The genetic make-up of an individual contributes to the susceptibility and response to viral infection. Although environmental, clinical and social factors have a role in the chance of exposure to SARS-CoV-2 and the severity of COVID-191,2, host genetics may also be important. Identifying host-specific genetic factors may reveal biological mechanisms of therapeutic relevance and clarify causal relationships of modifiable environmental risk factors for SARS-CoV-2 infection and outcomes. We formed a global network of researchers to investigate the role of human genetics in SARS-CoV-2 infection and COVID-19 severity. Here we describe the results of three genome-wide association meta-analyses that consist of up to 49,562 patients with COVID-19 from 46 studies across 19 countries. We report 13 genome-wide significant loci that are associated with SARS-CoV-2 infection or severe manifestations of COVID-19. Several of these loci correspond to previously documented associations to lung or autoimmune and inflammatory diseases3,4,5,6,7. They also represent potentially actionable mechanisms in response to infection. Mendelian randomization analyses support a causal role for smoking and body-mass index for severe COVID-19 although not for type II diabetes. The identification of novel host genetic factors associated with COVID-19 was made possible by the community of human genetics researchers coming together to prioritize the sharing of data, results, resources and analytical frameworks. This working model of international collaboration underscores what is possible for future genetic discoveries in emerging pandemics, or indeed for any complex human disease.

Das Instrument der Roadmap schafft in einem ersten Schritt Transparenz darüber, welche Angebote und Ansprechpartner*innen zur Unterstützung von Personalverantwortlichen schon vorhanden sind und wo noch Handlungsbedarfe bestehen. Daran kann vor Ort angeknüpft und weitere Angebote und Ansprechpartner*innen ergänzt werden.

In Teil I des vorliegenden Dossiers wird zunächst ein Überblick über die vorliegende Literatur zur Gruppe der geringqualifizierten Beschäftigten, ihrer Beteiligung an Weiterbildung und Motivation zur Teilnahme daran präsentiert. Hierbei wird nach hemmenden und förderlichen Faktoren unterschieden. Im daran anschließenden Teil II werden die Perspektive Beschäftigter in der Nachqualifizierung und die empirisch untersuchten förderlichen Faktoren beleuchtet. Dazu wird zunächst das Studiendesign, das methodische Vorgehen und das Sample beschrieben und anschließend die aus der Studie abzuleitenden Ergebnisse präsentiert. Zusammengeführt werden die Ergebnisse in der Typisierung an- und ungelernter Beschäftigter in der Nachqualifizierung. Daraufhin werden in Teil III anhand der einzelnen Typen von Beschäftigten Empfehlungen für die Beratung abgeleitet und erläutert. Auf diese Weise soll den Beratungskräften der Initiative ProAbschluss, die sowohl Unternehmen als auch Beschäftigte direkt ansprechen können, eine Unterstützung geboten werden, um Beratung fein auszutarieren und noch zielgruppengerechter zu gestalten

Kapitel 1 des Dossiers beschreibt zunächst das Zulassungsverfahren für Externenprüfungen, erläutert im Rahmen der Datenauswertung verwendete Begrifflichkeiten und stellt das methodische Vorgehen vor. In Kapitel 2 wird die Bedeutung der Nachqualifizierung in Hessen insgesamt beleuchtet. Hier werden die Teilnahmen an Externenprüfungen im bundesdeutschen wie auch im Vergleich der Regionen des Bundeslandes dargestellt und Entwicklungen der vergangenen Jahre nachgezeichnet. Im Anschluss werden die Personen, die an einer Externenprüfung teilgenommen haben, anhand soziodemografischer Daten (Alter, Geschlecht, schulische Vorbildung) näher beschrieben und mit den Bezieherinnen und Beziehern des hessischen Qualifizierungsschecks verglichen. Das Kapitel endet mit einer Darstellung der Berufe, in denen in Hessen besonders viele Externenprüfungen absolviert wurden. Auch hier wird ein Vergleich mit den ProAbschluss‐Statistiken vorgenommen. Kapitel 3 beinhaltet regionalisierte Analysen für die zehn IHK‐Kammerbezirke in Hessen: Im ersten Teil wird das Ausmaß, in dem Berufsabschlüsse per Nachqualifizierung erlangt werden, für die Berufe mit den meisten Externenprüfungen verglichen. Im zweiten Teil folgt eine detaillierte Betrachtung der Situation innerhalb der einzelnen IHK‐Kammerbezirke, wobei verdeutlicht wird, in welchen Berufen vergleichsweise wenige Externenprüfungen absolviert werden. Das Dossier schließt mit einem Fazit in Kapitel 4.
GATA2 deficiency is a heterogeneous multi-system disorder characterized by a high risk of developing myelodysplastic syndrome (MDS) and myeloid leukemia. We analyzed the outcome of 65 patients reported to the registry of the European Working Group (EWOG) of MDS in childhood carrying a germline GATA2 mutation (GATA2mut) who had undergone hematopoietic stem cell transplantation (HSCT). At 5 years the probability of overall survival and disease-free survival (DFS) was 75% and 70%, respectively. Non-relapse mortality and relapse equally contributed to treatment failure. There was no evidence of increased incidence of graft-versus-host-disease or excessive rates of infections or organ toxicities. Advanced disease and monosomy 7 (−7) were associated with worse outcome. Patients with refractory cytopenia of childhood (RCC) and normal karyotype showed an excellent outcome (DFS 90%) compared to RCC and −7 (DFS 67%). Comparing outcome of GATA2mut with GATA2wt patients, there was no difference in DFS in patients with RCC and normal karyotype. The same was true for patients with −7 across morphological subtypes. We demonstrate that HSCT outcome is independent of GATA2 germline mutations in pediatric MDS suggesting the application of standard MDS algorithms and protocols. Our data support considering HSCT early in the course of GATA2 deficiency in young individuals.
NOSTRIN belongs to the family of F-BAR proteins, which are multi-domain adaptor proteins that have emerged as important regulators of membrane remodeling and actin dynamics in a variety of vital cellular processes. They have been analyzed structurally and biochemically and overexpression studies have revealed their potential in inducing membrane curvature and tubulation. Several studies have begun to decipher the function of individual proteins, but the understanding of F-BAR protein functions in vivo is still quite limited. The F-BAR protein NOSTRIN is mainly expressed in endothelial cells and has originally been described as interaction partner of the endothelial nitric oxide synthase (eNOS), modulating eNOS subcellular localization. The phenotypic characterization of NOSTRIN knockout mice revealed decreased nitric oxide (NO) and cGMP levels, an increase in systolic blood pressure and an impairment of the acetylcholine-induced, NO-dependent relaxation of aortic rings from mice with global as well as endothelial cell-specific knockout of the NOSTRIN gene (ECKO) . These findings implied that NOSTRIN plays a role in regulating NO production in vivo, but the underlying molecular mechanisms were unclear. Therefore, this study was aimed at addressing the mechanism causing the inhibited vasodilation specifically upon stimulation with acetylcholine in NOSTRIN KO and ECKO mice, and at exploring additional roles of NOSTRIN in the signal transduction of endothelial cells.
The major acetylcholine receptor that mediates vessel relaxation upon stimulation with acetylcholine is the muscarinic acetylcholine receptor subtype M3 (M3R). In the present study NOSTRIN was identified as novel interaction partner of the M3R and important factor for the correct spatial distribution and functionality of the M3R. Moreover, it provides the first example of an F-BAR protein regulating a GPCR. Confocal immunofluorescence microscopy analysis of isolated aortae from NOSTRIN KO and WT mice indicated that NOSTRIN was necessary for the proper subcellular localization of the M3R and targeted it to the plasma membrane. A series of pulldown experiments revealed a direct interaction of NOSTRIN with the M3R. The binding required the SH3 domain of NOSTRIN and the third intracellular loop of the M3R, which has a recognized role in receptor regulation. The interaction of NOSTRIN with the M3R was confirmed by co-localization of NOSTRIN and the M3R upon overexpression in mammalian cells. Expression levels of the M3R as well as eNOS were not affected by the loss of NOSTRIN in accordance with the finding, that NOSTRIN impacts on the acetylcholine/eNOS signaling axis through regulation of the subcellular trafficking of its binding partners.
Furthermore, there were first indications for a role of NOSTRIN in facilitating the carbachol-induced calcium response in M3R-expressing cells, suggesting that NOSTRIN might influence M3R activation. in the absence of NOSTRIN, the function of the M3R in mammalian cells overexpressing the M3R was markedly impaired, resulting in abolition of the calcium response to the M3R agonist carbachol. In accordance, the activated eNOS fraction associated with the Golgi complex was markedly reduced in aorta explants from NOSTRIN knockout and ECKO mice. Moreover, NOSTRIN knockout inhibited the carbachol-induced, activating phosphorylation of eNOS in murine aortae as well as primary mouse lung endothelial cells confirming its role as important regulator of eNOS activity in vivo.
This paper reports on Monte Carlo simulation results for future measurements of the moduli of time-like proton electromagnetic form factors, |GE | and |GM|, using the ¯pp → μ+μ− reaction at PANDA (FAIR). The electromagnetic form factors are fundamental quantities parameterizing the electric and magnetic structure of hadrons. This work estimates the statistical and total accuracy with which the form factors can be measured at PANDA, using an analysis of simulated data within the PandaRoot software framework. The most crucial background channel is ¯pp → π+π−,due to the very similar behavior of muons and pions in the detector. The suppression factors are evaluated for this and all other relevant background channels at different values of antiproton beam momentum. The signal/background separation is based on a multivariate analysis, using the Boosted Decision Trees method. An expected background subtraction is included in this study, based on realistic angular distribuations of the background contribution. Systematic uncertainties are considered and the relative total uncertainties of the form factor measurements are presented.
