Mit der Darstellbarkeit des Denkprozesses an sich beschäftigt sich der französische Lyriker und Philosoph Paul Valéry. Denn durch die Materialisierung dieser Prozesse, d. h. durch den Versuch der exakten Dokumentation, werden sie analysierbar und ihr Potential allererst begreifbar. Technische Genauigkeit wird hierbei zum methodischen Ideal. Gemäß dem Beitrag von Lucas Knierzinger sucht Valéry explizit nach einer präzisen, systematisierenden Notationsform, die mit dem variierenden Schreibfluss seiner "Cahiers" in Spannung tritt und den in der Handschrift sedimentierten Denkprozess in Erscheinung treten lässt. Mit Eggerts laboratorischen Versuchsanlagen vergleichbar, bezeichnet Valéry die Interaktion von Lesen und Sehen bei der anschließenden kritischen Betrachtung dieser faksimilierten Manuskriptseiten auch als analytische 'machine à lire'.
Specialized transporter proteins facilitate controlled uptake and extrusion of molecules across biological membranes that would otherwise be impermeable to them. The superfamily of solute carriers (SLC) comprises the second largest group of membrane proteins in humans, acting on a variety of small polar and non-polar molecules and ions. Because of their central role in metabolism, malfunctioning of these proteins often is pathogenic. The interest in SLC transporters as drug targets – as well as for drug delivery – has therefore increased in the past years. For many SLC subfamilies, however, structural and functional information remains scarce to date.
The here presented data provides important insights into different aspects of the transport mechanism of the SLC23 and SLC26 protein families. Importantly, we show that SLC23 nucleobase transporters, in contrast to what was been previously reported, work as uniporters rather than as proton-coupled symporters. In order to do so, we developed the first and only in vitro transport assay for the SLC23 family, which enables investigation of protein function in a defined environment. Moreover, we provide a hypothesis on the role of the extremely conserved negative charged substrate binding site residue found not only in the SLC23, but also SLC4 and SLC26 families. Based on a detailed analysis of binding and transport we conclude that this conserved negative charged has a relevance for protein stability rather than for substrate binding, which explains its conservation for all three protein families that otherwise differ in their substrate specificities and modes of transport. Lastly, we investigated the relevance of oligomerization for the SLC23 and SLC26 families, highlighting the importance of the STAS domain for forming active dimers in the SLC26 anion transporter family.
The Tanaidacea are ubiquitous and amongst the most abundant taxa in the deep sea. However, their diversity in submarine canyons remains largely unknown. Here, two new species and a new genus of Paratanaoidea are described. Paranarthrura cousteaui sp. nov. is distinguished by the combination of the following characters: post-cheliped sclerites not fused, presence of one seta in the maxilliped endite, one long midventral seta in cheliped, one penicillate seta in the basis of pereopods 4–6, uropod endopod bi-articulated and uropod exopod shorter than endopod article 1. This species was found at the upper reaches of three Portuguese canyons, Cascais, Setúbal and Nazaré Canyons, and the adjacent open slope, between 897 and 1001 m water depths. Tirana vallis gen. et sp. nov. presents a combination of the characters that define the other two genera of Paranarthrurellidae, Paranarthrurella and Armatognathia, but also unique characters within the family: the antenna, cheliped and uropod are more elongate than the rest of the species; the pereopods 4–6 carpus spines reach at least half of the length of the propodus and the propodus of pereopods 4–6 have ramified subdistal spines. This species was found at the middle reaches of Setúbal Canyon (3214–3219 m water depth).
The pathophysiological role of neural autoantibodies in acute psychotic disorders is receiving increased attention. However, there is still an ongoing debate, whether predominantly psychotic manifestations of autoimmune encephalitides exist that may remain undetected and, thus, untreated. Furthermore, it is discussed if such conditions can be diagnosed based on serum antibody results or if a reliable diagnosis requires additional cerebrospinal fluids (CSF) results. In this study, we screened pairs of serum and CSF samples from antipsychotic-naïve individuals with first-episode schizophrenic psychosis (FEP, n = 103), clinical high risk for psychosis (CHR, n = 47), and healthy volunteers (HV, n = 40) for eight different antibodies against various antigens that have been shown to be associated with autoimmune encephalitides: N-methyl-D-aspartate receptor (NMDAR, NR1 subunits only), glutamic acid decarboxylase (GAD65), leucine-rich glioma inactivated protein 1 (LGI1), contactin-associated protein-like 2 protein (CASPR2), α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor (AMPAR) subunit 1, AMPAR subunit 2, γ-aminobutyric acid-B receptors (GABABR), and glycine receptors. All patients were within the norm with regards to a careful neurological examination, a magnetic resonance imaging (MRI) of the brain, an electroencephalogram (EEG), and routine blood pathology. All CSF samples were autoantibody-negative. In three serum samples of individuals with FEP, we detected low-titer CASPR2 immunoglobulin (Ig) G antibodies (≤1:160, n = 2) and non-IgG antibodies against NMDAR (n = 1) (overall serum-autoantibody prevalence in FEP: 2.91%). However, the IgG titers were below the laboratory cut-off defined for positivity, and non-IgG antibodies are of no clinical relevance. This suggests that there were no cases of autoimmune encephalitis in our cohort. Our results highlight the importance and the high specificity of CSF analysis to reliably detect autoantibodies. They confirm the hypothesis that pure psychotic manifestations of antibody-associated autoimmune encephalitides without any additional neuropsychiatric findings are very rare. However, special attention must be paid to those presenting with atypical mental illnesses with additional neurological symptoms, evidence of clinically-significant cognitive involvement, profound sleep-wake perturbations, seizures, electroencephalographic, or magnetic resonance imaging pathologies to be able to identify cases with autoimmune-mediated psychiatric syndromes.
Ein durch einen chronischen Ethanolabusus vorgeschädigter Organismus erfährt nach Hämorrhagie/Reperfusions (H/R) Trauma eine vermehrte inflammatorische Antwort und höheres Ausmaß an hepatischer Schädigung. Dies ist mit häufigerem Auftreten von Komplikationen und verlängertem Intensivaufenthalt vergesellschaftet.
Andere sowie eigene Studien haben gezeigt, dass durch D-JNK-1, ein spezifisches zellgängiges, proteaseresistentes Peptid, welches die c-Jun-N-terminale Kinase hemmt, die Inflammationsreaktion und das Ausmaß der hepatischen Schädigung nach H/R vermindert werden kann.
Ziel dieser Arbeit war die Untersuchung, ob auch bei einem durch chronischen Ethanolabusus vorgeschädigten Organismus, die D-JNK-1 Gabe zu einer Abschwächung der Inflammation im Tiermodell führt. Des Weiteren sollte auch die Rolle des Transkriptionsfaktors NF-κB in der Inflammationskaskade unter D-JNK-1 Gabe bei der chronisch ethanolvorgeschädigter Leber genauer betrachtet werden.
Wir arbeiteten mit einem H/R Modell, sowie einem etablierten chronischen Ethanol Fütterungsregime mittels Lieber-DeCarli Diät. Die Versuchstiere erhielten nach der Durchführung von Hämorrhagie vor der Reperfusion eine D-JNK-1- oder NaCl-Gabe. Zwei Stunden nach Beendigung des Versuches wurden Leberenzyme, Lactatdehydrogenase und pro-inflammatorische Zytokine im Serum sowie die Konzentrationen der pro-inflammatorischen Mediatoren im Lebergewebe untersucht. Die Aktivität von NF-κB und c-Jun wurde durch Nachweis der relativen Proteinexpression mittels Westernblot quantifiziert. Zusätzlich wurde die NF-κB Aktivierung immunhistologisch durch die GFP Färbung untersucht. Der Leberschaden wurde histologisch mittels HE-Färbung nachgewiesen.
Durch eine 4-wöchige chronische ethanolhaltige Fütterung wird bei den Versuchstieren eine Steatosis hepatis, mit den entsprechenden strukturellen und funktionellen Veränderungen ausgelöst. Durch zusätzliche Durchführung der H/R-Prozedur wird eine relevante hepatische Schädigung sowie Inflammationsreaktion ausgelöst. NF-κB und c-Jun spielen in der Regulation der Inflammationsraktion nach H/R-Trauma eine wesentliche Rolle. Durch die chronische Ethanol-Fütterung kommt es ebenso wie durch die H/R-Prozedur zu einer Aktivierung von NF-κB und wenn auch in geringerem Ausmaß von c-Jun.
Die Intensität der durch H/R-Trauma ausgelösten Inflammationsreaktion und des Gewebeschadens in der durch chronischen ethanolgeschädigten Leber kann durch eine eimalige D-JNK-1 Gabe im Tiermodel nicht wesentlich beeinflusst werden. In der Kontrollgruppe kann durch die D-JNK-1 Applikation jedoch eine Abschwächung der posttraumatischen Inflammationsreaktion beobachtet werden.
Dies führt zu dem Schluss, dass nachdem im ersten Schritt die Grundlagen und Ursachen eines pathologischen Sachverhaltes untersucht wurden, im Anschluss an den zweiten Schritt, der weiterführenden Erforschung und Erarbeitung von therapeutischen Ansätzen, die erarbeiteten Ansätze in Hinsicht auf ihre Wirksamkeit bei Vorliegen weiterer Pathologien nochmals gesondert betrachtet werden müssen. Da das Zusammenspiel verschiedener pathologischer Zustände oftmals zur Zunahme der Komplexität führt und neue Sachverhalte aufzeigt. Jedoch bleibt es in weiterführenden Studien zu untersuchen, ob eine wiederholte Applikation des spezifischen c-Jun Inhibitors oder eine Erhöhung seiner Dosis sowie eine Modulation des Therapiezeitfensters die in vorherigen Studien beobachteten benefiziellen Effekte auch in diesem Modell herbeiführen können.
In der Arbeit wird das Certainty-Tool, eine Erweiterung für den Unity-basierten Teil des Stolperwege Projektes, vorgestellt. Dieses verfolgt die Idee des VAnnotatoR weiter und erlaubt die Visualisierung von informationeller Ungewissheit der im Stolperwege-Praktikum digital rekonstruierten Gebäude. Dabei inkorporiert das Tool das Konzept hinter BIM (Building Information Modelling), eine neuartige Methode der Planung in der AEC-Branche, welches ein Selbstbewusstsein von Informationen für Teile eines Gebäudes ermöglicht. Dabei werden im Certainty-Tool Stufen der informationellen Ungewissheit entwickelt und diese auf Teile des Gebäudes zugewiesen. Das Tool wird anhand einer digitalen Rekonstruktion des zerstörten Rothschild-Palais vorgestellt. Des Weiteren wurde eine Evaluation basierend auf der Usability Metric for User Experience durchgeführt und weiterführende Entwicklungen und Verbesserungen des Tools diskutiert.
