Exclusive measurements of quasi-free proton scattering reactions in inverse and complete kinematics

Quasi-free scattering reactions of the type (p, 2p)were measured for the first time exclusively in com-plete and inverse kinematics, using a 12C beam at an energy of ∼400MeV/u as a benchmark. This new technique has been developed to study the single-particle structure of exotic nuclei in experiments with radioactive-ion beams. The outgoing pair of protons and the fragments were measured simultaneously, enabling an unambiguous identification of the reaction channels and a redundant measurement of the kinematic observables. Both valence and deeply-bound nucleon orbits are probed, including those leading to unbound states of the daughter nucleus. Exclusive (p, 2p)cross sections of 15.8(18)mb, 1.9(2)mb and 1.5(2)mb to the low-lying 0p-hole states overlapping with the ground state (3/2−) and with the bound excited states of 11B at 2.125MeV (1/2−) and 5.02MeV (3/2−), respectively, were determined via γ-ray spectroscopy. Particle-unstable deep-hole states, corresponding to proton removal from the 0s-orbital, were studied via the invariant-mass technique. Cross sections and momentum distributions were ex-tracted and compared to theoretical calculations employing the eikonal formalism. The obtained results are in a good agreement with this theory and with direct-kinematics experiments. The dependence of the proton–proton scattering kinematics on the internal momentum of the struck proton and on its sep-aration energy was investigated for the first time in inverse kinematics employing a large-acceptance measurement.
Snake bite envenoming often results in disability or death of breadwinners of poor families in the rural tropics and the subtropics of Nepal. Identification of the medically relevant snake species, circumstances of venomous snake bites, prehospital care of their bites and human responses to snakes and snake bite is, therefore, crucial to enable victims or first aider to select the appropriate first aid measures, physicians to anticipate complications and to use appropriate treatment protocols as well as the local community to implement prevention strategies. Inadequate educational gaps exist in Nepal and hinder identification of snakes involved in bites. To fill this gap, I aim to provide an evidence-based list of medically relevant snake species. Snake specimens brought by patients bitten or their attendants from the tropical and subtropical regions in southeastern, southcentral, and southwestern Nepal to snake bite treatment centres over a period from 2010 through 2014, were taxonomically identified and medical records of envenoming were evaluated.
In Nepal, the epidemiology of snake bite is poorly known. Here I describe the ecological circumstances of proven krait (Bungarus spp.) and Russell´s Viper (Daboia russelii) bites to elucidate and examine, whether environmental circumstances or human behaviour contributed to envenoming. In a cross-sectional study, data about prehospital care, environmental circumstances of 46 krait and 10 Russell´s Viper bites were evaluated. Patients were interviewed using structured interview forms. Snake bite prone communities were surveyed to test people´s knowledge on snakes and their attitude towards venomous snakes in general.
Of 349 snakes involved in bites, 199 (57%) specimens were found to be medically relevant venomous snakes that included 11 species belonging to six genera and two families. Among them, Naja naja (n = 76, 22%), Bungarus caeruleus (n = 65, 19%) and Trimeresusurs albolabris (n = 10, 3%) were the most widely distributed snakes. Daboia russelii (n = 10, 3%) was found to be restricted to the southwestern part of Nepal. For B. walli, a previously poorly known species, 13 voucher specimens represent the first country records of this species as well as the first documented cases of involvement in snake bite envenoming by this species in Nepal.
Numerous snake bites (33%) occurred at night, during the rainy season, and are mainly due to Bungarus species, particularly B. caeruleus. Bites of cobras and Russell’s Vipers are a risk at daytime. Evaluation of data regarding the place where the bite happened, indicates that the snake bite risks appear to be as high in residential areas, in and around houses, as in rural areas. In cases of kraits (n = 46), 61% of the bites occurred while the victim was sleeping indoors, those of Russell´s Vipers mainly during agricultural activities in the fields. Analysis of socio-demographic data revealed that both krait and viper bites predominantly affected farmers or their family members. However, snake bites involved also people of higher socio-economic status, which suggests that it is not a health problem of poor people only living in the rural areas of Nepal.
A small number of snake bite victims (n = 7) sought help from traditional healers, but most patients went to hospitals for medical treatment using motorbikes (65%) or were transferred by ambulance cars (22%). As a first aid measure, most patients (78%) had used a tourniquet, which is of doubtful value and has often severe sequelae, instead of applying the WHO recommended pressure immobilisation bandage or local compression pad. The overall case fatality rate was calculated to be 10%, but up to 17% in cases of Bungarus spp. bites.
Rural community people were found to be extremely afraid of snakes, a major reason for indiscriminate killing of even harmless snakes, e.g., Lycodon aulicus, which were wrongly considered to be venomous. This is mainly due to the poor knowledge on snakes in general and on their role in providing ecological services, which may eventually lead to a decline in snake populations and even the extinction of rare species.
The results of the present study strongly emphasize that snake bite is an important public health issue in Nepal. There is an urgent need to improve the knowledge of people on snakes and to try changing their attitudes towards these reptiles, in addition to documenting the biodiversity and distribution of medically relevant snakes, the epidemiology and circumstances of their bites. Avoiding high-risk behaviour (e.g., killing of snakes), using screened doors and windows are some of the suggested measures preventing snake bite. Early and accurate identification of the snakes involved should help physicians to apply timely treatment, eventually referring the patient to the appropriate hospital. This also has important implications in developing public health and conservation strategies, to the benefit of the people of Nepal.
The c-MYC proto-oncogene is a regulator of fundamental cellular processes such as cell cycle progression and apoptosis. The development of novel c-MYC inhibitors that can act by targeting the c-MYC DNA G-quadruplex at the level of transcription would provide potential insight into structure-based design of small molecules and lead to a promising arena for cancer therapy. Herein we report our finding that two simple bis-triazolylcarbazole derivatives can inhibit c-MYC transcription, possibly by stabilizing the c-MYC G-quadruplex. These compounds are prepared using a facile and modular approach based on Cu(I) catalysed azide and alkyne cycloaddition. A carbazole ligand with carboxamide side chains is found to be microenvironment-sensitive and highly selective for "turn-on" detection of c-MYC quadruplex over duplex DNA. This fluorescent probe is applicable to visualize the cellular nucleus in living cells. Interestingly, the ligand binds to c-MYC in an asymmetric fashion and selects the minor-populated conformer via conformational selection.
IKKβ acts as a tumor suppressor in cancer-associated fibroblasts during intestinal tumorigenesis

Cancer-associated fibroblasts (CAFs) comprise one of the most important cell types in the tumor microenvironment. A proinflammatory NF-κB gene signature in CAFs has been suggested to promote tumorigenesis in models of pancreatic and mammary skin cancer. Using an autochthonous model of colitis-associated cancer (CAC) and sporadic cancer, we now provide evidence for a tumor-suppressive function of IKKβ/NF-κB in CAFs. Fibroblast-restricted deletion of Ikkβ stimulates intestinal epithelial cell proliferation, suppresses tumor cell death, enhances accumulation of CD4+Foxp3+ regulatory T cells, and induces angiogenesis, ultimately promoting colonic tumor growth. In Ikkβ-deficient fibroblasts, transcription of negative regulators of TGFβ signaling, including Smad7 and Smurf1, is impaired, causing up-regulation of a TGFβ gene signature and elevated hepatocyte growth factor (HGF) secretion. Overexpression of Smad7 in Ikkβ-deficient fibroblasts prevents HGF secretion, and pharmacological inhibition of Met during the CAC model confirms that enhanced tumor promotion is dependent on HGF–Met signaling in mucosa of Ikkβ-mutant animals. Collectively, these results highlight an unexpected tumor suppressive function of IKKβ/NF-κB in CAFs linked to HGF release and raise potential concerns about the use of IKK inhibitors in colorectal cancer patients.
Single-molecule super-resolution microscopy allows imaging of fluorescently-tagged proteins in live cells with a precision well below that of the diffraction limit. Here, we demonstrate 3D sectioning with single-molecule super-resolution microscopy by making use of the fitting information that is usually discarded to reject fluorophores that emit from above or below a virtual-'light-sheet', a thin volume centred on the focal plane of the microscope. We describe an easy-to-use routine (implemented as an open-source ImageJ plug-in) to quickly analyse a calibration sample to define and use such a virtual light-sheet. In addition, the plug-in is easily usable on almost any existing 2D super-resolution instrumentation. This optical sectioning of super-resolution images is achieved by applying well-characterised width and amplitude thresholds to diffraction-limited spots that can be used to tune the thickness of the virtual light-sheet. This allows qualitative and quantitative imaging improvements: by rejecting out-of-focus fluorophores, the super-resolution image gains contrast and local features may be revealed; by retaining only fluorophores close to the focal plane, virtual-'light-sheet' single-molecule localisation microscopy improves the probability that all emitting fluorophores will be detected, fitted and quantitatively evaluated.
Prehistoric dental treatments were extremely rare, and the few documented cases are known from the Neolithic, when the adoption of early farming culture caused an increase of carious lesions. Here we report the earliest evidence of dental caries intervention on a Late Upper Palaeolithic modern human specimen (Villabruna) from a burial in Northern Italy. Using Scanning Electron Microscopy we show the presence of striations deriving from the manipulation of a large occlusal carious cavity of the lower right third molar. The striations have a "V"-shaped transverse section and several parallel micro-scratches at their base, as typically displayed by cutmarks on teeth. Based on in vitro experimental replication and a complete functional reconstruction of the Villabruna dental arches, we confirm that the identified striations and the associated extensive enamel chipping on the mesial wall of the cavity were produced ante-mortem by pointed flint tools during scratching and levering activities. The Villabruna specimen is therefore the oldest known evidence of dental caries intervention, suggesting at least some knowledge of disease treatment well before the Neolithic. This study suggests that primitive forms of carious treatment in human evolution entail an adaptation of the well-known toothpicking for levering and scratching rather than drilling practices.
A novel series of ribonucleosides of 1,2,3-triazolylbenzyl-aminophosphonates was synthesized through the Kabachnik–Fields reaction using I2 as catalyst followed by copper-catalyzed cycloaddition of the azide–alkyne reaction (CuAAC). All structures of the newly prepared compounds were characterized by 1H NMR, 13C NMR, and HRMS spectra. The structures of 2e, 2f, 3d, and 3g were further confirmed by X-ray diffraction analysis. These compounds were tested against various strains of DNA and RNA viruses; compounds 4b and 4c showed a modest inhibitory activity against respiratory syncytial virus (RSV) and compound 4h displayed modest inhibitory activity against Coxsackie virus B4.
In the title compound, C20H24N2O4, both peptide bonds adopt a trans configuration with respect to the —N—H and —C=O groups. The dihedral angle between the aromatic rings is 53.58 (4)°. The molecular conformation is stabilized by an intramolecular N—H⋯O hydrogen bond. The crystal packing is characterized by zigzag chains of N—H⋯O hydrogen-bonded molecules running along the b-axis direction.
Ausgehend von dem gesellschaftlichen Problem des Übergewichts im Kindesalter wird die besondere Bedeutung und Verantwortung des Sportunterrichts für diese Klientel herausgestellt. Dabei wird die These vertreten, dass der Sportunterricht seinem Auftrag nur dann gerecht werden kann, wenn es gelingt, auch übergewichtigen Kindern positive Erfahrungen in Bezug auf Bewegung, Spiel und Sport zu vermitteln. Im Rahmen dieses sportpädagogischen Problemfeldes wurde zunächst ein Fragebogen konzipiert und validiert, der das Wohlbefinden als Indikator für positive Erfahrungen übergewichtiger Schüler mit dem von normalgewichtigen Kindern vergleicht (n = 336). Eine anschließende qualitative Untersuchung in Form von Leitfadeninterviews (mit acht übergewichtigen/adipösen Kindern) ergänzt und konkretisiert die Ergebnisse.
Als wesentliches Resultat konnte die Erkenntnis gewonnen werden, dass das Wohlbefinden – gemessen durch ein faktorenanalytisch generiertes Modell mit den drei Faktoren „Sportunterricht/Sportlehrer“ (Faktor I), „sportliches Selbstwertgefühl“ (Faktor II) und „Mitschüler/Schulzufriedenheit“ (Faktor III) – keine signifikanten Unterschiede zwischen den Gewichtsklassen zeigt (Faktor I p = .57; Faktor II p = .04; Faktor III p = .23). Übergewichtige Schüler fühlen sich demnach nicht weniger wohl als ihre normalgewichtigen Klassenkameraden, in der Skala sportliches Selbstwertgefühl erzielten sie sogar höhere Werte (Normalgewichtige m = 2,06 ± 0,96; Übergewichtige m = 2,27 ± 0,89). Trotz dieses positiven Befundes verspüren Übergewichtige durchaus so manche Unzufriedenheit. Die Frage nach der Wichtigkeit der bzw. der Zufriedenheit mit den Komponenten Sportunterricht, eigene sportliche Leistung, Zusammenarbeit mit den Mitschülern, Figur und Sportlehrer machte deutlich, dass den Übergewichtigen Figur und sportliche Leistung sehr wichtig sind, sie jedoch nur bedingt damit zufrieden sind. Die Unterschiede in den entsprechenden Skalen erwiesen sich als hoch signifikant (Figur p = .00 d = .28; sportliche Leistung p = .01 d = .29). Die Überprüfung der Frage F1.2 hinsichtlich der geschlechtsspezifischen Unterschiede in den Aussagen zum Wohlbefinden lieferte lediglich ein signifikantes Ergebnis (p = .01) mit mittlerem Effekt (d = .48). Übergewichtige Mädchen gaben im Faktor „Mitschüler/Schulzufriedenheit“ höhere Werte an (m = 3,27 ± 0,66) als übergewichtige Jungen (m = 2,93 ± 0,75). Daraus lässt sich schließen, dass sich die weiblichen Übergewichtigen besser von ihren Mitschülern verstanden und unterstützt fühlen und sie eine allgemein größere Schulzufriedenheit verspüren als die männliche Vergleichsgruppe.
Die Auswertung in Bezug auf die Herkunft der Schüler lieferte keine signifikanten Ergebnisse. Dieser Befund deutet auf eine gelungene Integration der ausländischen Schüler hin, die aber möglicherweise aufgrund des hohen Ausländeranteils im Stadtgebiet Offenbach nicht repräsentativ ist.
Die Auswertung der Interviews zeigte, dass der positive Selbstwert auf ein hohes Maß an sozialer Anerkennung zurückzuführen ist. Entgegen zahlreichen theoretischen Vorannahmen berichtete kein Kind von anhaltenden Diskriminierungen oder Schamgefühlen aufgrund seines Gewichts. Die Bedeutung der eigenen sportlichen Leistung zeichnete sich mehrfach als Schlüsselkriterium im Umgang mit der pädagogischen Herausforderung, dem Erschaffen eines Problembewusstseins, ohne den Selbstwert und die Freude am Sporttreiben zu trüben, ab. Übergewichtige Kinder messen der Leistung einen hohen Stellenwert bei und erkennen in der Hoffnung einer möglichen Verbesserung, dass eine Reduktion des Gewichtes vorteilhaft ist.

Am Anfang der folgenden Überlegungen steht eine Irritation: Sie resultiert aus der Bourdieu’schen Theorie von der Existenz unterschiedlicher Kapitalformen – des ökonomischen, sozialen und kulturellen Kapitals –, die im sozialen Miteinander mehr oder weniger konvertierbar sein sollten und vom Individuum zur sozialen Positionierung eingesetzt werden können. Zur Erklärung der sozialen Dynamik spätmittelalterlicher Höfe ist dieses Modell gleichermaßen einleuchtend und hilfreich. Es weist aber mindestens eine Bruchstelle auf: So gut es viele (wenn auch nicht alle) Prozesse und Strategien des Handelns im höfischen Kontext erklärt, ist die Dynamik erst einmal angelaufen, so lässt es doch die Frage nach dem Eintritt in das Spiel offen. Woher kommt das Kapital, das Bewegung über die ständige Konvertierung hinaus ermöglicht? Oder anders gefragt: Wie gelingt Neuankömmlingen der Eintritt? ...
The Federal Reserve’s muddled mandate to attain simultaneously the incompatible goals of maximum employment and price stability invites short-term-oriented discretionary policymaking inconsistent with the systematic approach needed for monetary policy to contribute best to the economy over time. Fear of liftoff—the reluctance to start the process of policy normalization after the end of a recession—serves as an example. Causes of the problem are discussed, drawing on public choice and cognitive psychology perspectives. The Federal Reserve could adopt a framework that relies on a simple policy rule subject to periodic reviews and adaptation. Replacing meeting-by-meeting discretion with a simple policy rule would eschew discretion in favor of systematic policy. Periodic review of the rule would allow the Federal Reserve the flexibility to account for and occasionally adapt to the evolving understanding of the economy. Congressional legislation could guide the Federal Reserve in this direction. However the Federal Reserve may be best placed to select the simple rule and could embrace this improvement on its own, within its current mandate, with the publication of a simple rule along the lines of its statement of longer-run goals.
Prostaglandin E2 (PGE2) favors multiple aspects of tumor development and immune evasion. Therefore, microsomal prostaglandin E synthase (mPGES-1/-2), is a potential target for cancer therapy. We explored whether inhibiting mPGES-1 in human and mouse models of breast cancer affects tumor-associated immunity. A new model of breast tumor spheroid killing by human PBMCs was developed. In this model, tumor killing required CD80 expression by tumor-associated phagocytes to trigger cytotoxic T cell activation. Pharmacological mPGES-1 inhibition increased CD80 expression, whereas addition of PGE2, a prostaglandin E2 receptor 2 (EP2) agonist, or activation of signaling downstream of EP2 reduced CD80 expression. Genetic ablation of mPGES-1 resulted in markedly reduced tumor growth in PyMT mice. Macrophages of mPGES-1-/- PyMT mice indeed expressed elevated levels of CD80 compared to their wildtype counterparts. CD80 expression in tumor-spheroid infiltrating mPGES-1-/- macrophages translated into antigen-specific cytotoxic T cell activation. In conclusion, mPGES-1 inhibition elevates CD80 expression by tumor-associated phagocytes to restrict tumor growth. We propose that mPGES-1 inhibition in combination with immune cell activation might be part of a therapeutic strategy to overcome the immunosuppressive tumor microenvironment.
Der interdisziplinäre Doktoranden-Workshop "Neue Tendenzen der Italienforschung zu Mittelalter und Renaissance" fand erstmalig vom 13. bis 15. November 2014 am Kunsthistorischen Institut in Florenz, Max-Planck-Institut, statt. Initiatoren und Organisatoren waren seitens der Geschichte INGRID BAUMGÄRTNER (Kassel) und KLAUS HERBERS (Erlangen-Nürnberg) sowie seitens der Kunstgeschichte ALESSANDRO NOVA (Florenz/Frankfurt am Main) und GERHARD WOLF (Florenz/Berlin). Ziel des gut besuchten Workshops war es, aktuelle Ansätze der Italienforschung gemeinsam zu kommentieren und kritisch zu würdigen. Nachwuchswissenschaftler/innen aus verschiedenen Disziplinen, insbesondere Geschichte, Kunstgeschichte und Philosophie, stellten ihre Projekte zur Diskussion. ...

Psychological ownership is a cognitive–affective construct based on individuals’ feelings of possessiveness towards and of being psychologically tied/attached to objects that are material (e.g. tools or work) and immaterial (e.g. ideas or workspace) in nature. Research suggests that psychological ownership could be influenced by various individual, organisational and contextual factors. The South African Employment Equity Act, which was implemented to grant equitable opportunities to previously disadvantaged employees, could be a significant contextual factor affecting psychological ownership, due to perceptions associated with inequality. Ethnicity may also act as a moderator for the relationship between perceptions of employment equity and psychological ownership. The objective of this study was to investigate the relationship between employment equity perceptions and psychological ownership and to explore whether ethnicity plays a moderating role in the relationship. A cross-sectional survey design was employed with a purposeful sample of 202 respondents employed in a large South African mining house. Pearson product–moment correlations and structural equation modelling confirmed that employment equity perceptions could predict the five components of psychological ownership. However, the results revealed that ethnicity has no moderating effect on the relationship between perceptions of employment equity and the emergence of psychological ownership. By implication, organisations that seek to retain employees targeted through equity initiatives need to find ways to enhance and develop the psychological ownership of these employees. The research contributes new insights into and knowledge of how contextual factors could influence employees’ psychological ownership.
Polygenic risk scores, based on risk variants identified in genome-wide-association-studies (GWAS), explain a considerable portion of the heritability for schizophrenia (SZ) and bipolar disorder (BD). However, little is known about the combined effects of these variants, although polygenic neuroimaging has developed into a powerful tool of translational neuroscience. In this study, we used genome wide significant SZ risk variants to test the predictive capacity of the polygenic model and explored potential associations with white matter volume, a key candidate in imaging phenotype for psychotic disorders.
By calculating the combined additive schizophrenia risk of seven SNPs (significant hits from a recent schizophrenia GWAS study), we show that increased additive genetic risk for SZ was associated with reduced white matter volume in a group of participants (n = 94) consisting of healthy individuals, SZ first-degree relatives, SZ patients and BD patients. This effect was also seen in a second independent sample of healthy individuals (n = 89). We suggest that a moderate portion of variance (~4%) of white matter volume can be explained by the seven hits from the recent schizophrenia GWAS.
These results provide evidence for associations between cumulative genetic risk for schizophrenia and intermediate neuroimaging phenotypes in models of psychosis. Our work contributes to a growing body of literature suggesting that polygenic risk may help to explain white matter alterations associated with familial risk for psychosis.
