Die Bedeutung der Apoptose und die zugrundeliegenden Mechanismen in verschiedenen pathophysiologischen Zuständen des Herzens sind noch weitgehend ungeklärt und es bleibt zu zeigen, daß die Apoptose-Signaltransduktion ähnlich reguliert wird, wie aus in vitro-Versuchen bekannt ist. Deshalb wurde die Apoptose in verschiedenen Tiermodellen kardialer Erkrankungen untersucht werden, um Hinweise auf die zugrundeliegende Signal-transduktion, durch Analyse der Proteine Bcl-2 und Bax, der finalen Exekutor-Caspase Caspase-3 oder p53 zu bekommen. Apoptose in der durch Hyperlipidämie induzierten Atherosklerose: In Aorten von 'Froxfield Heritable Hypercholesterolemic'-Kaninchen (genetische Hyperlipidämie) korrelierte die Apoptose von vaskulären glatten Muskelzellen und Makrophagen in fortgeschrittenen fibrösen Plaques mit einem 18-fachen Anstieg des proapoptotischen Bax. In Aorten Cholesterin gefütterter 'New Zealand White'-Kaninchen (0,25% Cholest., 12 Wochen) konnte eine erhöhte Baxexpression in Endothelzellen nachgewiesen werden, ohne daß morphologische Veränderungen zu beobachten waren. Die Apoptose in akut abgestoßenen allogenen Herztransplantaten (Rattenmodell) war von einer erhöhten Bax-Expression und einer totalen, posttranslationalen Degradation des antiapoptotischen Bcl-2 in ein spezifisches Degradationsprodukt durch eine Serinprotease gekennzeichnet. Die Rolle des wichtigen kardiovaskulären Mediators Stickstoffmonoxid (NO) auf die Apoptose wird kontrovers diskutiert. Da in der Zellkultur protektive Effekte von NO gezeigt werden konnten, wurde deren physiologische Relevanz in der durch Ischämie/Reperfusion induzierten Apoptose ex vivo im Langendorff-Rattenherzen untersucht. Es konnte gezeigt werden, daß Hemmung der endogenen NO-Synthese mit L-NG-Monomethyl-L-Arginin (LNMMA, 1mM) die Apoptose potenzierte und mit einer Aktivierung der Caspase-3 korrelierte. Bcl-2 und Bax wurden nicht reguliert. Untersuchung der Regulation der Proteinexpression der eNOS (endotheliale NO-Synthase) durch den proinflammatorischen/ proatherogenen Tumor-Nekrose-Faktor-[Alpha] (TNF[Alpha]) in der Endothelzellkultur (HUVEC) gaben Hinweise auf einen, die eNOS schützenden, Interaktionspartner. Zusammenfassend konnte in allen untersuchten Modellen für Herz(-Kreislauf)-Krankheiten Apoptose nachgewiesen werden, die jeweils spezifische Charakteristika zeigt, deren genauere Aufklärung interessante Ziele zukünftiger präventiver und therapeutischer Maßnahmen verspricht. Die Befunde weisen zudem auf antiapoptotische Effekte von NO - insbesondere durch die endotheliale NO-Synthaseaktivität - hin, deren genauere Charakterisierung dazu beitragen könnte, pathophysiologische Zustände der kardiovaskulären Biologie zu erklären.
The identification of specific genetic (presenilin-1 [PS1] and amyloid precursor protein [APP] mutations) and environmental factors responsible for Alzheimer's disease (AD) has revealed evidence for a shared pathway of neuronal death. Moreover, AD-specific cell defects may be observed in many other nonneuronal cells (e.g., lymphocytes). Thus, lymphocytes may serve as a cellular system in which to study risk factors of sporadic, as well as genetic AD in vivo. The aim of our present study was to clarify whether lymphocytes bearing genetic or sporadic risk factors of AD share an increased susceptibility to cell death. Additionally we examined whether a cell typespecific vulnerability pattern was present and how normal aging, the main risk factor of sporadic AD, contributes to changes in susceptibility to cell death. Here, we report that lymphocytes affected by sporadic or genetic APP and PS1 AD risk factors share an increased vulnerability to cell death and exhibit a similar cell type-specific pattern, given that enhanced vulnerability was most strongly developed in the CD4+ T-cell subtype. In this paradigm, sporadic risk factors revealed the highest impact on cell type-specific sensitivity of CD4+ T cells to apoptosis. In contrast, normal aging results in an increased susceptibility to apoptosis of both, CD4+ and CD8+ T cells.
In eukaryotes, double-stranded (ds) RNA induces sequence-specific inhibition of gene expression referred to as RNA interference (RNAi). We exploited RNAi to define the role of HER2/neu in the neoplastic proliferation of human breast cancer cells. We transfected SK-BR-3, BT-474, MCF-7, and MDA-MB-468 breast cancer cells with short interfering RNA (siRNA) targeted against human HER2/neu and analyzed the specific inhibition of HER2/neu expression by Northern and Western blots. Transfection with HER2/neu-specific siRNA resulted in a sequence-specific decrease in HER2/neu mRNA and protein levels. Moreover, transfection with HER2/neu siRNA caused cell cycle arrest at G0/G1 in the breast cancer cell lines SKBR-3 and BT-474, consistent with a powerful RNA silencing effect. siRNA treatment resulted in an antiproliferative and apoptotic response in cells overexpressing HER2/neu, but had no influence in cells with almost no expression of HER2/neu proteins like MDA-MB-468 cells. These data indicate that HER2/neu function is essential for the proliferation of HER2/neuoverexpressing breast cancer cells. Our observations suggest that siRNA targeted against human HER2/neu may be valuable tools as anti proliferative agents that display activity against neoplastic cells at very low doses.
Atherosklerotische Stenosen der Karotiden sind eine häufige Erkrankung mit variablem Verlauf und stellen durch ihre potentielle Emboligenität einen wichtigen Risikofaktor für zerebrale Ischämien dar. Klinische und paraklinische Parameter helfen, das individuelle Schlaganfallrisiko bei Patienten mit hochgradigen ACI-Stenosen einzuschätzen, das unmittelbar nach einem thrombembolischen Ereignis besonders hoch ist. Als histomorphologisches Korrelat dieser "Vulnerabilität" wird die Ruptur der fibrotischen Deckplatte der Plaque propagiert, die häufiger bei symptomatischen Patienten nachzuweisen ist. Sie korreliert mit der Infiltration der Gefäßwand durch aktivierte Leukozyten, die über molekulare und zelluläre Interaktionen die Zell- und Bindegewebskomposition der Plaque verändern können. Die strukturelle Integrität atherosklerotischer Läsionen beruht auf der extrazellulären Vernetzung von kollagenem Bindegewebe, das überwiegend von phänotypisch veränderten glatten Gefäßmuskelzellen produziert wird. Eine Hypothese besagt, dass die im Rahmen der Inflammation stattfindende Zunahme proapoptotischer Mediatoren über eine Ausdünnung der zellulären und bindegewebigen Strukturen zu einem Verlust an mechanischer Stabilität führt und somit eine symptomatische Ruptur begünstigt. Da der Nachweis einer Ruptur mit Exponierung des thrombogenen nekrotischen Kerns allerdings nur in einem Teil der symptomatischen Plaques und umgekehrt auch in einem Teil der asymptomatischen nachgewiesen werden kann, ist aber bislang unklar, ob o.g. Abläufe in der humanen Karotis-Atherosklerose tatsächlich mit einer klinischen Relevanz einhergehen. In der vorliegenden Arbeit wurde daher das Auftreten der Apoptose von glatten Gefäßmuskelzellen (mittels DNA in situ end labeling Technik, TUNEL-Färbung) in 38 prospektiv gesammelten Endarterektomie-Präparaten hochgradiger Karotisstenosen quantitativ erfasst und statistisch in Beziehung gesetzt zu Parametern der Plaque-Instabilität, klinisch definiert durch kürzliche (< 60 Tage zurückliegende) ischämische Ereignisse (n=19) und histopathologisch definiert über den Nachweis einer Plaque-Ruptur (n=14). Außerdem wurde eine morphometrische Analyse der einzelnen Plaque-Komponenten durchgeführt und deren Ergebnisse zu den zellulären und klinischen Parametern in Beziehung gesetzt. Die Morphometrie ergab keine signifikanten Unterschiede zwischen symptomatischen vs. asymptomatischen und rupturierten vs. unrupturierten Plaques was die Größe der fibrotischen Deckplatte, die durchschnittliche Dicke (Kern-Lumen-Distanz) und die dünnsten bzw. dicksten Stellen der Deckplatte anbelangt. Anzahl und Konzentration apoptotischer glatter Muskelzellen war deutlich (p<0,001) erhöht in symptomatischen, klinisch instabilen, Karotisplaques. Allerdings waren die Apoptose-Raten in Präparaten, die eine Plaque-Ruptur aufwiesen, nicht signifikant erhöht. Darüber hinaus fand sich kein Hinweis darauf, dass erhöhte Apoptose-Raten zu einem quantifizierbaren Verlust glatter Gefäßmuskelzellen in der fibrotischen Deckplatte führen. Auf dem Boden dieser Ergebnisse kann gefolgert werden, dass erhöhten Apoptose-Raten glatter Gefäßmuskelzellen in der humanen Atherosklerose offenbar eine tragende Bedeutung bei der Entwicklung thrombembolischer Ereignisse zukommt. Allerdings wird die Annahme, dass erhöhte Apoptose-Raten über einen Verlust an glatten Gefäßmuskelzellen Einfluss auf die morphometrischen Eigenschaften der fibrotischen Deckplatte atherosklerotischer Karotis-Läsionen nehmen und zu deren Ausdünnung führen durch die vorliegende Untersuchung nicht gestützt. Vielmehr scheint es plausibel, dass die Apoptose glatter Muskelzellen im Rahmen inflammatorischer Prozesse Einfluss auf die Komposition der Karotisplaque nimmt und so über eine Desintegration der zellulären und bindegewebigen Bestandteile zu reduzierter mechanischer Widerstandskraft und Rupturneigung führt.
The thesis entitled „Investigations on the significance of nucleo-cytoplasmic transport for the biological function of cellular proteins" aimed to unreveal molecular mechanisms in order to improve our understanding of the impact of nucleo-cytoplasmic transport on cellular functions. Within the scope of this work, it could be shown that regulated nucleo-cytoplasmic transport of a subfamily of homeobox transcription factors controlled their intra- and intercellular transport, and thereby influencing also their transcriptional activity. This study describes a novel regulatory mechanism, which could in general play an important role for the ordered differentiation of complex organisms. Besides cis-active transport Signals, also post-translational modifications can influence the localization and biological activity of proteins in trans. In addition to the known impact of phosphorylation on the transport and activity of STAT1, experimental evidence was provided demonstrating that acetylation affected the interaction of STAT1 with NF-kB p65, and subsequently modulated the expression of apoptosis-inducing NF-kB target genes. The impact of nucleo-cytoplasmic transport on the regulation of apoptosis was underlined by showing that the evolutionary conservation of a NES within the anti-apoptotic protein survivin plays an essential role for its dual function in the inhibition of apoptosis and ordered cell division. Since survivin is considered a bona fide cancer therapy target, these results strongly encourage future work to identify molecular decoys that specifically inhibit the nuclear export of survivin as novel therapeutics. In order to further dissect the regulation of nuclear transport and to efficiently identify transport inhibitors, cell-based assays are urgently required. Therefore, the cellular assay Systems developed in this work may not only serve to identify synthetic nuclear export and Import inhibitors but may also be applied in systematic RNAi-screening approaches to identify novel components of the transport machinery. In addition, the translocation based protease- and protein-interaction biosensors can be applied in various biological Systems, in particular to identify protein-protein interaction inhibitors of cancer relevant proteins. In summary, this work does not only underline the general significance of nucleo-cytoplasmic transport for cell biology, but also demonstrates its potential for the development of novel therapies against diseases like cancer and viral infections.
Bei allen Insekten mit holometamorphem Entwicklungsgang werden Größe und Gestalt im Verlauf der Ontogenese festgelegt. Die Larven unterscheiden sich oftmals extrem von den späteren Imagines. Dieser larvenspezifische Habitus ist von zahlreichen imaginifugalen Merkmalen geprägt, die während der Metamorphose beseitigt werden müssen. Diese Entwicklungsphase ist vergleichbar mit den Vorgängen während der Embryogenese, da nun in einem zweiten Schritt genetische, neuronale und hormonale Prozesse erneut aktiviert werden, die durch Proliferation, Zellwachstum und Zelltod (=Apoptose) den Abbau und Umbau sowie die Neogenese imaginipetaler Merkmale einleiten. Aufgrund des imagoähnlichen Phänotyps der Larvenstadien der Thysanoptera und der vermeintlichen „Rekonstruktion“ dieses adulten Status bezeichnete TAKAHASHI (1921) diese Form der Metamorphose als Remetabolie. Spontan initiierte Defekte während der Embryonalentwicklung zeigen allerdings darüber hinaus, dass eine Vorbereitung des Adultstatus bereits durch die Anlage imaginaler Stammzellcluster erfolgt (MORITZ 1997). Da Habitat und Ernährungsweise der Larvenstadien und Imagines keine Unterschiede aufweisen, sind typische, asymmetrische Mundgliedmaßen in beiden ontogenetischen Phasen nicht verwunderlich (MORITZ 1988, 1989). Interessant ist jedoch, warum zwischen zwei hoch spezialisierten Merkmalskomplexen ein Prozess der Destruktion liegt und wieso die Neogenese zu einem ontogenetisch vergleichbaren Resultat führt.
Identification and characterization of TNFalpha responsive genes in human breast cancer cells

One of the hallmarks of cancer is the escape of the transformed cells from apoptosis. Therefore, the identification of survival genes, allowing cancer cells to circumvent programmed cell death, could provide new diagnostic markers as well as targets for therapeutic intervention. A well known transcription factor regulating the balance between pro- and anti- apoptotic factors is NF-kappaB, which is strongly induced by tumor necrosis factor alpha (TNFalpha). When cells are stimulated by TNFalpha their response is biphasic with an initial NF-kappaB induction of survival genes which is overridden by the subsequent activation of initiator caspases triggering apoptosis. By combining gene trap mutagenesis with site specific recombination a strategy was developed, which enriches for genes induced by TNFalpha in the human breast cancer cell line MCF-7. The strategy relies on a one way gene expression switch based on Cre/loxP mediated recombination, which uncouples the expression of a marker gene from the trapped cellular promoter thereby enabling the recovery of genes that are only transiently induced by TNFalpha. The marker gene used in these experiments was a dominant negative variant of the TNFalpha-receptor associated protein FADD (dnFADD), which blocks the apoptotic branch of the TNFalpha induced signaling pathway. Initial experiments indicated that MCF-7 cells expressing high levels of dnFADD were insensitive to TNFalpha induced apoptosis and therefore suitable for the installment of a one way gene expression switch susceptible to Cre/loxP mediated recombination. A MCF-7 reporter clone harboring the recombinase dependent gene expression switch was infected with the gene trap retrovirus U3Cre, which inserts the Cre recombinase gene into a large collection of chromosomal sites. Insertion of Cre downstream of an active cellular promoter induces dnFADD expression from the gene expression switch enabling the cells to block TNFalpha triggered apoptosis. From a gene trap integration library containing approximately 2000000 unique proviral integrations, 69 unique TNFalpha inducible gene trap insertion sites were recovered in a two step selection procedure. Sequencing of the genomic regions adjacent to the insertion sites, which were obtained by inverse PCR (gene trap sequence tags, GTSTs), and data base analysis revealed that 42% of the GTSTs belonged to annotated genes, 13% to known cDNAs with open reading frames, 17% to Genscan predicted genes, 9% to ESTs, 9% to repetitive sequences and 10% to unannotated genomic sequence. Overall, 44% of the annotated genes recovered in this screen were directly or indirectly related to cancer, indicating that the gene trap strategy developed here is suitable for the identification of cancer relevant genes. Analysis of the expression patterns of the trapped and annotated genes in wild type cells revealed that 19 out of 24 genes were either up- or down- regulated by a factor of at least 1.45 by TNFalpha. A large fraction of the gene trap insertions were located upstream, in introns or in opposite orientation to annotated transcripts, indicating that the strategy efficiently recovers non-coding RNAs (ncRNAs). While the biological significance of these transcripts still needs to be elucidated, they fall into two main categories. The first category includes gene trap insertions upstream of genes, which could either represent regulatory RNAs interacting with promoter elements or transcripts driven by bidirectional promoters. The second includes inverse orientation gene trap insertions in introns of annotated genes suggesting the presence of natural antisense transcripts (NATs). Interestingly, more than 50% of all antisense integrations are located downstream of transcription start sites predicted by different algorithms supporting the existence of RNAs transcribed from the corresponding genomic regions. Intronic integrations on the coding strand could be derived from cryptic splicing, alternative promoter usage or additional, so far uncharacterized transcripts. Preliminary functional analysis of two genes recovered in this screen encoding the transcription factor ZFP67 and the FLJ14451 protein revealed that FLJ14451 but not ZFP67 inhibited anchorage independent growth in soft agar, suggesting that FLJ14451 might have some tumor suppressor functions. In summary, besides identifying a putative tumor suppressor protein, the present experiments have shown that gene trapping is useful in identifying non-coding transcripts in living cells and may turn out to be the method of choice in characterizing these transcripts whose functions are still largely unknown.
Der ubiquitäre Redoxregulator Thioredoxin-1 (Trx-1) hat wichtige Funktionen für den zellulären Redoxstatus, Zellwachstum und Apoptose. Reaktive Sauerstoffspezies (ROS) sind beteiligt an der Pathogenese kardiovaskulärer Erkrankungen wie der Atherosklerose und werden zunehmend in ihrer Rolle als intra- und extrazelluläre Signalmoleküle charakterisiert. Ein Ungleichgewicht zwischen der Entstehung von ROS und ihrem Abbau durch antioxidative Systeme führt zu oxidativem Stress, zur Oxidation von Proteinen und letztlich zum Zelltod. Daher wurde in dieser Doktorarbeit untersucht, wie reaktive Sauerstoffspezies Trx-1 in Endothelzellen regulieren, welchen Einfluss dies für die Endothelzellapoptose hat und welche Bedeutung Antioxidantien, Stickstoffmonoxid (NO) und Schubspannung haben. In dieser Arbeit wurde gezeigt, dass H2O2 konzentrationsabhängig die Expression von Trx-1 beeinflusst. Geringe Konzentrationen H2O2 wie 10 und 50 µM induzierten Trx-1-mRNA nach 3 Stunden. Auf Proteinebene fand sich dann nach 6 Stunden eine transiente Hochregulation von Trx-1. Diese geringen Konzentrationen von H2O2 wirkten antiapoptotisch. Dieser antiapoptotische Effekt war von der Trx-1 Proteinexpression abhängig. Im Gegensatz dazu kam es bei hohen Konzentrationen H2O2 zu einer Degradierung von Trx-1. Durch das Antioxidans NAC und NO konnte der Abbau von Trx-1 unter höheren H2O2-Konzentrationen verhindert werden. Untersuchungen zum Mechanismus des Degradierungsprozesses ergaben, dass Trx-1 durch die Aspartatprotease Cathepsin D abgebaut wird. Der protektive Effekt von NO auf die Trx-1 Expression konnte auch im Gewebe eNOS-defizienter Mäuse gezeigt werden, da bereits eNOS-defiziente Mäuse in den Nieren weniger Trx-1 Protein aufwiesen im Vergleich zu Wildtyp-Kontrollmäusen. Bei der Entstehung endothelialer Läsionen und der Stabilität atheromatöser Plaques spielt die Endothelzellapoptose vermutlich eine wichtige Rolle. Trx-1 schützt Endothelzellen vor Apoptose, wird jedoch unter oxidativem Stress abgebaut. Faktoren, die Trx-1 unter oxidativem Stress stabilisieren wie NAC und NO, kommt daher eine besondere Bedeutung für die Endothelzellhomöostase zu.
Betulinic acid is a natural product with a range of biological effects, for example potent antitumor activity. This anticancer property is linked to its ability to induce apoptotic cell death in cancer cells by triggering the mitochondrial pathway of apoptosis. In contrast to the cytotoxicity of betulinic acid against a variety of cancer types, normal cells and tissue are relatively resistant to betulinic acid, pointing to a therapeutic window. Compounds that exert a direct action on mitochondria present promising experimental cancer therapeutics, since they may trigger cell death under circumstances in which standard chemotherapeutics fail. Thus, mitochondrion-targeted agents such as betulinic acid hold great promise as a novel therapeutic strategy in the treatment of human cancers.

EFP1 is an ER stress-induced glycoprotein which interacts with the pro-apoptotic protein Par-4

We have isolated the rat ortholog of EFP1 (EF-hand binding protein 1) as a novel interaction partner of the pro-apoptotic protein Par-4 (prostate apoptosis response-4). Rat EFP1 contains two thioredoxin domains, the COOH-terminal one harboring a CGFC motif, and has a similar protein domain structure as members of the protein disulfide isomerase (PDI) family. In REF52.2 and CHO cells, EFP1 colocalized with the endoplasmic reticulum (ER) marker PDI. Furthermore, EFP1 possesses catalytic activity as demonstrated by an insulin disulfide reduction assay. Western blot analysis revealed two EFP1 protein bands of approximately 136 and 155 kDa, representing different glycosylation states of the protein. Complex formation between EFP1 and Par-4 was confirmed in vitro and in vivo by co-immunoprecipitation, dot blot overlay and pull-down experiments. In CHO cells, coexpression of EFP1 and Par-4 resulted in enhanced Par-4-mediated apoptosis, which required the catalytic activity of EFP1. Interestingly, EFP1 was specifically upregulated in NIH3T3 cells after induction of ER stress by thapsigargin, tunicamycin, and brefeldin A, but not by agents that induce oxidative stress or ER-independent apoptosis. Furthermore, we could show that the induction of apoptosis by Ca2+ stress-inducing agents was significantly decreased after siRNA oligonucleotide-mediated knockdown of Par-4. Our data suggest that EFP1 might represent a cell-protective enzyme that could play an important role in the decision between survival and initiation of Par-4-mediated apoptosis.
