In the mid-1990s, institutional investors entered the syndicated loan market and started to serve borrowers as lead arrangers. Why are non-banks able to compete for this role against banks? How do the composition of syndicates and loan pricing differ among lead arrangers? By using a dataset of 12,847 leveraged loans between 1997 and 2012, I aim to answer these questions. Non-banks benefit from looser regulatory requirements, have industry expertise which helps them in the screening and monitoring of borrowers and focus on firms that ask for loans only instead of additional cross-selling of other services. I can show that non-banks specialize on more opaque and less experienced borrowers, are more likely than banks to choose participants that help to reduce potentially higher information asymmetries and earn 105 basis points more than banks.
Although banks are at the center of systemic risk, there are other institutions that contribute to it. With the publication of the leveraged lending guideline in March 2013, the U.S. regulators show that they are especially worried about the private equity firms with their high-risk deals. Given these risks and the interconnectedness of the banks through the LBO loan syndicates, I shed light on the impact of a bank’s LBO loan exposure on its systemic risk. By using 3,538 observations between 2000 and 2013 from 165 global banks, I show that banks with higher LBO exposure also have a higher level of systemic risk. Other loan purposes do not show this positive relationship. The main drivers influencing this relationship positively are the bank’s interconnectedness to other LBO financing banks and its size. Lending experience with a specific PE sponsor, experience with leading LBO syndicates or a bank’s credit rating, however, lead to a lower impact of the LBO loan exposure on systemic risk.
Do economic fluctuations change the labour market attachment of mothers? How is the reentry process into the labour market after childbirth dependent on the country context women live in? Are these processes affected by occupational status? We address these questions using data from the National Longitudinal Study of Youth and the German Life History Study. Event history analyses demonstrate that in Germany and the United States, mothers who work in high occupational status jobs before birth return more quickly to their jobs and are less likely to interrupt their careers. During legally protected leave periods, mothers return at higher rates, exemplifying that family leaves strengthen mothers’ labour force attachment. Economic fluctuations mediate this latter finding, with different consequences in each country. In the United States, mothers tend to return to their jobs faster when unemployment is high. In Germany, mothers on family leave tend to return to their jobs later when unemployment is high. The cross-national comparison shows how similar market forces create distinct responses in balancing work and care.
The prefix cyber, prepended onto terms like war, peace, security, and so on, results in interesting word combinations which we construct with our spoken language. Many scholars, from political to social science, have discussed the terms and the semantics of it in order to understand the problem and to create some scientific value out of it. But this article will not be another endless discussion on whether cyberfoo exists somewhere in any computer network at the moment or not...
Objective: To investigate the accuracy, efficiency and radiation dose of a novel laser navigation system (LNS) compared to those of free-handed punctures on computed tomography (CT).
Materials and methods: Sixty punctures were performed using a phantom body to compare accuracy, timely effort, and radiation dose of the conventional free-handed procedure to those of the LNS-guided method. An additional 20 LNS-guided interventions were performed on another phantom to confirm accuracy. Ten patients subsequently underwent LNS-guided punctures.
Results: The phantom 1-LNS group showed a target point accuracy of 4.0 ± 2.7 mm (freehand, 6.3 ± 3.6 mm; p = 0.008), entrance point accuracy of 0.8 ± 0.6 mm (freehand, 6.1 ± 4.7 mm), needle angulation accuracy of 1.3 ± 0.9° (freehand, 3.4 ± 3.1°; p < 0.001), intervention time of 7.03 ± 5.18 minutes (freehand, 8.38 ± 4.09 minutes; p = 0.006), and 4.2 ± 3.6 CT images (freehand, 7.9 ± 5.1; p < 0.001). These results show significant improvement in 60 punctures compared to freehand. The phantom 2-LNS group showed a target point accuracy of 3.6 ± 2.5 mm, entrance point accuracy of 1.4 ± 2.0 mm, needle angulation accuracy of 1.0 ± 1.2°, intervention time of 1.44 ± 0.22 minutes, and 3.4 ± 1.7 CT images. The LNS group achieved target point accuracy of 5.0 ± 1.2 mm, entrance point accuracy of 2.0 ± 1.5 mm, needle angulation accuracy of 1.5 ± 0.3°, intervention time of 12.08 ± 3.07 minutes, and used 5.7 ± 1.6 CT-images for the first experience with patients.
Conclusion: Laser navigation system improved accuracy, duration of intervention, and radiation dose of CT-guided interventions.
Der Bundesgerichtshof hat mit Urteil vom 28. Januar 2015 entschieden, dass Kinder, die durch künstliche Befruchtung im Wege einer Samenspende gezeugt worden sind, gegen Reproduktionsmediziner und -kliniken einen Anspruch auf Auskunft über die Identität des Samenspenders haben können. Die Geltendmachung des Auskunftsanspruchs setzte kein bestimmtes Mindestalter der „Spenderkinder“ voraus. Der nachfolgende Beitrag analysiert die Konstruktion dieses Anspruchs vor dem Hintergrund eines durch neue Reproduktionstechnologien und gewandelte gesellschaftliche Vorstellungen veränderten Abstammungsrechts. Nach Methodenkritik und Rekonstruktion aus einer gesellschaftlich-institutionellen Perspektive eröffnen sich weitere Aussichten auf zukünftige Formen von Vaterschaft und ein entsprechend zu verwirklichendes Recht auf Kenntnis der eigenen Abstammung.
TO DERIVE OPTIMAL ORDER EXECUTION STRATEGIES THAT STRIVE TO MINIMIZE TRANSACTION COSTS, INVESTORS AS WELL AS AUTOMATED TRADING ENGINES MUST BE ABLE TO ANTICIPATE CHANGES IN THE AVAILABLE MARKET LIQUIDITY. BASED ON AN EVENT STUDY ON THE LIQUIDITY IMPACT OF AD-HOC DISCLOSURES, WE PROPOSE A NOVEL IT ARTIFACT THAT ALLOWS AUTOMATED TRADING ENGINES TO APPROPRIATELY REACT TO NEWS-RELATED LIQUIDITY SHOCKS. FURTHERMORE, WE PROVIDE A SIMULATIONBASED EVALUATION THAT SHOWS ITS ECONOMIC RELEVANCE.
Based on a cognitive notion of neo-additive capacities reflecting likelihood insensitivity with respect to survival chances, we construct a Choquet Bayesian learning model over the life-cycle that generates a motivational notion of neo-additive survival beliefs expressing ambiguity attitudes. We embed these neo-additive survival beliefs as decision weights in a Choquet expected utility life-cycle consumption model and calibrate it with data on subjective survival beliefs from the Health and Retirement Study. Our quantitative analysis shows that agents with calibrated neo-additive survival beliefs (i) save less than originally planned, (ii) exhibit undersaving at younger ages, and (iii) hold larger amounts of assets in old age than their rational expectations counterparts who correctly assess their survival chances. Our neo-additive life-cycle model can therefore simultaneously accommodate three important empirical findings on household saving behavior.
Chronic inflammation as an important epigenetic and environmental factor for putative tumorigenesis and tumor progression may be associated with specific activation of Toll-like receptors (TLR). Recently, carcinogenesis has been suggested to be dependent on TLR7 signaling. In the present study, we determined the role of both TLR7 and TLR8 expression and signaling in tumor cell proliferation and chemoresistance in pancreatic cancer. Expression of TLR7/TLR8 in UICC stage I-IV pancreatic cancer, chronic pancreatitis, normal pancreatic tissue and human pancreatic (PANC1) cancer cell line was examined. For in vitro/in vivo studies TLR7/TLR8 overexpressing PANC1 cell lines were generated and analyzed for effects of (un-)stimulated TLR expression on tumor cell proliferation and chemoresistance. TLR expression was increased in pancreatic cancer, with stage-dependent upregulation in advanced tumors, compared to earlier stages and chronic pancreatitis. Stimulation of TLR7/TLR8 overexpressing PANC1 cells resulted in elevated NF-κB and COX-2 expression, increased cancer cell proliferation and reduced chemosensitivity. More importantly, TLR7/TLR8 expression increased tumor growth in vivo. Our data demonstrate a stage-dependent upregulation of both TLR7 and TLR8 expression in pancreatic cancer. Functional analysis in human pancreatic cancer cells point to a significant role of both TLRs in chronic inflammation-mediated TLR7/TLR8 signaling leading to tumor cell proliferation and chemoresistance.
G-Protein-gekoppelte Rezeptoren (GPCR) sind im Immunsystem essentiell für die Verarbeitung von Signalen, die von Chemokinen, Lipiden und anderen Botenstoffen ausgehen. Ihre Existenz gewährleistet, dass Leukozyten sowohl unter physiologischen als auch unter pathophysiologischen Umständen ihren Funktionen als Immunzellen nachkommen können. Grundlegend wichtig für das angeborene Immunsystem sind die GPCR, die die Weiterleitung ihrer Signale über G-Proteine vom Typ Gi vermitteln. Die Migration, Adhäsion und Differenzierung von Leukozyten wird jedoch auch maßgeblich durch G12/13-gekoppelte Rezeptoren reguliert, wobei die kleine GTPase RhoA als Effektormolekül eine wichtige Rolle spielt. Die Bedeutung der G12/13-gekoppelten Signaltransduktion in Makrophagen ist allerdings weitgehend unverstanden. Mit Hilfe einer Mauslinie, in der speziell und ausschließlich in myeloiden Zellen die beiden G-Protein-Untereinheiten Gα12 und Gα13 durch ein konstitutiv aktives Cre-Rekombinase-System inaktiviert wurden (Lys-Gα12/Gα13-KO), sollte nun die Funktion und der genaue Mechanismus des G12/13-gekoppelten Signalweges in Monozyten und Makrophagen aufgeklärt werden und somit neue Erkenntnisse zur Rolle der GPCR im Immunsystem gewonnen werden.
Eine erste Untersuchung der peripheren Immunzellpopulationen des Lys-Gα12/Gα13-KO ergab, dass residente Gewebemakrophagen, im Speziellen die des Peritoneums, in ihrer Anzahl erhöht sind. In einer vertieften Analyse der residenten peritonealen Makrophagen (rpMP) konnte gezeigt werden, dass der Verlust von Gα12/13 zu Veränderung im Zytoskelett der Makrophagen führt. Die Zellen entwickeln einen Phänotyp mit besonders langen und verzweigten Filopodien und zeigen Ein-schränkungen in ihrer basalen Beweglichkeit.
Über diesen morphologischen Befund hinaus, konnte in einer Studie zur Gen-expression in diesen Zellen festgestellt werden, dass Gα12/13-defiziente Makrophagen verstärkt proinflammatorische Gene wie Nos2, die Cyclooxygenase 2 aber auch verschiedene Chemokine wie Cxcl10 oder Cytokine wie Il-6 oder Tnfα exprimieren. Ein ähnlicher Phänotyp in Bezug auf Morphologie und Genexpression wurde bei der Untersuchung von Makrophagen, die aus Knochenmark des Lys-Gα12/Gα13-KO generiert wurden, beobachtet.
Als vermutlich verantwortlicher G12/13-gekoppelter Rezeptor konnte der S1P-Rezeptor-subtyp 2 (S1P2) identifiziert werden. Mit Hilfe von Inhibitoren für die G12/13-gekoppelte Signaltransduktionskaskade konnte gezeigt werden, dass über die kleine GTPase RhoA die NF-κB-abhängige Genaktivität reguliert werden kann. Vermutlich aktiviert RhoA dazu die Rho Kinase ROCK, die wiederum das untergeordnete Effektormolekül Rac1 hemmen kann. Im Falle des Lys-Gα12/Gα13-KO führt eine reduzierte Aktivierung von RhoA insgesamt zu einer eingeschränkten Hemmung dieses Signalweges und im Folgenden zu einer außer Kontrolle geratenen Induktion entzündungsrelevanter Gene und damit einhergehend auch zu einer Veränderung des Milieus in der Bauchhöhle dieser Tiere.
Obwohl die Immunantwort in diesen Tieren auf klassische Pathogene wie Lipopolylsaccharide (LPS) unverändert ist, konnte ein Anstieg an peritonealen B-Zellen festgestellt werden. Diese B-Zellen, insbesondere B1 B-Zellen, sind als wichtige Produzenten von natürlichen Antikörpern gegen endogene Pathogene bekannt. Die Analyse von Plasma aus Lys-Gα12/Gα13-KO-Mäusen ergab einen erhöhten Titer für natürliche Antikörper wie beispielsweise gegen oxidierte Formen von atherogenen Lipoproteinen. Diese Erkenntnis führte zu der Frage, ob die ursprünglich pro-inflammatorischen Veränderungen der peritonealen Makrophagen einen indirekten, positiven Einfluss auf die Entwicklung einer Atherosklerose haben können. Interessanterweise sind die Tiere des Lys-Gα12/Gα13-KO signifikant vor Atherosklerose geschützt und die Existenz der natürlichen Antikörper in atherosklerotischen Läsionen wird als Hinweis für ihre protektive Rolle im Krankheitsverlauf angesehen. In einem therapeutischen Ansatz mit peritonealen Zellen konnte in Atherosklerose-gefährdeten Tieren die Progression dieser Gefäßerkrankung eingedämmt werden.
Die hier durchgeführte Studie hat durch in vitro und in vivo Versuche mit Lys-Gα12/Gα13-KO-Mäusen dazu beitragen, das Verständnis der Rolle der G12/13-gekoppelten Signaltransduktion im Immunsystem zu verbessern.
Die Komplexität der verschiedenen Funktionen einzelner Effektormoleküle einerseits und die Interaktionen unterschiedlicher Immunzellpopulationen andererseits lassen jedoch vermuten, dass noch weitreichende Untersuchungen an GPCR und G-Proteinen nötig sind, um diese für den Organismus bedeutsamen Informationssysteme voll-ständig zu verstehen und weiter therapeutisch nutzbar zu machen.
In der vorliegenden Doktorarbeit zur Untersuchung der Rolle der Superoxid-Dismutasen in P. anserina lieferten die durchgeführten Analysen folgende Ergebnisse:
1)Sowohl in P. anserina als auch in S. cerevisiae wurde eine gemeinsame Regulation von SODs nachgewiesen: Stämme, die die mitochondriale MnSod (PaSod3 bzw. ScSod2) überexprimieren zeigen eine erhöhte Cu/ZnSOD-Aktivität (PaSOD1 bzw. ScSOD1).
2)Es konnte keine SOD-Aktivität für die putativen SODs Pa_1_10620, Pa_1_10630 und Pa_1_6300 detektiert werden. Für Pa_1_10620, dessen Überexpression unter Standardbedingungen zu einer Lebensverlängerung führt, wird eine Funktion als mitochondriales ribosomales Protein angenommen.
3)Der ∆PaSod3-Stamm weist keinen Unterschied im Phänotyp, der Wuchsrate und der Lebensspanne unter Standardbedingungen zum Wildtyp auf. Paraquat-Stress führt allerdings zu einer Kurzlebigkeit des ∆PaSod3-Stammes, wohingegen diese Mutante eine höhere Resistenz gegenüber Wasserstoffperoxid aufweist als der Wildtyp.
4)Transkriptomanalysen des Wildtyps und der ∆PaSod3-Mutante lassen vermuten, dass eine Hochregulation von Detoxifizierungs- und Energie-abhängigen Prozessen die durch den Verlust der mitochondrialen PaSOD3 vermuteten negativen Auswirkungen kompensieren.
5)PaSod3_OEx-Stämme weisen unter Standardbedingungen aufgrund der erhöhten intrazellulären Wasserstoffperoxid-Menge, bedingt durch die vermehrte Umsetzung von Superoxid, diverse negative Auswirkungen auf: Eine reduzierte Wuchsrate, verkürzte Lebensspanne, geringere Fertilität, stärkere Pigmentierung, vermehrt fragmentierte Mitochondrien, mehr unprozessierte mitochondriale Proteine und weniger Komplex IV der Atmungskette als der Wildtyp. Zusätzlich wird vermehrt über die alternative Oxidase geatmet, um die ROS-Generierung zu reduzieren.
6)Oxidativer Stress in Form von Paraquat führt in PaSod3_OEx-Stämmen zu einer weiteren Verkürzung der medianen Lebensspanne, während die maximale Lebensspanne von PaSod3_OEx3-Stämmen im Vergleich zum Wildtyp sogar verlängert ist. Wasserstoff-peroxid resultiert in stark verringerten medianen und maximalen Lebensspannen beider PaSod3-überexprimierenden Stämme.
7)Die Anzucht auf Medium mit zusätzlichem Mangan (80 µM MnSO4) kann die beobachteten Defekte der PaSod3_OEx-Stämme fast vollständig auf Wildtyp-Niveau revertieren: Die Wuchsrate, die Lebensspanne, der Phänotyp, die Mitochondrien-morphologie, die Prozessierung mitochondrialer Proteine und die Atmung entsprechen dem Wildtyp. Lediglich die Fertilität erreicht nicht das Wildtyp-Niveau. Diese positiven Effekte von Mangan werden erzielt, da die erhöhte Wasserstoffperoxid-Menge in PaSod3_OEx-Stämmen entsprechend ihrer Entstehung detoxifiziert wird, denn Mangan führt zu einer gesteigerten Transkription bzw. Aktivität von Katalasen und Peroxidasen sowie zu einer erhöhten Peroxiredoxin-Menge.
8)Die Anzucht des Wildtyps unter Wasserstoffperoxid-Stress resultiert in einer Lebens-spannenverkürzung. Diese kann durch Supplementation mit Mangan revertiert werden. Unter diesen Bedingungen weisen u. a. Peroxidasen eine erhöhte Aktivität auf.
Insgesamt ließen die gewonnenen Daten den Schluss zu, dass das Genom von P. anserina für drei aktive SODs kodiert. Ein Verlust der einzigen mitochondrial lokalisierten SOD kann durch die Induktion von Energie-abhängigen Prozessen sowie von Detoxifizierungsprozessen kompensiert werden. Ferner weisen die durchgeführten Studien darauf hin, dass PaSod3_OEx-Stämme als Modell für erhöhte intrazelluläre Wasserstoffperoxid-Mengen in P. anserina verwendet werden können. Darüber hinaus wurde ein Zusammenhang zwischen Mangan und dem Detoxifizierungs-netzwerk in P. anserina nachgewiesen. Dabei können zwei Mechanismen zur Reduktion der Wasserstoffperoxid-Mengen unterschieden werden: Bei Vorhandensein ausreichender Mengen Mangan kommt es zu einer stärkeren Detoxifizierung von Wasserstoffperoxid. Ist Mangan allerdings limitiert und die Detoxifizierung kann nicht gesteigert werden, wird eine Umstellung der Atmung eingeleitet, um die neu entstehende ROS-Menge zu minimieren.
Die vorliegende kumulative Dissertation befasst sich mit der Erfassung der Behandlungsintegrität bestehend aus psychotherapeutischer Adhärenz, Kompetenz sowie der Behandlungsdifferenzierung im Rahmen der Psychotherapieforschung. Die Überprüfung, ob Behandlungen bzw. Interventionen so wie intendiert durchgeführt wurden, ist für die Sicherstellung valider Schlussfolgerungen aus einer klinischen Studie von hoher Relevanz.
Die erste Studie untersucht, ob die Erfassung der Behandlungsintegrität ökonomischer gestaltbar ist. Es zeigte sich, dass Beurteilungen der Adhärenz und Kompetenz basierend auf Sitzungssegmenten im Vergleich zu ganzen Sitzungen keine Unterschiede aufweisen hinsichtlich Reliabilität, Validität und Prädiktion des Behandlungserfolgs.
In der zweiten Studie wird die Entwicklung und Validierung einer Adhärenz- und Kompetenzskala vorgestellt. Diese Studie weist zudem auf die Verwendung im Rahmen der Aus- und Weiterbildung von Therapeuten hin.
Die dritte Studie zeigt, dass in Psychotherapiestudien die im Vergleich stehenden Behandlungsbedingungen gut voneinander unterscheidbar sein müssen. Für die Beschreibung der Behandlungsdifferenzierung und -spezifität wurde der Behandlungs-Spezifitäts-Index entwickelt, dessen Eignung bestätigt werden konnte.
Die vierte Studie überprüft, ob sich erfolgreiche von nicht erfolgreichen Therapien hinsichtlich der psychotherapeutischen Kompetenz, Adhärenz und psychotherapeutischen Beziehung unterscheiden. Es zeigte sich, dass Adhärenz eine Voraussetzung für kompetentes Vorgehen darstellt. Kompetenz beeinflusst die psychotherapeutische Beziehung maßgebend, die mitentscheidend für den (Miss-)Erfolg einer Behandlung zu sein scheint.
Insgesamt tragen die Ergebnisse zu einer differenzierteren, spezifischeren und ökonomischeren Erfassung der Behandlungsintegrität innerhalb der Psychotherapieforschung bei. Gleichzeitig erweitern sie den Fokus auf neue Ansätze für zukünftige Forschungen.
This article discusses the interrelation between transculturality and transmediality with an emphasis on processes of translation. It focuses on two examples of transcultural and transmedial writing taken from contemporary Cuban literature in Paris: Miguel Sales's recontextualization of Cuban popular music in Paris and William Navarrete's ekphrastic reinscription of his island into the realm of French romantic painting. The case studies are significant in this context because they show how cultural borders are simultaneously set and transgressed at medial crossings—between music and poetry, text, and image. Thus, cultural translations go hand in hand with medial transpositions that include forms of rewriting, recomposition, and revisualization. The connection between moving cultures and moving media also points to the question of “travelling memory” in diaspora.
Im Laufe dieser Bachelor-Arbeit wurden verschiedene GEM-Anordnungen systematisch auf ihr IBF-Verhalten hin untersucht. Neben der Reproduktion zuvor durchgeführter Messungen wurden auch neue GEM-Kombinationen getestet. Insbesondere lag der Fokus darauf, eine Verbesserung des IBFs gegenüber des Baseline-Setups zu erzielen. Dabei kamen neben der bisher verwendeten S und LP Folien auch SP Folien zum Einsatz. Die Messungen brachten jedoch kein Ergebnis hervor, welches als Verbesserung gegenüber der Ausgangslage angesehen werden könnte. Da mit SP GEMs zuvor wenig gearbeitet wurde, war es unter anderem ein Ziel, zu untersuchen, wie sich die Verwendung dieser GEMs auf den IBF auswirkt. Insbesondere war die Frage zu klären, ob durch ihre Verwendung der IBF des Baseline-Setups
verbessert werden kann. Zum besseren Verständnis wurde ebenfalls eine Variante, S-S-LPS, untersucht. Für dieses Setup konnte durch die Verwendung einer SP Folie auf Position 4 eine Verbesserung des IBF bewirkt werden, für das Baseline-Setup jedoch nicht. Ein wesentliches Ergebnis dieser Bachelor-Arbeit war, dass das Alignment der GEMs, entgegen bisheriger Annahmen, eine große praktische Relevanz hat. Die relative Orientierung zweier aufeinander folgender GEMs gleichen Lochabstands zueinander hat einen großen Ein
uss auf die lokale Ionentransmission. Eine genauere Untersuchung hat ergeben, dass man dem entgegenwirken kann, indem man aufeinander folgende GEMs um 90° gedreht einbaut. Aufgrund der Geometrie der Folien verhindert man dadurch, dass sich die Löcher zweier Folien direkt ßber- bzw. untereinander anordnen. Ein solcher Aufbau konnte durch eine geringfügige Modifikation der Testkammer erreicht werden.
Mit diesem veränderten Aufbau wäre es nun das Ziel gewesen, alle bisherigen Messungen zu wiederholen und auf Reproduzierbarkeit hin zu überprüfen. Die Wiederholung einer Messreihe mit um 90° gedrehten GEMs hat im Rahmen der Fehlertoleranzen reproduzierbare
Ergebnisse geliefert. Aus zeitlichen Gründen war es jedoch im Rahmen dieserArbeit nicht möglich, eine vollständige Wiederholung aller Messungen durchzuführen. Dies wurde zu einem späteren Zeitpunkt von anderen Personen getan.
Afghanistans ehemalige Mudschahedin haben es geschafft: Kritik an „heiligen Kriegern“, wie sie sich nun nennen, ist gleichzeitig Kritik am Islam, und das ist brandgefährlich in ihrem Land. Manch ein Kriegsverbrecher nutzt das Islam- Argument, um sich über das staatliche Gesetz zu stellen und damit unangreifbar zu machen. Säkulare Gruppierungen, die in der urbanen Bevölkerung weiter bestehen, würden es heute nicht mehr wagen, sich öffentlich so zu bezeichnen. In Regierung, Justiz und Gesellschaft ist der Druck, sich zum Islam zu bekennen, groß. Eine zuweilen absurd anmutende Konkurrenz darüber, wer am ‚islamischten‘ ist, führt zu immensem Druck auf Medien und Zivilgesellschaft, insbesondere auf Frauenrechtsgruppen, sich innerhalb des Islams zu positionieren und sich abzugrenzen von ‚unislamischen‘ Werten. Seit einigen Jahren dreht sich beispielsweise eine erhitzte gesellschaftliche Debatte um ein Gewaltschutzgesetz für Frauen, begleitet von einer medialen und religiösen Kampagne gegen Frauenschutzhäuser:
im Kern wird beiden vorgeworfen, antiislamisch zu sein. Frauenrechtsgruppen sehen sich gezwungen, juristisch und religiös zu begründen, dass das Gesetz keine Anteile aufweist, die dem Islam widersprechen.
Der hohe Rat der Ulema, 2002 von der Regierung eingesetzt und bezahlt, stellt die größte und einflussreichste religiöse Struktur in Afghanistan dar: er besteht aus 3000 Ulema und Mullas1 (davon ¾ Sunniten und ¼ Schiiten); viele sind gleichzeitig auch Richter, politische Berater, Lehrer oder Imame. Die meisten von ihnen gehören einer der Mudschahedin-Gruppierungen an. Auf nationaler Ebene berät der Rat die Regierung in religiösen Fragen, unterstützt zumeist ihre Entscheidungen und gewährt ihr so eine religiöse Legitimation; auf lokaler Ebene positionieren sich die Ulema und Mullas allerdings oft regierungskritisch und anti-westlich. In der gesellschaftlichen Debatte über Frauenrechte äußerten sie sich 2012 extrem konservativ, indem sie erklärten, Frauen seien weniger wert als Männer, sollten nicht ohne mahram (männlichen Verwandten) verreisen und bei Arbeit, Bildung und Freizeit den Kontakt zu Männern vermeiden. Ihre monatlichen Erklärungen auf nationaler Ebene sowie ihre Ansprachen in lokalen Moscheen haben großen Einfluss auf die gesellschaftliche Verhandlung von Normen. 2 Auch Saudi-Arabien versucht wachsenden Einfluss auf die religiöse Ausbildung in Afghanistan auszuüben, so z.B. durch den Bau und Betrieb eines religiösen Schulungszentrums in Kabul.3 Pakistanische religiöse Gelehrte sehen den Kampf der afghanischen Taliban gegen die westliche Intervention als berechtigt an, und erklären USA und NATO als allein verantwortlich für jegliche zivilen Opfer.
The degradation of natural forests to modified forests threatens subtropical and tropical biodiversity worldwide. Yet, species responses to forest modification vary considerably. Furthermore, effects of forest modification can differ, whether with respect to diversity components (taxonomic or phylogenetic) or to local (α-diversity) and regional (β-diversity) spatial scales. This real-world complexity has so far hampered our understanding of subtropical and tropical biodiversity patterns in human-modified forest landscapes. In a subtropical South African forest landscape, we studied the responses of three successive plant life stages (adult trees, saplings, seedlings) and of birds to five different types of forest modification distinguished by the degree of within-forest disturbance and forest loss. Responses of the two taxa differed markedly. Thus, the taxonomic α-diversity of birds was negatively correlated with the diversity of all plant life stages and, contrary to plant diversity, increased with forest disturbance. Conversely, forest disturbance reduced the phylogenetic α-diversity of all plant life stages but not that of birds. Forest loss neither affected taxonomic nor phylogenetic diversity of any taxon. On the regional scale, taxonomic but not phylogenetic β-diversity of both taxa was well predicted by variation in forest disturbance and forest loss. In contrast to adult trees, the phylogenetic diversity of saplings and seedlings showed signs of contemporary environmental filtering. In conclusion, forest modification in this subtropical landscape strongly shaped both local and regional biodiversity but with contrasting outcomes. Phylogenetic diversity of plants may be more threatened than that of mobile species such as birds. The reduced phylogenetic diversity of saplings and seedlings suggests losses in biodiversity that are not visible in adult trees, potentially indicating time-lags and contemporary shifts in forest regeneration. The different responses of taxonomic and phylogenetic diversity to forest modifications imply that biodiversity conservation in this subtropical landscape requires the preservation of natural and modified forests.
El propósito de este artículo es discutir, en un primero momento, en que medida los maestros vinculados a la Universidad de Salamanca y a su correspondiente Escuela de Salamanca contribuyeron para la validación de un saber relacionado a los descubrimientos que permitió pensar una nueva configuración geográfica de la Tierra. En un segundo momento, mostraremos como la universidad salmantina, junto con otras instituciones de saber, operaron como un centro de actividad ‘científica’ que estuve a servicio de los proyectos de la monarquía española.
Background: Sphingolipids constitute bioactive molecules with functional implications in liver homeostasis. Particularly, ablation of very long chain ceramides in a knockout mouse model has been shown to cause a severe hepatopathy.
Methods: We aimed to evaluate the serum sphingolipid profile of 244 patients with cirrhosis prospectively followed for a median period of 228±217 days via mass spectrometry.
Results: We thereby observed a significant decrease of long and very long chain ceramides, particularly of C24ceramide, in patients with increasing severity of cirrhosis (p<0.001). Additionally, hydropic decompensation, defined by clinical presentation of ascites formation, was significantly correlated to low C24ceramide levels (p<0.001) while a significant association to hepatic decompensation and poor overall survival was observed for low serum concentrations of C24ceramide (p<0.001) as well. Multivariate analysis further identified low serum C24ceramide to be independently associated to overall survival (standard beta = -0.001, p = 0.022).
Conclusions: In our current analysis serum levels of very long chain ceramides show a significant reciprocal correlation to disease severity and hepatic decompensation and are independently associated with overall survival in patients with cirrhosis. Serum sphingolipid metabolites and particularly C24ceramide may constitute novel molecular targets of disease severity, hepatic decompensation and overall prognosis in cirrhosis and should be further evaluated in basic research studies.
We previously reported that aberrant HH pathway activation confers a poor prognosis in rhabdomyosarcoma (RMS). Searching for new treatment strategies we therefore targeted HH signaling. Here, we identify a novel synthetic lethality of concomitant inhibition of HH and PI3K/AKT/mTOR pathways in RMS by GLI1/2 inhibitor GANT61 and PI3K/mTOR inhibitor PI103. Synergistic drug interaction is confirmed by calculation of combination index (CI < 0.2). Similarly, genetic silencing of GLI1/2 significantly increases PI103-induced apoptosis. GANT61 and PI103 also synergize to induce apoptosis in cultured primary RMS cells emphasizing the clinical relevance of this combination. Importantly, GANT61/PI103 cotreatment suppresses clonogenic survival, three-dimensional sphere formation and tumor growth in an in vivo model of RMS. Mechanistic studies reveal that GANT61 and PI103 cooperate to trigger caspase-dependent apoptosis via the mitochondrial pathway, as demonstrated by several lines of evidence. First, GANT61/PI103 cotreatment increases mRNA and protein expression of NOXA and BMF, which is required for apoptosis, since knockdown of NOXA or BMF significantly reduces GANT61/PI103-induced apoptosis. Second, GANT61/PI103 cotreatment triggers BAK/BAX activation, which contributes to GANT61/PI103-mediated apoptosis, since knockdown of BAK provides protection. Third, ectopic expression of BCL-2 or non-degradable phospho-mutant MCL-1 significantly rescue GANT61/PI103-triggered apoptosis. Fourth, GANT61/PI103 cotreatment initiate activation of the caspase cascade via apoptosome-mediated cleavage of the initiator caspase-9, as indicated by changes in the cleavage pattern of caspases (e.g. accumulation of the caspase-9 p35 cleavage fragment) upon addition of the caspase inhibitor zVAD.fmk. Thus, combined GLI1/2 and PI3K/mTOR inhibition represents a promising novel approach for synergistic apoptosis induction and tumor growth reduction with implications for new treatment strategies in RMS.

Zwei Frankfurter Arbeitsgruppen gelang es vor zehn Jahren erstmals, das Tier genetisch mit lichtaktivierbaren Ionenkanälen auszustatten und seine Bewegungen mit Licht zu steuern. Inzwischen studieren Forscher an dem durchsichtigen Wurm auch Prozesse, die für die medizinische Forschung bedeutsam sind – etwa die Entstehung und Behandlung genetisch bedingter Herz-Rhythmus-Störungen.
This paper undertakes a quantitative investigation of the effects of anticipated inflation on the distribution of household wealth and welfare. Consumer Finance Data on household financial wealth suggests that about a third of the US population holds all its financial assets in transaction accounts. The remaining two-third of the US population holds most of their financial assets outside transaction accounts. To account for this evidence, I introduce a portfolio choice in a standard incomplete markets model with heterogeneous agents. I calibrate the model economy to SCF 2010 US data and use this environment to study the distributive effects of changes in anticipated inflation. An increase in anticipated inflation leads households to reshuffle their portfolio towards real assets. This crowding-in of supply for real assets lowers equilibrium interest rates and thereby redistributes wealth from creditors to borrowers. Because borrowers have a higher marginal utility, this redistribution improves aggregate welfare. First, this paper shows that inflation acts not only a regressive consumption tax as in Erosa and Ventura (2002), but also as a progressive tax. Second, this paper shows that the welfare cost of inflation are even lower than the estimates computed by Lucas (2000) and Ireland (2009). Finally, this paper offers insights into why deflationary environments should be avoided.
Du, er und sie, wir, ihr und die Anderen, einige, viele, alle – und dazwischen Ich.
Wie können ästhetische Zugangsweisen und sinnlich-kreative Lernmethoden in den gesellschaftswissenschaftlichen Fächern aussehen? Vielleicht denken wir auf Anhieb an Rollenspiele im Geschichtsunterricht, Karten zeichnen in Erdkunde, geistliche Lieder im Religionsunterricht, eine Wahlsimulation in Politik und Wirtschaft – doch wie bald gehen uns die Ideen aus? Nun ist ein ästhetischer Zugang aber mehr als eine Sammlung von Methoden und Projekten; vielmehr kennzeichnet ihn ein bestimmter Blick auf die Inhalte des Unterrichts und auf die Menschen, die an diesem mitwirken. Einen solchen Blick zu gewinnen, ist Ziel des Workshops.
Das klingt sehr grundsätzlich – und ist daher am besten möglichst konkret zu erschließen. Thematisch heißt das in diesem Fall die Beschäftigung mit dem Leben zwischen Individualität und Sozialität, Selbstbestimmung und Abhängigkeit, mit Fragen von Identität(en), Alterität(en), Gruppen und sozialen Kontexten.
Methodisch meint "Beschäftigung" dabei durchaus auch ein Reflektieren, in erster Linie aber wird das unmittelbare Erfahren, Ausprobieren und Erfinden im Mittelpunkt stehen – z.B. beim Vortragen, Inszenieren, Zeichnen, Erzählen, Gestalten oder Spielen.
Der erwähnte, ganz besondere Blick auf die Dinge und Menschen verlangt nämlich nicht nur ein Beobachten derselben von außen, sondern ein tätiges Miteinander – dazwischen Ich!
In honeybees, reproductive females usually mate early in their life with more than 10 males in free flight, often within 10 minutes, and then store male gametes for up to five years. Because of the extreme polyandry and mating in free flight special adaptations in males are most likely. We present here the results of an investigation of the protein content of four types of male reproductive glands from the Western honeybee (Apis mellifera) drone, namely seminal vesicles (secretion in ejaculate), as well as bulbus, cornua and mucus glands (secretions for the mating plug). Using high resolution and accuracy mass spectrometry and a combination of database searching and de novo sequencing techniques it was possible to identify 50 different proteins in total, inside all mentioned glands, except in the mucus gland. Most of the proteins are unique for a specific gland type, only one of them (H9KEY1/ATP synthase subunit O) was found in three glands, and 7 proteins were found in two types of glands. The identified proteins represent a wide variety of biological functions and can be assigned to several physiological classes, such as protection, energy generation, maintaining optimal conditions, associated mainly with vesicula seminalis; signaling, cuticle proteins, icarpin and apolipoproteins located mainly in the bulbus and cornua glands; and some other classes. Most of the discovered proteins were not found earlier during investigation of semen, seminal fluid and tissue of reproductive glands of the bee drone. Moreover, we provide here the origin of each protein. Thus, the presented data might shed light on the role of each reproductive gland.
Este artículo presenta una lectura crítica de un trabajo central de Axel Honneth desde la teoría de la sujeción de Judith Butler. Intenta mostrar que, por la ausencia en su escrito de una consideración sobre el poder, el pensador alemán no logra cumplir satisfactoriamente su objetivo propuesto de enfrentar las posturas que cuestionan el potencial crítico del reconocimiento. La hipótesis que aquí se maneja es que esa ausencia está ligada a su definición del reconocimiento como lo contrario de las prácticas de dominio o sometimiento. Ahora bien, Honneth afirma que el escepticismo de esas posturas respecto del reconocimiento se basa en la idea de que toda praxis recognoscente reproduce de alguna manera el orden social dominante. El presente trabajo se propone entonces, cuestionar esta aseveración del autor advirtiendo que un análisis sobre el modo en que el poder actúa en las prácticas cotidianas de reconocimiento no necesariamente conlleva una renuncia de la función crítica del concepto para la teoría social. Más bien, como sugiere la noción butleriana (y foucaultiana) de crítica, sólo enmarcando al reconocimiento en el horizonte normativo que lo delimita puede convertirse en la base de la indagación social.
In silico Methoden spielen in der Wirkstoffentwicklung eine immer größere Bedeutung. Sie können eine Größe Hilfe in der Analyse des Targets oder beim Screening von neuen Liganden sein. Ihre Stärken liegen vorallem in der Zeit- und Kostenreduzierung während einer
Wirkstoffentwicklung.
Ziel der Arbeit war die Entwicklung neuer COX-2 Liganden mit Hilfe von in silico Methoden. Weil von der mCOX-2 keine Kristallstruktur in der PDB publiziert war, begann die Arbeit mit der Modellierung der mCOX-2. Dafür wurde aus der Sequenz der hCOX-2 aus UniProt mit der ID P35354 mit Hilfe der Kristallstruktur 3LN1 ein Homologie Modell entwickelt und im Anschluss über eine Validierungsmethode, den Ramachandran Plot, analysiert. Der Ramachandran Plot zeigte, dass 93.7% der Aminosäuren in favorisierten Regionen, 6.1% in
erlaubten Regionen, 0.2% in geduldeten Regionen und 0.0% in unerlaubten Regionen lagen. Mit diesem Modell wurde eine MD-Simulation durchgeführt, um die Energie des Modells zu
minimieren.
Die neuen Verbindungen wurden über drei verschiedene Ansätze designt. Im ersten Ansatz wurde die Software DOGS verwendet. Dabei handelt es sich um ein de novo Design Programm, welches nicht nur neue Verbindungen entwickelt, sondern auch deren Syntheseweg vorschlägt. Die vorgeschlagenen Verbindungen wurden über eine Docking-Studie analysiert, wobei die Verbindungen aus Abbildung 15 identifiziert werden konnten. Verbindung 22 wurde ohne weitere Variationen synthetisiert. Die Verbindungen 71 und 86 wurden aus der modellierten Verbindung 87, welche von DOGS vorgeschlagen wurde, weiterentwickelt. Dabei wurde Verbindung 71 als ein Fragment von Verbindung 85 entwickelt. Verbindung 86 wurde direkt aus Verbindung 87 entwickelt, wobei einige Variationen durchgenommen wurde. Hierbei sollte vorallem die Form von Verbindung 87 beibehalten werden.
Literatur verwendet, um ausgehend von den Verbindungen APHS und ASS neue COX-2 Inhibitoren zu entwickeln (siehe Abb. 16). Dabei wurden mehrere Verbindungen designt, wovon Verbindung 3 als ein leichter Inhibitor identifiziert werden konnte. 3 enthält keine für COX-Inhibitoren typische polare Gruppe, besitzt dafür aber eine Acetylgruppe, die gemäß in silico Untersuchungen in der Lage sein könnte, Ser530 in COX-2 zu acetylieren.
In der letzten Studie wurden mit Hilfe eines Fragment-basierten Designs neue Verbindungen entwickelt, wobei das Benzensulfonamid von Celecoxib aus der Kristallstruktur 1PTH extrahiert und mit kleinen Fragmente verknüpft wurde, welche zuvor über eine Docking-Studie analysiert wurden. Hieraus entwickelte sich Verbindung 35, die in einer kleinen SAR-Studie zu 70 optimiert werden konnte. Dabei konnte das Sulfonamid, welches typisch für Coxibe ist, gegen eine Carbonsäure ausgetauscht werden (69). Erst durch eine Vergrößerung der Verbindung um einen Benzyl-Rest am sekundären Amin von 69 führte zur potenten Verbindung 70.
Zusammenfassend konnten in dieser Arbeit fünf neue COX-2 Inhibitoren als Leitstrukturen entwickelt werden. Dabei kamen fortschrittliche in silico Methoden wie die De-Novo Design Software DOGS aber auch rationale Designmethoden zum Einsatz. Beide Methoden boten Vor- und Nachteile und haben jeweils zu guten Ergebnissen geführt. Bei der Entwicklung der vielversprechendsten Leitstrukturen 70 und 71 wurden die Vorteile beider Ansätze kombiniert.
In a political and economic perspective, the recent ECJ judgment on the OMT program is not more than a footnote, a short sideshow in a seemingly never-ending sequel of another dimension. Legally, however, I find the case quite remarkable. Unlike its Advocate General, the ECJ did not yield to the temptation to respond in kind to the FCC’s provocations. In particular, it avoids the issue of domestic vs. European constitutional identity that juxtaposed the FCC and the Advocate General. Instead, the ECJ has shown political responsibility and legal foresight in framing what could become a masterpiece of truly cooperative, other-regarding constitutional pluralism.
Bundesfinanzminister Wolfgang Schäubles Behauptung, Griechenland könne wegen Art. 125 AEUV nur außerhalb der Eurozone seine Schulden gegenüber anderen Euro-Staaten und EFSF bzw. ESM restrukturieren, beruht auf einem Denkfehler, wenn nicht gar auf einem Taschenspielertrick. Die Pringle-Rechtsprechung des EuGH zeigt: Das Europarecht schaufelt sich nicht sein eigenes Grab. Man muss es nicht erst umgehen, um die Ziele der Union wahrhaft zu verwirklichen.
Proteins of the secretin family form large macromolecular complexes, which assemble in the outer membrane of Gram-negative bacteria. Secretins are major components of type II and III secretion systems and are linked to extrusion of type IV pili (T4P) and to DNA uptake. By electron cryo-tomography of whole Thermus thermophilus cells, we determined the in situ structure of a T4P molecular machine in the open and the closed state. Comparison reveals a major conformational change whereby the N-terminal domains of the central secretin PilQ shift by ∼30 Å, and two periplasmic gates open to make way for pilus extrusion. Furthermore, we determine the structure of the assembled pilus.
The α-defensins, human neutrophil peptides (HNPs) are the predominant antimicrobial peptides of neutrophil granules. They are synthesized in promyelocytes and myelocytes as proHNPs, but only processed in promyelocytes and stored as mature HNPs in azurophil granules. Despite decades of search, the mechanisms underlying the posttranslational processing of neutrophil defensins remain unidentified. Thus, neither the enzyme that processes proHNPs nor the localization of processing has been identified. It has been hypothesized that proHNPs are processed by the serine proteases highly expressed in promyelocytes: Neutrophil elastase (NE), cathepsin G (CG), and proteinase 3 (PR3), all of which are able to process recombinant proHNP into HNP in vitro. We investigated whether serine proteases are in fact responsible for processing of proHNP in human bone marrow cells and in human and murine myeloid cell lines. Subcellular fractionation of the human promyelocytic cell line PLB-985 demonstrated proHNP processing to commence in fractions containing endoplasmic reticulum. Processing of 35S-proHNP was insensitive to serine protease inhibitors. Simultaneous knockdown of NE, CG, and PR3 did not decrease proHNP processing in primary human bone marrow cells. Furthermore, introduction of NE, CG, and PR3 into murine promyelocytic cells did not enhance the proHNP processing capability. Finally, two patients suffering from Papillon–Lefèvre syndrome, who lack active neutrophil serine proteases, demonstrated normal levels of fully processed HNP in peripheral neutrophils. Contradicting earlier assumptions, our study found serine proteases dispensable for processing of proHNPs in vivo. This calls for study of other protease classes in the search for the proHNP processing protease(s).
Hintergrund: Die Interaktion zwischen β-HCG und TSH in der Schwangerschaft stellt ein differentialdiagnostisches Problem dar, weil die Wertung von supprimierten TSH-Spiegeln erschwert wird. Dies kann im schlimmsten Fall zu Fehlinterpretationen führen. Ziel der vorliegenden Arbeit war, diese Interaktion an einem großen Kollektiv in einen zeitlichen Kontext mit dem Verlauf der Schwangerschaft zu bringen, da der Zeitpunkt des Screenings entscheidenden Einfluss auf die Höhe des TSH-Spiegels hat. Zusätzlich wurden anhand der vorliegenden Daten Referenzbereiche für Schwangere berechnet und der Einfluss einer Jodmedikation untersucht.
Patienten und Methoden: Aus einem unselektionierten Pool von Patientinnen eines nuklearmedizinischen Praxisverbundes wurden die Daten von 1283 schilddrüsengesunden Schwangeren zwischen 16 und 48 Jahren ausgewertet. Neben der TSH-Bestimmung lag ein Schwerpunkt auf dem zeitlichen Verlauf, so dass die Schwangeren in Untergruppen von je 2 Wochen analysiert wurden. Untersucht wurde der Einfluss der Jodmedikation auf die TSH-Werte. Am Ende erfolgte mit Hilfe der logarithmischen Transformation unter Verwendung der 2-Sigma-Grenzen die Bestimmung neuer TSH-Referenzbereiche für Schwangere.
Ergebnisse: Es zeigt sich zu Beginn der Schwangerschaft ein Anstieg der mittleren TSH-Werte von 1,22 mU/l in der 2. SSW bis auf 1,7 mU/l um die 7. SSW mit einem konsekutiven Abfall der TSH-Werte bis auf 0,9 mU/l bis zur 16. SSW (entsprechend 52,9%). Der größte Abfall findet sich in der 12. bis 14. SSW, also zum Zeitpunkt des ersten Screenings. Die Jodmedikation hat keinen maßgeblichen Einfluss auf den TSH-Wert. Die Berechnung schwangerschaftskorrigierter Referenzbereiche zeigt im ersten Drittel TSH-Werte von 0,08 – 3,67 mU/l, im 2. Drittel 0,04 – 2,88 mU/l und im 3. Trimenon 0,17 – 3,19 mU/l.
Schlussfolgerungen: Die Arbeit zeigt, dass die niedrigsten TSH-Werte zum Zeitpunkt des ersten Screenings zu erwarten sind und deswegen möglicherweise zu Fehlentscheidungen führen können. Ein relevanter Zusammenhang der Jodmedikation mit dem TSH-Wert lässt sich nicht nachweisen. Neue Referenzbereiche für Schwangere könnten hilfreich sein, dieses diagnostische Dilemma zu vermeiden.
Background: Clinical manifestations and outcomes of atherosclerotic disease differ between ethnic groups. In addition, the prevalence of risk factors is substantially different. Primary prevention programs are based on data derived from almost exclusively White people. We investigated how race/ethnic differences modify the associations of established risk factors with atherosclerosis and cardiovascular events.
Methods: We used data from an ongoing individual participant meta-analysis involving 17 population-based cohorts worldwide. We selected 60,211 participants without cardiovascular disease at baseline with available data on ethnicity (White, Black, Asian or Hispanic). We generated a multivariable linear regression model containing risk factors and ethnicity predicting mean common carotid intima-media thickness (CIMT) and a multivariable Cox regression model predicting myocardial infarction or stroke. For each risk factor we assessed how the association with the preclinical and clinical measures of cardiovascular atherosclerotic disease was affected by ethnicity.
Results: Ethnicity appeared to significantly modify the associations between risk factors and CIMT and cardiovascular events. The association between age and CIMT was weaker in Blacks and Hispanics. Systolic blood pressure associated more strongly with CIMT in Asians. HDL cholesterol and smoking associated less with CIMT in Blacks. Furthermore, the association of age and total cholesterol levels with the occurrence of cardiovascular events differed between Blacks and Whites.
Conclusion: The magnitude of associations between risk factors and the presence of atherosclerotic disease differs between race/ethnic groups. These subtle, yet significant differences provide insight in the etiology of cardiovascular disease among race/ethnic groups. These insights aid the race/ethnic-specific implementation of primary prevention.

Right up until its collapse, the Russian Empire was decidedly less urbanized and less advanced in agriculture in comparison not only with the West but also with East-Central Europe. ...
Background: The complex cellular networks within tumors, the cytokine milieu, and tumor immune escape mechanisms affecting infiltration and anti-tumor activity of immune cells are of great interest to understand tumor formation and to decipher novel access points for cancer therapy. However, cellular in vitro assays, which rely on monolayer cultures of mammalian cell lines, neglect the three-dimensional architecture of a tumor, thus limiting their validity for the in vivo situation.
Methods: Three-dimensional in vivo-like tumor spheroid were established from human cervical carcinoma cell lines as proof of concept to investigate infiltration and cytotoxicity of NK cells in a 96-well plate format, which is applicable for high-throughput screening. Tumor spheroids were monitored for NK cell infiltration and cytotoxicity by flow cytometry. Infiltrated NK cells, could be recovered by magnetic cell separation.
Results: The tumor spheroids were stable over several days with minor alterations in phenotypic appearance. The tumor spheroids expressed high levels of cellular ligands for the natural killer (NK) group 2D receptor (NKG2D), mediating spheroid destruction by primary human NK cells. Interestingly, destruction of a three-dimensional tumor spheroid took much longer when compared to the parental monolayer cultures. Moreover, destruction of tumor spheroids was accompanied by infiltration of a fraction of NK cells, which could be recovered at high purity.
Conclusion: Tumor spheroids represent a versatile in vivo-like model system to study cytotoxicity and infiltration of immune cells in high-throughput screening. This system might proof useful for the investigation of the modulatory potential of soluble factors and cells of the tumor microenvironment on immune cell activity as well as profiling of patient-/donor-derived immune cells to personalize cellular immunotherapy.
After reviewing the description of an unstable state in the framework of nonrelativistic Quantum Mechanics (QM) and relativistic Quantum Field Theory (QFT), we consider the effect of pulsed, ideal measurements repeated at equal time intervals on the lifetime of an unstable system. In particular, we investigate the case in which the ‘bare’ survival probability is an exact exponential (a very good approximation in both QM and QFT), but the measurement apparatus can detect the decay products only in a certain energy range. We show that the Quantum Zeno Effect can occur in this framework as well.
The Compressed Baryonic Matter (CBM) experiment will explore the phase diagram of strongly interacting matter in the region of high net baryonic densities. The matter at these extreme conditions will be produced and studied in heavy-ion collisions with a fixed target set-up.
The present work is dedicated to the main component of the CBM experiment - the Silicon Tracking System (STS). The STS comprises of 8 tracking stations with 1292 double sided silicon microstrip sensors. The STS has to enable the reconstruction of up to 1000 charged particle tracks per nucleus-nucleus interaction at the rate of up to 10 MHz, provide a momentum resolution of Δp/p =1%, and withstand the radiation load of up to 1 x 1014 neq/cm2 (neq — radiation dose of 1 MeV neutron equivalent). Self-triggering read-out electronics will be located on the periphery of the detecting planes, and connected to the sensors with low mass micro-cables.
During the R&D phase, as well as in the pre-series and series production phase, the characterization of the sensors, of the front-end electronics and of the complete detector modules has to be performed. It is evident that characterization of more than 1000 silicon microstrip sensors and later of complete detector modules is very time-consuming, and may even damage the objects if not performed carefully. One of the goals of this work was to develop a systematic procedure for the quality assurance for the double-sided silicon microstrip sensors. This includes static optical inspection and visual tests, passive electrical test (such as leakage current, bulk capacitance, inter-strip capacitance & resistances, bias resistance and coupling capacitance), radiation hardness and long-term stability. A strategy for the quality assurance of these sensors is presented, defining the various tests to be performed and the documentation of the results. The techniques and quality assurance criteria will be applied for the pre-series and series production.
With decreasing feature size and increase in functionality and structures, the classical mechanical probe approach for internal fault detection and functional testing faces increasing challenges. In the field of silicon based chips and sensors there is rarely any analysis on the topic of non-invasive or contact-less probing and characterization, despite the fact that the contact-less probing is becoming more and more important as the fabrication technologies become smaller and more susceptible to the parasitic impact of mechanical probes. The silicon micro-strip double sided sensors used in STS have a complex structure, such as 1024 metal electrodes, 2048 bias resistors, 2048 DC pads and 4098 AC pads for probing, several guard rings, and even more in the 6.2 cm x 6.2 cm prototype sensor. Photo-intrusive technique is the best solution for the characterization and investigation of crucial parameters related to the detector operation and its functionality. A photo-intrusive probing is a method in which a non-invasive pulsed laser of a desired wavelength is used to inject the photon into the bulk and resulting in electron-hole pairs (e-h). In a completely depleted silicon sensor the charge injected (or generated) by the pulsed laser beam could be detected as current and shall be used for characterization.
A non-invasive contact-less Laser Test System (LTS) was developed based on a pulsed laser to investigate properties of the silicon sensors. The set-up is able to inject charge locally and scan sensors(or detector modules) with a pulsed infra-red laser driven by a step motor. The LTS is designed to measure sensor response in an automatized procedure at several thousand positions across the sensor with focused infra-
red laser light (spot size = 12 μm , wavelength = 1060 nm). The duration (10 ns) and power (5 mW) of the laser pulses are selected such that the absorption of the laser light in the 300 μm thick silicon sensors produces a number of about 24000 electrons, which is similar to the charge created by the minimum ionizing particles (MIP) in these sensors. The set-up was used to developed characterization procedures to determine the charge sharing between strips, and to measure a qualitative uniformity of the sensor response over the whole active area. The prototype sensors which are tested with the set-up are small prototype sensors (256 strips, pitch = 50 μm on each side) and full-size detector modules (1024 strips/side and pitch = 58 μm). They are read-out using a self-triggering prototype read-out electronic ASIC called n-XYTER. Laser scans for amplitude response, charge sharing in the inter-strip region, and spot-size determination technique are reported. For the verification of the some design parameters, unique methods of determining coupling capacitance, and inter-strip capacitance have been developed. The modules were also tested with proton beams, and the charge sharing in the inter-strip region has been compared to the laser test results.

Diatoms originated from a secondary endosymbiosis that took place between a putative photosynthetic red algal ancestor and a heterotrophic eukaryote. Secondary endosymbiosis resulted in the formation of chloroplasts with four membranes. Centric diatoms (e.g. Thalassiosira pseudonana or Cyclotella meneghiniana) usually possess many small chloroplasts, while pennates (e.g. Phaeodactylum tricornutum) have several larger ones, or even only one which can occupy half of the cell volume...
Is postoperative imaging mandatory after meningioma removal? : results of a prospective study

Background: Routine postoperative imaging (PI) following surgery for intracranial meningiomas is common practice in most neurosurgical departments. The purpose of this study was to determine the role of routine PI and its impact on clinical decision making after resection of meningioma.
Methods: Patient and tumor characteristics, details of radiographic scans, symptoms and alteration of treatment courses were prospectively collected for patients undergoing removal of a supratentorial meningioma of the convexity, falx, tentorium, or lateral sphenoid wing at the authors’ institution between January 1st, 2010 and March 31st, 2012. Patients with infratentorial manifestations or meningiomas of the skull base known to be surgically difficult (e.g. olfactory groove, petroclival, medial sphenoid wing) were not included. Maximum tumor diameter was divided into groups of < 3cm (small), 3 to 6 cm (medium), and > 6 cm (large).
Results: 206 patients with meningiomas were operated between January 2010 and March 2012. Of these, 113 patients met the inclusion criteria and were analyzed in this study. 83 patients (73.5%) did not present new neurological deficits, whereas 30 patients (26.5%) became clinically symptomatic. Symptomatic patients had a change in treatment after PI in 21 cases (70%), while PI was without consequence in 9 patients (30%). PI did not result in a change of treatment in all asymptomatic patients (p<0.001) irrespective of tumor size (p<0.001) or localization (p<0.001).
Conclusions: PI is mandatory for clinically symptomatic patients but it is safe to waive it in clinically asymptomatic patients, even if the meningioma was large in size.

Dies ist der sechste Artikel unseres Blogfokus zu Flucht und Migration. In der Flüchtlingspolitik fällt häufig das Schlagwort „Sicherheit“. Dabei geht es meist um die (vermeintliche) Bedrohung der Aufnahmestaaten, seltener um die Sicherheit von Flüchtlingen. Um letztere angemessen zu adressieren, führt eine menschenrechtliche Perspektive weiter. Sie nimmt sowohl einige zentrale Fluchtursachen als auch Bedrohungen für Schutzsuchende während der Flucht in den Blick. Schließlich lassen sich so auch Gefahren ansprechen, die durch Verstöße gegen Flüchtlingsrechte in den Aufnahmestaaten drohen, etwa in Europa.
Although there is an increasing need for data in ecological studies, many datasets are still lost or not sufficiently visible due to a lack of appropriate data archives. With the West African Data and Metadata Repository, we present a secure long-term archive for a data-poor region allowing detailed documentation by metadata following the EML standard and giving data holders the opportunity to define levels of data access and conditions of use. This article gives an overview of structure, functions and content. The repository is online at the URL http://westafricandata.senckenberg.de.
In this paper we assemble an annual data set on broad and narrow money, prices, real economic activity and interest rates in Ireland from a variety of sources for the period 1933-2012. We discuss in detail how the data set is constructed and what assumptions we have made to do so. Furthermore, we estimate a simple SVAR model to provide some empirical evidence on the behaviour of these time series. Money supply shocks appear to be the most important drivers of both money and prices. Interest rate shocks, which capture monetary policy, play an important role driving output and, of course, interest rates. The GDP shocks, which raise prices, seem of less importance.
Since the 1970s, the overarching view in the literature has been that a Phillips curve relationship did not exist in Ireland prior to the 1979 exchange rate break with Sterling. It was argued that, as a small open economy, prices were determined externally. To test this relationship, we study the determination of inflation between 1926 and 2012, a longer sample period than any previously used. We find that the difference between unemployment and the NAIRU is a significant determinant of inflation both in the full sample and in the subsamples spanning the periods before and after the Sterling parity link.
Expectations of Sterling returning to Gold have been disregarded in empirical work on the US dollar – Sterling exchange rate in the early 1920s. We incorporate such considerations in a PPP model of the exchange rate, letting the probability of a return to gold follow a logistic function. We draw several conclusions: (i) the PPP model works well from spring 1919 to spring 1925; (ii) wholesale prices outperform consumer prices; (iii) allowing for a return to gold leads to a higher speed of adjustment of the exchange rate to PPP; (iv) interest rate differentials and the relative monetary base are crucial determinants of the expected return to gold; (v) the probability of a return to Gold peaked at about 72% in late 1924 and but fell to about 60% in early 1925; and (vi) our preferred model does not support the Keynes’ view that Sterling was overvalued after the return to gold.
Background: Inflammation, angiogenesis and oxidative stress have been implicated in the pathogenesis of various vascular diseases. Recent evidence suggests that dimethylfumarate (DMF), an antiposriatic and anti-multiple sclerosis agent, possesses anti-inflammatory, anti-oxidative and anti-angiogenic properties. Here, we analyze the influence of DMF on TNF-α-induced expression of the important pro-inflammatory and pro-atherogenic chemokine MCP-1 and investigate the underlying mechanisms of this expression.
Findings: We analyzed constitutive and TNF-α-induced expression of MCP-1 in human umbilical vascular endothelial cells (HUVEC) +/− DMF treatment via enzyme-linkes immunosorbent assay (ELISA). DMF significantly inhibited the protein expression levels in a time- and concentration-dependent manner. Furthermore, MCP-1 mRNA expression was also reduced in response to DMF, as demonstrated by RT-PCR. Thus, the regulation occurs at the transcriptional level. Interestingly, DMF prolonged the TNF-α-induced p38 and JNK phosphorylation in HUVEC, as demonstrated by Western blot analysis; however, the p38 and JNK inhibitor SB203580 did not affect the DMF-conveyed suppression of TNF-α-induced MCP-1 expression. DMF suppressed the TNF-α-induced nuclear translocation and phosphorylation (Serine 536) of p65 in these cells. These results were additionally approved by p65 luciferase promoter assays. Furthermore, we found that DMF slightly inhibited the early degradation of IκBα. In addition, we verified our results using other important inflammatory cytokines such as CCL-5, PDGF-BB, GM-CSF and IL-6.
Conclusion: DMF suppresses various TNF-α-induced pro-inflammatory and pro-atherogenic cytokines/chemokines in human endothelial cells. This action is regulated by reduced p65 activity and nuclear translocation, which can be explained in part by the reduced early degradation of IκBα and more important the reduced phosphorylation of p65 at Serine 536. These effects were independent of the p38, PI3K and p42/44 signaling pathways. As a result, DMF might be suitable for treating patients with vascular diseases.
Demographic change belongs to the mega-trends of the 20th and the 21st century. The ongoing aging process in major industrialized countries gives rise to the relative scarcity of raw labor and the relative abundance of physical capital. Standard macroeconomic models suggest that this depresses asset returns and increases wages which, in turn, provides incentives for more human capital accumulation. This thesis quantifies the macroeconomic effects of demographic change and reveals the importance of human capital adjustments for price and welfare effects within and across generations. Chapter 1 investigates the distributions of income, skills, and welfare in the German economy along the inter- and the intra-generational dimension. It shows that demographic change leads to a more capital- and skill-intensive economy and that high-school households loose compared to college households in terms of welfare. Chapter 2 disentangles the effect of demographic change on returns to risk-free and risky assets in the U.S. and measures the net effect on the equity premium. It shows that both returns decline while the equity premium increases slightly. Endogenous human capital adjustments are crucial for relatively small effects. Chapter 3 develops a method for computing transitional dynamics in heterogeneous agent models with aggregate risk if these transitions are induced by exogenous deterministic dynamics such as demographic change. The application of the method to a simple illustrative example shows a large reduction in total computing time while approximation errors are small.
Ubiquitin and the ubiquitin-like protein ATG8 are covalently attached to their respective targets via a coordinated cascade involving E1 activating, E2 conjugating and E3 ligating enzymes. Whereas ubiquitin is conferred to proteins as mono- and/or polymer(s) to alter their stability, localization and/or activity, the ubiquitin-like modifier (UBL) ATG8 is conjugated to the phospholipid phosphatidylethanolamine (PE). The best understood function of ATG8 is during autophagy where ATG8-PE conjugates are incorporated into both layers of incipient autophagosomes and serve as multipurpose docking sites for autophagosomal cargo receptors as well as regulatory factors (termed adaptors) that drive formation and maturation of autophagosomes. Mammalian cells harbor six ATG8 family members that can be subclassified into the LC3- and GABARAP-family and that can all be lipidated. However, it is currently unclear to what extent these proteins are functionally redundant or fulfil unique roles.
Cullin-RING ligase complexes (CRLs) are modular E3 ubiquitin ligases that comprise a RING-finger protein that associates with the ubiquitin-charged E2 enzyme, a substrate recruiting module as well as a cullin scaffold as a linker between RING protein and substrate adaptor. Whereas SCF (SKP1-CUL1-F-box protein) complexes, the most studied CRLs, harbor cullin-1 (CUL1) as scaffold and F-box proteins as substrate binding modules, CUL3-containing CRL complexes employ cullin-3 (CUL3), RING-box protein 1 (RBX1) and BTB proteins as substrate adaptors. Here, the BTB domain serves as binding interface for CUL3 and is usually complemented by an additional protein-protein interaction domain such as MATH or Kelch that mediates binding to the substrate of the E3 ligase complex.
Besides ubiquitylation, guanine nucleotide binding is another common way to regulate protein activity and signaling in cells. Here, small Rho GTPases cycle between active and inactive states by binding of the guanine nucleotides GTP or GDP with the help of regulatory proteins. Whereas GTPase-activating proteins (GAP) render RAC1 inactive by facilitating GTP hydrolysis, guanine exchange factors (GEF) such as T-lymphoma invasion and metastasis-inducing protein 1 (TIAM1) activate RAC1 by stimulating the exchange of GDP to GTP. Local control of RAC1 activity is essential to allow a specific cellular response to stimuli such as growth factors or migratory impulses.
This study reports an unexpected link between the GABARAP subfamily of mammalian ATG8 proteins, the ubiquitin proteasome system and RAC1 through the ubiquitylation of the RAC1 GEF TIAM1. The Kelch repeat and BTB domain-containing proteins 6 (KBTBD6) and 7 (KBTBD7) were established as heterodimeric substrate adaptors for CUL3. Interestingly, a thorough proteomic analysis revealed a number of putative substrates but, out of 11 substrate candidates tested, only the RAC1 GEF TIAM1 appeared to be influenced by depletion of CUL3KBTBD6/KBTBD7. Binding studies showed that KBTBD7 binds TIAM1 via the Kelch repeats and that this binding was markedly enhanced when CUL3 activation was abolished upon treatment with the neddylation inhibitor MLN4924. Also, total TIAM1 abundance was increased upon CUL3KBTBD6/KBTBD7 depletion and accumulation of TIAM1 upon proteasome inhibition suggested that TIAM1 is degraded via the proteasome. In vivo ubiquitylation assays and denaturing immunoprecipitations as well as mass spectrometrical analysis confirmed that CUL3KBTBD6/KBTBD7 ubiquitylates TIAM1 at two distinct lysines (K1404 and K1420) close to its C-terminus.
Previously, KBTBD6 and KBTBD7 were found as interactors of several members of the human ATG8 family of proteins in a proteomic study analyzing the human autophagy network. This association was confirmed in the present work. Furthermore, peptide array technology and mutational analysis revealed that KBTBD6 and KBTBD7 employ a classical ATG8-family interacting motif (AIM; also referred to as LC3-interacting region or LIR) as binding interface. The AIMs of KBTBD6 (W-V-R-V) and KBTBD7 (W-V-Q-V) fulfil the consensus AIM sequence motif (F/W/Y1-X2-X3-I/L/V4) and are preceded by several acidic residues and serines. A series of structural and cell biological experiments revealed a binding preference for the GABARAP subfamily of human ATG8 proteins and most importantly, a requirement of the GABARAP-KBTBD6 and -KBTBD7 interaction for TIAM1 ubiquitylation. The finding that TIAM1 binding to KBTBD6 and KBTBD7 AIM mutants was diminished raised the possibility that GABARAP binding mediates the recruitment of CUL3KBTBD6/KBTBD7 to membranes where TIAM1 is localized. Interestingly, colocalization of KBTBD6, GABARAPL1 and TIAM1 in punctuate structures could be observed. Since only a very small fraction of GABARAPL1 colocalized with LC3B, and colocalization between KBTBD6 and LC3B was not observed, these vesicular structures are most likely distinct from autophagosomes. Furthermore, TIAM1 ubiquitylation was reduced when GABARAP, but not LC3B, was depleted or when lipidation of GABARAP was prevented.
Stabilization of TIAM1 upon KBTBD6 and/or KBTBD7 depletion led to elevated TIAM1-dependent RAC1 activity, altered actin morphology with increased cortical actin and loss of vinculin foci. Re-introduction of wild-type KBTBD6 or KBTBD7 but not AIM mutants reverted all these phenotypes. Moreover, depletion of KBTBD6 or KBTBD7 in human breast cancer cells massively increased their invasiveness, whereas TIAM1 knockdown had the opposite outcome. All physiological effects of KBTBD6 and KBTBD7 depletion were inhibited by additional depletion of TIAM1 or RAC1 confirming that the phenotypes observed are indeed mediated by the CUL3KBTBD6/KBTBD7-TIAM1-RAC1 signaling pathway. Intriguingly, KBTBD6 and KBTBD7 were not subject to autophagosomal degradation, thereby establishing a new function for GABARAP proteins beyond autophagosomal degradation in providing a signaling platform for recruitment of the E3 ligase CUL3KBTBD6/KBTBD7 in close proximity to its substrate TIAM1, enabling localized ubiquitylation.
Local restricted control of RAC1 activity by ubiquitylation has been described for TIAM1-RAC1 signaling previously. Examples are HECT, UBA and WWE domain-containing protein 1 (HUWE1)-mediated TIAM1 ubiquitylation that occurs predominantly at cell-cell-junctions in response to hepatocyte growth factor stimulation in MDCKII cells or inhibition of RAC1 activity by the RAC1 GAP protein BCR (breakpoint cluster region) at the leading edge of astrocytes through binding to the TIAM1-Par (polarity) complex. SCFBTRC mediates ubiquitylation of TIAM1 in response to mitogens or DNA damage, though it has not been explored whether this regulation is spatially restricted. Thus, this study adds a novel layer of complexity to the spatial regulation of RAC1 signaling by implicating membrane-bound human ATG8 proteins in this process.
Also, this study is the first report specifically implicating the GABARAP proteins in cellular signaling events. It will be interesting to explore whether the concept of localized signaling mediated by GABARAPs applies to other substrates of CUL3KBTBD6/KBTBD7 and membranerelated signaling processes in which GABARAP proteins are involved. Controlling RAC1 activity at GABARAP-decorated membranes might also be important for trafficking events or autophagy since it was described that RAC1 has an inhibitory function on autophagy. Therefore, spatial restricted ubiquitylation of TIAM1 resulting in specific deactivation of RAC1 could promote the autophagic process when locally needed. Although the catalytic mTOR inhibitor Torin1 and the lysosomal H+ ATPase inhibitor BafilomycinA1 promoted TIAM1 ubiquitylation by increasing the pool of membrane-conjugated GABARAP, but other signals that stimulate GABARAP-KBTBD6/KBTBD7 association and subsequent TIAM1 ubiquitylation are to be identified. Besides, determining the KBTBD6/KBTBD7 binding site in TIAM1 or uncovering a deubiquitylating enzyme (DUB) that locally counteracts the ubiquitylation of TIAM1 will enable a better comprehension of the complete localized signaling cascade.

In a case study of a Greek government bond for the period 2009 to 2011 when Greece’s credit rating declined sharply, this study highlights the discretion that preparers have when estimating impairments. IFRS 9 relies more on management expectations and will lead to earlier impairments. However, these appear still delayed and low if compared to the fair value losses.
Attractive growth cone turning requires Igf2bp1-dependent local translation of β-actin mRNA in response to external cues in vitro. While in vivo studies have shown that Igf2bp1 is required for cell migration and axon terminal branching, a requirement for Igf2bp1 function during axon outgrowth has not been demonstrated. Using a timelapse assay in the zebrafish retinotectal system, we demonstrate that the β-actin 3’UTR is sufficient to target local translation of the photoconvertible fluorescent protein Kaede in growth cones of pathfinding retinal ganglion cells (RGCs) in vivo. Igf2bp1 knockdown reduced RGC axonal outgrowth and tectal coverage and retinal cell survival. RGC-specific expression of a phosphomimetic Igf2bp1 reduced the density of axonal projections in the optic tract while sparing RGCs, demonstrating for the first time that Igf2bp1 is required during axon outgrowth in vivo. Therefore, regulation of local translation mediated by Igf2bp proteins may be required at all stages of axon development.
Dilatometric studies on single crystalline barlowite – a structurally perfect spin-1/2 Kagome system

We present results of high-resolution thermal expansion measurements on single crystalline barlowite – a structurally perfect spin-1/2 kagome system. The data reveal strongly pronounced and anisotropic second-order phase transition anomalies at the Néel transition at TN = 16K. From these data, together with literature results on the specific heat, the uniaxial-pressure dependences of TN are derived. We find a rather large positive pressure coefficient for uniaxial pressure along the hexagonal c axis of ∂TN/∂pc = (2.3 ± 0.2) K/GPa and smaller negative in-plane pressure coefficient of ∂TN/∂pin-plane = -(0.6 ± 0.03) K/GPa. These effects result in a small positive pressure coefficient under hydrostatic-pressure conditions of ∂TN/∂phydr = (1.1 ± 0.2) K/GPa. Bond-lengths considerations indicate that inter-layer Cu-O bonds, being larger than those typically found in stable Cu-O complexes, are responsible for this behavior.

El artículo presenta cómo el filósofo alemán Theodor W. Adorno concibe el estado social de la música de la primera mitad del siglo XX. Para ello recurro a
una serie de conceptos utilizados por el autor y cuyas raíces están en el pensamiento filosófico de la modernidad. Con gran influencia de Walter Benjamin, cada concepto en Adorno es una constelación, es decir, una idea que debe ser desarrollada y que al relacionarse con otros conceptos se llega a una interpretación más abarcadora del problema a considerarse. Así pues, divido el escrito en dos partes: uno dedicado al análisis de la ideología y el otro al de reificación. Cada capítulo así mismo está dividido en distintos apartados, con el ánimo de dar una visión más amplia de cómo Adorno entendió el problema de la experiencia musical para al final dar cuenta de la importancia del pensamiento estético de Adorno al compararlo con otros autores que reflexionaron sobre el mismo tema.
Cyclic GMP (cGMP) signalling regulates multiple biological functions through activation of protein kinase G and cyclic nucleotide-gated (CNG) channels. In sensory neurons, cGMP permits signal modulation, amplification and encoding, before depolarization. Here we implement a guanylyl cyclase rhodopsin from Blastocladiella emersonii as a new optogenetic tool (BeCyclOp), enabling rapid light-triggered cGMP increase in heterologous cells (Xenopus oocytes, HEK293T cells) and in Caenorhabditis elegans. Among five different fungal CyclOps, exhibiting unusual eight transmembrane topologies and cytosolic N-termini, BeCyclOp is the superior optogenetic tool (light/dark activity ratio: 5,000; no cAMP production; turnover (20 °C) ∼17 cGMP s−1). Via co-expressed CNG channels (OLF in oocytes, TAX-2/4 in C. elegans muscle), BeCyclOp photoactivation induces a rapid conductance increase and depolarization at very low light intensities. In O2/CO2 sensory neurons of C. elegans, BeCyclOp activation evokes behavioural responses consistent with their normal sensory function. BeCyclOp therefore enables precise and rapid optogenetic manipulation of cGMP levels in cells and animals.
The ancestors to the Australian marsupials entered Australia around 60 (54-72) million years ago from Antarctica, and radiated into the four living orders Peramelemorphia, Dasyuromorphia, Diprotodontia and Notoryctemorphia. The relationship between the four Australian marsupial orders has been a long-standing question, because different phylogenetic studies were not able to consistently reconstruct the same topology. Initial in silico analysis of the Tasmanian devil genome and experimental screening in the seven marsupial orders revealed 20 informative transposable element insertions for resolving the inter- and intraordinal relationships of Australian and South American orders. However, the retrotransposon insertions support three conflicting topologies regarding Peramelemorphia, Dasyuromorphia and Notoryctemorphia, indicating that the split between the three orders may be best understood as a network. This finding is supported by a phylogenetic re-analysis of nuclear gene sequences, using a consensus network approach that allows depicting hidden phylogenetic conflict, otherwise lost when forcing the data into a bifurcating tree. The consensus network analysis agrees with the transposable element analysis in that all possible topologies regarding Peramelemorphia, Dasyuromorphia, and Notoryctemorphia in a rooted four-taxon topology are equally well supported. In addition, retrotransposon insertion data supports the South American order Didelphimorphia being the sistergroup to all other living marsupial orders. The four Australian orders originated within three million years at the Cretaceous-Paleogene boundary. The rapid divergences left conflicting phylogenetic information in the genome possibly generated by incomplete lineage sorting or introgressive hybridisation, leaving the relationship among Australian marsupial orders unresolvable as a bifurcating process million years later.
Krieg und physische Gewalt sind seit jeher präsente Themen der Frühmittelalterforschung, trotz gewisser konjunktureller Schwankungen. Über die letzten Jahre lässt sich eine intensivere Auseinandersetzung mit diesem Bereich beobachten, wohl ein Reflex auf aktuelle Ereignisse und die von ihnen ausgelösten wissenschaftlichen Debatten in stärker gegenwartsorientierten Disziplinen. Gerade in der deutschen Mittelalterforschung wird dabei eine Hinwendung zu einer "Kulturgeschichte des Krieges" (Hans-Henning Kortüm) vollzogen, was einerseits in den Schwierigkeiten begründet liegt, auf Basis des größtenteils sehr fragmentarischen Quellenmaterials "klassische" Militärgeschichte für das Frühmittelalter zu schreiben, andererseits aber auch Entwicklungen in den Kulturwissenschaften aufnimmt und den pazifistischen Grundtenor der deutschen Nachkriegsgesellschaft spiegelt. In diesen Kontext gehört das zu besprechende Buch von Laury Sarti, das auf ihrer 2012 bei Hans-Werner Goetz in Hamburg eingereichten Dissertation beruht. ...
Viruses rely completely on the hosts' machinery for translation of viral transcripts. However, for most viruses infecting humans, codon usage preferences (CUPrefs) do not match those of the host. Human papillomaviruses (HPVs) are a showcase to tackle this paradox: they present a large genotypic diversity and a broad range of phenotypic presentations, from asymptomatic infections to productive lesions and cancer. By applying phylogenetic inference and dimensionality reduction methods, we demonstrate first that genes in HPVs are poorly adapted to the average human CUPrefs, the only exception being capsid genes in viruses causing productive lesions. Phylogenetic relationships between HPVs explained only a small proportion of CUPrefs variation. Instead, the most important explanatory factor for viral CUPrefs was infection phenotype, as orthologous genes in viruses with similar clinical presentation displayed similar CUPrefs. Moreover, viral genes with similar spatiotemporal expression patterns also showed similar CUPrefs. Our results suggest that CUPrefs in HPVs reflect either variations in the mutation bias or differential selection pressures depending on the clinical presentation and expression timing. We propose that poor viral CUPrefs may be central to a trade-off between strong viral gene expression and the potential for eliciting protective immune response.
Secondary plant metabolites reveal numerous biological activities making them attractive as resource for drug development of human diseases. As the majority of cancer drugs clinically established during the past half century is derived from nature, cancer researchers worldwide try to identify novel natural products as lead compounds for cancer therapy. Natural products are considered as promising cancer therapeutics, either as single agents or in combination protocols, to enhance the antitumor activity of additional therapeutic modalities. Most natural compounds exert pleotrophic effects and modulate various signal transduction pathways. A better understanding of the complex mechanisms of action of natural products is expected to open new perspectives in coming years for their use alone or in combination therapies in oncology. Two major strategies to identify novel drug candidates from nature are the bioactivity-guided fractionation of medicinal plant extracts to isolate cytotoxic chemicals and the identification of small molecules inhibiting specific targets in cancer cells. In the present review, we report on our own efforts to unravel the molecular modes of action of phytochemicals in cancer cells and focus on resveratrol, betulinic acid, artesunate, dicentrine and camptothecin derivatives.

Activation of Mitochondrial complex II-dependent respiration is beneficial for α-Synucleinopathies

Parkinson’s disease and dementia with Lewy bodies are major challenges in research and clinical medicine world-wide and contribute to the most common neurodegenerative disorders. Previously, specific mitochondrial polymorphisms have been found to enhance clearance of amyloid-β from the brain of APP-transgenic mice leading to beneficial clinical outcome. It has been discussed whether specific mitochondrial alterations contribute to disease progression or even prevent toxic peptide deposition, as seen in many neurodegenerative diseases. Here, we investigated α-synuclein-transgenic C57BL/6J mice with the A30P mutation, and a novel A30P C57BL/6J mouse model with three mitochondrial DNA polymorphisms in the ND3, COX3 and mtRNAArg genes, as found in the inbred NOD/LtJ mouse strain. We were able to detect that the new model has increased mitochondrial complex II-respiration which occurs in parallel to neuronal loss and improved motor performance, although it exhibits higher amounts of high molecular weight species of α-synuclein. High molecular weight aggregates of different peptides are controversially discussed in the light of neurodegeneration. A favourable hypothesis states that high molecular weight species are protective and of minor importance for the pathogenesis of neurodegenerative disorders as compared to the extreme neurotoxic monomers and oligomers. Summarising, our results point to a potentially protective and beneficial effect of specific mitochondrial polymorphisms which cause improved mitochondrial complex II-respiration in α-synucleinopathies, an effect that could be exploited further for pharmaceutical interventions.

Dank einer entsprechenden Kurzbiographie des Sophisten Philostratos (soph. 2,7) steht der Name Hermogenes von Tarsos in der Forschung zur griechischen Kulturgeschichte der Kaiserzeit zumeist als Musterbeispiel für das schlussendliche Scheitern und damit einhergehende Absinken in die Unbedeutsamkeit eines vielversprechenden Sophisten nach einer kometenhaften Karriere. Trotz dieser wechselvollen Karriere des Redekünstlers haben, wenn auch nicht seine Reden, so doch zumindest einige seiner redetheoretischen Schriften nach einer erneuten Blüte ab der byzantinischen Epoche bis einschließlich ins 16.Jh. die Zeiten überdauert. Climate change and its impacts already pose considerable challenges for societies that will further increase with global warming (IPCC, 2014a, b). Uncertainties of the climatic response to greenhouse gas emissions include the potential passing of large-scale tipping points (e.g. Lenton et al., 2008; Levermann et al., 2012; Schellnhuber, 2010) and changes in extreme meteorological events (Field et al., 2012) with complex impacts on societies (Hallegatte et al., 2013). Thus climate change mitigation is considered a necessary societal response for avoiding uncontrollable impacts (Conference of the Parties, 2010). On the other hand, large-scale climate change mitigation itself implies fundamental changes in, for example, the global energy system. The associated challenges come on top of others that derive from equally important ethical imperatives like the fulfilment of increasing food demand that may draw on the same resources. For example, ensuring food security for a growing population may require an expansion of cropland, thereby reducing natural carbon sinks or the area available for bio-energy production. So far, available studies addressing this problem have relied on individual impact models, ignoring uncertainty in crop model and biome model projections. Here, we propose a probabilistic decision framework that allows for an evaluation of agricultural management and mitigation options in a multi-impact-model setting. Based on simulations generated within the Inter-Sectoral Impact Model Intercomparison Project (ISI-MIP), we outline how cross-sectorally consistent multi-model impact simulations could be used to generate the information required for robust decision making.
Using an illustrative future land use pattern, we discuss the trade-off between potential gains in crop production and associated losses in natural carbon sinks in the new multiple crop- and biome-model setting. In addition, crop and water model simulations are combined to explore irrigation increases as one possible measure of agricultural intensification that could limit the expansion of cropland required in response to climate change and growing food demand. This example shows that current impact model uncertainties pose an important challenge to long-term mitigation planning and must not be ignored in long-term strategic decision making.
Im Rahmen der vorliegenden Arbeit wurde einerseits der Einsatz lichtaktivierbarer Oligonukleotide zur Kontrolle der Leitfähigkeit entlang von DNA untersucht sowie neue photoaktivierbare Verbindungen für die Peptidchemie und für eine neu entwickelte Variante des SELEX (Systematic Evolution of Ligands by EXponetial enrichment) Verfahrens synthetisiert.
DNA vermittelte Ladungsübertragung verläuft entlang des gestapelten π-Systems der heteroaromatischen Nukleobasen. Die Leitfähigkeit von Oligonukleotiden reagiert daher empfindlich auf Störungen in der Watson-Crick-Basenpaarung. Die in der Arbeitsgruppe Heckel etablierte Technik, Nukleobasen an für die Basenpaarung relevanten Positionen mit photolabilen Schutzgruppen zu modifizieren, sollte daher mit Systemen der Ladungsübertragung in DNA kombiniert werden. Im Verlauf dieses Projekts wurden zwei literaturbekannte Varianten, in denen Ladungstransport über einen lichtinduzierten Redoxprozess zwischen Metallkomplexen ablaufen und über eine dabei unterdrückte Fluoreszenz optisch verfolgt werden sollte, als ungeeignete Systeme identifiziert. Durch den Wechsel zu elektrodengestützter Leitfähigkeitsmessung konnte der prinzipielle Effekt von Leitfähigkeit in perfekt gepaarter DNA und deutlich reduziertem Stromfluss in Oligonukleotiden mit Fehlpaarungen gezeigt werden. Beim Einsatz photolabil geschützter Oligonukleotide konnte jedoch auch in diesem System noch nicht der gewünschte Effekt gefunden werden.
Im zweiten Projekt dieser Arbeit wurden neue photolabile Verbindungen hergestellt, die Peptide nach ihrem Einbau in das Peptidrückgrat durch Zwei-Photonen-Anregung mit IR-Licht spalten sollen. Drei entsprechende Nitrodibenzofuran-Verbindungen und ein Cumarin-Baustein konnten erfolgreich synthetisiert werden. Die neuen Moleküle zeigten im Rahmen der Peptid-Festphasensynthese Stabilitätsprobleme. Diese Schwierigkeiten konnten durch Peptid-Kopplungen in Lösung umgangen werden. Mit Hilfe eines der hergestellten Bausteine wurden zwei Tripeptide hergestellt, die jeweils mit dem Farbstoff ATTO565 markiert und hinsichtlich ihrer photochemischen Eigenschaften charakterisiert wurden. Der neue Baustein zeigte neben den Eigenschaften als photospaltbare Gruppe, dass er gleichzeitig ein Quencher für den Farbstoff ATTO565 darstellt. Nach Belichtung stieg die Fluoreszenz um den Faktor 81 an. Die Aktivierung gelang wie erwartet mit Ein- und Zwei-Photonen-Anregung. In Kollaboration mit der Arbeitsgruppe von Prof. Heilemann konnten Antiköper mit einem der Tripeptide modifiziert werden und die Kompatibilität der Verbindung mit hochaufgelöster Einzelmolekül-Fluoreszenzmikroskopie demonstriert werden.
Im letzten in dieser Arbeit thematisierten Projekt wurden neue lichtspaltbare Verbindungen für eine Variante des SELEX-Prozesses hergestellt. Diese Verbindungen erlauben die temporäre Einführung einer Indol Modifikation an Alkin-modifizierte Oligonukleotide über die sogenannte Click-Chemie. Neue chemische Modifikationen wie die hier verwendeten Indole erhöhen die chemische Vielfalt der Oligonukleotide. Eine größere Vielfalt führt zu neuen potentiellen Wechselwirkungen gegenüber Verbindungen, gegen die mit Hilfe herkömmlicher SELEX-Verfahren keine Aptamere erzeugt werden konnten. Da die chemische Modifikation über eine photolabile Gruppe an die Oligonukleotide gebunden wird, kann sie photochemisch von der DNA gespalten werden, wodurch eine Interferenz der Modifikation mit den enzymatisch katalysierten Schritten innerhalb der SELEX ausgeschlossen werden kann.
Dass das Mittelalter "dunkel", gar "finster" gewesen sei, kann als handelsüblicher Topos gelten. Der stillschweigende Verweis etwa auf Autoritäten der Geistesgeschichte wie Luther, Voltaire oder Heine erübrigt jeglichen Beleg. Doch professionelle Mediävisten wagen, ein anderes Mittelalterbild zu zeichnen und werfen gleichzeitig ein Licht darauf, wie es zu diesem falschen Verständnis kam.
Das Folgende ist eine Auseinandersetzung mit dem Zeitungsartikel von Patrick Bahners (FAZ 5. 9. 2015) und zugleich mit seiner wichtigsten Autorität, Gerd Althoff (FmSt 48, 2014, S. 261-76: "Das Amtsverständnis Gregors VII. und die neue These vom Friedenspakt in Canossa"). Zu beiden Texten sind einige Bemerkungen vonnöten. Der vorliegenden Fassung meiner Darlegung gingen Versionen voraus, die an Jürgen Petersohn u.a., sowie jeweils erweitert an Nikolas Jaspert und zuletzt an Folker Reichert geschickt wurden. Ich stelle hiermit diese erweiterte Version (entgegen meiner ursprünglichen Absicht) auf Drängen von Kollegen und Freunden ins "Netz".
The small leucine-rich proteoglycan biglycan (Bgn) is a part of the extracellular matrix providing structure and enhancing fibril stability. In its soluble form, biglycan is able to bind and signal via the innate immune receptors Toll-like receptor (TLR) 2 and 4, thereby activating MAP-kinases and the NF-κB pathway. In macrophages soluble biglycan induces the secretion of several cytokines and chemokines, including TNF-α, CCL2, CXCL5 and CXCL13. A unique feature of biglycan is its ability to stimulate the secretion of mature IL-1β. By orchestrating TLR2 and 4 with the purinergic P2X4 and P2X7 receptor signalling biglycan triggers the activation of the NLRP3/ASC inflammasome, which in turn activates caspase-1 to cleave pro-IL-1β to mature IL-1β. Furthermore, in several inflammatory diseases an upregulated biglycan expression is found. Enhanced levels of biglycan could be measured in plasma and inflamed tissue. In mouse models of sepsis, lupus nephritis and renal ischemic reperfusion injury, biglycan-deficiency improved the disease outcome. Overexpression of soluble biglycan on the other hand increased immune cell infiltration into the kidney by inducing cytokine and chemokine expression in a TLR2/4-dependent manner. These studies emphasise its importance in inflammatory processes, especially in the kidney. Furthermore, the pro-inflammatory effects on macrophages and diseases established biglycan as a danger signalling molecule, yet its role as a soluble molecule in plasma was not further investigated.
Although an increase of soluble biglycan in the circulation could be seen in several inflammatory diseases, the source is not fully unravelled. Previously it could be shown that macrophages and dendritic cells secrete soluble biglycan after stimulation with IL-6 and TGF-β1. However, since these cell are resident in organs and do not circulate in the blood stream their contribution to soluble biglycan levels in plasma is likely minor. Therefore, monocytes as precursor of both macrophages and dendritic cells were investigated as a possible source of circulating biglycan. Analysis of blood from septic patients revealed elevated soluble biglycan levels as well as an increased number of monocytes. Isolated monocytes from healthy volunteers incubated with the inflammatory cytokines IL-1β, IL-6 and TGF-β1 displayed increased biglycan mRNA expression and secretion of soluble biglycan into the supernatant, revealing monocytes as a producer of soluble biglycan in blood. Therefore this work was directed to further investigate the influence of soluble biglycan on circulating monocytes, with regard to sepsis.
Monocytes can be classified into three subtypes, while the classical monocytes express CD14 (CD14++CD16low), intermediate monocytes express both CD14 and CD16 (CD14++CD16+) and non-classical monocytes express mainly CD16 (CD14lowCD16++). The intermediate and non-classical monocytes make up about 10 % of all monocytes and are referred to as CD16-positive subtypes. The CD16-positive monocytes express higher levels of TNF-α and IL-1β upon stimulation and display different migration behaviour. In most inflammatory diseases an expansion of CD16-positive monocytes is observed, especially an increased number of intermediate monocytes frequently correlate with disease severity and mortality. Since septic patients had increased circulating biglycan levels and augmented CD16-positive monocytes, a possible correlation between these two parameters was investigated. Using FACS analysis of biglycan-stimulated monocytes from healthy donors revealed a significant shift from classical to intermediate and non-classical monocytes. This shift was mediated by increased expression of CD14 and CD16 on mRNA and protein levels upon biglycan treatment. Furthermore, biglycan induced the mRNA expression of the adhesion molecules ICAM-1, VCAM-1 and ELAM-1 in CD14-positive monocytes. Four hours after biglycan stimulation an increased ICAM-1 protein expression on the cell surface of classical and intermediate monocytes was observed. Additionally, biglycan-treated CD14-positive monocytes rolled and attached to pre-stimulated endothelial cells to a greater extent compared to untreated monocytes. This demonstrates that biglycan not only triggers the expression of CD14 and CD16 but also induces a functional shift of monocytes. ...

Menschen mit Behinderung haben dieselben Menschenrechte wie alle anderen Menschen: eigentlich eine Selbstverständlichkeit. Zur Bekräftigung und Präzisierung dieser Tatsache haben die Vereinten Nationen (UN) 2006 die Behindertenrechtskonvention verabschiedet, die von 158 Staaten unterzeichnet wurde – 2009 auch von der Bundesrepublik Deutschland. Darin werden auch spezifische, vor allem behinderte Menschen betreffende Regelungen getroffen. Artikel 24 bezieht sich auf den Bereich Bildung: Um Diskriminierung zu vermeiden und Chancengleichheit zu verwirklichen, verpflichten sich die Vertragsstaaten, ein integratives bzw. inklusives Schulsystem einzurichten. Dabei liegt der Fokus nicht an der Behinderung an sich, sondern auf den Barrieren, die die soziale Teilhabe einschränken. Ziel ist eine Gesellschaft, in der niemand integriert werden muss, weil jeder von vornherein dazugehört. Eine Utopie? Auf alle Fälle ein großes Ziel, an dem stetig und auf vielen Ebenen gearbeitet werden muss. In Deutschland wird über das Thema lebhaft diskutiert. Das deutsche Schulsystem mit seiner frühen Aufteilung der Kinder auf verschiedene Schulformen tut sich teils schwer mit der Umsetzung der Konvention. Diese verbietet Förderschulen zwar keineswegs, sie schreibt jedoch einen gleichberechtigten Zugang aller zum Bildungswesen vor. Ein Rückbau der Förderschulen würde den inklusiven Prozess beschleunigen und mehr Mittel freisetzen, argumentieren Experten. Auch die Stadt Frankfurt hat sich auf den Weg gemacht: Im Rahmen des Programms „Modellregion Inklusion“ sollen Förderschulen in Förderzentren umgebaut und Ressourcen an die Regelschulen verlagert werden. Auf Einladung des UniReports diskutieren der Frankfurter Erziehungswissenschaftler Prof. Dieter Katzenbach und Martina Franke, Leiterin der Johann-Hinrich-Wichern-Schule (Frankfurt) mit Förderschwerpunkt Lernen.
The Central Nigerian Nok Culture has been well known for its elaborate terracotta sculptures and evidence of iron metallurgy since its discovery by British archaeologist Bernard Fagg in the 1940s. With a date in the first millennium BCE, both, sculptures and ironworking, belong to the earliest of their kind in sub-Saharan Africa. After a period of destruction of Nok sites by looting, scientific research resumed in 2006, when a team of archaeologists from Goethe University in Germany started to explore different Nok Culture aspects, one of which focused on chronology. Establishing a chronology for the Nok Culture employed two approaches: a comprehensive pottery analysis based on decoration and form elements and a wealth of radiocarbon dates from a large number of excavated sites. This volume presents the radiocarbon dates and the methods, data and results of the chronological pottery analysis, conducted within the scope of a dissertation project completed in 2015. Combining the two strands of information, a chronology emerges, dividing the Nok Culture into three phases from the middle of the second millennium BCE to the last centuries BCE and defining seven pottery groups that can be arranged to some extent in a chronological order.
Bayesian Networks are computer-based environmental models that are frequently used to support decision-making under uncertainty. Under data scarce conditions, Bayesian Networks can be developed, parameterized, and run based on expert knowledge only. However, the efficiency of expert-based Bayesian Network modeling is limited by the difficulty in deriving model inputs in the time available during expert workshops. This thesis therefore aimed at developing a simple and robust method for deriving conditional probability tables from expert estimates in a time-efficient way. The design and application of this new elicitation and conversion method is demonstrated using a case study in Xinjiang, Northwest China. The key characteristics of this method are its time-efficiency and the approach to use different conversion tables based on varying levels of confidence. Although the method has its limitations, e.g. it can only be applied for variables with one conditioning variable; it provides the opportunity to support the parameterization of Bayesian Networks which would otherwise remain half-finished due to time constraints. In addition, a case study in the Murray-Darling Basin, Australia, is used to compare Bayesian Network types and software to improve the presentation clarity of large Bayesian Networks. Both case studies aimed at gaining insights on how to improve the applicability of Bayesian Networks to support environmental management.
Background: Bats belong to one of the most species-rich orders within the Mammalia. They show a worldwide distribution, a high degree of ecological diversification as well as a high diversity of associated parasites and pathogens. Despite their prominent and unique role, the knowledge of their parasite-host-relationships as well as the mechanisms of co-evolutionary processes are, partly due to strict conservation regulations, scarce.
Methods: Juvenile specimens of the greater mouse-eared bat (Myotis myotis) from a roosting colony in Gladenbach (Hesse, Germany) were examined for their metazoan endo-and ectoparasite infections and pathogens. Morphometric data were recorded and the individuals were checked for Lyssavirus-specific antigen using a direct immunofluorescence test. For unambiguous species identification, the bats were analysed by cyt-b sequence comparison.
Results: Myotis myotis were parasitized by the six insect and arachnid ectoparasite species, i.e. Ixodes ricinus, Ischnopsyllus octactenus, Ichoronyssus scutatus, Steatonyssus periblepharus, Spinturnix myoti and Cimex dissimilis. Additionally, the nematode Molinostrongylus alatus and the cestode Vampirolepis balsaci were recorded. Each bat was parasitized by at least four species. The parasites showed partially extreme rates of infection, never recorded before, with more than 1,440 parasites per single host. Ichoronyssus scutatus, Steatonyssus periblepharus, Vampirolepis balsaci and Molinostrongylus alatus are recorded for the first time in Germany. A checklist for Europe is presented containing records of 98 parasite species of 14 Myotis species.
Conclusions: The Myotis myotis from Gladenbach (Hesse, Germany) were parasitized by a diverse parasite fauna with high infestation rates. We assume that in juvenile Myotis the number of parasites is generally higher than in adults due to only later acquired immune competence and behavioural adaptations. Our results revealed new insights into parasite fauna of M. myotis and European bats in general. The finding of endoparasitic cyclophyllidean cestodes that have a two-host lifecycle is, considering the stationary behaviour of the juvenile bats, rather unusual and suggests a non-predatory transmission mechanism (e.g. via autoinfection).
A new insight gained from the collated literature was that the European wide composition of the Myotis parasite fauna is dominated by a few specific taxonomic groups in Europe.
Terrestrische Säugetiere werden von unterschiedlichen Parasiten als Wirte genutzt. Dabei kann ihre Parasitenfauna je nach Art, Lebensweise, Verbreitung, Gesundheitszustand und Reproduktionsstatus des Wirts abweichen. Ein weiterer bestimmender Faktor, ist der Einfluss des Menschen in Form von Regulierungsmaßnahmen und Schaffung urbaner Lebensräume. Domestizierte Haustiere bzw. Nutztiere weisen daher in der Regel andere Parasiten auf als ihre wildlebenden Artgenossen. Gleichzeitig können sich sowohl Wildtiere als auch domestizierte Tiere und Menschen gegenseitig Parasitenarten teilen und wechselseitig aufeinander übertragen. Daraus resultierende Krankheiten werden als Zoonosen bezeichnet.
Insbesondere Fledermäuse (Unterordnung Microchiroptera) zeigen weltweit eine enorme Parasitendiversität, die noch weitgehend unerforscht ist. Ebenfalls Forschungsbedarf besteht für die Sandfloh-Gattung Tunga in Süd- und Mittelamerika in Hinblick auf ihr Wirtsspektrum, welches auch Menschen einschließt. Die Art Tunga penetrans und zahlreiche weitere Parasitenarten, parasitieren gleichzeitig auch bei Hunden. Daher stellen diese Wirte eine direkte Gesundheitsgefahr für Menschen in ihrer unmittelbaren Umgebung dar.
Die vorliegende Dissertation ist in kumulativer Form zusammengefasst und beinhaltet drei Einzelpublikationen sowie einen Reviewartikel.
Ziel war es, die Parasitendiversität von Hunden aus urbanen tropischen Gebieten und die Parasitendiversität des Großen Ameisenbären (Myrmecophaga tridactyla) mit Hilfe morphologischer und molekularbiologischer Methoden zu analysieren. Die jeweiligen Parasitenfaunen wurden in Hinblick auf die soziale bzw. solitäre Lebensweise der beiden Wirtsarten verglichen und ihr zoonotisches Potenzial bewertet.
Ein weiteres Ziel war die Zusammenfassung der Ektoparasitennachweise süd- und mittelamerikanischer Microchiroptera und für die europäischen Arten der Fledermaus-Gattung Myotis (hier Endo- und Ektoparasiten) auf Basis der verfügbaren Literatur. Des Weiteren sollten eigene Parasitennachweise aus Bolivien bzw. Deutschland erfolgen. Für die Nachweise aus Deutschland wurden M. myotis untersucht, deren Artzugehörigkeit vorher bestimmt wurde. Zusätzlich wurden diese Individuen auf humanpathogene Lyssaviren untersucht.
Die Nachweise erfolgten über molekularbiologische und morphologische Methoden.
As a surrogate of live cells, proteo-lipobeads are presented, encapsulating functional membrane proteins in a strict orientation into a lipid bilayer. Assays can be performed just as on live cells, for example using fluorescence measurements. As a proof of concept, we have demonstrated proton transport through cytochrome c oxidase.
2015 wurde das Grabungshaus der Abteilung Vorderasiatische Archäologie an der Goethe-Universität in Tell-Chuera, im Nordosten Syriens nur wenige Kilometer von der Grenze zur Türkei gelegen, zum großen Teil zerstört. Das Gebäude wurde 15 Jahre lang von Forschern und Studierenden der Uni im Rahmen eines Projektes zur frühen Urbanisation in Mesopotamien genutzt.
