Im Zuge des Braunkohlentagebaus Zukunft-West fand sich im Jahre 1980 in einem Matronenheiligtum in Eschweiler-Fronhoven (Städteregion Aachen) das ringsum abgebrochene Oberteil eines Weihealtars (Höhe 36 cm – Breite 47 cm – Tiefe 19 cm) aus hellgrauem Sandstein. Der fragmentarisch erhaltene Text aus der Zeit zwischen 150 und 230 n. Chr. wurde vom Ersteditor CH. B. RÜGER (in Majuskelschrift) wie folgt wiedergegeben...
CIL XIII 4136 : Rekonstruktion einer verlorenen Grabinschrift aus Irsch in der Gallia Belgica

Eine verlorene fragmentarische Grabinschrift römischer Zeitstellung aus Irsch bei Bitburg (CIL XIII 4136) lässt sich mit Hilfe des bekannten Namenmaterials relativ sicher ergänzen. Der Grabstein wurde von einem einheimischen Treverer namens [L.?] Ataco[niu]s Ammo[sus] für seinen verstorbenen Sohn [Am]mosius A[vitus?] gesetzt.
Working memory denotes the ability to retain stimuli in mind that are no longer physically present and to perform mental operations on them. Electro- and magnetoencephalography allow investigating the short-term maintenance of acoustic stimuli at a high temporal resolution. Studies investigating working memory for non-spatial and spatial auditory information have suggested differential roles of regions along the putative auditory ventral and dorsal streams, respectively, in the processing of the different sound properties. Analyses of event-related potentials have shown sustained, memory load-dependent deflections over the retention periods. The topography of these waves suggested an involvement of modality-specific sensory storage regions. Spectral analysis has yielded information about the temporal dynamics of auditory working memory processing of individual stimuli, showing activation peaks during the delay phase whose timing was related to task performance. Coherence at different frequencies was enhanced between frontal and sensory cortex. In summary, auditory working memory seems to rely on the dynamic interplay between frontal executive systems and sensory representation regions.
Das allgemeine Bild, sowohl beim materiellen wie auch beim prozessualen EU-Strafrecht, zeigt, dass trotz der positiven Schritte, die in den letzten Jahren zu verzeichnen sind, noch große Anstrengungen notwendig sein werden, damit man von einer zufrieden stellenden Achtung fundamentaler Strafrechtsprinzipien im Rahmen der EU sprechen kann. Der Beitrag versucht darauf aufmerksam zu machen und Wege zu zeigen, damit fundamentale Straf-rechtsprinzipien das zentrale Instrument einer notwendigen Korrektur werden, die die gemeinsame europäische Rechtskultur fördern können.
BACKGROUND: Inflammatory events after kidney transplantation (Tx) may lead to activation of the tryptophane-catabolizing enzyme indoleamine 2,3-dioxygenase followed by the formation of kynurenine (KYN). Post-transplant KYN serum levels in kidney allograft recipients were analyzed for their diagnostic value.
MATERIAL AND METHODS: This was a retrospective analysis of KYN levels (normal value: 2.7±0.6 nmol/ml) measured in 4083 blood samples collected from 355 kidney graft recipients in connection with uncomplicated courses, acute rejections (ARs), infections, and type of immunosuppression. We performed descriptive data analysis and analysis of variance.
RESULTS: In 212 recipients with immediately functioning grafts, the KYN levels dropped from pre-Tx 13.3±5.9 nmol/ml to nearly normal values at day 5 (5.8±3.0 nmol/ml). In patients with delayed graft function, the KYN reduction started only after the last hemodialysis treatment. With respect to ARs in recipients with creatinine values <300 µmol/l pre-AR, the increase of KYN levels depended on the severity of ARs (steroid-sensitive ARs: from 4.5±1.4 to 6.0±6.1 nmol/ml; steroid-resistant ARs: from 6.1±3.1 to 12.9±7.1 nmol/ml; vascular rejections: from 5.8±3.0 to 16.9±9.1 nmol/ml). In patients with creatinine values ≥300 µmol/l pre-AR, a further increase of the KYN level (from 10.1 to 13.2 nmol/ml) was only observed in severe, steroid-resistant ARs. With respect to infections evaluated, the KYN levels before diagnosis/start of treatment were 5.7±3.4 nmol/ml in asymptomatic CMV infections, 7.5±4.4 nmol/ml in CMV diseases, 8.3±3.3 nmol/ml in pneumonia, and 10.4±6.5 nmol/ml in bacterial sepsis.
CONCLUSIONS: Serum KYN seems to be a reliable diagnostic tool for the assessment of post-transplant inflammatory complications, already in an early stage, and for monitoring the efficacy of therapeutic interventions. Prospective studies are recommended.
Understanding land cover degradation patterns and the effects of geomorphological units on phytodiversity is important for guiding management decisions and restoration strategies in the Sahelian vulnerables zones. This paper describes land cover degradation by combining Landsat TM image analysis and field data measurements in the Gourouol catchment of the Sahelian zone of Burkina Faso. Erdas Imagine 9.2 and Arc-GIS.10 were applied. The change patterns were obtained by superposing land cover maps for 1992 and 2010. The field data were collected by the mean of inventories according to the Braun-Blanquet phytosociological relevés methods. Plot sizes were 50 m x 20 m for woody species and 10 m x 10 m for herbaceous species. Six land cover types were identified and mapped: cultivated lands, bared lands, lowlands, which all spatially increased; and shrub-steppes, grasslands and water bodies, which all spatially decreased. The dynamic patterns based on the geomorphological units were non-degraded lowlands, stable sand dunes and degraded glacis. High plant diversity was found in lowlands, whereas low diversity occurred in glacis. A significant dissimilarity was observed between communities. The Shannon diversity indices in plant communities were approximately close to ln(species richness). The Pielou indices were close to 1, indicating a species fairly good distribution. Our results showed a variation of land cover over time and the effects of geomorphological units on phytodiversity. Furthermore, this variation helps oppose land degradation in the Sahel.

Auf der theoretischen Grundlage des erweiterten kategorialen Rahmens ist ein breites Forschungsfeld entstanden, das Lehr-Lern-Prozesse aus der Perspektive der Biografie- und Lebenslaufforschung untersucht. Diese Ausgabe referiert auf diese Forschungslage. Sie knüpft damit an die Themenhefte des REPORT „Biografieforschung und biografisches Lernen“ und „Biografie und Bildung“ an, legt aber nun den Schwerpunkt auf die Verknüpfung von Lebenslauf und Biographie im Hinblick auf das Lernen Erwachsener in und außerhalb von Institutionen.
Effekte von Neonikotinoiden auf die Aktivität des Muskels M17 und das Lernverhalten der Honigbiene

Im Rahmen dieser Arbeit wurden Untersuchungen zur Auswirkung von Neonikotinoiden auf die Muskelspikeaktivität des Muskels M17 und auf das Lernvermögen in einer komplexen Aufgabe an der Honigbiene (Apis mellifera carnica) durchgeführt. Dabei wurden drei verschiedene Substanzen verwendet: Clothianidin, Thiacloprid und Imidacloprid. Neonikotinoide stehen häufig im Verdacht, für das Sterben von Bienenvölkern verantwortlich zu sein, da die Bienen bei ihrer Nahrungssuche an behandelten Pflanzen den Substanzen ausgesetzt sind und diese möglicherweise damit auch an ihr Volk weitergeben. Die vorliegende Arbeit soll dazu beitragen, diese mögliche Gefährdung der Honigbiene durch Neonikotinoide weiter aufzuklären und deren Risiken zu beurteilen. Der Hintergrund für die Versuche zur Muskelspikeaktivität waren vorangegangene Versuche, die Auswirkungen von Neonikotinoiden auf die Motorik von sich frei bewegenden Individuen dokumentierten. In der vorliegenden Arbeit wurde untersucht, ob sich diese Effekte auch an der Spikeaktivität des Muskels M17 widerspiegeln. Dafür wurden Elektromyogramme des Muskels M17 zu verschiedenen Zeitpunkten nach der Gabe der Substanzen erstellt und deren mediane Anzahl mit einer Kontrollgruppe verglichen.
In einem ersten Versuch wurden Clothianidin (1 µM), Thiacloprid (1 µM), Imidacloprid (1 µM) oder eine Kontrollsubstanz (PBS) in die Kopfkapsel appliziert. Beim Vergleich mit der Kontrollgruppe zeigten sich für Imidacloprid keine Auswirkungen. Clothianidin verursachte eine deutlich erhöhte mediane Spikerate des Muskels M17, während Thiacloprid diese absenkte, beides im Vergleich zur Kontrollgruppe. Auch bei einer um 30 Minuten versetzten Doppelapplikation von Clothianidin (1 µM) und Thiacloprid (10 µM) stellten sich diese Effekte ein, wobei Clothianidin eine dominante Rolle einzunehmen scheint. Diese Ergebnisse stehen im Einklang mit Untersuchungen zur Laufaktivität, welche sich ebenfalls durch Clothianidin erhöhte und durch Thiacloprid absenkte.
Ein weiteres Experiment untersuchte die Auswirkung einer akuten Fütterung mit Clothianidin (1 ng in 1 µl) bzw. Thiacloprid (250 ng in 1 µl) auf die Anzahl der Muskelaktionspotenziale. Auch hier zeigt sich eine deutliche Erhöhung der Spikeanzahl durch Clothianidin und eine Absenkung der Spikeanzahl durch Thiacloprid, was die Ergebnisse des ersten Versuchs nochmals bestätigt.
Des Weiteren wurden Untersuchungen zur Auswirkung einer chronischen Fütterung mit Clothianidin (50 ppb) bzw. Thiacloprid (5000 ppb) auf die Anzahl an Spikes durchgeführt. Die Bienen wurden dabei über mehrere Wochen im Volk mit den jeweiligen Substanzen gefüttert. Dabei zeigte sich, dass auch eine chronische Fütterung der Bienen mit Clothianidin ihre Anzahl an Muskelaktionspotenzialen deutlich erhöht, während eine chronische Fütterung mit Thiacloprid diese absenkt. In einer Kombination der chronischen Fütterung mit einer zusätzlichen akuten Fütterung des jeweils anderen Neonikotinoids zeigte sich, dass auch hier Clothianidin eine dominante Rolle gegenüber Thiacloprid einnimmt und auch keine synergistischen oder Gewöhnungseffekte der beiden Substanzen eintreten. Die chronisch eingefütterten Völker entwickelten sich dagegen wie die Kontrollvölker, sodass die hier beschriebenen Auswirkungen auf die Einzelbiene keine sichtbaren Effekte auf ganze Völker zu haben scheinen.
Zusätzlich wurden Versuche zum Duftlernen in einer komplexen Lernaufgabe, dem positiven Patterning, unter Einfluss von Clothianidin durchgeführt. Die Bienen müssen dabei zwischen unbelohnten Einzeldüften und einem belohnten Duftgemisch, bestehend aus den beiden Einzeldüften, unterscheiden. In verschiedenen Versuchsdurchläufen wurden 0,25 ng oder 1 ng Clothianidin (jeweils 1 µl) in den Thorax injiziert, entweder vor der Akquisitionsphase oder vor dem ersten Abruftest nach drei Stunden. In keinem der Versuchsdurchläufe zeigten sich Effekte des Clothianidins auf den Lernvorgang, weder in der Akquisitionsphase noch in den Abruftests. Somit wurden weder das Lernen an sich, noch die Konsolidierung und damit die Überführung des Gelernten in das Langzeitgedächtnis durch das Clothianidin beeinflusst. Allerdings könnte die Immunabwehr der Bienen nach längerer Einwirkung des Clothianidins (24 h) in der höheren Konzentration herabgesetzt sein, da viele Bienen starben.
Insgesamt ergaben sich Effekte von Clothianidin und Thiacloprid auf die Anzahl der Aktionspotenziale des Muskels M17, die sich aber im gesamten Volk nicht widerspiegeln. Der Lernvorgang in der hier durchgeführten komplexen Lernaufgabe wird durch Clothianidin nicht beeinflusst. Möglicherweise entsteht dieser Unterschied in den Ergebnissen durch eine Bindung der Substanzen an verschiedene Rezeptorsubtypen, die pharmakologisch unterschiedliche Eigenschaften besitzen und somit auch unterschiedliche Auswirkungen zeigen.

G protein coupled receptors (GPCRs) constitute the largest family of cell-surface receptors in mammals and are key players in signal transduction. By responding to a plethora of extracellular stimuli ranging from photons to amines to fatty acids to peptides and proteins, these receptors trigger intracellular signalling cascades and regulate a variety of cellular responses. Approximately 800 genes in humans encode GPCRs which are classified according to sequence conservation into rhodopsin-like, glutamate, adhesion, frizzled/taste2 and secretin receptors. GPCRs share a seven transmembrane domain fold undergoing a conformational change upon ligand binding which is translated to the intracellular surface of the receptor thereby allowing a heterotrimeric G protein to couple. Heterotrimeric G proteins consist of a Ga, Gb and Gg subunit and dissociate into their Ga and Gbg entities upon activation by a GPCR. Subsequently, distinct signalling cascades are triggered by each G protein protomer.
Membrane proteins and GPCRs in particular, are highly important targets in drug design and development as currently approximately 60% of all marketed drugs target membrane proteins. Although these classes of proteins are of high therapeutic interest, our understanding of their mechanism of action and structure remains limited. The first structure of a human GPCR was determined in 2007 and required the development of protein engineering and innovative crystallisation techniques. Since then, approximately 130 GPCR structures of less than 40 individual receptors have been determined providing insights into the structural arrangement of the transmembrane helices, ligand binding pockets and G protein interactions. Combined with spectroscopic methods, these studies allowed a more detailed understanding of the molecular aspects of GPCR activation and signalling. Despite the tremendous advances in GPCR structural biology, certain aspects of GPCR function still remain poorly understood. Due to their size and inherent flexibility, the interaction of protein and peptide ligands with their receptors remains a challenging aspect in the structural characterisation of GPCRs. Moreover, structural information on subtype selectivity of peptide ligands continues to be scarce. To contribute functional and structural information on the molecular mechanisms of peptide interactions with GPCRs, this thesis focused on characterising receptors from the chemoattractant cluster using radioligand binding assays as well as NMR spectroscopy.
The chemoattractant cluster mainly groups the kinin, angiotensin, anaphylatoxin chemotactic complement and apelin receptors according to conserved residues in their ligand binding cavities. All receptors in this cluster bind to peptide ligands deriving from high molecular weight protein precursors upon proteolytic processing. Comparable to the conserved binding pocket of the chemoattractant receptors, the peptide ligands display a certain sequence conservation although they differ strongly in size. The largest ligands used in this thesis are the anaphylatoxins complement 3a and 5a, comprising 77 or 74 residues, respectively. Due to their size and complex fold involving three intramolecular disulphide bonds, solid phase synthesis is impossible, which prompted us to develop a modified cell-free expression system to produce these ligands in tritiated form for subsequent functional characterisation of the complement receptors. To demonstrate the versatility of the developed system, it was applied to another disulphidebond containing peptide ligand, the 21 amino acid endothelin-1. We describe a reliable and multifaceted tool to generate custom labelled peptide ligands for the structural and functional characterisation of GPCRs. The system allows the production of custom radioligands, peptides labelled for NMR studies or with fluorescent amino acids.
Apart from the modulation of GPCR activity by orthosteric ligands, GPCR signalling has long been described to be regulated by allosteric ligands including peptides, small molecules and ions. In this thesis, the influence of sodium ions on the activity state of the chemoattractant cluster receptors and in particular on the apelin, bradykinin 2 and angiotensin II type 1 receptors was examined. In recent high resolution crystal structures an allosteric sodium ion pocket beneath the orthosteric ligand binding cavity was identified and residues contributing to the coordination of sodium ions are conserved throughout the chemoattractant cluster receptors. This allosteric sodium ion coordinated within the transmembrane domain bundle has been described to negatively influence the affinity of agonists but not of antagonists. It was found that sodium ions have distinct influences on the affinity state as well as the available number of binding sites of the chemoattractant receptors. In case of the apelin and bradykinin 2 receptors, sodium ions drastically reduced the number of available binding sites whereas the affinity of peptide ligands to the bradykinin 2 receptors remained constant and the ligand binding affinities to the apelin receptor were completely abolished. In contrast, the angiotensin II type 1 receptor affinity state towards the endogenous peptide ligand angiotensin II is highly dependent on the presence of sodium ions, whereas binding of the synthetic peptide antagonist Sar1-Ile8-angiotensin II remained unaffected by the sodium ion concentration. As differential effects irrespective of the efficacy class but dependent on the amino acid composition of the applied ligands are observed, it can be concluded that electrostatic interactions between charged residues of the peptide ligands and amino acids on the extracellular surface of the receptors are influenced by sodium ions thereby adding another layer of complexity on GPCR signalling.
To elucidate the structure-function relationship of ligand selectivity between the kinin receptors, the structure of desArg10-kallidin (DAK) bound to the bradykinin 1 receptor was determined using solid state NMR (SSNMR) in the course of this thesis. The kinin peptides DAK and bradykinin bind with high affinity and high selectivity to either the bradykinin 1 or bradykinin 2 receptor, respectively. The binding pockets of the receptors are highly conserved and the two peptide ligands only differ in one amino acid at their N- and C-termini whereas the remaining eight amino acids are fully conserved. DAK adopts a U-shaped structure when bound to the bradykinin 1 receptor which resembles a horse shoe-like conformation. Using 2D TEDOR spectroscopy it could furthermore be demonstrated that positively charged residues at the N-terminal part of the peptide engage in ionic interactions with negatively charged amino acids on the extracellular surface of the bradykinin 1 receptor. In contrast, bradykinin displays a distinct b-turn at the C-terminus and an S-shaped conformation of the N-terminal segment when bound to the bradykinin 2 receptor. By using SSNMR to study the binding mode of DAK on the bradykinin 1 receptor we could determine that subtype selectivity between the kinin receptors is conferred by distinct conformational restraints within the peptide ligands and by the formation of specific ionic interaction between charged residues on the peptide and receptor, respectively.
In brief, this thesis contributes structural and functional data on the binding mechanisms and binding mode of different peptide-ligand GPCRs helping to understand subtype selectivity and allosteric modulation of the chemoattractant cluster receptors. In addition, a versatile cell-free expression system was developed that allows the custom synthesis of isotopically labelled peptides containing disulphide bonds for the functional characterisation of GPCRs.
Under certain conditions, secondary organic aerosol (SOA) particles can exist in the atmosphere in an amorphous solid or semi-solid state. To determine their relevance to processes such as ice nucleation or chemistry occurring within particles requires knowledge of the temperature and relative humidity (RH) range for SOA to exist in these states. In the CLOUD experiment at CERN, we deployed a new in-situ optical method to detect the viscosity of α-pinene SOA particles and measured their transition from the amorphous viscous to liquid state. The method is based on the depolarising properties of laboratory-produced non-spherical SOA particles and their transformation to non-depolarising spherical liquid particles during deliquescence. We found that particles formed and grown in the chamber developed an asymmetric shape through coagulation. A transition to spherical shape was observed as the RH was increased to between 35 % at −10 ◦C and 80 % at −38 ◦C, confirming previous calculations of the viscosity transition conditions. Consequently, α-pinene SOA particles exist in a viscous state over a wide range of ambient conditions, including the cirrus region of the free troposphere. This has implications for the physical, chemical and ice-nucleation properties of SOA and SOA-coated particles in the atmosphere.
In vivo long-term monitoring of circulating tumor cells fluctuation during medical interventions

The goal of this research was to study the long-term impact of medical interventions on circulating tumor cell (CTC) dynamics. We have explored whether tumor compression, punch biopsy or tumor resection cause dissemination of CTCs into peripheral blood circulation using in vivo fluorescent flow cytometry and breast cancer-bearing mouse model inoculated with MDA-MB-231-Luc2-GFP cells in the mammary gland. Two weeks after tumor inoculation, three groups of mice were the subject of the following interventions: (1) tumor compression for 15 minutes using 400 g weight to approximate the pressure during mammography; (2) punch biopsy; or (3) surgery. The CTC dynamics were determined before, during and six weeks after these interventions. An additional group of tumor-bearing mice was used as control and did not receive an intervention. The CTC dynamics in all mice were monitored weekly for eight weeks after tumor inoculation. We determined that tumor compression did not significantly affect CTC dynamics, either during the procedure itself (P = 0.28), or during the 6-week follow-up. In the punch biopsy group, we observed a significant increase in CTC immediately after the biopsy (P = 0.02), and the rate stayed elevated up to six weeks after the procedure in comparison to the tumor control group. The CTCs in the group of mice that received a tumor resection disappeared immediately after the surgery (P = 0.03). However, CTC recurrence in small numbers was detected during six weeks after the surgery. In the future, to prevent these side effects of medical interventions, the defined dynamics of intervention-induced CTCs may be used as a basis for initiation of aggressive anti-CTC therapy at time-points of increasing CTC number.
Effiziente kryptographische Algorithmen sind ein wichtiger Grundstein für viele neue Anwendungen, wie zum Beispiel das Internet der Dinge (IoT) oder kontaktlose Zahlungssysteme. Daher ist es wichtig, dass neue Algorithmen mit verbesserten Sicherheitseigenschaften und speziellen Leistungseigenschaften entwickelt und analysiert werden. Ein Beispiel ist der aktuelle Trend zu leichtgewichtigen Algorithmen. Diese Entwicklungen erleichtern die Implementierung neuartiger Systeme und ermöglichen auch einen Schutz von bestehenden Systemen durch eine Anpassung auf den neuesten Stand der Technik. Neben der kryptologischen Analyse, ist die Bewertung von Implementierungs-Aspekten sehr wichtig, damit eine realistische Einschätzung der erzielbaren Leistung möglich ist.
Daher müssen für jeden neuen Algorithmus unterschiedliche Software- und Hardwarearchitekturen evaluiert werden. Die systematische Bewertung von Software-Implementierungen für unterschiedliche Hardware-Architekturen hat in den letzten Jahren große Fortschritte gemacht, zum Beispiel durch den SHA-3 Wettbewerb. Im Vergleich dazu ist die Evaluation für Hardware-Plattformen wie z.B. FPGAs weiterhin sehr zeitaufwendig und fehleranfällig. Dies liegt an vielen Faktoren, z.B. an den mannigfaltigen Möglichkeiten der verschiedenen Zieltechnologien. Ein möglicher Verbesserungsansatz besteht darin, die Bewertung mit einem abstrakteren Ansatz zu beginnen, um interessante Architekturen und Implementierungen anhand von theoretischen Eigenschaften auszuwählen.
Der erste Hauptbeitrag dieser Arbeit ist die Entwicklung einer abstrakten Bewertungsmethodik, die auf einem theoretischen Modell von getakteten Schaltungen basiert. Das Modell verbessert das Verständnis von Grundeigenschaften dieser Schaltungen und erleichtert auch die abstrakte Modellierung von Architekturen für einen spezifischen Algorithmus. Wenn mehrere verschiedene Architekturen für den gleichen Algorithmus ausgewertet werden, ist es auch möglich zu bestimmen, ob ein Algorithmus gut skaliert. Beispielsweise können Auswirkungen einer Verkleinerung des Datenpfades auf die Größe des Speicherverbrauchs analysiert werden. Basierend auf der entwickelten Methodik können wichtige Eigenschaften, wie der Speicherbedarf, die Anzahl an Taktzyklen oder die Pipeline-Tiefe systematisch bewertet werden. Damit kann eine grobe Schätzung für die Effektivtät einer Architektur abgeleitet werden.
Die Performance-Abschätzung wird auch durch ein theoretisches Konzept der Optimalität der Anzahl an Taktzyklen untermauert. Optimal in diesem Sinne ist eine Architektur, wenn sie verzögerungsfrei ist, d.h. keine Wartezyklen benötigt. Durch die Betrachtung von Datenabhängigkeiten zwischen den einzelnen Runden kann eine minimale und maximale Anzahl an Taktzyklen ermittelt werden. Eine Verletzung dieser Grenzen würde bedeuten, dass die Berechnung der Runden-Funktion nicht alle Ausgangs-Bits produziert hat, wenn diese für die nächste Runde benötigt werden und somit würden Wartezyklen entstehen.
Der zweite Beitrag der Dissertation nutzt die Analysemethodik für mehrere Hash-Funktion. Es werden sechs Hash-Funktionen bewertet: BLAKE, Grøstl, Keccak, JH, Skein und Photon. Die ersten fünf Hash-Funktionen sind die Finalisten des SHA-3 Wettbewerb. Die SHA-3 Finalisten haben eine hohe Sicherheit als oberstes Design-Ziel und nur in zweiter Linie eine hohe Performance. Im Gegensatz dazu wurde Photon für leichtgewichtige Anwendungen konzipiert, z.B. RFID-Tags. Dazu wurde auch die Sicherheit von Photon reduziert. Für jeden Algorithmus wird eine oder mehrere mögliche Organisationensformen des Speichers entwickelt. Als nächstes wird die Anzahl von Taktzyklen auf der Grundlage der Speicherorganisation ermittelt. Das generelle Ziel dabei ist die Entwicklung von Architekturen mit einer optimalen Anzahl von Taktzyklen. Die Diskussion konzentriert sich als nächstes auf verschiedene Möglichkeiten die Runden-Funktion optimal umzusetzen. Das Ergebnis der Evaluierung umfasst mindestens die Schätzung der minimalen Speicheranforderung, die analysierte Pipeline-Tiefe und den theoretischen Durchsatz für lange Nachrichten mit einer festgelegten Taktfrequenz. Diese Ergebnisse lassen eine Einschätzung über die mögliche Leistung der jeweiligen Architekturen zu.
Der dritte Beitrag der Arbeit besteht aus mehreren Implementierungs-Ergebnissen. Zunächst werden Ergebnisse für die SHA-3 Finalisten BLAKE, Grøstl, JH, Keccak und Skein gezeigt. Von den fünf Algorithmen haben alle außer Skein eine relativ hohe Performanz, während Skein abgeschlagen ist. Eine weitere Untersuchung konzentriert sich auf kleinere Implementierungen des SHA-3 Siegers Keccak. Dazu gehören auch nicht standardisierte Varianten mit einem kleineren Zustand. Diese kleineren Versionen werden mit ersten FPGA-Ergebnissen für die Photon Hash-Funktion verglichen. Eine wesentliche Erkenntnis davon ist, dass Keccak auch für FPGA-Anwendungen mit beschränktem Ressourcen-Bedarf prinzipiell sehr wettbewerbsfähig ist.
Im Rahmen dieser Arbeit wurden grundlegende Eigenschaften von GEM-Verstärkungsstrukturen untersucht. Dies waren der Einfluss des Alignmenteffektes auf die Reproduzierbarkeit von Messungen, die Elektronenextraktionseffizienz von GEMs im allgemeinen und die Auswirkungen von Druckschwankungen auf die Gasverstärkung. Weiterhin wurden verschiedene vierlagige GEM-Verstärkungssysteme mit einer MP-GEM an erster Stelle in Hinblick auf Ionenrückfluss und Energieauflösung untersucht.
Der Alignmenteffekt ist noch nicht vollkommen verstanden und verlangt weitere Untersuchungen. Was aber definitiv gesagt werden kann ist, dass das Drehen der GEMs um 90° die Reproduzierbarkeit der Messergebnisse sicherstellt.
Die unterschiedlichen Elektronenextraktionseffizienzen der verschiedenen GEM-Typen sind noch unverstanden. Auch wenn die grundsätzliche Zunahme der Extraktion mit zunehmenden Transferfeld verständlich ist, so bleibt vor allem das Verhalten einer LP-GEM in diesem Kontext bis jetzt unerklärlich.
Die Versuche mit einer MP-GEM an erster Stelle einer vierlagigen Verstärkungsstruktur haben sich als keine Verbesserung im Vergleich zu den S-Konfigurationen herausgestellt. Auch wenn manche gefundenen Einstellungen die Kriterien von einem IBF von weniger als 1 % und einem σ(55Fe) von weniger als 12 % gleichzeitig erfüllen, liegen diese Messpunkte so knapp an den definierten Grenzen, dass sie nicht für den Betrieb in der Spurendriftkammer von ALICE geeignet sind. Eine Erkenntnis, die trotzdem gewonnen werden konnte, ist, dass sich das Verhalten von verschiedenen Konfigurationen verstehen lässt. So ist die beste untersuchte Konfiguration die MP-LP-LP-S-Konfiguration gewesen, danach folgte die MP-S-LP-S und als schlechteste hat die MP-S-LP-SP-Konfiguration abgeschnitten. Dies ist genau die gleiche Reihenfolge, wie sie auch bei den S-Konfigurationen auftritt: S-LP-LP-S, dann S-S-LP-S und danach S-S-LP-SP. Ein wichtiger Schritt in einem guten Kompromiss zwischen Ionenrückfluss und σ(55Fe), scheinen zwei LP-GEMs an zweiter und dritter Stelle zu sein und weniger der Lochabstand der letzten GEM.
Die Druckabhängigkeit der Gasverstärkung hat einen großen Einfluss auf die Verstärkung und damit auf die Reproduzierbarkeit von Messungen. Bei einem Höhenunterschied von ca. 400 m ergibt sich eine Änderung der Verstärkung von ca. 35 %. Zusätzlich wird dieser Effekt von lokalen Wetterbedingungen überlagert. Der Einfluss des Luftdruckes kann jedoch mit dem Fit in Abbildung 43 berücksichtigt und damit herausgerechnet werden
In seinem unlängst erschienenen Buch „Citizen Science“ untersucht der Wissenschaftstheoretiker Peter Finke die Rolle von Laiinnen und Laien für die Wissenschaft. Sein Anliegen ist es, ihre Bedeutung für den Erkenntnisfortschritt wie auch für ein praxisbezogenes bürgerschaftliches Engagement darzulegen. Aus zahlreichen Blickwinkeln variiert Finke den Grundgedanken einer Kontinuität des Handelns von Laiinnen und Laien zu dem von Fachwissenschaftlerinnen und Fachwissenschaftlern, die durch die institutionalisierten Erscheinungsformen der Wissenschaft verschleiert wird. Demgegenüber sollen im vorliegenden Beitrag Aspekte der Diskontinuität hervorgehoben werden, die es zu berücksichtigen gilt, gerade wenn man von der Wichtigkeit einer Etablierung und Förderung von „Citizen Science“ überzeugt ist.
Rolf van Dick und Holger Horz haben in ihrem UniReport-Essay »Bestmögliche Bildung und Ausbildung für alle« gefordert und die Kritik von Hans Peter Klein und Julian Nida-Rümelin am aktuellen Akademisierungswahn scharf zurückgewiesen. Mesenchymal stem/stromal cells (MSCs) feature promising potential for cellular therapies, yet significant progress in development of MSC therapeutics and assays is hampered because of remarkable MSC heterogeneity in vivo and in vitro. This heterogeneity poses challenges for standardization of MSC characterization and potency assays as well as for MSC study comparability and manufacturing. This review discusses promising marker combinations for prospective MSC subpopulation enrichment and expansion, and reflects MSC phenotype changes due to environment and age. In order to address animal modelling in MSC biology, comparison of mouse and human MSC markers highlights current common ground of MSCs between species.
Intrinsically disordered protein (IDP) duplexes composed of two IDP chains cross-linked by bivalent partner proteins form scaffolds for assembly of multiprotein complexes. The N-terminal domain of dynein intermediate chain (N-IC) is one such IDP that forms a bivalent scaffold with multiple dynein light chains including LC8, a hub protein that promotes duplex formation of diverse IDP partners. N-IC also binds a subunit of the dynein regulator, dynactin. Here we characterize interactions of a yeast ortholog of N-IC (N-Pac11) with yeast LC8 (Dyn2) or with the intermediate chain-binding subunit of yeast dynactin (Nip100). Residue level changes in Pac11 structure are monitored by NMR spectroscopy, and binding energetics are monitored by isothermal titration calorimetry (ITC). N-Pac11 is monomeric and primarily disordered except for a single α-helix (SAH) at the N terminus and a short nascent helix, LH, flanked by the two Dyn2 recognition motifs. Upon binding Dyn2, the only Pac11 residues making direct protein-protein interactions are in and immediately flanking the recognition motifs. Dyn2 binding also orders LH residues of Pac11. Upon binding Nip100, only Pac11 SAH residues make direct protein-protein interactions, but LH residues at a distant sequence position and L1 residues in an adjacent linker are also ordered. The long distance, ligand-dependent ordering of residues reveals new elements of dynamic structure within IDP linker regions.
In addition to infectious viral particles, hepatitis B virus-replicating cells secrete high amounts of SVPs, which are ssembled by HBsAg in the shape of spheres and filaments but lack any capsid and genome. Filaments are characterized by a much higher amount of the surface protein LHBs as compared to spheres. Spheres are
released via the constitutive secretory pathway, while viral particles are ESCRT-dependently released via MVBs. The interaction of virions with the ESCRT machinery is mediated by α-taxilin that connects the PreS1 domain of LHBs with the ESCRT-component tsg101. Since viral particles and filaments contain a significant amount of LHBs, it is unclear whether filaments are secreted as spheres or released like viral particles. To study the release pathways of HBV filaments in the absence of viral particles, A core-deficient
HBV mutant (1.2×HBVΔCore) was generated by site-directed mutagenesis based on wt1.2x HBV. The start codon of core protein was mutated into stop codon, which was confirmed by DNA sequencing. Data from HBsAg ELISA, Western blot, immunofluorescence microscopy and immunoelectron microscopy showed that the lack of core protein did neither affect the production nor the secretion of HBV SVPs. The intracellular distribution of
LHBs and SHBs showed no difference between wtHBV and the core-deficient mutant expressing cells. Therefore, this system is suitable to investigate the release pathway of HBV filaments in the absence of viral particles. Confocal microscopy analysis of cells cotransfected core-deficient mutants with peYFPRab7 as marker for the endosomal/MVB pathway or with pGalT-eGFP as marker for the trans Golgi apparatus showed that YFP-Rab7, but not GalT-GFP, partially colocalized with LHBs. Furthermore, LHBs could be found in dilated MVBs by immune electron microscopy of ultrathin sections. This was confirmed by isolation of MVBs by cell fractionation using discontinuous sucrose gradient ultracentrifugation and percoll-based linear gradient ultracentrifugation, indicating that filaments enter MVBs in the absence of virion formation. Moreover, inhibition of MVB biogenesis by the small molecular inhibitor U18666A significantly abolished the release of filaments in a dose-dependent manner, but no inhibition could be observed in the production. In contrast, no inhibition on the secretion and production of spheres could be
detected. Inhibition of ESCRT-functionality by coexpression of transdominant negative mutants (Vps4A, Vps4B, CHMP3) abolished the release of filaments while secretion of spheres was not affected. These data indicate that in contrast Abstract 73 to spheres while are secreted via the secretory pathway, filaments are released via ESCRT/MVB pathway like infectious viral particles.
Oxygen vacancies in strontium titanate surfaces (SrTiO3) have been linked to the presence of a two-dimensional electron gas with unique behavior. We perform a detailed density functional theory study of the lattice and electronic structure of SrTiO3 slabs with multiple oxygen vacancies, with a main focus on two vacancies near a titanium dioxide terminated SrTiO3 surface. We conclude based on total energies that the two vacancies preferably inhabit the first two layers, i.e. they cluster vertically, while in the direction parallel to the surface, the vacancies show a weak tendency towards equal spacing. Analysis of the nonmagnetic electronic structure indicates that oxygen defects in the surface TiO2 layer lead to population of Ti ${{t}_{2g}}$ states and thus itinerancy of the electrons donated by the oxygen vacancy. In contrast, electrons from subsurface oxygen vacancies populate Ti eg states and remain localized on the two Ti ions neighboring the vacancy. We find that both the formation of a bound oxygen-vacancy state composed of hybridized Ti 3eg and 4p states neighboring the oxygen vacancy as well as the elastic deformation after extracting oxygen contribute to the stabilization of the in-gap state.
The impact of (long-term) drought acclimation and (short-term) heat stress and their combination on fast chlorophyll fluorescence induction curves (OJIP) and grain yield was tested using pot-grown plants of wild barley (Hordeum spontaneum) originating from Northern Egypt. Concerning agronomic traits, the main effect of drought was decreased biomass accumulation and grain yield, while heat specifically affected floral development. The treatments caused specific inhibitions of photosystem II (PSII) functionality. While heat stressed plants showed a reduction of maximum quantum efficiency of PSII (φP0), an indication of effects on oxygen evolving complex (OEC) functionality, and the connectivity of PSII units, these features were entirely missing in drought acclimated plants. Drought caused a reduction of the Performance Index (PIabs) and of the relative amplitude of the IP-phase of the OJIP induction curve (ΔVIP). Individuals suffering from a combination of drought and heat showed a better ability to recover photosynthetic electron transport after the relief of stress in comparison to heat stressed plants. However, this improved capacity to recover was not accompanied by an increased grain yield. Thus, we conclude that chlorophyll fluorescence measurements provide valuable physiological data; however, their use in agronomic studies for the prediction of agronomic traits should be done with some precaution.
Long-term potentiation (LTP) and long-term depression (LTD) are widely accepted to be synaptic mechanisms involved in learning and memory. It remains uncertain, however, which particular activity rules are utilized by hippocampal neurons to induce LTP and LTD in behaving animals. Recent experiments in the dentate gyrus of freely moving rats revealed an unexpected pattern of LTP and LTD from high-frequency perforant path stimulation. While 400 Hz theta-burst stimulation (400-TBS) and 400 Hz delta-burst stimulation (400-DBS) elicited substantial LTP of the tetanized medial path input and, concurrently, LTD of the non-tetanized lateral path input, 100 Hz theta-burst stimulation (100-TBS, a normally efficient LTP protocol for in vitro preparations) produced only weak LTP and concurrent LTD. Here we show in a biophysically realistic compartmental granule cell model that this pattern of results can be accounted for by a voltage-based spike-timing-dependent plasticity (STDP) rule combined with a relatively fast Bienenstock-Cooper-Munro (BCM)-like homeostatic metaplasticity rule, all on a background of ongoing spontaneous activity in the input fibers. Our results suggest that, at least for dentate granule cells, the interplay of STDP-BCM plasticity rules and ongoing pre- and postsynaptic background activity determines not only the degree of input-specific LTP elicited by various plasticity-inducing protocols, but also the degree of associated LTD in neighboring non-tetanized inputs, as generated by the ongoing constitutive activity at these synapses.
Die Kephalometrie stellt einen bedeutenden Diagnostikbestandteil der kieferorthopädischen Diagnostik und Behandlungsplanung dar, mit deren Hilfe es möglich ist, skelettale Ursachen dentaler Befunde zu evaluieren. Üblicherweise erfolgt die kephalometrische Analyse anhand eines Fernröntgenseitenbildes, das jedoch neben der unausweichlichen Strahlenbelastung des Patienten, der sich zumeist im jugendlichen Alter und im Wachstum befindet und damit besonders vulnerabel bezüglich ionisierender Strahlung ist, auch geometrische Verzerrungen und Verzeichnungen aufweist, die eine kephalometrische Analyse erschweren. Zudem erfolgt beim FRS eine dreidimensionale Analyse, die auf einer zweidimensionalen Ansicht basiert und daher nicht die Exaktheit einer 1:1 Analyse besitzen kann. Die Kephalometrie mittels Magnetinduktion stellt eine strahlenfreie Alternative zur Verfügung. Im Rahmen der vorliegenden Studie ist untersucht worden, inwieweit Untersucher mit dem auf einer elektromagnetischen Induktion basierenden noXrayCeph®- Gerätes in der Lage sind, genaue Messdaten zu generieren und zu reproduzieren. Beim noXrayCeph®-Gerät handelt es sich um eine Weiterentwicklung des 3-Space®-Isotrak®- Gerätes der Firma POLHEMUS. Im Rahmen der hier durchgeführten Studie konnte die Messgenauigkeit der kephalometrischen Messung auf Basis der Magnetinduktion sowie die Reproduzierbarkeit durch verschiedene Untersucher statistisch belegt werden. Weiterhin sollten mit der Durchführung einer Anwenderschulung die Anwenderfreundlichkeit des Gerätes und das einfache Erlernen der Handhabung eruiert werden. Hierfür wurden insgesamt drei differente Versuchsreihen aufgebaut. Die Messgenauigkeit des noXrayCeph®-Gerätes wurde anhand eines normierten Abstands von exakt 100 mm nachgewiesen. Außerdem konnten fünf unterschiedliche Untersucher jeweils 15mal eine kephalometrische Analyse mit dem noXrayCeph®-Gerät reproduzierbar durchführen. Dabei waren die Messergebnisse sowohl Untersucher sowie zeitunabhängig. Diese kephalometrischen Untersuchungen erfolgten hierbei an einem Kunststoffschädel, auf den 55 Messpunkte als kephalometrische Variablen übertragen worden waren. In einer nachfolgenden Anwenderschulung konnten sechs männliche und vier weibliche Probanden nachweisen, dass auch ohne eine Anwenderschulung eine hohe Reproduzierbarkeit der ermittelten Werte erreicht werden konnte. Ebenfalls wurde der Einfluss des Geschlechts, des Winkels des angesetzten 103 Messstiftes, der Handhabung dieses Messstiftes sowie des physiologischen Tremors auf die Messgenauigkeit und die Reproduzierbarkeit der Daten geprüft. Es ließ sich wiederholt zeigen, dass mit dem noXrayCeph®-System genaue und reproduzierbare Werte einer kephalometrischen Analyse gewonnen werden, die bei den unterschiedlichen Untersuchern in einem Rahmen tolerierbarer Schwankungen vergleichbar sind. Auch intraindividuell weisen die ermittelten Daten eine hohe Reproduzierbarkeit auf. In dieser Studie sind Einflüsse des Geschlechts, der Handhabung des Messstiftes sowie des Winkels dieses Stiftes ebenso wenig nachweisbar wie ein positiver Einfluss einer Vorab-Schulung der Probanden. Der bei den Probanden vorhandene, unterschiedlich ausgeprägte physiologische Tremor und die unterschiedliche Haltung der Messspitze hatten keinerlei Auswirkung auf die Reproduzierbarkeit der Messergebnisse. Zusammenfassend lässt sich aus der durchgeführten Studie somit ableiten, dass das noXrayCeph®-Verfahren geeignet ist für eine exakte kephalometrische Analyse. Die Patienten sind keiner Strahlenexposition wie bei einer Röntgenaufnahme ausgesetzt. Die Anwenderschulung zeigte außerdem, dass das Verfahren und der Umgang mit dem noXrayCeph®-Gerät bei der Messung einfach erlernbar und gut durchführbar ist. Die aus dem Röntgenverfahren resultierenden Probleme wie Verzerrungen und Überlagerungsfehler sowie die Strahlenexposition gerade bei jungen Patienten können mit diesem Verfahren umgangen und eliminiert werden. Somit stellt die Kephalometrie mit dem noXrayCeph®-Gerät eine fundierte Alternative in der Kieferorthopädie dar.
In the qualitative analysis of solutions of partial differential equations, many interesting questions are related to the shape of solutions. In particular, the symmetries of a given solution are of interest. One of the first more general results in this direction was given in 1979 by Gidas, Ni and Nirenberg... The main tool in proving this symmetry and monotonicity result is the moving plane method. This method, which goes back to Alexandrov’s work on constant mean curvature surfaces in 1962, was introduced in 1971 by Serrin in the context of partial differential equations to analyze an overdetermined problem...
In 2000 Italy replaced its traditional system of severance pay for public employees with a new system. Under the old regime, severance pay was proportional to the final salary before retirement; under the new regime it is proportional to lifetime earnings. This reform entails substantial losses for future generations of public employees, in the range of €20,000-30,000, depending on seniority. Using a difference-in-difference framework, we estimate the impact of this unanticipated change in lifetime resources, on the current consumption and wealth accumulation of employees affected by the reform. In line with theoretical simulations, we find that each euro reduction in severance pay reduces the average propensity to consume by 3 cents and increases the wealth-income ratio by 0.32. The response is stronger for younger workers and for households where both spouses are public sector employees.
Ibegin by providing some background to conceptions of responsibility. I note the extent of disagreement in this area, the diverse and cross-cutting distinctions that are deployed, and the relative neglect of some important problems. These facts make it difficult to attribute responsibility for climate change, but so do some features of climate change itself which I go on to illuminate. Attributions of responsibility are often contested sites because such attributions are fundamentally pragmatic, mobilized in the service of a normative outlook. We should be pluralists about responsibility and shape whatever conceptions can help to explain, guide, and motivate our responses to climate change. I sketch one such notion, ‘intervention-responsibility’, and argue that it should be ascribed to international regimes and organizations, states and other jurisdictions, individuals, and firms. Each has different capacities and thus different intervention-responsibilities responsibilities, but these differences are not always mirrored in public discussion. In particular, the moral responsibility of firms has been greatly neglected.
Reading is not only "cold" information processing, but involves affective and aesthetic processes that go far beyond what current models of word recognition, sentence processing, or text comprehension can explain. To investigate such "hot" reading processes, standardized instruments that quantify both psycholinguistic and emotional variables at the sublexical, lexical, inter-, and supralexical levels (e.g., phonological iconicity, word valence, arousal-span, or passage suspense) are necessary. One such instrument, the Berlin Affective Word List (BAWL) has been used in over 50 published studies demonstrating effects of lexical emotional variables on all relevant processing levels (experiential, behavioral, neuronal). In this paper, we first present new data from several BAWL studies. Together, these studies examine various views on affective effects in reading arising from dimensional (e.g., valence) and discrete emotion features (e.g., happiness), or embodied cognition features like smelling. Second, we extend our investigation of the complex issue of affective word processing to words characterized by a mixture of affects. These words entail positive and negative valence, and/or features making them beautiful or ugly. Finally, we discuss tentative neurocognitive models of affective word processing in the light of the present results, raising new issues for future studies.
Background: In German breast cancer care, the S1-guidelines of the 1990s were substituted by national S3-guidelines in 2003. The application of guidelines became mandatory for certified breast cancer centers. The aim of the study was to assess guideline adherence according to time intervals and its impact on survival.
Methods: Women with primary breast cancer treated in three rural hospitals of one German geographical district were included. A cohort study design encompassed women from 1996–97 (N = 389) and from 2003–04 (N = 488). Quality indicators were defined along inpatient therapy sequences for each time interval and distinguished as guideline-adherent and guideline-divergent medical decisions. Based on all of the quality indicators, a binary overall adherence index was defined and served as a group indicator in multivariate Cox-regression models. A corrected group analysis estimated adjusted 5-year survival curves.
Results: From a total of 877 patients, 743 (85 %) and 504 (58 %) were included to assess 104 developed quality indicators and the resuming binary overall adherence index. The latter significantly increased from 13–15 % (1996–97) up to 33–35 % (2003–04). Within each time interval, no significant survival differences of guideline-adherent and -divergent treated patients were detected. Across time intervals and within the group of guideline-adherent treated patients only, survival increased but did not significantly differ between time intervals. Across time intervals and within the group of guideline-divergent treated patients only, survival increased and significantly differed between time intervals.
Conclusions: Infrastructural efforts contributed to the increase of process quality of the examined certified breast cancer center. Paradoxically, a systematic impact on 5-year survival has been observed for patients treated divergently from the guideline recommendations. This is an indicator for the appropriate application of guidelines. A maximization of guideline-based decisions instead of the ubiquitous demand of guideline adherence maximization is advocated.
Untreated periodontal disease may influence general health. However, how may a physician, who is not trained in periodontal probing, detect untreated periodontitis?
Activated matrix metalloproteinase-8 (aMMP-
8) in saliva correlates with periodontal probing parameters. Thus, sensitivity and specificity of a chair-side test for aMMP-8 to detect periodontitis were evaluated. Thirty cases [untreated chronic periodontitis (ChP); 15 generalized moderate and 15 generalized severe] and 30 controls [probing depths (PD) ≤3 mm, vertical probing attachment level (PAL-V) ≤2 mm at <30 % of sites) were examined periodontally (PD, PAL-V, bleeding on probing). Subsequently, the aMMP-8 test was performed. The test kit
becomes positive with ≥25 ng/ml aMMP-8 in the sample.
The aMMP-8 test was positive in 87 % of ChP and in 40 % of controls. That corresponds to a sensitivity of 87 % and a specificity of 60 %. The sensitivity to detect generalized severe ChP was 93 % (60 % specificity). Backward
stepwise logistic regression analysis to explain positive
aMMP-8 tests identified exclusively ChP with an odds of 9.8 (p < 0.001). Positive results of the aMMP-8 test significantly correlate with generalized ChP. The aMMP-8 test may be used by physicians to detect periodontitis in their patients.
The endocannabinoids (EC), their synthetizing and metabolizing enzymes, and the cannabinoid (CB) receptors comprise the endocannabinoid system (ECS) that has been detected by Yasuo et al. (2010) in rodent and human brain areas essential for circadian rhythmic control and hormone secretion. The EC are secreted in the pars tuberalis formation (PT) of the pituitary gland and unfold their effect as ligands on cannabinoid receptors type 1 (CB1) in the pars distalis (PD). The CB1 is mostly expressed on folliculo-stellate (fs) cells of the PD. The fs cells execute regulative and supportive functions to adjacent hormone-producing cells (Allaerts and Venkelecom, 2005; Mitsuishi et al., 2013). The lipid and calcium binding protein Annexin A1 (Anx A1) and the cell membrane permeable compound nitric oxide (NO) have been detected in the fs cells (Woods et al., 1990; Devnath and Inoue, 2008). There are published findings indicating strong influence of Anx A1 and NO on hormone production (Taylor et al. 1993; Venkelecom et al, 1997). The hypothesis of this study is that the EC influence hormonal secretion by acting upon CB1 receptors on fs cells and thus activating or inhibiting Anx A1 and NO that directly affect adjacent glandular cells.
Prevalently cell models were used to carry out the experimental work. The TtT/GF and Tpit/F1 cell lines represent the fs cells, the AtT20/D16v stand for the ACTH-producing corticotroph (C) cells, and GH4C1 for the PRL-producing lactotroph (L) cells. Whenever comparison with an integrity model was possible tissue from C3H mice was used. Chemoluminescent and photometrical detection, enzyme-linked immunosorbent assay (ELISA), fluorescence-activated cell sorting (FACS), immunoblot (IB), immunocyto- and immunohisto-chemical analysis (ICC, IHC), in situ hybridization (ISH), and (q) PCR methods were used as assaying tools to investigate CB1, Anx A1, the Anx A1 receptor - Fpr-rs1, NO, ACTH, and PRL.
CB1 was detected on the fs, C, and L cell models. The presence of fatty acid amide hydrolase (FAAH, an EC degrading enzyme) was confirmed in the fs cells. Incubations of the fs cells with CB1 agonists (2-AG, AEA, WIN) and antagonist (otenabant) were performed and resulting increase of Anx A1, and inhibition of NO were detected. Anx A1 binding sites, known as formyl peptide like receptor – related sequence 1 (Fpr-rs1) were identified on the C and L cells. The hormone-producing cells were treated with a 2-AG, Anx A1, and NO and the resulting changes in the levels of ACTH and PRL were detected. Anx A1 acted stimulatory on ACTH in the C AtT20/D16v cell and inhibitory on PRL in the L GH4C1 cell. NO inhibited both ACTH and PRL release. Additional analysis of the levels of expression of mRNA for Anx A1 and Fpr-rs1 in murine PD tissue demonstrated that while the expression of the first was not influenced by time, the expression of the latter was activated during the subjective day.
The here presented study shows that EC influence the ACTH release stimulatory through activating Anx A1 and inhibiting NO. As for PRL, the EC unfold an inhibition through activating Anx A1, and stimulation through inhibiting NO. A clear regulatory linkeage between the EC and ACTH and PRL control is revealed, involving the fs cells with possible time-dependence.

The decay behavior of low-lying dipole states in 140Ce was investigated exploiting the γ3-setup at the HIγS facility using quasi-monochromatic photon beams. Branching ratios of individual excited states as well as average branching ratios to low-lying states have been extracted using γ – γ coincidence measurements. The comparison of the average branching ratios to QPM calculations shows a remarkable agreement between experiment and theory in the energy range from 5.0 to 8.5 MeV.
We provide a comprehensive classification of the proteoglycan gene families and respective protein cores. This updated nomenclature is based on three criteria: Cellular and subcellular location, overall gene/protein homology, and the utilization of specific protein modules within their respective protein cores. These three signatures were utilized to design four major classes of proteoglycans with distinct forms and functions: the intracellular, cell-surface, pericellular and extracellular proteoglycans. The proposed nomenclature encompasses forty-three distinct proteoglycan-encoding genes and many alternatively-spliced variants. The biological functions of these four proteoglycan families are critically assessed in development, cancer and angiogenesis, and in various acquired and genetic diseases where their expression is aberrant.
The dynamics of strange vector meson resonances (K* and K̄*) is investigated within the Parton-Hadron-String Dynamics (PHSD) transport approach. We present the time evolution of the production of K*− resonances from the QGP phase by quark fusion as well as from hadronic sources. We also give a brief overview of the modification of the K* through Kπ decay and K*N interaction in a hot and dense nuclear medium.
There are strong indications that particles containing secondary organic aerosol (SOA) exhibit amorphous solid or semi-solid phase states in the atmosphere. This may facilitate deposition ice nucleation and thus influence cirrus cloud properties. However, experimental ice nucleation studies of biogenic SOA are scarce. Here, we investigated the ice nucleation ability of viscous SOA particles.
The SOA particles were produced from the ozone initiated oxidation of α-pinene in an aerosol chamber at temperatures in the range from −38 to −10 ◦C at 5–15 % relative humidity with respect to water to ensure their formation in a highly viscous phase state, i.e. semi-solid or glassy. The ice nucleation ability of SOA particles with different sizes was investigated with a new continuous flow diffusion chamber. For the first time, we observed heterogeneous ice nucleation of viscous α-pinene SOA in the deposition mode for ice saturation ratios between 1.3 and 1.4 significantly below the homogeneous freezing limit. The maximum frozen fractions found at temperatures between −36.5 and −38.3 °C ranged from 6 to 20 % and did not depend on the particle surface area. Global modelling of monoterpene SOA particles suggests that viscous biogenic SOA particles are indeed present in regions where cirrus cloud formation takes place. Hence, they could make up an important contribution to the global ice nuclei (IN) budget.
The banking system is highly interconnected and these connections can be conveniently represented as an interbank network. This survey presents a systematic overview of the recent advances in the theoretical literature on interbank networks. We assess our current understanding of the structure of interbank networks, of how network characteristics affect contagion in the banking system and of how banks form connections when faced with the possibility of contagion and systemic risk. In particular, we highlight how the theoretical literature on interbank networks offers a coherent way of studying interconnections, contagion processes and systemic risk, while emphasizing at the same time the challenges that must be addressed before general results on the link between the structure of the interbank network and financial stability can be established. The survey concludes with a discussion of the policy relevance of interbank network models with a special focus on macroprudential policies and monetary policy.

Obwohl derartige Praktiken mit dem Grundsatz der Geschlechtergleichheit und den Grundlagen einer offenen Bürgergesellschaft kollidieren, spricht sich die Verf. im Ergebnis gegen ein strafrechtliches Verbot aus.
La distinzione fra apollineo e dionisiaco è ritornata di moda grazie a Friedrich Nietzsche, che se ne è servito nella sua famosa opera La nascita della tragedia dallo spirito della musica. Questo scritto, tuttavia, non persegue affatto l’intento di contribuire a comprendere questi concetti, ma si serve di questa distinzione per spiegare due aspetti di colonialismo d’insediamento in forma di stato, con particolare riferimento all’esempio dell’Africa Sud Occidentale e della sua capitale Windhoek. Come è noto, Apollo veniva considerato il Dio del sole e della ragione, mentre Dionisio era visto come il Dio dell’ebbrezza e dell’estasi. Nel presente contesto, l’apollineo rappresenta il sogno utopico di potere di stato coloniale, comportante il diritto assoluto all’uso della forza, a giudicare, a proteggere ed a praticare una politica attentamente pianificata, mentre il dionisiaco rappresenta, grosso modo, la mentalità pionieristica dei coloni e le loro tendenze anomiche, derivanti alla fin fine dall’illegittimità incontrastata dell’intero progetto coloniale. Nello stesso tempo questo scritto si avvale di un altro contrasto: giorno e notte. È usato in senso metaforico – ma non esclusivamente. E mentre il primo rispecchia “il regno della luce”, basato sul potere dello stato e su una vita pubblica che evidenzia le caratteristiche di società civile, la seconda rappresenta la fase del giorno in cui la notte scende ed il controllo da parte dello stato viene a cessare del tutto, mentre la «vita coloniale sotterranea» si risveglia.
Sentences with doubly center-embedded relative clauses in which a verb phrase (VP) is missing are sometimes perceived as grammatical, thus giving rise to an illusion of grammaticality. In this paper, we provide a new account of why missing-VP sentences, which are both complex and ungrammatical, lead to an illusion of grammaticality, the so-called missing-VP effect. We propose that the missing-VP effect in particular, and processing difficulties with multiply center-embedded clauses more generally, are best understood as resulting from interference during cue-based retrieval. When processing a sentence with double center-embedding, a retrieval error due to interference can cause the verb of an embedded clause to be erroneously attached into a higher clause. This can lead to an illusion of grammaticality in the case of missing-VP sentences and to processing complexity in the case of complete sentences with double center-embedding. Evidence for an interference account of the missing-VP effect comes from experiments that have investigated the missing-VP effect in German using a speeded grammaticality judgments procedure. We review this evidence and then present two new experiments that show that the missing-VP effect can be found in German also with less restricting procedures. One experiment was a questionnaire study which required grammaticality judgments from participants without imposing any time constraints. The second experiment used a self-paced reading procedure and did not require any judgments. Both experiments confirm the prior findings of missing-VP effects in German and also show that the missing-VP effect is subject to a primacy effect as known from the memory literature. Based on this evidence, we argue that an account of missing-VP effects in terms of interference during cue-based retrieval is superior to accounts in terms of limited memory resources or in terms of experience with embedded structures.
Haematopoietic stem cells (HSCs) require the right composition of microRNAs (miR) for proper life-long balanced blood regeneration. Here we show a regulatory circuit that prevents excessive HSC self-renewal by upregulation of miR-193b upon self-renewal promoting thrombopoietin (TPO)-MPL-STAT5 signalling. In turn, miR-193b restricts cytokine signalling, by targeting the receptor tyrosine kinase c-KIT. We generated a miR-193b knockout mouse model to unravel the physiological function of miR-193b in haematopoiesis. MiR-193b−/− mice show a selective gradual enrichment of functional HSCs, which are fully competent in multilineage blood reconstitution upon transplantation. The absence of miR-193b causes an accelerated expansion of HSCs, without altering cell cycle or survival, but by decelerating differentiation. Conversely, ectopic miR-193b expression restricts long-term repopulating HSC expansion and blood reconstitution. MiR-193b-deficient haematopoietic stem and progenitor cells exhibit increased basal and cytokine-induced STAT5 and AKT signalling. This STAT5-induced microRNA provides a negative feedback for excessive signalling to restrict uncontrolled HSC expansion.
Interleukin (IL)-22 is a STAT3-activating cytokine displaying characteristic AU-rich elements (ARE) in the 3'-untranslated region (3'-UTR) of its mRNA. This architecture suggests gene regulation by modulation of mRNA stability. Since related cytokines undergo post-transcriptional regulation by ARE-binding tristetraprolin (TTP), the role of this destabilizing protein in IL-22 production was investigated. Herein, we demonstrate that TTP-deficient mice display augmented serum IL-22. Likewise, IL-22 mRNA was enhanced in TTP-deficient splenocytes and isolated primary T cells. A pivotal role for TTP is underscored by an extended IL-22 mRNA half-life detectable in TTP-deficient T cells. Luciferase-reporter assays performed in human Jurkat T cells proved the destabilizing potential of the human IL-22-3'-UTR. Furthermore, overexpression of TTP in HEK293 cells substantially decreased luciferase activity directed by the IL-22-3'-UTR. Transcript destabilization by TTP was nullified upon cellular activation by TPA/A23187, an effect dependent on MEK1/2 activity. Accordingly, IL-22 mRNA half-life as determined in TPA/A23187-stimulated Jurkat T cells decreased under the influence of the MEK1/2 inhibitor U0126. Altogether, data indicate that TTP directly controls IL-22 production, a process counteracted by MEK1/2. The TTP-dependent regulatory pathway described herein likely contributes to the role of IL-22 in inflammation and cancer and may evolve as novel target for pharmacological IL-22 modulation.
We have analyzed the distribution of mitochondrial contact site and cristae organizing system (MICOS) complex proteins and mitochondrial intermembrane space bridging complex (MIB) proteins over (sub)complexes and over species. The MICOS proteins are associated with the formation and maintenance of mitochondrial cristae. Indeed, the presence of MICOS genes in genomes correlates well with the presence of cristae: all cristae containing species have at least one MICOS gene and cristae-less species have none. Mic10 is the most widespread MICOS gene, while Mic60 appears be the oldest one, as it originates in the ancestors of mitochondria, the proteobacteria. In proteobacteria the gene occurs in clusters with genes involved in heme synthesis while the protein has been observed in intracellular membranes of the alphaproteobacterium Rhodobacter sphaeroides. In contrast, Mic23 and Mic27 appear to be the youngest MICOS proteins, as they only occur in opisthokonts. The remaining MICOS proteins, Mic10, Mic19, Mic25 and Mic12, the latter we show to be orthologous to human C19orf70/QIL1, trace back to the root of the eukaryotes. Of the remaining MIB proteins, also DNAJC11 shows a high correlation with the presence of cristae. In mitochondrial protein complexome profiles, the MIB complex occurs as a defined complex and as separate subcomplexes, potentially reflecting various assembly stages. We find three main forms of the complex: A) The MICOS complex, containing all the MICOS proteins, B) a membrane bridging subcomplex, containing in addition SAMM50, MTX2 and the previously uncharacterized MTX3, and C) the complete MIB complex containing in addition DNAJC11 and MTX1.
