In Deutschland leidet ca. jeder zehnte Mensch über 40 Jahren an einer chronischen Einschränkung seiner Nierenfunktion. Nicht wenige davon sind im Laufe der Erkrankung auf eine Nierenersatztherapie angewiesen. Die Ursachen für eine Nierenschädigung sind vielfältig. Als neuartiger und vielversprechender Therapieansatz werden aktuell Mesenchymale Stamm-/Stromazellen (MSC) als Therapeutikum für diverse Nierenerkrankungen getestet. Erste Ergebnisse klinischer Phase-I-Studien zeigen, dass MSC sicher als Immunsuppressivum nach Nierentransplantation angewendet werden können. Auch für weitere Erkrankungen der Niere sind erste klinische Studien am Laufen. MSC gelten als regenerativ, immunsupprimierend und antientzündlich. Dennoch gibt es noch einige Limitationen. Nach der Transplantation der Zellen ist das Wachstum der Zellen oft eingeschränkt und es kommt zur vermehrten Apoptose. Auch wird immer wieder ein paradoxes und entzündungsförderndes Verhalten der MSC am Wirkort beobachtet. Ein wichtiger Lösungsansatz ist eine in vitro Vorbehandlung der MSC zur Modulierung der zellulären Eigenschaften. In dieser Arbeit wurden drei Substanzen und Arzneimittel auf ihre Fähigkeit untersucht, die entzündungshemmenden Eigenschaften der MSC zu verbessern und die entzündungsfördernden zu unterdrücken. Der Fokus lag hierbei auf dem Arzneimittel Niclosamid und den beiden bisher noch nicht zugelassenen Substanzen Berberin und Gedunin, die alle in vitro und in vivo bereits erste vielversprechende antientzündliche Wirkungen bewiesen haben. Für diese Arbeit wurden MSC aus Fettgewebe isoliert (ASC) und mit LPS oder einem Cytokin-Mix (Mischung aus TNF-α, IFN-γ und IL-1β) sowie den drei Substanzen stimuliert. Untersucht wurden im Anschluss die mRNA-Expressionen der gängigsten proinflammatorischen (TNF-α, IL-6, TLR-4, ICAM-1, HLA-DR) und antiinflammatorischen Marker (IDO, IL-10), sowie mittels ELISA die Protein-Freisetzung von IL-6 und IL-8. Die vielversprechendsten Ergebnisse ließen sich durch Berberin hervorrufen. Hier zeigte sich eine deutliche Senkung der IL-8-Konzentration im ELISA. Die Anwendung des Gedunin hatte keine signifikante Wirkung auf die ASC. Niclosamid hingegen scheint widererwarten sogar entzündungsfördernd zu wirken, in dem es die IL-6-, ICAM-1-mRNA-Expression steigerte und die IDO-mRNA-Expression absenkte. Unter den drei getesteten Subtanzen hat Berberin die beste Wirkung bewiesen. Nach weiterer Testung könnte eine Anwendung mit Berberin als in vitro Präkonditionierung von MSC vielversprechend sein. Die Verwendung von Niclosamid hingegen sollte vermieden werden, die Wirkung von Gedunin müsste genauer untersucht werden.
Das Schilddrüsenkarzinom (SK) ist die häufigste bösartige endokrine Tumorerkrankung. Während das nicht-metastasierte und nicht-mutierte papilläre Schilddrüsenkarzinom (PSK) und das follikuläre Schilddrüsenkarzinom (FSK) eine gute Heilungschance aufweisen, zeigen die mutierten und metastasierten Varianten des PSK und FSK sowie das anaplastische Schilddrüsenkarzinom (ASK) weiterhin eine schlechte Prognose. Die Entwicklung von Therapieresistenzen stellen hierbei ein Hauptproblem in der Behandlung des fortgeschrittenen Schilddrüsenkarzinoms dar.
In den letzten Jahren wurden in Studien zunehmend Tumor-initiierende Zellen (TIZ) beschrieben, welche eine kleine Subpopulation von Zellen mit der Fähigkeit zur Selbsterneuerung, Tumorinitiierung und Entwicklung von Therapieresistenzen von Tumoren darstellen. Die Existenz von TIZ wurde auch im SK nachgewiesen. Ein entscheidender Faktor für die Persistenz von TIZ ist die Hypoxie, welche über eine Veränderung des Tumormikromilieus und des Zellmetabolismus zur Entstehung von Therapieresistenzen beiträgt. Ein durch Hypoxie hochreguliertes Enzym ist die Carboanhydrase IX (CAIX). CAIX wird hauptsächlich von Tumorzellen exprimiert und katalysiert die Reaktion von Kohlendioxid zu Bicarbonat und einem Proton und trägt damit zur Säurepufferung der Tumorzelle bei. CAIX stellt somit einen entscheidenden Faktor für das Überleben von Tumorzellen in einem hypoxischen Milieu dar. Des Weiteren ist eine erhöhte Expression von CAIX mit einem schlechten Patienten-Outcome assoziiert, wie z.B. im Brustkrebs. Diese Eigenschaften machen CAIX zu einem attraktiven Angriffspunkt einer zielgerichteten Tumortherapie. Die vorliegende Studie hat zum Ziel, die Expression von CAIX sowie dessen biologische Rolle im Schilddrüsenkarzinom näher zu untersuchen.
Hierzu wurden Proben von 114 SK-Patienten immunhistochemisch auf eine CAIX-Expression untersucht und mit tumorfreiem Schilddrüsengewebe verglichen. Hierbei waren unterschiedliche SK-Subtypen vertreten. Für eine weitere Validierung der Expressionsdaten erfolgte die Auswertung eines Datasets von „The Cancer Genome Atlas“ (TCGA) mithilfe von cBioportal. Da die Hypoxie ein wichtiger Faktor für die Persistenz von TIZ ist, wurde die CAIX-Expression in Tumorsphären, ein in vitro Nachweis von TIZ-Aktivität, mittels der Durchflusszytometrie bestimmt und mit der CAIX-Expression von Monolayern verglichen. Als SK-Zelllinien wurden BCPAP (PSK), FTC 133 (FSK) und 8505 C (ASK) verwendet. Anschließend wurde mithilfe der Polymerasekettenreaktion und Immunofluoreszenzfärbung untersucht, ob eine CAIX-Expression in den Tumorsphären mit der Expression von bereits bekannten Stammzellmarkern, u.a. NANOG, assoziiert ist. Die Unterschiede der CAIX-Expression, nach Inkubation der Monolayer jeweils in Normoxie und Hypoxie, wurden mittels Durchflusszytometrie bestimmt. Mithilfe eines genetischen CAIX-Knockdowns sowie einer pharmakologischen Inhibition mit dem CAIX-Inhibitor Methazolamid (MZM) wurde die Tumorzellproliferation und -Sphärenbildung unter Normoxie und Hypoxie bestimmt. Zusätzlich wurde der Einfluss von MZM auf die Apoptose und den Zellzyklus untersucht.
Immunhistochemische Färbungen der Gewebeproben von SK-Patienten zeigten, dass die CAIX-Expression sowohl im PSK und FSK als auch im ASK im Vergleich zum tumorfreien Schilddrüsengewebe erhöht war. Des Weiteren zeigte die klinisch-pathologische Datenanalyse, dass eine erhöhte CAIX-Expression mit dem Auftreten von Lymphknotenmetastasen im differenzierten SK assoziiert war. Auch die Analyse des TCGA-Datasets bestätigte, dass eine erhöhte Expression der CAIX-mRNA mit einem fortgeschrittenen Tumorstadium, Fernmetastasen und mit einem kürzeren Gesamt-Überleben von SK-Patienten korrelierte. Die weiteren funktionellen in vitro Untersuchungen ergaben, dass die CAIX-Expression in den Tumorsphären im Vergleich zu Monolayern erhöht und mit einer erhöhten Expression von Stammzellmarkern assoziiert war. Ein genetischer CAIX-Knockdown und eine CAIX-Inhibition mit MZM führten über eine Induktion der Apoptose und eines Zellzyklusarrests zu einer verminderten Tumorzellproliferation und Sphärenbildung.
Zusammenfassend deuten die Ergebnisse darauf hin, dass CAIX ein vielversprechendes Zielmolekül für eine gezielte Tumortherapie des fortgeschrittenen SK ist. Um diese Hypothese bestätigen zu können, sind jedoch weitere prospektive Analysen von Patientenproben sowie funktionelle in vivo Untersuchungen am SK nötig.
Exploring strategies to improve the reverse beta-oxidation pathway in Saccharomyces cerevisiae

Microbes are the most diverse living organisms on Earth, with various metabolic adaptations that allow them to live in different conditions and produce compounds with different chemical complexity. Microbial biotechnology exploits the metabolic diversity of microorganisms to manufacture products for different industries. Today, the chemical industry is a significant energy consumer and carbon dioxide emitter, with processes that harm natural ecosystems, like the extraction of medium-chain fatty acids (MCFAs). MCFAs are used as precursors for biofuels, volatile esters, surfactants, or polymers in materials with enhanced properties.
However, their current extraction process uses large, non-sustainable monocultures of coconut and palm trees. Therefore, the microbial production of MCFAs can help reduce the current environmental impact of obtaining these products and their derivatives.
In nature, fatty acids are mostly produced via fatty acid biosynthesis (FAB). However, the reverse β-oxidation (rBOX) is a more energy-efficient pathway compared to FAB. The rBOX pathway consists of four reactions, which result in the elongation of an acyl-CoA molecule by two carbon units from acetyl-CoA in each cycle. In this work we used Saccharomyces cerevisiae, an organism with a high tolerance towards toxic compounds, as the expression host of the rBOX pathway to produce MCFAs and medium-chain fatty alcohols (MCFOHs).
In the first part of this work, we expanded the length of the products from expressing the rBOX in the cytosol and increased the MCFAs titres. First, we deleted the major glycerol-3-phosphate dehydrogenase (GPD2). This resulted in a platform strain with significantly reduced glycerol fermentation and increased rBOX pathway activity, probably due to an increased availability of NADH. Then, we tested different combinations of rBOX enzymes to increase the length and titres of MCFA. Expressing the thiolase CnbktB and β-hydroxyacyl-CoA dehydrogenase CnpaaH1 from Cupriavidus necator, Cacrt from Clostridium acetobutylicum and the trans-enoyl-CoA reductase Tdter (Treponema denticola) resulted in hexanoic acid as the main product.
Expressing Cncrt2 (C. necator) or YlECH (Y. lipolytica) as enoyl-CoA hydratases resulted in octanoic acid as the main product. Then, we integrated the octanoic (Cncrt2 or YlECH) and the hexanoic acid (Cacrt)-producing variants in the genome of the platform strain and we achieved titers of ≈75 mg/L (hexanoic acid) and ≈ 60 mg/L (octanoic acid) when growing these strains in a complex, highly buffered medium. These are the highest titers of octanoic and hexanoic acid obtained in S. cerevisiae with the rBOX. Additionally, we deleted TES1 and FAA2 to prevent competition for butyryl-CoA and degradation of the produced fatty acids, respectively.
However, these deletions did not improve MCFA titers. In addition, we tested two dual acyl-CoA reductase/alcohol dehydrogenases (ACR/ADH), CaadhE2 from C. acetobutylicum and the putative ACR/ADH EceutE from Escherichia coli, in an octanoyl-CoA-producing strain to produce MCFOH. As a result, we produced 1-hexanol and 1-octanol for the first time in S. cerevisiae with these two enzymes. Nonetheless, the titres were low (<10 mg/L and <2 mg/L, respectively), and four-carbon 1-butanol was the main product in both cases (>80 mg/L). This showed the preference of these two enzymes for butyryl-CoA.
In the second part of this work, we expressed the rBOX in the mitochondria of S. cerevisiae to benefit from the high levels of acetyl-CoA and the reducing environment in that organelle. First, in an adh-deficient strain, we mutated MTH1, a transcription factor regulating the expression of hexose transporters, and deleted GPD2. This resulted in a strain with a reduced Crabtree effect and, therefore, an increased carbon flux to the mitochondria. We partially validated the increased flux to the mitochondria by expressing the ethanol-acetyltransferase EAT1 from Kluyveromyces marxianus in this organelle. This resulted in a higher isoamyl acetate production in the MTH1-mutant strain. Isoamyl acetate is synthesised by Eat1 from acetyl-CoA and isoamyl alcohol, a product of the metabolism of amino acids in the mitochondria. Then, we targeted different butyryl-CoA-producing rBOX variants to the mitochondria, and we used the production of 1-butanol and butyric acid as a proof-of-concept. The strong expression of all the enzymes was toxic for the cell, and the highest butyric acid titres (≈ 50 mg/L) in the mitochondria from the rBOX were obtained from the weak expression of the pathway. The highest 1-butanol titers (≈ 5 mg/L) were obtained with the downregulation of the mitochondrial NADH-oxidase NDI1. However, this downregulation led to a non-desirable petite phenotype.
In summary, we produced hexanoic and octanoic acid for the first time in S. cerevisiae using the rBOX and achieved the highest reported titers of hexanoic and octanoic acid so far using this pathway in S. cerevisiae. In addition, we successfully compartmentalised the rBOX in the mitochondria. However, competing reactions, some of them essential for the viability of the cell, limit the use of this organelle for the rBOX.
Inorganic phosphate is one of the most abundant and essential nutrients in living organisms. It plays an indispensable role in energy metabolism and serves as a building block for major cellular components such as the backbones of DNA and RNA, headgroups of phospholipids and in posttranslational modifcations of many proteins. Disturbances in cellular phosphate homeostasis have a detrimental effect on the viability of cells. There- fore, both the import and export of phosphate is strictly regulated in eukaryotic cells. In the eukaryotic model organism Saccharomyces cerevisiae, the uptake of phosphate is carried out either by transporters with high affinity or by transporters with low affinity, depending on the cytosolic phosphate concentration. While structures are available for homologues of the high-affinity transporters, no structures of low-affinity transporters have been solved so far. Interestingly, only the low-affinity transporters have a regulatory SPX domain, which is found in various proteins involved in phosphate homeostasis.
In this work, structures of Pho90 from Saccharomyces cerevisiae, a low-affinity phosphate transporter, were solved by cryo-EM, providing insights into its transport mechanism. The dimeric structure resembles the structures of proteins of the divalent anion symporter superfamily (DASS) and of mammalian transporters of the solute carrier 13 (SLC13) family. The transmembrane domain of each protomer consists of 13 helical elements and can be subdivided into scaffold and transport domains. The structure of ScPho90 in the presence of phosphate shows the phosphate binding site within the transporter domain in an outward-open conformation with a bound phosphate ion and two sodium ions. In the absence of phosphate, an asymmetric dimer structure was determined, with one protomer adopting an inward-open conformation. While the dimer contact and the scaffold domain are identical in both conformations, the transport domain is rotated by about 30° and shifted by 11 Å towards the cytoplasmic side, leading to the accessibility of the binding pocket from the cytoplasm. Based on these findings and by comparison with known structures, a phosphate transport mechanism is proposed in the present work that involves substrate binding on the extracellular side, conformational change by a rigid-body motion of the transport domain, in an "elevator-like" motion, and substrate release into the cytoplasm. The regulatory SPX domain is not well resolved in the ScPho90 structures, so that no direct conclusions were drawn about its regulatory mechanism. The findings provide new insights into the function and mechanism of eukaryotic low-affinity phosphate transporters.
While eukaryotic cells express various phosphate import proteins, most eukaryotes have only a single highly conserved and essential phosphate exporter. These exporters show no sequence homology to other transporters of known structure, but also possess a regulatory SPX domain. In this work, the structural basis for eukaryotic phosphate export is investigated by elucidating the structures of the homologous phosphate exporters Syg1 from Saccharomyces cerevisiae and Xpr1 from Homo sapiens, using cryo-EM. The structures of ScSyg1 and HsXpr1 show a conserved homodimeric structure and the transmembrane part of each protomer consists of 10 TM helices. Helix TM1 establishes the dimer contact by means of a glycine zipper motif, which is a known oligomerization motif. Helices TM2-5 form a hydrophobic pocket that has density for a lipid molecule. Whether the lipid binding into the hydrophobic pocket has an allosteric effect on the phosphate export activity or only serves protein stabilization is not known. Helices TM5-10 form a six-helix bundle, which constitutes a putative phosphate translocation pathway in its center. This bundle is formed by the protein sequence annotated as EXS domain.
The respective phosphate translocation pathways of ScSyg1 and HsXpr1 show structural differences. While the translocation pathway in HsXpr1 is accessible from the cytoplasm, in ScSyg1 it is closed by a large loop of the SPX domain. Interestingly, this loop is not conserved in higher eukaryotes and is therefore not present in HsXpr1. Another difference are distinct conformations of helix TM9. In ScSyg1, TM9 adopts a kinked conformation, which results in the translocation pathway being open to the extracellular side. In contrast, TM9 adopts a straight conformation in HsXpr1, resulting in the placement of a highly conserved tryptophane residue in the middle of the translocation pathway. As a result, the translocation pathway in HsXpr1 is closed to the extracellular side.
Autophagy is an important degradation pathway mediating the engulfment of cellular material (cargo) into autophagosomes followed by degradation in autophagosomes.
Different stress stimuli, e.g. nutrient deprivation, oxidative stress or organelle damage, engage autophagy to maintain cellular homeostasis, recycle nutrients or remove damaged cell organelles. Autophagy not only degrades bulk cytoplasmic material but also selective autophagic cargo, for example lysosomes (lysophagy), mitochondria (mitophagy), ER (ER-phagy), lipid droplets (lipophagy), protein aggregates (aggrephagy) or pathogens (xenophagy). Selective autophagy pathways are regulated by selective autophagy receptors which bind to ubiquitinated cargo proteins and link them to LC3 on the autophagosomal membrane.
Ubiquitination is an essential post-translational modification controlling different cellular processes such as proteasomal and lysosomal degradation or innate immune signaling.
M1-linked (linear) poly-Ubiquitin (poly-Ub) chains are exclusively assembled by the E3 ligase linear ubiquitin chain assembly complex (LUBAC) and removed by the M1 poly-Ub-specific OTU domain-containing deubiquitinase with linear linkage specificity (OTULIN). In addition to key functions in innate immune signaling and nuclear factor-κB (NF-κB) activation, M1 ubiquitination is also implicated in the regulation of autophagy.
LUBAC and OTULIN control autophagy initiation and maturation and the autophagic clearance of invading bacteria via xenophagy. However, additional functions of LUBAC- and OTULIN-regulated M1 ubiquitination in autophagy are largely unknown and it also remains unexplored if LUBAC and OTULIN control other selective autophagy pathways in addition to xenophagy. This study aimed to unravel the role of LUBAC- and OTULIN-controlled M1 ubiquitination in bulk and selective autophagy in more detail.
In this study, characterization of OTULIN-depleted MZ-54 glioblastoma (GBM) cells revealed that OTULIN deficiency results in enhanced LC3 lipidation in response to autophagy induction and upon blockade of late stage autophagy with Bafilomycin A1 (BafA1). Furthermore, electron microscopy analysis showed that OTULIN-deficient cells have an increased number of degradative compartments (DGCs), confirming enhanced autophagy activity upon loss of OTULIN. APEX2-based autophagosome content profiling identified various OTULIN-dependent autophagy cargo proteins. Among these were the autophagy receptor TAX1BP1 which regulates different forms of selective autophagy (e.g. lysophagy, aggrephagy) and the glycan-binding protein galectin-3 which serves key functions in lysophagy, suggesting a role of OTULIN and M1 poly-Ub in the regulation of aggrephagy and lysophagy.

To study aggrephagy, protein aggregation was induced with puromycin which causes premature termination of translation and accumulation of defective ribosomal products (DRiPs). Loss of OTULIN increased the number of M1 poly-Ub-positive foci and insoluble proteins and reduced the levels of soluble TAX1BP1 and p62 in response to puromycin-induced proteotoxic stress.
Intriguingly, upon induction of lysosomal membrane permeabilization (LMP) with the lysosomotropic drug L-Leucyl-L-Leucine methyl ester (LLOMe), M1 poly-Ub strongly accumulated at damaged lysosomes and colocalized with TAX1BP1- and galectin-3-positive puncta. M1 poly-Ub-modified lysosomes formed a platform for NF-κB essential modulator (NEMO) and inhibitor of κB (IκB) kinase (IKK) complex recruitment and local NF-κB activation in a K63 poly-Ub- and OTULIN-dependent manner. Furthermore, inhibition of lysosomal degradation enhanced LLOMe-induced cell death, suggesting pro-survival functions of lysophagy following LMP. Enrichment of M1 poly-Ub at damaged lysosomes was also observed in human dopaminergic neurons and in primary mouse embryonic cortical neurons, confirming the importance of M1 poly-Ub in the response to lysosomal damage.
Together, these results identify OTULIN as a negative regulator of autophagy induction and the autophagic flux and reveal OTULIN-dependent autophagy cargo proteins.
Furthermore, this study uncovers novel and important roles of M1 poly-Ub in the response to lysosomal damage and local NF-κB activation at damaged lysosomes.
Die Appendizitis stellt mit einer Inzidenz von 115 pro 100000 Einwohnern in Deutschland eine der häufigsten Ursachen für ein akutes Abdomen dar. Bakterielle Infektionen sind ein wesentlicher Faktor für die postoperative Morbidität nach Appendektomie.
Ziel dieser prospektiven Studie war es, das Keimspektrum und insbesondere die Prävalenz resistenter Keime bei der akuten Appendizitis zu bestimmen und die Auswirkungen resistenter Keime auf das Auftreten infektiöser Komplikationen zu analysieren. Alle erwachsenen Patient*innen mit akuter Appendizitis, die zwischen April 2022 und Juli 2023 am Universitätsklinikum Frankfurt operativ behandelt wurden, wurden prospektiv eingeschlossen. Das Keimspektrum der Appendix und die Häufigkeit von MRE in Rektalabstrichen wurden analysiert. Die klinischen Daten wurden extrahiert, um die Korrelation des Keimspektrums mit dem Auftreten von postoperativen Komplikationen, Dauer und Art der Antibiotikatherapie und dem postoperativen Verlauf zu evaluieren. 30 Tage nach der Operation wurde ein Follow-up durchgeführt. Insgesamt wurden 105 Patient*innen in die Studie eingeschlossen.
In den Appendixabstrichen gelang ein Erregernachweis bei 67,6 % aller Fälle. Hierbei betrug die Prävalenz von Keimen mit Antibiotikaresistenz 43,8 %, Multipler-Antibiotikaresistenz (MAR) 27,6 % und bei 5,7 % der Fälle gelang ein MRE-Nachweis nach CDC im Appendixabstrich. Beim MRE-Screening konnten im Rektalabstrich bei 11,4 % der Patient*innen ein MRE nachgewiesen werden. In vier Fällen zeigte sich eine Übereinstimmung zwischen rektalen und appendikulären Abstrich, somit betrug die Sensitivität des MRE-Screenings für einen Nachweis multiresistenter Keime in der Appendix lediglich 33,3 % bei einer Spezifität von 86,7 %. In einer univariaten Analyse konnte das Vorliegen eines Diabetes mellitus als Risikofaktor für das Auftreten von Keimen mit Cefuroxim- Resistenz, Multipler-Antibiotikaresistenz (MAR) und MRE identifiziert werden.
Insgesamt erhielten 47,6 % aller Patient*innen postoperativ eine Antibiotikatherapie, von denen erfolgte bei 46 % eine Umstellung der empirischen Antibiose auf eine antibiogramm-gerechte Therapie nach Erhalt des mikrobiologischen Befundes. Insbesondere Patient*innen mit komplizierter Appendizitis erhielten postoperativ eine Antibiotikatherapie, wobei 28 % eine Antibiotikaeskalation benötigten.
Patient*innen, deren Appendixabstriche eine Resistenz gegen maximal ein Antibiotikum aufwiesen, wurden als nicht multiple Resistenz (Nicht-MAR) definiert und mit Patient*innen verglichen, deren Keime mit multipler Antibiotikaresistenz (MAR) waren. Die MAR-Gruppe zeigte im Vergleich zur Nicht-MAR-Gruppe eine höhere Inzidenz postoperativer Komplikationen, insbesondere eine erhöhte Inzidenz von Clavien-Dindo Grad 3 Komplikationen sowie von tiefen postoperativen Wundinfektionen (CDC Grad A3). Weiterhin benötigten Patient*innen der MAR-Gruppe häufiger eine postoperative Antibiotikatherapie und es erfolgte häufiger eine Eskalation der antibiotischen Therapie. Dies ging auch mit einem signifikant verlängerten Krankenhausaufenthalt einher.
Zusammenfassend kann festgestellt werden, dass eine bakterielle Infektion mit Multipler Antibiotikaresistenz bei der akuten Appendizitis häufig auftritt und die Morbidität nach Appendektomie beeinflusst. Die mikrobiologische Untersuchung mittels Appendixabstrich bei operativ behandelten Fällen von akuter komplizierter Appendizitis, die eine postoperative Antibiotikatherapie erfordern, könnte somit eine gezielte und kürzere Antibiotikatherapie ermöglichen. Dies könnte dazu beitragen, längere Krankenhausaufenthalte zu vermeiden, den unnötigen Einsatz unwirksamer Antibiotika zu reduzieren und die Kosten im Gesundheitswesen zu senken.
The PhD addresses the feasibility of reconstructing open charm mesons with the Compressed Baryonic Matter experiment, which will be installed at the FAIR accelerator complex at Darmstadt/Germany. The measurements will be carried out by means of a dedicated Micro Vertex Detector (MVD), which will be equipped with CMOS Monolithic Active Pixel Sensors (MAPS). The feasibility of reconstructing the particles with a proposed detector setup was studied.
To obtain conclusive results, the properties of a MAPS prototype were measured in a beam test at the CERN-SPS accelerator. Based on the results achieved, a dedicated simulation software for the sensors was developed and implemented into the software framework of CBM (CBMRoot). Simulations on the reconstruction of D0-mesons were carried out. It is concluded that the reconstruction of those particles is possible.
The PhD introduces the physics motivation of doing open charm measurements, represents the results of the measurements of MAPS and introduces the innovative simulation model for those sensors as much as the concept and results of simulations of the D0 reconstruction.
Ziel: Nebennierenraumforderungen können sonographisch mit guter Sensitivität und Spezifität erkannt werden. Ziel der vorliegenden Studie war es, die Wertig-keit der Kontrastmittel-Sonographie (CEUS = contrast enhanced ultrasound) unter Verwendung von Zeit-Intensitäts-Kurven für die Charakterisierung von Nebennierenraumforderungen zu bewerten.
Material und Methoden: 108 Patienten mit 116 Raumforderungen der Neben-niere erhielten einen Ultraschall der Nebenniere, inklusive einer Kontrastmittel-Sonographie mit dem Kontrastmittel SonoVue®. Die Kontrastmitteldynamik wurde mittels Zeit-Intensitätskurven aufgezeichnet. Anhand der Kontrastmittel-anflutungszeit wurden folgende vier Kontrastmittelanreicherungsmuster ver-wendet, welche in einer Pilotstudie1 entwickelt wurden: Anreicherungsmuster I = früh arterielle Kontrastmittelanreicherung, Anreicherungsmuster II = arterielle Kontrastmittelanreicherung, Anreicherungsmuster III = späte Kontrastmittelan-reicherung, Anreicherungsmuster IV = keine Kontrastmittelanreicherung. Zu-sätzlich erhielten alle Patienten eine Beurteilung der Nebenniere durch ein zweite Bildgebendes Verfahren (Computertomographie (CT) oder Magnetresonanztomographie (MRT)) sowie eine laborchemische Hormondiagnostik. In Fällen mit malignitätsverdächtigem Befund erfolgte eine Biopsie oder Adrenalektomie.
Ergebnisse: CEUS- Anreicherungsmuster I und II konnten bei allen Patienten mit primärem oder sekundärem Malignom der Nebenniere (n = 16) nachgewie-sen werden. Die Sensitivität der CEUS für die Diagnose einer malignen Neben-nierenraumforderung betrug 100% (95% CI [75; 100]) und die Spezifität 67% (95% CI [56; 75]). Bei 40 Nebennierenraumforderungen war die Histologie die Referenzmethode. In 68% dieser Nebennierenraumforderungen waren MRT/CT und CEUS kongruent in der Beurteilung der Dignität. Schlussfolgerung: Die Kontrastmittel-Sonographie ist eine nützliche ergän-zende Methode in der Aufarbeitung von Nebennierenraumforderungen mit exzellenter Sensitivität für die Diagnose eines Malignoms.
Most elements heavier than iron are synthesized in stars during neutron capture reactions in the r- and s-process. The s-process nucleosynthesis is composed of the main and weak component. While the s-process is considered to be well understood, further investigations using nucleosynthesis simulations rely on measured neutron capture cross sections as crucial input parameters. Neutron capture cross sections
relevant for the s-process can be measured using various experimental methods. A prominent example is the activation method relying on the 7Li(p,n)7Be reaction as a neutron source, which has the advantage of high neutron intensities and is able to create a quasi-stellar neutron spectrum at kBT = 25 keV. Other neutron sources able to provide quasi-stellar spectra at different energies suffer from lower neutron intensities. Simulations using the PINO tool suggest the neutron activation of samples with different neutron spectra, provided by the 7Li(p,n)7Be reaction, and a subsequent linear combination of the obtained spectrum-averaged cross sections
to determine the Maxwellian-averaged cross section (MACS) at various energies of astrophysical relevance. To investigate the accuracy of the PINO tool at proton energies between the neutron emission threshold at Ep = 1880.4 keV and 2800 keV,
measurements of the 7Li(p,n)7Be neutron fields are presented, which were carried out at the PTB Ion Accelerator Facility at the Physikalisch-Technische Bundesanstalt in Braunschweig. The neutron fields of ten different proton energies were measured.
The presented neutron fields show a good agreement at proton energies Ep = 1887, 1897, 1907, 1912 and 2100 keV. For the other proton energies, E p = 2000, 2200, 2300, 2500, and 2800 keV, differences between measurement and simulation were found and discussed. The obtained results can be used to benchmark and adapt the PINO tool and provide crucial information for further improvement of the neutron activation method for astrophysics.
An application for the 7Li(p,n)7Be neutron fields is presented as an activation experiment campaign of gallium, an element that is mostly produced during the weak s-process in massive stars. The available cross section data for the 69,71Ga(n,γ)
reactions, mostly determined by activation measurements, show differences up toa factor of three. To improve the data situation, activation measurements were carried out using the 7Li(p,n)7Be reaction. The neutron capture cross sections for
a quasi-stellar neutron spectrum at kBT = 25 keV were determined for 69Ga and 71Ga.
In this dissertation the formal abstraction and verification of analog circuit is examined. An approach is introduced that automatically abstracts a transistor level circuit with full Spice accuracy into a hybrid automaton (HA) in various output languages. The generated behavioral model exhibits a significant simulation speed-up compared to the original netlist, while maintaining an acceptable accuracy, and can be therefore used in various verification and validation routines. On top of that, the generated models can be formally verified against their Spice netlists, making the obtained models correct by construction.
The generated abstract models can be extended to enclose modeling as well as technology dependent parameter variations with little over approximations. As these models enclose the various behaviors of the sampled netlists, the obtained models are of significant importance as they can replace several simulations with just a single reachability analysis or symbolic simulation. Moreover, these models can be as well be used in different verification routines as demonstrated in this dissertation.
As the obtained models are described by HAs with linear behaviors in the locations, the abstract models can be as well compositionally linked, allowing thereby the abstraction of complex analog circuits.
Depending on the specified modeling settings, including for example the number of locations of the HA and the description of the system behavior, the accuracy, speedup, and various additional properties of the HA can be influenced. This is examined in detail in this dissertation. The underlying abstraction process is first covered in detail. Several extensions are then handled including the modeling of the HAs with parameter variations. The obtained models are then verified using various verification methodologies. The accuracy and speed-up of the abstraction methodology is finally evaluated on several transistor level circuits ranging from simple operational amplifiers up to a complex circuits.
