Die vorliegende Arbeit beschäftigt sich mit der Emission von Elektronen aus kleinen dissoziierenden Molekülen.
Die Frage, der hier nachgegangen werden soll: Wie läuft ein solcher Prozess, bei dem ein Molekül in seine atomaren Einzelteile zerbricht, tatsächlich ab? Während es Experimentalphysikern schon seit längerem möglich ist präzise Aussagen über den Zustand eines Systems vor und nach einer solchen „halben“ chemischen Reaktion zu machen, war es lange nicht möglich die Reaktion selbst zu be-obachten, da sie auf einer Zeitskala von einigen Femtosekunden (1 fs = 10-15 s) stattfindet. Eine Möglichkeit, solche Prozesse zu untersuchen, ist die Ionisation, also das Herauslösen eines Elektrons aus seinem gebunden Zustand im Molekül, und die anschließende Messung der kinetische Energie oder des Impulsvektors des Elektrons. Dadurch können Rückschlüsse auf die Bindungsenergie und die räumliche Verteilung der Elektronen im gebundenen Zustand gezogen werden. Wenn man in der Lage ist die Elektronen, die von einem dissoziierenden Molekül zu verschiedenen Zeitpunkten während des Dissoziationsprozesses emittiert werden, zu messen, so sollte es unter Umständen möglich sein, den Übergang von molekularen zu atomaren Orbitalen zu beobachten.
Zur Durchführung der Messungen wurde ein COLTRIMS-Multikoinzidenzimpulsspektrometer ver-wendet, mit welchem sowohl die kinetische Energie aller geladenen Reaktionsprodukte als auch deren vollständige Impulsvektoren koinzident gemessen werden können.
In einer Messung an Chlorwasserstoff wurde auf diese Weise die ultraschnelle Dissoziation angeregter neutraler Moleküle untersucht. Hierbei machte man sich den Umstand zunutze, dass die angeregten Zustände bei beliebigen internuklearen Abständen zerfallen und ein Auger-Elektron emittieren können.
Für den resonanten Auger-Zerfall der 2p-16σ-Zustände des Chlorwasserstoffmoleküls wurden unseres Wissens nach erstmals alle Komponenten der Impulsvektoren sowohl der Auger-Elektronen als auch der ionischen Reaktionsprodukte gemessen. Durch diese kinematisch vollständige Messung konnte der Prozess in bisher noch nie dagewesenem Detail untersucht werden. Zum ersten Mal konnte sowohl der angeregte Zustand nach der Absorption des Photons, als auch der elektronische Endzustand für jeden einzelnen Zerfall bestimmt werden. Aufgeschlüsselt nach diesen Zuständen konnten dann die Winkelverteilungen der Auger-Elektronen und die Aufteilung der Energie auf Elektron und Kernbewegung vermessen werden. Dies ist der vollständige Satz aller möglichen Beobachtungsgrößen, so dass die Daten über keine Größe mehr integriert wurden.
In einer Messung an H2O-Molekülen wurde ein Prozess untersucht, bei dem ein einfach geladenes angeregtes Molekül in zwei Fragmente dissoziiert und, wenn die Dissoziation bereits sehr weit fort-geschritten ist, ein weiteres Elektron emittiert. Hier konnte gezeigt werden, wie eine Anisotropie in der Elektronenwinkelverteilung eines dissoziierenden Moleküls mit Hilfe einer einfachen klassischen Simulation dazu verwendet werden kann, den Abstand zwischen einem Proton und einem angeregten Molekül, in welche das ursprüngliche Molekül dissoziiert, zu dem Zeitpunkt an dem das angeregte Molekül durch Autoionisation ein weiteres Elektron emittiert, zu bestimmen. Es wurde nachgewiesen, dass der Auger-Zerfall eines H2O+*-Ions, bei dem ein Sauerstoff 2s-Elektron aus dem Molekülverband entfernt wurde, erst bei sehr großen Abständen von mehreren 100 Ångström zwischen dem Proton und dem OH*-Molekül stattfindet.
Die dritte Messung an dem Ne2-Dimer beschäftigt sich mit der Frage, ob die in einem Molekül er-zeugten Vakanzen als lokalisiert oder delokalisiert zu betrachten sind. Wie bereits in vorhergegangenen Messungen dargelegt wurde, ist die Antwort auf die Frage für kovalente Moleküle eine Frage des Messprozesses. In Rahmen dieser Arbeit konnte nachgewiesen werden, dass dies auch für schwach gebundene Van-der Waals-Moleküle der Fall ist und deren Valenzelektronen ebenfalls unter bestimmten Umständen als delokalisiert angesehen werden müssen. Das ist insoweit überraschend, da die gängige Vorstellung eines solchen Moleküls die eines Systems aus einzelnen Atomen ist, welche nur durch kleine Ladungspolarisationen in den Elektronenschalen eine Bindung eingehen. Des Weiteren wurde gezeigt, dass sich der Prozess qualitativ mit einer einfachen Doppelspalt-Simulation erklären lässt, bei welcher ebene Wellen mit einem bestimmten Phasenversatz, der sich aus der Form der beteiligten Atomorbitale ergibt, von den beiden Kernen emittiert werden.
Die Dissoziation des Moleküls war selber nicht Gegenstand der Untersuchung, sondern wurde ausgenutzt um verschiedene elektronische Zustände, in welchen sich das Molekül nach der Ionisation befinden kann und von denen nur einer dissoziativ ist, zu unterscheiden.
Perioperative Blutungskomplikationen stellen, insbesondere für neurochirurgisch behandelte Patienten, eine schwerwiegende Komplikation dar und sind mit einer signifikant erhöhten Morbidität und Mortalität assoziiert [1]. Der perioperative Blutverlust ist einer der Hauptgründe für die Transfusion allogener Blutprodukte [2, 3]. Transfusionen von Erythrozytenkonzentraten (EK), Gefrorenem Frischplasma (GFP) und Thrombozytenkonzentraten (TK) sind dabei unabhängige Risikofaktoren für die Entwicklung thromboembolischer, pulmonaler, hämodynamischer oder septischer Komplikationen und selbst mit einer erhöhten perioperativen Morbidität und Mortalität assoziiert [3-10]. In der Literatur wird die Prävalenz von Blutungskomplikationen im Rahmen von intrakraniellen Eingriffen (Raten zwischen 0,8% bis 50%) sehr unterschiedlich beschrieben [1, 11-13]. Gleichwertige Voraussetzungen für die Bildung und Aufrechterhaltung eines stabilen Blutgerinnsels sind, neben einer effizienten primären Hämostase (Thrombozytenzahl und –funktion, Von-Willebrand-Faktor), auch die plasmatische Gerinnung (extrinsische und intrinsische Gerinnungsfaktoren) und der Schutz vor (Hyper)fibrinolyse [14].
Effekte eines zweijährigen Krafttrainings auf die Sprintleistung im Nachwuchsleistungssport Fußball

In der internationalen Forschung von Effekten durch ein Krafttraining auf sportrelevante Leistungsparameter im Nachwuchsleistungssport Fußball gibt es keine langfristigen Untersuchungen mit einer Interventionsdauer von mehr als einem Jahr. Generell finden sich kaum bis keine Untersuchungen mit experimentellem Charakter über diesen Zeitraum. Demnach kommt es in der Literatur oft zu reinen Spekulationen über mögliche langfristige Anpassungen aufgrund der ermittelten Ergebnisse über kurze Interventionszeiträume. Dies führt dazu, dass eine Aufklärung bezüglich langfristiger Anpassungen von Trainingsmaßnahmen durch diese Art von Untersuchungen nicht gegeben werden kann. Gerade vor dem Hintergrund koordinativ anspruchsvoller Leistungsparameter ist der Spekulationsfaktor um ein vielfaches höher, da die Anzahl leistungslimitierender Faktoren 1 Einleitung und Problemstellung höher ist. Weiterhin sind Längsschnittuntersuchungen im leistungsorientierten Sport kaum vorhanden, da in der Praxis oft eine gewisse Skepsis vorherrscht, wenn sich Trainingsinterventionen möglicherweise negativ auswirken bzw. ohne Auswirkungen bleiben können. Diese Problematik zeigt sich nicht nur bei Profifußballspielern in hohem Maße, sondern auch im Nachwuchsbereich leistungsorientierter Fußballspieler. Zusätzlich kommt im Nachwuchsbereich neben der erwähnten Problematik noch der reduzierte Trainingsumfang für das Fußballtraining hinzu, der aus Zeitproblemen wegen der Schulpflicht resultiert. Ziel dieser Arbeit ist es daher aufzuzeigen, inwiefern sich ein langfristig periodisiertes Krafttraining über ein bzw. zwei Jahre im leistungsorientierten Nachwuchsfußball auf die im Fußball leistungsdeterminierenden Schnelligkeitsparameter, dem Linearsprint über 30m und dem Richtungswechselsprint, auswirkt. Daraus sollen sich Empfehlungen für die Praxis ableiten, um die Trainingsqualität im leistungssportlich orientierten Fußball zu steigern. Dazu werden Nachwuchsfußballspieler zweier anerkannter Nachwuchsleistungszentren des Deutschen Fußballbunds (DFB) über zwei Jahre begleitet. Aus den Daten soll ersichtlich werden, inwiefern sich Maximalkraft- und Sprintleistungen bei Fußballspielern, die ein Krafttraining durchführen, im Vergleich zu denjenigen, die nur das reguläre Fußballtraining absolvieren, entwickeln. Diese Betrachtung wird über die im Fußball gängigen Altersklassen A-, B- und C-Junioren vorgenommen. Weiterhin soll sich zeigen, ob mögliche Unterschiede der Leistungsentwicklung zwischen den Altersklassen zu erwarten sind.
Im Zuge des Ukrainekonflikts hat der Westen Sanktionen gegen Russland verhängt. Doch angesichts der ausbleibenden Verhaltensveränderung, wird der Nutzen der Sanktionen in Frage gestellt. Wenngleich sinkende Öleinnahmen eher Wirkung zeigen könnten, dürfen wir nicht vergessen, dass Sanktionen auch eine symbolische Wirkung zur Einhaltung geltenden Rechts aufweisen. Deshalb sollte der Westen an ihnen festhalten.
The aim of the present overview article is to raise awareness of an essential aspect that is usually not accounted for in the modelling of electron transport for focused-electron-beam-induced deposition (FEBID) of nanostructures: Surface excitations are on the one hand responsible for a sizeable fraction of the intensity in reflection-electron-energy-loss spectra for primary electron energies of up to a few kiloelectronvolts and, on the other hand, they play a key role in the emission of secondary electrons from solids, regardless of the primary energy. In this overview work we present a general perspective of recent works on the subject of surface excitations and on low-energy electron transport, highlighting the most relevant aspects for the modelling of electron transport in FEBID simulations.
Often adopting a feminist perspective, the sociological literature on migrant domestic services (MDS) does not make explicit which feminist paradigm it speaks from. This article situates this literature within ongoing debates in feminist theory, in particular the tension between materialist and poststructuralist approaches. Then, it discusses the empirical relevance of each of those two paradigms on the example of the results of original research into the personalization of employment relationships in MDS.
The contribution proposes a new way of making sense of the diversity of feminist theories, distinguishing between modern and postmodern approaches. Indeed, since the 1980s, feminist theory in the US and Western Europe has undergone a ‘postmodern turn’, which renders previous typologies much less up-to-speed with recent developments in the field. Then, the article examines which paradigms are implicit in the sociological literature on MDS. Initially, personalization in MDS was mainly seen in materialist terms, as a way to maximize the quantity and quality of labour (including emotional labour) extracted from domestic workers. The emergence of postmodern approaches in feminist theory set off a progressive shift in MDS literature. First, this literature showed that personalization also fulfils identity functions for employers and
workers, then it widened its focus to include the affective dimensions of domestic labour (not to be confused with emotional labour). The final section shows how modern and postmodern feminist approaches can be combined within a single research, on the example of original research on personalization in MDS in Belgium and Poland. In particular, the contribution shows that the distinction between material functions of personalization on the one hand, and its emotional/identity functions on the other is not empirically operative. Indeed, migrant domestic workers generally use emotional/identity categories to frame material questions, and vice versa. This final part shows that, rather than representing incompatible approaches, modern and postmodern feminisms complete each other, in this case showing a fuller image of personalization processes in MDS.
Diese Rechtskolumne stellt in Form eines Essays den staatsrechtlichen Diskurs und einige seiner Akteure zur Thematik des realen Wandels der Lebensverhältnisse von Ehe und Familie dar und befasst sich mit den Schwierigkeiten dessen normativer Verarbeitung. Der relevante Verfassungstext wurde nicht geändert, umstritten ist die Auslegung und inwieweit sie sich verändern darf. Das Bundesverfassungsgericht erklärte seit 2009 mehr-fach die Ungleichbehandlungen zwischen Ehe- und Lebenspartnern für verfassungswidrig. Die Vereinigung der deutschen Staatsrechtslehrer debattierte auf der Staatsrechtslehrertagung 2013 in Greifswald über Ehe und Familie, wobei es, insbesondere von den Männern, emotiona-le Stellungnahmen gegen die Auflösung der Ehe zu hören gab. Es dreht sich jedoch in diesem Diskurs über Ehe und Familie nicht nur um die Gleichstellung von Lesben und Schwulen. Immer wird auch das Geschlechterverhältnis zwischen Männern und Frauen mitverhandelt. Mit dem oft verteidigten traditionellen Familienbild ist die Ehe als patriarchale Institution gemeint. Bis heute wirkmächtig geblieben ist das spezifisch deutsche Mütterlichkeitsideal: Bleibt Mutti nicht zuhau-se, leidet das Kind. Unsere europäischen Nachbarn teilen diese Einstellung nicht. Das Recht muss akzeptieren und aufnehmen, dass Menschen heute in vielfältigen Familienformen (zu denen unter anderen auch die traditionelle Kleinfamilie gehört) leben.
Das Bundesverfassungsgericht sieht sich wegen seiner Entscheidungen, insbesondere zur europäischen Integration sowie zur rechtlichen Gleichstellung der Ehe mit der eingetragenen Lebenspartnerschaft, in jüngster Zeit mit zunehmender Kritik aus den Reihen der politischen Akteure konfrontiert. Die Rechtskolumne stellt diese Kritik in einen historischen Kontext und zeigt, dass inhaltliche Konflikte zwischen dem Gericht und der Politik ein wiederkehrendes Phänomen darstellen. Daran anschließend werden in der Diskussion stehende Maßnahmen zur Begrenzung des gerichtlichen Einflusses analysiert, die sich aber im Ergebnis als wenig erfolgversprechend bzw. aufgrund ihrer negativen strukturellen Auswirkungen als nicht tragfähig erweisen.
Ilse Staff ist in mehrfacher Hinsicht eine Pionierin. Sie ist die erste Frau, die am Fachbereich Rechtswissenschaft der Universität Frankfurt habilitiert wurde, zugleich die erste Professorin an diesem Fachbereich sowie die erste Frau überhaupt, die sich im öffentlichen Recht im deutschsprachigen Raum habilitierte.
Der Beitrag zeichnet zunächst bedeutende private und berufliche Stationen des Lebens von Ilse Staff nach und widmet sich anschließend ihrem juristischen Werk, in dem ihre politischen Wertmaßstäbe sowohl in der Wahl der Themen wie auch in den inhaltlichen Positionen zum Ausdruck kommen: Ilse Staff setzte sich als eine der ersten kritisch mit der Rolle der Justiz im Dritten Reich auseinander und leistete damit einen maßgeblichen Beitrag zur rechtshistorischen Aufarbeitung der NS-Zeit. Darüber hinaus ist ihr oeuvre geprägt von der rechtsphilosophisch-verfassungstheoretischen Beschäftigung mit den Positionen bedeutender Staatsrechtslehrer der Weimarer Republik sowie umfassenden (rechtsvergleichenden) Auseinandersetzungen mit dem italienischen Staatsrecht bzw. der italienischen Staatslehre.
Die Biographie der Staatsrechtslehrerin Ilse Staff ist beeindruckend, offenbart aber auch jene Hindernisse, mit denen sich Frauen auf dem Weg in die Wissenschaft – auch heute noch – konfrontiert sehen. Der Beitrag analysiert deshalb abschließend, inwiefern diese von der Gen-derforschung identifizierten strukturellen Widerstände Einfluss auf die wissenschaftliche Karrie-re von Ilse Staff gehabt haben könnten.
Unraveling the activation mechanism of taspase1 which controls the oncogenic AF4–MLL fusion protein

We have recently demonstrated that Taspase1-mediated cleavage of the AF4–MLL oncoprotein results in the formation of a stable multiprotein complex which forms the key event for the onset of acute proB leukemia in mice. Therefore, Taspase1 represents a conditional oncoprotein in the context of t(4;11) leukemia. In this report, we used site-directed mutagenesis to unravel the molecular events by which Taspase1 becomes sequentially activated. Monomeric pro-enzymes form dimers which are autocatalytically processed into the enzymatically active form of Taspase1 (αββα). The active enzyme cleaves only very few target proteins, e.g., MLL, MLL4 and TFIIA at their corresponding consensus cleavage sites (CSTasp1) as well as AF4–MLL in the case of leukemogenic translocation. This knowledge was translated into the design of a dominant-negative mutant of Taspase1 (dnTASP1). As expected, simultaneous expression of the leukemogenic AF4–MLL and dnTASP1 causes the disappearance of the leukemogenic oncoprotein, because the uncleaved AF4–MLL protein (328 kDa) is subject to proteasomal degradation, while the cleaved AF4–MLL forms a stable oncogenic multi-protein complex with a very long half-life. Moreover, coexpression of dnTASP1 with a BFP-CSTasp1-GFP FRET biosensor effectively inhibits cleavage. The impact of our findings on future drug development and potential treatment options for t(4;11) leukemia will be discussed.
As the lowest in the caste hierarchy, Dalits in Indian society have historically suffered caste-based social exclusion from economic, civil, cultural, and political rights. Women from this community suffer from not only discrimination based on their gender but also caste identity and consequent economic deprivation. Dalit women constituted about 16.60 percent of India’s female population in 2011. Dalit women’s problems encompass not only gender and economic deprivation but also discrimination associated with religion, caste, and untouchability, which in turn results in the denial of their social, economic, cultural, and political rights. They become vulnerable to sexual violence and exploitation due to their gender and caste. Dalit women also become victims of abhorrent social and religious practices such as devadasi/jogini (temple prostitution), resulting in sexual exploitation in the name of religion. The additional discrimination faced by Dalit women on account of their gender and caste is clearly reflected in the differential achievements in human development indicators for this group. In all the indicators of human development, for example, literacy and longevity, Dalit women score worse than Dalit men and non-Dalit women. Thus, the problems of Dalit women are distinct and unique in many ways, and they suffer from the ‘triple burden’ of gender bias, caste discrimination, and economic deprivation. To gain insights into the economic and social status of Dalit women, our paper will delve more closely into their lives and encapsulate the economic and social situations of Dalit women in India. The analyses of human poverty and caste and gender discrimination are based on official data sets as well as a number of primary studies in the labor market and on reproductive health.
Ein kritischer Diskurs ist essentiell für die Wissenschaft. Das ist zwar banal, wird aber im gegenwärtigen Streit um „Münkler Watch“, einem Blog, in dem Studierende der Humboldt-Universität Berlin eine Vorlesung des Politikwissenschaftlers Prof. Herfried Münkler anonym kritisieren, häufig vergessen. Aber auch den Studierenden scheint es nicht um einen inhaltlichen Dialog, sondern um Aufmerksamkeit zu gehen.
Mit dem machtpolitischen Aufstieg Chinas treten Weltregionen in den Fokus, die in unseren Breitengraden vormals eher am Rande wahrgenommen wurden – insbesondere Asien. Neben der gestiegenen weltpolitischen Bedeutung eröffnen sich auch akademisch spannende Perspektiven, wie unlängst die zweite Global South Caucus Konferenz der ISA in Singapur zeigte.

Denn Ai erhielt kein 6-Monatsvisum, weil er „straffällig“ sei. Doch der Künstler wurde niemals angeklagt. Was steckt hinter der Verweigerung? Ein Kommentar.
In many European countries poverty migration and its impact on the European continent are currently widely discussed topics. Many seem to forget about grave migration problems taking place in Asia, where in Hong Kong, for example, the working and living conditions for approximately 320,000 foreign domestic workers (mostly women) are often intolerable.
In most habitats, vegetation provides the main structure of the environment. This complexity can facilitate biodiversity and ecosystem services. Therefore, measures of vegetation structure can serve as indicators in ecosystem management. However, many structural measures are laborious and require expert knowledge. Here, we used consistent and convenient measures to assess vegetation structure over an exceptionally broad elevation gradient of 866–4550m above sea level at Mount Kilimanjaro, Tanzania. Additionally, we compared (human)-modified habitats, including maize fields, traditionally managed home gardens, grasslands, commercial coffee farms and logged and burned forests with natural habitats along this elevation gradient. We distinguished vertical and horizontal vegetation structure to account for habitat complexity and heterogeneity. Vertical vegetation structure (assessed as number, width and density of vegetation layers, maximum canopy height, leaf area index and vegetation cover) displayed a unimodal elevation pattern, peaking at intermediate elevations in montane forests, whereas horizontal structure (assessed as coefficient of variation of number, width and density of vegetation layers, maximum canopy height, leaf area index and vegetation cover) was lowest at intermediate altitudes. Overall, vertical structure was consistently lower in modified than in natural habitat types, whereas horizontal structure was inconsistently different in modified than in natural habitat types, depending on the specific structural measure and habitat type. Our study shows how vertical and horizontal vegetation structure can be assessed efficiently in various habitat types in tropical mountain regions, and we suggest to apply this as a tool for informing future biodiversity and ecosystem service studies.
The Tarim River basin, located in Xinjiang, NW China, is the largest endorheic river basin in China and one of the largest in all of Central Asia. Due to the extremely arid climate, with an annual precipitation of less than 100 mm, the water supply along the Aksu and Tarim rivers solely depends on river water. This is linked to anthropogenic activities (e.g., agriculture) and natural and semi-natural ecosystems as both compete for water. The ongoing increase in water consumption by agriculture and other human activities in this region has been enhancing the competition for water between human needs and nature. Against this background, 11 German and 6 Chinese universities and research institutes have formed the consortium SuMaRiO (Sustainable Management of River Oases along the Tarim River; http://www.sumario.de), which aims to create a holistic picture of the availability of water resources in the Tarim River basin and the impacts on anthropogenic activities and natural ecosystems caused by the water distribution within the Tarim River basin. On the basis of the results from field studies and modeling approaches as well as from suggestions by the relevant regional stakeholders, a decision support tool (DST) will be implemented that will then assist stakeholders in balancing the competition for water, acknowledging the major external effects of water allocation to agriculture and to natural ecosystems. This consortium was formed in 2011 and is funded by the German Federal Ministry of Education and Research. As the data collection phase was finished this year, the paper presented here brings together the results from the fields from the disciplines of climate modeling, cryology, hydrology, agricultural sciences, ecology, geoinformatics, and social sciences in order to present a comprehensive picture of the effects of different water availability schemes on anthropogenic activities and natural ecosystems along the Tarim River. The second objective is to present the project structure of the whole consortium, the current status of work (i.e., major new results and findings), explain the foundation of the decision support tool as a key product of this project, and conclude with application recommendations for the region. The discharge of the Aksu River, which is the major tributary of the Tarim, has been increasing over the past 6 decades. From 1989 to 2011, agricultural area more than doubled: cotton became the major crop and there was a shift from small-scale to large-scale intensive farming. The ongoing increase in irrigated agricultural land leads to the increased threat of salinization and soil degradation caused by increased evapotranspiration. Aside from agricultural land, the major natural and semi-natural ecosystems are riparian (Tugai) forests, shrub vegetation, reed beds, and other grassland, as well as urban and peri-urban vegetation. Within the SuMaRiO cluster, focus has been set on the Tugai forests, with Populus euphratica as the dominant tree species, because these forests belong to the most productive and species-rich natural ecosystems of the Tarim River basin. At sites close to the groundwater, the annual stem diameter increments of Populus euphratica correlated with the river runoffs of the previous year. However, the natural river dynamics cease along the downstream course and thus hamper the recruitment of Populus euphratica. A study on the willingness to pay for the conservation of the natural ecosystems was conducted to estimate the concern of the people in the region and in China's capital. These household surveys revealed that there is a considerable willingness to pay for conservation of the natural ecosystems, with mitigation of dust and sandstorms considered the most important ecosystem service. Stakeholder dialogues contributed to creating a scientific basis for a sustainable management in the future.

Im Geheimbund "Gläserne Kette" sahen er und seine Mitstreiter sich als Propheten und "übermenschliche Wächter" aller Künste. Die quasi religiös inspirierte Glasarchitektur wurde schon wenige Jahre später im Staatlichen Bauhaus Weimar durch Gropius' ästhetische Rationalität abgelöst.
Recent research has revealed that learning behavior is associated with academic achievement at the college level, but the impact of specific learning strategies on academic success as well as gender differences therein are still not clear. Therefore, the aim of this study was to investigate gender differences in the incremental contribution of learning strategies over general cognitive ability in the prediction of academic achievement. The relationship between these variables was examined by correlation analyses. A set of t-tests was used to test for gender differences in learning strategies, whereas structural equation modeling as well as multi-group analyses were applied to investigate the incremental contribution of learning strategies for male and female students’ academic performance. The sample consisted of 461 students (mean age = 21.2 years, SD = 3.2). Correlation analyses revealed that general cognitive ability as well as the learning strategies effort, attention, and learning environment were positively correlated with academic achievement. Gender differences were found in the reported application of many learning strategies. Importantly, the prediction of achievement in structural equation modeling revealed that only effort explained incremental variance (10%) over general cognitive ability. Results of multi-group analyses showed no gender differences in this prediction model. This finding provides further knowledge regarding gender differences in learning research and the specific role of learning strategies for academic achievement. The incremental assessment of learning strategy use as well as gender-differences in their predictive value contributes to the understanding and improvement of successful academic development.
Filamentous, heterocyst-forming cyanobacteria exchange nutrients and regulators between cells for diazotrophic growth. Two alternative modes of exchange have been discussed involving transport either through the periplasm or through septal junctions linking adjacent cells. Septal junctions and channels in the septal peptidoglycan are likely filled with septal junction complexes. While possible proteinaceous factors involved in septal junction formation, SepJ (FraG), FraC, and FraD, have been identified, little is known about peptidoglycan channel formation and septal junction complex anchoring to the peptidoglycan. We describe a factor, SjcF1, involved in regulation of septal junction channel formation in the heterocyst-forming cyanobacterium Anabaena sp. strain PCC 7120. SjcF1 interacts with the peptidoglycan layer through two peptidoglycan-binding domains and is localized throughout the cell periphery but at higher levels in the intercellular septa. A strain with an insertion in sjcF1 was not affected in peptidoglycan synthesis but showed an altered morphology of the septal peptidoglycan channels, which were significantly wider in the mutant than in the wild type. The mutant was impaired in intercellular exchange of a fluorescent probe to a similar extent as a sepJ deletion mutant. SjcF1 additionally bears an SH3 domain for protein-protein interactions. SH3 binding domains were identified in SepJ and FraC, and evidence for interaction of SjcF1 with both SepJ and FraC was obtained. SjcF1 represents a novel protein involved in structuring the peptidoglycan layer, which links peptidoglycan channel formation to septal junction complex function in multicellular cyanobacteria. Nonetheless, based on its subcellular distribution, this might not be the only function of SjcF1.
In the initial stage of relativistic heavy-ion collisions, strong magnetic fields appear due to the large velocity of the colliding charges. The evolution of these fields appears as a novel and intriguing feature in the fluid-dynamical description of heavy-ion collisions. In this work, we study analytically the one-dimensional, longitudinally boost-invariant motion of an ideal fluid in the presence of a transverse magnetic field. Interestingly, we find that, in the limit of ideal magnetohydrodynamics, i.e., for infinite conductivity, and irrespective of the strength of the initial magnetization, the decay of the fluid energy density e with proper time τ is the same as for the time-honoured “Bjorken flow” without magnetic field. Furthermore, when the magnetic field is assumed to decay , where a is an arbitrary number, two classes of analytic solutions can be found depending on whether a is larger or smaller than one. In summary, the analytic solutions presented here highlight that the Bjorken flow is far more general than formerly thought. These solutions can serve both to gain insight on the dynamics of heavy-ion collisions in the presence of strong magnetic fields and as testbeds for numerical codes.
Esse artigo tem por objetivo analisar as contribuições de Axel Honneth para o atual debate das teorias da justiça, entre as quais a principal é a busca de princípios normativos encrustados na realidade social. Em sua obra O direito da liberdade, o autor indica a liberdade como o grande valor moderno. O medium da justiça seria uma liberdade de tipo social a qual estaria expressa nas instituições vinculadas às relações pessoais, ao mercado e ao universo político. Considerando a lacuna entre os princípios normativos de justiça indicados pelo autor e a realidade social este artigo propõe colocar em discussão as potencialidades e limites da própria reconstrução normativa como instrumento de análise do social, pautando especificamente o mercado de trabalho, a fim de colaborar à discussão das possibilidades de articular a norma compartilhada e a emergência de valores em vias de institucionalização.
Das Feld der interdisziplinäre Diskursforschung hat in den letzten Jahren zunehmend an Bedeutung gewonnen und sich zu einer etablierten Forschungsperspektive am Schnittpunkt von Sprache und Gesellschaft, von Wissen und Macht entwickelt. Die theoretische und methodische Vielgestaltigkeit dieser Forschungsperspektive führt allerdings insbesondere bei der Konzeption und Durchführung von Forschungsarbeiten solchen Zuschnitts immer wieder zu Unsicherheiten und Schwierigkeiten. Drei Werke, die – in unterschiedlicher Weise – auf das sich aus der Vielgestaltigkeit dieses Feldes ergebende Bedürfnis nach Systematisierung und Orientierung antworten, werden im Folgenden vorgestellt. Dabei gilt es deutlich herauszustellen, dass die vorgestellten Werke nicht als Methodenbücher oder Anleitungen zur "korrekten" Durchführung von diskursorientierten Forschungsarbeiten misszuverstehen, sondern vielmehr als Anregung und Verständigung über Fragen, Probleme und Richtungen der Diskursforschung auch über nationale und disziplinäre Grenzen hinweg zu lesen sind.
Dies ist der zweite Artikel unseres Blogfokus zu Flucht und Migration. In den medialen Auftritten von PolitikerInnen dominieren Darstellungen der Flüchtlinge als Probleme. Gleichzeitig lassen sich in den Medien Bemühungen beobachten, die darauf abzielen, die Flüchtlinge nicht als Problem – welcher Art auch immer, sondern als die Lösung für bestimmte Probleme zu verstehen. Beides ist problematisch und ersetzt nicht einen öffentlichen Diskurs über die Frage, ob unser Verständnis von Demokratie und Selbstbestimmung möglicherweise überholt ist und nach einer Neuauflage verlangt, welche das Verhältnis zwischen dem Selbstbestimmungsrecht von Nationalstaaten und den (Menschen-)Rechten von Personen neu bestimmt.

The technique of self absorption has been applied for the first time to study the decay pattern of low-lying dipole states of 140Ce. In particular, ground-state transition widths 0 and branching ratios 0/ to the ground state have been investigated in the energy domain of the pygmy dipole resonance. Relative self-absorption measurements allow for a model-independent determination of 0. Without the need to perform a full spectroscopy of all decay channels, also the branching ratio to the ground state can be determined. The experiment on 140Ce was conducted at the bremsstrahlung facility of the superconducting Darmstadt electron linear accelerator S-DALINAC. In total, the self-absorption and, thus, 0 were determined for 104 excited states of 140Ce. The obtained results are presented and discussed with respect to simulations of γ cascades using the DICEBOX code.
The method of relative self absorption is based on the technique of nuclear resonance fluorescence measurements. It allows for a model-independent determination of ground-state transition widths, natural level widths, and, consequently, of branching ratios to the ground state for individual excitations. Relative self–absorption experiments have been performed on the nuclei 6Li and 140Ce. In order to investigate the total level width for the 0+1, T = 1 level at 3563 keV in 6Li, a high-precision self-absorption measurement has been performed. In the case of 140Ce, self absorption has been applied for the first time to study decay widths of dipole-excited states in the energy regime of the pygmy dipole resonance.

Für Arbeitswege charakteristisch sind eine räumliche und zeitliche Konzentration in Richtung Arbeitsplatzzentren und zu Tagesrandzeiten sowie ein überdurchschnittlich hoher Anteil des motorisierten Individualverkehrs an der Verkehrsmittelnutzung. So tragen Arbeitswege bedeutend zur zyklischen Belastung der Verkehrsinfrastruktur sowie zu städtischen Verkehrs- und Umweltproblemen bei. Da die Verkehrsmittelnutzung vor allem auch auf Arbeitswegen unter stabilen Rahmenbedingungen außerdem in hohem Maße routinisiert abläuft, kommt sogenannten Umbruchsereignissen in Mobilitätsbiographien eine zentrale Bedeutung hinsichtlich einer Reflexion des Mobilitätsverhaltens zu. Aufgrund dessen wird in dieser Arbeit zum einen untersucht, inwiefern überregionale Wohnumzüge (zwischen Bremen, Hamburg und dem Ruhrgebiet) zu einer Veränderung der Verkehrsmittelnutzung auf Arbeitswegen beitragen. Darauf aufbauend wird mittels multivariaten binär-logistischen Regressionsmodellen untersucht, welche Bedeutung veränderte raumstrukturelle Rahmenbedingungen sowie a priori vorhandene Verkehrsmittelpräferenzen für diese Veränderungen haben. Die Ergebnisse zeigen zum einen, dass es im Anschluss an Wohnumzüge zu einer signifikanten Veränderung der Verkehrsmittelnutzung auf Arbeitswegen in Form von einer Anpassung dessen an die mobilitätsbezogenen Rahmenbedingungen des Zielortes des Wohnumzugs kommt. Anhand der multivariaten Regressionsmodelle kann darüber hinaus ein signifikanter Einfluss raumstruktur- und präferenzbezogener Faktoren auf die Veränderung der Verkehrsmittelnutzung auf Arbeitswegen nachgewiesen werden. Da sich der Einfluss dieser Faktoren je nach Verkehrsmittel zum Teil sehr unterschiedlich gestaltet, werden aus diesen Ergebnissen abschließend verkehrsmittelspezifische Maßnahmen, z.B. zur Gestaltung eines (betrieblichen) Mobilitätsmanagements, abgeleitet.

Background: Aging is associated with loss of balance and activity in daily life. It impacts postural control and increases the risk of falls. The current study was conducted to determine the feasibility and long-term impact of stochastic resonance whole-body vibration (SR-WBV) on static and dynamic balance and reaction time among elderly individuals.
Methods: A randomized crossover pilot study with blinding of the participants. Twenty elderly were divided into group A (SR-WBV 5 Hz, Noise 4/SR-WBV 1 Hz, Noise 1) or group B (SR-WBV 1 Hz, Noise 1/SR-WBV 5 Hz, Noise 1). Feasibility outcomes included recruitment, compliance and safety. Secondary outcomes were Semi-Tandem Stand (STS), Functional Reach Test (FRT), Expanded Timed Get Up-and-Go (ETGUG), walking under single (ST) & dual task (DT) conditions, hand and foot reaction time (RTH/RTF). Puri and Sen Rank-Order L Statistics were used to analyse carry-over effects. To analyse SR-WBV effects Wilcoxon signed-ranked tests were used.
Results: With good recruitment rate (55%) and compliance (attrition 15%; adherence 85%) rates the intervention was deemed feasible. Three participants dropped out, two due to knee pain and one for personal reasons. ETGUG 0 to 2 m (p = 0.143; ES: 0.36) and ETGUG total time (p = 0.097; ES: 0.40) showed medium effect sizes.
Conclusions: Stochastic resonance training is feasible in untrained elderly resulting in good recruitment and compliance. Low volume SR-WBV exercises over 12 training sessions with 5 Hz, Noise 4 seems a sufficient stimulus to improve ETGUG total time. The stimulation did not elicit changes in other outcomes.

Life-saving pig-to-human xenotransplantation is a promising technology with the potential to balance the shortage of human organs in allotransplantation. Before this approach is applied on solid vascularized organs, several barriers must be overcome. Patient safety is menaced by infectious porcine endogenous retroviruses (PERV) which are able to infect human cell lines in vitro. Successful infection with PERV is associated with diverse life-threatening consequences including gene disruption, tumorigenicity, immune suppression as well as PERV proliferation throughout the whole human body. This could cause a catastrophic xenozonoosis leading to the emergence of new forms of pathogens and pandemic diseases similar to AIDS. However, in vivo, there is hitherto no incidence of any infection with PERV in preclinical xenotransplantations performed in the past.
PERV infection of human peripheral blood mononuclear cells (huPBMC) is a critical issue discussed controversially in several studies. It is essential to address the sensitivity of huPBMC to infection by PERV since it is generally one of the first retroviral targets upon viral invasion and infection of the human body. To assess definitely if huPBMC are infected productively by PERV, target cells were challenged with the highest infectious PERV class, recombinant PERV-A/C, in different assays. Modern and standard methods to detect PERV at different stages of viral cycles were used to monitor PERV development upon contact with host cells. Indeed, PERV-A/C in supernatants of producer cell lines failed to infect mitogen-activated huPBMC. Neither retroviral reverse transcriptase (RT) nor viral RNA packaged in virus particles were observed in supernatants of cells exposed to viral supernatants. In addition, provirus was not detected in huPBMC until 56 days p. i. with PERV-A/C. Independently of the virus load applied, culture conditions of huPBMC or administration of polybrene as enhancer, PERV was unable to infect huPBMC. Results suggest that PERV in supernatants lack sufficient infectious potential to be productively generated in huPBMC.
In order to approximate xenotransplantation scenarios, different PERV producing cells including PHA-activated porcine PBMC (poPBMC) were adopted as virus source in co-cultivation studies with huPBMC. In this case, expression of viral RNA was successfully measured. However, RT activity did not increase until 28 days p. e. with PERV producer cells which indicates that viral particles devoid of infectious capacity were released from non-productively infected cells.
On the other hand, co-cultivation of both virus producer and virus recipients increases the contact pressure between PERV and target cells. Consequently, PERV was able to be detected at least as provirus in huPBMC. Although virions produced were not functional, presence of provirus in infected cells will sooner or later provoke expression of provirus. This could lead to chromosomal rearrangements as well as virus reinfection and insertional mutagenesis.
Ecotropic PERV-C displays a restricted host range to porcine cells. Given its ability to serve as template to form recombinant xenotropic PERV-A/C, PERV-C represents a potent hazard in the course of xenotransplantation. Thus, isolation and functional characterization of PERV-C in the genome of pigs in use and intended for xenotransplantation is necessary to analyze the genetics of these virions as well as to select animals lacking proviral PERV-C or to generate transgenic PERV-C negative donors.
PERV-C was isolated from the genome of a female SLAd/d haplotype pig via screening of a bacteriophage library which was constructed from the genomic DNA of poPBMC extracted from this PERV non-transmitting sow. Upon genetic complementation of provirus using a PCR fragment infectious ability of full-length PERV-C clones was investigated in cell culture. PERV-C clones were successfully reproduced in susceptible porcine cells as RT activity as well as viral RNA were detected in supernatants of infected cells 56 days p. i. Furthermore, presence of proviruses in challenged cells was confirmed by nested PCR.
PERV-C clones were also isolated from a bacteriophage library generated on genomic DNA of an Auckland island pig of the DPF colony, whose individuals display a PERV-null phenotype and are already in use for xenotransplantation, and of a Göttingen minipig, whose relatives serve as animal models to study human diseases. In contrast to PERV clones isolated from the female SLAd/d haplotype sow PERV-C clones of the Auckland island pig as well as of the Göttingen minipig were not functional and therefore unable to infect target cells. This confirms the PERV-null phenotype which renders these animals putative candidates as donors in xenotransplantation. On the other hand, presence of functional PERV-C in SLAd/d haplotype pigs exerts a negative impact on patient safety in xenotransplantation. The suitability of these animals as potent organ donors should be intensively investigated.
In conclusion, PERV of all classes pose a virological risk in xenotransplantation which should not be ignored. Since exclusion of all PERV from donor herds is impossible, generation of transgenic humanized animals lacking genomic infectious PERV represents the best strategy to guarantee patient safety in future life-saving pig-to-human xenotransplantation.
The structure of the 1:2 co-crystalline adduct C8H16N4·2C6H5BrO, (I), from the solid-state reaction of 1,3,6,8-tetraazatricyclo[4.4.1.13,8]dodecane (TATD) and 4-bromophenol, has been determined. The asymmetric unit of the title co-crystalline adduct comprises a half molecule of aminal cage polyamine plus a 4-bromophenol molecule. A twofold rotation axis generates the other half of the adduct. The primary inter-species association in the title compound is through two intermolecular O—H⋯N hydrogen bonds. In the crystal, the adducts are linked by weak non-conventional C—H⋯O and C—H⋯Br hydrogen bonds, giving a two-dimensional supramolecular structure parallel to the bc plane.
In the title compound, C23H19NO2, an oxazine Mannich base derivative, the oxazine ring has a half-chair conformation. The 2-hydroxynaphthalen-1-yl substituent is placed in an axial position. There is an intramolecular O-H...N hydrogen bond, forming an S(6) graph-set motif. In the crystal, molecules are connected by a pair of C-H...[pi] interactions into an inversion dimer, which is reinforced by another pair of weak C-H...[pi] interactions. The dimers are linked by a [pi]-[pi] interaction [centroid-centroid distance = 3.6268 (17) Å], consolidating a column along the a axis. Furthermore, the columns interact with each other by a weak C-H...[pi] interaction, generating a three-dimensional network.
In the crystal of the title co-crystalline adduct, C8H16N4·C8H9ClO, (I), prepared by solid-state reaction, the molecules are linked by intermolecular O—H⋯N hydrogen bonds, forming a D motif. The azaadamantane structure in (I) is slightly distorted, with N—CH2—CH2—N torsion angles of 10.4 (3) and −9.0 (3)°. These values differ slightly from the corresponding torsion angles in the free aminal cage (0.0°) and in related co-crystalline adducts, which are not far from a planar geometry and consistent with a D2d molecular symmetry in the tetraazatricyclo structure. The structures also differ in that there is a slight elongation of the N—C bond lengths about the N atom that accepts the hydrogen bond in (I) compared with the other N—C bond lengths. In the crystal, the two molecules are not only linked by a classical O—H⋯N hydrogen bond but are further connected by weak C—H⋯π interactions, forming a two-dimensional supramolecular network parallel to the bc plane.
The crystal structure of the title compound, C25H24N2O2, at 173 K has monoclinic (C2/c) symmetry. The molecule is located on a crystallographic twofold rotation axis with only half a molecule in the asymmetric unit. The imidazolidine ring adopts a twist conformation, with a twist about the ring C—C bond. The crystal structure shows the anticlinal disposition of the two (2-hydroxynaphthalen-1-yl)methyl substituents of the imidazolidine ring. The structure displays two intramolecular O—H⋯N hydrogen bonds, each forming an S(6) ring motif.
In the title ternary co-crystalline adduct, C7H14N4·2C6H5NO3, molecules are linked by two intermolecular O—H⋯N hydrogen bonds, forming a tricomponent aggregates in the asymmetric unit. The hydrogen-bond formation to one of the N atoms is enough to induce structural stereoelectronic effects in the normal donor→acceptor direction. In the title adduct, the two independent nitrophenol molecules are essentially planar, with maximum deviations of 0.0157 (13) and 0.0039 (13) Å. The dihedral angles between the planes of the nitro group and the attached benzene rings are 4.04 (17) and 5.79 (17)°. In the crystal, aggregates are connected by C—H⋯O hydrogen bonds, forming a supramolecular dimer enclosing an R66(32) ring motif. Additional C—H⋯O intermolecular hydrogen-bonding interactions form a second supramolecular inversion dimer with an R22(10) motif. These units are linked via C—H⋯O and C—H⋯N hydrogen bonds, forming a three-dimensional network.
The title compound, C21H26Cl2N2O2, was prepared in a solvent-free microwave-assisted synthesis, and crystallizes in the orthorhombic space group Pna21. The imidazolidine ring adopts an envelope conformation and its mean plane is almost perpendicular to the two pendant aromatic rings [dihedral angles = 84.61 (9) and 86.54 (9)°]. The molecular structure shows the presence of two intramolecular O—H⋯N hydrogen bonds between the phenolic hydroxy groups and imidazolidine N atoms. The two 3-chloro-6-hydroxy-2,4-dimethylbenzyl groups are located in a cis orientation with respect to the imidazolidine fragment. As a result, the lone pairs of electrons on the N atoms are presumed to be disposed in a syn conformation. This is therefore the first example of an exception to the `rabbit-ears' effect in such 2,2′-[imidazolidine-1,3-diylbis(methylene)]diphenol derivatives.

PRACTITIONERS AND RESEARCHERS ALIKE INCREASINGLY USE SOCIAL MEDIA MESSAGES AS AN ADDITIONAL SOURCE OF INFORMATION WHEN DEALING WITH STOCKS. BASED ON EMOTION THEORY AND AN ESTABLISHED SENTIMENT LEXICON, WE DEVELOP AND APPLY AN OPEN SOURCE DICTIONARY FOR THE ANALYSIS OF SEVEN DIFFERENT EMOTIONS IN 5.5 MILLION TWITTER MESSAGES ON 33 S&P 100 COMPANIES. WE FIND VARYING EXPLANATORY POWER OF DIFFERENT EMOTIONS (ESP. HAPPINESS, AND DEPRESSION) FOR COMPANY-SPECIFIC STOCK PRICE MOVEMENTS OVER A PERIOD OF THREE MONTHS.
Introduction: Aim of this study was to reduce blood loss caused by diagnostic blood sampling and to minimize the development of anemia in a high-risk group of mechanically ventilated medical intensive care patients. We therefore implemented a “blood-saving bundle” (BSB) combining a closed-loop arterial blood sampling system, smaller sampling tubes, reduced frequency of blood drawings, and reduced sample numbers.
Methods: The study included all patients from our medical ICU who were ventilated for more than 72 hours. Exclusion criteria were: acute or chronic anemia on admission, bleeding episode(s) during the ICU stay, or end-of-life therapy. The BSB was introduced in 2009 with training and educational support. Patients treated in 2008, before the introduction of the BSB, served as a control group (n = 41, 617 observation days), and were compared with patients treated in 2010 after the introduction of the BSB (BSB group, n = 50, 559 observation days). Primary endpoints were blood loss per day, and development of anemia. Secondary endpoints were numbers of blood transfusions, number of days on mechanical ventilation, and length of the ICU stay.
Results: Mean blood loss per ICU day was decreased from 43.3 ml (95% CI: 41.2 to 45.3 ml) in the controls to 15.0 ml (14.3 to 15.7 ml) in the BSB group (P < 0.001). The introduction of a closed-loop arterial blood sampling system was the major contributor to this effect. Mean hemoglobin concentrations showed no significant differences in both groups during the ICU stay. Hemoglobin values <9 g/dl, however, were recorded in 21.2% of observation days in the controls versus 15.4% in the BSB group (P = 0.01). Units of transfused red blood cells per 100 observation days decreased from 7 to 2.3 (P < 0.001). The mean number of ventilation days was 7.1 days (6.1 to 8.3 days) in the controls and 7.5 days (6.6 to 8.5 days) in the BSB group (P = NS). In total, patients in the BSB group stayed in ICU for a mean of 9.9 days (8.6 to 11.3 days), compared to a mean ICU stay of 13.0 days (10.9 to 15.4 days) in the control group (P = 0.014). Due to the longitudinal study design, however, we cannot exclude uncontrolled confounders affecting the transfusion frequency and mean ICU stay.
Conclusion: Our BSB could be easily implemented and was able to reduce diagnostic blood loss.

Municipal wastewater contains nutrients valuable for a reuse in agriculture and can be the source of a multitude of chemicals used in private households and industry, too. As many of these chemicals are incompletely degraded during wastewater treatment, their residues remain partly in sewage sludge and partly in treated wastewater. Concerns are linked particularly to the so called micropollutants, i.e. anthropogenic organic substances such as personal care products, pharmaceuticals and biocides, for which scarce data on their degradability and environmental fate and particularly on their ecotoxicity are available. Thus, when reusing treated wastewater and sewage sludge for irrigation or as soil amendment for a sustainable land and water management, these wastewater-borne pollutants may enter soil, groundwater and surface water. The present work therefore aimed at assessing potential ecotoxic effects on aquatic and terrestrial organisms of reusing treated wastewater and sewage sludge. To this end, established as well as newly developed experimental approaches were used to investigate the problem on several levels. Individual wastewater-borne substances, samples from field study sites and samples from a soil column experiment simulating prolonged wastewater irrigation were examined.
At the start of the experimental work, the ecotoxicity of climbazole was characterised towards five aquatic and five terrestrial test organisms. Climbazole is an azole antimycotic agent applied in cosmetics and anti-dandruff shampoos and was recently detected in relatively high concentrations in treated wastewater and sewage sludge. In the present work climbazole was found to be particularly toxic towards plants such as water lentils with effective concentrations comparable to those of agricultural azole fungicides. Dwarfism, that is reduced shoot elongation observed in plants, pointed at a specific, phytohormone inhibiting mode of action of climbazole. Furthermore, the expected influence of the soil pH on the phytotoxicity of climbazole was experimentally confirmed.
Based on the findings for climbazole, two additional azole antimycotics, ketoconazole and fluconazole, and the regularly in sewage sludge detected biocide benzyldimethyldodecyl-ammonium chloride (BDDA) were investigated for their toxicity towards plants.
In aqueous medium, an increasing phytotoxicity from fluconazole to BDDA, ketoconazole and climbazole was observed, while in soil, phytotoxicity increased from BDDA to ketoconazole, climbazole and fluconazole. The relatively low terrestrial toxicity of BDDA and ketoconazole probably resulted from their strong binding to soil as well as their good biodegradability. To render the exposure scenario more realistic, sewage sludge was co-applied with the four test substances in a parallel test run. However, as no detectable influence on their effective concentrations was found, it can be assumed that the current practice of assessing sewage sludge borne substances with biotests in standard soil is sufficiently realistic. In a further study, different advanced sludge-treatment technologies were assessed for their efficacy in reducing pollutants. Results from the present work indicated that effects assessed in terrestrial short term biotests only seldom correlated with the concentrations of certain pollutants. Rather, a negative correlation of the stability of the sludges, determined by the ratio of volatile to total solids, to their ecotoxicity was seen.
Another aspect of the present work was the design and performance of an experimental approach to assess the environmental risk of a long-term irrigation with treated wastewater concerning the quality of soil and water in a prospective way, i.e. before the installation at field scale. For the simulation of a continuous irrigation corresponding to approximately 30 years, a percolation apparatus was developed and four different soils were percolated with treated wastewater for three months. Acute and chronic biotests with nine test organisms from different trophic levels (green algae, water lentils and water fleas as well as oilseed rape, oats, bacteria, spring tails, enchytraeids and earthworms) were used to assess the soil percolates as well as the soils with and without percolation. These investigations were accompanied by a comprehensive chemical monitoring conducted by project partners. Results indicated that the soil passage, that is the percolation through the soil, generally improved the quality of the treated wastewater as habitat for aquatic organisms which was visible by a reduction of its phytotoxicity. However, in some cases it deteriorated the water quality, probably resulting from the leaching of metals from pre-contaminated soil. A deteriorated habitat quality of the soil after the percolation with treated wastewater was observed for several test organisms and soils. In the same, mainly peaty soils, the highest accumulation of wastewater-borne micropollutants and of zinc was measured. Yet, their concentrations did not correlate to the observed biological effects. Moreover, data on ecotoxicity were only available for a small fraction of the detected substances so that their concentrations could not successfully be used to predict expected biological effects.
The experimental approach used in the present work demonstrated to be an adequate tool to support the prospective evaluation of environmental risks of treated wastewater irrigation. Overall, it can be concluded that the reuse of treated wastewater on soil can improve the quality of treated wastewater but that this can come at the cost of deteriorating the quality of the soil. As these risks cannot be generalised, a comprehensive biotest battery as well as chemical analysis should be used to assess them on a case-specific basis for each respective wastewater and the respective soil.
In conclusion our data show, that Flightless I function is essential for striated muscle development in zebrafish. Myofibrillar bundling and focal adhesion formation represent the basis for this development, and are ultimately a prerequisite for cardiac trabeculation. Future analysis of Actin polymerization in trabeculation will provide addition knowledge about the sensitivity of the developing and adult heart to a disequilibrium in F-actin versus G-actin availability.
In this study we found a novel ErbB2-dependent cardiomyocyte maturation process which affects both cardiac chambers. It will be of great interest to further study the nature of the Memo1-GFP cell-cell junctions and other junction proteins in order to unravel the significance of this maturation process for heart development.
Interestingly we found, that memo1bns4 homozygous mutant animals, which we generated with CRISPR/Cas9 technology, develop indistinguishable from siblings, suggesting that zygotic memo1 expression is dispensable for zebrafish development. Future studies will address the question if maternal zygotic memo1bns4 mutants will develop a heart or vascular phenotype as reported form Memo1 knockout mice or as observed in memo1 morphants in this study.
In cultured C2 mouse skeletal muscle cells the Golgi-apparatus relocalizes dependent on centrosomal proteins and independent of microtubules. We describe here that zebrafish cardiomyocytes have a similar Golgi-complex distribution suggesting a similar differentiation-dependent reorganization. This striated muscle specific, fragmented Golgi distribution might be an advantage for these cells in order to shuttle vesicles through the densely packed sarcomere structures. Future studies could address the timing of the Golgi-reorganization in cardiomyocytes during development and possibly use this Golgi-zebrafish line as a tool to study cardiomyocyte maturation in disease models and in heart regeneration.
