We investigate US households’ direct investment in stocks, bonds and liquid accounts and their foreign counterparts, in order to identify the different participation hurdles affecting asset investment domestically and overseas. To this end, we estimate a trivariate probit model with three further selection equations that allows correlations among unobservables of all possible asset choices. Our results point to the existence of a second hurdle that stock owners need to overcome in order to invest in foreign stocks. Among stockholders, we show that economic resources, willingness to assume greater financial risks, shopping around for the best investment opportunities all increase the probability to invest in foreign stocks. Furthermore, we find that households who seek financial advice from relatives, friends and work contacts are less likely to invest in foreign stocks. This result corroborates the conjecture by Hong et al. (2004) that social interactions should discourage investment in foreign stocks, given their limited popularity. On the other hand, we find little evidence for additional pecuniary or informational costs associated with investment in foreign bonds and liquid accounts. Finally, we show that ignoring correlations of unobservables across different asset choices can lead to very misleading results.
We investigate the effects of both trust and sociability for stock market participation, the role of which has been examined separately by existing finance literature. We use internationally comparable household data from the Survey of Health, Ageing and Retirement in Europe supplemented with regional information on generalized trust from the World Value Survey and on specific trust to financial institutions from Eurobarometer. We show that trust and sociability have distinct and sizeable positive effects on stock market participation and that sociability is likely to partly balance the discouragement effect on stockholding induced by low generalized trust in the region of residence. We also show that specific trust in advice given by financial institutions represents a prominent factor for stock investing, compared to other tangible features of the banking environment. Probing further into various groups of households, we find that sociability can induce stockholding among the less well off in Sweden, Denmark, and Switzerland where stock market participation is widespread. On the other hand, the effect of generalized trust is strong in countries with limited participation and low average trust like Austria, Spain, and Italy, offering an explanation for the remarkably low participation rates of the wealthy living therein.
In the New-Keynesian model, optimal interest rate policy under uncertainty is formulated without reference to monetary aggregates as long as certain standard assumptions on the distributions of unobservables are satisfied. The model has been criticized for failing to explain common trends in money growth and inflation, and that therefore money should be used as a cross-check in policy formulation (see Lucas (2007)). We show that the New-Keynesian model can explain such trends if one allows for the possibility of persistent central bank misperceptions. Such misperceptions motivate the search for policies that include additional robustness checks. In earlier work, we proposed an interest rate rule that is near-optimal in normal times but includes a cross-check with monetary information. In case of unusual monetary trends, interest rates are adjusted. In this paper, we show in detail how to derive the appropriate magnitude of the interest rate adjustment following a significant cross-check with monetary information, when the New-Keynesian model is the central bank’s preferred model. The cross-check is shown to be effective in offsetting persistent deviations of inflation due to central bank misperceptions. We reconsider the issue of price discovery in spot and futures markets. We use a threshold error correction model to allow for arbitrage operations to have an impact on the return dynamics. We estimate the model using quote midpoints, and we modify the model to account for time-varying transaction costs. We find that the futures market leads in the process of price discovery. The lead of the futures market is more pronounced in the presence of arbitrage signals. Thus, when the deviation between the spot and the futures market is large, the spot market tends to adjust to the futures market.
The budget constraint requires that, eventually, consumption must adjust fully to any permanent shock to income. Intuition suggests that, knowing this, optimizing agents will fully adjust their spending immediately upon experiencing a permanent shock. However, this paper shows that if consumers are impatient and are subject to transitory as well as permanent shocks, the optimal marginal propensity to consume out of permanent shocks (the MPCP) is strictly less than 1, because buffer stock savers have a target wealth-to-permanent-income ratio; a positive shock to permanent income moves the ratio below its target, temporarily boosting saving. We use a novel disaggregate sectoral euro area dataset with a regional breakdown that allows explicit estimation of the sectoral component of price changes (rather than interpreting the idiosyncratic component as sectoral as done in other papers). Employing a new method to extract factors from over-lapping data blocks, we find for our euro area data set that the sectoral component explains much less of the variation in sectoral regional inflation rates and exhibits much less volatility than previous findings for the US indicate. Country- and region-specific factors play an important role in addition to the sector-specific factors. We conclude that sectoral price changes have a “geographical” dimension, as yet unexplored in the literature, that might lead to new insights regarding the properties of sectoral price changes.
This paper reviews the rationale for quantitative easing when central bank policy rates reach near zero levels in light of recent announcements regarding direct asset purchases by the Bank of England, the Bank of Japan, the U.S. Federal Reserve and the European Central Bank. Empirical evidence from the previous period of quantitative easing in Japan between 2001 and 2006 is presented. During this earlier period the Bank of Japan was able to expand the monetary base very quickly and significantly. Quantitative easing translated into a greater and more lasting expansion of M1 relative to nominal GDP. Deflation subsided by 2005. As soon as inflation appeared to stabilize near a rate of zero, the Bank of Japan rapidly reduced the monetary base as a share of nominal income as it had announced in 2001. The Bank was able to exit from extensive quantitative easing within less than a year. Some implications for the current situation in Europe and the United States are discussed.
Seit 2003 wird die Krankenhausvergütung in Deutschland schrittweise auf ein neues, leistungsorientiertes System auf Basis von Fallpauschalen (diagnosis related groups – DRG) umgestellt. Die Über gangsphase, in der gesonderte gesetzliche Regelungen gegolten hatten, sollte Ende 2008 auslaufen, weshalb der gesetzlichen Rahmen für die Nachkonvergenzzeit neu geregelt werden musste. Neben diesen technischen Regelungen und einem einheitlichen Bundesbasisfallwert standen auch die Reform der Investitionsfinanzierung, der weitere Ausbau des Wettbewerbes und die schlechte Finanzsituation der Krankenhäuser auf der Agenda des Krankenhausfinanzierungsreformgesetzes (KHRG). Aufgrund der heterogenen Interessenslagen von Bund und Ländern, gestaltete sich auch bei diesem Reformgesetz die Kompromisssuche schwierig. Herausgekommen ist ein Gesetz, dass die Reformpolitik der letzten Jahre weiterführt, die Aufgabe der zukünftigen Sicherstellung der bedarfsgerechten Versorgung aber nicht löst und die problematischen Auswirkungen des DRG-Systems negiert. Das Diskussionspapier zeichnet die wichtigsten Schritte des Gesetzgebungsprozesses nach, präsentiert und diskutiert die wichtigsten Inhalte des Gesetzes und ordnet das KHRG in den Reformkontext der letzten Jahre ein.
Die Europäische Aktiengesellschaft (SE) gestattet es bekanntlich, die Mitbestimmung der Arbeitnehmer im Wege einer Vereinbarungslösung zu regeln und die Größe des Aufsichtsrats zu reduzieren. Zahlreiche deutsche Unternehmen haben aus diesem Grund die Rechtsform der AG verlassen und diejenige der SE gewählt. Um dieser „Flucht aus der AG” zu begegnen, hat der aus sieben unabhängigen Hochschullehrern bestehende Arbeitskreis „Unternehmerische Mitbestimmung” einen Gesetzesvorschlag entwickelt, der eine Verhandlungslösung auch für die AG und die GmbH zulässt und eine arbeitsfähige Aufsichtsratsgröße vorsieht. Der im Folgenden abgedruckte Vorschlag ist in ZIP/Zeitschrift für Wirtschaftsrecht 2009, S. 885 ff veröffentlicht.

Der Buddha setzte vor seinem Tod keinen Nachfolger ein. Vielmehr forderte er seine Jünger auf, die Lehre sollte fortan ihr Meister sein. Dies hatte zur Folge, dass sich seine Nachfolger über die Worte des Buddha uneins wurden und dass sich verschiedene Schulen bildeten. Tatsächlich bindend sind für die gläubigen buddhistischen Laien fünf Verpflichtungen: nicht zu töten, nicht zu stehlen, nicht zu lügen, nicht die Ehe zu brechen und keine berauschenden Getränke zu genießen.
The documentation of life on Earth, that is, the inventorization of nature and the naming and classification of organisms found therein, is a major task for biologists today and a fundamental precondition for nature conservation efforts. This study aimed at contributing to the inventory of amphibians and reptiles in selected, previously understudied ecoregions of Bolivia. I strove to document diversity patterns and seek possible ecological and historical reasons for these patterns. Special attention was paid to the Chiquitano Region situated in the eastern lowlands of Bolivia in a climatic transition zone between the humid evergreen Amazon Forests and the deciduous thorn-scrub vegetation of the Gran Chaco. In congruence with its location in the transition zone, the Chiquitano Region displays a mosaic of habitats: The vegetation is dominated by the endemic Chiquitano Dry Forest, which is probably the largest extant patch of Seasonal Dry Tropical Forest, with enclaves of savanna, the western outliers of the Cerrado biome of central Brazil. Taxonomic revisions: The taxonomic data in this study are used as a tool to measure biodiversity, to assess biogeographic relationships, and to evaluate conservation needs. Since all is predicated on the taxonomic decisions made, an adequate taxonomy is essential, and taxonomy can be regarded as the foundation of this study. The methodology encompassed a variety of herpetological field techniques, such as different survey methods, preparation and documentation of voucher specimens, recording of frog calls, and herpetological laboratory techniques, such as morphology, molecular procedures with mtDNA, phylogenetic analyses, and bioacoustic analysis and descriptions of frog calls. A total of 1251 specimens belonging to 200 species were obtained during this study, including 87 amphibian and 123 reptile species. This constitutes about 36% of the herpetofauna currently known for Bolivia, about 34% of the amphibians currently known for Bolivia and about 40% of the reptiles, respectively. In the course of this study, a new species of frog was described from the study site Caparu in the eastern lowlands of Bolivia; this species, Hydrolaetare caparu Jansen, Gonzales & G. Köhler 2007, differs from the other two congeners in external morphology (e.g., lateral fringes and relative length of fingers, size of palmar tubercle, webbing of toes, and colouration) and advertisement call. Two new colubrid snake species were also described from the study site San Sebastián. Thus far, both are known only from the Chiquitano Region, Provincia Ñuflo de Chávez. Phalotris sansebastiani Jansen & G. Köhler 2008 differs from all the other species in the genus in having a triangular projection of the red snout colouration reaching onto the parietals. Xenopholis werdingorum Jansen, Gonzales & G. Köhler 2009 can be identified as a member of the genus Xenopholis by its vertebral morphology. It differs from the other two species of Xenopholis in having a unique uniform dorsal colour pattern, and from X. scalaris in having two prefrontals and a narrow septum within the neural spine and perpendicular to its long axis as evident in the x-ray images. A review of a small collection of pitvipers from different lowland localities and from the Inter-Andean dry valleys of the region of Pampagrande revealed one new species of Bothrops and one of Bothrocophias (both to be formally described elsewhere). The two pitviper species differ morphologically and genetically from their congeners. The results of a brief review of a small collection of frogs of the genus Scinax (Anura: Hylidae) from different localities in the lowlands, together with analyses of their bioacoustics, suggest an unknown cryptic diversity in Bolivian species of Scinax cf. fuscomarginatus and allies. However, further studies are necessary to clarify the taxonomic status of these populations. In addition, this study provides new data on the morphology (e.g., pholidosis) of snakes, many of them previously known only from few museum specimens. Keys to the Bolivian lizard species of Cercosaura and the Bolivian snake species of Chironius, Clelia, Liophis, Lystrophis, Phalotris, and Xenodon are presented here for the first time. New information on distribution includes many range extensions of amphibian and reptile species, such as five new country records (one frog species, four snake species) and six new departmental records (two frog species, four snake species). Observations on ecology and natural history: Several observations on ecology and natural history were made during field work. Visual signaling, an aspect of territorial behavior that was already known for several species of the genus Phyllomedusa, could be described for the first time for Phyllomedusa boliviana (Jansen & J. Köhler 2007). Furthermore, during audio surveys of an anuran community at the study site San Sebastián from 2005 to 2007, a decline of certain amphibian populations was observed in the rainy season 2006/2007 (Jansen et al., in press). This is possibly related to an extreme drought in the dry season of 2006 where 158 consecutive days without rainfall were recorded. In addition, a new method for measuring intensity of anuran choruses by means of a continuous sound pressure metre was developed (Jansen 2009). The method was suitable to detect calling phenology (during one night), as well as differences in calling activity (between two nights). Biodiversity and biogeographical relationships: Species lists were compiled at the six study sites Pampagrande, Los Volcanes, San Sebastián, Caparú, El Espinal und El Corbalan. The total amphibian and reptile species numbers observed ranged from 37 to 101 with the highest species numbers in San Sebastián (101) and Caparú (89) and the lowest in Los Volcanes (37) and El Espinal (41). A preliminary species list of the herpetofauna of the Chiquitano Region was presented, including 60 amphibian and 84 reptile species. The majority of the amphibians of the Chiquitano Region are classified predominantly as inhabitants of open formations (41 species, 68.3%). Interestingly, even the majority of species recorded from the Chiquitano Dry Forest (32 species) are usually associated with open formations (22 species, 66.7%), followed by the number of species associated with open and forest formations (8 species, 24.4%). Only two of the observed species (6.0%) are predominant forest dwellers. The amphibian assemblage of the Chiquitano Region is most similar in composition to that of the Cerrado biome: 46 species (76.7%) occur in the Cerrado as well, and three species are regarded as Cerrado endemics (5.0%). The Chiquitano Region shares considerably fewer amphibian species with the other biomes (Amazon: 22 species, 36.7%; Gran Chaco: 13 species, 21.7%; Caatinga: 16 species, 26.7%). The reptile assemblage also has significant affinities to the Cerrado, which can be seen in the high proportion of reptile species distributed in that biome (68 species; 81.0%). Affinities to the other biomes are as follows: Amazon (48 species, 57.1%), Chaco (37 species, 40.1%), and Caatinga (30 species, 35.7%). When arranged in mutually exclusive biome categories, reptiles and amphibians showed similar patterns so that the majority of both amphibians and reptiles of the Chiquitano Region can be regarded as widespread. The high proportion of reptile species probably endemic to this region (5 species, 6.0%) is remarkable (i.e. Tropidurus xanthochilus, Apostolepis phillipsi, Phalotris sansebastiani, Xenopholis werdingorum, and Micrurus diana). In an analysis of the biodiversity patterns and biogeographical relationships of the herpetofauna of the study sites, these sites were compared with literature data from 37 localities and included in a presence/absence matrix with a total of 657 amphibian and reptile species in the surrounding South American biomes Amazon, Cerrado and Gran Chaco. The biogeographic relationships between these sites were evaluated using the Coefficient of Biogeographic Resemblance (CBR), cluster analysis, and multidimensional scaling (MDS) of sites. The analyses were first conducted on amphibians and reptiles combined, and than group-specific each for amphibians, reptiles, lizards, and snakes, separately. A “bias-reduced analysis” was developed for a better understanding of the affinities of the amphibians. In this analysis, e.g., the distinct habitat types of the Chiquitano Region, the Chiquitano Dry Forest and the Cerrado were taken into account. Analyses of the biodiversity patterns revealed that the sites in the Amazon comprise highest species numbers, as expected, followed successively by the sites in the Cerrado biome and sites in-between the two biomes. Within the eastern lowlands of Bolivia, the Chiquitano Region is the most rich in species. Comparing it with the other South American sites, the Chiquitano Region has a surprisingly high alpha diversity, especially in amphibians. The microgeographic variation in species composition (beta diversity) in the Chiquitano Region is also remarkably high and obviously related to the mosaic character of the vegetation and habitats. However, the bias-reduced analysis revealed that the amphibian fauna of the open areas and savannas at Hacienda San Sebastián (with 36 species in the Cerrado and pastureland) was one of the most species-rich savanna sites known for amphibians in South America. Considering that the Hacienda San Sebastián site is only ca. 3300 ha (= 1.29 amphibian species per km2), this outcome is particularly suprising. The results of the analyses of the biogeographical relationships suggest that the herpetofauna of Bolivia’s lowlands, including the Beni, the Pantanal and the Chiquitano Region, is as distinct from the herpetofauna of the Gran Chaco, Amazon, and Cerrado as these biomes are from each other. The Chiquitano herpetofauna in particular represents a unique and well-defined herpetofaunal assemblage when compared to all surrounding localities and biomes. This is supported by high CBR-values, findings from the cluster analysis, as well as a clear separation of the Chiquitano sites in the MDS. Biogeographic relations exist in all the surrounding biomes, but are strongest to Cerrado, followed by the Amazon. This study strongly suggests that the Chiquitano herpetofauna is composite and has multiple affinities. This is congruent with a well-defined Chiquitano flora, avifauna and mammalian fauna, suggesting a similar history. The bias-reduced analysis revealed a more detailed picture of the biogeographic relations of the Chiquitano Region, especially the Chiquitano Dry Forest. I argue here that the Chiquitano Dry Forest herpetofauna is a “young”, and “former savanna herpetofauna”. Whereas the Chiquitano Dry Forest is rather poor in amphibian and reptile species, and endemics are lacking from this forest type, the isolated Cerrado enclaves are especially diverse in species and probably contain locally endemic species, such as Phalotris sansebastiani and Xenopholis werdingorum. The colonization of the young Chiquitano Dry Forest may have taken place from savannas by mainly open area species, and only briefly through the Amazon. The results emphasise the importance of bias-reduction in studies of biogeography, e.g., by using group-specific analyses or by taking into account criterias as area size and heterogeneity of compared sites. The different biogeographic patterns of reptiles and amphibians of the Andean valleys indicate a different history of these two groups. In regard to reptiles, dispersals and withdrawals into the valleys in warm humid and dry cool periods in the Pleistocene seem likely, supported by a relation between the valleys and the dry lowland (e.g., Chaco). However, it is more plausible that, during these climatic fluctuations, amphibians migrated to adjacent, more humid regions, such as Yungas. The study verified the known patterns of sister-species pairs in the Inter-Andean Dry Forest and the lowlands. Additionally, pairs of populations with slight differences in morphology were found in the valleys and in the lowlands (Cercosaura parkeri and Xenodon rhapdocephalus). Further studies must test the taxonomic status of these populations. The discovery of new species of Bothrops and Bothrocophias from the Andean valleys has several implications, and possible reasons for the high endemism in the dry valleys are discussed. Conservation and outlook: The high local alpha and beta diversity of the Chiquitano herpetofauna shows that this is a region of complex faunal interaction, which reflects the present heterogeneity of the region, but which is possibly also related to a complex geological and environmental history. The Chiquitano Region can be assessed as a region of distinct regional herpetofaunal diversity charaterised by small scale diversity patterns. It therefore merits recognition as a unique ecoregion, and conservation effort should be increased. Further research is necessary to solve the taxonomic problems addressed in this study. Moreover, future work should be directed towards the development and institution of longterm monitoring programs to evaluate the effects of climate change and changes in land-use on biodiversity, especially that of the Chiquitano Region.
The physiology of our most complex organ, the brain, is still not comprehensively understood. The brain basically serves the processing, storing and binding of external and internal information, and thereby generates amazing phenomena like the understanding of oneself as an individual entitiy. How exactly information is encoded and represented, how individual neurons or networks of neurons actually interact, is a gigantic puzzle, whose pieces were collected since many decades. Subject of scientific discussions are the basic spatiotemporal structures of neuronal representations. Suggestions and observations reach hereby from simple rate coding of individual neurons to synchronous activity of larger ensembles. To approach answers to these questions, our working group has used a combination of different recording techniques that allowed for the comparison of neuronal interactions on different spatial scales. We focused on prefrontal neuronal interactions during visual short-term memory. Herefore two rhesus monkeys had been trained to perform a visual short-term memory task. We measured and recorded their neuronal activity by means of a microelectrode matrix that could be inserted into the cortex via a closable chamber, which had been previously implanted above prefrontal cortex. The acquired signal was separated into two components: a high-frequency component, that represents the spiking output activity of few neurons in the vicinity of each electrode tip (multi-unit activity), and a low-frequency component, that results from dendritic input activity of larger neuronal assemblies (local field potential). From one of the experimental animals we also recorded mass signals of even larger neuronal populations by means of small silverball electrodes, that had been implated into the skull above prefrontal cortex (skull EEG) in the context of a pilot project. In the first subproject, we analyzed the selectivity of output signals with respect to the memorized stimulus and task performance. We compared selectivities of local recording sites (multi-unit activity) with the selectivities of patterns created by the combined activity of all recording sites, thus representing the activity of large and distributed ensembles. Local neuronal activity correlated with the course of the visual short-term memory task, but was not highly discriminative with respect to different visual stimuli. We could show that the population activity was significantly more specific. Concerning task performance, we obtained the same result, albeit less pronounced. Further analyses revealed that the patterns of distributed ensemble activity were only partly based on realtime coordination of neuronal activity, and in addition, did not remain stable across the time course of the short-term memory task. In the second subproject, we focused on the oscillatory behavior of the local field potential. After a time-frequency analysis, we studied different frequency bands concerning stimulus selectivity and task performance of the monkey. We hereby found significant modulations of oscillations in the beta- and gamma-frequency range, that correlated with different periods of the task. Especially for oscillations in beta- and low-gamma-range, we observed phase-locking of oscillations between different recording sites, which could play an important role as internal clock to coordinate spatially separate activity. Local high-gamma oscillations themselves seemed to be important for the maintenance of information. These results could be partly confirmed by mass signals of EEG. In sum, our results support the hypothesis that information is represented in the brain by means of concerted activity of spatially distributed neuronal ensembles. This activity again appears to be coordinated by oscillatory activity in beta- and low-gamma-frequency ranges. A deeper understanding of central nervous information processing could contribute to better treatment of diseases like Parkinson’s, Alzheimer’s as well as epilepsy, and neuropsychiatric disorders like schizophrenia.
In dieser Arbeit wurden die physiologische Funktion innerhalb der Ribosomenbiogenese und die physikalischen Interaktionen des nukleolären, essentiellen Proteins Nep1p in der Hefe Saccharomyces cerevisiae untersucht. Durch Hefe-Zwei-Hybrid-Experimente und biochemische Analysen konnte eine Homodimerisierung des Proteins festgestellt sowie eine strukturabgeleitete Dimerisierungsmutante identifiziert werden. Ebenfalls aus der Struktur des Nep1p-Homologs aus Methanocaldococcus jannaschii konnte eine Nop14p-Bindungsregion auf der der Dimerkontaktfläche abgewandten Seite des Hefeproteins vorhergesagt und nach in vitro-Mutagenese bestätigt werden. Innerhalb des Nop14-Proteins wurden zwei Domänen charakterisiert, die im Zwei-Hybrid-System mit Nep1p interagieren. Aus Strukturdaten in Kombination mit Hefe-Drei-Hybrid-Experimenten konnte die RNA-Bindungsregion an der Dimerkontaktfläche des Nep1-Proteins lokalisiert werden. In Drei-Hybrid-Selektionen wurden RNA-Sequenzen mit hoher Affinität zu dem M. jannaschii Nep1p identifiziert, die auf eine Bindung des Proteins bei Helix 35 der 16S rRNA schließen lassen. Aufgrund der hohen Konservierung dieser rRNA-Region ist eine Bindung des Hefeproteins an die 18S rRNA-Schleife von Nukleotid 1189-1196 sehr wahrscheinlich. Da Nep1p eine große Ähnlichkeit zu Proteinen der SPOUTFamilie von Methyltransferasen aufweist, war von einer rRNA-Methylierung im Verlauf der Ribosomenbiogenese als katalytische Funktion des Proteins auszugehen. Aus verschiedenen Drei-Hybrid-Experimenten zur RNA-Bindungungsspezifität ergab sich als mögliche Reaktion die N1-Methylierung des Nukleotids 1-Methyl-3-(3-Amino-3-Carboxypropyl)-Pseudouridin (m1acp3Y) 1191 der 18S rRNA. Durch eine spezifische radioaktive Markierung der acp-Gruppe konnte gezeigt werden, dass Nep1p keinen Einfluss auf die spätere Aminocarboxypropylmodifizierung hat. Diese findet auch bei einer Deletion der snoRNA35 statt, also auch an einem Uridin, und ist unabhängig von dem cytoplasmatischen Protein Tma20p. In RP-HPLC-Experimenten konnte nachgewiesen werden, dass die 18S rRNA einer Dnep1Dnop6-Doppelmutante ein Aminocarboxypropyl-modifiziertes Nukleosid enthält, dass sich in seinem Retensionsverhalten von dem m1acp3Y eines Wildtyps unterscheidet. Bei dem in diesem Stamm detektierten acp-modifizierten Nukleosid handelt es sich vermutlich um ein nicht-methyliertes acpY, was eine Funktion von Nep1p als N1-Methyltransferase des Nukleotids Y1191 der 18S rRNA höchst wahrscheinlich macht. Diese katalytische Funktion konnte in Zusammenarbeit mit Prof. Wöhnert auch für das M. jannaschii Nep1p gezeigt werden. Dass sowohl eine snr35- Deletion als auch eine 18S rRNA-Mutation des Nukleotids 1191 nicht letal sind, machte deutlich, dass die N1-Methylierung nicht die essentielle Funktion von Nep1p darstellen kann. Weiterhin konnte nachgewiesen werden, dass die Suppression der nep1-1ts-Mutante durch S-Adenosylmethionin nicht auf der Unterstützung der Methyltransferase-Aktivität des Proteins, sondern vermutlich eher auf einer generellen Stabilisierung des temperatursensitiven Proteins beruht. Sowohl im Hefe-Nep1p als auch im humanen Homolog wurden durch biochemische und genetische Experimente mehrere Phänotypen der Bowen-Conradi-Mutation (Aspartat 90 zu Glycin in ScNep1p) nachgewiesen. Diese lassen auf eine Aggregation des mutierten Proteins sowie eine dadurch bedingte Fehllokalisation innerhalb der Zelle schließen. Zusätzlich ist aber auch ein RNA-Bindungsdefekt durch den Aminosäureaustausch wahrscheinlich. Nichtsdestotrotz liegt offensichtlich ausreichend Nep1p-Protein vor, dass seine essentielle Funktion erfüllen kann, da die Mutation selbst zu keinem Wachstumsphänotyp führt. Erst bei einer partiellen Translationsrepression des mutierten Proteins unter Verwendung des artifiziellen Tetrazyklin-Aptamer-Systems ist ein verlangsamtes Wachstum von Hefezellen zu beobachten, was dieses System geeignet zur Analyse von möglichen Therapeutika macht.
Riboswitche – Vorbilder für die Konstruktion synthetischer RNA Schalter Riboswitche sind natürliche RNA Regulatorelemente. Sie sind in den nicht kodierenden Regionen von messenger RNAs (mRNAs) lokalisiert und beeinflussen die Expression nachfolgender Gene. Riboswitche bestehen aus zwei Domänen. Die Binde- oder Aptamerdomäne bildet eine Bindetasche, die einen Liganden ohne die Hilfe zusätzlicher Faktoren hoch spezifisch und affin binden kann. Die zweite Domäne, die sogenannte Expressionsplattform, interpretiert den Bindestatus der Aptamerdomäne und beeinflusst die Expression der nachfolgenden Gene. Liganden sind meist kleine, organische Moleküle wie Nukleotide, Aminosäuren oder Vitamine. Riboswitche regulieren Gene, die für die Synthese oder Verwertung ihres jeweiligen Liganden in der Zelle von Bedeutung sind. Kontrolliert wird die Genexpression meist durch Transkriptionstermination oder durch Maskierung der ribosomalen Bindestelle (SD = Shine Dalgarno Sequenz). Auch Eukaryoten nutzen das Prinzip der direkten RNA-Ligand-Interaktion zur Genregulation, wenn gleich in geringerem Ausmaß. In Pilzen und Pflanzen wird durch Ligandenbindung alternatives Spleißen von prä-mRNAs induziert, was entweder zur mRNA Degradation durch alternative Polyadenylierung oder der Repression der Translation durch alternative Leserahmen (uORFs) führt. Charakteristisch für eine Regulation über Riboswitche ist die direkte Wechselwirkung des niedermolekularen Liganden mit der RNA. In trans kodierte Proteinfaktoren sind aufgrund dieser direkten Bindung nicht notwendig. Dies macht natürliche Riboswitche zu geeigneten Vorbildern für die Entwicklung künstlicher RNA Schalter. Synthetische Riboswitche Aptamere sind kleine, synthetisch hergestellte, einzelsträngige RNA oder DNA Moleküle, die hochaffin und sehr spezifisch ein Zielmolekül binden können. Man kann Aptamere gegen nahezu jedes Molekül der Wahl über einen Prozess der in vitro Selektion gewinnen (SELEX = systematic evolution of ligands by exponential enrichment). Eine Eigenschaft der meisten Aptamere ist, dass sie ihre endgültige Struktur erst in Gegenwart des spezifischen Liganden ausbilden („induced fit“). Dies kann ausgenutzt werden, um RNA Aptamere als regulatorische Elemente einzusetzen. Hierzu inseriert man Aptamere in nicht translatierte Regionen einer mRNA. In Abwesenheit des Liganden bildet sich die Struktur nur teilweise aus und interferiert nicht mit zellulären Funktionen. Erst im Komplex mit einem Liganden kommt es zur effizienten Beeinflussung der Genexpression. Inseriert man ein regulatorisch aktives Aptamer in den 5’ nicht translatierten Bereich (5’UTR) einer eukaryotischen mRNA, erlaubt das Aptamer in der nicht ligandengebundenen Form die Translation nachfolgender Gene. Erst der Aptamer-Ligand-Komplex interferiert mit der Translationsinitiation. Ist das Aptamer nahe der cap-Struktur positioniert, behindert es die initiale Bindung des Ribosoms an die mRNA. Bei einer weiter stromabwärts gelegenen Insertion interferiert es mit dem Scannen der kleinen ribosomalen Untereinheit nach dem Startcodon. Die beste Regulationseffizienz wird hierbei bei einer Insertion direkt vor dem Startcodon erreicht. Es zeigte sich jedoch, dass nur eine sehr geringe Anzahl an Aptameren in der Lage ist, als RNA Schalter aktiv zu sein. Dies führte dazu, dass bis heute nahezu alle Systeme entweder auf dem Theophyllin oder dem Tetrazyklin Aptamer basieren. Ziele dieser Arbeit In dieser Arbeit sollte untersucht werden, warum nur wenige Aptamere regulatorisch aktiv sind und was diese von inaktiven Varianten unterscheidet. Dafür wurden ein Tetrazyklin und ein Neomycin Aptamer detailliert charakterisiert. Desweiteren wurden neue RNA-basierte Regulationssysteme aufgebaut und ihr regulatorischer Mechanismus analysiert. Innerhalb dieser Arbeit wurde dabei ein System zur aptamerabhängigen Regulation des prä-mRNA Spleißens in Hefe etabliert. Außerdem konnte das bekannte Translationssystem für die Regulation essentieller Gene in Hefe weiter entwickelt werden. Folgende Ergebnisse wurden in dieser Arbeit erhalten: 1.Das Tetrazyklin Aptamer – In vitro Charakterisierung eines synthetischen Riboswitches. Das Tetrazyklin Aptamer ist 69 Nukleotide lang. Es besteht aus drei Stämmen (P1, P2 und P3) sowie drei einzelsträngigen Bereichen (J1/2, J2/3 und die Schleife L3; siehe Abbildung 1, links). Die Domäne oberhalb von P2 ist nicht an der Ligandenbindung beteiligt und kann ausgetauscht werden. Die Stämme P1-P3 sind bereits vor Ligandenbindung ausgebildet. Tetrazyklin wird über die drei einzelsträngigen Bereiche gebunden (siehe Abbildung 1, rechts). Durch fluorimetrische und kalorimetrische Methoden wurde eine Bindekonstante von Tetrazyklin an das Aptamer von 770 pM ermittelt. Diese Affinität ist außergewöhnlich hoch. Vergleichbare Aptamere und natürliche Riboswitche binden niedermolekulare Liganden 10- bis 1000-fach schlechter. Wir konnten zeigen, dass hohe Affinität eine Grundvoraussetzung für die regulatorische Aktivität ist, da Aptamermutanten mit verschlechterten Bindekonstanten keine in vivo Aktivität mehr aufweisen sind (Seiten 19-29). Durch Größenausschlußchromatographie konnte gezeigt werden, dass das Tetrazyklin Aptamer durch Ligandenbindung keine größeren globalen Konformationsänderungen erfährt. Dies weist auf eine weitgehende Vorformung der Bindetasche bereits ohne Tetrazyklin hin. Bei Ligandenbindung nimmt das Aptamer eine pseudoknotenähnliche Tertiärstruktur an, welche wahrscheinlich für die inhibitorische Wirkung auf das Ribosom verantwortlich ist (Seiten 19-29). Im Laufe dieser Arbeit wurde die Kristallstruktur des Aptamers im Komplex mit Tetrazyklin in der Arbeitsgruppe von A. R. Ferré-D’Amaré gelöst. Die Struktur zeigt, dass die Stämme P1 und P3 aufeinander gestapelt sind (Abbildung 1, rechts). Stamm P2 bildet die Verlängerung einer irregulären Helix, die aus den einzelsträngigen Bereichen J1/2 und J2/3 gebildet wird. Nukleotide der Schleife L3 interagieren mit dieser irregulären Helix und bilden mit ihr zusammen die Bindetasche für Tetrazyklin. Diese hochauflösende Struktur diente uns in weiteren Arbeiten als Ausgangspunkt für die detaillierte Charakterisierung von ligandeninduzierten Änderungen (siehe 6.). 2. Das Tetrazyklin Aptamer ist in der Lage, prä-mRNA Spleißen in Hefe zu inhibieren. Der Aptamer-Tetrazyklin-Komplex kann nicht nur mit der Translationsinitiation, sondern auch mit dem Spleißen der prä-mRNA in Hefe interferieren (Seiten 31-37). Dazu wurde ein Hefe-Intron in den Leserahmen von GFP inseriert. Nur bei korrektem prä-mRNA Spleißen wird die reife mRNA aus dem Kern transportiert und GFP exprimiert. Für eine RNA-basierte Regulation des Spleißens wurde die Konsensussequenz der 5’ Spleißstelle in den Stamm P1 des Tetrazyklin Aptamers integriert. Dieser ist nicht an der Ligandenbindung beteiligt und seine Sequenz daher variabel. Es konnte gezeigt werden, dass in Abwesenheit von Tetrazyklin das Intron vom Spleißosom erkannt und entfernt wird. Die Expression des Gens ist dann möglich. Durch die Zugabe von Tetrazyklin wird das Spleißen inhibiert und GFP nicht länger exprimiert. Biochemische Strukturkartierungen der RNA in An- und Abwesenheit von Tetrazyklin zeigten, dass der Stamm P1 durch Ligandenbindung verfestigt wird. Die Ligandenbindung beeinflusst also nicht nur die Struktur der Bindetasche, sondern wird auch auf angrenzende Stammbereiche übermittelt. Durch Stabilisierung des Stammes P1 wird die 5’ Spleißstelle für das Spleißosom maskiert. Somit konnten wir den Mechanismus für die Aptamer basierte Regulation des prä-mRNA Spleißens aufklären. 3. Die Tetrazyklin Aptamer basierte Inhibition der Translationsinitiation ermöglicht die Regulation essentieller Gene in Hefe. Frühere Arbeiten zeigten, dass die Insertion mehrerer Aptamerkopien in den 5’UTR zu einem effizienten Abschalten der Genexpression führt. Dies wurde genutzt, um ein neuartiges System für die konditionale Expression essentieller Gene in Hefe zu etablieren. In Zusammenarbeit mit der Arbeitsgruppe von Prof. K.-D. Entian wurden Insertionskassetten für eine PCR-basierte chromosomale Integration von Tetrazyklin Aptameren unter Kontrolle verschieden starker Promotoren konstruiert. Dafür wurden 1-3 Kopien des Tetrazyklin Aptamers unter Kontrolle des hoch exprimierenden TDH3-Promoters und des etwas schwächeren ADH1-Promoters gestellt. Außerdem wurde eines HA-tag angefügt, um die Genexpression mittels Westernblot verfolgen zu können. Zur Überprüfung der chromosomalen Insertion diente eine Kanamycin-Resistenz. Das neue System wurde erfolgreich an von fünf essentiellen Genen getestet. Es zeigte sich, dass die Zugabe von Tetrazyklin zu einem schnellen und effizienten Abschalten aller getesteten Gene führt. Die Vorteile dieses neuartigen konditionalen Genexpressionssystems in Hefe liegen in der einfachen Handhabung und der Unabhängigkeit vom verwendeten Stamm. Es müssen keine in transkodierten Proteinfaktoren coexprimiert werden. Durch dieses System konnte zum ersten Mal die Aptamer-basierte Regulation endogener, essentieller Gene gezeigt werden (Seiten 49-57). 4. Die Kombination von in vitro Selektion und in vivo Screening ermöglicht die Identifikation neuer regulatorisch aktiver Aptamere – ein Neomycin Riboswitch. Nur wenige in vitro selektierte Aptamere sind als synthetischer Riboswitch aktiv. In unserer Arbeitsgruppe wurde daher ein in vivo Screeningsystem zur Identifizierung neuer Aptamere in Hefe entwickelt. Eine Bibliothek in vitro selektierter Aptamere wurde hierzu in den 5’UTR des GFP Gens kloniert und die Aktivität einzelner Kandidaten durch Vergleich der Fluoreszenz in An- und Abwesenheit des Liganden überprüft. Wir verwendeten eine Bibliothek aus Neomycin-bindenden Aptameren und analysierten 5000 Hefeklone. Hierbei konnten zehn Sequenzen isoliert werden, die abhängig von Neomycin die Initiation der Translation inhibieren. Das 33 Nukleotid lange Aptamer N1 zeigt eine 7,5-fache Regulation und wurde näher charakterisiert. Es besteht aus einer internen asymmetrischen und einer terminalen Schleife, die durch zwei GC Basenpaare getrennt sind. Enzymatische Strukturkartierung und Mutationsanalyse zeigten, dass beide einzelsträngigen Bereiche für die Ligandenbindung wichtig sind. Der abschließende Stamm ist nicht an der Ligandenbindung beteiligt und hat geringen Einfluss auf die regulatorische Aktivität. N1 kann außerdem gegen andere Aminoglykosidantibiotika diskriminieren (Seiten 39-47). Interessanterweise sind die regulatorisch aktiven Aptamere in der in vitro selektierten Bibliothek stark unterrepräsentiert und konnten durch zufälliges Sequenzieren nicht identifiziert werden. Dieses Beispiel verdeutlicht eindrucksvoll die Notwendigkeit eines Screenings in vivo. 5. Regulatorisch aktive Neomycin Aptamere unterscheiden sich von inaktiven durch eine größere thermische Stabilisierung bei Ligandenbindung. Durch weitere Mutationsanalysen von N1 konnte ein aktivitätsvermittelndes Element im Neomycin Riboswitch identifiziert werden. Dazu wurde entweder die terminale oder die interne asymmetrische Schleife mutiert. Es konnte gezeigt werden, dass die Sequenz der terminalen Schleife nur einen modulierenden Einfluss auf die Aktivität hat, wobei die Asymmetrie der internen Schleife (aber nicht deren exakte Sequenz) ausschlaggebend für die regulatorische Aktivität ist. Für weitere Analysen wurde N1 mit fünf mutierten Varianten und dem inaktiven Neomycin bindenden Aptamer R23 verglichen. Alle sieben Aptamer haben eine ähnliche Sekundärstruktur und Ligandenaffinität, zeigen aber unterschiedliche Aktivität in vivo. Durch Bestimmung des Schmelzpunktes der verschiedenen Aptamere in An- und Abwesenheit von Neomycin zeigte sich, dass aktive Aptamere thermisch deutlich mehr durch Ligandenbindung stabilisiert werden als inaktive. Dabei ist die thermische Stabilität der Aptamer-Neomycin-Komplexe ähnlich. Jedoch ist die Stabilität ohne Ligand bei aktiven Aptameren gegenüber inaktiven Varianten deutlich erniedrigt. Durch NMR spektroskopische Untersuchungen in Zusammenarbeit mit Prof. J. Wöhnert konnte bestätigt werden, dass aktive Aptamere weniger stark vorgeformt sind als inaktive. Das in den Mutationsanalysen identifizierte Element nimmt nicht an der Ligandenbindung teil, sondern dient als Schalter, der den freien Zustand das Aptamers destabilisiert. Damit sorgt es für den großen Unterschied in der thermischen Stabilität des freien und des gebundenen Zustandes aktiver Aptamere. Dies zeigt, dass Unterschiede in der Stabilität die regulatorische Aktivität vermitteln (Seiten 73-102). Laufende Arbeiten sollen nun klären, ob thermische Stabilisierung durch Ligandenbindung ein allgemeingültiger Vermittler von regulatorischer Aktivität ist. Dazu werden weitere Aptamere überprüft, welche in Abwesenheit des Liganden unterschiedlich stark strukturiert sind und eventuell durch Ligandenbindung unterschiedlich stabilisiert werden. Außerdem werden wir testen, ob es die gewonnen Erkenntnisse erlauben, durch rationelles Design synthetische Riboswitche zu verbessern oder inaktive Aptamere in aktive zu verwandeln. 6. Was macht ein Aptamer zu einem regulatorisch aktiven Riboswitch? Für das Tetrazyklin Aptamer konnten wir zeigen, dass zum einen eine extrem hohe Bindekonstante und zum anderen eine hoch komplexe Bindetasche für die regulatorische Aktivität entscheidend sind. Dabei ist die Bindetasche in Abwesenheit des Liganden stark vorstrukturiert und erfährt keine globalen strukturellen Änderungen (Seiten 19-29). In Zusammenarbeit mit der Arbeitsgruppe von Prof. J. Wachtveitl untersuchen wir den Einfluss von Bindekinetik und Lebensdauer des Aptamer-Tetrazyklin-Komplexes auf die regulatorische Aktivität. Dafür vergleichen wir das Tetrazyklin Aptamer mit drei regulatorisch inaktiven Mutanten. Für die Messungen nutzen wir die Eigenfluoreszenz des Tetrazyklins. Diese ist in wässriger Lösung geqenched und steigt bei Bindung an die RNA deutlich an. Erste Ergebnisse zeigen große Unterschiede zwischen den Aptameren in der Geschwindigkeit der Ligandenbindung. Außerdem zeigen sich geringe Unterschiede in der Lebensdauer der verschiedenen Komplexe. Durch NMR spektroskopische Untersuchungen in der Arbeitsgruppe von Prof. J. Wöhnert können die Veränderungen einzelner Basen bei Ligandenbindung untersucht werden. Hierbei zeigen erste Messungen am Tetrazyklin Aptamer, unterschiedliches Verhalten einzelner an der Bindung beteiligter Nukleotide. Eine detaillierte Aufklärung der ligandeninduzierten Veränderungen gewährt uns weitere Einblicke, warum das Tetrazyklin Aptamer als Riboswitch aktiv ist. Die regulatorische Aktivität Neomycin abhängiger Riboswitche wird durch thermische Stabilisierung bei Ligandenbindung vermittelt. Dabei zeigte sich, dass durch Neomycin neue Basenpaare und Basenstapelungen entstehen. Durch weiterführende strukturelle Untersuchungen sollen nun ligandeninduzierte Veränderungen in N1 detailliert geklärt werden. Größere globale Änderungen konnten bereits durch EPR Spektroskopie in Zusammenarbeit mit der Arbeitsgruppe von Prof. T. F. Prisner ausgeschlossen werden. Hierzu wurden in der Arbeitsgruppe von Prof. J. W. Engels spinmarkierte Neomycin Aptamere hergestellt und die Abstände der Sonden in An- und Abwesenheit von Neomycin bestimmt. Es zeigte sich, dass sich der Abstand der Spinmarkierungen durch Zugabe von Neomycin (oder anderen Aminoglykosiden) nicht ändert (Seiten 59-72). Dies weist auf eher lokale Änderungen in der Bindetasche hin. Durch NMR Spektroskopie in Zusammenarbeit mit der Arbeitsgruppe von Prof. J. Wöhnert werden im Moment die Strukturen verschiedener N1-Aminoglykosid-Komplexe gelöst. Dabei zeigt sich, dass in vivo aktive und inaktive Liganden eine ähnliche Struktur im Aptamer induzieren. Was die einzelnen Komplexe unterscheidet und damit die verschiedene Aktivität begründet ist Ziel der Analyse. Insgesamt konnte in dieser Arbeit ein Regulationssystem für die Aptamer-basierte Kontrolle des prä-mRNA Spleißens in Hefe entwickelt und das bestehende Translationssystem für die Applikation auf essentielle Gene angewendet werden. Außerdem wurden wichtige Punkte, warum Aptamere als Riboswitch funktionieren aufgeklärt. Damit legt diese Arbeit einen wertvollen Grundstein für die Weiterentwicklung RNA-basierter Genregulationselemente für die Anwendung in der synthetischen Biologie.

Is the digital future a blessing for philologists, especially those working the vast area of Germanic Languages & Literatures? Or does it rather come with problems that jeopardize philology, in the Germanic and the broader scope? This paper sets out to explore the status quo (1.) of digital source material in Germanic philology, ranging from medieval manuscripts to 21st century e-books and their, at times, highly restricted availability to the scientific community. Do we really face a quantum leap in terms of open access, or is this leap rather confined to those only who pay the exorbitant fees specialist libraries charge for the use of their rare manuscript and book collections? How about Google Books (2.)? Are we in danger of neglecting everything that is (still) missing there? And what is in it for the German scholar? Can we believe that in some years’ time we will be able to get our hands on every source text we desire within seconds since it is only a click away? This paper critically assesses the process and progress of the digitization of mankind’s written records (3.), focusing on problems to be overcome by e.g. medievalists wishing to consult certain source material. This is illustrated by means of the example of how it is not yet possible consult certain materials related to the Franciscan preacher Berthold von Regensburg († 1272). A short concluding summary (4.) highlights perspectives for further thinking and discussion.
Das Biodiversität und Klima Forschungszentrum (BiKF) hat sich selbst das Ziel gesetzt, mit seinen Arbeiten wissenschaftliche Beiträge zum Management klimabedingter relevanter Biodiversitätsveränderungen sowie zur Erfüllung internationaler Übereinkommen wie der EU-Fauna-Flora-Habitat-Richtlinie (EU-FFH), der EU-Wasserrahmenrichtlinie (EU-WRRL), der internationalen Biodiversitätskonvention (CBD) und der Klimarahmenkonvention (UNFCCC) zu liefern. Voraussetzung dafür ist ein Ergebnis- und Wissenstransfer zwischen den Forschenden des Zentrums und den Anspruchsgruppen aus Zivilgesellschaft, Politik und Wirtschaft (vgl. Biodiversität und Klima Forschungszentrum (BiKF) 2008: 1 und 40ff.). Um gesellschaftliche Wirkungen zu erzeugen, müssen die Interessen, Positionen und praktischen Wissensbestände der Anspruchsgruppen in die Forschungen aufgenommen werden und die Forschungsergebnisse in die gesellschaftliche Praxis (rück-) vermittelt werden. Um für das Biodiversität und Klima Forschungszentrum (BiKF) ein besseres Verständnis über die Entstehung und den Transfer von Wissen zwischen Forschung und Gesellschaft vorzubereiten und Ansatzpunkte für Interventionen in öffentliche Debatten aufzuzeigen, wurde eine problemorientierte Diskursfeldanalyse (DFA) zum übergreifenden Themengebiet Klimabedingte Biodiversitätsveränderungen durchgeführt (zur Methode siehe Infobox). Dabei wurden prioritäre Themen identifiziert sowie relevante Akteure und ihre Positionen typisiert. Diese Analyse dient somit als Orientierungshilfe für die strategische Ausrichtung des Zentrums; sie bietet eine strukturierte Grundlage für eine zentrumsinterne Diskussion des weiteren Vorgehens im Wissens- und Ergebnistransfer. Die Diskursfeldanalyse speist sich aus drei Detailanalysen zu den Diskursarenen ‚9. Vertragsstaatenkonferenz der CBD‘, ‚Nationale Biodiversitätsstrategie‘ und ‚Millennium Ecosystem Assessment‘. Diese werden nach einer kurzen Einführung zur Methode vorgestellt. In einem zweiten Schritt werden die über diese drei Arenen hinweg relevanten und für BiKF mögliche Ansatzpunkte bietenden Diskursstränge aufgezeigt. Diese illustrieren die Vielfalt an Themen und Positionen der Akteure innerhalb des Diskursfelds. Abschließend werden Schlussfolgerungen für die Struktur von Wissenskonflikten im Diskursfeld gezogen, die Orientierung geben sollen für eine weitere Auseinandersetzung des Forschungszentrums mit möglichen Querschnittsthemen und Zielen von Interventionen in den Diskurs.

More importantly, the bodies left by the serial killer are not likely to be intact and whole. What he leaves behind and what we, the audience, will get to see is the body in pieces, dismembered, scattered. A series of snapshots, partial views, and close-ups, inflicting cold sharp shocks, is all we may glimpse: the head of Benjamin Raspail floating in a jar of formaldehyde in The Silence of the Lambs, a finger removed by the serial killer from his landlord’s hand in Kalifornia, a ziploc bag of fingers recovered from a flooded drainpipe in When The Bough Breaks, a surgically severed hand used to leave misleading fingerprints on a wall at a crime scene in Seven.
Crystals of the title compound, C12H8N2·C7H8O2, were obtained during cocrystallization experiments of a compound with two hydrogen-bond donors (2-hydroxybenzyl alcohol) with another compound containing two hydrogen-bond acceptors (phenanthroline). Unexpectedly, the two molecules do not form dimers with two O—H ... N hydrogen bonds connecting the two molecules. However, one of the hydroxy groups forms a bifurcated hydrogen bond to both phenanthroline N atoms, whereas the other hydroxy group forms an O—H ... O hydrogen bond to a symmetry-equivalent 2-hydroxybenzyl alcohol molecule. In addition, the crystal packing is stabilized by Pi – Pi interactions between the two phenanthroline ring systems, with a centroid–centroid distance of 3.570 Å.
The title compound, C6H5NO2·C6H6O2, crystallizes with one pyridinium-2-carboxylate zwitterion and one molecule of benzene-1,2-diol in the asymmetric unit. The crystal structure is characterized by alternating molecules forming zigzag chains running along the a axis: the molecules are connected by O—H ... O and N—H ... (O,O) hydrogen bonds.
Die Frage, ob "Literatur als Kampfmittel" Theorie oder Realität in der DDR gewesen ist, lässt sich nicht eindeutig beantworten. Schließlich redet man über 40 Jahre DDR-Regime, in denen es kulturpolitische Zäsuren und Strategiewechsel gegeben hat. Generell gilt: Die Kulturpolitik der DDR war kunstfeindlich. Offiziell gab es keine Zensur, faktisch schon. Die Führung der DDR betrachtete ganz in der Tradition von Stalin und Chruschtschow KünstlerInnen als aktive Kämpfer für den Sozialismus. Der realistische Sozialismus war genreübergreifend die einzig legitime Form. Und man war gewillt, diese offen formulierten Anforderungen an Literaten auch mit Gewalt durchzusetzen. Trotzdem war die offizielle Literaturpolitik das eine, die literarische Praxis das andere. Die Literaturszene im Prenzlauer Berg etwa war erstaunlich autonom und im Aufbau-Verlag erschienen bemerkenswerte Titel. Die Ausbürgerung Biermanns 1976 führte zu einer historisch einmaligen Solidaritätsbewegung und einem Exodus der Ost-Literaten. Sicher ist: Die DDR ist Vergangenheit, aber in ihrer Literatur ist die Erinnerung an Staat und Gesellschaftssystem bewahrt.
Alan Croslands Film THE JAZZ SINGER wird in zahlreichen wissenschaftlichen Abhandlungen zum Übergang vom Stumm- zum Tonfilm als der Film beschrieben, der dem Tonfilm endgültig zum Durchbruch verhalf (vgl. Dibbets 1998, 197; Nowell-Smith 1998, 193; Henzel 2006, 47). Doch obwohl THE JAZZ SINGER maßgeblich dazu beigetragen hat, das neue Medium Tonfilm am Markt zu etablieren, handelt es sich bei diesem Film nicht um den ersten Tonfilm, wie gelegentlich behauptet wird (vgl. Ferrari 2004, 70), sondern lediglich um einen „Stummfilm mit einigen vertonten Einschüben“ (Dibbets 1998, 197). THE JAZZ SINGER ist ein so genannter part-talkie, ein Film also, der nur zum Teil vertont wurde. An ausgewählten Stellen enthält er „lippensynchrone Lieder und Dialog“ (Dibbets 1998, 197). Das expressive Spiel der Figuren und der Einsatz von Zwischentiteln hingegen erinnern an den Stummfilm. Doch nicht nur die Tatsache, dass THE JAZZ SINGER kein hundertprozentiger Tonfilm ist, steht laut Christoph Henzel einer Definition als erstem Tonfilm entgegen. Darüber hinaus ist er auch nicht der erste Film, der technisch in der Lage war, Bild und Ton zu synchronisieren. Vielmehr positioniert er sich als ein Ereignis unter vielen, als ein Glied innerhalb einer ganzen Reihe technischer Entwicklungen vom Stumm- zum Tonfilm (vgl. Henzel 2006, 48/49).
In der vorliegenden Arbeit wurden zehn Substanzen, die im Rahmen des Projekts INTAFERE analytisch in verschiedenen Gewässersystemen des Hessischen Rieds nachgewiesen werden konnten, ökotoxikologisch charakterisiert. Neben der Bestimmung der Akut- und der chronischen Toxizität wurde auch das endokrine Potential mit Hilfe eines rekombinanten Hefe-Assays ermittelt.Die akute Toxizität zeigt zwischen den verschiedenen Substanzen große Differenzen. Die drei Organophosphate TCPP, TBEP und TCEP zeigen selbst bei hohen Konzentrationen keine oder nur sehr geringe Effekte, während 4-NP, 4-t-OP, BPA, TDCPP, AHTN und Terbutryn mit LC50-Werten bis zu 5 mg/l eine höhere Toxizität besitzen.Im Hefe-Assay kann in mehreren Versuchswiederholungen das östrogene Potential von 4-NP (MW: 6,71x10-6 M), 4-t-OP (MW: 7,16x10-6 M) und BPA (MW: 4,88x10-6 M), sowie die antiandrogene Wirkung des Bisphenols (5,29x10-5 M) bestätigt werden. Im Gegensatz dazu können im Yeast Antiestrogen Screen zum ersten Mal Hinweise auf die antiöstrogene Wirkung von TCPP (MW: 6,86x10-5 M), Terbutryn (MW: 3,99x10-5 M), TDCPP (MW: 2,65x10-6 M) und TBP (MW: 2,28x10-5 M) aufgezeigt werden.TBP und TDCPP führen auch in der chronischen Exposition bei Potamopyrgus antipodarum zu einer Reduktion in der Embryonenzahl (mehrfach beobachtete NOEC beider Substanzen: 6,25 mg/kg), ein Effekt, der zunächst nicht von einer toxischen Wirkung unterschieden werden kann. Allerdings scheint sich ein antiöstrogener Wirkmechanismus auf die Schnecken bei den Versuchen zur Mischtoxizität zu bestätigen, da die gleichzeitige Exposition gegenüber BPA und 4-tOP zu einer geringfügigen Aufhebung des Effekts führt. Potamopyrgus reagiert ebenfalls mit einer Reduktion der Embryonenzahl bei der Exposition gegenüber AHTN (mehrfach beobachtete NOEC: 2 mg/kg), zeigt sich aber insensitiv gegenüber der Belastung mit Terbutryn.Die Exposition von Chironomus riparius gegenüber den verschiedenen Substanzen führt bis auf eine Ausnahme zu keinen signifikanten substanzbedingten Beeinträchtigungen. Einzig das s-Triazin Terbutryn verursacht eine hohe Mortalität mit einer NOEC (28 d) von 250 μg/kg. Eine Toxizität auf den Anneliden Lumbriculus variegatus zeigt sich lediglich bei derExposition gegenüber BPA (NOEC: 10 mg/kg; Biomasse, 28 d) und 4-NP (EC10: 6,88 mg/kg; 95% KI: 3,97-11,9; Reproduktion, 28 d). Die durchgeführten Versuche zur Toxizität von Mischungen zeigen eine größere Gefährdung auf als bei Vorliegen der Einzelsubstanzen in Konzentrationen unterhalb ihres Schwellenwertes zu vermuten wäre. Allerdings lassen sich die mit dem Hefe-Assay erzielten Ergebnisse aufgrund großer Variabilitäten zwischen den einzelnen Versuchswiederholungen nicht immer eindeutig mit Hilfe des additiven oder des unabhängigen Modells beschreiben, zeigen dabei jedoch trotzdem ein gegenüber den Einzelsubstanzergebnissen verändertes Risiko auf. Um diesem in der Risikobewertung Rechnung zu tragen, wird ein Verfahrensvorschlag entwickelt, in dem durch zusätzliche Sicherheitsfaktoren für PNECs ein Überschreiten des risikoanzeigenden Werts von 1 des PEC/PNEC-Quotienten einer Mischung verhindert werden kann.
It has become popular for journalists who are trying to sell newspapers, and politicians who are trying to solicit votes, to refer to this financial crisis as the worst since the Great Depression or WWII. I don’t know whether it is the worst or not so will leave that question to the historians and economists of the future once the storm has past. But it is indeed a “storm” as described by Vince Cable, Member of Parliament in his UK bestselling book entitled “The Storm – The World Economic Crisis and What it Means”. He describes this “storm” as a very destructive one displacing jobs, businesses, banks and whole economies from Iceland to the United Kingdom to the United States. I propose to offer a short chronology and summary of the causes of the current economic crisis. Then I will review several of the regulatory responses to the crisis focusing on the Turner Report, the de Larosière Group and certain US Treasury statements. I will offer my critiques of these proposals and then make some predictions of what the financial services industry may look like in the future.
Ezrin gehört zur Gruppe der Actin-Zytoskelett-bindenden ERM-Proteine und ist in seiner Funktion für Leukozyten gut beschrieben, wo es in Zusammenhang mit der interund intrazellulären Induktion von Apoptose gebracht wurde. Im Kontext der Astrozytenbiologie ist seine Expression mit astrozytärer Aktivierung, der Malignität von Astrozytomen und der Apoptose von Neuronen bei HIV-Encephalitis assoziiert. Die vorliegende Studie untersucht die Expression von Ezrin in Astrozyten bei verschiedenen Demenzerkrankungen mit der Frage nach krankheitsspezifischen astrozytären Aktivierungsphänomenen und einem möglichen Zusammenhang mit neuronaler Apoptose. Hierzu wurden 51 Gehirne von Demenzpatienten und 17 Gehirne von Normalkontrollen des Neurologischen Instituts (Edinger Institut) der Johann Wolfgang Goethe-Universität Frankfurt am Main histopathologisch untersucht. An Schnitten von jeweils 4-6 in Paraffin eingebetteten Gehirnarealen pro Fall wurden eine modifizierte Silberimprägnationsfärbung nach Bielschowsky und immunhistochemische Färbeverfahren für Ezrin, GFAP, HLA-DR, hyperphosphoryliertes Tau-Protein und Ubiquitin angefertigt und semiquantitativ evaluiert. Zur Darstellung von apoptotischen DNA-Strangbrüchen wurde die TUNEL-Methode verwendet. Die hier untersuchte Stichprobe entspricht in Hinsicht auf die Verteilung verschiedener Demenztypen weitgehend der neueren Literatur: Die häufigste Demenzerkrankung war die Alzheimersche Erkrankung mit 84% (N=43), gefolgt von der Demenz mit Lewy-Körperchen mit 35% (N=18), der vaskulären Demenz mit 18% (N=9) und der Frontal–Lobe-Dementia mit 6% (N=3). 47% der Fälle litten an einer Kombination von zwei Demenzerkrankungen. Die astrozytäre Ezrin-Expression zeigt keine signifikante Differenz zwischen verschiedenen Demenzformen. Dies trifft sowohl für singuläre Demenzerkrankungen als auch für Kombinationsdemenzerkrankungen zu. Die astrozytäre Ezrin-Expression korreliert hochsignifikant mit dem Schweregrad der Alzheimerschen Erkrankung, gemessen an der Dichte neuritischer Plaques in Anlehnung an die Klassifikation nach CERAD. Die astrozytäre Ezrin-Expression korreliert hochsignifikant positiv mit der Apoptose von Neuronen unabhängig vom Erkrankungstyp. Im Unterschied zur Ezrin-Immunoreaktivität, die nahezu ausschließlich bei Demenzfällen mit neuronaler Apoptose eine deutliche Erhöhung aufwies, zeigte sich die GFAP- und HLA-DR-Immunoreaktivität bei Demenzfällen generell im Vergleich zu Kontrollfällen deutlich erhöht. Zusammenfassend trägt die Untersuchung der astrozytären Ezrinexpression nicht zur Differentialdiagnose von Demenztypen bei, ist jedoch mit dem Schweregrad von Demenzerkrankungen korreliert. Ezrin fungiert offenbar als Indikator eines spezifischen astrozytären Aktivierungszustandes, der eine Assoziation mit schwerem Gewebeschaden in Form neuronaler Apoptose aufweist. Dieser Aktivierungszustand erwies sich in der vorliegenden Untersuchung als unabhängig vom Krankheitstyp, was für die These spricht, dass eurodegenerative Erkrankungen pathophysiologische Gemeinsamkeiten teilen könnten. Darüber hinaus weisen die Ergebnisse der Studie darauf hin, dass die Aktivierung von Astrozyten kein homogenes Phänomen darstellt, sondern in distinkten Formen assoziiert mit progressiver Gewebsschädigung des ZNS auftreten kann.
Untersuchung der Bedeutung von Kombinationstherapien bei triple-negativen Mammakarzinomzellen

Das Mammakarzinom umfasst eine heterogene Gruppe von Tumoren und lässt sich bei prognostischen Aussagen und der Auswahl von Therapiestrategien an spezifischen Kriterien wie dem Tumorgrad, dem Hormonrezeptorstatus und der Expression von HER2/neu-Rezeptoren klassifizieren (Schindler, 2008). Die so genannten “triple-negativen” Mammakarzinome tragen weder Östrogen-, noch Progesteron-, noch HER2/neu-Rezeptoren. Sie stellen aufgrund ihrer besonders aggressiven Wachstumsrate, ihrer verhältnismäßig ungünstigen Prognose sowie dem Fehlen zielgerichteter Therapien eine große klinische Herausforderung dar. Kombinierte Chemotherapien versprechen bis dato die größten Erfolgsaussichten. Allerdings gibt es bis heute noch keine spezifische systemische Therapie für triple-negative Mammakarzinome (Cleator et al. 2007). Demzufolge muss es das Ziel der aktuellen Forschung sein, die bestehenden Therapiekonzepte zu optimieren bzw. weitere Methoden für neue zielgerichtete Therapien zu entwickeln. In der aktuellen molekularbiologischen Forschung ist es mit Hilfe von neuartigen Strategien wie der RNA-Interferenz (RNAi) möglich, gezielt die Expression eines bestimmten Gens zu unterdrücken, wodurch dann beispielsweise das Wachstumsverhalten von Tumorzellen gehemmt werden kann. Wie vorausgegangene Studien gezeigt haben, fällt Polo-like Kinase 1 (Plk1) in Zellen mit hohem mitotischem Index und einer dadurch verstärkten Proliferation eine besondere Bedeutung zu. So liegt Plk1 in nahezu allen humanen Tumorarten überexprimiert vor (Strebhardt et al. 2006). Zudem besteht ein Zusammenhang zwischen Überexpression von Plk1 und einem Schutz der Tumorzellen vor Apoptose (Cogswell et al. 2000). Im Rahmen der hier vorgelegten Promotionsarbeit wurden diese Erkenntnisse aufgegriffen und mögliche Therapieformen geprüft. Dabei wurde die Applikation von siRNA zur Hemmung der Expression von Plk1 bei der bislang wenig untersuchten triple-negativen Mammakarzinomzelllinie EVSA-T getestet. Darüber hinaus wurde nach eventuellen synergistischen Effekten von siRNA in Kombination mit bereits etablierten Therapeutika, wie Paclitaxel oder Carboplatin gesucht. Synergistische Effekte könnten für die Krebstherapie von höchster Relevanz sein. Bei der Einzelbehandlung der triple-negativen EVSA-T-Zelllinie sowohl mit siRNA4 gegen Plk1, als auch mit Paclitaxel oder Carboplatin, wurde die Zellproliferation signifikant inhibiert. Mittels FACS-Analyse konnte einhergehend mit verminderter Plk1-Expression ein deutlich erhöhter Anteil von Zellen in der G2/M-Phase festgestellt werden. Desweiteren wurden für Apoptose typische Ereignisse, wie DNA-Kondensation, ein Schrumpfen der Zellsomata und die Aktivierung der Procaspasen 3 und -9 festgestellt. Im Vergleich zur Wirkung der Einzelsubstanzen wurden durch kombinierte Applikation von siRNA4 mit Paclitaxel oder Carboplatin synergistische Effekte bezüglich einer Hemmung der Zellproliferation beobachtet (CI < 1). Auch die Untersuchung der Zellzyklusverteilung ergab eine synergistische Verstärkung des G2/M-Arrest durch Kombination von siRNA4 mit Paclitaxel (CI < 1). Dies liefert wichtige Erkenntnisse für die Weiterentwicklung und Optimierung von Kombinationstherapien. So könnten potentielle Nebenwirkungen minimiert werden, indem statt einer definierten Konzentration einer Einzelsubstanz, zwei Therapeutika in geringeren Konzentrationen verabreicht werden. Weiterhin läßt sich anhand der durchgeführten Experimente der Schluss ziehen, dass in triplenegativen Brusttumoren eine tumorintrinsische Wachstumsregulation durch Plk1 stattfindet und dass siRNA4, welche gegen Plk1 gerichtet ist, in vitro in der Lage ist, die Proliferation triple-negativer Tumorzellen zu inhibieren. Um Plk1 als potentes Zielgen für die Therapie des triple-negativen Mammakarzinoms nachhaltig zu bestätigen und die Innovation von Kombinationstherapien voranzutreiben, müssten in weiterführenden Untersuchungen zunächst im Rahmen von Zellkulturexperimenten alternative triple-negative Zelllinien verwendet werden. Um die in vitro beobachteten Effekte auf die in vivo-Situation übertragen zu können, sollte ein Xenograft-Mausmodell etabliert werden. Nach erfolgversprechenden und quantitativ ausreichenden Tierexperimenten sowie vorklinischen Studien könnten beschriebene Therapieformen anschließend in klinischen Studien angewendet werden. Besonders positiv fällt dabei ins Gewicht, dass bis auf siRNA alle hier verwendeten Therapeutika bereits eine klinische Zulassung besitzen.
The recent financial crisis has led to a major debate about fair-value accounting. Many critics have argued that fair-value accounting, often also called mark-to-market accounting, has significantly contributed to the financial crisis or, at least, exacerbated its severity. In this paper, we assess these arguments and examine the role of fair-value accounting in the financial crisis using descriptive data and empirical evidence. Based on our analysis, it is unlikely that fair-value accounting added to the severity of the current financial crisis in a major way. While there may have been downward spirals or asset-fire sales in certain markets, we find little evidence that these effects are the result of fair-value accounting. We also find little support for claims that fair-value accounting leads to excessive write-downs of banks’ assets. If anything, empirical evidence to date points in the opposite direction, that is, towards overvaluation of bank assets.
The global financial crisis has lead to a renewed interest in discretionary fiscal stimulus. Advocates of discretionary measures emphasize that government spending can stimulate additional private spending — the so-called Keynesian multiplier effect. Thus, we investigate whether the discretionary spending announced by Euro area governments for 2009 and 2010 is likely to boost euro area GDP by more than one for one. Because of modeling uncertainty, it is essential that such policy evaluations be robust to alternative modeling assumptions and different parameterizations. Therefore, we use five different empirical macroeconomic models with Keynesian features such as price and wage rigidities to evaluate the impact of fiscal stimulus. Four of them suggest that the planned increase in government spending will reduce private spending for consumption and investment purposes significantly. If announced government expenditures are implemented with delay the initial effect on euro area GDP, when stimulus is most needed, may even be negative. Traditional Keynesian multiplier effects only arise in a model that ignores the forward-looking behavioral response of consumers and firms. Using a multi-country model, we find that spillovers between euro area countries are negligible or even negative, because direct demand effects are offset by the indirect effect of euro appreciation.
Recent evaluations of the fiscal stimulus packages recently enacted in the United States and Europe such as Cogan, Cwik, Taylor and Wieland (2009) and Cwik and Wieland (2009) suggest that the GDP effects will be modest due to crowding-out of private consumption and investment. Corsetti, Meier and Mueller (2009a,b) argue that spending shocks are typically followed by consolidations with substantive spending cuts, which enhance the short-run stimulus effect. This note investigates the implications of this argument for the estimated impact of recent stimulus packages and the case for discretionary fiscal policy.
This paper investigates the impact of IT standardization on bank performance based on a panel of 457 German savings banks over the period from 1996 to 2006. We measure IT standardization as the fraction of IT expenses for centralized services over banks' total IT expenses. Bank efficiency, in turn, is measured by traditional accounting performance indicators as well as by cost and profit efficiencies that are estimated by a stochastic frontier approach. Our results suggest that IT standardization is conducive to cost efficiency. The relation is positive and robust for small and medium-sized banks but vanishes for very large banks. Furthermore, our study confirms the often cited computer paradox by showing that total IT expenditures negatively impact cost efficiency and have no influence on bank profits. To the best of our knowledge, this paper is first to empirically explore whether IT standardization enhances efficiency by employing genuine data of banks' IT expenditures. Over the last four decades the literature on bond rating changes and its effects on security prices increased significantly with almost all studies not controlling for the respective reason for those. We therefore investigate the impact of rating events on the stock and the credit default swap (CDS) market incorporating rating reviews and rating changes together with the reason mentioned by the rating agency. Our results for the general effects are in line with prior findings but conditioning on the respective reason shows that the markets’ anticipation of rating actions is largely driven by events due to changes in firms’ operating performance. Furthermore, we provide empirical evidence for the hypothesis in prior literature that a surprise downgrade does not necessarily have to be bad news for stockholders when wealth is transferred from bondholders, but negative rating actions are always bad news for bondholders. The results additionally reveal increasing rating announcement effects by declining credit quality of firms for both rating reviews and changes. JEL Classification: D82, G14, G20. The objective of this paper is to test the hypothesis that in particular financially constrained firms lease a higher share of their assets to mitigate problems of asymmetric information. The assumptions are tested under a GMM framework which simultaneously controls for endogeneity problems and firms’ fixed effects. We find that the share of total annual lease expenses attributable to either finance or operating leases is considerably higher for financially strained as well as for small and fast-growing firms – those likely to face higher agency-cost premiums on marginal financing. Furthermore, our results confirm the substitution of leasing and debt financing for lessee firms. However, we find no evidence that firms use leasing as an instrument to reduce their tax burdens. In this paper we investigate the comparative properties of empirically-estimated monetary models of the U.S. economy. We make use of a new data base of models designed for such investigations. We focus on three representative models: the Christiano, Eichenbaum, Evans (2005) model, the Smets and Wouters (2007) model, and the Taylor (1993a) model. Although the three models differ in terms of structure, estimation method, sample period, and data vintage, we find surprisingly similar economic impacts of unanticipated changes in the federal funds rate. However, the optimal monetary policy responses to other sources of economic fluctuations are widely different in the different models. We show that simple optimal policy rules that respond to the growth rate of output and smooth the interest rate are not robust. In contrast, policy rules with no interest rate smoothing and no response to the growth rate, as distinct from the level, of output are more robust. Robustness can be improved further by optimizing rules with respect to the average loss across the three models.
Renewed interest in fiscal policy has increased the use of quantitative models to evaluate policy. Because of modeling uncertainty, it is essential that policy evaluations be robust to alternative assumptions. We find that models currently being used in practice to evaluate fiscal policy stimulus proposals are not robust. Government spending multipliers in an alternative empirically-estimated and widely-cited new Keynesian model are much smaller than in these old Keynesian models; the estimated stimulus is extremely small with GDP and employment effects only one-sixth as large.
We examined the neural signatures of stimulus features in visual working memory (WM) by integrating functional magnetic resonance imaging (fMRI) and event-related potential data recorded during mental manipulation of colors, rotation angles, and color–angle conjunctions. The N200, negative slow wave, and P3b were modulated by the information content of WM, and an fMRI-constrained source model revealed a progression in neural activity from posterior visual areas to higher order areas in the ventral and dorsal processing streams. Color processing was associated with activity in inferior frontal gyrus during encoding and retrieval, whereas angle processing involved right parietal regions during the delay interval. WM for color–angle conjunctions did not involve any additional neural processes. The finding that different patterns of brain activity underlie WM for color and spatial information is consistent with ideas that the ventral/dorsal “what/where” segregation of perceptual processing influences WM organization. The absence of characteristic signatures of conjunction-related brain activity, which was generally intermediate between the 2 single conditions, suggests that conjunction judgments are based on the coordinated activity of these 2 streams. We show that loanword adaptation can be understood entirely in terms of phonological and phonetic comprehension and production mechanisms in the first language. We provide explicit accounts of several loanword adaptation phenomena (in Korean) in terms of an Optimality-Theoretic grammar model with the same three levels of representation that are needed to describe L1 phonology: the underlying form, the phonological surface form, and the auditory-phonetic form. The model is bidirectional, i.e., the same constraints and rankings are used by the listener and by the speaker. These constraints and rankings are the same for L1 processing and loanword adaptation.
Experimental data shows that adult learners of an artificial language with a phonotactic restriction learned this restriction better when being trained on word types (e.g. when they were presented with 80 different words twice each) than when being trained on word tokens (e.g. when presented with 40 different words four times each) (Hamann & Ernestus submitted). These findings support Pierrehumbert’s (2003) observation that phonotactic co-occurrence restrictions are formed across lexical entries, since only lexical levels of representation can be sensitive to type frequencies.
The present study argues that variation across listeners in the perception of a non-native contrast is due to two factors: the listener-specic weighting of auditory dimensions and the listener-specic construction of new segmental representations. The interaction of both factors is shown to take place in the perception grammar, which can be modelled within an OT framework. These points are illustrated with the acquisition of the Dutch three-member labiodental contrast [V v f] by German learners of Dutch, focussing on four types of learners from the perception study by Hamann and Sennema (2005a).
The multiplicity fluctuations in A+A collisions at SPS and RHIC energies are studied within the HSD transport approach. We find a dominant role of the fluctuations in the nucleon participant number for the final fluctuations. In order to extract physical fluctuations one should decrease the fluctuations in the participants number. This can be done considering very central collisions. The system size dependence of the multiplicity fluctuations in central A+A collisions at the SPS energy range – obtained in the HSD and UrQMD transport models – is presented. The results can be used as a ‘background’ for experimental measurements of fluctuations as a signal of the critical point. Event-by-event fluctuations of the K/p , K/p and p/p ratios in A+A collisions are also studied. Event-by-event fluctuations of the kaon to pion number ratio in nucleus-nucleus collisions are studied for SPS and RHIC energies. We find that the HSD model can qualitatively reproduce the measured excitation function for the K/p ratio fluctuations in central Au+Au (or Pb+Pb) collisions from low SPS up to top RHIC energies. The forward-backward correlation coefficient measured by the STAR Collaboration in Au+Au collisions at RHIC is also studied. We discuss the effects of initial collision geometry and centrality bin definition on correlations in nucleus-nucleus collisions. This paper analyzes the risk properties of typical asset-backed securities (ABS), like CDOs or MBS, relying on a model with both macroeconomic and idiosyncratic components. The examined properties include expected loss, loss given default, and macro factor dependencies. Using a two-dimensional loss decomposition as a new metric, the risk properties of individual ABS tranches can directly be compared to those of corporate bonds, within and across rating classes. By applying Monte Carlo Simulation, we find that the risk properties of ABS differ significantly and systematically from those of straight bonds with the same rating. In particular, loss given default, the sensitivities to macroeconomic risk, and model risk differ greatly between instruments. Our findings have implications for understanding the credit crisis and for policy making. On an economic level, our analysis suggests a new explanation for the observed rating inflation in structured finance markets during the pre-crisis period 2004-2007. On a policy level, our findings call for a termination of the 'one-size-fits-all' approach to the rating methodology for fixed income instruments, requiring an own rating methodology for structured finance instruments. The genus Haroldiataenius Chalumeau, 1981 (Aphodiinae: Eupariini) from southern United States, Mexico, and Central America is revised and nine species are recognized. The subgeneric name Sayloria Chalumeau, 1981 is synonymized with Haroldiataenius (sensu stricto) and Ataenius sabinoi Cartwright, 1974 is synonymized with A. lucanus Horn, 1871. Five species are transferred to Haroldiataenius from the genus Ataenius Harold creating the following new combinations: H. convexus (Robinson), H. griffini (Cartwright), H. lucanus (Horn), H. saramari (Cartwright), and H. semipilosus (Van Dyke). One new species,Haroldiataenius buvexus is described from Texas, USA. A key to species of Haroldiataenius is included and pertinent morphological details are illustrated.
The New World euparine scarab genera Parataenius Balthasar, 1961 and Pseudataenius Brown, 1927 are revised. Ataenius brunneus Schmidt is transferred to the genus Parataenius becoming Parataenius brunneus (Schmidt), new combination. The monospecific genus Ataenioides Petrovitz, 1973, is synonymized with Pseudataenius Brown, 1927, (new synonymy) and the type species, Ataenioides gracilitarsis Petrovitz, is given the new combination Pseudataenius gracilitarsis (Petrovitz). New species of Parataenius are described from southern South America: Parataenius selvae, P. estero, and P. martinezi. Keys for species of both genera are presented and pertinent morphological details are illustrated.
Interesse für und Liebe zu den Vögeln und vor allem zu den Vogelstimmen entwickelten sich an getrennten Orten: Bei Hans-Heiner Bergmann in Marburg und Osnabrück, bei Hans-Wolfgang Helb in Erlangen und Kaiserslautern. Neue Forschungsmöglichkeiten und moderne Technik wie die Sonagraphie ließen aus den beiden Ornitho-Akustikern ein Kollegen-Team werden. Beide Stimmen-Sammlungen zusammen mündeten in gemeinsamen Publikationen, auch in dem Werk „Die Stimmen der Vögel Europas“.
Epidermal growth factor (EGF) receptor belongs to the broad family of enzymatic receptors called receptor tyrosine kinases (RTKs). Generally, the binding of a ligand to these receptors leads to activation of their intracellular kinase activity that sets in motion a cascade of signaling events. In order to ensure appropriate responses to physiological stimuli, the cell is endowed with the ability to regulate signal transduction via numerous mechanisms such as dephosphorylation of the RTK and its substrates as well as downregulation of the RTK. Activation of EGFR is a potent mitogenic (proliferative) and motogenic (cell motility) signal that plays crucial roles during embryonic development and maintenance of adult tissue. EGFR signaling is primarily regulated by ligand-induced receptor internalization with subsequent degradation in lysosomes. While the complex of proteins that are recruited to EGFR after its activation is well understood, proteins that interact with the receptor in the absence of ligand binding are still not systematically studied. With the goal of identifying novel binding partners of non-activated EGFR, a membrane based yeast-two hybrid screen (MYTH) was conducted. MYTH is based on the principle of in vivo reconstitution of the N-terminus (Nub) and C-terminus (Cub) halves of ubiquitin once brought into close proximity. A chimeric protein consisting of EGFR fused to Cub and a transcription factor was used as a bait to screen Nub-tagged cDNA library. Analysis of resultant yeast transformants revealed a total of 87 proteins to interact with EGFR. Of these only 11 were previously shown to bind to EGFR. A majority of the other proteins were shown to interact with the receptor by yeast retransformation. Fifteen were confirmed to bind to EGFR by coimmunoprecipitation assays in mammalian cells. One of the novel EGFR interactors identified in the screen was histone deacetylase 6 (HDAC6). This deacetylase is localized in the cytoplasm and known to deacetylate alpha-tubulin, HSP90 and cortactin. The juxtamembrane region of EGFR binds to the Cterminus of HDAC6. Functionally, overexpression of wild type HDAC6 stabilized ligand-induced degradation of the receptor. On the other hand, deacetylase deficient or EGFR binding compromised mutants of HDAC6 were able to stabilize EGFR only partially. Downmodulation of HDAC6 expression by RNAi markedly accelerated degradation of the receptor. Taken together, HDAC6 is a negative regulator of EGFR downregulation that is dependent on its deacetylase activity and ability to bind to the receptor. Imaging studies revealed that HDAC6 does not affect internalization of EGFR from the plasma membrane but rather influences the post-endocytic trafficking of the receptor-ligand complex to lysosomes. Pulse-chase experiments using fluorophoretagged EGF showed that EGFR is transported faster towards the peri-nuclear region and delivered to late endosomes rapidly in HDAC6 depleted cells. HDAC6 is demonstrated to act, at least partly, by regulating the acetylation of alpha-tubulin. Upon EGFR activation, acetylation of alpha-tubulin on lysine 40 is progressively increased as shown by mass spectrometry and immunoblotting. Forced expression of a dominant negative mutant of alpha-tubulin, but not wild type alpha-tubulin, led to reduced speed and processive movement of early endosomes in GFP-Rab5 expressing cells. In a surprising twist, EGFR is able to phosphorylate HDAC6 on Tyr570. Phosphorylation of Tyr570 and Ser568 leads to inactivation of the deacetylase function of HDAC6 as shown by in vivo and in vitro assays. In summary, HDAC6 diminishes EGFR downregulation by slowing the transport of intracellular vesicles. The inhibitory effect is removed once HDAC6 is phosphorylated on key residues. In line with these findings, two recent reports have shown that hyper-acetylation of alpha-tubulin induced by inhibition of HDAC6 increases the transport of brain derived neurotrophic factor and JNK interacting protein-1 in different cell systems. Acetylated microtubules are more efficient in recruiting motor proteins like kinesin-1 and dynein. These findings indicate that HDAC6 plays an important regulatory role in intracellular trafficking pathways. However, several outstanding issues still remain unresolved. How does acetylation of microtubules influence vesicular trafficking? In this regard, the temporal and spatial dynamics of alpha-tubulin acetylation following EGFR activation should be studied. Furthermore, whether HDAC6 affects the trafficking of other endocytic cargos and additional organelles is an interesting question to address.
Die vorliegende Arbeit analysierte die Behandlung von Patienten mit Infektionserkrankungen am Zentrum der Zahn-, Mund- und Kieferheilkunde des Klinikums der Johann Wolfgang Goethe-Universität, Frankfurt am Main (Carolinum). Dabei wurde vor allem untersucht, welche Auswirkungen die Reduzierung von Personalressourcen speziell für die Behandlung dieser Patienten ab dem Jahr 2000 hinsichtlich der Betreuung und Versorgung hatte. Die Studie war retrospektiv angelegt und wertete Daten aus den Jahren 1998 bis 2002 aus. Hierfür wurden alle im Archiv lagernden Karteikarten herangezogen und die Daten in eine dafür entwickelte FilemakerPro©-Datenbank übertragen. Im Untersuchungszeitraum nahmen 940 Patienten mit Infektionskrankheiten etwa 3700 Besuche wahr. Regelmäßig erschienen 25% der Patienten, auf sie entfielen 60% aller Besuche. Diese Gruppe wurde einer näheren Betrachtung unterzogen: Die Auswertung der erhobenen Daten zeigte, dass das Ziel der Absenkung der Patientenzahlen und die Anzahl der Behandlungstermine erreicht werden konnte, die systematische Betreuung der Patienten sich jedoch verschlechterte. Der Anteil der sanierten Patienten sank von 34% auf 18% ab, die Zahl der unsystematisch behandelten Patienten verdreifachte sich dagegen von 1999 auf 2000 und blieb bei diesem hohen Wert. Vorsorgebehandlungen nahmen maximal ein fünftel aller Behandlungen ein, mit abnehmender Systematik sank dieser Anteil gegen null. Begünstigender Faktor für eine Sanierung waren ein erstelltes OPG (ersatzweise ein Zahnfilmstatus) und die Erstellung einer Behandlungsplanung. Das Vorhandensein eines OPGs erhöhte aber nicht die Wahrscheinlichkeit für eine folgende Behandlungsplanung. Patienten die an das Carolinum von außerhalb überwiesen wurden, hatten eine größere Chance auf eine Sanierung. Letztlich wurden nur 4% aller regelmäßig erschienenen Patienten systematisch mit Recall betreut, zahnärztlich saniert wurden insgesamt jedoch 30% der behandelten Patienten. Entgegen allgemeiner Annahmen waren kurzfristig abgesagte oder nicht eingehaltene Termine die Ausnahme.
The selection of features for classification, clustering and approximation is an important task in pattern recognition, data mining and soft computing. For real-valued features, this contribution shows how feature selection for a high number of features can be implemented using mutual in-formation. Especially, the common problem for mutual information computation of computing joint probabilities for many dimensions using only a few samples is treated by using the Rènyi mutual information of order two as computational base. For this, the Grassberger-Takens corre-lation integral is used which was developed for estimating probability densities in chaos theory. Additionally, an adaptive procedure for computing the hypercube size is introduced and for real world applications, the treatment of missing values is included. The computation procedure is accelerated by exploiting the ranking of the set of real feature values especially for the example of time series. As example, a small blackbox-glassbox example shows how the relevant features and their time lags are determined in the time series even if the input feature time series determine nonlinearly the output. A more realistic example from chemical industry shows that this enables a better ap-proximation of the input-output mapping than the best neural network approach developed for an international contest. By the computationally efficient implementation, mutual information becomes an attractive tool for feature selection even for a high number of real-valued features.

W. G. Sebald's "The Rings of Saturn" resists genre classification: Neither novel nor mere travel literature, it combines report with fiction, historical anecdotes with melancholy musings, explanations of economic and political facts with accounts of personal anxieties. This vast material, however, is joined by a semiotic regard, a way of looking at the world attentive on signs and their use. Hence, some of the far-ranging narratives, observations and reflections, often barely connected by associations of the narrator, can be systematically linked by a semiotic methodology. In this article, semiotic categories like index, icon and symbol, encoding, decoding and recoding, change of meaning and pattern recognition are applied to different levels of the text. The semiotic approach thus integrates the disparate episodes in "The Rings of Saturn", revealing them as being linked by their interest in signs and signification. In this way, semiotics proves itself as a method for literary studies.
In this thesis we have studied the physics of different ultracold Bose-Fermi mixtures in optical lattices, as well as spin 1=2 fermions in a harmonic trap. To study these systems we generalized dynamical mean-field theory for a mixture of fermions and bosons, as well as for an inhomogeneous environment. Generalized dynamical mean-field theory (GDMFT) is a method that describes a mixture of fermions and bosons. This method consists of Gutzwiller mean-field for the bosons, and dynamical mean-field theory for the fermions, which are coupled on-site by the Bose-Fermi density-density interaction and possibly a Feshbach term which converts a pair of up and down fermions into a molecule, i.e. a boson. We derived the self-consistency equations and showed that this method is well-controlled in the limit of high lattice coordination number z. We develop real-space dynamical mean-field theory for studying systems in an inhomogeneous environment, e.g. in a harmonic trap. The crucial difference compared to standard DMFT is that we are taking into account that different sites are not equivalent to each other and thus take into account the inhomogeneity of the system. Different sites are coupled by the real-space Dyson equation. ...

Da Fugenelemente morphologisch zum Bestimmungs- und nicht zum Grundwort gehören, spielt das Grundwort keine Rolle bei der Entscheidung, ob die Kompositionsnahtstelle verfugt wird oder nicht […]. […] Dabei fällt auf – und hierauf spielt der Zweifelsfall Subjekt(+s+)pronomen im Titel an –, dass es zahlreiche Schwankungen gibt […]. Schwankungsfälle erfordern u. E. besondere Aufmerksamkeit, weil sie in der Regel Sprachwandel im Vollzug darstellen, d. h. den Übergang eines älteren zu einem jüngeren Sprachzustand markieren. […] Was die Fremdwortkomposita so interessant macht, ist die Tatsache, dass nichtnatives Material einem sehr nativen Verfahren […] unterzogen wird […]. In diesem Beitrag gehen wir wie folgt vor: Zunächst – in Kapitel 2 – definieren wir das Fugenelement und stellen kurz seine Entstehung, sein Inventar von immerhin sechs Allo-Formen und deren Distribution dar. Die deutschen Präteritopräsentia sind, indem alte Perfektformen das heutige Präsens stellen, aus mehreren Gründen als hochgradig irregulär zu betrachten. Hinzu kommt ein bisher nicht geklärter Umlaut bei vier (von heute sieben) dieser Verben: „müssen“, „dürfen“, „können“ und „mögen“. Bisherige Erklärungsversuche werden diesem Problem nicht gerecht: Zwar versuchen sie durchaus, den Umlaut im Präsens zu motivieren, doch vermögen sie es nicht, sein ausschließliches Vorkommen im Plural des Präsens zu erklären. Hier wird für die These argumentiert, dass es sich um einen (verbalen) Pluralumlaut handelt, der insbesondere auch im Nominalbereich gang und gäbe ist und dort zur gleichen Zeit einen massiven Ausbau (Morphologisierung) erfährt. Damit handelt es sich um einen sog. transkategorialen Marker. In deutschen Dialekten haben auch andere Verben zu solchen Pluralumlauten gegriffen.

Posebno se istražuje participska konstrukcija apsolutni nominativ (particip u nominativu + ime u nominativu) te se utvrđuju njezine sintaktičke funkcije.
This article compares the prototypical (i. e. the most frequent) surnames of three neighbouring regions: The Netherlands, Flanders, and Germany. It concentrates on the surname’s emergence, development, their lexical sources and their current distribution. The latter is documented by maps based on telephone or official registers. Only some of the regional differences can be explained by cultural or historical factors. An important result is that onomastic landscapes do not follow national or linguistic borders.

Tagung des Berufsverbandes der Fachärzte für Orthopädie und Unfallchirurgie ; 21. - 24.10.2009, Berlin Fragestellung: Ziel war die Evaluierung der funktionellen und radiologischen Ergebnisse nach osteosynthestischer Versorgung von Olecranonfrakturen mit einer zur Hakenplatte modifizierten Drittelrohrplatte. Methodik: In einem Zeitraum von 12 Monaten wurden 29 Patienten mit Olecranonfrakturen prospektiv erfasst und eine Osteosynthese mit einer zur Hakenplatte modifizierten Kleinfragment-Drittelrohrplatte durchgeführt. Eine additive Verschraubung von zusätzlichen Fragmenten erfolgte bei 6 Patienten (20%). Das mittlere Patientenalter zum Unfallzeitpunkt betrug 50 Jahre (Min 29/ Max 83). Unter Verwendung der Frakturklassifikation nach Schatzker stellte sich in 8 Fällen (28%) eine Querfraktur vom Typ A, in 6 Fällen (20%) eine Querfraktur mit Impaktion vom Typ B und in 15 Fällen (52%) eine Mehrfragmentfraktur vom Typ D dar. Bei 4 Patienten (14%) lag eine offene Fraktursituation vor. Nach im Mittel 7,2 Monaten (Min 6/ Max 8) wurde das funktionelle Outcome anhand des Mayo Elbow Performence Scores (MEPS), der visuellen Analogskala (VAS) und des Disabilities of the Arm, Shoulder and Hand Scores (DASH) bewertet sowie die radiologischen Befunde erhoben. Ergebnisse und Schlussfolgerungen: Das Ziel einer primär übungsstabilen Osteosynthese konnte bei allen Patienten erreicht werden. Unter Verwendung des MEPS wurden annähernd ausschließlich sehr gute (12 Fälle/41%) und gute (16 Fälle/55%) Ergebnisse erzielt. Lediglich bei einem Patienten ergab sich ein nur befriedigendes Ergebnis. Der mittlere Punktwert für den MEPS betrug 91,4 (Min 65/Max 100). In der VAS konnte ein Mittelwert von 8,2 Punkten (Min 7/Max 10) erzielt werden (0=keine Zufriedenheit, 10=volle Zufriedenheit). Der mittlere DASH-Wert betrug 16,2 Punkte (Min 0/Max 39). Der mittlere Bewegungsumfang betrug für Extension/Flexion bei einem durchschnittlichen Streckdefizit von 8° (Min 0°/Max 25°) und einer Beugung von 135° (Min 105°/Max 155°) 125° (Min 90°/Max 155°). Die Unterarmumwendbewegungen waren mit 175° (Min 165°/Max 180°) kaum eingeschränkt. Bei keinem Patienten gab es postoperative Komplikationen. Eine zur Hakenplatte modifizierte Drittelrohrplatte stellt im Gegensatz zu präformierten, winkelstabilen Implantaten eine kostengünstige Alternative zur Osteosynthese bei Olecranonfrakturen dar. Auch bei komplexen mehrfragmentären Frakturtypen und osteoporotischer Knochenqualität konnte so im nachuntersuchten Kollektiv eine sichere Frakturretention erzielt werden. Hinsichtlich des funktionellen Ergebnisses profitieren die Patienten von einer dadurch unmittelbar postoperativ möglichen, physiotherapeutischen Nachbehandlung. Das für die Hakenplatte benötigte Osteosynthesematerial (Kleinfragment-Drittelrohrplatte) ist nahezu überall verfügbar und lässt sich in kurzer Zeit der individuellen Anatomie und Frakturmorphologie anpassen. Eine Drahtmigration, wie bei der weit verbreiteten Zuggurtungsosteosynthese häufig beobachtet, ist ausgeschlossen.
Biodiversität und Klimawandel in der Debatte um den ökologischen Waldumbau – eine Diskursfeldanalyse

In der Umweltpolitik werden die Themenfelder Klimawandel und Biodiversität besonders betont. Klimaveränderung und Erhalt/Veränderungen der biologischen Vielfalt werden zumeist unabhängig voneinander betrachtet und nicht miteinander verknüpft. Debatten zum Klimawandel und zum Schutz der Biodiversität werden von Vertretern der Politik, Wirtschaft, Wissenschaft und Gesellschaft kontrovers geführt. Die im aktuellen Diskurs um den ökologischen Waldumbau erwähnten Zusammenhänge von Biodiversität und Klimawandel werden analysiert, da dieser Umbau darauf abzielt, Waldbestände mit naturnahen Strukturen und natürlichen Lebensabläufen zu schaffen. Aufgrund des Klimawandels sind trotz Wissenslücken Maßnahmen eines klimaanpassenden Umbaus sinnvoll. Erwartungsgemäß wird Biodiversität häufig thematisiert, etwa bezogen auf die Baumartenwahl und -mischung, aber auch die Verkürzung der Umtriebszeiten und Grenzen der Naturverjüngung. Hinweise für weitere Aktivitäten im LOEWE Biodiversität und Klima Forschungszentrum BiKF wurden identifiziert.
Background The effect of additional treatment strategies with antineoplastic agents on intraperitoneal tumor stimulating interleukin levels are unclear. Taurolidine and Povidone-iodine have been mainly used for abdominal lavage in Germany and Europe. Methods In the settings of a multicentre (three University Hospitals) prospective randomized controlled trial 120 patients were randomly allocated to receive either 0.5% taurolidine/2,500 IU heparin (TRD) or 0.25% povidone-iodine (control) intraperitoneally for resectable colorectal, gastric or pancreatic cancers. Due to the fact that IL-1beta (produced by macrophages) is preoperatively indifferent in various gastrointestinal cancer types our major outcome criterion was the perioperative (overall) level of IL-1beta in peritoneal fluid. Results Cytokine values were significantly lower after TRD lavage for IL-1beta, IL-6, and IL-10. Perioperative complications did not differ. The median follow-up was 50.0 months. The overall mortality rate (28 vs. 25, p = 0.36), the cancer-related death rate (17 vs. 19, p = .2), the local recurrence rate (7 vs. 12, p = .16), the distant metastasis rate (13 vs. 18, p = 0.2) as well as the time to relapse were not statistically significant different. Conclusion Reduced cytokine levels might explain a short term antitumorigenic intraperitoneal effect of TRD. But, this study analyzed different types of cancer. Therefore, we set up a multicentre randomized trial in patients undergoing curative colorectal cancer resection. Trial registration : ISRCTN66478538
Der Begriff des doing gender als interaktive Inszenierung des sozialen Geschlechts (gender) hat sich auch in der Linguistik etabliert und ist vor allem für die Sprachverwendung bzw.- Gesprächslinguistik fruchtbar gemacht worden. Doch selbst etwas so biologisch determiniert Erscheinendes wie weibliche und männliche Stimmen, ihre Höhe, ihre Verlaufsmuster, sind konstruierter, als man dies bisher für möglich gehalten hatte. Der am stärksten und radikalsten segregierte sprachliche Bereich, die Rufnamen, wurde für das Deutsche erst 2003 mit der Arbeit "Naming Gender" von Susanne Oelkers empirisch auf die Kodierung von Geschlecht hin untersucht. Erstmals wird systematisch nachgewiesen, dass und worin sich Frauen- und Männernamen phonologisch-strukturell voneinander unterscheiden, außerdem, dass wir diese Geschlechtszuordnungen auch bei uns unbekannten Namen vornehmen. Das heißt, es besteht ein kollektives Wissen darüber, wie weibliche und männliche Rufnamen beschaffen sind.

Die jüngste Expedition einer Gruppe Studierender im Jahr 2006 unter Leitung von Professor Hans-Heiner Bergmann (Universität Osnabrück) brachte es ans Tageslicht. In der Sarmaschlucht westlich des Baikal im russischen Sibirien kommt eine bisher unbekannte endemische Art aus der Ordnung der Sperlingsvögel (Passeriformes) vor, der Sarmensische Steppenschwirl (Locustella sarmensis, Bergmann 2006).
U radu se analizira latinski sintaktički utjecaj pri uporabi zamjenica u Marulićevu i Kašićevu prijevodu popularnoga srednjovjekovnog djela De imitatione Christi. Istražuju se ova sintaktička svojstava zamjenica: izricanje pripadanja 3. licu s pomoću genitiva ličnih zamjenica za 3. lice, uporaba posvojne zamjenice za 1. i 2. lice u odnosu na povratno-posvojnu zamjenicu svoj, uporaba povratno-posvojne zamjenice svoj u odnosu na posvojne za 3. lice, uporaba lične zamjenice za 1. i 2. lice u odnosu na povratnu zamjenicu, uporaba odnosnih zamjenica na početku rečenice te množina srednjega roda pokaznih, relativne (koji) i neodređenih (sav, svaki) zamjenica u značenju jednine. Na temelju promatranih kategorija autorice nastoje utvrditi sličnosti i razlike tih dvaju proznih prijevodnih tekstova te objasniti prevoditeljski postupak.

Representations of the unknown and the foreign can be found in every culture. Paralleling the method of constructing identity in relation to the Other, all cultures create myths about the ‘foreign’ in order to discern what the ‘native’ is, and thus often essentialize them as either good or bad, ultimately to vindicate one’s own actions and values. The nature of myths has it as such that they lend themselves to images, which are easily transformed into representations. Representations of the foreign in the United States follow the same purpose; they are propagated to define the nation’s identity and set it into political and cultural relation to other nations and civilizations. In this thesis’ context, then, representations of Asian Americans in American culture strengthen the imaginative bonds of American national identity manifesto. However, the interdependency of the Self and the Other clarifies and further entangles the subjects that constitute American national identity and in turn legitimizes the belated claim of Asian Americans to be included into it. Asian American literature is primarily concerned with these myths and (mis)representations that are influenced by Orientalist images in Western culture. Thus, Orientalism – a constructed myth about the Orient, which exists in art, books, and armchair theories of all kinds in the Western world – becomes the main motif for Asian American literature. If we construe this theory a little further then Asian American identity is formed in relation to Orientalist representations that need to be deconstructed first. From the outset, if Orientalism is considered as a produce of imperialism, it seems that time is a defining factor in Orientalism, both as an agent of change and as a factor of perspective. In reality, however, Orientalism seems resilient to time and change; the creation of the Madame Butterfly myth exemplifies what was created in 1887 had been perfected by 1900 and since then enjoys frequent comebacks until today. Thus, for Asian American artists and writers to dismantle Orientalist stereotypes begins a literally archaeological process: excavating the leftovers of American Orientalism, evaluating those finds, and re-relating them with their own cultural and historical actuality. Rather than producing a neat line of argumentation, the approaches on defining Asian American identity within the American national identity manifesto fall into unwieldy clusters and even get tangled up into self-contradictions. The methods of dismantling Orientalist stereotypes are manifold and range from total rejection over evocation and appropriation to reflection. In order to wrestle such disparate issues Orientalism produces in Asian American Literature into an organic whole, it was important to focus consistently on the over arching theme of American national identity. As this thesis aims to show, Orientalist issues that are dealt with in Asian American literature all point toward the greater aim of national inclusion. This thesis is grouped into two parts. PART I provides historical and theoretical background information necessary to understand Orientalist issues in contemporary Asian American literature. Analogous to Asian American writers that feel the necessity to bed their work into the correct historical frame in order to prevent misunderstanding, chapters two and three serve to couch my argument into the correct frame. The theoretical base work is laid with Edward Said’s Orientalism and its implementation on the American and Asian American context. PART II examines literary examples, applying the theorems discussed in PART I. Chapter four is a close analysis of the submissive Butterfly stereotype that has, since its appearance in late nineteenth century, moved, inspired and even outraged writers. Beginning with the literary development of Madame Butterfly, D. H. Hwang’s deconstructivist M. Butterfly gives new perspectives on Orientalism by redefining gender and racial roles. To complement my analysis, in chapter five, I try to trace current Asian American reactions to Orientalism. Texts by comedian Margaret Cho and poet Beau Sia serve as examples of analysis. As a result of the disparate narrative forms of the analyzed works and the unevenness of scholarship on twenty-first century, the analyses vary greatly in scope and detail. In choosing fairly young narrative forms like stand-up comedy and spoken word poetry I want to emphasize how Orientalism pertains to the question of Asian American identity. To close the circle of my discourse I will go back to where I start my thesis: Asian Americans and their position within America’s national identity discourse. It is noteworthy that until today, Asian American identity remains a hostage of these Orientalist stereotypes that mark the boundaries of their American identity.
Introduction Loss of intestinal integrity has been implicated as an important contributor to the development of excessive inflammation following severe trauma. Thus far, clinical data concerning the occurrence and significance of intestinal damage after trauma remain scarce. This study investigates whether early intestinal epithelial cell damage occurs in trauma patients and, if present, whether such cell injury is related to shock, injury severity and the subsequent inflammatory response. Methods Prospective observational cohort study in 96 adult trauma patients. Upon arrival at the emergency room (ER) plasma levels of intestinal fatty acid binding protein (i-FABP), a specific marker for damage of differentiated enterocytes, were measured. Factors that potentially influence the development of intestinal cell damage after trauma were determined, including the presence of shock and the extent of abdominal trauma and general injury severity. Furthermore, early plasma levels of i-FABP were related to inflammatory markers interleukin-6 (IL-6), procalcitonin (PCT) and C-reactive protein (CRP). Results Upon arrival at the ER, plasma i-FABP levels were increased compared with healthy volunteers, especially in the presence of shock (P < 0.01). The elevation of i-FABP was related to the extent of abdominal trauma as well as general injury severity (P < 0.05). Circulatory i-FABP concentrations at ER correlated positively with IL-6 and PCT levels at the first day (r2 = 0.19; P < 0.01 and r2 = 0.36; P < 0.001 respectively) and CRP concentrations at the second day after trauma (r2 = 0.25; P < 0.01). Conclusions This study reveals early presence of intestinal epithelial cell damage in trauma patients. The extent of intestinal damage is associated with the presence of shock and injury severity. Early intestinal damage precedes and is related to the subsequent developing inflammatory response.
Realität – Repräsentation – Öffentlichkeit : der Dokumentarfilm in der heutigen Fernsehlandschaft

In dieser Studie wurde die Frage aufgestellt, ob und wie durch einen Fernsehdokumentarfilm im Vergleich zu ethnographischen Darstellungen Wirklichkeit dokumentiert und Wissen über diese vermittelt werden kann. Die Beantwortung der Fragestellung wurde durch eine vergleichende Analyse einzelner Produktionsschritte, wie auch der dahinter stehenden Kriterien und Konventionen auf empirischer und theoretischer Ebene, angestrebt. Ergänzt wurde dies durch eine Evaluation der Rahmenbedingungen, die sich auf die Produktion und vereinzelt auch auf die Konventionen auswirken können.
Breaking tolerance to the natural human liver autoantigen cytochrome P450 2D6 by virus infection

Autoimmune hepatitis (AIH) is a chronic liver disease of unknown etiology, characterized by a loss of tolerance against hepatocytes leading to the progressive destruction of hepatic parenchyma and cirrhosis. Clinical signs for AIH are interface hepatitis and portal plasma cell infiltration, hypergammaglobulinemia, and autoantibodies. Based on serological markers AIH is defined in subtypes. The hallmark of AIH type 2 are type 1 liver/kidney microsomal autoantibodies (LKM-1), whereas AIH type 1 is characterized by the presence of anti-nuclear (ANA) and/or anti-smooth muscular (SMA) autoantibodies. The major autoantigen recognized specifically by LKM-1 autoantibodies was identified as the 2D6 isoform of the cytochrome P450 enzyme family (CYP2D6). Not much is known about the etiology and pathogenic mechanisms of AIH so far and most animal models available result in only transient hepatic liver damage after a rather complex initiation method. It was the aim of my project to generate a novel animal model for AIH that reflects the chronic and progressive destruction of the liver characteristic for the human disease while using a defined and feasible initiating event to further analyze the pathogenic mechanisms leading to the autoimmune-mediated destruction of the liver. Therefore, mice transgenically expressing the human CYP2D6 in the liver and wild-type mice were infected with a liver-tropic adenovirus expressing the human CYP2D6 (Ad-2D6). Selftolerance to CYP2D6 was broken in Ad-2D6-infected mice, resulting in persistent autoimmune liver damage, apparent by cellular infiltration, hepatic fibrosis and necrosis. Similar to type 2 AIH patients, Ad-2D6-infected mice generated LKM-1-like antibodies recognizing the same immunodominant epitope of CYP2D6. Taken together, we could introduce a new animal model that reflects the persistent autoimmune-mediated liver damage as well as the serological marker characteristic for AIH type 2 and we could demonstrate that chronic autoimmune diseases targeting the liver can be triggered by molecular mimicry occurring in the context of a hepatotropic viral infection.
In this work, we extend the Hegselmann and Krause (HK) model, presented in [16] to an arbitrary metric space. We also present some theoretical analysis and some numerical results of the condensing of particles in finite and continuous metric spaces. For simulations in a finite metric space, we introduce the notion "random metric" using the split metrics studies by Dress and al. [2, 11, 12].
Background: Mitochondrial DNA sequencing increasingly results in the recognition of genetically divergent, but morphologically cryptic lineages. Species delimitation approaches that rely on multiple lines of evidence in areas of co-occurrence are particularly powerful to infer their specific status. We investigated the species boundaries of two cryptic lineages of the land snail genus Trochulus in a contact zone, using mitochondrial and nuclear DNA marker as well as shell morphometrics.
Results: Both mitochondrial lineages have a distinct geographical distribution with a small zone of co-occurrence. In the same area, we detected two nuclear genotype clusters, each being highly significantly associated to one mitochondrial lineage. This association however had exceptions: a small number of individuals in the contact zone showed intermediate genotypes (4%) or cytonuclear disequilibrium (12%). Both mitochondrial lineage and nuclear cluster were statistically significant predictors for the shell shape indicating morphological divergence. Nevertheless, the lineage morphospaces largely overlapped (low posterior classification success rate of 69% and 78%, respectively): the two lineages are truly cryptic.
Conclusions: The integrative approach using multiple lines of evidence supported the hypothesis that the investigated Trochulus lineages are reproductively isolated species. In the small contact area, however, the lineages hybridise to a limited extent. This detection of a hybrid zone adds an instance to the rare reported cases of hybridisation in land snails.
Zwischen Bilateralismus und Multilateralismus : die Zentralasienpolitik der Volksrepublik China

Die Außenpolitik der Volksrepublik China (VRC) ist im Wandel begriffen. Nur ein – wenngleich wesentlicher – Ausdruck dessen ist die erst seit wenigen Jahren zu beobachtende Entwicklung, dass sich die VRC vermehrt multilateralen Institutionen zuwendet und diese mittlerweile zum Teil gar selbst initiiert. Dieser Trend stellt eine radikale Kehrtwende gegenüber dem dar, was die VRC seit ihrer Gründung 1949 noch bis weit in die 1990er-Jahre hinein mit Multilateralismus (ML) assoziiert hat – nämlich Mechanismen zur Strafung und Einengung der VRC. Doch welche praktische Bedeutung kommt multilateralen Wirkungsweisen innerhalb der chinesischen Außenpolitik in jüngster Zeit tatsächlich zu? Die vorliegende Magisterarbeit beschäftigt sich mit dieser Fragestellung, indem sie zu analysieren sucht, welche Rolle multilaterale Strukturen – im Gegensatz zu bilateralen – bei der Implementierung chinesischer außenpolitischer Interessen spielen. Dies geschieht anhand eines einzigartigen Fallbeispiels, und zwar der chinesischen Zentralasienpolitik, innerhalb der wiederum die Shanghai Cooperation Organization (SCO) eine herausragende Stellung einnimmt. Diese Organisation ist das einzige multilaterale Forum, an dem die VRC von Beginn an als der maßgebende Akteur im Hinblick auf ideelle, konzeptionelle und inhaltliche Belange teilgenommen hat. Entsprechend ist davon auszugehen, dass die Untersuchung ein Höchstmaß an Einblicken in den tatsächlich intendierten Zweck des chinesischen ML zu liefern vermag. Die Analyse zeigt, dass multilaterale Vorgehensweisen in quantitativer Hinsicht seit Gründung der SCO im Jahre 2001 stark zugenommen haben, und das auf allen vier identifizierten Hauptinteressensgebieten der VRC in Zentralasien: Sicherheit, Wirtschaft, Energie und Geopolitik. Mit Blick auf die qualitative Komponente ist indessen zu konstatieren, dass dem ML, den China in der Region praktiziert, einige negativ behaftete Charakteristika zu eigen sind. Die Attribute „flexibel“, „selektiv“, „flach“ und „instrumentalisiert“, die derzeit weitgehend als prägend für den chinesischen ML in seiner Gesamtheit angesehen werden, besitzen auch – und gerade – in Zentralasien ihre Gültigkeit. Insgesamt fällt auf, dass die VRC multilaterale Strategien nur dann verfolgt, wenn der kurzfristige Eigennutzen über den bilateraler Mechanismen hinausgeht. Und auch dann hört der Rückgriff auf ML dort auf, wo der eigene Gewinn ein Maximum erreicht. Somit bleiben die chinesischen Machthaber den Praxisbeweis einer integrativen Funktion von ML noch schuldig.
Medium range hydrological forecasts in mesoscale catchments are only possible with the use of hydrological models driven by meteorological forecasts, which in particular contribute quantitative precipitation forecasts (QPF). QPFs are accompanied by large uncertainties, especially for longer lead times, which are propagated within the hydrometeorological model system. To deal with this limitation of predictability, a probabilistic forecasting system is tested, which is based on a hydrological-meteorological ensemble prediction system. The meteorological component of the system is the operational limited-area ensemble prediction system COSMO-LEPS that downscales the global ECMWF ensemble to a horizontal resolution of 10 km, while the hydrological component is based on the semi-distributed hydrological model PREVAH with a spatial resolution of 500 m.
Earlier studies have mostly addressed the potential benefits of hydrometeorological ensemble systems in short case studies. Here we present an analysis of hydrological ensemble hindcasts for two years (2005 and 2006). It is shown that the ensemble covers the uncertainty during different weather situations with appropriate spread. The ensemble also shows advantages over a corresponding deterministic forecast, even under consideration of an artificial spread.
CD8 T cells are recognized key players in control of persistent virus infections, but increasing evidence suggests that assistance from other immune mediators is also needed. Here, we investigated whether specific antibody responses contribute to control of lymphocytic choriomeningitis virus (LCMV), a prototypic mouse model of systemic persistent infection. Mice expressing transgenic B cell receptors of LCMV-unrelated specificity, and mice unable to produce soluble immunoglobulin M (IgM) exhibited protracted viremia or failed to resolve LCMV. Virus control depended on immunoglobulin class switch, but neither on complement cascades nor on Fc receptor gamma chain or Fc gamma receptor IIB. Cessation of viremia concurred with the emergence of viral envelope-specific antibodies, rather than with neutralizing serum activity, and even early nonneutralizing IgM impeded viral persistence. This important role for virus-specific antibodies may be similarly underappreciated in other primarily T cell–controlled infections such as HIV and hepatitis C virus, and we suggest this contribution of antibodies be given consideration in future strategies for vaccination and immunotherapy.
Introduction Chemokines and their receptors control immune cell migration during infections as well as in autoimmune responses. A 32 bp deletion in the gene of the chemokine receptor CCR5 confers protection against HIV infection, but has also been reported to decrease susceptibility to rheumatoid arthritis (RA). The influence of this deletion variant on the clinical course of this autoimmune disease was investigated. Methods Genotyping for CCR5d32 was performed by PCR and subsequent electrophoretic fragment length determination. For the clinical analysis, the following extra-articular manifestations of RA were documented by the rheumatologist following the patient: presence of rheumatoid nodules, major organ vasculitis, pulmonary fibrosis, serositis or a Raynaud's syndrome. All documented CRP levels were analyzed retrospectively, and the last available hand and feet radiographs were analyzed with regards to the presence or absence of erosive disease. Results Analysis of the CCR5 polymorphism in 503 RA patients and in 459 age-matched healthy controls revealed a significantly decreased disease susceptibility for carriers of the CCR5d32 deletion (Odds ratio 0.67, P = 0.0437). Within the RA patient cohort, CCR5d32 was significantly less frequent in patients with extra-articular manifestations compared with those with limited, articular disease (13.2% versus 22.8%, P = 0.0374). In addition, the deletion was associated with significantly lower average CRP levels over time (median 8.85 vs. median 14.1, P = 0.0041) and had a protective effect against the development of erosive disease (OR = 0.40, P = 0.0047). Intriguingly, homozygosity for the RA associated DNASE2 -1066 G allele had an additive effect on the disease susceptibility conferred by the wt allele of CCR5 (OR = 2.24, P = 0.0051 for carrier of both RA associated alleles) Conclusions The presence of CCR5d32 significantly influenced disease susceptibility to and clinical course of RA in a German study population. The protective effect of this deletion, which has been described to lead to a decreased receptor expression in heterozygous patients, underlines the importance of chemokines in the pathogenesis of RA.
Mammalian retinae have rod photoreceptors for night vision and cone photoreceptors for daylight and colour vision. For colour discrimination, most mammals possess two cone populations with two visual pigments (opsins) that have absorption maxima at short wavelengths (blue or ultraviolet light) and long wavelengths (green or red light). Microchiropteran bats, which use echolocation to navigate and forage in complete darkness, have long been considered to have pure rod retinae. Here we use opsin immunohistochemistry to show that two phyllostomid microbats, Glossophaga soricina and Carollia perspicillata, possess a significant population of cones and express two cone opsins, a shortwave-sensitive (S) opsin and a longwave-sensitive (L) opsin. A substantial population of cones expresses S opsin exclusively, whereas the other cones mostly coexpress L and S opsin. S opsin gene analysis suggests ultraviolet (UV, wavelengths <400 nm) sensitivity, and corneal electroretinogram recordings reveal an elevated sensitivity to UV light which is mediated by an S cone visual pigment. Therefore bats have retained the ancestral UV tuning of the S cone pigment. We conclude that bats have the prerequisite for daylight vision, dichromatic colour vision, and UV vision. For bats, the UV-sensitive cones may be advantageous for visual orientation at twilight, predator avoidance, and detection of UV-reflecting flowers for those that feed on nectar.
