In-depth analyses of cancer cell proteomes are needed to elucidate oncogenic pathomechanisms, as well as to identify potential drug targets and diagnostic biomarkers. However, methods for quantitative proteomic characterization of patient-derived tumors and in particular their cellular subpopulations are largely lacking. Here we describe an experimental set-up that allows quantitative analysis of proteomes of cancer cell subpopulations derived from either liquid or solid tumors. This is achieved by combining cellular enrichment strategies with quantitative Super-SILAC-based mass spectrometry followed by bioinformatic data analysis. To enrich specific cellular subsets, liquid tumors are first immunophenotyped by flow cytometry followed by FACS-sorting; for solid tumors, laser-capture microdissection is used to purify specific cellular subpopulations. In a second step, proteins are extracted from the purified cells and subsequently combined with a tumor-specific, SILAC-labeled spike-in standard that enables protein quantification. The resulting protein mixture is subjected to either gel electrophoresis or Filter Aided Sample Preparation (FASP) followed by tryptic digestion. Finally, tryptic peptides are analyzed using a hybrid quadrupole-orbitrap mass spectrometer, and the data obtained are processed with bioinformatic software suites including MaxQuant. By means of the workflow presented here, up to 8,000 proteins can be identified and quantified in patient-derived samples, and the resulting protein expression profiles can be compared among patients to identify diagnostic proteomic signatures or potential drug targets.
This paper presents some results from a wider study which aims to define the features of German medical textbooks from a diachronic perspective. The medical textbook is presented here as a genre-class (Textsortenklasse) subsuming all texts written for didactic purposes in the field of medicine, i.e. texts aiming to present the theoretical and practical background required in this particular specialist professional field. Since the lingua franca of academic communication was Latin until the 18th century, the corpus used for this study mainly comprises surgery textbooks. The paper focuses solely on structural aspects of textbooks, seeking to show how these aspects functionally contribute to the realization of the general textual function and thereby constitute a historically established conventionalized scheme underlying genres. However, a thorough and effective description of a genre-class requires a much more extensive approach including more interrelated levels of analysis.
Der folgende Beitrag präsentiert die Problematik der Eponyme in der Fachsprache und beschäftigt sich mit deren Vorkommen in ausgewählten naturwissenschaftlichen Bereichen. Naturwissenschaften stellen eine reiche Quelle eponymischer Bezeichnungen dar. Für die Analyse wurden drei Disziplinen ausgewählt: Medizin, Chemie und Physik. Alle drei naturwissenschaftlichen Disziplinen wurden in deutscher Sprache analysiert und konfrontiert. Die Analyse konzentriert sich auf die Besonderheiten der Eponyme, d.h. Struktur, Gebrauch, Vorkommen, Kategorisierung und Evaluierung im Rahmen der ausgewählten Disziplinen. Mit der Problematik der Eponyme haben sich mehrere Autoren beschäftigt, z.B. Morton S. Freeman und Dorothy Auchter. In der Slowakei haben sich den Eponymen Mária Bujalková, Božena Džuganová, Gabriela Poláčková und Ivan Masár gewidmet. I. Background: Imbalances in female career promotion are a key factor of gender disparities at the workplace. They may lead to stress and stress-related diseases including burnout, depression or cardiovascular diseases. Since this problem cannot be generalized and varies between different fields, new approaches are needed to assess and describe the magnitude of the problem in single fields of work.
Methods: To construct a new index, operating figures of female and male medical students were collected for Germany in a period over 15 years and their progression throughout their studies towards specialization and academic chair positions. By the use of different female to male ratios (f:m), we constructed an index that describes the extend by which women can ascent in their academic career by using the field of academic medicine as an example.
Results: A medical student f:m ratio of 1.54 (52,366 female vs. 34,010 male) was found for Germany in 2013. In 1998, this f:m ratio was 0.999. In the same year (2013), the OB/GYN hospital specialists’ f:m ratio was 1.566 (3347 female vs. 2137 male physicians) and 0.577 (516 female vs 894 male physicians) for ENT hospital specialists, respectively. The f:m ratios concerning chairs of OB/GYN and ENT were 0.105 and 0.1, respectively. Then an index was generated that incorporated these operating figures with the student f:m ratio as denominator and the chair f:m ratio as numerator while the hospital specialist f:m ratio served as a corrector in the numerator in order to adjust to the attraction of a given field to female physicians. As a result, the index was 0.044 for OB/GYN and 0.113 for ENT instead of ideally ~1 in a completely gender harmonized situation.
Conclusion: In summary, a new index to describe female career advancement was established for academic medicine. By the use of this index, different academic and medical fields can now be compared to each other and future benchmarks could be proposed. Also, country differences may be examined using the proposed index and the success of specific funding programs.
