Electrospinning is a versatile and promising drug delivery technology for the development of tailor-made drug delivery systems for various clinical applications. By applying high voltages to drug-loaded polymer solutions, solid polymeric nanofibers can be generated, which encapsulate active pharmaceutical ingredients (APIs) into their polymer matrix. During the electrospinning process, the fibers are deposited on a collector and form a nonwoven network of drug-loaded polymer fibers. These fibers are spatially distributed in aligned or random orientation, providing the opportunity to design highly tunable structural and mechanical properties, which can be adapted to the biological requirements of the intended application site. The mechanically flexible fiber networks can therapeutically be administered to a multitude of pharmaceutical application sites. Their highly porous fiber structure exhibits a large surface-to-volume ratio, which is ideal for controlled drug release kinetics from the polymer matrix upon contact with biological fluids, such as tear fluid, saliva, mucus, wound exudate or gastro-intestinal fluid. For application at the target site, fiber mats are cut into patches. As the patch size determines the quantity of applied API, the electrospinning process must ensure homogeneous distribution of the API throughout the entire fiber mat area.
In this thesis, electrospinning was established as a formulation technology for the rational fabrication of tailor-made multifunctional drug carrier systems for local and site-specific drug delivery to the epithelial interfaces skin, oral mucosa as well as cornea. For adequate characterization and analysis of the drug delivery systems, a broad panel of robust and predictive analytical tools, based of novel investigation techniques for physicochemical characterization of electrospun fibers, was developed.
The initial part of the thesis thematically focuses on the development of predictive analytical techniques, to determine fiber morphology and physicochemical properties, as well as fiber composition and drug release. By designing two model formulations with contrasting properties, and subsequent analysis and characterization with a set of newly developed techniques and state-of-the-art methods, a comprehensive toolset has been made available and evaluated, aiming at advancing and standardizing respective techniques in the scientific field of electrospun drug delivery systems.
Starting with the initiation of the electrospinning formulation process, which often relies on empirical data rather than analytical methods to predict successful processability, analysis of rheological properties of electrospinning solutions was used to rationally detect the minimum polymer concentration required for electrospinning.
For analysis of fiber morphology, scanning electron microscopy is a common technique. However, little attention is given to underlying readout parameters. By analyzing the fiber orientation and diameter of the respective fibers, predictive results regarding mechanical properties could be obtained, which were subsequently confirmed by measuring elongation force with tensile testing. Confocal Raman microscopy, a label-free method for chemically- selective imaging of the fiber samples, was introduced as a complementary visualization technique, enabling the detection of fiber composition and drug distribution.
A novel technique for investigation of water contact angles on the fiber surface of highly hydrophilic polymers was introduced, which provides predictive data regarding interaction with body fluids and the resulting drug release kinetics. Subsequent release testing in a newly developed setup for analyzing drug release from electrospun fibers in low-volume body compartments, confirmed the anticipated drug release kinetics from measurement of the surface hydrophilicity.
By combining complementary analytical methods, including spectral composition analysis, morphology visualization, characterization of physico-chemical properties and drug release kinetics, as well as the application of multivariate data analysis, a robust and predictive toolset has been established, which can support comparability of future electrospinning studies and the translation from the lab bench into clinics.
Based on the analytical toolset, the main part of the thesis focuses on the development and preparation of electrospun platform drug delivery systems for application on epithelial barriers. Electrospun fiber mats are thin, flat, and mechanically flexible, which allows close adherence to epithelial surfaces and reduction of diffusion paths, which enables efficient drug delivery to the skin, oral mucosa, as well as the cornea.
Electrospun fibers bear a high potential for application as wound dressings, while simultaneously controlling the local delivery of APIs to the wound area. Their close resemblance to the extracellular matrix of human skin provides a suitable microenvironment for cellular proliferation and migration for wound closure. In this work, insulin, a fragile proteohormone with growth factor characteristics, was successfully encapsulated into the core of coaxially electrospun fibers, thus maintaining bioactivity throughout and after the electrospinning process. The shell has been designed from biocompatible polymers, which, upon contact with aqueous wound exudate, partially dissolve and form pores through which bioactive insulin is released in a controlled manner. The shell layer provides a hydrophilic surface for interaction with body fluids and skin cells, and possesses substantial mechanical strength, flexibility, and high tensile elongation required for application on wounds. The biocompatibility of the wound dressing was investigated by interaction with primary human dermal fibroblasts and keratinocytes, which displayed healthy cell morphologies without indicating any elevated levels of cytotoxicity markers.
To investigate the effect of insulin on cell migration, in vitro scratch assays on human skin cells were performed. Increased cellular migration speed and wound closure could be observed, indicating improved wound healing. Bio relevance of in vitro wound healing potential results was advanced by development of 3D ex vivo human epidermal skin wound models from reduction surgery donor material. These complex wound models were treated with electrospun insulin fibers and analyzed by proteome analysis to reveal significant increases in wound healing-associated signaling pathways, which could be attributed to a material-driven remarkably positive impact on wound healing of the electrospun fibers...
An empirical study of the per capita yield of science Nobel prizes : is the US era coming to an end?

We point out that the Nobel prize production of the USA, the UK, Germany and France has been in numbers that are large enough to allow for a reliable analysis of the long-term historical developments. Nobel prizes are often split, such that up to three awardees receive a corresponding fractional prize. The historical trends for the fractional number of Nobelists per population are surprisingly robust, indicating in particular that the maximum Nobel productivity peaked in the 1970s for the USA and around 1900 for both France and Germany. The yearly success rates of these three countries are to date of the order of 0.2–0.3 physics, chemistry and medicine laureates per 100 million inhabitants, with the US value being a factor of 2.4 down from the maximum attained in the 1970s. The UK in contrast managed to retain during most of the last century a rate of 0.9–1.0 science Nobel prizes per year and per 100 million inhabitants. For the USA, one finds that the entire history of science Noble prizes is described on a per capita basis to an astonishing accuracy by a single large productivity boost decaying at a continuously accelerating rate since its peak in 1972.
Transcranial electrical stimulation has widespread clinical and research applications, yet its effect on ongoing neural activity in humans is not well established. Previous reports argue that transcranial alternating current stimulation (tACS) can entrain and enhance neural rhythms related to memory, but the evidence from non-invasive recordings has remained inconclusive. Here, we measure endogenous spindle and theta activity intracranially in humans during low-frequency tACS and find no stable entrainment of spindle power during non-REM sleep, nor of theta power during resting wakefulness. As positive controls, we find robust entrainment of spindle activity to endogenous slow-wave activity in 66% of electrodes as well as entrainment to rhythmic noise-burst acoustic stimulation in 14% of electrodes. We conclude that low-frequency tACS at common stimulation intensities neither acutely modulates spindle activity during sleep nor theta activity during waking rest, likely because of the attenuated electrical fields reaching the cortical surface.
Im Folgenden wird Herders wechselseitiger Entwurf von Mensch und Natur in ein Verhältnis zur Klimakonzeption Kants gesetzt, wie dieser sie im Kontext seiner naturphilosophischen Überlegungen entwickelt. Dabei wird es nicht darum gehen, eine der beiden Positionen gegen die andere auszuspielen, sondern darum, die Denk- und Darstellbarkeit klimatischer Natur – noch vor ihrer Repräsentation in Diagrammen oder dem Versuch der Klimaprognose - auf den Grundlagen von zwei verschiedenen theoretischen Positionen zu rekonstruieren. Ein Hauptaugenmerk gilt der Frage, ob und inwiefern bei der Darstellung klimatischer Natur für beide Autoren Modelle eine Rolle spielen, und zwar in zwei Hinsichten: Erstens gilt die Aufmerksamkeit der Verwendung von Modellen, die rückwirkend als solche identifiziert werden können, von den Autoren aber nicht unbedingt als "Modell" bezeichnet wurden; zweitens der Verbindung zwischen den Darstellungsweisen klimatischer Natur in den Texten und ihrer Reflexion auf den Einsatz von Modellen, die sowohl bei Kant als auch bei Herder in der expliziten Beschäftigung mit der Funktion der Analogie zu finden ist.
"Wellenformen" : die Leistung mathematischer Modellbildung für Akustik, Physiologie und Musiktheorie

Im Jahr 1857 hält Hermann von Helmholtz einen Vortrag 'Ueber die physiologischen Ursachen der musikalischen Harmonien', in dem er erstmals Ergebnisse seiner akustischen und hörphysiologischen Forschungen einer akademischen Öffentlichkeit vorstellt. Dabei bilden die Untersuchungen und Experimente, die Helmholtz im Rahmen seiner Tätigkeit als Professor für Anatomie und Physiologie an der Universität Bonn durchgeführt hat, Grundlage und Ausgangspunkt einer umfassenden Neukonstitution von Wissenszusammenhängen, in deren Zuge ältere Wissensbestände arrondiert, im Lichte neuer Erkenntnisse bewertet, erweitert, neu gefasst und in ausgearbeiteter Form sechs Jahre später unter dem Titel 'Die Lehre von den Tonempfindungen' veröffentlicht werden. In den einleitenden Worten seines Vortrages aus dem Jahr 1857 verweist Helmholtz in diesem Kontext auf einen Aspekt, der ihm offenkundig von großer Signifikanz zu sein scheint:
"Es hat mich immer als ein wunderbares und besonders interessantes Geheimnis angezogen, dass gerade in der Lehre von den Tönen, in den physikalischen und technischen Fundamenten der Musik, die unter allen Künsten in ihrer Wirkung auf das Gemüth als die stoffloseste, flüchtigste und zarteste Urheberin unberechenbarer und unbeschreiblicher Stimmungen erscheint, die Wissenschaft des reinsten und consequentesten Denkens, die Mathematik, sich so fruchtbar erwies."
Bemerkenswert an dieser Aussage ist nicht, dass Musik und Mathematik in eine enge Beziehung zueinander gesetzt werden - hier kann Helmholtz auf eine über zweitausendjährige Tradition der wechselseitigen Elaboration beider Bereiche verweisen -, sondern dass Darlegungen, die auf die systematische Durchdringung der Zusammenhänge zwischen akustischen, physiologischen, psychischen, musiktheoretischen und ästhetischen Phänomenbereichen zielen, ihren Ausgangspunkt in der Mathematik nehmen. Diesen Leistungen, die mathematisches Denken für die Untersuchung und Explikation dieser Zusammenhänge erbringt, möchte der folgende Beitrag nachgehen. Es wird sich zeigen, dass eine spezifische mathematische Operation für das Verständnis von akustischen und physiologischen Prozessen modellbildend wirkt und über verschiedene Applikationswege hinweg neue Impulse der Systematisierung von Wissensbeständen setzt.
Simulationsmodelle

Mit der Entwicklung elektronischer Computer in den 1940er Jahren und höheren Programmiersprachen in den 1950er Jahren hält ein neuer Modelltyp Einzug in die Wissenschaften: Simulationsmodelle. Bekannteste Vertreter sind wohl Klima- und Wettermodelle, die mittlerweile Teil der Alltagskultur geworden sind. Kaum eine Natur- oder Technikwissenschaft kommt heute noch ohne Simulationsmodelle aus und neben der traditionellen Einteilung in Theorie und Empirie fügt sich die Simulation als 'dritte Methode' im Rahmen von 'Computational Departments' in die Wissenschaftslandschaft ein. Dabei ist der Begriff des Simulierens durchaus nicht eindeutig definiert. In einem weiten Sinne kann er im wissenschaftlichen Kontext für jegliche Form des Nachahmens und Imitierens verwendet werden: ein Crashtest im Labor simuliert einen Autounfall, ein Schiffsmodell im Strömungskanal bildet maßstabsgerecht ein Containerschiff nach und ein Ball-Stick-Model imitiert ein Molekül. Dennoch hat sich im wissenschaftlichen Kontext der Begriff des Simulierens auf die Computersimulation zentriert und in unterschiedliche Subkategorien ausdifferenziert:
– deterministische Simulationen basierend auf Differentialgleichungen
– stochastische Simulationen basierend auf stochastischen Differentialgleichungen oder
Zufallsläufeerzeugungsmethoden wie der Monte-Carlo-Simulation
– ereignisbasierte Simulationen, in denen bestimmte Ereignisse andere Ereignisse auslösen
– sogenannte 'Soft Computing'-Methoden wie Agentenbasierte Simulationen, Genetische
Programmierung, Evolutionäre Algorithmen oder Neuronale Netze.
Im vorliegenden Zusammenhang soll der Begriff des Simulierens jedoch einzig auf deterministische Simulationen bezogen werden. Diese Simulationsart ist nicht nur die weitest verbreitete in den Natur- oder Technikwissenschaften, sie ist auch die älteste und damit klassische Form der Simulation.
Im Folgenden wird Herders wechselseitiger Entwurf von Mensch und Natur in ein Verhältnis zur Klimakonzeption Kants gesetzt, wie dieser sie im Kontext seiner naturphilosophischen Überlegungen entwickelt. Zwischen 'Umwelt' und 'milieu' : zur Begriffsgeschichte von 'environment' in der Evolutionstheorie

Wohl kaum ein Begriff ist derzeit ähnlich aufgeladen, belegt und machtvoll wie der des auch ins Deutsche übergehenden 'environment'. Er ist allgegenwärtig, zum theoriestrategischen wie handlungsleitenden Instrument geworden und geht über seine angestammten Fachgebiete im Bereich der Biologie und der Ökologie sowie die Vielfalt umweltpolitischer Programme weit hinaus. Die ersten Schritte auf diesem Weg der Ausweitung, die den Begriff derzeit auf unterschiedlichsten Gebieten plausibel macht, sollen im Folgenden nachvollzogen werden. Dabei wird besonderes Augenmerk auf sein Verhältnis zu zwei benachbarten Begriffen gelegt werden, die zwar häufig zur Übersetzung herangezogen werden, dadurch aber ihre eigene Spezifik und historische Tiefe zu verlieren drohen: 'Umwelt' und 'milieu'. Werden diese drei Ausdrücke füreinander ein- oder gar miteinander gleichgesetzt, vermischen sich ihre in entscheidenden Punkten voneinander abweichenden Theorietraditionen. Ein begriffsgeschichtlicher Blick auf die Etablierung von 'environment' in der britischen Evolutionstheorie des 19. Jahrhunderts soll diese bislang viel zu selten beachteten Komplikationen verständlich machen - angesichts seiner momentanen Ausweitung, in der alle drei Begriffe selbstverständlich zu werden drohen.
Trotz der Annäherung der Begriffe 'Geschichte' und 'Evolution' unter dem Vorzeichen einer semantischen Verschiebung kulturellen Kapitals haben sich bis in die Gegenwart auch begriffliche Differenzen erhalten. So gibt es neben der unbekümmerten Anwendung von 'Geschichte' und 'Evolution' auf alle sich in der Zeit verändernden Dinge auch Versuche, die Begriffe terminologisch stark zu machen und in der Folge dessen 'Evolution' für den Bereich der Natur und 'Geschichte' für den der Kultur zu reservieren. Der Beitrag liefert eine historische Rekonstruktion dieser Entwicklung. Am Anfang stehen dabei einige quantitative sprachwissenschaftliche Beobachtungen zur Entwicklung der Häufigkeit der Begriffe in verschiedenen Textgattungen und zur gegenwärtigen Semantik durch einen Vergleich der häufigsten Genitivattribute. Im zweiten Abschnitt wird die Veränderung des Ausdrucks 'Geschichte' untersucht, zunächst in seiner terminologischen Bedeutung in den Geschichtswissenschaften, dann in seiner Ausweitung auf Gegenstände der Natur. Der dritte Abschnitt liefert eine analoge Untersuchung zu 'Evolution', ausgehend von den Naturwissenschaften und in der Ausweitung auf kulturelle Phänomene. Im vierten Abschnitt wird eine Verbindung der beiden Begriffe näher betrachtet, die im Sinne einer semantischen Verschränkung wirksam ist und sich unter anderem daraus ergibt, dass das Wort 'Evolution' eine teleologische Konnotation hat, die bei 'Geschichte' nicht vorliegt. Der fünfte Abschnitt schließlich beleuchtet die wissenschaftliche Stellung der Begriffe in der Gegenwart und erwägt die Aussichten ihrer terminologischen Differenzierung.
In the 20th century, the term "ecosystem" was one of the most important concepts for the biological discipline "ecology." Originally coined by the English botanist Arthur G. Tansley in an article from 1935, it is now a well-established term. The authors of the textbook 'Ecology' write, the ecosystem concept "has become a powerful tool for integrating ecology with other disciplines." But this only addresses the scientific resonance of the term. In the 1970s "ecosystem" also became an important concept for the environmental movement, for the term "ecosystem" describes nature as a whole entity, in which all things are linked together, forming a network of biotic and abiotic factors. In this sense, the "ecosystem" concept also took on a key role in the political ecology discourse. This article begins with a look at the political ecology discourse, and then focuses on the formation of the "ecosystem" concept. The terminological development of the term turns first to the linguistic definition of "ecosystem" before looking how the ecosystem became an established concept by transforming the object "lake" into the scientific object "ecosystem." Sections four and five further pursue the role of the ecosystem concept in the environmental discourse, based on the metaphor of "spaceship earth" on the one hand and of the "closing circle" on the other. Finally, the article contextualizes the "ecosystem" concept in conjunction with Claude Lefort's concept of "the political." As we will see, the political impact of the "ecosystem" concept inheres in the very term itself for it describes a wholeness that human beings are inevitably a part of even as their actions alter or disturb with the ecosystem fundamentally. In other words, human beings are both inside and outside of the "ecosystem" at the same time. This paradoxical situation is inevitably constituted by the concept "ecosystem", which is understood as a (nearly) closed system. Hence solutions to environmental problems aim at reintegrating human beings into the closed circle of the global ecosystem through technical constructions or through adapting to natural processes.
