For the environs of the Late Bronze Age fortification enclosure Iarcuri the hydro-morphological relief characteristics are combined with archaeological evidences. Target of the study is to evaluate the impact of settlement activities in the surroundings of Iarcuri on the development of the channel network. Data analysis is based on topographic map-derived and high resolution DEMs provided by LiDAR scanning; derivatives of the DEMs are used to characterize the different sub-catchments that show varying influences by the fortification ramparts. The tributaries reaching the receiving stream close to the central settlement area source close to the gates in the ramparts in the Late Bronze Age built-up areas. Additionally, also the geometry of these tributaries differs from that of other tributaries. The distinct character of the channel network with repeatedly occurring rectangular bends indicates the capture of channels, which developed as gullies along paths by retrogressive erosion.
Eine Erkrankung wird als monogen bezeichnet, wenn sie auf einen Gendefekt eines einzelnen Gens zurückzuführen ist. Durch einen angeborenen Gendefekt kann bei den sog. primären Immundefekten (PIDs) das Immunsystem von asymptomatisch bis lebensbedrohlich mehr oder weniger stark beeinträchtigt werden. Für lebensbedrohliche Immundefekte gilt die allogene Stammzelltransplantation eines passenden Spenders als einzig kurative Therapie. Weil jedoch für etwa 30 % aller Patienten kein passender Spender verfügbar ist, bietet die Gentherapie in Kombination mit einer autologen Stammzelltransplantation eine häufig lebensrettende Alternative. Dabei werden patienteneigene CD34+-Blutstammzellen isoliert, ex vivo mit einer funktionalen Kopie des defekten Gens genetisch modifiziert und anschließend zurück in den Patienten infundiert. Die dabei eingesetzten Genfähren basieren in der Regel auf viralen Vektoren, mit denen das gesunde Gen in die Patientenzellen eingeschleust wird. Retrovirale Vektoren wurden für die Gentherapie am häufigsten eingesetzt.
In mehreren klinischen Gentherapie-Studien zur Behandlung diverser PIDs kam es aufgrund insertionsbedingter Transaktivierung benachbarter Proto-Onkogene zur Leukämieentwicklung. Deswegen wurde gezielt an der Sicherheit retroviraler Genfähren gearbeitet. Insbesondere wurden die in der ersten Generation benutzten retroviralen Promotor/Enhancer-Elemente aus der U3-Region des 5’ LTRs deletiert (self-inactivating, SIN-Vektoren) und durch interne, gewebespezifische Promotoren ersetzt. Auf Genfallen basierende Vektoren (gene trap, GT-Vektoren) könnten eine sicherere Alternative zu den Standardvektoren bieten, weil sie zum einen auf den γ-retroviralen SIN-Vektoren basieren und zum anderen keinen internen Promotor enthalten, der zur Transaktivierung benachbarter Gene führen kann. Bei GT-Vektoren wird das integrierte Transgen von endogenen Promotoren kontrolliert, was zu einer robusteren Transgenexpression und zu einem erhöhten Sicherheitsprofil führen sollte.
Ziel dieser Arbeit war, GT-Vektoren hinsichtlich ihres Potentials als Vektoren für die Gentherapie zu bewerten. Dafür wurde zunächst die Gentransduktionseffizienz unterschiedlicher GT-Vektoren in murinen, embryonalen Stammzellen (mES-Zellen) untersucht. In einem klassischen GT-Vektor ist das Therapiegen von einem 5‘ liegenden Spleißakzeptor (SA) und einer 3‘ liegenden Polyadenylierungssequenz (pA) flankiert. Dies bewirkt, dass das Therapiegen nach Integration in ein exprimiertes Gen als Fusionstranskript mit den 5‘ liegenden endogenen Genfragmenten exprimiert wird. Sind diese kodierend, entsteht ein Fusionsprotein, das die Funktionalität des Therapiegens beeinträchtigen kann. Zu Vermeidung einer derartigen Konstellation wurden drei Strategien zur Verhinderung N-terminaler Fusionen getestet: Die Fusion (i) einer internen ribosomalen Eintrittsstelle (IRES) und (ii) eines viralen Proteinspaltungspeptids (T2A) an das 5‘ Ende des Therapiegens sowie (iii) die Insertion von drei Stop-Codons hinter den SA. Die Versuche in mES-Zellen zeigten, dass die GT-Variante mit den Stop-Codons am effizientesten war, weshalb sie für alle weiteren Ansätze verwendet wurde.
Dieser Arbeit vorangegangen war die Entwicklung einer Genfallenstrategie zur Korrektur des septischen Granulomatose (X-CGD) verursachenden gp91phox (CYBB)-Gendefektes in einer gp91phox-defizienten Leukämiezelllinie (PLB-XCGD). Obwohl Genfallen transduzierte PLB-XCGD-Zellen das Therapiegen gp91phox exprimierten, war diese Expression im Vergleich zu den mit einem positiven, Promotor-enthaltenden Kontrollvektor (FES-gp91phox) transduzierten Zellen sehr gering. Deswegen war es notwendig eine effiziente Selektionsstrategie für Genfallenereignisse in hämatopoetischen Zellen zu entwickeln. Eine Strategie basierte auf einem FKBP12/Thrombopoetinrezeptor-Fusionsprotein, dessen Expression in hämatopoetischen BaF3 Zellen eine 25-fache Anreicherung von Genfallen exprimierenden Zellen nach Zugabe des chemischen Liganden AP20187 ermöglichte. Allerdings konnte dieses System in primären, hämatopoetischen Zellen leider nicht etabliert werden.
Die andere Selektionsstrategie basierte auf dem X-SCID Krankheitsmodell, in dem IL2RG-Mutationen, einer Untereinheit verschiedenster Zytokinrezeptoren (z. B. des IL-2-Rezeptors), zu einem kompletten Verlust von T-Zellen und somit zur Reduktion funktionaler B-Zellen führen. Nach ex vivo Korrektur und Transplantation der korrigierten, autologen hämatopoetischen Stammzellen (HSZ), kann eine Expansion der IL2RG exprimierenden T-Zellen erzielt werden. Initiale Versuche wurden an der IL2RG-/--Zelllinie ED-7R in vitro durchgeführt. Nachdem über die durchflusszytometrische Analyse der pSTAT5-Expression eine Aktivierung des IL2RG-abhängigen Signalweges in GT-IL2RG-Genfallen transduzierten ED-7R-Zellen nachgewiesen werden konnte, wurde in einem X-SCID-Mausmodell (IL2RG-/-) überprüft, ob es zu der erwarteten Anreicherung von IL2RG exprimierenden T-Zellen nach Transplantation autologer GT-IL2RG transduzierter HSZ kommt. Dabei wurde sowohl die immunologische Rekonstitution der Mäuse als auch die Funktionalität der rekonstituierten Lymphozyten untersucht. In der GT-Gruppe konnte nach Transplantation genetisch modifizierter Zellen weder ein Unterschied der absoluten Zahl an Lymphozyten (B-Zellen, T-Zellen) im Blut, noch ein erhöhter Prozentsatz der verschiedenen Lymphozyten-subpopulationen in KM, Milz oder Thymus beobachtet werden. Lediglich im Thymus einer Maus aus der GT-Gruppe konnten IL2RG exprimierende Zellen nachgewiesen werden. Andererseits konnten aus der Milz transplantierter GT-Mäuse T-Zellen isoliert werden, die nach Interleukin-2-Stimulation STAT5-Phosphorylierung aufwiesen, was eine erfolgreiche obgleich geringe GT-IL2RG Transduktion belegt. Durch die Beurteilung des Engraftments, also des Anwachsens der transplantierten Spenderzellen im Empfängerorganismus, konnte gezeigt werden, dass die niedrigere IL2RG-Rekonstitutionseffizienz durch Genfallen nicht auf einem suboptimalen Engraftment, sondern auf einer zu geringen Anzahl an produktiven Genfallenereignissen beruht.
Zusammenfassend legen die Ergebnisse nahe, dass Genfallen zu diesem Zeitpunkt keine realistische Alternative gegenüber konventionellen Gentherapievektoren zur Korrektur monogener Bluterkrankungen bieten. Neue Entwicklungen, die eine Genkorrektur mittels sog. „Designer Endonukleasen“ vor Ort ermöglichen, werden sicherlich in der nahen Zukunft sämtliche, beliebig ins Genom integrierende Gentherapievektoren ersetzen.

Plant communities provide floral resource-landscapes for pollinators. Yet, it is insufficiently understood how these landscapes shape pollinator-mediated interactions among multiple plant species. Here, we study how pollinators and the seed set of plants respond to the distribution of a floral resource (nectar sugar) in space and across plant species, inflorescences and flowering phenologies. In a global biodiversity hotspot, we quantified floral resource-landscapes on 27 sites of 4 ha comprising 127,993 shrubs of 19 species. Visitation rates of key bird pollinators strongly depended on the phenology of site-scale resource amounts. Seed set of focal plants increased with resources of conspecific neighbours and with site-scale resources, notably with heterospecific resources of lower quality (less sugar per inflorescence). Floral resources are thus a common currency determining how multiple plant species interact via pollinators. These interactions may alter conditions for species coexistence in plant communities and cause community-level Allee effects that promote extinction cascades.
Die hier vorliegende Dissertation befasst sich mit der Synthese von Naturstoffen aus Xenorhabdus und Photorhabdus spp. Da 6,0 - 7,5% ihres Genoms Sekundärmetabolit Clustern zuzuordnen sind, gelten diese entomopathogenen Bakterien als vielversprechende Naturstoffproduzenten. Die Palette der von ihnen produzierten Naturstoffe reicht von Antibiotika über Insektizide bis hin zu potentiellen Zytostatika. Die im Rahmen dieser Arbeit synthetisierten und charakterisierten Substanzen lassen sich in vier Kategorien einteilen: kleine Sekundärmetabolite (Phurealipide), zyklische Makrolaktame (Xenotetrapeptide, GameXPeptide und Ambactin), zyklische Makrolaktone (Szentiamide, Xentrivalpeptide und Xenephematide) und methylierte lineare Peptide (Rhabdopeptide und Rhabdopeptid-ähnliche Moleküle).
In diesem Artikel wird am Beispiel des Fremdsprachenlernens der wissenschaftlich noch wenig bearbeitete Zusammenhang zwischen Identität und Lernen thematisiert. Annahme ist, dass trotz des bildungspolitischen Appells zum Lebenslangen Lernen das Lernen im Lebenslauf nicht kontinuierlich, sondern diskontinuierlich ist. Es wird die Frage gestellt, wie die Lernenden mit diesen Diskontinuitäten umgehen. Die Wahrnehmung, Beschreibung und Ordnung dieser Diskontinuitäten ist – so die These des Beitrags – abhängig vom Selbstbild als Lerner bzw. von dessen Identität. Drei Formen werden empirisch herausgearbeitet: die Identität als heroische(r) Lerner(in), die Identität als selbstverständliche(r) Lerner(in) und die Identität als potenzielle(r) Lerner(in). In allen drei Formen, die keineswegs ausschließend sind, sondern sich individuell im Lebenslauf abwechseln können und zugleich zeitbezogen unterschiedliche Verbindungen eingehen können, wird Kontinuität als lebenslanger Lerner her- bzw. dargestellt; und dies auch dann, wenn das Lernen diskontinuierlich verläuft.

Dieser Leitfaden ist im Rahmen des Projektes "IndUK – Individuelles Umwelthandeln und Klimaschutz – Ergebnisintegration und transdisziplinäre Verwertung von Erkenntnissen aus der SÖF-Forschung zu den sozialen Dimensionen von Klimaschutz und Klimawandel" entstanden. The Liikanen Group proposes contingent convertible (CoCo) bonds as a potential mechanism to enhance financial stability in the banking industry. Especially life insurance companies could serve as CoCo bond holders as they are already the largest purchasers of bank bonds in Europe. We develop a stylized model with a direct financial connection between banking and insurance and study the effects of various types of bonds such as non-convertible bonds, write-down bonds and CoCos on banks' and insurers' risk situations. In addition, we compare insurers' capital requirements under the proposed Solvency II standard model as well as under an internal model that ex-ante anticipates additional risks due to possible conversion of the CoCo bond into bank shares. In order to check the robustness of our findings, we consider different CoCo designs (write-down factor, trigger value, holding time of bank shares) and compare the resulting capital requirements with those for holding non-convertible bonds. We identify situations in which insurers benefit from buying CoCo bonds due to lower capital requirements and higher coupon rates. Furthermore, our results highlight how the Solvency II standard model can mislead insurers in their CoCo investment decision due to economically irrational incentives.
I assess how Basel III, Solvency II and the low interest rate environment will affect the financial connection between the bank and insurance sector by changing the funding patterns of banks as well as the investment strategies of life insurance companies. Especially for life insurance companies, the current low interest rate environment poses a key risk since declining returns on investments jeopardize the guaranteed return on life insurance contracts, a core component of traditional life insurance contracts in several European countries. I consider a contingent claim framework with a direct financial connection between banks and life insurers via bank bonds. The results indicate that life insurers' demand for bank bonds increases over the mid-term but ultimately declines in the long-run. Since life insurers are the largest purchasers of bank bonds in Europe, banks could lose one of their main funding sources. In addition, I show that shareholder value driven life insurers' appetite for risk increases when the gap between asset return and liability growth diminishes. To check the robustness of the findings, I calibrate a prolonged low interest rate scenario. The results show that the insurer's risk appetite is even higher when interest rates remain persistently low. A sensitivity analysis regarding industry-specific regulatory safety levels reveals that contagion between bank and life insurer is driven by the insurers' demand for bank bonds which itself depends on the regulatory safety level of banks.
Um nachvollziehen zu können, in welchem forschungspraktischen Kontext diese Studie steht, ist Folgendes zu erläutern: Nachdem Anfang der 2000er Jahre die ersten Schulen ihre Schulprogramme vorgelegt hatten, führte eine Forschungsgruppe unter der Leitung von Prof. Dr. Gruschka ein DFG-Projekt durch, dessen erste Ergebnisse im Jahr 2003 in den „Frankfurter Beiträgen zur Erziehungswissenschaft“ veröffentlicht wurden. Das von wissenschaftlicher Seite unbestätigte Postulat, Reformen könnten durch Schulprogrammarbeit initiiert werden, wurde in dieser Studie einer sachhaltigen Überprüfung unterzogen. Die Ausgangsfrage lautete: In welcher Weise bewirkt eine Kriseninduktion in Form administeriell verordneter Schulprogrammarbeit einen Prozess der inneren Schulreform? Um auf diese Frage eine wissenschaftlich begründete Antwort zu erhalten, wurden nicht nur Schulprogramme einzelner Schulen untersucht, sondern ebenso die von administerieller Seite angewandten Mittel, mit denen man die Krise zu induzieren gedachte. Zu zeigen war, in welcher in-haltlichen und formalen Weise sowohl die Kultusbürokratie wie auch die einzelnen Schulaufsichtsbehörden das Vorhaben begriffen und den Schulen vermittelten. Es ging in dieser Studie demnach nicht nur um das Schulprogramm selbst, sondern auch um den Induktionsprozess bzw. die einzelnen Vermittlungsinstanzen der Krisenin-duktion. Die letzte Instanz dieser Kette (bestehend aus Kultusbürokratie, Schulaufsichtsbehörde und Schulleiter) waren die Lehrer, die ja im Hinblick auf die Anfertigung des Schulprogramms die zentralen Akteure waren (bzw. zu sein hatten). Der zentrale Gegenstand dieser Studie – die Interviews mit den an der Schulprogrammarbeit beteiligten Lehrern – wurde im Rahmen dieses Forschungsprojekts erhoben. Die wissenschaftliche Rekonstruktion der Interviews ist also als eine Teilstudie des um-fassenderen DFG-Forschungsprojekts einzuordnen. Cloud microphysical processes involving the ice phase in tropospheric clouds are among the major uncertainties in cloud formation, weather and General Circulation Models (GCMs). The simultaneous detection of aerosol particles, liquid droplets, and ice crystals, especially in the small cloud-particle size range below 50 µm, remains challenging in mixed phase, often unstable ice-water phase environments. The Cloud Aerosol Spectrometer with Polarisation (CASPOL) is an airborne instrument that has the ability to detect such small cloud particles and measure their effects on the backscatter polarisation state. Here we operate the versatile Cosmics-Leaving- OUtdoor-Droplets (CLOUD) chamber facility at the European Organisation for Nuclear Research (CERN) to produce controlled mixed phase and other clouds by adiabatic expansions in an ultraclean environment, and use the CASPOL to discriminate between different aerosols, water and ice particles. In this paper, optical property measurements of mixed phase clouds and viscous Secondary Organic Aerosol (SOA) are presented. We report observations of significant liquid – viscous SOA particle polarisation transitions under dry conditions using CASPOL. Cluster analysis techniques were subsequently used to classify different types of particles according to their polarisation ratios during phase transition. A classification map is presented for water droplets, organic aerosol (e.g., SOA and oxalic acid), crystalline substances such as ammonium sulphate, and volcanic ash. Finally, we discuss the benefits and limitations of this classi- fication approach for atmospherically relevant concentration and mixtures with respect to the CLOUD 8–9 campaigns and its potential contribution to Tropical Troposphere Layer (TTL) analysis.
Background and aims: Clinical trials of therapy against chronic hepatitis C virus (HCV) infection including boceprevir (BOC) or telaprevir (TVR) plus pegylated interferon and ribavirin (PR) have reported considerably higher response rates than those achieved with PR alone. This study sought to evaluate the efficacy and safety of triple therapy including BOC or TVR in combination with PR in HIV/HCV-coinfected patients under real-life conditions.
Methods: In a multicentre study conducted in 24 sites throughout five European countries, all HIV/HCV-coinfected patients who initiated a combination of BOC or TVR plus PR and who had at least 60 weeks of follow-up, were analyzed. Sustained virologic response 12 weeks after the scheduled end of therapy date (SVR12) and the rate of discontinuations due to adverse events (AE) were evaluated.
Results: Of the 159 subjects included, 127 (79.9%) were male, 45 (34.4%) were treatment-naïve for PR and 60 (45.4%) showed cirrhosis. SVR12 was observed in 31/46 (67.4%) patients treated with BOC and 69/113 (61.1%) patients treated with TVR. Overall discontinuations due to AE rates were 8.7% for BOC and 8% for TVR. Grade 3 or 4 hematological abnormalities were frequently observed; anemia 7%, thrombocytopenia 17.2% and neutropenia 16.4%.
Conclusion: The efficacy and safety of triple therapy including BOC or TVR plus PR under real-life conditions of use in the HIV/HCV-coinfected population was similar to what is observed in clinical trials. Hematological side effects are frequent but manageable.
Multiple antenatal dexamethasone treatment alters brain vessel differentiation in newborn mouse pups

Antenatal steroid treatment decreases morbidity and mortality in premature infants through the maturation of lung tissue, which enables sufficient breathing performance. However, clinical and animal studies have shown that repeated doses of glucocorticoids such as dexamethasone and betamethasone lead to long-term adverse effects on brain development. Therefore, we established a mouse model for antenatal dexamethasone treatment to investigate the effects of dexamethasone on brain vessel differentiation towards the blood-brain barrier (BBB) phenotype, focusing on molecular marker analysis. The major findings were that in total brains on postnatal day (PN) 4 triple antenatal dexamethasone treatment significantly downregulated the tight junction protein claudin-5, the endothelial marker Pecam-1/CD31, the glucocorticoid receptor, the NR1 subunit of the N-methyl-D-aspartate receptor, and Abc transporters (Abcb1a, Abcg2 Abcc4). Less pronounced effects were found after single antenatal dexamethasone treatment and in PN10 samples. Comparisons of total brain samples with isolated brain endothelial cells together with the stainings for Pecam-1/CD31 and claudin-5 led to the assumption that the morphology of brain vessels is affected by antenatal dexamethasone treatment at PN4. On the mRNA level markers for angiogenesis, the sonic hedgehog and the Wnt pathway were downregulated in PN4 samples, suggesting fundamental changes in brain vascularization and/or differentiation. In conclusion, we provided a first comprehensive molecular basis for the adverse effects of multiple antenatal dexamethasone treatment on brain vessel differentiation.
Partial cross sections of the 89Y(p, γ )90Zr reaction have been measured to investigate the γ-ray strength function in the neutron–magic nucleus 90Zr. For five proton energies between E p = 3.65 MeV and E p = 4.70 MeV partial cross sections for the population of seven discrete states in 90Zr have been determined by means of in-beam γ-ray spectroscopy. Since these γ-ray transitions are dominantly of E1 character, the present measurement allows an access to the low-lying dipole strength in 90Zr. A γ-ray strength function based on the experimental data could be extracted, which is used to describe the total and partial cross sections of this reaction by Hauser–Feshbach calculations successfully. Significant differences with respect to previously measured strength functions from photoabsorption data point towards deviations from the Brink–Axel hypothesis relating the photo-excitation and de-excitation strength functions.
Modulation der Zytokinproduktion durch Interaktionen von IRAK4 und p47phox mit den TLR-Signalwegen

p47phox wird als ein in mehreren Strukturen involviertes Protein beschrieben, welches in seiner Funktion einen großen Einfluss auf das Immunsystem nehmen kann und damit zu schweren Erkrankungen führen kann. Wegen der Vielfalt der Interaktionen ist es schwierig die Effekte von p47phox einer bestimmten Wechselwirkung zuzuordnen. Dennoch ist es unserer Arbeitsgruppe gelungen eine negative Rückkopplung von p47phox auf die Produktion von IL-12p70 [1] festzustellen. Ausgehend von dieser Interaktion und der Phosphorylierung von p47phox durch IRAK4 [2], wurde daher in dieser Doktorarbeit der Zusammenhang einer Modulation der IL-12p70-Produktion durch IRAK4 näher untersucht.
Herausforderungen für die nationale, regionale und thematische Webarchivierung und deren Nutzung

Das World Wide Web ist als weltweites Informations- und Kommunikationsmedium etabliert. Neue Technologien erweitern regelmäßig die Nutzungsformen und erlauben es auch unerfahrenen Nutzern, Inhalte zu publizieren oder an Diskussionen teilzunehmen. Daher wird das Web auch als eine gute Dokumentation der heutigen Gesellschaft angesehen. Aufgrund seiner Dynamik sind die Inhalte des Web vergänglich und neue Technologien und Nutzungsformen stellen regelmäßig neue Herausforderungen an die Sammlung von Webinhalten für die Webarchivierung. Dominierten in den Anfangstagen der Webarchivierung noch statische Seiten, so hat man es heute häufig mit dynamisch generierten Inhalten zu tun, die Informationen aus verschiedenen Quellen integrieren. Neben dem klassischen domainorientieren Webharvesting kann auch ein steigendes Interesse aus verschiedenen Forschungsdisziplinen an thematischen Webkollektionen und deren Nutzung und Exploration beobachtet werden. In diesem Artikel werden einige Herausforderungen und Lösungsansätze für die Sammlung von thematischen und dynamischen Inhalten aus dem Web und den sozialen Medien vorgestellt. Des Weiteren werden aktuelle Probleme der wissenschaftlichen Nutzung diskutiert und gezeigt, wie Webarchive und andere temporale Kollektionen besser durchsucht werden können.
Seit einiger Zeit wird die Bedeutung der Kultur auch für die wirtschaftliche Entwicklung diskutiert. Zusammenhänge zwischen Kultur und Wirtschaft werden auch auf regionaler Ebene zunehmend thematisiert. Hintergrund für dieses regionale Interesse dürfte sein, dass Regionen trotz oder wegen der Globalisierung eherzunehmende Bedeutung haben. Nicht nur ökonomische,sondern auch nicht‐ökonomische Standort‐ und Wettbewerbsfaktoren sind danach für die regionale Wirtschaft und Wettbewerbsfähigkeit relevant; Kultur ist einer davon. Mit dieser Studie möchten wir einen Diskussionsbeitrag leisten, die Bedeutung (ausgewählter) regionaler Kulturvariabler für die Wirtschaftsentwicklung und Wettbewerbsfähigkeit exemplarisch für die beiden Metropolregionen FrankfurtRheinMain und Stuttgart quantitativ zu eruieren. Das erkenntnisleitende Interesse richtetsich auf eine erste Beantwortung folgender Fragen:
* Welche quantitative Bedeutung haben die (ausgewählten) Kulturvariablen für die regionale Wirtschaftsentwicklung und Wettbewerbsfähigkeit?
* Bestehen innerhalb einer Region Unterschiede bei der quantitativen Bedeutung dieser Kulturvariablen?
* Bestehen zwischen den beiden Metropolregionen Unterschiede in dieser Bedeutung? ...
The extracellular matrix is rapidly emerging as a prominent contributor to various fundamental processes of tumorigenesis. In particular, decorin, a member of the small leucine-rich proteoglycan gene family, is assuming a central role as a potent soluble tumor repressor. Decorin binds and antagonizes various receptor tyrosine kinases and inhibits downstream oncogenic signaling in several solid tumors. Among other functions, decorin evokes cell cycle arrest, apoptosis, and antimetastatic, and antiangiogenic programs. Recent work has revealed a paradigmatic shift in our understanding of the molecular mechanisms underlying its tumoricidal properties. Decorin adversely compromises the genetic signature of the tumor microenvironment and induces endothelial cell autophagy downstream of VEGFR2. Moreover, decorin selectively evokes destruction of tumor cell mitochondria downstream of Met through mitophagy. Acting as a partial agonist, decorin signals via proautophagic receptors and triggers procatabolic processes that parallel the classical tumoricidal properties of this multifaceted proteoglycan.
BACKGROUND: Plasminogen deficiency is a rare autosomal recessive disease, which is associated with aggressive periodontitis and gingival enlargement. Previously described treatments of plasminogen deficiency associated periodontitis have shown limited success. This is the first case report indicating a successful therapy approach consisting of a non-surgical supra- and subgingival debridement in combination with an adjunctive systemic antibiotic therapy and a strict supportive periodontal regimen over an observation period of 4 years.
CASE PRESENTATION: The intraoral examination of a 17-year-old Turkish female with severe plasminogen deficiency revealed generalized increased pocket probing depths ranging from 6 to 9 mm, bleeding on probing over 30%, generalized tooth mobility, and gingival hyperplasia. Alveolar bone loss ranged from 30% to 50%. Clinical attachment loss corresponded to pocket probing depths. Aggregatibacter actinomycetemcomitans, Porphyromonas gingivalis, Treponema denticola, Prevotella intermedia, Prevotella nigrescens and Eikenella corrodens have been detected by realtime polymerase chain reaction. Periodontal treatment consisted of full mouth disinfection and adjunctive systemic administration of amoxicillin (500 mg tid) and metronidazole (400 mg tid). A strict supportive periodontal therapy regimen every three month in terms of supra- and subgingival debridement was rendered. The reported therapy has significantly improved periodontal health and arrested disease progression. Intraoral examination at the end of the observation period 3.5 years after non-surgical periodontal therapy showed generalized decreased pocket probing depths ranging from 1 to 6 mm, bleeding on probing lower 30%, and tooth mobility class I and II. Furthermore, microbiological analysis shows the absence of Porphyromonas gingivalis, Prevotella intermedia and Treponema denticola after therapy.
CONCLUSION: Adjunctive antibiotic treatment may alter the oral microbiome and thus, the inflammatory response of periodontal disease associated to plasminogen deficiency and diminishes the risk of pseudomembrane formation and progressive attachment loss. This case report indicates that patients with plasminogen deficiency may benefit from non-surgical periodontal treatment in combination with an adjunctive antibiotic therapy and a strict supportive periodontal therapy regimen.
A fundamental question in evolutionary genetics concerns the extent to which adaptive phenotypic convergence is attributable to convergent or parallel changes at the molecular sequence level. Here we report a comparative analysis of hemoglobin (Hb) function in eight phylogenetically replicated pairs of high- and low-altitude waterfowl taxa to test for convergence in the oxygenation properties of Hb, and to assess the extent to which convergence in biochemical phenotype is attributable to repeated amino acid replacements. Functional experiments on native Hb variants and protein engineering experiments based on site-directed mutagenesis revealed the phenotypic effects of specific amino acid replacements that were responsible for convergent increases in Hb-O2 affinity in multiple high-altitude taxa. In six of the eight taxon pairs, high-altitude taxa evolved derived increases in Hb-O2 affinity that were caused by a combination of unique replacements, parallel replacements (involving identical-by-state variants with independent mutational origins in different lineages), and collateral replacements (involving shared, identical-by-descent variants derived via introgressive hybridization). In genome scans of nucleotide differentiation involving high- and low-altitude populations of three separate species, function-altering amino acid polymorphisms in the globin genes emerged as highly significant outliers, providing independent evidence for adaptive divergence in Hb function. The experimental results demonstrate that convergent changes in protein function can occur through multiple historical paths, and can involve multiple possible mutations. Most cases of convergence in Hb function did not involve parallel substitutions and most parallel substitutions did not affect Hb-O2 affinity, indicating that the repeatability of phenotypic evolution does not require parallelism at the molecular level.
Macrophages respond to the Th2 cytokine IL-4 with elevated expression of arachidonate 15-lipoxygenase (ALOX15). Although IL-4 signaling elicits anti-inflammatory responses, 15-lipoxygenase may either support or inhibit inflammatory processes in a context-dependent manner. AMP-activated protein kinase (AMPK) is a metabolic sensor/regulator that supports an anti-inflammatory macrophage phenotype. How AMPK activation is linked to IL-4-elicited gene signatures remains unexplored. Using primary human macrophages stimulated with IL-4, we observed elevated ALOX15 mRNA and protein expression, which was attenuated by AMPK activation. AMPK activators, e.g. phenformin and aminoimidazole-4-carboxamide 1-β-d-ribofuranoside inhibited IL-4-evoked activation of STAT3 while leaving activation of STAT6 and induction of typical IL-4-responsive genes intact. In addition, phenformin prevented IL-4-induced association of STAT6 and Lys-9 acetylation of histone H3 at the ALOX15 promoter. Activating AMPK abolished cellular production of 15-lipoxygenase arachidonic acid metabolites in IL-4-stimulated macrophages, which was mimicked by ALOX15 knockdown. Finally, pretreatment of macrophages with IL-4 for 48 h increased the mRNA expression of the proinflammatory cytokines IL-6, IL-12, CXCL9, and CXCL10 induced by subsequent stimulation with lipopolysaccharide. This response was attenuated by inhibition of ALOX15 or activation of AMPK during incubation with IL-4. In conclusion, limiting ALOX15 expression by AMPK may promote an anti-inflammatory phenotype of IL-4-stimulated human macrophages.
Previous studies used a text-fading procedure as a training tool with the goal to increase silent reading fluency (i.e., proficient reading rate and comprehension). In recently published studies, this procedure resulted in lasting reading enhancements for adult and adolescent research samples. However, studies working with children reported mixed results. While reading rate improvements were observable for Dutch reading children in a text-fading training study, reading fluency improvements in standardized reading tests post-training attributable to the fading manipulation were not detectable. These results raise the question of whether text-fading training is not effective for children or whether research design issues have concealed possible transfer effects. Hence, the present study sought to investigate possible transfer effects resulting from a text-fading based reading training program, using a modified research design. Over a period of 3 weeks, two groups of German third-graders read sentences either with an adaptive text-fading procedure or at their self-paced reading rate. A standardized test measuring reading fluency at the word, sentence, and text level was conducted pre- and post-training. Text level reading fluency improved for both groups equally. Post-training gains at the word level were found for the text-fading group, however, no significant interaction between groups was revealed for word reading fluency. Sentence level reading fluency gains were found for the text-fading group, which significantly differed from the group of children reading at their self-paced reading routine. These findings provide evidence for the efficacy of text-fading as a training method for sentence reading fluency improvement also for children.
Epigenetic silencing of transgene expression represents a major obstacle for the efficient genetic modification of multipotent and pluripotent stem cells. We and others have demonstrated that a 1.5 kb methylation-free CpG island from the human HNRPA2B1-CBX3 housekeeping genes (A2UCOE) effectively prevents transgene silencing and variegation in cell lines, multipotent and pluripotent stem cells, and their differentiated progeny. However, the bidirectional promoter activity of this element may disturb expression of neighboring genes. Furthermore, the epigenetic basis underlying the anti-silencing effect of the UCOE on juxtaposed promoters has been only partially explored. In this study we removed the HNRPA2B1 moiety from the A2UCOE and demonstrate efficient anti-silencing properties also for a minimal 0.7 kb element containing merely the CBX3 promoter. This DNA element largely prevents silencing of viral and tissue-specific promoters in multipotent and pluripotent stem cells. The protective activity of CBX3 was associated with reduced promoter CpG-methylation, decreased levels of repressive and increased levels of active histone marks. Moreover, the anti-silencing effect of CBX3 was locally restricted and when linked to tissue-specific promoters did not activate transcription in off target cells. Thus, CBX3 is a highly attractive element for sustained, tissue-specific and copy-number dependent transgene expression in vitro and in vivo.
Augmenting LTP-like plasticity in human motor cortex by spaced paired associative stimulation

Paired associative stimulation (PASLTP) of the human primary motor cortex (M1) can induce LTP-like plasticity by increasing corticospinal excitability beyond the stimulation period. Previous studies showed that two consecutive PASLTP protocols interact by homeostatic metaplasticity, but animal experiments provided evidence that LTP can be augmented by repeated stimulation protocols spaced by ~30min. Here we tested in twelve healthy selected PASLTP responders the possibility that LTP-like plasticity can be augmented in the human M1 by systematically varying the interval between two consecutive PASLTP protocols. The first PASLTP protocol (PAS1) induced strong LTP-like plasticity lasting for 30-60min. The effect of a second identical PASLTP protocol (PAS2) critically depended on the time between PAS1 and PAS2. At 10min, PAS2 prolonged the PAS1-induced LTP-like plasticity. At 30min, PAS2 augmented the LTP-like plasticity induced by PAS1, by increasing both magnitude and duration. At 60min and 180min, PAS2 had no effect on corticospinal excitability. The cumulative LTP-like plasticity after PAS1 and PAS2 at 30min exceeded significantly the effect of PAS1 alone, and the cumulative PAS1 and PAS2 effects at 60min and 180min. In summary, consecutive PASLTP protocols interact in human M1 in a time-dependent manner. If spaced by 30min, two consecutive PASLTP sessions can augment LTP-like plasticity in human M1. Findings may inspire further research on optimized therapeutic applications of non-invasive brain stimulation in neurological and psychiatric diseases.
The transport of air masses originating from the Asian monsoon anticyclone into the extratropical upper troposphere and lower stratosphere (Ex-UTLS) above potential temperatures Θ = 380K was identified during the HALO aircraft mission TACTS in August and September 2012. In-situ measurements of CO, O3 and N2O during TACTS Flight 2 on the 30 August 2012 show the irreversible mixing of aged with younger (originating from the troposphere) stratospheric air masses within the Ex-UTLS. Backward trajectories calculated with the trajetory module of the CLaMS model indicate that these tropospherically affected air masses originate from the Asian monsoon anticyclone. From the monsoon circulation region these air masses are quasi-isentropically transported above Θ = 380 K into the Ex-UTLS where they subsequently mix with stratospheric air masses. The overall trace gas distribution measured during TACTS shows that this transport pathway has a significant impact on the Ex-UTLS during boreal summer and autumn. This leads to an intensification of the tropospheric influence on the Ex-UTLS with ∆Θ > 30 K (relative to the tropopause) within three weeks during the TACTS mission. In the same time period a weakening of the tropospheric influence on the lowermost stratosphere (LMS) is determined. Therefore, the study shows that the transport of air masses originating from the Asian summer monsoon region within the lower stratosphere above Θ = 380K is of major importance for the change of the chemical composition of the Ex-UTLS from summer to autumn.
Die Debatte, wie die Organisation des universitären Lehrbetriebs an deutschen Hochschulen verbessert werden kann, wurde in den vergangenen Monaten intensiv medial begleitet. Eine ehrliche Diskussion über realistisch umsetzbare Reformen ist notwendig. Ich plädiere vor allem für eine Diversifizierung der Prüfungsleistungen, wodurch sich gleichzeitig die Vorbereitung auf Seminare verbessern könnte. Darüber hinaus wäre ein konsensorientierteres Verhältnis zwischen Dozenten_innen, Administration und Studentenschaft wünschenswert.

Nein, in vorliegender Form hätte diese Doktorarbeit nie veröffentlicht werden dürfen, weist sie doch, hochgerechnet, eine sicherlich im vierstelligen Bereich liegende Zahl an Fehlern, Nachlässigkeiten, ja Schlampereien auf. Ein Leser, der sich bis zum bitteren Ende durchgekämpft hat, dürfte fassungslos auf einen Kampf zurückblicken, den der Autor seinerseits mit der, nein: gegen die deutsche Sprache führte, um dabei an elementaren Regeln der Grammatik, Zeichensetzung und bisweilen sogar an der Orthografie zu scheitern. A. Willershausen ist mithin nicht nur dem Genetiv und Dativ, sondern der deutschen Sprache überhaupt feind. Ähnlich sieht’s im Lateinischen und Französischen aus; oft sind nicht einmal gedruckte Texte korrekt wiedergegeben. Und ganz schlimm wird’s, wenn Deutsches und Französisches zusammenstoßen: "Vermittlung Gui de Boulognes […] Verhandlungen Talleyrands de Périgord auf der Poitier-Kampagnes" (S. 81, Anm. 349) ‒ "Sainte-Maria-Madelaine" (S. 196). Diese zwei Beispiele müssen hierfür aus Platzgründen genügen; generell gilt, dass mein Monita-Leporello sehr, sehr lang ist und bei Bedarf eingesehen werden kann. Doch jedem, der sich der Mühe der Lektüre nur eines einzigen Kapitels unterzieht, dürften die vielen Fehlleistungen ohnehin auffallen. Die Verantwortung für all das liegt natürlich beim Autor, aber sicher sind hier auch kritische Fragen an den Doktorvater und den Zweitgutachter sowie an einen Verlag zu stellen, der die Vorlage offensichtlich unbesehen, geschweige denn lektoriert zum Druck freigab. ...

Nachdem der 26-jährige Ahmad Tarmimi Maliki im Mai 2014 ein mit einer Tonne Sprengstoff beladenes Fahrzeug in ein irakisches Militärgebäude steuerte und 25 Soldaten einer Elite-Einheit mit in den Tod riss, wurde er in den einschlägigen sozialen Netzwerken Südostasiens von tausenden IS-Sympathisanten als „Märtyrer“ gefeiert. At the sensor level many aspects, such as spectral power, functional and effective connectivity as well as relative-power-ratio ratio (RPR) and spatial resolution have been comprehensively investigated through both electroencephalography (EEG) and magnetoencephalography (MEG). Despite this, differences between both modalities have not yet been systematically studied by direct comparison. It remains an open question as to whether the integration of EEG and MEG data would improve the information obtained from the above mentioned parameters. Here, EEG (64-channel system) and MEG (275 sensor system) were recorded simultaneously in conditions with eyes open (EO) and eyes closed (EC) in 29 healthy adults. Spectral power, functional and effective connectivity, RPR, and spatial resolution were analyzed at five different frequency bands (delta, theta, alpha, beta and gamma). Networks of functional and effective connectivity were described using a spatial filter approach called the dynamic imaging of coherent sources (DICS) followed by the renormalized partial directed coherence (RPDC). Absolute mean power at the sensor level was significantly higher in EEG than in MEG data in both EO and EC conditions. At the source level, there was a trend towards a better performance of the combined EEG+MEG analysis compared with separate EEG or MEG analyses for the source mean power, functional correlation, effective connectivity for both EO and EC. The network of coherent sources and the spatial resolution were similar for both the EEG and MEG data if they were analyzed separately. Results indicate that the combined approach has several advantages over the separate analyses of both EEG and MEG. Moreover, by a direct comparison of EEG and MEG, EEG was characterized by significantly higher values in all measured parameters in both sensor and source level. All the above conclusions are specific to the resting state task and the specific analysis used in this study to have general conclusion multi-center studies would be helpful.
Last November, the media organisation of the „Islamic State“ (IS) published a video, the sole purpose of which was to prove that the „caliphate“ which the IS has established in June 2014 was in fact a proper state. The video highlighted a host of institutions in order to drive home the claim of real statehood, including examples like a working judiciary, a prison administration, a schooling system, and so on. At one point in the video, the IS claimed that it was also financially independent and had apt resources at its disposal, namely oil and gas.
However, while it is true that the IS controls a number of oil and gas fields in Syria as well as in Iraq, we have by now enough evidence to be rather sure that the economic base of the „caliphate“ is by no means sustainable...
This paper argues that it is necessary to focus on gender rather than exclusively on women in discussions on global poverty eradication. It argues firstly, that the drivers of poverty are complex and multifaceted leading to a least two different forms of deprivation – transitory and structural poverty – each requiring different forms of analysis and treatment. Transitory poverty can arise as a consequence of an event or shock that would diminish an individual’s capacity to retain or secure employment and where a State lacks an appropriate form of social protection. Structural poverty, on the other hand, arises where groups are excluded from the workforce on a more permanent basis due to a wide variety of factors of discrimination such as sex, race, ethnicity, and age. Focusing on the sex of an individual alone cannot explain why some are more likely to experience different forms of poverty than others. Policies that protect women against transitory poverty, such as care related allowances, are not sufficient to eradicate structural poverty. Secondly, structural poverty prompts an examination of gender roles and relations. Unlike the category of ‘women’, the concept of gender demands consideration of a wider range of intersecting factors that influence life chances. The structure of contemporary gender relations, where women continue to experience higher levels of violence, and carry the greatest burden of responsibility for non-market based production activities, create the social conditions where domination and dependence thrive, and where persistently high rates of poverty seem inevitable. Such circumstances are generated by human agency. Thus, thirdly, it argues that these circumstances can and should be changed through human action. Knowledge of these circumstances gives rise to moral obligations for both men and women to avoid upholding values and practices that lead to domination and dependence as a matter of basic justice.
Models propose an auditory-motor mapping via a left-hemispheric dorsal speech-processing stream, yet its detailed contributions to speech perception and production are unclear. Using fMRI-navigated repetitive transcranial magnetic stimulation (rTMS), we virtually lesioned left dorsal stream components in healthy human subjects and probed the consequences on speech-related facilitation of articulatory motor cortex (M1) excitability, as indexed by increases in motor-evoked potential (MEP) amplitude of a lip muscle, and on speech processing performance in phonological tests. Speech-related MEP facilitation was disrupted by rTMS of the posterior superior temporal sulcus (pSTS), the sylvian parieto-temporal region (SPT), and by double-knock-out but not individual lesioning of pars opercularis of the inferior frontal gyrus (pIFG) and the dorsal premotor cortex (dPMC), and not by rTMS of the ventral speech-processing stream or an occipital control site. RTMS of the dorsal stream but not of the ventral stream or the occipital control site caused deficits specifically in the processing of fast transients of the acoustic speech signal. Performance of syllable and pseudoword repetition correlated with speech-related MEP facilitation, and this relation was abolished with rTMS of pSTS, SPT, and pIFG. Findings provide direct evidence that auditory-motor mapping in the left dorsal stream causes reliable and specific speech-related MEP facilitation in left articulatory M1. The left dorsal stream targets the articulatory M1 through pSTS and SPT constituting essential posterior input regions and parallel via frontal pathways through pIFG and dPMC. Finally, engagement of the left dorsal stream is necessary for processing of fast transients in the auditory signal.
Photorhabdus are highly effective insect pathogenic bacteria that exist in a mutualistic relationship with Heterorhabditid nematodes. Unlike other members of the genus, Photorhabdus asymbiotica can also infect humans. Most Photorhabdus cannot replicate above 34°C, limiting their host-range to poikilothermic invertebrates. In contrast, P. asymbiotica must necessarily be able to replicate at 37°C or above. Many well-studied mammalian pathogens use the elevated temperature of their host as a signal to regulate the necessary changes in gene expression required for infection. Here we use RNA-seq, proteomics and phenotype microarrays to examine temperature dependent differences in transcription, translation and phenotype of P. asymbiotica at 28°C versus 37°C, relevant to the insect or human hosts respectively. Our findings reveal relatively few temperature dependant differences in gene expression. There is however a striking difference in metabolism at 37°C, with a significant reduction in the range of carbon and nitrogen sources that otherwise support respiration at 28°C. We propose that the key adaptation that enables P. asymbiotica to infect humans is to aggressively acquire amino acids, peptides and other nutrients from the human host, employing a so called “nutritional virulence” strategy. This would simultaneously cripple the host immune response while providing nutrients sufficient for reproduction. This might explain the severity of ulcerated lesions observed in clinical cases of Photorhabdosis. Furthermore, while P. asymbiotica can invade mammalian cells they must also resist immediate killing by humoral immunity components in serum. We observed an increase in the production of the insect Phenol-oxidase inhibitor Rhabduscin normally deployed to inhibit the melanisation immune cascade. Crucially we demonstrated this molecule also facilitates protection against killing by the alternative human complement pathway.
Pharmakophore sind ein zentrales Konzept der medizinischen Chemie. Im Liganden-basierten Design abstrahieren sie physikochemische Eigenschaften einer Menge aktiver Liganden und lassen dadurch Rückschlüsse auf die möglichen Interaktionen mit einem Target zu. Umgekehrt werden im Struktur-basierten Design Kristallstrukturen von Proteinen genutzt um zu modellieren, welche Eigenschaften die Bindetasche besitzt und welche Eigenschaften das entsprechende Gegenstück möglicher Liganden habe sollte. Diese Informationen können genutzt werden, um neue Substanzen zu identifizieren, welche die im Pharmakophore-Modell modellierten Interaktionen mit dem Target eingehen können. Durch die Abstraktion können hierbei sowohl Verbindungen mit neuen Grundgerüsten (scaffold) als auch mit veränderten funktionellen Gruppen gefunden werden. Im ersten Fall spricht man dabei von „scaffold hopping“, der letzte Fall ist eng verbunden mit dem Konzept des bioisosteren Ersatzes.
Im Rahmen dieser Arbeit wurden Pharmakophore genutzt, um in drei Studien neue Inhibitoren der Arachidonsäurekaskade zu finden. Die Arachidonsäurekaskade ist ein Stoffwechselweg in der aus Arachidonsäure eine Reihe von Lipidmediatoren synthetisiert wird. Viele dieser Mediatoren spielen eine entscheidende Rolle in Entzündungsprozessen und damit einhergehenden Krankheitsbildern. Es wurde außerdem eine neue Methode zur automatischen Generierung von Pharmakophor-Modellen aus einer Menge bekannter Liganden entwickelt.
In der ersten Studie wurde ein Struktur-basiertes Pharmakophor-Modell der Bindetasche der löslichen Epoxidhydrolase (sEH) erstellt, welches die möglichen, relevanten Interaktionsmöglichkeiten abbilden sollte. Dieses Pharmakophor-Modell wurde zum Screening einer Datenbank kommerziell erhältlicher Verbindungen genutzt und führte zu zwei Verbindungen mit IC50-Werten im niedrigen mikromolaren Bereich sowie einer dritten Verbindung mit einem bisher nicht für Inhibitoren der sEH beschriebenem Chemotyp. Zwar war diese Verbindung in höheren Konzentration unlöslich war, jedoch erreichten Derivate ebenfalls IC50-Werte im niedrigen mikromolaren Bereich und könnten als mögliche Startpunkte für eine neue Substanzklasse von sEH-Inhibitoren dienen.
In einer zweiten Studie wurde ein Liganden-basierter Ansatz gewählt um neue Inhibitoren der Leukotrien-A4 Hydrolase (LTA4H) zu suchen. Im Rahmen dieser Studie wurde außerdem eine neue Methode zur automatischen Generierung von Pharmakophor-Modellen entwickelt, welche auf einem wachsenden neuronalen Gas basiert, einer Methode aus dem Bereich des maschinellen Lernens. Die Methode wurde retrospektiv anhand mehrerer Benchmark-Datensätze validiert. Unter anderem wurde überprüft, inwiefern die Methode in der Lage ist die bioaktive Konformation eines Liganden vorherzusagen. Hierzu wurden, ausgehend von co-kristallisierten Liganden, automatisch Modelle generiert, welche im Anschluss genutzt wurden um Konformations-Datenbanken der Liganden zu durchsuchen. Je näher die beste gefundene Konformation an der co-kristallisierten Konformation lag, desto besser war das erzeugte Modell. Die entwickelte Methode war in nahezu allen Fällen in der Lage ein Modell zu erzeugen, mit welchem die durchschnittliche Abweichung zwischen co-kristallisierter und gefundener Konformation unter 2 Å lag. Im Rahmen der Studie wurde die neu entwickelte Methode auch in einem prospektiven Virtual Screening nach neuen Liganden der LTA4H genutzt. Hierzu wurden basierend auf 24 Kristallstrukturen mehrere Pharmakophor-Modelle für LTA4H-Liganden erstellt. Durch zusätzliche Nutzung des ESshape3D Fingerprints konnte außerdem die Form der Bindetasche der LTA4H erfasst werden. Diese Modelle wurden anschließend eingesetzt um eine Datenbank kommerziell erhältlicher Verbindungen zu durchsuchen und führten zur Identifizierung von zwei Substanzen mit IC50-Werten im unteren mikromolaren Bereich. Des Weiteren war die neue Methode in der Lage, den Bindemodus des genutzten Referenzinhibitors vorherzusagen, welcher durch Röntgenstrukturanalyse bestätigt wurde.
In zwei weiteren Studien wurde versucht, duale Inhibitoren der sEH und der 5-Lipoxygenase (5-LO) zu finden. Die erste dieser beiden Studien nutzte hierfür „duale“ Pharmakophor-Modelle: für beide Targets wurde basierend auf einer Vielzahl publizierter, aktiver Liganden eine Reihe von Pharmakophor-Modellen erstellt. Diese Modelle wurden paarweise miteinander verglichen; Modelle, welche eine ausreichend hohe Überlappung an Features besaßen, dienten als Ausgangspunkt für die Suche nach potentiell dualen Liganden. Durch die Suche in einer Fragment-Datenbank konnten neun Verbindungen identifiziert werden, welche eine Aktivität gegenüber einem der beiden Targets zeigten. Diese Verbindungen besaßen zum Teil noch nicht in der Literatur für sEH- oder 5 LO Inhibitoren beschriebene Strukturmerkmale. Eine der Verbindungen war außerdem in der Lage beide Targets im niedrigen mikromolaren Bereich zu inhibieren und könnte als Ausgangspunkt zur Entwicklung weiterer dualer 5-LO/sEH-Inhibitoren dienen. In der zweiten Studie wurde eine auf selbst-organisierenden Karten (SOM) basierende Methode genutzt um potentiell duale Liganden zu suchen. Hierzu wird je eine SOM mit repräsentativen (Sub-) Strukturen von Liganden beider Targets trainiert. Die DrugBank, eine Datenbank zugelassener Wirkstoffe, dient hierbei als Hintergrundverteilung und stellt den Raum wirkstoffartiger chemischer Strukturen dar. Durch einen automatischen Vergleich der trainierten SOMs können mögliche gemeinsame Substrukturen identifiziert werden. Die Anwendung dieser Methode auf bekannte Inhibitoren der sEH und der 5-LO identifizierte neun Fragmente, die auf einem der beiden Targets, sowie fünf Fragmente welche auf beiden Targets im niedrigen mikromolaren Bereich inhibierend wirken. Eine Substruktursuche nach einem dieser Fragmente in einer internen Datenbank lieferte eine Verbindung, welche beide Targets im nanomolaren Bereich inhibiert und eine vielversprechender Basis als Leitstruktur für die Entwicklung dualer 5-LO/sEH-Inhibitoren darstellt.
Zusammenfassend wurden in dieser Arbeit mehrere Ansätze vorgestellt wie Pharmakophore in der Wirkstoffsuche eingesetzt werden können. Im Rahmen mehrerer Virtual Screenings konnten eine Reihe neuer Inhibitoren gefunden werden, einige mit nicht zuvor beschriebenen Strukturmerkmalen für das jeweilige Target. Es wurde außerdem eine neue Methode zur automatischen Generierung von Pharmakophor-Modellen entwickelt, welche sowohl retrospektiv als auch prospektiv validiert wurde.
We compute the probability distribution P(N) of the net-baryon number at finite temperature and quark-chemical potential, μ, at a physical value of the pion mass in the quark-meson model within the functional renormalization group scheme. For μ/T < 1, the model exhibits the chiral crossover transition which belongs to the universality class of the O(4) spin system in three dimensions. We explore the influence of the chiral crossover transition on the properties of the net baryon number probability distribution, P(N). By considering ratios of P(N) to the Skellam function, with the same mean and variance, we unravel the characteristic features of the distribution that are related to O(4) criticality at the chiral crossover transition. We explore the corresponding ratios for data obtained at RHIC by the STAR Collaboration and discuss their implications. We also examine O(4) criticality in the context of binomial and negative-binomial distributions for the net proton number.
Protein folding in cells is regulated by networks of chaperones, including the heat shock protein 70 (Hsp70) system, which consists of the Hsp40 cochaperone and a nucleotide exchange factor. Hsp40 mediates complex formation between Hsp70 and client proteins prior to interaction with Hsp90. We used mass spectrometry (MS) to monitor assemblies formed between eukaryotic Hsp90/Hsp70/Hsp40, Hop, p23, and a client protein, a fragment of the glucocorticoid receptor (GR). We found that Hsp40 promotes interactions between the client and Hsp70, and facilitates dimerization of monomeric Hsp70. This dimerization is antiparallel, stabilized by post-translational modifications (PTMs), and maintained in the stable heterohexameric client-loading complex Hsp902Hsp702HopGR identified here. Addition of p23 to this client-loading complex induces transfer of GR onto Hsp90 and leads to expulsion of Hop and Hsp70. Based on these results, we propose that Hsp70 antiparallel dimerization, stabilized by PTMs, positions the client for transfer from Hsp70 to Hsp90.
BACKGROUND: Vermeulen et al. 2014 published a meta-regression analysis of three relevant epidemiological US studies (Steenland et al. 1998, Garshick et al. 2012, Silverman et al. 2012) that estimated the association between occupational diesel engine exhaust (DEE) exposure and lung cancer mortality. The DEE exposure was measured as cumulative exposure to estimated respirable elemental carbon in μg/m(3)-years. Vermeulen et al. 2014 found a statistically significant dose-response association and described elevated lung cancer risks even at very low exposures.
METHODS: We performed an extended re-analysis using different modelling approaches (fixed and random effects regression analyses, Greenland/Longnecker method) and explored the impact of varying input data (modified coefficients of Garshick et al. 2012, results from Crump et al. 2015 replacing Silverman et al. 2012, modified analysis of Moehner et al. 2013).
RESULTS: We reproduced the individual and main meta-analytical results of Vermeulen et al. 2014. However, our analysis demonstrated a heterogeneity of the baseline relative risk levels between the three studies. This heterogeneity was reduced after the coefficients of Garshick et al. 2012 were modified while the dose coefficient dropped by an order of magnitude for this study and was far from being significant (P = 0.6). A (non-significant) threshold estimate for the cumulative DEE exposure was found at 150 μg/m(3)-years when extending the meta-analyses of the three studies by hockey-stick regression modelling (including the modified coefficients for Garshick et al. 2012). The data used by Vermeulen and colleagues led to the highest relative risk estimate across all sensitivity analyses performed. The lowest relative risk estimate was found after exclusion of the explorative study by Steenland et al. 1998 in a meta-regression analysis of Garshick et al. 2012 (modified), Silverman et al. 2012 (modified according to Crump et al. 2015) and Möhner et al. 2013. The meta-coefficient was estimated to be about 10-20 % of the main effect estimate in Vermeulen et al. 2014 in this analysis.
CONCLUSIONS: The findings of Vermeulen et al. 2014 should not be used without reservations in any risk assessments. This is particularly true for the low end of the exposure scale.
The RAF family of kinases constitutes the members A, B and CRAF. They mediate RAS signaling by linking it to the MEK/ERK transduction module, which regulates cellular processes such as cell proliferation, migration, survival and cell death. As the RAS/RAF/MEK/ERK (MAPK) pathway is found to be activated in human cancers, the RAF kinases have been exploited as valuable therapeutic targets and RAF inhibitors show promising results in the clinic, esp. with tumors harboring an activating BRAFV600E mutation. However, RAF inhibitors paradoxically accelerate metastasis in RAS mutant and BRAF wildtype tumors. They also become ineffective over time in BRAFV600E tumors because of reactivation of downstream mitogen-activated protein kinase (MAPK) signaling by promoting RAF dimerization. Aims of the present work were 1) to investigate the role of ARAF kinase in the paradoxical activation of the enzymatic cascade by RAF inhibitors downstream of mutated RAS and 2) to study the consequences of the loss of ARAF function on signal transduction in vitro and in vivo (nude mice). We have engineered several cell lines that would allow the study of basal and RAF inhibitor induced effects on MAPK activation, tumor cell migration and invasion.
In summary, we were able to show that the RAF isoform ARAF has an obligatory role in promoting MAPK activity and tumor cell invasion in a cell type dependent manner. In these cell types, ARAF depletion prevented the activation of MAPK kinase 1 (MEK1) and extracellular signal-regulated kinase 1 and 2 (ERK1/2) and led to a significant decrease of protrusions growing out of tumor cell spheroids in a three-dimensional (3D) culture that were otherwise induced by BRAFV600E-specific or BRAF/CRAF inhibitors (GDC-0879 and sorafenib, respectively). RAF inhibitors stimulated homodimerization of ARAF and heteromerization of BRAF with CRAF and the scaffolding protein KSR1. However, induced oligomerization was not sufficient to activate MAPK signaling if ARAF was depleted. By employing full length recombinant kinases, we were able to show for the first time that the three RAF isoforms competed for the binding to MEK1. In cell culture models, the overexpression of dimer-deficient ARAF mutants impaired the interaction between ARAF and endogenous MEK1 and thus prevented the subsequent phosphorylation of MEK1 and ERK1/2. Our findings reveal a new role for ARAF in directly activating the MAPK cascade through homodimerization and thereby promoting tumor cell invasion, suggesting the conserved RAF-dimer interface as a target for RAS- and RAF mediated cancer therapy.
Collectively, we provide evidence for the dual role ARAF plays in controlling MAPK signaling and cancer as loss of ARAF promoted strong lung metastasis formation in nude mice. Preliminary data describing the underlying mechanisms behind ARAF-regulated metastases have been presented and discussed.
Mechanisms regulating protein degradation ensure the correct and timely expression of transcription factors such as hypoxia inducible factor (HIF). Under normal O2 tension, HIFα subunits are targeted for proteasomal degradation, mainly through vHL-dependent ubiquitylation. Deubiquitylases are responsible for reversing this process. Although the mechanism and regulation of HIFα by ubiquitin-dependent proteasomal degradation has been the object of many studies, little is known about the role of deubiquitylases. Here, we show that expression of HIF2α (encoded by EPAS1) is regulated by the deubiquitylase Cezanne (also known as OTUD7B) in an E2F1-dependent manner. Knockdown of Cezanne downregulates HIF2α mRNA, protein and activity independently of hypoxia and proteasomal degradation. Mechanistically, expression of the HIF2α gene is controlled directly by E2F1, and Cezanne regulates the stability of E2F1. Exogenous E2F1 can rescue HIF2α transcript and protein expression when Cezanne is depleted. Taken together, these data reveal a novel mechanism for the regulation of the expression of HIF2α, demonstrating that the HIF2α promoter is regulated by E2F1 directly and that Cezanne regulates HIF2α expression through control of E2F1 levels. Our results thus suggest that HIF2α is controlled transcriptionally in a cell-cycle-dependent manner and in response to oncogenic signalling.
In search for the elusive schizophrenia pathway, candidate genes for the disorder from a discovery sample were localized within the energy-delivering and ischemia protection pathway. To test the adult vascular-ischemic (AVIH) and the competing neurodevelopmental hypothesis (NDH), functional genomic analyses of practically all available schizophrenia-associated genes from candidate gene, genome-wide association and postmortem expression studies were performed. Our results indicate a significant overrepresentation of genes involved in vascular function (P<0.001), vasoregulation (that is, perivascular (P<0.001) and shear stress (P<0.01), cerebral ischemia (P<0.001), neurodevelopment (P<0.001) and postischemic repair (P<0.001) among schizophrenia-associated genes from genetic association studies. These findings support both the NDH and the AVIH. The genes from postmortem studies showed an upregulation of vascular-ischemic genes (P=0.020) combined with downregulated synaptic (P=0.005) genes, and ND/repair (P=0.003) genes. Evidence for the AVIH and the NDH is critically discussed. We conclude that schizophrenia is probably a mild adult vascular-ischemic and postischemic repair disorder. Adult postischemic repair involves ND genes for adult neurogenesis, synaptic plasticity, glutamate and increased long-term potentiation of excitatory neurotransmission (i-LTP). Schizophrenia might be caused by the cerebral analog of microvascular angina.
