der Fichtenanteil in den Monaten Dezember bis Februar mit minimal 18 % bis maximal 64 % auf äußerst niedrigem Niveau schwankt. Eine Umstellung auf die Winternadelnahrung, wie sie bei Wildvögeln bereits in den Herbstmonaten erfolgt, konnte nur anhand von Einzelproben nachgewiesen werden und verlief deutlich zeitversetzt (Februar). Günstige Witterungsbedingungen bis weit in die Wintermonate hinein (Verfügbarkeit der Krautschicht als Nahrungsquelle) und ernährungsphysiologische Defizite (geringe Verdauungsleistung) werden als Ursachen dafür diskutiert. Da das Hauptziel von solchen Artenschutzprogrammen die Ausbringung überlebensfähiger Individuen sein muss, ist zukünftig verstärkt darauf zu achten, dass die dafür notwendigen Grundlagen garantiert sind. Eine Voraussetzung dabei ist die Entwicklung umsetzbarer Fütterungsprogramme für Auswilderungszuchten und deren erfolgreiche Etablierung. Gelingen diese nicht, ist der weitere Einsatz von Zuchtvögeln für Ansiedlungsprojekte nicht zu befürworten.
Bei den im Rahmen des Forschungsverbundes erfolgten faunistischen Untersuchungen stand die Charakterisierung der unterschiedlichen, vor allem aber der wertvollen Biotoptypen der Braunkohlenfolgelandschaft mit Hilfe der dort lebenden Tierarten bzw. Artengemeinschaften im Vordergrund. Es wurden Tiergruppen bearbeitet, die zum einen ein hohes indikatorisches Potential besitzen, zum anderen unterschiedliche ökologische Hierachieebenen repräsentieren. Eine Auswertung der Ergebnisse erfolgte zumeist auf der Ebene der Biotoptypengruppen (siehe Heyde; Jakob; Köck; Reuter im gleichen Heft).
Die 'Weimarer Beiträge' haben in der Wissenschaftsgeschichte der DDR-Ästhetik eine wichtige Rolle gespielt. Sie waren Wegbegleiter einer hochschul- und wissenschaftspolitischen Innovation, die ebenso große Chancen wie Risiken barg. Mitte der sechziger Jahre setzte die Abteilung Ästhetik am Institut für Philosophie der Berliner Humboldt-Universität Stellung und Charakter der Ästhetik als philosophischer Disziplin aufs Spiel, ohne daß die Beteiligten wissen konnten, ob dies möglicherweise der erste Schritt zur allmählichen Selbstabschaffung war. Im Jahre 1965 wurde nach einiger Vorbereitung spruchreif, was sogleich unter dem Schlagwort "Auszug der Ästhetik aus der Philosophie " kursierte. Die Ästhetik machte sich (institutionell) selbständig und riskierte fortan die Marginalisierung im Feld der philosophischen Diskurse. Ein ähnlicher Vorgang vollzog sich in Leipzig. Zum Wintersemester 1963/64 wurde in Leipzig und Berlin ein neuer Studiengang Kulturwissenschaft eingerichtet, der bei den Studienbewerbern einen unerwartet starken Zulauf fand.
Der Studiengang Kulturwissenschaft verband eine philosophische Grundausbildung mit einer für alle Studierenden verbindlichen Spezialisierung (Ästhetik oder Kulturtheorie). Zugleich war ein einzelwissenschaftliches Nebenfach zu absolvieren, in Berlin zunächst beschränkt auf eine Kunstwissenschaft oder Philologie. Der Ansturm der Studenten war so groß, daß die kleinen Ästhetikabteilungen an den philosophischen Instituten hoffnungslos überfordert waren. Die Gründung selbständiger Institute für Ästhetik war nicht nur eine theoretische, sondern eine eminent praktische Frage. Das Netz der betrieblichen und kommunalen Kultureinrichtungen breitete sich damals über das ganze Land aus und war auf qualifizierte und kreative "Kulturarbeiter" angewiesen.
Wer Karla Kliches alltägliches Wirken als Redakteurin der Weimarer Beiträge längere Zeit beobachten konnte, den hat vor allem eine Haltung beeindruckt: diese Achtsamkeit gegenüber dem anderen, dieses Ernstnehmen von Autoren auch im kritischen Umgang mit ihnen, die Bewertung wissenschaftlicher Prosa nicht nur nach dispositionellen, sondern auch nach stilistischen Qualitäten, diese Wertschätzung neuer Gedanken - man muss es über Jahre hin erlebt haben, um all dies als etwas Seltenes und Kostbares zu schätzen zu wissen. Karla Kliches Umgangstugenden haben den Geist dieser Zeitschrift mitgeprägt
Immunomodulatory properties and molecular effects in inflammatory diseases of low-dose X-irradiation

Inflammatory diseases are the result of complex and pathologically unbalanced multicellular interactions. For decades, low-dose X-irradiation therapy (LD-RT) has been clinically documented to exert an anti-inflammatory effect on benign diseases and chronic degenerative disorders. By contrast, experimental studies to confirm the effectiveness and to reveal underlying cellular and molecular mechanisms are still at their early stages. During the last decade, however, the modulation of a multitude of immunological processes by LD-RT has been explored in vitro and in vivo. These include leukocyte/endothelial cell adhesion, adhesion molecule and cytokine/chemokine expression, apoptosis induction, and mononuclear/polymorphonuclear cell metabolism and activity. Interestingly, these mechanisms display comparable dose dependences and dose-effect relationships with a maximum effect in the range between 0.3 and 0.7 Gy, already empirically identified to be most effective in the clinical routine. This review summarizes data and models exploring the mechanisms underlying the immunomodulatory properties of LD-RT that may serve as a prerequisite for further systematic analyses to optimize low-dose irradiation procedures in future clinical practice.
Background: In a previous analysis (Int J Radiat Oncol Biol Phys 70:828-835,2010), we assessed whether an adjuvant supplementation with selenium (Se) improves Se status and reduces the radiation-induced side-effects of patients treated by adjuvant radiotherapy (RT) for cervical and uterine cancer. Now, a potential relation between the planning target volume (PTV) of the RT and the Se effect concerning radiation induced diarrhoea was evaluated in detail.
Methods: Whole blood Se concentrations had been measured in patients with cervical (n=11) and uterine cancer (n=70) after surgical treatment, during, and at the end of RT. Patients with initial Se concentrations of less than 84 μg/l were categorized as Se-deficient and randomized before RT to receive Se (as sodium selenite) per os on the days of RT, or to receive no supplement during RT. Diarrhoea was graded according to the Common Toxicity Criteria system (CTC, Version 2a). The evaluation of the PTV of the RT was ascertained with the help of a specialised computer-assisted treatment planning software used for radiation planning procedure.
Results: A total of 81 patients had been randomized for the initial supplementation study, 39 of which received Se [selenium group, SeG] and 42 serving as controls [control group, CG]. Mean Se levels did not differ between SeG and CG upon study initiation, but were significantly higher in the SeG compared to the CG at the end of RT. The actuarial incidence of at least CTC 2 radiation induced diarrhoea in the SeG was 20.5% compared to 44.5% in the CG (p=0.04). The median PTV in both groups was 1302 ml (916–4608). With a PTV of <= 1302 ml (n=41) the actuarial incidence of at least CTC 2 diarrhoea in the SeG was 22.3% (4 of 18 patients) compared to 34.8% (8 of 23 patients) in the CG (p=0.50). In patients with a PTV of > 1302 ml (n=40) the actuarial incidence of at least CTC 2 diarrhoea in the SeG was 19.1% (4 of 21 patients) versus 52.6% (10 of 19 patients) in the CG (p=0.046).
Conclusions: Se supplementation during RT was effective to improve blood Se status in Se-deficient cervical and uterine cancer patients, and reduces episodes and severity of RT-induced diarrhoea. This effect was most pronounced and significant in patients with large PTV (> 1302 ml).
The huge neutron fluxes offer the possibility to use research reactors to produce isotopes of interest, which can be investigated afterwards. An example is the half-lives of long-lived isotopes like 129I. A direct usage of reactor neutrons in the astrophysical energy regime is only possible, if the corresponding ions are not at rest in the laboratory frame. The combination of an ion storage ring with a reactor and a neutron guide could open the path to direct measurements of neutron-induced cross sections on short-lived radioactive isotopes in the astrophysically interesting energy regime.
The immune suppressive microenvironment affects efficacy of radio-immunotherapy in brain metastasis

The tumor microenvironment in brain metastases is characterized by high myeloid cell content associated with immune suppressive and cancer-permissive functions. Moreover, brain metastases induce the recruitment of lymphocytes. Despite their presence, T-cell-directed therapies fail to elicit effective anti-tumor immune responses. Here, we seek to evaluate the applicability of radio- immunotherapy to modulate tumor immunity and overcome inhibitory effects that diminish anti-cancer activity. Radiotherapy- induced immune modulation resulted in an increase in cytotoxic T-cell numbers and prevented the induction of lymphocyte-mediated immune suppression. Radio-immunotherapy led to significantly improved tumor control with prolonged median survival in experi- mental breast-to-brain metastasis. However, long-term efficacy was not observed. Recurrent brain metastases showed accumula- tion of blood-borne PD-L1+ myeloid cells after radio-immunother- apy indicating the establishment of an immune suppressive environment to counteract re-activated T-cell responses. This finding was further supported by transcriptional analyses indicat- ing a crucial role for monocyte-derived macrophages in mediating immune suppression and regulating T-cell function. Therefore, selective targeting of immune suppressive functions of myeloid cells is expected to be critical for improved therapeutic efficacy of radio-immunotherapy in brain metastases.
Background Endothelium-derived nitric oxide plays an important role for the bone marrow microenvironment. Since several important effects of nitric oxide are mediated by cGMP-dependent pathways, we investigated the role of the cGMP downstream effector cGMP-dependent protein kinase I (cGKI) on postnatal neovascularization. Methodology/Principal Findings In a disc neovascularization model, cGKI -/- mice showed an impaired neovascularization as compared to their wild-type (WT) littermates. Infusion of WT, but not cGKI -/- bone marrow progenitors rescued the impaired ingrowth of new vessels in cGKI-deficient mice. Bone marrow progenitors from cGKI -/- mice showed reduced proliferation and survival rates. In addition, we used cGKI alpha leucine zipper mutant (LZM) mice as model for cGKI deficiency. LZM mice harbor a mutation in the cGKI alpha leucine zipper that prevents interaction with downstream signaling molecules. Consistently, LZM mice exhibited reduced numbers of vasculogenic progenitors and impaired neovascularization following hindlimb ischemia compared to WT mice. Conclusions/Significance Our findings demonstrate that the cGMP-cGKI pathway is critical for postnatal neovascularization and establish a new role for cGKI in vasculogenesis, which is mediated by bone marrow-derived progenitors.
Vismodegib, an inhibitor of the Hedgehog signaling pathway, is an approved drug for monotherapy in locally advanced or metastatic basal cell carcinoma (BCC). Data on combined modality treatment by vismodegib and radiation therapy, however, are rare. In the present study, we examined the radiation sensitizing effects of vismodegib by analyzing viability, cell cycle distribution, cell death, DNA damage repair and clonogenic survival in three-dimensional cultures of a BCC and a head and neck squamous cell carcinoma (HNSCC) cell line. We found that vismodegib decreases expression of the Hedgehog target genes glioma-associated oncogene homologue (GLI1) and the inhibitor of apoptosis protein (IAP) Survivin in a cell line- and irradiation-dependent manner, most pronounced in squamous cell carcinoma (SCC) cells. Furthermore, vismodegib significantly reduced proliferation in both cell lines, while additional irradiation only slightly further impacted on viability. Analyses of cell cycle distribution and cell death induction indicated a G1 arrest in BCC and a G2 arrest in HNSCC cells and an increased fraction of cells in SubG1 phase following combined treatment. Moreover, a significant rise in the number of phosphorylated histone-2AX/p53-binding protein 1 (γH2AX/53BP1) foci in vismodegib- and radiation-treated cells was associated with a significant radiosensitization of both cell lines. In summary, these findings indicate that inhibition of the Hedgehog signaling pathway may increase cellular radiation response in BCC and HNSCC cells.
Purpose: Recent advances in the treatment algorithm of locally advanced rectal cancer (LARC) have significantly improved complete response (CR) rates and disease-free survival (DFS), but therapy resistance, with its substantial impact on outcomes and survival, remains a major challenge. Our group has recently unraveled a critical role of interleukin-1α (IL-1α) signaling in activating inflammatory cancer-associated fibroblasts (iCAFs) and mediating radiation-induced senescence, extracellular matrix (ECM) accumulation, and ultimately therapy resistance. We here summarize the recently initiated ACO/ARO/AIO-21 phase I trial, testing the IL-1 receptor antagonist (IL-1 RA) anakinra in combination with fluoropyrimidine-based chemoradiotherapy (CRT) for advanced rectal cancer.
Methods/Design: The ACO/ARO/AIO-21 is an investigator-driven, prospective, open-labeled phase I drug-repurposing trial assessing the maximum tolerated dose (MTD) of capecitabine administered concurrently to standard preoperative radiotherapy (45 Gy in 25 fractions followed by 9 Gy boost in 5 fractions) in combination with fixed doses of the IL-1RA anakinra (100 mg, days −10 to 40). Capecitabine will be administered using a 3 + 3 dose-escalation design (500 mg/m2 bid; 650 mg/m2 bid; 825 mg/m2 bid, respectively) from day 1 to day 40. Response assessment including digital rectal examination (DRE), endoscopy and pelvic magnetic resonance imaging (MRI) is scheduled 10 weeks after completion of CRT. For patients achieving clinical complete response (cCR), primary non-operative management is provided. In case of non-cCR immediate total mesorectal excision (TME) will be performed. Primary endpoint of this phase I trial is the MTD of capecitabine.
Discussion: Based on extensive preclinical research, the ACO/ARO/AIO-21 phase I trial will assess whether the IL-1RA anakinra can be safely combined with fluoropyrimidine-based CRT in rectal cancer. It will further explore the potential of IL-1 inhibition to overcome therapy resistance and improve response rates. A comprehensive translational research program will expand our understanding from a clinical perspective and may help translate the results into a randomized phase II trial.
Spatial and temporal processes shaping microbial communities are inseparably linked but rarely studied together. By Illumina 16S rRNA sequencing, we monitored soil bacteria in 360 stations on a 100 square meter plot distributed across six intra-annual samplings in a rarely managed, temperate grassland. Using a multi-tiered approach, we tested the extent to which stochastic or deterministic processes influenced the composition of local communities. A combination of phylogenetic turnover analysis and null modeling demonstrated that either homogenization by unlimited stochastic dispersal or scenarios, in which neither stochastic processes nor deterministic forces dominated, explained local assembly processes. Thus, the majority of all sampled communities (82%) was rather homogeneous with no significant changes in abundance-weighted composition. However, we detected strong and uniform taxonomic shifts within just nine samples in early summer. Thus, community snapshots sampled from single points in time or space do not necessarily reflect a representative community state. The potential for change despite the overall homogeneity was further demonstrated when the focus shifted to the rare biosphere. Rare OTU turnover, rather than nestedness, characterized abundance-independent β-diversity. Accordingly, boosted generalized additive models encompassing spatial, temporal and environmental variables revealed strong and highly diverse effects of space on OTU abundance, even within the same genus. This pure spatial effect increased with decreasing OTU abundance and frequency, whereas soil moisture – the most important environmental variable – had an opposite effect by impacting abundant OTUs more than the rare ones. These results indicate that – despite considerable oscillation in space and time – the abundant and resident OTUs provide a community backbone that supports much higher β-diversity of a dynamic rare biosphere. Our findings reveal complex interactions among space, time, and environmental filters within bacterial communities in a long-established temperate grassland.
The study of neutron-induced reactions is of high relevance in a wide variety of fields, ranging from stellar nucleosynthesis and fundamental nuclear physics to applications of nuclear technology. In nuclear energy, high accuracy neutron data are needed for the development of Generation IV fast reactors and accelerator driven systems, these last aimed specifically at nuclear waste incineration, as well as for research on innovative fuel cycles. In this context, a high luminosity Neutron Time Of Flight facility, n_TOF, is operating at CERN since more than a decade, with the aim of providing new, high accuracy and high resolution neutron cross-sections. Thanks to the features of the neutron beam, a rich experimental program relevant to nuclear technology has been carried out so far. The program will be further expanded in the near future, thanks in particular to a new high-flux experimental area, now under construction.
The n_TOF facility operates at CERN with the aim of addressing the request of high accuracy nuclear data for advanced nuclear energy systems as well as for nuclear astrophysics. Thanks to the features of the neutron beam, important results have been obtained on neutron induced fission and capture cross sections of U, Pu and minor actinides. Recently the construction of another beam line has started; the new line will be complementary to the first one, allowing to further extend the experimental program foreseen for next measurement campaigns.
Purpose: Medically recommended training often faces the dilemma that necessary mechanical intensities for muscle adaptations exceed patients' physical capacity. In this regard, blood flow restriction (BFR) training is becoming increasingly popular because it enables gains in muscle mass and strength despite using low-mechanical loads combined with external venous occlusion. Since the underlying mechanisms are still unknown, we applied invasive measurements during exercise with and without BFR to promote physiological understanding and safety of this popular training technique. Methods: In a randomized cross-over design, ten healthy men (28.1 ± 6.5 years) underwent two trials of unilateral biceps curls either with (BFR) and without BFR (CON). For analysis of changes in intravascular pressures, blood gases, oximetry and electrolytes, an arterial and a venous catheter were placed at the exercising arm before exercise. Arterial and venous blood gases and intravascular pressures were analyzed before, during and 5 min after exercise. Results: Intravascular pressures in the arterial and venous system were more increased during exercise with BFR compared to CON (p < 0.001). Furthermore, arterial and venous blood gas analyses revealed a BFR-induced metabolic acidosis (p < 0.05) with increased lactate production (p < 0.05) and associated elevations in [K+], [Ca2+] and [Na+] (p < 0.001). Conclusion: The present study describes for the first time the local physiological changes during BFR training. While BFR causes greater hypertension in the arterial and venous system of the exercising extremity, observed electrolyte shifts corroborate a local metabolic acidosis with concurrent rises in [K+] and [Na+]. Although BFR could be a promising new training concept for medical application, its execution is associated with comprehensive physiological challenges.
Im vorliegenden Beitrag soll untersucht werden, welche Akteur- und Zuschauerkonstellationen sich in Online-Communities aufweisen lassen, die Millionen individueller Nutzer durch Social Software miteinander verbinden. Sind spektatorische Situationen, in denen interagierende soziale Akteure auf einen Dritten treffen, der ihnen zuschaut, auch in medialisierten Formen des sozialen Verkehrs wirksam, in denen die Hauptbedingung der Face-to-Face-Kommunikation, die körperliche Kopräsenz von Akteuren, Co-Akteuren und zuschauenden Dritten, nicht mehr zwingend gegeben ist? Es hat sich erwiesen, daß die webbasierte Gruppendynamik nur dann angemessen zu erfassen ist, wenn die mehrstelligen Akteur- und Zuschauerkonstellationen analysiert werden, in denen sich die (Selbst-)Strukturierungsprozesse in sozialen Gruppen vollziehen. Die Herausforderungen des Web 2.0 haben Neuansätze zu sozial- und kulturwissenschaftlichen Akteurtheorien befördert; das Konzept der spektatorischen Situation, das sich auf Perspektiven- und Positionswechsel zwischen Ego-Akteuren, Alter-Akteuren und zuschauenden Dritten konzentriert, kann als eine Variante dieser neuen Akteurtheorien begriffen werden.
Tierfilme und -reportagen haben Konjunktur, neue Bildtechnologien ermöglichen immer differenziertere Einblicke in Verhaltens- und Lebensweisen bekannter und unbekannter Tierarten. Wir fühlen uns als teilnehmende Beobachter in scheinbar gewagtester Nähe. Das "Privatleben" der Tiere wird bis in den letzten Winkel verfolgt, die Entdeckung der Tierindividualitäten schreitet voran, Tiere bekommen ein Schicksal und wecken Empathie, der vor über 2000 Jahren entstandene Tier-Mensch-Verwandtschafts-Topos, dem noch jeder Gedanke an die Abstammung der Arten fern lag, wird neu belebt. Doch das kulturelle Interesse am Tier erschöpft sich nicht im Fasziniertsein durch perfekt visualisierte Verhaltensstudien oder durch neueste Lesarten der Tiermetaphorik, in der es stets weniger um Tiere als um Menschen geht. Der historische Wandel der menschlichen Tierbeziehung wird in der Tierethik reflektiert.
Seit der Antike umstrittene kognitive Fähigkeiten verschiedener Tierarten bilden heute einen einzelwissenschaftlich fundierten philosophischen Diskussions- und Forschungsgegenstand ("Der Geist der Tiere"). Die Biosemiotik macht große Fortschritte, der Tier-Mensch-Vergleich - bereits ein Kernstück antiker Anthropologie und Ethik - erfährt eine tiefgreifende Umwertung. Dabei steht nicht mehr von vornherein der Mensch im Mittelpunkt; vielmehr wird versucht, das Animalische in seiner Artenvielfalt als eine vielgestaltige eigene Lebensform zu beschreiben; diese ist nicht länger an Maßstäben einer Anthropozentrik zu messen, die im innersten teleologisch geblieben ist. Der Tier-Mensch-Vergleich wird heute nicht mehr so einseitig und ausschließlich auf kognitive Fähigkeiten bezogen. Vielmehr werden weitere Aspekte wie Emotionalität, Wertungs- bzw. Präferenzverhalten, moralanaloge Verhaltensweisen, Antriebsstrukturen, Soziabilität, Zeichenverhalten mit einbezogen. Diese Komplexität der Vergleichspunkte oder Vergleichseinheiten war erst mit dem neuzeitlichen Rationalismus immer mehr eingeschränkt worden. Die gegenwärtig wiedergewonnene Komplexität lässt nach historischen Vorbildern Ausschau halten; da verwundert es nicht, wenn eine Anthropologie ins Blickfeld rückt, die im Tier-Mensch-Vergleich einen ihrer wichtigsten Ausgangspunkte hatte: die antike Anthropologie.
Am 14. November 1956, vormittags gegen zehn Uhr, versammelten sich die Studenten des Philosophischen Instituts der Berliner Humboldt-Universität vor dem Haus Universitätsstraße 3b, dem Sitz des Instituts, zu einem kleinen Demonstrationszug, der angemeldet gewesen sein musste, denn er wurde von zwei Polizeimotorrädern, eines vorne, eines hinten, durch den Verkehr geleitet. Angeführt vom Institutsdirektor Georg Klaus bewegte sich der Zug über die Weidendammer Brücke die Friedrichstraße entlang bis zum Dorotheenstädtischen Friedhof an der Chausseestraße. Anlass war der 150. Todestag von Georg Wilhelm Friedrich Hegel, was den meisten der neugierig zuschauenden Passanten nur wenig sagte. An Hegels Grab wurden Blumen niedergelegt, Reden wurden nicht gehalten, einige Studenten ließen es sich nicht nehmen, auf dem Grabstein, einer alten Sitte gemäß, ein paar Münzen niederzulegen. Es herrschte eine fast fröhliche Stimmung, die von einem Gefühl der Genugtuung erfüllt war, hatte man doch über die Stalinistische Fronde gegen Hegel gesiegt. Ein paar Stunden später traf man sich wieder im vollbesetzten Audi Max, in dem Ernst Bloch einen Vortrag über Hegel und das offene System der marxistischen Philosophie hielt. Zehn Tage zuvor waren sowjetische Panzer in Budapest eingerückt, um eine exzessive Massenerhebung niederzuwalzen. Der Zorn der Massen hatte sich gegen das stalinistische Rákosi-Regime und seine Geheimpolizei gerichtet; die Russen machten jedoch die neue Regierung Imre Nagy für die inneren Unruhen verantwortlich. Die Nagy-Regierung wurde abgesetzt und kriminalisiert, darunter der streitbare Marxist und profunde Hegel-Kenner Georg Lukács. Der Vorgang führte bald auch in der Universitätsstraße 3b zu heftigen Diskussionen. Eine Anfang 1957 als Reforminitiative konzipierte studentische Streitschrift, die die ungarischen Ereignisse als eine Folge der systematischen Verhinderung einer freien öffentlichen Diskussion erklärte, zog die Aufmerksamkeit der SED-Zentrale auf das Institut und löste eine Kette von Parteiverfahren und Maßregelungen aus. Vier Studenten waren von mehrjähriger Studienunterbrechung zwecks Praxisbewährung betroffen, anderthalb Jahre später folgte ein von der Stasi vorbereiteter Schauprozess, der für drei weitere Studenten mit Haftstrafen bis zu sechs Jahren endete.
Der im vergangenen Jahr verstorbene britische Philosoph Bernard Williams hat 1993 sein Buch 'Shame and Necessity' vorgelegt. Williams verheißt eine Befreiung der Antike – vor allem aus den Banden einer universalistischen Fortschrittskonzeption von Evolution und Geschichte; nach deren Maßstäben müssen den Griechen "primitive Ideen des Handelns, der Verantwortung, der ethischen Motivation und der Gerechtigkeit" zugeschrieben werden, "die im Laufe der Geschichte durch ein wesentlich komplexeres und feineres Netz von Vorstellungen ersetzt worden sind, in denen sich eine reifere Form der ethischen Erfahrung niederschlägt".
Es geht Williams nicht um die Fortschreibung der Tradition nach dem Muster einer übergreifenden europäischen Entwicklungskontinuität, sondern um die Aufsprengung einer solchen Kontinuität. Er sieht sich in der Schuld der Cambridge Ritualists, die uns daran gewöhnt hätten, an die Gesellschaften des antiken Griechenland mit Methoden heranzutreten, die denen der Kulturanthropologen ähnlich seien und neue Wege zur Entzifferung der Mythen und Rituale gebahnt hätten. Die Anthropologie der Antike habe uns die Griechen fremd gemacht und einen Sinn für ihre Andersheit erzeugt, der erst erlaubt, konkret historisch nach Ähnlichkeiten und Unterschieden zu fragen. Die Antike aus einer übergreifenden Entwicklungskontinuität herauszusprengen, ist Mittel und Voraussetzung dafür, sie als "historische Formation" wiederzugewinnen. Auch wenn die antike Kultur ihren geschichtsphilosophisch privilegierten Status verloren hat, sieht Williams die Europäer in einem besonderen Verhältnis zu den "alten Griechen". Ohne blind zu sein für das, "was die Geschichte der europäischen Herrschaft zerstört oder beiseitegeschoben hat", sei das, "was wir von den Griechen lernen können", wenn wir sie mit neuen Augen sehen, Bestandteil eines soziokulturellen Selbstverständigungsprozesses, in dem Figuren des Europäischen zur Debatte stehen. Hauptgegenstand von Williams’ Buch ist die Relevanz antiker Personalitätsmodelle für die gegenwärtige Ethikdebatte.
Background: In intensive care units (ICU) octogenarians become a routine patients group with aggravated therapeutic and diagnostic decision-making. Due to increased mortality and a reduced quality of life in this high-risk population, medical decision-making a fortiori requires an optimum of risk stratification. Recently, the VIP-1 trial prospectively observed that the clinical frailty scale (CFS) performed well in ICU patients in overall-survival and short-term outcome prediction. However, it is known that healthcare systems differ in the 21 countries contributing to the VIP-1 trial. Hence, our main focus was to investigate whether the CFS is usable for risk stratification in octogenarians admitted to diversified and high tech German ICUs.
Methods: This multicentre prospective cohort study analyses very old patients admitted to 20 German ICUs as a sub-analysis of the VIP-1 trial. Three hundred and eight patients of 80 years of age or older admitted consecutively to participating ICUs. CFS, cause of admission, APACHE II, SAPS II and SOFA scores, use of ICU resources and ICU- and 30-day mortality were recorded. Multivariate logistic regression analysis was used to identify factors associated with 30-day mortality.
Results: Patients had a median age of 84 [IQR 82–87] years and a mean CFS of 4.75 (± 1.6 standard-deviation) points. More than half of the patients (53.6%) were classified as frail (CFS ≥ 5). ICU-mortality was 17.3% and 30-day mortality was 31.2%. The cause of admission (planned vs. unplanned), (OR 5.74) and the CFS (OR 1.44 per point increase) were independent predictors of 30-day survival.
Conclusions: The CFS is an easy determinable valuable tool for prediction of 30-day ICU survival in octogenarians, thus, it may facilitate decision-making for intensive care givers in Germany.

A recent global meta‐analysis reported a decrease in terrestrial but increase in freshwater insect abundance and biomass (van Klink et al., Science 368, p. 417). The authors suggested that water quality has been improving, thereby challenging recent reports documenting drastic global declines in freshwater biodiversity. We raise two major concerns with the meta‐analysis and suggest that these account for the discrepancy with the declines reported elsewhere. First, total abundance and biomass alone are poor indicators of the status of freshwater insect assemblages, and the observed differences may well have been driven by the replacement of sensitive species with tolerant ones. Second, many of the datasets poorly represent global trends and reflect responses to local conditions or nonrandom site selection. We conclude that the results of the meta‐analysis should not be considered indicative of an overall improvement in the condition of freshwater ecosystems.
Die vorliegende Arbeit beschäftigt sich mit der Erfassung und Beschreibung von elf Waldresten im südlich der Donau liegenden Teil des Wiener Beckens. Das Untersuchungsgebiet ist durch von Löss bedeckte Schotterterrassen geprägt. Die Waldreste wurden floristisch erfasst und syntaxonomisch bearbeitet. Zusätzlich wurde in den Franziszäischen Kataster von 1820 Einsicht genommen, um genauere Informationen über die damaligen Nutzungsformen der heutigen Waldflächen zu erhalten. Die Eichen- Hainbuchenwälder wurden in das Polygonato latifolii-Carpinetum, die Eichenwälder großenteils in das Euphorbio angulatae-Quercetum eingereiht. Innerhalb dieser Assoziation wurden vier Ausbildungen ausgewiesen (degradiert, frisch, mäßig trocken und trocken). Eine Aufnahme wurde zum Lithospermo purpurocaerulei-Quercetum pubescentis gestellt. Drei Aufnahmen aus Beständen, die deutlich durch die Robinie überprägt sind, wurden als Balloto-Robinietum klassifiziert. Das Areal dieser Eichen- und Eichen-Hainbuchenwälder ist altes Siedlungsgebiet und wird seit Jahrhunderten (z.T. Jahrtausenden) genutzt, weshalb der Wald nur noch in Form von Fragmenten erhalten ist. Weiters sind neophytische Baumarten wie die Robinie und der Götterbaum ein Problem für den Fortbestand dieser Wälder, da sie die natürliche Artenzusammensetzung der Bestände verändern.
NIMA (never-in-mitosis gene A)-related kinase 1 (Nek1) is shown to impact on different cellular pathways such as DNA repair, checkpoint activation, and apoptosis. Its role as a molecular target for radiation sensitization of malignant cells, however, remains elusive. Stably transduced doxycycline (Dox)-inducible Nek1 shRNA HeLa cervix and siRNA-transfected HCT-15 colorectal carcinoma cells were irradiated in vitro and 3D clonogenic radiation survival, residual DNA damage, cell cycle distribution, and apoptosis were analyzed. Nek1 knockdown (KD) sensitized both cell lines to ionizing radiation following a single dose irradiation and more pronounced in combination with a 6 h fractionation (3 × 2 Gy) regime. For preclinical analyses we focused on cervical cancer. Nek1 shRNA HeLa cells were grafted into NOD/SCID/IL-2Rγc−/− (NSG) mice and Nek1 KD was induced by Dox-infused drinking water resulting in a significant cytostatic effect if combined with a 6 h fractionation (3 × 2 Gy) regime. In addition, we correlated Nek1 expression in biopsies of patients with cervical cancer with histopathological parameters and clinical follow-up. Our results indicate that elevated levels of Nek1 were associated with an increased rate of local or distant failure, as well as with impaired cancer-specific and overall survival in univariate analyses and for most endpoints in multivariable analyses. Finally, findings from The Cancer Genome Atlas (TCGA) validation cohort confirmed a significant association of high Nek1 expression with a reduced disease-free survival. In conclusion, we consider Nek1 to represent a novel biomarker and potential therapeutic target for drug development in the context of optimized fractionation intervals.
The title compound, [Fe2(C5H5)2(C24H22BP2)(CO)4][FeCl4]·CHCl3, is an oxidation product of CpFe(CO)2PPh2BH3. One pair of phenyl rings attached to the two different P atoms are almost parallel, as are the other pair [dihedral angles = 8.7 (5) and 8.9 (5)°]. The planes of the two cyclopentadienyl rings are inclined by 26.8 (7)° with respect to each other. The carbonyl groups at each Fe atom are almost perpendicular [C-Fe-C = 92.6 (6) and 94.3 (5)°]. The geometric parameters of the molecule of the title compound, C14H16O2P2, are in the usual ranges. It is a meso compound with the two chiral P atoms having opposite configurations. The P-CH2-CH2-P chain adopts a trans conformation [torsion angle -178.59 (17)°]. The P=O bonds are almost coplanar with the adjacent phenyl ring [torsion angles = 3.8 (3) and 0.3 (3)°]. Whereas one of them is synclinal [torsion angle = -59.0 (2)°] to the central C-C bond, the other is anticlinal [torsion angle = 56.6 (2)°] to the central C-C bond. The dihedral angle between the two phenyl rings is 5.2 (3)°. The molecules are linked by weak C-H...O hydrogen bonds. They crystallize in rows running along the c axis. The title compound, [Li4O4(C12H8BO)4(C4H10O)4], features a Li4O4 cube. Each Li atom in the cube is additionally coordinated by a diethyl ether molecule and each O atom in the cube carries a 9-oxa-10-boraanthracene residue. The crystal studied was a non-merohedral twin [twin law (-1 0 0 / 0 0 1 / 0 1 0); the contribution of the major twin component refined to 0.553 (3)] emulating apparent tetragonal symmetry, whereas the actual crystal system is just orthorhombic.
Mol-ecules of the title compound, [Zn(8)(C(6)F(5))(8)O(4)(C(4)H(10)O)(4)], are located on a special position of site symmetry [Formula: see text]. As a result, there is just one quarter-mol-ecule in the asymmetric unit. The title compound features a Zn(4)O(4) cube. Each Zn atom in the cube carries a pentafluorophenyl substituent. Each O atom is bonded to a further Zn atom, which is connected to a pentafluorophenyl substituent and the O atom of a diethyl ether mol-ecule. All ether C atoms are disordered over two sets of sites with a site occupation factor of 0.51 (2) for the major occupied site.
Brain metastases are the most common intracranial tumor in adults and are associated with poor patient prognosis and median survival of only a few months. Treatment options for brain metastasis patients remain limited and largely depend on surgical resection, radio- and/or chemotherapy. The development and pre-clinical testing of novel therapeutic strategies require reliable experimental models and diagnostic tools that closely mimic technologies that are used in the clinic and reflect histopathological and biochemical changes that distinguish tumor progression from therapeutic response. In this study, we sought to test the applicability of magnetic resonance (MR) spectroscopy in combination with MR imaging to closely monitor therapeutic efficacy in a breast-to-brain metastasis model. Given the importance of radiotherapy as the standard of care for the majority of brain metastases patients, we chose to monitor the post-irradiation response by magnetic resonance spectroscopy (MRS) in combination with MR imaging (MRI) using a 7 Tesla small animal scanner. Radiation was applied as whole brain radiotherapy (WBRT) using the image-guided Small Animal Radiation Research Platform (SARRP). Here we describe alterations in different metabolites, including creatine and N-acetylaspartate, that are characteristic for brain metastases progression and lactate, which indicates hypoxia, while choline levels remained stable. Radiotherapy resulted in normalization of metabolite levels indicating tumor stasis or regression in response to treatment. Our data indicate that the use of MR spectroscopy in addition to MRI represents a valuable tool to closely monitor not only volumetrical but also metabolic changes during tumor progression and to evaluate therapeutic efficacy of intervention strategies. Adapting the analytical technology in brain metastasis models to those used in clinical settings will increase the translational significance of experimental evaluation and thus contribute to the advancement of pre-clinical assessment of novel therapeutic strategies to improve treatment options for brain metastases patients.

Background: MicroRNA-21 (miR-21) is up-regulated in tumor tissue of patients with malignant diseases, including hepatocellular carcinoma (HCC). Elevated concentrations of miR-21 have also been found in sera or plasma from patients with malignancies, rendering it an interesting candidate as serum/plasma marker for malignancies. Here we correlated serum miR-21 levels with clinical parameters in patients with different stages of chronic hepatitis C virus infection (CHC) and CHC-associated HCC.
Methodology/Principal Findings: 62 CHC patients, 29 patients with CHC and HCC and 19 healthy controls were prospectively enrolled. RNA was extracted from the sera and miR-21 as well as miR-16 levels were analyzed by quantitative real-time PCR; miR-21 levels (normalized by miR-16) were correlated with standard liver parameters, histological grading and staging of CHC. The data show that serum levels of miR-21 were elevated in patients with CHC compared to healthy controls (P<0.001); there was no difference between serum miR-21 in patients with CHC and CHC-associated HCC. Serum miR-21 levels correlated with histological activity index (HAI) in the liver (r = −0.494, P = 0.00002), alanine aminotransferase (ALT) (r = −0.309, P = 0.007), aspartate aminotransferase (r = −0.495, P = 0.000007), bilirubin (r = −0.362, P = 0.002), international normalized ratio (r = −0.338, P = 0.034) and γ-glutamyltransferase (r = −0.244, P = 0.034). Multivariate analysis revealed that ALT and miR-21 serum levels were independently associated with HAI. At a cut-off dCT of 1.96, miR-21 discriminated between minimal and mild-severe necroinflammation (AUC = 0.758) with a sensitivity of 53.3% and a specificity of 95.2%.
Conclusions/Significance: The serum miR-21 level is a marker for necroinflammatory activity, but does not differ between patients with HCV and HCV-induced HCC.
Artenreiche montane Rasengesellschaften auf Lawinenbahnen des Nationalparks Gesäuse (Österreich)

Das Ziel dieser Untersuchung war es, Erkenntnisse über die Bedeutung von Lawinenbahnen für die Biodiversität zu gewinnen. Dazu wurden im Nationalpark Gesäuse (Steiermark, Österreich) 16 Pflanzenbestände in drei ausgewählten Lawinenbahnen pflanzensoziologisch untersucht, vegetationsökologisch charakterisiert sowie mit Vegetationstabelle, bodenkundlichen Analysedaten und topographischen Parametern dokumentiert. Außerdem erfolgte eine Bestandsaufnahme der Schmetterlinge. Die Pflanzenartenvielfalt wurde mit verschiedenen Diversitäts-Indizes beurteilt. Die Ursachen für den Artenreichtum in den untersuchten Lawinenbahnen werden diskutiert.
Die Pflanzenbestände wurden überwiegend zum Origano-Calamagrostietum variae gestellt; eine Einzelaufnahme wurde dem Seslerio-Caricetum sem pervirentis zugeordnet. Im Ersteren dominieren Arten mit hemikryptophytischer Lebensform; Therophyten erreichen die zweithöchste Deckung. Die Pflanzengesellschaft wird von CSR-, CS- und C-Strategen aufgebaut; alle anderen Lebensstrategietypen haben keine Bedeutung. Das Origano-Calamagrostietum variae ist eine artenreiche, natürliche Dauergesellschaft, in der mäßiger Stress und schwache Störungen die Artenzusammensetzung maßgeblich bestimmen. Aktive Lawinenbahnen auf sehr flachgründigen, steinigen, nährstoffarmen, basenreichen Rendzinen in lokalklimatisch wärmebegünstigten steilen Hanglagen der montanen Höhenstufe zählen zu den arten-, blüten- und aspektreichsten und somit ökologisch wertvollsten Vegetationsformationen im Nationalparkgebiet. In den kräuterreichen Pflanzenbeständen wurden im Durchschnitt 71 Gefäßpflanzenarten und 5 Moosarten pro 20 m2 Aufnahmefläche gefunden. Mit dieser hohen Phytodiversität und dem Blütenreichtum ist auch eine große Artenvielfalt bei den Schmetterlingen verbunden. Die untersuchten Lawinenbahnen weisen nicht nur ein hohes Maß an Naturnähe auf, sie dürften auch der ursprüngliche Lebensraum einiger Pflanzenarten des Wirtschafts- und Extensivgrünlandes der Tallagen sein. Auf Grund ihrer hohen naturschutzfachlichen Bedeutung müssen Lawinenbahnen als Ganzes geschützt werden. Periodische oder episodische Lawinenabgänge sind Voraussetzung für die Existenz dieser störungsgeprägten Ökosysteme. Unterhalb von Lawinenbahnen dürfen keine Gebäude oder Infrastrukturen errichtet werden, weil diese Bautätigkeit Schutzobjekte erzeugt. Dadurch würde ein Bedarf für Schutzmaßnahmen entstehen, vor allem in Form von Lawinenverbauungen und Aufforstungen, wodurch Lawinenabgänge verhindert werden.
The ability of hematopoietic stem cells (HSCs) to self-renew is a prerequisite for the establishment of definitive hematopoiesis and life-long blood regeneration. Here, we report the single-stranded DNA-binding transcriptional regulator far upstream element (FUSE)-binding protein 1 (FUBP1) as an essential factor of HSC self-renewal. Functional inactivation of FUBP1 in two different mouse models resulted in embryonic lethal anemia at around E15.5 caused by severely diminished HSCs. Fetal and adult HSCs lacking FUBP1 revealed an HSC-intrinsic defect in their maintenance, expansion, and long-term blood reconstitution, but could differentiate into all hematopoietic lineages. FUBP1-deficient adult HSCs exhibit significant transcriptional changes, including upregulation of the cell-cycle inhibitor p21 and the pro-apoptotic Noxa molecule. These changes caused an increase in generation time and death of HSCs as determined by video-microscopy-based tracking. Our data establish FUBP1 and its recognition of single-stranded genomic DNA as an important element in the transcriptional regulation of HSC self-renewal.
We measured the neutron capture cross sections of 69Ga and 71Ga for a quasi-stellar spectrum at kBT = 25 keV and a spectrum with a peak energy at 90 keV by the activation technique at the Joint Research Centre (JRC) in Geel, Belgium. Protons were provided by an electrostatic Van de Graaff accelerator to produce neutrons via the reaction 7Li(p,n). The produced activity was measured via the γ emission of the product nuclei by high-purity germanium detectors. We present preliminary results.
Within the last 30 years the role of nitrogen in Central European forests has changed fundamentally from limiting resource to environmental problem. As the retrospective tracking of nutrient availability by soil chemical and biogeochemical measurements faces serious problems, bioindication based on understorey species composition is indispensable for monitoring broad-scale eutrophication. Based on a broad survey of more than 100,000 forest vegetation plots accessible in electronic data-bases from Germany and adjacent countries, we calculated unweighted average Ellenberg nutrient values (mN) as a proxy of plant-available macronutrients. Based on the quantiles of the frequency distribution of mN in a regionally stratified sample, we define five trophic classes, which can be used to compare dimensionless mN values. We studied spatial patterns of average nutrient values within 17 regions and compared the periods from 1899 to 1975 and 1976 to 2006. After 1975 eutrophic (mN > 5.67) and hypertrophic (mN > 6.28) conditions were common everywhere except in the Alps and Saxony-Anhalt, but very oligotrophic conditions (mN < 3.44) were still widespread in regions with nutrient-poor bedrock. Before 1975 mN of plots had been lower than after 1975 in all but the southeastern regions. Between the pre- and post-1975 data the proportion of hypertrophic plots increased from 5.7 to 11.8%, and that of very oligo-trophic plots decreased from 14.6 to 8.3%. To remove bias resulting from uneven distribution, the dataset was stratified by five tree layer dominance types, period and region and resampled. In pre-1975 plots medians of mN increased in the order Pinus sylvestris, Quercus spp., Picea abies, Fagus sylvatica and Alnus spp, whereas the increase of mN was highest in forest types with historically low nutrient values. Therefore, the widespread change in mN must be attributed to the pronounced vegetation changes in Quercus and Pinus stands, indicating the importance of land-use change, i.e. recovery of nutrient cycles after hundreds of years of exploitation through coppicing, grazing and litter use. The analysis confirms eutrophication as a megatrend of modern vegetation change and demonstrates the high research potential of linking vegetation plot databases across large regions.
The live attenuated yellow fever (YF) vaccine has an excellent record of efficacy and one dose provides long-lasting immunity, which in many cases may last a lifetime. Vaccination stimulates strong innate and adaptive immune responses, and neutralizing antibodies are considered to be the major effectors that correlate with protection from disease. Similar to other flaviviruses, such antibodies are primarily induced by the viral envelope protein E, which consists of three distinct domains (DI, II, and III) and is presented at the surface of mature flavivirions in an icosahedral arrangement. In general, the dominance and individual variation of antibodies to different domains of viral surface proteins and their impact on neutralizing activity are aspects of humoral immunity that are not well understood. To gain insight into these phenomena, we established a platform of immunoassays using recombinant proteins and protein domains that allowed us to dissect and quantify fine specificities of the polyclonal antibody response after YF vaccination in a panel of 51 vaccinees as well as determine their contribution to virus neutralization by serum depletion analyses. Our data revealed a high degree of individual variation in antibody specificities present in post-vaccination sera and differences in the contribution of different antibody subsets to virus neutralization. Irrespective of individual variation, a substantial proportion of neutralizing activity appeared to be due to antibodies directed to complex quaternary epitopes displayed on the virion surface only but not on monomeric E. On the other hand, DIII-specific antibodies (presumed to have the highest neutralizing activity) as well as broadly flavivirus cross-reactive antibodies were absent or present at very low titers. These data provide new information on the fine specificity as well as variability of antibody responses after YF vaccination that are consistent with a strong influence of individual-specific factors on immunodominance in humoral immune responses.
Author Summary: The live-attenuated yellow fever vaccine has been administered to more than 600 million people worldwide and is considered to be one of the most successful viral vaccines ever produced. Following injection, the apathogenic vaccine virus replicates in the vaccinee and induces antibodies that mediate virus neutralization and subsequent protection from disease. In principle, many different antibodies are induced by viral antigens, but it is becoming increasingly clear that only a subset of them is capable of inactivating the virus, and some antibody populations appear to dominate the immune response. However, to date there has been very little information on individual-specific variations of immunodominance and how such variations can affect the functionality of antibody responses. In our study, we addressed these issues and analyzed the fine specificities of antibodies induced by YF vaccination as well as the contribution of different antibody subsets to virus neutralization in 51 vaccinees. We demonstrate an extensive degree of individual variation with respect to immunodominance of antibody populations and their contribution to virus neutralization. Such variations can have an impact on vaccine-mediated protection, and thus insight into this phenomenon can provide leads for novel strategies in modern vaccine design.
Ventriculoperitoneal shunts equipped with a reservoir and a valve to manually switch off the shunt function can be used for intraventricular injections of therapeutics in patients suffering from a communicating hydrocephalus caused by leptomeningeal metastases. These shunt devices avoid the risk of injecting therapeutics through the distal leg of the shunt system into the intraperitoneal space, which may cause toxicity. Furthermore, regular intraventricular injections of chemotherapeutics help to maintain sufficient concentrations in the ventricular space. Therefore, ventriculoperitoneal shunts equipped with an on-off valve are a useful tool to reliably inject chemotherapeutics into the ventricles. In order to systematically assess feasibility, safety, and efficacy of this procedure, we performed a retrospective analysis of all patients with leptomeningeal metastases who had received a shunt system at our institution. In total, six adult patients had a ventriculoperitoneal shunt equipped with an on-off valve implanted. Out of these six patients, two patients subsequently received intraventricular injections of chemotherapeutics. The configuration of the valve setting and the intraventricular injections were easily feasible in the setting of a neuro-oncology department. The complication of a shunt leakage occurred in one patient following the first intraventricular injection. No extra-central nervous system (CNS) toxicities were observed. In summary, ventriculoperitoneal shunts with on-off valves are useful tools for reliable intraventricular administration of therapeutics.
In patients with glioblastoma, antiangiogenic therapy with bevacizumab (BEV) has been shown to improve progression-free survival (PFS), but not overall survival (OS). Especially in patients with an unusual infiltrative phenotype as seen in multifocal glioblastoma, the use of BEV therapy is still more controversial. Therefore, we prepared a retrospective case series with 16 patients suffering from a multifocal glioblastoma treated with BEV. We compared these patients to a matched control cohort of 16 patients suffering from glioblastoma with a single lesion treated with BEV. The objective of this study was to evaluate whether the course of disease differs in glioblastoma patients with a multifocal disease pattern compared to those with a single lesion only. Patients were treated with BEV monotherapy or BEV in combination with irinotecan or lomustine (CCNU). Response rates and PFS were similar in both groups. There was a trend for an unfavorable OS in the patient group with multifocal glioblastoma, which was expected due to the generally worse prognosis of multifocal glioblastoma. We investigated whether BEV therapy affects the invasive growth pattern as measured by the appearance of new lesions on magnetic resonance imaging (MRI). Under BEV therapy, there was a trend for a lower frequency of new lesions both in multifocal and solitary glioblastoma. Based on these results, BEV therapy at relapse appears to be justified to no lesser extent in multifocal glioblastoma than in solitary glioblastoma.
